ABSTRACT: Probiotics and synbiotics can mitigate the negative health consequences of early-life antibiotic exposure. This study aimed to determine whether supplementation with Bifidobacterium longum ssp. infantis 79 (B79) or synbiotics composed of B79 and 2′-fucosyllactose (2′-FL) could mitigate the negative impact of ceftriaxone exposure in early life. We found that antibiotic-treated mice exhibited lower BW, crypt depth, short-chain fatty acid content, and α-diversity indices at weaning, whereas they had increased relative abundance of opportunistic pathogens (such as Enterococcus and Staphylococcus) and decreased relative abundance of intestinal commensal bacteria. Supplementation with B79 and 2′-FL revived these antibiotic-induced negative effects and reduced the mRNA expression of IL-6, IL-12p40, and TNF-α in the spleen at weaning. Moreover, B79 and 2′-FL supplementation persistently improved crypt depth, propionic acid synthesis, and IgG and secretory IgA production, as well as revived the gut microbiota structure and composition in adulthood. Overall, our study suggests that early-life supplementation with B79 alone or in combination with 2′-FL can mitigate ceftriaxone-induced negative effects on the gut microbiota and intestinal and immune development of mice, and these improvements can partially last into adulthood.

Abstract Objective Malnutrition is a common issue in patients with head and neck squamous cell carcinoma (HNSCC), leading to compromised immune function and diminished treatment efficacy. This is particularly concerning for therapies targeting the PD-1 pathway, which are pivotal in cancer treatment. The aim of this retrospective study was to explore the potential benefits of immunonutrition in improving immune responses and clinical outcomes for these patients. Methods In the study, 49 individuals diagnosed with HNSCC were enrolled and divided into two groups: one group received specialized immunonutrition support designed to enhance immune function, while the other group received standard nutritional care. The researchers assessed immune function by evaluating CD4+ and CD8+ T cell counts, which are critical indicators of immune health. Additionally, clinical outcomes were monitored, focusing on infection rates and the duration of hospital stays. Results Patients who received immunonutrition showed a significant improvement in immune function, as indicated by higher levels of CD4+ and CD8+ T cells. Furthermore, these patients experienced shorter recovery times and lower infection rates compared to those receiving standard nutrition. Conclusion These findings suggest that immunonutrition may play a vital role in enhancing the effectiveness of cancer therapies, including PD-1 inhibitors. By supporting immune health, immunonutrition could potentially improve patient outcomes and quality of life during and after treatment. This study underscores the importance of integrating nutritional support into cancer care, particularly for patients with HNSCC. As the field of oncology continues to evolve, incorporating strategies that address nutritional deficiencies could be key to optimizing treatment efficacy and improving overall survival rates.

The COVID-19 pandemic has significantly impacted human life, posing serious physical and psychological threats, particularly to the elderly. While individuals of all ages are susceptible to contracting COVID-19, older people face a heightened risk of developing various diseases due to age-related immunophysiological changes and preexisting health conditions. The interplay between immune health and physical activity is believed to hold even greater significance during a pandemic. Recent findings from our research indicate that the intervention of square stepping exercise (SSE), characterized by a rhythmic and controlled stepping pattern, resulted in increased levels of Brain-Derived Neurotrophic Factor (BDNF) in the elderly. BDNF, known to influence not only nerve cells but also immune cells, suggests a potential link between SSE and immune system modulation. Consequently, this exercise regimen holds promise in counteracting age-related immunophysiological changes, fine-tuning immune responses, and mitigating the severity of potential new virus outcomes, such as ‘Disease X.’ This review aims to underscore the significance of integrating SSE as a home-based program, serving as a potent tool to enhance immune resilience, prepare for future potential pandemics, and empower older individuals during challenging times. Through the practice of SSE, older adults may strengthen their ability to navigate the challenges posed by pandemics and maintain a sense of control over their well-being.

Infant formula (IF) based on cow milk and goat milk is a substitute food for infants who are underfed with human milk. In our previous study, we reported the composition and physicochemical stability of IF based on milk from cows and goats and a combination of both milks. Here, we investigated the effects of these 3 IFs on intestinal immunity and short-chain fatty acid production (SCFAs) using human microbiota-associated (HMA) mice and selected human milk as a positive control. The results showed that goat milk-based IF is associated with a functional immune advantage, due to the rise in the levels of immune-related cytokines interleukin (IL)-2 and IL-10, decreased levels of intestinal permeability markers D-lactic acid and endotoxin, and increased mRNA levels of intestinal tight junction proteins occludin and claudin. In addition, the intestine of mice fed with goat milk -based IF contained 12.06 μmol/g acetate, 2.42 μmol/g propionate, and 1.72 μmol/g butyrate, which reached 69%, 79 %, and 60% of the levels in human milk, respectively. Our results indicate that goat milk-based IF improves intestinal immune function and promotes the production of intestinal SCFAs.

In patients with sepsis, immunosuppression is associated with poor prognosis, and immune monitoring during the early stages has prognostic value. This study aims to explore immunologic parameters associated with sepsis prognosis, potentially identifying patients who may benefit from immunotherapy, improving intensive care survival.Methods: A total of 65 patients with sepsis from the Department of Emergency Medicine were divided based on survival at 28 days (47 in the survival group, 18 in the non-survival group). Peripheral blood was collected to measure absolute lymphocyte count and T lymphocyte subpopulations, including the percentage and absolute count of total T cells, CD4+ T, CD8+ T, and NK cells, and the percentages of naïve CD4+ T, central memory CD4+ T, effector CD4+ T, effector memory CD4+ T, naïve CD8+ T, central memory CD8+ T, effector CD8+ T, effector memory CD8+ T, CD4+HLA-DR+ T, and CD8+HLA-DR+ T cells, and Tregs. The differences in these parameters between the two groups were compared and a regression model was constructed to identify possible risk factors for death in patients with sepsis.Results: The absolute lymphocyte count, absolute T cell count (CD3+, CD4+, and CD8+) and naïve CD4+ T cell percentage were significantly lower in the non-survival group. Conversely, Tregs were higher in patients who did not survive sepsis. In regression analysis, the absolute lymphocyte count and naïve CD4+ T cell percentage remained statistically significant. The receiver operating characteristic curve showed that a model based on the absolute lymphocyte count (435 cells/μL) and naïve CD4+ T cell percentage (20.25%) performed best in predicting sepsis prognosis.Conclusion: Monitoring of absolute lymphocyte count and analysis of T cell subtypes in the early phase of sepsis is predictive of outcome and may help identify those patients who would benefit from immunotherapy, improving survival.Keywords: sepsis, absolute lymphocyte count, T cell subpopulations, immune function

Abstract Background Acute myocardial infarction (AMI) is a leading cause of death and morbidity worldwide. Ferroptosis, a form of regulated cell death, plays a critical role in modulating immune functions during AMI. This study aimed to identify ferroptosis-related hub genes that could serve as potential therapeutic targets in the progression of AMI. Methods Bioinformatics was used to identify overlapping genes associated with ferroptosis and the infiltration of 22 immune cells by Cell-type Identification by Estimating Relative Subsets of RNA Transcript (CIBERSORT) analysis. The expression of ferroptosis-related genes in AMI was validated across independent datasets, clinical samples, and in vitro cellular experiments. The predictive value for heart failure was evaluated in the first dimension of principal component analysis (PCA) using receiver operating characteristic (ROC) analysis. Results The study identified 11 key ferroptosis-related genes significantly correlated with immune cell abundance. CIBERSORT analysis highlighted immune dysregulation in AMI. JDP2, DUSP1, TLR4, NFS1, and SLC1A5 were identified as potential biomarkers for AMI progression. Additionally, JDP2, DUSP1, and DDIT4 demonstrated strong predictive value for long-term heart failure. Conclusion This study highlights the potential association of ferroptosis-related genes with the pathogenesis of AMI, suggesting a role in the molecular mechanisms that may underlie acute coronary events.

