To address the challenges posed by the vast and complex knowledge information in cultural heritage design, such as low knowledge retrieval efficiency and limited visualization, this study proposes a method for knowledge extraction and knowledge graph construction based on graph attention neural networks (GAT). Using Tang Dynasty gold and silver artifacts as samples, we establish a joint knowledge extraction model based on GAT. The model employs the BERT pretraining model to encode collected textual knowledge data, conducts sentence dependency analysis, and utilizes GAT to allocate weights among entities, thereby enhancing the identification of target entities and their relationships. Comparative experiments on public datasets demonstrate that this model significantly outperforms baseline models in extraction effectiveness. Finally, the proposed method is applied to the construction of a knowledge graph for Tang Dynasty gold and silver artifacts. Taking the Gilded Musician Pattern Silver Cup as an example, this method provides designers with a visualized and interconnected knowledge collection structure.

A scientific method for evaluating the design of interfaces is proposed to address the unique characteristics and user needs of infrequent-contact public service interfaces. This method is significant for enhancing service efficiency and promoting the sustainable development of public services. Current interface evaluation methods are limited in scope and often fail to meet actual user needs. To address this, this study focuses on virtual museums, examining users’ aesthetic psychology and cognitive behavior in terms of layout aesthetics and visual cognitive characteristics, aiming to explore the relationship between the two. Interface layout aesthetic values and user visual cognitive measurements were obtained by using computational aesthetics methods and eye-tracking experiments. These served as input data for a new model. An improved gray H-convex correlation model utilizing the ICRITIC method is proposed to examine the mapping relationship between interface layout aesthetics and visual cognitive features. The results demonstrate that our new model achieves over 90% accuracy, outperforming existing models. For virtual museum interfaces, symmetry and dominance significantly influence user visual cognition, with the most notable correlations found between density and gaze shift frequency, simplicity and mean pupil diameter, and order and gaze shift frequency. Additionally, fixation duration, fixation count, and mean pupil diameter were inversely correlated with interface layout aesthetics, whereas gaze shift frequency and gaze time percentage were positively correlated.

Surface cracks and joint deteriorations are typical premature failures of urban cement concrete pavement. However, traffic loads on the urban pavement are much lower than those on highways. Limited research has been conducted to investigate the causes of accelerated damage in urban cement concrete roads. To investigate the foundation issues that may cause the accelerated damage of urban cement concrete pavements, in this study, field evaluations were conducted to assess pavement foundation support and drainage conditions. Field visual inspections, Ground Penetrating Radar (GPR) survey, Dynamic Cone Penetrometer (DCP) test, and the Core-Hole Permeameter (CHP) test were performed. In urban residential areas with inadequate subgrade bearing capacity, cement concrete pavements are prone to early damage. Foundations with a higher content of coarse particles exhibit a higher CBR value, which can extend the service life of the pavement. The compaction of foundation materials near sewer pipelines and manholes is insufficient, leading to non-uniform support conditions. Moreover, the permeability of the foundation material can influence the service life of pavement surface structures. Foundation materials with fewer fine particles enhance drainage performance, contributing to a longer service life for PCC pavements. In areas with inadequate drainage, water accumulation reduces the bearing capacity of the foundation, thereby accelerating pavement deterioration. The poor bearing capacity and drainage conditions of the foundation lead to cavities between the surface layer and foundation material thus yielding stress concentrations on the pavement surface, which cause the formation of pavement surface cracks.

There is an urgent need to find new solutions for the global dilemma of increasing antibiotic resistance in humans and animals. Modifying the performance of existing antibiotics using the nanocarrier drug delivery system (DDS) is a good option considering economic costs, labor costs, and time investment compared to the development of new antibiotics. Numerous studies on nanomedicine carriers that can be used for humans are available in the literature, but relatively few studies have been reported specifically for veterinary pharmaceutical products. Polymer-based nano-DDS are becoming a research hotspot in the pharmaceutical industry owing to their advantages, such as stability and modifiability. This review presents current research progress on polymer-based nanodelivery systems for veterinary antimicrobial drugs, focusing on the role of polymeric materials in enhancing drug performance. The use of polymer-based nanoformulations improves treatment compliance in livestock and companion animals, thereby reducing the workload of managers. Although promising advances have been made, many obstacles remain to be addressed before nanoformulations can be used in a clinical setting. Some crucial issues currently facing this field, including toxicity, quality control, and mass production, are discussed in this review. With the continuous optimization of nanotechnology, polymer-based DDS has shown its potential in reducing antibiotic resistance to veterinary medicines.

Biomaterials with exceptional biocompatibility and bioactivity are now pushing the boundaries of bone tissue engineering. In this study, natural Arabic gum biopolymer incorporating titanium dioxide nanoparticles (NAG + TiO2NP) nanocomposite film was fabricated. The FTIR and XRD analysis show the presence of functional groups assigned to NAG biopolymers and highly crystalline anatase TiO2NP. Well dispersed TiO2NP can be seen from SEM micrograph suggesting good interaction between TiO2NP filler and NAG biopolymer matrix to enhance the mechanical characteristics of nanocomposite film. The NAG + TiO2NP nanocomposite film exhibited strong bioactivity to form bone-like apatite and promoted the proliferation of MG-63 cells attributed to their excellent biocompatibility and non-toxicity. The NAG + TiO2NP nanocomposite film also displays high antibacterial activity with (36.33 ± 1.53) mm and (27.00 ± 2.00) mm inhibition zone were recorded against Staphylococcus aureus and Escherichia coli. The findings indicate that the NAG + TiO2NP nanocomposite film, with its improved mechanical properties, high swelling capacity, biodegradability, and non-toxicity, shows promise as a viable option for bone tissue regeneration materials.

