Los Macocos, Banda de Teatro, una de las agrupaciones más sólidas del teatro nacional, se creó en los años 80, por iniciativa de tres estudiantes de la Escuela Nacional de Arte Dramático: Daniel Casablanca, Joaquín Romero y Martín Salazar. Años más tarde, en 1998, se incorporan al grupo Marcelo Xicarts y Gabriel Wolf. La Banda formó parte del pujante movimiento de renovación teatral surgido desde la recuperación de la democracia, que se potenció y se nutrió de esa transformación fundamental en la vida social y política del país. Desde sus orígenes, los integrantes de La Banda de Teatro generaron sus propios espectáculos a partir de la creación colectiva como metodología de trabajo, tanto en lo que respecta al plano dramatúrgico como al escénico.

El presente artículo propone una aproximación a la escritura dramática producida por mujeres en Argentina entre la década del sesenta y del noventa con la finalidad de indagar cómo se configura una mirada crítica desde el género, un discurso femenino autónomo. En vistas a ello, nos referimos a algunas dramaturgas paradigmáticas - Griselda Gambaro, Aída Bortnik, Susana Torres Molina, Cristina Escofet y Diana Raznovich- que, desde diversas poéticas, intereses estéticos e ideológicos, contribuyeron a la representación y la autorrepresentación de la mujer como un sujeto plural, atravesado y habitado por una complejidad de voces y de inscripciones personales, sociales, genéricas, culturales. Para la descripción y el análisis de las piezas dramáticas más representativas en este sentido partimos de una perspectiva historicista que tiene en cuenta tanto el contexto histórico social como la dinámica propia del teatro argentino. En las décadas del sesenta y del setenta se verifica una notable limitación de la presencia de la mujer en el campo teatral nacional, no solo en lo que atañe a dramaturgas y directoras, sino también dentro de la gestión cultural, la recepción crítica y los jurados teatrales. Sin duda el convulsionado contexto sociopolítico obstaculizó el surgimiento y el desarrollo de un teatro propiamente femenino hasta el advenimiento de la democracia, momento en que comienzan a tratarse los hechos sociales teniendo en cuenta las diferencias genéricas que los atraviesan. Por último, observamos que, abordando directamente cuestiones de género o bien movilizadas por las urgencias de una sociedad en crisis, con la intención de esbozar posibles respuestas o simplemente de plantear interrogantes, las autoras a las que aludimos buscan alejarse de los estereotipos y los modelos arquetípicos, interpelar las narrativas, quebrar los mandatos de silencio impuestos por las dictaduras o por la sociedad patriarcal.

The Mangalarga Marchador (MM) horse breed has expressive importance in the Brazilian economy. Thus, the aim of this study was to investigate diversity in the MM breed. A database with a total of 3,193 genotyped horses was used (MM, n = 2,829; Andalusian – AND, n = 67; Pure Blood Lusitano – LUS, n = 43; English Thoroughbred – THO, n = 54; Arabian – ARA, n = 99; Campolina – CAM, n = 61; and Mangalarga – MAN, n = 40) for 13 microsatellite. Diversity parameters were estimates, such as mean number of alleles (Nma) and the number of rare alleles (AR), expected heterozygosity (He), F statistics, genetic distances, Hardy-Weinberg equilibrium test (HWE), population structure, and others. The Nma was 10.85, the AR was prevalent in the MM, and the He was 0.7402. In MM, the values of Fis (−0.0195), Fit (0.0566), Fst (0.0748), and deviations of HWE were observed. The genetic distances of the ARA and THO breeds with the other breeds were greater than the distances between the Brazilian breeds and between these and the breeds in the Iberian Peninsula. The population structure indicated that MM was substructured, yet there were some more genetically defined breeding farms. The genetic diversity is satisfactory for MM conservation, but the population is substructure, and parameters indicate moderate gene flow and the existence, though few, of crosses with other horse breeds. Immediate implementation of a genetic breeding program is required, especially seeking to conserve the structure of the MM breed as a well-defined genetic entity.

