Objective To observe the therapeutic effect of intragastric administration of empagliflozin on ventricular diastolic dysfunction in hypertensive rats and to explore its possible mechanism. Methods Among 25 male SD rats, 5 underwent abdominal aortic dissection and then the abdominal cavity was closed (sham operation group), the other 20 underwent abdominal aortic coarctation to make hypertensive ventricular diastolic dysfunction models (blood pressure was measured at the 8th weekend after surgery, and echocardiography was performed to detect the ventricular structure and function indexes of rats to evaluate whether the model was successful, and 15 were successfully made). Fifteen hypertensive ventricular diastolic dysfunction model rats were randomly divided into three groups groups A, B, and C (with 5 rats in each), and then underwent echocardiography again (before administration). Rats in the group A were orally administered 10 mg/ (kg. d) of empagliflozin for 4 consecutive weeks (after administration), rats in the group B were treated with normal saline once a day for 4 consecutive weeks, and rats in the group C were not treated. The rats in group A and group B underwent echocardiography immediately after the administration of medication, while the rats in the group C were euthanized and the myocardial tissues were separated. The inflammatory factors, oxidative stress indicators, and collagen fiber indicators in the myocardial tissues of rats in the three groups were detected by ELISA and Western blotting, respectively. Results Compared with the group B, rats of the group A had shorter left ventricular isovolumic relaxation time (IVRT), decreased left ventricular mass (LVM), and decreased left ventricular end systolic diameter (LVIDs) (all P<0. 05). The IVRT of rats in the group A was short, LVM and LVIDs decreased in comparison with those before administration (all P< 0. 05). Compared with the group B, the content of collagen type Ⅰ, collagen type Ⅲ, malondialdehyde (MDA) and transforming growth factor-β (TGF-β) was lower, the content of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) was higher, and the relative expression of NLRP3 was lower in the group A after administration (all P<0. 05). Conclusion Intragastric administration of empagliflozin can alleviate hypertensive ventricular diastolic dysfunction in rats, and the mechanism may be that empagliflozin reduces the expression of MDA, TGF-β, collagen type Ⅰ, collagen type Ⅲ and NLRP3 in myocardial tissues, and promotes the expression of SOD and GSH-PX. Aiming point optimization refers to the selection of the aiming point of a missile so that the strike effect on the target is at or near the best. As an example, a modified genetic algorithm is used to study the aiming point optimization problem of a multi-bullet missile striking the same target with different damage effects. Combined with the target vulnerability of the substation, suitable damage indicators and corresponding types of missile weapons are selected to establish the optimal aiming point solution model. In order to solve the defects of traditional genetic algorithm, an experience pool is established to store the dominant individuals to avoid the loss of dominant individuals and to retain the individuals with different genotypes to enhance the population diversity and avoid premature convergence of the algorithm. After calculations, the algorithm can effectively solve the problem of multi-bullet hybrid aiming point optimization, and the overall optimization result is better than the traditional algorithm, and has good convergence, which is of high application value. Objective To investigate the protective effects of empagliflozin, a highly selective SGLT-2 inhibitor, on cardiac function in mice with acute myocardial infarction (AMI) and to explore its mechanism. Methods Thirty-four 6- week-old male C57BL/6J mice were randomly divided into the sham group (n=11), model group (n=11) and treatment group (n=12) . AMI was induced in the model group and treatment group by left anterior descending (LAD) artery ligation. The mice in the sham group underwent the same procedure except that the LAD was not ligated. The mice in the treatment group were given empagliflozin (10 mg/kg/day, oral gavage) at 4 h after MI surgery for 7 days. The mice in the sham group and model group were given 0. 5% hydroxyethyl cellulose. The left ventricular internal dimension in diastole (LVIDd), left ventricular internal dimension in systole (LVIDs), fractional shortening (FS) and ejection fraction (EF) were measured by echocardiography in all three groups. TUNEL staining was used to measure cardiac apoptosis. Blood samples were collected on day 7 after AMI to measure serum levels of NLRP3, ASC, Caspase-1, IL-1β, IL-18 and CRP by ELISA. The expression levels of NLRP3, ASC, Caspase-1, cleaved Caspase-1, IL-1β, and IL-18 were analyzed by Western blotting. Results There was no difference in LVIDd among these groups (P>0. 05). Compared with the sham group, the treatment group and model group had increased LVIDs (both P<0. 05). Compared with the sham group, the treatment group and the model group had decreased EF and FS, increased serum levels of NLRP3, IL-1β, IL-18, CRP, and up-regulated protein levels of NLRP3, ASC, cleaved Caspase-1, IL-1β, and IL-18 in the cardiac tissues of mice, with much more significant changes in the model group (all P<0. 05). The mice in the model group had significantly higher cardiac apoptosis rate than the sham group and the treatment group (both P<0. 05), while there was no statistical difference between the sham group and the treatment group (P>0. 05). There was no significant difference in the serum levels of Caspase-1 between the sham group and the model group (P>0. 05). Similarly, no statistical difference was found in the protein level of Caspase-1 in the cardiac tissues of mice among all three groups (P>0. 05). Conclusion Empagliflozin improves cardiac function and reduces cardiomyocyte apoptosis in AMI mice, and its mechanism may be related to the inhibition of the NLRP3/ASC/Caspase-1 signaling pathway and reduction of the inflammatory response. 19 (34) orthogonal test was used to investigate the protective effect of trehalose, PEG2000 and mannitol on freeze — dried powder of Haudinaria manillensis crude extract wider different time and temperature conditions. The results showed that among the three protective agents, PEG2000 had the strongest protective effect and trehalose was the weakest. When the average activity within 24 h was used as the index, the optimal mass fraction combination of trehalose, PEG2000 and mannitol was 4%, 4% mid 2%. Taking the average activity after 15 d at ( —18 ± 2 ) ºC and the average activity after 15 d at (27 ± 3 ) °C as the indexes, the optimal mass fraction combi-nation of those agents was 2%, 4% and 2%. The intestinal microbial communities and abundance of Hirudo nipponia after starvation and refeed ing studied with high-throughput sequencing technology. The results showed that 562 384 effective sequences were obtained from 9 samples which could be clustered to 769 operational taxonomic units (OTUS). The number of OTUS in the starvation group, 7 days after refeeding group and 21 days after refeeding group was 444, 664 and 440, respectively, among which 289 OTUs were shared. The intestinal microbial diversity index of Hirudo nipponia increased after 7 days of refeeding, but decreased after 21 days. The intestinal tract of Hirudo nipponia had three dominant phylums, which were Bacteroidetes, Proteobacteria and Fimicutes. After refeeding, the relative abun dance of Bacteroidetes decreased at first and then increased, while the variation trend of relative abundance of Pro teobacteria and Firmicides were opposite. After 7 days of refeeding, there were 8 strains with significantly increased relative abundance, all of which belonged to Proteobacteria. Among them, Aeromonas and Yokenella had greater a bundance. These results suggested that there was a close connection between ingesting blood and bacterial diversity index and species composition in the Hirudo nipponia intestine.
