Consumers feeding at the aquatic–terrestrial ecosystem interface may obtain a mixture of aquatic and terrestrial diet resources that vary in nutritional composition. However, in lake riparian spiders, the relative significance of aquatic versus terrestrial diet sources remains to be explored. We investigated the trophic transfer of lipids and polyunsaturated fatty acids (PUFA) from emergent aquatic and terrestrial insects to spiders at varying distances from the shoreline of a subalpine lake in Austria, using differences in fatty acid profiles and compound-specific stable carbon (δ13C) and hydrogen (δ2H) isotopes. The omega-3 PUFA content of emergent aquatic insects was higher than that of terrestrial insects. Emergent aquatic insects contained on average 6.6 times more eicosapentaenoic acid (EPA) and 1.2 times more α-linolenic acid (ALA) than terrestrial insects, whereas terrestrial insects contained on average 2.6 times more linoleic acid (LIN) than emergent aquatic insects. Spiders sampled directly on the lake and in upland habitats had similar EPA contents, but this EPA was derived from different diet sources, depending on the habitat. The δ13CEPA and δ2HEPA values of ‘lake spiders' revealed an aquatic diet pathway (i.e. EPA of aquatic origin). In contrast, EPA of spiders collected in terrestrial habitats was depleted in both 13C and 2H compared to any potential food sources, and their ALA isotopic values, suggesting that EPA was partly bioconverted from its dietary precursor ALA (i.e. internal pathway). The δ2H values of fatty acids clearly indicated that diet sources differed depending on the spider's habitat, which was less evident from the δ13C values of the fatty acids. Our data highlight that spiders can use two distinct pathways (trophic versus metabolic) to satisfy their physiological EPA demand, depending on habitat use and dietary availability. published

1. Omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) are essential micronutrients for optimal functioning of cellular metabolism and for somatic growth of all vertebrates including fishes. In addition, n-3 LC-PUFA could also play a key role in response of fishes and other ectothermic vertebrates to changing temperatures. 2. An important, but largely overlooked, consequence of climate change is the reduced availability of dietary n-3 LC-PUFA in aquatic food webs. Changes in availability of dietary n-3 LC-PUFA have recently been proposed as a major driver of novel adaptations and diversification of consumers. Yet, there is only limited knowledge about how n-3 LC-PUFA depletion in aquatic food-webs will affect the performance of wild fishes. 3. Here we combine biochemistry and physiology at the cellular level with physiological and cognitive processes at the whole-animal level to test how ecologically relevant deprivation of n-3 LC-PUFA affects performance of wild juvenile Atlantic salmon (Salmo salar). 4. We found that juvenile salmon had a limited capacity to maintain the fatty acid profile of both muscle and brain under a n-3 LC-PUFA-deficient diet. Despite these findings, brain tissues showed remarkable functional stability in mitochondrial metabolism, and we found no effect of diet on learning ability. However, we found that mitochondrial efficiency in muscles and the somatic growth were reduced under a n-3 LC-PUFA-deficient diet. Importantly, we discovered that the somatic growth of juvenile salmon within both treatments decreased with increasing rate of DHA synthesis and retention. 5. Since DHA is essential for functioning of cellular metabolism, which together with body size are traits closely related to fitness of wild fishes, we suggest that the trade-off between growth rate and accumulation of DHA could play a critical role in resilience of juvenile salmon to the ongoing rapid environmental change.

The River Continuum Concept implies that consumers in headwater streams have greater dietary access to terrestrial basal resources, but recent studies have highlighted the dietary importance of high-quality algae. Algae provide consumers with physiologically important omega-3 (n-3) polyunsaturated fatty acids (PUFA), particularly eicosapentaenoic acid (EPA). However, terrestrial plants and most benthic stream algae lack the long-chain (LC) n-3 PUFA docosahexaenoic acid (DHA, 22:6n-3), which is essential for neural development in fish and other vertebrates. We sampled subalpine streams to investigate how the PUFA composition of neural (brain and eyes), muscle, and liver tissues of freshwater fish is related to their potential diets (macroinvertebrates, epilithon, fresh and conditioned terrestrial leaves). The PUFA composition of consumers was more similar to epilithon than to terrestrial leaves. Storage lipids of eyes most closely resembled dietary PUFA (aquatic invertebrates and algae). However, DHA and arachidonic acid (ARA, 20:4n-6) were not directly available in the diet but abundant in organs. This implies that algal PUFA were selectively retained or were produced internally via enzymatic PUFA conversion by aquatic consumers. This field study demonstrates the nutritional importance of algal PUFA for neural organs in aquatic consumers of headwater regions. Across ecosystems, resources vary in their nutritional composition and thus their dietary value to consumers. Animals can either access organic compounds, such as fatty acids, directly from diet or through internal biosynthesis, and the extent to which they use these two alternatives likely varies based on the availability of such compounds across the nutritional landscape. Cross-ecosystem subsidies of important dietary nutrients, like omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA), may provide consumers with the opportunity to relax the demands of synthesis and rely upon dietary flexibility rather than internal metabolic processes. Here, we examined how dietary flexibility and distance from a lake influenced the degree to which generalist insectivores relied upon dietary n-3 LC-PUFA from emergent aquatic insects versus n-3 LC-PUFA synthesized from precursor compounds found in terrestrial insects. We used bulk and compound-specific stable isotope analyses to understand spider and insectivorous bird (Blue Tit; Cyanistes caeruleus) reliance on aquatic and terrestrial resources, including dietary PUFA sources, along a riparian to upland gradient from a lake. We simultaneously investigated n-3 LC-PUFA synthesis ability in nestlings using 13C fatty acid labeling. We found that riparian spiders took advantage of emergent aquatic insect subsidies, deriving their overall diet and their n-3 PUFA from aquatic resources whereas nestling birds at all distances and upland spiders relied upon terrestrial resources, including PUFA. Our 13C labeling experiment demonstrated that nestling tits were able to synthesize the n-3 LC-PUFA docosahexaenoic acid from the dietary precursor α-linolenic acid, suggesting that they are not limited by aquatic resources to satisfy their LC-PUFA requirements. Overall, this study suggests that habitat generalist insectivores vary in the degree to which they can shift diet to take advantage of high-quality aquatic resources depending upon both their foraging flexibility and internal synthesis capacity.

