1. Upscale Synthesis of Magnetic Mesoporous Silica Nanoparticles and Application to Metal Ion Separation: Nanosafety Evaluation

Abstract

The synthesis of core–shell magnetic mesoporous nanoparticles (MMSNs) through a phase transfer process is usually performed at the 100–250 mg scale. At the gram scale, nanoparticles without cores or with multicore systems are observed. Iron oxide core nanoparticles (IO) were synthesized through a thermal decomposition procedure of α-FeO(OH) in oleic acid. A phase transfer from chloroform to water was then performed in order to wrap the IO nanoparticles with a mesoporous silica shell through the sol–gel procedure. MMSNs were then functionalized with DTPA (diethylenetriaminepentacetic acid) and used for the separation of metal ions. Their toxicity was evaluated. The phase transfer procedure was crucial to obtaining MMSNs on a large scale. Three synthesis parameters were rigorously controlled: temperature, time and glassware. The homogeneous dispersion of MMSNs on the gram scale was successfully obtained. After functionalization with DTPA, the MMSN-DTPAs were shown to have a strong affinity for Ni ions. Furthermore, toxicity was evaluated in cells, zebrafish and seahorse cell metabolic assays, and the nanoparticles were found to be nontoxic. We developed a method of preparing MMSNs at the gram scale. After functionalization with DTPA, the nanoparticles were efficient in metal ion removal and separation; furthermore, no toxicity was noticed up to 125 µg mL−1 in zebrafish.

Abstract

Dendritic mesoporous organosilica nanoparticles (DMON) are a new class of biodegradable nanoparticles suitable for biomolecule delivery. We studied the photochemical internalization (PCI) and photodynamic therapy (PDT) of DMON to investigate new ways for DMON to escape from the endosomes-lysosomes and deliver biomolecules into the cytoplasm of cells. We added photosensitizers in the framework of DMON and found that DMON were loaded with siRNA or FVIII factor protein. We made four formulations with four different photosensitizers. The photosensitizers allowed us to perform imaging of DMON in cancer cells, but the presence of the tetrasulfide bond in the framework of DMON quenched the formation of singlet oxygen. Fortunately, one formulation allowed us to efficiently deliver proapoptotic siRNA in MCF-7 cancer cells leading to 31% of cancer cell death, without irradiation. As for FVIII protein, it was loaded in two formulations with drug-loading capacities (DLC) up to 25%. In conclusion, DMON are versatile nanoparticles capable of loading siRNA and delivering it into cancer cells, and also loading FVIII protein with good DLC. Due to the presence of tetrasulfide, it was not possible to perform PDT or PCI.

Abstract

Background: In addition to their great optical properties, nanodiamonds (NDs) have recently proved useful for two-photon-excited photodynamic therapy (TPE-PDT) applications. Indeed, they are able to produce reactive oxygen species (ROS) directly upon two-photon excitation but not with one-photon excitation; Methods: Fluorescent NDs (FNDs) with a 100 nm diameter and detonation NDs (DNDs) of 30 nm were compared. In order to use the gems for cancer-cell theranostics, they were encapsulated in a bis(triethoxysilyl)ethylene-based (ENE) periodic mesoporous organosilica (PMO) shell, and the surface of the formed nanoparticles (NPs) was modified by the direct grafting of polyethylene glycol (PEG) and amino groups using PEG-hexyltriethoxysilane and aminoundecyltriethoxysilane during the sol–gel process. The NPs’ phototoxicity and interaction with MDA-MB-231 breast cancer cells were evaluated afterwards; Results: Transmission electronic microscopy images showed the formation of core–shell NPs. Infrared spectra and zeta-potential measurements confirmed the grafting of PEG and NH2 groups. The encapsulation of the NDs allowed for the imaging of cancer cells with NDs and for the performance of TPE-PDT of MDA-MB-231 cancer cells with significant mortality. Conclusions: Multifunctional ND@PMO core–shell nanosystems were successfully prepared. The NPs demonstrated high biocompatibility and TPE-PDT efficiency in vitro in the cancer cell model. Such systems hold good potential for two-photon-excited PDT applications.

