Abstract: LAM S5 is a polysulphated derivative of the glucan laminarian that inhibits basic fibroblast growth factor (bFGF) binding and the bFGF-stimulated proliferation of fetal bovine heart endothelial (FBHE) cells. This report demonstrates that LAM S5 has anti-angiogenic activity, as shown by inhibition of tubule formation by endothelial cells cultured on Matrigel and inhibition of vascularisation of the chick chorioallantoic membrane. In addition, LAM S5 caused a tumour growth delay of the murine RIF-1 tumour of 2.6 days (P = 0.01). Images Figure 2

Abstract: WOS: 000207179200012, PubMed ID: 15841263, The essential oil of Betula pendula Roth. buds was obtained using both hydrodistillation and microdistillation techniques and their chemical compositions were analyzed using both gas chromatography ( GC) and gas chromatography-mass spectrometry (GC-MS). Overall, more than 50 compounds were identified representing 80% and 92% for hydrodistillation and microdistillation, respectively. The main components ( by hydrodistillation and microdistillation, respectively) found were alpha-copaene (12% and 10%), germacrene D (11% and 18%) and delta-cadinene (11% and 15%) in the analyzed essential oils. The microdistillation technique proved to be a useful tool and compliant alternative when compared to hydrodistillation.

Abstract: OBJECTIVES This study sought to further understand the mechanisms underlying effect of spironolactone and assessed its impact on multiple plasma protein biomarkers and their respective underlying biologic pathways.BACKGROUND In addition to their beneficial effects in established heart failure (HF), mineralocorticoid receptor antagonists may act upstream on mechanisms, preventing incident HF. In people at risk for developing HF, the HOMAGE (Heart OMics in AGEing) trial showed that spironolactone treatment could provide antifibrotic and antiremodeling effects, potentially slowing the progression to HF.METHODS Baseline, 1-month, and 9-month (or last visit) plasma samples of HOMAGE participants were measured for protein biomarkers (n = 276) by using Olink Proseek-Multiplex cardiovascular and inflammation panels (Olink, Uppsala, Sweden). The effect of spironolactone on biomarkers was assessed by analysis of covariance and explored by knowledgebased network analysis. RESULTS A total of 527 participants were enrolled; 265 were randomized to spironolactone (25 to 50 mg/day) and 262 to standard care ("control"). The median (interquartile range) age was 73 years (69 to 79 years), and 26% were female. Spironolactone reduced biomarkers of collagen metabolism (e.g., COL1A1, MMP-2); brain natriuretic peptide; and biomarkers related to metabolic processes (e.g., PAPPA), inflammation, and thrombosis (e.g., IL17A, VEGF, and urokinase). Spironolactone increased biomarkers that reflect the blockade of the mineralocorticoid receptor (e.g., renin) and increased the levels of adipokines involved in the anti-inflammatory response (e.g., RARRES2) and biomarkers of hemostasis maintenance (e.g., tPA, UPAR), myelosuppressive activity (e.g., CCL16), insulin suppression (e.g., RETN), and inflammatory regulation (e.g., IL-12B).CONCLUSIONS Proteomic analyses suggest that spironolactone exerts pleiotropic effects including reduction in fibrosis, inflammation, thrombosis, congestion, and vascular function improvement, all of which may mediate cardiovascular protective effects, potentially slowing progression toward heart failure. Abstract: Udgivelsesdato: 2008-Feb-14 In contrast to some extensively examined food mutagens, for example, aflatoxins, N-nitrosamines and heterocyclic amines, some other food contaminants, in particular polycyclic aromatic hydrocarbons (PAH) and other aromatic compounds, have received less attention. Therefore, exploring the relationships between dietary habits and the levels of biomarkers related to exposure to aromatic compounds is highly relevant. We have investigated in the European Prospective Investigation into Cancer and Nutrition (EPIC) cohort the association between dietary items (food groups and nutrients) and aromatic DNA adducts and 4-aminobiphenyl-Hb adducts. Both types of adducts are biomarkers of carcinogen exposure and possibly of cancer risk, and were measured, respectively, in leucocytes and erythrocytes of 1086 (DNA adducts) and 190 (Hb adducts) non-smokers. An inverse, statistically significant, association has been found between DNA adduct levels and dietary fibre intake (P = 0.02), vitamin E (P = 0.04) and alcohol (P = 0.03) but not with other nutrients or food groups. Also, an inverse association between fibre and fruit intake, and BMI and 4-aminobiphenyl-Hb adducts (P = 0.03, 0.04, and 0.03 respectively) was observed. After multivariate regression analysis these inverse correlations remained statistically significant, except for the correlation adducts v. fruit intake. The present study suggests that fibre intake in the usual range can modify the level of DNA or Hb aromatic adducts, but such role seems to be quantitatively modest. Fibres could reduce the formation of DNA adducts in different manners, by diluting potential food mutagens and carcinogens in the gastrointestinal tract, by speeding their transit through the colon and by binding carcinogenic substances.

No. of bitstreams: 0 Previous issue date: 2022-01-19 Deep convection overshooting the lowermost stratosphere is well known for its role in the local stratospheric water vapour (WV) budget. While it is seldom the case, local enhancement of WV associated with stratospheric overshoots is often published. Nevertheless, one debatable topic persists regarding the global impact of this event with respect to the temperature-driven dehydration of air parcels entering the stratosphere. As a first step, it is critical to quantify their role at a cloud-resolving scale before assessing their impact on a large scale in a climate model. It would lead to a nudging scheme for large-scale simulation of overshoots. This paper reports on the local enhancements of WV linked to stratospheric overshoots, observed during the TRO-Pico campaign conducted in March 2012 in Bauru, Brazil, using the BRAMS (Brazilian version of the Regional Atmospheric Modeling System; RAMS) mesoscale model. Since numerical simulations depend on the choice of several preferred parameters, each having its uncertainties, we vary the microphysics or the vertical resolution while simulating the overshoots. Thus, we produce a set of simulations illustrating the possible variations in representing the stratospheric overshoots. To better resolve the stratospheric hydration, we opt for simulations with the 800gm horizontal-grid-point presentation. Next, we validate these simulations against the Bauru S-band radar echo tops and the TRO-Pico balloon-borne observations of WV and particles. Two of the three simulations' setups yield results compatible with the TRO-Pico observations. From these two simulations, we determine approximately 333-2000gt of WV mass prevailing in the stratosphere due to an overshooting plume depending on the simulation setup. About 70g% of the ice mass remains between the 380 and 385gK isentropic levels. The overshooting top comprises pristine ice and snow, while aggregates only play a role just above the tropopause. Interestingly, the horizontal cross section of the overshooting top is about 450gkm2 at the 380gK isentrope, which is similar to the horizontal-grid-point resolution of a simulation that cannot compute overshoots explicitly. In a large-scale simulation, these findings could provide guidance for a nudging scheme of overshooting hydration or dehydration. Abstract: Cytokinesis requires the constriction of ESCRT-III filaments on the side of the midbody, where abscission occurs. After ESCRT recruitment at the midbody, it is not known how the ESCRT-III machinery localizes to the abscission site. To reveal actors involved in abscission, we obtained the proteome of intact, post-abscission midbodies (Flemmingsome) and identified 489 proteins enriched in this organelle. Among these proteins, we further characterized a plasma membrane-to-ESCRT module composed of the transmembrane proteoglycan syndecan-4, ALIX and syntenin, a protein that bridges ESCRT-III/ALIX to syndecans. The three proteins are highly recruited first at the midbody then at the abscission site, and their depletion delays abscission. Mechanistically, direct interactions between ALIX, syntenin and syndecan-4 are essential for proper enrichment of the ESCRT-III machinery at the abscission site, but not at the midbody. We propose that the ESCRT-III machinery must be physically coupled to a membrane protein at the cytokinetic abscission site for efficient scission, uncovering common requirements in cytokinesis, exosome formation and HIV budding., ESCRT filaments drive the final abscission between two daughter cells but how they physically interact with the membrane is unclear. Using proteomics, the authors show that syndecan-4/syntenin/ALIX couples the ESCRT machinery to the abscission site and thus promotes efficient abscission.

