Enzymes derived from microbial sources have gained increasing popularity in industrial applications over the past decades. Despite the high production cost, alkaline proteases have wide applications in industries such as tanneries, food production, and detergents. In recent years, there has been a shift towards utilizing natural carbon sources for cultivating microorganisms and extracting proteases in order to reduce production costs. This study aimed to investigate the biochemical and kinetic properties of protease enzymes obtained from Aspergillus niger cultivated in a paper waste medium and compare with the enzyme produced in a basal medium. Glucose is a more favorable carbon source compared to cellulose, so paper waste was pretreated with cellulose-degrading bacteria to convert cellulose into smaller carbohydrates. After the growth of A. niger in basal and combinational media, the enzymatic properties were compared between the extracted enzymes by using casein as substrate. The results demonstrated that A. niger could produce protease enzymes in the paper waste medium similar to the basal medium with more than 5-fold cost saving. The specific activity of the enzymes isolated from the basal and paper waste media was calculated to be 184.95 ± 10.56 U ml -1 and 169.88 ± 11.05 U ml -1 , respectively. Carbon sources did not affect the optimum pH and temperature of the protease enzyme, which were found to be 8 and 37 °C, respectively. Biofilms are composed of complex multi-species in nature, potentially threatening drinking water safety. In this work, the formation of single- and multi-species fungal biofilms formed by Aspergillus niger (A. niger) and Aspergillus flavus (A. flavus), and the inactivation of mature biofilms using chlor(am)ine were firstly investigated. Results revealed that the antagonistic interaction occurred between A. niger and A. flavus. Chloramination at 20 mg/L for 30 min achieved 74.74 % and 76.04 % inactivation of A. flavus and multi-species biofilm, which were 1.69- and 1.84-fold higher than that of chlorine at the same condition. However, no significant difference was observed in the inactivation of A. niger biofilm between chlorine and monochloramine disinfection due to the lower amount of extracellular polymeric substance produced by it (p > 0.05). The inactivation of biofilm by monochloramine fitted the Weibull model well. According to the Weibull model, the monochloramine resistance of biofilm were as follows: A. flavus > multi-species > A. niger biofilm. Besides, an increase in reactive oxygen levels, damage of cell membrane, and leakage of intracellular substances in biofilms were observed after chlor(am)ination. More intracellular polysaccharides and proteins were leaked in chloramination inactivation (p < 0.05). This study explores the enhancement of phosphate rock (PR) solubilization through solid-state fermentation (SSF) by optimizing oxalic acid production using Aspergillus niger. Key process parameters, including the use of agro-industrial by-products (sugarcane bagasse (SCB), wheat bran (WB), soybean bran (SB)), pH levels, sucrose supplementation, and methanol addition, were systematically evaluated through sequential experimental designs. The results identified SCB and SB in a 1:1 ratio as the most effective substrate. Remarkably, the inclusion of methanol (7 %) and sucrose (0.5 %) resulted in a 3-fold increase in oxalic acid production. Under these optimized conditions, significant phosphorus solubilization of Bayóvar, Itafós, and Registro PRs was achieved, with Bayóvar rock releasing up to 12.1 g/kgds of soluble P (63.8 % efficiency). Additionally, the SSF process effectively released organic phosphorus from the agro-industrial substrates. In this study, the family GH10 xylanase AnXylA10 derived from Aspergillus niger JL15 strain was expressed in Pichia pastoris X33. The recombinant xylanase, reAnXylA10 exhibited optimal activity at 40 ℃ and pH 5.0. The hydrolysates generated from beechwood xylan using reAnXylA10 primarily consisted of xylobiose (X2) to xylohexaose (X6) and demonstrated remarkable antioxidant capacity. Furthermore, the rice xylanase inhibitory protein (riceXIP) was observed to competitively inhibit reAnXylA10, exhibiting an inhibition constant (K i ) of 140.6 nM. Molecular dynamics (MD) simulations of AnXylA10-riceXIP complex revealed that the α-7 helix (Q225-S238) of riceXIP intruded into the catalytic pocket of AnXylA10, thereby obstructing substrate access to the active site. Specifically, residue K226 of riceXIP formed robust interactions with E136 and E242, the two catalytic sites of AnXylA10, predominantly through high-occupied hydrogen bonds. Based on QTAIM, electron densities for the atom pairs K226 riceXIP @HZ1-E136 AnXylA10 @OE2 and K226 riceXIP @HZ3-E242 AnXylA10 @OE1 were determined to be 0.04628 and 0.02914 a.u., respectively. Binding free energy of AnXylA10-riceXIP complex was -59.0±7.6 kcal/mol, significantly driven by electrostatic and van der Waals forces. Despite their acidic pH, carbonated beverages can be contaminated by spoilage microorganisms. Thermal treatments, before and/or after carbonation, are usually applied to prevent the growth of these microorganisms. However, the impact of CO 2 on the heat resistance of spoilage microorganisms has never been studied. A better understanding of the combined impact of CO 2 and pH on the heat resistance of spoilage microorganisms commonly found in carbonated beverages might allow to optimize thermal treatment. Five microorganisms were selected for this study: Alicyclobacillus acidoterrestris (spores), Aspergillus niger (spores), Byssochlamys fulva (spores), Saccharomyces cerevisiae (vegetative cells), and Zygosaccharomyces parabailii (vegetative cells). A method was developed to assess the impact of heat treatments in carbonated media on microbial resistance. The heat resistances of the five studied species are coherent with the literature, when data were available. However, neither the dissolved CO 2 concentration (from 0 to 7 g/L), nor the pH (from 2.8 to 4.1) have an impact on the heat resistance of the selected microorganisms, except for As. niger, for which the presence of dissolved CO 2 reduced the heat resistance. Aspergillus niger (A. niger) spores can induce numerous health problems. Once the airflow-imposed drag force on an A. niger spore exceeds its binding force with the colony, the spore is detached. Turbulent flow may considerably increase the spore detachment. No method is currently available for prediction of the drag force on a spore and its detachment in turbulent flows. This investigation measured the turbulent velocities and detachment of A. niger colonies in a wind tunnel. Computational fluid dynamics (CFD) was employed to model an A. niger unit subjected to turbulent flow blowing. The top 1 % quantile instantaneous velocity of the turbulent flow was specified as the steady entry flow boundary condition for solving the peak velocity distribution and the peak drag forces onto spores. The predicted spore detachment ratios were compared with the measurement data for model validation. The results revealed that the spore detachment ratios with a turbulence intensity of 17 % to 20 % can be twice to triple the ratio with a