Aromatic amines (AA) are carcinogenic compounds that can enter the human body through many sources, one of the most important being tobacco smoke. They are excreted with the urine, from which they can be extracted and measured. To that end, hollow fiber-liquid-phase microextraction (HF-LPME) and parallel artificial liquid membrane extraction (PALME) were optimized for the analysis of representative aromatic amines, as alternatives to liquid–liquid extraction (LLE). Relevant extraction parameters, namely organic solvent, extraction time, agitation speed, and acceptor solution pH, were studied, and the two optimized techniques—HF-LPME: dihexyl ether, 45 min, 250 rpm, and pH 1; PALME: undecane, 20 min, 250 rpm and pH 1—were compared. Comparison of the optimized methods showed that significantly higher recoveries could be obtained with PALME than with HF-LPME. Therefore, PALME was further validated. The results were successful for nine different AA, with regression coefficients (R2) of at least 0.991, limits of detection (LOD) of 45–75 ng/L, and repeatability and peak area relative standard deviations (RSD) below 20%. Furthermore, two urine samples from smokers were measured as proof of concept, and 2-methylaniline was successfully quantified in one of them. These results show that PALME is a great green alternative to LLE. Not only does it use much smaller volumes of toxic organic solvents, and sample—enabling the study of samples with limited available volumes—but it is also less time consuming and labor intensive, and it can be automated. Migration of emerging contaminants (ECs) from pipes into water is a global concern due to potential human health effects. Nevertheless, a review of migration ECs from pipes into water distribution systems is presently lacking. This paper reviews, the reported occurrence migration of ECs from pipes into water distribution systems in the world. Furthermore, the results related to ECs migration from pipes into water distribution systems, their probable sources, and their hazards are discussed. The present manuscript considered the existing reports on migration of five main categories of ECs including microplastics (MPs), bisphenol A (BPA), phthalates, nonylphenol (NP), perfluoroalkyl, and polyfluoroalkyl substances (PFAS) from distribution network into tap water. A focus on tap water in published literature suggests that pipes type used had an important role on levels of ECs migration in water during transport and storage of water. For comparison, tap drinking water in contact with polymer pipes had the highest mean concentrations of reviewed contaminants. Polyvinyl chloride (PVC), polyamide (PA), polypropylene (PP), polyethylene (PE), and polyethylene terephthalate (PET) were the most frequently detected types of microplastics (MPs) in tap water. Based on the risk assessment analysis of ECs, levels of perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluorohexane sulfonate (PFHxS), and perfluorooctane sulfonate (PFOS) were above 1, indicating a potential non-carcinogenic health risk to consumers. Finally, there are still scientific gaps on occurrence and migration of ECs from pipes used in distribution systems, and this needs more in-depth studies to evaluate their exposure hazards on human health.

The analysis of fatty acid methyl esters (FAMEs) is of high relevance for monitoring and control of various industrial processes and biological systems. In this study, a novel, green analytical approach for the determination of 24 FAMEs from aqueous samples is proposed, which is based on a headspace solid-phase microextraction (SPME) arrow followed by gas chromatography coupled to tandem mass spectrometry (GC–MS/MS). The method was substantially accelerated to a run time of 44 min per sample by thorough optimization and automation of the relevant parameters. The limiting parameters, mostly based on expediting equilibrium attainment, were found to be parameters of extraction: material, pH, time, and temperature, which were optimized to divinylbenzene polydimethylsiloxane (DVB-PDMS), pH 2, 20 min, and 70 °C, respectively. The optimization and automation of the method led to low method detection limits (9–437 ng L−1) and high selectivity. Evaluation of the method on real samples was done by analyzing the aqueous phase of a bioreactor, whereby the matrix effect could be greatly reduced due to dilution and headspace sampling. The rapid, sensitive, selective, and matrix-reduced approach is found to be not only a novel method for water analysis but is promising for further applications, e.g., with solid and gaseous samples containing FAMEs. Complex multivariate datasets are generated in environmental non-target screening (NTS) studies covering different sampling locations and times. This study presents a comprehensive chemometrics-based data processing workflow to reveal hidden data patterns and to find a subset of discriminating features between samples. We used ANOVA-simultaneous component analysis (ASCA) to disentangle the influence of spatial and seasonal effects as well as their interaction on a multiclass dataset. The dataset was obtained by a Chemcatcher passive sampler (PS) monitoring campaign of three small streams and one major river over four sampling periods from spring to summer. Monitoring of small streams is important as they are impacted by non-point source introduction of organic micropollutants (OMPs). The use of a PS provides a higher representativeness of sampling, and NTS broadens the range of detectable OMPs. A comparison of ASCA results of target analysis and NTS showed for both datasets a dominant influence of different sampling locations and individual temporal pollution patterns for each river. With the limited set of target analytes, general seasonal pollution patterns were apparent, but NTS data provide a more holistic view on site-specific pollutant loads. The similarity of temporal pollution patterns of two geographically close small streams was revealed, which was not observed in undecomposed data analysis like principal component analysis (PCA). With a complementary partial least squares-discriminant analysis (PLS-DA) and Volcano-based prioritization strategy, 223 site- and 45 season-specific features were selected and tentatively identified.

