Abstract Humic acid (HA) can substantially enhance plant growth and improve soil health. Currently, the impacts of HA concentrations variation on the development and soil quality of Panax notoginseng (Sanqi) from the forest understorey are still unclear. In this study, exogenous HA was administered to the roots of Sanqi at varying concentrations (2, 4, and 6 ml/L). Subsequently, the diversity and community structure of bacteria and fungi were assessed through high-throughput sequencing technology. The investigation further involved analyzing the interplay among the growth of sanqi, soil edaphic factors, and the microbial network stability. Our finding revealed that moderate concentrations (4 ml/L) of HA improved the fresh/dry weight of Sanqi and NO3 -−N levels. Compared with control, the moderate concentrations of HA had a notable impact on the bacterial and fungal communities compositions. However, there was no significant difference in the α and β diversity of bacteria and fungi. Moreover, the abundance of beneficial bacteria (Bradyrhizobium) and harmful bacteria (Xanthobacteraceae) increased and decreased at 4 ml/L HA, respectively, while the bacterial and fungal network stability were enhanced. Structural equation model (SEM) revealed that the fresh weight of Sanqi and bacterial and fungal communities were the factors that directly affected the microbial network stability at moderate concentrations of HA. In conclusion, 4 ml/L of HA is beneficial for promoting Sanqi growth and soil quality. Our study provides a reference for increasing the yield of Sanqi and sustainable development of the Sanqi-pine agroforestry system.

Abstract Objectives Knoxia roxburghii is a member of the madder (Rubiaceae) family. This plant is cultivated in different areas of China and recognized for its medicinal properties, which leads to its use in traditional Chinese medicine. The incidence of root rot was 10–15%. In June 2023, the causal agent of root rot on K. roxburghii was identified as Fusarium oxysporum. To the best of our knowledge, this is the first report of the complete genome of F. oxysporum strain ByF01 that is the causal agent of root rot of K. roxburghii in China. The results will provide effective resources for pathogenesis on K. roxburghii and the prevention and control of root rot on this host in the future. Data description To understand the molecular mechanisms used by F. oxysporum to cause root rot on K. roxburghii, strain ByF01 was isolated from diseased roots and identified by morphological and molecular methods. The complete genome of strain ByF01 was then sequenced using a combination of the PacBio Sequel IIe and Illumina sequencing platforms. We obtained 54,431,725 bp of nucleotides, 47.46% GC content, and 16,705 coding sequences.

Abstract Background Rhizosphere and endophytic fungi play important roles in plant health and crop productivity. However, their community dynamics during the continuous cropping of Knoxia valerianoides have rarely been reported. K. valerianoides is a perennial herb of the family Rubiaceae and has been used in herbal medicines for ages. Here, we used high-throughput sequencing technology Illumina MiSeq to study the structural and functional dynamics of the rhizosphere and endophytic fungi of K. valerianoides. Results The findings indicate that continuous planting has led to an increase in the richness and diversity of rhizosphere fungi, while concomitantly resulting in a decrease in the richness and diversity of root fungi. The diversity of endophytic fungal communities in roots was lower than that of the rhizosphere fungi. Ascomycota and Basidiomycota were the dominant phyla detected during the continuous cropping of K. valerianoides. In addition, we found that root rot directly affected the structure and diversity of fungal communities in the rhizosphere and the roots of K. valerianoides. Consequently, both the rhizosphere and endophyte fungal communities of root rot-infected plants showed higher richness than the healthy plants. The relative abundance of Fusarium in two and three years old root rot-infected plants was significantly higher than the control, indicating that continuous planting negatively affected the health of K. valerianoides plants. Paris polyphylla is an important traditional medicinal plant of the Himalayan region. It is extensively used for the production of natural steroidal saponins and flavonoids. Although seed dormancy of wild plants can be broken to be artificially maintained and regenerated through micropropagation in the laboratory, the success of secondary metabolite production in higher quantities and the synthesis of superior plant metabolites have been very limited. In this study, we present differential metabolic profiling of P. polyphylla plants maintained for 8 years in natural and greenhouse conditions. Untargeted profiling of the metabolites through ultra-performance liquid chromatography-mass spectrometry/mass spectrometry (UPLC-MS/MS), followed by statistical analysis, identified secondary metabolites that were enriched in the naturally occurring plant roots compared with the greenhouse plant roots. A multivariate statistical analysis revealed the differential distribution of the compounds between the two groups. Overall, we identified 1,182 secondary metabolites, with 116 metabolites being differentially upregulated and 256 metabolites being downregulated. Moreover, 810 metabolites showed no significant variation under both growing conditions. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that the naturally forest-grown P. polyphylla plants were significantly enriched in steroidal saponins, lipids, vitamins, flavonoids, and flavonols. An analysis of the top 10 differentially upregulated secondary metabolites indicated a significantly enriched quantity of spirost-5-en-3,12-diol and kaempferol synthesis pathways, which are known to reduce the effect of free radicals scavengers inside the cell. In addition, veratramine alkaloids were also enriched under natural conditions. Our findings indicated that naturally maintained P. polyphylla plants are suitable for the extraction of medicinally important compounds. Our study established a causal relationship between the metabolic composition of the roots and their natural growth condition. This study highlighted the importance of environmental conditions in the biosynthesis of secondary metabolites of plants.

Abstract Phytoalexin sakuranetin functions in resistance against rice blast. However, the mechanisms underlying the effects of sakuranetin remains elusive. Here, we report that rice lines expressing resistance (R) genes were found to contain high levels of sakuranetin, which correlates with attenuated endocytic trafficking of plasma membrane (PM) proteins. Exogenous and endogenous sakuranetin attenuates the endocytosis of various PM proteins and the fungal effector PWL2. Moreover, accumulation of the avirulence protein AvrCO39, resulting from uptake into rice cells by Magnaporthe oryzae, was reduced following treatment with sakuranetin. Pharmacological manipulation of clathrin-mediated endocytic (CME) suggests that this pathway is targeted by sakuranetin. Indeed, attenuation of CME by sakuranetin is sufficient to convey resistance against rice blast. Our data reveals a mechanism of rice against M. oryzae by increasing sakuranetin levels and repressing the CME of pathogen effectors, which is distinct from the action of many R genes that mainly function by modulating transcription.

The microbial communities in rhizosphere soil play important roles in plant health and crop productivity. However, the microbial community structure of rhizosphere soil still remains unclear. In this study, the composition, diversity and function of the microbial communities in the rhizosphere soil of healthy and diseased plants were compared using Illumina MiSeq high-throughput sequencing. The Sobs (richness) and Shannon (diversity) indices of the soil microbial communities were higher in the rhizospheres of 2- and 3-year-old susceptible plants than in those of the healthy plants. With the increase in planting time, the numbers of fungi tended to decrease, while those of the bacteria tended to increase. Fungal diversity could be used as a biological indicator to measure the health of Knoxia roxburghii. The microbial composition and differential analyses revealed that the rhizosphere soil infested with fungi had a higher relative abundance at the phylum level in Ascomycota and Basidiomycota, while the bacteria had a higher relative abundance of Chloroflexi and a lower relative abundance of Actinobacteriota. At the genus level, the rhizosphere soil infested with fungi had relatively more abundant unclassified_f__Didymellaceae and Solicoccozyma and relatively less abundant Saitozyma and Penicillium. The bacterial genus norank_f__Gemmatimonadaceae was the most abundant, while Arthrobacter was less abundant. In addition, the abundance of Fusarium in the fungal community varied (p = 0.001). It tended to increase in parallel with the planting years. Therefore, it was hypothesized that the change in the community composition of Fusarium may be the primary reason for the occurrence of root rot in K. roxburghii, and the change in the abundance of Fusarium OTU1450 may be an indication of the occurrence of root rot in this species. The community function and prediction analyses showed that the pathogenic fungi increased with the increase in planting years. In general, soil fungi can be roughly divided into three types, including pathotrophs, symbiotrophs, and saprotrophs. Abstract Plants usually produce defence metabolites in non-active forms to minimize the risk of harm to themselves and spatiotemporally activate these defence metabolites upon pathogen attack. This so-called two-component system plays a decisive role in the chemical defence of various plants. Here, we discovered that Panax notoginseng, a valuable medicinal plant, has evolved a two-component chemical defence system composed of a chloroplast-localized β-glucosidase, denominated PnGH1, and its substrates 20(S)-protopanaxadiol ginsenosides. The β-glucosidase and its substrates are spatially separated in cells under physiological conditions, and ginsenoside hydrolysis is therefore activated only upon chloroplast disruption, which is caused by the induced exoenzymes of pathogenic fungi upon exposure to plant leaves. This activation of PnGH1-mediated hydrolysis results in the production of a series of less-polar ginsenosides by selective hydrolysis of an outer glucose at the C-3 site, with a broader spectrum and more potent antifungal activity in vitro and in vivo than the precursor molecules. Furthermore, such β-glucosidase-mediated hydrolysis upon fungal infection was also found in the congeneric species P. quinquefolium and P. ginseng. Our findings reveal a two-component chemical defence system in Panax species and offer insights for developing botanical pesticides for disease management in Panax species.

