Purpose: To predict the prognosis of craniopharyngioma in children by optical coherence tomography angiography (OCTA).Methods: We evaluated if the relationship between preoperative OCTA of the choroidal capillary density (CCD) and visual outcome continued over long-term visual recovery in 38 patients undergoing craniopharyngioma resection. Patients were evaluated 3 times: 1 week before surgery (Visit1), followed-up at 6–10 weeks (Visit2), and 9–15 months (Visit3) after surgery.Results: In total 38 patients (70 eyes) with craniopharyngiomas, which included 20 (52.6%) boys and 18 (47.4%)girls, the mean age was 11.8 ± 2.7 years (range: 6–18 years). The age (p = 0.71), gender (p = 1.00), mean refractive error (p = 0.55), and axial length (p = 0.23) of 38 normal volunteers (76 eyes) were matched. After surgery, the cross-compression of patients was relieved. The average visual acuity change in the normal CCD group was 0.07 ± 0.02; the average visual acuity change in the low CCD group was 0.01 ± 0.01, p < 0.001. Preoperative CCD value is related to the preoperative BCVA (p < 0.001), and the visual function after the long-term follow-up (9–15 months) (p < 0.001). The prognosis of CCD has the same trend as the BCVA. Further correlation analysis shows that CCD and BCVA are significantly correlated (r = 0.878; p < 0.001). The dynamic analysis of the soft landing of the lunar probe is very important to the design of the probe. The initial movement and attitude parameters of the probe during landing have a direct influence on the landing impact. In order to investigate the lunar probe soft-landing dynamic impact by different initial horizontal velocities, pitch angles, and inclinations of the lunar slope, an inertial force-based 7-DOF soft-landing dynamic model is applied under two conditions: the upward and downward slope landing surfaces. The impact on the dynamic characteristics of soft landing is analyzed in terms of body displacement, body overload, and the forces of the primary and secondary buffer struts due to the change of initial horizontal velocity and initial pitch angle of the probe. The result shows that, in 2-2 landing mode, the stress conditions on the primary and secondary struts are obviously impacted by initial horizontal velocity, and the initial pitch angle affects the body overload and the loading state of the secondary buffer strut. The body overload and landing impact could be significantly mitigated if the lunar probe’s horizontal landing speed is limited within 1 m/s, the pitch angle is limited within 12°, and the landing is along the uphill terrain with the inclination of the lunar slope less and equal to 9°. The analysis can directly determine the range of the horizontal speed and pitch attitude angle to ensure the safety of landing, and provide a reference for the reasonable control design of the lander’s horizontal speed and pitch attitude.

Purpose. To investigate the changes in thromboelastography (TEG) in patients with dyslipidemia to study its effect on the blood coagulation status. Methods. 131 patients hospitalized in Fujian Provincial Jinshan Hospital from January 2018 to December 2020 were selected, and 64 cases in the hyperlipidemia (HL) group and 67 cases in the non-HL group were set according to whether their blood lipids were abnormal. By measuring the changes of each parameter of TEG in patients, the relevant parameters R value, K value, α angle, and MA value were calculated. And routine blood coagulation (PT, APTT, INR, FIB, and TT) and routine blood (platelet count) tests were performed on all study subjects to analyze the changes of each index of the coagulation function and each parameter of TED in both groups and explore the clinical value of TEG on HL diseases. Results. Compared with the non-HL group, R and K values decreased, and angle and MA values increased in the HL group (P0.05). Abstract. Platelets play vital roles in hemostasis, inflammation, and vascular biology. Platelets are also active participants in the immune responses. As vertebrates, zebrafish have a highly conserved hematopoietic system in the developmental, cellular, functional, biochemical, and genetic levels with mammals. Thrombocytes in zebrafish are functional homologs of mammalian platelets. Here, we summarized thrombocyte development, function, and related research techniques in zebrafish, and reviewed available zebrafish models of platelet-associated disorders, including congenital amegakaryocytic thrombocytopenia, inherited thrombocytopenia, essential thrombocythemia, and blood coagulation disorders such as gray platelet syndrome. These elegant zebrafish models and methods are crucial for understanding the molecular and genetic mechanisms of thrombocyte development and function, and provide deep insights into related human disease pathophysiology and drug development.

