Research in rodents has shown that dietary vitamin A reduces body fat by enhancing fat mobilisation and energy utilisation; however, their effects in growing dogs remain unclear. In the present study, we evaluated the development of body weight and body composition and compared observed energy intake with predicted energy intake in forty-nine puppies from two breeds (twenty-four Labrador Retriever (LAB) and twenty-five Miniature Schnauzer (MS)). A total of four different diets with increasing vitamin A content between 5·24 and 104·80 μmol retinol (5000-100 000 IU vitamin A)/4184 kJ (1000 kcal) metabolisable energy were fed from the age of 8 weeks up to 52 (MS) and 78 weeks (LAB). The daily energy intake was recorded throughout the experimental period. The body condition score was evaluated weekly using a seven-category system, and food allowances were adjusted to maintain optimal body condition. Body composition was assessed at the age of 26 and 52 weeks for both breeds and at the age of 78 weeks for the LAB breed only using dual-energy X-ray absorptiometry. The growth curves of the dogs followed a breed-specific pattern. However, data on energy intake showed considerable variability between the two breeds as well as when compared with predicted energy intake. In conclusion, the data show that energy intakes of puppies particularly during early growth are highly variable; however, the growth pattern and body composition of the LAB and MS breeds are not affected by the intake of vitamin A at levels up to 104·80 μmol retinol (100 000 IU vitamin A)/4184 kJ (1000 kcal).

Objective: To develop morphometric equations for prediction of body composition and create a body fat index (BFI) system to estimate body fat percentage in overweight and obese cats., Design: Prospective evaluation study., Animals: 76 overweight or obese cats ≥ 1 year of age., Procedures: Body condition score (BCS) was determined with a 5-point scale, morphometric measurements were made, and dual-energy x-ray absorptiometry (DEXA) was performed. Visual and palpation-based evaluation of various body regions was conducted, and results were used for development of the BFI system. Best-fit multiple regression models were used to develop equations for predicting lean body mass and fat mass from morphometric measurements. Predicted values for body composition components were compared with DEXA results., Results: For the study population, prediction equations accounted for 85% of the variation in lean body mass and 98% of the variation in fat mass. Values derived from morphometric equations for fat mass and lean mass were within 10% of DEXA values for 55 of 76 (72%) and 66 of 76 (87%) cats, respectively. Body fat as a percentage of total body weight (ie, body fat percentage) predicted with the BCS and BFI was within 10% of the DEXA value for 5 of 39 (13%) and 22 of 39 (56%) cats, respectively., Conclusions and Clinical Relevance: The BFI system and morphometric equations were considered accurate for estimation of body composition components in overweight and obese cats of the study population and appeared to be more useful than BCS for evaluation of these patients. Further research is needed to validate the use of these methods in other feline populations.

Objective: To develop morphometric equations for prediction of body composition and create a body fat index (BFI) to estimate body fat percentage in overweight and obese dogs., Design: Prospective evaluation study., Animals: 83 overweight or obese dogs ≥ 1 year of age., Procedures: Body condition score (BCS) was assessed on a 5-point scale, morphometric measurements were made, and visual and palpation-based assessments and dual-energy x-ray absorptiometry (DEXA) were performed. Equations for predicting lean body mass, fat mass, and body fat as a percentage of total body weight (ie, body fat percentage) on the basis of morphometric measurements were generated with best-fit statistical models. Visual and palpation-based descriptors were used to develop a BFI. Predicted values for body composition components were compared with DEXA-measured values., Results: For the study population, the developed morphometric equations accounted for 98% of the variation in lean body mass and fat mass and 82% of the variation in body fat percentage. The proportion of dogs with predicted values within 10% of the DEXA values was 66 of 83 (80%) for lean body mass, 56 of 83 (68%) for fat mass, and 56 of 83 (67%) for body fat percentage. The BFI accurately predicted body fat percentage in 25 of 47 (53%) dogs, whereas the value predicted with BCS was accurate in 6 of 47 (13%) dogs., Conclusions and Clinical Relevance: Morphometric measurements and the BFI appeared to be more accurate than the 5-point BCS method for estimation of body fat percentage in overweight and obese dogs. Further research is needed to assess the applicability of these findings to other populations of dogs.

Objective: To evaluate subchondral joint space width measurements (SJSW) and CT-osteoabsorptiometry (CTOAM) measurements in the elbow of dogs with naturally occurring fragmented medial coronoid process (FMCP) preoperatively and 6 months postoperatively, and to compare these to normal dogs., Study Design: Prospective randomized clinical trial., Animals: Large breed dogs <1 year of age (n = 15)., Methods: Arthroscopic FMCP debridement was performed, followed by randomization to a proximal ulnar ostectomy with IM pin (PUO). CTs were performed pre-operatively and 6 months post-operatively. CTOAM and SJSW measurements were acquired in 7 locations within the joint and compared to an age-matched normal control group. Pearson correlation was performed on SJSW and CTOAM measurements. A linear mixed model determined the effect of disease and treatment on SJSW and CTOAM measurements., Results: Fifteen dogs (28 elbows) with FMCP participated (11 arthroscopic FMCP removal, 17 with additional PUO). Data were normally distributed. Pearson correlation between CTOAM and SJSW measurements showed moderate to strong negative correlation in the control dogs. Preoperatively, affected elbows had lower medial compartment and higher lateral coronoid process CTOAM values than normal elbows. After treatment, CTOAM values of the medial compartment increased to normal. Treatment with PUO did not affect SJSW or CTOAM., Conclusions: Our data agree with these previous studies suggesting lower subchondral plate mineralization in dogs affected by FMCP. This prospective clinical study examined the effect of obesity and subsequent weight loss on oxygenation and ventilation during deep sedation in pet dogs. Data from nine dogs completing a formalised weight loss programme were evaluated. Dual-energy X-ray absorptiometry (DEXA) was used to quantify body fat mass prior to and after weight loss. Dogs were deeply sedated and positioned in dorsal recumbency. Sedation was scored using a semi-objective scheme. As part of the monitoring of sedation, arterial oxygen partial pressure (PaO2) and arterial carbon dioxide partial pressure (PaCO2) were measured after 10 min in dorsal recumbency. Oxygen saturation of haemoglobin (SpO2) was monitored continuously using pulse oximetry, starting oxygen supplementation where indicated (SpO2<90%) via a face mask. Morphometric measurements were taken from DEXA images and compared before and after weight loss. Several oxygen indices were calculated and correlated with body fat variables evaluated by DEXA. All body fat variables improved significantly after weight loss. PaO2 increased from 27.9±19.2 kPa to 34.8±24.4 kPa, while FiO2 decreased from 0.74±0.31 to 0.66±0.35. Morphometric measurements improved significantly after weight loss. PaO2/FiO2 (inspired oxygen fraction) and Pa/AO2 (ratio of PaO2 to alveolar PO2) also improved significantly, but there was no change in f-shunt and PaCO2 after weight loss. On multiple linear regression analysis, all oxygen indices were negatively associated with thoracic fat percentage. In conclusion, obesity decreases oxygenation in dogs during deep sedation. Objective: To evaluate the influence of acidifying or alkalinizing diets on bone mineral density and urine relative supersaturation (URSS) with calcium oxalate and struvite in healthy cats., Animals: 6 castrated male and 6 spayed female cats., Procedures: 3 groups of 4 cats each were fed diets for 12 months that differed only in acidifying or alkalinizing properties (alkalinizing, neutral, and acidifying). Body composition was estimated by use of dual energy x-ray absorptiometry, and 48-hour urine samples were collected for URSS determination., Results: Urine pH differed significantly among diet groups, with the lowest urine pH values in the acidifying diet group and the highest values in the alkalinizing diet group. Differences were not observed in other variables except urinary ammonia excretion, which was significantly higher in the neutral diet group. Calcium oxalate URSS was highest in the acidifying diet group and lowest in the alkalinizing diet group; struvite URSS was not different among groups. Diet was not significantly associated with bone mineral content or density., Conclusions and Clinical Relevance: Urinary undersaturation with calcium oxalate was achieved by inducing alkaluria. Feeding an alkalinizing diet was not associated with URSS with struvite. Bone mineral density and calcium content were not adversely affected by diet; therefore, release of calcium from bone caused by feeding an acidifying diet may not occur in healthy cats.

