Several members of the transforming growth factor beta (TGF‐β) superfamily regulate the proliferation, differentiation, and function of bone‐forming osteoblasts and bone‐resorbing osteoclasts. However, it is still unknown whether Nodal, a member of the TGF‐β superfamily, serves a function in bone cells. In this study, we found that Nodal did not have any function in osteoblasts but instead negatively regulated osteoclast differentiation. Nodal inhibited RANKL‐induced osteoclast differentiation by downregulating the expression of pro‐osteoclastogenic genes, including c‐fos, Nfatc1, and Blimp1, and upregulating the expression of antiosteoclastogenic genes, including Bcl6 and Irf8. Nodal activated STAT1 in osteoclast precursor cells, and STAT1 downregulation significantly reduced the inhibitory effect of Nodal on osteoclast differentiation. These findings indicate that Nodal activates STAT1 to downregulate or upregulate the expression of pro‐osteoclastogenic or antiosteoclastogenic genes, respectively, leading to the inhibition of osteoclast differentiation. Global energy policies have urged power consumption reductions for electronic devices. However, this is inconsistent with the users' need for enhanced device performance. In this work, converse flexoelectricity is used to reduce power consumption while maintaining the superior display image quality of liquid crystal display (LCD). The first 1 Hz‐refresh‐rate and flicker‐free fringe‐field switching (FFS) LCD for a 13.4‐inch laptop are reported. A noticeable brightness deviation is inevitable when driving voltage polarity is altered during low‐frequency driving, which is caused by the flexoelectric effect and voltage‐holding failure. The nature of the converse flexoelectric contribution of mesogenic systems with the optimum flexoelectric coefficients and molecular polarity, and developed materials based on findings is investigated. The quality of static images and videos at 1‐ and 10 Hz refresh rates, respectively, is maintained, and the measured power consumptions are reduced by 83% and 47% compared to that at a 60 Hz refresh rate, and minimal flicker level is measured by −68 dB. Human monocyte chemoattractant protein‐1 (MCP‐1) in mice has two orthologs, MCP‐1 and MCP‐5. MCP‐1, which is highly expressed in osteoclasts rather than in osteoclast precursor cells, is an important factor in osteoclast differentiation. However, the roles of MCP‐5 in osteoclasts are completely unknown. In this study, contrary to MCP‐1, MCP‐5 was downregulated during receptor activator of nuclear factor kappa B ligand (RANKL)‐induced osteoclast differentiation and was considered an inhibitory factor in osteoclast differentiation. The inhibitory role of MCP‐5 in osteoclast differentiation was closely related to the increase in Ccr5 expression and the inhibition of IκB degradation by RANKL. Transgenic mice expressing MCP‐5 controlled by Mx‐1 promoter exhibited an increased bone mass because of a decrease in osteoclasts. This result strongly supported that MCP‐5 negatively regulated osteoclast differentiation. MCP‐5 also prevented severe bone loss caused by RANKL. [ABSTRACT FROM AUTHOR]

MXenes possess the characteristics required for high‐performance supercapacitors, such as high metallic conductivity and electrochemical activity, but their full potential remains unrealized owing to their tendency to self‐restack when fabricated into an electrode. Designing an MXene interlayer with an effective intercalant has, therefore, become an important criterion to alleviate the restacking issue while also synergistically interact with MXene to further improve its electrochemical activity. This study reports the intercalation of 1D π‐d conjugated coordination polymer (Ni‐BTA) within the Ti3C2Tx nanosheet for the fabrication of a highly efficient supercapacitor electrode. Ni‐BTA, which consists of a nickel center and 1,2,4,5‐benzenetetramine (BTA) organic chain, is uniformly intercalated by direct synthesis on the abundant oxygen terminals on the Ti3C2Tx nanosheet surface. The intercalated Ni‐BTA acts as an effective charge carrier transportation pathway through its 1D stretched delocalized π‐d electrons while participating in pseudocapacitive activity with the Ni centers. As a result, the Ni‐BTA/MXene film exhibits excellent rate performance and a gravimetric specific capacitance of 264.4 F g−1 at 5 mV s−1. This magnitude is retained up to 94.6% after 10 000 cycles. Cav3.1 T‐type Ca2+ channels play pivotal roles in neuronal low‐threshold spikes, visceral pain, and pacemaker activity. Phosphorylation has been reported to potently regulate the activity and gating properties of Cav3.1 channels. However, systematic identification of phosphorylation sites (phosphosites) in Cav3.1 channel has been poorly investigated. In this work, we analyzed rat Cav3.1 protein expressed in HEK‐293 cells by mass spectrometry, identified 30 phosphosites located at the cytoplasmic regions, and illustrated them as a Cav3.1 phosphorylation map which includes the reported mouse Cav3.1 phosphosites. Site‐directed mutagenesis of the phosphosites to Ala residues and functional analysis of the phospho‐silent Cav3.1 mutants expressed in Xenopus oocytes showed that the phospho‐silent mutation of the N‐terminal Ser18 reduced its current amplitude with accelerated current kinetics and negatively shifted channel availability. Remarkably, the phospho‐silent mutations of the C‐terminal Ser residues (Ser1924, Ser2001, Ser2163, Ser2166, or Ser2189) greatly reduced their current amplitude without altering the voltage‐ dependent gating properties. In contrast, the phosphomimetic Asp mutations of Cav3.1 on the N‐ and C‐terminal Ser residues reversed the effects of the phospho‐ silent mutations. Collectively, these findings demonstrate that the multiple phosphosites of Cav3.1 at the N‐ and C‐terminal regions