Although sweet tea is rich in bioactive polysaccharides, the knowledge regarding their structures, bioactivities, and gut microbial metabolism is still limited. Therefore, in order to promote the application of sweet tea polysaccharide (STP) in the food industry, the pressurized hot water extraction (PHWE) of STP was optimized, and its structural properties and biological effects as well as microbial fermentation characteristics were investigated. The maximum extraction yield (4.64 % ± 0.03 %) of STP extracted by PHWE was obtained under the optimal conditions. Both homogalacturonan and arabinogalactan might exist as major polysaccharide fragments in STP. Additionally, STP exerted obviously in vitro antioxidant, anti-diabetic, and immunostimulatory effects, which might be related to its chemical properties, such as uronic acids, conjugated polyphenolics, and esterification degree. Furthermore, STP could be consumed by intestinal microbiota, and its fermentability was about 54 % at the end stage of fecal fermentation. Indeed, STP could modulate the microbial composition via improving the growth of several beneficial microbes, causing the release of beneficial short-chain fatty acids. Nowadays, few investigations on the process parameters of grafted starch synthesized using electron transfer atom transfer radical polymerization (ARGET ATRP) and its applications in warp sizing and paper-making are presented. Therefore, this study aimed to survey the appropriate process parameters of bromoisobutyryl esterified starch- g -poly(acrylic acid) (BBES- g -PAA) synthesized by the ARGET ATRP, and also aimed to provide a new biobased BBES- g -PAA adhesive. The appropriate synthesis process parameters were 1.2, 0.32, and 0.6 in the molar ratios of vitamin C, CuBr 2 , and pentamethyldivinyltriamine to BBES, respectively, at 40 °C for 5 h. The BBES- g -PAA samples with a grafting ratio range of 4.63–14.14 % exhibited bonding forces of 57.8–64.6 N to wool fibers [55.5 N (BBES) and 53.8 N (ATS)], and their films showed breaking elongations of 3.29–3.80 % [2.74 % (BBES) and 2.49 % (ATS)] and tensile strengths of 29.1–25.4 MPa [30.4 MPa (BBES) and 34.7 MPa (ATS)]. Compared with BBES, significantly increased bonding forces and film elongations, and decreased film strengths for the BBES-g-PAA samples with grafting ratios ≥10.54 % were displayed (p < 0.05). The time (100–42 s) taken for the BBES- g -PAA films was significantly shorter than that of ATS (246 s) and BBES (196 s) films (p < 0.05), corresponding to better desizability. The influence of the quaternization and butyrylation on the sizing properties of biological starch macromolecule was evaluated for acquiring a new starch bio-based size [quaternized-butyrylated starch (QBS)]. The sizing properties of granular QBS samples were investigated in comparison with acid-thinned starch (ATS) and quaternized starch (QS). The QBS samples with a DS range of 0.029–0.0779 exhibited bonding strengths of 17.0–18.3 cN/tex to cotton fibers [15.5 cN/tex (ATS, DS = 0) and 16.6 cN/tex (QS, DS = 0.0240)] and 31.0–34.3 cN/tex to polyester fibers [28.0 cN/tex (ATS) and 30.1 cN/tex (QS)], and their films showed breaking elongations of 2.99–3.51% [2.59% (ATS) and 2.81% (QS)] and tensile strengths of 36.5–32.1 MPa [38.1 MPa (ATS) and 37.3 MPa (QS)]. Compared with QS, significantly increased bonding strengths as well as obviously decreased strengths and increased elongations of the films for the QBS samples with the total DS ≥ 0.0635 were exhibited. As increasing the modification levels from 0.029 to 0.0779, QBS presented paste stabilities from 90.4% to 85.7% which met with the requirement in warp sizing, and displayed higher desizing efficiencies (93.2–93.8%) than ATS (91.5%) and QS (90.2%). Based on these results, the amphiphilic quaternization-butyrylation was a good means for starch to acquire good sizing properties. Aquatic products with high moisture and protein content are susceptible to bacterial growth and spoilage. Searching for efficient and safe natural antibacterial agents to preserve aquatic products has been concerned widely. In this study, ε-poly-lysine-epigallocatechin gallate/sodium alginate-chitosan nanoparticles (ε-PL-EGCG/SA-CS NPs) were prepared using sodium alginate and chitosan as wall materials and ε-PL-EGCG as core material. The size of nanoparticles was about 200 nm and the encapsulation efficiency was 78.2%. Transmission electron microscopy (TEM) images confirmed the prepared spherical nanoparticles. Fourier transform infrared spectroscopy (FTIR) and multifunctional polycrystalline X-ray diffraction (XRD) spectra indicated that ε-PL-EGCG was encapsulated in the nanoparticles. Thermo-gravimetric analysis (TGA) illustrated that the thermal stability of encapsulated ε-PL-EGCG was improved more than that of bare ε-PL-EGCG. In addition, in vitro release assays showed that the ε-PL-EGCG was released continuously over 36 h. Bacteria inhibition results showed that the ε-PL-EGCG/SA-CS NPs significantly inhibited specific spoilage bacteria E3 that screened out of aquatic products, Escherichia coli and Staphylococcus aureus. A novel strategy adaptive to 3D printing of stereo-complexed polylactide matrix for simultaneous toughness and stiffness was designed. Stereo-complexation is a potent way to enhance both aqueous stability and heat resistance of polylactide, but also aggravates brittleness problem of polylactide. Though poly(butyleneadipate- co -terephthalate) elastomer with epoxidized compatibilizer improved stiffness and toughness of common polylactide, their effectiveness on mechanical and crystallization properties of stereo-complexed polylactide remained unknown. More importantly, incorporation of above techniques into 3D printing kept a fundamental challenge. Both stereo-complexation of polylactide and co valent coupling of polylactide and poly(butyleneadipate-co-terephthalate) by epoxidized compatibilizer are easy to occur when preparing the filaments for printing, impeding the following 3D printing procedure. The hypothesis for this research is that controlled hierarchical crystallization and reaction in three thermal processes could ensure simultaneous toughness and stiffness, and complete stereo-complexation in polylactide matrices. Reinforcing effects of a selected epoxidized compatibilizer, POSS (epoxy)8 , on crystallinities, thermal properties, mechanical properties and morphologies were systematically studied. Flammability and poor toughness of unmodified PLA limit its applications in various fields. Though ammonium polyphosphate (APP) is a green and effective flame retardant, it has poor compatibility with the matrix, leading to a decrease in mechanical properties. Stereo-complexation greatly improves the strength and heat resistance of traditional PLA. However, the effect of flame retardants on the formation of stereo-complexed crystals and the impact of stereo-complexation on flame retardancy have not been studied previously. In this research, PDLA chains were first in-situ reacted with APP particles for improved interfacial compatibility. By utilizing the characteristic of PLA enantiomers that can form stereo-complexed crystals, near-complete stereo-complexed PLA fibers with flame retardancy were produced via clean and continuous melt spinning. The compatibility between PDLA-g-APP and PLLA matrix was studied by SEM, rheological analyses and DSC. Strength and flexibility of the fibers were simultaneously enhanced compared to traditional PLA due to the synergistic effect of interfacial compatibility and stereo-complexation. Compared to traditional PLA, the peak heat release rate and total heat release in microcalorimetry test were reduced by 33 % and 22 %, respectively. The flame-retardant fibers achieved a V-0 rating in the UL-94 test, and an increase in LOI value from 