Understanding the relationship between incursions of insect pests and established populations is critical to implementing effective control. Studies of genetic variation can provide powerful tools to examine potential invasion pathways and longevity of individual pest outbreaks. The major fruit fly pest in eastern Australia, Queensland fruit fly Bactrocera tryoni (Froggatt), has been subject to significant long‐term quarantine and population reduction control measures in the major horticulture production areas of southeastern Australia, at the species southern range limit. Previous studies have employed microsatellite markers to estimate gene flow between populations across this region. In this study, we used an independent genetic marker, mitochondrial DNA (mtDNA) sequences, to screen genetic variation in established and adjacent outbreak populations in southeastern Australia. During the study period, favorable environmental conditions resulted in multiple outbreaks, which appeared genetically distinctive and relatively geographically localized, implying minimal dispersal between simultaneous outbreaks. Populations in established regions were found to occur over much larger areas. Screening mtDNA (female) lineages proved to be an effective alternative genetic tool to assist in understanding fruit fly population dynamics and provide another possible molecular method that could now be employed for better understanding of the ecology and evolution of this and other pest species.

Tramps ants are a constant biosecurity threat to Australia. Red Imported Fire Ant Solenopsis invicta was detected in Sydney, New South Wales in 2014, and a successful eradication program was undertaken. Stratified surveillance was conducted as part the program, and allowed for scenario tree analysis to statistically support the claim of freedom. Our analysis demonstrated a mean probability of freedom was 93.1%, 99.5% and > 99.9% after one, two or three surveys for two nests within 2 km of the detection. We demonstrated > 99.9% probability of freedom after three surveys if for a threshold value of five nests were present. Our analysis was the basis of declaring Sydney free from S. invicta. Sydney was declared free 18 months after the first detection of S. invicta.

DNA polymerases Pol ε and Pol δ replicate DNA during cell division. However, in some cancers, defective proofreading due to acquired POLE/POLD1 exonuclease domain mutations causes markedly elevated somatic mutation burdens with distinctive mutational signatures. Germline POLE/POLD1 mutations cause familial cancer predisposition. Here, we sequenced normal tissue and tumor DNA from individuals with germline POLE/POLD1 mutations. Increased mutation burdens with characteristic mutational signatures were found in normal adult somatic cell types, during early embryogenesis and in sperm. Thus human physiology can tolerate ubiquitously elevated mutation burdens. Except for increased cancer risk, individuals with germline POLE/POLD1 mutations do not exhibit overt features of premature aging. These results do not support a model in which all features of aging are attributable to widespread cell malfunction directly resulting from somatic mutation burdens accrued during life.

Tephritid fruit flies are major economic pests for fruit production and are an impediment to international trade. Different host fruits are known to vary in their suitability for fruit flies to complete their life cycle. Currently, international regulatory standards that define the likely legal host status for tephritid fruit flies categorize fruits as a natural host, a conditional host, or a nonhost. For those fruits that are natural or conditional hosts, infestation rate can vary as a spectrum ranging from highly attractive fruits supporting large numbers of fruit flies to very poor hosts supporting low numbers. Here, we propose a Host Suitability Index (HSI), which divides the host status of natural and conditional hosts into five categories based on the log infestation rate (number of flies per kilogram of fruit) ranging from very poor (100). Infestation rates may be determined by field sampling or cage infestation studies. We illustrate the concept of this index using 21 papers that examine the host status of fruits in five species of polyphagous fruit flies in the Pacific region: Bactrocera tryoni Froggatt, Bactrocera dorsalis (Hendel), Bactrocera latifrons (Hendel), Zeugodacus cucurbitae (Coquillett), and Ceratitis capitata (Wiedemann) (Diptera: Tephritidae). This general-purpose index may be useful in developing systems approaches that rely on poor host status, for determining surveillance and detection protocols for potential incursions, and to guide the appropriate regulatory response during fruit fly outbreaks.

The mammalian intestine is colonized by trillions of microorganisms that have co-evolved with the host in a symbiotic relationship. Although the influence of the gut microbiota on intestinal physiology and immunity is well known, mounting evidence suggests a key role for intestinal symbionts in controlling immune cell responses and development outside the gut. Although the underlying mechanisms by which the gut symbionts influence systemic immune responses remain poorly understood, there is evidence for both direct and indirect effects. In addition, the gut microbiota can contribute to immune responses associated with diseases outside the intestine. Understanding the complex interactions between the gut microbiota and the host is thus of fundamental importance to understand both immunity and human health.

Introduction: Rapid diagnosis of M. tuberculosis directly from sputum samples is a challenging process. This study aimed to design and evaluate a multiplex-PCR method for direct diagnosis of M. tuberculosis from sputum specimens. Materials and Methods: 46 suspected tuberculosis patients and 25 apparently healthy individuals were enrolled in the study. Sputa were collected from the study population and processed by cold ZN stain. DNA was extracted from each sample and processed by Multiplex PCR and Genotype Mycobacteria CM. Results: Out of the 46 Tuberculosis suspected patients, 22 (47.8%) revealed positive Acid fast ba- cilli (AFB), while 19 (41.3%) showed positive by both multiplex PCR and Genotype Mycobacte- ria CM. The overall sensitivity of multiplex PCR and smear microscopy were 100% while the specificity were 100, and 86.3%, respectively. Conclusion: Multiplex PCR method using two different sets of primers in combination with other diagnostic tools such as X-Rays and smear Microscopy are cheap, rapid and reliable methods for the diagnosis of M. tuberculosis from clinical samples and are able to identify most of the smear positive cases with valuable accuracy.

The CD34 cell surface antigen is widely expressed in tissues on cells with progenitor-like properties and on mature vascular endothelia. In adult human bone marrow, CD34 marks hematopoietic stem and progenitor cells (HSPCs) starting from the bulk of hematopoietic stem cells with long-term repopulating potential (LT-HSCs) throughout expansion and differentiation of oligopotent and unipotent progenitors. CD34 protein surface expression is typically lost as cells mature into terminal effectors. Because of this expression pattern of HSPCs, CD34 has had a central role in the evaluation or selection of donor graft tissue in HSC transplant (HSCT). Given its clinical importance, it is surprising that the biological functions of CD34 are still poorly understood. This enigma is due, in part, to CD34's context-specific role as both a pro-adhesive and anti-adhesive molecule and its potential functional redundancy with other sialomucins. Moreover, there are also critical differences in the regulation of CD34 expression on HSPCs in humans and experimental mice. In this review, we highlight some of the more well-defined functions of CD34 in HSPCs with a focus on proposed functions most relevant to HSCT biology.