Objective: To investigate the protective effects of different doses of Fomes officinalis Ames polysaccharides (FOP) on intestinal flora and immune function in exercise-induced immunosuppression (EIS) rats. Methods: Fifty SPF male SD rats aged 7 to 8 weeks were randomly divided into control group (NC group), exercise training group (Ex group), exercise training+low-dose FOP group (LFOP group, 40 mg/(kg·d)), exercise training+medium-dose FOP group (MFOP group, 60 mg/(kg·d)) and exercise training+high-dose FOP group (HFOP group, 80 mg/(kg·d)). Ex group and FOP group were trained on the treadmill for 6 weeks to construct EIS model. LFOP, MFOP and HFOP groups were given FOP after each training. After the intervention, serum IgG, IgM, IL-6, IL-10, INF-γ, TNF-α, LPS levels and intestinal short-chain fatty acid content were detected by enzyme-linked immunosorbent assay. The number of serum CD4+ and CD8+ was detected by cell analyzer. 16S rDNA was used to detect the structural changes of intestinal flora. Objective: To investigate the impact of giant salamander protein peptide (CGS) and its Maillard reaction product (CGSXyU) on lowering blood glucose and lipid levels in type Ⅱ diabetic mice and enhancing immunomodulatory functions in immunosuppressed mice, and anti-fatigue effects in normal mice. Methods: Mice were randomly assigned to a blank control group, a model control group, a positive control group (200 mg/kg metformin), a low-dose sample group (CGS/CGSXyU-L, 50 mg/kg), and a high-dose sample group (CGS/CGSXyU-H, 200 mg/kg). Following the successful induction of diabetes with 100 mg/kg alloxan, the mice underwent gavage treatment for 3 weeks, after which their fasting blood glucose levels, serum total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) levels were measured. Mice were randomly assigned to a blank control group, a model control group, a positive control group (50 mg/kg levamisole), a low-dose sample group (CGS/CGSXyU-L, 50 mg/kg), and a high-dose sample group (CGS/CGSXyU-H, 200 mg/kg). Abstract To assess the safety and efficacy of fecal microbiota transplantation (FMT) as an adjunctive therapeutic intervention for IgA nephropathy (IgAN). Fifteen patients with IgA nephropathy were recruited based on inclusion and exclusion criteria and underwent FMT using enteric microbial capsules. Clinical indicators, intestinal microbiota and metabolomic profiles, as well as changes in serum immune cells and cytokines, were monitored before and after FMT. No severe adverse reactions were observed in the subjects. After FMT, there was a reduction in the 24-h urinary protein quantification in subjects. The relative abundances of Phocaeicola_vulgatus, Bacteroides_uniformis, Prevotella_copri, Phocaeicola_dorei, Bacteroides_ovatus, Bacteroides_xylanisolvens, Parabacteroides _distasonis, Bifidobacterium_pseudocatenulatum, Bacteroides_sp._HF-162, and Bifidobacterium_longum changed after FMT. In terms of intestinal metabolites, the levels of acylcarnitine18:0 (ACar.18:0), cotinine, N-arachidonoyl-L-serine, phosphatidylcholine (PC. (18:3e/22:6)), serotonin, and fumagillin showed significant changes. Flow cytometry analysis showed the absolute count of plasma B cells decreased in subjects, and this change correlated with alterations in the intestinal microbiota and metabolites. This study preliminarily evaluates the safety and efficacy of FMT in patients with IgAN. No significant adverse reactions were observed, and the administration of FMT alongside ACEI/ARB therapy was effective in reducing urinary protein levels in patients with IgAN, a process that may be associated with B-cell immunity.

Objective To investigate the mechanism of human leukocyte antigen-G (HLA-G) expression and T cells activation in triple-negative breast cancer(TNBC) cells to exert immunosuppressive function. Methods From three groups of breast cancer cell lines（MDA-MB-231，HCC1937， MDA-MB-468）,real-time quantitative polymerase chain reaction (RT-qPCR) and Western blot were detected and screened that the breast cancer cell line was HCC1937,which with high expression of HLA-G, and low expression of fibrinogen-like protein 1 (FGL1), programmed death factor ligand 1 (PD-L1), as well as sialic acid-binding immunoglobulin-like lectin 15 (SIGLEC15) . HCC1937 cells were divided into NC group (control group) and KD group (knockdown group), and RNA interference (RNAi) lentiviral vectors were constructed and transfected into KD cells to knock down HLA-G gene, and the verification was carried out. HCC1937 cells were treated with HLA-G antibody blocking/nonblocking, and Jurkat cells were treated with CD3 and CD28 activation/non-activation, respectively, and the two types of cells were co-cultured into 8 groups: NC--, KD--, NC+-, KD+-, NC-+, KD-+, NC++, and KD++ groups. Cell scratch assay, MTT and ELISA method were used to detect cell migration rate, proliferation rate, and interferon (IFN)-γ and interleukin (IL)-2 levels in cell supernatants, respectively. Results RT-qPCR showed that the HLA-G gene was successfully knocked down in HCC1937 cells. Compared with the NC++ group, the migration rate of the KD++ group was significantly increased (P＜0.05), but there was no statistical difference in the cell proliferation rate between each pair of co-culture groups (P＞0.05). The concentration of IFN-γ in the KD++ group was lower than that in the NC++ group and the NC-+ group (P＜0.05), but there was no significant difference in the concentration of IL-2 between each pair of co-culture groups (P＞0.05). Conclusion HLA-G in TNBC cells may promote the secretion of IFN-γ by activing T cells, and IFN-γ can inhibit the immune function of T cells and promote tumor metastasis after HLA-G decreased."

Patient outcomes were classified based on survival or mortality. Biomarkers, including sIL-7R levels and illness severity scores, were documented. All statistical analyses, including predictive modeling and comparisons were carried out using SPSS v.23.0 software and R software.Results: On the fifth day post-admission, sIL-7R levels significantly decreased in both the survival and death groups, compared with levels on day one (2.09 ± 0.65 vs 1.07 ± 0.53 ng/mL, P < 0.01). There was a significant correlation between the sIL-7R level and the CD3+ T-lymphocyte count (CD3+) (r = 0.44) and lymphocyte count (LYM) (r = 0.42). The combination of the sIL-7R level with the Sequential Organ Failure Assessment (SOFA) score demonstrated optimal predictive value for clinical outcomes in patients with sepsis, demonstrated by an area under the receiver operating characteristic curve of 0.998.Conclusion: sIL-7R levels are correlated with CD3+ and LYM counts. Additionally, the combination of serum sIL-7R level and SOFA score provides a robust method for predicting sepsis outcomes.Keywords: immune function, prognosis, sepsis, soluble interleukin-7 receptor

Objective: To clarify the specific mechanisms of action of raw Phellodendron chinense Schneid. (RPC) and saltwater-processed PC (SPC) in the treatment of rats with a kidney-yin deficiency pattern (KYDP). Methods: Healthy rats were administered hydrocortisone to establish a KYDP model. The rats were divided into seven groups: blank control, model, positive control (Liuwei Dihuang pills), high-dose RPC, low-dose RPC, high-dose SPC, and low-dose SPC. Enzyme-linked immunosorbent assay was used to measure the levels of cAMP, cGMP, TRH, TSH, T3, T4, IFN-γ, TNF-α, and testosterone in the serum and the levels of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in the liver. TRH mRNA expression in the rat hypothalamus was measured using RT-PCR. THRα1+2 protein expression in the hypothalamus of rats was measured using Western blot. Immunohistochemistry was performed to determine the expression levels of FAS, FasL, and TSHR. Flow cytometry was used to determine CD4+ and CD8+ T lymphocyte levels. Illumina MiSeq sequencing technology was used to evaluate the diversity of intestinal flora in KYDP rats. Results: The cAMP/cGMP ratio was significantly higher in the model group than in the blank control group (P = .048). Compared with the model group, after administration, the levels of the above-mentioned serum and liver indexes decreased, except that of testosterone. The CD4+/CD8+ ratio also decreased. Compared with the RPC group, the levels of T3, IFN-γ, FAS, FasL, and TSHR in the SPC group decreased whereas that of testosterone increased. Additionally, immune function and intestinal flora diversity improved in the SPC group. SPC proved to be more effective in improving liver energy metabolism in KYDP rats than RPC. Conclusion: SPC had a better therapeutic effect on KYDP than RPC. The underlying mechanism of action may be related to improvements in liver energy metabolism, immune function, and intestinal flora diversity.