Abstract

(1) Background: Due to human activities, greenhouse gas (GHG) concentrations in the atmosphere are constantly rising, causing the greenhouse effect. Among GHGs, carbon dioxide (CO2) is responsible for about two-thirds of the total energy imbalance which is the origin of the increase in the Earth’s temperature. (2) Methods: In this field, we describe the development of periodic mesoporous organosilica nanoparticles (PMO NPs) used to capture and store CO2 present in the atmosphere. Several types of PMO NP (bis(triethoxysilyl)ethane (BTEE) as matrix, co-condensed with trialkoxysilylated aminopyridine (py) and trialkoxysilylated bipyridine (Etbipy and iPrbipy)) were synthesized by means of the sol-gel procedure, then characterized with different techniques (DLS, TEM, FTIR, BET). A systematic evaluation of CO2 adsorption was carried out at 298 K and 273 K, at low pressure. (3) Results: The best values of CO2 adsorption were obtained with 6% bipyridine: 1.045 mmol·g−1 at 298 K and 2.26 mmol·g−1 at 273 K. (4) Conclusions: The synthetized BTEE/aminopyridine or bipyridine PMO NPs showed significant results and could be promising for carbon capture and storage (CCS) application.

Abstract

Abstract New therapy development is critically needed for ovarian cancer. We used the chicken egg CAM assay to evaluate efficacy of anticancer drug delivery using recently developed biodegradable PMO (periodic mesoporous organosilica) nanoparticles. Human ovarian cancer cells were transplanted onto the CAM membrane of fertilized eggs, resulting in rapid tumor formation. The tumor closely resembles cancer patient tumor and contains extracellular matrix as well as stromal cells and extensive vasculature. PMO nanoparticles loaded with doxorubicin were injected intravenously into the chicken egg resulting in elimination of the tumor. No significant damage to various organs in the chicken embryo occurred. In contrast, injection of free doxorubicin caused widespread organ damage, even when less amount was administered. The lack of toxic effect of nanoparticle loaded doxorubicin was associated with specific delivery of doxorubicin to the tumor. Furthermore, we observed excellent tumor accumulation of the nanoparticles. Lastly, a tumor could be established in the egg using tumor samples from ovarian cancer patients and that our nanoparticles were effective in eliminating the tumor. These results point to the remarkable efficacy of our nanoparticle based drug delivery system and suggests the value of the chicken egg tumor model for testing novel therapies for ovarian cancer.

Abstract

We report herein the preparation of mixed periodic mesoporous organosilica nanoparticles (E-Pn 75/25 and 90/10 PMO NPs) by sol-gel co-condensation of E-1,2-bis(triethoxysilyl)ethylene ((E)-BTSE or E) with previously synthesized disilylated tert-butyl 3,5-dialkoxybenzoates bearing either sulfide (precursor P1) or carbamate (precursor P2) functionalities in the linker. The syntheses were performed with cetyltrimethylammonium bromide (CTAB) as template in the presence of sodium hydroxide in water at 80 °C. The nanomaterials have been characterized by Transmission Electron Microscopy (TEM), nitrogen-sorption measurements (BET), Dynamic Light Scattering (DLS), zeta-potential, Thermogravimetric Analysis (TGA), FTIR, 13C CP MAS NMR and small angle X-ray diffraction (p-XRD). All the nanomaterials were obtained as mesoporous rodlike-shape nanoparticles. Remarkably, E-Pn 90/10 PMO NPs presented high specific surface areas ranging from 700 to 970 m2g−1, comparable or even higher than pure E PMO nanorods. Moreover, XRD analyses showed an organized porosity for E-P1 90/10 PMO NPs typical for a hexagonal 2D symmetry. The other materials showed a worm-like mesoporosity.

Abstract

Periodic Mesoporous Organosilica Nanoparticles (PMONPs) are nanoparticles of high interest for nanomedicine applications. These nanoparticles are not composed of silica (SiO2). They belong to hybrid organic−inorganic systems. We considered using these nanoparticles for CO2 release as a contrast agent for High Intensity Focused Ultrasounds (HIFU). Three molecules (P1−P3) possessing two to four triethoxysilyl groups were synthesized through click chemistry. These molecules possess a tert-butoxycarbonyl (BOC) group whose cleavage in water at 90−100 °C releases CO2. Bis(triethoxysilyl)ethylene E was mixed with the molecules Pn (or not for P3) at a proportion of 90/10 to 75/25, and the polymerization triggered by the sol-gel procedure led to PMONPs. PMONPs were characterized by different techniques, and nanorods of 200−300 nm were obtained. These nanorods were porous at a proportion of 90/10, but non-porous at 75/25. Alternatively, molecules P3 alone led to mesoporous nanoparticles of 100 nm diameter. The BOC group was stable, but it was cleaved at pH 1 in boiling water. Molecules possessing a BOC group were successfully used for the preparation of nanoparticles for CO2 release. The BOC group was stable and we did not observe release of CO2 under HIFU at lysosomal pH of 5.5. The pH needed to be adjusted to 1 in boiling water to cleave the BOC group. Nevertheless, the concept is interesting for HIFU theranostic agents.

