Recent data suggest that only a small fraction of severe malaria heritability is explained by the totality of genetic markers discovered so far. The extensive genetic diversity within African populations means that significant associations are likely to be found in Africa. In their series of multi-site genome-wide association studies (GWAS) across sub-Saharan Africa, the Malaria Genomic Epidemiology Network (MalariaGEN) observed specific limitations and encouraged country-specific analyses. Here, we present findings of a GWAS of Cameroonian participants that contributed to MalariaGEN projects (n = 1103). Several sub-Saharan African countries rely on irrigation for food production. This study examined the impact of environmental modifications resulting from irrigation on the ecology of aquatic stages of malaria vectors in a semi-arid region of western Kenya. Mosquito larvae were collected from irrigated and non-irrigated ecosystems during seasonal cross-sectional and monthly longitudinal studies to assess habitat availability, stability, and productivity of anophelines in temporary, semipermanent, and permanent habitats during the dry and wet seasons. The duration of habitat stability was also compared between selected habitats. Emergence traps were used to determine the daily production of female adult mosquitoes from different habitat types. Malaria vectors were morphologically identified and sibling species subjected to molecular analysis. Data was statistically compared between the two ecosystems. After aggregating the data, the overall malaria vector productivity for habitats in the two ecosystems was estimated. Immatures of the malaria vector (Anopheles arabiensis) Patton (Diptera: Culicidae) comprised 98.3% of the Anopheles in both the irrigated and non-irrigated habitats. The irrigated ecosystem had the most habitats, higher larval densities, and produced 85.8% of emerged adult females. These results showed that irrigation provided conditions that increased habitat availability, stability, and diversity, consequently increasing the An. arabiensis production and potential risk of malaria transmission throughout the year. The irrigated ecosystems increased the number of habitats suitable for Anopheles breeding by about 3-fold compared to non-irrigated ecosystems. These results suggest that water management in the irrigation systems of western Kenya would serve as an effective method for malaria vector control.

Background Malaria control in Kenya is based on case management and vector control using long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS). However, the development of insecticide resistance compromises the effectiveness of insecticide-based vector control programs. The use of pesticides for agricultural purposes has been implicated as one of the sources driving the selection of resistance. The current study was undertaken to assess the status and mechanism of insecticide resistance in malaria vectors in irrigated and non-irrigated areas with varying agrochemical use in western Kenya. Methods The study was carried out in 2018–2019 in Homa Bay County, western Kenya. The bioassay was performed on adults reared from larvae collected from irrigated and non-irrigated fields in order to assess the susceptibility of malaria vectors to different classes of insecticides following the standard WHO guidelines. Characterization of knockdown resistance (kdr) and acetylcholinesterase-inhibiting enzyme/angiotensin-converting enzyme (Ace-1) mutations within Anopheles gambiae s.l. species was performed using the polymerase chain reaction (PCR) method. To determine the agricultural and public health insecticide usage pattern, a questionnaire was administered to farmers, households, and veterinary officers in the study area. Results Anopheles arabiensis was the predominant species in the irrigated (100%, n = 154) area and the dominant species in the non-irrigated areas (97.5%, n = 162), the rest being An. gambiae sensu stricto. In 2018, Anopheles arabiensis in the irrigated region were susceptible to all insecticides tested, while in the non-irrigated region reduced mortality was observed (84%) against deltamethrin. In 2019, phenotypic mortality was decreased (97.8–84% to 83.3–78.2%). In contrast, high mortality from malathion (100%), DDT (98.98%), and piperonyl butoxide (PBO)-deltamethrin (100%) was observed. Molecular analysis of the vectors from the irrigated and non-irrigated areas revealed low levels of leucine-serine/phenylalanine substitution at position 1014 (L1014S/L1014F), with mutation frequencies of 1–16%, and low-frequency mutation in the Ace-1R gene (0.7%). In addition to very high coverage of LLINs impregnated with pyrethroids and IRS with organophosphate insecticides, pyrethroids were the predominant chemical class of pesticides used for crop and animal protection. Conclusion Anopheles arabiensis from irrigated areas showed increased phenotypic resistance, and the intensive use of pesticides for crop protection in this region may have contributed to the selection of resistance genes observed. The susceptibility of these malaria vectors to organophosphates and PBO synergists in pyrethroids offers a promising future for IRS and insecticide-treated net-based vector control interventions. These findings emphasize the need for integrated vector control strategies, with particular attention to agricultural practices to mitigate mosquito resistance to insecticides. Graphic abstract