Abstract Background The aim of this study is to investigate the impact of arginine on immune function and postoperative complications in colorectal cancer (CRC) patients. Methods We conducted a comprehensive search to identify eligible RCTs in various databases, such as PubMed, Cochrane Library, EMBASE, Web of Science, MEDLINE, China National Knowledge Infrastructure (CNKI), Wanfang, VIP Medicine Information System (VIP), and Chinese Biomedical Database (CBM). This study aimed to examine IgA, IgG, and IgM levels as well as CD4+ and CD8+ counts as well as the CD4+/CD8+ ratio. Anastomotic leaking, length of stay (LOS), and surgical site infection (SSI) were included as secondary outcomes. Stata (StataCorp, version 14.0) was utilized for data analysis. To ensure the results were reliable, we used meta-regression, sensitivity analysis, and publication bias analysis. Results A total of 24 publications (including 1883 patients) out of 681 that were retrieved fulfilled the inclusion criteria. The arginine group showed notable improvements in humoral immunity, with gains in IgA (SMD=0.45, 95% CI: 0.30-0.60), IgG (SMD=0.80, 95% CI: 0.64-0.96), and IgM (SMD=0.66, 95% CI: 0.39-0.93). With regards to cellular immunity, the arginine group exhibited a substantial increase in the CD4+ T cell count (SMD = 1.03, 95% CI: 0.67-1.38) compared to the control group. However, the CD4+/CD8+ ratio decreased significantly (SMD=1.37, 95% CI: 0.88-1.86) in the same arginine group, indicating a change in the balance between these two cell types. Additionally, the CD8+ T cell count showed a notable decrease (SMD=-0.70, 95% CI: -1.09 to -0.32) in the arginine group when compared to the control group. Anastomotic leakage was also considerably lower in the arginine group (SMD=-0.05, 95% CI: -0.08 to -0.02), the rate of SSIs was lower (RR = -0.02, 95% CI: -0.05-0), and the length of time patients spent in the hospital was shorter (SMD=-0.15, 95% CI: -0.38 to -0.08). Conclusions After radiation treatment for CRC, arginine improves immune function and decreases the risk of infection problems. Abstract Background The microbiota and metabolites in the gastrointestinal tracts of female animals at different reproductive periods are very important to the growth, development, and health of themselves and their offspring. However, the changes in the gastrointestinal microbiota and metabolites throughout reproductive period of different sheep breeds and their effects on the growth and development of offspring lambs are still unclear. Hence, this study presents an assessment of the reproductive hormone levels, immune levels, rumen microbiota, and metabolites in Hu sheep and Suffolk ewes at different reproductive periods and their effects on the growth and development of offspring lambs. Results Hu sheep and Suffolk during non-pregnancy, pregnancy, and lactation were used as the research objects to determine reproductive and immune indexes of ewes at different periods, analyze rumen microbiome and metabolome, and track the growth performance and development of offspring lambs. The results showed that the reproductive hormone and immune levels of Hu sheep and Suffolk underwent adaptive changes across different reproductive periods. Compared with non-pregnancy, the microbial energy metabolism and lipid metabolism function decreased during Hu sheep pregnancy, and energy metabolism function decreased during lactation. In Suffolk, energy metabolism, glycan biosynthesis, and metabolism function were enhanced during pregnancy, and the metabolism of cofactors and vitamins was enhanced during lactation. Prevotella increased in Suffolk during pregnancy and lactation (P < 0.05) and was positively correlated with the birth weight and body size of the lambs (P < 0.05). Moreover, the abundances of Butyrivibrio and Rikenellaceae_RC9_gut_group during pregnancy were positively correlated with the intestinal immunity of the offspring lambs (P < 0.05), thereby regulating the intestinal immunity level of the lambs. Metabolomic analysis revealed that the protein digestion, absorption, and amino acid metabolism of Hu sheep were enhanced during pregnancy, which provided amino acids for the growth and development of pregnant ewes and fetuses and was significantly correlated with the birth weight, body size, and intestinal immunity of lambs (P < 0.05). Simultaneously, there was an increase in acetate and propionate during the pregnancy and lactation period of both Hu sheep and Suffolk, providing energy for ewes during reproductive period. Moreover, the microbiota during the lactation period was significantly correlated with the milk quality and lambs daily gain (P < 0.05). Conclusions This study revealed the characteristic succession changes in the rumen microbiota and its metabolites at different reproductive periods in sheep breeds and their regulation of reproductive hormone and immune levels and identified their potential effects on the growth and development of offspring lambs. The findings provide valuable insights into the health and feeding management of different sheep breeds during the reproductive stage. Video Abstract

Abstract Introduction This study analyzed and discussed the characteristics of peripheral blood lymphocytes (PBLs) in patients with endometrial carcinoma (EC) to explore the PBLs’ clinical application value. Methods This single-center case‒control study analyzed the clinical data of patients with EC and uterine fibroids who underwent surgery at the First Affiliated Hospital of Chongqing Medical University between October 2018 and October 2023 retrospectively. The Center for Clinical Molecular Medical Detection of our hospital performed PBLs detection using flow cytometry, and recorded the detection results in the electronic medical records system. Between-group and subgroup comparisons of PBLs in patients with EC were analyzed using t-test or Mann–Whitney U test. The effect of surgery on PBLs in patients with EC was assessed using a paired t-test or the Wilcoxon signed rank test. Results The immune function of patients with EC was significantly lower than that of healthy people (P < 0.05) and those with benign uterine diseases (P < 0.05) and was related to body mass index (BMI), hypertension, diabetes, and blood lipids (P < 0.05). In patients with EC, the PBLs counts decreased significantly after surgery (P < 0.05) and more kinds of lymphocytes were affected in the laparoscopic surgery group than in the open surgery group. Conclusions With the decrease of PBLs counts, the immune status of patients with EC is impaired. Metabolic syndrome (Mets), including obesity, hypertension, diabetes, and high blood lipids, also affects the immune function of patients with EC. And for EC patients, the effect of laparoscopic surgery is greater than that of open surgery. PBLs has the potential to be one of indicator during the diagnosis and treatment of EC. Trial registration This study was retrospectively registered by the Ethics Committee of the First Affiliated Hospital of Chongqing Medical University (approval number K2023-578).

Deoxynivalenol (DON) is a mycotoxin that is produced by various species of Fusarium and is ubiquitous in food and feed. At low concentrations, it can cause metabolic disorders in animals and humans and, at high concentrations, it can lead to pathological changes in the body. The impact of DON on human/animal health and animal productivity has thus attracted a great deal of attention around the world. DON causes severe damage to the intestine, including compromised intestinal barrier, mucosal damage, weakened immune function, and alterations in gut microbiota composition. These effects exacerbate intestinal infections and inflammation in livestock and poultry, posing adverse effects on overall health. Furthermore, research into biological methods for DON detoxification is a crucial avenue for future studies. This includes the utilization of adsorption, enzymatic degradation, and other biological approaches to mitigate DON’s impact, offering new strategies for prevention and treatment of DON-induced diseases. Future research will focus on identifying highly efficient detoxifying microorganisms or enzymes to reduce DON levels in food and feed, thereby mitigating its risks to both animals and human health.

