Abstract Background In patients with prostate cancer (PCa), imaging with gastrin-releasing peptide receptor (GRPR) ligands is an alternative to PSMA-targeted tracers, particularly if PSMA expression is low or absent. [99mTc]Tc-N4-BTG is a newly developed GRPR-directed probe for conventional scintigraphy and single photon emission computed tomography (SPECT) imaging. The current study aims to investigate the safety, biodistribution and dosimetry of [99mTc]Tc-N4-BTG in patients with biochemical recurrence (BCR) of PCa. Results No adverse pharmacologic effects were observed. Injection of [99mTc]Tc-N4-BTG resulted in an effective dose of 0.0027 ± 0.0002 mSv/MBq. The urinary bladder was the critical organ with the highest mean absorbed dose of 0.028 ± 0.001 mGy/MBq, followed by the pancreas with 0.0043 ± 0.0015 mGy/MBq, osteogenic cells with 0.0039 ± 0.0005 mGy/MBq, the kidneys with 0.0034 ± 0.0003 mGy/MBq, and the liver with 0.0019 ± 0.0004 mGy/MBq, respectively. No focal tracer uptake suggestive of PCa recurrence could be revealed for any of the patients. Conclusion [99mTc]Tc-N4-BTG appears to be a safe diagnostic agent. Compared to GRPR-targeted PET tracers, this 99mTc-labelled SPECT agent could contribute to a broader application and better availability of this novel approach. Further research to assess its clinical value is warranted.

Abstract Background Oral squamous carcinoma (OSCC) is often diagnosed at late stages and bone erosion or invasion of the jawbone is frequently present. Computed tomography (CT) and magnetic resonance imaging (MRI) are known to have high diagnostic sensitivities, specificities, and accuracies in detecting these bone affections in patients suffering from OSCC. To date, the existing data regarding the impact of cone-beam computed tomography (CBCT) have been weak. Therefore, this study aimed to investigate whether CBCT is a suitable tool to detect bone erosion or invasion in patients with OSCC. Methods We investigated in a prospective trial the impact of CBCT in the diagnosis of bone erosion or invasion in patients with OSCC who underwent surgery. Every participant received a CBCT, CT, and MRI scan during staging. Imaging modalities were evaluated by two specialists in oral and maxillofacial surgery (CBCT) and two specialists in radiology (CT and MRI) in a blinded way, to determine whether a bone affection was present or not. Reporting used the following 3-point system: no bony destruction (“0”), cortical bone erosion (“1”), or medullary bone invasion (“2”). Histological examination or a follow-up served to calculate the sensitivities, specificities, and accuracies of the imaging modalities. Results Our results revealed high diagnostic sensitivities (95.6%, 84.4%, and 88.9%), specificities (87.0%, 91.7%, and 91.7%), and accuracies (89.5%, 89.5%, and 90.8%) for CBCT, CT, and MRI. A pairwise comparison found no statistical difference between CBCT, CT, and MRI. Conclusion Our data support the routine use of CBCT in the diagnosis of bone erosion and invasion in patients with OSCC as diagnostic accuracy is equal to CT and MRI, the procedure is cost-effective, and it can be performed during initial contact with the patient.