Inferences from genetic association studies rely largely on the definition and description of the underlying populations that highlight their genetic similarities and differences. The clustering of human populations into subgroups (population structure) can significantly confound disease associations. This study investigated the fine-scale genetic structure within Cameroon that may underlie disparities observed with Cameroonian ethnicities in malaria genome-wide association studies in sub-Saharan Africa. Genotype data of 1073 individuals from three regions and three ethnic groups in Cameroon were analyzed using measures of genetic proximity to ascertain fine-scale genetic structure. Model-based clustering revealed distinct ancestral proportions among the Bantu, Semi-Bantu and Foulbe ethnic groups, while haplotype-based coancestry estimation revealed possible longstanding and ongoing sympatric differentiation among individuals of the Foulbe ethnic group, and their Bantu and Semi-Bantu counterparts. A genome scan found strong selection signatures in the HLA gene region, confirming longstanding knowledge of natural selection on this genomic region in African populations following immense disease pressure. Signatures of selection were also observed in the HBB gene cluster, a genomic region known to be under strong balancing selection in sub-Saharan Africa due to its co-evolution with malaria. This study further supports the role of evolution in shaping genomes of Cameroonian populations and reveals fine-scale hierarchical structure among and within Cameroonian ethnicities that may impact genetic association studies in the country.

Glossina morsitans morsitans are vectors of Animal African Trypanosomiasis (nagana) and Human African Trypanosomiasis (sleeping sickness). Glossina uses odorant receptors (Ors) expressed within the olfactory receptor neurons to identify chemical cues within the environment, locate hosts and find larviposition sites and mating partners. Most Ors have been identified in insects’ olfactory tissues but little is known about the potential functions of Ors in non-olfactory organs. In the present study, we determine the expression profile of 46 Ors in male and female G. m. morsitans brain, gut and reproductive tissues using quantitative polymerase chain reaction. Our results indicate that 38 Or genes were detected in the male brain and gut, 37 in reproductive tissues while the number identified in the female included 44 in brain, 22 in gut and 40 in reproductive tissue. The Or genes were abundantly distributed in both male and female brain, gut and reproductive tissues at different fold expression levels. Quantitative real time PCR revealed that 4 Or genes (GmmOr19, GmmOr28, GmmOr30, GmmOr31) were significantly highly expressed in the male G. m. morsitans brain while the gut had 6 Ors (GmmOr7, GmmOr12, GmmOr32, GmmOr34, GmmOr42, GmmOr44) and the testis and accessory glands had 3 Or genes (GmmOr32, GmmOr44, GmmOr46) significantly highly expressed. In the female G. m. morsitans, the Ors that were significantly expressed were 2 in the brain (GmmOr25, GmmOr27), 3 both in the gut (GmmOr17, GmmOr33, GmmOr35) and reproductive tissues (GmmOr9, GmmOr22, GmmOr27). This results provides insight on the expression profile of Or genes in non-olfactory organs (brain, gut and reproductive tissues) in male and female G. m. morsitans and open avenues to investigate the possible role of the Or genes that were significantly expressed, which could provide targets that could be explored to control Glossina and the devastating trypanosomiasis disease.