Abstract Purpose To explore the influence of circulating lymphocyte subsets, serum markers, clinical factors, and their impact on overall survival (OS) in locally advanced nasopharyngeal carcinoma (LA-NPC). Additionally, to construct a nomogram predicting OS for LA-NPC patients using independent prognostic factors. Methods A total of 530 patients with LA-NPC were included in this study. In the training cohort, Cox regression analysis was utilized to identify independent prognostic factors, which were then integrated into the nomogram. The concordance index (C-index) was calculated for both training and validation cohorts. Schoenfeld residual analysis, calibration curves, and decision curve analysis (DCA) were employed to evaluate the nomogram. Kaplan-Meier methods was performed based on risk stratification using the nomogram. Results A total of 530 LA-NPC patients were included. However, the impact of the Chinese visceral adiposity index (CVAI) on the efficacy of SARS-CoV-2 vaccines remains poorly understood. This study aims to explore the impact of CVAI on the production of neutralizing antibodies (NAb) in inactivated SARS-CoV-2 vaccines and the potential mechanism, thereby optimizing vaccination guidance.Methods: In this cross-sectional study, 206 health workers (completed two SARS-CoV-2 vaccination on February 8th and March 10th, 2021, respectively) were recruited. All baseline anthropometric parameters of the participants were collected, and venous blood samples were obtained 6 weeks later to measure peripheral innate immune cells, inflammatory cytokines, and NAb titers against SARS-CoV-2. CVAI were calculated according to the formula and divided participants into two groups depending on CVAI median.Results: The median NAb titer among healthcare workers was 12.94 AU/mL, with an efficacy of 87.86% for the SARS-CoV-2 vaccine. NAb titers were lower in the CVAI dysfunction group than in the CVAI reference group (median: 11.40 AU/mL vs 15.57 AU/mL), the hsCRP levels (median: 0.50 mg/L vs 0.30 mg/L) and peripheral monocyte count (mean: 0.47 × 109/L vs 0.42 × 109/L) in the CVAI dysfunction group were higher than in the CVAI reference group. Additionally, CVAI showed positive correlations with hsCRP, monocytes, lymphocytes, and B-lymphocytes, and a negative correlation with NAb titers.Conclusion: CVAI may inhibit SARS-CoV-2 neutralizing antibody expression through inducing immune dysfunction and chronic inflammation. Abstract Bronchopulmonary dysplasia (BPD) is one of the most prevalent and severe chronic lung diseases in premature infants. The objective of the current study was to screen for key BPD‐associated genes and pathways by transcriptomic analysis from clinical patients and animal models. In our study, the differentially expressed genes were screened from 58 children with 14‐day BPD and 40 normal children in the GSE32472 dataset of the Gene Expression Omnibus database. Then, we identified four hub genes (Cd3e, Cd3g, Cd247, and Itk) and a signaling pathway (T cell receptor signaling pathway) by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis and protein‐protein interaction analysis. The differential expression of the relevant pathways and gene sets among the groups was verified via GSEA analysis. Subsequently, a rat model of BPD with hyperoxia‐induced lung injury was established, and the transcriptome sequencing of the whole lung tissue was performed. A similar analysis was done on the sequencing data of the hub genes and associated pathway screening to verify the accuracy. Ultimately, quantitative polymerase chain reaction was performed to validate the transcriptomics data of core gene expression in the rat model. Our study revealed that the downregulation of the expression of the above four key genes in the course of BPD leads to a decrease in the function of T cell receptor signaling pathways, it causes immune dysfunction and increases the severity of lung inflammation as well as susceptibility to other respiratory infectious diseases.

ABSTRACT: This study investigated the effect of Macleaya cordata extract (MCE) on the performance, gut health, and microbiota of laying hens. A total of 192 thirty-wk-old Hyline brown laying hens were randomly divided into 4 treatment groups. The CON group received a basal diet, while the low (MCE250), medium (MCE350), and high (MCE450) dose groups were supplemented with 250, 350, and 450 mg/kg MCE, respectively. The egg weight and Haugh unit demonstrated a linear and quadratic increase with the MCE dose during the initial 4-wk period of the experiment (P < 0.05). Furthermore, the dietary supplementation of MCE led to a significant enhancement in eggshell thickness and Haugh unit at wk 8 and the data showed a statistically significant linear and quadratic increase (P < 0.05). Serum cytokine assay showed that dietary supplementation of MCE led to linear and quadratic increases in IL-4 and IL-10 level (P < 0.05). Dietary supplementation of 350 and 450 mg/kg MCE was observed to result in linear and quadratic increase in serum lysozyme levels (P < 0.05). The addition of MCE to the diet resulted in a linear and quadratic increase in the levels of sIgA in the jejunum and ileum (P < 0.05). In terms of gene expression, the addition of MCE to the diet resulted in linear and quadratic increases in the expression of IL-10, IgA, Serpinb14, Serpinb14B, and OIH (P < 0.05). The expression of jejunal genes pIgR and IL-4 was observed to increase in a linear and quadratic manner, respectively, following the dietary addition of 350 mg/kg MCE and IL-1β decreased in a linear manner (P < 0.05). Moreover, these favorable effects were maximized at medium dosage (350 mg/kg) of MCE addition, and intestinal microbial composition in the control and MCE350 groups was assessed. 350 mg/kg MCE increased the relative abundance of Bryobacter and Parasutterella and decreased the relative abundance of Erysipelatoclostridium in the cecum (P < 0.05). Spearman correlation analysis revealed that Bryobacter, Parasutterella, Skermanella, and Erysipelatoclostridium were associated with nonspecific immune functions (P < 0.05). In conclusion, 350 mg/kg MCE supplementation elevated the immune response, and upregulated the expression of genes related to protein production in eggs, thereby improving egg quality. These effects may be associated with changes in the microbiota, specifically Bryobacter, Parasutterella, and Erysipelatoclostridium.

Alpinia oxyphylla, a perennial herb belonging to the Zingiberaceae family, has a long history of traditional medicinal use. The present study evaluated the efficacy of different concentrations of Alpinia oxyphylla essential oil (AEO) on the growth performance, serum antioxidation capacities, immune function, apparent digestibility of nutrients, and gut microbiota in fattening pigs. A total of 120 pigs were divided into five treatments, with six replicates each and four pigs per replicate. The pigs were fed a basal diet or basal diet with chlortetracycline (CTC) alone or AEO at 250, 500, and 1,000 mg/kg (referred to as groups AEO1, AEO2, and AEO3, respectively) for 35 days, preceded by a 7-day pre-feed period. Introduction Sleep quality is increasingly acknowledged as critical for immune function and overall health. Poor sleep can disrupt immune response, elevating inflammation and susceptibility to infection and chronic diseases. Given the prevalence of sleep disturbances across populations, understanding their impact on immune health and overall wellness is essential. Purpose of Work This review aims to examine the relationship between sleep quality and immune function by synthesizing findings on how poor sleep contributes to immune suppression, heightened inflammation, and mental health disturbances, particularly in vulnerable populations. This review also explores possible interventions to mitigate these effects. Material and Methods A literature search was conducted on databases including PubMed and Google Scholar, focusing on studies from 2012 to 2023. Articles assessing sleep quality, immune markers, and the mediating roles of stress and depression were reviewed. Criteria included studies with both subjective and objective assessments of sleep, immune function, and relevant mental health markers. Summary Findings indicate that poor sleep quality negatively impacts immune health, primarily through increased systemic inflammation and decreased immune cell functionality. Depression and stress further exacerbate these effects, particularly in populations with chronic illnesses or high psychological distress. Targeted interventions that improve sleep quality and manage stress may bolster immune resilience and overall health. Future research should continue to address these interactions across diverse populations to develop targeted interventions.

Selenium is an important micronutrient that plays key functions in antioxidant defense, immune function, and thyroid health. However, its benefits and risks are closely linked to selenium methylation, a process that significantly influences its biological activity and potential toxicity. This review critically analyse the role of selenium methylation in modulating selenium’s effects on health, highlighting its impact on both protective and adverse outcomes. it also discusses the current understanding of selenium’s essential functions and toxicity risks, emphasizing the complexities of Se methylation. The review addresses notable research gaps and controversies, including inconsistent findings on selenium’s cancer-preventive properties, the diverse biological activities of different selenium species, and the challenges in determining optimal selenium intake levels. By synthesizing existing knowledge and identifying key areas for further investigation, this review aims to advance readers’ understanding of selenium’s multifaceted roles that guide future research and public health strategies.

Quercetin (QRT), a potent flavonoid, holds immense mechanistic potential in enhancing various aspects of broiler chicken physiological status. This comprehensive study explores the profound еffеcts of QRT on behavior, productive performance, and immune functions, unraveling the underlying mechanisms behind its efficacy. Four hundred, one-day-old Cobb 500 chicks were placed randomly into 4 supplementation groups (100 birds /group and five replicates) and provided diets enriched with varying concentrations of QRT (0, 200, 400, and 600 ppm) for six weeks. Visual scanning revealed significant (P > 0.05) improvements in feeding and body care behaviors, accompanied by reduced instances of idleness and walking in the QRT-supplemented groups. Moreover, QRT supplementation exerted a substantial (P > 0.05) positive influence on weight gain, feed intake and the final body weight of the broilers. In-depth evaluation of immune parameters, QRT supplementation elicited significant (P > 0.05) enhancements in immune functions, including improved spleen, thymus, and bursa indices, enhanced secretion of immunoglobulin M (IgM) and immunoglobulin A (IgA), and stimulated humoral immunity against sheep red blood cells (SRBCs). Furthermore, QRT displays potent antioxidant properties, as showed by diminished splenic malondialdehyde levels and augmented activity of antioxidative stress enzymes. Remarkably, QRT supplementation elicited dose-dependent upregulation (P < 0.001) of key immune-related genes, such as intеrlеukin-4 (IL-4), interferon-γ (INF-γ), Toll-like receptor 2 (TLR2), and tumor necrosis factor-α (TNF-α) in the splenic tissue. Collectively, these mechanistic insights underscore the profound impact of QRT as a functional feed additive, fostering enhanced behavior, performance, and immune function in broiler chickens, while delivering robust antioxidant fortification.