turbulence intensity of approximately 1 %, when the average velocity for blowing remains the same. The proposed CFD model can accurately predict the detachment ratios of the A. niger spores. ENVIRONMENTAL IMPLICATION: Some people are sensitive to the Aspergillus niger (A. niger) spores, and excessive exposure can cause nasal congestion, skin tingling, coughing, and even asthma. Turbulent flow can considerably increase the spore detachment, due to the increased airflow-imposed drag force on the spores during turbulence. This investigation developed a numerical model to solve for the peak velocity distribution and the peak drag forces onto spores in turbulent flows to predict the spore detachment. Nano-biotechnology is quickly developing as an important field of modern research, generating the most promising applications in medicine and agriculture. Biosynthesis of silver nanoparticles using biogenic or green approach provide ecofriendly, clean and effective way out for the synthesis of nanoparticles. The main aim of the study was to synthesize silver nanoparticles (AgNPs) from Aspergillus niger, Aspergillus flavus and Pencillium chrysogenum using a green approach and to test the antifungal activity of these synthesized AgNPs against a variety of pathogenic fungi. The characterization of samples was done by using UV-visible spectroscopy, SEM (scanning electron microscopy), FTIR (Fourier transmission infrared spectroscopy), and XRD (X-ray diffractometry). The investigation confirmed the creation of AgNPs by the fungi Aspergillus niger, Aspergillus flavus and Pencillium chrysogenum, as evidenced by prominent plasmon absorbance bands at 420 and 450 nm.The biosynthesized AgNPs were 80-100 nm in size, asymmetrical in shape and became spherical to sub-spherical when aggregated. Agar well diffusion method was performed to evaluate the antifungal activity of AgNPs against various plant pathogenic fungi. An efficient and strong antifungal activity was shown by these biosynthesized nanoparticles against serious plant pathogenic fungi, viz. Aspergillus terreus, Fusarium oxysporum, Penicillium citrinum, Rhizopus stolonifer and Mucor mucedo. The biosynthesized AgNPs at various concentrations caused significant zone of inhibition in the test fungal pathogens. Silver nanoparticles (AgNPs) biosynthesized from Aspergillus niger at highest concentrations showed maximum zone of inhibition against Penicillium citrinum (19.33 ± 0.57 mm) followed by Rhizopus stolonifer (17.66 ± 0.57), Aspergillus terreus (16.33 ± 1.54 mm), Fusarium oxysporum (14.00 ± 1.00 mm) and Mucor mucedo (13.33 ± 1.15 mm) respectively. In recent years, with the booming of the edible mushroom industry, chitin production has become increasingly dependent on fungi and other non-traditional sources. Fungal chitin has advantages including superior performance, simpler separation processes, abundant raw materials, and the absence of shellfish allergens. As a kind of edible mushroom, flammulina velutipes (F. velutipes) also has the advantages of wide source and large annual yield. This provided the possibility for the extraction of chitin. Here, a procedure to extract chitin from F. velutipes waste be presented. This method comprises low-concentration acid pretreatment coupled with consolidated bioprocessing with Aspergillus niger. Characterization by SEM, FTIR, XRD, NMR, and TGA confirmed that the extracted chitin was β-chitin. To achieve optimal fermentation of F. velutipes waste (80 g/L), ammonium sulfate and glucose were selected as nitrogen and carbon sources (5 g/L), with a fermentation time of 5 days. The extracted chitin could be further deacetylated and purified to obtain high-purity chitosan (99.2 % ± 1.07 %). This chitosan exhibited a wide degree of deacetylation (50.0 % ± 1.33 % - 92.1 % ± 0.97 %) and a molecular weight distribution of 92-192 kDa. Disposal and recycling of heavy metal-enriched biomass is the key to measure the success of phytoremediation. This study employed innovative approach to use Aspergillus niger (A. niger) for the treatment of Cd-contaminated Helianthus annuus L. (sunflower) stalk after phytoremediation. Single-factor results showed that the removal of Cd at an initial pH of 3 was superior to sucrose and inoculation amount. 67.67% of Cd was removed by A. niger leaching system after 11 days based on response surface methodology optimum conditions (sucrose: 76.266 g L -1 ; inoculation amount: 10%; initial pH: 3), while the concentrations of nitrogen, phosphorus and potassium (N, P and K) of sunflower stalk were unaffected. While physicochemical pretreatment effectively enhanced the bioleaching efficiency, it also resulted in significant loss of P and K elements, thereby reducing the value of biomass for recycling and utilization. Cinnamaldehyde displays strong antifungal activity against fungi such as Aspergillus niger, but its precise molecular mechanisms of antifungal action remain inadequately understood. In this investigation, we applied chemoproteomics and bioinformatic analysis to unveil the target proteins of cinnamaldehyde in Aspergillus niger cells. Additionally, our study encompassed the examination of cinnamaldehyde's effects on cell membranes, mitochondrial malate dehydrogenase activity, and intracellular ATP levels in Aspergillus niger cells. Our findings suggest that malate dehydrogenase could potentially serve as an inhibitory target of cinnamaldehyde in Aspergillus niger cells. By disrupting the activity of malate dehydrogenase, cinnamaldehyde interferes with the mitochondrial tricarboxylic acid (TCA) cycle, leading to a significant decrease in intracellular ATP levels. Following treatment with cinnamaldehyde at a concentration of 1 MIC, the inhibition rate of MDH activity was 74.90 %, accompanied by an 84.5 % decrease in intracellular ATP content. Furthermore, cinnamaldehyde disrupts cell membrane integrity, resulting in the release of cellular contents and subsequent cell demise. Inactivation of flbA in Aspergillus niger results in thinner cell walls, increased cell lysis, abolished sporulation, and an increased secretome complexity. A total of 36 transcription factor (TF) genes are differentially expressed in ΔflbA. Here, seven of these genes (abaA, aslA, aslB, azf1, htfA, nosA, and srbA) were inactivated. Inactivation of each of these genes affected sporulation and, with the exception of abaA, cell wall integrity and protein secretion. The impact on secretion was strongest in the case of ΔaslA and ΔaslB that showed increased pepsin, cellulase, and amylase activity. Biomass was reduced of agar cultures of ΔabaA, ΔaslA, ΔnosA, and ΔsrbA, while biomass was higher in liquid shaken cultures of ΔaslA and ΔaslB. The ΔaslA and ΔhtfA strains showed increased resistance to H 2 O 2 , while ΔaslB was more sensitive to this reactive oxygen species. Developing efficient microbiological methods to convert polysaccharide-rich materials into fermentable sugars, particularly monosaccharides, is vital for advancing the bioeconomy and producing renewable chemicals and energy sources. This study focused on optimizing the production conditions of an enzyme cocktail from Aspergillus niger ATCC 9642 using solid-state fermentation (SSF) and assessing its effectiveness in saccharifying mango