ObjectiveSporadic early-onset colorectal cancer (EOCRC) has bad prognosis, yet is poorly represented by cell line models. We examine the key mutational and transcriptomic alterations in an organoid biobank enriched in EOCRCs.DesignWe established paired cancer (n=32) and normal organoids (n=18) from 20 patients enriched in microsatellite-stable EOCRC. Exome and transcriptome analysis was performed.ResultsWe observed a striking diversity of molecular phenotypes, including PTPRK-RSPO3 fusions. Transcriptionally, RSPO fusion organoids resembled normal colon organoids and were distinct from APC mutant organoids, with high BMP2 and low PTK7 expression. Single cell transcriptome analysis confirmed the similarity between RSPO fusion organoids and normal organoids, with a propensity for maturation on Wnt withdrawal, whereas the APC mutant organoids were locked in progenitor stages. CRISPR/Cas9 engineered mutation of APC in normal human colon organoids led to upregulation of PTK7 protein and suppression of BMP2, but less so with an engineered RNF43 mutation. The frequent co-occurrence of RSPO fusions with SMAD4 or BMPR1A mutation was confirmed in TCGA database searches. RNF43 mutation was found in organoid from a leukaemia survivor with a novel mutational signature; and organoids with POLE proofreading mutation displayed ultramutation. The cancer organoid genomes were stable over long culture periods, while normal human colon organoids tended to be subject to clonal dominance over time.ConclusionsThese organoid models enriched in EOCRCs with linked genomic data fill a gap in existing CRC models and reveal distinct genetic profiles and novel pathway cooperativity.

The centromere is a specialized region on the chromosome that directs equal chromosome segregation. Centromeres are usually not defined by DNA sequences alone. How centromere formation and function are determined by epigenetics is still not fully understood. Active centromeres are often marked by the presence of centromeric-specific histone H3 variant, centromere protein A (CENP-A). How CENP-A is assembled into the centromeric chromatin during the cell cycle and propagated to the next cell cycle or the next generation to maintain the centromere function has been intensively investigated. In this review, we summarize current understanding of how post-translational modifications of CENP-A and other centromere proteins, centromeric and pericentric histone modifications, non-coding transcription and transcripts contribute to centromere function, and discuss their intricate relationships and potential feedback mechanisms.

The mammalian intestine is colonized by trillions of microorganisms that have co-evolved with the host in a symbiotic relationship. The presence of large numbers of symbionts near the epithelial surface of the intestine poses an enormous challenge to the host because it must avoid the activation of harmful inflammatory responses to the microorganisms while preserving its ability to mount robust immune responses to invading pathogens. In patients with inflammatory bowel disease, there is a breakdown of the multiple strategies that the immune system has evolved to promote the separation between symbiotic microorganisms and the intestinal epithelium and the effective killing of penetrant microorganisms, while suppressing the activation of inappropriate T cell responses to resident microorganisms. Understanding the complex interactions between intestinal microorganisms and the host may provide crucial insight into the pathogenesis of inflammatory bowel disease as well as new avenues to prevent and treat the disease.

Cellular DNA damage caused by reactive oxygen species is repaired by the base excision repair (BER) pathway which includes the DNA glycosylase MUTYH. Inherited biallelic MUTYH mutations cause predisposition to colorectal adenomas and carcinoma. However, the mechanistic progression from germline MUTYH mutations to MUTYH-Associated Polyposis (MAP) is incompletely understood. Here, we sequenced normal tissue DNAs from 10 individuals with MAP. Somatic base substitution mutation rates in intestinal epithelial cells were elevated 2 to 5-fold in all individuals, except for one showing a 33-fold increase, and were also increased in other tissues. The increased mutation burdens were of multiple mutational signatures characterised by C>A changes. Different mutation rates and signatures between individuals were likely due to different MUTYH mutations or additional inherited mutations in other BER pathway genes. The elevated base substitution rate in normal cells likely accounts for the predisposition to neoplasia in MAP. Despite ubiquitously elevated mutation rates, individuals with MAP do not display overt evidence of premature ageing. Thus, accumulation of somatic mutations may not be sufficient to cause the global organismal functional decline of ageing.Summary

Introduction: The memory impairment that is characteristic of amnestic mild cognitive impairment (aMCI) is often accompanied by difficulties in executive functioning, including planning. Though planning deficits in aMCI are well documented, their neural correlates are largely unknown, and have not yet been investigated with functional magnetic resonance imaging (fMRI).Objectives: The aim of this study was to: (1) identify differences in brain activity and connectivity during planning between people with aMCI and cognitively healthy older adults, and (2) find whether planning-related activity and connectivity are associated with cognitive performance and symptoms of apathy.Methods: Twenty-five people with aMCI and 15 cognitively healthy older adults performed a visuospatial planning task (Tower of London; ToL) during fMRI. Task-related brain activation, spatial maps of task-related independent components, and seed-to-voxel functional connectivity were compared between the two groups and regressed against measures of executive functions (Trail Making Test difference score, TMT B-A; Digit Symbol Substitution Test, DSST), delayed recall (Rey Auditory Verbal Learning Test), and apathy (Apathy Evaluation Scale).Results: People with aMCI scored lower on task-switching (TMT B-A), working memory (DSST), and planning (ToL). During planning, people with aMCI had less activation in the bilateral anterior calcarine sulcus/cuneus, the bilateral temporal cortices, the left precentral gyrus, the thalamus, and the right cerebellum. Across all participants, higher planning-related activity in the supplementary motor area, the retrosplenial cortex and surrounding areas, and the right temporal cortex was related to better delayed recall. There were no betweengroup differences in functional connectivity, nor were there any associations between connectivity and cognition. We also did not find any associations between brain activity or connectivity and apathy.Conclusion: Impaired planning in people with aMCI appears to be accompanied by lower activation in a diffuse cortico-thalamic network. Across all participants, higher planning-related activity in parieto-occipital, temporal, and frontal areas was related to better memory performance. The results point to the relevance of planning deficits for understanding aMCI and extend its clinical and neurobiological signature.

Fiji leaf gall has been a major disease in the Australian sugarcane industry, causing extensive crop losses in the 1970–1990 period. The disease has been managed via a number of measures including varietal resistance, disease-free plant sources, termination of heavily-infested crops, quarantine and rogueing of disease stools. These measures have been highly effective and reduced detected disease incidence to zero in several districts. Over 32,000 crop inspections have been undertaken since the last known diseased crop was sighted in the Central Queensland region. All disease inspection data were collated and analysed for this region, with modelling undertaken to determine the probability of the disease being present but remaining undetected. These analyses suggest that it is very highly unlikely to be present; a proposal is made therefore to declare ‘area freedom from Fiji leaf gall’ for the Central Queensland region.