play crucial roles in the regulation of the channel activity and voltage‐dependent gating properties. Background: The association between diabetes mellitus (DM) and bone diseases is acknowledged. However, the mechanistic pathways leading to the alveolar bone (AB) destruction remain unclear. This study aims to elucidate the mechanical forces (MF)-induced AB destruction in DM and its underlying mechanism.Methods: In vivo periodontal tissue responses to MF were evaluated in rats with diabetes. In vitro human periodontal ligament (PDL) cells were either treated with advanced glycation end products (AGEs) alone or with AGEs and MF.Results: In vivo, the transcription of VEGF-A, colony stimulating factor-1 (CSF-1), and Ager was upregulated in diabetes, whereas changes in DDOST and Glo1 mRNAs were negligible. DM induced VEGF-A protein in the vascular cells of the PDL and subsequent angiogenesis, but DM itself did not induce osteoclastogenesis. MF-induced AB resorption was augmented in DM, and such augmentation was morphologically substantiated by the occasional undermining resorption as well as the frontal resorption of the AB by osteoclasts. The mRNA levels of CSF-1 and vascular endothelial growth factor (VEGF) during MF application were highly elevated in diabetes, compared with those of the normal counterparts. In vitro, AGEs treatment elevated Glut-1 and CSF-1 mRNA levels via the p38 and JNK pathways, whereas OGT and VEGF levels remained unchanged. Compressive MF especially caused upregulation of VEGF, CSF-1, and Glut-1 levels, and such upregulation was further enhanced by AGEs treatment.Conclusions: Overloaded MF and AGEs metabolites may synergistically aggravate AB destruction by upregulating CSF-1 and VEGF. Therefore, regulating the compressive overloading of teeth, as well as the levels of diabetic AGEs, may prove to be an effective therapeutic modality for managing DM-induced AB destruction. Stretchable organic electronics open opportunities for novel applications in next‐generation intelligent electronics, such as human‐friendly, skin‐attachable, and implantable devices. Stretchable transparent electrodes with appropriate electrical, mechanical, and optical properties as well as high conformability to curved surfaces and human skin are key components of wearable and implantable devices. Herein, a silver nanowire (AgNW)‐embedded chitosan biopolymer hybrid film is presented as a high‐performance stretchable transparent electrode. The introduction of an organic surface modifier and embedding the AgNW network results in remarkably improved adhesion between the nanowires and substrate; consequently, the electrical properties and mechanical performance of the stretchable films are improved simultaneously. The fabricated hybrid films show high transmittance (89.0% at 550 nm), low sheet resistance (8.4 ohm sq−1), and limited resistance change under tensile strains. Moreover, these stretchable transparent electrodes with versatile applicability are integrated into multifunctional stretchable electronic devices such as heaters, strain sensors, and light‐emitting devices, which show excellent thermal, strain sensing, and light‐emitting properties, respectively. In addition, they conform to curved surfaces or human skin and maintain their mechanical robustness. The proposed strategy offers new opportunities for stretchable and biocompatible organic electronics, particularly for skin‐attachable, visually imperceptible thermal management devices, strain sensors, and light‐emitting devices. 2D transition metal carbides and nitrides, namely MXenes, are normally synthesized in acidic solutions and are delaminated in basic solutions. This results in versatile materials with unique physical/chemical properties suitable for various practical applications. However, solution‐based chemical treatments can affect the chemical structures of MXenes, which accelerates the oxidation reactions and degrades their intrinsic properties. Here, long‐term stable Ti3C2Tx dispersion in deep eutectic solvents (DESs) that resisted oxidation degradation for up to 28 weeks is demonstrated. As an anti‐oxidative dispersion medium, DESs helped prevent oxidation of Ti3C2Tx layers due to hydrogen bond accepting and donating molecules passivated surface of the Ti3C2Tx. In addition, DES molecules in bulk solution can also be hydrated in the presence of water, which stabilizes reactive oxygen by forming stable DES‐water cluster. Therefore, the use of DESs enhanced the delamination of the Ti3C2Tx nanosheets, while preventing oxidation of the nanosheets in solution and even in their dried state. As a result, thick and thin films of Ti3C2Tx fabricated using DESs exhibited stable sheet resistance in comparison with pristine‐Ti3C2Tx. In addition, Ti3C2Tx dispersed in DESs can be applied as electrodes for electrochemical capacitors, in which they showed higher chemical stability and better performance than pristine Ti3C2Tx. Introduction: The association between nutrition and oral health has been studied in the elderly. This study aimed to examine the impacts of undernutrition and the mothers' socioeconomic and oral health statuses on the incidence of dental caries in Korean preschool children. Methods: Data of 610 children aged 3‐5 years and their mothers who underwent oral examinations and responded to the questionnaires in the 6th KNAHNES were used. Caries prevalence was measured by dft and dt among the primary teeth in children and DMFT among mothers. Dietary reference intake values were used to evaluate nutritional status, a nutritional quality index and the mean nutrient adequacy ratio. Complex sample correlation analysis was performed by using children's dft and dt statuses as dependent variables. Multilevel linear regression was applied to investigate the impacts of undernutrition and mothers' socioeconomic and oral health status on children's dft and dt statuses. Statistical significance was set as P <.05. Results: Factors significantly related to dft in children were age and food. Food insecurity was the only factor significantly associated with dt in children. Children's sex and mother's DMFT were likely to be relevant to dft in children. Children's age and a nutritional quality index value less than 1 were also likely to be relevant to the dt of children. Conclusions: Children with an undernourished status had a higher caries experience. A comprehensive community dental health promotion programme should be developed to prevent the incidence of dental caries in vulnerable undernourished children. [ABSTRACT FROM AUTHOR]