19.4 % to 28.2 %. In this study, zein-loaded cinnamaldehyde (Cin@ZN) nanoparticles were incorporated into Chitosan (CS)/dialdehyde carboxymethyl cellulose (DCMC) matrix to fabricate the active food packaging materials possessing outstanding antioxidant and antibacterial properties. The research investigated how varying levels of Cin@ZN nanoparticles affected the morphology, microstructure, physicochemical properties of CS/DCMC composite films. The inclusion of Cin@ZN could significantly improve the mechanical strength, reduce the water vapor and oxygen permeability of CS/DCMC composite films and endow films with UV-light blocking properties. It's worth noting that the antibacterial and antioxidant capacities of CS/DCMC films had an astonishing enhancement with Cin@ZN blending, in which ABTS scavenging ratio of the composite films (100 mg) with different Cin@ZN contents reached >90 %. Furthermore, CS/DCMC/Cin@ZN 35 % composite film has the ability to efficiently protect strawberries from microbial damage and decelerate the spoilage rate of strawberries under ambient condition. This study revealed the influence of phosphorylation-dimethylammonium chloride acyloxylation (PDACA) on the desizability, film properties, paste stability, and adhesion of biological starch macromolecules. A new starch-based sizing agent, phosphorylated-dimethylammonium chloride acyloxylated starch (PDACAS), was synthesized with degrees of substitution (DS) ranging from 0.033 to 0.065. Compared to control phosphorylated-quaternized starch (PQS, 87.4 %), the desizing efficiency of cotton yarns sized with PDACAS was ~94 %, exceeding the industrial minimum requirement of 90 %. The PDACAS film tensile properties were as follows: elongation at break of 3.31 %–3.78 %, bending endurance of 1131–1537 cycles, and tensile strength of 35.83–28.31 MPa, compared with those of acid-thinned starch (ATS) film (2.74 %, 957 cycles, and 38.12 MPa). The PDACAS had paste stability of ~92 %, compared with 83.3 % for ATS. The bonding forces (an indicator of adhesion to fibers) ranged from 107.1 N to 125.3 N for cotton roving, and 128.3 N to 148.7 N for polyester/cotton roving, which were significantly better than those of ATS (95 N for cotton and 117.9 N for polyester/cotton roving). Overall, PDACA treatment effectively avoided the adverse effect of high DS quaternization on the desizability of PQS and imparted good film properties, paste stability, and adhesion to starch. A novel strategy adaptive to 3D printing of PLA matrix for complete stereo-complexation was designed. Stereo-complexation has been demonstrated for its effectiveness in simultaneously improving aqueous stability and heat resistance of PLA. However, current techniques could not be directly incorporated into 3D printing of stereo-complexed PLA since stereo-complexed crystallites are easily formed before printing. High printing temperatures are thus required but decompose PLA materials at the same time. The hypothesis for this research is that controllable hierarchical crystallization in three thermal processes, the filament preparation, 3D printing and post annealing, could ensure feasibility of the strategy and a 100% stereo-complexation level in PLA matrices. Effects of extrusion, ambient and annealing temperatures on material structures were analyzed via WAXD, DSC and DMA. Resistance to hydrolysis and heat of the 3D printed PLA matrix was evaluated under practical conditions. It was showed that homo-crystallites anchored molecular chains of PLA during the post-annealing process for a high retention of tensile properties, while stereo-complexed crystallites provided stronger intermolecular interactions for improved hydrolytic and thermal resistance. The objective of this study was to survey the influence of poly(acrylic acid- co -butyl acrylate) [P(AA- co -BA)] branches on the sizing properties (i.e., paste stability, adhesion, properties of film and sized warps) of biological macromolecule (corn starch) for further promoting the properties of bromoisobutyryl esterified starch (BBES) and developing a new biobased sizing agent [BBES- g -P(AA- co -BA)]. The sizing properties of BBES- g -P(AA- co -BA) were estimated in comparison with acid-hydrolyzed starch (AHS) and BBES. Compared with the two starches, BBES- g -P(AA- co -BA) displayed higher paste stability, bonding forces to both polyester and polyester/cotton roving, film elongation and water solubility, as well as lower film strength. And the increased grafting ratio exhibited positive effects on these properties of BBES- g -P(AA- co -BA). The properties containing increase in strength, loss in extension, abrasion resistance and hairiness of polyester and polyester/cotton blended yarns sized with BBES- g -P(AA- co -BA), were superior to those sized with AHS and BBES, respectively, indicating that the incorporation of P(AA- co -BA) branches onto BBES could further promote the sizing qualities of both yarns. The conversion of renewable lignin with low-cost and high carbon content properties into porous carbon materials for supercapacitor applications has caught considerable interest. Herein, two dimensional lignin-derived carbon nanosheets (N-LHPC) with hierarchically porous structures were facilely synthesized via a novel metal-lignin assembly strategy and their performances for supercapacitor applications were investigated. During the carbonization process, the uniformly distributed Zn facilitates the coordinating development of micropores structure and the generated MgO embedded in the carbon matrix acts as a template to produce mesoporous structure after acid washing. Moreover, the melamine addition promotes the development of mesopores by formation of lamellae structure and realizes the N doping in the carbon materials. Therefore, the obtained N-LHPC presents an excellent specific capacitance of 235.75 F/g at 0.5 A/g owing to its hierarchical pore structure as well as the N/O functional groups. Moreover, at the power density of 450 W/kg, the N-LHPC achieves a maximum energy density of 14.75 Wh/kg, showing great application potential in energy storage. The metal-lignin assembly strategy followed by N-doping proposed in this paper provides N-LHPC materials with hierarchical nanostructure, good electron/ion transfer properties, and abundant pseudocapacitive active species, which improve the capacitance performances of the N-LHPC. Interferon (IFN) alpha/beta receptor 1 (IFNAR1) is indispensable for antiviral responses and the immune regulation. Dysregulation of the IFNAR1-mediaetd signaling pathways leads to deleterious autoimmune diseases such as systemic lupus erythematosus (SLE). QX006N, a humanized therapeutic monoclonal antibody, specifically targets human IFNAR1 and is in the clinical trial phase for treating SLE, but the molecular mechanism underlying the QX006N-mediated recognition of IFNAR1 remains unclear. Here, we report the high neutralization activities of QX006N against IFNAR1-mediated signal transduction. Meanwhile, we determine the structures of the fragment antigen-binding domain (Fab) of QX006N (QX006N-Fab) and QX006N-Fab in complex with the subdomains 1–3 of IFNAR1 (IFNAR1-SD123) at 2.87 Å and 2.68 Å resolutions, respectively. In the structure of the QX006N-Fab/IFNAR1-SD123 complex, QX006N-Fab only recognizes the SD3 subdomain of IFNAR1 by the hydrophobic, hydrogen-bonding and electrostatic interactions. Compared with the structure of the IFN/IFNAR1/IFNAR2 complex, the binding of QX006N-Fab to IFNAR1-SD3 blocks its association with IFN due to steric hindrance, which inhibits the IFN/IFNAR1/IFNAR2 complex formation for signal transduction. The results of this study provide the structural evidence for the specific targeting of IFNAR1 by the therapeutic antibody QX006N and pave the way for the rational design of antibody drugs