Thyroid hormone receptor interactor 13 (TRIP13) is an ATPase that has been found to be overexpressed in many tumors. The aim of this study was to investigate the role of TRIP13 and its mechanism of action in lung cancer. The expression of TRIP13 was examined in lung cancer tissues and corresponding normal lung tissues by western blotting. TRIP13 was overexpressed or knocked down by transient transfection or siRNA interference in lung cancer cells, respectively. The expression of key proteins associated with the Wnt signaling pathway and epithelial-mesenchymal transition (EMT) was assessed. The interaction between TRIP13 and low-density lipoprotein receptor-related protein 6 (LRP6) was examined by co-immunoprecipitation and laser confocal immunofluorescence. Moreover, this study determined the proliferative and invasive ability of cells through colony formation, cell proliferation, and Matrigel invasion assays. The expression of TRIP13 was higher in lung cancer tissues than in normal lung tissues (p = 0.002), and this correlated with poor patient prognosis (p < 0.001). In addition, overexpression of TRIP13 enhanced the levels of active β-catenin and target proteins of the Wnt signaling pathways (p < 0.05). This study found that TRIP13 can co-localize and bind with LRP6. Furthermore, overexpression of TRIP13 caused the upregulation of N-cadherin, Snail, and vimentin, and the downregulation of E-cadherin (p < 0.05). The aforementioned results were reversed after knocking down the expression of TRIP13 (p < 0.05). In clinical research on complementary and integrative medicine, experts and scientists have often pursued a research agenda in spite of an incomplete understanding of the needs of end users. Consequently, the majority of previous clinical trials have mainly assessed the efficacy of interventions. Scant data is available on their effectiveness. Comparative effectiveness research (CER) promises to support decision makers by generating evidence that compares the benefits and harms of the best care options. This evidence, more generalizable than the evidence generated by traditional randomized controlled trials (RCTs), is better suited to inform real-world care decisions. An emphasis on CER supports the development of the evidence base for clinical and policy decision-making. Whereas in most areas of complementary and integrative medicine data on comparative effectiveness is scarce, available acupuncture research already contributes to CER evidence. Sucrose synthase (SS), sucrose phosphate synthase (SPS), soluble acid invertase (SAI) and neutral invertase (NI) are the key enzymes of sucrose metabolism, which activities play key role in sugar accumulation. In the present study, sugarcane variety GT28 was used as the plant material, effects of ethephon, an ethylene releasing agent, on the key enzymes of sucrose metabolism and ethylene production in relation to sucrose accumulation in sugarcane stalk and leaves were analyzed. The plants were treated by foliar spray of 400 mg/L ethephon at late growth stage. The results showed that ethylene production in leaves was positively while that in stalk internodes was negatively correlated with the maturity of leaves and stalks. The activities of SS, SPS and NI in stalk and leaves were positively while that of NI was negatively correlated with the maturity of stalks and leaves. High concentration of ethephon significantly inhibited the ethylene production and the activities of SS, SPS and NI but increased the activity of SAI enzyme in immature leaves, while promoted the ethylene production and the activities of SS, SPS and NI in maturing and matured leaves and the activity of SAI in maturing leaves but inhibited the activity of SAI in matured leaves. The active duration of ethylene releasing and the effects of ethephon on the key enzymes in leaves lasted for about 5-10 days. The ethylene production was much lower in internodes than in leaves. High concentrations of ethephon significantly increased the ethylene production in all the three different internodes in a short time but longer for immature internode, and it significantly improved the activities of SS, SPS in all the internodes and that of NI in immature and maturing internodes, but inhibited the activity of SAI in the three internodes and that of NI in matured internode. It is suggested that high concentration of ethephon may accelerate the leaf maturity and aging of the matured leaves by increasing ethylene production which could promote the activities of SS, SPS and NI in maturing and matured leaves, and the activity of SAI in immature and maturing leaves, speeding up the sucrose synthesis in maturing and matured leaf and the sucrose transportation from leaves to immature and maturing internodes, resulting in faster sugar accumulation in immature and maturing internodes of the stalks and higher sucrose content in whole cane. Background: Acupuncture is commonly used to treat migraine. We assessed the efficacy of acupuncture at migraine-specific acupuncture points compared with other acupuncture points and sham acupuncture. Methods: We performed a multicentre, singleblind randomized controlled trial. In total, 480 patients with migraine were randomly assigned to one of four groups (Shaoyang-specific acupuncture, Shaoyang-nonspecific acupuncture, Yangming-specific acupuncture or sham acupuncture [control]). All groups received 20 treatments, which included electrical stimulation, over a period of four weeks. The primary outcome was the number of days with a migraine experienced during weeks 5–8 after randomization. Our secondary outcomes included the frequency of migraine attack, migraine intensity and migrainespecific quality of life. Results: Compared with patients in the control group, patients in the acupuncture groups reported fewer days with a migraine during weeks 5–8, however the differences between treatments were not significant (p > 0.05). There was a significant reduction in the number of days with a