BACKGROUND: The plant root system is critical for the absorption of water and nutrients, and have a direct influence on growth and yield. In cucumber, a globally consumed crop, the molecular mechanism of root development remains unclear, and this has implications for developing stress tolerant varieties. This study sought to determine the genetic patterns and related genes of cucumber root weight. A core cucumber germplasms population was used to do the GWAS analysis in three environments. RESULTS: Here, we investigated four root-weight related traits including root fresh weight (RFW), root dry weight (RDW), ratio of root dry weight to root fresh weight (RDFW) and the comprehensive evaluation index, D-value of root weight (DRW) deduced based on the above three traits for the core germplasm of the cucumber global repository. According to the D-value, we identified 21 and 16 accessions with light and heavy-root, respectively. We also found that the East Asian ecotype accessions had significantly heavier root than other three ecotypes. The genome-wide association study (GWAS) for these four traits reveals that 4 of 10 significant loci (gDRW3.1, gDRW3.2, gDRW4.1 and gDRW5.1) were repeatedly detected for at least two traits. Further haplotype and expression analysis for protein-coding genes positioned within these 4 loci between light and heavy-root accessions predicted five candidate genes (i.e., Csa3G132020 and Csa3G132520 both encoding F-box protein PP2-B1 for gDRW3.1, Csa3G629240 encoding a B-cell receptor-associated protein for gDRW3.2, Csa4G499330 encodes a GTP binding protein for gDRW4.1, and Csa5G286040 encodes a proteinase inhibitor for gDRW5.1). CONCLUSIONS: We conducted a systematic analysis of the root genetic basis and characteristics of cucumber core germplasms population. We detected four novel loci, which regulate the root weight in cucumber. Our study provides valuable candidate genes and haplotypes for the improvement of root system in cucumber breeding.

Background: Brassica napus L. (B. napus) is susceptible to waterlogging stress during different cultivation periods. Therefore, it is crucial to enhance the resistance to waterlogging stress to achieve a high and stable yield of B. napus. Results: Here we observed significant differences in the responses of two B. napus varieties in root under waterlogging stress. The sensitive variety (23651) exhibited a more pronounced and rapid reduction in cell wall thickness and root integrity compared with the tolerant variety (Santana) under waterlogging stress. By module clustering analysis based on transcriptome data, we identified that cell wall polysaccharide metabolism responded to waterlogging stress in root. It was found that pectin content was significantly reduced in the sensitive variety compared with the tolerant variety. Furthermore, transcriptome analysis revealed that the expression of two homologous genes encoding polygalacturonase-inhibiting protein 2 (PGIP2), involved in polysaccharide metabolic pathways, was highly upregulated in root of the tolerant variety under waterlogging stress. BnaPGIP2s probably confer waterlogging resistance by inhibiting the activity of polygalacturonases (PGs), which in turn reduces the degradation of the pectin backbone polygalacturonic acid. Conclusions: Our findings demonstrate that cell wall polysaccharides in root plays a vital role in response to the waterlogging stress and provide a theoretical foundation for breeding waterlogging resistance in B. napus varieties. Understanding the mechanisms that regulate plant root growth under soil drying is an important challenge in root biology. We observed that moderate soil drying promotes wheat root growth. To understand whether metabolic and hormonic changes are involved in this regulation, we performed transcriptome sequencing on wheat roots under well-watered and moderate soil drying conditions. The genes upregulated in wheat roots under soil drying were mainly involved in starch and sucrose metabolism and benzoxazinoid biosynthesis. Various plant hormone-related genes were differentially expressed during soil drying. Quantification of the plant hormones under these conditions showed that the concentrations of abscisic acid (ABA), cis-zeatin (CZ), and indole-3-acetic acid (IAA) significantly increased during soil drying, whereas the concentrations of salicylic (SA), jasmonic (JA), and glycosylated salicylic (SAG) acids significantly decreased. Correlation analysis of total root length and phytohormones indicated that CZ, ABA, and IAA are positively associated with wheat root length. These results suggest that changes in metabolic pathways and plant hormones caused by moderate soil drying help wheat roots grow into deeper soil layers. [ABSTRACT FROM AUTHOR]

We consider two spectral problems on an equilateral rooted tree with the standard (continuity and Kirchhoff's type) conditions at the interior vertices (except of the root if it is interior) and Dirichlet conditions at the pendant vertices (except of the root if it is pendant). For the first (Neumann) problem we impose the standard conditions (if the root is an interior vertex) or Neumann condition (if the root is a pendant vertex) at the root, while for the second (Dirichlet) problem we impose the Dirichlet condition at the root. We show that for caterpillar trees the spectra of the Neumann problem and of the Dirichlet problem uniquely determine the shape of the tree. This article explores the impact of salinity on two different maize cultivars and their ability to tolerate salt stress. The study reveals that the more salt-tolerant cultivar is better at excluding sodium (Na+) and retaining potassium (K+), which are crucial for minimizing Na+ uptake. The researchers also discovered that the salt-tolerant cultivar experienced a net influx of hydrogen ions (H+) under severe salt stress, while the salt-sensitive cultivar did not. This suggests that net H+ influx is an important but often overlooked trait in salt tolerance. Nitrogen (N) is essential for sugar beet (Beta vulgaris L.), a highly N-demanding sugar crop. This study investigated the morphological, subcellular, and microRNA-regulated responses of sugar beet roots to low N (LN) stress (0.5 mmol/L N) to better understand the N perception, uptake, and utilization in this species. The results showed that LN led to decreased dry weight of roots, N accumulation, and N dry matter production efficiency, along with damage to cell walls and membranes and a reduction in organelle numbers (particularly mitochondria). Meanwhile, there was an increase in root length (7.2%) and branch numbers (29.2%) and a decrease in root surface area (6.14%) and root volume (6.23%) in sugar beet after 7 d of LN exposure compared to the control (5 mmol/L N). Transcriptomics analysis was confirmed by qRT-PCR for 6 randomly selected microRNAs, and we identified 22 differentially expressed microRNAs (DEMs) in beet root under LN treatment. They were primarily enriched in functions related to binding (1125), ion binding (641), intracellular (437) and intracellular parts (428), and organelles (350) and associated with starch and sucrose metabolism, tyrosine metabolism, pyrimidine metabolism, amino sugar and nucleotide sugar metabolism, and isoquinoline alkaloid biosynthesis, as indicated by the GO and KEGG analyses. Among them, the upregulated miR156a, with conserved sequences, was identified as a key DEM that potentially targets and regulates squamosa promoter-binding-like proteins (SPLs, 104889216 and 104897537) through the microRNA-mRNA network. Overexpression of miR156a (MIR) promoted root growth in transgenic Arabidopsis, increasing the length, surface area, and volume. In contrast, silencing miR156a (STTM) had the opposite effect. Notably, the fresh root weight decreased by 45.6% in STTM lines, while it increased by 27.4% in MIR lines, compared to the wild type (WT). It can be inferred that microRNAs, especially miR156, play crucial roles in sugar beet root's development and acclimation to LN conditions. They likely facilitate active responses to N deficiency through network regulation, enabling beet roots to take up nutrients from the environment and sustain their vital life processes. Litter and root decomposition is an important source of soil organic matter and nutrients. To ascertain the contribution of litter and root to natural grassland nutrients in rocky desertification areas, from March 2017 to January 2018, the continuous soil column method, collector method, and litter decomposition method were used to study the soil nutrients, litter and root biomass, decomposition, and nutrient release of potential, moderate, and severe rocky desertification grasslands, as well as their responses to rocky desertification. The results showed that the litter and root decomposition rate showed a trend of being first fast and then slow, and the decomposition rate of litter and root was greater than 50% after 300 days. The annual litter decomposition rates of potential, moderate, and severe rocky desertification grasslands were 69.98%, 62.14%, and 49.79%, respectively, and the annual decomposition rates of root were 73.64%, 67.61%, and 64.09%, respectively. With a deepening degree of rocky desertification, the litter and root decomposition rate decreased. The decomposition coefficients, k, of litter in potential, moderate, and severe rocky desertification grasslands were 1.128, 0.896, and 0.668, respectively, and the decomposition coefficients, k, of root were 1.152, 1.018, and 0.987, respectively. The nutrient release processes of litter