The T cell co-stimulatory molecule OX40 and its cognate ligand OX40L have attracted broad research interest as a therapeutic target in T cell-mediated diseases. Accumulating preclinical evidence highlights the therapeutic efficacy of both agonist and blockade of the OX40–OX40L interaction. Despite this progress, many questions about the immuno-modulator roles of OX40 on T cell function remain unanswered. In this review we summarize the impact of the OX40–OX40L interaction on T cell subsets, including Th1, Th2, Th9, Th17, Th22, Treg, Tfh, and CD8+ T cells, to gain a comprehensive understanding of anti-OX40 mAb-based therapies. The potential therapeutic application of the OX40–OX40L interaction in autoimmunity diseases and cancer immunotherapy are further discussed; OX40–OX40L blockade may ameliorate autoantigen-specific T cell responses and reduce immune activity in autoimmunity diseases. We also explore the rationale of targeting OX40–OX40L interactions in cancer immunotherapy. Ligation of OX40 with targeted agonist anti-OX40 mAbs conveys activating signals to T cells. When combined with other therapeutic treatments, such as anti-PD-1 or anti-CTLA-4 blockade, cytokines, chemotherapy, or radiotherapy, the anti-tumor activity of agonist anti-OX40 treatment will be further enhanced. Ketamine exposure can lead to selective neuroapoptosis in the developing brain. p66ShcA, the cellular adapter protein expressed selectively in immature neurons, is a known pro-apoptotic molecule that triggers neuroapoptosis when activated. Sprague-Dawley rats at postnatal day 7 were subcutaneously injected in the neck with ketamine 20 mg/kg, six times at 2-hour intervals. At 0, 1, 3, and 6 hours after final injection, western blot assay was used to detect the expression of cleaved caspase-3, p66ShcA, and phosphorylated p66ShcA. We found that the expression of activated p66ShcA and caspase-3 increased after ketamine exposure and peaked at 3 hours. The same procedure was performed on a different group of rats. At the age of 4 weeks, spatial learning and memory abilities were tested with the Morris water maze. Latency to find the hidden platform for these rats was longer than it was for control rats, although the residence time in the target quadrant was similar. These findings indicate that ketamine exposure resulted in p66ShcA being activated in the course of an apoptotic cascade during the neonatal period. This may have contributed to the deficit in spatial learning and memory that persisted into adulthood. The plant microbiome is a key determinant of health and productivity. However, it is still difficult to understand the structural composition of the bacterial and fungal microbiomes of diseased and healthy plants, especially the spatial dynamics and phylogenies of endophytic and rhizosphere microbial communities. We studied the differentiation and variability in the rhizosphere and endosphere microbiomes of healthy and diseased cotton from north and south of the Tianshan Mountains using the methods of PCR-based high-throughput sequencing and real-time quantitative PCR. The endophytic and rhizosphere bacterial abundances in the diseased plants were greater than those of healthy plants. The numbers of endophytic and rhizosphere fungi associated with diseased plants were greater than those associated healthy plants (p < 0.05). Endophytic and rhizosphere bacteria did not share common OTUs. The dominant rhizosphere bacteria were Proteobacteria (29.70%), Acidobacteria (23.14%), Gemmatimonadetes (15.17%), Actinobacteria (8.31%), Chloroflexi (7.99%), and Bacteroidetes (5.15%). The dominant rhizosphere fungi were Ascomycota (83.52%), Mortierellomycota (7.67%), Basidiomycota (2.13%), Chytridiomycota (0.39%), and Olpidiomycota (0.08%). The distribution of dominant bacteria in different cotton rhizosphere soils and roots differed, with the dominant bacteria Pseudomonas (15.54%) and Pantoea (9.19%), and the dominant fungi Alternaria (16.15%) and Cephalotrichum (9.10%) being present in the greatest numbers. At sampling points in different ecological regions, the total numbers of cotton endophytic and rhizosphere microbiome OTUs from southern to northern Xinjiang showed an increasing trend. There were significant differences in the composition and diversity of rhizosphere microbes and endophytes during the entire cotton growth period and in representative ecological regions (p < 0.01), whereas rhizosphere microbes and endophytes showed no significant differences among the four growth periods and in representative ecological regions. RB41, H16, Nitrospira, and Sphingomonas play important roles in the microbial ecology of cotton rhizosphere soil. Pseudomonas accounted for a large proportion of the microbes in the cotton rhizosphere soil. This study provides an in-depth understanding of the complex microbial composition and diversity associated with cotton north and south of the Tianshan Mountains.

A high-quality on-water landscape can improve the quality of cities and promote tourism development. However, current research on urban rivers has primarily focused on the riverside perspective, whereas few studies investigated the visual quality from an on-water perspective or conducted quantitative evaluations. This paper established a quantitative landscape index system by using a deep learning based semantic segmentation model to analyze human visual perception. A random forest model was used to analyze the nonlinear correlation between quantitative indicators and public scores, and an analysis and prediction model suitable for assessing the visual quality of an urban river on-water landscape was developed. This model provided high prediction accuracy and could rank the importance of the impact factors. The urban construction level, destructive index, hard revetment visibility, and green visibility index substantially affected the visual quality of the on-water landscape. The green visibility index was positively correlated, and the other three factors were negatively correlated with the visual quality. This model represents an intelligent approach for evaluating the visual perception and visual quality of the on-water landscape, enabling researchers and policymakers to analyze waterscapes from a new perspective and with high efficiency.

Bmp2 is essential for dentin development and formation. Bmp2 conditional knock-out (KO) mice display a similar tooth phenotype of dentinogenesis imperfecta (DGI). To elucidate a foundation for subsequent functional studies of cross talk between mRNAs and lncRNAs in Bmp2-mediated dentinogenesis, we investigated the profiling of lncRNAs and mRNAs using immortalized mouse dental Bmp2 flox/flox (iBmp2fx/fx) and Bmp2 knock-out (iBmp2ko/ko) papilla cells. RNA sequencing was implemented to study the expression of the lncRNAs and mRNAs. Quantitative real-time PCR (RT-qPCR) was used to validate expressions of lncRNAs and mRNAs. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases were used to predict functions of differentially expressed genes (DEGs). Protein–protein interaction (PPI) and lncRNA–mRNA co-expression network were analyzed by using bioinformatics methods. As a result, a total of 22 differentially expressed lncRNAs (16 downregulated vs 6 upregulated) and 227 differentially expressed mRNAs (133 downregulated vs. 94 upregulated) were identified in the iBmp2ko/ko cells compared with those of the iBmp2fx/fx cells. RT-qPCR results showed significantly differential expressions of several lncRNAs and mRNAs which were consistent with the RNA-seq data. GO and KEGG analyses showed differentially expressed genes were closely related to cell differentiation, transcriptional regulation, and developmentally relevant signaling pathways. Moreover, network-based bioinformatics analysis depicted the co-expression network between lncRNAs and mRNAs regulated by Bmp2 in mouse dental papilla cells and symmetrically analyzed the effect of Bmp2 during dentinogenesis via coding and non-coding RNA signaling.