In burn injuries, risk factors and limitations to treatment success are difficult to assess clinically. However, local cellular responses are characterized by specific gene-expression patterns. MicroRNAs (miRNAs) are single-stranded, non-coding RNAs that regulate mRNA expression on a posttranscriptional level. Secreted through exosome-like vesicles (ELV), miRNAs are intracellular signalers and epigenetic regulators. To date, their role in the regulation of the early burn response remains unclear. Here, we identified 43 miRNAs as potential regulators of the early burn response through the bioinformatics analysis of an existing dataset. We used an established human ex vivo skin model of a deep partial-thickness burn to characterize ELVs and miRNAs in dermal interstitial fluid (dISF). Moreover, we identified miR-497-5p as stably downregulated in tissue and dISF in the early phase after a burn injury. MiR-218-5p and miR-212-3p were downregulated in dISF, but not in tissue. Target genes of the miRNAs were mainly upregulated in tissue post-burn. The altered levels of miRNAs in dISF of thermally injured skin mark them as new biomarker candidates for burn injuries. To our knowledge, this is the first study to report miRNAs altered in the dISF in the early phase of deep partial-thickness burns.

RATIONALE Compound-specific stable isotope analysis (CSIA) is a powerful tool for a better understanding of trophic transfer of dietary molecules in and across ecosystems. Hydrogen isotope values (δ2 H) in consumer tissues have potential to more clearly distinguish dietary sources than 13 C or 15 N values within and among habitats, but have not been used at the fatty acid level for ecological purposes. METHODS Here we demonstrate a new online high-capacity gas chromatography-isotope ratio mass spectrometry technique (2 H-CSIA) that offers accurate and reproducible determination of δ2 H values for a range of fatty acids from organisms of aquatic food webs. RESULTS We show that lipid extracts obtained from aquatic sources, such as biofilms, leaves, invertebrates, or fish muscle tissue, have distinctive δ2 H values that can be used to assess sources and trophic interactions, as well as dietary allocation and origin of fatty acids within consumer tissue. CONCLUSIONS The new 2 H-CSIA method can be applied to evaluate sources and trophic dynamics of fatty acids in organisms ranging from food web ecology to migratory connectivity.

In recent decades, much conceptual thinking in trophic ecology has been guided by theories of nutrient limitation and the flow of elements, such as carbon and nitrogen, within and among ecosystems. More recently, ecologists have also turned their attention to examining the value of specific dietary nutrients, in particular polyunsaturated fatty acids (PUFA), among which the omega-3 PUFA, especially eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) play a central role as essential components of neuronal cell membranes in many organisms. This review focuses on a new neuro-ecological approach stemming from the biochemical (mechanistic) and physiological (functional) role of DHA in neuronal cell membranes, in particular in conjunction with G-protein coupled receptors (GPCRs). We link the co-evolution of these neurological functions to metabolic dependency on dietary omega-3 PUFA. We outline ways in which deficiencies in dietary DHA supply may affect, cognition, vision, and behaviour, and ultimately, the biological fitness of consumers. We then review emerging evidence that changes in access to dietary omega-3 PUFA may ultimately have profound impacts on trophic interactions leading to potential changes in community structure and ecosystem functioning that, in turn, may affect the supply of DHA within and across ecosystems, including the supply for human consumption.

Application of a multilayered coating architecture based on two alternating hard materials provides a strategy to enhance the mechanical properties of hard coatings for the metal cutting industry. Within this work, the mechanical behavior of CVD TiN/TiBN multilayer coatings was correlated with their microstructure and compared to the respective single-layers. Multilayers with different bilayer periods (1400, 800, 300 and 200 nm) were prepared in an industrial-scale thermal CVD plant by alternate variation of the feed gas composition. Complementary X-ray diffraction, Raman spectroscopy and transmission electron microscopy investigations confirmed a dominating face-centered cubic TiN structure, accompanied by hexagonal TiB2 and amorphous TiB in the B containing coatings. The addition of B and a decreasing bilayer period resulted in a decreasing grain size and increasing hardness. Micromechanical bending tests revealed an increase of strength and fracture toughness with addition of B and increasing layer number. While the highest strength was observed in a TiBN single-layered coating, the multilayered TiN/TiBN with a bilayer period of 200 nm was the hardest and at the same time toughest of the investigated coatings.

12 pages, 8 figures, 3 tables, 1 appendix, Dietary uptake is a key step in conveying both toxic mercury (Hg; particularly as highly bioavailable methylmercury, MeHg) and essential dietary biochemicals, such as polyunsaturated fatty acids (PUFA), across trophic levels within aquatic food webs. Using stable isotopes and fatty acids we evaluated the role of food sources in size-fractioned plankton and littoral macroinvertebrates for the bioaccumulation of total Hg and MeHg in six oligotrophic and one mesotrophic Swedish lakes with differing concentrations of dissolved organic carbon (DOC). We found that the consumption of both algal and terrestrial diets (assessed by PUFA and long-chain saturated fatty acids, respectively) predicted >66% of the Hg concentration variability in meso- (100–500 μm) and macrozooplankton (>500 μm) in oligotrophic lakes. Data included are related to the research article "Isolation of biofunctional bovine immunoglobulin G from milk- and colostral whey with mixed-mode chromatography at lab and pilot scale" (Heidebrecht et al., 2018) [1]. Data show individual bovine whey proteins in flow-through and elution fractions using different chromatographic resins as well as different binding and elution conditions. The relevant analytical methods for individual protein detection were SDS-PAGE and reversed phase- high performance liquid chromatography. The focus of the data is on the two mixed mode materials MEP HyperCel™ and Capto™-multimodal chromatography. Resins were used individually, in series and at different scale. Data provide information at which binding and elution conditions it is possible to isolate bovine IgG from milk and colostral whey and at which purity.