and root were different, and the release mode ultimately manifests as "release". In rocky desertification grasslands, the organic carbon (OC), total nitrogen (TN), total phosphorus (TP), and total potassium (TK) released by litter and root decomposition were 18.93–263.03 g·m−2·yr−1, 1.79–5.59 g·m−2·yr−1, 0.18–0.47 g·m−2·yr−1, and 0.66–3.70 g·m−2·yr−1, respectively. The contribution of root to soil nutrients was greater than that of litter. The degree of rocky desertification was negatively correlated with the biomass, decomposition rate, and nutrient return amount of litter and root. The bottle gourd [Lagenaria siceraria (Molina) Standl.] is often utilized as a rootstock for watermelon grafting. This practice effectively mitigates the challenges associated with continuous cropping obstacles in watermelon cultivation. The lower ground temperature has a direct impact on the rootstocks' root development and nutrient absorption, ultimately leading to slower growth and even the onset of yellowing. However, the mechanisms underlying the bottle gourd's regulation of root growth in response to low root zone temperature (LRT) remain elusive. Understanding the dynamic response of bottle gourd roots to LRT stress is crucial for advancing research regarding its tolerance to low temperatures. In this study, we compared the physiological traits of bottle gourd roots under control and LRT treatments; root sample transcriptomic profiles were monitored after 0 h, 48 h and 72 h of LRT treatment. LRT stress increased the malondialdehyde (MDA) content, relative electrolyte permeability and reactive oxygen species (ROS) levels, especially H2O2 and O2−. Concurrently, LRT treatment enhanced the activities of antioxidant enzymes like superoxide dismutase (SOD) and peroxidase (POD). RNA-Seq analysis revealed the presence of 2507 and 1326 differentially expressed genes (DEGs) after 48 h and 72 h of LRT treatment, respectively. Notably, 174 and 271 transcription factors (TFs) were identified as DEGs compared to the 0 h control. We utilized quantitative real-time polymerase chain reaction (qRT-PCR) to confirm the expression patterns of DEGs belonging to the WRKY, NAC, bHLH, AP2/ERF and MYB families. Collectively, our study provides a robust foundation for the functional characterization of LRT-responsive TFs in bottle gourd roots. Plant leaves are a source of essential phenolic compounds, which have numerous health benefits and can be used in multiple applications. While various techniques are available for recovering bioactive compounds from by-products, more data are needed on enzyme-assisted extraction (EAE). The aim of this study was to compare EAE and solid–liquid extraction (SLE), to evaluate the impact on bioactive compounds' extraction yield, phytochemical composition, and the antioxidant, antimicrobial, and antidiabetic properties of Aralia cordata leaves and roots, sea buckthorn Hippophae rhamnoides, and hemp Cannabis sativa leaves. The results indicate that EAE with Viscozyme L enzyme (EAE_Visc) extracts of the tested plant leaves possess the highest yield, antioxidant activity, and total phenolic content. Moreover, the EAE_Visc extract increased by 40% the total sugar content compared to the control extract of A. cordata root. Interestingly, the sea buckthorn leaf extracts exhibited α-glucosidase inhibitory activity, which reached an almost 99% inhibition in all extracts. Furthermore, the sea buckthorn leaves SLE and EAE_Visc extracts possess antibacterial activity against Staphylococcus aureus. Additionally, scanning electron microscopy was used to examine changes in cell wall morphology after EAE. Overall, this study shows that EAE can be a promising method for increasing the yield and improving the functional properties of the resulting extracts in a fast and sustainable way compared to SLE. Background: Studies have shown that plant endophytic microbial communities are ubiquitous and closely related to plant growth and health. To clarify the mechanism of the melon varieties with high resistant to wilt, the endophytic microbial compositions and metabolites in roots of melon varieties with high resistant ability to wilt were analyzed. Results: The results showed that the abundances of Firmicutes, Ascomycota, Bacillus, Bradyrhizobium, Amycolatopsis, Actinospica, and Catenulispora all increased in roots of wilt high resistant melon varieties (MT) which compared to wilt susceptible melon varieties (MS). Meanwhile, Ochrobactrum, Bordetella, Roseateles, Staphylococcus, Acidovorax, Amycolatopsis, Catenulispora, Promicromonospora, and Gymnopilus were the unique endophytic microbes in roots of MT. Moreover, in comparison with the MS varieties, the functions of Defense mechanisms, Secondary metabolites biosynthesis, transport and catabolism, Nucleotide transport and metabolism, Signal transduction mechanisms, Coenzyme transport and metabolism, Carbohydrate transport and metabolism and Amino acid transport and metabolism all increased in roots of MT varieties. Additionally, the nucleotide metabolism and biosynthesis of cofactors metabolic pathways were also significantly increased in roots of MT varieties. On the other hand, the untargeted metabolome results showed that Biosynthesis of various plant secondary metabolites, Nucleotide metabolism and Biosynthesis of cofactors metabolic pathways were significantly increased in the expression of MT varieties; and the content of metabolic compounds such as flavonoids, Cinnamic acid compounds, Organic acid compounds, and Nucleotides were increased. In addition, the correlation between microbiome and metabolome indicates a significant correlation between the two. Conclusions: All above results suggested that higher abundant antagonistic microbes and metabolic functions of endophytes in roots of wilt high resistant melon varieties (MT) were the important mechanisms for their high resistance to wilt. Background: Understanding the root canal morphology is essential for the success of root canal treatment. Therefore, this study aimed to evaluate and analyze the root canal configuration of maxillary premolars using Cone Beam Computed Tomography in the Pakistani subpopulation. Method: This cross-sectional study utilized CBCT scans from two distinct centres: Aga Khan University in Karachi and Jinnah MRI and Body Scans in Lahore. The CBCT images were visualized using GALAXIS version 1.9 (SICAT GmbH and Co. KG, Bonn, Germany), integrated within the Sirona Dental System (D-64625 Bensheim, Germany). The scanning parameters were standardized at 85 kV, 7 mA, with a 15-s exposure time and a voxel size of 0.16 mm. A total of 707 CBCT scans were collected, encompassing 2180 maxillary premolars. Root canal configurations were classified based on (Ahmed et al. Int Endod J. 2017;50(8):761–70). Statistical analyses were performed using SPSS version 26, employing the Chi-square test with a significance level set at p < 0.05. Results: The distribution of root canal morphologies varied significantly with age and gender. Among maxillary premolars, 50% exhibited the typical configuration of 2MPMB1 L1 (two roots, single canal in each buccal and lingual root), while 26% of maxillary right second premolars displayed 1MPM1 (one root, one canal). Overall, 1MPM1 accounted for 27.4% of the total cases in the second premolars. There was no statistically significant relationship between age and root canal distribution in either first premolars (p = 0.338) or second premolars (p = 0.833). Regarding gender, a significant difference was observed in the distribution of right maxillary 1st premolars (p = 0.022*), with a higher prevalence among females. Conclusion: This study offers significant insights into the anatomical variations of root canals in maxillary premolars across diverse regional subpopulations in Pakistan. While specific root canal configurations were prevalent, the findings indicate no statistically significant correlation between age and root canal morphology in maxillary premolars. Understanding how plant nutrient acquisition strategies respond to grazing at the community level is critical understanding ecosystem structure and functioning in grasslands. However, few studies have simultaneously compared the difference in above‐ground (leaf) and below‐ground (root) nutrient‐acquisition strategies in response to long‐term grazing, especially at the regional scale.Here, we measured a set of leaf and fine‐root traits that correspond to the fast‐slow economic spectrum at the community level in 10 experimental sites from paired grazed and ungrazed grasslands across a soil nutrient gradient covering three major types of grasslands in northern China.We found that patterns of variations of leaf and fine‐root traits were consistent with both a leaf and root economic spectrum at the community level for both grazed and non‐grazed plots. Grazing had minor effect on community‐level leaf nutrient‐acquisition strategies but strongly influenced community‐level root nutrient‐acquisition strategies. Specifically, root nutrient‐acquisition strategies were shifted to more exploitative resource use in grazed communities. Moreover, soil nutrient contributed to the changes in both leaf and root nutrient‐acquisition strategies, which tended towards a more resource acquisition strategy with increasing soil nutrient levels. Grazing significantly interacted with soil nutrient to affect root nutrient‐acquisition strategies, and grazing contributed more to root nutrient‐acquisition strategies than soil nutrient.Synthesis. Our results demonstrated completely inconsistent responses of community‐level