In a previous study, carnitine supplementation to piglets during the suckling period resulted in an increased total muscle fibre number at weaning in piglets of low birth weight. The objective of the present study was to investigate whether this effect is maintained until market age and whether this would attenuate the negative consequences of low birth weight on carcass and meat quality. Using a split-plot design with litter as block, sex as whole plot and treatment as subplot, the effects of early-postnatal l-carnitine supplementation on female and castrated male piglets of low birth weight were investigated on a total of 56 German Landrace piglets from 14 litters. From days 7 to 27 of age piglets were orally supplemented once daily with 400 mg of l-carnitine dissolved in 1 ml of water or received an equal volume of water without carnitine. From weaning (day 28) until slaughter (day 166 of age) all pigs were fed standard diets. At weaning, carnitine-supplemented piglets had a twofold increased concentration of free carnitine (P < 0.001) and a lower concentration of non-esterified fatty acids (P < 0.05) in blood plasma indicating that carnitine became bioavailable and increased fatty acid utilization during the period of supplementation. Growth performance was not influenced by treatment in any growth period. Dual-energy X-ray absorptiometry revealed no differences in body composition between groups in weeks 12, 16 and 20 of age. LW at slaughter, carcass weight, measures of meat yield and fat accretion, as well as body composition by chemical analyses and dissection of primal cuts did not differ between treatments. No differences between control and carnitine-treated pigs in total fibre number (P = 0.85) and fibre cross-sectional area (P = 0.68) in m. semitendinosus (ST) measured at slaughter could be observed. The carnitine group tended to exhibit a smaller proportion of slow-twitch oxidative fibres (P = 0.08), a greater proportion of fast-twitch glycolytic fibres (P = 0.11), and increased specific lactate dehydrogenase activity (P = 0.09) in ST indicating a more glycolytic muscle metabolism. Compared with the controls, a lower pH24 value was observed (P = 0.05) in ST muscle of carnitine-supplemented pigs, which - in castrates only - was associated with an increased drip loss (P < 0.01). Meat quality traits in m. longissimus were not influenced by treatment. In conclusion, our hypothesis that early-postnatal carnitine supplementation to piglets of low birth weight permanently increases myofibre number and improves later carcass and meat quality could not be confirmed by this experiment.

The objective of the study was to evaluate whether magnetic resonance imaging (MRI) offers the opportunity to reliably analyze body composition of pigs in vivo. Therefore, the relation between areas of loin eye muscle and its back fat based on MRI images were used to predict body composition values measured by dual energy X-ray absorptiometry (DXA). During the study, a total of 77 pigs were studied by MRI and DXA, with a BW ranging between 42 and 102 kg. The pigs originated from different extensive or conventional breeds or crossbreds such as Cerdo Iberico, Duroc, German Landrace, German Large White, Hampshire and Pietrain. A Siemens Magnetom Open was used for MRI in the thorax region between 13th and 14th vertebrae in order to measure the loin eye area (MRI-LA) and the above back fat area (MRI-FA) of both body sides, whereas a whole body scan was performed by DXA with a GE Lunar DPX-IQ in order to measure the amount and percentage of fat tissue (DXA-FM; DXA-%FM) and lean tissue mass (DXA-LM; DXA-%LM). A linear single regression analysis was performed to quantify the linear relationships between MRI- and DXA-derived traits. In addition, a stepwise regression procedure was carried out to calculate (multiple) regression equations between MRI and DXA variables (including BW). Single regression analyses showed high relationships between DXA-%FM and MRI-FA (R 2 = 0.89, √MSE = 2.39%), DXA-FM and MRI-FA (R 2 = 0.82, √MSE = 2757 g) and DXA-LM and MRI-LA (R 2 = 0.82, √MSE = 4018 g). Only DXA-%LM and MRI-LA did not show any relationship (R 2 = 0). As a result of the multiple regression analysis, DXA-LM and DXA-FM were both highly related to MRI-LA, MRI-FA and BW (R 2 = 0.96; √MSE = 1784 g, and R 2 = 0.95, √MSE = 1496 g). Therefore, it can be concluded that the use of MRI-derived images provides exact information about important 'carcass-traits' in pigs and may be used to reliably predict the body composition in vivo.

A number of parameters have been used to assess the impact ofcoccidiosis on chickens in clinical settings as well as in experimental studies. However, a rapid way to determine body composition would be useful to evaluate or compare responses to coccidia and could give further insight into the metabolic impact of infection. The current study evaluates the use of dual X-ray absorptiometry (DEXA) to determine the impact of coccidiosis on body composition in chicks receiving inoculations with single or mixed species of Eimeria. Chicks infected with Eimeria maxima, Eimeria acervulina, or Eimeria tenella had altered parameters of body composition as measured by DEXA at 6 days postinfection (PI). The greatest effects were noted in birds infected with E. acervulina or E. maxima, where lean mass and fat were reduced from control values about 75% and 85%, respectively. In chicks infected with E. tenella, tissue and fat were reduced about 10%. Bone mineral content (BMC) was about 75% of control values in birds infected with E. acervulina or E. maxima, but only E. acervulina altered bone mineral density (BMD). The decreases in BMC and BMD are likely due to malabsorption. In chicks receiving a mixed coccidian infection, all DEXA parameters were significantly decreased at 8 days PI compared with age-matched controls. As with single infections, BMD and BMC were significantly depressed (P < 0.05). Values of all DEXA parameters were near 92% of control values by day 16 PI. Analysis of all birds in the current study indicates DEXA tissue weight slightly underestimated the gravimetrically measured weight by about 3%. The current results demonstrate that DEXA is a potentially important tool for the rapid evaluation of the effect of coccidiosis on broiler chicks and suggest it can be useful for evaluation of vaccines and other disease controls.

Objective: To assess dual-energy x-ray absorptiometry (DXA) for evaluating effects of diet and environment on bone mineral density in Hermann's tortoises (Testudo hermanni)., Animals: 26 Hermann's tortoises within 1 month after hatching., Procedures: Group 1 was housed in an artificial setting and fed naturally growing vegetation. Group 2 was housed in an artificial setting and fed vegetables grown for human consumption. Group 3 was maintained in an outside enclosure and fed naturally growing vegetation. After 10 months, pyramidal growth, body weight, and adverse conditions were assessed. Bone mineral density (BMD) of the axial and appendicular skeleton, shell, vertebral column, and pelvis was measured via DXA., Results: Group 2 had the highest mean ± SD body weight (65.42 ± 30.85 g), followed by group 1 (51.08 ± 22.92 g) and group 3 (35.74 ± 7.13 g). Mean BMD of the shell varied significantly among groups (group 1, 0.05 ± 0.03 g/cm(2)•m; group 2, 0.09 ± 0.15 g/cm(2)•m; and group 3, undetectable). The BMD of the axial and appendicular skeleton, vertebral column, and pelvis did not differ significantly among groups. Pyramidal growth was highest in group 1 and not evident in group 3., Conclusions and Clinical Relevance: Tortoises raised in artificial conditions did not have deficits in BMD, compared with results for outdoor-housed hibernating tortoises. Supplemental calcium was apparently not necessary when an adequate photothermal habitat and plant-based diet were provided. Higher BMD of captive-raised tortoises was morphologically associated with a higher incidence of pyramidal growth in captive-raised groups.

Objective: To compare quantitative magnetic resonance (QMR), dual-energy x-ray absorptiometry (DXA), and deuterium oxide (D2O) methods for measurement of total body water (TBW), lean body mass (LBM), and fat mass (FM) in healthy dogs and to assess QMR accuracy., Animals: 58 Beagles (9 months to 11.5 years old)., Procedures: QMR scans were performed on awake dogs. A D2O tracer was administered (100 mg/kg, PO) immediately before dogs were sedated, which was followed by a second QMR or DXA scan. Jugular blood samples were collected before and 120 minutes after D2O administration., Results: TBW, LBM, and FM determined via QMR were not significantly different between awake or sedated dogs, and means differed by only 2.0%, 2.2%, and 4.3%, respectively. Compared with results for D2O dilution, QMR significantly underestimated TBW (10.2%), LBM (13.4%), and FM (15.4%). Similarly, DXA underestimated LBM (7.3%) and FM (8.4%). A significant relationship was detected between FM measured via D2O dilution and QMR (r(2) > 0.89) or DXA (r(2) > 0.88). Even though means of TBW and LBM differed significantly between D2O dilution and QMR or DXA, values were highly related (r(2) > 0.92)., Conclusions and Clinical Relevance: QMR was useful for determining body composition in dogs and can be used to safely and rapidly acquire accurate data without the need for sedation or anesthesia. These benefits can facilitate frequent scans, particularly in geriatric, extremely young, or ill pets. Compared with the D2O dilution method, QMR correction equations provided accurate assessment over a range of body compositions.