to combat IFNAR1-related autoimmune diseases. To enhance the flexibility of starch film adhesion on yarns, sizing lipids (saponified lipid or emulsified lipid) must be added during the sizing process. However, different types of sizing lipids may have diverse combinations with starch to impact enzyme desizing. Therefore, this study investigated the effects of saponified lipid and emulsified lipid commonly used in warp sizing on the hydrolysis of starch. Additionally, the desizing efficiency and chain structure of desizing residues were analyzed. Experimental results demonstrated that the existence of saponified lipid or emulsified lipid led to a reduction in the degree of hydrolysis (1.1 % and 2.6 %, respectively) compared to the original corn starch. Notably, saponified lipid exhibited a relatively strong negative impact. Furthermore, the desizing efficiency decreased after adding emulsified lipid (1.2 %) or saponified lipid (2.9 %). Starch-lipid V-type complexes and physical hindrance could inhibit the enzyme desizing, resulting in a larger wavelength of maximum absorbance for desizing residues, along with higher molecular weight, z-average radius of gyration, and an increased proportion of long chains. The presence of saponified lipid significantly negatively influenced desizing, possibly due to the smaller particle size and propensity for complex formation with starch. Natural plant polysaccharides are macromolecular substances with a wide range of biological activities. They have a wide range of biological activities, especially play an important role in the treatment of inflammatory bowel disease. The molecular weight of polysaccharides, the composition of monosaccharides and the connection of glycosidic bonds will affect the therapeutic effect on inflammatory bowel disease. Traditional Chinese medicine plant polysaccharides and various types of plant polysaccharides reduce the levels of inflammatory cytokines IL-1β, IL-6, IL-8 and IL-17, increase the level of anti-inflammatory factor IL-10, regulate NF-κB signaling pathway, and NLRP3 inflammasome to relieve colitis. At the same time, they can play a protective role by regulating the balance of intestinal flora in mice with colitis and increasing the abundance of probiotics to promote the metabolism of polysaccharide metabolites SCFAs. This review summarizes the research on the treatment of inflammatory bowel disease by many natural plant polysaccharides, and provides a theoretical basis for the later treatment of polysaccharides on inflammatory bowel disease. Polysaccharides, being a natural, active, and biodegradable polymer, have garnered significant attention due to their exceptional properties. These properties make them ideal for creating multifunctional hydrogels that can be used as wound dressings for skin injuries. Polysaccharide hydrogel has the ability to both simulate the natural extracellular matrix, promote cell proliferation, and provide a suitable environment for wound healing while protecting it from bacterial invasion. Polysaccharide hydrogels offer a promising solution for repairing damaged skin. This review provides an overview of the mechanisms involved in skin damage repair and emphasizes the potential of polysaccharide hydrogels in this regard. For different skin injuries, polysaccharide hydrogels can play a role in promoting wound healing. The mechanical mismatch between soft hydrated tissues and sutures has become a common negative impact on wound healing process. A novel method of coating multilayer polymer shells is thus reported to improve the mechanical performance of hydrogel sutures. It is suitable for tissue patching and shows advantages of convenient, efficient, and biosafety. Specifically, a precursor hydrogel (Cu@CMC) consisted of carboxymethyl chitosan and copper modified by carbon dots was used as the inner sheath, and then bonding the precursor hydrogel sheath with toughening polyethylene glycol network by anchoring sites composited from rigid chitosan shell integrated a whole structure. Subsequently, the whole system was soaked with EtOH, and rapid dehydration of EtOH was used to accelerate the entanglement process between the two coatings by constricting the molecular chains. Finally, an ideal suture (Cu-fiber) with both toughness and rigidness was obtained. The data showed that the tensile strength and biosafety of the hydrogel sutures prepared by the new strategy were significantly improved, and the skin, liver and vessel of rodents can be sutured without secondary damage. The cell surface of lactic acid bacteria (LAB) plays an essential role in cell-cell and cell-host interactions. Exopolysaccharides (EPSs) are produced on the cell surface of LAB or in the surrounding medium and are considered to be in favor of the strain- specific probiotic surface characteristics. In this work, the structure features of EPS from Lacticaseibacillus paracasei S-NB were analyzed preliminarily, and the genes involved in EPS biosynthesis of S-NB strain were hypothesized and annotated, and their role in phenotypic characteristics were demonstrated by gene deletion analysis. Four mutant strains with deletion of crucial genes involved in EPS synthesis were analyzed for strain characteristics that are closely related to their ability to interact with the host intestinal epithelium cells, including strain surface characteristics and viability under the gastrointestinal stress conditions (both acid and bile stress). Furthermore, the adherence and immunomodulatory properties of wild-type S-NB and its mutant strains were compared using Caco-2 and RAW 264.7 cell lines, respectively. Taken together, the results indicated the importance of genes associated with EPS biosynthesis in L. paracasei S-NB as a determinant in strain surface characteristics and cell-host interaction, especially for S-NB_2176 (responsible for EPS polymerization) and S-NB_2175 (responsible for CpsD/CapB family tyrosine-protein kinase). Activation of lignin by demethylation for improving the reactivity has attracted extensive attentions. However, it still faces many challenges, such as the unsatisfied increase of hydroxyl content and the undesired cracking of linear linkages. Here, the efficient demethylations for significantly increasing the hydroxyl content and protecting the structure of industrial lignin were explored using lewis acid as modification reagent. As BBr 3 was used, the phenolic hydroxyl content (Ar-OH) was increased by 80.65 %, but the lignin structure might be destroyed. About 75 % of the β- O -4 linkages could be fortunately retained by using AlCl 3. This method could also be used for the demethylation of alkaline poplar lignin with up to 171.67 % increase of Ar-OH (from 1.80 to 4.89 mmol/g). After activation, the antioxidant properties were improved 4.64-fold and 2.58-fold for 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals, respectively. The treatment of traumatic spinal cord injury (SCI) remains challenging as the neuron regeneration is impaired by irregular cavity and apoptosis. An injectable in situ gelling hydrogel is therefore developed for the local delivery of cannabidiol (CBD) through a novel method based on polyelectrolyte (PEC) interaction of sodium carboxymethylcellulose (CMC) and chitosan (CS). It can be injected into the spinal cord cavity with a 26-gauge syringe before gelation, and gelled after 110 ± 10 s. Of note, the in-situ forming hydrogel has mechanical properties similar to spinal cord. Moreover, the CBD-loaded hydrogels sustain delivery of CBD for up to 72 h, resulting in reducing apoptosis in SCI by enhancing mitochondrial biogenesis. Importantly, the CBD-loaded hydrogels raise neurogenesis more than pure hydrogels both in vivo and in vitro, further achieving significant recovery of motor and urinary function in SCI