Evidence of cerebellar abiotrophy (CA) was found in a six-month-old Arabian filly with signs of incoordination, head tremor, wobbling, loss of balance and falling over, consistent with a cerebellar lesion. Normal hematology profile blood test and cerebrospinal fluid analysis excluded infectious encephalitis, and serological testing for Sarcocystis neurona was negative. The filly was euthanized. Postmortem X-ray radiography of the cervical cephalic region identified not abnormalities, discounting spinal trauma. The histopathological analysis of serial transverse cerebellar sections by electron microscopy revealed morphological characteristics of apoptotic cells with pyknotic nuclei and degenerate mitochondria, cytoplasmic condensation and areas with absence of Purkinje cells, matching with CA histopathological characteristics. The indirect DNA test for CA was positive in the filly, and DNA test confirmed the CA carrier state in the parents and the recessive inheritance of the disease. El objetivo del trabajo de Tesis Doctoral consistió en estudiar el polimorfismo y el poligenismo de los genes de clase II DRB del Complejo Principal de Histocompatibilidad en Equinos (ELA). La variabilidad genética de las regiones promotoras y de la región de reconocimiento del antígeno (exón 2) se analizó mediante los métodos de PCR-RFLP y PCR-SSCP. Se detectaron tres nuevos alelos del exón 2 definidos por PCR-RFLP, los que se confirmaron por clonado y secuenciación de los productos de amplificación. Además, el número de variantes detectadas en cada animal permitió inferir la presencia de al menos tres copias de genes DRB en los haplotipos equinos analizados. Estos datos se confirmaron a través de análisis filogenético y de segregación. El clonado y secuenciación de la región reguladora (URR) de los genes DRB permitió caracterizar la organización del promotor proximal. Los resultados obtenidos mostraron la presencia en dirección 5´ - 3´ de las cajas conservadas W, X, Y, CCAAT y TATA. Esta estructura es semejante a la reportada en los genes ortólogos de otras especies de mamíferos y evidenciaría que la región analizada corresponde a un gen DRB funcional. Por otra parte, el análisis del polimorfismo del promotor permitió identificar cinco alelos definidos por SSCP. El conjunto de resultados obtenidos mostró que los genes ELA-DRB presentan las principales características descriptas para los genes de Clase II de mamíferos: existencia de copias múltiples, alto grado de polimorfismo, alta tasa de sustituciones no sinónimas en los sitios de reconocimiento del antígeno, y conservación de secuencia y estructura del promotor proximal.

Copy number variation (CNV) has been proved to be widespread in human, animal and plant genomes. Together with single nucleotide polymorphisms (SNPs), CVNs play a key role in genetic diversity. In this study, genome-wide detection of CNVs was performed based on SNP data from 24 Criollo Argentino horses genotyped with the GGP Equine70k array. Overall, 165 CNVs meeting stringent quality control criteria were identified and then aggregated into 87 CNV regions (CNVRs), representing a horse genome coverage of 13.69 Mb. Functional analysis of CNVRs allowed the identification of 337 genes implicated in a wide range of biological functions such as signal transducer activity (olfactory receptors), receptor activities and binding. Furthermore, enrichment analysis showed that the most represented protein classes (over 25%) were immunoglobulin receptor subfamily, immunoglobulins and major histocompatibility complex antigen (beta-2-microglobulin). La diversidad de los genes del complejo principal de histocompatibilidad (MHC) es clave en su función en la presentación de antígenos en el sistema inmune. La tipificación indirecta basada en microsatélites (STR) del MHC aporta información de la variabilidad genética y de la estructura poblacional y es una estrategia para conocer procesos selectivos y evolutivos. Con el fin de caracterizar una población de caballos de raza Árabe, se identificaron los haplotipos del MHC sobre la base de tres microsatélites (UM011, DRB2-STR2 y COR112) abarcando una región de ~1Mpb. El ADN genómico se extrajo de sangre y pelo de 30 caballos de cinco haras de la provincia de Buenos Aires y los STRs se amplificaron con cebadores fluorescentes para tipificar en secuenciador automático. Se estimaron parámetros poblacionales y de diversidad y la detección de haplotipos se realizó mediante análisis de segregación y con el programa de reconstrucción de haplotipos PHASE. Se reconocieron 