The International Rome Committee defines Disorders of Gut-Brain Interactions (DGBI) based upon distinct combinations of chronic and/or recurrent unexplained gastrointestinal symptoms. Yet patients often experience overlapping DGBI. Patients with DGBI frequently also suffer from extraintestinal symptoms, including fatigue, sleep disturbances, anxiety, and depression. Patients with overlapping DGBI typically experience more severe GI symptoms and increased psychosocial burden. Concerning the pathophysiology, DGBI are associated with disruptions in gut motility, function of the brain and enteric neurons, immune function, and genetic markers, with recent findings revealing gut microbiome alterations linked to these mechanisms of DGBI. Emerging evidence summarized in this review suggests that the microbiome influences various established disease mechanisms of different DGBI groups. Overall, changes in the gastrointestinal microbiome do not seem to be linked to a specific DGBI subgroup but may play a key role in the manifestation of different DGBI and, subsequently, overlap of DGBI. Understanding these shared mechanisms and the role of the gastrointestinal microbiome, particularly for overlapping DGBI, might aid in developing more precise diagnostic criteria and treatment strategies while developing personalized interventions that target specific mechanisms to improve patient outcomes.

Hepatocellular carcinoma is the predominant histologic variant of hepatic malignancy and has become a major challenge to global health. The increasing incidence and mortality of hepatocellular carcinoma has created an urgent need for effective prevention, diagnosis, and treatment strategies. This is despite the impressive results of multiple treatments in the clinic. However, the unique tumor immunosuppressive microenvironment of hepatocellular carcinoma increases the difficulty of treatment and immune tolerance. In recent years, the application of nanoparticles in the treatment of hepatocellular carcinoma has brought new hope for tumor patients. Nano agents target tumor-associated fibroblasts, regulatory T cells, myeloid suppressor cells, tumor-associated macrophages, tumor-associated neutrophils, and immature dendritic cells, reversed the immunosuppressive microenvironment of hepatocellular carcinoma. In addition, he purpose of this review is to summarize the advantages of nanotechnology in guiding surgical excision, local ablation, TACE, standard chemotherapy, and immunotherapy, application of nano-vaccines has also continuously enriched the treatment of liver cancer. This study aims to investigate the potential applications of nanotechnology in the management of hepatocellular carcinoma, with the ultimate goal of enhancing therapeutic outcomes and improving the prognosis for patients affected by this malignancy.

The maintenance of host health and immune function is heavily dependent on the gut microbiota. However, the precise contribution of individual microbial taxa to regulating the overall functionality of the gut microbiome remains inadequately investigated. Chickens are commonly used as models for studying poultry gut microbiota, with high-throughput 16S rRNA sequencing has emerged as a valuable tool for assessing both its composition and functionality. The interactions between the gut’s microbial community and its host significantly influence health outcomes, disease susceptibility, and various mechanisms affecting gastrointestinal function. Despite substantial research efforts, the dynamic nature of this microbial ecosystem has led to inconsistencies in findings related to chicken gut microbiota, which is largely attributed to variations in rearing conditions. Consequently, the interaction between the chickens’ gut microflora and its host remains inadequately explored. This review highlights recent advances in understanding these relationships, with a specific focus on microbial composition, diversity, functional mechanisms, and their potential implications for improving poultry production.

The aim of this research was to investigate the effects of adding moringa polysaccharides (MOP) on the growth performance, immune function, rumen tissue morphology, and rumen microbial community in early-weaned goat kids. Twenty-one 7-day-old Leizhou male goat kids weighing (3.05 ± 0.63) kg, were randomly divided into a control group (CON group), a low-dose group (LOW group), and a high-dose group (HIG group). MOP was added to the goat kids’ milk replacer (MR) at 0, 0.15, and 0.3% (on dry matter basis),fed until 60 days of age, and four goat kids in each group with body weights close to the mean of each group were selected for slaughter. The results showed that, compared to the CON group, the MOP groups significantly improved final body weight, body measurements, daily weight gain, and feed intake of the early weaned goat kids; significantly reduced the content of propionic acid, butyric acid, valeric acid, and ammoniacal nitrogen; and in addition, the addition of MOP could significantly increase the height of rumen nipple, the content of immunoglobulin G (IgG) in the serum. The HIG group significantly increased rumen pH, rumen muscularis layer thickness, rumen wall thickness, and serum immunoglobulin A (IgA), and immunoglobulin M (IgM). In conclusion, the addition of MOP positively impacted the growth performance, serum immune function, and rumen tissue morphology in early-weaned goat kids.

Abstract Background Minimal access breast surgery (MABS) is commonly employed in the management of breast cancer, but there is limited research on the postoperative immune function associated with MABS. Objective This study aimed to assess the postoperative immune function in breast patients who underwent MABS or conventional open breast surgery (COBS). Methods We retrospectively analyzed the medical records of 829 breast cancer patients treated with either MABS or COBS at a single hospital between January 2020 and June 2023. Among them, 116 matched pairs were obtained through 1:1 propensity score matching (PSM). Flow cytometry was used to measure the percentages of CD3+, CD4+, and CD8+ cells, as well as the CD4+/CD8+ ratio, on three different time points: preoperative day 1 (PreD1), postoperative day 1 (PostD1), and postoperative day 7 (PostD7). Results Both the MABS and COBS groups demonstrated a significant reduction in the percentages of CD3+, CD4+, and CD8+ cells, along with the CD4+/CD8+ ratio, from PreD1 to PostD1. Interestingly, the MABS group showed a reversal of these parameters, returning to preoperative levels by PostD7. Conversely, the COBS group showed an increase in these parameters from PostD1 to PostD7, but they still remained significantly lower than preoperative levels at PostD7. Conclusion MABS treatment may result in reduced postoperative immune suppression and faster recovery of preoperative immune function compared to COBS in patients.

Objective To investigate the effect of stereotactic minimally invasive surgery on immune level and activities of daily living in elderly patients with glioma. Methods Ninety elderly patients with glioma admitted to the First Affiliated Hospital of Harbin Medical University from March 2020 to February 2022 were selected as the study subjects and divided into two groups according to the random number table, 45 cases for each group. The control group was treated with conventional craniotomy and the observation group was treated with stereotactic minimally invasive surgery. The clinical efficacy, activities of daily living (ADL) score, National Institutes of Health Stroke Scale (NIHSS) score, peripheral blood immune parameters T helper (Th) 1, Th2 and Th17 levels were compared between the two groups. Results The overall response rate in the observation group was higher than that in the control group. The recurrence rate at 1 year after surgery was lower than that in the control group. After treatment, the ADL score in the observation group was higher than that in the control group, the NIHSS score was lower than that in the control group, the Th1 level was higher than that in the control group, and the Th2 and Th17 levels were lower than those in the control group. Abstract Background The research aims to explore the characteristics of intestinal flora, nutritional status and immune function in patients with different types of obese colon cancer. Methods A retrospective analysis is conducted on 64 cases of obese colon cancer diagnosed from June 2018 to January 2020. According to the histological staging of the cancer, they are classified into adenocarcinoma, adenosquamous carcinoma and undifferentiated carcinoma, with corresponding cases of 24, 22 and 18, respectively. The intestinal flora (Bifidobacterium, Lactobacillus, Enterococcus faecalis, Escherichia coli, and yeast), nutritional status (Hb, Alb, PA, TFN, and PNI), immune function (IgG, IgM, IgA, CD4 +, CD8 +, and CD4 +/CD8 +) are analyzed in the different groups of patients. Survival curves are evaluated by Kaplan-Meier method and log-rank test for tumour death, local recurrence, and distant metastasis. Results There were no statistically significant differences in intestinal flora (Bifidobacterium, Lactobacillus, Enterococcus faecalis, Escherichia coli, and yeast), nutritional status (Hb, Alb, PA, TFN, and PNI) and immune function (IgG, IgM, IgA, CD4 +, CD8 +, and CD4 +/CD8 +) between different groups. There was a significant correlation between intestinal flora, nutritional status and immune function for all three. The survival curves of tumour death, local recurrence and distant metastasis in different groups of obese colon cancer patients were statistically significant. The tumor mortality rate, local recurrence, and distant metastasis rate in adenocarcinoma were 78.65%, 54.25% and 48.26% respectively. Conclusion There are differences in intestinal flora, nutritional status and immune function among different types of obese colon cancer patients, but adenocarcinoma has the least benefit in intestinal flora, poor nutritional status, and weakest immune function.