Background: The sudden onset of P. falciparum resistance to antimalarial drugs requires careful surveillance of African parasite population. Genomics tools are implemented to detect evolutionary changes that could impact on malaria control and elimination strategies. Here, we evaluated the genome-wide pattern of selection and sequence variation of P. falciparum populations from Côte d’Ivoire. Methods: The study was carried out in three localities of Abidjan from 2013 to 2014. We collected 70 blood samples following a written consent from patients above two years of age. After extracting P. falciparum DNA from isolates, we performed Whole Genome Sequencing and used population genomics approaches to investigate genetic diversity, complexity of infection and identify loci under positive directional selection. Results: We observed an excess of rare variants in the population showing a clear mutation process in the isolates. Moderate Fst estimates (0.3) was detected for surfin genes expressed at the surface of infected erythrocytes and released merozoites. Seven iHS regions that had at least two SNPs with a score > 3.2 were identified. These regions code for genes that have been under strong directional selection. Two of these genes were the chloroquine resistance transporter ( crt) on chromosome 7 and the dihydropteroate reductase (dhps) on chromosome 8 despite their official proscription for malaria treatment since 2007. There was also a recent selective sweep in the erythrocyte membrane protein ( Pfemp1) . Conclusion: Population genomics revealed selective drug pressure and balancing selection on protective immunity target genes, demonstrating his effectiveness to follow up adaptive evolution of parasite populations and adopt appropriate strategies to control malaria in Côte d’Ivoire. Background Histidine-rich protein 2 (HRP2)-based malaria rapid diagnostic tests (RDTs) are effective and widely used for the detection of wild-type Plasmodium falciparum infections. Although recent studies have reported false negative HRP2 RDT results due to pfhrp2 and pfhrp3 gene deletions in different countries, there is a paucity of data on the deletions of these genes in Tanzania. Methods A community-based cross-sectional survey was conducted between July and November 2017 in four regions: Geita, Kigoma, Mtwara and Ruvuma. All participants had microscopy and RDT performed in the field and provided a blood sample for laboratory multiplex antigen detection (for Plasmodium lactate dehydrogenase, aldolase, and P. falciparum HRP2). Samples showing RDT false negativity or aberrant relationship of HRP2 to pan-Plasmodium antigens were genotyped to detect the presence/absence of pfhrp2/3 genes. Results Of all samples screened by the multiplex antigen assay (n = 7543), 2417 (32.0%) were positive for any Plasmodium antigens while 5126 (68.0%) were negative for all antigens. The vast majority of the antigen positive samples contained HRP2 (2411, 99.8%), but 6 (0.2%) had only pLDH and/or aldolase without HRP2. Overall, 13 samples had an atypical relationship between a pan-Plasmodium antigen and HRP2, but were positive by PCR. An additional 16 samples with negative HRP2 RDT results but P. falciparum positive by microscopy were also chosen for pfhrp2/3 genotyping. The summation of false negative RDT results and laboratory antigen results provided 35 total samples with confirmed P. falciparum DNA for pfhrp2/3 genotyping. Of the 35 samples, 4 (11.4%) failed to consistently amplify positive control genes; pfmsp1 and pfmsp2 and were excluded from the analysis. The pfhrp2 and pfhrp3 genes were successfully amplified in the remaining 31 (88.6%) samples, confirming an absence of deletions in these genes. Conclusions This study provides evidence that P. falciparum parasites in the study area have no deletions of both pfhrp2 and pfhrp3 genes. Although single gene deletions could have been missed by the multiplex antigen assay, the findings support the continued use of HRP2-based RDTs in Tanzania for routine malaria diagnosis. There is a need for the surveillance to monitor the status of pfhrp2 and/or pfhrp3 deletions in the future.

The aims of this study were to follow up on the establishment of Cotesia vestalis in the coastal regions of Kenya, assessing its contribution in the management of the diamondback moth (Plutella xylostella), parasitism rates, and cultural practices affecting its establishment. Post-release surveys were carried out in five counties in Kenya, Kitui, Kajiado, Kwale, Machakos and Makueni, between 2015 and 2016. The results showed that the overall parasitism rate of C. vestalis in 2015 ranged between 0% and 37.86% while that in 2016 ranged from 0% to 32.19% in the different counties. Farmers carry out routine sprays, either weekly or fortnightly, with different synthetic insecticides. Pyrethroids (60.56%) constituted most of the insecticides used, while only 3.18% of the products used were plant or microorganism- based. Farmers did most of their cabbage production during the rainy season, with production being greatly reduced during the dry seasons. The sampled diamondback moths from Kajiado, Kitui, Makueni, Kwale and Malawi had ≥98% similarity to Cotesia vestalis, as shown by bioinformatics analyses using PCR amplified products of 700 bp, obtained for the mitochondrial COI gene. Alignment showed highly conserved regions, and the phylogenetic analysis revealed two close lineages corresponding to Cotesia vestalis (Genbank accession: FJ154897) and Cotesia spp. (acc. HM430398). Data provide a clear indication that the parasitoid became established in its release sites in Kenya, although parasitism rates are still low probably due to heavy pesticide use and climatic factors.