Abstract

(1) Background: Nanomedicine has recently emerged as a promising field, particularly for cancer theranostics. In this context, nanoparticles designed for imaging and therapeutic applications are of interest. We, therefore, studied the encapsulation of upconverting nanoparticles in mesoporous organosilica nanoparticles. Indeed, mesoporous organosilica nanoparticles have been shown to be very efficient for drug delivery, and upconverting nanoparticles are interesting for near-infrared and X-ray computed tomography imaging, depending on the matrix used. (2) Methods: Two different upconverting-based nanoparticles were synthesized with Yb3+-Er3+ as the upconverting system and NaYF4 or BaLuF5 as the matrix. The encapsulation of these nanoparticles was studied through the sol-gel procedure with bis(triethoxysilyl)ethylene and bis(triethoxysilyl)ethane in the presence of CTAB. (3) Results: with bis(triethoxysilyl)ethylene, BaLuF5: Yb3+-Er3+, nanoparticles were not encapsulated, but anchored on the surface of the obtained mesoporous nanorods BaLuF5: Yb3+-Er3+@Ethylene. With bis(triethoxysilyl)ethane, BaLuF5: Yb3+-Er3+ and NaYF4: Yb3+-Er3+nanoparticles were encapsulated in the mesoporous cubic structure leading to BaLuF5: Yb3+-Er3+@Ethane and NaYF4: Yb3+-Er3+@Ethane, respectively. (4) Conclusions: upconversion nanoparticles were located on the surface of mesoporous nanorods obtained by hydrolysis polycondensation of bis(triethoxysilyl)ethylene, whereas encapsulation occurred with bis(triethoxysilyl)ethane. The later nanoparticles NaYF4: Yb3+-Er3+@Ethane or BaLuF5: Yb3+-Er3+@Ethane were promising for applications with cancer cell imaging or X-ray-computed tomography respectively.

Abstract

The aim of the present work is the development of highly efficient targeting molecules to specifically address mesoporous silica nanoparticles (MSNs) designed for the photodynamic therapy (PDT) of prostate cancer. We chose the strategy to develop a novel compound that allows the improvement of the targeting of the cation-independent mannose 6-phosphate receptor, which is overexpressed in prostate cancer. This original sugar, a dimannoside-carboxylate (M6C-Man) grafted on the surface of MSN for PDT applications, leads to a higher endocytosis and thus increases the efficacy of MSNs.

Abstract

(1) Background: Nanomedicine has recently emerged as a new area of research, particularly to fight cancer. In this field, we were interested in the vectorization of pepstatin A, a peptide which does not cross cell membranes, but which is a potent inhibitor of cathepsin D, an aspartic protease particularly overexpressed in breast cancer. (2) Methods: We studied two kinds of nanoparticles. For pepstatin A delivery, mesoporous silica nanoparticles with large pores (LPMSNs) and hollow organosilica nanoparticles (HOSNPs) obtained through the sol–gel procedure were used. The nanoparticles were loaded with pepstatin A, and then the nanoparticles were incubated with cancer cells. (3) Results: LPMSNs were monodisperse with 100 nm diameter. HOSNPs were more polydisperse with diameters below 100 nm. Good loading capacities were obtained for both types of nanoparticles. The nanoparticles were endocytosed in cancer cells, and HOSNPs led to the best results for cancer cell killing. (4) Conclusions: Mesoporous silica-based nanoparticles with large pores or cavities are promising for nanomedicine applications with peptides.

Abstract

International audience; The world of biomedical research is in constant evolution, requiring more and more conditions and norms through pre-clinic and clinic studies. Nanodiamonds (NDs) with exceptional optical, thermal and mechanical properties emerged on the global scientific scene and recently gained more attention in biomedicine and bioanalysis fields. Many problematics have been deliberated to better understand their in vitro and in vivo efficiency and compatibility. Light was shed on their synthesis, modification and purification steps, as well as particle size and surface properties in order to find the most suitable operating conditions. In this review, we present the latest advances of NDs use in bioapplications. A large variety of subjects including anticancer and antimicrobial systems, wound healing and tissue engineering management tools, but also bioimaging and labeling probes are tackled. The key information resulting from these recent works were evidenced to make an overview of the potential features of NDs, with a special look on emerging therapeutic and diagnosis combinations.

