Perfluorooctane sulfonate (PFOS) is an increasingly important environmental pollutant that is pervasive in the environment. A number of studies have focused on the toxicological effects of PFOS on model fish species (zebrafish and medaka), but little is known about the impact of PFOS on commercially important marine fish. Thus, the present study examined transcriptome responses to PFOS exposure in the liver of juvenile Cynoglossus semilaevis, an important farmed flatfish in China. Then, in response to PFOS challenges, 1695 and 5244 genes were identified as significantly increased and depressed, respectively. Significant expression changes were observed in immune-related genes (cytokine-cytokine receptor interaction, T-helper [Th]17 cell differentiation, and the chemokine nuclear factor-kappa B and T-cell receptor signaling pathways), indicating that immunotoxicity is a key aspect of the effects of PFOS on C. semilaevis. Exposure to PFOS also altered the gene expression levels of hormones (inhibin, insulin, somatostatin, and glucagon), which could lead to severe metabolic and endocrine dysfunction. As expected from previous studies, several phase I and phase II detoxification enzymes were significantly up-regulated, which could facilitate the biotransformation and detoxification of PFOS in C. semilaevis. Triazophos (TP) is a widespread pollutant in aquatic environments. A sex-specific metabolic response in green-lipped mussel Perna viridis to TP exposure was observed in our previous study, and this led us to investigate the mechanisms associated with its toxicity. P. viridis were subjected to chronic exposure (15 days) to TP at 35 μg/L to compare the sex-biased transcriptomic profiles in the gonads of male and female mussels. We identified 632 differentially expressed genes (DEGs) (348 up-regulated and 284 down-regulated) in TP-exposed males, and only 61 DEGs (9 up-regulated and 52 down-regulated) in TP-exposed females. Many DEGs were found to be involved in the nervous, reproductive endocrine, oxidative stress, and immune systems of P. viridis. Additionally, enzymatic activity analysis indicated TP induced neurotoxic effects and oxidative damage to the mussels. Our results demonstrate that the stress response and molecular mechanisms of TP toxicology are different between female and male mussels.

Di-(2-ethylhexyl) phthalate (DEHP) is currently the most frequently detected phthalic acid esters (PAEs) compounds and can induce diverse toxicities on aquatic organisms. To understand the molecular responses of fish to DEHP, we performed transcriptomic profiles in liver of tilapia (Oreochromis niloticus) which were exposed to environmental concentration of DEHP. A total of 30.10 Mb and 30.16 Mb clean reads were retrieved from the control and DEHP treated libraries, respectively. De-novo assembly of all the clean reads obtained 58,585 unigenes. After comparing the two libraries, 2814 and 1790 genes were identified as significantly increased and depressed, respectively. Gene ontology (GO) classification system and Kyoto Encyclopedia of Genes and Genomes (KEGG) database analysis demonstrated that DEHP significantly disturbed the expression level of genes associated with immunity, endocrine and reproductive system, lipid metabolism and so on. Quantitative real-time PCR was performed to validate the results of RNA-sequencing (RNA-seq) analysis. The resulting data provide new insights for exploring the molecular basis of tilapia response to DEHP exposure.

Di-n-butyl phthalate, one of the most easily detected pollutants of phthalate esters in the environment, has been added to the priority list of hazardous substances by many countries. As one of low molecular weight phthalates, Di-n-butyl phthalate may have a great adverse potency on various aquatic organisms. In this study, the juvenile red snapper, Lutjanus argentimaculatus, was exposed to the concentrations of Di-n-butyl phthalate (20 μg L−1, 100 μg L−1 and 500 μg L−1) for 15 days. EROD activities and CYP1A levels were measured in liver and gill tissues. In gills, the similar effect has been found to inhibit or induce EROD activities and CYP1A levels, and there existed a good correlation between them. Whereas in the case of the liver, a moderate correlation was observed between EROD activities and CYP1A levels, which was mainly due to the inhibited EROD activities and the CYP1A levels with no significant difference by day 15. In conclusion, this study revealed the similar and different effects of cytochrome P450 enzymes on fish in the time-, concentration-, and tissue-dependent Di-n-butyl phthalate exposure. Furthermore, as the adverse effects indicated between CYP1A levels and EROD activities, metabolic mechanisms of phthalates in different tissues should be highly emphasized in future studies.

Perfluorooctane sulfonate (PFOS) is an increasingly important environmental pollutant, which has been detected almost everywhere in the environment. Despite the widespread presence of PFOS, much less notice is taken of its toxicology effects on marine bivalves. Thus, the transcriptome response to PFOS treatment (nominal concentration of 20 mg/L) in hepatopancreas of a sentinel organism, Ruditapes philippinarum was examined. Compared with the control group, 32,149 unigenes were up-regulated and 26,958 unigenes down-regulated. Notably, significant gene expression changes were found in carbohydrate metabolism, energy metabolism, amino acid metabolism, lipid metabolism and protein biosynthesis, indicating the metabolic disruptions caused by PFOS in R. philippinarum. Additionally, numerous other differentially expressed genes were involved in immune system, antioxidant defense system and detoxification metabolism. In summary, transcriptome profiling of R. philippinarum after exposure to PFOS provided molecular support for our current understanding of the detrimental toxicity of PFOS on marine bivalves.