Abstract Background We recently introduced radiohybrid (rh)-based minigastrin analogs e.g., DOTA-rhCCK-18 (DOTA-D-Dap(p-SiFA)-(D-γ-Glu)8-Ala-Tyr-Gly-Trp-Nle-Asp-Phe-NH2), that revealed substantially increased activity retention in the tumor. However, one major drawback of these first generation rh-based cholecystokinin-2 receptor (CCK-2R) ligands is their elevated activity levels in the kidneys, especially at later time points (24 h p.i.). Therefore, this study aimed to reduce kidney retention with regard to a therapeutic use via substitution of negatively charged D-glutamic acid moieties by hydrophilic uncharged polyethylene glycol (PEG) linkers of various length ((PEG)4 to (PEG)11). Furthermore, the influence of differently charged silicon-based fluoride acceptor (SiFA)-moieties (p-SiFA: neutral, SiFA-ipa: negatively charged, and SiFAlin: positively charged) on in vitro properties of minigastrin analogs was evaluated. Out of all compounds evaluated in vitro, the two most promising minigastrin analogs were further investigated in vivo. Results CCK-2R affinity of most compounds evaluated was found to be in a range of 8–20 nM (by means of apparent IC 50), while ligands containing a SiFA-ipa moiety displayed elevated IC 50 values. Lipophilicity was noticeably lower for compounds containing a D-γ-glutamate (D-γ-Glu) moiety next to the D-Dap(SiFA) unit as compared to their counterparts lacking the additional negative charge. Abstract The possibility to stimulate the production of some nutraceutical properties of fenugreek (Trigonella foenum-graecum L.) sprouts by non-thermal plasma (NTP) processing of the seeds in different conditions was studied. The non-thermal plasma used in this work was a surface dielectric barrier discharge. Two types of processing were performed: direct NTP treatment and NTP with a cover treatment, to simulate the processing of packaged seeds. For all treatments, the effect of pre-soaking of the seeds was studied as well. The analyses of the seeds after processing indicated an increase of the hydrophilicity of their surface for NTP direct treatment as resulted from the water contact angle measurements, which could be due to the strong etching evidenced by scanning electron microscopy imaging. Longwall mining shearers equipped with drum-type mechanisms have become widely applied equipment in underground coal exploitation. The constructive evolution, quality, reliability, and functional performance of these machines, influenced by technological advancements in materials and manufacturing, is nonetheless overshadowed by their adaptability to various geological and mining conditions, particularly concerning the parameters of the cutting mechanism. A judicious correlation of geometric, dimensional, kinematic, and functional parameters with the characteristics of the coal seam provides a vast field for research. For this reason, we have approached this type of equipment, with special emphasis on the working components, in a study where computer-based methods can prove their utility.

Abstract Riverbed sediments have recently been found to be an important reservoir for microplastics. But the hydrogeological factors that control the abundance of microplastics are complex and conceptual frameworks priorising the parameters affecting their transport and retention during deep riverbed filtration are still missing. In this study a series of saturated column experiments was conducted to investigate the vertical distribution patterns of secondary polystyrene fragments (100–2000 μm) in dependence on their particle size, grain size of the sediment, seepage velocity and duration of infiltration flow. The columns with a length of 50 cm were operated with flow velocities between 1.8 m d−1 and 27 m d−1. Invasive samples obtained after the experiments were density separated and then depth profiles of microplastic concentrations were retrieved using fluorescence imaging analysis. Most polystyrene particles were retained in the upper 20 cm and 15 cm of the medium gravel and coarse sand sediments, respectively. Through the high particle retention riverbed sediments can act as a temporary sink or long term retention site for the transport of microplastic particles (MPPs) from streams to oceans. A small fraction of particles ranging from 100 to 500 μm in size was observed down to infiltration depths of 50 cm suggesting that MPPs at the pore scale have the potential to be advectively transferred via hyporheic exchange or induced bank filtration into coarse riverbed sediments and alluvial aquifers. MPP abundance over column depth follows an exponential relationship with a filter coefficient that was found to depend significantly on the flow rate, MPP and sediment grain size, as indicated by multiple linear regression (R2 = 0.92). The experimentally derived empirical relation allows to estimate particle abundances of initially negatively buoyant MPP in riverbed sediments by surface water infiltration.