Laser ablation in liquids has been established as a scalable preparation method of nanoparticles for various applications. Particularly for materials prone to oxidation, it is established to suppress oxidation by using organic solvents as a liquid medium. While this often functionalizes the nanoparticles with a carbon shell, the related chemical processes that result from laser-induced decomposition reactions of the organic solvents remain uncertain. Using a systematic series of C₆ solvents complemented by n-pentane and n-heptane during the nanosecond laser ablation of gold, the present study focuses on the solvent-dependent influence on gas formation rates, nanoparticle productivity, and gas composition. Both the permanent gas and hydrogen formation was found to be linearly correlated with ablation rate, ΔHvₐp, and pyrolysis activation energy. Based on this, a decomposition pathway linked to pyrolysis is proposed allowing the deduction of first selection rules for solvents that influence the formation of carbon or permanent gases. in press

Compound-specific stable isotope analysis (CSIA) is a unique analytical technique for determining small variations in isotope ratios of light isotopes in analytes from complex mixtures. A problem of CSIA using gas chromatography (GC) and liquid chromatography-isotope ratio mass spectrometry (LC-IRMS) is that any structural information of the analytes is lost due to the processes involved in determining the isotope ratio. We measured the carbon isotope composition of amino acids and glucose in the cestode Schistocephalus solidus and in liver and muscle tissues of its second intermediate host, the three-spined stickleback (Gasterosteus aculeatus), over the course of 90 days in a controlled infection experiment. Similar linear regressions of δ13C values over time and low trophic fractionation of essential amino acids indicate that the parasite assimilates nutrients from sources closely connected to the liver metabolism of its host. Biosynthesis of glucose in the parasite might occur from the glucogenic precursors alanine, asparagine and glutamine and with an isotope fractionation of − 2 to – 3 ‰ from enzymatic reactions, while trophic fractionation of glycine, serine and threonine could be interpreted as extensive nutrient conversion to fuel parasitic growth through one-carbon metabolism. Trophic fractionation of amino acids between sticklebacks and their diets was slightly increased in infected compared to uninfected individuals, which could be caused by increased (immune-) metabolic activities due to parasitic infection. Our results show that compound-specific stable isotope analysis has unique opportunities to study host and parasite physiology.

Some aromatic amines (AA) have been classified as carcinogens to humans. After entering the body, mainly through tobacco smoke, they can be detected in urine. Thus, their trace analysis as biomarkers in biofluids is of high relevance and can be achieved with gas chromatography (GC–MS), usually after derivatization. This study compares three gas chromatographic methods for the analysis of ten iodinated derivatives of AA: GC–MS in single-ion monitoring (SIM) mode with (1) electron ionization (GC-EI-MS) and (2) negative chemical ionization (GC-NCI-MS), and (3) GC-EI-MS/MS in multiple reaction monitoring (MRM) mode using electron ionization. All methods and most analytes showed good coefficients of determination (R2 > 0.99) for broad linear ranges covering three to five orders of magnitude in the picogram-per-liter to nanogram-per-liter range, with one and two exceptions for (1) and (2) respectively. Excellent limits of detection (LODs) of 9–50, 3.0–7.3, and 0.9–3.9 pg/L were observed for (1), (2), and (3) respectively, and good precision was achieved (intra-day repeatability α = 0.05) higher concentrations among smokers. Background Stone wool is an inorganic mineral insulation material increasingly used to reduce the climate impact of buildings. The acute and chronic ecotoxicological potential of stone wool eluates have been studied in a battery of standardized laboratory ecotoxicological tests. The experiments were conducted with stone wool test materials in fibrous and milled form, with and without the presence of organic binder. For the preparation of eluates, the OECD protocol on the transformation/dissolution of metals and metal compounds was applied. The resulting eluates were used in acute tests, i.e., bioluminescence test with Aliivibrio fischeri (DIN EN ISO 11348-1:2009), algae growth test with Desmodesmus subspicatus (OECD No. 201) and immobilization test with Daphnia magna (OECD No. 202), as well as chronic tests, i.e., the Daphnia magna reproduction test (OECD No. 211) and the nematode growth and reproduction test with Caenorhabditis elegans (ISO 10872:2010). Results While no acute or chronic ecotoxicological effects of the eluates were observed for fibrous stone wool material, the milled test materials showed some chronic effects on aquatic invertebrates. Depending on the test materials and concentrations of milled stone wool used in the eluate preparation, these chronic effects included significant stimulation or inhibition of daphnid reproduction and nematode growth. The chemical analysis conducted in parallel to the ecotoxicological assessment indicated no leaching of organic substances from the applied binder or mineral oils and no formation of nanoparticles by the milling of stone wool. Furthermore, ICP-MS and ICP-OES analysis of eighteen elements revealed that only aluminum and nickel could be quantified in the eluates, at concentrations of approximately 750 µg/L and 7 µg/L, respectively. Conclusions Based on the present ecotoxicological assessment, eluates from stone wool fibers cannot be considered as chemically hazardous to the aquatic environment. However, additional investigations of the ecotoxicological potential of the milled material and the environmental exposure of stone wool products are necessary for a complete evaluation of potentially negative effects of stone wool insulation materials.