Enterococcus faecium (E. faecium) is widely used in foods and is known as a probiotic to treat or prevent diarrhea in pets and livestock. However, the poor resistance of E. faecium to high temperature processing procedures limits its use. Strain domestication is a low-cost and effective method to obtain high-temperature-resistant strains. In this study, heat treatment was performed from 45 °C to 70 °C and the temperature was gradually increased by 5 °C every 3 days. After domestication, the survival rates of the high temperature adaptation strain RS047-wl under 65 °C water bath for 40 min was 11.5 times higher than WT RS047. Moreover, the saturated fatty acid (SFA) contents in cell membrane and the cell volume significantly increased in the RS047-wl. The combined transcriptomic, metabolomic, and proteomics analysis results showed a significant enhancement of cell wall and membrane synthesis ability in the RS047-wl. In conclusion, one of the main factors contributing to the improved high temperature resistance of RS047-wl was its enhanced ability to synthesize cell wall and membrane, which helped maintain normal cell morphology. Developing a high-temperature-resistant strain and understanding its mechanism enables it to adapt to high temperatures. This lays the groundwork for its future development and application.

Without affecting cell viability, epigallocatechin gallate (EGCG), gallocatechin gallate (GCG), theaflavine-3,3′-digallate (TFDG), or theasinensin A (TSA) have been found to effectively reduce intracellular melanin content and tyrosinase (TYR) activity. However, studies on the anti-melanogenic mechanism of the above samples remain weak, and the activities of these samples in regulating melanogenesis at the molecular level lack comparison. Using B16F10 cells with the α-melanocyte-stimulating hormone (α-MSH) stimulation and without the α-MSH stimulation as models, the effects of EGCG, GCG, TFDG, or TSA on cell phenotypes and expression of key targets related to melanogenesis were studied. The results showed that α-MSH always promoted melanogenesis with or without adding the four samples. Meanwhile, the anti-melanogenic activities of the four samples were not affected by whether the α-MSH was added in the medium or not and the added time of the α-MSH. On this basis, the 100 µg/mL EGCG, GCG, TFDG, or TSA did not affect the TYR catalytic activity but inhibited melanin formation partly through downregulating the melanocortin 1 receptor (MC1R), microphthalmia-associated transcription factor (MITF), and the TYR family. The downregulation abilities of catechins on the TYR family and MITF expression were stronger than those of dimers at both the transcription and translation levels, while the ability of dimers to downregulate the MC1R expression was stronger than that of catechins at both the transcription and translation levels to some extent. The results of molecular docking showed that these four samples could stably bind to MC1R protein. Taken together, this study offered molecular mechanisms for the anti-melanogenic activity of the EGCG, GCG, TFDG, and TSA, as potential effective components against the UV-induced tanning reactions, and a key target (MC1R) was identified.

The evolution and mechanisms of tectonic subsidence in the Xisha area are poorly investigated, especially the spatiotemporal distribution features and reasons for the variations in tectonic subsidence. In this study, multi-channel seismic data and stratigraphic and lithologic features of wells are used to examine tectonic subsidence in the Xisha area from the Paleogene to Quaternary. The largest tectonic subsidence in the Xisha area is located in the Changchang Depression, with a maximum subsidence of 5.4 km, while the smallest tectonic subsidence is located on the Guangle Uplift and Xisha Uplift, which are close to 1.0 km and 1.5 km, respectively. Two rapid tectonic subsidence phases were mainly in the Oligocene, and from Middle to Late Miocene, with maximum subsidence rates of 0.45 m/ky and 0.32 m/ky, respectively. Five phases for the tectonic subsidence are proposed since the Paleogene based on our data. (1) The slow subsidence phase during the Eocene (53.5–32 Ma) was due to the transchronicity of the basement in the pro-rifted stage. (2) The rapid subsidence phase was common in the south and north margins of Qiongdongnan Basin, because of the faults triggered by the inherited stretched and thinned of crust in the Oligocene from 32 to 23.3 Ma. (3) The interim phase followed the rapid subsidence phase was in the Early Miocene (23.3–15.5 Ma) and marked the end of the rifted stage. (4) The accelerated rise phase started from the Middle Miocene (15.5 Ma) to the Late Miocene (5.5 Ma), and the reversal of the Red River Fault Zone may be tied to the acceleration of the tectonic subsidence. (5) The transitional phase started in the Pliocene (5.5 Ma) and lasts to the present. As the Red River Fault Zone changed from sinistral to dextral movement, the stress field of the study area has changed. Our results are helpful to better understand the spatiotemporal coupling relationship between tectonic subsidence and regional geological evolution in the Xisha area, South China Sea.

Salmonella enterica serovar typhimurium (S. Typhimurium) is a common Gram-negative foodborne pathogenic bacterium that causes gastrointestinal disease in humans and animals. It is well known that adhesins and invasins play crucial roles in the infection mechanism of S. Typhimurium. S. Typhimurium STM0306 has been denoted as a putative protein and its functions have rarely been reported. In this study, we constructed the STM0306 gene mutant strain of S. Typhimurium and purified the recombinant STM0306 from Escherichia coli. Deletion of the STM0306 gene resulted in reduced adhesion and invasion of S. Typhimurium to IPEC-J2, Caco-2, and RAW264.7 cells. In addition, STM0306 could bind to intestinal epithelial cells and induced F-actin modulation in IPEC-J2 cells. Furthermore, we found that STM0306 activated the nuclear factor kappa B (NF-κB) signaling pathway and increased the mRNA expression of pro-inflammatory cytokines such as IL-1β, TNF-α, as well as chemokine CXCL2, thus resulting in cellular inflammation in host cells. In vivo, the deletion of the STM0306 gene led to reduced pathogenicity of S. Typhimurium, as evidenced by lower fecal bacterial counts and reduced body weight loss in S. Typhimurium infected mice. In conclusion, the STM0306 of S. Typhimurium is an important adhesin/invasin involved in the pathogenic process and cellular inflammation of the host.