Abstract

Periodic Mesoporous Organosilica Nanoparticles (PMONPs) are nanoparticles of high interest for nanomedicine applications. These nanoparticles are not composed of silica (SiO2). They belong to hybrid organic&ndash, inorganic systems. We considered using these nanoparticles for CO2 release as a contrast agent for High Intensity Focused Ultrasounds (HIFU). Three molecules (P1&ndash, P3) possessing two to four triethoxysilyl groups were synthesized through click chemistry. These molecules possess a tert-butoxycarbonyl (BOC) group whose cleavage in water at 90&ndash, 100 &deg, C releases CO2. Bis(triethoxysilyl)ethylene E was mixed with the molecules Pn (or not for P3) at a proportion of 90/10 to 75/25, and the polymerization triggered by the sol-gel procedure led to PMONPs. PMONPs were characterized by different techniques, and nanorods of 200&ndash, 300 nm were obtained. These nanorods were porous at a proportion of 90/10, but non-porous at 75/25. Alternatively, molecules P3 alone led to mesoporous nanoparticles of 100 nm diameter. The BOC group was stable, but it was cleaved at pH 1 in boiling water. Molecules possessing a BOC group were successfully used for the preparation of nanoparticles for CO2 release. The BOC group was stable and we did not observe release of CO2 under HIFU at lysosomal pH of 5.5. The pH needed to be adjusted to 1 in boiling water to cleave the BOC group. Nevertheless, the concept is interesting for HIFU theranostic agents.

Abstract

1) Background: Nanomedicine has recently emerged as a promising field, particularly for cancer theranostics. In this context, nanoparticles designed for imaging and therapeutic applications are of interest. We, therefore, studied the encapsulation of upconverting nanoparticles in mesoporous organosilica nanoparticles. Indeed, mesoporous organosilica nanoparticles have been shown to be very efficient for drug delivery, and upconverting nanoparticles are interesting for near-infrared and X-ray computed tomography imaging, depending on the matrix used. (2) Methods: Two different upconverting-based nanoparticles were synthesized with Yb3+-Er3+ as the upconverting system and NaYF4 or BaLuF5 as the matrix. The encapsulation of these nanoparticles was studied through the sol-gel procedure with bis(triethoxysilyl)ethylene and bis(triethoxysilyl)ethane in the presence of CTAB. (3) Results: with bis(triethoxysilyl)ethylene, BaLuF5: Yb3+-Er3+, nanoparticles were not encapsulated, but anchored on the surface of the obtained mesoporous nanorods BaLuF5: Yb3+-Er3+@Ethylene. With bis(triethoxysilyl)ethane, BaLuF5: Yb3+-Er3+ and NaYF4: Yb3+-Er3+nanoparticles were encapsulated in the mesoporous cubic structure leading to BaLuF5: Yb3+-Er3+@Ethane and NaYF4: Yb3+-Er3+@Ethane, respectively. (4) Conclusions: upconversion nanoparticles were located on the surface of mesoporous nanorods obtained by hydrolysis polycondensation of bis(triethoxysilyl)ethylene, whereas encapsulation occurred with bis(triethoxysilyl)ethane. The later nanoparticles NaYF4: Yb3+-Er3+@Ethane or BaLuF5: Yb3+-Er3+@Ethane were promising for applications with cancer cell imaging or X-ray-computed tomography respectively., This research was funded by ANR NanoptPDT. Abstract

International audience; Mesoporous organosilica nanoparticles (PHT-PMO) have been prepared from an octa-triethoxysilylated Zn phthalocyanine precursor. These PHT-PMO nanoparticles had no dark toxicity but high phototoxicity when irradiated at 650 nm, and remarkable near-infrared phototoxicity when excited at 760 and 810 nm. The PHT-PMO were then aminated to promote electrostatic complexation with siRNA. Transfection experiments were performed upon NIR irradiation and photochemical internalization was very efficient, leading to 65% luciferase extinction in MCF-7 cancer cells expressing stable luciferase.