Abstract: Free tropospheric ozone (O3) trends in the Central East China (CEC) and export regions are investigated for 2008–2017 using the IASI (Infrared Atmospheric Sounding Interferometer) O3 observations and the LMDZ-OR-INCA model simulations, including the most recent Chinese emission inventory. The observed and modelled trends in the CEC region are −0.07 ± 0.02 and −0.08 ± 0.02 DU yr−1, respectively, for the lower free troposphere (3–6 km column) and −0.05 ± 0.02 and −0.06 ± 0.02 DU yr−1, respectively, for the upper free troposphere (6–9 km column). The statistical p value is smaller to 0.01 for all the derived trends. A good agreement between the observations and the model is also observed in the region, including the Korean Peninsula and Japan and corresponding to the region of pollution export from China. Based on sensitivity studies conducted with the model, we evaluate, at 60 % and 52 %, the contribution of the Chinese anthropogenic emissions to the trend in the lower and upper free troposphere, respectively. The second main contribution to the trend is the meteorological variability (34 % and 50 %, respectively). These results suggest that the reduction in NOx anthropogenic emissions that has occurred since 2013 in China led to a decrease in ozone in the Chinese free troposphere, contrary to the increase in ozone at the surface. We designed some tests to compare the trends derived by the IASI observations and the model to independent measurements, such as the In-service Aircraft for a Global Observing System (IAGOS) or other satellite measurements (Ozone Monitoring Instrument (OMI)/Microwave Limb Sounder (MLS)). These comparisons do not confirm the O3 decrease and stress the difficulty in analysing short-term trends using multiple data sets with various sampling and the risk of overinterpreting the results.

Abstract: Refractory pollutants often exist in wastewater after photocatalysis. Here, experiments, gas chromatography-mass spectrometry (GC–MS) analysis and density functional theory (DFT) simulations were used to explain the different levels of photocatalytic degradation of organics in papermaking wastewater associated with two photocatalytic processes with the goal of improving environmental sustainability. The photocatalytic experiments of flocculation and SBR effluents of papermaking wastewater were studied in a reactor with nano-TiO2 colloid, followed by GC–MS analysis, and the different degradation levels of organics were explained based on DFT adsorption and photocatalysis simulations. The experimental results show that the total COD removal rates were 65.8% and 90.8% for the flocculation-photocatalysis and SBR-photocatalysis processes, and the resin acids and fatty acids in wastewater were classified as easily degradable pollutants and esters as refractory pollutants. The adsorption and photocatalysis simulations show that compared with dehydroabietic acid and pentadecanoic acid, the adsorption of glycerol monostearate by nano-TiO2 colloid is difficult, the reaction pathways for photocatalysis are complex and the band gap of TiO2 photocatalyst after adsorption is wide. The experimental and simulation results indicate that the different degradation levels are attributed to the difficulty of the adsorption and photocatalytic reactions of glycerol monostearate compared with the reactions of the other two easily degradable pollutants. Thus, the different photocatalytic degradation levels of organic pollutants in the two photocatalytic processes of papermaking wastewater indicate that sustainable photocatalytic technology could be applied to different types of industrial wastewater.

Abstract: International audience; ConspectusThe chemical study of materials from natural history and cultural heritage, which provide information for art history, archeology, or paleontology, presents a series of specific challenges. The complexity of these ancient and historical materials, which are chemically heterogeneous, the product of alteration processes, and inherently not reproducible, is a major obstacle to a thorough understanding of their making and long-term behavior (e.g., fossilization). These challenges required the development of methodologies and instruments coupling imaging and data processing approaches that are optimized for the specific properties of the materials. This Account discusses how these characteristics not only constrain their study but also open up specific innovative avenues for providing key historical information. Synchrotron methods have extensively been used since the late 1990s to study heritage objects, in particular for their potential to provide speciation information from excitation spectroscopies and to image complex heritage objects and samples in two and three dimensions at high resolution. We examine in practice how the identification of key intrinsic chemical specificities has offered fertile ground for the development of novel synchrotron approaches allowing a better stochastic description of the properties of ancient and historical materials. These developments encompass three main aspects: (1) The multiscale heterogeneity of these materials can provide an essential source of information in the development of probes targeting their multiple scales of homogeneity. (2) Chemical alteration can be described in many ways, e.g., by segmenting datasets in a semiquantitative way to jointly inform morphological and chemical transformation pathways. (3) The intrinsic individuality of chemical signatures in artifacts triggers the development of specific strategies, such as those focusing on weak signal detection. We propose a rereading of the advent of these new methodologies for analysis and characterization and examine how they have led to innovative strategies combining materials science, instrument development, history, and data science. In particular, we show that spectral imaging and the search for correlations in image datasets have provided a powerful way to address what archeologists have called the uncertainty and ambiguity of the material record. This approach has implications beyond synchrotron techniques and extends in particular to a series of rapidly developing approaches that couple spectral and spatial information, as in hyperspectral imaging and spatially resolved mass spectrometry. The preeminence of correlations holds promise for the future development of machine learning methods for processing data on historical objects. Beyond heritage, these developments are an original source of inspiration for the study of materials in many related fields, such as environmental, geochemical, or life sciences, which deal with systems whose alteration and heterogeneity cannot be neglected.

Abstract: Background Multidrug-resistant (MDR) Mycobacterium tuberculosis complex strains not detected by commercial molecular drug susceptibility testing (mDST) assays due to the RpoB I491F resistance mutation are threatening the control of MDR tuberculosis (MDR-TB) in Eswatini. Methods We investigate the evolution and spread of MDR strains in Eswatini with a focus on bedaquiline (BDQ) and clofazimine (CFZ) resistance using whole-genome sequencing in two collections ((1) national drug resistance survey, 2009–2010; (2) MDR strains from the Nhlangano region, 2014–2017). Results MDR strains in collection 1 had a high cluster rate (95%, 117/123 MDR strains) with 55% grouped into the two largest clusters (gCL3, n = 28; gCL10, n = 40). All gCL10 isolates, which likely emerged around 1993 (95% highest posterior density 1987–1998), carried the mutation RpoB I491F that is missed by commercial mDST assays. In addition, 21 (53%) gCL10 isolates shared a Rv0678 M146T mutation that correlated with elevated minimum inhibitory concentrations (MICs) to BDQ and CFZ compared to wild type isolates. gCL10 isolates with the Rv0678 M146T mutation were also detected in collection 2. Conclusion The high clustering rate suggests that transmission has been driving the MDR-TB epidemic in Eswatini for three decades. The presence of MDR strains in Eswatini that are not detected by commercial mDST assays and have elevated MICs to BDQ and CFZ potentially jeopardizes the successful implementation of new MDR-TB treatment guidelines. Measures to limit the spread of these outbreak isolates need to be implemented urgently.

