Abstract

The department of Medical Informatics of the University Hospital Münster and the Gesakon GmbH (an university offspring) initiated the cooperative development of an electronic health record (EHR) called "akteonline.de" in 2000. From 2001 onwards several clinics of the university hospital have already offered this EHR (within pilot projects) as an additional service to selected subsets of their patients. Based on the experiences of those pilot projects the system architecture and the basic data model underwent several evolutionary enhancements, e.g. implementations of electronic interfaces to other clinical systems (considering for example data interchange methods like the Clinical Document Architecture - standardized within the HL7 group - and also interfacing architectures of German GP systems, such as VCS and D2D). "akteonline.de" in its current structure supports patients as well as health care professionals and aims at providing a collaborative health information system which perfectly supports the clinical workflow even across institutional boundaries and including the patient himself. Since such an EHR needs to strictly fulfill high data security and data protection requirements, a complex authorization and access control component has been included. Furthermore the EHR data are encrypted within the database itself and during their transfer across the internet.

271. Assigning dispatching rules using a genetic algorithm to solve a hybrid flow shop scheduling problem

Abstract

The paper shows how assigning different predefined dispatching rules for a predefined number of times at any points in time using a genetic algorithm can solve a hybrid flow shop scheduling problem with sequence-dependent setup times, using the example of a company producing printed circuit boards. The paper describes the implementation of the genetic algorithm, analyses the results for four different data sets and compares them with the results of applying standard dispatching rules. Using a genetic algorithm to assign dispatching rules achieves better solutions than simple dispatching rules often used in industrial practice. The genetic algorithm already achieves good solutions after a few seconds up to a few minutes, which means that this approach can be used for real-time decision making in a cyber-physical system in the age of Industry 4.0.

Maintaining focus during time-lapse experiments remains an important challenge in optical microscopy. In this paper we present an automated focusing system for fluorescence microscopy that can be used in time-lapse experiments. We provide software for flexibly operating the autofocus system and a complete alignment guide. The system is evaluated using a time-lapse experiment with temperature variations on Tetraspeck beads.

Abstract

Preisdifferenzierung, ob räumlich, zeitlich, quantitativ oder qualitativ, gehört seit Jahrzehnten zum Alltag. Durch die zunehmende Digitalisierung und die damit einhergehende hohe Verfügbarkeit von Informationen, insbesondere von Kundendaten, eröffnen sich neue Möglichkeiten, Preisdifferenzierung dynamisch und individuell zu gestalten. Während im Onlinehandel der Preis oft dynamisch differenziert wird, ist dies im stationären Detailhandel zurzeit selten der Fall. Ein Grund dafür ist, dass die Akzeptanz einer dynamisch individualisierten Preisdifferenzierung und deren Einflussfaktoren auf die Kundenloyalität weitgehend unerforscht sind. In der vorliegenden Arbeit wird diese Fragestellung mittels quantitativer Methoden untersucht. Im Rahmen der empirischen Untersuchung zeigt sich, dass Transparenz und Nachvollziehbarkeit bezüglich Preisgestaltung wichtige Parameter zur Erhöhung der Akzeptanz von dynamisch individualisierten Ansätzen sind. Allerdings wurde auch eine Intensions-Verhaltens-Lücke festgestellt: Ein beachtlicher Anteil der Kunden, die Kundenprogramme aus Gründen des Datenschutzes (Stichwort «gläserne Kunden») ablehnen, sind dennoch auf sozialen Medien aktiv. Die Ergebnisse zeigen weiter, dass Kunden zwischen 18 und 35 Jahren, die sogenannten Millennials, deutlich empfänglicher für individualisierte Preise sind als Kunden fortgeschrittenen Alters. Allerdings ist die Teilnahmerate an Kundenloyalitäts-Programmen bei Millennials deutlich tiefer als bei älteren Kunden, was die Notwendigkeit einer Anpassung traditioneller Kundenbindungsprogramme an die veränderten Kundenbedürfnisse deutlich macht. Die zunehmende Individualisierung und Dynamisierung erfordern zusätzlich eine flexible Systemarchitektur, die in der Lage ist, sowohl interne Positionssysteme als auch Batch- und Real-Time-Analysesysteme in ein Gesamtsystem zu integrieren.

KMT2A rearrangements are associated with a poor clinical outcome in infant, pediatric, and adult acute lymphoblastic and myeloid leukemia. Here, we present a protocol to reconstruct chromosomal translocations with different partner genes of KMT2A in vitro. We describe steps for patient-specific single guide RNA (sgRNA) design, optimized sgRNA in vitrotranscription, detailed purification of hematopoietic stem and progenitor cells (HSPCs) from umbilical cord blood (UCB), and CRISPR-Cas9 editing of the test cell line K562 as well as UCB HSPCs. The provided methodology is donor independent.