Although energy‐storage devices based on Li ions are considered as the most prominent candidates for immediate application in the near future, concerns with regard to their stability, safety, and environmental impact still remain. As a solution, the development of all‐solid‐state energy‐storage devices with enhanced stability is proposed. A new eco‐friendly polymer electrolyte has been synthesized by incorporating lithium trifluoromethanesulfonate into chemically modified methyl cellulose (LiTFS–LiSMC). The transparent and flexible electrolyte exhibits a good conductivity of near 1 mS cm−1. An all‐solid‐state supercapacitor fabricated from 20 wt % LiTFS–LiSMC shows comparable specific capacitances to a standard liquid‐electrolyte supercapacitor and an excellent stability even after 20 000 charge–discharge cycles. The electrolyte is also compatible with patterned carbon, which enables the simple fabrication of micro‐supercapacitors. In addition, the LiTFS–LiSMC electrolyte can be recycled and reused more than 20 times with negligible change in its performance. Purpose: Kilovoltage intrafraction monitoring (KIM) allows for real‐time image guidance for tracking tumor motion in six‐degrees‐of‐freedom (6DoF) on a standard linear accelerator. This study assessed the geometric accuracy and precision of KIM used to guide patient treatments in the TROG 15.01 multi‐institutional Stereotactic Prostate Ablative Radiotherapy with KIM trial and investigated factors affecting accuracy and precision. Methods: Fractions from 44 patients with prostate cancer treated using KIM‐guided SBRT were analyzed across four institutions, on two different linear accelerator models and two different beam models (6 MV and 10 MV FFF). The geometric accuracy and precision of KIM was assessed from over 33 000 images (translation) and over 9000 images (rotation) by comparing the real‐time measured motion to retrospective kV/MV triangulation. Factors potentially affecting accuracy, including contrast‐to‐noise ratio (CNR) of kV images and incorrect marker segmentation, were also investigated. Results: The geometric accuracy and precision did not depend on treatment institution, beam model or motion magnitude, but was correlated with gantry angle. The centroid geometric accuracy and precision of the KIM system for SABR prostate treatments was 0.0 ± 0.5, 0.0 ± 0.4 and 0.1 ± 0.3 mm for translation, and −0.1 ± 0.6°, −0.1 ± 1.4° and −0.1 ± 1.0° for rotation in the AP, LR and SI directions respectively. Centroid geometric error exceeded 2 mm for 0.05% of this dataset. No significant relationship was found between large geometric error and CNR or marker segmentation correlation. Conclusions: This study demonstrated the ability of KIM to locate the prostate with accuracy below other uncertainties in radiotherapy treatments, and the feasibility for KIM to be implemented across multiple institutions. Bromelain (BR), a protease extracted from Ananas comosus, reportedly possesses pharmacological activities including the reduction of thrombogenesis, and antihypertensive, and antimicrobial effects. This study aimed to investigate the potential effects of BR on oral cancer cells. The effect of BR on the viability of Ca9‐22 and SCC25 cells was determined using the MTT assay. These cells were also treated with different doses of BR, and Western blotting was conducted to monitor apoptosis. Finally, flow cytometry analysis was performed to identify sub‐G1 populations of oral cancer cells. After treatment, the viability of both Ca9‐22 and SCC25 cells was markedly reduced, in a dose‐dependent manner. BR induced poly (ADP‐ribose) polymerase (PARP) and lamin A/C degradation, and generated cleavage products. Flow cytometry analysis showed that BR treatment significantly increased the sub‐G1 population. Our findings therefore indicate that BR has potential as a novel, natural anticarcinogenic medicine. Gestational vitamin D insufficiency is related with increased risks of various diseases and poor health outcomes later in life. Telomere length at birth or early in life is known to be a predictor of individual health. Both vitamin D and telomere length are related with various health conditions, and vitamin D concentrations are associated with leukocyte telomere lengths in women. We investigated the association between maternal vitamin D concentrations and newborn leukocyte telomere lengths. This cross-sectional study included 106 healthy pregnant women without adverse obstetric outcomes and their offspring. We examined the maternal age, weight before pregnancy, health behaviours, and nutritional intakes, along with each newborn's sex and birthweight, and we measured maternal height, telomere length, total white blood cell count, and glycosylated haemoglobin as covariates. Pearson's correlation coefficients were calculated to evaluate the relationship between the baseline variables and newborn leukocyte telomere lengths. To confirm that there was an independent association between newborn leukocyte telomere lengths and maternal vitamin D concentrations, we performed a stepwise multiple linear regression analysis. Newborn leukocyte telomere lengths correlated positively with maternal leukocyte telomere lengths ( r = .76, p < .01), maternal 25-hydroxyvitamin D concentrations ( r = .72, p < .01), maternal energy intakes ( r = .22, p = .03), and newborn body weights ( r = .51, p < .01). In the multivariate model, newborn leukocyte telomere lengths were associated with maternal vitamin D concentrations (β = .33, p < .01). These findings suggest that the maternal vitamin D concentration during pregnancy may be a significant determinant of the offspring's telomere length. [ABSTRACT FROM AUTHOR]