Abstract Background Laparoscopic partial hepatectomy inevitably decrease patient immune function. Propofol has been shown to have immunomodulatory effects but is associated with hemodynamic side effects. Despite studies showing a negligible impact of remimazolam tosylate on hemodynamics, it has not been reported for partial hepatectomy patients. Its influence on immune function also remains unexplored. This study sought to investigate the differences in immune function and intraoperative hemodynamics between patients who underwent laparoscopic partial hepatectomy with remimazolam tosylate and those who underwent laparoscopic partial hepatectomy with propofol. Methods This was a single-center, randomized controlled trial involving 70 patients, who underwent elective laparoscopic partial hepatectomy. The patients were randomly divided into two groups: the remimazolam group (group R) and the propofol group (group P). In this study, the primary outcomes assessed included the patient’s immune function and hemodynamic parameters, and the secondary outcomes encompassed the patient’s liver function and adverse events. Results Data from 64 patients (group R, n = 31; group P, n = 33) were analyzed. The differences in the percentages of CD3+, CD4+, CD8+, and NK cells and the CD4+/CD8+ ratio between the two groups were not statistically significant at 1 day or 3 days after surgery. Compared with those in group P, the MAP and HR at T2 and the MAP at T1 in group R were significantly increased(P 0.05). Conclusion The effects of remimazolam tosylate on the immune function of patients after partial hepatectomy are comparable to those of propofol. Additionally, its minimal effect on hemodynamics significantly decreases the incidence of hypotension during anesthesia induction, thereby enhancing overall perioperative safety. Trial registration The trial was registered on May 9, 2022 in the Chinese Clinical Trial Registry, registration number ChiCTR2200059715 (09/05/2022).

This study aimed to investigate the impact of adequate energy supply on these indicators in COPD patients.Methods: COPD patients meeting specific criteria were recruited and categorized into energy-adequate and energy-deficient groups based on their energy supply. Comparable demographic factors such as age, gender, smoking and drinking history, COPD duration, inhaled drug classification, and home oxygen therapy application were observed. Notable differences were found in BMI and inhaled drug use between the two groups.Results: The energy-adequate group exhibited significant improvements in various health indicators, including lymphocyte count, hemoglobin, CRP, total cholesterol, prealbumin, albumin, PNI, SII, SIRI, CAR, and CONUT scores in the secondary auxiliary examination. These positive changes suggest a notable enhancement in nutritional, immune, and inflammatory status.Conclusion: This research highlights the substantial benefits of adequate energy supply in elderly COPD patients. The observed improvements in nutritional, immune, and inflammatory markers underscore the importance of addressing energy needs to positively influence disease-related outcomes in this population. Abstract Eucommia ulmoides is a traditional Chinese herbal medicine, with pharmacological effects such as lowering blood pressure and enhancing immune function. The effects of dietary Eucommia ulmoides polysaccharide (EUP) on immune function and meat quality were studied in Songliao Black Pigs. Blood lymphocyte counts and percentage, concentrations of serum total protein and of albumin increased, whereas those of urea nitrogen and triglyceride decreased. White blood cell and lymphocyte counts, and serum IgA, IgE, IgG2 a and IFN-γ increased. Average daily weight gain, slaughter weight, lean meat rate and cooked meat rate increased, whereas pH24, feed-weight ratio, fat rate, yellowness (b#) and centrifugal dehydration rate decreased. Transcriptome sequencing of longissimus dorsi muscle detected 32 differentially expressed genes (DEGs), of which 26 were up-regulated and 6 down-regulated. A total of 19 genes were differentially expressed in the four groups, 18 of which were up-regulated. The DEGs included ADAMTS4, PER1, STAC, SERPINE1, FASN, THRSP, SP7 and KRT80 and the protein interaction network showed 20 up-regulated nodes, three down-regulated nodes and 14 DEGs. GO functional annotation and enrichment analysis showed that 34 items were significantly enriched, including transferase activity, actin binding, acetyl coenzyme A, acyl coenzyme A metabolism, adipose tissue development and acyl glycerol homeostasis. KEGG pathway analysis showed that the AMPK and PPAR signaling pathways were enriched. Dietary Eucommia polysaccharide enhanced immune function in Songliao Black Pigs, improved growth and carcass performance, increased the expression of genes related to meat quality traits and improved meat quality.

Abstract Background The alteration of the immune microenvironment in the axillary metastatic lymph nodes of luminal A breast cancer patients is still unclear. Methods Postsurgical tissues from the enrolled luminal A BCs were divided into five categories: primary BC lesion at stage N0 (PL1), primary BC lesion at stage N1 (PL2), negative axillary lymph node at stage N0 BC (LN1), negative axillary lymph node at stage N1 BC (LN2), and positive axillary lymph node at stage N1 BC (LN3). The frequencies of positive immune markers (CD4, CD8, PD1, PD-L1, T-cell immunoglobulin and mucin domain 3 (TIM3), and forkhead box protein 3 (Foxp3)) in the above tissues were quantified by AKOYA Opal Polaris 7 Color Manual IHC Detection Kit. Results A total of 50 female patients with luminal A BC were enrolled in this study. Among these patients, 23 had stage N1 disease, and 27 had stage N0 disease. Background. Exercise-induced muscle damage (EIMD) is a temporary response to intense or prolonged exercise that can cause muscle pain, inflammation, and impaired muscle function. Antioxidant supplementation is a proposed strategy to reduce EIMD symptoms by targeting reactive oxygen and nitrogen species (RONS) involved in the process. Objective. The study aimed to investigate the effect of Selenomethionine supplementation on malondialdehyde (MDA) and C-reactive protein (CRP) levels resulting from Exercise-Induced Muscle Damage (EIMD). Materials and methods. This study used a randomized pretest-posttest control group design, involving a total of 32 male recreational students from the State University of Surabaya (Indonesian: Universitas Negeri Surabaya), (age 19.25 ± 0.76 years, body mass 63.16 ± 3.38 kg, height 167.28 ± 4.54 cm, body fat 19.60% ± 4.57%). The participants were randomly assigned to the selenomethionine group (SEM, 100 µg/day) or placebo group (PLA, corn starch 100 mg/day) for a period of 28 days (4 weeks). On days 1 (baseline) and 29, participants underwent a single bout of EIMD. Blood samples were collected 24 hours post-EIMD to measure MDA and CRP concentrations in plasma. The statistical analysis was conducted using paired sample t-test. Results. The placebo group experienced a significant increase in plasma MDA and CRP concentrations after EIMD compared with baseline values (p < 0.05). However, the SeMet group showed lower plasma MDA and CRP levels than the placebo group. Conclusions. Daily Selenomethionine supplementation for 28 days has been found to reduce oxidative stress by lowering MDA levels in the blood and to decrease inflammation by reducing CRP levels post-exercise-induced muscle damage. This indicates a lower risk of EIMD due to reduced oxidative stress and inflammation.