We chose the strategy to develop a novel compound that allows the improvement of the targeting of the cation-independent mannose 6-phosphate receptor, which is overexpressed in prostate cancer. This original sugar, a dimannoside-carboxylate (M6C-Man) grafted on the surface of MSN for PDT applications, leads to a higher endocytosis and thus increases the efficacy of MSNs. Abstract

International audience; (1) Background: Nanomedicine has recently emerged as a new area of research, particularly to fight cancer. In this field, we were interested in the vectorization of pepstatin A, a peptide which does not cross cell membranes, but which is a potent inhibitor of cathepsin D, an aspartic protease particularly overexpressed in breast cancer. (2) Methods: We studied two kinds of nanoparticles. For pepstatin A delivery, mesoporous silica nanoparticles with large pores (LPMSNs) and hollow organosilica nanoparticles (HOSNPs) obtained through the sol-gel procedure were used. The nanoparticles were loaded with pepstatin A, and then the nanoparticles were incubated with cancer cells. (3) Results: LPMSNs were monodisperse with 100 nm diameter. HOSNPs were more polydisperse with diameters below 100 nm. Good loading capacities were obtained for both types of nanoparticles. The nanoparticles were endocytosed in cancer cells, and HOSNPs led to the best results for cancer cell killing. (4) Conclusions: Mesoporous silica-based nanoparticles with large pores or cavities are promising for nanomedicine applications with peptides.

Abstract

The synthesis of hollow organosilica nanoparticles is described from bis triethoxysilylpropyldisulfide and bis(trimethoxylsilylpropyl)methylamine. The nanoparticles were monodisperse with a mean diameter of 90 nm, and the adsorption−desorption properties of these nanoparticles were investigated with nitrogen at 77 K, n-hexane, and water. The nanoparticles were nonporous at 77 K as shown by N2 sorption analyses. n-Hexane interacted reversibly through physisorption with the nanoparticles, whereas water showed a strong nonreversible interaction, probably because of chemisorption with nitrogen atom sites. Amoxicillin antibiotic was loaded with these nanoparticles and the loading capacity was 30%. Ag+ ions were also adsorbed with amoxicillin, and the multifunctional nanoparticles were efficient in inhibiting bacterial growth with a synergy between the antibiotic delivery and Ag+ release.

Abstract

International audience; In this review, the use of mesoporous silica nanoparticles for photodynamic therapy (PDT) applications is described for the year 2017. Since the pioneering work in 2009, nanosystems involving mesoporous silica nanoparticles have gained in complexity with a sophisticated core–shell system able to perform multi-imaging and multi-therapies, not only for cancer diseases but also for anti-microbial therapy, atherosclerosis, or Alzheimer disease. Near-infrared, excitation light based on up-converting systems, X-rays or persistent luminescent systems are described for deeper tissue treatments.

International audience; The sol‐gel synthesis of hollow organosilica nanoparticles incorporating amino groups with uniform size are described. These nanoparticles were successfully prepared via a microemulsion method. Abstract

International audience; Three dimensional sub-micron resolution has made two-photon nanomedicine a very promising medical tool for cancer treatment since current techniques cause significant side effects for lack of spatial selectivity. Two-photon-excited (TPE) photodynamic therapy (PDT) has been achieved via mesoporous nanoscaffolds, but the efficiency of the treatment could still be improved. Herein, we demonstrate the enhancement of the treatment efficiency via gold-mesoporous organosilica nanocomposites for TPE-PDT in cancer cells when compared to mesoporous organosilica particles. We performed the first comparative study of the influence of the shape and spatial position of gold nanoparticles (AuNPs) with mesoporous silica nanoparticles (MSN) functionalized with thiol groups and doped with a two-photon electron donor (2PS). The resulting multifunctional nanocarriers displayed TPE-fluorescence and were imaged inside cells. Furthermore, mesoporous organosilica NPs decorated gold nanospheres (AuNSs) induced 63 percent of selective killing on MCF-7 breast cancer cells. This study thus provides insights for the design of more effective multifunctional two-photon-sensitive nanocomposites via AuNPs for biomedical applications.

Abstract

International audience; We report the synthesis of 20-nm-sized mesoporous silica nanoparticles encapsulating three different porphyrin photosensitizers (PS). Synthesized by the sol–gel method, in the presence of cetyltrimethylammonium bromide surfactant and tetraorthosilicate as silica source, we developed those nanoparticles with the aim of treating cancer cells using photodynamic therapy. The colloidal stability, due to the PEG-silane chain grafted on the surface,was demonstrated at 37 C in cell culture medium. Then, nanoparticles were functionalized by silylated squarate mannose and used in MCF-7 breast cancer cells for one-photon therapy. Promising results were obtained after 5 h of incubation.