Objective— HO-1 (heme oxygenase-1) induction may prevent or reduce ischemia-reperfusion injury. We previously evaluated its in vivo induction after a single systemic administration of heme arginate in peripheral blood mononuclear cells. The current trial was designed to assess the pharmacological tissue induction of HO-1 in the human heart with heme arginate in vivo. Approach and Results— Patients planned for conventional aortic valve replacement received placebo (n=8), 1 mg/kg (n=7) or 3 mg/kg (n=9) heme arginate infused intravenously 24 hours before surgery. A biopsy of the right ventricle was performed directly before aortic cross-clamping and after cross-clamp release. In addition, the right atrial appendage was partially removed for analysis. HO-1 protein and mRNA concentrations were measured in tissue samples and in peripheral blood mononuclear cells before to and up to 72 hours after surgery. No study medication-related adverse events occurred. A strong, dose-dependent effect on myocardial HO-1 mRNA levels was observed (right ventricle: 7.9±5.0 versus 88.6±49.1 versus 203.6±148.7; P =0.002 and right atrium: 10.8±8.8 versus 229.8±173.1 versus 392.7±195.7; P =0.001). This was paralleled by a profound increase of HO-1 protein concentration in atrial tissue (8401±3889 versus 28 585±10 692 versus 29 022±8583; P P P =0.041). No effect on plasma HO-1 protein levels could be detected. Conclusions— Myocardial HO-1 mRNA and protein can be dose-dependently induced by heme arginate. 1. The ecological role of emergent aquatic insects from lakes in exporting dietary polyunsaturated fatty acids (PUFA) across the freshwater-land interface is still poorly understood.2. In this field study, we explored the seasonal biomass export of emergent insects from three subalpine lakes and investigated how lipids of emergent insects were related to lake bathymetry, lipids of organic matter in lake sediments (i.e., basal resources), and the taxonomic composition of insects.3. The total lipid and PUFA fluxes of emergent insects were strongly related to taxonomy and lake bathymetry, but weakly associated with the PUFA content of the uppermost lake sediment layers. PUFA flux estimates of the dominant taxon, Chironomidae, from the shallowest lake (3 m depth; 125 g PUFA m−2 season−1) were considerably higher than those from the deepest lake (33 m depth; 56 g PUFA m−2 season−1), due to the higher per area biomass of emergent insects from this shallow lake. Insect taxonomy also affected the composition of PUFA transfer to land: Chironomidae were richer in ω-6 PUFA, such as linoleic acid (18:2n-6) and arachidonic acid (20:4n-6), whereas Ephemeroptera and Trichoptera contained more ω-3 PUFA, especially α-linolenic acid (18:3n-3) and eicosapentaenoic acid (20:5n-3).4. Climate change scenarios predict increases in temperature and organic matter supply from land to water, which affect trophic transfer of nutrients and contaminants in aquatic food webs. How essential nutrients, such as polyunsaturated fatty acids (PUFA), and potentially toxic contaminants, such as methylmercury (MeHg), at the base of aquatic food webs will be affected under climate change scenarios, remains unclear. The objective of this outdoor mesocosm study was to examine how increased water temperature and terrestrially-derived dissolved organic matter supply (tDOM; i.e., lake browning), and the interaction of both, will influence MeHg and PUFA in organisms at the base of food webs (i.e. seston; the most edible plankton size for zooplankton) in subalpine lake ecosystems. Lakes and streams in mountain regions are important contributors to carbon (C) fluxes. However, detailed carbon balances of the stream‐lake continuum are rare. Combining eddy covariance (EC) measurements of lake‐atmosphere net ecosystem CO2 exchange with measurements of fluvial C fluxes (dissolved organic C, DOC; particulate organic C, POC; dissolved inorganic C, DIC), and in‐lake sedimentation, we here present a detailed annual C balance of an oligotrophic clearwater lake in the eastern European Alps. The C flux into the lake was 1522 Mg C year−1 by DIC (93%), DOC (6%), and POC (0.7%). The largest C losses were fluvial exports (1595 Mg C year−1) of DIC (93%), DOC (6%), and POC (0.8%), while sedimentation accounted for 7.3 Mg C year−1. The residual of all fluvial and sedimentation fluxes revealed the lake as a net sink of atmospheric CO2 of 77 Mg C year−1. The EC measurement confirmed a small positive or negative contribution of atmospheric exchange to the lake C balance. In‐lake transfer among C pools was only significant for the flux from DIC to POC (8.4 Mg C year−1), which, following our model, is the transfer through primary production in summer. Fluvial DOC and DIC fluxes were controlled by discharge; POC retention and sedimentation depended on the meteorological season and in‐lake residence time. Following our findings, we conclude that hydrology acted as the most important control for the C balance of this clearwater mountain lake by controlling inflow, outflow, and sedimentation fluxes.

International audience; Microbial parasites have only recently been included in planktonic food web studies, but their functional role in conveying dietary energy still remains to be elucidated. Parasitic fungi (chytrids) infecting phytoplankton may constitute an alternative trophic link and promote organic matter transfer through the production of dissemination zoospores. Particularly, during proliferation of inedible or toxic algal species, such as large Cyanobacteria fostered by global warming, parasites can constitute an alternative trophic link providing essential dietary nutrients that support somatic growth and reproduction of consumers. Using phytoplankton-parasites associations grown under laboratory controlled conditions we assessed the fatty acids and biochemical composition of species with different nutritional quality and followed the metabolic pathway from the algal host and their parasites zoospores using compound specific stable isotope analysis. This study demonstrated that chytrids are trophic upgraders able to retain essential nutrients that can be transferred to upper trophic levels both in terms of organic matter quantity and nutritional quality. Through the production of zoospores, nutritionally important long-chain polyunsaturated fatty acids that can be consequently assimilated by consumers. We conclude that parasitism at the base of aquatic food webs may represent a crucial trophic link for dietary nutrients and essential biomolecules alternative to herbivory or bacterivory, which can be particularly crucial during the proliferation of inedible or nutritionally inadequate algal species fostered by climate change.

The metabolite α-ketoglutarate is membrane-impermeable, meaning that it is usually added to cells in the form of esters such as dimethyl α-ketoglutarate (DMKG), trifluoromethylbenzyl α-ketoglutarate (TFMKG) and octyl α-ketoglutarate (O-KG). Once these compounds cross the plasma membrane, they are hydrolyzed by esterases to generate α-ketoglutarate, which remains trapped within cells. Here, we systematically compared DMKG, TFMKG and O-KG for their metabolic and functional effects. All three compounds similarly increased the intracellular levels of α-ketoglutarate, yet each of them had multiple effects on other metabolites that were not shared among the three agents, as determined by mass spectrometric metabolomics. While all three compounds reduced autophagy induced by culture in nutrient-free conditions, TFMKG and O-KG (but not DMKG) caused an increase in baseline autophagy in cells cultured in complete medium. O-KG (but neither DMKG nor TFMK) inhibited oxidative phosphorylation and exhibited cellular toxicity. Altogether, these results support the idea that intracellular α-ketoglutarate inhibits starvation-induced autophagy and that it has no direct respiration-inhibitory effect.