peels through a simple, rapid, and efficient one-step process. A rotatable central composite design was employed to determine optimal conditions of moisture, time, and pH for enzyme production in SSF medium. The optimized enzyme cocktail exhibited cellulase activity (CMCase) at 6.28 U/g, filter paper activity (FPase) at 3.29 U/g, and pectinase activity at 117.02 U/g. These optimal activities were achieved with an SSF duration of 81 h, pH of 4.66, and a moisture content of 59%. The optimized enzyme cocktail effectively saccharified the mango peels without the need for chemical agents. The maximum saccharification yield reached approximately 81%, indicating efficient conversion of mango peels into sugars. The enzyme cocktail displayed consistent thermal stability within the tested temperature range of 30-60°C. Notably, the highest sugar release occurred within 36 h, with glucose, arabinose, galactose, and xylose being the primary monosaccharides released during saccharification. This study highlights the potential application of Aspergillus niger ATCC 9642 and SSF for enzymatic production, offering a simple and high-performance process for monosaccharide production. Prolyl endopeptidase from Aspergillus niger (An-PEP) is an enzyme that recognizes C-terminal peptide bonds of amino acid chains and cleaves them by hydrolysis. An aqueous two-phase system (ATPS) was used to separate An-PEP from fermentation broth. Through single factor experiments, the ATPS containing 16 % (w/w) PEG2000 and 15 % (w/w) (NH 4 ) 2 SO 4 at pH 6.0 obtained the recovery of 79.74 ± 0.16 % and the purification coefficient of 7.64 ± 0.08. It was then used to produce soy protein isolate peptide (SPIP) by hydrolysis of soy protein isolate (SPI), and SPIP-Ferrous chelate (SPIP-Fe) was prepared with SPIP and Fe 2+ . The chelation conditions were optimized by RSM, as the chelation time was 30 min, chelation temperature was 25 °C, SPIP mass to VC mass was two to one and pH was 6.0. The obtained chelation rate was 82.56 ± 2.30 %. The change in the structures and functional features of SPIP before and after chelation were investigated. The FTIR and UV-Vis results indicated that the chelation of Fe 2+ and SPIP depended mainly on the formation of amide bonds. The fluorescence, SEM and amino acid composition analysis results indicated that Fe 2+ could induce and stabilize the surface conformation and change the amino acid distribution on the surfaces of SPIP. The chelation of SPIP and Fe 2+ resulted in the enhancement of radical scavenging activities and ACE inhibitory activities. This study investigates the effects of SCG embedded into biodegradable polymer blends and aimed to formulate and characterise biomass-reinforced biocomposites using spent coffee ground (SCG) as reinforcement in PHB/PLA polymer blend. The effect of SCG filler loading and varying PHB/PLA ratios on the tensile properties and morphological characteristics of the biocomposites were examined. The results indicated that tensile properties reduction could be due to its incompatibility with the PHB/PLA matrixSCG aggregation at 40 wt% content resulted in higher void formation compared to lower content at 10 wt%. A PHB/PLA ratio of 50/50 with SCG loading 20 wt% was chosen for biocomposites with treated SCG. Biological treatment of SCG using Phanerochaete chrysosporium CK01 and Aspergillus niger DWA8 indicated P. chrysosporium CK01 necessitated a higher moisture content for optimum growth and enzyme production, whereas the optimal conditions for enzyme production (50-55 %, w/w) differed from those promoting A. niger DWA8 growth (40 %, w/w). SEM micrographs highlighted uniform distribution and effective wetting of treated SCG, resulting in improvements of tensile strength and modulus of biocomposites, respectively. Detachment of fungal spores from growing colonies results in human exposure. Thus far, the distribution of the binding forces of the spores in a fungal unit is unknown, so that precise prediction of the spores detachment is quite challenging. This investigation used centrifugal separation to measure the binding forces of the spores. Aspergillus niger (A. niger) colonies on a culture plate were placed in a centrifuge, the detached spores were counted, and this number was used to obtain the distribution of binding forces. Next, the air-blowing of an A. niger unit was modeled by computational fluid dynamics (CFD). A spore was judged to be detached if the air-imposed drag force was greater than the binding force. For model validation, the predicted spore detachment ratios were compared with the ratios measured in a wind tunnel test. The results revealed that the binding forces of the spores obeyed the log-normal distribution. The binding forces of the distal spores from colonies with a growth age of 66 h ranged from 0 nN to 4.0 nN and had a mean of 0.65 nN. Excessive sodium significantly inhibits citric acid fermentation by Aspergillus niger during the recycling of citric acid wastewater. This study aimed to elucidate the inhibition mechanism at the interface of physiology and transcriptomics. The results showed that excessive sodium caused a 22.3 % increase in oxalic acid secretion and a 147.6 % increase in H + -ATPase activity at the 4 h fermentation compared to the control. Meanwhile, a 13.1 % reduction in energy charge level and a 15.2 % decline in NADH content were found, which implied the effects on carbon metabolism and redox balance. In addition, transcriptomic analysis revealed that excessive sodium altered the gene expression profiles related to ATPase, hydrolase, and oxidoreductase, as well as pathways like glyoxylate metabolism, and transmembrane transport. Aspergilli can be used to produce food but can spoil it as well. Both food production and spoilage are initiated by germination of the conidia of these fungi that have been introduced by inoculation and contamination, respectively. Germination of these spores includes activation, swelling, establishment of cell polarity, and formation of a germ tube. So far, only quantitative single-species germination studies of fungal spores have been performed. Here, spore germination of the food spoilage fungus Aspergillus niger was studied quantitatively in mono-culture or when mixed with other food-relevant aspergilli (Aspergillus nidulans, Aspergillus terreus, Aspergillus clavatus, and Aspergillus oryzae). In the presence of the germination inducing amino acids proline or alanine, but not in the case of the lowly inducing amino acid arginine, the incidence of swelling and germ tube formation was reduced when 35,000 extra conidia of Aspergillus niger were added to wells containing 5000 of these spores. Adding 35,000 spores of one of the other aspergilli also did not have an effect on germination in the presence of arginine, but the germination inhibition was stronger when compared to the extra A. niger spores in the case of alanine. A similar effect was obtained with proline. Together, results show that the germination of A. niger conidia is impacted by the density of its own spores and that of other aspergilli under favorable nutritional conditions. Environmental hormones have attracted more attention because of their adverse impact on the health and ecological security of