Effect-directed analysis (EDA) that combines effect-based methods (EBMs) with high-performance thin-layer chromatography (HPTLC) is a useful technique for spatial, temporal, and process-related effect evaluation and may provide a link between effect testing and responsible substance identification. In this study, a yeast multi endocrine-effect screen (YMEES) for the detection of endocrine effects is combined with HPTLC. Simultaneous detection of estrogenic, androgenic, and gestagenic effects on the HPTLC plate is achieved by mixing different genetically modified Arxula adeninivorans yeast strains, which contain either the human estrogen, androgen, or progesterone receptor. Depending on the yeast strain, different fluorescent proteins are formed when an appropriate substance binds to the specific hormone receptor. This allows to measure hormonal effects at different wavelengths. Two yeast cell application approaches, immersion and spraying, are compared. The sensitivity and reproducibility of the method are shown by dose-response investigations for reference compounds. The spraying approach indicated similar sensitivities and higher precisions for the tested hormones compared to immersion. The EC10s for estrone (E1), 17β-estradiol (E2), 17α-ethinylestradiol (EE2), 5α-dihydrotestosterone (DHT), and progesterone (P4) were 95, 1.4, 10, 7.4, and 15 pg/spot, respectively. Recovery rates of E1, E2, EE2, DHT, and P4 between 88 and 120% show the usability of the general method in combination with sample enrichment by solid phase extraction (SPE). The simultaneous detection of estrogenic, androgenic, and gestagenic effects in wastewater and surface water samples demonstrates the successful application of the YMEES in such matrices. This promising method allows us to identify more than one endocrine effect on the same HPTLC plate, which saves time and material. The method could be used for comparison, evaluation, and monitoring of different river sites and wastewater treatment steps and should be tested in further studies.

Perovskite nanomaterials such as LaMnO3, LaFeO3, and LaCoO3 were synthesized in a spray flame from metal nitrates dissolved in combustible liquids. The addition of low-boiling solvents such as 2-ethylhexanoic acid (2-EHA) to the ethanol-based solutions supports the formation of phase-pure particles with unimodal particle-size distribution in the 10-nm range attributed to enhanced evaporation through micro-explosions. Nevertheless, in many cases, a second particle mode with sizes of a few hundred nanometers is formed. In this paper, we investigate two possible reasons for the appearance of large particles. Firstly, we analyze the effect of the oxygen dispersion gas flow applied in the two-fluid nozzle on the droplet size distributions of burning sprays using phase Doppler anemometry. We identified that an increase of the dispersion gas flow significantly decreases the number concentration of large droplets (>30 μm), which causes a significant increase of the BET surface area of as-synthesized LaMnO3 and LaCoO3 with increasing dispersion gas flow from 60 m2/g (5 slm dispersion gas) to 100 m2/g (8 slm). Secondly, the esterification in the mixture of solvents towards ethyl-2-ethylhexanoate, which is associated with the release of water as a byproduct, was analyzed by GC/MS. The ester concentration in the iron-containing solution was found to be up to nine times higher than in cobalt or manganese precursor solutions. Simultaneously, the produced LaFeO3 materials show lower BET surface areas and the increasing dispersion gas flow has a minor effect on this material than on the cobalt and manganese perovskite cases. We attribute this to the fact that water formed during esterification forces the hydrolysis of iron nitrate and the formation of large particles within the droplets.

A GCxGC-MS system was employed with a non-polar × mid-polar column set for the metabolic non-target analysis of Cobetia marina, the model bacteria for marine biofouling. C. marina was treated with ozone to investigate the intracellular metabolic state change under oxidative stress. A minimal inhibitory concentration test was involved to guarantee that the applied ozone dosages were not lethal for the cells. In this study, non-target analyses were performed to identify the metabolites according to the NIST database. As a result, over 170 signals were detected under normal living conditions including 35 potential metabolites. By the comparison of ozone-treated and non-treated samples, five compounds were selected to describe observed trends of signals in the contour plots. Oleic acid exhibited a slight growth by increasing ozone dosage. In contrast, other metabolites such as the amino acid l-proline showed less abundance after ozone treatment, which was more evident once ozone dosage was raised. Thus, this work could provide a hint for searching for up/downregulating factors in such environmental stress conditions for C. marina.