above‐ and below‐ground resource acquisition strategies to long‐term grazing, and below‐ground acquisition strategies were more sensitive to long‐term grazing. The yield of sweet potato [Ipomoea batatas (L.) Lam] can be easily threatened by drought stress. Typically, early stages like the seedling stage and tuber-root expansion stage are more vulnerable to drought stress. In this study, a highly drought-tolerant sweet potato cultivar "WanSu 63" was subjected to drought stress at both the seedling stage (15 days after transplanting, 15 DAT) and the tuber-root expansion stage (45 DAT). Twenty-four cDNA libraries were constructed from leaf segments and root tissues at 15 and 45 DAT for Next-Generation Sequencing. A total of 663, 063, and 218 clean reads were obtained and then aligned to the reference genome with a total mapped ratio greater than 82.73%. A sum of 7119, 8811, 5463, and 930 differentially expressed genes were identified from leaves in 15 days (L15), roots in 15 days (R15), leaves in 45 days (L45), and roots in 45 days (R45), respectively, in drought stress versus control. It was found that genes encoding heat shock proteins, sporamin, LEA protein dehydrin, ABA signaling pathway protein gene NCED1, as well as a group of receptor-like protein kinases genes were enriched in differentially expressed genes. ABA content was significantly higher in drought-treated tissues than in the control. The sweet potato biomass declined sharply to nearly one-quarter after drought stress. In conclusion, this study is the first to identify the differentially expressed drought-responsive genes and signaling pathways in the leaves and roots of sweet potato at the seedling and root expansion stages. Due to the high demands of data communication, the broadcasting system streams the data daily. This service not only sends out the message to the correct participant but also respects the security of the identity user. In addition, when delivered, all the information must be protected for the party who employs the broadcasting service. Currently, Attribute-Based Broadcast Encryption (ABBE) is useful to apply for the broadcasting service. (ABBE) is a combination of Attribute-Based Encryption (ABE) and Broadcast Encryption (BE), which allows a broadcaster (or encrypter) to broadcast an encrypted message, including a predefined user set and specified access policy to install the authorization mechanism. It is desirable to hide all the information when producing in the ciphertext, which has not been considered in the previous works of ABBE. Motivated by the above issue, we devise a solution to achieve anonymity for the ABBE scheme, which not only hides the access structures but also anonymizes the user's identity. In this work, we propose two schemes as Anonymous Key Policy (AKP)-ABBE and Anonymous Ciphertext Policy (ACP)-ABBE with supporting multiple access structures by using OR / AND gates. Specifically, we present the generic constructions of AKP/ACP-ABBE on the building block of the Inner Product Encryption (IPE ), which enables the hidden user's identity and complex OR / AND -Gate access structure. [Objective] To explore the differences of vegetation growth characteristics between different plant communities and gully positions. [Methods] The vegetation growth characteristics of 5 typical plant communities were systematically studied in Zhifanggou small watershed of Ansai, Shaanxi Province.The investigated characteristics included species diversity, coverage, aboveground biomass, litter storage and underground biomass. [Results] (1) The above ground biomass, litter storage and root reduction coefficient were significantly affected by vegetation type, tree cutting ditch > shrub cutting ditch > herb cutting ditch, and the coverage was shrub cutting ditch > herb cutting ditch > tree cutting ditch. (2) The species diversity index and coverage of the right side of gully (sunny to semi-sunny slopes) were smaller than those on the left side (shade to semi-shade slopes); (3) Under the comprehensive influence of hydrothermal conditions, there was no obvious difference in vegetation growth characteristics between gully bottom and the original slope; (4) The accumulation of litter on the original slope was greater than that of different positions of gully. [Conclusion] The research results provide data foundation and theoretical support for understanding of the potential effects of vegetation restoration on the development of gully and evaluating the role of vegetation in mitigating gully erosion.

Abstract Background Studies have shown that plant endophytic microbial communities are ubiquitous and closely related to plant growth and health. To clarify the mechanism of the melon varieties with high resistant to wilt, the endophytic microbial compositions and metabolites in roots of melon varieties with high resistant ability to wilt were analyzed. Results The results showed that the abundances of Firmicutes, Ascomycota, Bacillus, Bradyrhizobium, Amycolatopsis, Actinospica, and Catenulispora all increased in roots of wilt high resistant melon varieties (MT) which compared to wilt susceptible melon varieties (MS). Meanwhile, Ochrobactrum, Bordetella, Roseateles, Staphylococcus, Acidovorax, Amycolatopsis, Catenulispora, Promicromonospora, and Gymnopilus were the unique endophytic microbes in roots of MT. Moreover, in comparison with the MS varieties, the functions of Defense mechanisms, Secondary metabolites biosynthesis, transport and catabolism, Nucleotide transport and metabolism, Signal transduction mechanisms, Coenzyme transport and metabolism, Carbohydrate transport and metabolism and Amino acid transport and metabolism all increased in roots of MT varieties. Additionally, the nucleotide metabolism and biosynthesis of cofactors metabolic pathways were also significantly increased in roots of MT varieties. On the other hand, the untargeted metabolome results showed that Biosynthesis of various plant secondary metabolites, Nucleotide metabolism and Biosynthesis of cofactors metabolic pathways were significantly increased in the expression of MT varieties; and the content of metabolic compounds such as flavonoids, Cinnamic acid compounds, Organic acid compounds, and Nucleotides were increased. In addition, the correlation between microbiome and metabolome indicates a significant correlation between the two. Conclusions All above results suggested that higher abundant antagonistic microbes and metabolic functions of endophytes in roots of wilt high resistant melon varieties (MT) were the important mechanisms for their high resistance to wilt. We consider two spectral problems on an equilateral rooted tree with the standard (continuity and Kirchhoff's type) conditions at the interior vertices (except of the root if it is interior) and Dirichlet conditions at the pendant vertices (except of the root if it is pendant). For the first (Neumann) problem we impose the standard conditions (if the root is an interior vertex) or Neumann condition (if the root is a pendant vertex) at the root, while for the second (Dirichlet) problem we impose the Dirichlet condition at the root. We show that for caterpillar trees the spectra of the Neumann problem and of the Dirichlet problem uniquely determine the shape of the tree. Also, we present an example of co-spectral snowflake graphs.

Summary: Barley (Hordeum vulgare) is an important global cereal crop and a model in genetic studies. Despite advances in characterising barley genomic resources, few mutant studies have identified genes controlling root architecture and anatomy, which plays a critical role in capturing soil resources.Our phenotypic screening of a TILLING mutant collection identified line TM5992 exhibiting a short‐root phenotype compared with wild‐type (WT) Morex background. Outcrossing TM5992 with barley variety Proctor and subsequent SNP array‐based bulk segregant analysis, fine mapped the mutation to a cM scale. Exome sequencing pinpointed a mutation in the candidate gene HvPIN1a, further confirming this by analysing independent mutant alleles.Detailed analysis of root growth and anatomy in Hvpin1a mutant alleles exhibited a slower growth rate, shorter apical meristem and striking vascular patterning defects compared to WT. Expression and mutant analyses of PIN1 members in the closely related cereal brachypodium (Brachypodium distachyon) revealed that BdPIN1a and BdPIN1b were redundantly expressed in root vascular tissues but only Bdpin1a mutant allele displayed root vascular defects similar to Hvpin1a.We conclude that barley PIN1 genes have sub‐functionalised in cereals, compared to Arabidopsis (Arabidopsis thaliana), where PIN1a sequences control root vascular patterning. This study analyzed issues surrounding the concept of "stem" in Turkish linguistics. It began by establishing a general framework based on existing definitions of "root" and "stem" in the literature. Subsequently, it analyzed the treatment and examples of "stem" presented in Turkish grammar books published from 1940 to the present. The analysis revealed inconsistencies in the scope and definition of "stem," with terms like "root," "base," "derived root," "closed stem" and "closed form unit" often used interchangeably in examples. These inconsistencies stem not only from varying terminological preferences but also from the challenge of handling words derived from roots that do not exist independently. While some researchers propose adopting the term "base" to address these limitations, modern linguists increasingly favor terms like "form unit" and "word unit."