Fluorescent dust marking is widely used in tephritid sterile insect programs to differentiate mass reared sterile individuals from the wild individuals. While fluorescent dust marking is economical, it does not guarantee 100% marking success. In programs releasing millions of sterile flies, even a very low rate of misclassification of sterile individuals (due to a failure of dust marking) could greatly over-estimate the number of resident wild individuals. This could be an important outcome since, in some horticultural production areas, detection of as few as two wild individuals trigger suspension of exports. In this study, we sought to test the accuracy of fluorescent dust marking for the identification of sterile Bactrocera tryoni (Froggatt) during a sterile release technique (SIT) program in New South Wales, Australia. We used DNA microsatellite data to as an independent discriminator between wild from sterile flies. Our results confirm earlier suspicions that a small proportion of sterile flies remain unmarked during these releases. We also found that wild flies can be misclassified as sterile due to the presence of acquired dust grains on the body. These results indicate that any B. tryoni SIT program should not rely on fluorescent dust alone to unambiguously identify sterile released individuals. We show that additional methods, such as carbon isotope ratio measurements, are practical for use in B. tryoni SIT programs.

Following the loss of two key pesticides, dimethoate and fenthion, producers and regulators need to develop new fruit fly management options. A systems approach is a likely measure in the post dimethoate and fenthion era. The risk of fruit fly can be mitigated at the regional, district and farm level. Some measures such as awareness and exclusion are common across all three levels. Pest control measures will be based on an understanding of climate, biology and pesticides. In this present paper, 28 different risk mitigation options that can be used in a systems approach in Australia are discussed.

Highlights • In aMCI, apathy severity was associated with lower FA in widespread WM pathways. • WM aberrations are related to apathy severity after controlling for depression. • Disruptions related to apathy severity are not limited to frontal-subcortical area., Apathy is recognized as a prevalent behavioral symptom of amnestic Mild Cognitive Impairment (aMCI). In aMCI, apathy is associated with an increased risk and increases the risk of progression to Alzheimer’s Disease (AD). Previous DTI study in aMCI showed that apathy has been associated with white matter alterations in the cingulum, middle and inferior longitudinal fasciculus, fornix, and uncinate fasciculus. However, the underlying white matter correlates associated with apathy in aMCI are still unclear. We investigated this relationship using whole-brain diffusion tensor imaging (DTI). Twenty-nine aMCI patients and 20 matched cognitively healthy controls were included. Apathy severity was assessed using the Apathy Evaluation Scale Clinician version. We applied the tract-based spatial statistics analyses to DTI parameters: fractional anisotropy (FA), mean diffusivity, axial diffusivity, and radial diffusivity to investigate changes in white matter pathways associated with the severity of apathy. No significant difference was found in any of the DTI parameters between aMCI and the control group. In aMCI, higher severity of apathy was associated with lower FA in various white matter pathways including the left anterior part of inferior fronto-occipital fasciculus/uncinate fasciculus, genu and body of the corpus callosum, superior and anterior corona radiata, anterior thalamic radiation of both hemispheres and in the right superior longitudinal fasciculus/anterior segment of arcuate fasciculus (p

This paper describes chronic features of neuroangiostrongyliasis (NAS), a long-term outcome of the disease that has not been adequately described. Current and past literature is predominantly limited to acute manifestations of NAS, and mention of chronic, ongoing clinical symptoms is usually limited to brief notes in a discussion of severe cases. This study investigated the long-term outcomes in ten individuals who were diagnosed with acute neuroangiostrongyliasis in Hawaii between 2009 and 2017. The study demonstrates a significant number of persons in Hawaii sustain residual symptoms for many years, including troublesome sensory paresthesia (abnormal spontaneous sensations of skin experienced as ‘burning, pricking, pins and needles’; also described as allodynia or hyperesthesia) and extremity muscle pains. As a consequence, employment and economic hardships, domestic relocations, and psychological impairments affecting personal relationships occurred. The study summarizes common features of chronic disease, sensory paresthesia and hyperesthesia, diffuse muscular pain, insomnia, and accompanying emotional distress; highlights the frequently unsuccessful endeavours of individuals struggling to find effective treatment; proposes pathogenic mechanisms responsible for prolonged illness including possible reasons for differences in disease presentation in Hawaii compared to Southeast Asia.

Larval diets used for artificial rearing can have a significant effect on insect biology. The Queensland fruit fly (aka ‘Qfly’), Bactrocera tryoni (Froggatt) (Diptera: Tephritidae), is one of the greatest challenges for fruit growers in Australia. The sterile insect technique (SIT) is being developed to manage outbreaks in regions that remain free of Qfly and to reduce populations in regions where this species is endemic. Factory scale rearing is essential for SIT; however, artificial larval diets are known to affect the microbiome of Qfly, which may then affect fly performance. In this study, high-throughput Illumina sequencing was used to assess the Qfly microbiome in colonies reared, for five generations from nature, on two common artificial diets (carrot and gel). At generation five (G5), the microbiome was assessed in larvae, pupae, adult males and adult females and standard fly quality control parameters were assessed together with additional performance measures of mating propensity and survival under nutritional stress. At the genus level, bacterial communities were significantly different between the colonies reared on the two larval diets. However, communities converged at Phyla to family taxonomic levels. Interestingly, pathogenic bacterial genera (e.g., Morganella, Citrobacter, Providencia, Burkholderia) were highly abundant in all developmental stages of Qfly reared on the gel diet, when compared to the carrot diet. Despite abundance of these commonly pathogenic genera, a greater percentage of egg hatching, heavier pupal weight and a higher percentage of fliers were found in the Qfly reared on the gel diet. Mating propensity and survival under nutritional stress was similar for adult Qfly that had been reared on the two larval diets. Overall, our findings demonstrate that the artificial larval diet strongly influences the microbiome and quality control measures of Qfly, with likely downstream effects on performance of flies released in SIT programs.