human. Biodegradation is still an efficient tactics to remove environmental hormones, but human intestinal microbes remain to be elucidated in the role of their degradation. In the present work, we intended to perform the in vitro experiment for investigating the degradation of 17β-estradiol, the main environmental estrogen, by human intestinal microflora Aspergillus niger RG13B1. Its degradation led to the production of eighteen metabolites characterized by 1 H, 13 C, and 2D NMR, and HRMS spectra, including nine new (1-9) and nine known metabolites (10-18). Based on their structures, the degradation pathway of 17β-estradiol mediated by A. niger RG13B1 involved hydroxylation, oxidation, methylation, acetylation, and dehydrogenation, especially infrequent lactylation, and the key degradation enzymes were found in the gene cluster of A. niger. In addition, we found that metabolite 12 interacted with amino acid residues Lys37, Gln39, Lys93, and Asn115 of NF-κB p65 to suppress expressions of inflammatory genes or proteins, exerting its anti-inflammatory effect. In this paper, for the first time, photopolymers were synthesized from glycerol acrylates with different numbers of functional groups, 2-hydroxy-3-phenoxypropyl acrylate, glycerol dimethacrylate or glycerol trimethacrylate, without and with the addition of vanillin styrene. The photocuring kinetics were monitored by real-time photorheometry. The mechanical, rheological, thermal, antimicrobial and shape-memory properties of the photopolymers were investigated. All polymers synthesized demonstrated antibacterial activity against Escherichia coli and Staphylococcus aureus, as well as antifungal activity against Aspergillus flavus and Aspergillus niger. 2-Hydroxy-3-phenoxypropyl acrylate-based polymers showed thermoresponsive shape-memory behavior. They were able to maintain their temporary shape below the glass transition temperature and return to their permanent shape above the glass transition temperature. The paper deals with the possible use of fungi to decontaminate polluted waters. Specifically, the focus is the selection of the most promising fungal strain capable of bioaccumulating arsenic, which is a globally widespread environmental contaminant. To this aim, allochthonous fungal strains from the ColD UNIGE JRU MIRRI strains collection were selected. Their capability to survive and accumulate this kind of pollutant was evaluated and compared with that of an autochthonous fungi set directly isolated from the arsenic polluted water. A preliminary screening at various concentrations of arsenic (0, 200, 400, 800, 1600 μg L−1) revealed that the best performing strains were Aspergillus niger and Penicillium expansum among the autochthonous strains and Aspergillus niger and Penicillium expansum among the allochthonous strains. Moreover, all the strains were subjected to bioaccumulation tests at a 1600 μg L−1 concentration. Local and allochthonous fungal strains showed different behaviors: the allochthonous strains grew rapidly and in a sustained way but without expressing any bioaccumulation activity. On the contrary, the indigenous fungi, despite a moderate growth, showed a good bioaccumulation capacity (in particular, Aspergillus niger). Cellulose nanocrystals are an important class of bio‐based crystalline nanostructures, finding application in several technological fields, including paper and textile coating, biocomposite engineering, biocatalysts immobilization, etc. This study explores enzymatic hydrolysis of Avicel, using endoglucanase from Aspergillus niger, to find an environmentally friendly method to extract cellulose nanocrystals from cellulose sources. Enzymatic hydrolysis has the advantage of reduced energy consumption and higher environmental friendliness compared to acid hydrolysis. In this work, we report for the first time very high nanocrystals yield by combining mechanical pretreatment of the cellulose starting material with a ball miller and endoglucanase hydrolysis, as a result of an extensive optimization of reaction conditions. In particular, a ball milling pretreatment carried out for 50 minutes at 3 Hz, allowed to isolate enzymatic CNCs with 76 % yield and with crystallinity as high as 75 %. The materials were characterized by X‐Ray diffractometry, attenuated total reflectance Fourier transform infrared (ATR‐FTIR) spectroscopy, dynamic light scattering, zeta potential and field emission scanning electron microscopy (FE‐SEM). Their characteristics were compared with the properties of sulfated CNCs, prepared from Avicel by sulfuric acid hydrolysis. This study examined the fermentation duration in relation to the effectiveness of tannin levels in mangrove (Sonneratia alba) fruit during the fermentation process using Aspergillus niger. The tested durations were 7, 10, 13, and 16 days in anaerobic conditions. The outcomes measured included crude fat, crude fibre, and nitrogenfree extract digestibility. Microbial biomass, protozoa population, and methane gas production were also recorded as responses to the rumen microbial activity. The results showed that 16 days of fermentation gave the highest average for all responses. The protozoa population reached around 4.07 × 105 cells/ml and methane gas amounted to 33.9 ml/g of dry matter (DM). This is caused by a decrease in the anti-nutrient tannin content in mangrove (S. alba) fruit due to fermentation by A. niger according to treatment. The conclusion of this research is that the 16-day fermentation treatment (T4) of mangrove fruit is the optimal time to be used as a source of concentrate feed for livestock in terms of the increase in nutritional value and gastrointestinal microbes represented by total protozoa population, crude fibre digestibility, crude fat digestibility, and the highest nitrogen-free extract digestibility. Glucose oxidase (GOx) catalyzes the oxidation of D -glucose to D -glucono-1,5-lactone and has a wide range of applications in various industries. However, the strict substrate specificity of GOx hampers its application in the conversion of other abundant sugars such as D -xylose. In this study, the substrate preference of GOx from Aspergillus niger (AnGOx) was engineered using a semi-rational design approach. The mutant T110V/F414L exhibited a 5.7-fold increase in D -xylose oxidation activity compared to that of the wild-type enzyme, which was attributed to its enhanced affinity for the substrate. Molecular dynamics simulations indicated that the T110V and F414L mutations may mitigate the non-productive binding of D -xylose at the entrance of the substrate-binding pocket, and therefore, are beneficial for providing access of its C1 hydroxyl group to the catalytic residues. Moreover, the mutant simultaneously oxidized D -xylose and D -glucose in the corncob hydrolysate to the corresponding aldonic acids when coupled with catalase. These findings provide new insights into substrate recognition by GOx and offer a new method for the utilization of D -xylose from lignocellulosic feedstocks. [ABSTRACT FROM AUTHOR]