Abstract Introduction Metronomic capecitabine used as an adjuvant therapy improves survival in patients with locoregionally advanced nasopharyngeal carcinoma (LA-NPC). This therapeutic approach may also contribute to improving immune function, consequently enhancing overall therapeutic efficacy. Aim We aimed to evaluate the effect of metronomic capecitabine as adjuvant therapy on immune function and survival in cases of LA-NPC. Subjects and methods 28 patients with LA-NPC were enrolled in the study and equally assigned to two groups of 14 each: experimental and control group. The experimental group received induction chemotherapy + concurrent chemotherapy + adjuvant chemotherapy as well as oral capecitabine at a dose of 650 mg/m² of body surface area twice daily for 1 year, with the option to discontinue in case of intolerance. The control group did not receive additional chemotherapy or targeted drugs after the induction chemotherapy + concurrent chemoradiotherapy; however, they were followed up regularly. Changes in immune function and survival were compared between the two groups. Results The median follow-up time was 43.5 months. One year after adjuvant chemotherapy, the experimental group showed higher levels of CD8 + cells, CD28 + CD8 + cells, and activated CD8 + cells compared to the control group (P 0 0.05 ). Conclusion Metronomic capecitabine chemotherapy was observed to induce an immunomodulatory effect in LA-NPC. Trial registration NCT02958111, date of registration 04-11-2016.

Abstract Background Low immune function after laparoscopic total gastrectomy puts patients at risk of infection-related complications. Low-dose naloxone (LDN) can improve the prognosis of patients suffering from chronic inflammatory diseases or autoimmune diseases. The use of LDN during perioperative procedures may reduce perioperative complications. The purpose of this study was to examine the effects of LDN on endogenous immune function in gastric cancer patients and its specific mechanisms through a randomized controlled trial. Methods Fifty-five patients who underwent laparoscopic-assisted total gastrectomy were randomly assigned to either a naloxone group (n = 23) or a nonnaloxone group (n = 22). Patients in the naloxone group received 0.05 µg/kg-1.h− 1naloxone from 3 days before surgery to 5 days after surgery via a patient-controlled intravenous injection (PCIA) pump, and patients in the nonnaloxone group did not receive special treatment. The primary outcomes were the rates of postoperative complications and immune function assessed by NK cell, CD3+ T cell, CD4+ T cell, CD8+ T cell, WBC count, neutrophil percentage, and IL-6 and calcitonin levels. The secondary outcomes were the expression levels of TLR4 (Toll-like receptor), IL-6 and TNF-α in gastric cancer tissue. When piglets are exposed to pathogens for a long period, the immune system organs, among them the spleen, play a major role in combating the stress caused by those pathogens. In the present study, the effect on splenocyte function was investigated in a model of weaned piglets in which stress was induced by multiple low doses of lipopolysaccharide (LPS).

【Objective】The experiment was carried out to study the effects of potassium magnesium sulfate (PMS) on sow performance, plasma biochemical indicators, antioxidant capacity and immune function in late pregnancy.【Method】By using simple factor design of the experiment, sixty sows (Landrace×Yorkshire) with 3-4 parity, good health and similar expected delivery in the third trimester of pregnancy (85th day of pregnancy) were randomly divided into 4 groups (15 replicates in each group and 1 sow in each replicate). The experimental groups during gestation were supplemented with PMS 0 (CK), 5, 10 and 15 g/d for each sow, respectively. After delivery, all sows during lactation were fed the same commercial lactation feed. The experiment ended on the 18th day of lactation.【Result】(1) Supplemental PMS had no significant effects on litter size, healthy litter size, back fat thickness, labor, body weight, feed intake in sows during lactation, and newborn weight of piglets, number of weaned piglets and number of deaths of lactating piglets. Compared with the control group, the fecal score of sows during late pregnancy after PMS supplementation showed a linear increase (Plinear < 0.05) and 10 g/d group had significant effects (PANOVA < 0.05); After supplementation with PMS, it showed a linear increase (Plinear < 0.05) in newborn litter weight, litter weight of weaned piglets, average litter gain of lactating piglets and daily gain of lactating piglets (PANOVA < 0.05). Compared with the control group, the newborn litter weight, litter weight of weaned piglets, average litter gain of lactating piglets and daily gain of lactating piglets in the 10 g/d group were increased by 16.27%, 24.42%, 35.14% and 31.64%. Except for newborn litter weight, all other indicators showed quadratic effect (PQuadratic < 0.05) and significant differences were found between groups (PANOVA < 0.05); (2) It showed a linear increase in the contents of triglyceride (TG), cholesterol (CHO), low-density lipoprotein (LDL) and insulin (INS) in plasma of sows supplemented with PMS (Plinear < 0.05). The contents of TG, CHO, LDL and INS in the 10 g/d group increased by 50.00%, 28.35%, 25.00% and 31.98% compared with the control group; TG and LDL had a quadratic effect (PQuadratic < 0.05). (3) Linear changes were found in contents of immunoglobulin G (IgG), immunoglobulin A (IgA), immunoglobulin M (IgM) and glutathione peroxidase (GSH-Px) (Plinear < 0.05) in the plasma of sows after supplementation with PMS, and they were increased by 17.10%, 20.34%, 8.00% and 15.04% in the 10 g/d group, compared with the control group. (4) It showed a linear increase (PANOVA < 0.05) in fat content of colostrum of sow with PMS supplementation (Plinear < 0.05), with significant increase. Non-fat milk solids, lactose and protein were linearly increased (Plinear < 0.05) and had an increasing trend (0.05 < PANOVA < 0.1). The fat content of colostrum supplemented with 10 g/d were increased by 51.42% compared with the control group.【Conclusion】PMS supplementation in late preganancy can significantly improve the fecal scores of sows, relieve sow constipation, and improve sow performance and body health, with 10 g/d supplementation as the optimal dose.

With trends towards low fishmeal and higher plant ingredient-based diets for shrimp, there is consequently greater demand on feed additives to bolster the protein and nutrient profile quality for achieving more efficiency. Also, there is a need to enhance innate immunity and resilience to infection under intensive shrimp production scenarios. This investigation tested incremental levels 0, 0.2; 0.4; 0.6 and 0.8 % of a commercial yeast cell wall extract mainly comprising β-glucan and mannan oligosaccharides (MOS) (YM, YeaMOS, Angel Yeast, Yichang, Hubei, China) for post-larval shrimp Penaeus vannamei in isonitrogenous and iso-lipidic diets with proteinaceous plant ingredients amounting to 50 % contribution of the diet formulation. After 60 d, growth and feed utilization performance were not significantly affected in shrimp in terms of weight gain or FCR, but due to much higher survival, total biomass in groups of shrimps fed with dietary YM was higher compared to the control treatment. Background and aims: Whole grain consumption is widely recognized as a vital component of a balanced diet. Dietary fiber has been well-documented to play a crucial role in these health benefits attributed to whole grain intake. However, population-based evidence directly linking whole grain consumption to anti-inflammatory effects, especially in the context of immune-mediated inflammation, remains limited. We hypothesized that whole grain consumption promotes health by modulating immune-mediated inflammation. Methods and results: This study was designed as a real-world, population-based randomized controlled trial. We compared the effects of whole grain versus refined grain consumption on immune-mediated inflammation through staple food substitution, while participants maintained their usual dietary practices. The results demonstrated that whole grain consumption significantly reduced circulating levels of pro-inflammatory cytokines IL-22 and IL-23 compared to refined grain consumption. These reductions were associated with optimized short-chain fatty acid profiles and changes in CD4+ T cell subset distributions. Conclusions: The findings suggest that the anti-inflammatory effects of whole grain consumption in middle-aged and elderly populations are mediated by targeting specific CD4+ T cell subsets, in addition to modulating both upstream short-chain fatty acid composition and downstream expression of the pro-inflammatory cytokines IL-22 and IL-23.

Yeast peptides, which are small-molecule active peptides extracted from yeast proteins, are known for their antibacterial, antioxidant, and anti-inflammatory properties. However, the effects of yeast peptide on suckling calves remain unclear. In this study, the effects of yeast peptide supplementation on the growth, diarrhea incidence, and immune function of calves during the suckling period were determined. Thirty newborn calves were randomly divided into two groups: the control group (CON) and the treatment group (AP), which received fresh pasteurized milk supplemented with yeast peptides (5 g/day). The experiment lasted for 49 days (7–56 days of age). The dry matter intake, body weight, diarrhea status, immune function, antioxidant capacity, and anti-inflammatory activity of the calves were analyzed. The AP group had higher dry matter intake, daily weight gain, and feed efficiency than the CON group (P < 0.05). Moreover, the duration and frequency of diarrhea were significantly lower in the AP group than in the CON group (P < 0.05). Furthermore, the immune, antioxidant, and anti-inflammatory capabilities of the AP group were significantly higher than those of the CON group (P < 0.05). These findings provide valuable insights for the improvement of early health management during calf rearing.

