Cilj ovog istraživanja bio je istražiti terapeutski potencijal resveratrola u kombinaciji s cisplatinom na rast tumora u hipertermalnim uvjetima u miša, odnosno djelovanje ove kombinacije na inhibiciju angiogeneze i rasta tumora te na polarizaciju makrofaga. Uz to, istraženo je može li višemodalni pristup s hipertermijom i resveratrolom dokinuti otpornost tumorskih stanica na cisplatinu kroz modulaciju aktivnosti histonskih deacetilaza (HDAC) i proteina toplinskog stresa te tako doprinijeti izravnoj toksičnosti na tumorske stanice. Tumor je izazvan injiciranjem 1×106 stanica EAT subkutano (sc) u bedro miševima soja Swiss albino. Miševi su obrađeni davanjem resveratrola per os (po) u dozi od 50 mg/kg tijekom pet dana u nizu s početkom 2. dana od injiciranja tumora i/ili cisplatinom koja je injicirana intaperitonealno (ip) u dozama od 2,5 mg/kg 10. i 12. dan te 5 mg/kg 15. dan. Neposredno nakon obrade miševi su izloženi sustavnoj hipertermiji tijekom 15 minuta pri temperaturi od 41 °C. Rezultati pokazuju da primjena resveratrola i cisplatine s hipertermijom jest učinkovit pristup u terapiji tumora. Uspješno inhibira angiogenezu i rast tumora te modulira polarizaciju makrofaga prema fenotipu M1. Također, dokida otpornost tumorskih stanica na cisplatinu kroz modulaciju aktivnosti HDAC-a i razine šaperona HSP-70 i HSP-90 te tako doprinosi povećanju životnog vijeka miševa. The aim of this study was to investigate the therapeutic potential of resveratrol in combination with cisplatin on mouse tumor growth in hyperthermal conditions, i.e. the effect of this combination on inhibition of angiogenesis and tumor development, and on macrophage polarization. In addition, it was investigated whether a multimodal approach with hyperthermia and resveratrol could abolish cisplatin resistance in tumor cells through modulation of histone deacetylases (HDAC) activity and heat shock proteins, contributing to direct toxicity on tumor cells. The tumor was induced by injecting 1×106 EAT cells subcutaneously (sc) into the thighs of Swiss albino mice. Mice were treated with resveratrol per os for five consecutive days beginning on day 2 after tumor injection and/or by injecting cisplatin intraperitoneally (ip) at a dose of 2.5 mg/kg on days 10 and 12 and a dose of 5 mg/kg on day 15. Immediately thereafter, mice were exposed to systematic hyperthermia for 15 min at a temperature of 41 °C. Results show that the administration of resveratrol and cisplatin with hyperthermia is an effective approach in tumor therapy. It successfully inhibits angiogenesis and tumor growth and modulates macrophage polarization to the M1 phenotype. Also, it abolishes tumor cells' cisplatin resistance through modulation of HDAC activity and concentration of HSP-70 and HSP-90 chaperones, contributing to the increased lifespan of mice.

Chemotherapy-induced nausea and vomiting has a major adverse impact in câncer patients, and this has led to considerable research in this field. There are a great number of clinical trials focusing on acute emesis and, significant strides forward have heen made with this problem. However, the pathophysiology of delayed emesis is still poorly understood and the control of it remains a therapeutic challenge. To prevent and treat chemotherapy induced nausea and vomiting, a wide variety of drugs is available. In order to prevent acute emesis induced by highly emetogenic chemotherapy, the use of both 5-HT3 receptor antagonist plus dexamethasone is the choice. For delayed emesis, metoclopramide plus dexamethasone should be considered for those patients with satisfactory control of acute episodes and for those with poor acute emesis control, the combination of 5-HT3 receptor antagonist plus dexamethasone should be used. The prevention of acute emesis induced by moderately emetogenic chemotherapy involves the use of a combination of a 5-HT3 receptor antagonist plus dexainethasone, while more well-conducted studies are necessary to define the best anti-emetic therapy for delayed emesis, since the is no study comparing the different treatments in a proper manner. Náuseas e vômitos decorrentes de quimioterapia têm um impacto negativo importante em pacientes com câncer, mas isso acabou levando a um considerável avanço em pesquisa neste campo. A grande maioria dos estudos se concentra no tratamento da emese aguda, onde se fez grandes avanços; por outro lado, a fisiopatologia da emese tardia não é tão bem entendida, constituindo-se num desafio terapêutico. A literatura mostra que o tratamento para emese aguda induzida por regimes altamente emetogênicos é a associação de antagonistas do receptor 5-HT3 mais dexametasona. Na emese tardia, metoclopramida e dexametasona é o tratamento de escolha para pacientes com bom controle dos episódios de emese prévio. Para aqueles com mau controle, sugere-se antagonistas do receptor 5-HT3 associados a dexametasona. Na prevenção da emese aguda induzida por quimioterápicos moderadamente emetogênicos, o regime mais efetivo é a combinação dos antagonistas dos receptores 5-HT3 e dexametasona. Na emese tardia, há a necessidade de mais estudos, pois até agora, não existem estudos comparando claramente os diversos tratamentos entre si.

Highlights Cisplatin is an antineoplastic agent used malignant diseases. Cisplatin ototoxicity is generally bilateral, irreversible, and progressive. Genistein is a phytoestrogen. Genistein functions as antioxidant and cell cycle inhibitor by inhibiting DNA topoisomerase. Genistein showed positive effects on ototoxicity with its antioxidant. Objective Cisplatin is an antineoplastic agent used in adults and children for the treatment of various malignant diseases. It can cause irreversible ototoxicity. Genistein is a phytoestrogen. Genistein functions as an antioxidant and cell cycle inhibitor by inhibiting the DNA topoisomerase and tyrosine protein kinase enzymes. The protective effect of genistein in preventing cisplatin-induced ototoxicity and levels of the oxidative stress was investigated. Methods 32 Sprague Dawley rats were used in 4 groups (control, cisplatin, cisplatin + genistein, genistein). Otoacoustic emission measurements of the distortion product were performed on the 1st, 2nd and 5th days of the test protocol. Serum malondialdehyde, superoxide dismutase, catalase, glutathione peroxidase, total antioxidant status, total oxidant status and oxidative stress index measurements were made. Results The hearing of the cisplatin + genistein group was found to be better than that of the cisplatin group. While the malondialdehyde, total oxidant status and oxidative stress index parameters decreased significantly in the cisplatin + genistein group compared to the cisplatin group, superoxide dismutase increased significantly (p < 0.05). Conclusion Genistein showed positive effects against ototoxicity with its antioxidant effect. Level of evidence Level 3. Resumo DESTAQUES A cisplatina é um agente antineoplásico usado em lesões malignas. A ototoxicidade da cisplatina é geralmente bilateral, irreversível e progressiva. A genisteína é um fitoestrógeno. A genisteína funciona como antioxidante e inibidor do ciclo celular ao inibir a topoisomerase do DNA. A genisteína apresentou efeitos positivos sobre a ototoxicidade com seu efeito antioxidante. Objetivo A cisplatina é um agente antineoplásico usado em adultos e crianças para o tratamento de diversas lesões malignas. Pode causar ototoxicidade irreversível. A genisteína é um fitoestrógeno que funciona como antioxidante e inibidor do ciclo celular ao inibir as enzimas DNA topoisomerase e tirosina-quinase. O efeito protetor da genisteína na prevenção da ototoxicidade induzida pela cisplatina e os níveis de estresse oxidativo foram investigados. Método Trinta e dois ratos Sprague Dawley foram usados em 4 grupos (controle, cisplatina, cisplatina + genisteína, genisteína). As medidas das emissões otoacústicas por produto de distorção foram tomadas nos dias 1, 2 e 5 do protocolo do teste. Foram medidos os níveis séricos de malondialdeído, superóxido dismutase, catalase, glutationa peroxidase, estado antioxidante total, estado oxidante total e índice de estresse oxidativo. Resultados A audição do grupo cisplatina + genisteína foi melhor do que a do grupo cisplatina. Enquanto os parâmetros malondialdeído, estado oxidante total e índice de estresse oxidativo diminuíram significantemente no grupo cisplatina + genisteína em comparação com o grupo cisplatina, o superóxido dismutase mostrou aumento significantemente (p < 0,05). Conclusão A genisteína apresentou efeitos positivos contra a ototoxicidade com seu efeito antioxidante. Nível de evidência Nível 3.

O estudo das propriedades e interações de drogas quimioterápicas possui um papel vital no desenvolvimento de novos tratamentos oncológicos e no aprimoramento dos que estão atualmente em uso. A cisplatina (Pt(NH3)2Cl2) e outras drogas a base de platina são medicamentos amplamente utilizados para tratar diversos tipos de câncer, incluindo testículo, ovário, pescoço, entre outros. O presente trabalho investigou a interação da cisplatina com células de glioblastoma humano da linhagem U87 a fim de descobrir se o uptake era distinto entre elas. Entender e estudar como se dá o uptake de um droga é de suma importância para a compreensão dos mecanismos de resistências associados a interação dela com as células cancerosas. As células foram tratadas com diferentes concentrações de cisplatina (10, 20 e 100 µM) por 24 horas. A análise foi realizada utilizando a técnica de µ-PIXE, com um feixe de prótons de 2,2 MeV e tamanho de aproximadamente 1,2 x 1,2 µm2 , que forneceu informação elementar da amostra e permitiu a criação de mapas elementares 2D com resolução espacial micrométrica. Medindo os sinais do Cloro e da Platina, foi possível identificar células individuais na amostra e observar quais delas possuíram um maior uptake de cisplatina, respectivamente. Além disso, foi notado que na amostra de 100 µM houve a presença de um alto sinal de Ferro correlacionando com o sinal da Platina, que não estava presente em concentraçõe menores de cisplatina. Tal resultado pode ser evidência de ferroptose, isto é, um tipo de morte de celular dependente de ferro, completamente independente da apoptose convencional. Estudos recentes já mostraram que a cisplatina pode induzir ferroptose caso a dose exceda um determinado limite. Todos os experimentos foram realizados no Laboratório de Implantação Iônica com um acelerador Tandem de 3MV. Esse trabalho também demonstrou a versatilidade e aplicabilidade da técnica de µ-PIXE e como ela pode ser utilizada para conduzir análises e experimentos multidisciplinares. The study of chemotherapy drug’s properties plays a vital role in the development of new cancerrelated treatments and the improvement of those currently in use. Cisplatin (Pt(NH3)2Cl2) and other platinum-based drugs are widely used medications that treat several different cancers, including, but not limited to, ovarian, testicular, and neck. The present work investigated the interaction of cisplatin with human glioblastoma U87 cells in order to find out if the uptake was distinct between them. Cells were treated with cisplatin at a concentration of 10, 20, and 100 µM for 24 hours. The analysis was done using the µ-PIXE technique, with a 2.2 MeV proton beam of 1.2 x 1.2 µm2 spot size, which gave us elemental information about the sample and allowed us to create 2D maps with micrometric spacial resolution. By measuring the chlorine and platinum signals, we were able to identify individual cells in the sample and detect which ones had a greater cisplatin uptake, respectively. Understanding the drug’s uptake is crucial for unraveling the mechanisms of resistance that are involved in its interaction with cancer cells. Moreover, we noticed that the 100 µM sample had a high iron signal correlating with the platinum signal, which was not present at lower treatment concentrations. This result could be a piece of evidence for ferroptosis, i.e. a type of iron-dependant cell death completely independent from regular apoptosis. Recent studies have already shown that cisplatin can induce ferroptosis in high treatment concentrations. All experiments were conducted at the Ion Implantation Laboratory with a 3MV Tandem accelerator. This work also highlighted the versatility and applicability of the µ-PIXE technique and how it can be used to perform interdisciplinary analysis.