rats. Thus, it suggested that CMC/CS/CBD hydrogels could be used as promising biomaterials for tissue engineering and SCI. CBD-loaded CMC/CS PEC hydrogels with injectable and in situ gelling are developed. An arabinogalactan named JSP-1a was isolated from Jasmine tea processing waste by DEAE Sepharose FF and Sephacryl S-200 HR chromatography. Polysaccharide JSP-1a, with an average molecular weight of 87.5 kDa, was composed of galactose (59.60 %), arabinose (33.89 %), mannose (4.81 %), and rhamnose (1.70 %). JSP-1a was found to be a type II arabinogalactan comprising the main backbone of 1, 6-linked Gal p residues, and the side chain containing α-T-Ara f , α-1,5-Ara f , β-T-Gal p , β-1,3-Gal p , and β-1,4-Man p residues was attached to the O-3 position of β-1,3,6-Gal p residues. Evidence from bioactivity assays indicated that JSP-1a possessed potent immunomodulatory effects on RAW264.7 macrophages: treatment with JSP-1a increased phagocytosis, activated NF-κB p65 translocation, and promoted the production of NO, reactive oxygen species (ROS), the tumor necrosis factor (TNF)-α, and interleukin (IL)-6. Furthermore, inhibition of Toll-like receptor 4 caused the suppression of NO release and cytokines secretion, which indicated that TLR-4/NF-κB pathway might play a significant role in JSP-1a-induced macrophages' immune response. In this study, heparin-like polysaccharides were successfully produced by sulfation of carboxymethylcellulose sodium, then a fully biobased bilayer composed of sulfated carboxymethylcellulose sodium (SCMC) and chitosan (CS) was composited on the surface of Poly (L-lactic acid) (PLA) through layer-by-layer (LBL) assembly for the potential blood-contact application such as bioresorbable vascular scaffold. The preliminary structure and bioactivity of SCMC with different degree of sulfation were investigated, and the SCMC with best performance was selected. The surface chemical compositions, morphologies and wettability of SCMC/CS multilayer-modified PLA films were researched by X-ray photoelectron spectrometer, scanning electron microscopy and water contact angle meter. A series of anticoagulation tests of SCMC/CS multilayer-modified PLA films were performed. In term of (SCMC/CS) 15 multilayer-modified PLA film, the protein adsorption and plate adhesion decreased by 44.6 % and 71.5 %, respectively, the activated partial thromboplastin time prolonged by 11.9 times and thrombin time exceed 300 s, the contact activation and hemolysis rate significantly reduced compared with unmodified PLA film. Besides, this modified PLA films performed good cytocompatibility to L929 fibroblast cells, excellent anti-inflammatory and antibacterial abilities. In this work, we reported an in situ exopolysaccharide (in situ -EPS1) containing rare fucose produced by Lactobacillus helveticus MB2-1 in Sayram ketteki yoghurt, which made it unique. Its fine structure was characterized by GPC, HPLC, FT-IR, GC–MS,1HNMR and 13CNMR together with two-dimensional (2D) NMR spectra. The results revealed that in situ -EPS1 was a new heteropolysaccharide with molecular weight of 1.06 × 105 Da, and was composed of mannose, rhamnose, glucose, galactose and fucose with the following repeating units. In this study, a green and efficient enrichment method for the four majors active diterpenoid components: pimelotide C, pimelotide A, simplexin, and 6α,7α-epoxy-5β-hydroxy-12-deoxyphorbol-13-decanoate in the buds of Wikstroemia chamaedaphne was established using macroporous resin chromatography. The adsorption and desorption rates of seven macroporous resins were compared using static tests. The D101 macroporous resin exhibited the best performance. Static and dynamic adsorption tests were performed to determine the enrichment and purification of important bioactive diterpenoids in the buds of W. chamaedaphne. Diterpenoid extracts were obtained by using D101 macroporous resin from the crude extracts of W. chamaedaphne. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis demonstrated that most of the diterpenoids were enriched in diterpenoid extracts. These results confirmed that diterpenoids in the buds of W. chamaedaphne could be enriched using macroporous resin technology, and the enriched diterpenoid extracts showed more efficient activation of the latent human immunodeficiency virus. This study provides a novel strategy for discovering efficient and low-toxicity latency-reversing agents and a potential basis for the comprehensive development and clinical application of the buds of W. chamaedaphne. Solar desalination is an effective solution to address the global water scarcity issue. However, biofouling poses a significant challenge for solar evaporators due to the presence of bacteria in seawater. In this study, an anti-biofouling evaporator was constructed using the synergistic antibacterial effect of lignin and silver nanoparticles (AgNPs). The AgNPs were easily synthesized using lignin as reductant under mild reaction conditions. Subsequently, the Lignin-AgNPs solution was integrated into polyacrylamide hydrogel (PAAm) without any purification steps, resulting in the formation of Lignin/AgNPs-PAAm (LAg-PAAm). Under the combined action of AgNPs and the hydroquinone groups present in oxidized lignin, LAg-PAAm achieved over 99 % disinfection efficiency within 1 h, effectively preventing biofilm formation in pore channels of solar evaporators. The anti-biofouling solar evaporator demonstrated an evaporation rate of 1.85 kg m−2 h−1 under 1 sun irradiation, and maintained stable performance for >30 days due to its high efficient bactericidal effect. Polysaccharides have gained attention as valuable supplements and natural medicinal resources, particularly for their anti-tumor properties. Their low toxicity and potent anti-tumor effects make them promising candidates for cancer prevention and treatment. The tumor microenvironment is crucial in tumor development and offers potential avenues for novel cancer therapies. Research indicates that polysaccharides can positively influence the tumor microenvironment. However, the structural complexity of most anti-tumor polysaccharides, often heteropolysaccharides, poses challenges for structural analysis. To enhance their pharmacological activity, researchers have modified the structure and properties of natural polysaccharides based on structure–activity relationships, and they have discovered that many polysaccharides exhibit significantly enhanced anti-tumor activity after chemical modification. This article reviews recent strategies for targeting the tumor microenvironment with polysaccharides and briefly discusses the structure–activity relationships of anti-tumor polysaccharides. It also summarises the main chemical modification methods of polysaccharides and discusses the impact of chemical modifications on the anti-tumor activity of polysaccharides. Many dietary polysaccharides have been shown to protect against various harmful external stimuli by protecting the integrity of the intestinal barrier. Arabinogalactan (AG) is a high molecular weight polysaccharide composed of arabinose and galactose, which has good immunomodulatory, antioxidant and intestinal conditioning activities. Gastrointestinal injury caused by cisplatin (CP) is an inevitable damage during CP chemotherapy. This research explored the ameliorative effect of AG on cisplatin-induced intestinal toxicity and its possible molecular targets and mechanisms. The results showed that AG (200, 400 mg/kg) could significantly reverse the intestinal histopathological changes and oxidative stress injury caused by CP. Meantime, AG could target the IRE1α/JNK axis to inhibit the expression of apoptosis-related proteins and block the apoptotic cascade, thus reducing intestinal damage. In vitro , AG (10, 20, and 40 μg/mL) could regulate the