To understand consumer dietary requirements and resource use across ecosystems, researchers have employed a variety of methods, including bulk stable isotope and fatty acid composition analyses. Compound-specific stable isotope analysis (CSIA) of fatty acids combines both of these tools into an even more powerful method with the capacity to broaden our understanding of food web ecology and nutritional dynamics. Here, we provide an overview of the potential that CSIA studies hold and their constraints. We first review the use of fatty acid CSIA in ecology at the natural abundance level as well as enriched physiological tracers, and highlight the unique insights that CSIA of fatty acids can provide. Next, we evaluate methodological best practices when generating and interpreting CSIA data. We then introduce three cutting-edge methods: hydrogen CSIA of fatty acids, and fatty acid isotopomer and isotopologue analyses, which are not yet widely used in ecological studies, but hold the potential to address some of the limitations of current techniques. Finally, we address future priorities in the field of CSIA including: generating more data across a wider range of taxa; lowering costs and increasing laboratory availability; working across disciplinary and methodological boundaries; and combining approaches to answer macroevolutionary questions. Determining the transfer and transformation of organic matter in food webs is a fundamental challenge that has implications for sustainable management of ecosystems. Fatty acids (FA) offer a potential approach for resolving complex diet mixtures of organisms because they provide a suite of molecular tracers. Yet, uncertainties in the degree of their biochemical modification by consumers, due to selective retention or metabolism, have limited their application. Here, we consolidated 316 controlled feeding studies of aquatic ectotherms (fishes and invertebrates) involving 1404 species–diet combinations to assess the degree of trophic modification of FA in muscle tissue. We found a high degree of variability within and among taxa in the %FA in consumer muscle tissue versus %FA in diet regression equations. Most saturated FA had weak relationships with the diet ( r 2 < 0.30) and shallow slopes ( m < 0.30), suggesting a lack of retention in muscle when fed in increasing amounts. Contrarily, several essential FA, including linoleic (18:2n-6) and α-linolenic acid (18:3n-3), exhibited significant relationships with the diet ( m > 0.35, r 2 > 0.50), suggesting supply limitations and selective retention in muscle by consumers. For all FA, relationships strengthened with increasing taxonomic specificity. We also demonstrated the utility of new correction equations by calculating the potential contributions of approximately 20 prey items to the diet of selected species of generalist fishes using a FA mixing model. Our analyses further reveal how a broad range of fishes and invertebrates convert or store these compounds in muscle tissue to meet physiological needs and point to their power in resolving complex diets in aquatic food webs. The electronic surroundings of phosphorus and lithium atoms in the ionic conductor lithium dihydrogen phosphate (LDP) have been studied by single-crystal nuclear magnetic resonance (NMR) spectroscopy at room temperature. From orientation-dependent NMR spectra of a large homegrown LDP single crystal, the full 31P chemical shift (CS) and 7Li quadrupole coupling (QC) tensor was determined, using a global fit over three rotation patterns. The resulting CS tensor is characterized by its three eigenvalues: &delta, 11 P A S = ( 67.0 &plusmn, 0.6 ) ppm, &delta, 22 P A S = ( 13.9 &plusmn, 1.5 ) ppm, and &delta, 33 P A S = ( &minus, 78.7 &plusmn, 0.9 ) ppm. All eigenvalues have also been verified by magic-angle spinning NMR on a polycrystalline sample, using Herzfeld&ndash, Berger analysis of the rotational side band pattern. The resulting 7Li QC tensor is characterized by its quadrupolar coupling constant &chi, = Q 33 P A S = ( &minus, 71 &plusmn, 1 ) kHz and the two eigenvalues Q 11 P A S = ( 22.3 &plusmn, 0.9 ) kHz, and Q 22 P A S = ( 48.4 &plusmn, 0.8 ) kHz. The initially unknown orientation of the mounted crystal, expressed by the orientation of the rotation axis in the orthorhombic crystal frame, was included in the global data fit as well, thus obtaining it from NMR data only.

The established industrial processes used for the growth of TiCxN1-x coatings by chemical vapor deposition (CVD) suffer from substantial limitations, either in regard of brittle phase formation or restriction in the C/(C + N) ratio. Using the alternative C precursor C2H6 allows to overcome these issues. Thus, within this work, the microstructure, phase composition, micro-mechanical and thermo-physical properties of CVD TiCxN1-x coatings grown with C2H6 were investigated. Through adjustment of the C2H6 and N2 flow in the feed gas, the C/(C + N) ratio in the coatings was varied between pure TiN and TiC0.80N0.20. All coatings are characterized by a single-phase face centered cubic structure. The 〈110〉 fiber texture present in all coatings becomes more pronounced with increasing C content. None of the investigated coatings showed thermal cracks on the surface. The thermal conductivity decreases with addition of C from 45 ± 5 W/mK in TiN to 32 ± 3 W/mK in all ternary TiCxN1-x coatings. TiC0.47N0.53 exhibits the highest hardness (30.0 ± 1.4 GPa), while TiC0.63N0.36 turned out as the stiffest coating with a Young's modulus of 576 ± 23 GPa. The fracture stress sF and toughness KIC are superior in coatings with moderate C and N content, with TiC0.63N0.37 being the strongest (sF = 7.7 ± 0.4 GPa) and TiC0.47N0.53 (KIC = 4.4 ± 0.3 MPa m1/2) the toughest within this series. Coatings with moderate to high C content were found to exhibit a microstructure provoking a lower thermal conductivity and improved mechanical properties compared to those with a low C/(C + N) ratio.

Hard protective coatings are commonly subjected to temperatures exceeding 1000 °C, which has significant influence on their thermo-physical properties and the associated performance in application. Within the present work, temperature dependent physical properties of coatings within the Ti(B,N) system grown by chemical vapor deposition were correlated with their chemical composition. High-energy X-ray diffraction experiments in inert atmosphere proved that TiN, TiB2 and ternary TiBxNy coatings with varying B contents are thermally stable up to 1000 °C. In-plane strains of TiN and TiBxNy coatings diminish during heating, whereas TiB2 exhibits compressive strain enhancement up to the deposition temperature. Nanocrystalline TiB2 exhibits more pronounced grain growth during annealing compared to coarse grained columnar TiN. Within the investigated coatings, the mean thermal expansion coefficient decreases as the B content increases. The same trend was observed for the thermal conductivity, which correlates with the grain size of the coatings.