A two-year field trial was conducted in order to assess techniques to control ginger blast and explore the effects of fumigants on soil bacterial microorganisms and ginger metabolites. This study examined the effects of dazomet and chloropicrin on the control of ginger blast and their influence on ginger yield in Luoping County, Yunnan Province, China. The results showed that in 2022, the control effectiveness of dazomet and chloropicrin treatments on ginger blast was 84.33% and 94.67%, respectively. The corresponding yields were 50,154.40 kg/hm2 and 50,296.90 kg/hm2. In 2023, the control effectiveness of dazomet and chloropicrin treatments on ginger blast were 86.33% and 93.67%, respectively, and the yields were 65,115.83 kg/hm2 and 65,337.93 kg/hm2. In both years, the incidence of ginger blast in the control group reached nearly 100%, leading to the near extinction of the crop. Additionally, in 2023, 16S rRNA high-throughput sequencing and non-targeted metabolomics techniques were used to analyze the effects of the fumigants on soil bacterial microorganisms and the metabolites in ginger. The results showed that the diversity and richness of soil bacterial communities were lower than those in the control group at 0 and 120 days after treatment with two fumigants, but the relative abundances of beneficial bacteria such as Pseudomonas increased at 60 days, and the relative abundances of Actinobacteria, Gemmatimonadetes, and Bacillus increased at 120 days. The abundance of Firmicutes also increased after 120 days of chloropicrin treatment. The non-targeted metabolic LC–MS results showed that the production of phenols and terpenoids was upregulated after dazomet and chloropicrin treatments. The contents of amino acids and their derivatives were also upregulated. This upregulation of metabolites was beneficial to the flavor quality of ginger and enhanced its anti-inflammatory, anti-tumoral, and antioxidant effects.

The main driving factors of river ecological environment were analyzed to reveal the response mechanism of river ecosystem to ecological environmental factors. The results showed that the driving factors of river water quality were resistivity, COD and reoxidation potential, the driving factors of soil environment along river banks were total phosphorus, total nitrogen and pH, and the driving factors of plant nutrition along river banks were total potassium and total nitrogen. The contribution rates of water quality, soil and plant to river ecological environment health were 43, 51 and 70%, respectively. The comprehensive ecological environment of Menghuo River, Gongyihai, Arulendi River, Yiniu River, Dahonggou River and Nanya River is superior to each other, with the comprehensive proximity index of 0.6258, 0.5908, 0.5524, 0.5265, 0.5195 and 0.3889, respectively. Each detection index can accurately and truly invert the ecological environment health status of rivers in the protected areas. Drought stress (DS) is one of the important abiotic stresses facing cash crops today. Drought can reduce plant growth and development, inhibit photosynthesis, and thus reduce plant yield. In this experiment, we investigated the protective mechanism of AMF on plant photosynthetic system by inoculating Paris polyphylla var. yunnanensis(P.py) with a clumping mycorrhizal fungus (AMF) under drought conditions. The drought environment was maintained by weighing AMF plants and non-AMF plants. The relative water content (RWC) of plant leaves was measured to determine its drought effect. DS decreased the RWC of plants, but AMF was able to increase the RWC of plants. chlorophyll a fluorescence curve measurements revealed that DS increased the OKJIP curve of plants, but AMF was able to reduce this trend, indicating that AMF increased the light absorption capacity of plants. DS also caused a decrease in plant Y(I) and Y(II). ETRI and ETRII, and increased Y(NO) and Y(NA) in plants, indicating that DS caused photosystem damage in plants. For the same host, different AMFs did not help to the same extent, but all AMFs were able to help plants reduce this damage and contribute to the increase of plant photosynthesis under normal water conditions.

The Shangri-La region, located in southwest China, is known for its dry climate and intense UltraViolet (UV) radiation due to its high altitude. Research conducted in the past has demonstrated that UV radiation, particularly UV-B radiation, enhances plants' ability to withstand cold temperatures, diseases, and arid conditions. Indeed, the Cabernet Sauvignon grape variety, predominantly cultivated in Shangri-La where the UV intensity reaches 21–22w/m2, has displayed remarkable resilience in recent decades. However, the specific impact of UV-B radiation on Cabernet Sauvignon grape leaves has yet to be fully understood. Consequently, our objective is to investigate the influence of UV radiation on grapes grown at high altitudes. To gain insights into the short-term effects on grape leaves, we conducted a comprehensive investigation involving anatomical and photosynthetic changes. Additionally, we analyzed the transcriptomic and proteomic regulation associated with these changes following exposure to UV-B radiation at intensities ranging from 15–20w/m2 for durations of 3 and 7 days.The findings of our study demonstrated that exposure to UV-B radiation, particularly at intensities surpassing 10w/m2, led to noticeable changes in the morphology of epidermal and palisade tissue cells. Furthermore, after a 3-day exposure to UV-B, we observed an upregulation of proteins responsible for cytoskeletal organization and cell wall synthesis. Conversely, following a 7-day treatment, proteins associated with disease resistance and antioxidants showed increased expression. Transcriptomic analysis revealed a significant enrichment of proteins and genes linked to pathogen infection, defense mechanisms, and antioxidant processes. Overall, our research highlighted the positive impact of intense UV radiation at high altitudes on enhancing disease resistance and environmental adaptability in Cabernet Sauvignon grape plants.

Panax notoginseng is a traditional Chinese medicinal herb. With its wide applications in medicine, food products, health care, cosmetics and other industries, identification and detection of P. notoginseng have become critical. In this study, we developed a real-time polymerase chain reaction (qPCR) assay with specific and internal control probes based on chloroplast psbA-trnH gene and 18S rDNA to identify and detect P. notoginseng in different products. A specific primer and probe set was designed by comparing psbA-trnH sequence among P. notoginseng, Panax ginseng, Panax quinquefolius and Panax japonicus. Besides, the 18S rDNA gene was used as an internal control. The qPCR condition was optimized with the sensitivity to be 0.005 ng/μL and high specificity. There was no cross-reaction among 18 closely related common Chinese herbs. Detection results of 28 commercially available P. notoginseng products were consistent with the standardized sequencing results. This study provides a rapid, specific and sensitive assay for detection of P. notoginseng in different products.