Background: Radiotherapy of locally advanced head and neck cancer represents a major clinical challenge. Any treatment intensification aiming at improved treatment outcomes poten­tially results in a higher toxicity. The search for optimal treatment schedule involving conventional or altered fractionation of radiotherapy and the frequency and dose of concomitant cisplatin or other systemic agents has been spanning over several decades. Purpose: To evaluate long-term outcomes and toxicity of accelerated chemoradiotherapy of locally advanced squamous cell carcinoma of the head and neck (LA SCCHN). Patients and methods: Forty patients with stage III and IVA (TNM, 7th Ed.) LA SCCHN were treated with accelerated radiotherapy with a total dose of 67.5 Gy in 6 weeks delivered with simultaneous integrated boost intensity-modulated radiotherapy (SIB IMRT) and concomitant weekly cisplatin 40 mg/m2. Five-year outcomes and early and late toxicity were evaluated. Results: With the median follow-up of 47.8 months, a 5-year locoregional control rate (LCR) was 56.5%, distant control rate (DCR) was 87% and 5-year progression-free survival (PFS) and overall survival (OS) were 37 and 45%, respectively. Cisplatin cumulative dose of ≥ 200 mg/m2 was administered in 83% of patients. Grade ≥ 2 late toxicity with dietary change was observed in 21 (53%) patients. Human papillomavirus (HPV) status determined by p16 immunohistochemistry was the only significant factor in 5-year treatment outcomes analysis with LCR 100 vs. 41% (P < 0.01), DCR 100 vs. 78% (P = 0.154), PFS 80 vs. 23% (P = 0.01) and OS 80 vs. 34% (P = 0.03) for HPV positive oropharyngeal cancer (OPC) and other HPV negative LA SCCHN. Conclusion: High proportion of patients with LA SCCHN received an adequate cumulative dose of concurrent cisplatin with accelerated radiotherapy with SIB IMRT. This study demonstrated that chemoradiotherapy with weekly cisplatin resulted in favorable local control rate and survival in patients with HPV+ OPC.

Cisplatin (CP) is a commonly used, powerful antineoplastic drug, having numerous side effects. Casticin (CAS) is considered as a free radical scavenger and a potent antioxidant. The present research was planned to assess the curative potential of CAS on CP persuaded renal injury in male albino rats. Twenty four male albino rats were distributed into four equal groups. Group-1 was considered as a control group. Animals of Group-2 were injected with 5mg/kg of CP intraperitoneally. Group-3 was co-treated with CAS (50mg/kg) orally and injection of CP (5mg/kg). Group-4 was treated with CAS (50mg/kg) orally throughout the experiment. CP administration substantially reduced the activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), glutathione S-transferase (GST), glutathione reductase (GSR), glutathione (GSH) content while increased thiobarbituric acid reactive substances (TBARS), and hydrogen peroxide (H2O2) levels. Urea, urinary creatinine, urobilinogen, urinary proteins, kidney injury molecule-1 (KIM-1), and neutrophil gelatinase-associated lipocalin (NGAL) levels were substantially increased. In contrast, albumin and creatinine clearance was significantly reduced in CP treated group. The results demonstrated that CP significantly increased the inflammation indicators including nuclear factor kappa-B (NF-κB), tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6) levels and cyclooxygenase-2 (COX-2) activity and histopathological damages. However, the administration of CAS displayed a palliative effect against CP-generated renal toxicity and recovered all parameters by bringing them to a normal level. These results revealed that the CAS is an effective compound having the curative potential to counter the CP-induced renal damage. Resumo A cisplatina (CP) é uma droga antineoplásica poderosa, comumente usada, com vários efeitos colaterais. Casticin (CAS) é considerado um eliminador de radicais livres e um potente antioxidante. A presente pesquisa foi planejada para avaliar o potencial curativo da CAS em lesão renal induzida por PC em ratos albinos machos. Vinte e quatro ratos albinos machos foram distribuídos em quatro grupos iguais. O Grupo 1 foi considerado grupo controle. Os animais do Grupo 2 foram injetados com 5 mg / kg de PB por via intraperitoneal. O Grupo 3 foi cotratado com CAS (50 mg / kg) por via oral e injeção de CP (5 mg / kg). O Grupo 4 foi tratado com CAS (50 mg / kg) por via oral durante todo o experimento. A administração de CP reduziu substancialmente as atividades de catalase (CAT), superóxido dismutase (SOD), peroxidase (POD), glutationa S-transferase (GST), glutationa redutase (GSR), glutationa (GSH), enquanto aumentou as substâncias reativas ao ácido tiobarbitúrico (TBARS) e níveis de peróxido de hidrogênio (H2O2). Os níveis de ureia, creatinina urinária, urobilinogênio, proteínas urinárias, molécula 1 de lesão renal (KIM-1) e lipocalina associada à gelatinase de neutrófilos (NGAL) aumentaram substancialmente. Em contraste, a albumina e a depuração da creatinina foram significativamente reduzidas no grupo tratado com PC. Os resultados demonstraram que a CP aumentou significativamente os indicadores de inflamação, incluindo fator nuclear kappa-B (NF-κB), fator de necrose tumoral-α (TNF-α), interleucina-1β (IL-1β), interleucina-6 (IL-6) níveis e atividade da ciclooxigenase-2 (COX-2) e danos histopatológicos. No entanto, a administração de CAS apresentou um efeito paliativo contra a toxicidade renal gerada por CP e recuperou todos os parâmetros, trazendo-os a um nível normal. Estes resultados revelaram que o CAS é um composto eficaz com potencial curativo para combater o dano renal induzido por CP.

At present, nuclear condensation and fragmentation have been estimated also using Hoechst probes in fluorescence microscopy and flow cytometry. However, none of the methods used the Hoechst probes for quantitative spectrofluorometric assessment. Therefore, the aim of the present study was to develop a spectrofluorometric assay for detection of nuclear condensation and fragmentation in the intact cells. We used human hepatoma HepG2 and renal HK-2 cells cultured in 96-well plates treated with potent apoptotic inducers (i.e. cisplatin, staurosporine, camptothecin) for 6-48 h. Afterwards, the cells were incubated with Hoechst 33258 (2 mu g/mL) and the increase of fluorescence after binding of the dye to DNA was measured. The developed spectrofluorometric assay was capable to detect nuclear changes caused by all tested apoptotic inducers. Then, we compared the outcomes of the spectrofluorometric assay with other methods detecting cell impairment and apoptosis (i.e. WST-1 and glutathione tests, TUNEL, DNA ladder, caspase activity, PARP-1 and JNKs expressions). We found that our developed spectrofluorometric assay provided results of the same sensitivity as the TUNEL assay but with the advantages of being fast processing, low-cost and a high throughput. Because nuclear condensation and fragmentation can be typical markers of cell death, especially in apoptosis, we suppose that the spectrofluorometric assay could become a routinely used method for characterizing cell death processes. V současné době se k detekci jaderné kondenzace a fragmentace využívají fluorescenční sondy Hoechst. Žádná z metod však nepoužívá sondy Hoechst pro kvantitativní spektrofluorometrické hodnocení. Cílem této studie bylo vyvinout spektrofluorometrický test pro detekci jaderné kondenzace a fragmentace v intaktních buňkách. Použili jsme buňky lidského hepatomu HepG2 a renální HK-2 buňky kultivované v 96-jamkových inkubovanými s apoptotickými induktory (tj. cisplatinou, staurosporinem, kamptotecinem) po dobu 6-48 hodin. Poté byly buňky inkubovány s Hoechst 33258 (2 ug/ml) a bylo detekováno zvýšení intenzity fluorescence po navázání barviva na DNA. Vyvinutý spektrofluorometrický test byl schopen detekovat jaderné změny způsobené všemi testovanými apoptotickými induktory. Poté jsme výsledky spektrofluorometrického testu porovnali s dalšími metodami detekujícími poškození buněk a apoptózu (tj. testy WST-1 a glutathionu, TUNEL, DNA ladder, aktivita kaspázy, exprese PARP-1 a JNKs). Zjistili jsme, že náš vyvinutý spektrofluorometrický test poskytuje výsledky se stejnou citlivostí jako test TUNEL, ale s výhodami rychlého zpracování, nízkých nákladů a vysokého výkonu. Protože jaderná kondenzace a fragmentace mohou být typickými markery buněčné smrti, zejména při apoptóze, předpokládáme, že spektrofluorometrický test by se mohl stát rutinně používanou metodou pro charakterizaci procesů buněčné smrti.