Glossina spp. (Diptera: Glossinidae) vector trypanosomes, the causal pathogens of trypanosomiasis in humans and animals. Glossina spp. chemosensory mechanism is relevant to understanding the behavioral responses of these insects to environmental cues. Odorant binding proteins (OBPs) are involved in either olfactory or nonolfactory cue sensation depending on their spatial and temporal expression. In most insects, the OBPs are ubiquitously expressed, although the levels are higher in the antennae. This study was aimed at determining the differential gene expression patterns of OBP genes in the developmental stages (larval, pupal, teneral) and differentially starved adults of Glossina brevipalpis (Newstead) collected from Shimba Hills National Park, Kenya. Tsetse flies (Glossina) are vectors of African trypanosomiasis. Olfaction plays a critical role in Glossina behavior, including larviposition, feeding, and reproduction. Odorant receptors (ORs) are important in insect chemoreception as they bind volatile odorants and transport them to olfactory receptor neurons to elicit behavioral response. To better understand Glossina chemoreception, we used quantitative polymerase chain reaction to examine the expression levels of ORs in female and male Glossina morsitans morsitans Wiedemann, 1850 (Diptera: Glossinidae) antennae and legs. Results showed that G. m. morsitans ORs code for a transmembrane domain and are involved in odorant binding. The ORs had homologs in Drosophila, mosquitoes, other Glossina species, and the reduced number of tsetse ORs could be linked to its restricted blood-feeding diet. The OR genes were more highly expressed in antennae than the legs with GmmOR33 and GmmOR45 transcript levels being high in the female and male antennae, respectively, whereas GmmOR26 and GmmOR34 levels were high in female and male G. m. morsitans legs, respectively. These findings identified sex- and tissue-specific G. m. morsitans ORs. The expression levels of OR genes in female and male G. m. morsitans could be conserved in function with the antenna being the main olfactory organ. Thus, this study provides a blueprint to explore the functional roles of tsetse ORs with the potential to identify molecular targets that can be used to control the vector based on disruption of its chemosensory system.

The mosquito species Culex pipiens is a potential vector of several pathogens infecting humans and occurs in two distinct bioforms, pipiens and molestus. Traditional morphological identification fails to separate the bioforms of Cx. pipiens despite their behavioural differences since they are morphologically indistinguishable. However, molecular methods can identify the two bioforms. The bioform molestus thrives in urban environments and bite all kinds of vertebrates, whereas bioform pipiens is more rural and mainly feed on birds.Mosquito samples submitted in a citizen science project from people experiencing mosquito problems in South Sweden were analyzed to determine the geographical distribution of the molestus bioform of Cx. pipiens. Mosquito specimens were identified to species by DNA barcoding of the cytochrome C oxidase subunit I (COI) gene and the bioforms were determined with the CQ11 microsatellite marker. To establish other differences between the bioforms, part of the CPIJ001674 gene was sequenced. Culex pipiens f molestus, was present both within and outside of urban areas in several sites in southern Sweden. In one site, hybrids between the two bioforms were found. Culex pipiens f molestus has previously been found in urban areas in Sweden, but the detection of the bioform in several rural areas was surprising, indicating that it may be more widely spread than previously thought.

Bacterial wilt caused by Ralstonia solanacearum is considered among the most damaging diseases of potato in Sub-Saharan Africa and the most significant biotic constraint of potato production alongside late blight. Unlike late blight,which can be managed by chemical means, R. solanacearum can only be managed through cultural methods and clean seed. Laboratory testing to certify seed before planting is required to confirm the absence of the pathogen in Kenya. A loop-mediated isothermal amplification (LAMP) assaywas developed using the UDP-(3-O-acyl)-N-acetylglucosamine deacetylase gene (IpxC) to screen seed potato for R. solanacearum strains. The assay was assessed using DNA extracted from R. solanacearum and other soil and potato pathogens to demonstrate specificity and sensitivity. The LAMP assay was validated using field samples from different potato growing regions of Kenya collected over two growing seasons and compared with established nucleic acid and protein-based assays. The IpxC LAMP assay was found to be specific and sensitive to R. solanacearum, detecting as low as 2.5 pg/ml of R. solanacearum DNA. Of the 47 potentially infected field samples collected, both IpxC LAMP and quantitative polymerase chain reaction (PCR) detected R. solanacearum DNA in 90% of the samples, followed by conventional PCR (86%) and ELISA (75%). Using real-time RT-PCR, we screened stool samples from children aged &lt, 5 years presenting with diarrhea and admitted to Kilifi County Hospital, coastal Kenya, pre- (2003 and 2013) and post-rotavirus vaccine introduction (2016 and 2019) for five viruses, namely rotavirus group A (RVA), norovirus GII, adenovirus, astrovirus and sapovirus. Of the 984 samples analyzed, at least one virus was detected in 401 (40.8%) patients. Post rotavirus vaccine introduction, the prevalence of RVA decreased (23.3% vs. 13.8%, p &lt, 0.001) while that of norovirus GII increased (6.6% vs. 10.9%, p = 0.023). The prevalence of adenovirus, astrovirus and sapovirus remained statistically unchanged between the two periods: 9.9% vs. 14.2%, 2.4% vs. 3.2 %, 4.6% vs. 2.6%, (p = 0.053, 0.585 and 0.133), respectively. The median age of diarrhea cases was higher post vaccine introduction (12.5 months, interquartile range (IQR): 7.9&ndash, 21 vs. 11.2 months pre-introduction, IQR: 6.8&ndash, 16.5, p &lt, 0.001). In this setting, RVA and adenovirus cases peaked in the dry months while norovirus GII and sapovirus peaked in the rainy season. Astrovirus did not display clear seasonality. In conclusion, following rotavirus vaccine introduction, we found a significant reduction in the prevalence of RVA in coastal Kenya but an increase in norovirus GII prevalence in hospitalized children.