Urban renewal urgently needs to find a new media tool to extensively promote public participation. Professionals also need strong and powerful communication tools for the public. Mass media has the ability to influence human perceptions and behaviors, but its role has been overlooked. This study aims to arouse professionals’ attention to mass media and promote interdisciplinary cooperation through empirical evidence. By observing the performance of participatory urban gardening projects in Chinese newspapers, we highlight the positive effect of mass media on participatory landscape revitalization. We selected two projects in China as samples, collected newspaper reports on them during 2017–2021, and analyzed the textual framing and report communication based on communication theory. According to the result, mass media has four positive effects that not only affect the public but also contribute to participatory landscape revitalization development. Based on the results, we discuss the consistence of views of the mass media and landscape architecture. This study suggests that landscape architecture needs to actively collaborate with public media to better leverage the role of landscape in sustainable urban transformation.

In recent years, malignant breast cancer metastasis has caused a great increase in mortality. Research on the genetic and molecular mechanisms of malignant breast cancer has continued to deepen, and targeted therapy has become the general trend. Among them, competing endogenous RNA (ceRNA)-related molecules have received much attention. Homeobox transcript antisense RNA (HOTAIR) has been reported to function extensively as a ceRNA in breast cancer. Notably, miR-203 and Caveolin 1 (CAV1) have also been found to play a role in breast cancer. However, the relationship between the three remains unclear. In this study, we present a new mechanic through bioinformatics tool and basic experiments: the HOTAIR/miR-203/CAV1 axis, which complemented the role network of HOTAIR as a ceRNA, thus, it will provide a novel potential idea for breast cancer research and therapy.

Platelets play an essential role in thrombosis and hemostasis. Abnormal hemostasis can cause spontaneous or severe post-traumatic bleeding. Bernard-Soulier syndrome (BSS) is a rare inherited bleeding disorder caused by a complete quantitative deficiency in the GPIb-IX-V complex. Multiple mutations in GP9 lead to the clinical manifestations of BSS. Understanding the roles and underlying mechanisms of GP9 in thrombopoiesis and establishing a proper animal model of BSS would be valuable to understand the disease pathogenesis and to improve its medical management. Here, by using CRISPR-Cas9 technology, we created a zebrafish gp9SMU15 mutant to model human BSS. Disruption of zebrafish gp9 led to thrombocytopenia and a pronounced bleeding tendency, as well as an abnormal expansion of progenitor cells. The gp9SMU15 zebrafish can be used as a BSS animal model as the roles of GP9 in thrombocytopoiesis are highly conserved from zebrafish to mammals. Utilizing the BSS model, we verified the clinical GP9 mutations by in vivo functional assay and tested clinical drugs for their ability to increase platelets. Thus, the inherited BSS zebrafish model could be of benefit for in vivo verification of patient-derived GP9 variants of uncertain significance and for the development of potential therapeutic strategies for BSS.

The bone microenvironment is crucial for the growth and development of different types of osteocytes. Small extracellular vesicles (sEVs) secreted by bone mesenchymal stem cells are delivered to target cells where their contents regulate biological functions. Here, we evaluated the osteogenic effects and mechanism of sEVs derived from Plastrum testudinis-preconditioned bone mesenchymal stem cells (PT-sEV). The osteogenic effects of PT-sEV were evaluated by the differentiation of osteoblasts and the alternation of bone quality and quantity in ovariectomized rats. The specific mechanism was explored by high-throughput sequencing and verified by transfection with the corresponding miRNA mimic and inhibitor. RNA-sequence identified a unique enrichment of a set of miRNAs in PT-sEV compared with sEVs derived from untreated BMSCs. Overexpression or inhibition in vitro indicated that the osteogenic inducing potential of sEVs was mainly attributable to miR-330-5p, one of the most dramatically downregulated miRNAs in the PT-sEV fraction. Dual luciferase reporter assays showed that miR-330-5p negatively regulated osteogenesis by directly binding to the 3′ untranslated region of Tnc. Additional experiments showed that Tnc regulated Wnt/β-catenin signaling, and rescue experiment showed that miR-330-5p could restore β-catenin expression; additionally, animal experiments indicated that Wnt signaling was inactivated in the ovariectomized rats. These data demonstrated the regenerative potential of PT-sEV, which induced osteogenic differentiation of pre-osteoblasts, leading to bone formation. This process was achieved by delivering miR-330-5p, which regulated Tnc to control Wnt/β-catenin signaling.

The bifurcations or transitions of the classical nonlinear systems with finite degrees of freedom exhibit some dynamical behaviors that are similar to those in the phase transitions of many-body systems. A dynamical type of them in the form of a sudden transition of the amplitudes between two harmonic oscillations, which are stable under periodically driving force, is not so common to see. We show that a sudden transition of this type can be realized in a system of coupling a microresonator that supports a mechanical vibration to another microresonator, and the system was experimentally demonstrated as a phonon laser. If the pump laser power or the intercavity coupling strength is adjusted to a critical value, the cavity field patterns will suddenly change, together with a jump of the stabilized mechanical amplitude. Such transition can be applied to a precise measurement of the optomechanical coupling constant, which is hard to measure due to its proportion to a tiny zero-point mechanical fluctuation amplitude.