Panax notoginseng is a perennial plant well known for its versatile medicinal properties, including hepatoprotective, antioxidant, anti-inflammatory, anti-tumor, estrogen-like, and antidepressant characteristics. It has been reported that plant age affects the quality of P. notoginseng. This study aimed to explore the differential metabolome and transcriptome of 2-year (PN2) and 3-year-old (PN3) P. notoginseng plant root samples. Principal component analysis of metabolome and transcriptome data revealed major differences between the two groups (PN2 vs. PN3). A total of 1813 metabolites and 28,587 genes were detected in this study, of which 255 metabolites and 3141 genes were found to be differential (p < 0.05) between PN2 vs. PN3, respectively. Among differential metabolites and genes, 155 metabolites and 1217 genes were up-regulated, while 100 metabolites and 1924 genes were down-regulated. The KEGG pathway analysis revealed differentially enriched metabolites belonging to class lipids (“13S-hydroperoxy-9Z, 11E-octadecadionic acid”, “9S-hydroxy-10E, 12Z-octadecadionic acid”, “9S-oxo-10E, 12Z-octadecadionic acid”, and “9,10,13-trihydroxy-11-octadecadionic acid”), nucleotides and derivatives (guanine and cytidine), and phenolic acids (chlorogenic acid) were found to be enriched (p < 0.05) in PN3 compared to PN2. Further, these differentially enriched metabolites were found to be significantly (p < 0.05) regulated via linoleic acid metabolism, nucleotide metabolism, plant hormone signal transduction, and arachidonic acid metabolism pathways. Furthermore, the transcriptome analysis showed the up-regulation of key genes MAT, DMAS, SDH, gallate 1-beta-glucosyltransferase, and beta-D-glucosidase in various plants’ secondary metabolic pathways and SAUR, GID1, PP2C, ETR, CTR1, EBF1/2, and ERF1/2 genes observed in phytohormone signal transduction pathway that is involved in plant growth and development, and protection against the various stressors. This study concluded that the roots of a 3-year-old P. notoginseng plant have better metabolome and transcriptome profiles compared to a 2-year-old plant with importantly enriched metabolites and genes in pathways related to metabolism, plant hormone signal transduction, and various biological processes. These findings provide insights into the plant’s dynamic biochemical and molecular changes during its growth that have several implications regarding its therapeutic use.

The occurrence of root-knot nematode disease has seriously constrained the development of the Chinese herbal medicine industry. China is one of the largest producers of Polygonatum sibiricum and Peucedanum praeruptorum in the world, but the unidentified root-knot nematodes have become important pests of these two Chinese herbal medicines in China. Both morphological characteristics and molecular identification were used to identify the nematodes. The identification results showed that Meloidogyne incognita and M. arenaria were the causal species of root-knot nematode infection in P. sibiricum, and M. hapla was the causal species of the infection in P. praeruptorum. Through investigation, this is the first report of M. incognita and M. arenaria infecting P. sibiricum, and M. hapla infecting P. praeruptorum, in China. The two Chinese herbs are being severely damaged by various root-knot nematodes, and this damage should be taken seriously.

Panax notoginseng (Sanqi) is a precious traditional Chinese medicine that is commonly cultivated using conventional management methods in agricultural systems in Yunnan, China, where it faces the challenge of continuous cropping obstacles (CCOs). However, the existence of Sanqi CCOs in Sanqi–pine agroforestry systems remains unclear. Here, we applied three types of exogenous organic acids (phthalic acid, palmitic acid, and phthalic + palmitic mixed organic acids) mainly derived from the root exudates of Sanqi to simulate the CCOs; then, we compared the effects on plant growth, soil physicochemical properties, soil microbes, and soil metabolites. We found that organic acid concentrations of >250 mg/kg reduced the fresh weight of Sanqi and the levels of total nitrogen, ammonium nitrogen, soil water content, total phosphorus, and pH value; these concentrations also increased the soil bacterial and fungal α-diversity. The type of organic acid, as opposed to the concentration and duration of treatment, had a vital impact on the structure of the bacterial and fungal community in Sanqi soils. Moreover, the organic acid concentrations of >250 mg/kg also decreased the complexity and stability of the bacterial and fungal network. In addition, the metabolic pathways in the soils under different organic acids included 17 differential metabolites (DMs), which were involved in steroid hormone biosynthesis. The structural equation models (SEMs) revealed that plant growth, soil edaphic factors, and soil metabolites had direct or indirect influences on soil microbial communities under different organic acid conditions. Our results suggest that any phthalic or palmitic acid concentrations at concentrations >250 mg/kg are detrimental to multiple aspects of Sanqi cultivation, confirming the presence of Sanqi CCOs in Sanqi–pine agroforestry systems.

Panax notoginseng (P. notoginseng) is an invaluable perennial medicinal herb. However, the roots of P. notoginseng are frequently subjected to severe damage caused by root-knot nematode (RKN) infestation. Although we have observed that P. notoginseng possessed adult-plant resistance (APR) against RKN disease, the defense response mechanisms against RKN disease in different age groups of P. notoginseng remain unexplored. We aimed to elucidate the response mechanisms of P. notoginseng at different stages of development to RKN infection by employing transcriptome, metabolome, and histochemistry analyses. Our findings indicated that distinct age groups of P. notoginseng may activate the phenylpropanoid and flavonoid biosynthesis pathways in varying ways, leading to the synthesis of phenolics, flavonoids, lignin, and anthocyanin pigments as both the response and defense mechanism against RKN attacks. Specifically, one-year-old P. notoginseng exhibited resistance to RKN through the upregulation of 5-O-p-coumaroylquinic acid and key genes involved in monolignol biosynthesis, such as PAL, CCR, CYP73A, CYP98A, POD, and CAD. Moreover, two-year-old P. notoginseng enhanced the resistance by depleting chlorogenic acid and downregulating most genes associated with monolignol biosynthesis, while concurrently increasing cyanidin and ANR in flavonoid biosynthesis. Three-year-old P. notoginseng reinforced its resistance by significantly increasing five phenolic acids related to monolignol biosynthesis, namely p-coumaric acid, chlorogenic acid, 1-O-sinapoyl-D-glucose, coniferyl alcohol, and ferulic acid. Notably, P. notoginseng can establish a lignin barrier that restricted RKN to the infection site. In summary, P. notoginseng exhibited a potential ability to impede the further propagation of RKN through the accumulation or depletion of the compounds relevant to resistance within the phenylpropanoid and flavonoid pathways, as well as the induction of lignification in tissue cells.

Panax notoginseng is highly used in traditional Chinese medicine, and its root is valued for ginsenoside contents. However, evidence suggests that other plant parts such as fruit, leaves, and stem are also potential sources of bioactive compounds. Therefore, this study aimed at providing insight into the differential accumulation of metabolites in fruit, leaf, root, and stem tissues collected from P. notoginseng. A total of 808 metabolites from 11 major metabolite classes were identified. Furthermore, 32 Ginsenosides with six conserved Ginsenosides Rg1, Rf, St-3, R1, Ro, Rc, and five conserved notoginsenosides R1, K, M, E, Rb1, were identified with differential accumulation in fruit, leaf, stem, and root tissues. Several other metabolites known for their potential roles in human health were also identified as differentially accumulated in the tissues. Furthermore, the accumulation pattern of ginsenosides in different tissues is highly suggestive of utilizing fruit, leaf, and stem tissues along with roots for value addition of P. notoginseng.

Plant nucleotide-binding and leucine-rich repeat (NLR) proteins are immune sensors that detect pathogen effectors and initiate a strong immune response. In many cases, single NLR proteins are sufficient for both effector recognition and signaling activation. These proteins possess a conserved architecture, including a C-terminal leucine-rich repeat (LRR) domain, a central nucleotide-binding (NB) domain, and a variable N-terminal domain. Nevertheless, many paired NLRs linked in a head-to-head configuration have now been identified. The ones carrying integrated domains (IDs) can recognize pathogen effector proteins by various modes; these are known as sensor NLR (sNLR) proteins. Structural and biochemical studies have provided insights into the molecular basis of heavy metal-associated IDs (HMA IDs) from paired NLRs in rice and revealed the co-evolution between pathogens and hosts by combining naturally occurring favorable interactions across diverse interfaces. Focusing on structural and molecular models, here we highlight advances in structure-guided engineering to expand and enhance the response profile of paired NLR-HMA IDs in rice to variants of the rice blast pathogen MAX-effectors (Magnaporthe oryzae AVRs and ToxB-like). These results demonstrate that the HMA IDs-based design of rice materials with broad and enhanced resistance profiles possesses great application potential but also face considerable challenges.