Here, we sequenced normal tissue DNA from individuals with germline POLE or POLD1 exonuclease domain mutations. Increased mutation burdens with characteristic mutational signatures were found to varying extents in all normal adult somatic cell types examined, during early embryogenesis and in sperm. Mutation burdens were further markedly elevated in neoplasms from these individuals. Thus human physiology is able to tolerate ubiquitously elevated mutation burdens. Indeed, with the exception of early onset cancer, individuals with germline POLE and POLD1 exonuclease domain mutations are not reported to show abnormal phenotypic features, including those of premature ageing. The results, therefore, do not support a simple model in which all features of ageing are attributable to widespread cell malfunction directly resulting from somatic mutation burdens accrued during life.

Innate lymphoid cells (ILCs) are a recently identified subset of leukocytes that play a central role in pathogen surveillance and resistance, modulation of immune response, and tissue repair. They are remarkably similar to CD4+ T-helper subsets in terms of function and transcription factors required for their development but are distinguished by their lack of antigen-specific receptors. Despite their similarities, the absence of a surface T-cell receptor (TCR) and presence of ILCs and precursors in adult bone marrow has led to speculation that ILCs and T cells develop separately from lineages that branch at the point of precursors within the bone marrow. Considering the common lineage markers and effector cytokine profiles shared between ILCs and T cells, it is surprising that the status of the TCR loci in ILCs was not fully explored at the time of their discovery. Here, we demonstrate that a high proportion of peripheral tissue ILC2s have TCRγ chain gene rearrangements and TCRδ locus deletions. Detailed analyses of these loci show abundant frameshifts and premature stop codons that would encode nonfunctional TCR proteins. Collectively, these data argue that ILC2 can develop from T cells that fail to appropriately rearrange TCR genes, potentially within the thymus.

Tephritid fruit flies are among the most destructive horticultural pests posing risks to Australia's multi-billion-dollar horticulture industry. Currently, there are 11 pest fruit fly species of economic concern in Australia. Of these, nine are native to this continent (Bactrocera aquilonis, B. bryoniae, B. halfordiae, B. jarvisi, B. kraussi, B. musae, B. neohumeralis, B. tryoni and Zeugodacus cucumis), while B. frauenfeldi and Ceratitis capitata are introduced. To varying degrees these species are costly to Australia's horticulture through in-farm management, monitoring to demonstrate pest freedom, quarantine and trade restrictions, and crop losses. Here, we used a common species distribution model, Maxent, to assess climate suitability for these 11 species under baseline (1960-1990) and future climate scenarios for Australia. Projections indicate that the Wet Tropics is likely to be vulnerable to all 11 species until at least 2070, with the east coast of Australia also likely to remain vulnerable to multiple species. While the Cape York Peninsula and Northern Territory are projected to have suitable climate for numerous species, extrapolation to novel climates in these areas decreases confidence in model projections. The climate suitability of major horticulture areas currently in eastern Queensland, southern-central New South Wales and southern Victoria to these pests may increase as climate changes. By highlighting areas at risk of pest range expansion in the future our study may guide Australia's horticulture industry in developing effective monitoring and management strategies.

Background Antibiotic use is a well-known risk factor for acquisition of drug-resistant bacteria and community antibiotic prescribing can drive high rates of resistance within the hospital setting. Owing to concerns over increasing fluoroquinolone (FQ) resistance among Gram-negative organisms at UAB Hospital, our stewardship program implemented a pre-authorization policy. The goal of this study was to assess the relationship between hospital fluoroquinolone use and antibiotic resistance. Methods In 2006, the inpatient formulary was consolidated to only ciprofloxacin and moxifloxacin with implementation of guidelines for use to limit inpatient prescribing. Any use outside of these guidelines required approval from an infectious diseases physician. Organism-specific data were obtained from the clinical microbiology database and FQ use was obtained from the hospital database. Correlations were calculated using Pearson’s coefficient. Results From 1998 to 2004, FQ use peaked at 173 days of therapy (DOT)/1,000 patient-days, but has remained below 60 DOT/1,000 patient-days since restriction implementation (Figure 1). FQ susceptibility was documented for five common Gram-negative isolates, P. aeruginosa, Acinetobacter spp., Enterobacter cloacae, E. coli, and K. pneumoniae, over an 18-year period (1998–2016). Common hospital acquired pathogens, including Pseudomonas aeruginosa, Acinetobacter spp. and Enterobacter cloacae improved in their susceptibilities to fluoroquinolones. Acinetobacter went from 35% to over 50% susceptible in the preceding 10 years after the policy. Pseudomonas improved from 50% susceptible to over 70% and Enterobacter improved from less than 50% to over 90% susceptible. Interestingly this improvement was not seen for E. coli which continued to show a decline in susceptibility from over 90% to near 60% in 2016. Conclusion In a large academic hospital setting, FQ susceptibility for common hospital-acquired GNRS improved significantly with the introduction of a restricted use program. A continued decline in E. coli FQ susceptibility suggests resistance rates may be driven by outpatient and community antibiotic use and thus, outpatient stewardship programs are necessary to prevent further spread of FQ resistance. Radiation is a well understood and established technology which has been researched and reviewed extensively for over 100 years. The capability of irradiation to substantially improve the international bilateral trade of horticultural commodities has only just been gaining recognition, particularly over the last 10 years. The poor rate of adoption of irradiation as a method of disinfesting food can be attributed to the conservative attitude of some importing countries and consumer perception, particularly when it's associated with the word “nuclear”. The world's leading food health and safety bodies including the World Health Organisation, Food Standards Australia New Zealand and the United Nations Food and Agriculture Organizations have all reported that food irradiation is a safe and effective treatment to a wide range of insect pests and kills bacteria. Educating consumers about benefits and safety of irradiated food is considered the key to irradiation becoming accepted widely amongst Australia's international trading partners. Irradiated fresh fruit and vegetables are not accepted by several importing Asian countries or the European Union but there are several other countries that do use and accept phytosanitary irradiated treated fruits. As a technology, irradiation offers a range of benefits for fresh horticultural food producers, exporters and importers. Low-dose irradiation provides a rapid treatment that does not alter the temperature of the commodity, is free of residues, fruit remains nutritionally adequate, and can enhance shelf life and colouring of some fruits. Some research has reported that there is a reduction in vitamin A and C in treated fruits at higher dose rates. One of Australia's major phytosanitary pests of concern for international trade is the Queensland Fruit Fly Bactrocera tryoni (Froggatt) (Diptera: Tephritidae). Methyl bromide and cold treatment are treatment methods commonly used for international trade. However, methyl bromide is a greenhouse gas and has occupational health concerns. Cold treatment takes several weeks to complete and is unsuitable for air freight. Time frames for lemons, for instance, vary from fourteen to sixteen days (for Qfly) plus temperature ramp down and then ramp up after treatment; ramping often takes three days each. Given the limitations of other conventional post-harvest treatments, irradiation is well suited for fruit fly disinfestation of commodities to be airfreighted to countries demanding fresh fruit with no residues.

