Starch, as a typical polysaccharide with natural spherical morphology, is not only a preferred precursor for preparing carbon materials but also a model polymer for investigating thermochemical evolution mechanisms. However, starch usually suffers from severe foaming and low carbon yield during direct pyrolysis. Herein, we report a simple and eco-friendly dry strategy, by maleic anhydride initiating the esterification of starch, to design carbon microspheres against the starch foaming. Moreover, the influence of ester grafting on the pyrolytic behavior of starch is also focused. The formation of ester groups in precursor guarantees the structural stability of starch-based intermediate because it can promote the accumulation of unsaturated species and accelerate the water elimination during pyrolysis. Meanwhile, the esterification and dehydration reactions greatly deplete the primary hydroxyl groups in the starch molecules and thus the rapid levoglucosan release is inhibited, which well keeps the spherical morphology of starch and ensures the high carbon yield. In further exploration as anode materials for Lithium-ion batteries, the obtained carbon microspheres exhibit good cyclability and rate performance with a reversible capacity of 444 mAh g−1 at 50 mA g−1. This work provides theoretical fundamentals for the controllable thermal transformation of biomass towards wide applications.

Effect of temperature (300–500 °C), carrier gas flow rate (0.2–0.8 L min−1), and the heating rate (10–20 °C min−1) on the final bio-oil production from the Salvinia auriculata (SA) and Ulva lactuca (UL) was optimized using response surface methodology (RSM), established by a central composite design (CCD). The maximum bio-oil yield from UL was 34.8%, reached at 500 °C, 0.2 L min−1 nitrogen flow rate, and 10 °C min−1 heating rate. However, in the case of SA feedstock, the highest bio-oil yield was 32.3% at 400 °C, 0.5 L min−1 nitrogen flow rate, and 20 °C min−1 heating rate. Both bio-oil samples contained saturated and unsaturated hydrocarbons; but the average hydrocarbon chain length in UL bio-oil (C4–C16) was relatively shorter than bio-oil from SA (C6– C24). The bio-oil from A. filiculoides exhibited higher HHV values than that of UL derived bio-oil due to its relative large carbon and hydrogen concentration and small oxygen content. Although both the bio-oils showed different heating values, the UL biooil had more appropriate properties, i.e. lower viscosity and density.

A route is present for preparing porous titanium scaffolds with open and interconnected pore morphology through calcium thermal in-situ reduction of TiO precursor. Titanium and dioxide titanium powders, with ammonium carbonate as the space holder, were used to prepare porous precursor whose main phase is TiO by a sintering process. Then calcium vapor was conduced to reduce the precursor and CaO was removed by leaching to obtain porous titanium structures. The morphology, pore structure, phase analysis of porous titanium precursor and porous titanium scaffolds were investigated by means of XRD, SEM and EDS. The porosity was observed by Archimedes method. By this route, the porous titanium with multi-gradient pore sizes ranging from a few microns to several hundred microns was formed. The porous titanium is of multi-gradient pore size distribution of 100 μm bigger, 10 μm∼100 μm and 10 μm smaller. The micro-pores on the pore walls increases connectivity between the macro-pores throughout the porous titanium interior. The porous titanium obtained by this method has a porosity of 50–65% and a titanium content more than 99%.

China’s Mars rover Zhurong successfully landed on Mars on 15 May 2021, and it is currently conducting an exploration mission on the Red Planet. This paper develops slip estimation models for the Mars rover Zhurong based on the data drive approach. Data were obtained by Zhurong’s validator ground indoor tests, and the test site was equipped with a low-gravity simulation device and simulated Mars soil to simulate the Mars conditions as much as possible. The obtained slip models trained by BP and GA-BP algorithms were applied to estimate Zhurong’s longitudinal (slip_x) and lateral slip (slip_y) on Mars, and the slip estimation values were used to display Zhurong’s actual driving path. The analyzed results prove that the GA-BP slip models perform better than the BP models, and can both be applied for correcting Zhurong’s path. The proposed models have high potential in guiding the path planning and monitoring of the slip for the Mars rover Zhurong.