Introduction Plasmodium falciparum relies on gametocytogenesis to transmit from humans to mosquitoes. Gametocyte development 1 (Pfgdv1) is an upstream activator and epigenetic controller of gametocytogenesis. The emergence of drug resistance is a major public health concern and this requires the development of new strategies that target the transmission of malaria. As a putative drug target, Pfgdv1 has not been characterized to identify its polymorphisms and alleles under selection and how such polymorphisms influence protein structure. Methods This study characterized single-nucleotide polymorphisms (SNPs) in primary sequences (n = 30) of Pfgdv1 gene generated from thirty blood samples collected from patients infected with P. falciparum and secondary sequences (n = 216) retrieved from PlasmoDB. ChromasPro, MUSCLE, Tajima's D statistic, SLAC, and STRUM were used in editing raw sequences, performing multiple sequence alignment (MSA), identifying signatures of selection, detecting codon sites under selection pressure, and determining the effect of SNPs, respectively. Results MSA of primary and secondary sequences established the existence of five SNPs, consisting of four non-synonymous substitutions (nsSNPs) (p.P217H, p.R398Q, p.H417N, and p.D497E), and a synonymous substitution (p.S514S). The analysis of amino acid changes reveals that p.P217H, p.R398Q, and p.H417N comprise non-conservative changes. Tajima's D statistic showed that these SNPs were under balancing selection, while SLAC analysis identified p.P217H to be under the strongest positive selection. . Further analysis based on thermodynamics indicated that p.P217H has a destabilizing effect, while p.R398Q and p.D497E have stabilizing effects on the protein structure. Conclusions The existence of four nsSNPs implies that Pfgdv1 has a minimal diversity in the encoded protein. Selection analysis demonstrates that these nsSNPs are under balancing selection in both local and global populations. However, p.P217H exhibits positive directional selection consistent with previous reports where it showed differentiatial selection of P. falciparum in low and high transmission regions. Malaria causes about half a million deaths annually, with Plasmodium falciparum being responsible for 90% of all the cases. Recent reports on artemisinin resistance in Southeast Asia warrant urgent discovery of novel drugs for the treatment of malaria. However, most bioactive compounds fail to progress to treatments due to safety concerns. Drug repositioning offers an alternative strategy where drugs that have already been approved as safe for other diseases could be used to treat malaria. This study screened approved drugs for antimalarial activity using an in silico chemogenomics approach prior to in vitro verification. All the P. falciparum proteins sequences available in NCBI RefSeq were mined and used to perform a similarity search against DrugBank, TTD and STITCH databases to identify similar putative drug targets. Druggability indices of the potential P. falciparum drug targets were obtained from TDR targets database. Functional amino acid residues of the drug targets were determined using ConSurf server which was used to fine tune the similarity search. This study predicted 133 approved drugs that could target 34 P. falciparum proteins. A literature search done at PubMed and Google Scholar showed 105 out of the 133 drugs to have been previously tested against malaria, with most showing activity. For further validation, drug susceptibility assays using SYBR Green I method were done on a representative group of 10 predicted drugs, eight of which did show activity against P. falciparum 3D7 clone. Seven had IC50 values ranging from 1 μM to 50 μM. This study also suggests drug-target association and hence possible mechanisms of action of drugs that did show antiplasmodial activity. The study results validate the use of proteome-wide target similarity approach in identifying approved drugs with activity against P. falciparum and could be adapted for other pathogens.