The transformation efficiency (TE) was improved by a series of special chemical and physical methods using immature embryos from the cultivar Fielder, with the PureWheat technique. To analyze the reaction of immature embryos infected, which seemed to provide the necessary by Agrobacterium tumefaciens in PureWheat, a combination of scanning electron microscopy (SEM), complete transcriptome analysis, and metabolome analysis was conducted to understand the progress. The results of the SEM analysis revealed that Agrobacterium tumefaciens were deposited under the damaged cortex of immature embryos as a result of pretreatment and contacted the receptor cells to improve the TE. Transcriptome analysis indicated that the differentially expressed genes were mainly enriched in phenylpropanoid biosynthesis, starch and sucrose metabolism, plant–pathogen interaction, plant hormone signal transduction, and the MAPK (Mitogen-activated protein kinase) signaling pathway. By analyzing the correlation between differentially expressed genes and metabolites, the expression of many genes and the accumulation of metabolites were changed in glucose metabolism and the TCA cycle (Citrate cycle), as well as the amino acid metabolism; this suggests that the infection of wheat embryos with Agrobacterium is an energy-demanding process. The shikimate pathway may act as a hub between glucose metabolism and phenylpropanoid metabolism during Agrobacterium infection. The downregulation of the F5H gene and upregulation of the CCR gene led to the accumulation of lignin precursors through phenylpropanoid metabolism. In addition, several metabolic pathways and oxidases were found to be involved in the infection treatment, including melatonin biosynthesis, benzoxazinoid biosynthesis, betaine biosynthesis, superoxide dismutase, and peroxidase, suggesting that wheat embryos may be under the stress of Agrobacterium and, thus, undergo an oxidative stress response. These findings explore the physiological and molecular changes of immature embryos during the co-culture stage of the PureWheat technique and provide insights for Agrobacterium-mediated transgenic wheat experiments.

The inhibition of tyrosinase (TYR) activity is an effective measure to inhibit melanin synthesis. At present, there are many methods with discrepant details that study the TYR inhibitory activity of samples. Under the same experimental conditions, this paper systematically studies whether enzyme species and sample addition methods are the key factors that determine the TYR inhibitory activity of samples. TYRs extracted from B16F10 cells, apple and mushroom, called BTYR, ATYR and MTYR, respectively, were selected to implement this study. Results showed that TYR inhibitory activities of samples were obviously affected by the above two factors. It was necessary to select the appropriate enzyme according to the problems to be explained. It was speculated that indirectly inhibitory activity reflected the comprehensive effects of samples on TYR catalytic activity and intracellular TYR synthesis pathway, while directly inhibitory activity reflected the effects of samples on TYR catalytic activity. Additionally, kojic acid could be used as a positive control for both B16F10 cells and MTYR models. The TYR inhibitory activity of β-arbutin was complicated and fickle, while that of epigallocatechin gallate (EGCG) was universal and stable, which is to say, EGCG always inhibited TYR activity in a dose-dependent manner. In conclusion, the TYR inhibitory activities of samples were affected by enzyme species and sample addition methods. Compared with the unstable β-arbutin, EGCG was more valuable for clinical research.

17α-Methyltestosterone (MT), a synthetic environmental endocrine disruptor with androgenic effects, has been shown to disrupt the reproductive system and inhibit germ cell maturation in Gobiocypris rarus. To further investigate the regulation of gonadal development by MT through the hypothalamic-pituitary-gonadal (HPG) axis, G. rarus were exposed to 0, 25, 50, and 100 ng/L of MT for 7, 14, and 21 days. We analyzed its biological indicators, gonadotropin-releasing hormone (GnRH), gonadotropins, reproduction-related gene expression, and brain tissue transcriptome profiles. We found a significant decrease in the gonadosomatic index (GSI) in G. rarus males exposed to MT for 21 days compared to the control group. GnRH, follicle-stimulating hormone (FSH), and luteinizing hormone (LH) levels, as well as the expressions of the gnrh3, gnrhr1, gnrhr3, fshβ, and cyp19a1b genes, were significantly reduced in the brains of both male and female fish when exposed to 100 ng/L MT for 14 days compared to the controls. Therefore, we further constructed four RNA-seq libraries from 100 ng/L MT-treated groups of male and female fish, obtaining 2412 and 2509 DEGs in male and female brain tissue, respectively. Three common pathways were observed to be affected in both sexes after exposure to MT, namely, nicotinate and nicotinamide metabolism, focal adhesion, and cell adhesion molecules. Furthermore, we found that MT affected the PI3K/Akt/FoxO3a signaling pathway through the upregulation of foxo3 and ccnd2, and the downregulation of pik3c3 and ccnd1. Therefore, we hypothesize that MT interferes with the levels of gonadotropin-releasing hormone (GnRH, FSH, and LH) in G. rarus brains through the PI3K/Akt/FoxO3a signaling pathway, and affects the expression of key genes in the hormone production pathway (gnrh3, gnrhr1 and cyp19a1b) to interfere with the stability of the HPG axis, thus leading to abnormal gonadal development. This study provides a multidimensional perspective on the damaging effects of MT on fish and confirms that G. rarus is a suitable model animal for aquatic toxicology.