Aims Cannulated surgical instruments may retain biologic debris after routine cleaning and sterilization. Residual debris after cleaning is assumed to be sterile; however, there is no experimental basis for this assumption. The purpose of this study was to determine the sterility of retained biodebris found within cannulated surgical instruments after autoclave sterilization. Materials and Methods Fifteen cannulated drill bits were used to drill pig scapulae to create a plug of bone that was exposed to a mixture of Bacillus cereus, Pseudomonas aeruginosa, and methicillin-resistant Staphylococcus aureus for 60, 120, or 180 minutes prior to sterilization. The drill bits were autoclave sterilized using standard settings. The “sterilized” bone cores were then incubated in solution and streak-plated on blood agar. Results All 3 positive controls were positive for the experimental bacteria. Two negative controls were positive for contaminant bacteria. A B. cereus strain was recovered from 1 of the experimental group drill bits in the 180-minute group. Pulsed-field gel electrophoresis confirmed that the recovered B. cereus strain was identical to the experimental inoculate. Conclusion Retained biodebris in cannulated drills may not be sterile after standard autoclave sterilization. In addition, delay of surgical instrument reprocessing may increase the risk of resistant contamination.

Background Enterococcus species frequently cause health care–associated bacteremia, with high attributable mortality. The benefit of consultation with infectious disease (ID) specialists has been previously illustrated with Staphylococcus aureus bacteremia. Whether ID consultation (IDC) improves mortality in enterococcal bacteremia is unknown. Methods This is a retrospective cohort single-center study from January 1, 2015, to June 30, 2016, that included all patients >18 years of age admitted with a first episode of Enterococcus bacteremia. Patients were excluded if death or transfer to palliative care occurred within 2 days of positive blood culture. Results Two hundred five patients were included in the study, of whom 64% received IDC. Participants who received IDC were more likely to undergo repeat cultures to ensure clearance (99% vs 74%; P, We report a retrospective cohort study of the effect of infectious diseases consultation (IDC) on thirty-day mortality in patients with Enterococcus bacteremia. We found a 65% decrease in 30-day in-hospital mortality, illustrating that IDC may be essential in improving survival.

Bactrocera cucumis is a lesser known native fruit fly in Australia. There are 44 reported hosts including cucurbits, papaws and tomatoes with a strong preference for cucurbits. Adult flies are not attracted to the known male lures such as cuelure, methyl eugenol or zingerone. They are weakly attracted to protein lures and a recently discovered cucumber volatile. For this reason, surveillance for this fruit fly remains problematic. Its range is predominantly coastal and appears to be centred north of Rockhampton in Queensland. It is known in the Northern Territory, Queensland and in New South Wales north of 29 o S latitude. Internationally, it is not reported in any Pacific countries or other parts of the world.

Bactrocera cucumis is one of many fruit fly species of concern for Australia's international trade partners. This species is one of the lesser known native fruit flies in Australia and its distribution and hosts have been reviewed. Eggs are larger than in most other species with a short hatching period. The adults and larvae are also larger than most other Australian fruit flies. Adults are long lived and have a high rate of reproduction. Stridulation and pheromones appear not to be used in the mating process. Adults can be managed using a range of measures including chemosterilants, the sterile insect technique and chemical sprays. Produce can be disinfested using a range of post-harvest treatments. Current export protocols include hot air and chilling treatments, irradiation, methyl bromide fumigation and winter window.

Background: Accurately tracing nosocomial transmission of coronavirus disease 2019 (COVID-19) is critical to developing effective infection prevention policies. Given the high prevalence and variable incubation period of SARS-CoV-2 infection, the utility of traditional contact tracing is limited. We describe a nosocomial outbreak in which whole-genome sequencing (WGS) was pivotal to identifying the primary case. Methods: This study was conducted at a New York City academic hospital. The index case was identified on August 13, 2020, and the last case on September 9, 2020. Hospital policy required all inpatients to be screened for COVID-19 on admission by SARS-CoV-2 molecular amplification testing. All healthcare workers (HCWs) were required to wear masks and eye protection for patient care. After a patient (patient 1), who tested SARS-CoV-2 negative on admission, was positive on preprocedure screening on hospital day 9, contact tracing was initiated. Two patients (patients 2 and 3) and 13 HCWs with high-risk exposures (HREs) to patient 1 were quarantined and referred for testing. Additional surveillance testing was performed on 18 inpatients and 84 HCWs on the affected unit. Patients 2 and 3 and 3 HCWs (HCW-1, -2, and -3), only 1 of whom had a high-risk exposure to patient 1, tested positive. WGS was performed to further investigate this outbreak. Results: The outbreak variant (clade 20A) was found in samples from 6 patients and 2 HCWs. Patients 2 and 3 were roommates of patient 1 in the 2 days before patient 1’s positive test, and they did not consistently wear masks in the room. HCW-1 placed a peripheral IV in patient 1 the day before patient 1’s positive test without wearing eye protection. Four additional cases in this cluster (patients 4–6 and HCW-4) were identified by surveillance WGS of positive tests. A review indicated that patient 1 was located ~3 m (~10 feet) away from patient 4 in the emergency department (ED) for 6 hours on hospital day 1, when the admission SARS-CoV-2 test from patient 4 was not positive. No epidemiologic link was found to patient 5 or 6 or HCW-4. The specimen from HCW-2 was inadequate for WGS. The specimen from HCW-3 was not linked to this cluster. Conclusions: This complex nosocomial outbreak highlights the importance of WGS in understanding transmission events. Patient 4 was not identified by traditional contact tracing but was linked to patient 1 and was recognized as the primary case through WGS, having likely infected patient 1 in the ED. Based on these findings, we focused our corrective actions on more promptly isolating suspected COVID-19 cases in the ED, increasing inpatient masking, and improving HCW adherence to universal eye protection.Funding: NoDisclosures: None

The study determined the composition, abundance, and distribution of phytoplankton in Lake Mainit. Plankton samples were collected in eight sampling stations four times in 2018. A total of 26 phytoplankton taxa in four groups – Bacillariophyceae/Myxophyceae, Chlorophyceae, Cyanophyceae and Euglenophyceae were identified. Aulacoseira was ranked as the most abundant taxa, followed by Zygnema, Fragilaria, Trichodesmium, and Ulothrix. ANOVA revealed no significant (P < 0.05) difference in the number of phytoplankton between the eight stations during the first sampling. In the second sampling, Bunga showed a significantly (P < 0.05) lower number of phytoplankton than the other stations. The third sampling also showed no significant (P < 0.05) difference in the number of phytoplankton between stations. During the fourth sampling, Tagbuyawan had by far (P < 0.05) the highest number of phytoplankton. Species diversity (H’) of phytoplankton was stable at Magticao, Jaliobong, Dinarawan, Kalinawan, and Bunga. The dominance of some species (Aulacoseira, Fragilaria, and Zygnema) affected the diversity and distribution of plankton. Spectrophotometric readings recorded an average Chl-a of 0.75 µg/L. The lake was still considered oligotrophic. Remediation measures and further studies on the lake towards the primary variation of plankton are recommended. Management efforts should be strengthened to support the sustainability of life in the lake.