Purpose: Although current computed tomography (CT) systems can scan the head in a very short time, patient motion sometimes still induces artifacts. If motion occurs, one has to repeat the scan; to avoid motion, sedation or anesthesia is sometimes applied. Methods: The authors propose a method to iteratively estimate and compensate this motion during the reconstruction. In every iteration, the rigid motion was estimated view-by-view and then used to update the system matrix. A multiresolution scheme was used to speed up the convergence of this joint estimation of the image and the motion of the subject. A final iterative reconstruction was performed with the last motion estimate. Results: The method was evaluated on simulations, patient scans, and a phantom study. The quality of the reconstructed images was improved substantially after the compensation. In simulation and phantom studies, root-mean-square error was reduced and mean structural similarity was increased. In the patient studies, most of the motion blurring in the reconstructed images disappeared after the compensation. Conclusions: The proposed method effectively eliminated motion-induced artifacts in head CT scans. Since only measured raw data are needed for the motion estimation and compensation, the proposed method can be applied retrospectively to clinical helical CT scans affected by motion. The morphology and conductivity of poly(styrenephosphonate- b-methylbutylene) block copolymers containing ionic liquids are investigated. The block copolymers display a series of well-defined self-assembled morphologies, i.e., lamellae, gyroid, hexagonal cylinder (HEX), body-centered cubic, and A15 lattice, in the absence and presence of ionic liquids. The observation of an equilibrium A15 lattice for linear diblock copolymers, in contrast to a number of theoretical studies, is ascribed to the packing frustration accompanied by prevailing electrostatic interactions. The samples having the A15 lattice show a substantially higher morphology factors (0.83-0.96) than those with the HEX phases (0.42-0.69). This indicates the structural advantage of the A15 lattice with a well-defined 3D symmetry over the 2D HEX structure for constructing less tortuous ion-conduction pathways. ABSTRACT Mammalian sterile 20-like kinase 2 (Mst2) plays a central role in the Hippo pathway, controlling cell proliferation, differentiation, and apoptosis during development. However, the roles of Mst2 in osteoclast and osteoblast development are largely unknown. Here, we demonstrate that mice deficient in Mst2 exhibit osteoporotic phenotypes with increased numbers of osteoclasts and decreased numbers of osteoblasts as shown by micro-computed tomography (µCT) and histomorphometric analyses. Osteoclast precursors lacking Mst2 exhibit increased osteoclastogenesis and Nfatc1, Acp5, and Oscar expression in response to receptor activator of NF-κB ligand (RANKL) exposure. Conversely, Mst2 overexpression in osteoclast precursors leads to the inhibition of RANKL-induced osteoclast differentiation. Osteoblast precursors deficient in Mst2 exhibit attenuated osteoblast differentiation and function by downregulating the expression of Runx2, Alpl, Ibsp, and Bglap. Conversely, ectopic expression of Mst2 in osteoblast precursors increases osteoblastogenesis. Finally, we demonstrate that the NF-κB pathway is activated by Mst2 deficiency during osteoclast and osteoblast development. Our findings suggest that Mst2 is involved in bone homeostasis, functioning as a reciprocal regulator of osteoclast and osteoblast differentiation through the NF-κB pathway. © 2015 American Society for Bone and Mineral Research. Carcinoembryonic antigen-related cell adhesion molecule (CEACAM) 1 and CEACAM20, immunoglobulin superfamily members, are predominantly expressed in intestinal epithelial cells (IECs) and co-localized at the apical surface of these cells. We here showed that the expression of mouse CEACAM1 and CEACAM20 at both mRNA and protein levels was markedly reduced in IECs of the small intestine by the treatment of mice with antibiotics against Gram-positive bacteria. The expression of both proteins was also decreased in IECs of the small intestine from germ-free mice, compared with that from control specific-pathogen-free mice. Exposure of intestinal organoids to IFN-γ markedly increased the expression of either CEACAM1 or CEACAM20, whereas the exposure to TNF-α increased the expression of the former protein, but not that of the latter. In contrast, the expression of CEACAM20, but not of CEACAM1, in intestinal organoids was markedly increased by exposure to butyrate, a short-chain fatty acid produced by bacterial fermentation in the intestine. Collectively, our results suggest that Gram-positive bacteria promote the mRNA expression of CEACAM1 or CEACAM20 in the small intestine. Background The 1550 nm Er:Glass fractional laser is widely used for the treatment of atrophic acne scar. A novel fractional radiofrequency microneedle device has recently emerged as an alternative for treating acne scars. Objectives To evaluate the clinical efficacy and safety of a Er:Glass fractional laser and fractional radiofrequency microneedle device in the treatment of facial atrophic acne scars and to assess the difference between the treatment