For achieving successful chemotherapy against cancer, designing biocompatible drug delivery systems (DDSs) with long circulation times, high cellular endocytosis efficiency, and targeted drug release is of upmost importance. Herein, a well-defined PEG-b-P(MASSChol-co-MANBoc) block copolymer bearing redox-sensitive cholesteryl-side group was prepared via reversible addition-fragmentation chain transfer (RAFT) polymerization (with non-redox PEG-b-P(MACCChol-co-MAN-DCA) as the reference), and 1,2-dicarboxylic-cyclohexene acid (DCA) was then grafted onto the hydrophobic block to endow it with charge-convertible characteristics under a tumor microenvironment. The amphiphilic copolymer could be assembled into polymeric spherical micelles (SSMCs) with polyethylene glycol (PEG) as the corona/shell, and anti-cancer drug doxorubicin (DOX) was successfully encapsulated into the micellar core via strong hydrophobic and electrostatic interactions. This nanocarrier showed high stability in the physiological environment and demonstrated “smart” surface charge conversion from negative to positive in the slightly acidic environment of tumor tissues (pH 6.5~6.8), as determined by dynamic light scattering (DLS). Moreover, the cleavage of a disulfide bond linking the cholesterol grafts under an intracellular redox environment (10 mM GSH) resulted in micellar dissociation and accelerated drug release, with the non-redox-responsive micelles (CCMCs) as the control. Additionally, a cellular endocytosis and tumor proliferation inhibition study against MCF-7 tumor cells demonstrated the enhanced endocytosis and tumor cell inhibitory efficiency of dual-responsive SSMCs/DOX nanomedicines, revealing potentials as multifunctional nanoplatforms for effective oncology treatment.

Heart (right) failure is the most frequent cause of death in patients with pulmonary arterial hypertension. Although historically, increased right ventricular afterload has been considered the main contributor to right heart failure in such patients, recent evidence has suggested a potential role of load-independent factors. Here, we tested the hypothesis that resistin–like molecule α (RELMα), which has been implicated in the pathogenesis of vascular remodeling in pulmonary artery hypertension, also contributes to cardiac metabolic remodeling, leading to heart failure. Recombinant RELMα (rRELMα) was generated via a Tet-On expression system in the T-REx 293 cell line. Cultured neonatal rat cardiomyocytes were treated with purified rRELMα for 24 h at a dose of 50 nM. Treated cardiomyocytes exhibited decreased mRNA and protein expression of peroxisome proliferator-activated receptor gamma coactivator 1α (PGC-1α) and transcription factors PPARα and ERRα, which regulate mitochondrial fatty acid metabolism, whereas genes that encode for glycolysis-related proteins were significantly upregulated. Cardiomyocytes treated with rRELMα also exhibited a decreased basal respiration, maximal respiration, spare respiratory capacity, ATP-linked OCR, and increased glycolysis, as assessed with a microplate-based cellular respirometry apparatus. Transmission electron microscopy revealed abnormal mitochondrial ultrastructure in cardiomyocytes treated with rRELMα. Our data indicate that RELMα affects cardiac energy metabolism and mitochondrial structure, biogenesis, and function by downregulating the expression of the PGC-1α/PPARα/ERRα axis.

The paper reports the use the sol-gel auto-combustion method for the preparation of nanocrystalline La1-xSrxFeO3 (x = 0¸0.7). The physical and chemical properties, microstructure, morphology and magnetic properties of the particles thus formed were studied. The X-ray diffraction (XRD) results indicated that Sr2+ instead of perovskite La are attributed to the lattice forming solid solution La1-xSrxFeO3, which has single orthogonal perovskite structure, space group Pnma, and does not show any impurity peaks. Fourier-transform infrared (FT-IR) spectroscopy confirmed the presence of Fe-O-Fe bond antisymmetric stretching vibration of FeO6 regular octahedron. The scanning electron microscope (SEM) analysis showed that introduction of Sr2+ ions can inhibit the growth of size of the particles thereby forming particles with spherical shape and better dispersion properties. Abstract Background Blastocystis is a common protist that can infect domestic and wild animals worldwide. Yak (Bos grunniens), an ancient species which can survive in alpine regions, has supplied necessities to local residents in plateau areas for generations. However, the infections with Blastocystis in yaks has been ignored for a long time. In the present study, the infections and genotypes of Blastocystis spp. in domestic yaks from Qinghai Province (northwestern China) were explored. Results Of 1027 faecal samples collected from yaks in seven regions of Qinghai Province, northwestern China, the total prevalence of Blastocystis was 27.07% (278/1027) targeting the small subunit ribosome rRNA (SSU rRNA) gene. This protist was detected in yaks within each examined age group, geographical origin and season. Significant difference in prevalence was found in yaks from different geographical origins. The highest prevalence (48.94%) was observed in animals from Haixi county. Sequence analysis revealed three animal-specific subtypes (ST10, ST12 and ST14) of Blastocystis spp. in these yaks, with ST10 being the predominant subtype widely distributed in all investigated regions, seasons and age groups. Interestingly, this is the first report about subtype ST12 infecting yaks. Conclusions To our knowledge, this is the first systematic report on Blastocystis prevalence in yaks from China, and the findings provide fundamental data for establishing effective control measures for this protist in yaks as well as other animals in China.

