The neural mechanisms associated with the limited capacity of working memory (WM) has long been studied, but it is still unclear which neural regions are associated with the precision of visual WM. Here, an orientation recall task for estimating the trial-wise precision of visual WM was performed and then repeated two weeks later in an fMRI scanner. Results showed that activity in frontal and parietal regions during WM maintenance scaled with WM load, but not with the precision of WM (i.e., recall error in radians). Conversely, activity in the lateral occipital complex (LOC) during WM maintenance was not affected by memory load, but rather, correlated with WM precision on a trial-by-trial basis. Moreover, activity in LOC also correlated with the individual participant's precision of WM from a separate behavioral experiment. Interestingly, a region within the prefrontal cortex, the inferior frontal junction (IFJ), exhibited greater functional connectivity with LOC when the WM load increased. Together, our findings provide unique evidence that the LOC supports visual WM precision, while communication between the IFJ and LOC varies based on WM load demands. These results suggest an intriguing possibility that distinct neural mechanisms may be associated with general content (load) or detailed information (precision) of WM., (Copyright © 2019 The Author(s). Working memory (WM) refers to the process of temporally maintaining and manipulating input information. WM is the global workspace of cognitive functions, however, with severely restricted capacity and precision. Previous cognitive and computational models discussed the methods of calculating capacity and precision of WM and the reason why they are so limited. It still remains debated which model is the best across all datasets, and whether there exists upper limits of items. Besides, sensory cortices and the frontal-parietal loop are suggested to represent WM memorandum. Yet recently, the sensory recruitment hypothesis that posits an important role of sensory cortices in WM is strongly argued. Meanwhile, whether the prefrontal cortex shows sustained activity or bursting γ oscillations is intensely debated as well. In the future, disentangling the contribution to WM of feedforward γ vs feedback α/β oscillations, and/or dopamine vs serotonin systems, is critical for understanding the neural mechanisms underlying WM. It will further do help to recognize the basis for the psychiatric (e.g. schizophrenia) or neurological (e.g. Alzheimer's disease) disorders, and potentially to develop effective training and intervening methods.

In the present study, we searched for causal evidence linking activity in the bilateral primary somatosensory cortex (SI), posterior parietal cortex (PPC), and prefrontal cortex (PFC) with behavioral performance in vibrotactile working memory. Participants performed a vibrotactile delayed matching-to-sample task, while single-pulse transcranial magnetic stimulation (sp-TMS) was applied over these cortical areas at 100, 200, 300, 600, 1600, and 1900 ms after the onset of vibrotactile stimulation (200 ms duration). In our experiments, sp-TMS over the contralateral SI at the early delay (100 and 200 ms) deteriorated the accuracy of task performance, and over the ipsilateral SI at the late delay (1600 and 1900 ms) also induced such deteriorating effects. Furthermore, deteriorating effects caused by sp-TMS over the contralateral DLPFC at the same maintenance stage (1600 ms) were correlated with the effects caused by sp-TMS over the ipsilateral SI, indicating that information retained in the ipsilateral SI during the late delay may be associated with the DLPFC. Taken together, these results suggest that both the contralateral and ipsilateral SIs are involved in tactile WM, and the contralateral DLPFC bridges the contralateral SI and ipsilateral SI for goal-directed action.

Neural activity in the dorsolateral prefrontal cortex (DLPFC) has been suggested to integrate information from distinct sensory areas. However, how the DLPFC interacts with the bilateral primary somatosensory cortices (SIs) in tactile-visual cross-modal working memory has not yet been established. In the present study, we applied single-pulse transcranial magnetic stimulation (sp-TMS) over the contralateral DLPFC and bilateral SIs of human participants at various time points, while they performed a tactile-visual delayed matching-to-sample task with a 2-second delay. sp-TMS over the contralateral DLPFC or the contralateral SI at either an sensory encoding stage [i.e. 100 ms after the onset of a vibrotactile sample stimulus (200-ms duration)] or an early maintenance stage (i.e. 300 ms after the onset), significantly impaired the accuracy of task performance; sp-TMS over the contralateral DLPFC or the ipsilateral SI at a late maintenance stage (1600 ms and 1900 ms) also significantly disrupted the performance. Furthermore, at 300 ms after the onset of the vibrotactile sample stimulus, there was a significant correlation between the deteriorating effects of sp-TMS over the contralateral SI and the contralateral DLPFC. These results imply that the DLPFC and the bilateral SIs play causal roles at distinctive stages during cross-modal working memory, while the contralateral DLPFC communicates with the contralateral SI in the early delay, and cooperates with the ipsilateral SI in the late delay., (Copyright © 2018 Elsevier B.V. The activity in sensory cortices and the prefrontal cortex (PFC) throughout the delay interval of working memory (WM) tasks reflect two aspects of WM-quality and quantity, respectively. The delay activity in sensory cortices is fine-tuned to sensory information and forms the neural basis of the precision of WM storage, while the delay activity in the PFC appears to represent behavioral goals and filters out irrelevant distractions, forming the neural basis of the quantity of task-relevant information in WM. The PFC and sensory cortices interact through different frequency bands of neuronal oscillation (theta, alpha, and gamma) to fulfill goal-directed behaviors.