Objective: To compare quantitative magnetic resonance (QMR), dual-energy x-ray absorptiometry (DXA), and deuterium oxide (D2O) dilution methods for measurement of total body water (TBW), lean body mass (LBM), and fat mass (FM) in healthy cats and to assess QMR precision and accuracy., Animals: Domestic shorthair cats (58 and 32 cats for trials 1 and 2, respectively)., Procedures: QMR scans of awake cats performed with 2 units were followed by administration of D2O tracer (100 mg/kg, PO). Cats then were anesthetized, which was followed by QMR and DXA scans. Jugular blood samples were collected before and 120 minutes after D2O administration., Results: QMR precision was similar between units (coefficient of variation < 2.9% for all measures). Fat mass, LBM, and TBW were similar for awake or sedated cats and differed by 4.0%, 3.4%, and 3.9%, respectively, depending on the unit. The QMR minimally underestimated TBW (1.4%) and LBM (4.4%) but significantly underestimated FM (29%), whereas DXA significantly underestimated LBM (9.2%) and quantitatively underestimated FM (9.3%). A significant relationship with D2O measurement was detected for all QMR (r(2) > 0.84) and DXA (r(2) > 0.84) measurements., Conclusions and Clinical Relevance: QMR was useful for determining body composition in cats; precision was improved over DXA. Quantitative magnetic resonance can be used to safely and rapidly acquire data without the need for anesthesia, facilitating frequent monitoring of weight changes in geriatric, extremely young, or ill pets. Compared with the D2O dilution method, QMR correction equations provided accurate data over a range of body compositions.

Two health problems have plagued captive common marmoset (Callithrix jacchus) colonies for nearly as long as those colonies have existed: marmoset wasting syndrome and metabolic bone disease. While marmoset wasting syndrome is explicitly linked to nutrient malabsorption, we propose metabolic bone disease is also linked to nutrient malabsorption, although indirectly. If animals experience negative nutrient balance chronically, critical nutrients may be taken from mineral stores such as the skeleton, thus leaving those stores depleted. We indirectly tested this prediction through an initial investigation of digestive efficiency, as measured by apparent energy digestibility, and serum parameters known to play a part in metabolic bone mineral density of captive common marmoset monkeys. In our initial study on 12 clinically healthy animals, we found a wide range of digestive efficiencies, and subjects with lower digestive efficiency had lower serum vitamin D despite having higher food intakes. A second experiment on 23 subjects including several with suspected bone disease was undertaken to measure digestive and serum parameters, with the addition of a measure of bone mineral density by dual-energy X-ray absorptiometry (DEXA). Bone mineral density was positively associated with apparent digestibility of energy, vitamin D, and serum calcium. Further, digestive efficiency was found to predict bone mineral density when mediated by serum calcium. These data indicate that a poor ability to digest and absorb nutrients leads to calcium and vitamin D insufficiency. Vitamin D absorption may be particularly critical for indoor-housed animals, as opposed to animals in a more natural setting, because vitamin D that would otherwise be synthesized via exposure to sunlight must be absorbed from their diet. If malabsorption persists, metabolic bone disease is a possible consequence in common marmosets. Kyphosis is an idiopathic disease characterized by abnormal, outward spinal curvature. A spontaneous outbreak and subsidence of kyphosis over a 4-mo period in the University of Wisconsin Swine Research and Teaching Center herd coincided with an accidental omission of vitamin D(3) in 1 of 2 premixes used in sow diets. This controlled experiment was conducted to determine whether vitamin D deletion from premixes used in sow diets would induce kyphosis in their offspring. Crossbred (Landrace × Large White), multiparous sows (n = 8) were fed corn-soybean meal diets supplemented with either 325 IU vitamin D(3)/kg (+D) or 45 IU vitamin D(3)/kg (-D) diet from breeding through lactation. The vitamin D concentrations duplicated formulations of diets fed during the earlier spontaneous outbreak. At weaning (approximately 4 wk), pigs were fed diets devoid of supplemental vitamin D and formulated to supply either 120% of the Ca and P requirements (HCaP) or 80% of the Ca and P requirements (LCaP) until wk 9. At wk 9, all pigs were fed the HCaP diet until wk 13. No evidence of kyphosis was observed in pigs at weaning. Pigs produced by -D sows and fed LCaP diets exhibited a 17% incidence (4/23 pigs) of kyphosis at wk 9. At wk 13, the incidence of kyphosis had increased to 32% (6/19 pigs). Unexpectedly at wk 13, pigs produced by +D sows and fed LCaP diets exhibited a 26% incidence (5/19 pigs) of kyphosis. None of the pigs fed HCaP diets from wk 4 to 13 displayed kyphosis, regardless of maternal diets. Evidence of kyphosis was detected at a younger age if pigs were produced by sows fed -D diets. Whole body and femur bone mineral content determined with dual energy X-ray absorptiometry were reduced (P < 0.05) in pigs fed LCaP vs. HCaP diets, but pigs produced by -D sows were more severely affected. Femur bending moments were reduced (P < 0.05) at wk 9 and 13 in pigs fed LCaP vs. HCaP diets. At wk 13, pigs produced by -D sows and fed LCaP diets had reduced (P < 0.05) bone mineral density and femur yield bending moment compared with pigs from +D sows fed LCaP diets. In conclusion, the 20 to 30% incidence of kyphosis induced by altering vitamin D, Ca, and P concentrations in maternal and nursery diets mimics the incidence observed in spontaneous outbreaks in afflicted herds. A reproducible vitamin D-induced kyphosis in young pigs offers a suitable model to study skeletal tissue characteristics, fetal skeletal tissue development, and potential treatments for pigs and human patients afflicted by this disease.

A total of 72 male weaned pigs were used in a 110-day study to investigate the effect of feeding genetically modified (GM) Bt MON810 maize on selected growth and health indicators. It was hypothesised that in pigs fed Bt maize, growth and health are not impacted compared with pigs fed isogenic maize-based diets. Following a 12-day basal period, pigs (10.7 ± 1.9 kg body weight (BW); ∼40 days old) were blocked by weight and ancestry and randomly assigned to treatments: (1) non-GM maize diet for 110 days (non-GM), (2) GM maize diet for 110 days (GM), (3) non-GM maize diet for 30 days followed by GM maize diet up to day 110 (non-GM/GM) and (4) GM maize diet for 30 days followed by non-GM maize diet up to day 110 (GM/non-GM). BW and daily feed intake were recorded on days 0, 30, 60 and 110 (n = 15). Body composition was determined by dual energy X-ray absorptiometry (n = 10) on day 80. Following slaughter on day 110, organs and intestines were weighed and sampled for histological analysis and urine was collected for biochemical analysis (n = 10). Serum biochemistry analysis was performed on days 0, 30, 60, 100 and 110. Growth performance and serum biochemistry were analysed as repeated measures with time and treatment as main factors. The slice option of SAS was used to determine treatment differences at individual time points. There was no effect of feeding GM maize on overall growth, body composition, organ and intestinal weight and histology or serum biochemistry on days 60 and 100 and on urine biochemistry on day 110. A treatment × time interaction was observed for serum urea (SU; P < 0.05), creatinine (SC; P < 0.05) and aspartate aminotransferase (AST; P < 0.05). On day 30, SU was lower for the non-GM/GM treatment compared with the non-GM, GM and GM/non-GM treatments (P < 0.05). On day 110, SC was higher for the non-GM/GM and GM/non-GM treatments compared with non-GM and GM treatments (P < 0.05). Overall, serum total protein was lower for the GM/non-GM treatment compared with the non-GM/GM treatment (P < 0.05). The magnitude of change observed in some serum biochemical parameters did not indicate organ dysfunction and the changes were not accompanied by histological lesions. Long-term feeding of GM maize to pigs did not adversely affect growth or the selected health indicators investigated.