Abstract Background Soybean establishes a mutualistic interaction with nitrogen-fixing rhizobacteria, acquiring most of its nitrogen requirements through symbiotic nitrogen fixation. This crop is susceptible to water deficit; evidence suggests that its nodulation status—whether it is nodulated or not—can influence how it responds to water deficit. The translational control step of gene expression has proven relevant in plants subjected to water deficit. Results Here, we analyzed soybean roots’ differential responses to water deficit at transcriptional, translational, and mixed (transcriptional + translational) levels. Thus, the transcriptome and translatome of four combined-treated soybean roots were analyzed. We found hormone metabolism-related genes among the differentially expressed genes (DEGs) at the translatome level in nodulated and water-restricted plants. Also, weighted gene co-expression network analysis followed by differential expression analysis identified gene modules associated with nodulation and water deficit conditions. Protein-protein interaction network analysis was performed for subsets of mixed DEGs of the modules associated with the plant responses to nodulation, water deficit, or their combination. Conclusions Our research reveals that the stand-out processes and pathways in the before-mentioned plant responses partially differ; terms related to glutathione metabolism and hormone signal transduction (2 C protein phosphatases) were associated with the response to water deficit, terms related to transmembrane transport, response to abscisic acid, pigment metabolic process were associated with the response to nodulation plus water deficit. Still, two processes were common: galactose metabolism and branched-chain amino acid catabolism. A comprehensive analysis of these processes could lead to identifying new sources of tolerance to drought in soybean.

Objective To analyze the spatial relationship between the roots of maxillary anterior premolars and the maxillary sinus, thus providing an anatomical basis for timing, planning, surgical approaches, and implant selection at this site. Methods Cone beam CT (CBCT) images were collected from 264 patients (aged 20-65 years) who visited the Ruihua Dental Clinic between January 2017 and March 2023. The minimum distance from the apex of the maxillary anterior premolar roots to the lower wall of the maxillary sinus was measured on the coronal plane. The classification of the vertical relationship between the tooth root and the lower wall of the maxillary sinus was performed, and comparisons were made bilaterally, between genders, and among different age groups. Results The minimum distance (Q50) from the apex of the first maxillary premolar root to the lower wall of the maxillary sinus was 7.34 mm for the single-root type, 7.80 mm for the buccal root of the double-root type, and 7.36 mm for the palatal root. For the second maxillary premolar, the median distance was 2.56 mm for the single root type, 1.73 mm for the buccal root type, and 1.23 mm for the palatal root type. There was a significant difference in the shortest distance from the apex of the right second maxillary premolar single root to the lower wall of the maxillary sinus among the different age groups (PP>0.05). The most common vertical relationship between the apex of the maxillary anterior premolar roots and the lower wall of the maxillary sinus was noncontact. There was no significant difference in the vertical relationship classification between the single-root and double-root types (P>0.05). Conclusion Most maxillary first premolar roots can provide sufficient bone height, which makes it easy to achieve immediate implantation. The maxillary second premolar root frequently involves insufficient bone, which is necessary to make full use of the bone wall of the extraction socket or the sinus floor cortical bone to achieve initial stability. The vertical relationship between the premolar root and maxillary sinus was influenced by age and dental position. Younger age groups often exhibit inadequate bone height, and the indication for immediate implantation should be carefully considered. The number of roots does not significantly affect the relationship between the sinus and root; however, double-rooted premolars offer more support for immediate implantation and socket healing due to the small root diameter and bony separation between the roots.

Introduction: The brachial plexus is a variable nerve plexus in its formation and branching pattern. Variations in the brachial plexus are not uncommon, and its area of supply and associations with other adjacent anatomical structures require clinical and surgical attention. Aim: To find out any anatomical variations in the formation and branching pattern of the brachial plexus in human foetuses. Materials and Methods: A morphometric prospective observational study was conducted from January 2020 to July 2022, on 30 stillborn or Intrauterine Dead (IUD) human foetuses were taken in the study. Gestation age from 28 to 40 weeks of gestation were included in the study, while those with gross anomalies or morphological anomalies of the cranium and vertebral column were excluded. The dissection includes vertical midline incision of the skin from the external occipital protuberance to the lower limit of the thoracic vertebra (T12). The skin was cut in the midline and reflected laterally. All superficial and deep muscles of the neck were dissect out to clear the cervical and thoracic parts of the vertebral column. The vertebral column was cut and opened to visualise the spinal cord and spinal nerves. Based on gestational age, the foetuses were divided into three groups: four in the first group (28-31 weeks), twelve in the second group (32-35 weeks), and fourteen in the third group (36-40 weeks) were divided for descriptive purposes. The gender of foetuses was determined based on the external genitalia. Results: A total of 60 brachial plexuses were dissected, of which 49 (81.66%) had the usual anatomical formation of the trunks: the upper trunk formed by C5 and C6 roots, the middle trunk by the C7 root, and the lower trunk by the C8 and T1 roots. Eleven (18.34%) plexuses presented variations in trunk formation. Specifically, 8 (13.3%) brachial plexuses were of the prefix type, where the upper trunk was formed by the C5 and C6 roots with an additional contribution from the C4 root; there was inter-branch communication between C6 and C7 in one (1.7%) case; the middle and lower trunks united by C7, C8, and T1 roots formed the lower trunk in another (1.7%) case, and finally, one (1.7%) case exhibited the post-fix type. Conclusion: In this study, out of the 30 foetuses examined, 11 showed variations, which is not uncommon. The prefix type of brachial plexus is quite common among the possible variations, although anatomists and neurosurgeons cannot ignore the possibility of other variations, like post-fix type or communication between the roots of the brachial plexuses.

Abstract Peanut (Arachis hypogaea L.) is an important oilseed crop worldwide. However, soil salinization becomes one of the main limiting factors of peanut production. Therefore, developing salt-tolerant varieties and understanding the molecular mechanisms of salt tolerance is important to protect peanut yield in saline areas. In this study, we selected four peanut varieties with contrasting response to salt challenges with T1 and T2 being tolerance and S1 and S2 being susceptible. High-throughput RNA sequencing resulted in more than 314.63 Gb of clean data from 48 samples. We identified 12,057 new genes, 7,971of which have functional annotations. KEGG pathway enrichment analysis of uniquely expressed genes in salt-tolerant peanut revealed that upregulated genes in the root are involved in the MAPK signaling pathway, fatty acid degradation, glycolysis/gluconeogenesis, and upregulated genes in the shoot were involved in plant hormone signal transduction and the MAPK signaling pathway. Na+ content, K+ content, K+/ Na+, and dry mass were measured in root and shoot tissues, and two gene co-expression networks were constructed based on weighted gene co-expression network analysis (WGCNA) in root and shoot. In this study, four key modules that are highly related to peanut salt tolerance in root and shoot were identified, plant hormone signal transduction, phenylpropanoid biosynthesis, starch and sucrose metabolism, flavonoid biosynthesis, carbon metabolism were identified as the key biological processes and metabolic pathways for improving peanut salt tolerance. The hub genes include genes encoding ion transport (such as HAK8, CNGCs, NHX, NCL1) protein, aquaporin protein, CIPK11 (CBL-interacting serine/threonine-protein kinase 11), LEA5 (late embryogenesis abundant protein), POD3 (peroxidase 3), transcription factor, and MAPKKK3. There were some new salt-tolerant genes identified in peanut, including cytochrome P450, vinorine synthase, sugar transport protein 13, NPF 4.5, IAA14, zinc finger CCCH domain-containing protein 62, beta-amylase, fatty acyl-CoA reductase 3, MLO-like protein 6, G-type lectin S-receptor-like serine/threonine-protein kinase, and kinesin-like protein KIN-7B. The identification of key modules, biological pathways, and hub genes in this study enhances our understanding of the molecular mechanisms underlying salt tolerance in peanuts. This knowledge lays a theoretical foundation for improving and innovating salt-tolerant peanut germplasm.