modalities depending on facial compartment. Methods A total of 40 patients were equally randomized into two groups. Each group of 20 patients received three treatments at 4-week interval using Er:Glass fractional laser or fractional radiofrequency microneedle device. Results Scar severity scores ( ECCA grading scale) improved by a mean of 25.0% and 18.6% in groups A and B, respectively (both P < 0.01). The difference in the degree of improvement was not statistically significant between the groups after three sessions of treatment. There were no significant side effects. Conclusion Atrophic acne scars improved in both groups without significant side effects. Additionally, the fractional laser was a more effective treatment option for acne scars, but the fractional radiofrequency microneedle device offered good adherence and short downtime. Objective: To evaluate the risk for postoperative inguinal hernia according to the presence of patent processus vaginalis in an adult population. Methods: Medical records of 205 patients who underwent robot-assisted laparoscopic radical prostatectomy from May 2007 to November 2011 were reviewed. Age, prostate-specific antigen, prostate volume, body mass index, operative time and history of previous abdominal surgery were evaluated. The existence of patent processus vaginalis was also evaluated for the development of postoperative inguinal hernia. Results: Postoperative inguinal hernia occurred in 20 out of 410 (4.9%) groins (17/205 patients; 8.3%), and patent processus vaginalis was observed in 49 out of 410 (11.9%) groins. In the normal groin group, inguinal hernia occurred in seven out of 361 (1.9%) groins. However, in the patent processus vaginalis group, it occurred in 13 out of 49 (26.5%) groins. On univariate analysis using Cox proportional hazards model, age, body mass index, history of previous abdominal surgery and patent processus vaginalis were significant risk factors. Among them, patent processus vaginalis significantly increased the risk of postoperative inguinal hernia in multivariate analysis (hazard ratio 22.37). In the patent processus vaginalis group, inguinal hernia developed at 12.9 ± 9.2 months after robot-assisted laparoscopic radical prostatectomy and 15 ± 7.4 months in the normal groin group. Inguinal hernia-free ratios were significantly lower in the patent processus vaginalis group than the normal groin group ( P < 0.001). Conclusions: The existence of patent processus vaginalis represents an important risk factor for postoperative inguinal hernia in adults. We study the opto-electrical properties of Natronomonas pharaonis sensory rhodopsin II (NpSRII) by using a near-field microwave microprobe (NFMM) under external light illumination. To investigate the possibility of application of NFMM to biological macromolecules, we used time dependent properties of NPSRII before/after light activation which has three distinct states - ground-state, M-state, and O-state. The diagnostic ability of NFMM is demonstrated by measuring the microwave reflection coefficient ( S11) spectrum of NpSRII under steady-state illumination in the wavelength range of 350-650 nm. Moreover, we present microwave reflection coefficient S11 spectra in the same wavelength range for two fast-photocycling rhodopsins: green light-absorbing proteorhodopsin (GPR) and Gloeobacter rhodopsin (GR). In addition the frequency sweep shift can be detected completely even for tiny amounts of sample (∼10-3 OD of rhodopsin). Triplicate groups of wild and farmed purebred offspring and their reciprocal hybrids in olive flounder, Paralichthys olivaceus (body weight: 3.8-4.9 g), were fed to apparent satiation for 112 d. Body weight, feed intake, and feed efficiency were analyzed as repeated measures, with initial weight as a covariate. The effect of cross type on body weight and feed intake was highly significant ( P < 0.0001). Final body weight was highest in the farmed purebreds and lowest in the wild purebreds among the four crosses. The farmed purebreds had the highest feed intake during the initial 28 d. Feed efficiency was independent of cross type ( P > 0.1). At the end of the feeding trial, the n-3 highly unsaturated fatty acid content was highest in the hybrid cross type from farmed females and wild males, and consequently, crossbreeding could be advantage for the trait. The serum alanine aminotransferase activity was highest in the wild purebreds among crosses. This result may be involved in growth inferiority of wild purebreds, following the deficiencies in artificial diet adaptability. We suggest that the growth differences between the purebred wild and farmed olive flounder result from the differences in relative feed intake, and appear to be based on additive genetic variation due to mid-parent values of hybrid. Interleukin-3 (IL-3) is produced under various pathological conditions and is thought to be involved in the pathogenesis of inflammatory diseases; however, its function in bone homeostasis under normal conditions or nature of the downstream molecular targets remains unknown. Here we examined the effect of IL-3 on osteoclast differentiation from mouse and human bone marrow-derived macrophages (BMMs). Although IL-3 can induce osteoclast differentiation of multiple myeloma bone marrow cells, IL-3 greatly inhibited osteoclast differentiation of human BMMs isolated from healthy donors. These inhibitory effects of IL-3 were only observed at early time points (days 0 and 1). IL-3 inhibited the expression of c-Fos and NFATc1 in BMMs treated with RANKL. However, IL-3-mediated inhibition of osteoclast differentiation was not completely reversed by ectopic expression of c-Fos or NFATc1. Importantly, IL-3 induced inhibitor of DNA binding/differentiation (Id)1 in hBMMs, while Id2 were sustained during osteoclast differentiation of mBMMs treated with IL-3. Ectopic