Abstract Background Recently, we introduced Stroma-AReactive-Invasion-Front-Areas (SARIFA) as a novel hematoxylin–eosin (H&E)-based histopathologic prognostic biomarker for various gastrointestinal cancers, closely related to lipid metabolism. To date, no studies on SARIFA, which is defined as direct tumor-adipocyte-interaction, beyond the alimentary tract exist. Hence, the objective of our current investigation was to study the significance of SARIFA in pT3a prostate cancer (PCa) and explore its association with lipid metabolism in PCa as lipid metabolism plays a key role in PCa development and progression. Methods To this end, we evaluated SARIFA-status in 301 radical prostatectomy specimens and examined the relationship between SARIFA-status, clinicopathological characteristics, overall survival, and immunohistochemical expression of FABP4 and CD36 (proteins closely involved in fatty-acid metabolism). Additionally, we investigated the correlation between SARIFA and biochemical recurrence-free survival (BRFS) and PSMA-positive recurrences in PET/CT imaging in a patient subgroup. Moreover, a quantitative SARIFA cut-off was established to further understand the underlying tumor biology. Results SARIFA positivity occurred in 59.1% (n = 178) of pT3a PCas. Our analysis demonstrated that SARIFA positivity is strongly associated with established high-risk features, such as R1 status, extraprostatic extension, and higher initial PSA values. Additionally, we observed an upregulation of immunohistochemical CD36 expression specifically at SARIFAs (p = 0.00014). Kaplan–Meier analyses revealed a trend toward poorer outcomes, particularly in terms of BRFS (p = 0.1). More extensive tumor-adipocyte interaction, assessed as quantity-dependent SARIFA-status on H&E slides, is also significantly associated with high-risk features, such as lymph node metastasis, and seems to be associated with worse survival outcomes (p = 0.16). Moreover, SARIFA positivity appeared to be linked to more distant lymph node and bone metastasis, although statistical significance was slightly not achieved (both p > 0.05). Conclusions This is the first study to introduce SARIFA as easy-and-fast-to-assess H&E-based biomarker in locally advanced PCa. SARIFA as the histopathologic correlate of a distinct tumor biology, closely related to lipid metabolism, could pave the way to a more detailed patient stratification and to the development of novel drugs targeting lipid metabolism in pT3a PCa. On the basis of this biomarker discovery study, further research efforts on the prognostic and predictive role of SARIFA in PCa can be designed.

La zone collinaire de la ville de Kinshasa sud est confrontée chaque jour qui passe à des multiples impacts environnementaux négatifs. Parmi ceux-ci, il nous faut relever des aléas environnementaux qui occupent une place de choix, aléas qui constituent des sources des vulnérabilités humaines. Cette recherche qui porte sur la ville de Kinshasa se fonde sur des observations au départ d’une enquête sociodémographique relative à la gestion des terres urbaines des aléas qu’ils induisent révèlant que 60 % des parcelles ont déjà été touchées par les érosions, que 55,2 % des parcelles sont directement et indirectement menacées par les érosions, que 25 % des parcelles sont gravement affectées par des inondations au niveau de lit majeur des rivières en saisons pluvieuses et que 15 % des parcelles sont exposés aux ensablements au cours de la période du déroulement de cette enquête. Ces différentes impacts environnementaux négatifs occasionnent des dégâts immenses qui entravent les équilibre écologique et le développement durable la contrée sous-étude. Selon des reponses recueillies auprès des personnes vivant sur ces terres marginales de la ville de Kinshasa sud, les causes qui sont à la base de ces aléas environnementaux négatifs et des autres vulnérabilités survenant dans cette partie de la ville sont entre autres liées à la mauvaise orientation topographique des habitations (20,15 %), à l’inexistence de réseaux de drainage des eaux (47,86 %,) et à l’absence de puisards dans les parcelles (15,65 % ). Toutes ces causes sont dues, soit à l’absence des etudes avant l’occupation, au non suivi de la politique d’habitat, soit à l’anarchie dans la distribution des parcelles, soit ainsi à l’administration dualiste des Services de l’Etat dans le domaine foncier.