IRE1α/JNK axis, inhibit apoptosis, and restore the antioxidant defense system damaged by CP to play a protective role in the intestine. In addition, 4-phenylbutyrate (4-PBA), a specific inhibitor of endoplasmic reticulum stress, was used to verify that AG also affected protein expression levels by regulating the IRE1α/JNK pathway-mediated endoplasmic reticulum stress signaling pathway, thereby alleviating CP-induced gastrointestinal dysfunction. In this study, cellulose-based superabsorbent hydrogel was synthesized from sodium carboxymethyl cellulose (CMC-Na), acrylic acid (AA), and 2-acrylamido-2-methylpropanesulfonic acid (AMPS) to enhance its water absorbency and salt tolerance for soil-conditioning applications in areas suffering from drought and soil salinization. Superabsorbent hydrogels (SHs) were prepared by CMC-Na and AMPS successfully, using chemical graft technology. Structure, morphology, thermal stability, and water absorbency of SHs were deduced. The cellulose-based hydrogel showed a high salt tolerance that the maximum water absorbency reached 604 and 119% in distilled water and saline water, respectively. The swelling behavior in aqueous solvents indicated that the water absorption of hydrogels was improved with the increasing ratio of CMC-Na. All SHs exhibited adsorption of nitrogen with the maximum adsorption of ammonia nitrogen 30 mg·g−1 and the presence of hydrogels could slow down the loss of nutrients in the soil. This study provided a feasible strategy that AMPS was substituted by CMC-Na to synthesize SHs with strong water absorbency and high salt tolerance which could be efficiently applied in agriculture as a soil conditioner. In order to study the mechanism of high viscosity of Sayram ketteki yoghurt, the growth, acidification properties, in situ exopolysaccharides (EPS) production of Lactobacillus helveticus MB2-1 in milk medium were investigated. The microstructure of the yoghurt was analyzed. The characteristics of in situ EPS produced by this strain in yoghurt were studied by high-performance liquid chromatography (HPLC), Fourier-transform infrared spectroscopy (FT-IR), ultraviolet-visible (UV–vis) analysis. The amount of in situ EPS produced could be up to 689.47 mg/L. The micrographs of Sayram ketteki yoghurt demonstrated that the in situ EPS secreted by ropy L. helveticus MB2-1 were closely connected with proteins, effectively filling the three-dimensional network structure of casein clusters, thereby resulting in high viscosity of yoghurt. Besides, the molecular weight of in situ EPS was 9.34 × 104 Da, and the in situ EPS was determined to be a new heteropolysaccharide, containing fucose, which made it unique. Moreover, the set yoghurts added with in situ EPS were demonstrated fine effects on the texture improvement. These results illustrated that L. helveticus MB2-1 could be set as a good starter and the in situ EPS could be considered as a probiotic stabilizer substitute for fermented dairy products. This study investigated the supramolecular structure features and immunomodulatory effects of two exopolysaccharide fractions (EPS1 and EPS2) from Paecilomyces cicada TJJ1213 in vitro. AFM images revealed that EPS1 and EPS2 displayed different morphological features at different concentrations. Congo red and XRD assay further proved that EPS1 and EPS2 mainly exhibited amorphous structure with random coil conformation in solution. Furthermore, the immunomodulatory effect of EPSs was investigated on RAW264.7 cells. Results showed that EPS1 and EPS2 could enhance the phagocytic activity and induce the NO production and could also significantly up-regulate the mRNA expression of iNOS, TNF-α, IL-6, IL-1β, IFN-γ and IL-4. Western blot assay analysis demonstrated that EPSs increased protein expression of TLR4 and the nuclear translocation of NF-κB p50/p65. Additionally, the phosphorylation levels of MAPKs proteins (p38, ERK and JNK) were also remarkably increased. Diabetic wound healing is a substantial clinical challenge, characterized by delayed angiogenesis and unresolved inflammation. Lentinan, a polysaccharide extracted from shiitake mushrooms, has the potential to regulate both macrophage polarization and angiogenesis, though this aspect remains inadequately explored. To facilitate lentinan's clinical utility, we have developed a GelMA hydrogel encapsulated with lentinan (10 μM), offering a controlled release mechanism for sustained lentinan delivery at the wound site. Application of the lentinan-encapsulated delivery system topically significantly expedites wound closure compared to control groups. Furthermore, histological examination demonstrates enhanced neovascularization and reduced inflammation in lentinan-treated wounds, as evidenced by increased M2 macrophage infiltration. Moreover, our results indicated that lentinan-induced AMPK activation promotes DAF16 expression, enhancing the resistance of macrophages and HUVECs to oxidative stress in high-glucose environments, thereby promoting M2 macrophage polarization and angiogenesis. All these findings underscore lentinan's capacity to modulate macrophage polarization and angiogenesis via the AMPK/DAF16 pathway, ultimately facilitating the healing of diabetic wounds. The genus Schisandra , a member of the Magnoliaceae family, is a well-known tonic traditional Chinese medicine with a long history of traditional medicinal and functional food used in China. Polysaccharides are one of its main active constituents, which have a wide range of bioactivities, such as anti-inflammatory, anti-tumor, neuroprotection, anti-diabetes, hepatoprotection, immunomodulation, and anti-fatigue. In this paper, we review the extraction, isolation, purification, structural characterization, bioactivities, as well as structure-activity relationship of polysaccharides from the genus Schisandra. Ganoderma is a well-known medicinal macrofungal genus, of which several species have been thoroughly studied from the medicinal perspective, but most species are rarely involved in. In this study, we focus on the polysaccharides extracted from Ganoderma boninense and their antioxidant activity. Ganoderma boninense is a serious pathogen of oil palms that are cultivated commercially in Southeast Asia. Response surface methodology was conducted to optimize the liquid medium composition, and the mycelia biomass reached 7.063 g/L, that is, 1.4-fold compared with the seed medium. The crude and purified polysaccharides extracted from the fermentation broth showed well 1,1-diphenyl-2-picrylhydrazyl and 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging abilities, and the scavenging abilities of purified polysaccharides reached 94.47 % and 99.88 %, respectively. Six fractions of polysaccharides were extracted and purified from fruiting bodies, mycelia and fermentation broth separately with the elution buffers of distilled water and 0.1 M NaCl solution. Generally, the polysaccharides from fruiting bodies showed stronger protective effect on H 2 O 2 -induced HepG2 cell oxidative damage than other fractions. A total of five to seven monosaccharides were identified in the six fractions of polysaccharides. The correlation analysis revealed that the content of fucose was significantly correlated with the antioxidant activity of polysaccharides, while xylose showed negative correlation results. GH 11 endo -β-1,4-xylanase (Xy) was a crucial enzyme for xylooligosaccharides (XOS) production. The lower reusability and higher cost of purification has limited the industrial application of Xy. Addressing these challenges, our study utilized various immobilization techniques, different supports and forces for Xy immobilization. This study presents a new method in the development of Fe 3 O 4 @PDA@MOF-Xy which is immobilized via multi-point interaction forces, demonstrating a significant