Reversed-phase liquid chromatography (RPLC) used for water analysis is not ideal for the analysis of highly polar and ionic contaminants because of low retention. Capillary electrophoresis (CE), on the other hand, is perfectly suited for the separation of ionic compounds but rarely applied in environmental analysis due to the weak concentration sensitivity when coupled to mass spectrometry (MS). However, novel interface designs and MS technology strongly improve the sensitivity. Here, a method is presented enabling the screening of anionic micropollutants in drinking water without sample pretreatment by coupling of CE to an Orbitrap mass spectrometer by a nanoflow sheath liquid interface. Targeted analysis of halogenated acetic acids, trifluoromethanesulfonic acid, and perfluorooctanoic and perfluorooctanesulfonic acid was conducted in drinking water samples which were chlorinated for disinfection. A bare fused silica capillary with an optimized background electrolyte (BGE) for separation consisting of 10% acetic acid with 10% isopropanol with large volume sample injection and optimized interface parameters offer limits of quantification in the range of 0.993) and repeatability (14% standard deviation in area). Concentrations of the target analytes ranged from 0.1 to 6.2 μg/L in the water samples. Masses corresponding to halogenated methanesulfonic acids have been found as suspects and were subsequently verified by standards. Mono-, dichloro-, and bromochloro methanesulfonic acid were quantified in a range of 0.2 to 3.6 μg/L. Furthermore, five sulfonic acids, four organosulfates, and the artificial sweeteners acesulfame and cyclamate as well as inorganics such as halides, halogenates, phosphate, and sulfate could be determined as suspects among more than 300 features in a non-targeted screening. Overall, this approach demonstrates the great potential of CE-nanoESI-MS for the screening of ionic contaminants in environmental samples, complementary to chromatographic approaches. Advancements in column technology resulted in smaller particles and more efficient phases. In parallel, the use of columns with reduced dimensions is becoming more common. This means the effective column volume is also decreased, thereby making the systems more susceptible to effects of band broadening due to extra-column volume. Despite these trends and the fact that a growing number of miniaturized liquid chromatography systems are being offered commercially, manufacturers often stick to the modular concept with dedicated units for pumps, column oven, and detectors. This modular design results in long connection capillaries, which leads to extra-column band broadening and consequently prevents the exploitation of the intrinsic efficiency of state-of-the-art columns. In particular, band broadening post column has a considerable negative effect on efficiency. In this study, mass flow and concentration-dependent detectors were examined for their influence on band broadening using a micro-LC system. A mass spectrometric detector, an evaporative light scattering detector, two UV detectors, and a previously undescribed fluorescence detector were compared. The influence on efficiency is compared using plate height vs linear velocity data and peak variance. It is shown that an increase in the inner diameter after the post-column transfer capillary leads to significant loss in plate height. Comparing the UV detectors, it could be shown that the dispersion was reduced by 38% by the reduction of the post-column volume. The largest variance was found for the evaporative light scattering detector, which was 368% higher compared to the variance of the detector with the least effect on band broadening.

With the increasing demand for ultrapure water in the pharmaceutical and semiconductor industry, the need for precise measuring instruments for those applications is also growing. One critical parameter of water quality is the amount of total organic carbon (TOC). This work presents a system that uses the advantage of the increased oxidation power achieved with UV/O3 advanced oxidation process (AOP) for TOC measurement in combination with a significant miniaturization compared to the state of the art. The miniaturization is achieved by using polymer-electrolyte membrane (PEM) electrolysis cells for ozone generation in combination with UV-LEDs for irradiation of the measuring solution, as both components are significantly smaller than standard equipment. Conductivity measurement after oxidation is the measuring principle and measurements were carried out in the TOC range between 10 and 1000 ppb TOC. The suitability of the system for TOC measurement is demonstrated using the oxidation by ozonation combined with UV irradiation of defined concentrations of isopropyl alcohol (IPA).

Wastewater treatment plants (WWTPs) may represent point sources for microplastic discharge into the environment. Quantification of microplastic in effluents of WWTPs has been targeted by several studies although standardized methods are missing to enable a comparability of results. This study discusses theoretical and practical perspectives on best practices for microplastic sampling campaigns of WWTPs. One focus of the study was the potential for synergies between thermoanalytical and spectroscopic analysis to gain more representative sampling using the complementary information provided by the different analytical techniques. Samples were obtained before and after sand filtration from two WWTPs in Germany using cascade filtration with size classes of 5,000 - 100 µm, 100 - 50 µm, and 50 - 10 µm. For spectroscopic methods samples were treated by a Fenton process to remove natural organic matter, whereas TED-GC-MS required only sample extraction from the filter cascade. µFTIR spectroscopy was used for the 100 µm and 50 µm basket filters and µRaman spectroscopy was applied to analyze particles on the smallest basket filter (10 µm). TED-GC-MS was used for all size classes as it is size independent. All techniques showed a similar trend, where PE was consistently the most prominent polymer in WWTP effluents. Based on this insight, PE was chosen as surrogate polymer to investigate whether it can describe the total polymer removal efficiency of tertiary sand filters. The results revealed no significant difference (ANOVA) between retention efficiencies of tertiary sand filtration obtained using only PE and by analyzing all possible polymers with µFTIR and µRaman spectroscopy. Findings from this study provide valuable insights on advantages and limitations of cascade filtration, the benefit of complementary analyses, a suitable design for future experimental approaches, and recommendations for future investigations.

Reaction mechanisms of sulfate radical with bromide and organic substances can be largely altered at elevated temperature in heat-activated persulfate process due to different activation energies. This study investigated kinetics and stoichiometry of bromide oxidation in the heat-activated persulfate process. We postulated that reaction of sulfate radical with persulfate, which has relatively low reaction rate (6.3 × 105 M−1s−1), may become an important reaction at elevated temperatures. Persulfate decomposition was inhibited in the presence of sulfate radical scavenger phenol at low temperature (40 °C) whereas phenol affected persulfate decomposition only at 50 °C, which can be explained by a high activation energy (estimated activation energy: 311 kJ/mol). Bromate formation was inhibited in the presence of phenol. However, phenol oxidation was not affected by the presence of Br−, even though Br− reacts faster with sulfate radicals than phenol and was present in excess. This indicated that formed reactive bromine species such as Br and Br2 − oxidize phenol under reformation of bromide. Formation of insoluble products was also observed indicating polymerization of phenol. This can be explained by lack of dissolved oxygen under conditions of heat-activation.