Background Quizalofop-p-ethyl (QPE), a unitary R configuration aromatic oxyphenoxypropionic acid ester (AOPP) herbicide, was widely used and had led to detrimental environmental effects. For finding the QPE-degrading bacteria and promoting the biodegradation of QPE, a series of studies were carried out. Results A QPE-degrading bacterial strain YC-XJ1 was isolated from desert soil and identified as Methylobacterium populi, which could degrade QPE with methanol by cometabolism. Ninety-seven percent of QPE (50 mg/L) could be degraded within 72 h under optimum biodegradation condition of 35°C and pH 8.0. The maximum degradation rate of QPE was 1.4 mg/L/h, and the strain YC-XJ1 exhibited some certain salinity tolerance. Two novel metabolites, 2-hydroxy-6-chloroquinoxaline and quinoxaline, were found by high-performance liquid chromatography/mass spectroscopy analysis. The metabolic pathway of QPE was predicted. The catalytic efficiency of strain YC-XJ1 toward different AOPPs herbicides in descending order was as follows: haloxyfop-p-methyl ≈ diclofop-methyl ≈ fluazifop-p-butyl > clodinafop-propargyl > cyhalofop-butyl > quizalofop-p-ethyl > fenoxaprop-p-ethyl > propaquizafop > quizalofop-p-tefuryl. The genome of strain YC-XJ1 was sequenced using a combination of PacBio RS II and Illumina platforms. According to the annotation result, one α/β hydrolase gene was selected and named qpeh1, for which QPE-degrading function has obtained validation. Based on the phylogenetic analysis and multiple sequence alignment with other QPE-degrading esterases reported previously, the QPEH1 was clustered with esterase family V. Conclusion M. populi YC-XJ1 could degrade QPE with a novel pathway, and the qpeh1 gene was identified as one of QPE-degrading esterase gene.How to cite: Li X, Wang J, Wu W, et al. A rapid preparation method of TiO2-x by thermal treatment using H2TiO3 as the precursor was proposed compared with the static hydrogenation thermal treatment process. Its adsorption properties of arsenic under visible light were explored and investigated as well. Various colors of TiO2-x were prepared and characterized via XRD, TEM, BET, and so on. The results indicate that the method of rapid preparation is feasible. The TiO2-x exhibits a larger particle size that varied from 10 nm to 2 μm, and deeper color products were obtained as the treatment temperature increased from 600 to 900 °C. Light yellow TiO2-x was prepared after increasing the temperature from 600 to 900 °C, Ti4O7 and Ti6O11 with a dark color were formed under a H2 atmosphere at 1500 °C. The arsenic adsorption performances of some samples under visible light were tested, and reveal a high efficiency of TiO2-x in the photocatalytic oxidation arsenic adsorption under visible light, the conversion ratio of As(III) photocatalytic oxidation fluctuates around 2.85 mgAsg-1 h-1. In the absence of visible light, the adsorption capacities for As(III) and As(V) are 3.7 and 42.7 mg/g, respectively, at pH = 3. Under visible light condition, the adsorption capacity of As(III) increases sharply to 15.6 mg/g, which provides the foundation for a new application of TiO2-x in the field of arsenic adsorption.

Runt‑related transcription factor 3 (RUNX3) is a candidate tumor suppressor, and its inactivation may play a crucial role in the carcinogenesis process of numerous cancer types, including esophageal squamous cell carcinoma (ESCC). We previously revealed that RUNX3 inactivation was correlated with lymph node metastasis (LNM) and ESCC recurrence. However, the exact mechanisms of this process are still under investigation. Background Gene therapy remains a significant challenge due to lots of barriers limiting the genetic manipulation technologies. As for non-viral delivery vectors, they often suffer insufficient performance due to inadequate cellular uptake and gene degradation in endosome or lysosome. The importance of overcoming these conserved intracellular barriers is increasing as the delivery of genetic cargo. Results A surface-functionalized non-viral vector involving the biomimetic mannitol moiety is initiated, which can control the cellular uptake and promote the caveolae-mediated pathway and intracellular trafficking, thus avoiding acidic and enzymatic lysosomal degradation of loaded gene internalized by clathrin-mediated pathway. Different degrees of mannitol moiety are anchored onto the surface of the nanoparticles to form bio-inspired non-viral vectors and CaP-MA-40 exhibits remarkably high stability, negligible toxicity, and significantly enhanced transgene expression both in vitro and in vivo. Conclusions This strategy highlights a paradigmatic approach to construct vectors that need precise intracellular delivery for innovative applications.

Background Morphine, a µ-opioid receptor (MOR) agonist, has been shown to be related to the activity of cancer cells, and a higher morphine dosage reduces the survival time of patients with lung cancer. However, the effect of morphine on the malignant behavior of lung cancer cells remains unclear. The aim of this study was to investigate the specific molecular mechanism by which morphine regulates the malignant biological behavior of non-small cell lung cancer. Methods Immunofluorescence staining and Western blot analyses were performed to detect MOR expression. H460 non-small cell lung cancer cells were used in this study, and cell proliferation, the cell cycle and apoptosis were evaluated using Cell Counting Kit-8 (CCK-8) and flow cytometry assays, respectively. Cell migration and invasion were detected using wound healing and Transwell assays. The effect of morphine on lung cancer development in vivo was examined by performing a xenograft tumor assay following morphine treatment. Results Morphine promoted the growth of H460 cells both in vivo and in vitro. Morphine enhanced cell migration and invasion, modified cell cycle progression through the S/G2 transition and exerted an antiapoptotic effect on H460 cells. Additionally, morphine increased Rous sarcoma oncogene cellular homolog (Src) phosphorylation and activated the phosphoinositide 3 kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) pathway. Treatment with the MOR antagonist methylnaltrexone (MNTX) and the Src inhibitor protein phosphatase 1 (PP1) reduced the phosphorylation induced by morphine. Furthermore, MNTX, PP1, and the PI3K/AKT inhibitor deguelin reversed the antiapoptotic effect of morphine on lung cancer cells. Conclusion Morphine promotes the malignant biological behavior of H460 cells by activating the MOR and Src/mTOR signaling pathways.

