Different from conventional ferromagnetic methods, producing a spin-polarized current through electrical ways in spintronic devices can greatly increase operating speed, reduce power consumption, and improve device integration. Inspired by recent experimental progress on the synthesis of a heptauthrene molecule, we investigate its spin-dependent transport contacted with Au electrodes through first-principles calculations. By applying a gate voltage, the transmission can be switched between completely spin-up and spin-down polarized states, achieving an electrically controlled dual-spin filter. Furthermore, a fine tuning on the spin polarization, between 100% and − 100%, can also be realized, where the transport with any ratio of spin-up to spin-down electron quantities can be realized beyond the traditional devices. The peculiar transmission spectra and their shift are found to play crucial roles, where transmission peaks distribute on both sides of the Fermi level with opposite spin components. Such a spin-polarization modulating effect is found to be robust to the molecule-electrode contacting site, indicating it is an intrinsic feature of such systems. Importance: Intra-arterial therapies(IATs) are promising options for unresectable hepatocellular carcinoma(HCC). Stratifying the prognostic risk before administering IAT is important for clinical decision-making and for designing future clinical trials. Objective: To develop and validate a machine learning(ML)-based decision support model(MLDSM) for recommending IAT modalities for unresectable HCC. Design, setting, and participants: Between October 2014 and October 2022, a total of 2,959 patients with HCC who underwent initial IATs were enroled retrospectively from 13 tertiary hospitals. These patients were divided into the training cohort (n = 1700), validation cohort (n = 428), and test cohort (n = 200). Main outcomes and measures: Thirty-two clinical variables were input, and five supervised ML algorithms, including eXtreme Gradient Boosting (XGBoost), Categorical Gradient Boosting (CatBoost), Gradient Boosting Decision Tree (GBDT), Light Gradient Boosting Machine (LGBM) and Random Forest (RF), were compared using the areas under the receiver operating characteristic curve (AUC) with the DeLong test. Results: A total of 1856 patients were assigned to the IAT alone Group(I-A), and 1103 patients were assigned to the IAT combination Group(I-C). The 12-month death rates were 31.9% (352/1103) in the I-A group and 50.4% (936/1856) in the I-C group. For the test cohort, in the I-C group, the CatBoost model achieved the best discrimination when 30 variables were input, with an AUC of 0.776 (95% confidence intervals [CI], 0.833–0.868). In the I-A group, the LGBM model achieved the best discrimination when 24 variables were input, with an AUC of 0.776 (95% CI, 0.833–0.868). According to the decision trees, BCLC grade, local therapy, and diameter as top three variables were used to guide clinical decisions between IAT modalities. An oxidant-controlled divergent synthesis of a pyrrolidone fused pyrimido[1,2-b]indazole skeleton was developed through selective cyclization of an in situ generated enone intermediate and 1H-indazol-3-amine. The one-pot, metal-free process formed three C–N bonds, one C–C bond, and a tetrasubstituted carbon stereocenter containing a hydroxyl group. This method not only allowed for the synthesis of over 60 new pyrrolidone fused pyrimido[1,2-b]indazole derivatives, but was also compatible with the transformation of complex active molecules and the derivation of target products. Delay/disruption tolerant networking (DTN) is proposed as a networking architecture to overcome challenging space communication characteristics for reliable data transmission service in presence of long propagation delays and/or lengthy link disruptions. Bundle protocol (BP) and Licklider Transmission Protocol (LTP) are the main key technologies for DTN. LTP red transmission offers a reliable transmission mechanism for space networks. One of the key metrics used to measure the performance of LTP in space applications is the end-to-end data delivery delay, which is influenced by factors such as the quality of spatial channels and the size