Continuous cropping of the important achlorophyllous medicinal orchid Gastrodia elata Blume causes an imbalance in soil microecology leading to soil-borne diseases. However, the impacts on different land covers remain largely unknown. Hence, this study aimed to investigate changes in the soil nutrient composition and the global microbial community structure in rhizospheres of G. elata cultivated on a barren slope (HPGJ) and under a forest (LXT) using integrated shotgun metagenomics and an analysis of soil chemical properties. High-throughput sequencing revealed an increase in the abundance of Proteobacteria, Actinobacteria, Mucoromycota, Basidiomycota, and Ascomycota, which drive N- and C-cycling genes in HPGJ and LXT. Notably, the fungal community was significantly improved in the HPGJ (from 0.17% to 23.61%) compared to the LXT (from 0.2% to 2.04%). Consequently, mineral cycling was enhanced in the HPGJ, resulting in a more improved soil nutrient composition than in the LXT. The soil chemical properties analysis unveiled a significant increase in the contents of the total nitrogen, NO3−-N, organic matter, total carbon, organic carbon, total sulfur, and total phosphorus in the HPGJ, while no changes were recorded in the LXT. It was noteworthy that the abundance of pathogenic microorganisms increased significantly in the HPGJ compared to the LXT. Our results provide supporting data to optimize G. elata cultivation on slopes.

Chimonanthus praecox is an aromatic plant that flowers in winter. The composition of the floral volatiles of C. praecox is influenced by different blooming stages, circadian rhythms and species. However, the relationship between floral volatiles and plant endophytic fungi has not received much research attention. Here, we used high-throughput sequencing technology to compare and analyze the changes in the structure and diversity of the endophytic fungal communities in C. praecox under different circadian rhythms (7:00 a.m., 1:00 p.m., and 7:00 p.m.) and in different blooming stages (unopened flowers and opened flowers). The endophytic fungi of C. praecox consisted of nine phyla, 34 classes, 79 orders, 181 families, 293 genera, and 397 species, and Ascomycota was the dominant phylum. Under a diurnal rhythm, the diversity (Chao1 and Shannon indices) of endophytic fungi gradually decreased in the unopened flowers, while an increasing and then decreasing trend was found for the opened flowers. In the different blooming stages, the endophytic fungal diversity was significantly higher at 7:00 a.m. in the unopened flowers compared to the opened flowers. Humidity was the key factors that significantly affected the endophytic fungal diversity and community. Moreover, 11 endophytic fungi were significantly positively or negatively correlated with seven floral volatiles. In conclusion, the community structure and diversity of endophytic fungi in C. praecox were affected by the different blooming stages and circadian rhythms, and a correlation effect related to floral volatiles was found, but there are other possible reasons that were not tested. This study provides a theoretical basis for elucidating the interrelationships between endophytic fungi, floral volatiles, and environmental factors in C. praecox.

The rhizosphere bacteria Bacillus velezensis GJ-7, as a biological control agent (BCA), has significant biological control effects on Meloidogyne hapla, and has strong colonization ability in the root of Panax notoginseng. In this study, we conducted a comparative transcriptome analysis using P. notoginseng plant roots treated with B. velezensis GJ-7 or sterile water alone and in combination with M. hapla inoculation to explore the interactions involving the P. notoginseng plant, B. velezensis GJ-7, and M. hapla. Four treatments from P. notoginseng roots were sequenced, and twelve high-quality total clean bases were obtained, ranging from 3.57 to 4.74 Gb. The Gene Ontology (GO) classification and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment showed that numerous DEGs are involved in the phenylpropane biosynthesis pathway and the MAPK signaling pathway in the roots of P. notoginseng with B. velezensis GJ-7 treatments. The analysis results of the two signaling pathways indicated that B. velezensis GJ-7 could enhance the expression of lignin- and camalexin-synthesis-related genes in plant roots to resist M. hapla. In addition, B. velezensis GJ-7 could enhance plant resistance to M. hapla by regulating the expression of resistance-related genes and transcription factors (TFs), including ETR, ERF, ChiB, WRKY22, and PR1. The expression of plant disease resistance genes in the roots of P. notoginseng with different treatments was validated by using real-time quantitative PCR (qRT-PCR), and the results were consistent with transcriptome sequencing. Taken together, this study indicated that B. velezensis GJ-7 can trigger a stronger defense response of P. notoginseng against M. hapla.

Epiverta chelonia (Mader 1933; Coleoptera: Coccinellidae) is an important economically and scientifically valuable insect. In this study, the first complete mitochondrial genome of E. chelonia was sequenced and characterized using next-generation sequencing techniques. The circular mitogenome of E. chelonia consists of 17,347 bp including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a control region (D-loop). The base composition was AT-biased (75.77%). Bayesian Inference and Maximum likelihood phylogenetic trees strongly supported the monophyly of Coccinellinae. Also, E. chelonia was supported as the sister group of Subcoccinella vigintiquatuorpunctata, within Epilachninae. Thus, the E. chelonia mitochondrial genome will be a fundamental resource for understanding the molecular phylogenetic relationships of the species-rich family Coccinellidae of Coleoptera.

Round spot is a destructive disease that limits of Panax notoginseng production in China. However, the genetic diversity of its etiological agent Mycocentrospora acerina has yet to be studied. In this work, firstly, we developed 32 M. acerina polymorphic microsatellite markers using MISA and CERVUS 3.0 and selected 14 for further analysis. Then, we studied the genetic diversity of 187 isolates collected from P. notoginseng round spot using simple sequence repeat markers and polyacrylamide gel electrophoresis. The genetic diversity ranged from 0.813 to 0.946, with 264 alleles detected at the 14 microsatellite loci. The expected average heterozygosity was 0.897.

The integrity of the packaging of a liquid foodstuff makes it difficult to detect spoilage. Therefore, it is important to develop a sensitive, fast and real-time material for liquid food detection. CMC, as lignocellulose derivatives and starch are widely used in the food industry. In this study, starch films with pH-responsive properties are successfully prepared from full-component starch and corn amylopectin (CA) by adding CMC. The effects of CMC on the mechanical properties, morphology characteristics, physical and chemical structures, stability and pH responsiveness of the starch films are analyzed. The starch/CMC-1.0 g composite films display good electrical conductivity and reduce the resistance of the composite film by two orders of magnitude. The composite films have pH response ability; in the simulation of orange juice spoilage experiment, the CA/CMC composite film has a more sensitive current response and was more suitable for the application to liquid food quality detection. Additionally, the starch/CMC composite films have potential applications for rapid detection and real-time monitoring of the safety of liquid food.

ABSTRACT Negative plant-soil feedback (NPSF) due to the buildup of soilborne pathogens in soil is a major obstacle in sustainable agricultural systems. Beneficial rhizosphere microfloras are recruited by plants, and mediating this has become a strategic priority to manipulate plant health. Here, we found that foliar infection of Panax notoginseng by Alternaria panax changed plant-soil feedback from negative to positive. Foliar infection modified the rhizosphere soil microbial community and reversed the direction of the buildup of the soilborne pathogen Ilyonectria destructans and beneficial microbes, including Trichoderma, Bacillus, and Streptomyces, in rhizosphere soil. These beneficial microbes not only showed antagonistic ability against the pathogen I. destructans but also enhanced the resistance of plants to A. panax. Foliar infection enhanced the exudation of short- and long-chain organic acids, sugars, and amino acids from roots. In vitro and in vivo experiments validated that short- and long-chain organic acids and sugars play dual roles in simultaneously suppressing pathogens but enriching beneficial microbes. In summary, foliar infection could change root secretion to drive shifts in the rhizosphere microbial community to enhance soil health, providing a new strategy to alleviate belowground disease in plants through aboveground inducement. IMPORTANCE Belowground soilborne disease is the main factor limiting sustainable agricultural production and is difficult to manage due to the complexity of the soil environment. Here, we found that aboveground parts of plants infected by foliar pathogens could enhance the secretion of organic acids, sugars, and amino acids in root exudates to suppress soilborne pathogens and enrich beneficial microbes, eventually changing the plant and soil feedback from negative to positive and alleviating belowground soilborne disease. This is an exciting strategy by which to achieve belowground soilborne disease management by manipulating the aboveground state through aboveground stimulation.