N-containing heterocycles (NCHs) are largely used as precursors for pharmaceuticals and can enter the environment. Some NCHs have been shown to be toxic, persistent, and very mobile in the environment. Thus, they have received increasing attention in the past years. However, the analysis of these polar compounds in environmental samples is still a challenge for liquid chromatography. This paper investigates the use of mixed-mode liquid chromatography (MMLC), which has reversed-phase and ion exchange characteristics for measurements of NCHs in water. NCHs with low pKa (i.e., Ka (i.e., > 5) interact by a mixture of reversed-phase/ion exchange/HILIC mechanism. It was also shown that the presented method performs well in the quantification of the majority of the selected NCHs in surface water with MDLs between 3 and 6 μg/L, a low matrix effect and recoveries in the range of 77–96% except for pyridazine exhibiting 32% were achieved. The method was successfully employed to follow the degradation of NCHs in ozonation.

High-resolution mass spectrometry is widely used in many research fields allowing for accurate mass determinations. In this context, it is pretty standard that high-resolution profile mode mass spectra are reduced to centroided data, which many data processing routines rely on for further evaluation. Yet information on the peak profile quality is not conserved in those approaches; i.e., describing results reliability is almost impossible. Therefore, we overcome this limitation by developing a new statistical parameter called data quality score (DQS). For the DQS calculations, we performed a very fast and robust regression analysis of the individual high-resolution peak profiles and considered error propagation to estimate the uncertainties of the regression coefficients. We successfully validated the new algorithm with the vendor-specific algorithm implemented in Proteowizard’s msConvert. Moreover, we show that the DQS is a sum parameter associated with centroid accuracy and precision. We also demonstrate the benefit of the new algorithm in nontarget screenings as the DQS prioritizes signals that are not influenced by non-resolved isobaric ions or isotopic fine structures. The algorithm is implemented in Python, R, and Julia programming languages and supports multi- and cross-platform downstream data handling.

The anthropogenic entry of organic micropollutants into the aquatic environment leads to a potential risk for drinking water resources and the drinking water itself. Therefore, sensitive screening analysis methods are needed to monitor the raw and drinking water quality continuously. Non-target screening analysis has been shown to allow for a more comprehensive investigation of drinking water processes compared to target analysis alone. However, non-target screening is challenging due to the many features that can be detected. Thus, data processing techniques to reduce the high number of features are necessary, and prioritization techniques are important to find the features of interest for identification, as identification of unknown substances is challenging as well. In this study, a drinking water production process, where drinking water is supplied by a water reservoir, was investigated. Since the water reservoir provides surface water, which is anthropogenically influenced by wastewater treatment plant (WWTP) effluents, substances originating from WWTP effluents and reaching the drinking water were investigated, because this indicates that they cannot be removed by the drinking water production process. For this purpose, ultra-performance liquid chromatography coupled with an ion-mobility high-resolution mass spectrometer (UPLC-IM-HRMS) was used in a combined approach including target, suspect and non-target screening analysis to identify known and unknown substances. Additionally, the role of ion-mobility-derived collision cross sections (CCS) in identification is discussed. To that end, six samples (two WWTP effluent samples, a surface water sample that received the effluents, a raw water sample from a downstream water reservoir, a process sample and the drinking water) were analyzed. Positive findings for a total of 60 substances in at least one sample were obtained through quantitative screening. Sixty-five percent (15 out of 23) of the identified substances in the drinking water sample were pharmaceuticals and transformation products of pharmaceuticals. Using suspect screening, further 33 substances were tentatively identified in one or more samples, where for 19 of these substances, CCS values could be compared with CCS values from the literature, which supported the tentative identification. Eight substances were identified by reference standards. In the non-target screening, a total of ten features detected in all six samples were prioritized, whereby metoprolol acid/atenolol acid (a transformation product of the two β-blockers metoprolol and atenolol) and 1,3-benzothiazol-2-sulfonic acid (a transformation product of the vulcanization accelerator 2-mercaptobenzothiazole) were identified with reference standards. Overall, this study demonstrates the added value of a comprehensive water monitoring approach based on UPLC-IM-HRMS analysis. Coffee is typically brewed by extracting roasted and milled beans with hot water, but alternative methods such as cold brewing became increasingly popular over the past years. Cold-brewed coffee is attributed to health benefits, fewer acids, and bitter substances. But the preparation of cold brew typically needs several hours or even days. To create a cold-brew coffee within a few minutes, we present an approach in which an ultrashort-pulsed laser system is applied at the brewing entity without heating the powder suspension in water, efficiently extracting caffeine and aromatic substances from the powder. Already 3 min irradiation at room temperature leads to a caffeine concentration of 25 mg caffeine per 100 ml, comparable to the concentrations achieved by traditional hot brewing methods but comes without heating the suspension. Furthermore, the liquid phase’s alkaloid content, analyzed by reversed-phase liquid chromatography coupled to high-resolution mass spectrometry, is dominated by caffeine and trigonelline and is comparable to traditional cold-brewed coffee rather than hot-brewed coffee. Furthermore, analyzing the head-space of the prepared coffee variants, using in-tube extraction dynamic head-space followed by gas chromatography coupled to mass spectrometry, gives evidence that the lack of heating leads to the preservation of more (semi-)volatile substances like pyridine, which provide cold-brew coffee its unique taste. This pioneering study may give the impetus to investigate further the possibility of cold-brewing coffee, accelerated by more than one order of magnitude, using ultrafast laser systems.