The synthesis of a zinc porphyrin derivative possessing eight triethoxysilyl groups was performed through a CuAAC-click reaction. This porphyrin was covalently entrapped in ethenylene-bridged mesoporous organosilica nanoparticles which efficiently allowed performing doxorubicin delivery and two-photon imaging of breast cancer cells.

Abstract

Research in nanomedicine has grown rapidly over the past few years and is playing a key role in the development of effective treatments for ophthalmological purpose. Retinoblastoma is an intraocular tumor triggered by genetic mutation in young children and photodynamic therapy (PDT) is currently developed as a promising non-mutagenic approach to treat this cancer. In this work, we have first identified two receptors, MRC2 and CD209, which are highly expressed by retinoblastoma. Then, we developed mesoporous silica nanoparticles (MSN) grafted with the antibodies anti-MRC2 and/or anti-CD209 for retinoblastoma PDT and imaging.

Abstract

We report a two-photon-actuated cancer cell killing system that kills the cancer cells via drug delivery through multifunctional mesoporous silica nanogates. Two-photon-sensitive mesoporous organosilica (M2PS) nanocarriers were synthesized via the co-condensation of a silica precursor and a two-photon electron donor. The nanogates were constructed using a fast one-pot process at room temperature on the drug-loaded M2PS nanoparticles (NPs) with the bis(3-triethoxysilylpropyl)disulfide precursor. One and two-photon-actuated cargo releases from the M2PS nanogates were successfully monitored in aqueous solutions. Furthermore, the cellular uptake in MCF-7 cells was demonstrated via two-photon fluorescence imaging of the NPs, which were then applied successfully for drug delivery in cells.

Abstract

International audience; In this work, we describe the synthesis of new Mixed Periodic Mesoporous Organosilica Nanoparticles (MPMO NPs), combining the co-condensation of a tetra-trialkoxysilylated twophoton photosensitizer with bis-(triethoxysilyl)phenylene or ethylene. Novel gold core-MPMO shell systems are also described. The MPMO NPs are analyzed and characterized by multiple techniques, and are very efficient for anti-cancer drug delivery combined with two-photon therapy in MCF-7 breast cancer cells, leading down to 76% cancer cell death. MPMO NPs are thus very promising for nanomedicine applications.

Abstract

International audience; The combination of the copper-catalyzed alkyneazide cycloaddition (CuAAC) reactionwith sol-gel processing enables the versatile preparation of sol-gel materials under different shapes with targeted functionalities through a diversity-oriented approach. In this account, the development of the CuAAC reaction under anhydrous conditions for the synthesis of sol-gel precursors and for the assembling of magnetic nanoparticles on self-assembled monolayers is related, as well as the use of the classical CuAAC methodologies for the functionalization of mesoporous silica nanoparticles and microdots arrays. Coupling CuAAC and Sol-Gel will result in simplified preparations of multifunctional materials with controlled morphologies.

Abstract

We report the study of the functionalization of small sized MSNs with mannose. Classical procedures which were efficient with 100 nm diameter MSNs led to aggregation of small sized MSNs. Therefore, we grafted long chain amino silane and PEG-silane on the surface of the MSNs and reacted squarate mannose with NH2 groups. Although the MSN suspension was stable in EtOH, immediate aggregation was noticed in PBS which is attributed to remaining amino groups on the surface of the MSNs. We therefore grafted silylated mannose and PEG-silane on the MSN surface. Depending on the conditions, reversible aggregation was observed in PBS when heating to 37 °C and cooling to RT, or stable suspensions were obtained. The stable mannose-functionalized MSN suspensions were used for the imaging of retinoblastoma cells.

Abstract

Versatile heavy metal ions removal is performed via iron oxide core mesoporous silica shell nanocontainers functionalized with diethylene triamine pentaacetic acid. Magnetic separation from aqueous media is obtained for a dozen of the most toxic heavy metal ions with remarkable efficiencies. Furthermore, this study demonstrates enhancement of the adsorption capacities with the combination of porous and surface functionalization of the nanocontainers.

Abstract

International audience; Mesoporous silica nanoparticles (MSN) are functionalized in the walls with an original fluorophore with a high two-photon absorption cross-section. The pores of the MSN filled with anticancer drug are blocked with a pseudo-rotaxane constituted by an azobenzene stalk and a β-cyclodextrin moiety. After incubation of the nanosystem with MCF-7 breast cancer cells, two-photon irradiation at low power is used to image the cells. At high power, cancer cell killing is observed due to the two-photon-triggered opening of the pores through FRET and the release of the anticancer drug from the MSN.