Currently, heavy metal pollution is a challenge that needs to be solved urgently, and there is a broad prospect for food waste(FW) utilization. In this paper, activated hydrochar (AHC) made from FW according to actual proportion was produced by hydrothermal carbonization (HTC) and activated with K2CO3. The effects of the AHC on Cu(II) adsorption were investigated using static adsorption experiments. The results indicated that the sample fabricated at 160°C (FW-160) for 6 h had the best adsorption performance with the maximum adsorption amount of Cu(II) (36.77 mg/g). The oxygenated functional group on the surface of FW-160 was observed. The pseudo-second-order kinetic model fitted the experimental results better, which proved that chemisorption was the main adsorption mechanism, and the surface complexations, including oxygenated functional groups, were the predominant factors in hydrochar(HC) sorption for Cu(II). The adsorption amount of the sample simulating rotten FW with Aspergillus niger (FW-160A) was 24.82 mg/g. Through the HTC process, the rotten FW can be made into heavy metal absorbents which can be stored for long term, and finally realize the reduction of volume and cost of storage. Vanadium is a rare metal with excellent properties. The recovery of vanadium from low-grade stone coal and vanadium-containing solid wastes using microbial leaching, has become an increasingly popular technology. In this paper, Vanadium was obtained from stone coal using Aspergillus niger as the leaching strain, and the process of vanadium bioleaching was strengthened. Compared with plasma treatment and ultrasonic pretreatment of stone coal, the leaching effect of stone coal after roasting at 900 °C was the best, and the leaching rate of vanadium was as high as 94.52 %. The vanadium bioleaching mechanism was explored by SEM-EDS, XRD, XPS and FTIR analysis. The findings indicated that Aspergillus niger has the ability to adhere to the surface of stone coal, resulting in subsequent corrosion and degradation. The roasting treatment of stone coal and bioleaching of Aspergillus niger jointly lead to the decrease of crystallinity, the increase of interplanar spacing and the decrease of stability of stone coal. Meanwhile, the valence state and binding form of vanadium changed, and V (III) was oxidized to V (IV) and V (V). The mineral structure may be distorted as a result of the distinctive peak's vibratory stretching and the influence of functional categories that include oxygen, which in turn causes the change of the coordination form of the internal metal ions, which affects the symmetry of the mineral and facilitates the dissolution of vanadium from the stone coal. Dyes can be considered the main contaminants of textile effluents, which have a heterocyclic complicated structure. Disperse dyes are one of the most commonly used classes of dyes with an insoluble nature and low molecular weight. In this study, the elimination of disperse dyes from aqueous medium was investigated by Aspergillus niger fungi using in vivo and in vitro methods. According to the in vitro decolorization results, the fungi showed high biosorption capacity for three different disperse dyes. At an initial dye concentration of 330 mg/l , the adsorption capacity of A. niger measured 225.3 mg/g for Disperse Yellow 23, 127.4 mg/g for Disperse Blue 183, and 176.3 mg/g for Disperse Red 1. Complete dye removal was achieved at lower initial concentrations after a 6-h operation. The results showed that the biosorption process favored acidic conditions, and the adsorption capacity and rate were higher at pH 4.5. Moreover, the kinetics of the adsorption process for all dyes followed the pseudo–second-order kinetic model. An in vivo decolorization test was also performed, in which A. niger was inoculated in a dye-containing medium to investigate the possible biodegradation of the dye by fungi during its growth. The biodegradation process was studied by Fourier transform infrared spectroscopy (FTIR) and high-performance liquid chromatography (HPLC) methods. The results obtained showed a maximum biodegradation of 49.77% for Disperse Red 1, whereas the decolorization for other dyes was mainly based on biosorption. Practical Applications: The industrial wastewater treatment plans in most dyeing houses consist of preliminary coagulation and flocculation tanks followed by biological wastewater treatment by activated sludge. According to the results of this study, A. niger could be applicable in both wastewater treatment steps in industrial plans. A. niger, as a nonharmful microorganism, could be used as a good adsorbent in the coagulation tanks, replacing the chemical coagulants. The adsorbed dye in A. niger's body mass could easily be separated because of its bulky structure. Because dye removal in this method is nearly complete, the effluent could be used in the same dyeing process without much refining process. Using A. niger in biological aerobic procedures could also be practicable according to the results. A. niger could grow in the aerobic condition of biological tanks and simultaneously degrade and adsorb dye molecules. On the other side, A. niger is commercially used for the production of citric acid, and some enzymes and the body mass produced by these industries could be considered for dye removal processes. The use of A. niger as a biosorbent or degradant for dyes still could be studied in several aspects. The study of the degradation mechanism of different dyes by precise tests like liquid chromatography (LC)-mass is an ongoing study by the current team. Biopolymers embedded with nanoparticles of metal oxides (MOs) demonstrate a wide range of bio-functions. Chitosan-incorporated MOs are an interesting class of support matrices for enhancing the biological function, compared to other support matrices. Therefore, the importance of this study lies in exploiting chitosan as a carrier not of one metal as in previous studies, but of two metals in the form of a nanocomposite to carry out several biological functions. The coprecipitation approach was employed to synthesize chitosan/Fe2O3/ZnO-nanocomposite in the present research. The characterization of chitosan/Fe2O3/ZnO-nanocomposite was performed to find out the morphology and dispersion properties of chitosan/Fe2O3/ZnO-nanocomposite. The X-ray diffraction (XRD) investigation revealed that these were crystalline. Fourier transforms infrared (FTIR) spectrum bands were viewed at 400/cm and 900/cm, due to the stretching vibration of Fe and Zn oxygen bond. TEM showed that chitosan/Fe2O3/ZnO-nanocomposite was of 20–95 nm in size. chitosan/Fe2O3/ZnO-nanocomposite exhibited inhibitory potential against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Candida albicans with inhibition zones of 25 ± 0.1, 28 ± 0.2, 27 ± 0.1, and 27 ± 0.2 mm, respectively while didn't inhibited Aspergillus niger. MIC value of nanocomposite was 15.62 ± 0.33 µg/mL for C. albicans, B. subtilis and E. coli, while it was 62.50 ± 0.66 µg/mL for Pseudomonas aeruginosa. Ranged values of nanocomposite MBC (15.62 ± 0.33 to 125 ± 1 µg/mL) were attributed to all tested bacteria. Different concentrations of chitosan/Fe2O3/ZnO-nanocomposite MBC (25, 50, and 75%) reflected anti-biofilm activity against E. coli (85.0, 93.2, and 96.0%), B. subtilis (84.88, 92.21, and 96.99%), S. aureus 81.64, 90.52, and 94.64%) and P. aurogenosa (90.11, 94.43, and 98.24%), respectively. The differences in the levels of antimicrobial