The aim of the present work was to develop a new scalable and cost-efficient process to isolate bovine immunoglobulin G from colostral whey with high purity and minimal loss of activity. The mixed mode material Mercapto-Ethyl-Pyridine-Hypercel™ was identified appropriate for direct capture of immunoglobulin G. The binding mechanism is primarily based on hydrophobic interactions at physiological conditions. As compared to immunoglobulin G, all other low molecular whey proteins such as α-Lactalbumin or β-Lactoglobulin, except lactoperoxidase, are more hydrophilic and were therefore found in the flow-through fraction. In order to remove lactoperoxidase as an impurity the column was combined in series with a second mixed mode material (Capto™- with N-benzoyl-homocysteine as ligand) using the same binding conditions. At pH 7.5 the carboxyl group of this ligand is negatively charged and can hence bind the positively charged lactoperoxidase, whose isoelectric point is at pH 9.6. After sample application, the columns were eluted separately. By combining the two columns it was possible to obtain immunoglobulin G with a purity of >96.1% and yield of 65-80%. The process development was carried out using 1 mL columns and upscaling was performed in three steps up to a column volume of 8800 mL for the Hypercel™ column and 3000 mL for the Capto™- column. At this scale it is possible to obtain 130-150 g pure immunoglobulin G from 3 L colostrum within five hours, including the regeneration of both columns. Additionally, the impact of freeze-drying on the isolated immunoglobulin G was studied. The nativity of the freeze dried immunoglobulin was above 95%, which was proven by reversed phase liquid chromatography and validated by differential scanning calorimetry. The activity of immunoglobulin G was preserved over the isolation process and during drying as measured by enzyme-linked immunosorbent assay. In conclusion, by applying the proposed isolation process, it becomes feasible to obtain pure, active and stable imunnunoglobulin G at large scale.

Objective Aim of this study was to analyze whether children with objectively measured second‐hand cigarette smoke (SHS) exposure suffer from a more severe course of disease when hospitalized with lower respiratory tract infection (LRTI) due to respiratory syncytial virus (RSV). Methods This prospective study was conducted at the Department of Pediatrics, Wilhelminen‐Hospital, Vienna, Austria in children aged below 1 year without a history of preceding lung disease and with acute symptoms of LRTI and a positive nasopharyngeal swab for RSV. On admission, urinary cotinine was measured as a marker of recent SHS and clinical severity of LRTI was assessed by oxygen saturation SpO2 and the “admission clinical severity score” (CSSA). Parents/caregivers were asked to complete a customized questionnaire assessing risks for SHS and demographic characteristics. Results After inclusion of 217 patients, data of 185 patients with a mean (SD) age of 106 days (80) were analyzed. Twenty‐five patients (13.5%) were “cotinine‐positive” (COT+) defined as a urinary cotinine level of ≥7 μg/L. SpO2 on admission was significantly lower in children recently exposed to SHS defined objectively by COT+ (94.8% ±2.0) in urine on admission compared to children not recently exposed (COT−) (96.8% ±3.0; P

Aquatic macroinvertebrates play an important functional role in energy transfer in food webs, linking basal food sources to upper trophic levels that include fish, birds, and humans. However, the trophic coupling of nutritional quality between macroinvertebrates and their food sources is still poorly understood. We conducted a field study in subalpine streams in Austria to investigate how the nutritional quality (measured by long‐chain polyunsaturated fatty acids, LC‐PUFAs) in macroinvertebrates changes relative to their basal food sources. Samples of macroinvertebrates, periphyton, and leaves were collected from 17 streams in July and October 2016 and their fatty acid (FA) composition was analyzed. Periphyton FA varied strongly with time and space, and their trophic effect on macroinvertebrate FA differed among functional feeding groups. The match between periphyton FA and macroinvertebrate FA decreased with increasing trophic levels, but LC‐PUFA content increased with each trophic step from periphyton to grazers and finally predators. Macroinvertebrates fed selectively on, assimilated, and/or actively controlled their LC‐PUFA, especially eicosapentaenoic acid (EPA, 20 : 5ω3) relative to their basal food sources in the face of spatial and temporal changes. Grazer FA profiles reflected periphyton FA with relatively good fidelity, and especially their EPA feeding strategy was primarily linked to periphyton FA variation across seasons. In contrast, shredders appeared to preferentially assimilate more EPA over other FA, which was determined by the availability of high‐quality food over seasons. Predators may more actively control their LC‐PUFA distribution with respect to different quality foods and showed less fidelity to the basal FA profiles in plants and prey. Overall, grazers and shredders showed relatively good fidelity to food FA profiles and performed as both “collectors” and “integrators” for LC‐PUFA requirements across seasons, while predators at higher trophic levels were more “integrators” with added metabolic complexity leading to somewhat more divergent FA profiles. These results are potentially applicable for other aquatic consumers in freshwater and marine ecosystems.

Summary Belatacept is a second-generation cytotoxic T lymphocyte antigen (CTLA)-4 immunoglobulin (Ig) fusion protein approved for immunosuppression in renal transplant recipients. It was designed intentionally to interrupt co-stimulation via CD28 by binding to its ligands B7·1 and B7·2. Experimental evidence suggests a potential additional mechanism for CTLA-4 Ig compounds through binding to B7 molecules expressed on antigen-presenting cells (APCs) and up-regulation of indoleamine 2,3-dioxygenase (IDO), an immunomodulating enzyme that catalyzes the degradation of tryptophan to kynurenine and that down-regulates T cell immunity. So far it remains unknown whether belatacept up-regulates IDO in transplant recipients. We therefore investigated whether belatacept therapy enhances IDO activity in liver transplant recipients enrolled in a multi-centre, investigator-initiated substudy of the Phase II trial of belatacept in liver transplantation (IM103-045). Tryptophan and kynurenine serum levels were measured during the first 6 weeks post-transplant in liver transplant patients randomized to receive either belatacept or tacrolimus-based immunosuppression. There was no significant difference in IDO activity, as indicated by the kynurenine/tryptophan ratio, between belatacept and tacrolimus-treated patients in per-protocol and in intent-to-treat analyses. Moreover, no evidence was found that belatacept affects IDO in human dendritic cells (DC) in vitro. These data provide evidence that belatacept is not associated with detectable IDO induction in the clinical transplant setting compared to tacrolimus-treated patients.