Doping at A/B-sites can effectively improve the physical properties of perovskite ferrites. In this study, the performance of Bi0.8Ca0.2-xSrxFeO3 was investigated using the sol-gel autocombustion method. The results show that with an increase in x value, the grain size first increases and then decreases. With an increase in the doping concentration, the reunion phenomenon reduces, but the smoothness and flatness of the samples are also destroyed. Moreover, with increasing Sr2+ concentration, particle growth is inhibited, resulting in smaller particle sizes. When the Sr2+ concentration increases to a certain value, the environment becomes advantageous for particle growth and therefore the particle size increases. For x=0.13, small particles are obtained. With increasing Sr2+ concentration, the spatial modulation of the spin structure is destroyed, the lattice distorts, and the magnetic force is freed. Mössbauer spectrum measurements shows that when the Ca2+ concentration is greater than the Sr2+ concentration, Sr2+ ions replace Ca2+ ions; as a result, A-A magnetic superexchange diminishes, leading to a decrease in the hyperfine field. Appropriate doping of BiFeO3 can improve its coercivity and refine its grains and result in a larger magnetic force.

Antibiotic resistance genes (ARGs), prevalent across multiple environmental media, threaten human health worldwide and are considered emerging environmental contaminants. Earthworm gut, a niche for bacteria to survive, represents a potential reservoir for ARGs in soil. However, the compositions of ARGs in the earthworm gut microbiota remain elusive, especially under field conditions. In this study, we applied high-throughput quantitative PCR to profile the ARGs in the gut microbiota of earthworms after chronic exposure to fertilizers. To elucidate the factors that impact the ARGs composition, the bacterial community of gut microbiota, mobile genetic elements (MGEs), soil (nutrients, heavy metals, and antibiotics) and the properties of gut content (pH and nutrients) were analyzed. A total of 98 subtypes among 9 major types of ARGs, and 3 different MGEs were detected in the gut microbiota of earthworms. Organic fertilizer (sewage sludge and chicken manure) application significantly increased the diversity and abundance of ARGs. Of the 1123 identified operational taxonomic units (OTUs) at 97% similarity cutoff, most of them were assigned to Firmicutes (55.5%) and Proteobacteria (33.6%) in earthworm gut microbiota. Long-term organic fertilization slightly changed the microbiota composition, but did not impact the diversity. Partial redundancy analysis (pRDA) revealed that bacterial community, combined with environmental factors (soil and gut content properties) and MGEs, explained 72% of the variations of ARGs in the earthworm gut. Furthermore, the co-occurrence pattern between ARGs and MGEs indicated that horizontal gene transfer via MGEs may occur in the earthworm gut. These findings improve the current understanding of the dynamics of soil fauna-associated ARGs and the gut microbiota of earthworms may be an underappreciated hotspot for ARGs in the environment. The parasites from buffaloes in Guizhou province, southwest China, were suspected to be sarcocysts through morphologic observation. 18S rRNA and the subunit I of cytochrome oxidase (cox1) sequences were amplified, then implemented by phylogenetic analysis. Phylogenetic analyses of 18S rRNA and cox1 genes both indicated the low degree of genetic diversity among the isolated sequences and other sequences of Sarcocystis fusiformis. The result shows that the sarcocysts from these buffaloes in Guizhou province S. fusiformis. This study is the first molecular characterization of S. fusiformis from Chinese buffaloes that used two genetic markers (18S rRNA and cox1 sequences) in China.

Optomechanical systems are suitable for realizing the ground-state cooling of macroscopic objects. Based on a dynamical approach that goes beyond the validity of the standard linearization approach, we simulate the detailed cooling processes for a membrane-in-middle optomechanical system. In addition to the cooling results, we especially study the cooling speed, which is indicated by how soon the first minimum thermal phonon number is reached. Their relevance to the system parameters provides essential knowledge about how to achieve the best and/or fastest cooling under various combinations of different driving fields.

Istradefylline as a selective adenosine A2A-receptor antagonist is clinically used to treat Parkinson’s disease and improve dyskinesia in its early stages. However, its crystal form, as an important factor in the efficacy of the drug, is rarely studied. Herein, three kinds of crystal forms of istradefylline prepared from ethanol (form I), methanol (form II), and acetonitrile (form III) are reported by use of a crystal engineering strategy. These three crystal forms were characterized and made into tablets for dissolution testing. Both the solubility and the dissolution rates were also determined. The dissolution rate of form I and form III is significantly higher than form II at pH 1.2 (87.1%, 58.2%, and 87.7% for form I, form II, and form III, respectively), pH 4.5 (88.1%, 58.9%, and 87.1% for form I, form II, and form III, respectively) and pH 6.8 (87.5%, 58.2%, and 86.0% for form I, form II, and form III, respectively) at 60 min. Considering the prepared solution and the proper dissolution profile, form I is anticipated to possess promising absorption for bioavailability.

As a common herbicide in farmland, there has been wide concern over quinclorac residue because of its potential risks to the environment and human health. For the detection and monitoring of quinclorac residue in the environment, enzyme-linked immunoassay (ELISA) and time-resolved fluoroimmunoassay (TRFIA) were established. The half-maximal inhibition concentrations (IC50) of ELISA and TRFIA were 0.169 mg/L and 0.087 mg/L with a linear range (IC20–IC80) of 0.020–1.389 mg/L and 0.004–1.861 mg/L, respectively. Compared with ELISA, the limit of detection (LOD, IC20) and IC50 of TRFIA improved approximately 5-fold and 2-fold. The cross-reaction rates for the quinclorac analogs were less than 2%. The average recoveries of quinclorac in river water, paddy water, paddy soil, and brown rice samples were 77.3–106.1%, with RSDs of 1.7–12.5%. More importantly, the results of the two methods were consistent with that of the referenced method of UPLC-MS/MS (R2 > 0.98). ELISA and TRFIA both showed good detection performance and could meet the requirements of the quantitative determination of quinclorac. Therefore, the proposed ELISA and TRFIA could be applied to the rapid and sensitive detection and monitoring of quinclorac residue in the environment.