advancement in protein loading capacity (80.67 mg/g), and exhibiting remarkable tolerance to acidic and alkaline conditions. This method significantly improved Xy reusability and efficiency for industrial applications, maintaining 60 % activity over 10 cycles. Approximately 23 % XOS production was achieved by Fe 3 O 4 @PDA@MOF-Xy. Moreover, the yield of XOS from cobcorn xylan using this system was 1.15 times higher than that of the free enzyme system. This study assessed the potential prebiotic characteristics of the previously reported Lactiplantibacillus plantarum extracellular polysaccharide (EPS-T1) with immunological activity. EPS-T1 was a novel heteropolysaccharide composed of glucose and galactose (1.00:1.21), with a molecular weight of 1.41 × 106 Da. The monosaccharide composition, molecular weight, fourier transform infrared, and 1H NMR analysis showed that EPS-T1 was well tolerated in the simulated oral cavity, gastric fluid, and small intestinal fluid environments, and was not easily degraded. Meanwhile, EPS-T1 could effectively be used as a carbon source to promote the growth of beneficial Lactobacillus species (Lactobacillus delbrueckii ssp. Bulgaricus , Streptococcus thermophilus , Lacticaseibacillus rhamnose GG, Lactiplantibacillus plantarum , Lacticaseibacillus paracasei and Lactobacillus reuteri). After 24 h of fecal fermentation, EPS-T1(5 mg/mL) effectively reduced the relative abundance of harmful bacteria such as the Escherichia-Shigella , Citrobacter , Fusobacterium , Parasutterella , and Lachnoclostridium. While, the level content of beneficial flora (Bacteroides , Blautia , Phascolarctobacterium , Bifidobacterium , Parabacteroides , and Subdoligranulum) were significantly increased. In addition, EPS-T1 was able to significantly promote the enrichment of short-chain fatty acids such as acetic acid, propionic acid and butyric acid. Colitis can significantly impact daily life. This study utilized DSS to induce acute colitis in mice and examined the regulatory effect of arabinogalactan (AG). The findings demonstrated that AG intake effectively alleviated the phenotype of DSS-induced colitis in mice and protected against small intestine damage. Furthermore, AG suppressed the secretion of pro-inflammatory factors TNF-α and IL-1β, while promoting the secretion of anti-inflammatory factor IL-10. It also inhibited the secretion of LPS in serum and MPO in colon tissue. Additionally, AG regulated the NF-κB/MAPK/PPARγ signaling pathway and inhibited the NLRP3 inflammasome signaling pathway, thereby ameliorating DSS-induced colitis inflammation in mice. AG also influenced the metabolism of short-chain fatty acids, particularly butyrate, in the intestinal tract of mice. Moreover, AG modulated and enhanced the composition of intestinal flora in mice with colitis, increasing the diversity of dominant flora and promoting the growth of beneficial bacteria. Pancreatic cancer (PC) has a poor prognosis due to chemotherapy resistance and unfavorable drug transportation. Albumin conjugates are commonly used as drug carriers to overcome these obstacles. However, membrane-bound glycoprotein mucin 4 (MUC4) has emerged as a promising biomarker among the genetic mutations affecting albumin conjugates therapeutic window. Human serum albumin-conjugated arsenic trioxide (HSA-ATO) has shown potential in treating solid tumors but is limited in PC therapy due to unclear targets and mechanisms. This study investigated the transport mechanisms and therapeutic efficacy of HSA-ATO in PC cells with different MUC4 mutation statuses. Results revealed improved penetration of ATO into PC tumors through conjugated with HSA. However, MUC4 mutation significantly affected treatment sensitivity and HSA-ATO uptake both in vitro and in vivo. Mutant MUC4 cells exhibited over ten times higher IC50 for HSA-ATO and approximately half the uptake compared to wildtype cells. Further research demonstrated that ALPL activation by HSA-ATO enhanced transcytosis in wildtype MUC4 PC cells but not in mutant MUC4 cells, leading to impaired uptake and weaker antitumor effects. Periodontitis is a chronic inflammation of the periodontium caused by a persistent bacterial infection, resulting in destruction of the supporting structures of teeth. Analysis of microbial composition in saliva can inform periodontal status. Actinobacillus actinomycetemcomitans (Aa), Porphyromonas gingivalis (Pg), and Streptococcus mutans (Sm) are among reported periodontal pathogens, and were used as model systems in this study. Our atomic force microscopic (AFM) study revealed that these pathogens are biological nanorods with dimensions of 0.6–1.1 μm in length and 500–700 nm in width. Current bacterial detection methods often involve complex preparation steps and require labeled reporting motifs. Employing surface-enhanced Raman spectroscopy (SERS), we revealed cell-type specific Raman signatures of these pathogens for label-free detection. It overcame the complexity associated with spectral overlaps among different bacterial species, relying on high signal-to-noise ratio (SNR) spectra carefully collected from pure species samples. To enable simple, rapid, and multiplexed detection, we harnessed advanced machine learning techniques to establish predictive models based on a large set of raw spectra of each bacterial species and their mixtures. Using these models, given a raw spectrum collected from a bacterial suspension, simultaneous identification of all three species in the test sample was achieved at 95.6 % accuracy. This sensing modality can be applied to multiplex detection of a broader range and a larger set of periodontal pathogens, paving the way for hassle-free detection of oral bacteria in saliva with little to no sample preparation. Herein, a novel magnetic cross-linked chitosan CS-BA@Fe 3 O 4 was rationally synthesized by cross-linked with epichlorohydrin and coated with Fe 3 O 4 to the acylated chitosan, which was prepared by the reaction of chitosan with benzenetricarboxylic anhydride. The as-obtained absorbent was characterized by FTIR, XRD, VSM, TGA, TEM, BET, SEM and EDS. The results showed that the maximum adsorption capacities of CR and CV were 471.46 ± 16.97 mg/g and 515.91 ± 25.12 mg/g at 318.15 K, respectively. The main adsorption mechanisms were H-bonding and electrostatic interaction. The kinetic data were in good agreement with the pseudo-second-order model and closed to adsorption equilibrium at 30 min. Thermodynamic studies showed that the adsorption on CS-BA@Fe 3 O 4 were spontaneous and endothermic. More importantly, the adsorbent exhibited excellent regeneration properties after 6 cycles and remarkable stability under harsh environments including strong acid, strong alkali, multi-salt and mixed dyes conditions. Carbon dioxide (CO 2) assisted synthesis of water-soluble silver nanoparticle with a narrow particle size distribution is reported here based on the phase-inversion procedure. Bio-derived chitosan (CS) is used to stabilize the metal nanoparticles according to its abundant functional groups. Formic acid is employed as both a solvent (for the polymer) and a reductant for in-situ reducing the silver precursor along with the solvent evaporation. CO 2 is utilized to combine with the amino groups of CS, reducing the viscosity of chitosan/formic acid solution and limiting the formation of hydrogen bonds. This promotes the stabilization and reduction efficiency of silver nanoparticles. In particular, 100% of Ag metal nanoparticles with the size of 7.5 ± 2.3 nm is successfully synthesized with the assistance of CO 2. Interestingly, the synthesized CS/Ag nanocomposites are water-soluble owing to the formation of carbamate groups. This study aimed to explore the in vitro simulated digestion and fecal fermentation behaviors of two purified exopolysaccharide