expression of NFATc1 in Id2-deficient BMMs completely reversed the inhibitory effect of IL-3 on osteoclast differentiation. Furthermore, inflammation-induced bone erosion was markedly inhibited by IL-3 administration. Taken together, our results suggest that IL-3 plays an inhibitory role in osteoclast differentiation by regulating c-Fos and Ids, and also exerts anti-bone erosion effects. J. Cell. Individual pharmacokinetic differences for herb-drug interaction have been associated with genetic variations of the multidrug resistance (MDR) gene. A high level expression of MDR protein increases cellular efflux and might decrease drug sensitivity. This study investigated the drug efflux activity difference of human MDR1 triallelic variant 2677G/T/A (rs2032582), as a nonsynonymous 893Ala/Ser/Thr, using Xenopus laevis oocytes and MDR1 overexpressing LLC-PK1 cells. Two MDR1 variants (2667T/893Ser and 2667A/893Thr) were generated using human MDR1 cDNA (2677G/893Ala). No significant difference in the expression of MDR1 893Ala/Ser/Thr was found in X. laevis oocytes. However, the MDR1 2667A/893Thr variant interestingly showed a significant decrease of efflux activity for both digoxin and daunorubicin compared with those of 893Ala and 893Ser variants. In further investigation assessing the inhibitory effects of three herbal extracts on MDR1, 893Ala and 893Ser showed significant decreases of efflux activities in treatments with P. cocos ( p = 0.005 for 893Ser) and D. dasycarpus ( p = 0.0009 for 893Ala; p = 0.002 for 893Ser) in X. laevis oocytes. The results in this study suggest that herbal medicines could interact with other drugs and change the therapeutic effects depending on the genetic polymorphisms of individuals. For the study, researchers used olive flounder from two sources from three commercial flounder farms in Korea as parental stock. They found that the parentage for 95.8 percent of the offspring was successfully designated to a single parental couple utilizing eight microsatellite DNA markers.

Purpose: PRESAGE® is a dosimeter made of polyurethane, which is suitable for 3D dosimetry in modern radiation treatment techniques. Since an ideal dosimeter is radiologically water equivalent, the authors investigated water equivalency and the radiological properties of three different PRESAGE® formulations that differ primarily in their elemental compositions. Two of the formulations are new and have lower halogen content than the original formulation. Methods: The radiological water equivalence was assessed by comparing the densities, interaction probabilities, and radiation dosimetry properties of the three different PRESAGE® formulations to the corresponding values for water. The relative depth doses were calculated using Monte Carlo methods for 50, 100, 200, and 350 kVp and 6 MV x-ray beams. Results: The mass densities of the three PRESAGE® formulations varied from 5.3% higher than that of water to as much as 10% higher than that of water for the original formulation. The probability of photoelectric absorption in the three different PRESAGE® formulations varied from 2.2 times greater than that of water for the new formulations to 3.5 times greater than that of water for the original formulation. The mass attenuation coefficient for the three formulations is 12%-50% higher than the value for water. These differences occur over an energy range (10-100 keV) in which the photoelectric effect is the dominant interaction. The collision mass stopping powers of the relatively lower halogen-containing PRESAGE® formulations also exhibit marginally better water equivalency than the original higher halogen-containing PRESAGE® formulation. Furthermore, the depth dose curves for the lower halogen-containing PRESAGE® formulations are slightly closer to that of water for a 6 MV beam. In the kilovoltage energy range, the depth dose curves for the lower halogen-containing PRESAGE® formulations are in better agreement with water than the original PRESAGE® formulation. Conclusions: Based on the results of this study, the new PRESAGE® formulations with lower halogen content are more radiologically water equivalent overall than the original formulation. This indicates that the new PRESAGE® formulations are better suited to clinical applications and are more accurate dosimeters and phantoms than the original PRESAGE® formulation. While correction factors are still needed to convert the dose measured by the dosimeter to an absorbed dose in water in the kilovoltage energy range, these correction factors are considerably smaller for the new PRESAGE® formulations compared to the original PRESAGE® and the existing polymer gel dosimeters. Abstract: When deprived of an anchorage to the extracellular matrix, fibroblasts arrest in the G1 phase with inactivation of Cdk4/6 and Cdk2 and destruction of Cdc6, the assembler of prereplicative complexes essential for S phase onset. How cellular anchorages control these kinases and Cdc6 stability is poorly understood. Here, we report that in rat embryonic fibroblasts, activation of mammalian target of rapamycin complex 1 by a Tsc2 mutation or overexpression of a constitutively active mutant Rheb overrides the absence of the anchorage and stabilizes Cdc6 at least partly via activating Cdk4/6 that induces Emi1, an APC/CCdh1 ubiquitin ligase inhibitor. The article highlights a study on the role played by the orphan nuclear receptor small heterodimer partner (SHP;NR0B2) in osteoblastic bone formation. SHP usually regulates a range of cellular events like cell proliferation, differentiation and metabolism and during osteoblast differentiation, it is regulated by bone morphogenetic protein 2, which is important in bone formation. The study found that SHP plays a positive regulator in bone formation as it supplement osteoblast differentiation by regulating Runx2 activity.