17α-Methyltestosterone (17MT), a synthetic organic compound commonly found in sewage waters, can affect reproduction in aquatic animals, such as tilapia and yellow catfish. In the present study, male Gobiocypris rarus were exposed to 25, 50, and 100 ng/L of 17α-methyltestosterone (17MT) for 7 days. We first analyzed miRNA- and RNA-seq results to determine miRNA-target gene pairs and then developed miRNA-mRNA interactive networks after 17MT administration. Total weights, total lengths, and body lengths were not significantly different between the test groups and control groups. The paraffin slice method was applied to testes of G. rarus in the MT exposure and control groups. We found that there were more mature sperm (S) and fewer secondary spermatocytes (SSs) and spermatogonia (SGs) in the testes of control groups. As 17MT concentration increased, fewer and fewer mature sperm (S) were observed in the testes of male G. rarus. The results showed that FSH, 11-KT, and E2 were significantly higher in individuals exposed to 25 ng/L 17MT compared with the control groups. VTG, FSH, LH, 11-KT, and E2 were significantly lower in the 50 ng/L 17MT exposure groups compared to the control groups. VTG, FSH, LH, 11-KT, E2, and T were significantly lower in the groups exposed to 100 ng/L 17MT. High-throughput sequencing revealed 73,449 unigenes, 1205 known mature miRNAs, and 939 novel miRNAs in the gonads of G. rarus. With miRNA-seq, 49 (MT25-M vs. Con-M), 66 (MT50-M vs. Con-M), and 49 (MT100-M vs. Con-M) DEMs were identified in the treatment groups. Five mature miRNAs (miR-122-x, miR-574-x, miR-430-y, lin-4-x, and miR-7-y), as well as seven differentially expressed genes (soat2, inhbb, ihhb, gatm, faxdc2, ebp, and cyp1a1), which may be associated with testicular development, metabolism, apoptosis, and disease response, were assayed using qRT-PCR. Furthermore, miR-122-x (related to lipid metabolism), miR-430-y (embryonic development), lin-4-x (apoptosis), and miR-7-y (disease) were differentially expressed in the testes of 17MT-exposed G. rarus. This study highlights the role of miRNA-mRNA pairs in the regulation of testicular development and immune response to disease and will facilitate future studies on the miRNA-RNA-associated regulation of teleost reproduction.

It has been indicated that amyloid β (Aβ) plaques can be accumulated within the basement membranes of cerebrovascular smooth muscle cells (CVSMCs) and stimulate the induction of cerebral amyloid angiopathy (CAA). However, the exact mechanism(s) of which small molecules including callistephin mitigate the formation of Aβ aggregation and associated CAA is not well-understood. Therefore, in the present study, Aβ1–42 samples in the aggregation buffer were co-incubated for 36 h without or with of callistephin and the protein aggregation features along with the associated cytotoxicity against CVSMCs as the core components of cerebral arterial wall were explored by different biochemical and cellular methods. Fluorescence (ThT, Nile red) and CD techniques indicated the inhibition of Aβ1–42 fibrillization in the presence of callistephin. Cellular assays revealed that cytotoxicity of Aβ1–42 samples aged in the aggregation buffer with callistephin was much less against CVSMCs than Aβ1–42 amyloid alone through regulation of membrane leakage and downregulation of TNF-α and IL-6 at protein level. In conclusion, these data may provide useful information about the possible mechanisms by which callistephin can show its protective effect against CAA.

The product line approach enables the rapid building of software products by reusing core assets. How to manage the commonality and variability of product family is the core problem of product line engineering. The feature modeling approach is one of the main approaches to solve this problem. However, this approach does not define the internal functions of features. Some researchers use formal descriptions to define the function of features, but they do not consider the connection between feature specification and feature model. These product family modeling approaches result in product family models that suffer from inconsistent descriptions, which are difficult to resolve manually. To this end, this paper proposes a computer-aided approach to construct an internally consistent product family model. The method performs an inconsistency check during the formalization of each feature in the given initial feature model, pushing the evolution of the feature model and feature specification. The creation and evolution of feature models are achieved through a template-based feature modeling approach. The method is implemented by means of templates, each of which contains two parts of knowledge: the elements required for a class of feature model, and a way to convert the given elements into a formal description. An inconsistency check is achieved through a rule-based detection approach. Each rule contains two parts: a class of conditions that the feature model and the feature specification satisfy, and the actions to resolve inconsistencies once the conditions are satisfied. We have developed a tool. The tool guides the user to build an internally consistent product family model that contains the feature model and feature specifications. Experiments have shown that our approach can provide effective automation support for the process of formal product family modeling.

This paper introduces a high-performance self-powered nitrogen dioxide gas sensor based on Pd-modified ZnO/MoSe2 nanocomposites. Poly(vinyl alcohol) (PVA) nanofibers were prepared by high-voltage electrospinning and tribological nanogenerators (TENGs) were designed. The output voltage of TENG and the performance of the generator at different frequencies were measured. The absolute value of the maximum positive and negative voltage exceeds 200 V. Then, the output voltage of a single ZnO thin-film sensor, Pd@ZnO thin-film sensor and Pd@ZnO/MoSe2 thin-film sensor was tested by using the energy generated by TENG at 5 Hz, when the thin-film sensor was exposed to 1–50 ppm NO2 gas. The experimental results showed that the sensing response of the Pd@ZnO/MoSe2 thin-film sensor was higher than that of the single ZnO film sensor and Pd@ZnO thin-film sensor. The TENG-driven response rate of the Pd@ZnO/MoSe2 sensor on exposure to 50 ppm NO2 gas was 13.8. At the same time, the sensor had good repeatability and selectivity. The synthetic Pd@ZnO/MoSe2 ternary nanocomposite was an ideal material for the NO2 sensor, with excellent structure and performance.