Bactrocera tryoni (Froggatt) (Diptera: Tephritidae), commonly called 'Queensland fruit fly' in Australia, and Mediterranean fruit fly (Ceratitis capitata Wiedemann) (Diptera: Tephritidae) are the two most economically important fruit fly in Australia with B. tryoni in the east and Mediterranean fruit fly in the west. The two species coexisted for several decades, but it is believed that B. tryoni displaced Mediterranean fruit fly. In southeastern Australia, this was deemed inadequate for export market access, and a large fruit fly free zone (fruit fly exclusion zone) was developed in 1996 where B. tryoni was eradicated by each state department in their portion of the zone. This zone caused an artificial restricted distribution of B. tryoni. When the fruit fly exclusion zone was withdrawn in Victoria and New South Wales in 2013, B. tryoni became endemic once again in this area and the national distribution of B. tryoni changed. For export markets, B. tryoni is now deemed endemic to all eastern Australian states, except for the Greater Sunraysia Pest-Free Area. All regulatory controls have been removed between eastern states, except for some small zones, subject to domestic market access requirements. The eastern Australian states now form a B. tryoni endemic trading group or block. All Australian states and territories maintain legislation to regulate the movement of potentially infested host fruit into their states. In particular, eastern states remain active and regulate the entry of commodities possibly infested with Mediterranean fruit fly. The combination of regulatory controls limits the chances of Mediterranean fruit fly entering eastern states, and if it did, Mediterranean fruit fly is unlikely to establish in the opposition to a well-established B. tryoni population.

Australia is an island nation and a primary producer of agricultural and horticultural products. There is a large diversity of plant biosecurity threats which could adversely impact on Australia's production and exports. Surveillance has traditionally been used to monitor pests and optimise production. Increasingly surveillance is being used for early detection of exotic incursions, demonstration of eradication of incursions and pest freedom from exotic or endemic pests. These newer uses of surveillance utilise general and specific surveillance: surveillance data is maintained in electronic databases. Specific surveillance is a targeted surveillance search used by industry or state regulators for a specific pest to support pest freedom or other trade standards. The plant biosecurity surveillance cycle shows the flow of surveillance operations. In this paper, this cycle is demonstrated by case studies including pre border and the northern Australian at-border surveillance for the Australian-Asian interface. Within Australia, the multiple plant pest surveillance program was established in most capital cities where there are high flows of population and produce. As an industry example, the cotton industry surveillance program, particularly for cotton leaf curl, demonstrates how plant biosecurity surveillance operates within an industry. Asiatic citrus canker is another example of industry pertinent surveillance. Finally, surveillance for the purpose of declaring pest freedom areas is reviewed using fruit flies and currant lettuce aphid as examples.

Anthropogenic climate change is a major factor driving shifts in the distributions of pests and invasive species. The Queensland fruit fly, Bactrocera tryoni (Froggatt) (Qfly), is the most economically damaging insect pest of Australia’s horticultural industry, and its management is a key priority for plant protection and biosecurity. Identifying the extent to which climate change may alter the distribution of suitable habitat for Qfly is important for the development and continuation of effective monitoring programs, phytosanitary measures, and management strategies. We used Maxent, a species distribution model, to map suitable habitat for Qfly under current climate, and six climate scenarios for 2030, 2050 and 2070. Our results highlight that south-western Australia, northern regions of the Northern Territory, eastern Queensland, and much of south-eastern Australia are currently suitable for Qfly. This includes southern Victoria and eastern Tasmania, which are currently free of breeding populations. There is substantial agreement across future climate scenarios that most areas currently suitable will remain so until at least 2070. Our projections provide an initial estimate of the potential exposure of Australia’s horticultural industry to Qfly as climate changes, highlighting the need for long-term vigilance across southern Australia to prevent further range expansion of this species.

Innate lymphoid cells (ILCs) are critical for host defense and tissue repair but can also contribute to chronic inflammatory diseases. The transcription factor RORα is required for ILC2 development but is also highly expressed by other ILC subsets where its function remains poorly defined. We previously reported that Rorasg/sg bone marrow chimeric mice (C57BL/6J) were protected from Salmonella-induced intestinal fibrosis due to defective ILC3 responses. In this study, single-cell RNA analysis of ILCs isolated from inflamed tissues indicates that RORα perturbation led to a reduction in ILC3 lineages. Furthermore, residual Rorasg/sg ILC3s have decreased expression of key signature genes, including Rorc and activating cytokine receptors. Collectively, our data suggest that RORα plays a key role in preserving functional ILC3s by modulating their ability to integrate environmental cues to efficiently produce cytokines.

Tephritid fruit flies are among the most destructive horticultural pests and pose risks to Australia’s multi-billion-dollar horticulture industry. Currently, there are 11 pest fruit fly species of economic concern present in various regions of Australia. Of these, nine are native to this continent (Bactrocera aquilonis, B. bryoniae, B. halfordiae, B. jarvisi, B. kraussi, B. musae, B. neohumeralis, B. tryoniandZeugodacus cucumis), whileB. frauenfeldiandCeratitis capitataare introduced. To varying degrees these species are costly to Australia’s horticulture through in-farm management, monitoring to demonstrate pest freedom, quarantine and trade restrictions, and crop losses. Here, we used a common species distribution modelling approach, Maxent, to assess habitat suitability for these 11 species under current and future climate scenarios. These projections indicate that the Wet Tropics is likely to be vulnerable to all 11 species. The east coast of Australia will likely remain vulnerable to multiple species until at least 2070. Both the Cape York Peninsula and Northern Territory are also likely to be vulnerable, however, extrapolation to novel climates in these areas decrease confidence in model projections. The climate suitability of current major horticulture regions in north-western Australia, the Northern Territory, southern-central regions of New South Wales and southern Victoria to these pests is projected to increase as climate changes. Our study highlights areas at risk of pest range expansion in the future, to guide Australia’s horticulture industry in developing effective monitoring and management strategies.