Humic substances (HS), a natural polymer isomeric mixture, is an important component of terrestrial ecosystem. Its promotion effect on plant growth has been widely studied in recent years. Multiple signaling pathways play an important role in the process of HS promoting plant growth and development. The article summarizes the signaling pathway transduction and crosstalk mechanisms of HS in promoting the growth and development of plant roots and shoots. During the promotion of root growth, HS mainly involves the transduction of Indole-3-acetic acid (IAA), nitric oxide (NO), abscisic acid (ABA), reactive oxygen species (ROS) and Ca2+ signal, and the complex crosstalk between them; During the promotion of shoot growth, HS mainly involves the transmission of cytokinin (CTK) and ABA signaling pathways, and the complex crosstalk occurs between nitrate and CTK signaling pathways. HS regulates plant physiological metabolism through a complex metabolic network, involving multiple complicated signaling pathways and crosstalk processes. Among them, (PM) H+- ATPase is the key node of HS promoting root and shoot growth, and multiple signaling pathways mediate the signal transduction of cascade amplification via (PM) H+- ATPase. NO signal has a complex crosstalk process with multiple signaling pathways that regulate root and shoot growth, and is an important crosstalk signaling molecule involved in the promotion of plant growth by HS. There are other signaling pathways involved in the growth and development of roots and shoots triggered by HS, among which the defense signaling pathways salicylicacid (SA) and jasmonic acid (JA) are regulated by signaling pathways IAA, NO, ABA and CTK. Due to the dispersion, heterogeneity and complexity of biochemical research of HS, the relevant mechanism of the short-term promoting effect of HS on plant growth and the overall action mechanism on plant growth and development during the whole growth stage have not yet been clarified. Therefore, further in-depth analysis is needed to reveal the signal transduction, crossover, crosstalk and integration pathways involved in the growth promotion process, and the complex nutritional and metabolic pathways. The result of our research could provide theoretical basis for studying the action mechanism of HS physiological activity and its application in agricultural production.

Summary Single‐cell proteomics (SCP) is an emerging approach to resolve cellular heterogeneity within complex tissues of multi‐cellular organisms. Here, we demonstrate the feasibility of SCP on plant samples using the model plant Arabidopsis thaliana. Specifically, we focused on examining isolated single cells from the cortex and endodermis, which are two adjacent root cell types derived from a common stem cell lineage. From 756 root cells, we identified 3763 proteins and 1118 proteins/cell. Ultimately, we focus on 3217 proteins quantified following stringent filtering. Of these, we identified 596 proteins whose expression is enriched in either the cortex or endodermis and are able to differentiate these closely related plant cell types. Collectivity, this study demonstrates that SCP can resolve neighboring cell types with distinct functions, thereby facilitating the identification of biomarkers and candidate proteins to enable functional genomics. Proso millet (Panicum miliaceum L.) is resilient to abiotic stress, especially to drought. However, the mechanisms by which its roots adapt and tolerate salt stress are obscure. In this study, to clarify the molecular mechanism of proso millet in response to drought stress, the physiological indexes and transcriptome in the root of seedlings of the proso millet cultivar 'Yumi 2' were analyzed at 0, 0.5, 1.0, 1.5, and 3.0 h of stimulated drought stress by using 20% PEG-6000 and after 24 h of rehydration. The results showed that the SOD activity, POD activity, soluble protein content, MDA, and O2−· content of 'Yumi 2' increased with the time of drought stress, but rapidly decreased after rehydration. Here, 130.46 Gb of clean data from 18 samples were obtained, and the Q30 value of each sample exceeded 92%. Compared with 0 h, the number of differentially expressed genes (DEGs) reached the maximum of 16,105 after 3 h of drought, including 9153 upregulated DEGs and 6952 downregulated DEGs. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses revealed that upregulated DEGs were mainly involved in ATP binding, nucleus, protein serine/threonine phosphatase activity, MAPK signaling pathway–plant, plant–pathogen interactions, and plant hormone signal transduction under drought stress, while downregulated DEGs were mainly involved in metal ion binding, transmembrane transporter activity, and phenylpropanoid biosynthesis. Additionally, 1441 TFs screened from DEGs were clustered into 64 TF families, such as AP2/ERF-ERF, bHLH, WRKY, NAC, MYB, and bZIP TF families. Genes related to physiological traits were closely related to starch and sucrose metabolism, phenylpropanoid biosynthesis, glutathione metabolism, and plant hormone signal transduction. In conclusion, the active oxygen metabolism system and the soluble protein of proso millet root could be regulated by the activity of protein serine/threonine phosphatase. AP2/ERF-ERF, bHLH, WRKY, NAC, MYB, and bZIP TF families were found to be closely associated with drought tolerance in proso millet root. Tobacco cembranoids, known for their anti-insect and antifungal properties, were shown to be mainly present on the surface of leaves and flowers, being biosynthesized by their trichomes. It remains unclear whether they could be biosynthesized in other organs without trichomes. Cembratrien-ol synthases (CBTSs) catalyze the conversion of GGPP to CBT-ols and thus play an important role in cembranoid biosynthesis. This study identified the CBTS family genes in tobacco and examined their spatiotemporal expression patterns. The CBTS genes showed diverse expression patterns in tobacco organs, with the majority highly expressed in leaves and a few highly expressed in flowers. The expression of CBTS genes were also correlated with the development of tobacco plants, and most of them showed the highest expression level at the budding stage. Furthermore, their expression is mediated by the JA (jasmonate) signaling in all tobacco organs. Several CBTS genes were found to be highly expressed in tobacco roots that have no trichomes, which prompted us to determine the cembranoid production in roots and other organs. GC-MS and UPLC assays revealed that cembranoids were produced in all tobacco organs, which was supported by the bioactivity assay results that almost all these CBTS enzymes could catalyze CBT-ol biosyntheis in yeast, and that the content ratio of CBT-ols and CBT-diols in tobacco roots was different to that in leaves. This work sheds insights into the expression profiles of tobacco CBTS genes and provides a feasibility to engineer tobacco roots for industrial production of cembranoids. [ABSTRACT FROM AUTHOR]

[Objective] The aims of this study are to clarify the anti-scourability difference of three herbaceous root-soil system and its relationship with influencing factors in the loess hilly region of southern Ningxia, and to provide a theoretical basis for improving the quality of ecological construction and optimizing the allocation of vegetation in the study area. [Methods] The common herbaceous root-soil system of Thymus mongolicus, Potentilla acaulis and Stipa bungeana were taken as research objects of the study area. The indoor undamaged soil trench scouring tests was carried out. LA-S root sanning analysis system was used to study the anti-scourability of different herbs root-soil system in 0—15 cm soil layer and their relationship with root and soil. [Results] (1) The anti-scourability of three herbaceous root-soil system in 0—15 cm soil layer decreased in the order: Potentilla acaulis (53.7 L·h/g) ＞Thymus mongolicus (36.5 L·h/g) ＞Stipa bungeana (14.2 L·h/g) . Potentilla acaulis root-soil system anti-scourability was the strongest. (2) The average root length density, surface area density, volume density and specific root length of three herbaceous root-soil system in 0—15 cm soil layer were 4.25～10.56 cm/cm³, 42.95～111.51 mm²/cm³, 5.04～17.94 mm³/cm³, 35.06～67.96 m/g, respectively. The root length density and other parameters of Thyme mongolicus were significantly larger than other herbs. ( 3) The organic carbon contents of three herbaceous root soils in 0—15 cm soil layer decreased in the order: Thyme mongolicus (1.437%) ＞Potentilla acaulis (1.290%) ＞Stipa bungeana (0.430%) . The root soil clay content of Thyme mongolicus was 1.14 times and 1.20 times that of Potentilla acaulis and Stipa bungeana. The soil aggregate parameters MWD, GMD, and R＞0.25 of three herbaceous were 13.80～18.23 mm, 7.15～11.11 mm, and 43.06%～70.05%, respectively. All above indicators of Potentilla acaulis was the largest. (4) Root and soil could directly or indirectly, individually or jointly affect anti-scourability of three herbaceous root-soil system. Clay, silt, sand and aggregates of Thyme mongolicus soil directly affected soil anti-scourability, and most of its roots indirectly affected soil anti-scourability by affecting soil mechanical composition and organic carbon content. Soil silt and most fine roots of Potentilla acaulis directly affected soil anti-scourability, and 0～0.5 mm roots, soil clay and sand also indirectly affected anti-scourability by affecting soil aggregates. Soil