Introduction: Cisplatin is one of the main chemotherapeutic agents used for the treatment of many types of cancer. However, ototoxicity, one of the most serious side effects of cisplatin, restricts its usage. Objective: We aimed to investigate the protective effects of whortleberry extract against cisplatin-induced ototoxicity by evaluating hearing and histopathological cochlear damage and by measuring the biochemical parameters affected byoxidative stress. Methods: Forty-eight male rats were included in the study after performing Distortion Product Otoacoustic Emission test to confirm that their hearing levels were normal. The rats were randomly divided into six groups: the control group, the sham group, and, which received only whortleberry extract, only cisplatin, cisplatin + 100 mg whortleberry extract, cisplatin + 200 mg whortleberry extract, respectively. Audiologic investigation was performed by performing the Distortion Product Otoacoustic Emission test at the beginning and at the eighth day of the study. Cardiac blood samples were collected for biochemical analysis, and the rats were sacrificed to obtain cochlear histopathological specimens on the eighth day. Results: The results revealed that whortleberry protects hearing against cisplatin-induced ototoxicity independent of the dose. However, high doses of whortleberry extract are needed to prevent histopathological degeneration and oxidative stress. Conclusion: The results obtained in this study show that whortleberry extract has a protective effect against cisplatin-induced ototoxicity. Resumo: Introdução: A cisplatina é um dos principais agentes quimioterápicos utilizados para o tratamento de muitos tipos de câncer. No entanto, a ototoxicidade, um dos efeitos colaterais mais graves da cisplatina, restringe seu uso. Objetivo: Nosso objetivo foi investigar os efeitos protetores do extrato de uva-do-monte contra a ototoxicidade induzida por cisplatina, avaliar o dano auditivo e histopatológico coclear e medir os parâmetros bioquímicos afetados pelo estresse oxidativo. Método: Foram incluídos no estudo 48 ratos machos após teste de emissão otoacústica evocada por produto de distorção para confirmar que seus níveis de audição eram normais. Os ratos foram divididos aleatoriamente em seis grupos: o grupo controle, o grupo simulado, o que recebeu apenas extrato de uva-do-monte, o que recebeu apenas cisplatina, o que recebeu cisplatina + 100 mg de extrato de uva-do-monte e o que recebeu cisplatina + 200 mg de extrato de uva-do-monte, respectivamente. A investigação audiológica foi feita através do teste de emissão otoacústica de produto de distorção no início e no oitavo dia do estudo. As amostras de sangue cardíaco foram coletadas para análise bioquímica e os ratos foram sacrificados para obtenção de espécimes histopatológicos cocleares no oitavo dia. Resultados: Os resultados revelaram que o extrato de uva-do-monte protege a audição contra a ototoxicidade induzida por cisplatina, independentemente da dose. No entanto, são necessárias doses elevadas do extrato para evitar a degeneração histopatológica e o estresse oxidativo. Conclusão: Os resultados obtidos neste estudo mostram que o extrato de uva-do-monte tem um efeito protetor contra a ototoxicidade induzida por cisplatina. Cilj ovog istraživanja bio je istražiti sinergistički učinak sustavne hipertermije uz nanokristale resveratrola i cisplatinu na inhibiciju brzorastućeg EAT, preživljavanje miševa, polarizaciju makrofaga te sposobnost nanokristala resveratrola u smanjenju toksičnih učinaka cisplatine i hipertermije u jetri i bubregu miševa. Miševima soja Swiss albino izazvan je tumor injiciranjem 1x106 stanica EAT subkutano u bedro, a zatim su obrađeni injiciranjem nanokristala resveratrola u dozi 50 mg/kg per os tijekom 5 dana u nizu te cisplatine u dozi 2,5 mg/kg 10. i 12. dan te u dozi 5 mg/kg 14. dan uz izlaganje sustavnoj hipertermiji pri 41°C. Dobiveni rezultati pokazuju da samostalna primjena nanokristala resveratrola i izlaganje hipertermiji vodi rastu tumora dok primjena nanokristala resveratrola i cisplatine uz izlaganje sustavnoj hipertermiji ostvaruje izraziti antitumorski učinak inhibicijom rasta EAT u solidnom obliku, ali ne pridonosi znatnijem povećanju preživljavanja životinja. Injiciranje cisplatine pri hipertermalnim uvjetima povećava životni vijek miševa za 47% u odnosu na kontrolu. Temeljem dobivenih rezultata možemo zaključiti da nanokristali resveratrola mogu polučiti antioksidacije i/ili prooksidacijske učinke u različitim mikrookolišnim uvjetima, ali i sudjelovati u povećanju makrofagne plastičnosti i polarizacije te zaštititi bubreg i jetru od toksičnosti izazvane cisplatinom. The aim of this study was to investigate the synergistic effect of systemic hyperthermia with resveratrol nanocrystals and cisplatin on the inhibition of fast-growing EAT, mouse survival, macrophage polarization, and the ability of resveratrol nanocrystals to reduce the toxic effects of cisplatin and hyperthermia. In Swiss albino mice tumor were induced by injecting 1x106 EAT cells subcutaneously into the thigh and then treated by injecting resveratrol nanocrystals at a dose of 50 mg/kg per os for 5 consecutive days and cisplatin at a dose of 2,5 mg/kg day 10 and 12 and at a dose of 5 mg/kg on day 14 with exposure to systemic hyperthermia at 41°C. The results show that the use of resveratrol nanocrystals alone and exposure to hyperthermia leads to tumor growth, while the use of resveratrol and cisplatin nanocrystals with exposure to systemic hyperthermia has a pronounced antitumor effect by inhibiting EAT growth in solid form, but does not significantly increase animal survival. Injection of cisplatin under hyperthermal conditions increased the lifespan of mice by 47% compared to control. Based on the obtained results, we can conclude that resveratrol nanocrystals have antioxidant and/or prooxidative effects in various microenvironmental conditions, but also participate in increasing macrophage plasticity and polarization and protect the kidney and liver from cisplatin-induced toxicity.

Cilj ovog istraživanja bio je istražiti združeno djelovanje citostatika cisplatine i resveratrola na inhibiciju angiogeneze i razvoja tumora, povećanje životnog vijeka domaćina, te istražiti aktivnost makrofaga u tkivu tumora kroz analizu inducibilne dušik oksid sintaze. Uz to cilj je bio utvrditi kako višekratno djelovanje citostatika djeluje na razinu proteina toplinskog stresa i da li obrada resveratrolom modulira njihovu aktivnost vodeći dokidanju rezistencije tumorskih stanica na citostatik. Tumor je izazvan injiciranjem 1x106 stanica EAT subkutano (sc) u bedro miševa soja Swiss albino. Obrada životinja s EAT započeta je unosom resveratrola per os u dozi od 50 mg/kg tijekom 5 dana u nizu s početkom 2. dana od injiciranja tumora dok je cisplatina injicirana u dozi od 2,5 mg/kg 10. i 12. dana te dozom od 5 mg/kg 15. dana. Dobiveni rezultati pokazuju da resveratrol združen s cisplatinom može inhibirati proces angiogeneze, usporiti rast tumora te povećati životni vijek nositelja tumora jačanjem imunomodulatorne aktivnosti domaćina kroz modulaciju proteina toplinskog stresa HSP-90 i HSP-70 i smanjuje štetne učinke cisplatine na normalne stanice i tkiva, što ukazuje na mogućnost kliničke uporabe ove kombinacije The aim of this study was to investigate the combined effect of cytostatic cisplatin and resveratrol on inhibition of angiogenesis and tumor development, increase of host lifespan, and to investigate macrophage activity in tumor tissue through analysis of inducible nitric oxide synthase. In addition, the aim was to determine how the repeated action of cytostatics affects the level of heat shock proteins and whether resveratrol treatment modulates their activity leading to the abolition of tumor cell resistance to cytostatics. The tumor was induced by injecting 1x106 EAT cells subcutaneously (sc) into the thighs of Swiss albino mice. Treatment of animals with EAT was initiated by administration of resveratrol per os at a dose of 50 mg/kg for 5 consecutive days beginning on day 2 after tumor injection while cisplatin was injected at a dose of 2.5 mg/kg on days 10 and 12 and at a dose of 5 mg/kg on day 15. The results show that cisplatin combined with resveratrol can inhibit angiogenesis, slow tumor growth, and increase host lifespan by enhancing host immunomodulatory activity by modulating heat shock proteins HSP-90 and HSP70 and reducing the adverse effects of cisplatin on normal cells and tissues, which suggest the possibility of clinical use of this combination.

Introduction Platinum-based chemotherapeutics play an important role in the treatment of cancer at different levels and are the most cited ototoxic agents when scientific evidence is analyzed. Objective To present scientific evidence based on a systematic literature review, PRISMA, in order to systematize information on the ototoxic effects of using antineoplastic drugs. Methods For the selection of studies, the combination based on the Medical Subject Heading Terms (MeSH) was used. The Medline (Pubmed), LILACS, SciELO, SCOPUS, WEB OF SCIENCE and BIREME databases were used, without restriction of language, period, and location. Evaluation of the quality of the articles was carried out, which included articles with a minimum score of 6 in the modified scale of the literature. The designs of the selected studies were descriptive, cohort, and cross-sectional, which were related to the research objective. Results Three articles were included in this systematic review. The ototoxicity caused by cisplatin alone varied from 45% to 83.3%, while that caused by the use associated with carboplatin varied from 16.6% to 75%. There was a significant variation in the cumulative doses of these antineoplastic agents, both in isolated and in combination. Auditory changes, especially at high frequencies, were evident after completion of treatment. Conclusion Auditory changes after the use of platinum-based antineoplastic drugs were found, however, there was an important heterogeneity regarding the frequency of ototoxicity and the cumulative dose of the drugs used. Resumo Introdução Os quimioterápicos à base de cisplatina desempenham papel importante no tratamento do câncer em diversos níveis e são os agentes ototóxicos mais citados quando analisadas evidências científicas. Objetivo Apresentar evidências científicas com base em revisão sistemática da literatura (PRISMA), com o intuito de sistematizar informações sobre os efeitos ototóxicos do uso de medicamentos antineoplásicos. Método Para a seleção dos estudos foi usada a combinação baseada no Medical Subject Heading Terms (MeSH). Foram usadas as bases de dados Medline (Pubmed), Lilacs, SciELO, Scopus, Web of Science e Bireme, sem restrição de idioma, período e localização. Foi feita avaliação da qualidade dos artigos, na qual se incluíam artigos com nota mínima 6 na escala modificada da literatura. Os desenhos dos estudos selecionados foram do tipo descritivo, coorte e transversal, os quais estavam relacionados com o objetivo da pesquisa. Resultados Três artigos foram admitidos para esta revisão sistemática. A ototoxicidade causada pela cisplatina isoladamente variou de 45% a 83,3%, enquanto a causada pelo uso associado com a carboplatina variou de 16,6% a 75%. Verificou-se uma variação significativa nas doses cumulativas desses antineoplásicos, tanto no uso isolado quanto em combinação. Alterações auditivas, principalmente em altas frequências, foram evidenciadas após finalização do tratamento. Conclusão Alterações auditivas após o uso de medicamentos antineoplásicos à base de platina foram constatadas, porém verificou-se uma importante heterogeneidade quanto à frequência da ototoxicidade e a dose cumulativa das drogas usadas.

Uvod: U ovoj studiji su ispitivani efekti ekstrakta biljke Satureja hortensis L. (SHL) na biheviorlane manifestacije neurotoksičnosti izazvane cisplatinom kod pacova. Materijal i metode: Studija je sprovedena na 35 pacova Wistar albino soja, starosti 2 meseca, telesne mase 200-250 g, podeljenih u 7 grupa: kontrolna, cisplatina, cisplatina+ SHL 50 mg/kg, cisplatina+SHL 100 mg/kg, cisplatina+SHL 200 mg/kg, cisplatina+silimarin 100mg/kg i SHL 200 mg/kg. Cisplatina je primenjivana u pojedinačnoj dozi (7.5 mg/kg), dok je antioksidantna suplementacija sprovođena oralno tokom 10 dana. Bihevioralni testovi su izvođeni desetog dana protokola, nakon čega su životinje žrtvovane u cilju dobijanja uzoraka hipokampusa za određivanje markera oksidativnog stresa i apoptoze. Rezultati: Rezultati bihevioralnog ispitivanja su pokazali anksiogeni efekat cisplatine koji je bio ublažen primenom SHL ekstrakta (100 mg/kg) pri čemu je slabiji odgovor bio u grupama tretiranim ekstraktom SHL u dozama od 50 i 200 mg/kg. Takođe, smanjenje neurotoksičnosti izazvano cisplatinom, u smislu povećanja markera oksidativnog stresa i apoptoze, je ostvareno primenom ekstrakta u dozi od 100 mg/kg. Neuroprotektivni efekat SHL je bio sličan dejstvu silimarina u istoj dozi. Ekstrakt SHL nije uzrokovao značajne promene ispitivanih parametara kada je primenjivan samostalno. Zaključak: Anksiogeni efekat, kao manifestacija neurotoksičnosti izazvane cisplatinom, je u bihevioralnim testovima bio uspešno umanjen oralnom primenom ekstrakta SHL u optimalnoj dozi (100 mg/kg). Korisni efekti ove vrste antioksidantne suplementacije su takođe potvrđeni smanjenjem oksidativnog oštećenja i apoptotskih mehanizama. Introduction: In this study we evaluated effects of Saturejа hortensis L. (SHL) extract on behavioral manifestations of cisplatin-induced neurotoxicity in rats. Material and methods: The study was performed on 35 Wistar albino rats, 2 months old, weighting 200-250 g, divided into 7 groups: control, cisplatin, cisplatin+SHL 50 mg/kg, cisplatin+SHL 100 mg/kg, cisplatin+SHL 200 mg/kg, cisplatin+silymarin 100 mg/kg, and SHL 200 mg/kg. Cisplatin was administered in a single dose of 7.5 mg/kg and antioxidant supplementation was performed orally for 10 days. Behavioral testing was performed on 10th day and then the animals were sacrificed in order to obtain the samples for oxidative stress and apoptotic markers in hippocampus. Results: The results of behavioral testing showed cisplatin-induced anxiogenic effect, that was diminished with SHL extract in a dose of 100mg/kg, with much weaker responses to SHL extracts in doses of 50 and 200 mg/kg. Also, the attenuation of cisplatin-induced neurotoxicity, by means of increased oxidative stress and apoptotic markers, was successfully achieved with SHL extract in a dose of 100 mg/kg. The neuroprotective effect of SHL was similar to silymarin action in the same dose. When applied alone, SHL extract induced no significant effect in any of parameters followed. Conclusion: Cisplatin-induced neurotoxicity manifested by anxiogenic response in behavioral testing can be successfully attenuated by oral administration of SHL extract in an optimal dose (100 mg/kg). The beneficial effects of this kind of antioxidant supplementation were also confirmed by decreased oxidative damage and apoptotic mechanisms.