Triazophos, as a lipophilic organophosphate pesticide, displays higher bioaccumulation in the gonads of shellfish. To study the reproductive toxicity of triazophos, we applied metabolomics to characterize the gender-specific metabolic responses in mussel Perna viridis exposed to triazophos. Metabolites were differently altered by triazophos in ovaries of mussel at different concentrations and time intervals, while basically similar metabolic response patterns were observed in male mussels at the two tested concentrations after exposure for 24 and 48 h. The significant changes of metabolites in ovaries of mussel exhibited the disturbances in energy metabolism and osmotic regulation, while in male samples triazophos only affected the energy metabolism. Moreover, glycine, sn-glycero-3-phosphocholine, ethanol, aspartate, etc. exhibited consistent variation tendency in both male and female individuals. While the changes of homarine, betaine, taurine, hypotaurine, malonate, β-alanine, succinate, and choline showed obviously gender-specific responses. Overall, this study confirmed the gender-specific responses in gonad of P. viridis to triazophos exposure.

As a principal extracellular iron storage molecule, ferritin plays an important role in the iron-withholding strategy of innate immunity and detoxification system. In this study, we cloned and characterized another ferritin from Venerupis philippinarum (designated as VpFer2), in addition to one previously reported (VpFer1). VpFer2 possessed all the conserved features critical for the fundamental structure and function of ferritin H subunit. VpFer1 and VpFer2 mRNA were both found to be most abundantly expressed in hepatopancreas. Vibrio challenge could significantly up-regulate the mRNA expression of VpFers, and VpFer2 showed more sensitive to Vibrio anguillarum infection. For heavy metals exposure, the expression level of VpFer1 was significantly induced by Cd at 48 h, but kept relatively constant after exposure to Cu. With regards to VpFer2, the expression level dropped significantly at 24 h, then began to increase to the peak value at 48 h under Cd exposure, while Cu exposure constantly depressed the expression level of VpFer2 throughout the time course. Similarly, VpFer2 seemed to be more sensitive to heavy metals exposure than VpFer1 as its mRNA level changed by higher magnitudes. All these results suggested that VpFers may be important proteins involved in host immune defense and heavy metals detoxification. The diverse expression patterns of VpFers demonstrated that VpFer2 was an early and sensitive responder to environmental stress in V. philippinarum.

As pattern recognitionreceptors, the C1q-domain-containing (C1qDC) proteins play an important role in the pathogen recognition and complement pathway activation. In the present study, four novel C1q domain containing proteins (designated as VpC1qDC1, VpC1qDC2, VpC1qDC3 and VpC1qDC4) were cloned and characterized from clam Venerupis philippinarum. The four VpC1qDCs all possessed the conserved features critical for the fundamental structure and function of the C1q family. The four VpC1qDCs genes showed differential response profiles after exposure to the water soluble fraction of No.0 diesel oil (WSFD). More notably, VpC1qDC1 and VpC1qDC3 were more sensitive to low concentration of WSFD, as their mRNA level changed by higher magnitudes. In addition, VpC1qDC2 and VpC1qDC4 displayed notable increases with larger amplitude to high concentration of WSFD. All these results suggested that the transcriptional response of VpC1qDCs genes were probably a protective mechanism of the cell to oils pollution. The diverse expression patterns of VpC1qDCs demonstrated that VpC1qDC1 and VpC1qDC3 were sensitive responders to environmental stress in V. philippinarum.

Green-lipped mussels (Perna viridis) were exposed to waterborne tributyltin chloride (TBTCl) at different concentrations (0.2, 0.4 and 0.8 μg L(-1)) for up to 72 h. Accumulated TBTCl in Perna viridis correlated linearly with the exposure concentrations of 0.2 μg L(-1)(R(2) = 0.772), 0.4 μg L(-1)(R(2) = 0.952), and 0.8 μg L(-1)(R(2) = 0.909). The results of superoxide dismutase (SOD), glutathione peroxidase (GPx) and malondialdehyde (MDA) all decreased in gill tissues after 24 h of exposure, but the hepatic SOD and the hepatic GPx showed either little or no effect on exposure of TBTCl solutions. Analysis using the Spearman rank correlation coefficient showed the hepatic GPx activity appeared to have a significant negative correlativity (R(s) = -0.42) with the exposed TBTCl concentrations, and the hepatic MDA was significantly negatively correlated (R(s) = -0.33) with the tissue TBTCl concentrations. Conversely, a significant positive correlation (R(s) = 0.60) was shown between the gill MDA contents and exposure time. This study illustrates oxyradical scavenger GPx best correlated with stress level of pollutants among the various antioxidant parameters.