Abstract: NFATc1 is a master regulator of RANKL-induced osteoclast differentiation and herein we investigate the regulatory mechanism of NFATc1 in osteoclast activation. Inactivation of NFATc1 strongly attenuates RANKL-induced bone resorption and overexpression of a constitutively active form of NFATc1 in osteoclasts induces formation of actin rings and resorption pits on dentin slices. We demonstrate that NFATc1 binds directly to the promoter regions of its target genes and induces expression of various genes, including LTBP3, ClC7, cathepsin K, MMP9, and c-Src, which are key players in bone resorption. Thus, NFATc1 is essential for RANKL-induced osteoclast activation via up-regulation of osteoclast-activating genes. [Copyright &y& Elsevier]

We characterized 52 new microsatellite markers isolated from (GT) n and (CT) n microsatellite-enriched genomic libraries of the olive flounder ( Paralichthys olivaceus). All markers were polymorphic, with eight to 30 (mean 15.1) alleles detected in 30 individuals from a single natural population. Observed heterozygosity ranged from 0.20 to 1.0. Segregation analysis within a mapping family revealed non-amplifying null alleles at six loci. This study assessed the association of hepatitis B virus (HBV) and hepatitis C virus (HCV) infection with non-Hodgkin's lymphoma in a highly HBV-endemic area. The prevalence of either HBV or HCV infection in 235 patients with non-Hodgkin's lymphoma was compared with that of an age- and sex-matched hospital control group of 235 patients. The prevalence of HBV infection was higher in B-cell non-Hodgkin's lymphoma (15.5%) than control (8.1%), but the prevalence of HCV infection in the non-Hodgkin's lymphoma patients (2.1%) and control group (3%) was similar. HBV prevalence increased significantly with age in the B-cell non-Hodgkin's lymphoma patients. The presence of HBV proteins and DNA in lymphoma tissues and peripheral blood mononuclear cells (PBMCs) from HBV-infected non-Hodgkin's lymphoma patients was also investigated using immunohistochemistry and PCR. HBV DNA was frequently detected in PBMCs from HBV-infected non-Hodgkin's lymphoma patients, but HBV antigens were not. Therefore, HBV infection, but not HCV infection, was associated with B-cell non-Hodgkin's lymphoma in Korea, suggesting a possible role for HBV in the development of non-Hodgkin's lymphoma. J. Med. Purpose: To correlate the incidence and rate of placental migration with the mode of delivery, pregnancy outcome, and maternal complication according to the location (anterior versus posterior) and type of placenta previa (PP). Methods: We prospectively evaluated the placentas of 98 pregnant women with PP or low-lying placenta (LLP) at the prenatal sonographic examination performed between 20 and 27 gestational weeks. We divided the pregnant women into groups according to type and location of PP. Follow-up sonographic examination was performed between 32 and 37 weeks. We compared incidence of migration to the normal position and calculated the migration rate as the migrated distance divided by the weeks of interval between 2 sonographic examinations. We compared the incidences of cesarean section, fetal outcome, and maternal complications during the pregnancy. Results: The incidence of migration in the group of anterior placentas was significantly higher than that in the group of posterior placentas. The mean migration rate in the anterior group was 2.6 mm/week, whereas that in the posterior group was 1.6 mm/week. The migration rate of incomplete PP was significantly higher than that of LLP. Incidence of cesarean section for nonmigrated PP was significantly higher in the posterior group. The incidences of premature delivery and vaginal spotting were also significantly higher in the posterior group. Conclusion: Anterior PP and LLP may migrate more often and faster than posterior PP. Abstract: We recently reported that a histidine (H191) in the S3–S4 loop of domain I is critical for nickel inhibition of the Cav3.2 T-type Ca2+ channel. As in Cav3.2, two histidine residues are commonly found in the IS3–IS4 loops of mammalian Cav2.3 Ca2+ channels, which are also blocked by low micromolar concentrations of nickel. We show here by site-directed mutagenesis and electrophysiology that both residues contribute to the nickel sensitivity of Cav2.3, with H183 being more critical than H179. These findings strongly suggest that both H179 and H183 in the IS3–IS4 loop are essential structural determinants required for nickel sensitive inhibition of the Cav2.3. [Copyright &y& Elsevier]

T-type Ca2+ channels play essential roles in numerous cellular processes. Recently, we reported that phorbol-12-myristate-13-acetate (PMA) potently enhanced the current amplitude of Cav3.2 T-type channels reconstituted in Xenopus oocytes. Here, we have compared PMA modulation of the activities of Cav3.1, Cav3.2 and Cav3.3 channels, and have investigated the underlying mechanism. PMA augmented the current amplitudes of the three T-type channel isoforms, but the fold stimulations and time courses differed. The augmentation effects were not mimicked by 4α-PMA, an inactive stereoisomer of PMA, but were abolished by preincubation with protein kinase C (PKC) inhibitors, indicating that PMA augmented T-type channel currents via activation of oocyte PKC. The stimulation effect on Cav3.1 channel activity by PKC was mimicked by endothelin when endothelin receptor type A was coexpressed with Cav3.1 in the Xenopus oocyte system. Pharmacological studies combined with fluorescence imaging revealed that the surface density of Cav3.1 T-type channels was not significantly changed by activation of PKC. Abstract: High voltage-activated Ca2+ channel expression and gating is controlled by their β subunits. Although the sites of interaction are known at the atomic level, how β modulates gating remains to be determined. Using a chimeric approach, β subunit regulation was conferred to a low voltage-activated channel. Regulation was dependent on a rigid linker connecting the α1 interaction domain to IS6. Chimeric channels also revealed a role for IS6 in channel gating. Taken together, these results support a direct coupling model where β subunits alter movements in IS6 that occur as