Successful optimization of plant-derived compounds into control of nuisance insects would benefit from scientifically validated targets. However, the close association between the genotypic responses and physiological toxicity effects mediated by these compounds remains underexplored. In this study, we evaluated the sublethal dose effects of proanthocyanidins (PAs) sourced from green tea (Camellia sinensis) on life history traits of Anopheles gambiae (sensu stricto) mosquitoes with an aim to unravel the probable molecular targets. Based on the induced phenotypic effects, genes selected for study targeted juvenile hormone (JH) biosynthesis, signal transduction, oxidative stress response and xenobiotic detoxification in addition to vitellogenesis in females. Our findings suggest that chronic exposure of larval stages (L3/L4) to sublethal dose of 5 ppm dramatically extended larval developmental period for up to 12 days, slowed down pupation rates, induced abnormal larval-pupal intermediates and caused 100% inhibition of adult emergence. Further, females exhibited significant interference of fecundity and egg hatchability relative to controls (p < 0.001). Using reverse transcription quantitative polymerase chain reaction (RT-qPCR), our findings show that PA-treated larvae exhibited significant repression of AgamJHAMT (p < 0.001), AgamILP1 (p < 0.001) and AgamCYP6M2 (p < 0.001) with up-regulation of Hsp70 (p < 0.001). Females exposed as larvae demonstrated down-regulation of AgamVg (p = 0.03), AgamILP1 (p = 0.009), AgamCYP6M2 (p = 0.05) and AgamJHAMT (p = 0.02). Our findings support that C. sinensis proanthocyanidins affect important vectorial capacity components such as mosquito survival rates and reproductive fitness thus could be potentially used for controlling populations of malaria vectors.

Tsetse flies (Glossina) depend on their olfactory system to identify host, larviposition sites and mates. Previous studies have characterized olfactory organs and evaluated the odorant receptor (OR) expression to host-derived chemicals. However, no studies thus far have investigated the odorant co-receptor (Orco) associated with these olfaction processes in tissues and developmental stages in tsetse flies. In this study, quantitative polymerase chain reaction was done with Glossina morsitans morsitans, Glossina pallidipies and Glossina fuscipes fuscipes tissues (antennae and legs) and developmental stage (larvae and pupae) to quantify Orco expression levels and G. m. morsitans OR genes in G. m. morsitans larvae and pupae. Our findings indicate that expression of Orco were elevated in G. f. fuscipes male antennae and legs relative to G. f. fuscipes female antennae and relative to male antennae in G. m. morsitans and G. pallidipies. However, in female G. m. morsitans antennae, expression of Orco was higher than in female G. pallidipies and G. f. fuscipes. Orco levels were also significantly higher in pupal stages of G. f. fuscipes and G. pallidipies relative to the three larval stages and G. m. morsitans pupae. Two G. m. morsitans OR genes (GmmOR20 and GmmOR28) were highly expressed in G. m. morsitans larvae and pupae respectively. This study demonstrate that the Orco gene was significantly expressed in adult male and female tissues as well as in the developmental stages of Glossina species. The G. m. morsitans OR genes reported in larvae and pupae could mean co-expression of Orco and ORs in developmental stages. These findings pinpointed the olfactory differences that exists amongst G. m. morsitans, G. pallidipies and G. f. fuscipes, and provide valuable information to the probable role of olfactory genes in Glossina contributing further to the understanding of olfactory processes and the evolution of olfactory genes in tsetse flies. Background Anopheles arabiensis and A. gambiae (sensu stricto) are the most prolific Afrotropical malaria vectors. Population control efforts of these two vectors have been hampered by extremely diverse larval breeding sites and widespread resistance to currently available insecticides. Control of mosquito larval stages using bioactive compounds of plant origin has the potential to suppress vector populations leading to concomitant reduction in disease transmission rates. In this study, we evaluated the efficacy of Camellia sinensis crude leaf extract and its fraction against the larvae of A. arabiensis and A. gambiae (s.s.). Methods Late third/early fourth instar larvae (L3/L4) of A. arabiensis and A. gambiae (s.s.) were exposed to increasing doses of C. sinensis leaf extract and its active fraction for 72 h, with mortality rates recorded every 24 h in both control and test groups. Ultra performance liquid chromatography electron spray ionization quadruple time of flight coupled with mass spectrometry (UPLC/ESI-Qtof/MS) was used to determine the main active constituents in the fraction. Results The major bioactive chemical constituents in the C. sinensis leaf extract were identified to be proanthocyanidins. Glossina pallidipes salivary gland hypertrophy virus (GpSGHV; family Hytrosaviridae) is a dsDNA virus exclusively pathogenic to tsetse flies (Diptera; Glossinidae). The 190 kb GpSGHV genome contains 160 open reading frames and encodes more than 60 confirmed proteins. The asymptomatic GpSGHV infection in flies can convert to symptomatic infection that is characterized by overt salivary gland hypertrophy (SGH). Flies with SGH show reduced general fitness and reproductive dysfunction. Although the occurrence of SGH is an exception rather than the rule, G. pallidipes is thought to be the most susceptible to expression of overt SGH symptoms compared to other Glossina species that are largely asymptomatic. Although Glossina salivary glands (SGs) play an essential role in GpSGHV transmission, the functions of the salivary components during the virus infection are poorly understood. In this study, we used mass spectrometry to study SG proteomes of G. pallidipes and G. m. morsitans, two Glossina model species that exhibit differential GpSGHV pathologies (high and low incidence of SGH, respectively). A total of 540 host proteins were identified, of which 23 and 9 proteins were significantly up- and down-regulated, respectively, in G. pallidipes compared to G. m. morsitans. Whereas 58 GpSGHV proteins were detected in G. pallidipes F1 progenies, only 5 viral proteins were detected in G. m. morsitans. Unlike in G. pallidipes, qPCR assay did not show any significant increase in virus titers in G. m. morsitans F1 progenies, confirming that G. m. morsitans is less susceptible to GpSGHV infection and replication compared to G. pallidipes. Based on our results, we speculate that in the case of G. pallidipes, GpSGHV employs a repertoire of host intracellular signaling pathways for successful infection. In the case of G. m. morsitans, antiviral responses appeared to be dominant. These results are useful for designing additional tools to investigate the Glossina-GpSGHV interactions.