fractions (EPS1 and EPS2) from Paecilomyces cicadae TJJ1213 and its effects on human gut microbiota composition. Results showed that EPS1 and EPS2 could not be digested by saliva-gastrointestinal. After fecal fermentation, however, the molecular weight of EPS1 and EPS2 significantly decreased, and the molar ratios of the monosaccharide composition remarkably changed, indicating that EPS1 and EPS2 could be degraded and consumed by human gut microbiota. Notably, EPS1 and EPS2 could significantly modulate the composition, via increasing the relative abundances of Bacteroides and Phascolarctobacterium and decreasing the pathogenic bacteria Escherichia-Shigella, Klebsiella and Fusobacterium. In addition, EPS1 and EPS2 also could promote the production of short-chain fatty acids during fermentation for 24 h. These results suggested that EPS from Paecilomyces cicadae TJJ1213 can be used as a functional food to improve health and prevent diseases by promoting gut health. The wall-associated kinases (WAKs) and WAK-like kinases (WAKLs) form a group of receptor-like kinases (RLKs) with extracellular domains tightly linked to the cell wall. The WAKs/WAKLs have been known to be involved in plant growth, development, and stress responses. However, the functions of WAKs/WAKLs are less well known in cotton. In this study, 58, 66, and 99 WAK / WAKL genes were identified in Gossypium arboreum , G. raimondii , and G. hirsutum , respectively. Phylogenetic analysis showed they were classified into five groups, with two groups specific to cotton. Collinearity analysis revealed that segmental and tandem duplications resulted in expansion of the WAK/WAKL gene family in cotton. Moreover, the Ka/Ks ratios indicated this family was exposed to purifying selection pressure during evolution. The structures of the GhWAK / WAKL genes and encoded proteins suggested the functions of WAKs/WAKLs in cotton were conserved. Transient expression of four WAK/WAKL-GFP fusion constructs in Arabidopsis protoplasts indicated that they were localized on the plasma membrane. The cis -elements in the GhWAK / WAKL promoters were responsive to multiple phytohormones and abiotic stresses. Expression profiling showed that GhWAK / WAKL genes were induced by various abiotic stresses. As the most abundant aromatic biopolymer, lignin presents great potential to produce valuable materials and chemicals. However, its large-scale value-added application is still facing many practical challenges and one of them is the unstable properties caused by lignin heterogeneity. Herein, we developed a novel "one-step-three-fractions" fractionation strategy to reduce lignin heterogeneity using aqueous two-phase system (ATPS) composed of (NH 4) 2 SO 4 and ethanol. In contrast to conventional step-wise fractionation processes, the proposed process subdivided heterogeneous lignin into three homogeneous fractions in only one step: the first fraction (F1) dissolved in the ethanol-rich top layer; the second fraction (F2) dissolved in the salt-rich bottom layer and the last fraction (F3) insoluble in both two layers. F2 presented the lowest molecular weight followed by F1 while F3 showed the highest molecular weight. With the increase of molecular weight, the contents of guaiacyl unit and β-O-4 linkage increased while the content of hydrophilic groups (carboxyl and aromatic hydroxyl) decreased significantly. Moreover, the ATPS exhibited satisfactory recyclability and the fractionation approach could be applied to different types/sources of lignin. Consequently, the work indicates that ATPS is a novel and effective way to fractionate lignin and reduce its molecular weight polydispersity and structural heterogeneity in one step. Fe/Mn bimetallic carbon materials were synthesized by combining oat and urea, followed by and carbonization processes, the activity and mechanism of the obtained materials in activating peroxymonosulfate (PMS) for sulfamethoxazole (SMX) degradation were determined. Data suggested that the obtained material (CN@FeMn-10-800) showed the optimal performance for SMX degradation under the1:8:0.05:0.05 mass ratios of oat/urea/Fe/Mn. Around 91.2 % SMX (10 mg L−1) was removed under the conditions of 0.15 g L−1 CN@FeMn-10-800 and 0.20 g L−1 PMS. The CN@FeMn-10-800 showed great adaptability under different conditions, satisfactory activation repeatability and versatility. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) demonstrated that core-shell structure with rich porous of CN@FeMn-10-800 was achieved. Quenching test and electron paramagnetic resonance (EPR) indicated that surface bound oxygen and singlet oxygen (1O 2) were the dominate reactive groups in this system. X-ray photoelectron spectroscopy (XPS) suggested that graphite N, Fe0, Fe 3 C and Mn(II) were the dominant active sites. Influenza remains a global health concern due to its potential to cause pandemics as a result of rapidly mutating influenza virus strains. Existing vaccines often struggle to keep up with these rapidly mutating flu viruses. Therefore, the development of a broad-spectrum peptide vaccine that can stimulate an optimal antibody response has emerged as an innovative approach to addressing the influenza threat. In this study, an immunoinformatic approach was employed to rapidly predict immunodominant epitopes from different antigens, aiming to develop an effective multiepitope influenza vaccine (MEV). The immunodominant B-cell linear epitopes of seasonal influenza strains hemagglutinin (HA) and neuraminidase (NA) were predicted using an antibody-peptide microarray, involving a human cohort including vaccinees and infected patients. On the other hand, bioinformatics tools were used to predict immunodominant cytotoxic T-cell (CTL) and helper T-cell (HTL) epitopes. Subsequently, these epitopes were evaluated by various immunoinformatic tools. Epitopes with high antigenicity, high immunogenicity, non-allergenicity, non-toxicity, as well as exemplary conservation were then connected in series with appropriate linkers and adjuvants to construct a broad-spectrum MEV. Moreover, the structural analysis revealed that the MEV candidates exhibited good stability, and the docking results demonstrated their strong affinity to Toll-like receptors 4 (TLR4). In addition, molecular dynamics simulation confirmed the stable interaction between TLR4 and MEVs. Three injections with MEVs showed a high level of B-cell and T-cell immune responses according to the immunological simulations in silico. Furthermore, in-silico cloning was performed, and the results indicated that the MEVs could be produced in considerable quantities in Escherichia coli (E. coli). A feature of the Chinese soft-shelled turtle (Pelodiscus sinensis) is seasonal spermatogenesis; however, the underlying molecular mechanism is not well clarified. Here, we firstly cloned and characterized P. sinensis DKKL1 , and then performed comparative genomic studies, expression analysis, and functional validation. P. sinensis DKKL1 had 2 putative N-glycosylation sites and 16 phosphorylation sites. DKKL1 also had classic transmembrane structures that were extracellularly localized. DKKL1 's genetic distance was close to turtles, followed by amphibians and mammals, but its genetic distance was far from fishes. DKKL1 genes from different species shared distinct genomic characteristics. Meanwhile, they were also relatively conserved among themselves, at least from the perspective of classes. Notably, the transcription factors associated with spermatogenesis were also identified, containing CTCF , EWSR1 , and FOXL2. DKKL1 exhibited sexually dimorphic expression only in adult gonads, which was significantly higher than that in other somatic tissues (P < 0.001), and was barely expressed in embryonic gonads. DKKL1 transcripts showed a strong signal in