the channel transits between closed, open, and inactivated states. Neural cell adhesion molecule (NCAM) influences proliferation and differentiation of neuronal cells. However, only a little is known about the downstream effects of NCAM signalling, such as alterations in gene transcription, which are associated with cell fate choice. To examine whether NCAM plays a role in cell fate choice during hippocampal neurogenesis, we performed a gain-of-function study, using a retroviral vector which contained full-length NCAM140 cDNA and the marker gene EGFP , and found that NCAM140 promoted neurogenesis by activating proneural transcription activators with concurrent inhibition of gliogenesis. The enhanced transcript levels of proneural transcription factors in NCAM140-transduced cells were down-regulated by treatment of the cells with mitogen-activated protein kinase kinase (MEK) inhibitor PD098059. Overall, these findings suggest that NCAM140 may facilitate hippocampal neurogenesis via regulation of proneurogenic transcription factors in an extracellular signal-regulated kinase (ERK)-dependent manner. Although T-type Ca2+ channels have been implicated in numerous physiological functions, their regulations by protein kinases have been obscured by conflicting reports. We investigated the effects of protein kinase C (PKC) on Cav3.2 T-type channels reconstituted in Xenopus oocytes. Phorbol-12-myristate-13-acetate (PMA) strongly enhanced the amplitude of Cav3.2 channel currents (∼3-fold). The augmentation effects were not mimicked by 4α-PMA, an inactive stereoisomer of PMA, and abolished by preincubation with PKC inhibitors. Molecular diversity in T-type Ca2+ channels is produced by expression of three genes, and alternative splicing of those genes. Prompted by differences noted between rat and human Cav3.3 sequences, we searched for splice variants. We cloned six variants, which are produced by splicing at exon 33 and exon 34. Expression of the variants differed between brain regions. The electrophysiological properties of the variants displayed similar voltage-dependent gating, but differed in their kinetic properties. The functional impact of splicing was inter-related, suggesting an interaction. We conclude that alternative splicing of the Cav3.3 gene produces channels with distinct properties. Abstract Expression of rat α1G, human α1H and rat α1I subunits of voltage-activated Ca2 + channels in HEK-293 cells yields robust Ca2 + inward currents with 1.25 mm Ca2 + as the charge carrier. Both similarities and marked differences are found between their biophysical properties. Currents induced by expression of α1G show the fastest activation and inactivation kinetics. The α1H and α1I currents activate and inactivate up to 1.5- and 5-fold slower, respectively. No differences in the voltage dependence of steady state inactivation are detected. Currents induced by expression of α1G and α1H deactivate with time constants of up to 6 ms at a test potential of - 80 mV, but currents induced by α1I deactivate about three-fold faster. Recovery from short-term inactivation is more than three-fold slower for currents induced by α1H and α1I in comparison to α1G. In contrast to these characteristics, reactivation after long-term inactivation was fastest for currents arising from expression of α1I and slowest in cells expressing α1H calcium channels. The calcium inward current induced by expression of α1I is increased by positive prepulses while currents induced by α1H and α1G show little ( < 5%) or no facilitation. The data thus provide a characteristic fingerprint of each channel’s activity, which may allow correlation of the α1G, α1H and α1I induced currents with their in vivo counterparts. Abstract Voltage-dependent activity around the resting potential is determinant in neuronal physiology and participates in the definition of the firing pattern. Low-voltage-activated T-type Ca2 + channels directly affect the membrane potential and control a number of secondary Ca2 + -dependent permeabilities. We have studied the ability of the cloned T-type channels (α1G,H,I) to carry Ca2 + currents in response to mock action potentials. The relationship between the spike duration and the current amplitude is specific for each of the T-type channels, reflecting their individual kinetic properties. Typically the charge transfer increases with spike broadening, but the total Ca2 + entry saturates at different spike durations according to the channel type: 4 ms for α1G; 7 ms for α1H; and > 10 ms for α1I channels. During bursts, currents are inhibited and/or transiently potentiated according to the α1 channel type, with larger effects at higher frequency. The inhibition may be induced by voltage-independent transitions toward inactivated states and/or channel inactivation through intermediate closed states. The potentiation is explained by an acceleration in the channel activation kinetics. Relatively fast inactivation and slow recovery limit the ability of α1G and α1H channels to respond to high frequency stimulation ( > 20 Hz). In contrast, the slow inactivation of α1I subunits allows these channels to continue participating in high frequency bursts (100 Hz). Robot kinematics is an extensively researched subject and several well developed techniques for the analysis of robot manipulators and spatial mechanisms exist. Most of these methods are based on point transformations. However, far less research is in evidence for line transformations. Lines and screws are more fundamental to velocity analysis and hence line transformations are believed to be better suited for the kinematic and static analysis of manipulators. In this article, the kinematics for serial chain manipulators using line transformations is investigated. Dual number quaternions are used as spatial transformation operators since they allow compact representation of both rotations and translations. The article presents a study which shows the association of trans fatty acids (TFAs) and attention-deficit/hyperactivity disorder (ADHD) in female adolescents. It states that to assess ADHD symptoms, Conners-Wells Adolescent Self-Report Scale was used. Results show that high TFAs intake can act as a possible pathophysiology in ADHD development.