sand and 0～1.0 mm roots of Stipa bungeana directly affected soil anti-scourability, and its specific fine roots also indirectly affected anti-scourability by affecting soil aggregates and organic carbon content. [Conclusion] There are obvious differences in the anti-scourability of the three kinds of herbaceous root-soil system. Roots and soil can affect their anti-scourability. The anti-scourability of different herbaceous (vegetation) should be considered in the ecological construction in the loess hilly area of southern Ningxia, and the scientific configuration should be carried out on the basis of improving the relevant influencing factors. Results: A total of 60 brachial plexuses were dissected, of which 49 (81.66%) had the usual anatomical formation of the trunks: the upper trunk formed by C5 and C6 roots, the middle trunk by the C7 root, and the lower trunk by the C8 and T1 roots. Eleven (18.34%) plexuses presented variations in trunk formation. Specifically, 8 (13.3%) brachial plexuses were of the prefix type, where the upper trunk was formed by the C5 and C6 roots with an additional contribution from the C4 root; there was inter-branch communication between C6 and C7 in one (1.7%) case; the middle and lower trunks united by C7, C8, and T1 roots formed the lower trunk in another (1.7%) case, and finally, one (1.7%) case exhibited the post-fix type. Conclusion: In this study, out of the 30 foetuses examined, 11 showed variations, which is not uncommon. The prefix type of brachial plexus is quite common among the possible variations, although anatomists and neurosurgeons cannot ignore the possibility of other variations, like post-fix type or communication between the roots of the brachial plexuses. Aims: Fokienia hodginsii is a threatened conifer tree species, known as the dominant nursery-grown species capable of colonizing the challenging woodland environments in southern China due to its strong root penetrating ability. The ecological phenotype of Fokienia hodginsii is not well documented during its breeding process, which limits the potential planting area and its ecological function. This study aims to understand how Fokienia hodginsii associates with microbes to conduct its key ecological function and provide a theoretical basis for further improving the forest nursery management of Fokienia hodginsii. Methods: This study explored the ecological traits of 11 main Fokienia hodginsii provenances in a homogeneous garden experiment by analyzing their nutrient utilization strategies and associated microbial features in the rhizosphere soil and roots. Results: The study found that the paramount difference in the rhizosphere soil among provenances is in Ca and Fe content. Some microbial communities, namely Crenarchaeota, Verrucomicrobiota, and Desulfobacterota, were positively correlated with the amounts of the soil nutrient elements, whereas Abditibacteriota and Dependentiae were negatively correlated. The abundance of N- and Fe-related bacteria in the Fu Jian Chang Ting (FJCT) provenance was significantly higher than that in other provenances, while the C-, P-, K-, and Mg-related fungal communities, respectively, had higher abundances in the FJCT, Fu Jian Long Yan (FJLY), Fu Jian Gu Tian (FJGT), and Fu Jian Xian You (FJXY) provenances than the others. The impacts of the Gui Zhou Li Ping (GZLP), Hu Nan Dao Xian (HNDX), Jiang Xi Shang Yao (JXSY), and Guang Dong Shi Xing (GDSX) provenances on the rhizosphere soil are similar, but the differences in nutrient utilization arise from the plant itself. Conversely, the root nutrient contents of the FJCT, Fu Jian You Xi (FJYX), Fu Jian An Xi (FJAX), FJLY, Fu Jian De Hua (FJDH), FJGT, and FJXY provenances are highly correlated with soil nutrient features. Conclusions: For the native provenances, their economic traits are better than the exotic provenances. The native provenances are more sensitive to local soil conditions, so they should benefit more from human interventions, rendering them more suitable for artificial cultivation. The growth of the exotic provenances is less affected by the soil environment, making them better suited for the ecological transformation of forest stands and soil improvement. Peanut (Arachis hypogaea L.) is an important oilseed crop worldwide. However, soil salinization becomes one of the main limiting factors of peanut production. Therefore, developing salt-tolerant varieties and understanding the molecular mechanisms of salt tolerance is important to protect peanut yield in saline areas. In this study, we selected four peanut varieties with contrasting response to salt challenges with T1 and T2 being tolerance and S1 and S2 being susceptible. High-throughput RNA sequencing resulted in more than 314.63 Gb of clean data from 48 samples. We identified 12,057 new genes, 7,971of which have functional annotations. KEGG pathway enrichment analysis of uniquely expressed genes in salt-tolerant peanut revealed that upregulated genes in the root are involved in the MAPK signaling pathway, fatty acid degradation, glycolysis/gluconeogenesis, and upregulated genes in the shoot were involved in plant hormone signal transduction and the MAPK signaling pathway. Na+ content, K+ content, K+/ Na+, and dry mass were measured in root and shoot tissues, and two gene co-expression networks were constructed based on weighted gene co-expression network analysis (WGCNA) in root and shoot. In this study, four key modules that are highly related to peanut salt tolerance in root and shoot were identified, plant hormone signal transduction, phenylpropanoid biosynthesis, starch and sucrose metabolism, flavonoid biosynthesis, carbon metabolism were identified as the key biological processes and metabolic pathways for improving peanut salt tolerance. The hub genes include genes encoding ion transport (such as HAK8, CNGCs, NHX, NCL1) protein, aquaporin protein, CIPK11 (CBL-interacting serine/threonine-protein kinase 11), LEA5 (late embryogenesis abundant protein), POD3 (peroxidase 3), transcription factor, and MAPKKK3. There were some new salt-tolerant genes identified in peanut, including cytochrome P450, vinorine synthase, sugar transport protein 13, NPF 4.5, IAA14, zinc finger CCCH domain-containing protein 62, beta-amylase, fatty acyl-CoA reductase 3, MLO-like protein 6, G-type lectin S-receptor-like serine/threonine-protein kinase, and kinesin-like protein KIN-7B. The identification of key modules, biological pathways, and hub genes in this study enhances our understanding of the molecular mechanisms underlying salt tolerance in peanuts. Soil salinization poses a significant threat to agricultural productivity, necessitating innovative agronomic strategies to mitigate its impact. This study focuses on improving salt stress resistance in tomato plants through the application of silicon (Si) in roots. A greenhouse experiment was carried out under normal conditions (control, and 1 and 4 mM Si) and under salinity stress (salt control, and 1 and 4 mM Si). Various parameters were analyzed in leaves and roots. Under normal conditions, tomato plants grown in non-saline conditions exhibited some toxicity when exposed to Na2SiO3. As for the experiments under salt stress conditions, Si mitigated oxidative damage, preserving root cell membrane integrity. The concentration of malondialdehyde was reduced by 69.5%, that of proline was reduced by 56.4% and there was a 57.6% decrease in catalase activity for tomato plants treated with 1 mM Si under salt stress. Furthermore, Fe uptake and distribution, under salt conditions, increased from 91 to 123 mg kg−1, the same concentration as that obtained for the normal control. In all cases, the lower dose produced better results under normal conditions than the 4 mM dose. Background: In complex teeth like maxillary premolars, endodontic treatment success depends on a complete comprehension of root canal anatomy. The research on mandibular premolars' root canal anatomy has been extensive and well-documented in existing literature. However, there appears to be a notable gap in available data concerning the root canal anatomy of maxillary premolars. This study aimed to explore the root canal morphology of maxillary premolars using cone-beam computed tomography (CBCT) imaging, considering age and gender variations. Methods: From 500 patient CBCT scans, 787 maxillary premolar teeth were evaluated. The sample was divided by gender and age (10–20, 21–30, 31–40, 41–50, 51–60, and 61 years and older). Ahmed et al. classification system was used to record root canal morphology. Results: The most frequent classifications for right maxillary 1st premolars were 2MPM1 B1 L1 (39.03%) and 1MPM1 (2.81%), while the most frequent classifications for right maxillary 2nd premolars were 2MPM1 B1 L1 (39.08%) and 1MPM1 (17.85%). Most of the premolars typically had two roots (left maxillary first premolars: 81.5%, left maxillary second premolars: 82.7%, right maxillary first premolars: 74.4%, right maxillary second premolars: 75.7%). Left and right maxillary 1st premolars for classes 1MPM1 and 1MPM1–2−1 showed significant gender differences. For classifications 1MPM1 and 1MPM1–2−1, age-related changes were seen in the left and right maxillary first premolars. Conclusion: This study provides novel insights into the root canal anatomy of maxillary premolars within the Saudi population, addressing a notable gap in the literature specific to this demographic. Through CBCT imaging and analysis of large sample sizes, the