activities may depend on the type of examined microbes. Antioxidant activity of chitosan/Fe2O3/ZnO-nanocomposite was recorded with excellent IC50 values of 16.06 and 32.6 µg/mL using DPPH and ABTS scavenging, respectively. Wound heal by chitosan/Fe2O3/ZnO-nanocomposite was achieved with 100% compared to the untreated cells (76.75% of wound closer). The cytotoxicity outcomes showed that the IC50 of the chitosan/Fe2O3/ZnO-nanocomposite was 564.32 ± 1.46 µg/mL normal WI-38 cells. Epothilones are one of the common prescribed anticancer drugs for solid tumors, for their exceptional binding affinity with β-tubulin microtubule, stabilizing their disassembly, causing an ultimate arrest to the cellular growth. Epothilones were initially isolated from Sornagium cellulosum, however, their extremely slow growth rate and low yield of epothilone is the challenge. So, screening for a novel fungal endophyte dwelling medicinal plants, with higher epothilone productivity and feasibility of growth manipulation was the objective. Aspergillus niger EFBL-SR OR342867, an endophyte of Latania loddegesii, has been recognized as the heady epothilone producer (140.2 μg/L). The chemical structural identity of the TLC-purified putative sample of A. niger was resolved from the HPLC, FTIR and LC–ESI–MS/MS analyses, with an identical molecular structure of the authentic epothilone B. The purified A. niger epothilone B showed a resilient activity against MCF-7 (0.022 μM), HepG-2 (0.037 μM), and HCT-116 (0.12 μM), with selectivity indices 21.8, 12.9 and 4, respectively. The purified epothilone B exhibited a potential anti-wound healing activity to HepG-2 and MCF-7 cells by ~ 54.07 and 60.0%, respectively, after 24 h, compared to the untreated cells. The purified epothilone has a significant antiproliferative effect by arresting the cellular growth of MCF-7 at G2/M phase by ~ 2.1 folds, inducing the total apoptosis by ~ 12.2 folds, normalized to the control cells. The epothilone B productivity by A. niger was optimized by the response surface methodology, with ~ 1.4 fold increments (266.9 μg/L), over the control. The epothilone productivity by A. niger was reduced by ~ 2.4 folds by 6 months storage as a slope culture at 4 °C, however, the epothilone productivity was slightly restored with ethylacetate extracts of L. loddegesii, confirming the plant-derived chemical signals that partially triggers the biosynthetic genes of A. niger epothilones. So, this is the first report emphasizing the metabolic potency of A. niger, an endophyte of L. loddegesii, to produce epothilone B, that could be a new platform for industrial production of this drug. Objective: Due to wide range of biological activities exhibited by pyran derivatives such as antibacterial, antioxidant, anticancer and other activities, we have synthesized and investigated the biological activity of some pyran derivatives. Methods: A series of 4-hydroxy-6-methyl-5,6-dihydro-2H-pyran-2-one based azo dyes (IIIa–IIIf) have been synthesized by a conventional diazo coupling reaction. The antioxidant activity was determined via DPPH assay and Antimicrobial activity through disc diffusion test. Results and Discussion: Compound (IIIb) has shown excellent radical scavenging activity with an IC50 value of 20.22 ± 7.36 µg/mL compared to the standard. Furthermore, compounds (IIIc) and (IIId) showed a good antibacterial and antifungal activity against Escherichia coli and Aspergillus niger compared to other prepared azo compounds. Invasive fungal infections (IFI) pose a significant health burden, leading to high morbidity, mortality, and treatment costs. This study aims to develop and characterize nanomicelles for the codelivery of posaconazole and hemp seed oil for IFI via the oral route. The nanomicelles were prepared using a nanoprecipitation method and optimized through the Box Behnken design. The optimized nanomicelles resulted in satisfactory results for zeta potential, size, PDI, entrapment efficiency, TEM, and stability studies. FTIR and DSC results confirm the compatibility and amorphous state of the prepared nanomicelles. Confocal laser scanning microscopy showed that the optimized nanomicelles penetrated the tissue more deeply (44.9µm) than the suspension (25µm). The drug-loaded nanomicelles exhibited sustained cumulative drug release of 95.48 ± 3.27% for 24 h. The nanomicelles showed significant inhibition against Aspergillus niger and Candida albicans (22.4 ± 0.21 and 32.2 ± 0.46 mm, respectively). The pharmacokinetic study on Wistar rats exhibited a 1.8-fold increase in relative bioavailability for the nanomicelles compared to the suspension. These results confirm their therapeutic efficacy and lay the groundwork for future research and clinical applications, providing a promising synergistic antifungal nanomicelles approach for treating IFIs. Among the several threats to humanity by anthropogenic activities, contamination of the environment by heavy metals is of great concern. Upon entry into the food chain, these metals cause serious hazards to plants and other organisms including humans. Use of microbes for bioremediation of the soil and stress mitigation in plants are among the preferred strategies to provide an efficient, cost-effective, eco-friendly solution of the problem. The current investigation is an attempt in this direction where fungal strain PH1 was isolated from the rhizosphere of Parthenium hysterophorus which was identified as Aspergillus niger by sequence homology of the ITS 1 and ITS 4 regions of the rRNA. The strain was tested for its effect on growth and biochemical parameters as reflection of its potential to mitigate Pb stress in Zea mays exposed to 100, 200 and 500 µg of Pb/g of soil. In the initial screening, it was revealed that the strain has the ability to tolerate lead stress, solubilize insoluble phosphate and produce plant growth promoting hormones (IAA and SA) and other metabolites like phenolics, flavonoids, sugar, protein and lipids. Under 500 µg of Pb/g of soil, Z. mays exhibited significant growth retardation with a reduction of 31% in root length, 30.5% in shoot length, 57.5% in fresh weight and 45.2% in dry weight as compared to control plants. Inoculation of A. niger to Pb treated plants not only restored root and shoot length, rather promoted it to a level significantly higher than the control plants. Association of the strain modulated the physio-hormonal attributes of maize plants that resulted in their better growth which indicated a state of low stress. Additionally, the strain boosted the antioxidant defence system of the maize there by causing a significant reduction in the ascorbic acid peroxidase (1.5%), catalase (19%) and 1,1-diphenyl-2 picrylhydrazyl (DPPH) radical scavenging activity (33.3%), indicating a lower stress condition as compared to their non-inoculated stressed plants. Based on current evidence, this strain can potentially be used as a biofertilizer for Pb-contaminated sites where it will improve overall plant health with the hope of achieving better biological and agricultural yields. Eucommiae Cortex (EC) is frequently used alone or in combination with other active ingredients to treat a range of illnesses. An efficient technical instrument for changing cheap or plentiful