No. of bitstreams: 0 Previous issue date: 2020-11-01 Clonal plantations of Eucalyptus are among the most productive forests in the world, with intensification of silviculture and genetic breeding doubling the wood mean annual increments over the past four decades. The TECHS Project demonstrated that even with intensive silviculture, wood production varies by more than two-fold across environmental gradients, and growth of highly selected clones differs by more than two-fold within a site. Wood production accounts for less than half of the photosynthesis of a forest, and we tested two hypotheses about the relation between wood production and the entire carbon balance for five genotypes across four of the TECHS sites, varying in temperature and water availability. We hypothesized that the influence of the environment on carbon fluxes and partitioning related to gross primary production would be consistent across genotypes. We also hypothesized that carbon flux and partitioning would be more sensitive to water stress than temperature. Annual average temperatures ranged from 18 to 27 °C, and annual rainfall ranged from about 600 to 1500 mm yr−1. Water stress was further tested by reduction in rainfall within sites using troughs to capture about 30% of incoming rain. The geographic gradient led to a six-fold range in wood net primary production during the two years of measurement (from age 1.5 to 3.5 years, the period of maximum current annual increment). Gross primary production (GPP) differed only by two-fold, highlighting very large differences among sites in partitioning: wood net primary production (NPP) accounted for 44% of GPP on sites with higher GPP, and only 34% of GPP on lower GPP sites. The average differences for wood NPP among clones was also large, with about half of the differences among clones relating to differences in GPP, and half to differences in the partitioning to wood NPP. The clones showed similar partitioning patterns across sites, supporting our first hypothesis. Differences across sites and clones in partitioning of GPP to wood NPP related inversely to belowground allocation. Belowground partitioning of carbon increased with increasing temperature and increasing water stress. Our second hypothesis was rejected, as patterns across sites related somewhat more strongly to temperature than to water stress. Overall, this ecophysiological investigation in the TECHS Project underscored the importance of understanding how carbon budgets differ across sites (even with intensive silviculture), and why clones can largely differ in wood production. Abstract: In this work, a multi-scale model established from the concept of periodic homogenization is utilized to predict the cyclic and time-dependent response of thermoplastic-based woven composites. The macroscopic behaviour of the composite is determined from finite element simulations of the representative unit cell of the periodic microstructure, where the local non-linear constitutive laws of the components are directly integrated, namely, the matrix and the yarns. The thermoplastic matrix is described by a phenomenological multi-mechanisms constitutive model accounting for viscoelasticity, viscoplasticity and ductile damage. For the yarns, a hybrid micromechanical–phenomenological constitutive model accounting for anisotropic damage and anelasticity induced by the presence of a diffuse micro-crack network is utilized. The capabilities of the overall multi-scale model are validated by comparing the numerical predictions with experimental data. Further illustrative examples are also provided, where the composite undergoes time-dependent deformations under uni-axial and non-proportional multi-axial loading paths. The multi-scale model is also employed to analyze the influence of the local deformation processes on the macroscopic response of the composite. Abstract: International audience; Despite its major role as a global climate regulator, little is known about the recent evolution of the North Atlantic Ocean, especially prior to the fifties, principally because of the lack of long-term instrumental data. Moreover, the North Atlantic Ocean is undergoing rapid changes at the physical scale leading to modifications at the biological scale increasing interest to monitor the environment. The phytoplankton is the base of the ocean life and its perturbation can lead to further changes in the food chain. Recording its dynamics implies to observe both its spatial and temporal variations. This study brings forward the use of the Ba/Ca ratio from shells of the long-living bivalve Arctica islandica collected in the Northwestern Atlantic Ocean near Saint-Pierre and Miquelon (SPM) as a recorder of the past and present phytoplankton dynamics. A high inter and intra-reproducibility and synchrony between Ba/Cashell profiles were demonstrated allowing the reconstruction of a 124 years annually-resolved Ba/Cashell master chronology (from 1893 to 2016). This master chronology was positively correlated to the surface chlorophyll a measured by satellite sensor at regional (around SPM) and global scales (Northwestern Atlantic).

Abstract: International audience; This paper focuses on hydrogen production for green mobility applications (other applications are currently under investigation). Firstly, a brief state of the art of hydrogen generation by hydrolysis with magnesium is shown. The hydrolysis performance of Magnesium powder ball–milled along with different additives (graphite and transition metals TM = Ni, Fe, and Al) is taken for comparison. The best performance was observed with Mg–10 wt.% g mixtures (95% of theoretical hydrogen generation yield in about 3 min). An efficient solution to control this hydrolysis reaction is proposed to produce hydrogen on demand and to feed a PEM fuel cell. Tests on a bench fitted with a 100 W Proton Exchange Membrane (PEM) fuel cell have demonstrated the technological potential of this solution for electric assistance applications in the field of light mobility.

Abstract: International audience; Purpose: To describe retinal alterations detected by swept-source optical coherence tomography (SS-OCT) in paediatric patients with Usher syndrome type 1 (USH1) and to compare these findings to previously published reports.Methods: Thirty-two eyes from 16 patients (11 males and 5 females) with a genetic diagnosis of USH1 because of MYO7A mutations underwent SS-OCT. Patients ranged in age from 4 to 17 years (mean, 11,13 ± 4,29). The subfoveal and macular area were analysed with SS-OCT at 1050 nm using 12 radial scans of 12.0 mm. Structural abnormalities were evaluated and correlated with best-corrected visual acuity (BCVA).Results: The most common qualitative retinal abnormality was external layer damage in macular area. Specific alterations included external limiting membrane loss/disruption (27 eyes; 84.4%), disruption of the Myoid zone (27 eyes; 84.4%); Ellipsoid zone disruption (28 eyes; 87.5%), and loss of the outer segments (29 eyes; 90.6%). The damage of the retinal pigment epithelium was divided according to the loss of the different layers: phagosome zone (30 eyes; 93.8%), melanosome zone (29 eyes; 90.6%) and mitochondria zone (0 eyes; 0%). The presence of cystoid macular oedema (CMO) was significantly correlated with alterations in photoreceptors. Disruption or absence of the myoid and ellipsoid zones of the photoreceptors were the only variables independently associated with decreased BCVA.Conclusions: The findings of this study suggest that the physiopathologic basis of early-stage Usher syndrome (USH) may be changes in the outer retinal layer, particularly the photoreceptors, which in turn may cause alterations-such as CMO-in the inner retinal layers. Accordingly, monitoring the condition of photoreceptors during follow-up may be advisable for the early detection of pathologic changes.

Abstract: Multiple studies have suggested that ω-3 fatty acid intake may have a protective effect on cancer risk; however, its true association with cancer risk remains controversial. We performed an umbrella review of meta-analyses to summarize and evaluate the evidence for the association between ω-3 fatty acid intake and cancer outcomes. We searched PubMed, Embase, and the Cochrane Database of Systematic Reviews from inception to December 1, 2018. We included meta-analyses of observational studies that examined associations between intake of fish or ω-3 fatty acid and cancer risk (gastrointestinal, liver, breast, gynecologic, prostate, brain, lung, and skin) and determined the level of evidence of associations. In addition, we appraised the quality of the evidence of significant meta-analyses by using the Grading of Recommendations Assessment, Development and Evaluation (GRADE) system. We initially screened 598 articles, and 15 articles, including 57 meta-analyses, were eligible. Among 57 meta-analyses, 15 reported statistically significant results. We found that 12 meta-analyses showed weak evidence of an association between ω-3 fatty acid intake and risk of the following types of cancer: liver cancer (n = 4 of 6), breast cancer (n = 3 of 14), prostate cancer (n = 3 of 11), and brain tumor (n = 2 of 2). In the other 3 meta-analyses, studies of endometrial cancer and skin cancer, there were no assessable data for determining the evidence levels. No meta-analysis showed convincing, highly suggestive, or suggestive evidence of an association. In the sensitivity analysis of meta-analyses by study design, we found weak associations between ω-3 fatty acid intake and breast cancer risk in cohort studies, but no statistically significant association in case-control studies. However, the opposite results were found in case of brain tumor risk. Although ω-3 fatty acids have been studied in several meta-analyses with regard to a wide range of cancer outcomes, only weak associations were identified in some cancer types, with several limitations. Considering the nonsignificant or weak evidence level, clinicians and researchers should cautiously interpret reported associations between ω-3 fatty acid consumption and cancer risks. Abstract: Forest production efficiency (FPE) metric describes how efficiently the assimilated carbon is partitioned into plants organs (biomass production, BP) or—more generally—for the production of organic matter (net primary production, NPP). We present a global analysis of the relationship of FPE to stand-age and climate, based on a large compilation of data on gross primary production and either BP or NPP. FPE is important for both forest production and atmospheric carbon dioxide uptake. We find that FPE increases with absolute latitude, precipitation and (all else equal) with temperature. Earlier findings—FPE declining with age—are also supported by this analysis. However, the temperature effect is opposite to what would be expected based on the short-term physiological response of respiration rates to temperature, implying a top-down regulation of carbon loss, perhaps reflecting the higher carbon costs of nutrient acquisition in colder climates. Current ecosystem models do not reproduce this phenomenon. They consistently predict lower FPE in warmer climates, and are therefore likely to overestimate carbon losses in a warming climate., Many models assume a universal carbon use efficiency across forest biomes, in contrast to assumptions of other process-based models. Here the authors analyse forest production efficiency across a wide range of climates to show a positive relationship with annual temperature and precipitation, indicating that ecosystem models are overestimating forest carbon losses under warming.