Uvod: Rutinska primena do sada otkrivenih preparata platine u onkologiji ima svoje nedostatke i ograničenja, a to je njihovo toksično dejstvo, ograničena rastvorljivost i pojava rezistencije nakon duže primene. Da bi se prevazišli nedostaci derivata platine, a posebno toksičnost, sintetisan je veliki broj kompleksa platine(II) kao što su karboplatina, oksaliplatina, nedaplatina, lobaplatina i heptaplatina (4). Još veći pomak je napravljen sintezom kompleksa koji sadrže dva ili više jona platine povezanih mostnim ligandima, tzv. polinuklearni kompleksi platine ali i oni su imali izvesna ograničenja u primeni. Materijal i metode: Ova studija je sprovedena po tipu eksperimentalne studije na materijalu humanog i animalnog porekla in vitro. Sintetisali smo odgovarajuće platina(II) komplekse, [{Pt(en)Cl}2(µ-1,7-phen)}(ClO4)2 i [{Pt(en)Cl}2(µ-4,7-phen)}(ClO4)2, gde je 1,7-phen mostni ligand 1,7-fenantrolin, odnosno 4,7-fenantrolin. Iste komplekse smo metadama elementarne mikroanalize i spektroskopije okarakterisali. Antitumorski efekat sintetisanih kompleksa in vitro smo dokazali MTT testom, analizom ćelija obojenih Annexin-om V i propidijum jodidom, analizom molekula uključenih u proces apoptoze (Bax, Bcl-2, kaspaza-3) i analizom ćelijskog ciklusa. Rezultati: U našem istraživanju nakon šo smo okarakterisali dva kompleksa K1 i K2 dokazali smo da oba kompleksa, naročito K1 imaju izraženiji citotoksični efekat na ćelijske linije karcinoma dojke in vitro kako humanog tako i animalnog porekla. Sa druge strane naši rezultati pokazuju da efekat ovih komplesa na zdrave fibroblaste iz linije MRC-5 je manje cititoksičan u odnosu na konvencionalne hemioterapeutike iz grupe derivata platine. Ova činjenica bi mogla biti pretpostavka da bi novosintetisani lekovi na bazi dinuklearnih kompleksa platine bili manje toksični a samim tim i bezbedniji za rutinsku primenu. Zaključak: Dinuklearni kompleksi platine(II) koji su upotrebljeni u ovoj tezi su rastvorni u vodi i po strukturi slični oksaliplatini koja se koristi za lečenje obolelih od karcinoma dojke, a naročito agresivnih oblika ove bolesti. Pokazali smo da novosintetisani dinuklearni kompleksi platine(II) pokazuju izraženiji citotoksični efekat na ćelije karcinoma dojke in vitro u poređenju sa cisplatinom i oksaliplatinom. Ovim eksperimentalnim radom ukazali smo na nove dinuklearne komplekse platine(II)124 koji bi se mogli koristiti u narednim istraživanjima u oblasti koordinovanja platine sa ligandima od farmakološkog značaja. Introduction: The routine use of platinum preparations discovered so far in oncology has its drawbacks and limitations, which is their toxic effect, limited solubility and the appearance of resistance after prolonged administration. To overcome the disadvantages of platinum derivatives, and in particular toxicity, a number of platinum (II) complexes such as carboplatin, oxaliplatin, nedaplatin, lobaplatin, and heptaplatin have been synthesized. A further shift was126 made by the synthesis of complexes containing two or more platinum ions linked by bridge ligands, the so-called polynuclear platinum complexes but they also had some limitations in their application. Material and Methods: This study was conducted by type of experimental study on material of human and animal origin in vitro. We synthesized the corresponding platinum (II) complexes, [{Pt (en) Cl} 2 (µ-1,7-phen)} (ClO4) 2 and [{Pt (en) Cl} 2 (µ-4,7-phen )} (ClO4) 2, where the 1,7-phen bridge ligand is 1,7-phenanthroline and 4,7-phenanthroline, respectively. We characterized the same complexes by the methods of elemental microanalysis and spectroscopy. The in vitro antitumor effect of the synthesized complexes was demonstrated by MTT assay, analysis of Annexin V and propidium iodide stained cells, analysis of molecules involved in apoptosis (Bax, Bcl-2, caspase-3) and cell cycle analysis. Results: In our study, after characterizing the two K1 and K2 complexes, we demonstrated that both complexes, especially K1, have a more pronounced cytotoxic effect on breast cancer cell lines in vitro of both human and animal origin. On the other hand, our results show that the effect of these complexes on healthy MRC-5 fibroblasts is less cytotoxic than conventional platinumderived chemotherapeutics. This fact might suggest that newly synthesized drugs based on dinuclear platinum complexes would be less toxic and therefore safer for routine use. Conclusion: The dinuclear platinum (II) complexes used in this thesis are water-soluble and oxaliplatin-like in structure for the treatment of breast cancer patients, and in particular for aggressive forms of this disease. We have shown that the newly synthesized dinuclear platinum (II) complexes exhibit a more pronounced cytotoxic effect on breast cancer cells in vitro compared with cisplatin and oxaliplatin. Through this experimental work, we have identified novel dinuclear platinum (II) complexes that could be used in future studies in the field of platinum coordination with ligands of pharmacological importance.

Introduction: The use of electron microscopy in the study of the inner ear has allowed us to observe minute details of the hair cells, especially in ototoxicity studies; however, the preparation of this material is a difficult and delicate task. In an attempt to simplify the handling of these materials, two agents, toluidine blue and ethylenediamine tetra-acetic acid were tested, in addition to the elimination of osmium tetroxide during the preparation of albino guinea pig cochleae. We also tested the applicability of these methodologies in an ototoxicity protocol. Objective: To verify the quality of the images obtained with and without the use of ethylenediamine tetra-acetic acid, toluidine blue and osmium tetroxide in the preparation of cochleae of albino guinea pigs for the scanning electron microscopy. Methods: Three groups of cochleae were used. In Group 1, 10 cochleae were prepared with the usual methodology, dissecting the optical capsule without decalcification and using osmium tetroxide as a post-fixative agent. In Group 2, we prepared 10 cochleae decalcified with ethylenediamine tetra-acetic acid, injecting toluidine blue in the endolymphatic space to facilitate the identification of the organ of Corti. In Group 3, we used 4 cochleae of guinea pigs that received 3 doses of cisplatin (7.5 mg/kg, D1-D5-D6), two prepared according to the methodology used in Group 1 and two with that used in Group 2. Scanning electron microscopy images were obtained from the organ of Corti region of the basal turn of each cochlea. Results: The organ of Corti was more easily identified with the use of toluidine blue. The dissection of the cochlea was more accurate in the decalcified cochleae. The quality of the images and the preservation of the organ of Corti obtained with the two methodologies were similar. Conclusion: The proposed modifications resulted in images of similar quality as those observed using the traditional methodology. Resumo Introdução: O emprego da microscopia eletrônica no estudo da orelha interna permitiu observar detalhes minuciosos das células ciliadas especialmente em estudos de ototoxicidade. Entretanto, o preparo desse material é trabalhoso e delicado. Para simplificar a manipulação desses materiais, testou-se o uso de dois agentes, azul de toluidina e ácido etilenodiamino tetra-acético, além da retirada do tetróxido de ósmio na preparação de cócleas de cobaias albinas. Testamos também a aplicabilidade dessas metodologias em um protocolo de ototoxicidade. Objetivo: Verificar a qualidade das imagens obtidas com e sem o uso de ácido etilenodiamino tetra-acético, azul de toluidina e tetróxido de ósmio na preparação de cócleas de cobaias albinas para a microscopia eletrônica de varredura. Método: Foram utilizados três grupos de cócleas. No Grupo 1 preparou-se 10 cócleas com a metodologia usual, dissecando a cápsula ótica sem descalcificac¸ão e utilizando tetróxido de ósmio como pós-fixador. No Grupo 2 preparamos 10 cócleas descalcificadas com ácido etilenodiamino tetra-acético, injetando azul de toluidina no espac¸o endolinfático para facilitar a identificação do órgão de Corti. No Grupo 3 utilizamos 4 cócleas de cobaias que receberam 3 doses de cisplatina (7,5 mg/kg, D1-D5-D6), duas preparadas com a metodologia do Grupo 1 e duas com a do Grupo 2. Foram obtidas imagens da microscopia eletrônica de varredura da região do órgão de Corti do giro basal de cada cóclea. Resultados: O órgão de Corti foi mais facilmente identificado com o azul de touidina. A dissecção da cóclea foi mais precisa nas cócleas descalcificadas A qualidade das imagens e a preservac¸ão do órgão de Corti obtidas com as duas metodologias foi similar. Conclusão: As modificações propostas resultaram em imagens de qualidade similar as observadas com o uso da metodologia tradicional.

Resumo: Devido a crise de legitimidade desencadeada pela invasão napoleônica da Espanha em 1808, diferentes projetos políticos surgiram nas colônias hispano-americanas. No Rio da Plata, enquanto Buenos Aires emergiu como um pólo revolucionário, a cidade portuária de Montevidéu optou por projetos políticos monárquicos e monarquistas. Este artigo examina a relação entre o comércio trans-imperial e monarquismo no Rio da Prata, e especialmente em Montevidéu. Os projetos monárquicos no Rio da Prata estavam intimamente associados à manutenção de redes de comércio trans-imperiais e à manutenção da ordem política e econômica do período colonial. O monarquismo representava a continuidade dos princípios legais do antigo regime, proporcionava segurança e estabilidade ao comércio trans-imperial e, acima de tudo, impedia as mudanças econômicas, políticas e sociais propostas pelos projetos revolucionários. Além disso, este trabalho contribui para os debates sobre os múltiplos significados de soberania e autonomia política durante a era das revoluções na América ibérica. Abstract: With the crisis of legitimacy triggered by Napoleon’s invasion of the Iberian Peninsula in 1808, different reactions and political projects emerged in the Spanish American colonies. In Rio de la Plata, while Buenos Aires broke away from Spanish rule, the port city of Montevideo opted for royalist and monarchical political projects in opposition to Buenos Aires. This article examines the relationship between trans-imperial trade and monarchism in Rio de la Plata. Moreover, this piece contributes to debates on the multiple and contested meanings of sovereignty during the age of revolution in Iberian America. Monarchism in Rio de la Plata was intimately associated with the maintenance of trans-imperial networks of trade and the maintenance of the political and economic order of the colonial period. Monarchism represented the continuity of the old regime’s legal principles, provided safety and stability for trade trans-imperial trade, and above all, prevented the economic, political, and social changes proposed by revolutionary projects.