Objective: To assess effects of foods fortified with docosahexaenoic acid (DHA)-rich fish oil on cognitive, memory, psychomotor, immunologic, and retinal function and other measures of development in healthy puppies., Design: Evaluation study., Animals: 48 Beagle puppies., Procedures: Puppies were assigned to 3 groups after weaning (n = 16/group) and received 1 of 3 foods (low-DHA, moderate-DHA, or high-DHA food) as their sole source of nutrition until 1 year of age. Visual discrimination learning and memory tasks, psychomotor performance tasks, and physiologic tests including blood and serum analysis, electroretinography, and dual-energy x-ray absorptiometry were performed at various time points. Anti-rabies virus antibody titers were evaluated 1, 2, 4, and 8 weeks after vaccination at 16 weeks of age., Results: Foods had similar proximate analysis results but varied in concentration of DHA from fish oil; the high-DHA food also contained higher concentrations of vitamin E, taurine, choline, and l-carnitine than did other foods. The high-DHA group had significantly better results for reversal task learning, visual contrast discrimination, and early psychomotor performance in side-to-side navigation through an obstacle-containing maze than did the moderate-DHA and low-DHA groups. The high-DHA group had significantly higher anti-rabies antibody titers 1 and 2 weeks after vaccination than did other groups. Peak b-wave amplitudes during scotopic electroretinography were positively correlated with serum DHA concentrations at all evaluated time points., Conclusions and Clinical Relevance: Dietary fortification with fish oils rich in DHA and possibly other nutrients implicated in neurocognitive development following weaning improved cognitive, memory, psychomotor, immunologic, and retinal functions in growing dogs.

Objectives: To compare the ability of N-terminal telopeptide (NTx) assays and dual-energy x-ray absorptiometry (DEXA) to detect bone resorption in dogs with nonneoplastic bone lysis and evaluate the correlation between these diagnostic tools., Study Design: Prospective, cross-sectional clinical study., Animals: Dogs (n = 35; 39 femoral heads) that had femoral head and neck ostectomy and 6 cadaver specimens from healthy immature small dogs., Methods: Small dogs with avascular necrosis (n = 12), a reference group of small dogs (7), large dogs with degenerative joint disease (DJD; 10), and large dogs with trauma (10) were studied in addition to 6 femoral heads harvested from 6 small immature and healthy dogs euthanatized for reasons unrelated to this study. Densitometric measurements of femoral heads, urine NTx excretion, and serum NTx concentration were compared between groups., Results: Avascular necrosis resulted in a decrease in bone mineral density (BMD) (0.18 ± 0.01 g/cm(2;) P < .01) of the femoral head and elevation of serum NTx (159.3 ± 59.4 nM; P = .03) compared to small dog controls (0.28 ± 0.02 g/cm(2) ; 18.7 ± 1.83 nM, respectively), but did not seem to affect urine NTx. DJD in large dogs did not seem to affect any of the densitometric parameters evaluated. BMD (P = .03) and serum NTx (P = .04) were lower in small compared to large dogs. Serum NTx and densitometric measurements correlate inversely with each other (P = .001) but neither test correlated with urine NTx (P = .8-.9)., Conclusion: Serum NTx levels vary with dog size but seem to correlate better with BMD better than urine NTx excretion in dogs with nonneoplastic bone resorption., (© Copyright 2012 by The American College of Veterinary Surgeons.)

Objective: To evaluate gene transfer in an equine metacarpal IV (MCIV) ostectomy model using adenoviral vectors encoding the human bone morphogenetic protein-2 and protein-7 gene (Ad-BMP-2/-7)., Experimental Animals: Healthy adult horses (n = 15)., Methods: A plate stabilized, critical size 1.5 cm ostectomy was created in left and right MCIV. The ostectomy site was injected with either Ad-green fluorescent protein (Ad-GFP) or Ad-hBMP-2/-7 at completion of surgery; the same treatment was assigned to both the left and right forelimb of each horse (n = 5 horses/group). Bone healing was evaluated radiographically every 2 weeks for 16 weeks. Horses in a pilot study (n = 5) were used as untreated controls for radiographic evaluation to 8 weeks. After euthanasia at 16 weeks bone healing was evaluated using dual energy X-ray absorptiometry (DEXA) and histomorphometry. Data were analyzed using an ANOVA or Kruskal-Wallis test. Level of significance was P < .05., Results: At 4 and 6 weeks, the Ad-GFP group had a significantly lower percentage defect ossification compared with the untreated control group. There was no significant difference between untreated and Ad-hBMP-2/-7 groups at any time point and no significant difference in bone healing radiographically, histologically, or using DEXA between any groups at 16 weeks., Conclusions: Ad-hBMP-2/-7 did not improve bone healing in horses at 16 weeks., (© Copyright 2012 by The American College of Veterinary Surgeons.)

A 2.3-y-old female cynomolgus macaque (Macaca fascicularis) presented with a broken right tibia and fibula. Radiographs showed multiple cyst-like defects in all long bones. We suspected that both fractures were pathologic because they occurred through these defects. Ultrasonography, MRI, and dual X-ray absorptiometry revealed that the defects were filled with soft tissue. Grossly, the bones were abnormal in shape, and a gelatinous material filled the defects and the surrounding marrow cavity. Histologically, the gelatinous material was composed of fibrin and cartilage; few normal bone cells were seen. Genetic testing revealed extra material on the short arm of chromosome 8 in all tissues examined, but no copy number alterations of likely clinical significance were observed, and no abnormalities were found that were unique to the lesions. In light of the clinical signs and radiographic and pathologic findings, polyostotic fibrous dysplasia was diagnosed. This report represents the first documented case of fibrous dysplasia in a cynomolgus macaque.

Objective: To evaluate the precision and accuracy of assessing bone mineral density (BMD) by use of mean gray value (MGV) on digitalized and digital images of conventional and digital radiographs, respectively, of ex vivo bovine and equine bone specimens in relation to the gold-standard technique of dual-energy x-ray absorptiometry (DEXA)., Sample: Left and right metatarsal bones from 11 beef cattle and right femurs from 2 horses., Procedures: Bovine specimens were imaged by use of conventional radiography, whereas equine specimens were imaged by use of computed radiography (digital radiography). Each specimen was subsequently scanned by use of the same DEXA equipment. The BMD values resulting from each DEXA scan were paired with the MGVs obtained by use of software on the corresponding digitalized or digital radiographic image., Results: The MGV analysis of digitalized and digital x-ray images was a precise (coefficient of variation, 0.1 and 0.09, respectively) and highly accurate method for assessing BMD, compared with DEXA (correlation coefficient, 0.910 and 0.937 for conventional and digital radiography, respectively)., Conclusions and Clinical Relevance: The high correlation between MGV and BMD indicated that MGV analysis may be a reliable alternative to DEXA in assessing radiographic bone density. This may provide a new, inexpensive, and readily available estimate of BMD.

Body condition scoring (BCS) is a subjective semiquantitative method of assessing body fat and muscle by palpation of key anatomic features. A previously published BCS system for rhesus macaques (Macaca mulatta) uses a scale comprising both whole and half units, in which the midrange represents optimal body condition (3.0), lower values represent emaciated to lean conditions (1.0 to 2.0), and higher values (4.0 to 5.0) indicate excessive body fat. A valid BCS system is well described, relevant to the species, has agreement within and between raters, and is consistent with objective measures. Here we correlate the subjective BCS assigned during physical exam with percentage body fat as determined by dual-energy X-ray absorptiometry (DEXA). Adult rhesus monkeys from an indoor-housed breeding colony were evaluated by the veterinary staff and assigned to 1 of 9 BCS score groups to give a minimum of 6 animals in each group. DEXA was used to obtain objective body composition measurements for macaques in each BCS group. Animals in the 'optimal' BCS group (3.0) had 25% body fat on average. Each full unit change in BCS was associated with an approximate 10% change in body fat percentage for macaques in the 2.0-to-5.0 BCS range. Absolute body fat in animals with BCS of 1.0 or 1.5 may be too low for accurate assessment by DEXA.