Land-use conversion affects the composition and assembly of plant-associated microbiomes, which in turn affects plant growth, development, and ecosystem functioning. However, agroforestry systems, as sustainable land types, have received little attention regarding the dynamics of different plant-associated microbes. In this study, we used high-throughput sequencing technology to analyze the assembly mechanisms and the driving factors of pine- and Panax notoginseng (P.n.)-associated microbiomes during the conversion of different pine forests (Pinus kesiya var. langbianensis and Pinus armandii) into P.n.-pine agroforestry systems. The results showed that the conversion of pure pine forest into P.n.-pine agroforestry systems significantly altered the diversity of pine-associated fungi rather than the community structure, and the community structure of P.n.-associated fungi rather than the diversity. Additionally, plant-associated fungi were more responsive to land-use change than bacteria. Main effect analysis revealed that compartment rather than genotype was the driving factor of pine- and P.n.-associated microbiomes, but P.n. cultivation also significantly affected the assembly of pine-associated microbiomes. In addition, there was a transfer of P.n. endophytes to pine trees in agroforestry systems and the beneficial microbiomes (Massilia, Marmoricola, Herbaspirillum, etc.) were enlarged in pine roots. Therefore, the diversity of the assembly mechanisms of P.n.- and pine-associated microbiomes played an important role in the P.n.--pine agroforestry systems and were the basis for the sustainable development of the P.n.--pine agroforestry systems.

ABSTRACT Harnessing indigenous soil microbial suppression is an emerging strategy for managing soilborne plant diseases. Soil moisture is a vital factor in soil microbiomes, but its role in the regulation of microbial suppression is poorly understood. Here, we investigated the correlation of root rot disease of Panax notoginseng with rhizosphere microbial communities mediated by soil moisture gradients from 55% to 100% field capacity (FC); then, we captured the disease-suppressive and disease-inductive microbiomes and validated their functions by a culture experiment with synthetic microbiotas containing keystone species. We found that proper soil moisture at 75% to 95% FC could maintain a disease-suppressive microbiome to alleviate root rot disease. However, extremely low or high soil moistures (>95% FC or

A comparative study was conducted among the flesh (SOF) and pericarp (SOP) of Stauntonia obovatifoliola, a wild edible fruit in China. The nutrient composition of both these tissues was firstly quantified, and liquid-liquid extraction was then used to separate their methanolic extracts to get petroleum ether, chloroform, ethyl acetate, n-butanol, and residual aqueous fractions, which were evaluated for their total phenol content (TPC), total flavonoid content (TFC), antioxidant capacities, and α-glucosidase and acetylcholinesterase inhibition abilities. Finally, high-performance liquid chromatography (HPLC) was used to analyze their phytochemical composition. The results revealed the excellent nutritional properties of both SOF and SOP, especially SOP (total dietary fiber, 15.50 g/100 g; total amino acids, 0.80 g/100 g; vitamin C, 18.00 mg/100 g; Ca, 272.00 mg/kg; K, 402.00 mg/100 g). For both tissues, their ethyl acetate fractions showed the highest TPC (355.12 and 390.99 mg GAE/g DE) and TFC (306.58 and 298.48 mg RE/g DE). Surprisingly, the ethyl acetate fraction of SOP exhibited the strongest DPPH and ABTS radical scavenging capacity with 1046.94 and 1298.64 mg Trolox/g, respectively, which were higher than that of controls Vc and BHT. In contrast, their chloroform fractions exhibited the strongest ferric reducing antioxidant power (1903.05 and 1407.11 mg FeSO4/g DE) and oxygen radical absorbance capacity (951.12 and 1510.21 mg Trolox/g DE). In addition, the ethyl acetate fraction of SOF displayed superior α-glucosidase inhibition ability with the IC50 value of 0.19 mg/mL, which was comparable to control acarbose. In comparison, the ethyl acetate fraction of SOP had the best acetylcholinesterase inhibition ability with the IC50 value of 0.47 mg/mL. The HPLC analysis results demonstrated that the ethyl acetate fraction of SOP showed significantly higher phenolic content, particularly for phenolic acids (p-hydroxybenzoic acid, 8.00 ± 0.65 mg/g) and flavonoids (epicatechin, 28.63 ± 1.26 mg/g), as compared to other samples. The above results suggest that Stauntonia obovatifoliola, especially its pericarp, had excellent nutrient compositions, bioactive properties and phytochemical characteristics, and had the potential to be developed as natural functional food.

The objective of the research was to investigate the digestion and fecal fermentation characteristics of the flowers of Juglans regia (FJR), by using in vitro simulated digestion model (oral, gastric, and intestine) as well as colonic fermentation. As a result, the contents of most active substances and functional activities of FJR were decreased as the digestion proceeded, and showed a trend of first increasing and then decreasing in the fecal fermentation phase. In the oral digestion phase, the total phenolic and total flavonoid contents were released most with the values of 11.43 and 9.41 μg/mg, respectively. While in the gastric digestion phase, the antioxidant abilities, α-glucosidase and α-amylase inhibitory abilities were the weakest. By using high-performance liquid chromatography, 13 phenolic acids and 3 flavonoids were detected. Of these, the highest number of identified compounds were found in the undigested and the oral digestion stages, which were mainly salicylic acid, epicatechin, 3,5-dihydroxybenoic acid, vanillic acid, and protocatechuic acid. However, great losses were observed during the gastric and intestinal digestion stages, only epicatechin, salicylic acid, and protocatechuic acid were found. Surprisingly, fecal fermentation released more abundant phenolic substances compared to gastric and intestinal digestion. Additionally, FJR reduced the pH values in the colonic fermentation system, significantly promoted the production of short-chain fatty acids, and regulated the microbe community structure by improving the community richness of beneficial microbiota. This indicated that FJR had the benefit to improve the microorganismal environment in the intestine. Further Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that FJR could suppress the metabolic pathways related to diseases, such as infectious diseases, metabolic diseases and neurodegenerative diseases. In conclusion, although the bioactivities of FJR decreased significantly after in vitro gastrointestinal digestion and fecal fermentation, it still maintained certain antioxidant and hypoglycemic ability in vitro. This study described the detailed changes in the active compounds and bioactivities of FJR during in vitro gastrointestinal digestion and fecal fermentation, and its effects on microbiota composition and SCFAs levels in feces. Our results revealed the potential health benefits of FJR, and could provide a reference for its further research and development.

The objective of the research was to investigate and compare the bioactivities and bioaccessibility of the polyphenols (PPs) from Dendrobium officinale (DO) and probiotic fermented Dendrobium officinale (FDO), by using in vitro simulated digestion model under oral, gastric and intestinal phases as well as colonic fermentation. The results indicated that FDO possessed significantly higher total phenolic contents (TPC) and total flavonoid contents (TFC) than DO, and they were released most in the intestinal digestion phase with 6.96 ± 0.99 mg GAE/g DE and 10.70 ± 1.31 mg RE/g DE, respectively. Using high-performance liquid chromatography (HPLC), a total of six phenolic acids and four flavonoids were detected. In the intestinal phase, syringaldehyde and ferulic acid were major released by DO, whereas they were p-hydroxybenzoic acid, vanillic acid, and syringic acid for FDO. However, apigenin and scutellarin were sustained throughout the digestion whether DO or FDO. As the digestive process progressed, their antioxidant ability, α-amylase and α-glucosidase inhibitory activities were increased, and FDO was overall substantially stronger in these activities than that of DO. Both DO and FDO could reduce pH values in the colonic fermentation system, and enhance the contents of short-chain fatty acids, but there were no significantly different between them. The results of the 16S rRNA gene sequence analysis showed that both DO and FDO could alter intestinal microbial diversity during in vitro colonic fermentation. In particular, after colonic fermentation for 24 h, FDO could significantly improve the ratio of Firmicutes to Bacteroidetes, and enrich the abundancy of Enterococcus and Bifidobacterium (p < 0.05), which was most likely through the carbohydrate metabolism signal pathway. Taken together, the PPs from DO and FDO had good potential for antioxidant and modulation of gut bacterial flora during the digestive processes, and FDO had better bioactivities and bioaccessibility. This study could provide scientific data and novel insights for Dendrobium officinale to be developed as functional foods.