Receptor Toll-like 3 (TLR3) je transmembranski protein čija je ekspresija prvotno otkrivena na stanicama imunološkog sustava. TLR3 je eksprimiran i u stanicama brojnih zdravih i tumorskih tkiva, između ostalog tumora glave i vrata gdje ima dvojaku ulogu. Aktivacija TLR3 može pokrenuti preživljenje i progresiju tumora, ali i apoptozu stanica tumora. U ovom smo radu željeli istražiti je li, i na koji način, moguće potaknuti smrt stanica tumora kombinirajući agonist TLR3, poly(I:C), sa standardnim liječenjem cisplatinom uz zračenje. Naši rezultati pokazali su da poly(I:C) u kombinaciji s cisplatinom inhibira rast stanica tumora, potiče apoptozu aktivacijom kaspaza -3 i -7 i smanjivanjem ekspresije gena inhibitora apoptoze, potiče stvaranje reaktivnih oblika kisika i inducira zaustavljanje staničnog ciklusa u fazi G2/M. Navedeni rezultati ukazuju na to da poly(I:C) sinergistički s cisplatinom smanjuje preživljenje stanica tumora te povećava osjetljivost stanica tumora na zračenje, što ga čini vrijednom terapijskom opcijom u liječenju tumora glave i vrata. Toll-like receptor 3 (TLR3) is a transmembrane protein initially found to be expressed in cells of the immune system, but later also found in the cells of numerous normal and tumor tissues, including the head and neck tumors. TLR3 activation can initiate tumor cell survival and progression, but also tumor cell apoptosis. We have investigated whether it is possible to induce tumor cell death by combining TLR3 agonist poly(I:C) with standard treatment with cisplatin and irradiation. Our results showed that poly (I:C) in combination with cisplatin decreases tumor cell growth, promotes apoptosis via caspase -3 and -7 activation and via suppression of genes that inhibit apoptosis, induces ROS formation and cell cycle arrest in G2/M phase. These results suggest that poly(I:C) with cisplatin synergistically reduces the survival and increases the radiosensitivity of tumor cells, thus showing it is a valuable therapeutic option in the treatment of head and neck tumors.

Uvod: Nedostaci cisplatine i drugih registrovanih citostatika (neželjena dejstva, pojava rezistetnosti i visoka toksičnost) otvorila je mogućnost istraživanja u pravcu sinteze novih jedinjenja sa istom ili većom efikasnošću i smanjenom toksičnošću. Kardiotoksičnost je jedna od najčešćih i najozbiljnijh komplikacija primene hemoterapeutika u terapiji karcinoma. Efekti cisplatine i novih metalnih antineoplastičnih agenasa na srce i koronarnu cirkulaciju su gotovo nepoznati. Cilj: Cilj ovog istraživanja je bio ispitivanje i upoređivanje efekata cisplatine (referentna supstanca) i dietil-estra liganda (S,S)etilendiamin-N,N-di-2-(3- cikloheksil)dipropanoat-dihidrohlorida i njegovih kompleksa sa Pt(IV) i Pd(II) na modelu izolovanog srca pacova perfundovanog po Langendorff-ovom modelu. Materijal i metode: Istraživanje je dizajnirano kao eksperimentalna studija sprovedena in vivo i ex vivo. Životinje u eksperimentalnim grupama su tokom četiri nedelje dobijale intraperitonealno po jednu dozu (4 mg/kg telesne mase) ispitivanog jedinjenja, dok su životinje u kontrolnim grupama jednom nedeljno dobijale jednaku zapreminu fiziološkog rastvora ili dimetil sulfoksida (DMSO). Određivani su sledeći parametri srčanog rada: srčana frekvencija - HR, maksimalna stopa promene pritiska u levoj komori - LV(dP/dt)max, minimalna stopa promene pritiska u levoj komori - LV(dP/dt)min, sistolni - SLVP i dijastolni pritisak u levoj komori - DLVP i koronarni protok – CF. Iz uzoraka plazme i koronarnog venskog efluenta spektrofotometrijski su određivani NO2-, TBARS, O2ˉ i H2O2, dok su iz lizata eritrocita na isti način mereni enzimi antioksidacione zaštite (SOD, CAT i GPH). Rezultati: Rezultati ovog istraživanja pokazuju da u primenjenoj dozi i dužini ekspozicije nijedno od ispitivanih jedinjenja nije oštetilo funkciju izolovanog srca pacova tj. nisu ostvarili kardiodepresivni efekat, a sa druge strane novi ligand (L3) ali i njegovi kompleksi sa četvorovalentnom platinom (K3) i dvovalentnim paladijumom (PdL3) imaju respektabilan i obećavajući potencijal ka mobilizaciji antioksidacionih enzima. Zaključak: Dobijeni rezultati istraživanja daju originalan i veoma važan doprinos razumevanju razvoja kardiotoksičnosti izazvane hemoterapeuticima, kao i ispitivanju kardiotoksičnog potencijala potencijalno novih antikancerskih lekova. Introduction: Deficiencies of cisplatin and other registered cytostatics (side effects, resistance and high toxicity) have opened up the possibility of research towards the synthesis of new compounds with the same or greater efficacy and reduced toxicity. Cardiotoxicity is one of the most common and serious complications of the use of chemotherapeutics in cancer therapy. The effects of cisplatin and new metal antineoplastic agents on the heart and coronary circulation are almost unknown. Aim: The aim of this study was to investigate and compare the effects of cisplatin (reference substance) and diethyl-ester ligand (S,S)ethylenediamine-N,N-di-2-(3-cyclohexyl)dipropanoate dihydrochloride and its complexes with Pt(IV) and Pd(II) on an isolated rat heart model perfused according to the Langendorff technique. Material and methods: The investigation was designed as an experimental study conducted in vivo and ex vivo. Animals in the experimental groups received intraperitoneally one dose (4 mg/kg body weight) of the test compound over four weeks, whereas animals in the control groups received an equal volume of saline or dimethyl sulfoxide (DMSO) once a week. The following parameters of heart function rate were determined: heart rate - HR, maximal rate of pressure development in left ventricle - LV(dP/dt)max, minimum rate of pressure development in left ventricle - LV(dP/dt)min, systolic - SLVP and diastolic pressure in left ventricle - DLVP and coronary flow - CF. NO2-, TBARS, O2ˉ and H2O2 were determined spectrophotometrically from plasma and coronary venous effluent samples, while antioxidant protection enzymes (SOD, CAT аnd GPH) were measured on the same way from erythrocyte lysates. Results: The results of this study show that at the administered dose and exposure length, none of the compounds tested impaired the function of the isolated rat heart, ie. have not achieved cardio-depressant effects, while on the other hand, the new ligand (L3) and also its complexes with four-valent platinum (K3) and divalent palladium (PdL3) have a respectable and promising potential for mobilizing antioxidant enzymes. Conclusions: The findings of present research provide an original and very important contribution to understanding the development of cardiotoxicity induced by chemotherapeutics, as well as examining the cardiotoxic properties of potentially new anticancer drugs.