Multiple environment phenotypes may be utilized to implement genomic prediction in plant breeding, while it is unclear about optimal utilization strategies according to its different availability. It is necessary to assess the utilization strategies of genomic prediction models based on different availability of multiple environment phenotypes. Here, we compared the prediction accuracy of three genomic prediction models (genomic prediction model (genomic best linear unbiased prediction (GBLUP), genomic best linear unbiased prediction (GFBLUP), and multi-trait genomic best linear unbiased prediction (mtGBLUP)) which leveraged diverse information from multiple environment phenotypes using a rice dataset containing 19 agronomic traits in two disparate seasons. We found that the prediction accuracy of genomic prediction models considering multiple environment phenotypes (GFBLUP and mtGBLUP) was better than the classical genomic prediction model (GBLUP model). The deviation of prediction accuracy of between GBLUP and mtGBLUP or GFBLUP was associated with the phenotypic correlation. In summary, the genomic prediction models considering multiple environment phenotypes (GFBLUP and mtGBLUP) demonstrated better prediction accuracy. In addition, we could utilize different genomic prediction strategies according to different availability of multiple environment phenotypes.

Accurate short-term forecasting of intensive rainfall has high practical value but remains difficult to achieve. Based on deep learning and spatial–temporal sequence predictions, this paper proposes a hierarchical dynamic graph network. To fully model the correlations among data, the model uses a dynamically constructed graph convolution operator to model the spatial correlation, a recurrent structure to model the time correlation, and a hierarchical architecture built with graph pooling to extract and fuse multi-level feature spaces. Experiments on two datasets, based on the measured cumulative rainfall data at a ground station in Fujian Province, China, and the corresponding numerical weather grid product, show that this method can model various correlations among data more effectively than the baseline methods, achieving further improvements owing to reversed sequence enhancement and low-rainfall sequence removal.

Acute myeloid leukemia (AML) is a highly heterogeneous disease that requires fine-grained risk stratification for the best prognosis of patients. As a class of small non-coding RNAs with important biological functions, microRNAs play a crucial role in the pathogenesis of AML. To assess the prognostic impact of miR-20b on AML in the presence of other clinical and molecular factors, we screened 90 AML patients receiving chemotherapy only and 74 also undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) from the Cancer Genome Atlas (TCGA) database. In the chemotherapy-only group, high miR-20b expression subgroup had shorter event-free survival (EFS) and overall survival (OS, both P < 0.001); whereas, there were no significant differences in EFS and OS between high and low expression subgroups in the allo-HSCT group. Then we divided all patients into high and low expression groups based on median miR-20b expression level. In the high expression group, patients treated with allo-HSCT had longer EFS and OS than those with chemotherapy alone (both P < 0.01); however, there were no significant differences in EFS and OS between different treatment subgroups in the low expression group. Further analysis showed that miR-20b was negatively correlated with genes in “ribosome,” “myeloid leukocyte mediated immunity,” and “DNA replication” signaling pathways. ORAI2, the gene with the strongest correlation with miR-20b, also had significant prognostic value in patients undergoing chemotherapy but not in the allo-HSCT group. In conclusion, our findings suggest that high miR-20b expression is a poor prognostic indicator for AML, but allo-HSCT may override its prognostic impact.

According to the dynamic characteristics of the adhesion desorption process between gecko-like polyurethane setae and the contact surface, the microcontact principle of an elastic sphere and plane is established based on the Johnson–Kendall–Robert model. On this basis, combined with the cantilever beam model, microscale adhesive contact models in the case of a single and an array of setae are obtained. The contact process is numerically simulated and verified by the adhesion desorption test. After that, the effects of external preload, the elastic modulus of setae material, the surface energy, and the surface roughness on the contact force and depth during the dynamic contact process of setae are studied. The results show that the error between the simulation and test is 15.9%, and the simulation model could reflect the real contact procedure. With the increase in preload, the push-off force of the setae array would grow and remain basically constant after reaching saturation. Increasing the elastic modulus of setae material would reduce the contact depth, but have little effect on the maximum push-off force; with the increase in the surface energy of the contact object, both the push-off force between the objects and the contact depth during desorption would increase. With the increase in wall roughness, the push-off force curve of the setae array becomes smoother, but the maximum push-off force would decrease. By exploring the dynamic mechanical characteristics of the micro angle of setae, the corresponding theoretical basis is provided for the numerical simulation of the adsorption force of macro materials.

The Chern insulator manifests itself via the surface quantized Hall current and magnetoelectric effect. The manipulation of surface magnetizations enables a control of the dissipationless chiral transport and thus allows for potential applications of topological magnetoelectric devices with low-energy consumption. Here, we present experimental studies of bias-modulated switching the magnetic states utilizing the magnetoelectric coupling in a Chern insulator. This is achieved via applying a d.c. bias across the source and drain at various magnetic states, during which an effective magnetic field is developed to switch the quantum anomalous Hall state towards its opposite. Comprehensive transport studies show that the switch efficiency is proportional to the amplitude and applying time of the bias, depends on the initial magnetic state, but is insensitive to the electric polarity. Our results provide an efficient scheme to manipulate the Chern insulator and understanding on the electric breakdown of chiral edge states.