Background Cell invasion is a hallmark of metastatic cancer, leading to unfavorable clinical outcomes. In this study, we established two highly invasive lung cancer cell models (A549-i8 and H1299-i8) and identified mesoderm-specific transcript (MEST) as a novel invasive regulator of lung cancer. We aim to characterize its biological function and clinical significance in lung cancer metastasis. Methods Transwell invasion assay was performed to establish high-invasive lung cancer cell model. Immunohistochemistry (IHC) was used to detect MEST expression in tumor tissues. Mass spectrometry and bioinformatic analyses were used to identify MEST-regulated proteins and binding partners. Co-immunoprecipitation assay was performed to detect the interaction of MEST and VCP. The biological functions of MEST were investigated in vitro and in vivo. Immunofluorescence staining was conducted to explore the colocalization of MEST and VCP. Results MEST overexpression promoted metastasis of lung cancer cells in vivo and in vitro by activating NF-κB signaling. MEST increased the interaction between VCP and IκBα, which accelerated IκBα degradation and NF-κB activation. Such acceleration was abrogated by VCP silencing, indicating that MEST is an upstream activator of the VCP/IκBα/NF-κB signaling pathway. Furthermore, high expressions of MEST and VCP were associated with poor survival of lung cancer patients. Conclusion Collectively, these results demonstrate that MEST plays an important role in driving invasion and metastasis of lung cancer by interacting with VCP to coordinate the IκBα/NF-κB pathway. Targeting the MEST/VCP/IκBα/NF-κB signaling pathway may be a promising strategy to treat lung cancer.

The nonlinear refraction of silver nanoparticles (AgNPs) in n-hexane was studied by using the closed-aperture Z-scan technique with a 532 nm nanosecond laser. It was found that, the nonlinear refraction of AgNPs shows the coexistence and transformation from self-focusing to self-defocusing. Specifically, self-focusing occurs at low excitation intensity, self-defocusing occurs at high excitation intensity, and coexistence of self-focusing and self-defocusing occurs at relatively moderate excitation intensity. The experimental results were analysed and discussed in terms of third-order and fifth-order nonlinear refractive effect. Specifically, the self-focusing is caused by the positive third-order nonlinear refraction, the self-defocusing is induced by the negative fifth-order nonlinear refraction, and the transformation from the self-focusing to self-defocusing at medium excitation intensity is caused by the competition of third-order and fifth-order nonlinear refraction. Finally, the third-order refractive index and fifth-order refractive index were obtained.

Utilizing the in-situ reducing method, a reduced graphene oxide (rGO) paper for 13~14 um was obtained and exhibits great Terahertz (THz) absorbing capability. The absorption of rGO paper exceeds 43% in the whole frequency band from 0.3~1.2 THz and maximum absorption reaches 59.33% at 1.13 THz. This makes it possible to realize ultra-thin broadband THz absorbing materials.

The intercalation of methotrexate(MTX) into layered double hydroxides(LDHs) via an exfoliation-reassembly process was performed and the resulting hybrids were then characterized by X-ray diffractiometry(XRD), Fourier transform infrared(FTIR) spectroscopy, thermogravimetry(TG) and differential scanning calorimetry(DSC), etc. In the synthesis procedure, LDHs particles were firstly delaminated, and the reassembly process was performed to get MTX intercalated LDH(MTX/LDHs) hybrids. The reassembly conditions, such as reassembly time and different reassembly solvent, were studied emphatically. It was found that 20 min is the optimum time and formamide is the most suitable solvent in the reassembly process. At last, the controlled release property and the drug-loading capacity were examined emphatically to reveal that MTX/LDHs hybrids reassembled in formamide exhibit the highest drug-loading capacity. The cytotoxicity of the hybrids was estimated by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, thiazolyl blue tetrazolium bromide(MTT) assays on the human lung cancer cells(A549), with the results showing that MTX/LDHs hybrids have effective suppress role on the proliferation of cancer cells, and the hybrids obtained from the formamide solution present much better anti- cancer effect than the other samples.

Stimuli-responsive materials are promising paradigm applied to construct diagnostic and therapeutic intracellular controlled release vectors, while highlighting many challenges and opportunities. In this paper, six α-cyclodextrin-based supramolecular nanovectors were constructed and the efficacy of amine groups, stimuli-responsive profiles and endocytic mechanisms were investigated. The results indicated that the designed supermolecules can compact DNA to form stable complexes and display low cytotoxicity. Among them, PRPEI-2 with suitable PEI amine group exhibited enhanced transfecting performance, high dilution stability, nice serum compatibility, and good acid-responsive profiles to enable endosome escape, significantly higher than commercially available transfecting agent PEI25000, the most effective vector studied to date. The endocytic uptake mechanisms involved in the transfection was mainly through clathrin-mediated pathway, which is closely associated with and can be improved by endosome escape. Moreover, PRPEI-2/DNA polyplex can be effectively expressed in vivo even after 48 h via only single tail-vein injection, and the gene expression and main tissue distribution appeared in the testis, liver, brain and spleen. These excellent characteristics demonstrated that the supramolecular PRPEI-2 represents an excellent prospect as stimuli-responsive nanovectors for gene diagnosis and therapy.