The phenylpropenoid glycosides rosavin, rosarin, rosin summarised as ‘total rosavins’ are quality indicators of Rhodiola rosea L. Here we report the systematic quantification of these glycosides and the aglycon cinnamyl alcohol in various provenances cultivated homogenously at 1580 m altitude. (I) We examined rhizomes versus roots in cultivation year 9 (20 provenances). Extracts (70% ethanol) contained 0.5–4.3 mg/mL (rhizomes) and 0.4–2.5 mg/mL (roots) total rosavins. The rosavin/cinnamyl alcohol ratio (average rhizomes 8.1:1, roots 6.2:1), was more influenced by plant origin than plant part. We additionally analysed salidroside including its aglycon tyrosol. The rosavins/salidroside ratios (including aglycons) of 6.4:1 (rhizome) and 5.5:1 (root) indicated authentic R. rosea independent of plant part and origin. (II) Rhizomes extracts (8 provenances) from various dates during years 6 and 7 contained 0.4–4.8 mg/mL total rosavins corresponding to 0.2–3.0% in the dry drug. Age (decreasing trend) and provenance impact were superposed by seasonal variation with a phenylpropenoid peak in spring. Rosavin/cinnamyl alcohol ratios in North European plants were 3–8:1, for Alpine/Pyrenean plants 9–14:1. (III) Compared to our previous dataset from cultivation in South England, on average, there was no major difference between phenylpropenoid values from cultivation at 65 m and 1580 m apart from moderately more cinnamyl alcohol at higher altitude. As per provenance, absolute values were site dependent but phenylpropenoid glycoside/aglycon ratios consistent. In conclusion, this is the first systematic investigation for a high altitude cultivation of R. rosea demonstrating the major extent plant part, season and age influence absolute phenylpropenoid values. In contrary, relative phenolic compound profiles are mostly origin-determined, independent of cultivation conditions and site, and could be used for authentication in quality control.

Freshwater salmonids gain increasing popularity as diet fish and are an important dietary source of omega-3 polyunsaturated fatty acids (PUFA) for humans. The aim of this study was to investigate effects of alternative feeds from domestic sources on fish growth and lipid composition of the sparctic Alsatian charr (Salvelinus alpinus X fontinalis), commonly used in freshwater aquaculture in Europe. In this fish feeding experiment, we used a control feed (32% fish meal; feed F1) and experimental diets containing generally only half the amount of fish meal, which was replaced in equal parts by poultry (F2), two different pork by-product meals (F3 and F4), plant-based feeds (F5), or by a feed containing 25% fish and 15% poultry meal (F6) that had a similar caloric value compared to the other feeds. Six hundred charrs of similar initial weight (ca. 90 g) were randomly distributed into 12 tanks (50/tank) of 1.4 m3, supplied with subalpine spring water, and fed one of the feeds. Fish biomass development was the highest in fish fed diet F6, followed by F1, F2, and F5. Pork by-product meals as partial replacement of fish meal resulted on average in a 25% lower biomass gain compared to charr feeding on poultry by-product meal (F6). The use of poultry or pork by-product meals as partial fish meal replacements did not significantly change the total lipids or fatty acids retained in these fish. This study shows that these alternative feeds, with similar lipid sources, had no significant effect on the fatty acid composition in Alsatian charr, however, poultry by-product meal as partial replacement of marine fish meal clearly enhanced charr biomass by 15% relative to conventional fish meals.

CrTaN coatings grown by cathodic arc evaporation have recently been reported to exhibit high hardness, good thermal stability and promising tribological properties. Despite the known grain refining and thus hardening effect of boron on transition metal nitrides, detailed studies on quaternary CrTaBN coatings are lacking in the literature. Thus, within this work, Cr0.69Ta0.20B0.11N coatings were grown by cathodic arc evaporation and the effect of the applied bias voltage on the microstructure and mechanical properties of the coatings was investigated. All coatings were grown on cemented carbide substrates and crystallize in an fcc-CrxTa1-xN solid solution without indication for additional crystalline boron containing compounds. Increasing the negative bias voltage results in a decreased surface roughness, higher compressive residual stress and improved hardness. Starting at ~1200 °C, the powdered fcc-CrxTa1-xN solid solution decomposes into h-Cr2N and h-Ta5N4, followed by the formation of h-Ta2N and bcc-Cr at 1400 °C. An annealing treatment at 1100 °C for 15 min has no significant influence on the phase composition of the coatings on cemented carbide substrates. Annealing at 1200 °C however induces a reaction between the coating and the substrate. As a result, fcc-TaC and bcc-Cr form, which is accompanied by loss of nitrogen and deterioration of the mechanical properties.

Ti(Al)SiN protective hard coatings are promising candidates for the metal cutting industry, due to their favorable properties such as a high hardness and good oxidation resistance. Within the scope of this work, the microstructure of cathodic arc evaporated TiN, Ti84Si16N, Ti81Al3Si16N and Ti70Al14Si16N was correlated with their mechanical properties. X-ray diffraction (XRD) as well as scanning electron microscopy investigations of the cross-sections revealed a pronounced grain refinement for Ti(Al)SiN compared to TiN. The texture of the coatings was analyzed by XRD pole-figures, showing a change in the preferred orientation in growth direction from (111) for TiN to (200) for Ti(Al)SiN. Hardness values were found to strongly increase from 29 GPa to 40 GPa upon Si addition, but remained constant for further alloying with Al. Micromechanical bending tests allowed to gain a detailed insight into the fracture properties of the investigated coatings and revealed an enhanced fracture stress and fracture toughness (KIC) for Ti(Al)SiN compared to TiN. Hereby, the KIC value of 2.4 ± 0.3 MPa m1/2 for TiN could be increased to a maximum of 3.3 ± 0.2 MPa m1/2 for Ti70Al14Si16N. The present findings provide a systematic overview of the microstructure and mechanical properties of arc evaporated Ti(Al,Si)N coatings, thus allowing to select the most suitable coatings for different applications in the cutting industry.