Rapid and accurate detection of Camellia oleifera fruit is beneficial to improve the picking efficiency. However, detection faces new challenges because of the complex field environment. A Camellia oleifera fruit detection method based on YOLOv7 network and multiple data augmentation was proposed to detect Camellia oleifera fruit in complex field scenes. Firstly, the images of Camellia oleifera fruit were collected in the field to establish training and test sets. Detection performance was then compared among YOLOv7, YOLOv5s, YOLOv3-spp and Faster R-CNN networks. The YOLOv7 network with the best performance was selected. A DA-YOLOv7 model was established via the YOLOv7 network combined with various data augmentation methods. The DA-YOLOv7 model had the best detection performance and a strong generalisation ability in complex scenes, with mAP, Precision, Recall, F1 score and average detection time of 96.03%, 94.76%, 95.54%, 95.15% and 0.025 s per image, respectively. Therefore, YOLOv7 combined with data augmentation can be used to detect Camellia oleifera fruit in complex scenes. This study provides a theoretical reference for the detection and harvesting of crops under complex conditions.

Curvularia lunata (Wakker) Boed, the causal agent of leaf spot in maize, is prone to mutation, making it difficult to control. RNAi technology has proven to be an important tool of genetic engineering and functional genomics aimed for crop improvement. MicroRNAs (miRNAs), which act as post-transcriptional regulators, often cause translational repression and gene silencing. In this article, four small RNA (sRNA) libraries were generated from two maize genotypes inoculated by C. lunata; among these, ltR1 and ltR2 were from the susceptible variety Huangzao 4 (HZ), ltR3 and ltR4, from the resistant variety Luyuan (LY), and 2286, 2145, 1556 and 2504 reads were annotated as miRNA in these four sRNA libraries, respectively. Through the combined analysis of high-throughput sequencing, microarray hybridization and degradome, 48 miRNAs were identified as being related to maize resistance to C. lunata. Among these, PC-732 and PC-169, two new maize miRNAs discovered, were predicted to cleave mRNAs of metacaspase 1 (AMC1) and thioredoxin family protein (Trx), respectively, possibly playing crucial roles in the resistance of maize to C. lunata. To further confirm the role of PC-732 in the interaction of maize and C. lunata, the miRNA was silenced through STTM (short tandem target mimic) technology, and we found that knocking down PC-732 decreased the susceptibility of maize to C. lunata. Precisely speaking, the target gene of PC-732 might inhibit the expression of disease resistance-related genes during the interaction between maize and C. lunata. Overall, the findings of this study indicated the existence of miRNAs involved in the resistance of maize to C. lunata and will contribute to rapidly clarify the resistant mechanism of maize to C. lunata.

Potassium (K+) is essential for plant growth and stress responses. A deficiency in soil K+ contents can result in decreased wheat quality and productivity. Thus, clarifying the molecular mechanism underlying wheat responses to low-K+ (LK) stress is critical. In this study, a tandem mass tag (TMT)-based quantitative proteomic analysis was performed to investigate the differentially abundant proteins (DAPs) in roots of the LK-tolerant wheat cultivar “KN9204” at the seedling stage after exposure to LK stress. A total of 104 DAPs were identified in the LK-treated roots. The DAPs related to carbohydrate and energy metabolism, transport, stress responses and defense, and post-translational modifications under LK conditions were highlighted. We identified a high-affinity potassium transporter (TaHAK1-4A) that was significantly up-regulated after the LK treatment. Additionally, TaHAK1-4A was mainly expressed in roots, and the encoded protein was localized in the plasma membrane. The complementation assay in yeast suggested that TaHAK1-4A mediates K+ uptake under extreme LK conditions. The overexpression of TaHAK1-4A increased the fresh weight and root length of Arabidopsis under LK conditions and improved the growth of Arabidopsis athak5 mutant seedlings, which grow poorly under LK conditions. Moreover, silencing of TaHAK1-4A in wheat roots treated with LK stress decreased the root length, dry weight, K+ concentration, and K+ influx. Accordingly, TaHAK1-4A is important for the uptake of K+ by roots exposed to LK stress. Our results reveal the protein metabolic changes in wheat induced by LK stress. Furthermore, we identified a candidate gene potentially relevant for developing wheat lines with increased K+ use efficiency.

Nitric oxide (NO) is an active and critical nitrogen oxide in the microbe-driven nitrogen biogeochemical cycle, and is of great interest to medicine and the biological sciences. As a gas molecule prior to oxygen, NO respiration represents an early form of energy generation via various reactions in prokaryotes. Major enzymes for endogenous NO formation known to date include two types of nitrite reductases in denitrification, hydroxylamine oxidoreductase in ammonia oxidation, and NO synthases (NOSs). While the former two play critical roles in shaping electron transport pathways in bacteria, NOSs are intracellular enzymes catalyzing metabolism of certain amino acids and have been extensively studied in mammals. NO interacts with numerous cellular targets, most of which are redox-active proteins. Doing so, NO plays harmful and beneficial roles by affecting diverse biological processes within bacterial physiology. Here, we discuss recent advances in the field, including NO-forming enzymes, the molecular mechanisms by which these enzymes function, physiological roles of bacterial NOSs, and regulation of NO homeostasis in bacteria.