Background: While MRI is the primary diagnostic tool for the diagnosis of spondylodiscitis, the role of [18F]-fluorodeoxyglucose ([18F]FDG) PET/CT is gaining prominence. This study aimed to determine the frequency of [18F]FDG PET/CT usage and its impact on the in-hospital mortality rate in patients with spondylodiscitis, particularly in the geriatric population. Methods: We conducted a Germany-wide cross-sectional study from 2019 to 2021 using an open-access, Germany-wide database, analyzing cases with ICD-10 codes M46.2-, M46.3-, and M46.4- (‘Osteomyelitis of vertebrae’, ‘Infection of intervertebral disc (pyogenic)’, and ‘Discitis unspecified’). Diagnostic modalities were compared for their association with in-hospital mortality, with a focus on [18F]FDG PET/CT. Results: In total, 29,362 hospital admissions from 2019 to 2021 were analyzed. Of these, 60.1% were male and 39.9% were female, and 71.8% of the patients were aged 65 years and above. The overall in-hospital mortality rate was 6.5% for the entire cohort and 8.2% for the geriatric subgroup (p < 0.001). Contrast-enhanced (ce) MRI (48.1%) and native CT (39.4%) of the spine were the most frequently conducted diagnostic modalities. [18F]FDG PET/CT was performed in 2.7% of cases. CeCT was associated with increased in-hospital mortality (OR = 2.03, 95% CI: 1.90–2.17, p < 0.001). Cases with documented [18F]FDG PET/CT showed a lower frequency of in-hospital deaths (OR = 0.58, 95% CI: 0.18–0.50; p = 0.002). This finding was more pronounced in patients aged 65 and above (OR = 0.42, 95% CI: 0.27–0.65, p = 0.001). Conclusions: Despite its infrequent use, [18F]FDG PET/CT was associated with a lower in-hospital mortality rate in patients with spondylodiscitis, particularly in the geriatric cohort. This study is limited by only considering data on hospitalized patients and relying on the assumption of error-free coding. Further research is needed to optimize diagnostic approaches for spondylodiscitis.

Background: The purpose of this study was to establish a standardized structured workflow to compare findings from high-resolution, optimized reconstructions from post-mortem computed tomography (pmCT) with autopsy results in the detection of fractures of the laryngohyoid complex in strangulation victims. Method: Forty-two strangulation cases were selected, and pmCT scans of the laryngohyoid complex were obtained. Both pmCT scans and autopsy reports were analyzed using a structured template and compared using Cohen’s kappa coefficient (κ) and the McNemar test. The study also compared the prevalence of ossa sesamoidea and non-fusion of the major and minor horns of the hyoid bone between both diagnostic methods. Results: The detection of fractures showed a very good correlation between autopsy and pmCT results (κ = 0.905), with the McNemar test showing no statistically significant difference between the two methods. PmCT identified 28 sesamoid bones, 45 non-fusions of the major horns, and 47 non-fusions of the minor horns of the hyoid bone, compared to four, six, and zero, respectively, identified by autopsy (p < 0.0001). Conclusions: Autopsy and pmCT findings correlate well and can be used in a complementary manner. PmCT is superior to autopsy in identifying dislocations and detecting anatomical variations in the laryngohyoid complex, which can lead to misinterpretations during autopsy. Therefore, we do not advocate replacing autopsy with pmCT but propose using a structured workflow, including our standardized reporting template, for evaluating lesions in the laryngohyoid complex.

Introduction/Justification: We present a case of a 71-year-old male patient diagnosed with myoepithelial carcinoma of the pelvis and perineum, who underwent fibroblast activation protein (FAP)-directed radioligand therapy and presented long lasting tumor retention as detected by late whole-body scans. Report: The patient had been previously submitted to neoadjuvant radiation therapy and surgery, with tumoral relapse at the surgical margins in the gluteal region. Lesions displayed a slow and progressive sarcomatoid growth pattern, with sacral osteolytic involvement, accompanied with obturatory and left common iliac lymph node metastases. Immunotherapy was initiated but discontinued due to grade IV diarrhea. With progressive disease evident on FDG PET/CT and no other viable chemotherapy indicated, FAPI radioligand therapy was proposed, given significant tracer uptake observed in FAP-directed PET/CT. The patient received an intravenous injection of 200 mCi of [177Lu]Lu-FAP-RTX, which was well tolerated, with no acute side effects reported. Blood tests remained within normal range. Beyond partial hair loss,no other adverse events were observed. Imaging studies including whole-body planar and SPECT/CT scans revealed intense tracer retention in the sacral mass and mild to moderate retention in the lymph node metastases up to 15 days post-treatment. Notably, indicative of a response to FAP-directed radioligand therapy, there was a resolution of drainage from a fistula in the intergluteal region. Follow-up imaging is still pending at the moment. Conclusion: This case is the first report on favorably sustained tumor retention of the radiopharmaceutical in a carcinoma patient undergoing FAP-directed radioligand therapy. With tumor response assessment still pending, longer follow-up and detailed observation is still necessary for a better understanding of potential benefits and side effects of FAP-directed radioligand therapy, especially in patients undergoing subsequent treatment cycles.