24 haplotipos; entre ellos, 12 se verificaron por segregación en los registros de pedigree del Stud Book. Los resultados obtenidos demostraron la existencia de ligamiento con alelos conocidos de genes de clase II del equine leukocyte antigen (ELA) y la cantidad de haplotipos identificada permitió expandir la valoración de la diversidad del MHC equino. Esta metodología constituye una herramienta de utilidad y un método alternativo para la tipificación del MHC en linajes de caballos y en estudios poblacionales relacionados con la inmunidad., Genetic diversity of the major histocompatibility complex (MHC) is essential for the antigen recognition and presentation during the immune response. Indirect MHC typing by microsatellites (STR) provides data on genetic diversity and population structure, and provides knowledge of selective and evolutionary processes. In order to characterize a sample of Arab horses, we identified MHC haplotypes based on three STRs (UM011, DRB2-STR2 and COR112) covering a region of ~1Mpb. Genomic DNA was extracted from blood and hair of 30 horses from five farms of the province of Buenos Aires. STRs were amplified with fluorescent primers and typed in an automated sequencer. Population and diversity parameters were estimated. Haplotype detection was performed by segregation analysis and with the PHASE program for reconstructing haplotypes. Of the 24 haplotypes identified, 12 were verified by segregation in the Stud Book pedigree records. Our results showed linkage with known equine leukocyte antigen (ELA) class II alleles. The number of haplotypes identified allowed to expand the estimates of diversity in the equine MHC. Our objective was to obtain Equine Arteritis Virus M protein in prokaryotic system to test it as immunogen. LP02/C Equine Arteritis Virus cDNA was used as template to obtain and clone this protein. Equine Arteritis M protein was cloned in the expression vector pQE30 and the recombinant plasmid pQE30/M was transformed in Escherichia Coli M15 cells. The OD600 values of the IPTG-induced M15-pQE30/M culture showed an inhibition of the kinetics growth compared with the non-induced M15-pQE30/M and positive M15-pQE40/DHFR cultures. Several factors such as growth temperature, IPTG concentration and different inductors were analyzed but any of them showed an improvement in protein expression. Instead of E. coli M15strain, a new strain (E. coli BL21) was used and transformed with the pQE30/M. This resolved in part the growth inhibition observed in E. coli M15 cells, but no the recovery yield of the protein. So, as all gene products that affect cells kinetics growth are considered to be toxic, we argue that the lower yields in M protein recovery could be attributed to an associated toxicity of EAV-M protein from LP02/C strain in this expression system., Facultad de Ciencias Veterinarias (FCV)

Canine chronic superficial keratitis (CSK) is an inflammatory corneal disease that primarily occurs in German shepherd dogs (GSDs). Several studies support the hypothesis that CSK is an immune-mediated disease. To investigate the genetic factors associated with CSK development, the upstream regulatory regions (URRs) of the DLA-DRB, -DQA and -DQB genes were genotyped in 60 dogs, including 32 CSK animals. LD analysis identified two blocks (r2 ≤ 45), with two DLA-DRB1 and five DLA-DQB1 haplotypes. Analysis of DLA-URR alleles/haplotypes showed a significant association between DQB1*-154 [C/T] (p = 0.016) and CSK, suggesting that the T variant may increase the risk for developing CSK disease (OR = 3, 95% CI = 1.25–7.68). When haplotype associations were performed, the URR-DQB*CATT haplotype was significantly associated with CSK (p = 0.016), increasing the risk of develop this disease over two-fold (OR = 3, 95%, CI = 1.25–7.68). These results showed that dogs homozygous at DRB1*69 [C/T] had a risk for developing CSK disease that was over four times the risk for heterozygotes. This genetic association supports the previous clinical, histological and pharmacological studies that suggest that CSK is an immune-mediated disease, and this association could potentially be used to identify susceptible animals. Equine lentivirus receptor-1 (ELR1) has been characterized as the specific functional receptor that mediates equine infectious anemia virus (EIAV) entrance to horse macrophages. This receptor is tumor necrosis factor receptor superfamily member 14 (TNFRSF14). The aim of this study was to investigate the occurrence of allelic variants in the coding sequence of equine TNFRSF14 gene by