of cross-layer packets. In this paper, an end-to-end reliable data delivery delay model of LTP red transmission is proposed using a roulette wheel algorithm, and the roulette wheel algorithm is more in line with the typical random characteristics in space networks. The proposed models are validated through real data transmission experiments on a semi-physical testing platform. Furthermore, the impact of cross-layer packet size on the performance of LTP reliable transmission is analyzed, with a focus on bundle size, block size, and segment size. Comprehensive Summary: A [2 + 1 + 1 + 1] cyclization reaction has been developed for the synthesis of multisubstituted β‐pyrrolidinones from commercially available aryl methyl ketones, primary amines, and ethyl nitroacetate. In this I2–DMSO‐meditated process, the C—NO2 bond of ethyl nitroacetate is cleaved, affording a C1 synthon, and the formation of two C—C and two C—N bonds and a quaternary carbon center are constructed in one pot. Background: The long-term survival benefit of immune checkpoint inhibitors (ICIs) in neoadjuvant and adjuvant settings is unclear for colorectal cancers (CRC) and gastric cancers (GC) with deficiency of mismatch repair (dMMR) or microsatellite instability-high (MSI-H). Methods: This retrospective study enrolled patients with dMMR/MSI-H CRC and GC who received at least one dose of neoadjuvant ICIs (neoadjuvant cohort, NAC) or adjuvant ICIs (adjuvant cohort, AC) at 17 centers in China. Patients with stage IV disease were also eligible if all tumor lesions were radically resectable. Results: In NAC (n = 124), objective response rates were 75.7% and 55.4%, respectively, in CRC and GC, and pathological complete response rates were 73.4% and 47.7%, respectively. The 3-year disease-free survival (DFS) and overall survival (OS) rates were 96% (95%CI 90–100%) and 100% for CRC (median follow-up [mFU] 29.4 months), respectively, and were 84% (72–96%) and 93% (85–100%) for GC (mFU 33.0 months), respectively. In AC (n = 48), the 3-year DFS and OS rates were 94% (84–100%) and 100% for CRC (mFU 35.5 months), respectively, and were 92% (82–100%) and 96% (88–100%) for GC (mFU 40.4 months), respectively. Among the seven patients with distant relapse, four received dual blockade of PD1 and CTLA4 combined with or without chemo- and targeted drugs, with three partial response and one progressive disease. Conclusion: With a relatively long follow-up, this study demonstrated that neoadjuvant and adjuvant ICIs might be both associated with promising DFS and OS in dMMR/MSI-H CRC and GC, which should be confirmed in further randomized clinical trials. The porcine reproductive and respiratory syndrome virus (PRRSV) is a highly significant infectious disease that poses a substantial threat to the global pig industry. In recent years, the NADC30-like strain has gradually emerged as prevalent in China, causing a profound impact on the country's pig farming industry. Therefore, it is important to conduct an in-depth study on the characteristics and gene functions of the NADC30-like strain. An infectious cDNA clone is an indispensable tool for investigating the functions of viral genes. In this current study, we successfully isolated a NADC30-like strain and constructed its full-length infectious cDNA clone. A growing body of evidence indicates that the G protein-coupled bile acid receptor, TGR5, plays a critical role in multiple physiological processes ranging from metabolic disorders to cancers. However, the biological functions of TGR5 in cervical cancer (CC) have not been elucidated. Here, using TGR5 knockout mice, we found that a deficiency of TGR5 leads to greater sensitivity to the progression of cervical inflammation. Activation of TGR5 by its specific ligands significantly attenuated the malignant behavior of CC cells. In addition, we found that TGR5 can negatively modulate the expression of lncRNA HCP5 by blocking its transcription activation when mediated by p65. HCP5 was highly expressed in CC tissues, which was positively correlated with the poor prognosis of CC patients. HCP5 knockdown notably restrained CC cell proliferation, colony formation, and migration in vitro, and inhibited tumor growth in vivo. Furthermore, HCP5 can