A frequency stabilization scheme for CO2 lasers using external modulation via an electrooptic modulator was developed. The saturation signal of an OsO4 line detected in transmission of the Fabry-Perot cavity was used to obtain a stability of 0.1 Hz on a 100-s time scale as well as a reproducibility of up to 10 Hz. The present scheme performed better than a previous setup for which modulation was applied through piezoelectric transducers. The stabilized laser was also found to be easily tunable and not frequency-modulated.

This study investigated the effects of incorporating grape pomace (GP) into fish diets on the growth performance, physiological parameters, and biochemical composition of carp (Cyprinus carpio L.). A total of 180 carp, with an average initial weight of 65 g, were reared in a recirculating aquaculture system (RAS). They were divided into a control group (C), fed with a diet containing 0% GP, and two experimental groups (V1 and V2), fed with diets containing 5% and 10% GP, respectively. The experiment lasted for eight weeks. Results revealed that most growth parameters were not significantly affected by GP inclusion, except for a notable difference in the hepatosomatic index (HSI), indicating an impact on liver size relative to body weight. The biochemical analysis of carp meat showed significant differences in moisture, protein, fat, collagen, and salt content between the control and experimental groups. Furthermore, the oxidative status assessment indicated that GP supplementation modulates oxidative stress and lipid peroxidation pathways in carp, enhancing their antioxidant defenses and overall health. Microbiological examination of the carp intestinal content showed that GP inclusion in fish diets influenced microbial parameters, particularly affecting the abundance of aerobic germs and Enterobacteriaceae.

Abstract Background Over the last years, several strategies have been reported to improve the metabolic stability of minigastrin analogs. However, currently applied compounds still reveal limited in vitro and in vivo stability. We thus performed a glycine scan at the N-terminus of DOTA-MGS5 (DOTA-d-Glu-Ala-Tyr-Gly-Trp-(N-Me)Nle-Asp-1-Nal) to systematically analyze the peptide structure. We substituted N-terminal amino acids by simple PEG spacers and investigated in vitro stability in human serum. Furthermore, we evaluated different modifications on its tetrapeptide binding sequence (H-Trp-(N-Me)Nle-Asp-1-Nal-NH2). Results Affinity data of all glycine scan peptides were found to be in a low nanomolar range (4.2–8.5 nM). However, a truncated compound lacking the d-γ-Glu-Ala-Tyr sequence revealed a significant loss in CCK-2R affinity. Substitution of the d-γ-Glu-Ala-Tyr-Gly sequence of DOTA-γ-MGS5 (DOTA- d-γ-Glu-Ala-Tyr-Gly-Trp-(N-Me)Nle-Asp-1-Nal-NH2) by polyethylene glycol (PEG) spacers of different length exhibited only a minor influence on CCK-2R affinity and lipophilicity. However, in vitro stability of the PEG-containing compounds was significantly decreased. In addition, we confirmed that the tetrapeptide sequence H-Trp-Asp-(N-Me)Nle-1-Nal-NH2 is indeed sufficient for high CCK-2R affinity. Conclusion We could demonstrate that a substitution of d-γ-Glu-Ala-Tyr-Gly by PEG spacers simplified the peptide structure of DOTA-MGS5 while high CCK-2R affinity and favorable lipophilicity were maintained. Nevertheless, further optimization with regard to metabolic stability must be carried out for these minigastrin analogs.

As the population ages, the prevalence of heart failure and individuals wearing an implanted cardiac device is increasing. The combination of different underlying pathophysiologies and (the combination of) implanted cardiac devices can become a challenge with regard to the determination of cause and manner of death in such individuals. Additionally, heart disease is frequently associated with mental disease, ranging from anxiety and depression to suicidality and suicide (attempts). At the same time, the correct diagnosis of cause and manner of death is the basis for quality assurance, further therapeutic advances, legal safety, and suicide prevention. By that, an interdisciplinary field between legal medicine, clinicians, and law enforcement opens up. In this field, the different participants can simultaneously benefit from and need each other. For example, legal medicine experts need investigatory results and clinical expertise for the interpretation of readout data of implanted cardiac devices in order to correctly determine the cause of death. A correctly determined cause of death can assist law enforcement and help clinicians to further improve various therapeutic approaches based on correct mortality data collection. In addition, it is the basis for identification of suicides of device carriers, allowing psychological and psychiatric experts to better understand the burden of mental disease in this particular cohort. Against this interdisciplinary background, this manuscript summarizes information about psychiatric comorbidities and suicidality while being on a device. Thereby, basic information on complications and malfunctions of implanted cardiac devices, device-associated deaths with particular emphasis on device manipulation is displayed as basic information needed for correct determination of the cause of death. Also, legal and ethical issues in this field are outlined. The final result is a proposal of an interdisciplinary assessment workflow for a conjoint approach to improve the diagnosis of deaths associated with implanted cardiac devices. It will allow for a differentiation between an individual who died with or due to the device.