Background:Acupuncture stimulation at GV26 during the acute phase of cerebral ischaemia can effectively reduce brain damage induced by ischaemic injury. However, the time course of the effects of acupuncture stimulation has not yet been thoroughly studied.Objective:To investigate the effects of manual acupuncture (MA) on glutamic acid (Glu) and γ-aminobutyric acid (GABA) expression in the cerebrospinal fluid of rats with middle cerebral artery occlusion (MCAO) and determine whether there is a temporal effect of acupuncture on the treatment of cerebral ischaemia.Methods:We performed thread occlusion of the right middle cerebral artery in rats to establish an animal model of MCAO. Simultaneously, during acupuncture treatment, microdialysis was used to continuously and dynamically observe immediate alterations in amino acid metabolism with acupuncture stimulation after cerebral ischaemia in vivo in this rat model of MCAO.Results:We found that, in comparison with an untreated MCAO group, Glu content was significantly decreased during the first acupuncture stimulation and during the course of the acupuncture treatment in the MCAO+MA group (MCAO vs MCAO+MA: day 1, P=0.032; day 2, P=0.021; day 3, P=0.017). These findings were also seen after the end of treatment when acupuncture was no longer applied (MCAO vs MCAO+MA: day 7, P=0.009). Measurements of GABA content following cerebral ischaemic injury showed that GABA peaks 24 hours after damage, falls thereafter and decreases to baseline levels on day 7. In the MCAO+MA group, GABA content on days 1 to day 2 was lower than in the MCAO group (MCAO+MA vs MCAO: day 1, P=0.003; day 2, P=0.001), although it was higher than in the control group (MCAO+MA vs control: day 1, P=0.024; day 2, P=0.009). GABA content on day 3 and day 7 was higher in the MCAO+MA group than in the MCAO group and the control group (MCAO+MA vs MCAO: day 3, P=0.008; day 7, P=0.013; MCAO+MA vs control: day 3, P=0.002; day 7, P=0.009).Conclusion:Acupuncture stimulation at GV26 can effectively decrease excessive release of Glu induced by ischaemia and maintain the endogenous inhibitory activity of GABA. This phenomenon was seen during the entire course of acupuncture treatment and continued for some time after the end of acupuncture treatment.

Polychlorinated biphenyls (PCBs) are a group of persistent organic pollutants (POPs) widely existing in the environment. Arthrobacter sp. YC-RL1 is a biphenyl-degrading bacterium that shows metabolic versatility towards aromatic compounds. A 2-hydroxy-6-oxo-6-phenylhexa-2, 4-dienoate (HOPDA) hydrolase (BphD) gene involved in the biodegradation of biphenyl was cloned from strain YC-RL1 and heterologously expressed in Escherichia coli BL21 (DE3). The recombinant BphDYC-RL1 was purified and characterized. BphDYC-RL1 showed the highest activity at 45 °C and pH 7. It was stable under a wide range of temperature (20–50 °C). The enzyme had a Km value of 0.14 mM, Kcat of 11.61 s−1, and Vmax of 0.027 U/mg. Temperature dependence catalysis exhibited a biphasic Arrhenius Plot with a transition at 20 °C. BphDYC-RL1 was inactivated by SDS, Tween 20, Tween 80, Trition X-100, DTT, CHAPS, NBS, PMSF, and DEPC, but insensitive to EDTA. Site-directed mutagenesis of the active-site residues revealed that the catalytic triad residues (Ser115, His275, and Asp247) of BphDYC-RL1 were necessary for its activity. The investigation of BphDYC-RL1 not only provides new potential enzyme resource for the biodegradation of biphenyl but also helps deepen our understanding on the catalytic process and mechanism.

Polyaniline/poly(methylmethacrylate) (PANI/PMMA) microfiber coating were successfully deposited on carbon steel by electro-spinning and drop-cast methods, and their corrosion protection performances were evaluated in H2SO4. The results show the corrosion protection property of coating is outstanding with a certain amount of PANI (25 wt.%), especially for electro-spun coating. The reason of superior corrosion protection function of PANI electro-spun coating was investigated, which is primarily attributed to its better barrier protection, behaved as a lower porosity and enhanced adhesive ability. The outstanding corrosion protection performance of PANI/PMMA microfiber coatings encourages the wider application of electro-spun coatings in the corrosion protection area.