The bone mineral density (BMD) and the bone mineral content (BMC) in the bone tissue of the bilateral first phalanges of horses' thoracic limbs were analysed. The research material consisted of isolated pastern bones derived from 22 horses. The research was conducted with the use of a Norland model Excell Plus densitometer (Fort Atkinson WI, USA), using affinited beam X-ray technology and an animal research programme (Research Scan, 3.9.6. version) at the following parameters: scanning resolution of 1.5 x 1.5 mm, scanning speed 60 mm/s. The differences between BMC and BMD values in bilateral first phalanges in the thoracic limbs in horses were found to be nonsignificant. It also appeared that there are statistically significant positive correlations between values of the left and right bone of both analysed variables.

Intermittent administration of parathyroid hormone (PTH) is an anabolic therapy for osteoporotic conditions in humans. This study evaluated the effects of equine PTH fragment (ePTH-1-37) administration on bone metabolism in 12 healthy horses. Six horses each were treated once daily for 120days with subcutaneous injections of 0.5μg/kg ePTH-1-37 or placebo. Blood was collected to determine ionized calcium (Ca(++)), total Ca (Ca(T)), inorganic phosphorus, serum equine osteocalcin (eOC), carboxy-terminal telopeptide of type I collagen (ICTP), bone-specific alkaline phosphatase, and carboxy-terminal cross-linked telopeptide of type I collagen. Bone mineral density (BMD) was determined with dual X-ray absorptiometry of the metacarpus and calcaneus. Significantly higher blood Ca(++) and plasma Ca(T) concentrations were measured 5h after ePTH-1-37 administration compared to placebo. Higher serum eOC concentrations were found for ePTH-1-37 treatment at days 90 (P<0.05) and 120 (P=0.05). Significantly higher serum ICTP levels were observed with ePTH-1-37 treatment at days 60 and 90. For both study groups, BMD increased significantly in the calcaneus. Long-term use of ePTH-1-37 seemed to have no negative effects on bone metabolism in healthy horses. Background: The metabolic syndrome is common in populations exposed to a typical Western diet. There is a lack of an animal model that mimics this condition., Methods: We fed 15 cynomolgus monkeys ad libitum a high-sugar high-fat (HSHF) diet for 33 weeks. Body weight, body composition, serum lipids, and insulin were measured at baseline and at 33 weeks., Results: The animals tolerated the HSHF diet very well. In the intervention group, total serum cholesterol and low-density lipoprotein cholesterol were 3- and 5-fold higher, respectively, at 33 weeks as compared with their baseline levels. Serum high-density lipoprotein cholesterol and triglycerides were not significantly affected. Dual-energy X-ray absorptiometry (DXA) analysis of the intervention group indicated that the trunk fat mass increased by 187% during this period., Conclusions: Cynomolgus monkeys should be a useful model for investigating the interactions of diet and other factors such as genetics in the development of the metabolic syndrome., (© 2011 John Wiley & Sons A/S.)

Body condition scoring (BCS) systems primarily assess body fat. Both overweight and underweight animals may have loss of lean tissue that may not be noted using standard BCS systems. Catabolism of lean tissue can occur rapidly, may account for a disproportionate amount of body mass loss in sick cats and can have deleterious consequences for outcome. Therefore, along with evaluation of body fat, patients should undergo evaluation of muscle mass. The aims of the present study were first to evaluate the repeatability and reproducibility of a 4-point feline muscle mass scoring (MMS) system and second to assess the convergent validity of MMS by dual-energy X-ray absorptiometry (DXA). MMS was as follows: 3, normal muscle mass; 2, slight wasting; 1, moderate wasting; 0, severe wasting. For the first aim, forty-four cats were selected for evaluation based on age and BCS, and for the second aim, thirty-three cats were selected based on age, BCS and MMS. Cats were scored by ten different evaluators on three separate occasions. Body composition was determined by DXA. Inter- and intra-rater agreement were assessed using kappa analysis. Correlation between MMS and BCS, age, percentage lean body mass and lean body mass (LBM) was determined using Spearman's rank-order correlation. The MMS showed moderate inter-rater agreement in cats that scored normal or severely wasted (κ = 0.48-0.53). Intra-rater agreement was substantial (κ = 0.71-0.73). The MMS was significantly correlated with BCS (r 0.76, P < 0.0001), age (r - 0.75, P < 0.0001), LBM (g) (r 0.62, P < 0.0001) and percentage LBM (r - 0.49, P < 0.0035). Additional investigation is needed to determine whether the MMS can be refined and to assess its clinical applicability.

Migrating blackcaps (Sylvia atricapilla) were used to test the predictions that (1) the rebuilding of the digestive tract, as reflected by mass-specific consumption of food on the first 2-3 days of a stopover, is faster in birds with access to drinking water than in birds without, and (2) that adipose tissue and pectoral muscles grow faster and to a greater extent in birds with unlimited access to water. We simulated migratory stopover in two experiments. In Experiment I, each of 31 birds was randomly assigned to one of three experimental groups for 6 days. Along with mealworms (∼64% water) ad libitum, Group 1 received drinking water ad libitum; Group 2 had 0.5 h/day access to water; and Group 3 had no access to water. In Experiment II, 30 birds were offered a mixed diet for insectivorous birds (∼33% water) ad libitum for 6 days, while randomly assigned to two groups: (1) Water ad libitum-control; and (2) 30 min access to water twice a day. We measured lean mass and fat mass using dual energy X-ray absorptiometry, as well as body mass (m(b)), pectoral muscle index (PMI), and daily intake of food and water. Mean daily water intake was significantly different among the groups in both experiments. However, the availability of drinking water positively affected the rates of gain of lean and fat mass only in birds fed with the mixed, relatively dry diet. Furthermore, mass-specific daily food intake was affected by the availability of drinking water only in the mixed diet experiment, in which birds with unlimited access to drinking water reached an asymptote, 1 day earlier than birds in the water-restricted group. We suggest that in birds consuming diets with low water content, the lack of sufficient drinking water may result in slower rebuilding of the digestive tract, or may influence biochemical processes in the gut that result in slower growth of tissue. Although blackcaps obtained sufficient water from preformed and metabolic water to renew lost tissues when eating mealworms, given access to water, the birds drank prodigiously. Our results also suggest that if drinking water is unavailable to migrating blackcaps, their choices are restricted to water-rich foods, which may constrain their rate of feeding and thus the rate at which they deposit fat. Consequently, drinking water may have an important influence on birds' migratory strategies with respect to habitat selection, use of energy, and the saving of time.

During migration, birds undergo alternating periods of fasting and re-feeding that are associated with dynamic changes in body mass (m(b)) and in organ size, including that of the digestive tract. After arrival at a migratory stopover site, following a long flight, a bird must restore the tissues of its digestive tract before it can refuel. In the present study we examined how the availability of dietary protein influences refueling of migrating blackcaps (Sylvia atricapilla) during a migratory stopover. We tested the following predictions in blackcaps deprived of food and water for 1-2 days to induce stopover behavior: (1) birds provided with a low-protein diet will gain m(b), lean mass and fat mass, and increase in pectoral muscle size slower than do birds fed a high-protein diet; (2) since stopover time is shorter in spring, birds will gain m(b) and build up fat tissue and lean tissue faster than in autumn; and (3) if low dietary protein limits a bird's ability to gain m(b) and fat reserves, then birds that do not obtain enough protein will initiate migratory restlessness (Zugunruhe) earlier than will birds with adequate dietary protein. These predictions were tested by providing captured migrating blackcaps with semisynthetic isocaloric diets differing only in their protein content. Each day, we measured m(b), and food intake; also lean mass and fat mass were measured using dual energy X-ray absorptiometry. In addition, we monitored nocturnal activity with a video recording system. In both spring and autumn, birds fed diets containing either 3 or 20% protein increased in m(b), lean mass and fat mass at similar rates during the experiment. However, the group receiving 3% protein ate more than did the group receiving 20% protein. In support of our predictions, m(b), lean mass, fat mass, and intake of food all were higher in spring than in autumn. We also found that in spring all birds had higher levels of migratory restlessness, but birds fed 3% protein were less active at night than were birds fed 20% protein, possibly an adaptation conserving energy and protein. We conclude that protein requirements of migrating blackcaps during stopover are lower than expected, and that birds can compensate for low dietary protein by behavioral responses, i.e. hyperphagia and decreased migratory restlessness, that ensure rapid refueling.