A retaining wall with sandy fill was considered as the research object in order to study the nonlimiting active earth pressure under the rotation about the base (RB mode). Rankine’s and Coulomb’s earth pressure theories are no longer applicable to the above conditions (RB mode and nonlimiting active earth pressure). In order to improve the traditional earth pressure calculation methods (Rankine and Coulomb), a calculation method using curvilinear thin layer elements is presented with overall considerations of wall displacement, soil arching effect, and friction angle exertion coefficient to deduce the nonlimit active earth pressure under RB. Additionally, the calculation results were in good agreement with model test data (from Fang and Smita). Moreover, a parametric analysis was carried out. It was revealed that the developed value of the shear strength decreased with the depth, and the active earth pressure distribution curve was linear and nonlinear in the upper and lower halves, respectively.

As an opportunistic pathogen, Escherichia coli (E. coli) forms biofilm that increases the virulence of bacteria and antibiotic resistance, posing a serious threat to human and animal health. Recently, ginkgetin (Gin) has been discovered to have antiinflammatory, antioxidant, and antitumor properties. In the present study, we evaluated the antibiofilm and antibacterial synergist of Gin against E. coli. Additionally, Alamar Blue assay combined with confocal laser scanning microscope (CLSM) and crystal violet (CV) staining was used to evaluate the effect of antibiofilm and antibacterial synergist against E. coli. Results showed that Gin reduces biofilm formation, exopolysaccharide (EPS) production, and motility against E. coli without limiting its growth and metabolic activity. Furthermore, we identified the inhibitory effect of Gin on AI-2 signaling molecule production, which showed apparent anti-quorum sensing (QS) properties. The qRT-PCR also indicated that Gin reduced the transcription of curli-related genes (csgA, csgD), flagella-formation genes (flhC, flhD, fliC, fliM), and QS-related genes (luxS, lsrB, lsrK, lsrR). Moreover, Gin showed obvious antibacterial synergism to overcome antibiotic resistance in E. coli with marketed antibiotics, including gentamicin, colistin B, and colistin E. These results suggested the potent antibiofilm and novel antibacterial synergist effect of Gin for treating E. coli infections.

The two-electron oxygen reduction reaction (2e−ORR) pathway electrochemical synthesis to H2O2 has the advantages of low investment and environmental protection and is considered to be a promising green method. Herein, the oxidized Mo2TiC2 MXene (O-Mo2TiC2) was successfully synthesized by a facile hydrothermal method as an electrocatalyst in electrocatalytic H2O2 production. The O-Mo2TiC2 achieved the 90% of H2O2 selectivity and 0.72 V vs. RHE of the onset potential. Moreover, O-Mo2TiC2 showed high charge transfer ability and long-term stable working ability of 40 h. This significantly enhanced electrocatalytic H2O2 production capacity is assigned the oxidation treatment of Mo2TiC2 MXene to generate more oxygen-containing groups in O-Mo2TiC2. This work provides a promising catalyst candidate for the electrochemical synthesis of H2O2.

Product derivation is the process of building a specific product from a software product line. Effective product derivation can improve software reuse productivity. Existing methods can only obtain abstract feature models, lacking detailed specifications of individual features. They are more about deriving code assets or class diagram templates without precise model descriptions for specific products. This article proposes a product derivation approach to obtain a formal specification of a specific product based on the feature model and formal specification. We use the integration ordering and behavior preserving integration techniques to integrate the formal specification for each feature pair. The method is divided into two steps. First, it determines the feature formal specification integration ordering based on the feature model. Then, the behavior-preserving integration will be conducted for pairs, including declaration integration, functional scenario path generation, and function integration based on path matching. Behavior preserving integration guarantees consistent behavior to ensure the quality of the formal specification after integration. Finally, we developed a support tool to conduct a case study. The tool first guides the user to perform feature functional scenario path matching, then performs functional integration based on the matching results and repeats the above steps to generate a product model. The result indicates that our method facilitates the derivation process and improves the quality of the generated models.

The major facilitator superfamily (MFS) is the largest secondary transporter family and is responsible for transporting a broad range of substrates across the biomembrane. These proteins are involved in a series of conformational changes during substrate transport. To decipher the transport mechanism, it is necessary to obtain structures of these different conformations. At present, great progress has been made in predicting protein structure based on coevolutionary information. In this study, AlphaFold2 was used to predict different conformational structures for 69 MFS transporters of E. coli after the selective mutation of residues at the interface between the N- and C-terminal domains. The predicted structures for these mutants had small RMSD values when compared to structures obtained using X-ray crystallography, which indicates that AlphaFold2 predicts the structure of MSF transporters with high accuracy. In addition, different conformations of other transporter family proteins have been successfully predicted based on mutation methods. This study provides a structural basis to study the transporting mechanism of the MFS transporters and a method to probe dynamic conformation changes of transporter family proteins when performing their function.

Drought decreases the inorganic phosphate (Pi) supply of soil, resulting in Pi starvation of plants, but the molecular mechanism of how plants, especially the perennial trees, are tolerant to drought stress and Pi starvation, is still elusive. In this study, we identified an AP2/ERF transcription factor gene, PalERF2, from Populus alba var. pyramidalis, and it was induced by both mannitol treatment and Pi starvation. Overexpressing and knocking-down of PalERF2 both enhanced and attenuated tolerance to drought stress and Pi deficiency compared to WT, respectively. Moreover, the overexpression of PalERF2 up-regulated the expression levels of Pi starvation-induced (PSI) genes and increased Pi uptake under drought conditions; however, its RNAi poplar showed the opposite phenotypes. Subsequent analysis indicated that PalERF2 directly modulated expressions of drought-responsive genes PalRD20 and PalSAG113, as well as PSI genes PalPHL2 and PalPHT1;4, through binding to the DRE motifs on their promoters. These results clearly indicate that poplars can recruit PalERF2 to increase the tolerance to drought and also elevate Pi uptake under drought stress.