Abstract

A new coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has recently emerged to cause a human pandemic. Although molecular diagnostic tests were rapidly developed, serologic assays are still lacking, yet urgently needed. Validated serologic assays are needed for contact tracing, identifying the viral reservoir, and epidemiologic studies. We developed serologic assays for detection of SARS-CoV-2 neutralizing, spike protein-specific, and nucleocapsid-specific antibodies. Using serum samples from patients with PCR-confirmed SARS-CoV-2 infections, other coronaviruses, or other respiratory pathogenic infections, we validated and tested various antigens in different in-house and commercial ELISAs. We demonstrated that most PCR-confirmed SARS-CoV-2-infected persons seroconverted by 2 weeks after disease onset. We found that commercial S1 IgG or IgA ELISAs were of lower specificity, and sensitivity varied between the 2 assays; the IgA ELISA showed higher sensitivity. Overall, the validated assays described can be instrumental for detection of SARS-CoV-2-specific antibodies for diagnostic, seroepidemiologic, and vaccine evaluation studies.

Abstract

In recent years, the advancing digitization and internationalization of business processes led to increasing inter-organizational collaboration. In such collaborative processes, different organizations work together towards a single objective. Usually, subprocesses carried out by one collaborator are beyond the domain of influence of all other collaborators. This leads to uncertainty regarding the execution of the collaborative business process. If collaborators still want to engage in the process, trust is needed. Several studies identified blockchain and distributed ledger technologies as a promising tool to enhance trust in business processes. Therefore, this paper proposes and analyzes a taxonomy of blockchain-based trust design patterns from a process-centric perspective. Process engineers can utilize the taxonomy as an overview of how the blockchain technology can enhance trust in collaborative processes.

Abstract

Recently, business processes started to become more collaborative by nature. Prominent examples of collaborative processes can be found in domains such as supply chain management, e-commerce, and in the realm of online multisided platforms. These types of collaborations are characterized by the fact that a collaborator's subprocess execution is usually beyond the control of other collaborators. This property leads to the occurrence of uncertainty in the processes. If uncertainty is present in a collaboration, then trust is needed. This article proposes a method to identify uncertainty within collaborative business processes and to add a trust layer to their modeled representation. The trust layer allows process collaborators to make implicit uncertainties explicit within a process model. The proposed methodology is not meant to be applied not only in a postprocessing fashion, but also for the specification of new collaborative processes with trust-aware design principles.

Abstract

Middle East respiratory syndrome coronavirus (MERS-CoV) is enzootic in dromedary camels across the Middle East and Africa. Virus-induced pneumonia in humans results from animal contact, with a potential for limited onward transmission. Phenotypic changes have been suspected after a novel recombinant clade (lineage 5) caused large nosocomial outbreaks in Saudi Arabia and South Korea in 2016. However, there has been no functional assessment. Here we perform a comprehensive in vitro and ex vivo comparison of viruses from parental and recombinant virus lineages (lineage 3, n = 7; lineage 4, n = 8; lineage 5, n = 9 viruses) from Saudi Arabia, isolated immediately before and after the shift toward lineage 5. Replication of lineage 5 viruses is significantly increased. Transcriptional profiling finds reduced induction of immune genes IFNB1, CCL5, and IFNL1 in lung cells infected with lineage 5 strains. Phenotypic differences may be determined by IFN antagonism based on experiments using IFN receptor knock out and signaling inhibition. Additionally, lineage 5 is more resilient against IFN pre-treatment of Calu-3 cells (ca. 10-fold difference in replication). This phenotypic change associated with lineage 5 has remained undiscovered by viral sequence surveillance, but may be a relevant indicator of pandemic potential. MERS-CoV is enzootic in dromedary camels, can spread to humans but undergoes limited onward transmission. Here, Schroeder et al. compare clinical isolates of MERS-CoV in vitro and show that the predominantly circulating recombinant lineage 5 possess a fitness advantage over parental lineage 3 and 4 due to reduced activation of innate immune signaling. Abstract

Super-resolved structured illumination microscopy (SR-SIM) is among the fastest fluorescence microscopy techniques capable of surpassing the optical diffraction limit. Current custom-build instruments are able to deliver two-fold resolution enhancement with high acquisition speed. SR-SIM is usually a two-step process, with raw-data acquisition and subsequent, time-consuming post-processing for image reconstruction. In contrast, wide-field and (multi-spot) confocal techniques produce high-resolution images instantly. Such immediacy is also possible with SR-SIM, by tight integration of a video-rate capable SIM with fast reconstruction software. Here we present instant SR-SIM by VIGOR (Video-rate Immediate GPU-accelerated Open-Source Reconstruction). We demonstrate multi-color SR-SIM at video frame-rates, with less than 250 ms delay between measurement and reconstructed image display. This is achieved by modifying and extending high-speed SR-SIM image acquisition with a new, GPU-enhanced, network-enabled image-reconstruction software. We demonstrate high-speed surveying of biological samples in multiple colors and live imaging of moving mitochondria as an example of intracellular dynamics., Sequential acquisition and image reconstruction in super-resolved structured illumination microscopy (SR-SIM) is time-consuming. Here the authors optimise both acquisition and reconstruction software to achieve multicolour SR-SIM at video frame-rates with reconstructed images displaying with only milliseconds delay during the experiment.