Quantitative magnetic resonance (QMR) is a nuclear magnetic resonance-based method for measuring the fat, lean, and water content of the total body of the live animal. The purpose of this study was to evaluate the use of QMR for measuring the body composition of chickens while comparing QMR results to those obtained by dual x-ray absorptiometry (DXA) and chemical analysis (CA). A total of 191 birds were scanned live (nonanesthetized) by QMR, killed, and then scanned by DXA. The birds were Ross 708 broiler chickens and ranged in weight from 786 to 3,130 g. In addition, 48 of the carcasses were chemically analyzed for total body lipid, water, and ash content. Compared with CA, QMR underestimated the percentage of total body fat by 34% whereas DXA overestimated the percentage of fat by 50% (10.35 ± 3.35 by CA vs. 6.73 ± 3.90 by QMR and 15.55 ± 4.01 by DXA; P < 0.05). Both QMR and DXA measurements of percentage total body fat were highly correlated with the CA measurement (R(2) = 0.94 and 0.68, respectively). Both QMR and DXA estimates of total body water were close to the CA measurement (1,166 ± 277 g by CA vs. 1,214 ± 279 g by QMR and 1,217 ± 255 g by DXA; P > 0.05), with R(2) values of 0.90 and 0.91, respectively. Based on regression analysis, when prediction equations were applied to the entire group of birds, the QMR and DXA measurements of total body water and total body lean mass were in good agreement, with no significant difference (1,125 ± 244 g vs. 1,135 ± 246 g and 1,377 ± 311 g vs. 1,403 ± 309 g, respectively; P > 0.05) and highly correlated (R(2) = 0.97 for both). Likewise, the QMR measurement of total body fat agreed closely with that measured by DXA (164 ± 48 g and 167 ± 47 g, respectively) and was highly correlated (R(2) = 0.72). The results of this study demonstrate that with proper calibration, both QMR and DXA can provide accurate measurements of the body composition of chickens. The major advantage of the QMR method is that no anesthesia is required, thus facilitating the ease of measurement and repeated measurements.

Objective: To compare results of body condition scoring by use of a 9-point scale with body composition determined by dual-energy x-ray absorptiometry (DEXA) in indoor-confined neutered domestic shorthair (DSH) pet cats. Animals-72 indoor-confined, adult neutered DSH pet cats (38 females and 34 males)., Procedures: All cats underwent a physical examination including assessment of body weight (BW), body condition score (BCS; 1 = emaciated, 5 = ideal, and 9 = grossly obese), and girth. Urinalysis, CBC, and serum biochemical analysis were also performed. After the cats were confirmed healthy, they were anesthetized for body composition measurement via DEXA. Lean body mass, fat mass, and percentage body fat (%BF) were then evaluated., Results: The correlation between %BF and BCS (r = 0.87) was superior to the correlations between %BFand BW (r = 0.74) and between %BF and girth (r = 0.78). Values for %BF differed significantly between all pairs of BCSs except BCSs 8 and 9. Within a BCS, the %BF was similar for male and female cats. The mean %BF for cats with a BCS of 5 was 32, which exceeded the upper reference limit of %BF generally considered ideal (30)., Conclusions and Clinical Relevance: The 9-point BCS scale appears useful for assessing %BF in DSH pet cats. Nevertheless, study findings could indicate a need for redefining the ideal BCS for inactive neutered cats to include a BCS of 4.

Dual energy X-ray absorptiometry (DEXA) is a reference method for assessing body composition but is seldom `accessible in veterinary settings. Computed tomography (CT) can provide similar body composition estimates and we propose that it can be used in body composition studies in animals. We compared CT and DEXA data from 73 healthy adult neutered domestic cats. Three approaches for measuring adipose tissue percentage from full-body CT scans were explored. By examining the frequency distribution of voxels by Hounsfield unit (HU) value, it is possible to calculate a fat index (Fat%) that is in close agreement with the fat percentages obtained from DEXA scans. Fat% values obtained by the best of the methods had a mean difference of 0.96% (95% confidence interval 0.33-1.59%) from the DEXA results. Fat% obtained by the other two methods were characterized by good correlation but poor agreement and in one of the methods, the difference between the values from the two modalities was proportional to their mean. By using CT, it is possible to obtain body composition estimates that are in close agreement with those available using DEXA. While the significance of individual Fat% measurements obtained from CT can be difficult to interpret and to compare between centers, CT can contribute to research studies concerned either with nutrition or with obesity-related disorders., (© 2010 Veterinary Radiology & Ultrasound.)

The metacarpal bone mineral density of 136 healthy feedlot beef cattle of four different breeds (Charolaise, Limousine, Irish Crossbreed and Slovakian Crossbreed) raised and fed on standard conditions was measured by means of a dual-energy X-ray absorptiometry technique in an ex vivo study design. The average reference values (mean ± SD) of bone mineral density (BMD) for animals aged between 12 and 22 months and weighing between 236 and 546 kg have been reported and the effects of (i) breed, (ii) gender, (iii) age and (iv) body weight on bone mineral density have been considered. A significant difference (i) among different breeds and (ii) between genders resulted, whereas a high correlation between bone density and (iii) age and (iv) body weight was detected within the same breed and gender, with body weight being the most important factor affecting BMD. In the first part of this methodological study eleven metacarpi of 9 skeletally normal horses were examined from 4 directions by dual energy x-ray absorptiometry (DXA). The differences between the dorsopalmar-palmarodorsal and lateromedial-mediolateral (opposite sites) bone mineral density (BMD) values were found to be nonsignificant. In the second part of the study the precision of the Norland XR-26 densitometer was tested by measuring 34 metacarpal bones and 34 proximal phalanges, each of them three times, from a single direction. The difference between the individual measurements of the first phalanges and of the metacarpal bones originating from the right or the left side of the same horse were not significant, nor did the age or breed have a significant effect on BMD or bone mineral content (BMC). However, both BMD and BMC are greater in the metacarpal bones than in the proximal phalanges and are higher in geldings than in mares or to stallions, while the BMD or BMC values of mares and stallions did not differ from each other significantly. These data point to the necessity of further BMD studies in a higher number of patients.

The effect of dietary chlortetracycline (CTC) on the bone mineral density (BMD) of growing pigs was assessed using quantitative computed tomography (qCT) and dual energy X-ray absorptiometry (DXA). Pigs of 28 and 84 days old were given either drug-free (n=48) or CTC-fortified (800 ppm) diets for 56 days. At day 28, eight control pigs were selected for qCT and DXA, and their second lumbar vertebrae were removed. Eight control and eight CTC-treated animals were selected at days 84 and 140, respectively, and six pigs from each of these groups were assessed at day 168. Several CTC-exposure variables had significant effects (P<0.05) on the relationships between qCT-, trabecular- and vertebral body-BMD (R2 increments of 0.03 and 0.01, respectively) and between DXA-, trabecular- and vertebral body-BMD (R2 increments of 0.40 and 0.10, respectively). The findings of this study demonstrate that the qCT method is more reliable than the DXA technique in monitoring tetracycline-induced changes in porcine vertebral BMD. The magnitude of the effect of the drug treatment on the DXA-BMD estimation was surprisingly high, suggesting that DXA should not be used to monitor such changes in growing pigs. In contrast, the marginal influence of drug treatment on the qCT-BMD assessment suggests that this methodology is useful in this context. Objective: To assess performance of a portable bioimpedance monitor for measurement of body composition in dogs., Animals: 24 client-owned dogs., Procedures: Percentage body fat was measured via dual-energy x-ray absorptiometry (DEXA) and with a portable bioimpedance monitor, and body condition score (BCS) was measured by use of a 9-integer scale., Results: Although the precision of the bioimpedance monitor was good, this varied among dogs. Body position (standing vs sternal) had no effect on bioimpedance results. There was a significant association between results determined via DEXA and bioimpedance, but this association was weaker than between DEXA and BCS. When agreement was assessed via Bland-Altman plot, the bioimpedance monitor under- and overestimated values at high and low body fat percentages, respectively. In 9 dogs, body fat measurements were taken before and after weight loss to determine the proportional loss of tissue mass during weight management. There was a significant difference in the estimated percentage of weight lost as fat between the DEXA and bioimpedance methods., Conclusions and Clinical Relevance: Although percentage body fat measured by use of a portable bioimpedance monitor correlated well with values determined via DEXA, the imprecision and inaccuracy in dogs with high percentage body fat could make the monitor inappropriate for clinical practice.