Abstract: Cellulose nanofiber films (CNFF) were treated via a welding process using ionic liquids (ILs). Acid-base-conjugated ILs derived from 1,5-diazabicyclo[4.3.0]non-5-ene [DBN] and 1-ethyl-3-methylimidazolium acetate ([emim][OAc]) were utilized. The removal efficiency of ILs from welded CNFF was assessed using liquid-state nuclear magnetic resonance (NMR) spectroscopy and Fourier transform infrared spectroscopy (FTIR). The mechanical and physical properties of CNFF indicated surface plasticization of CNFF, which improved transparency. Upon treatment, the average CNFF toughness increased by 27%, and the films reached a Young's modulus of ∼5.8 GPa. These first attempts for IL "welding" show promise to tune the surfaces of biobased films, expanding the scope of properties for the production of new biobased materials in a green chemistry context. The results of this work are highly relevant to the fabrication of CNFFs using ionic liquids and related solvents. acceptedVersion

Abstract: A thorough understanding of the heritability, genetic correlations and additive and non-additive variance components of tree growth and wood properties is a requisite for effective tree breeding. This knowledge is essential to maximize genetic gain, that is, the amount of increase in trait performance achieved annually through directional selection. Understanding the genetic attributes of traits targeted by breeding is also important to sustain decade-long genetic progress, that is, the progress made by increasing the average genetic value of the offspring as compared to that of the parental generation. In this study, we report quantitative genetic parameters for fifteen growth, wood chemical and physical traits for the world-famous Eucalyptus urograndis hybrid (E. grandis × E. urophylla). These traits directly impact the optimal use of wood for cellulose pulp, paper, and energy production. A population of 1,000 trees sampled in a progeny trial was phenotyped directly or following the development and use of near-infrared spectroscopy calibration models. Trees were genotyped with 33,398 SNPs and 24,001 DArT-seq genome-wide markers and genomic realized relationship matrices (GRM) were used for parameter estimation with an individual-tree additive-dominant mixed model. Wood chemical properties and wood density showed stronger genetic control than growth, cellulose and fiber traits. Additive effects are the main drivers of genetic variation for all traits, but dominance plays an equally or more important role for growth, singularly in this hybrid. GRM´s with >10,000 markers provided stable relationships estimates and more accurate parameters than pedigrees by capturing the full genetic relationships among individuals and disentangling the non-additive from the additive genetic component. Low correlations between growth and wood properties indicate that simultaneous selection for wood traits can be applied with minor effects on genetic gain for growth. Conversely, moderate to strong correlations between wood density and chemical traits exist, likely due to their interdependency on cell wall structure such that responses to selection will be connected for these traits. Our results illustrate the advantage of using genomewide marker data to inform tree breeding in general and have important consequences for operational breeding of eucalypt urograndis hybrids. Abstract: Recent years have seen an increased interest towards utilizing bio-based and biodegradable materials for barrier packaging applications. Most of the above said materials usually have certain shortcomings that discourage their adoption as a preferred material of choice. Nanocellulose falls into such category. It has excellent barrier against grease, mineral oils and oxygen, but poor tolerance against water vapor, which makes it unsuitable to be used at high humidity. In addition, nanocellulose suspensions' high viscosity and yield stress already at low solids content, and poor adhesion to substrates create additional challenges for high-speed processing. Polylactic acid (PLA) is another potential candidate that has a reasonably high tolerance against water vapor, but rather poor barrier against oxygen. The current work explores the possibility to combine both these materials into thin multilayer coatings onto paperboard. A custom-built slot-die was used to coat either microfibrillated cellulose (MFC) or cellulose nanocrystals (CNCs) onto pigment-coated baseboard in a continuous process. These were subsequently coated with PLA using a pilot scale extrusion coater. Low-density polyethylene (LDPE) was used as a reference extrusion coating. Cationic starch pre-coating and corona treatment improved the adhesion at nanocellulose/baseboard and nanocellulose/PLA interfaces, respectively. Water vapor transmission rate for nanocellulose + PLA coatings remained lower than the control PLA coating, even at a high relative humidity of 90% (38 oC). The multilayer coating had 98% lower oxygen transmission rate compared to just PLA coated baseboard and heptane vapor transmission rate reduced by 99% in comparison to baseboard. Grease barrier for nanocellulose + PLA coatings increased 5-fold compared to nanocellulose alone and 2-fold compared to PLA alone. This approach of processing nanocellulose and PLA into multiple layers utilizing slot-die and extrusion coating in tandem has the potential to produce a barrier packaging paper that is both 100% bio-based and biodegradable. publishedVersion

Abstract: International audience; Metal-dependent histone deacetylases (HDACs) are key epigenetic regulators that represent promising therapeutic targets for the treatment of numerous human diseases. Yet the currently FDA-approved HDAC inhibitors nonspecifically target at least several of the 11 structurally similar but functionally different HDAC isozymes, which hampers their broad usage in clinical settings. Selective inhibitors targeting single HDAC isozymes are being developed, but precise understanding in molecular terms of their selectivity remains sparse. Here, we show that HDAC8-selective inhibitors adopt a L-shaped conformation required for their binding to a HDAC8-specific pocket formed by HDAC8 catalytic tyrosine and HDAC8 L1 and L6 loops. In other HDAC isozymes, a L1–L6 lock sterically prevents L-shaped inhibitor binding. Shielding of the HDAC8-specific pocket by protein engineering decreases potency of HDAC8-selective inhibitors and affects catalytic activity. Collectively, our results unravel key HDAC8 active site structural and functional determinants important for the design of next-generation chemical probes and epigenetic drugs.

Abstract: The development of new biocompatible, biodegradable functionalized biopolymers that can serve as scaffold for tissue regeneration or work as carriers for different bioactive molecules such as drugs, proteins, and enzymes remains a continuous challenge that need to be extensively explored. For this purpose, three water-soluble cellulose derivatives; namely 4(celluloseamino) butyric acid (CABA) 2(celluloseamino) succinic acid (CASA), and 3(celluloseamino) propane sulfonic acids (CAPSA) were synthesized from microcrystalline cellulose (MCC) via esterification with tosyl chloride that was followed by nucleophilic substitution by the proper aminoalkyl acid derivative. The products were characterized by elemental analyses, FTIR, 13C NMR spectroscopy. The thermal stability, surface morphology, and the elemental composition of the new ampholytic biopolymers were also studied by TGA, EDX-SEM. The new ampholytic cellulose derivatives were evaluated for their in vitro cytotoxicity on normal human retina cell line (RPE1) by MTT assay.