The effects of soybean oil (20%, v/w) and extraction time (30, 60, or 90 min) on volatile compounds in cinnamon bark extract were investigated. The relative content and odor activity values (OAVs) of volatile compounds were measured by Gas Chromatography-Mass Spectrometer (GC-MS). The results showed that a total of 26 and 27 volatile compounds were detected in the water extract and the aqueous phase of the water/oil extraction, respectively. Hexanal, nonanal, cinnamaldehyde, D-limonene, 1-octen-3-ol, linalool, and anethole were the major aroma-active compounds, accounting for 85% of the total substance content. Cinnamaldehyde had the highest contribution rate to the aroma of the water extract (26%), whereas anethole has the highest contribution rate to the aroma of the oil/water extract (30%). Whether or not the extraction medium contained soybean oil, the relative content of aroma-active compounds in the aqueous phase decreased with increased extraction time, and the relative content of these compounds in the aqueous phase further decreased when soybean oil was present. This should be due to the high hydrophobicity of these compounds, which were prone to dissolving in the oil layer during the extraction process, resulting in a decrease in the relative content of aroma-active compounds in the aqueous phase.

Tic disorders (TD) are a group neuropsychiatric disorders with childhood onset characterized by tics, i.e. repetitive, sudden, and involuntary movements or vocalizations; and Tourette syndrome (TS) is the most severe form of TD. Their clinical manifestations are diverse; and are often associated with various psychopathological and/or behavioral comorbidities, including attention deficit hyperactivity disorder (ADHD), obsessive-compulsive disorder (OCD), anxiety, depression, and sleep disorders. Individual severity and response to treatment are highly variable, and there are some refractory cases, which are less responsive to conventional TD treatment. TD/TS are also common in the Chinese pediatric population. To help improve the understanding of TD for pediatricians and other health professionals, and to improve its diagnosis and treatment in China, the Chinese Child Neurology Society (CCNS) has developed an Expert Consensus on Diagnosis and Treatment of TD in China, which is based on our clinical experience and the availability therapeutic avenues. It is focused on clinical diagnosis and evaluation of TD and its comorbidities, psychological and educational intervention, nonpharmacological therapy, pharmacological treatment, including traditional Chinese medicine and acupuncture, as well as prognosis in children with TD in China. A summary of the current status of TD and up-to-date diagnosis and treatment recommendations for TD in China is presented here.

In many countries, floods are the leading natural disaster in terms of damage and losses per year. Early prediction of such events can help prevent some of those losses. Artificial neural networks (ANN) show a strong ability to deal quickly with large amounts of measured data. In this work, we develop an ANN for outputting flood inundation maps based on multiple discharge inputs with a high grid resolution (4 m × 4 m). After testing different neural network training algorithms and network structures, we found resilience backpropagation to perform best. Furthermore, by introducing clustering for preprocessing discharge curves before training, the quality of the prediction could be improved. Synthetic flood events are used for the training and validation of the ANN. Historical events were additionally used for further validation with real data. The results show that the developed ANN is capable of predicting the maximum flood inundation extents. The mean squared error in more than 98 and 86% of the total area is smaller than 0.2 m2 in the prediction of synthetic events and historical events, respectively.

Resistin and resistin-like molecules are pleiotropic cytokines that are involved in inflammatory diseases. Our previous work suggested that resistin has the potential to be used as a biomarker and therapeutic target for human pulmonary arterial hypertension. However, data are limited on the distribution of resistin in healthy human organs. In this study, we used our newly developed anti-human resistin (hResistin) antibody to immunohistochemically detect the expression, localization, and intracellular/extracellular compartmentalization of hResistin in a full human tissue panel from healthy individuals. The potential cross reactivity of this monoclonal anti-hResistin IgG1 with normal human tissues also was verified. Results showed that hResistin is broadly distributed and principally localized in the cytoplasmic granules of macrophages scattered in the interstitium of most human tissues. Bone marrow hematopoietic precursor cells also exhibited hResistin signals in their cytoplasmic granules. Additionally, hResistin labeling was observed in the cytoplasm of nervous system cells. Notably, the cytokine activity of hResistin was illustrated by positively stained extracellular material in most human tissues. These data indicate that our generated antibody binds to the secreted hResistin and support its potential use for immunotherapy to reduce circulating hResistin levels in human disease. Our findings comprehensively document the basal expression patterns of hResistin protein in normal human tissues, suggest a critical role of this cytokine in normal and pathophysiologic inflammatory processes, and offer key insights for using our antibody in future pharmacokinetic studies and immunotherapeutic strategies.

Employing natural-based renewable sugar and saccharide resources to construct functional biopolymer mimics is a promising research frontier for green chemistry and sustainable biotechnology. As the mimics/analogues of natural glycoproteins, synthetic glycopolypeptides attracted great attention in the field of biomaterials and nanobiotechnology. This review describes the synthetic strategies and methods of glycopolypeptides and their analogues, the functional self-assemblies of the synthesized glycopolypeptides, and their biological applications such as biomolecular recognition, drug/gene delivery, and cell adhesion and targeting, as well as cell culture and tissue engineering. Future outlook of the synthetic glycopolypeptides was also discussed.

State-of-the-art facial expression methods outperform human beings, especially, thanks to the success of convolutional neural networks (CNNs). However, most of the existing works focus mainly on analyzing an adult’s face and ignore the important problems: how can we recognize facial expression from a baby’s face image and how difficult is it? In this paper, we first introduce a new face image database, named BabyExp, which contains 12,000 images from babies younger than two years old, and each image is with one of three facial expressions (i.e., happy, sad, and normal). To the best of our knowledge, the proposed dataset is the first baby face dataset for analyzing a baby’s face image, which is complementary to the existing adult face datasets and can shed some light on exploring baby face analysis. We also propose a feature guided CNN method with a new loss function, called distance loss, to optimize interclass distance. In order to facilitate further research, we provide the benchmark of expression recognition on the BabyExp dataset. Experimental results show that the proposed network achieves the recognition accuracy of 87.90% on BabyExp.