The objectives of this clinical study were firstly, to assess the effects of breed/genetic group on body composition in dogs using Dual-X-ray absorptiometry (DEXA) as the reference method and secondly, to check, in clinical field conditions, if methods easy to apply in veterinary practice [bioelectrical impedance (BIA), morphometric equations, body condition score and body mass index] can give similar body composition results to DEXA results, using canine breeds with very different genetic and morphologic backgrounds. 19 dogs from 6 breeds with different genetic origin were used. Results showed that breed differences exist regarding body composition in dogs. Body condition score and morphometric equation should be developed by breed or by groups of breeds for an accurate estimation of body composition of the various breeds of dogs with a different genetic background or morphology. Any of the other tested methodologies (BIA or morphometric equations) gave results in agreement with DEXA value, when dogs with different genetic background and morphologic characteristics are used, but BIA, eventually in combination with morphometric measurements, could be a method to develop to estimate fat-free mass in dogs of different breeds., (Copyright 2009 Elsevier Ltd. Objective: To identify morphometric characteristics of the pelvic limb musculature associated with the development of cranial cruciate ligament (CCL) deficiency in Labrador Retrievers., Study Design: Cross-sectional clinical study., Animals: Pure-bred female (n=18) and male (n=12) Labrador Retrievers with (n=16) and without (n=14) CCL deficiency., Methods: Muscle conformation of pelvic limbs was evaluated by physical examination, radiography (widths of quadriceps, hamstring, and gastrocnemius were expressed relative to tibial length and to each other), and dual-energy X-ray absorptiometry (DEXA, lean contents of quadriceps, hamstring, and gastrocnemius were expressed relative to tibial length and to each other). Pelvic limbs of dogs without CCL deficiency were classified as normal (n=28 limbs), whereas those with CCL deficiency were considered diseased (n=18 limbs) or sound contralateral to CCL deficiency (n=10 limbs). Variables were compared between groups using mixed models analysis of variance, with P<.05 considered significant., Results: The ratios of quadriceps width to tibial length (P=.008), hamstring width (P=.013), and gastrocnemius width (P=.005) on lateral radiographs were lower in diseased limbs than controls. The mass of hamstring muscles in CCL deficient limbs was similar to that of normal limbs. The ratio of the lean content of gastrocnemius to hamstring muscles was greater in diseased (P=.007) and sound contralateral (P=.013) limbs than in normal limbs., Conclusions: Atrophy associated with CCL deficiency may predominantly affect the quadriceps muscle. Dominance of the gastrocnemius muscle over active restraints to the cranial tibial thrust may be associated with predisposition to CCL deficiency in Labrador Retrievers., Clinical Relevance: If confirmed, this dynamic imbalance between muscle groups of the rear limbs could serve as a basis for screening programs and preventive rehabilitation.

Two three-month-old, intact female Abyssinian cats were presented with a history of lameness, constipation and ataxia. The cats had been fed a diet composed almost exclusively of meat. Both showed severe osteopenia and multiple pathological fractures on radiography. Following euthanasia of the more severely affected cat, postmortem examination revealed changes consistent with nutritional secondary hyperparathyroidism and fibrous osteodystrophy, such as cortical thinning, massive connective tissue invasion in the diaphysis of long bones, and hypertrophy of the chief cells in both parathyroid glands. After introducing a balanced commercial diet to the surviving cat, bone mineralisation improved from the baseline value, and at subsequent examinations at three, six and 22 weeks later, as indicated by bone mineral density measurements obtained by dual-energy X-ray absorptiometry and computed tomography.

The aim of the study was to evaluate the bone mineral density, as well as the biomechanic and morphometric changes in the femur of ovariectomised rabbits. Twenty-four six-month-old New Zealand rabbits were randomly divided into an ovariectomy (n = 12) and a sham (n = 12) group. Six rabbits in each group were euthanatized at eight and 16 weeks after surgery, and the femora were resected. The morphometric data were obtained from tomographic images. Periosteal and endosteal diameters and cortical thickness were measured. Total cross- sectional, cortical and medullary areas were also measured. The bone mineral content, the bone area and the bone mineral density were measured from the proximal, distal and midshaft of the femur as well as the total femur by dual energy X-ray absorptiometry. Employing the three-point bending method, the ultimate force, stiffness and work-to-failure were measured. The mechanical data were normalised to obtain intrinsic biomechanical properties such as ultimate stress, elastic modulus, and toughness, all of which are independent of size and shape. The results indicated that the femur was both larger and weaker 16 weeks after surgery in the ovariectomised group. Results also suggest that the rabbit might be a useful animal model for investigation of diseases related to oestrogen loss such as human postmenopausal osteoporosis. However, additional studies with advanced techniques at several time points via in vivo animal studies, and precision and predictability analyses should be designed to standardise the rabbit as a model for osteoporosis.

The objective of this study was to confirm that weight loss after treatment with mitratapide (Yarvitan®) is loss of adipose tissue. Obese dogs were treated with the recommended treatment schedule of mitratapide. Dual-energy X-ray absorptiometry (DEXA) was done before and after the treatment schedule. Body weight, feed consumption and pelvic circumference were recorded and a glucose tolerance test was performed. Dualenergy X-ray absorptiometry measurements showed an impressive loss of fat tissue, corresponding to a mean loss of approximately 41.6% of the body fat mass recorded before treatment. After treatment with mitratapide, the mean body fat percentage had returned within the normal range. At the end of the study, the dogs had lost on average 14.2% of their body weight and 15.2% of their pelvic circumference compared to baseline. The results also suggest that losing weight with mitratapide might help to reverse insulin resistance.

Prior to starting a weight loss programme, target weight (TW) is often estimated, using starting body condition score (BCS). The current study assessed how well such estimates perform in clinical practice. Information on body weight, BCS and body composition was assessed before and after weight loss in 28 obese, client-owned dogs. Median decrease in starting weight per BCS unit was 10% (5-15%), with no significant difference between dogs losing moderate (1-2 BCS points) or marked (3-4 BCS points) amounts of weight (P=0.627). Mean decrease in body fat per BCS unit change was 5% (3-9%). A model based on a change of 10% of starting weight per unit of BCS above ideal (5/9) most closely estimated actual TW, but marked variability was seen. Therefore, although such calculations may provide a guide to final TW in obese dogs, they can either over- or under-estimate the appropriate end point of weight loss.

Thirty-nine healthy boid snakes representing six different species (Python regius, Boa constrictor, Python reticulatus, Morelia viridis, Epicrates cenchria, and Morelia spilota) were examined using computed tomography (CT) to characterize the normal appearance of the respiratory tissue. Assessment was done subjectively and densitometry was performed using a defined protocol. The length of the right lung was calculated to be 11.1% of the body length, without a significant difference between species. The length of the left lung in proportion to the right was dependent on the species examined. The most developed left lung was in P. regius (81.2%), whereas in B. constrictor, the left lung was vestigial or absent (24.7%). A median attenuation of -814.6 HU and a variability of 45.9 HU were calculated for all species with no significant difference between species. Within the species, a significantly higher attenuation was found for P. regius in the dorsal and cranial aspect of the lung compared with the ventral and caudal part. In B. constrictor, the reduced left lung was significantly hyperattenuating compared with the right lung. Results of this study emphasize the value of CT and provide basic reference data for assessment of the snake lung in these species. Veterinary Radiology &

Obesity is one of the most common medical diseases in cats, but there remains little information on success of weight loss regimes in obese client-owned cats. No information currently exists on body composition changes during weight loss in clinical cases. Twelve obese client-owned cats undertook a weight loss programme incorporating a high-protein low fat diet. Body composition was quantified by dual-energy X-ray absorptiometry, before and after weight loss. Mean (+/-standard deviation) weight loss was 27+/-6.8% of starting weight, and mean rate of weight loss was 0.8+/-0.32% per week. Mean energy allocation during weight loss was 32+/-7.0 kcal/kg target weight. Mean composition of tissue lost was 86:13:1 (fat:lean:bone mineral). The proportion of lean tissue loss was positively associated with overall percentage of weight loss (simple linear regression, r(2)=44.2%, P=0.026). Conventional weight loss programmes produce safe weight loss, but lean tissue loss is an inevitable consequence in cats that lose significant proportions of their starting body weight.