Summary Background In September, 2017, human monkeypox re-emerged in Nigeria, 39 years after the last reported case. We aimed to describe the clinical and epidemiological features of the 2017–18 human monkeypox outbreak in Nigeria. Methods We reviewed the epidemiological and clinical characteristics of cases of human monkeypox that occurred between Sept 22, 2017, and Sept 16, 2018. Data were collected with a standardised case investigation form, with a case definition of human monkeypox that was based on previously established guidelines. Diagnosis was confirmed by viral identification with real-time PCR and by detection of positive anti-orthopoxvirus IgM antibodies. Whole-genome sequencing was done for seven cases. Haplotype analysis results, genetic distance data, and epidemiological data were used to infer a likely series of events for potential human-to-human transmission of the west African clade of monkeypox virus. Findings 122 confirmed or probable cases of human monkeypox were recorded in 17 states, including seven deaths (case fatality rate 6%). People infected with monkeypox virus were aged between 2 days and 50 years (median 29 years [IQR 14]), and 84 (69%) were male. All 122 patients had vesiculopustular rash, and fever, pruritus, headache, and lymphadenopathy were also common. The rash affected all parts of the body, with the face being most affected. The distribution of cases and contacts suggested both primary zoonotic and secondary human-to-human transmission. Two cases of health-care-associated infection were recorded. Genomic analysis suggested multiple introductions of the virus and a single introduction along with human-to-human transmission in a prison facility. Interpretation This study describes the largest documented human outbreak of the west African clade of the monkeypox virus. Our results suggest endemicity of monkeypox virus in Nigeria, with some evidence of human-to-human transmission. Further studies are necessary to explore animal reservoirs and risk factors for transmission of the virus in Nigeria. Funding None.

The Queensland fruit fly (Qfly; Bactrocera tryoni Froggatt) is a major horticultural pest species in Australia and considered a serious quarantine concern, both domestically and internationally. When a Qfly outbreak is declared in the main horticultural regions of southeast Australia, a 15 km radius suspension zone is established (area of 707 km2) around the epicentre of the outbreak. This suspension radius was originally adopted in the early 1990s. Here, we analysed trapping data in New South Wales (Australia) from 2002 to 2010 to evaluate the suitability of the 15 km suspension radius. There were 439 Qfly incursions recorded in Qfly-free zones, but only 9.9% of these incursions resulted in a declared outbreak. For these outbreaks, we conducted two distinct analyses. First, we modelled the distribution of infestation radii from the outbreaks and predicted the suspension distance using multiple levels of risk protection (e.g. probit 8.7, probit 9). Second, we fitted various probability distributions to individual fly trapping data and then estimated upper confidence limits for suspension distances, based on dispersal patterns of Qfly. Overall, the predicted suspension radii were substantially smaller than the current 15 km radius. Our results concur with other studies suggesting that Qfly are relatively poor colonisers. We propose that the current 15 km could be reduced substantially to 3 or 4 km.

Neuroticism and genetic variation in the serotonin-transporter (SLC6A4) and catechol-O-methyltransferase (COMT) gene are risk factors for psychopathology. Alterations in the functional integration and segregation of neural circuits have recently been found in individuals scoring higher on neuroticism. The aim of the current study was to investigate how genetic risk factors impact functional network organization and whether genetic risk factors moderate the association between neuroticism and functional network organization. We applied graph theory analysis on resting-state fMRI data in a sample of 120 women selected based on their neuroticism score, and genotyped two polymorphisms: 5-HTTLPR (S-carriers and L-homozygotes) and COMT (rs4680-rs165599; COMT risk group and COMT non-risk group). For the 5-HTTLPR polymorphism, we found that subnetworks related to cognitive control show less connections with other subnetworks in S-carriers compared to L-homozygotes. The COMT polymorphism moderated the association between neuroticism and functional network organization. We found that neuroticism was associated with lower efficiency coefficients in visual and somatosensory-motor subnetworks in the COMT risk group compared to the COMT non-risk group. The findings of altered topology of specific subnetworks point to different cognitive-emotional processes that may be affected in relation to the genetic risk factors, concerning emotion regulation in S-carriers (5-HTTLPR) and emotional salience processing in COMT risk carriers. OBJECTIVE: Serrated polyps (hyperplastic polyps, sessile or traditional serrated adenomas), which can arise in a sporadic or polyposis setting, predispose to colorectal cancer (CRC), especially those with microsatellite instability (MSI) due to MLH1 promoter methylation (MLH1(me+)). We investigate genetic alterations in the serrated polyposis pathway. DESIGN: We used a combination of exome sequencing and target gene Sanger sequencing to study serrated polyposis families, sporadic serrated polyps and CRCs, with validation by analysis of The Cancer Genome Atlas (TCGA) cohort, followed by organoid-based functional studies. RESULTS: In one out of four serrated polyposis families, we identified a germline RNF43 mutation that displayed autosomal dominant cosegregation with the serrated polyposis phenotype, along with second-hit inactivation through loss of heterozygosity or somatic mutations in all serrated polyps (16), adenomas (5) and cancer (1) examined, as well as coincidental BRAF mutation in 62.5% of the serrated polyps. Concurrently, somatic RNF43 mutations were identified in 34% of sporadic sessile/traditional serrated adenomas, but 0% of hyperplastic polyps (p=0.013). Lastly, in MSI CRCs, we found significantly more frequent RNF43 mutations in the MLH1(me+) (85%) versus MLH1(me-) (33.3%) group (p

Centromeres, the specialized chromosomal regions for recruiting kinetochores and directing chromosome segregation, are epigenetically marked by a centromeric histone H3 variant, CENP-A. To maintain centromere identity through cell cycles, CENP-A diluted during DNA replication is replenished. The licensing factor M18BP1(KNL-2) is known to recruit CENP-A to holocentromeres. Here, we show that RbAp46/48(LIN-53), a conserved histone chaperone, is required for CENP-A(HCP-3) localization in holocentric Caenorhabditis elegans. Indeed, RbAp46/48(LIN-53) and CENP-A(HCP-3) localizations are interdependent. RbAp46/48(LIN-53) localizes to the centromere during metaphase in a CENP-A(HCP-3)- and M18BP1(KNL-2)-dependent manner, suggesting CENP-A(HCP-3) loading may occur before anaphase. RbAp46/48(LIN-53) does not function at the centromere through histone acetylation, H3K27 trimethylation, or its known chromatin-modifying complexes. RbAp46/48(LIN-53) may function independently to escort CENP-A(HCP-3) for holocentromere assembly but is dispensable for other kinetochore protein recruitment. Nonetheless, depletion of RbAp46/48(LIN-53) leads to anaphase bridges and chromosome missegregation. This study unravels the holocentromere assembly hierarchy and its conservation with monocentromeres.