This study examined the population genetic structure of the major malaria vector, Anopheles arabiensis mosquitoes, in Ethiopia and Eritrea. Ethiopia and Eritrea have great geographical diversity, with high mountains, rugged plateaus, deep gorges, and rolling plains. The plateau is bisected diagonally by the Great Rift Valley into the Northwestern Highlands and the Southeastern Highlands. Five A. arabiensis populations from the Northwestern Highlands region and two populations from high-altitude sites in the Great Rift Valley were genotyped using six microsatellite markers to estimate the genetic diversity and population genetic structure of A. arabiensis. We found that A. arabiensis populations from the Northwestern Highlands and the Great Rift Valley region showed a similar level of genetic diversity. The genetic differentiation (FST) of the five mosquito populations within the Northwestern Highlands region was 0.038 (P , .001), while the two populations within the Great Rift Valley showed little genetic differentiation (FST ¼ 0.007, P , .01). The degree of genetic differentiation between the Northwestern Highlands region and the Great Rift Valley region was small but statistically significant (FST ¼ 0.017, P , .001). The population genetic structure of A. arabiensis in the study area did not follow the isolation-by-distance model (r 2 ¼ 0.014, P . .05). The low FST estimates for A. arabiensis populations in Ethiopia and Eritrea are consistent with the general population genetic structure of this species in East Africa based on other molecular markers.

Tricorn protease is an archaeal protease acting downstream of the proteasome and together with its interacting aminopeptidases, degrades oligopeptides to free amino acids thus playing an important role in protein turnover. This study reports a wide distribution of tricorn protease and its homologs in archaea and bacteria. The homologs were identified through a combination of PSI-BLAST, orthology clustering and domain predictions. Functionally important sites were identified through multiple sequence alignment conducted by MAFFT v. 7. The aligned sequences were used to predict the phylogenetic relationship of tricorn protease and its homologs using MEGA v. 7. The functional associations of tricorn protease were predicted through STRING network v.10.0. This study identified several tricorn protease homologs in archaea and in all the bacterial phyla complete with β-propeller, PDZ and catalytic domains. However, in eukaryotes, tricorn protease-like homologs seemed limited to viridiplantae, stramenopile and in a basal metazoa and were classified as non-peptidase homologs with unknown functions. Conserved domain architecture retrieval revealed detectable homology of tricorn protease C-terminal half with the carboxyl-terminal proteases with similar PDZ domains. Therefore, this study predicts functional conservation of tricorn core catalytic domain in prokaryotes and given its role in cellular functions, targeting this protein or its functional homologs in prokaryotic pathogens could lead to development of alternative therapeutic agents.

Africa's Bane Tsetse are blood-feeding, fast-flying flies that transmit a range of Trypanosoma spp. protozoan pathogens, which cause sleeping sickness in humans and their nagana in their livestock. The International Glossina Genome Initiative (p. 380 ) sequenced the genome of Glossina morsitans and identified the genes for many attributes of the tsetse's remarkable biology, including viviparity and the expression of analogs of mammalian milk proteins. Tsetse are host to several specific symbionts that appear to synthesize essential nutrients for the fly and also to hitherto undiscovered parasitoid-derived viruses. Deeper exploration of this genome will reveal what makes these fly species so host- and trypanosome specific.