Cigarette butts (CBs) are some of the most abundant waste items in the environment and may contain high levels of different toxic chemicals, such as aromatic amines (AAs). However, to this date, there is no comprehensive study on the role of CBs in the emission of AAs into the environment. The present study investigated for the first time the concentration levels of 10 primary aromatic amines (PAAs), including ANL, p-TOL, m-TOL, o-TOL, 2,6-DMA, o-ASD, 2-NA, 1-NA, 3-ABP, and 4-ABP that were measured and compared in unsmoked cigarette, freshly smoked CBs, and CBs collected from urban streets (named here aged CBs). Permanently charged and ionizable organic compounds (IOC) are a large and diverse group of compounds belonging to many contaminant classes, including pharmaceuticals, pesticides, industrial chemicals, and natural toxins. Sorption and mobility of IOCs are distinctively different from those of neutral compounds. Due to electrostatic interactions with natural sorbents, existing concepts for describing neutral organic contaminant sorption, and by extension mobility, are inadequate for IOC. Predictive models developed for neutral compounds are based on octanol-water partitioning of compounds (Kow) and organic-carbon content of soil/sediment, which is used to normalize sorption measurements (KOC). We revisit those concepts and their translation to IOC (Dowand DOC) and discuss compound and soil properties determining sorption of IOC under water saturated conditions. Highlighting possible complementary and/or alternative approaches to better assess IOC mobility, we discuss implications on their regulation and risk assessment. The development of better models for IOC mobility needs consistent and reliable sorption measurements at well-defined chemical conditions in natural porewater, better IOC-, as well as sorbent characterization. Such models should be complemented by monitoring data from the natural environment. Interpretation of stable isotope data is of upmost importance in ecology to build sound models for the study of animal diets, migration patterns and physiology. However, our understanding of stable isotope fractionation and incorporation into consumer tissues is still limited. We therefore measured the δ13C values of individual amino acids over time from muscle and liver tissue of three-spined sticklebacks (Gasterosteus aculeatus) on a high protein diet. The δ13C values of amino acids in the liver quickly responded to small shifts of under ± 2.0‰ in dietary stable isotope compositions on 30-day intervals. We found on average no trophic fractionation in pooled essential (muscle, liver) and non-essential (muscle) amino acids. Negative Δδ13C values of − 0.7 ± 1.3‰ were observed for pooled non-essential (liver) amino acids and might indicate biosynthesis from small amounts of dietary lipids. Trophic fractionation of individual amino acids is reported and discussed, including unusual Δδ13C values of over + 4.9 ± 1.4‰ for histidine. Arginine and lysine showed the lowest trophic fractionation on individual sampling days and might be useful proxies for dietary sources on short time scales. We suggest further investigations using isotopically enriched materials to facilitate the correct interpretation of ecological field data.

Microplastics analysis in solid environmental samples like sediments and soils requires complex preparation steps to eliminate interfering matrix components. This is often combined with the use of highly concentrated, expensive, and environmentally harmful chemicals. These sample preparation methods can be very time-consuming. Hence, with regard to green analytical chemistry, reduction of emissions and hazards, these sample preparation methods should be modified or avoided, and more environmentally friendly methods should be developed. The characterization and evaluation of the optimized hydrophobicity-water/air-based enrichment cell for microplastics (µSEP) are presented in this study. The separation mechanism of µSEP is based on the hydrophobic adhesion of microplastic on finely dispersed upstreaming air bubbles, serving as a sustainable and practical microplastic sample preparation method. The separation mechanism requires only water and air to considerably reduce the matrix for microplastic analysis. Using soil samples as an example, the applicability of the µSEP sample preparation method for the analysis of microplastics by Raman microspectroscopy (µRaman) and thermal extraction-desorption gas chromatography-mass spectrometry (TED-GC–MS) is demonstrated. Within a sample preparation time of 60 min, for polyethylene terephthalate (PET), an average recovery of 77% ± 11%, for polystyrene (PS), 42% ± 11%, and for polypropylene (PP) 65% ± 12% was determined. Due to the complex manufacturing process of therapeutic monoclonal antibodies, it is hardly possible to produce an identical copy of the original product (originator). Consequently, follow-on products (biosimilars) must demonstrate their efficacy being similar to the originator in terms of structure and function. During this process, a variety of analytical methods are required for this purpose. This study focuses on three particularly relevant analytical techniques: high-resolution mass spectrometry, fragment crystallisable (Fc) affinity chromatography, and two-dimensional peptide mapping. Each analytical method proved able to identify specific differences between originator and biosimilar. High-resolution mass spectrometry was used to characterize the glycan pattern. It was shown that a trastuzumab biosimilar did not have the G0:G0F sugar modification identified in the originator. The application of affinity chromatography to rituximab showed that originator and biosimilar interacted differently with the immobilized Fc receptor. Furthermore, 2D-HPLC peptide mapping demonstrated the influence of orthogonality of separation dimensions, leading to differentiation of a rituximab originator and biosimilar.