The sterile insect technique (SIT) has been used to suppress or eradicate fruit flies. It is critical to be able to identify sterile and wild flies so that informed decisions can be made during eradication activities. The current dye marking approach can be flawed on a small number of occasions, and a genetic method is needed to test suspect misidentified samples. As a proof of concept, a single multiplex PCR with nine microsatellite markers was used to study the genetic structure of Queensland fruit flies Bactrocera tryoni (Froggatt) in 11 locations in southern New South Wales. Cluster analysis demonstrated that one cluster was exclusive to the sterile mass-reared flies. A second distinct cluster was exclusive for one site in a wetter cooler area. The other sites were admixture of two main clusters. These nine microsatellite markers could be used to distinguish laboratory-reared flies from field flies. The mass-reared flies would need to be reanalysed after each introduction of wildness.

Black locust colonization of floodplains can exacerbate the problems associated with increasing nitrogen inputs into the riparian area and river ecosystem. In this study we compare the nitrogen budgets of two sites of floodplain ecosystem, one is colonized by invasive black locust and the other is forested by the indigenous willow species. Our data report the considerably higher N-flow rates, nitrogen storage and the faster total N-related ecosystem processes in the Robinia site in comparison to the indigenous Salix site of the study floodplain. Black locust affects the nitrogen cycle in the study riparian ecosystem through the N-fixation, high N-content litterfall and rapid litter decomposition that results in the increase of the nitrogen flow into the river and contaminates the water. Black locust appeared to reduce the conservation values of colonized areas and adjacent river. Restoration strategies that remove the black locust from invaded sites and prevent the further invasion will slow down the contamination. However, high availability of nitrogen in soil and soil degradation might have a long-term effect on floodplain ecosystem after the restoration.

Background: Pre-emptive management of the water supply can reduce hospital-onset legionellosis associated with building water systems. In 2014, an outbreak of Legionella pneumonia occurred in a 1,150-bed academic medical center (with ∼0.557 km2 or ∼6 million ft2 of space) among hematology-oncology patients. A comprehensive water safety and management program was implemented after the outbreak was controlled. We describe our experience implementing this prevention program. Methods: After the Legionella outbreak was controlled, an air and water safety committee (AWSC) was established and cochaired by the healthcare epidemiologist and the chief facilities officer. The AWSC established protocols for proactive environmental testing and the development of the an infection control risk assessment (ICRA) dedicated to water safety known as the water system construction and renovation risk assessment (WSCRRA).The water system management plan (WSMP) was developed (prior to the publication of ASHRAE 188) to direct the risk assessments and mitigation of any risks throughout the campus. Results: The WSMP identifies critical control measures, points, and limits that need to be maintained to control and monitor Legionella growth in the water systems. A control point is any step in a process at which biological, chemical, or physical factors can be controlled. The UAB Medicine WSMP includes 7 control points that are monitored on a daily basis. Examples of these control steps include monitoring of the hot water temperatures at the water heaters and distal outlets, managing the levels of mono-chloramines and chlorines in the water system, and managing water system components. To validate the efficacy of the WSMP, >610 water samples are collected from 19 hospital buildings over a year to be tested for Legionella. The results of water testing have shown significant decrease in distal site positivity due to managing and controlling these control points. This WSMP also evaluated the efficacy of 2 different methods for disinfecting water systems on campus as a corrective measure to Legionella growth. These methods are hyperchlorination and temporary copper silver ionization; based on the culture results of the water samples collected post disinfection, WSMP data show that the copper silver ionization method was more effective than hyperchlorination in controlling Legionella growth and decreasing the distal site positivity. Conclusions: The WSMP has provided ongoing management of building water systems and proactive actions around construction and renovation projects that involve water systems to prevent healthcare-acquired legionellosis. Early detection of Dacinae (Diptera: Tephritidae) incursions is vital for quick and effective management. A key method for early detection is the implementation of trapping networks in risk areas. Australia relies on the use of extensive trapping networks at, and surrounding, major ports and fruit production areas to identity any fruit fly incursions. Therefore, improvements to the trap efficiency of these networks may result in earlier detection of incursions and hence more rapid response times for management and/or eradication. The current Australian standard, the Lynfield trap, is expensive and does not stack readily for transport. Here, we test the effectiveness of a new trap type, the Probodelt cone trap, compared to the Lynfield trap. We ran three separate trials: an experimental trial in an inland orchard and two field trials using the Sydney trapping network. At the orchard site, Lynfield and Probodelt traps had similar capture rates (within 10%) for Bactrocera tryoni. For the Sydney trapping network, no significant differences were found in the number of species caught or trap rates for B. tryoni, Bactrocera cacuminata, Dacus absonifacies, and Dacus aequalis. Thus, the Probodelt cone trap was at least comparable to the Lynfield trap. Given the economic advantages of the Probodelt cone trap (e.g. reduced shipping and storage costs), the use of the Probodelt trap may be preferred over the Lynfield trap.

Item does not contain fulltext Pathogenic mutations in TMPRSS3, which encodes a transmembrane serine protease, cause non-syndromic deafness DFNB8/10. Missense mutations map in the low density-lipoprotein receptor A (LDLRA), scavenger-receptor cysteine-rich (SRCR), and protease domains of the protein, indicating that all domains are important for its function. TMPRSS3 undergoes proteolytic cleavage and activates the ENaC sodium channel in a Xenopus oocyte model system. To assess the importance of this gene in non-syndromic childhood or congenital deafness in Turkey, we screened for mutations affected members of 25 unrelated Turkish families. The three families with the highest LOD score for linkage to chromosome 21q22.3 were shown to harbor P404L, R216L, or Q398X mutations, suggesting that mutations in TMPRSS3 are a considerable contributor to non-syndromic deafness in the Turkish population. The mutant TMPRSS3 harboring the novel R216L missense mutation within the predicted cleavage site of the protein fails to undergo proteolytic cleavage and is unable to activate ENaC, thus providing evidence that pre-cleavage of TMPRSS3 is mandatory for normal function.