The uniaxial compression stress dependence of the phase transitions and dielectric properties of a potassium niobate (KNbO3) ferroelectric crystal were investigated using Landau–Ginsburg–Devonshire phenomenological theory. The results showed that (1) uniaxial stress along [001]c could induce a spontaneous polarization change from the [001]c direction to the [010]c or [100]c directions and from the [011]c, [101]c directions to the [110]c direction; (2) the cubic to tetragonal and the tetragonal to orthorhombic phase transition temperatures increased, while the orthorhombic to rhombohedral phase transition temperature decreased as the uniaxial stress was increased; and (3) spontaneous polarization increased as the uniaxial stress was increased for the tetragonal and orthorhombic phases. The uniaxial stress dependence of the dielectric susceptibility also showed that a flatter Gibbs free energy profile corresponds with a greater expandability, contractibility, and rotationality of the polarization (which indicates a higher dielectric susceptibility), while a steeper Gibbs free energy profile corresponds with a reduced expandability, contractibility, and rotation of the polarization (which indicates a lower dielectric susceptibility).

The degradation of nitrobenzene and its intermediate aniline from wastewater by constructed wetlands coupled with the micro-electric field (CW-MEF) technology was studied. The results showed that the CW-MEF system had good degradation. With the increase of influent concentration of nitrobenzene, the removal rate of the anode was excellent which remained above 86%, but the degradation of CW-MEF for COD decreased. In different stages, the power generation capacity was different. In the second stage, the power generation voltage reached 430 V and the average power density was 85.07 MW m-3, while the maximum reached 87.47 MW m-3. Through high-throughput sequencing analysis, the A1 sludge layer contained 36% of thick-walled bacteria and 20% of bacteroides, the A2 contained about 20% of campylobacter green, and the A3 contained 10% of green campylobacter, pachyphyte and bacteroides.

Bisphenol A (BPA) is a widespread pollutant threatening the ecosystem and human health. An effective BPA degrader YC-JY1 was isolated and identified as Sphingobium sp. The optimal temperature and pH for the degradation of BPA by strain YC-JY1 were 30 &deg, C and 6.5, respectively. The biodegradation pathway was proposed based on the identification of the metabolites. The addition of cytochrome P450 (CYP) inhibitor 1-aminobenzotriazole significantly decreased the degradation of BPA by Sphingobium sp. YC-JY1. Escherichia coli BL21 (DE3) cells harboring pET28a-bisdAB achieved the ability to degrade BPA. The bisdB gene knockout strain YC-JY1&Delta, bisdB was unable to degrade BPA indicating that P450bisdB was an essential initiator of BPA metabolism in strain YC-JY1. For BPA polluted soil remediation, strain YC-JY1 considerably stimulated biodegradation of BPA associated with the soil microbial community. These results point out that strain YC-JY1 is a promising microbe for BPA removal and possesses great application potential.

Polyethylenimine (PEI), a kind of cationic non-viral gene delivery vector, is capable of stable and efficient transgene expression for gene delivery. However, low transfection efficiency in vivo along with high toxicity limited the further application of gene therapy in the clinic. To enhance gene transfection performance and reduce cytotoxicity of polyethylenimine, branched polyethylenimine-derived cationic polymers BPEI25 k-man-S/L/M/H with different grafting degree with mannitol moieties were prepared and the transfection efficiency was evaluated. Among them, BPEI25 k-man-L showed the best transfection efficiency, lower toxicity, and significantly enhanced long-term systemic transgene expression for 96 h in vivo even at a single-dose administration. The results of cellular uptake mechanism and western-blot experiments revealed that the mannitol modification of BPEI25 k induced and up-regulated the phosphorylation of caveolin-1 and thus enhanced the caveolae-mediated cellular uptake. This class of gene delivery system highlights a paradigmatic approach for the development of novel and safe non-viral vectors for gene therapy.

The abscisic acid (ABA), as a pivotal plant hormone, plays a key role in controlling the life cycle and adapting to the environmental stresses. The receptors of ABA are the Pyrabactin Resistance /Pyrabactin Resistance -Like/Regulatory Component of ABA Receptors (PYR/PYL/RCAR, PYLs for simplicity), which regulate the Protein Phosphatase 2Cs (PP2Cs) in the signal pathway. As an important ABA mimicking ligand, Pyrabactin shows the activation function to parts of members of PYLs, such as PYR1 and PYL1. Due to the antagonism of Pyrabactin to PYL2, it was used as a probe to discover a part of ABA receptors. Since then, many researchers have been trying to find out the determinants of the selective regulation of PYLs and PP2Cs interaction. However, the roles of residues on the selective regulation of PYR1/PYL2 and PP2Cs interaction induced by Pyrabactin are still ambiguous. This research investigated the selective activation mechanism of Pyrabactin through the sequence alignment, molecular docking, molecular dynamics simulation, and binding free energy calculation. Furthermore, the electrostatic and hydrophobic interaction differences induced by Pyrabactin and agonists were compared. The results indicate that Leu137/Val114, Ser85/Ser89, and Gly86/Gly90 from the pocket and gate of PYR1/PYL2 are the vital residues for the selective activation of Pyrabactin. Meanwhile, the electrostatic interaction between PP2Cs and PYLs complexed with agonists was improved. This mechanism provides strong support for the design of selective agonists and antagonists.