Parasitism is commonly recognised as a consumer strategy, although, the interaction of parasites in communities and ecosystems are generally poorly understood. As parasites are integral parts of food webs, analysis of the trophic interactions between parasites and hosts was assessed through comparison of stable isotope ratios of carbon (13C/12C) and nitrogen (15N/14N). Largemouth yellowfish (Labeobarbus kimberleyensis) infected with the Asian tapeworm (Schyzocotyle acheilognathi) were collected from the Vaal Dam. Signatures of δ13C and δ15N were assessed in host muscle and liver tissue, and cestodes using an elemental analyser coupled with an isotope ratio-mass spectrometer (EA-IRMS). Hosts were enriched by 4.1‰ in the heavy nitrogen isotope with respect to the S. acheilognathi and therefore occupy a higher trophic position than the parasite. Comparison of δ13C indicates that dietary sources of carbon in cestodes are derived from the host liver. Comparison of stable isotope signatures between Paradiplozoon ichthyoxanthon (another common parasite of the Largemouth yellowfish in the Vaal River) and S. acheilognathi showed that the monogenean was enriched by 5.3‰ in 15N which accounts for a difference of almost two trophic positions. Isotope differences in the host–parasite system considered indicate that differences can be related to the mode of nutrient acquisition employed by host and parasites. Cestodes, being depleted in both 13C and 15N relative to the host and monogenean (P. ichthyoxanthon), indicate that S. acheilognathi assimilates nutrients derived from the host metabolism which are released from the liver.

Chlorine dioxide (ClO2) is used for disinfection of water and pollutant control. Recently it was shown that free available chlorine (FAC) is formed during the oxidation of organic compounds with ClO2. To investigate the moieties in natural organic matter (NOM) responsible for the FAC formation, reactions of ClO2 with phenols and olefins were investigated. Thereby primary yields of the inorganic by-products chlorite, chloride, FAC, and chlorate were determined and compared with Suwannee River natural organic matter (SRNOM). The reaction of ClO2 with the studied model compounds (vanillin, hydroquinone, indigotrisulfonate, and dimedone) revealed very different inorganic by-products. The main inorganic by-product of all investigated compounds was chlorite (>50% per consumed ClO2). Besides chlorite, vanillin formed 35% chloride and 4% chlorate, indigotrisulfonate 25% chloride, 7% FAC and 3% chlorate and dimedone 50% chloride and 4% chlorate. Hydroquinone formed exclusively chlorite (90%). Yields of FAC formed by ClO2 oxidation of SRNOM were significant (22%), which principally could result in halogenated organic by-products. Additionally, chlorite (63%), chloride (8%) and chlorate (5%) were formed. The yield of FAC decreased significantly after pre-ozonation, which can be explained by ozone induced formation of poly-hydroxylated aromatic compounds, which may mainly form chlorite in analogy to hydroquinone.

Regarding endocrine disrupting chemicals, an impact on aquatic organisms at already very low concentrations is observed. Consequently, there is a need for sensitive analytical methods. Currently, no instrumental analysis methods exist that fulfil this requirement. To overcome these limitations, effect-based methods (EBMs) are one option for screening and quantification. Determination of agonistic effects caused by estrogens are the focus of current research activities due to environmental impacts and regulatory measures. However, antagonistic effects have to be considered to be able to correlate effects to single compounds, which is still necessary due to regulatory requirements. Since not much is known about relevant antagonists, this review highlights the perspectives and limitations of EBMs. The relevance of antagonistic acting compounds shows the potential of a systematic identification by effect-directed analysis. Collected data of already known antagonists are presented and discussed in a harmonized fashion as basis for future studies.

The analysis of microplastics is mainly performed using Fourier transformation infrared spectroscopy/microscopy (FTIR/ μFTIR). However, in contrast to most aspects of the analysis process, for example, sampling, sample preparation, and measurement, there is less known about data evaluation. This particularly critical step becomes more and more important if a large number of samples has to be handled. In this context, it is concerning that the commonly used library searching is not suitable to identify microplastics from real environmental samples automatically. Therefore, many spectra have to be rechecked by the operator manually, which is very time-consuming. In this study, a new fully automated robust microplastics identification method is presented that assigns over 98% of microplastics correctly. The main concept of this new method is to detect and numerically describe the individual vibrational bands within an FTIR absorbance spectrum by curve fitting, which leads to a very compact and highly characteristic peak list. This list allows very accurate and robust library searching. The developed approach is based on the already published microplastics identification algorithm (μIDENT) and extends and improves the field of application to μFTIR data with a special focus on relevant broad, overlapped, or complex vibrational bands.