function as the molecular sponge for miR-139-5p to upregulate DNA damage-induced transcript 4 (DDIT4) in CC cells. Murine xenograft studies demonstrated that TGR5 suppressed the tumor formation of CC cells and downregulated HCP5 and DDIT4 while increasing miR-139-5p in the xenografts. Taken together, these findings, for the first time, indicate that TGR5 inhibits CC progression by regulating the HCP5/miR-139-5p/DDIT4 axis, suggesting that it may represent a novel and potent target for CC treatment. Background: In this study, we comprehensively profiled the T‐cell receptor (TCR) repertoire of the tumor and adjacent normal tissue in patients with HBV‐associated hepatocellular carcinoma (HCC) and determined the baseline characteristics and clinical significance of TCR. Methods: High‐throughput sequencing was used to determine the profile of complementarity‐determining region 3 (CDR3) of the TCR‐β chain variable (TRBV) in the tumor and normal tissue samples of 14 HCC patients. At the same time, TRBV diversity and differences in expression between tumor and normal tissues were investigated. The cumulative frequency of top 100 CDR3 (CF100), clonality, and Shannon entropy as indices to evaluate diversity, Results: The diversity of TRBV CDR3 showed no significant difference between tumor and normal tissues. Of the 58 V gene segments in TRBV, TRBV16 and TRBV7‐6 had a significantly higher frequency in the tumor group than in the normal group (p < 0.05). The frequency of 14 J gene segments showed no significant difference between tumor and normal tissues. In contrast, the frequency of 22 TRBVx/BJx combinations was significantly higher in the tumor than in the normal tissue. In addition, the length and type of TRBV CDR3 were similar in tumor and normal tissues, and a Gaussian distribution was observed in both groups. Conclusion: This study provided a large amount of information about the TCR lineage in HBV‐associated HCC, laying the foundation for further research. In addition, the fact that the immune repertoire (TRBV CDR3) hardly differs between tumor and adjacent normal tissue provides a new clue for exploring the mechanism of the liver as an organ with immune privileges. Background: A cost-effective Escherichia coli expression system has gained popularity for producing virus-like particle (VLP) vaccines. However, the challenge lies in balancing the endotoxin residue and removal costs, as residual endotoxins can cause inflammatory reactions in the body. Results: In this study, porcine parvovirus virus-like particles (PPV-VLPs) were successfully assembled from Decreased Endotoxic BL21 (BL21-DeE), and the effect of structural changes in the lipid A of BL21 on endotoxin activity, immunogenicity, and safety was investigated. The lipopolysaccharide purified from BL21-DeE produced lower IL-6 and TNF-α than that from wild-type BL21 (BL21-W) in both RAW264.7 cells and BALB/c mice. Additionally, mice immunized with PPV-VLP derived form BL21-DeE (BL21-DeE-VLP) showed significantly lower production of inflammatory factors and a smaller increase in body temperature within 3 h than those immunized with VLP from BL21-W (BL21-W-VLP) and endotoxin-removed VLP (ReE-VLP). Moreover, mice in the BL21-DeE-VLP immunized group had similar levels of serum antibodies as those in the BL21-W-VLP group but significantly higher levels than those in the ReE-VLP group. Furthermore, the liver, lungs, and kidneys showed no pathological damage compared with the BL21-W-VLP group. Conclusion: Overall, this study proposes a method for producing VLP with high immunogenicity and minimal endotoxin activity without chemical or physical endotoxin removal methods. Effects of Cr alloying on phase stability, magnetism, and electronic structures in both body‐centered cubic (bcc) and face‐centered cubic (fcc) phases and on the transformation from fcc to bcc are studied by first‐principles calculations. Results show that the doped Cr atoms in fcc and bcc phases choose distinct occupation models. This phenomenon can be understood from the amount of electron density of states close to Fermi energy. For magnetism, Cr tends to be antiferromagnetically coupled with the surrounding Fe in the studied phases. The magnetic moment of Fe is greater than that of Cr in bcc, but the order is reversed in fcc. The moment of Fe is dictated by the distance between it and the doped Cr in bcc, whereas it is dominated by spatial orientation with Cr in fcc. For phase stability, it is found that the alloying of Cr prefers destabilizing bcc while tends to stabilize fcc, leading to a strong inhibition of phase transition from fcc to bcc. Notably, the role in the fcc phase is more prominent than that in bcc, which can be associated with the antiferromagnetism between Fe and Cr in fcc. [ABSTRACT FROM AUTHOR]