Tsetse flies use olfactory and gustatory responses, through odorant and gustatory receptors (ORs and GRs), to interact with their environment. Glossina morsitans morsitans genome ORs and GRs were annotated using homologs of these genes in Drosophila melanogaster and an ab initio approach based on OR and GR specific motifs in G. m. morsitans gene models coupled to gene ontology (GO). Phylogenetic relationships among the ORs or GRs and the homologs were determined using Maximum Likelihood estimates. Relative expression levels among the G. m. morsitans ORs or GRs were established using RNA-seq data derived from adult female fly. Overall, 46 and 14 putative G. m. morsitans ORs and GRs respectively were recovered. These were reduced by 12 and 59 ORs and GRs respectively compared to D. melanogaster. Six of the ORs were homologous to a single D. melanogaster OR (DmOr67d) associated with mating deterrence in females. Sweet taste GRs, present in all the other Diptera, were not recovered in G. m. morsitans. The GRs associated with detection of CO2 were conserved in G. m. morsitans relative to D. melanogaster. RNA-sequence data analysis revealed expression of GmmOR15 locus represented over 90% of expression profiles for the ORs. The G. m. morsitans ORs or GRs were phylogenetically closer to those in D. melanogaster than to other insects assessed. We found the chemoreceptor repertoire in G. m. morsitans smaller than other Diptera, and we postulate that this may be related to the restricted diet of blood-meal for both sexes of tsetse flies. However, the clade of some specific receptors has been expanded, indicative of their potential importance in chemoreception in the tsetse., Author Summary Tsetse flies navigate their environments using chemosensory receptors, which permit them to locate hosts, mating partners, and resting and larviposition sites. The genome of G. m. morsitans was interrogated for coding genes of odorant receptors (ORs) and gustatory receptors (GRs) that express in antennae and maxillary palp, and detect the volatile and soluble chemical signals. The signals are then transmitted to the central nervous system and translated to phenotypes. Majority of these genes in G. m. morsitans were spread across different scaffolds, but a few were found to occur in clusters, which suggested possible co-regulation of their expression. The number of ORs and GRs were much reduced in the G. m. morsitans genome, including the apparent loss of receptors for sugar when compared to selected Diptera. There was also an apparent numerical expansion of some receptors, presumably to maximize on their restricted blood-meal diet. The annotation of the chemoreceptor package of G. m. morsitans provides a resource for investigating key activities of tsetse flies that could be exploited for their control.

Background Rickettsia africae, the etiological agent of African tick bite fever, is widely distributed in sub-Saharan Africa. Contrary to reports of its homogeneity, a localized study in Asembo, Kenya recently reported high genetic diversity. The present study aims to elucidate the extent of this heterogeneity by examining archived Rickettsia africae DNA samples collected from different eco-regions of Kenya. Methods To evaluate their phylogenetic relationships, archived genomic DNA obtained from 57 ticks a priori identified to contain R. africae by comparison to ompA, ompB and gltA genes was used to amplify five rickettsial genes i.e. gltA, ompA, ompB, 17kDa and sca4. The resulting amplicons were sequenced. Translated amino acid alignments were used to guide the nucleotide alignments. Single gene and concatenated alignments were used to infer phylogenetic relationships. Results Out of the 57 DNA samples, three were determined to be R. aeschlimanii and not R. africae. One sample turned out to be a novel rickettsiae and an interim name of “Candidatus Rickettsia moyalensis” is proposed. The bonafide R. africae formed two distinct clades. Clade I contained 9% of the samples and branched with the validated R. africae str ESF-5, while clade II (two samples) formed a distinct sub-lineage. Conclusions This data supports the use of multiple genes for phylogenetic inferences. It is determined that, despite its recent emergence, the R. africae lineage is diverse. This data also provides evidence of a novel Rickettsia species, Candidatus Rickettsia moyalensis., Author Summary Rickettsia africae is a bacterium mainly vectored by Amblyomma and Rhipicephalus species of ticks. It is the etiological agent of African tick bite fever (ATBF), a spotted fever rickettsiosis that presents as an acute febrile illness characterized by petecheal skin hemorrhages, from which the name is derived. This bacterium is probably the most important in sub-Saharan Africa, including Kenya, in terms of incidence and prevalence. This notwithstanding, the disease is poorly understood and is often mistreated as malaria, and therefore qualifies as a highly neglected disease. This study examined the genetic relationships of R. africae collected from diverse eco-regions of Kenya. We present data that indicate high genetic diversity in Kenya’s R. africae and corroborate a recent study that reported similar genetic diversity in R. africae samples collected from a localized area in western Kenya. Importantly, we describe a divergent lineage and propose the name Candidatus Rickettsia moyalensis.