Aim. Our study is aimed at investigating whether Lipopolysaccharide- (LPS-) treated podocytes could polarize naive CD4+ T cells into different subsets in vitro. Materials and Methods. Podocytes and mouse bone marrow-derived dendritic cells (BMDCs) were first cultured with 25 μg/ml LPS for 6 hours, respectively. Then, naive CD4+ T cells were cocultured with the LPS-treated podocytes or BMDCs at a ratio of 1 : 1 or 1 : 1 : 1. After 48 hours, we collected the suspended cells and supernatant from all groups to measure T helper (Th)17 cells, regulatory T (Treg) cells, and cytokine concentration. Results. We observed the expression of CD80 and major histocompatibility complex class II molecule (MHC II) in podocytes but did not found the upregulation of them after treating podocytes with LPS. LPS-treated podocytes could induce naive CD4+ T cells to Th17 cells and Treg cells with a higher ratio of Th17/Treg than BMDCs. Possible interaction between podocytes and BMDCs may exist in the induction process of Th17 cells and Treg cells. Conclusion. Our study proved that CD80 and MHC II were constitutively expressed in podocytes but not upregulated by LPS. LPS-treated podocytes could polarize naive CD4+ T cells into Th17 and Treg cells and affect the Th17/Treg balance and may incline to cause a Th17 response.

The amoxicillin-UCCs-2/TPP nanoparticles constructed with ureido-modified chitosan derivative UCCs-2 and sodium tripolyphosphate (TPP) played an important role to deliver drug to achieve more efficacious and specific eradication of Helicobacterpylori (H. pylori) in vitro. In this study, the anti-H. pylori effectiveness in vivo and uptake mechanism was investigated in details, including the effect of temperature, pH values and the addition of competitive substrate urea on uptake. Compared with unmodified nanoparticles, a more efficacious and specific anti-H. pylori activities were obtained in vivo by using this biological chitosan derivative UCCs-2. Histological staining and immunological analysis verified that the amoxicillin-UCCs-2/TPP nanoparticles could diminish the proinflammatory cytokines levels and alleviate the inflammatory damages caused by H. pylori infection. The uredio-modified nanoparticles also have favorable gastric retention property, which is beneficial for the oral drug delivery to targeted eradicate H. pylori infection in stomach. These findings suggest that this targeted drug delivery system may serve for specific treatment of H. pylori infection both in vitro and in vivo, which can also be used as promising nanocarriers for other therapeutic reagents to target H. pylori.

An Al-Mg-Si alloy 6061 and an Al-Zn-Mg alloy 7A52 were joined by friction stir welding successfully. Pre- and post- heat treatment were employed to improve the strength of the weld. The results show a best weld joint with the lowest hardness of 100 HV in 6061 matrix, being achieved by post-solid-solution and subsequent two-stage artificial aging for the whole weld joint of the 7A52 and 6061 solid solution. Under this condition, the weld nugget zone (WNZ) is stronger than 6061 matrix but it has lower hardness than 7A52 matrix. The hardness of WNZ is contributed by the combination of &eta, &prime, and L precipitates, dynamically changes along with the ratios between the number of &eta, and L precipitates. The higher the number density of &eta, precipitates, the hardness of WNZ is closer to that of the 7A52 matrix. Otherwise, the higher number density of L precipitates, the hardness of WNZ is closer to that of 6061 matrix. The coexistence of &eta, and L precipitates is a direct result from the mixture of 7A52 and 6061 alloys achieved by stirring. Precipitates identification and composition analysis reveal a dynamic WNZ with constituent transition in hardness and composition.

The acidic environment of the stomach is a threat to the curative effect of antimicrobial drugs for the eradication of Helicobacter pylori (H. pylori) in the infected area. The conventional clinical formulations of antibiotics have low specificity to H. pylori, which disrupts the normal balance of intestinal microbiomes. Therefore, oral drug delivery system with better stability at low pH as well as higher specificity to target H. pylori would provide more effective strategy to eradicate H. pylori and reduce the side effect of antibiotics. Based on the construction of UreI-mediated targeted drug delivery system developed by our group, in this work, using urea-modified UCCs-2 as targeting moiety to the UreI channel protein which is specifically expressed on H. pylori, pH-sensitive amoxicillin-loaded AMX-PLGA/UCCs-2 nanoparticles produced by UCCs-2 and PLGA for targeted treatment of H. pylori infection were established. The nanoparticles were prepared by double emulsion-solvent evaporation method. To achieve a promising drug delivery system with favorable pH-sensitive properties, we adopted an orthogonal design to obtain the optimal formulation. The results showed that the optimized AMX-PLGA/UCCs-2 nanoparticles were in a favorable pH sensitive manner and exhibited low cytotoxicity, higher specificity and better anti-H. pylori efficiency than amoxicillin and non-targeting AMX-PLGA/Cs nanoparticle both in vitro and in vivo, which can protect the antimicrobial drugs against acidic environment and deliver them to targeted eradicate H. pylori in the infected location. The cellular uptake mechanism showed that AMX-PLGA/UCCs-2 nanoparticles are an effective UreI-mediated targeted drug delivery system for anti-H. pylori treatment, which can also be used as promising nanocarriers for oral delivery of other therapeutic drugs to targeted treat H. pylori.