screening for single-nucleotide polymorphisms (SNPs) in different equine populations. Forty seven horse samples were randomly selected from a reservoir of EIAV-seropositive and seronegative samples collected from different outbreaks and regions of Argentina. DNA samples were scanned via PCR and direct sequencing of exon 3 and exon 5 of TNFRSF14 gene. A total of 21 SNPs were identified, of which 11 were located in coding sequences. Within exon 5, four SNPs caused nonsynonymous substitutions, while two other SNPs caused synonymous substitutions in crucial residues (Ser112 and Thr114) implicated in the interaction with EIAV. Despite some of exon 5 variants occurred exclusively in EIAV-positive or EIAV-negative horses, critical residues for the function of the mature protein were conserved, accounting for selective pressures in favor of preserving the specific function of TNFRSF members and the host immune response. To our knowledge, this is the first report of the existence of allelic variations involving some crucial amino acid residues in horse ELR1. Further, it could be an initial step to test the possible functional relevance and relationship of these variants with EIAV infection and disease progression as well as to develop preventive strategies. Sequence information was obtained on the variation of the ELA-DRB upstream regulatory region (URR) after polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) cloning and sequencing of ≈ 220 bp upstream of the first exon of horse DRB genes. The sequence of the proximal URR of equine DRB is composed of highly conserved sequence motifs, showing the presence of the W, X, Y, CAAT and TATA conserved boxes of major histocompatibility complex (MHC) class II promoters. Five different polymorphic horse DRB promoter sequences were detected in five horse breeds. The results demonstrate the existence of polymorphism in the nucleotide sequences of the ELA-DRB URR, located in the functionally important conserved consensus sequences, the X2 box, the Y box and the TATA box, while conservation were observed in X1 and CAAT boxes. The nucleotide diversity among horse URRs was intermediate between that seen within human and mouse DRB promoters, suggesting the existence of another important source of variability in ELA-DRB genes. In addition, phylogenetic comparisons, identity analysis and sequence organization suggested that the reported sequences would correspond to an expressed ELA-DRB locus. However, further information about the functional significance of these promoter polymorphisms will probably be acquired through expression studies on the different sequences. Fil: Diaz, Silvina. Polymorphisms at Major Histocompatibility Complex (MHC) genes have been associated with resistance/susceptibility to infectious diseases in domestic animals. The aim of this investigation was to evaluate whether polymorphisms of the DRA gene the Equine Lymphocyte Antigen is associated with susceptibility to Equine Arteritis Virus (EAV) infection in horses in Argentina. The equine DRA gene was screened for polymorphisms using Pyrosequencing® Technology which allowed the detection of three ELA-DRA exon 2 alleles. Neither allele frequencies nor genotypic differentiation exhibited any statistically significant (P-values = 0.788 and 0.745) differences between the EAV-infected and no-infected horses. Fisher’s exact test and OR calculations did not show any significant association. As a consequence, no association could be established between the serological condition and ELA-DRA. The sequence of the proximal upstream regulatory region (URR) of the bovine DRB genes was amplified using oligonucleotide primers designed from the consensus among DRB sequences from different species. The obtained DNA sequence was 234 bp long and composed of highly conserved sequence motifs, showing the same organization as the HLA-DRB, H2-IAb, H2-IEb and ELA-DRB genes. In this paper we report the sequence of the proximal upstream regulatory region (URR) of the bovine DRB genes. These are the first sequence data on the upstream regulatory regions of MHC class II genes in cattle. The reverse oligonucleotide primer (5'-GAGAAATACAGACACACCATGC-3') was designed from the consensus among DRB sequences from different species: BoLA-DRB (accession numbers U77067-68, U78548 and D45357), SLA-DRB (M55165-6), HLA-DRB (S72812 and L07838-40), H2-IEb and H2-IAb (X86151-6). As a forward primer we used the oligonucleotide proposed by Turco et al. (1990) (5'-TGTTTCAGAAAAGGACCTTC-3'), which was designed from the consensus among