complex and diverse nature of root canal morphology in these teeth among Saudi individuals is elucidated. To improve nitrogen use efficiency (NUE) during rice cultivation, it is essential to comprehend the morphological and physiological traits of rice roots. However, in high-fertility black soil regions of Northeast China, the effects of combining biochar application with water-saving irrigation (WSI) conditions on rice root development and nitrogen utilization are still unknown. To address this knowledge gap, a combination of field experiments and 15N tracer micro-area investigations was conducted in this study. Four treatments were implemented: (i) controlled irrigation without biochar application (CB0); (ii) controlled irrigation with 2.5 t ha−1 biochar application (CB1); (iii) controlled irrigation with 12.5 t ha−1 biochar application (CB2); and (iv) controlled irrigation with 25 t ha–1 biochar application (CB3). Flooded irrigation conditions without biochar treatment (FB0) were used as the control. The primary objective of this research was to identify the mechanisms by which combined WSI conditions and biochar application affect rice root development and nitrogen utilization. Biochar application enhanced rice root morphological and physiological characteristics. Optimal biochar application increased the longest root length (RL), root volume (RV), root fresh weight (RFW), root active absorption area, root bleeding intensity, and root activity (RA) of rice while also optimizing the root–shoot ratio and facilitating nitrogen absorption by roots. These changes in root morphological and physiological characteristics facilitated the absorption of fertilizer-15N and soil nitrogen by rice roots, ultimately leading to improvements in rice yields and NUEs. Notably, the rice yields, NUE, nitrogen agronomic efficiency (NAE), and nitrogen partial factor productivity (NPFP) of CB2 plants were 16.45%, 39.42%, 24.48%, and 16.45% higher than those of FB0 plants, respectively. These results highlight the effectiveness of biochar application as a strategy to ensure food security and enhance NUE under WSI conditions. Cinmethylin is a potential pre-emergence herbicide that could be used to control grass weeds in winter cereals. To determine the herbicidal activity of cinmethylin against common gramineous weeds in wheat fields in China and its level of safety on wheat, we conducted the following experiments: (i) assessing the efficacy of cinmethylin against 11 grass weeds and (ii) determining its safety against 19 wheat varieties. The results showed that cinmethylin had good herbicidal efficacy against annual bluegrass (Poa annua L.), shortawn foxtail (Alopecurus aequalis Sobol.), slender meadow foxtail (Alopecurus myosuroides Huds.), Japanese foxtail (Alopecurus japonicus Steud.), Italian ryegrass (Lolium multiflorum Lam.), British timothy (Phleum paniculatum Huds.), Asia Minor bluegrass (Polypogon fugax Nees ex Steud.), Helictotrichon tibeticum (Roshev.) Holub., and wild oat (Avena fatua L.), with a GR50 (the herbicide dose resulting in 50% growth inhibition) value of 4.50–99.21 g a.i. ha−1 in plant height and 1.43–70.34 g a.i. ha−1 in fresh weight. However, cinmethylin cannot control Japanese brome (Bromus japonicus L.) or Tausch's goatgrass (Aegilops tauschii Coss.) at a dose of 200 g a.i. ha−1. Different wheat varieties varied in their phytotoxicity to cinmethylin. Overall, there is a phytotoxicity risk when using cinmethylin on wheat, mainly to wheat roots, with a reduction in root length of 40.81–64.09% at a dose of 400 g a.i. ha−1. These findings indicate that the pre-emergence herbicide cinmethylin provides good efficacy against most grass weeds and may possess potential for weed management in wheat fields. Simple Summary: The aim of this study was to analyze and compare the morphology of root canals in the carnassial teeth of German shepherds and mixed-breed dogs. Knowledge of the morphology of root canals is essential for a successful endodontic procedure. It has been determined that an apical delta, which represents a complex structure that consists of multiple cavities whose purpose is to allow the passing of nerves and blood vessels from the pulp cavity to the apex of the root, was present in 247 roots. There are four main types of apical delta. The most common type in superior fourth premolars was type II, with up to 10 apical ramifications, whereas type IIIA, with 10–20 apical ramifications, was most commonly present in inferior first molars. Root canal treatment of carnassial teeth in dogs is a common endodontic technique which aims to re-establish or maintain the health of the periapical tissues. In total, 43 dogs were used in this study. Root canal morphology was evaluated in 86 superior fourth premolars and 86 inferior first molars. Apical delta was present in 247 roots, while obliteration of the root canal was found in 11 roots. The most common type of apical delta of the roots of superior fourth premolars was type II, with up to 10 apical ramifications, while type IIIA, with 10–20 apical ramifications, was most commonly present in the roots of inferior first molars. Considering that knowledge of the morphology of root canals is essential for a successful endodontic procedure, the aim of this study was to analyze and compare the morphology of root canals in the carnassial teeth of German shepherds and mixed-breed dogs. Apical resection for the purpose of endodontic therapy of the superior fourth premolar and the inferior first molar is indicated at a length of 4 to 6 mm from the anatomical tip of the roots, which would completely remove the apical delta of these two teeth. The research was carried out between the months of November 2020 and March 2021 and aimed to estimate the crop coefficient, root depth and abatement factor, depending on thermal time. The research area belongs to the UBPC Grito de Yara, Granma, Cuba, which is located at 200 25' 02' N Latitude and 760 53' 27' O Longitude. The estimation of the crop coefficient, root growth and exhaustion fraction was carried out using curvilinear models as a function of degree days. The estimation of root growth in the bean crop, using the three models evaluated, depending on the degree days, is reliable and is a very useful indicator for irrigation programming, since it facilitates the timely estimation of the sheet of water from the depth of the root. Growing degree days explain the allowable depletion levels of soil water for bean cultivation; In this regard, it was confirmed that the curvilinear model is accurate, which allows irrigation management, using information on degree days, to avoid water stress. Summary: Phloem sieve elements (PSE), the primary conduits collaborating with neighboring phloem pole pericycle (PPP) cells to facilitate unloading in Arabidopsis roots, undergo a series of developmental stages before achieving maturation and functionality. However, the mechanism that maintains the proper progression of these differentiation stages remains largely unknown.We identified a gain‐of‐function mutant altered phloem pole pericycle 1 Dominant (app1D), producing a truncated, nuclear‐localized active form of NAC with Transmembrane Motif 1‐like (NTL9). This mutation leads to ectopic expression of its downstream target CALLOSE SYNTHASE 8 (CalS8), thereby inducing callose accumulation, impeding SE differentiation, impairing phloem transport, and inhibiting root growth.The app1D phenotype could be reproduced by blocking the symplastic channels of cells within APP1 expression domain in wild‐type (WT) roots. The WT APP1 is primarily membrane‐tethered and dormant in the root meristem cells but entries into the nucleus in several cells in PPP near the unloading region, and this import is inhibited by blocking the symplastic intercellular transport in differentiating SE.Our results suggest a potential maintenance mechanism involving an APP1‐CalS8 module, which induces CalS8 expression and modulates symplastic communication, and the proper activation of this module is crucial for the successful differentiation of SE in the Arabidopsis root. Starch extraction from tubers and roots has long been an essential process, playing a crucial role in diverse industries ranging from alimentary to pharmacology. This review explores the different methods employed in starch extraction, including traditional techniques and the most innovative mechanical strategies. The methods show a good improvement in many aspects, such as an improvement in the efficiency of the process and an improvement in the yield, showing a value of 10.0–65.0% depending on the starch source. On the other hand, solvents such as NaOH are used in many mechanical processes for alkaline digestion to improve the extraction time. Ethanol and K2S2O5 concentrations of 0.5% and 0.8% were used to prevent oxidation and modify some properties of the extracted starch. The use of many solvents has improved the optimization of the processes, providing the final extracted starch with more advantages and better quality. However, using enzymes such as cellulase in new and innovative ways has provided more advantages and a better efficiency and yield than the other methods. Each method has its advantages and challenges, highlighting the importance of understanding the diversity of different approaches and their impact on the yield, sustainability, environmental considerations, and quality of the extracted starch. As the world looks for more ecological approaches, this