Cathodic polarization antifouling deserves attention because of its environmentally friendly nature and good sustainability. It has been proven that cathodic voltages applied on metal substrates exhibit outstanding antifouling effects. However, most metals immersed in marine environment are protected by insulated anticorrosive coatings, restricting the cathodic polarization applied on metals. This study developed a conducting polypyrrole (PPy)/acrylic resin coating (σ = 0.18 Scm−1), which can be applied in cathodic polarization antifouling. The good stability and electro-activity of PPy in the negative polarity zone in alkalescent NaCl solution were verified by linear sweep voltammetry (LSV), chronoamperometry (CA), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS), demonstrating the feasibility of PPy as cathodic polarization material. Furthermore, the antifouling effects of PPy/acrylicresin coating on 24-h old Escherichia coli bacteria (E. coli) which formed on PPy/acrylic resin-coated plastic plate were measured under different cathodic potentials and treatment time, characterized by fluorescent microscope. The results suggest that at cathodic potential around −0.5 V (vs. saturated calomel electrode (SCE)), there was little trace of attached bacteria on the substrate after 20 min of treatment. PPy/acrylicresin-coated substrates were also subjected to repeated cycles of biofilm formation and electrochemical removal, where high removal efficiencies were maintained throughout the total polarization process. Under these conditions, the generation of hydrogen peroxide is believed to be responsible for the antifouling effects because of causing oxidative damage to cells, suggesting the potential of the proposed technology for application on insulated surfaces in various industrial settings.

A flash synthesis method of titanium suboxides (TSO) TinO2n-1 nanoparticles with Magneli phases by thermal plasma using rutile-phase metatitanic acid (H2TiO3) as the raw material was proposed and investigated, comparing with common sintering hydrogenation process of H2TiO3. TSO powders with diverse colors were prepared and characterized by means of XRD, SEM and TEM. The results showed Magneli phase nanoparticles with perfect spherical shape and particle size of 20–100 nm were rapidly synthesized by thermal plasma treatment under Ar and Ar/H2 atmospheres. Typical Magneli phases of Ti5O9 and Ti4O7 with peacock blue color were formed under argon atmosphere. An increase of hydrogen content in the atmosphere made nano-H2TiO3 efficiently reduced to form a different TSO phases with diverse colors of dark blue to blue black. Notably, parts of nanoparticles were of carbon shell/Magneli phase core structure. The UV–visible absorptivity and electric conductivity of samples were tested and the results indicate that thermal plasma is an efficient way to improve the optical and conductive properties of materials. But the electric resistivity of nano Magneli phase powders still keep 105–106 Ω.cm.

Streptochlorin, a small indole alkaloid isolated from marine Streptomyces sp., exhibits a wide range of potent biological activities. An efficient and economic synthetic protocol for streptochlorin has been developed and validated, 4 steps from indole in a total yield of 45%, and further applied for the synthesis of its analogues. Biological testing showed that most of the target compounds exhibited potential antifungal activity in the primary assays, especially compounds 6, 7 and 9c were the most active ones, representing effective activity against the phytopathogenic fungi screened in preliminary test and might be explored for the study of mode of action in the future.

Background: Biodegradation is a reliable approach for efficiently eliminating persistent pollutants such as chlorpyrifos. Despite many bacteria or fungi isolated from contaminated environment and capable of degrading chlorpyrifos, limited enzymes responsible for its degradation have been identified, let alone the catalytic mechanism of the enzymes. Results: In present study, the gene cpd encoding a chlorpyrifos hydrolase was cloned by analysis of genomic sequence of Paracoccus sp. TRP. Phylogenetic analysis and BLAST indicated that CPD was a novel member of organophosphate hydrolases. The purified CPD enzyme, with conserved catalytic triad (Ser155-Asp251-His281) and motif Gly-Asp-Ser-Ala-Gly, was significantly inhibited by PMSF, a serine modifier. Molecular docking between CPD and chlorpyrifos showed that Ser155 was adjacent to chlorpyrifos, which indicated that Ser155 may be the active amino acid involved in chlorpyrifos degradation. This speculation was confirmed by site-directed mutagenesis of Ser155Ala accounting for the decreased activity of CPD towards chlorpyrifos. According to the key role of Ser155 in chlorpyrifos degradation and molecular docking conformation, the nucleophilic catalytic mechanism for chlorpyrifos degradation by CPD was proposed. Conclusion: The novel enzyme CPD was capable of hydrolyze chlorpyrifos and Ser155 played key role during degradation of chlorpyrifos.