Pulmonary adenocarcinoma is one of the leading causes of death in the world associated with cancer, but eradication or control of this disease is not possible yet. Treatment with chemotherapeutic drugs such as cisplatin has been successful, leading to increased survival of some patients. Its mode of action occurs through direct binding to DNA preventing replication, leading to cell death. However, the development of cisplatin resistance represents a serious clinical problem. Studies focusing on new molecules that are more effective in treating cancer have gained notoriety, such as the antitumor properties of organocalcogen compounds that are used in organic syntheses. The identification of compounds with therapeutic potential is usually initiated in in vitro studies, as it allows the study of human cells in a larger number of combinations, with different parameters. Nevertheless, monolayer (2D) cell culture does not reproduce the spatial complexity of neoplastic cells, thus does not allow the same interactions between cellular and extracellular environments, for example. Thus, there is great interest in cellular models that reproduce more accurate pathophysiological in vitro characteristics found in vivo, such as three-dimensional cultivation. Three-dimensional cultures (spheroids) allow cells to perform cell-cell or cell-matrix interactions. These interactions lead to increased cell differentiation, activate cell signaling due to extracellular matrix components, modify the gene expression pattern previously observed in 2D culture, and alter the expression of molecules involved in matrix and cell-cell adhesion. In three-dimensional culture the cells are arranged in several layers giving a biological barrier to drug diffusion, which will have to diffuse between these layers, similar to what happens in vivo. In cancer, this culture has been widely used, for example, in research that tests drug efficiency. Thus, the general objective of this work was to develop a three-dimensional cellular model using a lung adenocarcinoma lineage and to use this model to screen for molecules with antitumor potential. Protocols were first tested using either matrigel or agarose gel for spheroid formation. After establishment, the morphometric parameters were evaluated by image analysis and the quantified area. In addition, spheroids were treated with nine organocalcogen compounds and compared with cisplatin chemotherapy, cytotoxicity was evaluated by the mtt assay, all compounds and cisplatin were diluted in DMSO. In this study, we made spheroid formation reliable using an A549 human adenocarcinoma cell line only in the tested agarose gel protocol. All compounds and cisplatin significantly decreased cell viability when compared with the control group after 48 hours. Compound 3 had the lowest IC50 value among all compounds tested after 48h exposure. Considering the data presented here, we demonstrate that a three-dimensional adenocarcinoma model is suitable for screening for possible antitumor agents. O adenocarcinoma pulmonar é uma das principais causas de morte no mundo associadas ao câncer, mas a erradicação ou controle dessa doença está longe de ser realidade. O tratamento com quimioterápicos como a cisplatina vem apresentando sucesso, levando ao aumento da sobrevida de alguns pacientes. Seu modo de ação ocorre através da ligação direta ao DNA impedindo a replicação, levando a morte celular. Porém, o desenvolvimento de resistência à cisplatina representa um sério problema clínico. Estudos com foco em novas moléculas mais efetivas no tratamento do câncer vêm ganhando notoriedade, como as propriedades antitumorais dos compostos organocalcogênios que são utilizados em sínteses orgânicas. A identificação de compostos com potencial terapêutico normalmente é iniciada no cultivo celular, pois permite o estudo de células humanas em um número maior de combinações, com diferentes parâmetros. Apesar disso, a cultura de células em monocamada (2D) não reproduz a complexidade espacial das células neoplásicas, assim, não permite as mesmas interações entre os ambientes celular e extracelular, por exemplo. Assim, há um grande interesse em modelos celulares que reproduzam características in vitro fisiopatológicas mais acuradas encontradas in vivo, como, o cultivo tridimensional. Culturas tridimensionais (esferoides) permitem que as células realizem interações célula-célula ou célula-matriz. Essas interações levam ao aumento da diferenciação celular, ativam a sinalização celular devido a componentes da matriz extracelular, modificam o padrão de expressão gênica previamente observado em cultura 2D e alteram a expressão de moléculas envolvidas na adesão de matriz e célula-célula. Na cultura tridimensional as células estão dispostas em várias camadas conferindo uma barreira biológica à difusão de fármacos, que terão que se difundir entre essas camadas, semelhante ao que ocorre in vivo. No câncer, essa cultura tem sido amplamente utilizada, por exemplo, em pesquisas que testam a eficiência de medicamentos. Dessa maneira o objetivo geral deste trabalho foi a padronização do modelo de cultivo celular tridimensional, na linhagem de adenocarcinoma de pulmão A549, e utilizar esse modelo no rastreio de moléculas com potencial antitumoral. Primeiramente foram testados protocolos que utilizaram ou matrigel ou gel de agarose para a formação dos esferoides. Após a padronização, os parâmetros morfométricos foram avaliados por análise de imagem e a área quantificada. Adicionalmente, os esferoides foram tratados com nove compostos organocalcogênios e comparados com o quimioterápico cisplatina, a citotoxicidade foi avaliada pelo ensaio de mtt, todos os compostos e a cisplatina foram diluídos em DMSO. Neste estudo, tornamos confiável a formação dos esferoides usando uma linhagem celular de adenocarcinoma humano A549 somente no protocolo de gel de agarose testado.Todos os compostos e a cisplatina diminuíram significativamente a viabilidade celular quando comparados com o grupo controle após 48 horas. O composto 3 apresentou o menor valor de IC50 entre todos os compostos testados após 48h de exposição. Considerando os dados aqui apresentados, demonstramos que um modelo tridimensional de adenocarcinoma é adequado para o rastreio de possíveis agentes antitumorais.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A quimioterapia envolvendo fármacos a base de Pt(II), como a cisplatina, tem demonstrado resultados efetivos no combate a células cancerosas, apesar dos severos efeitos colaterais devido à alta toxicidade associada com a baixa seletividade desses agentes antitumorais. Uma potencial alternativa para contornar essa problemática ou reduzir os efeitos colaterais consiste da utilização dos sistemas carreadores de fármacos que podem, além de proteger e carrear altas doses de fármaco, também promover a sua liberação lenta e direcionada para os sítios tumorais. Nessa perspectiva, utilizando o método da dinâmica molecular, esse trabalho teve como objetivo a investigação e a caracterização do comportamento dinâmico de complexos de inclusão cddp@NHC, constituídos pela cisplatina (cddp) encapsulada em modelos de nanohorns de carbono (NHC), em solução aquosa. Com relação a esses nanocarreadores a base de carbono, avaliou-se tanto a potencialidade de modelos pristinos, que se diferenciavam em função do ângulo de abertura do cone, quanto de modelos oxidados (NHCox) e reduzidos (NHCh) que se distinguiam em termos do tamanho das nanowindows funcionalizadas resultantes de diferentes graus de oxidação e redução. Os resultados indicaram a presença de até 36 moléculas de água no interior das cavidades dos NHC pristinos, dependendo no ângulo de cone e da presença da cddp. Algumas dessas moléculas de solvente foram expulsas com a inclusão da cddp, apesar de mais de 10 terem permanecido confinadas. Em contraste, complexos com modelos de NHCox e NHCh comportaram até 17 e 15 moléculas de solvente, respectivamente. O aumento do grau de oxidação também se mostrou relevante para a ampliação da área acessível ao solvente e, consequentemente, para a redução da hidrofobicidade dos NHC. A obstrução das nanowindows funcionalizadas impossibilitou o efeito da liberação da cddp. Como reportado em estudos experimentais, as energias livre de ligação (ΔbG) calculadas apontaram que a formação de todos os complexos de inclusão estudados foi termodinamicamente favorável em solução aquosa, o que sugeriu a potencialidade dessas nanoestruturas como carreadoras de fármacos. Enquanto para os complexos com o NHC pristino mais favorável a ΔbG média foi de -35 e -79 kcal.mol⁻¹ para a inclusão de uma e duas cddp, no caso daqueles com os NHCox e NHCh, os referidos valores foram de -47 e -77 kcal.mol⁻¹ e de -40 e -74 kcal.mol⁻¹ respectivamente, sendo que a maior parte dessa estabilidade foi proveniente das contribuições de van der Waals. Chemotherapy involving Pt(II)-based drugs, such as cisplatin, has demonstrated effective results against cancer cells, despite the severe side effects due to the high toxicity associated with the low selectivity of these antitumor agents. A potential alternative to overcome or decrease the side effects is to use drug delivery systems, which can provide protection against degradation, carry high doses of the anticancer drug and promote its slow and targeted release to the tumor sites. Herein, by using the molecular dynamics method, the purpose of this work was to investigate and characterize the dynamical behavior of the inclusion complexes cddp@CNH, formed by the encapsulated cisplatin (cddp) inside carbon nanohorns (CNH) models, in aqueous solution. In relation to these nanocarriers based on carbon atoms, it was evaluated the potentiality of both pristine models with different opening cone angles, and oxidized (CNHox) and reduced (CNHh) models, which were distinguished in terms of the size of the functionalized nanowindows generated by oxidation and reduction stages. The results indicated the presence of up to 36 water molecules inside the empty CNH cavity, depending on the cone angle and the presence of the cddp. Some of these solvent molecules are expelled out to the bulky upon cddp inclusion, although more than 10 molecules remain confined. In contrast, the complexes including CNHox and CNHh prototypes carried up to 17 and 15 solvent molecules, respectively. The increase in the degree of oxidation was also relevant for the expansion of the solvent accessible surface area, and consequently, for the hydrophobicity reduction of CNH. The effect of cddp release was made impossible by the obstruction of functionalized nanowindows. Moreover, as reported in experimental studies, the calculated binding free energy (ΔbG) pointed out that the inclusion complexes formation between CNH structures and up to two cddp molecules was thermodynamically favorable in aqueous media, which suggests the potential of this carbon nanostructures as drug carriers. While for the complexes with the most likely pristine CNH the average ΔbG was -35 and -79 kcal mol⁻¹ for inclusion of one and two cddp, respectively, for the complexes with CNHox and CNHh the referred values of the average ΔbG were -47 e -77 kcal mol⁻¹ and -40 e -74 kcal mol⁻¹ respectively, with most of the complex stability coming from the van der Waals contributions.

Introduction: Ototoxicity is a health problem appearing after powerful treatments in serious health conditions. It is sometimes inevitable when treatment of the serious disease is required. Cisplatin is an antineoplastic agent which was investigated previously to reveal increased nitrogen and reactive oxygen radicals that damages hair cells, resulting in ototoxicity. N-acetylcysteine, previously shown to decrease ototoxicity caused by different agents, is known to be a powerful in vitro antioxidant. Probably N-acetylcysteine, in addition to its antioxidant effect, blocks a cascade where reactive oxygen species result in apoptosis in the cochlea. Objectives: The possible preventive effect of N-acetylcysteine in cisplatin ototoxicity was studied with auditory brain stem responses, otoacoustic emissions, and histopathological investigation of the cochlea in a scanning electron microscopy. Methods: This study was conducted on 21 Wistar Albino rats in four groups. 1 mL/kg/day three times in total intraperitoneal (i.p.) Saline (n = 5), 500 mg/kg/day i.p. three times in total N-acetylcysteine (n = 5), i.p. 15 mg/kg cisplatin alone (single dose) (n = 5) and i.p. 15 mg/kg cisplatin plus 500 mg/kg/day N-acetylcysteine (n = 6) were administered. The rats were anesthetized to study the hearing tests before and after the experiment. The rats were sacrificed to investigate the cochleas by scanning electron microscopy. Results: Auditory brain stem responses and otoacoustic emissions values were attenuated in the cisplatin group. The group that received N-acetylcysteine in addition to cisplatin had better auditory brain stem responses thresholds and otoacoustic emissions. The samples obtained from the cisplatin group showed surface irregularities, degeneration areas, and total or partial severe stereocilia losses. The changes were milder in the cisplatin + N-acetylcysteine group. Conclusion: Cisplatin ototoxicity can be detected by auditory brain stem responses and otoacoustic emissions testing in rats. N-acetylcysteine may protect the cochlear cells from histopathological changes. We concluded that N-acetylcysteine given 4 h after cisplatin injection has a potential otoprotective effect against cisplatin ototoxicity. which suggests it could be used in clinical trials. Resumo: Introdução: A ototoxicidade é um problema que pode ocorrer após certos tipos de tratamentos para condições graves de saúde. Às vezes é inevitável quando o tratamento da doença é necessário. A cisplatina é um agente antineoplásico cujo uso em pesquisas anteriores demonstrou aumentar os radicais livres de nitrogênio e espécies reativas de oxigênio que danificam as células ciliadas e resultam em ototoxicidade. Por outro lado, a N-acetilcisteína, que já demonstrou diminuir a ototoxicidade causada por diferentes agentes, é conhecida por ser um potente antioxidante in vitro. Provavelmente a N-acetilcisteína, além de seu efeito antioxidante, bloqueia uma cascata onde espécies reativas de oxigênio resultam em apoptose na cóclea. Objetivos: Estudar o possível efeito preventivo da N-acetilcisteína na ototoxicidade por cisplatina por meio de potencial evocado auditivo de tronco encefálico, emissões otoacústicas e investigação histopatológica da cóclea por microscopia eletrônica de varredura. Método: Este estudo foi realizado em 21 ratos albinos Wistar, separados em quatro grupos. Foram administrados: 1 mL/kg/dia intraperitoneal (i.p.) de solução salina (n = 5), três vezes no total; 500 mg/kg/dia i.p. de N-acetilcisteína (n = 5), três vezes no total; 15 mg/kg i.p. (dose única) somente de cisplatina (n = 5) e 15 mg/kg i.p. de cisplatina e 500 mg/kg/dia i.p. de N-acetilcisteína (n = 6). Os ratos foram anestesiados para estudo dos testes auditivos antes e depois do experimento. Os ratos foram sacrificados para investigação da cóclea por microscopia eletrônica de varredura. Resultados: Os potenciais evocados auditivos de tronco encefálico e os valores das emissões otoacústicas estavam atenuados no grupo cisplatina. O grupo que recebeu N-acetilcisteína além da cisplatina apresentou melhores limiares de respostas auditivas do tronco encefálico e emissões otoacústicas. As amostras obtidas do grupo cisplatina apresentaram irregularidades de superfície, áreas de degeneração, com perdas graves totais ou parciais de estereocílios. As alterações foram mais leves no grupo cisplatina + N-acetilcisteína. Conclusão: A ototoxicidade por cisplatina pode ser detectada por meio de potenciais evocados auditivos de tronco encefálico e pelo teste de emissões otoacústicas em ratos. A N-acetilcisteína pode proteger as células cocleares contra alterações histopatológicas. Concluímos que a N-acetilcisteína administrada 4 horas após a injeção de cisplatina tem potencial efeito otoprotetor contra a ototoxicidade por cisplatina e pode ser utilizada em ensaios clínicos. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES Cisplatin is a widely used chemotherapeutic to treat several types of tumors on both adult and pediatric subjects. Although cisplatin is widely used, its use is associated with serious toxic effects, including nephrotoxicity and hepatotoxicity. Thus, the present study investigated the effects caused by cisplatin acute administration on newborn Wistar rats and the differences in the drug toxicity compared to adult rats. The study was carried out using male and female Wistar rats 10 and 60 day-old (CEUA nº 2699300315). Rats received cisplatin at doses of 5 and 10 mg/kg, by the intraperitoneal (ip.) route, whereas the control group received saline (0.9%, ip.). After 24 h of cisplatin/saline administration, the animals were killed and samples of blood, liver, and kidneys were collected for ex vivo analyses. Markers of renal (urea) and hepatic damage (alanine aminotransferase and aspartate aminotransferase activities) were determined in the serum samples. Parameters of oxidative stress (levels of reactive oxygen species, lipid peroxidation, carbonyl protein, non-protein thiols (NPSH)); activities of antioxidant enzymes (superoxide dismutase, catalase (CAT), glutathione peroxidase, glutathione- S-transferase and glutathione reductase) and activities of sulfhydryl enzymes (δ-aminolevulinate dehydratase (δ-ALA-D) and Na +, K + - ATPase) were determined in samples of liver and kidney of rats. The levels of proteins related to oxidative stress and apoptosis were also determined in samples of liver and kidney of rats at both ages. The cisplatin acute administration caused hepatorenal toxicity in both neonatal and adult rats. However, the pattern and the severity of the damages were different among the ages and tissues. Newborn rats presented greater oxidative metabolic damage when compared to adult rats, characterized by an increase in reactive species and carbonyl protein levels, lower NPSH content, and greater inhibition of δ-ALA-D and CAT activities. In addition, a faster molecular response was found in the protein levels involved with apoptosis and response to oxidative stress. In conclusion, the data from this study show that the pattern of cisplatin toxic effects was different among the ages and tissues of rats. The present study also revealed that newborn rats are more sensitive to treatment with cisplatin for at least the first 24 h and further showed that liver damage, at least in the beginning, was greater than renal damage in adult rats. Thus, the present study demonstrated that there are differences in the body response to acute exposure of cisplatin among adult and newborn rats, reinforcing the need for further studies on children response to chemotherapy and possible effective strategies to minimize the damage caused by cisplatin. A cisplatina é um quimioterápico amplamente utilizado para tratar diversos tipos de tumores, tanto em adultos quanto em crianças. Apesar de bastante difundido, o uso da cisplatina está relacionado a efeitos tóxicos graves, dentre eles a nefrotoxicidade e a hepatotoxicidade. Desta forma, a presente dissertação investigou os efeitos causados pela administração aguda de cisplatina em ratos com 10 dias e as diferenças na resposta ao fármaco, no que se refere à toxicidade, quando comparada a ratos de 60 dias. Para a realização deste modelo experimental foram utilizados ratos Wistar machos e fêmeas, com 10 e 60 dias (CEUA nº 2699300315). Os animais receberam a cisplatina nas doses de 5 e 10 mg/kg pela via intraperitoneal (i.p.), enquanto o grupo controle recebeu a solução salina (0,9%; i.p.). Após 24 h da administração de cisplatina/salina, os animais foram mortos e o sangue, o fígado e os rins foram coletados para as análises ex vivo. Nas amostras de soro foram avaliados os parâmetros de dano renal (ureia) e hepático (atividade das enzimas alanina aminotransferase, aspartato aminotransferase). Enquanto que nas amostras de fígado e rim foram determinados: os marcadores de estresse oxidativo (níveis de espécies reativas de oxigênio, peroxidação lipídica, carbonilação de proteínas e tióis não-proteicos (NPSH)); e as atividades das enzimas antioxidantes (superóxido dismutase, catalase (CAT), glutationa peroxidase, glutationa-S-transferase e glutationa redutase); assim como as atividades das enzimas sulfidrílicas δ-aminolevulinato desidratase (δ-ALA-D) e Na+, K+- ATPase. Os níveis renais e hepáticos das proteínas relacionadas com o estresse oxidativo e com a apoptose também foram determinados. A administração aguda de cisplatina causou toxicidade hepatorrenal em ratos recém-nascidos e adultos. No entanto, o padrão e a intensidade dos danos foram diferentes entre as idades e os tecidos. Os ratos recém-nascidos apresentaram um dano oxidativo mais acentuado em relação aos ratos adultos, caracterizado por um aumento nos níveis de espécies reativas e na proteína carbonila, um menor conteúdo de NPSH e uma maior inibição nas atividades da δ-ALA-D e CAT. Além disso, foi observada uma resposta molecular mais rápida nos níveis proteicos envolvidos com apoptose e resposta ao estresse oxidativo. Em conclusão, os dados deste estudo mostram que o padrão de efeitos tóxicos da cisplatina foi diferente entre as idades e os tecidos dos ratos. Além disso, o presente estudo revelou que os ratos recém-nascidos são mais sensíveis ao tratamento com a cisplatina, pelo menos nas primeiras 24 horas e ainda mostrou que, pelo menos no início, o dano hepático foi maior do que o renal em ratos adultos. Assim, o presente estudo demonstrou que existem diferenças na resposta corporal à exposição aguda da cisplatina entre ratos adultos e recém-nascidos, reforçando a necessidade de mais estudos sobre a resposta da criança à quimioterapia e possíveis estratégias efetivas para minimizar os danos causados pela cisplatina.