Abstract: International audience; Pentameric ligand-gated ion channels (pLGICs) constitute a widespread class of ion channels, present in archaea, bacteria, and eukaryotes. Upon binding of their agonists in the extracellular domain, the transmembrane pore opens, allowing ions to go through, via a gating mechanism that can be modulated by a number of drugs. Even though high-resolution structural information on pLGICs has increased in a spectacular way in recent years, both in bacterial and in eukaryotic systems, the structure of the open channel conformation of some intensively studied receptors whose structures are known in a nonactive (closed) form, such as Erwinia chrysanthemi pLGIC (ELIC), is still lacking. Here we describe a gammaproteobacterial pLGIC from an endo-symbiont of Tevnia jerichonana (sTeLIC), whose sequence is closely related to the pLGIC from ELIC with 28% identity. We provide an X-ray crystallographic structure at 2.3 Å in an active conformation, where the pore is found to be more open than any current conformation found for pLGICs. In addition, two charged restriction rings are present in the vestibule. Functional characterization shows sTeLIC to be a cationic channel activated at alkaline pH. It is inhibited by divalent cations, but not by quaternary ammonium ions, such as tetramethylammonium. Additionally, we found that sTeLIC is allosterically potentiated by aromatic amino acids Phe and Trp, as well as their derivatives, such as 4-bromo-cinnamate, whose cocrystal structure reveals a vestibular binding site equivalent to, but more deeply buried than, the one already described for benzodiazepines in ELIC.

Abstract: Concentrations of nine organophosphate flame retardants (OPFRs) and eight polybrominated diphenyl ethers (PBDEs) were measured in samples of indoor dust (n = 85) and air (n = 45) from Australian houses, offices, hotels, and transportation (buses, trains, and aircraft). All target compounds were detected in indoor dust and air samples. Median ∑9OPFRs concentrations were 40 μg/g in dust and 44 ng/m3 in indoor air, while median ∑8PBDEs concentrations were 2.1 μg/g and 0.049 ng/m3. Concentrations of FRs were higher in rooms that contained carpet, air conditioners, and various electronic items. Estimated daily intakes in adults are 14000 pg/kg body weight/day and 330 pg/kg body weight/day for ∑9OPFRs and ∑8PBDEs, respectively. Our results suggest that for the volatile FRs such as tris(2-chloroethyl) phosphate (TCEP) and TCIPP, inhalation is expected to be the more important intake pathway compared to dust ingestion and dermal contact.

Abstract: Although eucalypts are the most planted hardwood trees worldwide, the majority of them are frost sensitive. The recent creation of frost-tolerant hybrids such as Eucalyptus gundal plants (E. gunnii × E. dalrympleana hybrids), now enables the development of industrial plantations in northern countries. Our objective was to evaluate the impact of cold on the wood structure and composition of these hybrids, and on the biosynthetic and regulatory processes controlling their secondary cell-wall (SCW) formation. We used an integrated approach combining histology, biochemical characterization and transcriptomic profiling as well as gene co-expression analyses to investigate xylem tissues from Eucalyptus hybrids exposed to cold conditions. Chilling temperatures triggered the deposition of thicker and more lignified xylem cell walls as well as regulation at the transcriptional level of SCW genes. Most genes involved in lignin biosynthesis, except those specifically dedicated to syringyl unit biosynthesis, were up-regulated. The construction of a co-expression network enabled the identification of both known and potential new SCW transcription factors, induced by cold stress. These regulators at the crossroads between cold signalling and SCW formation are promising candidates for functional studies since they may contribute to the tolerance of E. gunnii × E. dalrympleana hybrids to cold.

Abstract: International audience; Leishmaniasis is a parasitic reticuloendotheliosis whose pathogen is a zooflagellate belonging to the genus Leishmania transmitted by the bite of an infected phlebotome. Recently, a unique secretory lipase from the human pathogen Leishmania donovani Ldlip3 has been identified and characterized. This lipase has a high identity with a putative triacylglycerol lipase of Leishmania major (Lmlip2). In the present study, Lmlip2 was expressed in the eukaryotic heterologous expression system Pichia pastoris as tagged enzyme of 308 amino acids. Maximal protein production was reached after 2 days of fermentation. Optimal Lmlip2 lipase activity was measured using the pH stat technique at pH 8 at 26 °C using vinyl esters and triacylglycerols (true lipids) as substrates. Moreover, biochemical characterization of Lmlip2 contained in culture supernatant, illustrates that L. major secreted lipase is active and stable at low temperatures especially 26°and prefer neutral pH; concerning substrate specificityLmlip2 presents a preference for short chains lipid substrates vinyl esters such as VC2, VC3 and VC4 likewise, it is capable to hydrolyze long chain triacylglycerols like olive oil. Metal ions and surfactants tested in this study decrease Lmlip2 activity. Further studies are needed to clarify the relation between the lipase activity and the virulence. Thus, it could lead to the identification of novel targets to block cutaneous Leishmaniasis in human hosts.

[Proc Natl Acad Sci U S A. 2018]; International audience; Carbon monoxide dehydrogenase/acetyl-CoA synthase (CODH/ ACS) is a five-subunit enzyme complex responsible for the carbonyl branch of the Wood-Ljungdahl (WL) pathway, considered one of the most ancient metabolisms for anaerobic carbon fixation, but its origin and evolutionary history have been unclear. While traditionally associated with methanogens and acetogens, the presence of CODH/ACS homologs has been reported in a large number of un-cultured anaerobic lineages. Here, we have carried out an exhaustive phylogenomic study of CODH/ACS in over 6,400 archaeal and bacterial genomes. The identification of complete and likely functional CODH/ACS complexes in these genomes significantly expands its distribution in microbial lineages. The CODH/ACS complex displays astounding conservation and vertical inheritance over geological times. Rare intradomain and interdomain transfer events might tie into important functional transitions, including the acquisition of CODH/ACS in some archaeal methanogens not known to fix carbon, the tinkering of the complex in a clade of model bacterial acetogens, or emergence of archaeal-bacterial hybrid complexes. Once these transfers were clearly identified, our results allowed us to infer the presence of a CODH/ACS complex with at least four subunits in the last universal common ancestor (LUCA). Different scenarios on the possible role of ancestral CODH/ACS are discussed. Despite common assumptions, all are equally compatible with an autotrophic, mixotrophic, or heterotrophic LUCA. Functional characterization of CODH/ACS from a larger spectrum of bacterial and archaeal lineages and detailed evolutionary analysis of the WL methyl branch will help resolve this issue.

Abstract: International audience; Peroxide sensing is essential for bacterial survival during aerobic metabolism and host infection. Peroxide stress regulators (PerRs) are homodimeric transcriptional repressors with each monomer typically containing both structural and regulatory metal-binding sites. PerR binding to gene promoters is controlled by the presence of iron in the regulatory site, and iron-catalyzed oxidation of PerR by H2O2 leads to the dissociation of PerR from DNA. In addition to a regulatory metal, most PerRs require a structural metal for proper dimeric assembly. We present here a structural and functional characterization of the PerR from the pathogenic spirochete Leptospira interrogans, a rare example of PerR lacking a structural metal-binding site. In vivo studies showed that the leptospiral PerR belongs to the peroxide stimulon in pathogenic species and is involved in controlling resistance to peroxide. Moreover, a perR mutant had decreased fitness in other host-related stress conditions, including at 37 °C or in the presence of superoxide anion. In vitro, leptospiral PerR could bind to the perR promoter region in a metal-dependent manner. The crystal structure of the leptospiral PerR revealed an asymmetric homodimer, with one monomer displaying complete regulatory metal coordination in the characteristic caliper-like DNA-binding conformation and the second monomer exhibiting disrupted regulatory metal coordination in an open non-DNA-binding conformation. This structure showed that leptospiral PerR assembles into a dimer in which a metal-induced conformational switch can occur independently in the two monomers. Our study demonstrates that structural metal binding is not compulsory for PerR dimeric assembly and for regulating peroxide stress.