The calmodulin-binding transcription activator (CAMTA) is a Ca2+/CaM-mediated transcription factor (TF) that modulates plant stress responses and development. Although the investigations of CAMTAs in various organisms revealed a broad range of functions from sensory mechanisms to physiological activities in crops, little is known about the CAMTA family in wheat (Triticum aestivum L.). Here, we systematically analyzed phylogeny, gene expansion, conserved motifs, gene structure, cis-elements, chromosomal localization, and expression patterns of CAMTA genes in wheat. We described and confirmed, via molecular evolution and functional verification analyses, two new members of the family, TaCAMTA5-B.1 and TaCAMTA5-B.2. In addition, we determined that the expression of most TaCAMTA genes responded to several abiotic stresses (drought, salt, heat, and cold) and ABA during the seedling stage, but it was mainly induced by drought stress. Our study provides considerable information about the changes in gene expression in wheat under stress, notably that drought stress-related gene expression in TaCAMTA1b-B.1 transgenic lines was significantly upregulated under drought stress. In addition to providing a comprehensive view of CAMTA genes in wheat, our results indicate that TaCAMTA1b-B.1 has a potential role in the drought stress response induced by a water deficit at the seedling stage.

This study proposes a new damage identification method for hangers of arch bridges using the static deflection difference at the anchorage point of the hanger and the tie-beam. The relationship between the ratio of cable tension loss and the deflection difference at the anchorage point is studied. For the first time, the deflection difference influence matrix for hanger damage identification is defined. The static deflection change parameter is formulated as a function of the deflection difference influence matrix and the ratio of cable tension loss. The study shows that the percentage of cable tension loss can be obtained from the changes in static deflection at the anchorage point and the deflection difference influence matrix. Therefore, by observing a plot of the ratio of cable tension loss, the damage locations can be identified conveniently. Numerical and laboratory studies show that this method can accurately locate the damaged hanger of through-arch bridges under various scenarios. The proposed damage detection method has a clear theoretical base and is simple to operate, and it is suitable for practical engineering.

3’-Phosphoadenosine 5’-monophosphate (pAp) is a byproduct of sulfate assimilation and coenzyme A metabolism. pAp can inhibit the activity of 3′-phosphoadenosine 5′-phosphosulfate (PAPS) reductase and sulfotransferase and regulate gene expression under stress conditions by inhibiting XRN family of exoribonucleases. In metazoans, plants, yeast, and some bacteria, pAp can be converted into 5’-adenosine monophosphate (AMP) and inorganic phosphate by CysQ. In some bacteria and archaea, nanoRNases (Nrn) from the Asp-His-His (DHH) phosphoesterase superfamily are responsible for recycling pAp. In addition, histidinol phosphatase from the amidohydrolase superfamily can hydrolyze pAp. The bacterial enzymes for pAp turnover and their catalysis mechanism have been well studied, but these processes remain unclear in archaea. Pyrococcus yayanosii, an obligate piezophilic hyperthermophilic archaea, encodes a DHH family pApase homolog (PyapApase). Biochemical characterization showed that PyapApase can efficiently convert pAp into AMP and phosphate. The resolved crystal structure of apo-PyapApase is similar to that of bacterial nanoRNaseA (NrnA), but they are slightly different in the α-helix linker connecting the DHH and Asp-His-His associated 1 (DHHA1) domains. The longer α-helix of PyapApase leads to a narrower substrate-binding cleft between the DHH and DHHA1 domains than what is observed in bacterial NrnA. Through mutation analysis of conserved amino acid residues involved in coordinating metal ion and binding substrate pAp, it was confirmed that PyapApase has an ion coordination pattern similar to that of NrnA and slightly different substrate binding patterns. The results provide combined structural and functional insight into the enzymatic turnover of pAp, implying the potential function of sulfate assimilation in hyperthermophilic cells.

Rhus potaninii Maxim is an economically and medicinally important tree species in China. It produces galls (induced by aphids) with a high abundance of tannins. Here, we discuss the histology, cellular structures and their distribution, and the macromolecular components of secretive glandular trichomes on the leaves of R. potaninii. A variation in the density of glandular trichomes and tomenta was found between the adaxial and abaxial sides of a leaf in different regions and stages of the leaf. The glandular trichomes on R. potaninii trees comprise a stalk with no cellular structure and a head with 8–15 cells. Based on staining, we found that the secretion of glandular trichomes has many polysaccharides, phenolic compounds, and acidic lipids but very few neutral lipids. The dense glandular trichomes provide mechanical protection for young tissues; additionally, their secretion protects the young tissues from pathogens by a special chemical component. According to transcriptome analysis, we found enhanced biosynthetic and metabolism pathways of glycan, lipids, toxic amino acids, and phenylpropanoids. This shows a similar tendency to the staining. The numbers of differentially expressed genes were large or small; the averaged range of upregulated genes was greater than that of the downregulated genes in most subpathways. Some selectively expressed genes were found in glandular trichomes, responsible for the chitinase activity and pathogenesis-related proteins, which all have antibacterial activity and serve for plant defense. To our knowledge, this is the first study showing the components of the secretion from glandular trichomes on the leaf surface of R. potaninii.