The bioaccumulation of mercury (Hg) and polyunsaturated fatty acids (PUFA) in zoobenthos varies across aquatic food webs. In this field study, contents of total Hg (THg), methylmercury (MeHg) and PUFA were investigated in zoobenthos of Chinese reservoirs and Swedish lakes, with contrasting environmental characteristics and algal diet sources, which can result in difference of Hg and PUFA in zoobenthos from these two habits. Using PUFA as dietary biomarkers of algae in zoobenthos, we evaluated effects of environmental factors and algal diet sources on the accumulation of THg, MeHg, and the highly required PUFA eicosapentaenoic (EPA, 20: 5n-3) and docosahexaenoic acid (DHA, 22:6n-3) in zoobenthos. Average THg and MeHg in zoobenthos were higher in Chinese reservoirs than Swedish lakes (p0.05). Average EPA content of zoobenthos was similar in these two habitats (p0.05), yet average DHA content of zoobenthos was higher in Chinese reservoirs than Swedish lakes (p0.05). Total Hg and MeHg contents of zoobenthos in Swedish lakes were predicted by environmental factors; e.g., negatively with pH and positively with dissolved organic carbon (DOC) concentrations, yet had no significant relationship with the algal dietary. In Chinese reservoirs, however, no environmental factor correlated well with THg contents in zoobenthos, and only DOC concentrations showed positive correlation with MeHg contents in zoobenthos. Besides, the algal dietary was also positively correlated with MeHg contents in zoobenthos. EPA and DHA contents of zoobenthos in Swedish lakes primarily associated with algal diet. By contrast, in Chinese reservoirs, EPA and DHA contents of zoobenthos were affected by both environmental factors and algal diet.

1. Streams and rivers provide crucial sources of long-chain polyunsaturated fatty acids (LC-PUFA, a subset of PUFA with ≥20 C in their acyl chains) for freshwater and terrestrial animals. LC-PUFA are primarily produced by particular algal taxa and are subsequently transferred to fish and higher consumers, including humans. Although research on the trophic transfer of LC-PUFA in streams is very limited, studies on the PUFA composition of organisms at various trophic levels within stream food webs are well documented. Here, we systematically analyze the literature to synthesise LC-PUFA distribution and retention in stream ecosystems. 2. In general, stream food webs are highly enriched in omega-3 LC-PUFA, in particular eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3). Algae from temperate rivers had more EPA compared with algae from subtropical rivers. Stream invertebrates from both temperate and subtropical rivers preferentially retain algal EPA and their PUFA tend to vary with algal PUFA. Invertebrates may be capable of regulating their fatty acid (FA) composition, but this ability is limited, to the extent that they must obtain physiologically important LC-PUFA primarily from algae. 3. DHA is selectively and highly retained in all fish taxa irrespective of their feeding sites (i.e. temperate or subtropical rivers). However, DHA content is variable among stream fish taxa and is likely linked to resource utilisation and their endogenous metabolism. Freshwater salmonids have a very high DHA content, which suggests they preferentially retain this FA or biosynthesise it from EPA. The PUFA profiles of other fish taxa more closely reflect their dietary PUFA supply. However, so far very few studies have considered the differences in the PUFA biosynthesis of fish taxa in rivers or compared the dietary PUFA effect on different fish taxa. 4. The supply of LC-PUFA from streams and rivers is known to be affected by altered light availability, greater nutrient inputs and other anthropogenic activities, and any decrease in algal LC-PUFA production will negatively affect the consumers that are nutritionally dependent on these molecules. Further investigations are needed to understand how environmental changes affect LC-PUFA production and retention in stream ecosystems, and how to protect streams from threats to this essential ecosystem service.

Polyunsaturated fatty acids (PUFA), especially long-chain (i.e., ≥20 carbons) polyunsaturated fatty acids (LC-PUFA), are fundamental to the health and survival of marine and terrestrial organisms. Therefore, it is imperative that we gain a better understanding of their origin, abundance, and transfer between and within these ecosystems. We evaluated the natural variation in PUFA distribution and abundance that exists between and within these ecosystems by amassing and analyzing, using multivariate and analysis of variance (ANOVA) methods, >3000 fatty acid (FA) profiles from marine and terrestrial organisms. There was a clear dichotomy in LC-PUFA abundance between organisms in marine and terrestrial ecosystems, mainly driven by the C18 PUFA in terrestrial organisms and omega-3 (n-3) LC-PUFA in marine organisms. The PUFA content of an organism depended on both its biome (marine vs terrestrial) and taxonomic group. Within the marine biome, the PUFA content varied among taxonomic groups. PUFA content of marine organisms was dependent on both geographic zone (i.e., latitude, and thus broadly related to temperature) and trophic level (a function of diet). The contents of n-3 LC-PUFA were higher in polar and temperate marine organisms than those from the tropics. Therefore, we conclude that, on a per capita basis, high latitude marine organisms provide a disproportionately large global share of these essential nutrients to consumers, including terrestrial predators. Our analysis also hints at how climate change, and other anthropogenic stressors, might act to negatively impact the global distribution and abundance of n-3 LC-PUFA within marine ecosystems and on the terrestrial consumers that depend on these subsidies.

The functional properties of whey protein isolate (WPI) based films and coatings have been the subject of several studies. However, quantitative information on the molecular interactions is rare. The solubility studies presented here allow qualitative statements to be made about the molecular interactions in WPI-based films. The other objective of this study was to determine the cross-linking density (CLD). Swelling studies were performed on WPI-based films containing different amounts of denatured WPI. The results of the swelling studies showed that the degree of denaturation has a significant influence on the CLD, which is directly proportional to the number of disulfide bonds in the WPI-based network. As a result, the structural stability of the polymer network was improved. The swelling studies showed that there was a significant linear increase in CLD of 1.17·10 −4 mol cm −3 on going from 50% to 100% denatured WPI-based films (0.22·10 −4 mol cm −3 → 1.39·10 −4 mol cm −3 ). The solubility study showed that WPI film solubility is mainly influence by non-covalent bonds up to a degree of denaturation of 75%. In general, there was a correlation between CLD and the mechanical properties of the films. The Younǵs modulus increased by 320% on going from 25% to 100% denatured WPI-based films (26.70 MPa → 85.30 MPa). The CLD had only a minor effect on the barrier properties of the WPI-based films. Thus, this scientific paper provides new knowledge for researchers and material developers because qualitative information about the covalent intermolecular interactions in whey protein isolate based films and coatings has been obtained for the first time.