The objective was to determine signalment-related differences in bone mineral content (BMC) and bone mineral density (BMD) in dogs. Unilateral appendicular bones were harvested from 62 canine cadavers. Mid-diaphyseal regions of interest (ROIs) were scanned using a Hologic DXA device Braincon, Vienna, Austria). BMC and BMD were calculated within this region. Middle-aged dogs (3-10 years) revealed the highest BMC and BMD levels. Mean BMC and BMD were higher in males compared to females. Furthermore, body-weight of the male dogs was significantly higher compared to the females (P < 0.0001). Body weight and bone length were significantly associated with BMC and BMD (P < or = 0.023) in all bones but the radius. These data suggest that BMC and BMD appear to be highest in male large-breed dogs with a body weight greater than 30 kg. These results may help determine risk factors in fracture development and healing.

The effects of dirlotapide on body weight (BW) reduction were investigated in overweight Labradors in two parallel-design studies. Study A involved 42 dogs randomized to 0.0, 0.025, 0.05, 0.1, 0.2 or 0.4 mg dirlotapide/kg/day orally for 4 weeks. Study B involved 72 dogs randomized to nine treatments: placebo (24 weeks); dirlotapide (24 weeks) followed by placebo (28 weeks); or dirlotapide (52 weeks); on diets containing 5%, 10% or 15% fat. Dirlotapide dose (initially 0.1 mg/kg) was adjusted monthly during 24-week weight-loss and subsequent 28-week weight-stabilization phases. Food was offered above maintenance energy requirements (MERx 1.1-1.2) based on initial BW. Body composition (body fat, lean tissue and bone mineral content) was monitored using dual-energy X-ray absorptiometry. After treatment, dogs that had received dirlotapide for 52 weeks were fed 90% of quantity consumed at week 52. In study A, BW and food intake decreased asymptotically with dose: mean weekly weight loss exceeded 1% at 0.1-0.4 mg/kg. In study B, dirlotapide resulted in significant mean weekly weight loss (>0.8%) and decreased food intake over 24 weeks compared with placebo (P = 0.0001) for all diets. Food restriction minimized post-treatment weight rebound. Dirlotapide administered daily to dogs for up to 52 weeks was clinically safe and resulted in sustained weight reduction.

Computed tomography and a 3-point bending test were performed on the metacarpal bones of adult production pigs to test the hypothesis that bone strength is strongly correlated with areal bone mineral density (BMD) in this population. The aim of the study was to subject material from adult production pigs grouped by BMD to 3-point bending, to test this hypothesis and determine any correlations. In all, 168 individual computed tomography scans and mechanical tests were performed on the collected material. For evaluation purposes, the material was divided into the categories low, medium, and high BMD (<1, 1 to 1.4, and >1.4 g/cm(2), respectively). The results showed a difference in the maximum load, in the stress at maximum load, and stiffness among each BMD group (P < 0.001) and in elastic modulus between the low BMD group and the 2 other groups (P < 0.001). A correlation between both intrinsic and extrinsic measures of bone strength and BMD was thus demonstrated. The projected change in each of the variables reported, for a 0.1 g/cm(2) alteration in BMD (within the BMD range evaluated in this study), is as follows: maximum load, 708 N; stress at maximum load, 50 N/mm(2); stiffness, 391.6 N/mm; and elastic modulus, 108 N/mm(2) (P < 0.001). The results confirm the relationship between BMD and bone strength and indicate that BMD screening can be used in fracture risk assessments in production pigs.

A high dietary fat intake may be an important environmental factor leading to obesity in some animals. The mechanism could be either an increase in caloric intake and/or a decrease in energy expenditure. To test the hypothesis that high fat diets result in decreased resting energy expenditure (REE), we measured REE using indirect calorimetry in 10-adult intact male Labrador Retrievers, eating weight-maintenance high-fat (HF, 41% energy, average daily intake: 8018 +/- 1247 kJ/day, mean +/- SD) and low-fat (LF, 14% energy, average daily intake: 7331 +/- 771 kJ/day) diets for a 30-day period. At the end of each dietary treatment, body composition measurements were performed using dual-energy X-ray absorptiometry. The mean +/- SD REE was not different between diets (4940 +/- 361 vs. 4861 +/- 413 kJ/day on HF and LF diets respectively). Measurements of fat-free mass (FFM) and fat mass (FM) also did not differ between diets (FFM: 26.8 +/- 2.3 kg vs. 26.3 +/- 2.5 kg; FM: 3.0 +/- 2.3 vs. 3.1 +/- 1.5 kg on HF and LF diets respectively). In summary, using a whole body calorimeter, we found no evidence of a decrease in REE or a change in body composition on a HF diet compared with LF diet.

The aim of the present study was to determine the efficacy of the bone markers osteocalcin (OC) and carboxyterminal cross-linked telopeptide of type-I collagen (ICTP) in evaluating new bone formation in the dog, using commercially available immunoassay kits. Dogs were randomly divided into three groups and a circular external skeletal fixation system (CESF) was mounted on the tibia. In the first group a distraction osteogenesis procedure of the crus was performed. The second group received an osteotomy without crural lengthening, whereas the third group served as a sham-operated control. Bone formation was assessed using densitometric image analysis of crural radiographs. Despite significant differences in the amount of newly formed bone, this finding was not reflected in the plasma levels of OC and ICTP. In conclusion, OC and ICTP were not efficacious as markers of bone formation and resorption during osteogenesis in this canine model.

This study compared the effects of a moderate carbohydrate-high fiber (MC-HF) food and a low carbohydrate-low fiber (LC-LF) food on glycemic control in cats with diabetes mellitus. Sixty-three diabetic cats (48 male castrated, 15 female spayed) were randomly assigned to be fed either a canned MC-HF (n = 32) food or a canned LC-LF (n = 31) food for 16 weeks. Owners were blinded to the type of diet fed. CBC, urinalysis, serum chemistry panel, fructosamine concentration and thyroxine concentration were determined on initial examination, and a complete blood count, serum chemistry panel, urinalysis and serum fructosamine concentration were repeated every 4 weeks for 16 weeks. Insulin doses were adjusted as needed to resolve clinical signs and lower serum fructosamine concentrations. Serum glucose (P = 0.0001) and fructosamine (P = 0.0001) concentrations significantly decreased from week 0 to week 16 in both dietary groups. By week 16, significantly more of the cats fed the LC-LF food (68%, 22/31), compared to the cats fed the MC-HF food (41%, 13/32), had reverted to a non-insulin-dependent state (P = 0.03). Cats in both groups were successfully taken off of insulin regardless of age, sex, type of insulin administered or duration of clinical disease before entering the study. There was no significant difference in the initial or final mean body weights or in the mean change in body weight from week 0 to week 16 between dietary groups. Diabetic cats in this study were significantly more likely to revert to a non-insulin-dependent state when fed the canned LC-LF food versus the MC-HF food.

Following cranial cruciate ligament transection and extracapsular stabilization, dual-energy X-ray absorptiometry was used to analyze bone mineral content and lean tissue mass in the surgical and nonsurgical legs (n=14) at 0, 2, 4, and 8 weeks, and to evaluate bone mineral content and bone mineral density (BMD) of the proximal, mid-, and distal tibia of both the surgical and nonsurgical legs (n=15) at 0, 5, and 10 weeks. There was significant loss of bone mineral content and lean tissue in the surgical leg compared to the nonsurgical leg. Significant loss in bone mineral content and BMD was detected in the tibia of the surgical leg and was most pronounced in the metaphyseal region.