Abstract

International audience; A therapy of cancer cells: Two-photon-triggered camptothecin delivery (see picture) with nanoimpellers was studied in MCF-7 breast cancer cells. A fluorophore with a high two-photon absorption cross-section was first incorporated in the nanoimpellers. Fluorescence resonance energy transfer (FRET) from the fluorophore to the azobenzene moiety was demonstrated.

Abstract

International audience; An original fluorophore engineered for two-photon excitation or a porphyrin derivative were entrapped in the silica shell of magnetic porous silica nanoparticles during the synthesis of the silica moiety without damaging the structure of the organic part. The mild conditions involved allowed obtaining microporous or mesoporous silica magnetic nanoparticles, respectively. Mannose was grafted on the surface of the nanoparticles to target MCF-7 breast cancer cells. The studies of magnetic properties of these hybrid nanoparticles showthat they present a blocking temperature at 190 K. The nano-objects designed with the twophoton fluorophore were efficient for two-photon imaging of MCF-7 cancer cells, whereas the nanoobjects with the photosensitizer efficiently killed cancer cells. The presence of the mannose moiety was demonstrated to improve both imaging and therapy properties.

Abstract

Retinoblastoma is a rare cancer triggered by genetic mutation that forms in the eyes of children. In industrialized countries, 95% of patients are cured by chemotherapy and conservative treatments. However these treatments can increase the risk of secondary tumors in patients with a constitutional alteration of the retinoblastoma gene Rb1. Photodynamic therapy (PDT) represents a therapeutic approach and may reduce the incidence of secondary tumors. PDT is an established cancer treatment based on the light activation of a photosensitizing agent thus generating cytotoxic reactive oxygen species that cause cellular damage. We focused on mesoporous silica nanoparticles (MSN) for one-photon excited PDT combined with drug delivery and carbohydrate targeting applied on retinoblastoma. We demonstrated that bitherapy (camptothecin delivery and PDT) performed with MSN was efficient in inducing retinoblastoma cell death. Alternatively, MSN designed for two-photon excited PDT were also studied and irradiation in near-infrared at low fluency efficiently killed retinoblastoma cancer cells. These data provide new evidences of the potential of functionalized and targeted MSN for treatment of retinoblastoma and could lead to propose a non-invasive therapy with reduced side effects.

Abstract

International audience; Photodynamic therapy has emerged as an alternative to chemotherapy and radiotherapy for cancer treatment. Nanoparticles have recently been proposed as effective carriers for photosensitizers. Depending on their chemical composition, these can be used for diagnosis and therapy due to the selective accumulation of the photosensitizer in cancer cells in vitro or in tumors in vivo. Multifunctional nanoplatforms combining several applications within the same nano-object emerge as potential important theranostic tools. This review, based on the chemical nature of the nanoparticles will discuss recent advances in the area of non polymeric nanoparticles for photodynamic therapy applications.

Abstract

The synthesis of mesoporous silica nanoparticles (MSN) covalently encapsulating fluoresceine or a photosensitizer, functionalized with galactose on the surface is described. Confocal microscopy experiments demonstrated that the uptake of galactose-functionalized MSN by colorectal cancer cells was mediated by galactose receptors leading to the accumulation of the nanoparticles in the endosomal and lysosomal compartments. The MSN functionalized with a photosensitizer and galactose were loaded with the anti-cancer drug camptothecin. Those MSN combining drug delivery and photodynamic therapy were tested on three cancer cell lines and showed a dramatic enhancement of cancer cell death compared to separate treatments.

Abstract

In this work, we focused on mesoporous silica nanoparticles (MSN) for one photon excitated photodynamic therapy (OPE-PDT) combined with drug delivery and carbohydrate targeting applied on retinoblastoma, a rare disease of childhood. We demonstrate that bitherapy (camptothecin delivery and photodynamic therapy) performed with MSN on retinoblastoma cancer cells was efficient in inducing cancer cell death. Alternatively MSN designed for two-photon excited photodynamic therapy (TPE-PDT) were also studied and irradiation at low fluence efficiently killed retinoblastoma cancer cells.

Abstract

International audience; Mesoporous silica nanoparticles containing clickable functions (azide or alkyne, 1 or 10 mol%) were synthesized by the sol-gel method directly from molecular precursors using CTAB as structure directing agent. Their structures were discussed on the basis of TEM, DLS, SAXS, N2-sorption analyses, FTIR, Raman and elemental analysis. The success of their functionalization by pyrene derivatives using CuAAC reactions opens new perspectives in the synthesis of functional MSNs for applications as nanomachines.