Abstract: A complèter : pagination et WOS; International audience; Large wood (LW, or pieces of dead wood longer than 1 m and thicker than 10 cm in diameter) is a key element in forested streams, but its abundance has decreased worldwide as a result of snagging and clearing of riparian forests. Therefore, many restoration projects introduce LW into stream channels to enhance geomorphology, biotic communities, and ecosystem functioning. Because LW enhances the retention of organic matter and sediments, its restoration can reduce siltation in receiving reservoirs, although so far little information on this subject is available. We studied the effects of restoring the natural loading of LW in four streams in the Aiako Harria Natural Park (the Basque Country, Spain) in pool abundance, channel form, and storage of organic matter and sediments. In all reaches log jams induced the formation of new geomorphic features and changes in physical habitat, especially an increase in the number and size of pools and in the formation of gravel bars and organic deposits. The storage of organic matter increased 5- to 88-fold and streambed level rose 7 ± 4 to 21 ± 4 cm on average, resulting in the storage of 35.2 ± 19.7 to 711 ± 375 m3 (733–1400 m3 ha− 1 y− 1) of sediment per reach. Extrapolation of these results to the entire drainage basin suggests that basinwide restoration of LW loading would enhance the retention potential of stream channels by 66,817 ± 27,804 m3 (1075 m3 ha− 1 y− 1) of sediment and by 361 t (5.32 T ha− 1 y− 1) of organic matter, which represents 60% of the estimated annual inputs of sediments to the downstream Añarbe Reservoir and almost twice as much as the annual input of organic matter to the entire river network. Therefore, basinwide restoration of LW loading is a potentially important tool to manage catchments that feed reservoirs, where retention of sediments and organic matter can be considered important ecosystem services as they reduce reservoir siltation.

Abstract: International audience; Adding catalyst could improve the yields and qualities of bio-gas and bio-oil, and realize the oriented production. Results showed that the catalytic gas-production capacities of CaO were higher than those of Fe2O3, and the bio-gas yield at 800°C reached a maximum of 35.1%. Because the polar cracking active sites of CaO reduced the activation energy of the pyrolysis reaction and resulted in high catalytic cracking efficiencies. In addition, the quality of bio-oil produced by CaO was superior to that by Fe2O3, although the bio-oil yield of CaO was relatively weak. The light bio-fuel oriented catalytic pyrolysis could be realized when adding different catalysts. At 800°C, CaO was 45% higher than Fe2O3 in aspect of H2 production while Fe2O3 was 103% higher than CaO in aspect of CH4 production. Therefore, CaO was more suitable for H2 production and Fe2O3 was more suitable for CH4 production.

Abstract: The nucleotidyl cyclase toxin ExoY is one of the virulence factors injected by the Pseudomonas aeruginosa type III secretion system into host cells. Inside cells, it is activated by an unknown eukaryotic cofactor to synthesize various cyclic nucleotide monophosphates. ExoY-like adenylate cyclases are also found in Multifunctional-Autoprocessing Repeats-in-ToXin (MARTX) toxins produced by various Gram-negative pathogens. Here we demonstrate that filamentous actin (F-actin) is the hitherto unknown cofactor of ExoY. Association with F-actin stimulates ExoY activity more than 10,000 fold in vitro and results in stabilization of actin filaments. ExoY is recruited to actin filaments in transfected cells and alters F-actin turnover. Actin also activates an ExoY-like adenylate cyclase MARTX effector domain from Vibrio nigripulchritudo. Finally, using a yeast genetic screen, we identify actin mutants that no longer activate ExoY. Our results thus reveal a new sub-group within the class II adenylyl cyclase family, namely actin-activated nucleotidyl cyclase (AA-NC) toxins., The ExoY toxin is injected by Pseudomonas aeruginosa into host cells, where it is activated by an unknown host factor. Here the authors identify such factor as filamentous actin.

Abstract: International audience; OBJECTIVES:Resistance to fluoroquinolones (FQs) in Mycobacterium tuberculosis (Mtb) is mainly due to mutations in DNA gyrase (GyrA2B2), with the most common substitutions located at positions 90 and 94 in GyrA. Two clinical MDR Mtb (MDR-TB) strains harbouring an A90E or D94N substitution in GyrA were found to be surprisingly susceptible to FQs (ofloxacin MIC ≤2 mg/L). We studied the impact of the additional GyrA substitutions found in these strains (T80A and T80A + A90G, respectively) on FQ susceptibility.METHODS:Mutants of interest were generated by site-specific mutagenesis of GyrA alleles. WT and mutant TB DNA gyrase subunits were overexpressed in Escherichia coli and purified, and the in vitro susceptibility to FQs of their DNA supercoiling reaction was studied.RESULTS:IC50s of mutant gyrase complexes bearing GyrA D94N and A90E were 3- to 36-fold higher than WT IC50s, whereas IC50s of gyrase bearing T80A + A90G + D94N and T80A + A90E were close to the WT IC50s.CONCLUSIONS:We demonstrated that substitutions T80A and A90G restore FQ susceptibility when associated with a substitution implicated in high-level FQ resistance. Line probe assay misclassification of MDR-TB strains as pre-XDR or XDR can be corrected by sequence analysis of gyrA.

Abstract: BACKGROUND Circulating tumor cells (CTCs) are biomarkers for noninvasively measuring the evolution of tumor genotypes during treatment and disease progression. Recent technical progress has made it possible to detect and characterize CTCs at the single-cell level in blood. CONTENT Most current methods are based on epithelial cell adhesion molecule (EpCAM) detection, but numerous studies have demonstrated that EpCAM is not a universal marker for CTC detection because it fails to detect both carcinoma cells that undergo epithelial-mesenchymal transition (EMT) and CTCs of mesenchymal origin. Moreover, EpCAM expression has been found in patients with benign diseases. A large proportion of the current studies and reviews about CTCs describe EpCAM-based methods, but there is evidence that not all tumor cells can be detected using this marker. Here we describe the most recent EpCAM-independent methods for enriching, isolating, and characterizing CTCs on the basis of physical and biological characteristics and point out the main advantages and disadvantages of these methods. SUMMARY CTCs offer an opportunity to obtain key biological information required for the development of personalized medicine. However, there is no universal marker of these cells. To strengthen the clinical utility of CTCs, it is important to improve existing technologies and develop new, non–EpCAM-based systems to enrich and isolate CTCs.