One of the most critical steps in nontarget screening of organic micropollutants (OMP) in complex environmental samples is handling of massive data obtained from liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS). Multivariate chemometric methods have brought about great progress in processing big data obtained from high-dimensional chromatographic systems. This work aimed at a comprehensive evaluation of two LC-Q-Orbitrap mass spectrometry full-scan data sets for target and nontarget screening of OMPs in drinking and wastewater samples, respectively. For each data set, following segmentation in the chromatographic dimension, at first multivariate curve resolution alternating least-squares (MCR-ALS) was employed for simultaneous resolution of global matrices. The chromatographic peaks and the corresponding mass spectra of OMP were fully resolved in the presence of highly co-eluting irrelevant and interfering peaks. Then partial least-squares-discriminant analysis was conducted to investigate the behavior of MCR-ALS components in different water classes and selection of most relevant components. Further prioritization of features in wastewater before and after ozonation and their reduction to 24 micropollutants were then obtained by univariate statistics. In liquid chromatography coupled to isotope ratio mass spectrometry (LC-IRMS), analytes are separated on an LC system and consecutively oxidized to CO2, which is required for the determination of compound-specific carbon isotope ratios. Oxidation is performed in an online reactor by sulfate radicals. Reaction conditions in the interface depend on the flow conditions determined by the LC method and the flow rates and concentrations of oxidation agent and phosphoric acid added in the interface. To determine accurate isotope ratios, a quantitative conversion of the carbon contained in the analyte to the CO2 measurement gas is a prerequisite. Oxidation efficiencies are not commonly evaluated during method development, although certain analytes are known to be difficult to be oxidized by sulfate radicals. For the assessment of the oxidation efficiency of the LC-IRMS system, three different approaches were evaluated. (1) Residual organic carbon in the eluent stream of the interface was determined to calculate oxidation yields depending on the initial analyte concentration. (2) The IRMS response was calibrated to an inorganic carbon reference material to determine oxidation efficiencies with the help of the IRMS as a detector. (3) The oxidation temperature was deliberately reduced while monitoring the δ13C and signal intensity. The common assumption that a linear relation of IRMS signal to analyte concentration is an indicator for complete oxidation in LC-IRMS could be disproved. All three approaches can be applied for future method development in LC-IRMS, monitoring of existing flow injection applications, as well as for verification of complete oxidation in established LC-IRMS methods.

Taste and odor compounds are organic chemicals produced via biochemical processes, and their presence, even at low nanogram-per-liter concentrations, can make water useless for drinking purposes. In this work, a very sensitive and completely automated analytical procedure, based on solid-phase microextraction, has been developed and optimized for determination of seven taste and odor compounds in water media, well below their odor threshold. The selected analytes were isopropyl-3-methoxypyrazine, 2-isobutyl-3-methoxypyrazine, geosmin, 2-methylisoborneol, 2,4,6-trichloroanisole, 2,4,6-bromoanisole, and beta-ionone. Compared with a conventional approach, the recently introduced PAL SPME Arrow showed a significant enhancement in sensitivity and also outstanding robustness and stability. Three commercially available fiber coatings, as well as experimental parameters of the headspace extraction procedure, such as extraction temperature, time, and ionic strength of the aqueous sample, were investigated to optimize the method. The linearity of the response was assessed over a three-orders-of-magnitude range, with R2 values higher than 0.9914. The method was satisfactorily precise, with RSDs less than 11% at the second lowest calibration point (10–26 ng L−1). The calculated LODs (S/N = 3) were below odor thresholds of the target analytes and varied between 0.05 and 0.6 ng L−1, for just 10 mL of water sample. An original and spiked river water sample was also analyzed, and relative recoveries of 75–116% were achieved. Based on these analytical performance characteristics, and compared with other published methods, the present method can be considered as the most sensitive wholly automated approach for determination of taste and odor compounds in water.

Due to its ubiquitous presence in wastewaters, wastewater treatment plant effluents and even surface waters, the removal of the pharmaceutical ibuprofen from water is of special interest. Ozonation is widely applied for the treatment of micropollutants in wastewater treatment plants and is already known to also degrade ibuprofen. However, the formation of a wide range of transformation products during such oxidation steps might affect the aquatic environment. This study focuses on the acute ecotoxicological impact of the ibuprofen ozonation products on the two model organisms Daphnia magna and Desmodesmus subspicatus. For the identification of possibly ecotoxic products, a new workflow combining ecotoxicological testing, analytical methods and toxicity prediction was applied. Examination at different pH conditions with increasing ozone doses can point to respective products for further systematic examination. Seven ozonation products were confirmed in this study, two of them for the first time. Five previously postulated products were rejected. For pH 7 the inhibition of green algae growth was observed for mixtures oxidized with low ozone doses, while at pH 3 the mixtures with higher ozone doses caused toxic effects on the mobility of daphnids. Together with the analytical measurements in combination with ecotoxicity prediction, six products were identified which might have caused the toxic effect on green algae. However, no assignment to the observed toxic effects on daphnids was possible. The gained results indicate that mixture toxicity might play a role in oxidation processes and needs to be considered in ozonation studies concerning the ecotoxicological impact. Furthermore, the different observed toxicity for the two organisms underlines the importance of using multiple test systems for a comprehensive evaluation of the ecotoxicity during ozonation processes.