organic chemicals into rare or costly counterparts is biotransformation. It combines EC with biotransformation techniques with the aim of producing some novel active ingredients, using different strains of bacteria that were introduced to biotransform EC in an aseptic environment. The high-quality strains were screened for identification after the fermentation broth was found using HPLC, and the primary unidentified chemicals were separated and purified in order to be structurally identified. Strain 1 was identified as Aspergillus niger and strain 2 as Actinomucor elegans; the main transformation product A was identified as pinoresinol (Pin) and B as dehydrodiconiferyl alcohol (DA). The biotransformation of EC utilizing Aspergillus niger and Actinomucor elegans is reported for the first time in this study’s conclusion, resulting in the production of Pin and DA. Background: The booming palm oil industry is in line with the growing population worldwide and surge in demand. This leads to a massive generation of palm oil mill effluent (POME). POME is composed of sterilizer condensate (SC), separator sludge (SS), and hydro-cyclone wastewater (HCW). Comparatively, SS exhibits the highest organic content, resulting in various environmental impacts. However, past studies mainly focused on treating the final effluent. Therefore, this pioneering research investigated the optimization of pollutant removal in SS via different aspects of bioremediation, including experimental conditions, treatment efficiencies, mechanisms, and degradation pathways. Methods: A two-level factorial design was employed to optimize the removal of chemical oxygen demand (COD) and turbidity using Aspergillus niger. Bioremediation of SS was performed through submerged fermentation (SmF) under several independent variables, including temperature (20–40 °C), agitation speed (100–200 RPM), fermentation duration (72–240 h), and initial sample concentration (20–100%). The characteristics of the treated SS were then compared to that of raw sludge. Results: Optimal COD and turbidity removal were achieved at 37 °C 100 RPM, 156 h, and 100% sludge. The analysis of variance (ANOVA) revealed a significant effect of selective individual and interacting variables (p < 0.05). The highest COD and turbidity removal were 97.43% and 95.11%, respectively, with less than 5% error from the predicted values. Remarkably, the selected optimized conditions also reduced other polluting attributes, namely, biological oxygen demand (BOD), oil and grease (OG), color, and carbon content. The present work aimed to use cocoa pod husk (CPH) and its extracted pectin as a potential substrate for the production of pectinase and to test the enzyme produced in the clarification process of apple juice. CPH with a particle size of <0.84 mm was employed for pectinase production by a selected strain of Aspergillus niger NRRL 2270. The optimization of the physicochemical conditions of the production medium led to an enzymatic activity of 602.03 U/g dry CPH, which was obtained under the following conditions: 110.25 g/L of CPH, 5% w/v pectin extract, 0.05 g/L of yeast extract, incubation at 28 °C, and pH 4, representing a 176% increase in enzymatic activity under the evaluated conditions. The production kinetics of pectinase showed maximum enzymatic activity at 96 h. Subsequently, the enzymatic extract was precipitated, microfiltered, and ultrafiltrated, resulting in 4852.50 U/mg of specific activity. The enzymatic activity after recovery and purification processes corresponded to 819 U/g dry CPH. Finally, a clarification stage of apple juice was carried out, in which the produced pectinase (CauPec) showed turbidity of 448.89 NTU compared to 417.89 NTU for the commercial enzyme and a viscosity of 1.86 cP, CauPec, and 1.19 cP, commercial pectinase, as well as soluble solids of 8.0 for commercial pectinase and 8.73 for CauPec. Therefore, it can be concluded that CPH and its pectin extract were excellent substrates for the production of pectinases, whose formulation is highly stable and can be applied in the clarification of apple juice. Six corroded glazed pottery figurines, which excavated from the Qibi Ming Tomb of the Tang Dynasty in Xianyang, were selected to study the corrosion mechanism. Optical microscopy (OM), scanning electron microscopy equipped with energy dispersive X-ray spectrometry (SEM–EDS), Energy X-ray fluorescence spectrometry (EDX), and micro-Raman spectroscopy were applied to analyze the pristine composition of the glaze layer and pottery substrate, the composition and microstructure of corrosion products. The results indicate that lead-glazed pottery figurines are low-temperature PbO-CaO-SiO2 glaze, with Pb as the main flux, and Cu, Fe as the main colorant. The corrosion products include PbCO3, CaCO3, PbCO3·PbCl2, PbCl2, PbO2, and Si-rich layer. There are Aspergillus niger and Aspergillus sclerotiorum on the surface of glazed pottery. These microorganisms produce organic acids and mold spots on the surface of the pottery figurines, which in turn cause corrosion pits to form on the surface of the figurines. Chemical corrosion reactions mainly include dissolution–precipitation, combination reaction, and oxidation reaction. This article describes the corrosion process of lead-glazed pottery figurines and establishes corresponding corrosion models. Corn straw is one kind of agricultural by-product containing 70–80% insoluble dietary fiber (IDF). In order to develop corn straw dietary fiber, this study was conducted to increase soluble dietary fiber (SDF) yield and improve the structure, functional and prebiotic properties of IDF and SDF from corn straw treated by alkali oxidation treatment, enzymatic hydrolysis, microbial fermentation and the combination of these methods. The results demonstrated that the yield of SDF was significantly increased from 2.64% to 17.15% after corn straw was treated by alkali oxidation treatment + Aspergillus niger fermentation + cellulase hydrolysis, compared with untreated corn straw. The SDF extracted from corn straw treated by alkali oxidation treatment + Aspergillus niger fermentation + cellulase hydrolysis (F-SDF) exhibited a honeycomb structure, low crystallinity (11.97%), good antioxidant capacity and high capacities of water holding, water solubility and cholesterol absorption and promoted short-chain fatty acids production by chicken cecal microbial fermentation in vitro. F-SDF enhanced the antibacterial activity against Escherichia coli and Staphylococcus aureus proliferations of Lactobacillus plantarum when it was used as a substrate for Lactobacillus plantarum fermentation. The process of carbohydrate metabolism and genetic information transfer is an important part of the study on the effects of the external environment on microbial growth and development. As one of themost significant environmental parameters, pH has an important effect on mycelial growth. In this study, the effects of environmental pH on the growth and nutrient composition of Aspergillus niger (A. niger) filaments were determined. The pH values of the mediumwere 5, 7, and 9, respectively, and themolecularmechanismwas further investigated by transcriptomics andmetabolomicsmethods. The results showed that pH 5 and 9 