Abstract: The study of silicon (Si) isotopes in ocean island basalts (OIB) has the potential to discern between different models for the origins of geochemical heterogeneities in the mantle. Relatively large (several per mil per atomic mass unit) Si isotope fractionation occurs in low-temperature environments during biochemical and geochemical precipitation of dissolved Si, where the precipitate is preferentially enriched in the lighter isotopes relative to the dissolved Si. In contrast, only a limited range (tenths of a per mil) of Si isotope fractionation has been observed from high-temperature igneous processes. Therefore, Si isotopes may be useful as tracers for the presence of crustal material within OIB mantle source regions that experienced relatively low-temperature surface processes in a manner similar to other stable isotope systems, such as oxygen. Characterizing the isotopic composition of the mantle is also of central importance to the use of the Si isotope system as a basis for comparisons with other planetary bodies (e.g., Moon, Mars, asteroids). Here we present the first comprehensive suite of high-precision Si isotope data obtained by MC-ICP-MS for a diverse suite of OIB. Samples originate from ocean islands in the Pacific, Atlantic, and Indian Ocean basins and include representative end-members for the EM-1, EM-2, and HIMU mantle components. On average, Si isotope compositions of OIB are in general agreement with previous estimates for the Si isotope composition of bulk silicate Earth. Nonetheless, some small systematic variations are present; specifically, most HIMU-type (Mangaia; Cape Verde; La Palma, Canary Islands) and Iceland OIB are enriched in the lighter isotopes of Si, consistent with recycled altered oceanic crust and lithospheric mantle in their mantle sources., Comment: 23 pages, 5 figures, 2 tables

Abstract: International audience; Polyhydroxyalkanoates (PHA) are bacterial polyesters usually produced from costly sugars or volatile fatty acids (VFAs). In this work, two processing waters rich in vegetable proteins and reducing sugars, i.e., a mixture of saccharose and stachyose in Leguminous Processing Water (LPW) and a mixture of glucose and fructose in Fruit Processing Water (FPW), were tested as growth medium for PHA production in a two-stage fermentation with a unique marine bacterial species: Halomonas i4786. In preliminary shake flask experiments, it was shown that the two media can effectively support the bacterial growth and the accumulation of PHA (evaluated using Nile Red staining). In batch cultivation mode in a 5-L fermentor, PHA productivities of 1.6 g L−1 and 1.8 g L−1 were further achieved within 72 h, in LPW and FPW respectively. Polymer characterization by Differential Scanning Calorimetry and Steric Exclusion Chromatography indicated that the two substrates led to the biosynthesis of polymers with different chain length, distribution and crystallinity. To summarize, these results show that by-products derived from local agri-food industry can be used as a user-adapted and cost-effective source to produce bio-sourced and biodegradable plastic materials

Abstract: Creep-age-forming of a thick Al-Cu-Li sheet is studied. An industrial stamping press is used to form a double curvature panel at a reduced scale. This forming, which includes several relaxation steps, is modelled using ABAQUS. A material model describing an elasto-viscoplastic behaviour with anisotropy effect has been identified and implemented in ABAQUS using Fortran subroutine. The numerical model is validated by comparing experiments and numerical results in terms of deformed shapes and an improved forming cycle is suggested. Abstract: Anthocyanins, water soluble polyphenols, have been associated with several beneficial health effects. The aim of this study was to determine how the baking process and food matrix affect anthocyanin stability and bioaccessibility in bakery products in order to develop functional foods. Three well known regularly consumed bakery products (buns, breadsticks and biscuits) were enriched with anthocyanin (AC) isolated from grape skin alone or in combination with docosahexaenoic acid (AC + DHA) to reveal knowledge on AC as active ingredients in real food systems rather than pure compounds. Anthocyanin amounts added to the formulations of buns, breadsticks and biscuits were 34 mg per 100 g, 40 mg per 100 g and 37 mg per 100 g, respectively. The effect of processing, storage and the food matrix on AC stability and bioaccessibility was investigated. In addition, the sensory properties of bakery products were evaluated. Breadsticks enriched with AC and AC + DHA received the lowest scores in the pre-screening sensory test. Therefore breadsticks were excluded from further analysis. AC retentions, which were monitored by determination of malvidin 3-O-glucoside, in the bun and biscuit after baking were 95.9% (13.6 mg per 100 g) and 98.6% (15.2 mg per 100 g), respectively. Biscuits and buns enriched only with AC showed significantly higher anthocyanin bioaccessibilities (57.26% and 57.30%, respectively) than the same ones enriched with AC + DHA. AC stability in enriched products stored for 21 days was significantly lower than in products stored for 7 days (p < 0.05). However, this loss can be accepted as negligible since more than 70% of AC was retained in all the products.

Abstract: Maintaining a proper pH level is crucial for successful cell culturing. Mammalian cells are commonly cultured in incubators, where the cell culture medium is saturated with a mixture of air and 5% carbon dioxide (CO2). Therefore, to keep cell culture medium pH in an acceptable level outside these incubators, a suitable CO2 concentration must be dissolved in the medium. However, it can be very difficult to control and measure precisely local concentration levels. Furthermore, possible undesired concentration gradients generated during long-term cell culturing are almost impossible to detect. Therefore, we have developed a computational model to estimate CO2 transport in silicone-based microfluidic devices. An extensive set of experiments was used to validate the finite element model. The model parameters were obtained using suitable measurement set-ups and the model was validated using a fully functional cell cultivation device. The predictions obtained by the simulations show very good responses to experiments. It is shown in this paper how the model helps to understand the dynamics of CO2 transport in silicone-based cell culturing devices possessing different geometries, thus providing cost-effective means for studying different device designs under a variety of experimental conditions without the need of actual testing. Finally, based on the results from the computational model, an alternative strategy for feeding CO2 is proposed to accelerate the system performance such that a faster and more uniform CO2 concentration response is achieved in the area of interest. acceptedVersion

Abstract: A sensitive electrochemical molecularly imprinted sensor was developed for the detection of estradiol, by electropolymerization of p-aminothiophenol functionalized gold nanoparticles in the presence of estradiol as template molecule. The extraction of the template leads to the formation of cavities that are able to recognize and bind estradiol with high affinity. The performance of the developed sensor for the detection of estradiol was investigated by linear sweep voltammetry using a hexacyanoferrate/hexacyanoferrite solution as redox probe. The molecularly imprinted sensor exhibits a broad linear range, between 3.6 fM and 3.6 nM and a limit of quantification of 1.09 fM. Compared to the non-imprinted sensor, the imprinted sensor exhibits high affinity for the binding of estradiol. Moreover, selectivity studies, performed towards binding of testosterone, a hormone with similar chemical structure, proved high sensor selectivity. Furthermore, the molecularly imprinted sensor was applied for the analysis of spiked river samples with good recoveries. Abstract: The performances of carboxylate- and cyclam-modified graphite felt electrodes prepared by different electrografting methods for trace analysis of copper (II) were compared to determine the influence of the immobilization process of the linkers on the sensor properties. The derivatization performed by cathodic reduction of diazonium salts and by anodic oxidation of amines in organic and aqueous media was first evaluated by cyclic voltammetry and X-ray photoelectron spectroscopy analyses, showing a higher surface coverage for the reduction process. Cyclam was subsequently attached on the COOH-modified graphite felts by a coupling reaction. The modified electrodes were then employed in a flow analytical system for trace analysis of copper (II) ions. The influence of the surface coverage and the nature of the linker on the electrochemical signal obtained by linear sweep stripping voltammetry analysis after a preconcentration step performed at open circuit was highlighted. The selectivity estimated in the presence of lead used as a common ion interferent was higher when a selective receptor was used and depends on the nature of the linker.