Cancer cells are able to adapt to different stress factors such as hypoxia, which is caused by insufficient tumor vascularization. An increased acetylation status of histones H3 and H4 in UKF-NB-3 and UKF-NB-4 neuroblastoma cell lines was found to be a mechanism of adaptation of these cells to hypoxia. An increase in acetylation of histones H3 and H4 is suggested to cause changes in the structure of chromatin that lead to activation of gene transcription. In addition, cultivation of tested neuroblastoma cells under hypoxic conditions changes expression of proteins of a transcription factor N-myc, which is essential for development of neuroblastomas. This transcription factor is also responsible for a metabolic adaptation of neuroblastoma cells, increases their aggressiveness and its expression leads to a worse prognosis of the disease. Stanice melanoma čovjeka izrazito su otporne na djelovanje protutumorskih lijekova. Stoga je važno razviti nove terapije, a jedna od njih je ciljana terapija inhibitorima tirozin kinaza i mogućnost njihove kombinacija s protutumorskim lijekovima. Stoga je u ovom radu na modelu četiri stanične linije melanoma čovjeka istražena mogućnost liječenja melanoma kombinacijom dasatiniba s protutumorskim lijekovima paklitakselom, vinkristinom ili cisplatinom. Preživljenje stanica nakon izlaganja navedenim spojevima ili njihovim kombinacijama određeno je testom MTT, dok je računanjem kombinacijskog indeksa procijenjen zajednički učinak. Pokazano je da su stanice RPMI-7951 i MeWo osjetljive na paklitaksel, vinkristin i cisplatinu, za razliku od stanica SK-MEL-3 koje pokazuju otpornost na sva tri protutumorska lijeka i stanica A375 koje su otporne na paklitaksel i vinkristin, a osjetljive na cisplatinu. Stanice RPMI-7951 pokazale su se osjetljivim na dasatinib, a njegova kombinacija sa paklitakselom, vinkristinom i cisplatinom daje antagonistički učinak. Kod stanica A375 uočena je slaba osjetljivost na dasatinib, a kombinacija sa cisplatinom pokazala je sinergistički učinak. Na temelju određivanja količine Src/pSrc(Y418) metodama SDS-PAGE i Western blot i rezultata preživljenja nakon utišavanja kinaze Src u stanicama RPMI-7951 zaključeno je da pSrc(Y418) nije meta dasatiniba te da su potrebna daljnja istraživanja u cilju otkrivanja biomarkera za predviđanje osjetljivost stanica melanoma na dasatinib. Human melanoma cells are highly resistant to antitumor drugs. Therefore, it is important to develop new therapies such as targeted therapy with tyrosine kinases inhibitors, as well as combining those inhibitors with antitumor drugs. In this research the potential of treating melanoma with the combination of dasatinib and antitumor drugs paclitaxel, vincristine or cisplatin in four human melanoma cell lines was investigated. Cell survival was determined by MTT test, and combined effect was assessed by calculation of combination index. Cell lines RPMI-7951 and MeWo were shown to be sensitive to paclitaxel, vincristine and cisplatin, SKMEL- 3 cells were shown to be resistant, while A375 cells were shown to be resistant to paclitaxel and vincristine but sensitive to cisplatin. RPMI-7951 cells demonstrated sensitivity to dasatinib, and its combination with paclitaxel, vincristine and cisplatin was shown to have antagonistic effect. A375 cells demonstrated a mild sensitivity to dasatinib, and its combination with cisplatin was shown to have a synergistic effect. On the basis of the amount of Src/pSrc(Y418) determined by SDS-PAGE and Western blot and the survival of RPMI-7951 cells upon silencing Src kinase, it is concluded that pSrc(Y418) is not a dasatinib target, emphasizing the need for identification of biomarkers that might predict sensitivity to dasatinib in melanoma cells.

Dissertação de mestrado em Genética Molecular, Despite all the advances made in cancer therapy during the last decades, cancer remains a death sentence for many patients. Ruthenium, per se, presents anticancer features which allow its targeting towards cancer cells, making it interesting in the research for novel therapeutic agents. In an attempt of searching for new anticancer agents, a family of ruthenium-derived compounds was synthesized, using a multifunctional approach in the design of the new molecules that exploits different tumor properties, in order to increase the specific targeting of these compounds to cancer cells. Here, we aimed at studying the anticancer effect of these compounds and identifying possible cellular targets in breast and colorectal cancer cells. We evaluated the effect of these agents on proliferation, viability, cellular mobility, cytoskeleton stability, as well as their effects on β-actin and GLUT1 expression. We observed that our compounds are more potent in these models than the classical chemotherapeutic agent cisplatin, as they show lower IC50 values. Our compounds inhibited cell proliferation and diminish the ability of breast and colorectal cancer cells to form colonies. We could also observe that these agents induce structural changes in the cytoskeleton, namely in Factin filaments, which is accompanied by alterations on the expression of β-actin in both cancer models. These drugs seem to inhibit the migratory ability of breast cancer cells although not showing statistical significance in colorectal cancer cells. Moreover, exposure to our compounds induce changes in GLUT1 expression in cancer cells, which might be associated to the different levels of sensitivity of cancer cells to these agents. Overall, we demonstrate that the multifunctional approach used in the design of the compounds increases their anticancer activity. The first steps towards the comprehension of the cellular and molecular effects of this family of compounds were taken with this study, although further studies are needed in order to better understand the cellular effects of these anticancer agents and identify their cellular targets., Apesar dos inúmeros avanços feitos nas últimas décadas no âmbito das terapias contra o cancro, esta doença representa ainda uma sentença de morte para vários pacientes. O ruténio apresenta certas propriedades anticancerígenas que permitem o seu direcionamento para as células tumorais, o que o torna interessante na investigação de novos agentes terapêuticos. Numa tentativa de descoberta de novos agentes anticancerígenos, uma família de compostos derivados do ruténio foi sintetizada, utilizando uma abordagem multifuncional na síntese destas novas moléculas e que tenta tirar partido de diferentes propriedades tumorais para a entrega dirigida destes compostos. Com este trabalho pretendemos estudar os efeitos anticancerígenos destes compostos e identificar possíveis alvos celulares, utilizando células derivadas de cancros da mama e colorretal. Avaliou-se os efeitos destes agentes na proliferação, viabilidade e mobilidade destas células, assim como os seus efeitos na estabilidade do citoesqueleto, na expressão de 􀈾-actina e de GLUT1. Nestes modelos, os compostos apresentam uma maior potência do que a cisplatina, um agente quimioterápico clássico, uma vez que têm valores de CI50 inferiores. Constatou-se que os compostos inibem a proliferação celular e diminuem a capacidade de formação de colónias de células de cancros de mama e colorretal. Descobriu-se ainda que estes agentes alteram a estrutura do citoesqueleto, nomeadamente ao nível dos filamentos de F-actina, o que está associado à alteração da expressão de 􀈾-actina nas linhas celulares utilizadas. Para além disso, estes compostos parecem inibir a capacidade migratória das células de cancro da mama, não interferindo de forma significativa nas linhas celulares de cancro colorrectal. Verifica-se também que a exposição aos compostos altera a expressão de GLUT1, o que poderá estar associado a diferenças na sensibilidade das células. Em suma, demonstrou-se que a abordagem multifuncional utilizada no desenho dos compostos melhorou a sua atividade anticancerígena. Neste trabalho foram dados os primeiros passos no sentido da caracterização dos efeitos celulares e moleculares desta família de compostos, apesar de serem necessários mais estudos para melhor identificar os efeitos e os alvos celulares destes agentes.