HLA-DRB sequences corresponding to the promoter region of the DRB genes. A polymerase chain reaction (PCR) was carried out in a total volume of 25 μl comprising 2.5 mM MgCl 2 , 20 mM Tris-HCl (pH = 8.4), 50 mM KCl, 100 μM of each dNTP, 0.5 μM of each primer, 1.0 unit Taq polymerase (Gibco BRL, Life-Technologies, Grand Island, NY) and 50-100 ng DNA template. The genomic DNA was extracted from whole blood from one animal of the Saavedreno Creole breed. The amplification profile consisted of 1 min at 94 °C, followed by 30 cycles of 45 s at 94 °C, 45 s at 55 °C and 45 s at 72 °C, with a final extension of 3 min at 72 °C. The amplification products were cloned into a dT-tailed pGEM-T easy vector (Promega, Madison, WI), and three clones of each PCR product were sequenced on an Applied Biosystems 377 automated sequencer (BioResource Center, Cornell University, Ithaca, NY), using a T7 universal primer. All the sequenced clones exhibited 100% sequence similarity to each other. The nucleotide sequence of the proximal upstream control region of the BoLA-DRB genes (GenBank accession number AY040327) was 234 bp long and composed of highly conserved sequence motifs that included, from the 5' to the 3' direction, W, X, Y, CCAAT and TATA-like boxes (Fig. 1), showing the same organization of the conserved regulatory elements as the HLA-DRB, H2-IAb, H2-IEb and ELA-DRB genes (e.g. Louis et al., 1993, 1994; Singal et al., 1993; Singal & Qiu, 1994). Furthermore, the BoLA-DRB URR nucleotide sequence had higher identity with HLA-DRB sequences than with HLA-DQB sequences (data not shown). This suggests that the sequence AY040327 corresponds to a BoLA-DRB promoter. However, we are still unable to assign this sequence to a specific BoLA-DRB gene, as at least three BoLA-DRB genes have been reported to exist (Andersson et al., 1986; Muggli-Cocket & Stone, 1988). BoLA-DRB3 is the most expressed DRB gene in cattle, while DRB1 is expressed at a low level and DRB2 is a pseudogene (Burke et al., 1991).

A peptide-based indirect ELISA was developed to detect antibodies against Equine arteritis virus (EAV). Two peptides for epitope C of protein GP5 and fragment E of protein M were designed, synthesized, purified and used as antigens either alone or combined. Ninety-two serum samples obtained from the 2010 Equine viral arteritis outbreak, analyzed previously by virus neutralization, were evaluated by the ELISA here developed. The best resolution was obtained using peptide GP5. The analysis of the inter- and intraplate variability showed that the assay was robust. The results allow concluding that this peptide-based ELISA is a good alternative to the OIE-prescribed virus neutralization test because it can be standardized between laboratories, can serve as rapid screening, can improve the speed of diagnosis of EAV-negative horses and can be particularly useful for routine surveillance in large populations., Instituto de Genética Veterinaria, Facultad de Ciencias Veterinarias

Equine Arteritis Virus (EAV) has been shown to induce apoptosis in vitro but the induction of this mechanism has not been previously associated with any viral gene product. In this work, we found a cytotoxicity effect of the EAV gP5 protein on baculovirus-insect cells and a low yield of protein recovery. Besides, different morphological features by electron transmission microscopy, DNA fragmentation in agarose gel, TUNEL analysis and caspase 3 activity were found. Genetic variation in the equine leucocyte antigen-DRB (ELA-DRB) second exon was investigated using polymerase chain reaction (PCR) amplification, restriction fragment length polymorphism (RFLP) of PCR products (PCR-RFLP) and deoxyribonucleic acid (DNA) sequencing. Eight distinct PCR-RFLP patterns could be identified in the studied Argentine Creole (AC) horses. The number of observed patterns per individual ranged from four to six, thus confirming the presence of multiple DRB copies in AC horses. Three PCR-RFLP alleles and three new sequences were identified. The estimated rates of synonymous and non-synonymous substitutions among ELA-DRB exon 2 sequences were higher within the antigen recognition site (ABS) than on the non-ABS. Phylogenetic analysis showed that the nucleotide sequences clustered in two main groups, while some sequences were not included in either group. Finally, the identification of the number of alleles per animal, the phylogenetic and segregation analyses allowed us to explain the number of ELA-DRB loci. However, it was not possible to identify specific alleles with specific loci.