review shows the importance of critically evaluating the yield, efficiency, and environmental implications of the extraction methods, providing us with more ways of evaluating the methods used for starch extraction. The ecological impact is a crucial point when evaluating the innovation of a new extraction process, which is why methods such as ultrasound and pulsed electric-field-assisted techniques have been proposed. These methods have been presented as sustainable techniques called green technologies, offering more approaches and different advantages than the other methods. The presence of epistasis in complex traits can be significant and affect the selection of segregating populations undesirably. The purpose of this study was to determine the epistatic genetic components that influence root and shoot traits of common bean and identify their effect on the performance of segregating populations. The field experiment consisted of 49 treatments (backcross progenies, parents and segregating populations in the F2 and F3 generations). The cross P1-BAF53 (Andean) × P2-IPR 88 Uirapuru (Mesoamerican) as reference. Six traits were taken into consideration: root distribution (%), first pod length (cm), number of grains (plot), plant height (cm), reproductive cycle (days) and number of basal branches (plot). For root distribution, first pod length and number of grains, the additive and additive × additive components were significant. On the contrary, for plant height, reproductive cycle and number of basal branches, no interaction component was observed. These results can be explained by the magnitude of variance determined for these traits. The additive × additive epistasis had a negative influence on the mean performance of segregating progenies. Contrariwise, transgressive segregation effects on plant height were observed as a function of genetic dominance deviation. Thus, additive × additive epistasis can hamper the success of a breeding program, for being associated with low-variability traits. This fact may be related with the number of crosses and/or the choice of genetically more distant parents for the program, and possibly with the presence of linked genes. [ABSTRACT FROM AUTHOR]

Protein disulfide isomerase (PDI, EC 5.3.4.1) is a thiol-disulfide oxidoreductase that plays a crucial role in catalyzing the oxidation and rearrangement of disulfides in substrate proteins. In plants, PDI is primarily involved in regulating seed germination and development, facilitating the oxidative folding of storage proteins in the endosperm, and also contributing to the formation of pollen. However, the role of PDI in root growth has not been previously studied. This research investigated the impact of PDI gene deficiency in plants by using 16F16 [2-(2-Chloroacetyl)-2,3,4,9-tetrahydro-1-methyl-1H-pyrido[3,4-b]indole-1-carboxylic acid methyl ester], a small-molecule inhibitor of PDI, to remove functional redundancy. The results showed that the growth of Arabidopsis roots was significantly inhibited when treated with 16F16. To further investigate the effects of 16F16 treatment, we conducted expression profiling of treated roots using RNA sequencing and a Tandem Mass Tag (TMT)-based quantitative proteomics approach at both the transcriptomic and proteomic levels. Our analysis revealed 994 differentially expressed genes (DEGs) at the transcript level, which were predominantly enriched in pathways associated with "phenylpropane biosynthesis", "plant hormone signal transduction", "plant−pathogen interaction" and "starch and sucrose metabolism" pathways. Additionally, we identified 120 differentially expressed proteins (DEPs) at the protein level. These proteins were mainly enriched in pathways such as "phenylpropanoid biosynthesis", "photosynthesis", "biosynthesis of various plant secondary metabolites", and "biosynthesis of secondary metabolites" pathways. The comprehensive transcriptome and proteome analyses revealed a regulatory network for root shortening in Arabidopsis seedlings under 16F16 treatment, mainly involving phenylpropane biosynthesis and plant hormone signal transduction pathways. This study enhances our understanding of the significant role of PDIs in Arabidopsis root growth and provides insights into the regulatory mechanisms of root shortening following 16F16 treatment. Humic substances (HS), a natural polymer isomeric mixture, is an important component of terrestrial ecosystem. Its promotion effect on plant growth has been widely studied in recent years. Multiple signaling pathways play an important role in the process of HS promoting plant growth and development. The article summarizes the signaling pathway transduction and crosstalk mechanisms of HS in promoting the growth and development of plant roots and shoots. During the promotion of root growth, HS mainly involves the transduction of Indole-3-acetic acid (IAA), nitric oxide (NO), abscisic acid (ABA), reactive oxygen species (ROS) and Ca2+ signal, and the complex crosstalk between them; During the promotion of shoot growth, HS mainly involves the transmission of cytokinin (CTK) and ABA signaling pathways, and the complex crosstalk occurs between nitrate and CTK signaling pathways. HS regulates plant physiological metabolism through a complex metabolic network, involving multiple complicated signaling pathways and crosstalk processes. Among them, (PM) H+- ATPase is the key node of HS promoting root and shoot growth, and multiple signaling pathways mediate the signal transduction of cascade amplification via (PM) H+- ATPase. NO signal has a complex crosstalk process with multiple signaling pathways that regulate root and shoot growth, and is an important crosstalk signaling molecule involved in the promotion of plant growth by HS. There are other signaling pathways involved in the growth and development of roots and shoots triggered by HS, among which the defense signaling pathways salicylicacid (SA) and jasmonic acid (JA) are regulated by signaling pathways IAA, NO, ABA and CTK. Due to the dispersion, heterogeneity and complexity of biochemical research of HS, the relevant mechanism of the short-term promoting effect of HS on plant growth and the overall action mechanism on plant growth and development during the whole growth stage have not yet been clarified. Therefore, further in-depth analysis is needed to reveal the signal transduction, crossover, crosstalk and integration pathways involved in the growth promotion process, and the complex nutritional and metabolic pathways. Introduction: Potato is the one of the important crop in the world. This plant in terms of consumption rate in the world levels is after rice and wheat. Thus, this crop is considered the maintain food security. However, potato influences the abiotic stress particularly drought and cold. The C-repeat binding factors (CBFs), also termed dehydration-responsive element-binding protein 1 (DREB1) family members, play crucial roles in the acquisition of stress tolerance, but in potato, the underlying mechanisms of stress tolerance remain elusive. The cold and drought stresses are the most abiotic stresses across the world causing to decrease of the crop yield. To gain insight into these mechanisms, CBF1 gene was selected from four potato and was assessed their expression under drought and cold stress conditions. Materials and methods: In order to investigate the effect of cold and drought on growth and development on potato, a experiment was conducted at the the Seed and Plant Improvement Institute located at Karaj, Iran. The experimental design used was randomized complete block design with three replications under drought and cold stress (normal and stressed). Gene expression of CBF1 gene was performed in different tissues (root, stem, leaves, and tuber) under control condiotin. Further, analysis of expression in root and leaves under stress condition. After 3 weeks after cultivated in Karaj, the water stress was induced by irrigation suppression in 2 weeks. sampling was performed 5 weeks after sprouting under drought. Under cold stress, analysis of expression in root and leaves was performed at the 24h and 48h after 3 weeks of sprouting. CBF1 gene was selected as one of important gene in the investigation of molecular mechanism under cold and heat stress in potato. Results and discussion: Average induced expression in response to cold suggested a correlation between potato CBF1 expression and the acquisition of cold tolerance in root. Whereas, the expression of this gene was decreased in leaves. Under drought stress, the expression of CBF1 gene was up-regulated in root. However, the expression rate of CBF1 decreased under stress. Responses that varied between sections may reflect tissue-specific stress tolerance mechanisms, suggesting an effect of ecological context on the development of CBF1 mediated stress tolerance in potato. In this study, the investigation of transcription factor binding sites, co-expressed genes, and gene ontology of CBF1 in potato was performed. This gene is involved in multiple biological processes and pathways. The analysis of promoter showed that CBF1 gene had the highest and the lowest number of the TFBS in MYB and WRKY, respectively. The present study revealed that the CBF1 gene expression in root was the higher than leaves. Conclusion: Overall, it can be concluded that CBF1 gene can contribute to cold and drought tolerance in potato and can be used for genetic and breeding manipulation to improve tolerance to stress.