Our previous studies on scalp-recorded event-related potentials (ERPs) showed that somatosensory N140 evoked by a tactile vibration in working memory tasks was enhanced when human subjects expected a coming visual stimulus that had been paired with the tactile stimulus. The results suggested that such enhancement represented the cortical activities involved in tactile-visual crossmodal association. In the present study, we further hypothesized that the enhancement represented the neural activities in somatosensory and frontal cortices in the crossmodal association. By applying independent component analysis (ICA) to the ERP data, we found independent components (ICs) located in the medial prefrontal cortex (around the anterior cingulate cortex, ACC) and the primary somatosensory cortex (SI). The activity represented by the IC in SI cortex showed enhancement in expectation of the visual stimulus. Such differential activity thus suggested the participation of SI cortex in the task-related crossmodal association. Further, the coherence analysis and the Granger causality spectral analysis of the ICs showed that SI cortex appeared to cooperate with ACC in attention and perception of the tactile stimulus in crossmodal association. The results of our study support with new evidence an important idea in cortical neurophysiology: higher cognitive operations develop from the modality-specific sensory cortices (in the present study, SI cortex) that are involved in sensation and perception of various stimuli.

Background: Repetitive transcranial magnetic stimulation has been employed to treat drug dependence, reduce drug use and improve cognition. The aim of the study was to analyze the effectiveness of intermittent theta-burst stimulation (iTBS) on cognition in individuals with methamphetamine use disorder (MUD). Methods: This was a secondary analysis of 40 MUD subjects receiving left dorsolateral prefrontal cortex (L-DLPFC) iTBS or sham iTBS for 20 times over 10 days (twice-daily). Changes in working memory (WM) accuracy, reaction time, and sensitivity index were analyzed before and after active and sham rTMS treatment. Resting-state EEG was also acquired to identify potential biological changes that may relate to any cognitive improvement. Results: The results showed that iTBS increased WM accuracy and discrimination ability, and improved reaction time relative to sham iTBS. iTBS also reduced resting-state delta power over the left prefrontal region. This reduction in resting-state delta power correlated with the changes in WM. Conclusions: Prefrontal iTBS may enhance WM performance in MUD subjects. iTBS induced resting EEG changes raising the possibility that such findings may represent a biological target of iTBS treatment response. The present study aimed to explore the effect of intermittent theta-burst stimulation (iTBS) on visual working memory for people suffering from methamphetamine use disorder (MUD). Five sessions of iTBS were carried over the left dorsolateral prefrontal cortex (DLPFC) or the vertex as a sham control, with each session in one day. Orientation free-recall tasks were conducted before the iTBS stimulation, after the first and fifth sessions of stimulation. Results showed that when compared with the sham group, a single session of iTBS over the left DLPFC improved participants' working memory performance. Specifically, iTBS over the left DLPFC increased the working memory capacity and such effects enlarged with multiple sessions. The present finding suggested that iTBS over DLPFC could be a promising intervention method to enhance the cognitive function of addicts with MUD. [ABSTRACT FROM AUTHOR]

Attention can be deployed among external sensory stimuli or internal working memory (WM) representations, and recent primate studies have revealed that these external and internal selections share a common neural basis in the prefrontal cortex (PFC). However, it remains to be elucidated how PFC implements these selections, especially in humans. The present study aimed to further investigate whether PFC responded differentially to the peripheral and central retrospective cues (retro-cues) that induced attention selection among WM representations. To achieve this, we combined magnetoencephalography (MEG, Experiment 1) and transcranial magnetic stimulation (TMS, Experiment 2) with an orientation-recall paradigm. Experiment 1 found that a peripheral retro-cue with 100% reliability had a greater benefit on WM performance than a central retro-cue, while this advantage of peripheral over central cues vanished when the cue reliability dropped to 50% (non-informative). MEG source analysis indicated that the 100% peripheral retro-cue elicited earlier (∼125 ms) PFC responses than the central retro-cue (∼275 ms). Meanwhile, Granger causality analysis showed that PFC had earlier (0–200 ms) top-down signals projecting to the superior parietal lobule (SPL) and the lateral occipital cortex (LOC) after the onset of peripheral retro-cues, while these top-down signals appeared later (300–500 ms) after the onset of central retro-cues. Importantly, PFC activity within this period of 300–500 ms correlated with the peripheral advantage in behavior. Moreover, Experiment 2 applied TMS at different time points to test the causal influence of brain activity on behavior and found that stimulating PFC at 100 ms abolished the behavioral benefit of the peripheral retro-cue, as well as its advantage over the central retro-cue. Taken together, our results suggested that the advantage of peripheral over central retro-cues in the mnemonic domain is realized through faster top-down control from PFC, which challenged traditional opinions that the top-down control of attention on WM required at least 300 ms to appear.