El teatro realizado en la calle interrumpe el devenir de las practicas cotidianas y re- significa el espacio publico, al tiempo que flexibiliza las relaciones entre realidad y ficcion modificando notablemente la percepcion del espectador. Nos proponemos analizar los principios esteticos e ideologicos de los espectaculos de calle de Los Calandracas -agrupacion paradigmatica del teatro callejero de la postdictadura argentina-, en tanto nos permiten comprender las concepciones filosoficas y artisticas del movimiento teatral callejero y comunitario en nuestro pais. En una epoca globalizada y “desideologizada”, y luego de la crisis institucional, politica y economica que estremecio a la sociedad argentina en 2001, estosgrupos propician una mayor articulacion con los movimientos sociales para recuperar el sentido politico, cultural e identitario a partir de la concepcion del arte como accion transformadora. El teatro aparece entonces como una herramienta de concienciacion, de resistencia y fortalecimiento de los lazos comunitarios.

South American camelids comprise the wild species guanaco and vicuña and their respective domestic relatives llama and alpaca. The aim of the present study was to determine by DNA analysis to which of these species belong a herd of camelids confiscated from a llama breeder but alleged to be alpacas by the prosecution, and to evaluate the usefulness of mitochondrial and autosomal DNA markers to solve judicial cases involving camelid taxa. Cytochrome b and cytochrome oxidase I mitochondrial genes and 7 STR were analyzed in 25 confiscated samples. Mitochondrial results were inconclusive because 18 of the sequestered samples presented haplotypes that corresponded to the guanaco haplogroup and the remaining seven belonged to a vicuña linage. Microsatellite data of casework samples and llama reference samples revealed different genetic profiles by the presence of private alleles at two microsatellites suggesting that the confiscated animals could be alpaca, or at least alpaca hybrids instead of pure llama. We investigated the association of equine arteritis virus (EAV) infection and three short tandem repeat (STR) polymorphisms located within or in close proximity to equine lymphocyte antigen (ELA) region. We used a case-control design as a first approach before proceeding to select candidate genes. One hundred and sixty-five Silla Argentino horses were taken in 2002 from positive serological detections of EAV in Argentina, to determine whether STR genotypes were correlated to genetic susceptibility to EVA. Allele frequency distribution did not show significant differences between both groups (P = 0.0781). Demodectic mange is a well-known parasitic skin disease characterized by the presence of a larger than normal number of Demodex mites (Demodex canis) in the skin of dogs. Recent research has suggested that major histocompatibility complex (MHC) class II expression is higher in the skin of dogs suffering from demodicosis than in normal ones. We have investigated whether canine Dog Leukocyte Antigen (DLA) class II alleles are associated with canine juvenile generalized demodicosis (JGD). In the present study, the analysis of microsatellite markers (FH2202, FH2975 and FH2054) linked to DLA was made in Boxer, Argentinean Mastiff and mixed breed dogs. DNA samples from 56 dogs affected with the disease and 60 breed-matched controls collected in Argentina were analysed. A highly significant association, in some of the analysed markers, in all breeds with the presence of demodicosis was observed with P0.05 and odds ratio (OR)or =5. The results of this study suggest that an underlying DLA association exists with demodicosis in dogs and that this may represent an important immunological risk factor in the aetiology of this condition. This information could be used in the future to develop diagnostic tests to prevent canine JGD.

In the construction of a modern Argentina, inmigration plays a very important role. In the case of theatre, two different movements -Teatro Abierto and communitarian theatre- are fundamental in the way they stage the problematics of this inmigration. We will analyse this phenomenon through the study of three plays - Gris de ausencia by Roberto Cossa, For export by Patricio Esteve and Lejana tierra prometida by Ricardo Halac -, and the production of the Grupo comunitario Catalinas Sur, Venimos de muy lejos. En la construcción de la Argentina moderna la inmigración ha jugado un rol fundamental. En diversas fases de su historia, los movimientos de Teatro Abierto y de teatro comunitario se ocuparon de retratar la problemática de dicha inmigración, como la pérdida de la propia identidad, el exilio político, los exilios interiores, el desarraigo y la soledad. Teniendo en cuenta estas premisas analizaremos algunas piezas paradigmáticas de Teatro Abierto en lo que atañe a esta temática -Gris de ausencia, de Roberto Cossa, For export, de Patricio Esteve y Lejana tierra prometida, de Ricardo Halac-, así como también Venimos de muy lejos en tanto paradigma ideológico y estético del Grupo comunitario Catalinas Sur. Nos proponemos analizar estas piezas tanto desde el punto de vista semántico como a nivel formal, teniendo en cuenta cómo se plantea la búsqueda y la reconstrucción de la propia identidad desde poéticas escénicas y actorales diversas.

