AHP-3a, a triple-helix acidic polysaccharide isolated from Alpinia officinarum Hance, was evaluated for its anticancer and antioxidant activities. The physicochemical properties and structure of AHP-3a were investigated through gel permeation chromatography, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. The weight-average molecular weight of AHP-3a was 484 kDa, with the molar percentages of GalA, Gal, Ara, Xyl, Rha, Glc, GlcA, and Fuc being 35.4%, 21.4%, 16.9%, 11.8%, 8.9%, 3.1%, 2.0%, and 0.5%, respectively. Based on the results of the monosaccharide composition analysis, methylation analysis, and NMR spectroscopy, the main chain of AHP-3a was presumed to consist of (1→4)-α-D-GalpA and (1→2)-α-L-Rhap residues, which is a pectic polysaccharide with homogalacturonan (HG) and rhamnogalacturonan-I (RG-I) structural domains containing side chains. In addition, the results of the antioxidant activity assay revealed that the ability of AHP-3a to scavenge DPPH, ABTS, and OH free radicals increased with an increase in its concentration. Moreover, according to the results from the EdU, wound healing, and Transwell assays, AHP-3a can control the proliferation, migration, and invasion of HepG2 and Huh7 hepatocellular carcinoma cells without causing any damage to healthy cells. Sea anemone venom, abundant in protein and peptide toxins, serves primarily for predatory defense and competition. This study delves into the insulin-like peptides (ILPs) present in sea anemones, particularly focusing on their role in potentially inducing hypoglycemic shock in prey. We identified five distinct ILPs in Exaiptasia diaphana, exhibiting varied sequences. Among these, ILP-Ap04 was successfully synthesized using solid phase peptide synthesis (SPPS) to evaluate its hypoglycemic activity. When tested in zebrafish, ILP-Ap04 significantly reduced blood glucose levels in a model of diabetes induced by streptozotocin (STZ) and glucose, concurrently affecting the normal locomotor behavior of zebrafish larvae. Furthermore, molecular docking studies revealed ILP-Ap04's unique interaction with the human insulin receptor, characterized by a detailed hydrogen-bonding network, which supports a unique mechanism for its hypoglycemic effects. Our findings suggest that sea anemones have evolved sophisticated strategies to activate insulin receptors in vertebrates, providing innovative insights into the design of novel drugs for the treatment of diabetes. In recent years, remote sensing analysis has gained significant attention in visual analysis applications, particularly in segmenting and recognizing remote sensing images. However, the existing research has predominantly focused on single-period RGB image analysis, thus overlooking the complexities of remote sensing image capture, especially in highly vegetated land parcels. In this paper, we provide a large-scale vegetation remote sensing (VRS) dataset and introduce the VRS-Seg task for multi-modal and multi-temporal vegetation segmentation. The VRS dataset incorporates diverse modalities and temporal variations, and its annotations are organized using the Vegetation Knowledge Graph (VKG), thereby providing detailed object attribute information. To address the VRS-Seg task, we introduce VRSFormer, a critical pipeline that integrates multi-temporal and multi-modal data fusion, geometric contour refinement, and category-level classification inference. The experimental results demonstrate the effectiveness and generalization capability of our approach. Lodging is one of the most important factors affecting the high and stable yield of wheat worldwide. Solid-stemmed wheat has higher stem strength and lodging resistance than hollow-stemmed wheat does. There are many solid-stemmed varieties, landraces, and old varieties of durum wheat. However, the transfer of solid stem genes from durum wheat is suppressed by a suppressor gene located on chromosome 3D in common wheat, and only hollow-stemmed lines have been created. However, synthetic hexaploid wheat can serve as a bridge for transferring solid stem genes from tetraploid wheat to common wheat. In this study, the F1, F2, and F2:3 generations of a cross between solid-stemmed Syn-SAU-119 and semisolid-stemmed Syn-SAU-117 were developed. A single dominant gene, which was tentatively designated Su-TdDof and suppresses stem solidity, was identified in synthetic hexaploid wheat Syn-SAU-117 by using genetic analysis. By using bulked segregant RNA-seq (BSR-seq) analysis, Su-TdDof was mapped to chromosome 7DS and flanked by markers KASP-669 and KASP-1055 within a 4.53 cM genetic interval corresponding to 3.86 Mb and 2.29 Mb physical regions in the Chinese Spring (IWGSC RefSeq v1.1) and Ae. tauschii (AL8/78 v4.0) genomes, respectively, in which three genes related to solid stem development were annotated. Su-TdDof differed from a previously reported