As estacas foram feitas de brotações de 4 a 6cm de comprimento de mudas de P. caribaea var. hondurensis Morelet com corte bisel na base sendo as acículas basais eliminadas. A base das estacas foram submetidas aos tratamentos por 2 segundos com os seguintes tratamentos: 1- NAA 2000mg L-1; 2- NAA 4000mg L-1; 3- NAA 6000mg L-1; 4- NAA 2000mg L-1 + PBZ 100mg L-1; 5- NAA 4000mg L-1 + PBZ; 6- NAA 6000mg L-1 + PBZ; 7- IBA 2000mg L-1; 8- IBA 4000mg L-1; 9- IBA 6000mg L-1; 10-IBA 2000mg L-1 + PBZ; 11- IBA 4000mg L-1 + PBZ; 12- IBA 6000mg L-1 + PBZ e testemunha. Após os tratamentos as estacas foram plantadas em tubetes contendo 50% de palha de arroz carbonizada e 50% de vermiculita. As avaliações realizadas aos 60 dias após o plantio mostraram que estacas de P. caribaea tratadas com IBA levaram a maior porcentagem de estacas enraizadas que aquelas tratadas com NAA, sendo o mais efetivo, IBA a 4000mg L-1 em conjunto com 100mg L-1 de paclobutrazol. This work evaluated the rooting of Pinus caribaea var. hondurensis Morelet cuttings under the action of different levels of plant growth regulators. The cuttings consisted of 4-6 cm long shoots of P. caribaea var. hondurensis Morelet with their basal needles removed. The basal part of the cuttings were treated for 2 seconds with the following treatments: 1- NAA 2000mg L-1; 2- NAA 4000mg L-1; 3- NAA 6000mg L-1; 4- NAA 2000mg L-1 + PBZ 100mg L-1; 5- NAA 4000mg L-1 + PBZ; 6- NAA 6000mg L-1 + PBZ; 7- IBA 2000mg L-1; 8- IBA 4000mg L-1; 9- IBA 6000mg L-1; 10-IBA 2000mg L-1 + PBZ; 11- IBA 4000mg L-1 + PBZ; 12- IBA 6000mg L-1 + PBZ; and a control. After receiving the treatment, the cuttings were planted in tubes containing 50% carbonized rice hulls and 50% vermiculite. The evaluations, performed 60 days after planting, showed that P. caribaea var. hondurensis cuttings treated with IBA produced a higher percentage of rooted cuttings than those treated with NAA; the most effective treatment was IBA 4000mg L-1 plus 100mg L-1 paclobutrazol. Abstract: International audience; The health problems amongst musical instrument makers working on the White mulberry (Morus alba), lead to investigate the toxicity potential of wood extractives. Five solvents of different polarity and two methods of extractions were used to ensure a thorough, clear and classified compound removal. The compounds were identified using qualitative GC-mass and the toxicity were determined by studying their effect on termite survival (Reticulitermes flavipes). The results indicated the presence of different compound categories like phenols, higher hydrocarbons, fatty acids, sterols and aromatic components in the extractives. These compounds were not removable by just one solvent and the order of removal was polarity related. The extractives were found toxic towards termites. Resorcinol, which corresponded to the highest proportion of peaks in the GC-mass spectra, has been assigned as the possible compound responsible for the allergic reactions by the artisans.

Abstract: International audience; Fetal testis is a major target of endocrine disruptors (EDs). During the last 20 years, we have developed an organotypic culture system that maintains the function of the different fetal testis cell types and have used this approach as a toxicological test to evaluate the effects of various compounds on gametogenesis and steroidogenesis in rat, mouse and human testes. We named this test rat, mouse and human fetal testis assay. With this approach, we compared the effects of six potential EDs ((mono-(2-ethylhexyl) phthalate (MEHP), cadmium, depleted uranium, diethylstilboestrol (DES), bisphenol A (BPA) and metformin) and one signalling molecule (retinoic acid (RA)) on the function of rat, mouse and human fetal testis at a comparable developmental stage. We found that the response is similar in humans and rodents for only one third of our analyses. For instance, RA and MEHP have similar negative effects on gametogenesis in the three species. For another third of our analyses, the threshold efficient concentrations that disturb gametogenesis and/or steroidogenesis differ as a function of the species. For instance, BPA and metformin have similar negative effects on steroidogenesis in human and rodents, but at different threshold doses. For the last third of our analyses, the qualitative response is species specific. For instance, MEHP and DES affect steroidogenesis in rodents, but not in human fetal testis. These species differences raise concerns about the extrapolation of data obtained in rodents to human health risk assessment and highlight the need of rigorous comparisons of the effects in human and rodent models, when assessing ED risk.

Abstract: This chapter reports the mechanical properties of starch&hyphen;based nanocomposite materials, considering that the starchy material can be used either as the continuous polymeric phase (matrix), or the nanodispersed phase (filler), or both. The first section of the chapter discusses the reinforcing effect of the different fillers used to improve the mechanical properties of thermoplastic starch, that is, inorganic clays, carbon nanotubes, and polysaccharide&hyphen;based (cellulose, starch) nanoparticles and attempts to give a critical comparison of the different reinforcements The second section talks about the reinforcing effect of starch nanocrystals, as well as the mechanisms proposed to explain it.

Abstract: International audience; The present work was focused on elucidating biochemical changes in the model bacterium Escherichia coli exposed to ionic silver mediated stress, at a single-cell scale. In order to achieve this, in situ synchrotron Fourier-transform infrared (sFTIR) microspectroscopy was performed, for the first time, on individual cells by attenuated total reflectance (ATR) combined with the use of zinc-selenide hemisphere for high spatial resolution. In a first part, the potential of the method was evaluated on bacteria subjected to a lethal 100 μM AgNO3 concentration for 2 h compared to untreated 100 % viable cells. Differences in cell composition were assessed for the C–H stretching and protein spectral regions, indicating that the inhibitory action was targeted against both fatty acids and proteins. Transmission electron microscopy (TEM) confirmed morphological damages of the cell ultrastructure. The relevance of ATR-sFTIR microspectroscopy for highlighting the heterogeneity in Ag+-mediated effects within a given bacterial population was also pointed out. Abstract: Background The usual treatments for pleural malignancies are mostly palliative. In contrast, peritoneal malignancies are often treated with a curative intent by cytoreductive surgery and intraperitoneal chemotherapy. As pressure has been shown to increase antitumor efficacy, we applied the concept of high-pressure intracavitary chemotherapy to the pleural space in a swine model. Methods Cisplatin and gemcitabine were selected because of their antineoplasic efficacy in vitro in a wide spectrum of cancer cell lines. The pleural cavity of 21 pigs was filled with saline solution; haemodynamic and respiratory parameters were monitored. The pressure was increased to 15-25 cm H2O. This treatment was associated with pneumonectomy in 6 pigs. Five pigs were treated with chemotherapy under pressure. Results The combination of gemcitabine (100 mg/l) and cisplatin (30 mg/l) was highly cytotoxic in vitro. The maximum tolerated pressure was 20 cm H20, due to haemodynamic failure. Pneumonectomy was not tolerated, either before or after pleural infusion. Five pigs survived intrapleural chemotherapy associating gemcitabine and cisplatin with 20 cm H2O pressure for 60 min. Conclusions High-pressure intrapleural chemotherapy is feasible in pigs. Further experiments will establish the pharmacokinetics and determine whether the benefit already shown in the peritoneum is also obtained in the pleura.

Abstract: Vibrational properties of wood are affected by several parameters, of which extractives can be one of the most important ones. Wood for European musical instruments has been often studied, but traditional Middle Eastern ones had been left unnoticed. In this study white mulberry (Morus alba L.), the main material for long-necked lutes in Iran, was extracted by five solvents of various polarities (water included). Free-free bar forced vibrations were used to measure longitudinal (L) loss tangent (tanδ), storage (elastic) modulus (E′) and specific modulus (E′/γ) in the acoustic range. Their anisotropy between the 3 axes of orthotropy was determined by dynamic mechanical analysis. Native wood had a quite low E L′/γ but its tanδ was smaller than expected, and the anisotropy of tanδ and E′/γ was very low. Removal of extractives caused tanδ to increase and moduli to decrease. Acetone, the most effective solvent on damping despite a moderate extraction yield, increased tanδ L by at least 20% but did not modify E′/γ as much. When used successively, its effects masked those of solvents used afterwards. Anisotropy of E′/γ was nearly unchanged after extraction in methanol or hot water, while tanδ was much more increased in R than in T direction. Results suggest that in white mulberry, damping is governed more by nature and localization of extractives rather than by their crud abundance.