In this study, the novel mixed-phase TiO2 nanocrystals (s-TiO2) with nearly equivalent anatase/rutile ratio were fabricated in the reagent of sec-butanol at the relatively low temperature of 80 °C by using a facile one-step condensing reflux method. The photocatalytic water splitting hydrogen production performance of s-TiO2 nanocrystals is close to that of commercial TiO2 (P25), and its photocatalytic degradation performance is about four times that of P25. The energy-level staggered interfaces and surface bridged hydroxyl groups significantly increased due to the anatase/rutile mixed-phase crystal structure and high specific surface area, which might generate the synergistic effect for the improvement of photocatalytic degradation.

It is highly desired that synthesis of photothermal agents with near-infrared (NIR) absorption, excellent photostability, and high photothermal conversion efficiency are essential for potential applications. In this work, three (D-A) conjugated polymers (PBABDF-BDTT, PBABDF-BT, and PBABDF-TVT) based on aza-heterocycle, bis(2-oxo-7-azaindolin-3-ylidene)benzodifurandione (BABDF) as the strong acceptor, and benzodithiophene-thiophene (BDTT), bithiophene (BT), and thiophene-vinylene-thiophene (TVT) as the donors, were designed and synthesized. The conjugated polymers showed significant absorption in the NIR region and a maximum absorption peak at 808 nm by adjusting the donor and acceptor units. Their photothermal properties were also investigated by using poly(ethylene glycol)-block-poly(hexyl ethylene phosphate) (mPEG-b-PHEP) to stabilize the conjugated polymers. Photoexcited conjugated polymer (PBABDF-TVT) nanoparticles underwent non-radiative decay when subjected to single-wavelength NIR light irradiation, leading to an excellent photothermal conversion efficiency of 40.7%. This work indicated the aza-heterocycle BABDF can be a useful building block for constructing D-A conjugated polymer with high conversion efficiency.

Attachment of conceptus to the endometrial luminal epithelium (LE) is a critical event for early placentation in Eutheria. Since the attachment occurs at a particular site within the uterus, a coordinated communication between three spatially distinct compartments (conceptus and endometrial LE from two anatomical regions of the uterus to which conceptus attaches and does not attach) is essential but remains to be fully characterized. Using the laser capture microdissection (LCM) technique, we firstly developed an approach that can allow us to pair the pig conceptus sample with its nearby endometrial epithelium sample without losing the native spatial information. Then, a comprehensive spatio-temporal transcriptomic profile without losing the original conceptus-endometrium coordinates was constructed. The analysis shows that an apparent difference in transcriptional responses to the conceptus exists between the endometrial LE from the two anatomically distinct regions in the uterus. In addition, we identified the communication pathways that link the conceptus and endometrial LE and found that these pathways have important roles in conceptus attachment. Furthermore, a number of genes whose expression is spatially restricted in the two different anatomical regions within the uterus were characterized for the first time and two of them (SULT2A1 and MEP1B) may cooperatively contribute to establish conceptus attachment in pigs. The results from our study have implications in understanding of conceptus/embryo attachment in pigs and other large polytocous species.

Curcumin is a naturally occurring polyphenolic compound that is commonly used in both medicine and food additives, but its low aqueous solubility and poor bioavailability hinder further clinical applications. For assessing the effect of the glycosylation of curcumin on its aqueous solubility, two glycosyltransferase genes (BsGT1 and BsGT2) were cloned from the genome of the strain Bacillus subtilis ATCC 6633 and over-expressed in Escherichia coli. Then, the two glycosyltransferases were purified, and their glycosylation capacity toward curcumin and its two analogues was verified. The results showed that both BsGT1 and BsGT2 could convert curcumin and its two analogues into their glucosidic derivatives. Then, the structures of the derivatives were characterized as curcumin 4′-O-β-D-glucoside and two new curcumin analogue monoglucosides namely, curcumoid-O-α-D-glucoside (2a) and 3-pentadienone-O-α-D-glucoside (3a) by nuclear magnetic resonance (NMR) spectroscopy. Subsequently, the dissolvability of curcumin 4′-O-β-D-glucoside was measured to be 18.78 mg/L, while its aglycone could not be determined. Furthermore, the optimal catalyzing conditions and kinetic parameters of BsGT1 and BsGT2 toward curcumin were determined, which showed that the Kcat value of BsGT1 was about 2.6-fold higher than that of BsGT2, indicating that curcumin is more favored for BsGT2. Our findings effectively apply the enzymatic approach to obtain glucoside derivatives with enhanced solubility.

Due to multiple charge transport pathways, adjustable layer spacing, compositional flexibility, low manufacturing cost, and absorption of visible light, layered double hydroxides (LDHs) are a promising material for wastewater treatment. In this study, LDH films and Fe-doped LDH films with different metal ions (Ni, Al, Fe) on the surface of conductive cloth were successfully prepared and applied for the photocatalytic degradation of wastewater containing methyl orange and Ag ions under visible-light irradiation. The chemical state of Fe ions and the composition of LDHs on methyl orange photodegradation were investigated. The experimental results showed that LDH films exhibited high photocatalytic activity. The photocatalytic activity of LDH films on methyl orange improved in the mixed wastewater, and the Fe-doped NiAl−LDH films exhibited best visible-light photocatalytic performance. The analysis showed that Ag ions in the mixed wastewater were reduced by the LDH films and subsequently deposited on the surface of the LDH films. The Ag nanoparticles acted as electron traps and promoted the photocatalytic activity of the LDH films on methyl orange. Thus, we have demonstrated that prepared LDH films can be used in the treatment of mixed wastewater and have broad application prospects in environmental remediation and purification processes.