The polymorphism of equine lymphocyte antigen (ELA) class II DRA gene had been detected by polymerase chain reaction-single-strand conformational polymorphism (PCR-SSCP) and reference strand-mediated conformation analysis. These methodologies allowed to identify 11 ELA-DRA exon 2 sequences, three of which are widely distributed among domestic horse breeds. Herein, we describe the development of a pyrosequencing-based method applicable to ELA-DRA typing, by screening samples from eight different horse breeds previously typed by PCR-SSCP. This sequence-based method would be useful in high-throughput genotyping of major histocompatibility complex genes in horses and other animal species, making this system interesting as a rapid screening method for animal genotyping of immune-related genes. In order to detect switching and/or manipulation of samples, the owner of a stallion asked our lab to perform a DNA test on a positive doping urine sample. The objective was to compare the urine DNA profile versus blood and hair DNA profiles from the same stallion. At first, 10 microsatellite markers were investigated to determine the horse identity. No results were obtained when horse specific markers were typed in the urine sample. In order to confirm the species origin of this sample we analyzed the mitochondrial cytochrome b gene. This analysis from blood and hair samples produced reproducible and clear PCR-RFLP patterns and DNA sequence match with those expected for horse, while the urine sample results were coincident with human. These results allowed us to exclude the urine sample from the questioned stallion and determine its human species origin, confirming the manipulation of urine sample. Fil: Diaz, Silvina. We report the nucleotide sequence and genetic diversity of four Equine Arteritis Virus (EAV) ORF 5 and 6 from Argentina isolates, obtained from asymptomatic virus-shedding stallions. Nucleic acid recovered from the isolates were amplified by RT-PCR and sequenced. Nucleotide and deduced amino acid sequences from the Argentine isolates were compared with 17 sequences available from the GenBank. Phylogenetic analysis revealed that the Argentine isolates grouped together in a definite cluster near European strains. Despite the greater genetic variability among ORF 5 from different isolates and strains of EAV, phylogenetic trees based on ORF 5 and 6 are similar. Both trees showed that virus sequences from America and Europe segregate into distinct clades based on sequence analysis of either ORF 5 or 6. The second exon of equine leucocyte antigen (ELA)-DQB genes was amplified from genomic DNA of 32 Argentine Creole horses by PCR. Amplified DNA was analysed by PCR-restriction fragment length polymorphism (RFLP) and PCR-single-strand conformation polymorphism (SSCP). The PCR-RFLP analysis revealed two HaeIII patterns, four RsaI patterns, five MspI patterns and two HinfI patterns. EcoRI showed no variation in the analysed sample. Additional patterns that did not account for known exon 2 DNA sequences were observed, suggesting the existence of novel ELA-DQB alleles. PCR-SSCP analysis exhibited seven different band patterns, and the number of bands per animal ranged from four to nine. Both methods indicated that at least two DQB genes are present. The presence of more than two alleles in each animal showed that the primers employed in this work are not specific for a unique DQB locus. The improvement of this PCR-RFLP method should provide a simple and rapid technique for an accurate definition of ELA-DQB typing in horses.

The genetic diversity and structure of 72 Appaloosa horses belonging to a closed breeding population from an ecological reserve in Buenos Aires, Argentina, was investigated using eight microsatellite markers from the International Society for Animal Genetics panel. Our data showed that this Appaloosa horse population had an elevated degree of genetic diversity (He = 0.746) and did not present a significant increase of homozygous individuals (FIS~0). However, the short tandem repeats, AHT5, ASB2, HTG10 and VHL20, were not in Hardy-Weinberg equilibrium (P-value < 0.05). Genetic relationships between this population and other well known horse breeds showed that Appaloosa horses from Argentina could have had their origin in the horses of the Nez Perce's people in Idaho while other Appaloosa horses may have had influences from Andalusian and Lusitano breeds. This closed breeding population conserves an important degree of Appaloosa genetic diversity and notwithstanding its particular breeding characteristics, represents a valuable genetic resource for conservation., Instituto de Genética Veterinaria