Resumo: O mesotelioma deciduóide é um subtipo raro de mesotelioma epitelióide, inicialmente encontrado apenas no peritoneu de mulheres jovens, sem relação com exposição a asbestos. No entanto, desde as primeiras descrições tem sido igualmente diagnosticado na pleura e no pericárdio e pode estar associado aos asbestos. De um modo geral, tem pior prognóstico quando comparado com o mesotelioma epitelióide. Há apenas 45 casos descritos na literatura até à data, 22 destes localizados na cavidade pleural. Os autores descrevem um caso de mesotelioma deciduóide pleural numa mulher de 40 anos com toracalgia pleurítica direita, sem história de exposição aos asbestos, submetida a quimioterapia seguida de ressecção cirúrgica, que veio a falecer no pós-operatório. Abstract: Deciduoid Mesothelioma is a rare variant of epithelioid mesothelioma; it was initially thought that it only occurred in the peritoneum of young women and had nothing to do with asbestos exposure. However, since these early findings it has also been observed in the pleura and the pericardium, with possible association to asbestos. In general the prognosis is poor compared to epithelioid mesothelioma. 45 cases have been reported in the literature up to now, 22 of these were located in the pleural cavity. The authors describe a case of deciduoid pleural mesothelioma in a 40-year-old-woman who presented with right pleuritic chest pain, with no history of asbestos exposure, treated with chemotherapy followed by surgery and who died postoperatively. Mesenchymal stem cells (MSCs) are known for their important properties involving multilineage differentiation potential., trophic factor secretion and localization along various organs and tissues. On the dark side, MSCs play a distinguished role in tumor microenvironments by differentiating into tumor-associated fibroblasts or supporting tumor growth via distinct mechanisms. Cisplatin (CIS) is a drug widely applied in the treatment of a large number of cancers and is known for its cytotoxic and genotoxic effects, both in vitro and in vivo. Here we assessed the effects of CIS on MSCs and the ovarian cancer cell line OVCAR-3, by MTT and comet assays. Our results demonstrated the resistance of MSCs to cell death and DNA damage induction by CIS, which was not observed when OVCAR-3 cells were exposed to this drug.

Tese de doutoramento em Biociências, na especialidade de Toxicologia, apresentada ao Departamento de Ciências da Vida da Faculdade de Ciências e Tecnologia da Universidade de Coimbra Um tumor é constituído por diferentes tipos de células, entre as quais as células estaminais tumorais. Células estaminais tumorais apresentam características únicas como rápida proliferação, plasticidade e pluripotência, tornando-as de elevado interesse clínico. As células estaminais tumorais são assim apontadas como sendo células iniciadoras de tumores bem como responsáveis da reincidência da doença. É também consensual que as células estaminais tumorais são mais resistentes às terapias convencionais, como a quimioterapia e radioterapia, quando comparando com a população heterogénea de células tumorais normais. Este trabalho pretendeu esclarecer as características desta subpopulação de células estaminais tumorais e o que as tornam mais resistentes à morte celular quando comparando com as células mais diferenciadas. O modelo celular que usámos, células P19 de carcinoma embrionário tem como vantagem a utilização de uma só linha celular, podendo-se obter células P19 estaminais de carcinoma (P19SCs) ou células P19 de carcinoma mais diferenciado (P19dCs) que nos permitirão investigar diferentes resultados observados relativamente ás terapias cancerigenas. Como tal, pretendemos verificar as diferenças fisiológicas mitocondriais bem como a sua capacidade de resistir e detectar danos de DNA, factores que contribuem para a resistência à morte celular das P19SCs quanto comparadas com as P19dCs. A mitocôndria está ligada a diversos processos celulares para além do seu papel na produção de energia. Neste trabalho, a mitocôndria demonstrou ser crucial nos processos celulares de heterogeneidade, de auto-renovação e de diferenciação, processos chaves no potencial tumorogénico das células cancerígenas. De facto, existe uma relação entre o metabolismo e o destino celular escolhido. As células alteraram as suas preferências metabólica, bem como a proliferação, auto-renovação e diferenciação a fim de minimizar o stress, manter um potencial estaminal máximo e estimularem a sobrevivência. P19SCs demonstraram ser essencialmente glicolíticas no processo de obtenção de energia e apresentaram mitocôndrias de características distintas. Quando forçadas a mudar o seu metabolismo energético para um metabolismo mais oxidativo, um processo de diferenciação é iniciado com consequente redução da proliferação celular. Neste trabalho as drogas quimioterapêuticas cisplatina, etoposídeo e doxorubicina foram utilizadas como agentes de danos de DNA, de modo a estudarmos a resistência das células estaminais cancerígenas, bem como esclarecer que vias podem estar envolvidas nesta resistência descrita na literatura. Com o nosso trabalho verificámos que cada uma destas drogas têm diferentes tipos de efeitos nas células P19SCs e nas P19dCs. De facto, as células mais diferenciadas parecem ser mais afectadas por todas as drogas (mais danos de DNA) em que o mecanismo de apoptose dependente da via das caspases se encontra activo. As P19SCs tratadas com doxorubicina parecem igualmente afectadas quando comparando com as P19dCs, demonstrando que, contrariamente ás outras duas drogas testadas, a doxorubicin provoca danos mais severos, até mesmo para as P19SCs. Nas P19SCs tratadas com cisplatina que não apresentam danos de ADN de cadeia dupla (mais severos) e que estão presentes nas células mais diferenciadas, vias como paragem no ciclo celular, activação da proteína PARP-1 envolvida na reparação celular, bem como a activação do mecanismo de autofagia (mecanismos não observados nas células mais diferenciadas), contribuem para a resistência destas aos danos de ADN provocados pela cisplatina (maioritariamente danos de cadeia simples). Para além destas vias, um mecanismo de diferenciação e manutenção do potencial de pluripotência pode também estar associado ao mecanismo de resistência. Nas P19SCs tratadas com etoposídeo que não apresentam danos de ADN de cadeia dupla (mais severos) detectados nas P19dCs, apesar da paragem do ciclo celular ser igual em ambas os tipos de células estudadas, a expressão da proteina PARP-1 bem como o mecanismo de autofagia estar activo contribuem para a resistência destas aos danos de ADN provocados pelo etoposídeo. Aliás a resistência das células estaminais ao etoposídeo pode passar pela manutenção do potencial de pluripotência. Em conclusão podemos afirmar que as P19SCs e as P19dCs reagem de modo diferente a cada umas das drogas, e que estas diferenças podem justificar e esclarecer a resistência das células estaminais tumorais a um dos tratamentos mais utilizados no cancro, a quimioterapia. Cancer stem cells (CSCs) are amongst the heterogeneous group of cells constituting tumors. CSCs present unique characteristics such as their high proliferative ability, plasticity and pluripotency, making these cells clinically relevant. In recent years, CSCs have been pointed out as tumor initiators and as responsible for disease recurrence. Moreover, is also consensual that CSCs are more resistant to conventional anti-cancer therapies, namely chemotherapy and radiotherapy, when compared with other cells constituting the tumor. This work was designed to clarify the characteristics that turn this subpopulation of cells more resistant to cell death when compared to their differentiated counterparts. To achieve this, we used P19 embryonic cancer cells that constitute a great model as it allows the study to be performed with a single cell line from which one can obtain both P19 cancer stem cells (P19SCs) and P19 differentiated cancer cells (P19dCs). By working with these P19 cell groups, we intended to investigate the distinct outcomes when applying different anti-cancer therapies. In this regard, our study focused on the identification of the main mitochondrial physiological alterations between P19dCs and P19SCs, as well as the characterization of each cell group capacity to resist and detect DNA damage. This knowledge comes with high relevance as together these attributes are thought to contribute to CSCs resistance to cell death. In addition to their role in energy production, mitochondria have been associated with important roles in different cell processes. Along this work, this organelle was shown to be crucial in heterogeneity cell processes, self-renewal, and differentiation, all of which contribute for the tumorigenic potential of cancer cells. Our results support the existence of a link between metabolism and cell faith. Cells were shown to alter their metabolic preferences, proliferation as well as selfrenewal and differentiation in order to minimize stress, maintain a maximum stemness potential and stimulate survival. P19SCs exhibited a more glycolytic metabolism and mitochondria from this cell group had particular characteristics that, when forced to an oxidative metabolism, leads to the initiation of a differentiation process resulting in a reduction of cell proliferation. The study of CSCs resistance to chemotherapeutic drugs was performed with cisplatin, etoposide and doxorubicin aiming the characterization and clarification of the pathways involved in the resistance described in the literature. Interestingly, the obtained results revealed that each one of the drugs used in the study hads different effects in P19SCs when compared to P19 cells differentiated with retinoic acid. In fact, differentiated cells were more affected by the drugs used as these cells present more DNA damage, in which caspase-dependent apoptosis pathway was activated. P19SCs were similarly affected to doxorubicin treatment when compared to P19dCs, revealing that in contrast to the other drugs used, cell damage induced by doxorubicin may be too severe impairing P19SCs resistance ability. Nevertheless, in contrast with more differentiated cells, P19SCs treated with cisplatin did not present double strand DNA damage, highly severe. Furthermore, mechanisms such as cell cycle arrest, PARP-1 activation, which is associated with cell repair, and autophagy activation, were not observed in P19dCs in contrast to P19SCs suggesting that the described mechanisms may contribute for the later cell group resistance to cisplatin-induced DNA damage. Additionally, the maintenance of a pluripotency potential or even a diferentiation processess might also be related with P19SCs resistance to cisplatin.. Similarly to our observations for cisplatin, etoposide treatment did not result in double strand DNA damage in P19SCs, however these cells showed identical cell cycle arrest pattern to their differentiated counterparts. Despite this similarity, P19SCs have higher PARP-1 expression, which together with autophagy activation might explain the observed resistance to DNA damage upon treatment with etoposide. Additionally, the maintenance of a pluripotency potential might also be related with P19SCs resistance to this drug. In summary, we conclude that P19SCs operate differently when subjected to distinct treatments and when compared to P19dCs. Overall, our observations allow for a better knowledge of the mechanism involved in cancer stem cell resistance to chemotherapeutics as it allows the understanding of the described resistance of cancer stem cells to anti-cancer treatments applied. O trabalho propõe a discussão sobre a construção do vocabulário político moderno utilizado na província do Rio Grande e na Banda Oriental durante o processo de formação das Cortes de Portugal, no processo de independência brasileiro, durante a elaboração da constituição nacional, na anexação da Província Cisplatina ao território brasileiro e no início da guerra de independência em relação ao Brasil. Termos como povo, nação, constituição e soberania, além de outros como opinião pública e autonomia política são considerados à luz dos eventos políticos no território brasileiro e oriental entre os anos de 1821 e 1825. Também a relação que se estabeleceu entre o império brasileiro e a condição da Província Cisplatina, principalmente sobre a relativa autonomia política, são consideradas nessa pesquisa. The paper proposes a discussion on the construction of modern political vocabularies used in the province of Rio Grande and Eastern Band during the process of formation of the Cortes of Portugal, the Brazilian independence process during the drafting of the national constitution, the annexation of cisplatin province to Brazil and at the beginning of the war of independence in relation to Brazil. Terms such as people, nation, constitution and sovereignty, as well as opinion public and political autonomy are considered in the light of political events in Brazil and eastern territory between the years 1821 and 1825. Also the relationship established between the Brazilian Empire and the condition of cisplatin province manly on the relative political autonomy is considered in this research.