Rapeseed straw, bagasse, and walnut peel have a large amount of resource reserves, but there are few technologies for high value-added utilization. In the research of biochar, walnut green husk is rarely used as raw material. In addition, the three main components of biomass (lignin, cellulose, and hemicellulose) are present in similar proportions, and the differences between the physical and chemical properties of biochar prepared with similar amounts of biomass raw materials are not clear. Using three kinds of biomass of the same quality as raw materials, biochar was prepared via pyrolysis at 400 °C, and activated carbon was prepared via CO2 activation at 800 °C. The results showed that the pore numbers of the three kinds of biochar increased after activation, resulting in the increase of the specific surface area. The resulting numbers were 352.99 m2/g for sugarcane bagasse biochar (SBB)-CO2, 215.04 m2/g for rapeseed straw biochar (RSB)-CO2, and 15.53 m2/g for walnut green husk biochar (WGB)-CO2. Ash increased the amount of carbon formation, but a large amount of ash caused biochar to form a perforated structure and decreased the specific surface area (e.g., WGB), which affected adsorption ability. When the three main components were present in similar proportions, a high content of cellulose and lignin was beneficial to the preparation of biochar. The adsorption value of MB by biochar decreased with the increase of biomass ash content. After activation, the maximum adsorption value of MB for bagasse biochar was 178.17 mg/g, rapeseed straw biochar was 119.25 mg/g, and walnut peel biochar was 85.92 mg/g when the concentration of methene blue solution was 300 mg/L and the biochar input was 0.1 g/100 mL at room temperature. The adsorption of MB by biochar in solution occurs simultaneously with physical adsorption and chemical adsorption, with chemical adsorption being dominant. The optimal MB adsorption by SBB-CO2 was dominated by multimolecular-layer adsorption. This experiment provides a theoretical basis for the preparation of biochar and research on its applications in the future.

To assess the effects of microbial fermentation on Gynostemma pentaphyllum leaves (GPL), four probiotics were used to ferment GPL (FGPL) for 7 days. At different stages of fermentation, changes in the active components and biological activities of FGPL were determined. The findings suggest that short-term fermentation with probiotics can enhance both the content and bioactivity of active components in GPL. However, prolonged fermentation may lead to a decline in these aspects. Among them, the best effect was observed with SWFU D16 fermentation for 2 days. This significantly improved the total phenolic and total flavonoid content, antioxidant capacity, and inhibitory ability against α-glucosidase activity with an increase of 28%, 114.82%, 7.42%, and 31.8%, respectively. The high-performance liquid chromatography (HPLC) analysis results also supported this trend. Untargeted metabolomics analysis revealed metabolite changes between GPL and FGPL and the key metabolites associated with these functional activities. These key metabolites are mainly organic acids, flavonoids, carbohydrates, terpenoids, and other substances. KEGG analysis demonstrated that microbial metabolism in diverse environments and carbon metabolism were the most significantly enriched pathways. Among them, 3-(3-hydroxyphenyl) propanoic acid, d-glucose, gallic acid, gluconic acid, l-lactic acid, and l-malic acid were mostly involved in the microbial metabolism of diverse environmental pathways. In contrast, D-glucose, gluconic acid, and l-malic acid were mainly related to the carbon metabolism pathway. This study revealed the positive effect of probiotic fermentation on GPL and its potential metabolism mechanism, which could provide supporting data for further research.

Biomass carbon dots (BCDs) have the advantages of being nontoxic, low cost and simple to prepare, have excellent optical properties, good biocompatibility and stability, and therefore have broad application prospects in areas such as heavy metal ion detection and optoelectronic devices. Herein, a simple, green, solvent-free method of preparing BCDs was developed. CDs with certain fluorescence properties were prepared by a solvent-free pyrolysis method at different temperatures using two abundant components (cellulose and lignin) of biomass resources as carbon sources. Both the cellulose CDs prepared at 300°C and the lignin CDs prepared at 350°C exhibited high quantum yields of 11.7% and 23.4%, respectively, a result that was mainly due to the high degree of graphitization. The analysis and results demonstrated the selectivity of CDs for the detection of various metal ion solutions. In particular, CDs are sensitive to Fe3+ and can be used as a fluorescent sensor for the detection of Fe3+, providing a more efficient, sustainable alternative for metal ion detection.

This study aimed to investigate the impact of probiotic fermentation on the active components and functions of Perilla frutescens leaves (PFL). PFL was fermented for 7 days using six probiotics (Lacticaseibacillus Paracasei SWFU D16, Lactobacillus Plantarum ATCC 8014, Lactobacillus Rhamnosus ATCC 53013, Streptococcus Thermophilus CICC 6038, Lactobacillus Casei ATCC 334, and Lactobacillus Bulgaricus CICC 6045). The total phenol and flavonoid contents, antioxidant abilities, as well as α-glucosidase and acetylcholinesterase inhibition abilities of PFL during the fermentation process were evaluated, and its bioactive compounds were further quantified by high-performance liquid chromatography (HPLC). Finally, non-targeted ultra-HPLC–tandem mass spectroscopy was used to identify the metabolites affected by fermentation and explore the possible mechanisms of the action of fermentation. The results showed that most of the active component contents and functional activities of PFL exhibited that it first increased and then decreased, and different probiotics had clearly distinguishable effects from each other, of which fermentation with ATCC 53013 for 1 day showed the highest enhancement effect. The same trend was also confirmed by the result of the changes in the contents of 12 phenolic acids and flavonoids by HPLC analysis. Further metabolomic analysis revealed significant metabolite changes under the best fermentation condition, which involved primarily the generation of fatty acids and their conjugates, flavonoids. A total of 574 and 387 metabolites were identified in positive ion and negative ion modes, respectively. Results of Spearman’s analysis indicated that some primary metabolites and secondary metabolites such as flavonoids, phenols, and fatty acids might play an important role in the functional activity of PFL. Differential metabolites were subjected to the KEGG database and 97 metabolites pathways were obtained, of which biosyntheses of unsaturated fatty acids, flavonoid, and isoflavonoid were the most enriched pathways. The above results revealed the potential reason for the differences in metabolic and functional levels of PFL after fermentation. This study could provide a scientific basis for the further study of PFL, as well as novel insights into the action mechanism of probiotic fermentation on the chemical composition and biological activity of food/drug.

The interactions between non-host roots and pathogens may be key to the inhibition of soilborne pathogens in intercropping systems. Fennel (Foeniculum vulgare) can be intercropped with a wide range of other plants to inhibit soilborne pathogens in biodiversity cultivation. However, the key compounds of fennel root exudates involved in the interactions between fennel roots and pathogens are still unknown. Here, a greenhouse experiment confirmed that intercropping with fennel suppressed pepper (Capsicum annuum) blight disease caused by Phytophthora capsici. Experimentally, the roots and root exudates of fennel can effectively interfere with the infection process of P. capsici at rhizosphere soil concentrations by attracting zoospores and inhibiting the motility of the zoospores and germination of the cystospores. Five terpene compounds (D-limonene, estragole, anethole, gamma-terpenes, and beta-myrcene) that were identified in the fennel rhizosphere soil and root exudates were found to interfere with P. capsica infection. D-limonene was associated with positive chemotaxis with zoospores, and a mixture of the five terpene compounds showed a strong synergistic effect on the infection process of P. capsici, especially for zoospore rupture. Furthermore, the five terpene compounds can induce the accumulation of reactive oxygen species (ROS), especially anethole, in hyphae. ROS accumulation may be one of the antimicrobial mechanisms of terpene compounds. Above all, we proposed that terpene compounds secreted from fennel root play a key role in Phytophthora disease suppression in this intercropping system.