Bufalin is a cardiotonic steroid and a key active ingredient of the Chinese medicine ChanSu. It has significant anti-tumor activity against many malignancies, including hepatocellular carcinoma (HCC). Previous studies have shown that human bodies contain an endogenous bufalin-like substance. This study aimed to confirm whether the endogenous bufalin-like substances is bufalin and further detect the differences between HCC and control groups of endogenous bufalin concentration by the high-performance liquid chromatography coupled tandem mass spectrometry (HPLC-MS/MS). The results confirmed the endogenous bufalin-like substance is bufalin. Totally, 227 serum samples were collected: 54 from HCC patients and 173 from healthy volunteers constituting a control group. Both the test group and the control group contained bufalin in serum, revealing that bufalin is indeed an endogenous substance. The bufalin concentration was 1.3 nM in HCC patients and 5.7 nM in normal people (P < 0.0001). These results indicate that human bodies contain endogenous bufalin, and it may be negatively correlated with the incidence of HCC.

The spin-orbit torque (SOT) induced magnetic switching makes metal/magnetic insulators bilayers preferred in the energy efficient spintronic applications. Here the authors show SOT switching in W/TmIG bilayers and reveal the dimension crossover of SOT as a function of TmIG thickness.

Osteosarcoma (OS) is the most common non-hematologic malignant tumor of bone in children. It is usually characterized by a high risk of developing lung metastasis and poor prognosis. Extracellular vesicles (EVs) are cell-derived nanoparticles with a small size of 50–200 nm in diameter. As a communicator, the contents of the EVs secreted via either fusing with lysosomes for degradation and recycling or fusing with the cell plasma membrane into the extracellular environment, which play an important role in regulating the tumor microenvironment of OS and mediating the Wnt/β-catenin and TGF-β signalings. Increasing evidences suggest that EVs have significant role in OS growth, progression, metastasis and drug resistance. In this study, the roles of EVs in the physiology and pathogenesis of OS and the potential attractive therapeutic target for the treatment of OS were reviewed. Spin-polarized carriers could show an extra Hall component when moving through certain real-space topological spin textures. Here, He et al. report an exchange bias experienced by the topological spin textures living at the interface between a topological insulator and an adjacent antiferromagnet, suggesting a chiral spin texture is induced.

Objective: Cerebral ischemia reperfusion injury (CIRI) is a major cause of ischemic stroke (IS) deterioration. Considering the intricate mechanism of the pathological process of CIRI, most drugs only work on one target. The neurovascular unit (NVU) puts forward the concept of neuroprotection from nerve protection to global stabilization. The NVU plays an important role in maintaining the brain microenvironment. This would promote neuronal survival and overall neurological recovery, which would likely lead to the reduction of mortality rate. Previous studies have shown that 4-methoxy benzyl alcohol (4-MA) ameliorated neurological score and cerebral infarct volume and reduced the concentration of Evans blue (EB) in brain tissue. In this research, we investigated the effects of 4-MA on NVU microenvironment improvement in rats impaired by middle cerebral artery occlusion/reperfusion (MCAO/R). Methods: First, we established a rat model of middle cerebral artery occlusion (MCAO) so as to use Western blot analysis, immunofluorescence and transmission electron microscopy (TEM) evaluating the NVU's protection of 4-MA. Then we established a primary cortical neuron model of oxygen glucose deprivation and re-oxygenation (OGD/R) with the objective of identifying whether 4-MA exhibited anti-oxidant and anti-apoptotic effects on neurons. Results: NVU ultra structural changes were improved by 4-MA. Immunofluorescence and western blot showed that 4-MA protected NVUs through enhancement of the expression of the symbolic neuronal proteins Microtubule Associated Protein-2(MAP-2), and attenuation of protein expression of Asy symbolic protein Glial Fibrillary Acidic Protein(GFAP). Furthermore, in the OGD/R model of I/R injury in vitro, 4-MA significantly increased Superoxide dismutase(SOD), Nitric Oxide(NO), B-cell lymphoma-2(Bcl-2), decreased Bcl-2-Associated X(Bax) and increased Bcl-2/Bax. Conclusion: 4-MA can play the role of anti-ischemic stroke drug by ameliorating the microenvironment of NVUs while its neuroprotective effects will contribute towards the inhibition of the antioxidant and anti-apoptotic activities.

Adjuvants are considered a necessary component for HBV therapeutic vaccines but few are licensed in clinical practice due to concerns about safety or efficiency. In our recent study, we established that a combination protocol of 3-day pretreatments with GM-CSF before a vaccination (3 × GM-CSF+VACCINE) into the same injection site could break immune tolerance and cause over 90% reduction of HBsAg level in the HBsAg transgenic mouse model. Herein, we further investigated the therapeutic potential of the combination in AAV8–1.3HBV-infected mice. After 4 vaccinations, both serum HBeAg and HBsAg were cleared and there was a 95% reduction of HBV-positive hepatocytes, in addition to the presence of large number of infiltrating CD8+ T cells in the livers. Mechanistically, the HBV-specific T-cell responses were elicited via a 3 × GM-CSF+VACCINE-induced conversion of CCR2-dependent CD11b+ Ly6Chi monocytes into CD11b+CD11c+ DCs. Experimental depletion of Ly6Chi monocytes resulted in a defective HBV-specific immune response thereby abrogating HBV eradication. This vaccination strategy could lead to development of an effective therapeutic protocol against chronic HBV in infected patients.