significantly inhibited filament growth and polysaccharide accumulation of A. niger. Further, themyceliumbiomass of A. niger and the crude polysaccharide content was higher when the medium's pH was 7. The DEGs related to ribosome biogenesis were themost abundant, and the downregulated expression of genes encoding XRN1, RRM, and RIO1 affected protein translation, modification, and carbohydrate metabolism in fungi. The dynamic changes of pargyline and choline were in response to the oxidative metabolism of A. niger SICU-33. The ribophorin_I enzymes and DL-lactate may be important substances related to pH changes during carbohydrate metabolism of A.niger SICU-33. The results of this study provide useful transcriptomic andmetabolomic information for further analyzing the bioinformatic characteristics of A. niger and improving the application in ecological agricultural fermentation. Immobilization of enzymes on aminated supports using the glutaraldehyde chemistry may involve three different interactions, cationic, hydrophobic, and covalent interactions. To try to understand the impact this heterofunctionality, we study the physical adsorption of the beta-galactosidase from Aspergillus niger , on aminated supports (MANAE) and aminated supports with one (MANAE-GLU) or two molecules of glutaraldehyde (MANAE-GLU-GLU). To eliminate the chemical reactivity of the glutaraldehyde, the supports were reduced using sodium borohydride. After enzyme adsorption, the release of the enzyme from the supports using different NaCl concentrations, Triton X100, ionic detergents (SDS and CTAB), or different temperatures (4 °C to 55 °C) was studied. Using MANAE support, at 0.3 M NaCl almost all the immobilized enzyme was released. Using MANAE-GLU, 0.3 M, and 0.6 M NaCl similar results were obtained. However, incubation at 1 M or 2 M NaCl, many enzyme molecules were not released from the support. For the MANAE-GLU-GLU support, none of the tested concentrations of NaCl was sufficient to release all enzyme bound to the support. Only using high temperatures, 0.6 M NaCl, and 1 % CTAB or SDS, could the totality of the proteins be released from the support. As the critical process for quality formation of ripened Pu-erh tea (RpPT), pile-fermentation mechanism has been widely studied by using omics approach, chemical analysis, microbial distribution and their inoculated fermentation tests. Despite several reviews about chemical compositions and health benefits in RpPT and other dark teas, more comprehensive study is requisite to summarize the pile-fermentation mechanism of RpPT and application prospects of relevant microbes. This paper aims to elaborate dominant microbes, recent advances in single- and multi-omics approaches, and pathways of characteristic components during the pile-fermentation. Microbial effects are summarized and compared, which would enhance their application potential in RpPT production. At least 93 fungi and 53 bacteria have been isolated from RpPT related to the pile-fermentation. Aspergillus niger is regarded as dominant microorganism in pile-fermentation for its higher detection frequency and relative abundance. Aspergillus , Penicillium, Rasamsoni, Rhizomucor and Bacillus genera promote RpPT quality formation. The low mycotoxins contamination risk in pile-fermentation and inoculated fermentation indicates microbial application potential in RpPT production. Several microbes are confirmed to promote formation of characteristic components including theabrownins, puerins, teadenol A, gallic acid, lovastatin, gamma-aminobutyric acid and several flavonols such as quercetin. Historical records indicate that Xianyang Ancient Ferry Site, dating back over three millennia to the late Shang Dynasty, has been an important transportation hub in ancient times. The use of combined wooden stakes and stones is a special method for flood control among ancient flood prevention techniques. This research aims to evaluate the current preservation condition of these ancient wooden stakes. The dating of the wooden stakes at the site ranges between 940 ± 30 BP and 320 ± 30 BP. The fiber cross-section and microstructure images of the wooden samples indicate that the wood is attributed to Platycladus orientalis. SEM images indicate decay and deterioration of the wooden samples, with the presence of mold spores within the cavities. XRD, FTIR, and TG/DSC spectra of historical wooden samples collected from the site and fresh Platycladus orientalis wood samples demonstrate a significant decrease of cellulose crystallinity in the historical samples, leading to severe deterioration of the wood at the site. Five dominant mold species were identified, i.e., Aspergillus niger, Aspergillus flavus, Penicillium citrinum, Trichoderma, and Phanerochaete chrysosporium, which accelerate the degradation of cellulose and other polysaccharides. This research provides scientific evidence for the selection of reinforcement materials and guides curators and conservators to formulate conservation strategies in the future. Palm leaf manuscripts (PLMs), venerable historical artefacts containing Buddhist scriptures, history, mathematics and literature, which are carried by palm leaves (Corypha umbraculifera) and are highly susceptible to microbial degradation during prolonged storage. This degradation results in significant alterations to both the appearance and material properties of PLMs, but the precise mechanism underlying this deterioration remains a mystery. To this end, the present study focused on ancient PLMs from Xishuangbanna Dai Autonomous Prefecture in Yunnan Province, China. The objective of present study was to isolate, culture and identify the microorganisms typically found in these manuscripts and to use them to biodegrade the carriers of PLMs. Detailed observations of the biodegradation behavior of these microorganisms on the carriers of PLMs were carried out, together with characterizations of the hierarchical structure and mechanical properties of the leaf fiber cell walls. This comprehensive analysis provided insights into the deterioration mechanisms of the carriers of PLMs. The study revealed the presence of the common fungus Aspergillus niger on ancient PLMs. Aspergillus niger can secrete cellulase, lipase, and acidic substances after colonizing on the carriers of PLMs. These substances sequentially damage the carrier's epidermal cells, mesophyll cells, and leaf fibers, leading to the separation of different tissue structures. At the molecular level, the lipids on the surface of the leaves were degraded initially, and sequential depolymerization of hemicellulose, amorphous cellulose, and crystalline cellulose occurred. Additionally, this study firstly applied nanoindentation technology in the research of PLMs. The mechanical properties of the cell walls underwent notable alterations due to the modifications in chemical and crystalline structure of the carriers of PLMs upon the biodegradation of Aspergillus niger. Specifically, the hardness and elastic modulus of leaf fiber cell walls showed an initial increasing and then decreasing trend, consistent with the trend of cellulose crystallinity, which also provided a new reference for assessing the degree of deterioration of PLMs. [ABSTRACT FROM AUTHOR]