The Bacillus velezensis GJ-7 strain isolated from the rhizosphere soil of Panax notoginseng showed high nematicidal activity and therefore has been considered a biological control agent that could act against the root-knot nematode Meloidogyne hapla. However, little was known about whether the GJ-7 strain could produce volatile organic compounds (VOCs) that were effective in biocontrol against M. hapla. In this study, we evaluated the nematicidal activity of VOCs produced by the fermentation of GJ-7 in three-compartment Petri dishes. The results revealed that the mortality rates of M. hapla J2s were 85% at 24 h and 97.1% at 48 h after treatment with the VOCs produced during GJ-7 fermentation. Subsequently, the VOCs produced by the GJ-7 strain were identified through solid-phase micro-extraction gas chromatography mass spectrometry (SPME-GC/MS). Six characteristic VOCs from the GJ-7 strain fermentation broth were identified, including 3-methyl-1-butanol, 3-methyl-2-pentanone, 5-methyl-2-hexanone, 2-heptanone, 2,5-dimethylpyrazine, and 6-methyl-2-heptanone. The in vitro experimental results from 24-well culture plates showed that the six volatiles had direct-contact nematicidal activity against M. hapla J2s and inhibition activity against egg hatching. In addition, 3-methyl-1-butanol and 2-heptanone showed significant fumigation effects on M. hapla J2s and eggs. Furthermore, all six of the VOCs repelled M. hapla J2 juveniles in 2% water agar Petri plates. The above data suggested that the VOCs of B. velezensis GJ-7 acted against M. hapla through multiple prevention and control modes (including direct-contact nematicidal activity, fumigant activity, and repellent activity), and therefore could be considered as potential biocontrol agents against root-knot nematodes.

Root-knot nematodes (RKNs) are soil-borne pathogens that severely affect Panax notoginseng growth and productivity. Thus, there is an urgent need for biological control agents or green nematicides to control root-knot nematodes. Rhizosphere bacteria can effectively control RKNs through different mechanisms. In this study, the three rhizosphere Bacillus strains, isolated from the root of P. notoginseng, were evaluated for the nematicidal activity and biological control efficacy against root-knot nematodes. In addition, we also evaluated the colonization ability of the two bacterial strains with significant biocontrol effect and dynamic regulation of genes related to systemic resistance in P. notoginseng. The rhizosphere Bacillus velezensis GJ-7 and Bacillus cereus NS-2 showed high nematicidal activity against Meloidogyne hapla in vitro and significantly reduced the number of root galls in three different control experiments. The results of colonization experiments showed that the strains GJ-7 and NS-2 colonized P. notoginseng root rapidly and stably. Additionally, the colonization of the strains NS-2 and GJ-7 activated the defense-responsive genes in P. notoginseng. These results indicated that the B. cereus strain NS-2 and B. velezensis strain GJ-7 have the potential for successful ecological niche occupation and enhance plant resistance and therefore could be considered as potential biocontrol agents against root-knot nematodes.

This study attempts to disclose the relationship between self-esteem, interpersonal trust, and social anxiety of college students and provide an empirical reference for enhancing their self-esteem, cultivating their interpersonal trust, and reducing their social anxiety. Specifically, 673 college students were randomly sampled and measured against the self-esteem scale (SES), interpersonal trust scale (TS), and interaction anxiousness scale (IAS). The results show that self-esteem does not vary significantly with genders and origins; social anxiety does not vary significantly with origins but differs significantly between genders; the social anxiety of males is much lower than that of females; interpersonal trust differs significantly between genders and between origins. ABSTRACT Management of crop root rot disease is one of the key factors in ensuring sustainable development in agricultural production. The accumulation of autotoxins and pathogens in soil has been reported as a primary driver of root rot diseases; however, less is known about the correlation of plants, their associated pathogens and microbiome mediated by autotoxins as well as the contributions autotoxins make to the occurrence of root rot disease. Here, we integrated metabolomic, transcriptomic, and rhizosphere microbiome analyses to identify the root cell wall degradants cellobiose and d-galacturonic acid as being induced by the autotoxic ginsenoside Rg1 of Panax notoginseng, and we found that exogenous cellobiose and d-galacturonic acid in addition to Rg1 could aggravate root rot disease by modifying the rhizosphere microbiome. Microorganisms that correlated positively with root rot disease were enriched and those that correlated negatively were suppressed by exogenous cellobiose, d-galacturonic acid, and Rg1. In particular, they promoted the growth and infection of the soilborne pathogen Ilyonectria destructans by upregulating pathogenicity-related genes. Cellobiose showed the highest ability to modify the microbiome and enhance pathogenicity, followed by Rg1 and then d-galacturonic acid. Collectively, autotoxins damaged root systems to release a series of cell wall degradants, some of which modified the rhizosphere microbiome so that the host plant became more susceptible to root rot disease. IMPORTANCE The accumulation of autotoxins and pathogens in soil has been reported as a primary driver of root rot disease and one of the key factors limiting sustainable development in agricultural production. However, less is known about the correlation of plants, their associated pathogens, and the microbiome mediated by autotoxins, as well as the contributions autotoxins make to the occurrence of root rot disease. In our study, we found that autotoxins can damage root systems, thus releasing a series of cell wall degradants, and both autotoxins and the cell wall degradants they induce could aggravate root rot disease by reassembling the rhizosphere microbiome, resulting in the enrichment of pathogens and microorganisms positively related to the disease but the suppression of beneficial microorganisms. Deciphering this mechanism among plants, their associated pathogens, and the microbiome mediated by autotoxins will advance our fundamental knowledge of and ability to degrade autotoxins or employ microbiome to alleviate root rot disease in agricultural systems.

The pyrolysis of biomass is an efficient means of utilizing biomass resources. Biomass can be converted into various high-performance chemicals and functional materials through pyrolysis. However, current pyrolysis technologies suffer from low conversion rates and single products, so the preparation of nitrogen compounds with high economic value remains a challenge. The walnut shell was soaked in three nitrogen-containing compound solutions before carbonization to produce high-value-added nitrogen-containing chemicals (with a nitrogen content of 59.09%) and biochar for the adsorption of polycyclic aromatic hydrocarbons (PAHs). According to biochar analysis, biochar has a porous structure with a specific surface area of 1161.30 m2/g and a high level of rocky desertification. The surface forms a dense pyrrole structure, and the structure produces π–π interactions with naphthalene molecules, exhibiting excellent naphthalene adsorption with a maximum capacity of 214.98 mg/g. This study provides an efficient, rapid, and environmentally friendly method for producing nitrogen-containing chemicals with high-added value and biochar.

Carbon dots (CDs), as a new type of photoluminescent nanomaterial, have attracted extensive attention in various fields because of their unique luminescence properties. However, CDs will exhibit fluorescence quenching in the solid state or aggregate state, which limits their application. In this paper, a unique strategy is proposed to regulate solutions to achieve multicolour fluorescence of CDs in the solid state. We report the successful preparation of orange, green and blue solid fluorescent CDs using citric acid, urea and phenylethylamine as precursors and methanol, ethanol and water as solvents, respectively. The solid-state fluorescence of CDs may be caused by the linkage of the phenylethyl structure to the surface of CDs during formation, which effectively disperses the CDs and prevents π–π interactions between graphitized nuclei. Meanwhile, multicolour solid fluorescent CDs are realized by adjusting the solvent in the preparation process. Based on the excellent fluorescence properties of CDs, orange, green and blue light-emitting diodes (LEDs) are prepared. A white LED (WLED) can be obtained by mixing the three colours of solid fluorescent CDs, which shows the application potential of CDs in display lighting equipment.

Biomass carbon dots (CDs) have the characteristics of being green, nontoxic, inexpensive, and simple to prepare, and they can be used in luminescence-related fields. In this study, red, green, and blue luminescent CDs were synthesised by a simple hydrothermal method under alkaline, neutral, and acidic conditions using TA as carbon source and o-phthalaldehyde as blend. The unique optical properties of these CDs are due to the differences in their degrees of conjugation, which can be controlled by the pH value of the reaction system. These three kinds of biomass CDs have good applications in light-emitting diodes (LEDs). By mixing biomass CDs with epoxy resin, warm, and cold white LEDs with Commission Internationale de l’Elcairage (CIE) coordinates (0.35, 0.36) were successfully constructed on extremely stable multicolour CDs. This study shows that these biomass CDs are a promising material for white LED lighting.