Fatty acids in muscle tissue and eggs of female Atlantic salmon Salmo salar spawners were analysed to evaluate the dietary quality of their final feeding areas in the Baltic Sea. The final likely feeding area was identified by comparing stable carbon and nitrogen isotope composition of the outermost growth region (final annulus) of scales of returned S. salar with that of reference S. salar caught from different feeding areas. Some overlap of stable-isotope reference values among the three areas, in addition to prespawning fasting, decreased the ability of muscle tri-acylglycerols to discriminate the final likely feeding area and the area's dietary quality. Among three long-chained polyunsaturated fatty acids, docosahexaenoic acid (DHA; 22:6n-3), eicosapentaenoic acid (EPA; 20:5n-3) and arachidonic acid (ARA; 20:4n-6), the proportions of ARA in total lipids of spawning S. salar muscle and eggs showed a significant negative correlation with increasing probability of S. salar having returned from the Baltic Sea main basin (i.e. the Baltic Sea proper). The results suggest that ARA in muscle and eggs is the best dietary indicator for dietary characteristics of final marine feeding area dietary characteristics among S. salar in the Baltic Sea.

Acute kidney injury (AKI) remains a major clinical event with high mortality rates. We previously identified renal miR-182 as the main driver of post-transplantation AKI. Therefore, we tested the causal inference of miR-182 by inhibiting its renal expression in vivo. In 45 rats AKI was induced by right nephrectomy and contralateral clamping of the renal pedicle for 40 minutes. Systemically administered antisense oligonucleotide (ASO) inhibited miR-182 in the kidneys up to 96 hours. The maximum creatinine elevation was on day 2 after injury (mg/dL; median and interquartile range): ASO 2.5mg/kg: 1.9 (1.3; 3.2), ASO 25mg/kg: 2.8 (0.7; 5.0), mismatch oligonucleotide (MM) 25mg/kg: 5.7 (5,0; 5.8), saline: 4.4 (3.5; 5.8) (P = 0.016, analysis of variance). Blinded semiquantitative histologic evaluation of renal biopsies showed better preserved morphology in both ASO groups than saline- and MM-treated kidneys (median and interquartile range of overall injury scores): ASO both concentrations 1 (1, 1), saline 3 (3, 3) and MM 3 (3, 3) (P0.001, analysis of variance). ASO facilitated cell proliferation, metabolism, and angiogenesis on a genome-wide level. ASO when applied in normothermic kidney machine perfusion reduced renal miR-182 expression by more than two magnitudes. In summary, we showed that in vivo inhibition of miR-182 by ASO improved kidney function and morphology after AKI. This technique may be applicable to reduce the high rate of AKI in the human renal transplantation setting.

Algae are higher-quality food resources than allochthonous plant litter for stream invertebrates, in part, because of their higher content of polyunsaturated fatty acids (PUFAs). We tested the hypothesis that algal biofilms on the surfaces of leaf litter improve the nutritional quality of terrestrial inputs for invertebrate shredders. We used a laboratory feeding experiment with 2 light levels (open and shaded) and 2 nutrient regimes (ambient and enriched) to manipulate the algal biofilms that form on leaf surfaces (Lophostemon confertus). We assessed how these treatments affected the fatty acid (FA) composition of these biofilms and the somatic growth of a stream invertebrate shredder (Anisocentropus bicoloratus, Trichoptera) that feeds on the conditioned leaf litter. Shredders reached a significantly larger size when nutrients were added, and leaf mass loss was significantly greater in these treatments than in treatments without nutrients. Aquaculture can affect the polyunsaturated fatty acids (PUFA) and mercury (Hg) in fish by altering their diet. Here, planktivorous (silver carp and bighead carp), omnivorous and carnivorous fish with different dietary strategies were selected from two reservoirs, one with on-going aquaculture (WJD) and another without aquaculture (HF) in Southwest China. We compared the total mercury (THg), methylmercury (MeHg) contents and PUFA profiles of fish and their potential diets in these two reservoirs. THg and MeHg contents in omnivorous and carnivorous fish were lower from the WJD Reservoir, which is related to the lower THg and MeHg contents in the artificial fish food. THg and MeHg contents in silver carp from the WJD Reservoir were lower than those from the HF Reservoir, while they were similar in bighead carps from the two reservoirs. The Hg variation in planktivorous fish were inconsistent with that in plankton. THg contents in phyto- and zooplankton from the HF Reservoir were higher than those from the WJD Reservoir, yet their MeHg contents were similar. Artificial fish food which contained higher total PUFA eicosapentaenoic (EPA; 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), significantly increased the total PUFA and EPA + DHA contents in carnivorous fish, but had less effect on that in omnivorous fish from the WJD Reservoir. Eutrophication caused by aquaculture reduced total PUFA and EPA + DHA contents of plankton in WJD, yet did not reduce those in planktivorous fish. The impacts of aquaculture on Hg and PUFA accumulated in fish were varied among different fish species, and the mechanism needs further exploration.

We present a displacement-sensitive sensor comprising a microelectromechanical (MEMS) chip and organic optoelectronic components capable of measuring the heart and respiration rate on humans. The MEMS sensor relies on the inertial deflection of a small silicon oscillator. The readout of the deflection is optical and works via modulation of the light flux passing through the MEMS. Organic optoelectronics are used as light source and detector, since these offer a homogeneous light distribution and a more compact package in a future integration. Two types of MEMS, differing in their resonance frequency, were designed and characterised in combination with both organic and inorganic optoelectronics prior to measuring heart and respiration rate. Subsequently, by measurements on the neck, pulse and respiration rate were successfully measured.

This paper reports on a MEMS gradiometer consisting of two independent laterally oscillating masses on a single chip with integrated optical readout. The symmetrical design of the two masses offers high accuracy and low cost by using conventional MEMS batch fabrication technology. The sensing principle is based on laterally displacement of the masses actuated by Lorentz forces which modulates a light flux passing through a stationary mask and the moving mask integrated in the masses. Phase and intensity detected by photodiodes reveals information about the uniformity of an external applied magnetic field, hence, enables the measurement of gradient-, homogeneous-and offset zradient maanetic fields. A quadrupol magnetic field created from four identical neodymium bar magnets had been characterised to demonstrate the sensor's working principle.