In this study, Lagenaria siceraria (Molina) Standl polysaccharides (LSP) was prepared using the water-alcohol precipitation method to evaluate its effects on growth performance, slaughter performance, cytokines, immune organ indices, cecal short-chain fatty acids (SCFAs), and microbial community structure in broiler chickens when added to the basal diet. Seventy-five broiler chickens were selected and randomly divided into five groups, with 15 chickens per group. All groups were fed a basal diet for 7 days. From 7 days of age, the control group continued to receive the basal diet, while the positive drug group was fed a diet supplemented with Astragalus polysaccharides (APS, 100 g/kg) in addition to the basal diet. The experimental groups were fed diets containing different concentrations of LSP (50, 100, and 200 g/kg) in addition to the basal diet, and the supplementation continued for 42 days. Fungal probiotics have the potential as feed additives, but less has been explored in ruminant feed up to date. This study aimed to determine the effect of compound probiotics (CPs) with Aspergillus oryzae 1, Aspergillus oryzae 2 and Candida utilis on Hu sheep’s growth performance, rumen fermentation and microbiota. A total of 120 male Hu sheep, aged 2 months and with the body weight of 16.95 ± 0.65 kg were divided into 4 groups. Each group consisted of 5 replicates, with 6 sheep per replicate. Group A was the control group fed with the basal diet. Group B, C and D was supplemented with the basal diet by adding 400, 800 and 1,200 grams per ton (g/t) CPs, respectively. The feeding trial lasted for 60 days after a 10-day adaptation period. The results showed that the average daily gain (ADG) of sheep in the CPs groups were significantly higher, the feed/gain were significantly lower than those in group A in the later stage and the overall period. The addition of CPs increased the economic benefit. The levels of CD4+ and the CD4+/CD8+ ratio in the CPs groups were higher than those in Group A. The levels of GSH, IgG, IL-2, IL-6, and IFN-γ in group C were significantly elevated compared with group A. Group B showed a significant increase in rumen NH3-N and cellulase activity. There was no difference in VFAs content between group A and group B, however, with the increasing addition of CPs, the butyric acid and isobutyric acid content tended to decrease. The rumen microbiota analysis indicated that the CPs addition increased the Firmicutes and Proteobacteria abundances, decreased the Bacteroidetes abundance. The correlation analysis showed that Prevotella was negatively correlated with ADG, and the addition of 400 CPs in group B reduced Prevotella’s relative abundance, indicating CPs increased sheep growth by decreasing Prevotella abundance. The CPs addition reduced caspase-3, NF-κB and TNF-α expression in liver, jejunum and rumen tissues. In conclusion, the addition of CPs increased the sheep production performance, reduced inflammation, improved rumen and intestinal health. Considering the above points and economic benefits, the optimal addition of CPs as an additive for Hu sheep is 800 g/t.

Abstract (301 words): Introduction: The effects of low energy availability (LEA) on the immune system are poorly understood. This study examined the effects of 14 days of LEA on immune cell redox balance and inflammation at rest and in response to acute exercise, and exercise performance in female athletes. Methods: Twelve female endurance athletes (age: 26.8 ± 3.4 yrs, maximum oxygen uptake (V˙O2max): 55.2 ± 5.1 mL × min−1 × kg−1) were included in a randomized, single-blinded crossover study. They were allocated to begin with either 14 days of optimal energy availability diet (OEA, 52 ± 2 kcal × kg fat free mass (FFM)−1 × day−1) or LEA diet (22 ± 2 kcal × kg FFM−1 × day−1), followed by 3 days of refueling (OEA) with maintained training volume. Peripheral blood mononuclear cells (PBMCs) were isolated, and plasma obtained at rest before and after each dietary period. The PBMCs were used for analysis of mitochondrial respiration and H2O2 emission and specific proteins. Exercise performance was assessed on cycle by a 20-min time trial and time to exhaustion at an intensity corresponding to ∼110 % V˙O2max). Results: LEA was associated with a 94 % (P = 0.003) increase in PBMC NADPH oxidase 2 protein content, and a 22 % (P = 0.013) increase in systemic cortisol. This review comprehensively examines the impact of anesthesia and surgical interventions on the immune function of cancer patients postoperatively. Recent studies have shown that surgery and its accompanying anesthesia management can significantly influence immune function in cancer patients, potentially affecting their prognosis. This review synthesizes clinical studies and basic research to summarize the specific effects of anesthesia methods, drugs, postoperative analgesia, intraoperative transfusion, surgical techniques, and trauma extent on the immune function of cancer patients post-surgery. Additionally, this review discusses optimization strategies based on current research, aiming to refine anesthesia and surgical management to maximize the preservation and enhancement of postoperative immune function in cancer patients, with the potential to improve clinical outcomes.

Objective: This research utilized network pharmacology to investigate the potential of Fuzheng Qudu prescription (FZQDP) in treating lung cancer (LC). Methods: The components and their targets of FZQDP were analyzed for their relationship with LC-related targets using bioinformatics tools. Mouse Lewis lung carcinoma (LLC) cells were cultured in vitro and treated with FZQDP or cisplatin (DDP) before applying the MTT assay to determine FZQDP concentrations, and the IC50 value. According to the IC50 value, the effect of FZQDP on apoptosis and cell cycle was detected by flow cytometry. Mouse tumor growth was recorded using live animal imaging, and measurements of tumor and spleen weight were used to calculate the tumor inhibition rate and spleen index. The effects on mouse liver and kidneys were observed by analyzing levels of AST, ALT, BUN, and CRE in blood and hematoxylin and eosin (H & E) stained sections. Additionally, levels of IL-2, IL-10, IL-6, and IFN-γ in serum, along with the frequencies of CD4+ and CD8+ T cells in the spleen, were measured using Mouse multiple Cytokine Assay and flow cytometry, respectively. Results: SRC, STAT3, MAPK3, and MAPK1 could be crucial targets of FZQDP in the treatment of LC. FZQDP demonstrated inhibition of LC cell proliferation and tumor growth, as well as enhancement of apoptosis and induction of G2 phase cell cycle arrest. Furthermore, FZQDP led to elevated levels of IL-2 and IFN-γ, increased frequencies of CD4+ T cells and decreased levels of IL-6 and IL-10. Importantly, FZQDP did not exhibit any noticeable hepatotoxic or nephrotoxic effects in mice. Conclusion: FZQDP may target multiple signaling pathways to treat LC. In a LC mouse model, FZQDP was found to inhibit tumor growth and improve immune function.

ABSTRACT: Mycoplasma gallisepticum (MG) can cause chronic respiratory disease (CRD) in chickens, which has a significant negative economic impact on the global poultry sector. Respiratory flora is the guardian of respiratory health, and its disorder is closely related to respiratory immunity and respiratory diseases. As a common probiotic in the chicken respiratory tract, Lactobacillus salivarius (L. salivarius) has potential antioxidant, growth performance enhancing, and anti-immunosuppressive properties. However, the specific mechanism through which L. salivarius protects against MG infection has not yet been thoroughly examined. This study intends to investigate whether L. salivarius could reduce MG-induced tracheal inflammation by modulating the respiratory microbiota and metabolites. The results indicated that L. salivarius reduced MG colonization significantly and alleviated the anomalous morphological changes by using the MG-infection model. L. salivarius also reduced the level of Th1 cell cytokines, increased the level of Th2 cell cytokines, and ameliorated immune imbalance during MG infection. In addition, L. salivarius improved the mucosal barrier, heightened immune function, and suppressed the Janus kinase/Signal transducer, and activator of transcription (JAK/STAT) signaling pathway. Notably, MG infection changed the composition of the respiratory microbiota and metabolites, and L. salivarius therapy partially reversed the aberrant respiratory microbiota and metabolite composition. Our results highlighted that these findings demonstrated that L. salivarius played a role in MG-mediated inflammatory damage and demonstrated that L. salivarius, by altering the respiratory microbiota and metabolites, could successfully prevent MG-induced inflammatory injury in chicken trachea.