Carbon dots (CDs) are widely used nanomaterials that not only exhibit good biocompatibility and photostability, but also benefit from a simple preparation process and easy functionalization, making them promising for broad applications in the fields of heavy metal ion detection and optoelectronic devices. Based on the excellent optical properties of CDs and the current situation of increasing energy shortages, this paper selects the natural polyphenolic compound tannic acid (TA) found in biomass materials as the carbon source and innovatively adopts a simple and convenient solvent-free pyrolysis method without auxiliary reagents or solvents. The CDs with good water solubility and certain fluorescence properties were directly prepared under the condition of high temperature, and the obtained CDs exhibited blue fluorescence, and a high QY of 35.4% was obtained at 300 °C. The analysis and results demonstrate the selectivity of these CDs for the detection of various metal ion solutions. In particular, these CDs are sensitive to Ni2+ and can be used as fluorescent sensors for the efficient and sustainable detection of Ni2+, whereas previous sensors were often specific to Fe3+ and Hg2+. Thus, a new sensing technique has been developed for the detection of Ni2+ to achieve more sensitive and rapid detection.

Panax notoginseng is a medicinal plant in China, the flowers of which have high medicinal value. To study the differences in the floral fragrance compounds of P. notoginseng flowers (bionic wild cultivation) from the forests of Yunnan Province, the floral fragrance compounds from four varieties of P. notoginseng flowers (four-forked seven leaves, three-forked seven leaves, four-forked five–seven leaves, and three-forked five–six leaves) were compared and analyzed via headspace solid phase microextraction combined with gas chromatography–mass spectrometry methods. A total of 53 floral fragrance compounds from the P. notoginseng flowers were divided into eight categories, mainly consisting of terpenes, alkynes, aromatic hydrocarbons, and alcohols. Moreover, high contents of 3-carene, germacrene D, (−)-α-gurjunene, valencene, (+)-γ-gurjunene, menogene, and aromandendrene were identified from the flowers of different P. notoginseng varieties. Interestingly, floral fragrance compounds such as 3-carene, valencene, aromandendrene, menogene, and (+)-γ-gurjunene were first reported in the flowers of P. notoginseng. Cluster analysis showed that P. notoginseng with four-forked and three-forked leaves clustered into two subgroups, respectively. In addition, principal component analysis showed that (+)-γ-gurjunene, (+)-calarene, copaene, 1,8,12-bisabolatriene, γ-elemene, (–)-aristolene, caryophyllene, 3-carenes, and 2,6-dimethyl-1,3,6-heptatriene can be used to distinguish the floral fragrance components of four P. notoginseng flower species. This study provides a theoretical basis for elucidating the floral fragrance compounds emitted from the flowers of different P. notoginseng varieties in an agroforestry system.

Chemical control plays a crucial role in pest management but has to face challenges due to insect resistance. It is important to discover alternatives to traditional pesticides. The spider mite Tetranychus pueraricola (Ehara & Gotoh) (Acari: Tetranychidae) is a major agricultural pest that causes severe damage to many crops. GC16 is a new agent that consists of a mixture of Calcium chloride (CaCl2) and lecithin. To explore the acaricidal effects and mode of action of GC16 against T. pueraricola, bioassays, cryogenic scanning electron microscopy (cryo-SEM) and transmission electron microscopy (TEM) were performed. GC16 had lethal effects on the eggs, larvae, nymphs, and adults of T. pueraricola, caused the mites to dehydrate and inactivate, and inhibited the development of eggs. GC16 displayed contact toxicity rather than stomach toxicity through the synergistic effects of CaCl2 with lecithin. Cryo-SEM analysis revealed that GC16 damaged T. pueraricola by disordering the array of the cuticle layer crest. Mitochondrial abnormalities were detected by TEM in mites treated by GC16. Overall, GC16 had the controlling efficacy on T. pueraricola by cuticle penetration and mitochondria dysfunction and had no effects on Picromerus lewisi and Harmonia axyridis, indicating that GC16 is likely a new eco-friendly acaricide.

Panax notoginseng is an important functional health product, and has been used worldwide because of a wide range of pharmacological activities, of which the taproot is the main edible or medicinal part. However, the technologies for origin discrimination still need to be further studied. In this study, an ICP-MS/MS method for the accurate determination of 49 elements was established, whereby the instrumental detection limits (LODs) were between 0.0003 and 7.716 mg/kg, whereas the quantification limits (LOQs) were between 0.0011 and 25.7202 mg/kg, recovery of the method was in the range of 85.82% to 104.98%, and the relative standard deviations (RSDs) were lower than 10%. Based on the content of multi-element in P. notoginseng (total of 89 mixed samples), the discriminant models of origins and cultivation models were accurately determined by the neural networks (prediction accuracy was 0.9259 and area under ROC curve was 0.9750) and the support vector machine algorithm (both 1.0000), respectively. The discriminant models established in this study could be used to support transparency and traceability of supply chains of P. notoginseng and thus avoid the fraud of geographic identification.

Nitrogen-fixing bacteria play a significant role in tropical forest ecosystems. However, little is known about the comprehensive effects of altitude gradient (1000–2600 m), seasons (October, January, April, and July), and slope aspects (east and west) on the abundance and diversity of nitrogen-fixing bacteria in subtropical forest. Q-PCR and PCR-DGGE methods were performed to explore the abundance and diversity of nitrogen-fixing bacteria, respectively, in the Ailao Mountain subtropical forest. Our results showed that the abundance of nitrogen-fixing bacteria was highest in October and December, whereas it was lowest in April and July. Moreover, there was no difference in the total number of soil nitrogen-fixing bacteria on the eastern and western slopes. The diversity of soil nitrogen-fixing bacteria is higher at low and medium altitudes, but lower at high and medium altitudes with increasing altitude, and similar variation in the eastern and western slopes as well. Moreover, the most influential factors affecting the abundance of nitrogen-fixing bacteria was NH4+-N and herbal coverage, while those most affecting the diversity of nitrogen-fixing bacteria were NH4+-N and NO3−-N. In addition, permutational multivariate analysis demonstrated that the season had the greatest effects on the abundance of nitrogen-fixing, whereas altitude had the greatest effects on the diversity of nitrogen-fixing bacteria. These findings provide evidence that the variation in nitrogen-fixing bacteria is affected by multiple factors (altitudes, seasons and slope aspects) in the subtropical forests of Yunnan, China.

Bioorganic fertilizers can alleviate (a) biotic stresses and sustainably increase crop yields. The effect of bioorganic fertilizers on the rhizosphere bacterial community of Panax notoginseng and soil metabolism remains unknown. Here, we tracked the changes in the soil physicochemical properties, bacterial microbiota responses, and soil metabolic functions after the addition of a bioorganic fertilizer in a P. notoginseng field. The application of a bioorganic fertilizer reduced the soil acidification, improved the organic matter, and increased the contents of the total/available soil nutrients. Soil amendment with a bioorganic fertilizer significantly affected the structure of the rhizosphere bacterial community, leading to the enrichment of specific bacterial consortia such as Rhodanobacter, Arthrobacter, Sphingomonas, Devosia, Pseudolabrys, Luteimonas, Lysobacter, Nitrosospira, and Nakamurella. Previously, many of these genera have been associated with nutrient cycling, plant productivity, and disease suppression. Metabolome analysis further highlighted that the bioorganic fertilizer treatment significantly reduced phenolic acids and flavonoids and enhanced organic acids, saccharides and alcohols, and amino acids. This result indicates a high survival of bacterial microbiota in the rhizosphere and an availability of nutrients for P. notoginseng growth. This work showed that the application of bioorganic fertilizers significantly improves soil health status, alters soil metabolic functions, and stimulates a specific subset of rhizosphere microbiota for nutrient cycling and disease protection in P. notoginseng.