sperm, while faint signals were detected in other male germ cells. DKKL1 in adult testes progressively increased per month (P < 0.05), displaying a seasonal expression trait. DKKL1 was significantly downregulated in testes cells after the sex hormones (17β-estradiol and 17α-methyltestosterone) and Wnt/β-catenin inhibitor treatment (P < 0.05). Likewise, the Wnt/β-catenin inhibitor treatment dramatically repressed CTCF, EWSR1, and FOXL2 expression. Conversely, they were markedly upregulated after the 17β-estradiol and 17α-methyltestosterone treatment, suggesting that the three transcription factors might bind to different promoter regions, thereby negatively regulating DKKL1 transcription in response to the changes in the estrogen and androgen pathways, and positively controlling DKKL1 transcription in answer to the alterations in the Wnt/β-catenin pathway. Knockdown of DKKL1 significantly reduced the relative expression of HMGB2 and SPATS1 (P < 0.01), suggesting that it may be involved in seasonal spermatogenesis of P. sinensis through a positive regulatory interaction with these two genes. The CONSTANS-like (COL) genes, as a core transcription factor in the photoperiod regulation pathway, play a key role in plant reproduction development. However, their molecular characterization has rarely been studied in Pinus tabuliformis. Here, 10 PtCOL genes were identified in the P. tabuliformis genome and multiple sequence alignments have indicated that the PtCOL proteins contained highly conserved B-BOX1 and CCT domains. Sequence similarity analysis showed that PtCOL1 and PtCOL3 had the higher similarity with Norway spruce COLs (PaCOL2 and PaCOL1) and Arabidopsis COLs (AtCOL3, 4 and 5), respectively. Phylogeny and gene structure analyses revealed that PtCOLs were divided into three subgroups, each with identical or similar distributions of exons, introns, and motifs. Moreover, 10 PtCOL s were distributed on 6 chromosomes and PtCOL9 has syntenic gene pairs in both Ginkgo biloba and Sequoiadendron giganteum. Interestingly, in transcriptome profiles, most PtCOL s exhibited a diurnal oscillation pattern under both long (LD) and short (SD) day conditions. Additionally, PtCOL s were highly expressed in needles and female cones, and showed different spatial expression patterns. Among the ten PtCOL s, PtCOL1/3 heterologous overexpression Arabidopsis displayed a delayed-flowering phenotype under SD, indicating that they are likely to play a crucial role in the reproductive development. Additionally, PtCOL1 and PtCOL3 were not only capable of interacting with each other, but they were each capable of interacting with themselves. Furthermore, PtCOL1 and PtCOL3 were also involved in the MADS-box protein-protein interaction (PPI) network in P. tabuliformis cone development. Direct interactions of PtDAL11 with PtCOL1/3 impeded PtCOL1/3 translocation into the nucleus. In summary, this study provided comprehensive understanding for the functions of the PtCOL gene family and revealed their biological roles in the photoperiod-dependent P. tabuliformis cone development. Facing the increasing electromagnetic interference (EMI) pollution in the living environment, it is a new trend to explore an efficient EMI shielding material with facile fabrication and a wide range of application scenarios. A hydrophobic composite paper composed of silver nanowires (AgNWs) and kapok microfibers cellulose (MFC) was modified by methyl trimethoxy silane (MTMS) through a simple method. As a result, the composite paper has a good EMI shielding effectiveness (EMI SE) of 61.7 dB with electrical conductivity of 695.41 S/cm. The modification of MTMS improved the thermal stability performance of composite paper, which also increased its water contact angle to 113°. The free silver ions (Ag+) released from AgNWs can kill surrounding microbial bacteria, endowing the composite paper with good antibacterial property. A polyhydroxyalkanoate (PHA) magnetic microsphere was designed for one-step purification and immobilization of a novel carbonyl reductase (RLSR5) from recombinant Escherichia coli lysate. The hydrophobic core of this microsphere was composed of a highly biocompatible polymer, poly(3-hydroxybutyrate- co -3-hydroxyhexanoate) (PHBHHx), in which magnetic Fe 3 O 4 particles were embedded during solvent evaporation. The hydrophilic shell of the fusion protein formed by PHA particle-binding protein (PhaP) and RLSR5 (PR) was expressed in recombinant E. coli. The magnetic core of Fe 3 O 4 @PHBHHx directly purified the hydrophilic shell from the E. coli lysate, and the two self-assembled to form Fe 3 O 4 @PHBHHx-PR through hydrophobic and hydrophilic interactions, eliminating the separation of the fusion protein. The microstructure, magnetic properties, morphology, size, and dispersion of Fe 3 O 4 @PHBHHx-PR were investigated by XRD, VSM, SEM, TEM, elemental mapping and DLS. It was found that Fe 3 O 4 @PHBHHx-PR correctly assembled, with a well dispersed spherical structure at the nanoscale and superparamagnetism properties. The amount of RLSR5 immobilized on PHA microspheres reached 121.9 mg/g. The Fe 3 O 4 @PHBHHx-PR was employed to synthesize (R)-tolvaptan with 99 % enantiomeric excess and 97 % bioconversion efficiency, and the catalyst maintained 78.6 % activity after 10 recovery cycles. Carboxymethyl chitosan and resistant starch exhibit good performance in diabetes regulation. We prepared carboxymethyl chitosan - resistant starch complex. Test the properties of composite resistant starch by using X-ray diffraction, water contact angle, infrared spectroscopy, and scanning electron microscopy, interactions with intestinal microbiota and mouse experiments were also conducted. The results indicated that the composite resistant starch had a good effect on promoting the proliferation of probiotics on Bifidobacterium and a significant inhibitory effect on Escherichia coli than resistant starch (P < 0.05). After administration, the water intake and weight of diabetic mice were significantly reduced. The blood glucose of diabetic mice was also reduced, and oral glucose tolerance showed that the glucose degradation rates of composite resistant starch were significantly improved compared to model mice. Cholesterol, triglycerides, high-density lipoprotein and low-density lipoprotein were significantly lower than those in the diabetes group (P < 0.05). Grain shape and plumpness affect barley yield. Despite numerous studies on shrunken endosperm mutants in barley, their molecular mechanism and application potential in the food industry are largely unknown. Here, map-based cloning, co-segregation analyses, and allelic variant validation revealed that the loss of HORVU6Hr1G037950 encoding an ADP-glucose transporter caused the shrunken endosperm in sex1. Haplotype analysis suggested that hap4 in the promoter sequence was positively related to the hundred-grain weight showing a breeding potential. A pair of near-isogenic lines targeting HORVU6Hr1G037950 was produced and characterized to investigate molecular mechanisms that SEX1 regulates endosperm development. Results presented that the absence of the SEX1 gene led to the decrease of starch content and A-type granules size, the increase of β-glucan, protein, gelatinization temperature, soluble sugar content, amylopectin A chains, and B1 chains. Enzymatic activity, transcriptome and metabolome analyses revealed the loss of SEX1 results in an impaired ADP-glucose-to-starch conversion process, consequently leading to higher soluble sugar contents and lower starch accumulation, thereby inducing a shrunken-endosperm phenotype in sex1. Taken together, this study provides new insights into barley grain development, and the elevated protein and β-glucan contents of the whole meal in sex1 imply its promising application in the food industry.