Nanoparticles have been used to improve the delivery and the efficiency of the photosensitizer as they allow its encapsulation without loss of activity. Using targeting strategies, they can also allow the selective accumulation of the photosensitizer in the cancer tissues. In this review, based on the chemical nature of the nanoparticles, the different methods for their syntheses are described from the pioneering works to the latest achievements. Indeed the nanosystems can be conjugated to a biomolecule or an antibody to target receptors over-expressed in cancer cells and/or angiogenic vascular endothelial cells. Numerous in vivo and in vitro applications have been described. Multifunctional nanoplatforms combining several applications within the same nano-object emerge as potential important theranostic tools.

Abstract

A new approach to the synthesis of multifunctional nanoparticles was developed by using covalent anchoring of cyano-bridged coordination polymer Ni2+/[Fe(CN)6]3− to the surface of two-photon dye-doped mesoporous silica nanoparticles. The obtained hybrid nanoparticles were studied by infrared (IR) spectroscopy, nitrogen adsorption (BET), X-ray diffraction, transmission electron microscopy (TEM), high resolution transmission electron microscopy (HRTEM), luminescence, and magnetic analysis. The synthesis leads to homogeneously dispersed uni-shaped nanoparticles of around 100 nm in length that are coated with cyano-bridged metallic coordination polymer nanoparticles. These hybrid nanoparticles combine effective two-photon excited fluorescence, porosity, high transverse nuclear relaxivity values (i.e. the magnetic resonance imaging efficiency) and superparamagnetic properties.

Abstract

International audience; The synthesis of silicalites and Mesoporous Silica Nanoparticles (MSN), which covalently incorporate original water-soluble photosensitizers for PDT applications is described. PDT was performed on MDA-MB-231 breast cancer cells. All the nanoparticles showed significant cell death after irradiation, which was not correlated with 1O2 quantum yield of the nanoparticles. Other parameters are involved and in particular the surface and shape of the nanoparticles which influence the pathway of endocytosis. Functionalization with mannose was necessary to obtain the best results with PDT due to an active endocytosis of mannose-functionalized nanoparticles. The quantity of mannose on the surface should be carefully adjusted as a too high amount of mannose impairs the phototoxicity of the nanoparticles. Fluorescein was also encapsulated in MCM-41 type MSN in order to localize the nanoparticles in the organelles of the cells by confocal microscopy. The MSN were localized in lysosomes after active endocytosis by mannose receptors.

Abstract

New nanotools for the imaging of cancer cells have been synthesized. Two-photon dye-doped 3-aminopropyltriethoxysilane-grafted mesoporous silica nanoparticles (MSN) have been grafted with folic acid (FA) functionalized PEG groups. Amine-PEG groups were first reacted with an activated ester derivative of FA. A mixture of FA- and hydroxyl-PEG has then been reacted with the amino groups at the surface of the particles. Cell culture experiments performed with MCF7 and HeLa cancer cells demonstrated that these functionalized MSN showed a low cytotoxicity even after a 24 hours incubation time at high concentrations. These modified MSN are promising for applications in the field of two-photon imaging and their potentiality for photodynamic therapy is currently being investigated.

Abstract

The chemical attachment of enantiomerically pure trianglamines to silicon wafers through isocyanate functions was performed. The self-assembled monolayer was analyzed by ATR-FTIR, ellipsometry, AFM, contact angle measurements. An AFM tip was functionalized by enantiomerically pure trianglamine using the same pathway. Chemical force microscopy showed chiral recognition between the functionalized tip and the functionalized wafer.

Abstract

The synthesis and functionalisation of zeolite nanoparticles designed for laser-polarized 129Xe NMR experiments in solution are described. These nanoparticles were functionalized by using original synthesis pathways through semicarbazide COCHO chemistry in order to ensure anchoring of a peptide for targeting biological receptors and attachment of PEG chains for in vivo experiments. Results demonstrated that accessibility of the dissolved noble gas to the micropores was maintained after functionalization. γ Scintigraphy with 111In linked to the zeolite particles has been investigated in order to follow the behaviour of the zeolite nanoparticles in mice. These preliminary 111In scintigraphy experiments showed the localization of the nanoparticles after injection in mice and their biodistribution, as a first proof-of-concept towards in vivo129Xe MRI.