solid stem suppressor gene based on its origin and position. Su-TdDof would provide a valuable example for research on the suppression phenomenon. The flanking markers developed in this study might be useful for screening Ae. tauschii accessions with no suppressor gene (Su-TdDof) to develop more synthetic hexaploid wheat lines for the breeding of lodging resistance in wheat and further cloning the suppressor gene Su-TdDof. Although cities only account for 3% of the global land area, they have disproportionately contributed 70% of total anthropogenic CO2 emissions; the main issue in estimating urban anthropogenic CO2 emissions is their large uncertainty. Tower-based atmospheric CO2 observations and simulations in urban areas have been frequently used as an independent approach to constrain and evaluate greenhouse gas emissions from city to regional scales, where only daytime CO2 observations and simulations are used considering the consensus that the large bias in simulating nighttime planetary boundary layer heights (PBLH) and atmospheric CO2 concentration will cause overestimation/underestimation in CO2 emission inversions. The above strategy of only using daytime observations makes the numbers of available concentration observations largely decrease even with the fact that tower-based atmospheric CO2 observations are sparsely distributed and conducted. Here, to solve the issue of large bias in nighttime CO2 simulations, we conducted four months of atmospheric CO2 observations from January to April in 2019, and raised an approach by coupling emission heights with dynamic PBLH variations in a WRF-STILT model. We found (1) the overestimation of simulated nighttime CO2 concentration decreased by 5–10 ppm, especially between 0:00 and 7:00. (2) The statistics for nighttime simulations were largely improved by using a revised model and posteriori emissions. The regression slopes of daily averages were 0.93 and 0.81 for the default model using a priori emissions and the revised model using the same a priori emissions, and the slope largely improved to 0.97 for the revised model using posteriori emissions. Moreover, the correlation coefficient also increased from 0.29 and 0.37 to 0.53; these results indicate our revised model obviously calibrated the bias in both nighttime and daily CO2 concentration simulations. In general, it is strongly recommended to use the revised WRF-STILT model in future inversion studies, which can effectively reduce the overestimation of nighttime spikes and make full use of nighttime observations. A method based on capacitively coupled contactless conductivity detection (C4D), which has been proven effective for the rapid detection of available soil potassium content, was firstly proposed to apply to soil nutrient detection. By combining a detection signal spectrum analysis, geographic information system (GIS) data, and a cluster analysis, a soil nutrient management system to match the detection device was developed. This system included six modules: soil sample information management, electrophoresis analysis, quantitative calculation, nutrient result viewing, cluster analysis, and nutrient distribution map generation. The soil samples, which were collected from an experimental field in Xuchang City of Henan Province, were analyzed using the C4D and flame photometer methods. The results showed that the detection results for the soil samples obtained via the two methods were in good agreement. C4D technology was feasible for the detection of the soil available nutrients and had the advantages of a high timeliness, low sample volume, and low pollution. The soil nutrient management system adopted the hierarchical clustering method to classify the grid cells of the experimental field according to the nutrient detection results. A soil nutrient distribution map displayed the spatial difference in nutrients. This paper provides a systematic solution for soil nutrient zone management that includes nutrient detection, signal analysis, data management for the nutrient zone, and field nutrient distribution map generation to support decision making in variable fertilization. Autoimmune thyroid diseases (AITDs), which include Hashimoto's thyroiditis (HT) and Graves' disease (GD), have a higher prevalence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in the literature. The effects of AITD-associated cytokines on SARS-CoV-2 infection-mediating molecule levels might be involved in the pathogenesis of susceptibility. We speculated that hydrogen sulfide (H2S) might attenuate this process since H2S has antiviral effects. Using immunohistochemistry, we found that angiotensin-converting enzyme-II (ACE2) expression was higher in the HT group and neuropilin 1 (NRP1) expression was higher in HT and GD groups than in the normal group, while transmembrane protease serine type 2 (TMPRSS2) expression