migraine during weeks 13–16 in all acupuncture groups compared with control (Shaoyang-specific acupuncture v. control: difference −1.06 [95% confidence interval (CI) −1.77 to −0.5], p = 0.003; Shaoyang-nonspecific acupuncture v. control: difference −1.22 [95% CI −1.92 to −0.52], p < 0.001; Yangming-specific acupuncture v. control: difference −0.91 [95% CI −1.61 to −0.21], p = 0.011). We found that there was a significant, but not clinically relevant, benefit for almost all secondary outcomes in the three acupuncture groups compared with the control group. We found no relevant differences between the three acupuncture groups. Interpretation: Acupuncture tested appeared to have a clinically minor effect on migraine prophylaxis compared with sham acupuncture. We evaluated the sensitivity and dose dependency of radiation-induced injury in hematopoietic stem/progenitor cells. Adult C57BL/6 mice were daily exposed to 0, 2, 10, 50, and 250 mGy γ-ray for 1 month in succession, respectively. The damage of hematopoietic stem/progenitor cells in bone marrow were investigated within 2 hours (acute phase) or at 3 months (chronic phase) after the last exposure. Daily exposure to over 10 mGy γ-ray significantly decreased the number and colony-forming capacity of hematopoietic stem/progenitor cells at acute phase, and did not completely recover at chronic phase with 250 mGy exposure. Interestingly, the daily exposure to 10 or 50 mGy γ-ray decreased the formation of mixed types of colonies at chronic phase, but the total number of colonies was comparable to control. Immunostaining analysis showed that the formation of 53BP1 foci in c-kit+ stem/progenitor cells was significantly increased with daily exposure to 50 and 250 mGy at acute phase, and 250 mGy at chronic phase. Many genes involved in toxicity responses were up- or down-regulated with the exposures to all doses. Our data have clearly shown the sensitivity and dose dependency of radiation-induced injury in hematopoietic stem/progenitor cells of mice with daily exposures to 2 ~ 250 mGy γ-ray. While exploring the process of CO/CO2 electroreduction (COxRR) is of great significance to achieve carbon recycling, deciphering reaction mechanisms so as to further design catalytic systems able to overcome sluggish kinetics remains challenging. In this work, a model single-Co-atom catalyst with well-defined coordination structure is developed and employed as a platform to unravel the underlying reaction mechanism of COxRR. The as-prepared single-Co-atom catalyst exhibits a maximum methanol Faradaic efficiency as high as 65% at 30 mA/cm2 in a membrane electrode assembly electrolyzer, while on the contrary, the reduction pathway of CO2 to methanol is strongly decreased in CO2RR. In-situ X-ray absorption and Fourier-transform infrared spectroscopies point to a different adsorption configuration of *CO intermediate in CORR as compared to that in CO2RR, with a weaker stretching vibration of the C–O bond in the former case. Theoretical calculations further evidence the low energy barrier for the formation of a H-CoPc-CO– species, which is a critical factor in promoting the electrochemical reduction of CO to methanol. Deciphering the reaction mechanisms of CO/CO2 electroreduction to methanol remains challenging. Here the authors report the higher electron density of single-Co-atom center, along with a different adsorption configuration of *CO, is crucial for promoting the CO electroreduction to methanol. More accurate source analysis of potentially toxic elements (PTEs) in atmospheric fallout that endanger biodiversity and human health remains needed. This study determined the concentrations of seven PTEs, including Pb, Cd, As, Cu, Zn, Ni, and Cr, by inductively coupled plasma mass spectrometry (ICP-MS), and the sources of PTE pollution were quantified using multivariate statistical analysis, including principal component analysis (PCA), cluster analysis (CA), and Pearson correlation analysis, and Moran index was applied for mutual verification and supplementation. PCA and CA revealed that the same mixed sources of Pb, Cd, As, Cu, and Zn were found in the atmospheric dust fall in the study area, while Ni and Cr had the same source of pollution. Pearson correlation analysis provided that there were strong correlations between Pb–Cd, Pb-As, Pb-Cu, Cd-As, Cd-Cu, As-Cu, and Ni–Cr, indicating commonality between the two sources of heavy metal pollution. Additionally, the Moran index showed that strong spatial correlations were observed between Pb, Cd, As, Cu, and Zn, whose sources were mainly related to non-ferrous metal processing smelter smelting slag sites and an environmental company in the study area. Objective: To examine the influence of widely used protein affinity tags and the tobacco PR1a signal peptide (SP) on detection, purification and bioactivity analyses of the small oomycete apoplastic effector SCR96 in planta. Results: Through agroinfiltration, the phytotoxic effector SCR96 of Phytophthora cactorum was expressed in Nicotiana benthamiana leaf apoplast as a fusion protein carrying single affinity tag (His, HA or FLAG) at either C- or N-terminus. Leaf necrosis caused by different affinity-tagged SCR96 varied among tags and replicates. All of tagged proteins can be detected by antibodies against SCR96. All of SCR96 fusions except N-terminally fused 6His-tagged protein were detected using tag antibodies, indicating that 6His tag may be degraded when fused at N-terminus. Interestingly, C-terminal His- and FLAG-tagged SCR96 maintained the biological activity after purification. In the substitution assay of SCR96 SP, we observed that PR1a SP can lead chimeric SCR96 expression in N. benthamiana, but the replacement totally disrupted its bioactivity. Conclusion: C-terminal His or FLAG tag, along with its original SP, is efficient enough to enable detection and purification of functional SCR96 from N. benthamiana leaf apoplast, which would facilitate plant–pathogen interaction studies.