A bifunctional tunable metamaterial composed of pattern metal structure, graphene, and strontium titanate (STO) film is proposed and studied numerically and theoretically. The dual plasmon-induced transparency (PIT) window is obtained by coupling the bright state cut wire (CW) and two pairs of dark state dual symmetric semiring resonators (DSSRs) with different parameters. Correspondingly, slow light effect can also be realized. When shifting independently, the Fermi level of the graphene strips, the amplitudes of the two PIT transparency windows and slow light effect can be tuned, respectively. In addition, when independently tuning the temperature of the metamaterial, the frequency of the dual PIT windows and slow light effect can be tuned. The physical mechanism of the dual-PIT was analyzed theoretically by using a three-harmonic oscillator model. The results show that the regulation function of the PIT peak results from the change of the oscillation damping at the dark state DSSRs by tuning conductivity of graphene. Our design presents a new structure to realize the bifunctional optical switch and slow light.

The remediation of heavy metal contaminated wastewater using microalgae has attracted more and more attention. In this study, the removal of copper (II) from piggery digestate by Desmodesmus sp. CHX1 was investigated. Results showed that the optimal conditions for copper (II) removal by Desmodesmus sp. CHX1 were pH of 6.00, microalgae inoculation concentration of 0.41 g/L and treatment time of 4 d, with the copper removal efficiency of 88.35%. The biosorption process of copper (II) by Desmodesmus sp. CHX1 under optimal conditions well followed the pseudo second order model, with the rate constant and the theoretical and experimental biosorption capacity of 0.15 mg/(L·h) and 0.85 mg/L, respectively. The isotherms adsorption of copper was described well by the Langmuir model, biosorption maybe dominated the adsorption process. The affinity of Cu2+ by functional groups in the microalgal cell walls was the main mechanism for copper removal by Desmodesmus sp. CHX1.

Transparent and flexible chitosan-based membranes containing antibacterial drugs were prepared through a casting/solvent evaporation method from suspension of chitosan floccule. To enhance mechanical properties, glycerin was incorporated into the floccule suspension to conjugate chitosan as a plasticizer. The mechanism of membrane formation is attributed to inter- and intro-hydrogen bonding between chitosan and glycerol molecules. The results showed that incorporation of glycerol has a significant influence on the properties of the chitosan membranes. With the increase of glycerol content, the tensile strength, swelling rate, water vapor permeability and wettability of membranes were significantly improved. In vitro enzymatic degradation revealed that the chitosan membrane had long-term stability regardless of the glycerol content. To enhance antibacterial properties, tetracycline hydrochloride (TH) and silver sulfadiazine (AgSD), representing the water-soluble and water-insoluble drug models, were integrated into the membranes, respectively. The results of controlled-release efficacy and inhibition zone indicate that the glycerol toughened chitosan membranes containing drugs have a promising future in treatment of bacterial infection as wound dressing.

In a polymer electrolyte membrane electrolyzer cell (PEMEC), the two electrode compartments are separated by a polymer membrane. Liquid water is fed to the anode side, forming oxygen gas on the anode, and hydrogen gas on the cathode side, respectively. The respective designs of the flow field patterns are important to obtain a uniform distribution of flow, in combination with low-pressure drops, during operation. In this study, oxygen production in a manifold electrolyzer is investigated using the Computational Fluid Dynamics (CFD) method. The flow is considered unsteady, three-dimensional, and two-phase. A mixture model is applied to simulate the water consumption and oxygen production. The anode side is analyzed and after the reaction, gas bubbles are produced inside the lower surface of the electrode. The simulation is performed for 5 s. The results show that from the moment of start to 2 s is an important time for the formation of pressure and velocity balance inside the manifolds. In 0.75 s, the oxygen reaches its highest concentration and, due to its physical nature, begins to move from low to high levels in the manifold. Increasing the rate of oxygen production and reducing the pressure drop in the system can be controlled by the number of different channels.

Three novel regular acceptor-donor1-acceptor-donor2(A-D1-A-D2) terpolymers were prepared via embedding a second donor(D2) unit into the traditional D-A backbone to manipulate the energy levels and molecular geometry with no complex synthesis or solubility loss. In these A-D1-A-D2 terpolymers, benzodithiophene(BDT, D1) and diketopyrrolopyrrole(DPP, A) were selected as the basic skeleton, and the dithienopyrrole(DTPy), carbazole(CZ) and fluorine(FL) units with different electron donating ability were chosen as the second donor unit(D2). The HOMO energy levels can be effectively modulated by only varying D2 unit because of the push-pull interaction between donor and acceptor units. Versus the D-A bipolymer PDPP-BDT, incorporation of the D2 unit into the copolymers can distinctly lower the highest occupied molecular orbital(HOMO) levels to –5.47 eV for PDDPP-BDT-DTPy, –5.38 eV for PDDPP-BDT-CZ and –5.23 eV for PDDPP-BDT-FL, which shows the strong dependence on electron-donating ability. Density functional theory(DFT) simulation and X-ray diffraction(XRD) measurements also reveal the effect of the D2 units on the molecular geometry of the terpolymers and their molecular packing. Notably, a PDDPP-BDT-DTPy combined with a thiophene ring and forked tail pendant away from the backbone had less back-bone torsion and more compact packing than the other two counterparts. These results demonstrate that embedding a second donor(D2) unit into the backbone to construct an A-D1-A-D2 structure can be an effective and direct strategy to manipulate the energy levels and molecular geometry and develop organic semiconducting materials.