was lower in HT and GD groups. When culturing primary thyrocytes with cytokines or sodium hydrosulfide (NaHS) plus cytokines, we found that ACE2 and NRP1 mRNA levels were upregulated while TMPRSS2 levels were downregulated by interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α). After pretreatment with NaHS in thyrocytes, ACE2 and NRP1 expression were downregulated compared to IFN-γ or TNF-α treatment, and NaHS had no effect on TMPRSS2 expression. Our findings suggested that IFN-γ and TNF-α, which are elevated in AITDs, promoted ACE2 and NRP1 expression and inhibited TMPRSS2 expression. This study aimed to apply transcriptomics to determine how Molor-Dabos-4 (MD-4) protects healthy rats against indomethacin (IND)-induced gastric ulcers and to identify the mechanism behind this protective effect. Rats were pretreated with MD-4 (0.3, 1.5, or 3 g/kg per day) for 21 days before inducing gastric ulcers by oral administration with indomethacin (30 mg/kg). Unulcerated and untreated healthy rats were used as controls. Effects of the treatment were assessed based on the ulcer index, histological and pathological examinations, and indicators of inflammation, which were determined by enzyme-linked immunosorbent assay. Transcriptomic analysis was performed for identifying potential pharmacological mechanisms. Eventually, after identifying potential target genes, the latter were validated by quantitative reverse-transcription polymerase chain reaction (qRT-PCR). After pretreatment with MD-4, gastric ulcers, along with other histopathological features, were reduced. MD-4 significantly (p < 0.05) increased the superoxide dismutase (SOD) levels in ulcers and reduced pepsin, TNF-α, and IL-6 levels. RNA-seq analysis identified a number of target genes on which MD-4 could potentially act. Many of these genes were involved in pathways that were linked to anti-inflammatory and antioxidant responses, and other protective mechanisms for the gastric mucosa. qRT-PCR showed that altered expression of the selected genes, such as Srm, Ryr-1, Eno3, Prkag3, and Eef1a2, was consistent with the transcriptome results. MD-4 exerts protective effects against IND-induced gastric ulcers by reducing inflammatory cytokines and pepsin and increasing the expression of SOD levels. Downregulation of Srm, Ryr-1, Eno3, Prkag3, and Eef1a2 genes involved in regulating arginine and proline metabolism, calcium signaling pathway, HIF-1 signaling pathway, oxytocin signaling pathway, and legionellosis are possibly involved in MD-4-mediated protection against gastric ulcers. Ixeris chinensis (Thunb.) Nakai (IC) is a folk medicinal herb used in Mongolian medical clinics for the treatment of hepatitis and fatty liver diseases even though its pharmacological mechanism has not been well characterized. This study investigated the hepatoprotective mechanism of IC on mice with nonalcoholic fatty liver disease (NAFLD) by integrating gut microbiota and metabolomic analysis. A high-fat diet (HFD) was used to develop nonalcoholic fatty liver disease, after which the mice were treated with oral IC (0.5, 1.5 and 3.0 g/kg) for 10 weeks. HFD induced NAFLD and the therapeutic effects were characterized by pathological and histological evaluations, and the serum indicators were analyzed by ELISA. The gut microbial and metabolite profiles were studied by 16S rRNA sequencing and untargeted metabolomic analysis, respectively. The results showed that the administration of IC resulted in significant decreases in body weight; liver index; serum biomarkers such as ALT, TG, and LDL-C; and the liver inflammatory factors IL-1β, IL-6, and TNF-α. The 16S rRNA sequencing results showed that administration of IC extract altered both the composition and abundance of the gut microbiota. Untargeted metabolomic analysis of liver samples detected a total of 212 metabolites, of which 128 were differentially expressed between the HFD and IC group. IC was found to significantly alter the levels of metabolites such as L-glutamic acid, pyridoxal, ornithine, L-aspartic acid, D-proline, and N4-acetylaminobutanal, which are involved in the regulation of glutamine and glutamate, Vitamin B6 metabolism, and arginine and proline metabolic pathways. Correlation analysis indicated that the effects of the IC extract on metabolites were associated with alterations in the abundance of Akkermansiaceae, Lachnospiraceae, and Muribaculaceae. Our study revealed that IC has a potential hepatoprotective effect in NAFLD and that its function might be linked to improvements in the composition of gut microbiota and their metabolites. As LoRaWAN is one of the most popular long-range wireless protocols among low-power IoT applications, more and more focus is shifting towards security. In particular, physical layer topics become relevant to improve the security of LoRaWAN nodes, which are often