Non-thermal plasma (NTP) has proven to be a green method in the agricultural field for the stimulation of germination, growth, and production of nutraceutical compounds in some cases. However, the process is far from being fully understood and depends on the targeted plant species and the NTP used. In this work, we focus on the production of alfalfa sprouts from NTP-treated seeds under different voltage conditions. A flexible electrode configuration was used to produce the NTP, which can also be placed on packages for in-package treatments. The surface of the seeds was analyzed, indicating that the microstructure was strongly affected by NTP treatment. Biometric measurements evidenced the possibility of stimulating the sprout growth in some conditions by up to 50% compared to the sprouts obtained from untreated seeds. Biochemical traits for the sprouts obtained in different processing conditions were also studied, such as the concentrations of chlorophyll pigments, flavonoids and polyphenols, and antioxidant activity. Most NTP treatments led to inhibitory effects, proving the strong dependence between NTP treatment and targeted plant species.

This study aims to investigate a marlstone and claystone succession located at the southern end of the Eastern Carpathians, a region where oceanic anoxic event 2 (OAE2) has not been pointed out so far. Toward the upper half of this succession, a 17-cm-thick black shale was identified. The investigated depositional interval lies within the late Cenomanian–early Turonian, encompassing the UC3d up to UC7 nannofossil zones. The δ13C values fluctuated between 2.06‰ and 3.89‰, showing a positive isotope excursion that was assigned to OAE2. The δ13C isotope curve displays the following intervals: pre-excursion, first build-up, trough, second build-up, plateau, and post-excursion. Within the second build-up interval of OAE2, a substantial shift in CaCO3 values, accompanied by high concentrations of total organic carbon and a significant decline in the abundance and diversity of calcareous nannofossil assemblages, was observed. The nannofossil turnover related to OAE2 climax revealed predominance of Watznaueria barnesiae and temporary disappearance from the record of surface-water higher fertility taxa, such as Biscutum constans, Zeugrhabdotus erectus, and Discorhabdus ignotus. Above OAE2, peaks of Eprolithus floralis, followed by increased abundance of Eiffellithus turriseiffelii and Nannoconus spp., were identified. In the lower part of the studied succession (i.e., the upper Cenomanian UC3d nannofossil subzone), during the pre-excursion characterized by low δ13C values and less negative δ18O values, a small group of nannofossils more related to mid- and high-paleolatitudes, such as Crucibiscutum salebrosum, Repagulum parvidentatum, and Seribiscutum primitivum, is present, always showing a low abundance.

Abstract In cirrhotic patients with hepatocellular carcinoma (HCC), right-sided radioembolization (RE) with Yttrium-90-loaded microspheres is an established palliative therapy and can be considered a “curative intention” treatment when aiming for sequential tumor resection. To become surgical candidate, hypertrophy of the left liver lobe to > 40% (future liver remnant, FLR) is mandatory, which can develop after RE. The amount of radiation-induced shrinkage of the right lobe and compensatory hypertrophy of the left lobe is difficult for clinicians to predict. This study aimed to utilize machine learning to predict left lobe liver hypertrophy in patients with HCC and cirrhosis scheduled for right lobe RE, with external validation. The results revealed that machine learning can accurately predict relative and absolute volume changes of the left liver lobe after right lobe RE. This prediction algorithm could help to estimate the chances of conversion from palliative RE to curative major hepatectomy following significant FLR hypertrophy.