limited in terms of computational power and communication resources. To this end, e.g., detection methods for wireless attacks improve the integrity and robustness of LoRaWAN access. Further, wireless physical layer techniques have potential to enhance key refreshment and device authentication. In this work, we aim to provide a comprehensive review of various vulnerabilities, countermeasures and security enhancing features concerning the LoRaWAN physical layer. The acquisition of nutrient data on a precise scale has played a vital role in nutrient management processes for soils. However, the lack of rapid precise and multi-index detection techniques for soil macronutrient contents hinders both rational fertilization and cost reduction. In this paper, a rapid detection method and device were devised, combining capillary electrophoresis (CE) and capacitively coupled contactless conductivity detection (C4D), and presented to detect macronutrient contents of soil. The device consisted of a capillary channel, C4D detector, high-voltage system, etc. It separated macronutrient ions using capillary electrophoresis and then measured the ion concentration based on the C4D principle. Lime concretion black soil samples from a complete field were collected and detected. NO3−, NH4+, H2PO4− and K+ in sample solutions could be detected in 5 min with relative standard deviations (RSDs) from 1.0 to 7.51%. The injection voltage was set to 10 kV for 5 s, and the separation voltage was set to 14 kV. This demonstrated the excellent performance of the C4D device on the detection of soil macronutrients, which could help to guide fertilization operations more effectively. [ABSTRACT FROM AUTHOR]

Papaver somniferum L. is an important medical plant that produces analgesic drugs used for the pain caused by cancers and surgeries. Recent studies have focused on the expression genes involved in analgesic drugs biosynthesis, and the real-time quantitative polymerase chain reaction (RT-qPCR) technique is the main strategy. However, no reference genes have been reported for gene expression normalization in P. somniferum. Herein, nine reference genes (actin (ACT), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), cyclophilin 2 (CYP2), elongation factor 1-alpha (EF-1α), glyceraldehyde-3-phosphate dehydrogenase 2, cytosolic (GAPC2), nuclear cap-binding protein subunit 2 (NCBP2), protein phosphatase 2A (PP2A), TIP41-like protein (TIP41), and tubulin beta chain (TUB)) of P. somniferum were selected and analyzed under five different treatments (cold, drought, salt, heavy metal, and hormone stress). Then, BestKeeper, NormFinder, geNorm, and RefFinder were employed to analyze their gene expression stability. The results reveal that NCBP2 is the most stable reference gene under various experimental conditions. The work described here is the first report regarding on reference gene selection in P. somniferum, which could be used for the accurate normalization of the gene expression involved in analgesic drug biosynthesis. The fifth generation (5G) and beyond wireless communications will transform many exciting applications and trigger massive data connections with private, confidential, and sensitive information. The security of wireless communications is conventionally established by cryptographic schemes and protocols in which the secret key distribution is one of the essential primitives. However, traditional cryptography-based key distribution protocols might be challenged in the 5G and beyond communications because of special features such as device-to-device and heterogeneous communications, and ultra-low latency requirements. Channel reciprocity-based key generation (CRKG) is an emerging physical layer-based technique to establish secret keys between devices. This article reviews CRKG when the 5G and beyond networks employ three candidate technologies: duplex modes, massive multiple-input multiple-output (MIMO) and mmWave communications. We identify the opportunities and challenges for CRKG and provide corresponding solutions. To further demonstrate the feasibility of CRKG in practical communication systems, we overview existing prototypes with different IoT protocols and examine their performance in real-world environments. MIL-53(Al)-graphene oxide (GO) nanocomposites of different GO to MIL-53(Al) mass ratios (1% to 25% GO) were synthesized and tested for removal of arsenite (As(III)), which is a well-known groundwater contaminant. The properties of MIL-53(Al)-GO nanocomposites were characterized using X-ray Diffraction (XRD), Fourier Transform Infrared (FT-IR) Spectroscopy, Brunauer-Emmett-Teller (BET) surface area measurements, and Scanning Electron Microscopy (SEM). Batch experiments were performed on MIL-53(Al)-GO nanocomposites for As(III) adsorption in aqueous solutions to investigate adsorption kinetics and isotherm behavior under varying environmental conditions. The effects of solution pH (2 to 11), initial As(III) concentrations (10–110 mg/L), adsorbent dosage (0.2–3.0 g/L), and temperature (298–318 K) on As(III) adsorption were investigated. MIL-53(Al)-GO nanocomposites showed higher adsorption of As(III) than pristine MIL-53(Al) and GO individually. As (III) removal was optimized at a ratio of 3% GO in the MIL-53(Al)-GO nanocomposite, with an adsorption capacity of 65 mg/g. The adsorption kinetics and isotherms followed pseudo-second-order and Langmuir isotherm models, respectively. Overall, these results suggest that MIL-53(Al)-GO nanocomposite holds a significant promise for use in the remediation of As (III) from groundwater and other aqueous solutions. Marine drugs have developed rapidly in recent decades. Cone snails, a group of more than 700 species, have always been one of the focuses for new drug discovery. These venomous snails capture prey using a diverse array of unique bioactive neurotoxins, usually named as conotoxins or conopeptides. These conotoxins have proven to be valuable pharmacological probes and potential drugs due to their high specificity and affinity to ion channels, receptors, and transporters in the nervous systems of target prey and humans. Several research groups, including ours, have examined the venom gland of cone snails using a combination of transcriptomic and proteomic sequencing, and revealed the existence of hundreds of conotoxin transcripts and thousands of conopeptides in each Conus species. Over 2000 nucleotide and 8000 peptide sequences of conotoxins have been published, and the number is still increasing quickly. However, more than 98% of these sequences still lack 3D structural and functional information. With the rapid development of genomics and bioinformatics in recent years, functional predictions and investigations on conotoxins are making great progress in promoting the discovery of novel drugs. For example, ω-MVIIA was approved by the U.S. Food and Drug Administration in 2004 to treat chronic pain, and nine more conotoxins are at various stages of preclinical or clinical evaluation. In short, the genus Conus , the big family of cone snails, has become an important genetic resource for conotoxin identification and drug development.

Most previous studies have focused on analgesic and anti-cancer activities for the conotoxins identified from piscivorous and molluscivorous cone snails, but little attention has been devoted to insecticidal activity of conotoxins from the dominant vermivorous species. As a representative vermivorous cone snail, the Chinese tubular cone snail ( Conus betulinus ) is the dominant Conus species inhabiting the South China Sea. We sequenced related venom transcriptomes from C. betulinus using both the next-generation sequencing and traditional Sanger sequencing technologies, and a comprehensive library of 215 conotoxin transcripts was constructed. In our current study, six conotoxins with potential insecticidal activity were screened out from our conotoxin library by homologous search with a reported positive control (alpha-conotoxin ImI from C. imperialis ) as the query. Subsequently, these conotoxins were synthesized by chemical solid-phase and oxidative folding for further insecticidal activity validation, such as MTT assay, insect bioassay and homology modeling. The final results proved insecticidal activities of our achieved six conotoxins from the transcriptome-based dataset. Interestingly, two of them presented a lot of high insecticidal activity, which supports their usefulness for a trial as insecticides in field investigations. Daphniphyllum calycinum (DC), a main component of Chinese patent drug Fengliao-Changweikang, is effectively used to cure bowel disease in the clinic. It was recorded that DC possessed slight toxicity, which was caused by the alkaloids existing in its extract. Unfortunately, to date the toxicity level and toxic constituents are still unclear. The present study is designed to illustrate the acute toxicity and induced organ damages of the total alkaloids as well as to determine the toxic constituents. Based on the above studies, not only was the acute toxicity determined but also hepatic toxicity was characterized by increased plasma biomarkers of ALT and AST and liver cell inflammatory infiltrate as well necrosis that was firstly observed. Significantly, deoxycalyciphylline B exhibited exactly the same hepatic toxicity so it was identified as the main toxic constituent in DC. An obvious dose-effect relationship between the toxic compound and induced hepatic injuries was also observed. Moreover, the Chinese patent drug Fengliao-Changweikang contained low levels of the toxic compound, compared with the total alkaloids. Therefore, this Chinese patent drug could be regarded to be safe in this point of view.