Aberrant signaling in tumor cells induces nonmetabolic functions of some metabolic enzymes in many cellular activities. As a key glycolytic enzyme, the nonmetabolic function of hexokinase 2 (HK2) plays a role in tumor immune evasion. However, whether HK2, dependent of its nonmetabolic activity, plays a role in human pancreatic ductal adenocarcinoma (PDAC) tumorigenesis remains unclear. Here, we demonstrated that HK2 acts as a protein kinase and phosphorylates IκBα at T291 in PDAC cells, activating NF‐κB, which enters the nucleus and promotes the expression of downstream targets under hypoxia. HK2 nonmetabolic activity‐promoted activation of NF‐κB promotes the proliferation, migration, and invasion of PDAC cells. These findings provide new insights into the multifaceted roles of HK2 in tumor development and underscore the potential of targeting HK2 protein kinase activity for PDAC treatment. Wheat cultivar Zhongmai 895 was earlier found to carry YR86 in an 11.6 Mb recombination-suppressed region on chromosome 2AL when crossed with Yangmai 16. To fine-map the YR86 locus, we developed two large F2 populations from crosses Emai 580/Zhongmai 895 and Avocet S/Zhongmai 895. Remarkably, both populations exhibited suppressed recombination in the same 2AL region. Collinearity analysis across Chinese Spring, Aikang 58, and 10+ wheat genomes revealed a 4.1 Mb chromosomal inversion spanning 708.5-712.6 Mb in the Chinese Spring reference genome. Molecular markers were developed in the breakpoint and were used to assess a wheat cultivar panel, revealing that Chinese Spring, Zhongmai 895, and Jimai 22 shared a common sequence named InvCS, whereas Aikang 58, Yangmai 16, Emai 580, and Avocet S shared the sequence named InvAK58. The inverted configuration explained the suppressed recombination observed in all three bi-parental populations. Normal recombination was observed in a Jimai 22/Zhongmai 895 F2 population, facilitating mapping of YR86 to a genetic interval of 0.15 cM corresponding to 710.27-712.56 Mb falling within the inverted region. Thirty-three high-confidence genes were annotated in the interval using the Chinese Spring reference genome, with six identified as potential candidates for YR86 based on genome and transcriptome analyses. With the emergence of novel variants, Omicron variant caused a different clinical picture than the previous variants and little evidence was reported regarding perioperative outcomes after Omicron variants. The aim of the study was to evaluate the postoperative outcomes of gastrointestinal cancer patients following Omicron variants infection and also to determine the timing of surgery after infection recovery. A total of 124 patients who underwent gastrointestinal cancer surgery with prior SARS-CoV-2 infection between December 2022 and February 2023 were retrospectively reviewed. 174 cases underwent the same operation during December 2018 and February 2019 as control group. SARS-CoV-2-infected patients were further categorized into three groups based on infected time (1–3 weeks; 4–6 weeks; and ≥ 7 weeks). 90.3% of SARS-CoV-2-infected patients had mild symptoms. The COVID-19 vaccination rate was 71.0%, with a full vaccination rate of 48.4%. There were no significant differences in 30-day morbidity and mortality. There was also no significant difference in pulmonary complications, cardiovascular complications, and surgical complications between the three different diagnosis time groups. In conclusion, reducing waiting time for elective surgery was safe for gastrointestinal cancer patients in the context of an increased transmissibility and milder illness severity with Omicron variant. Biological soil crusts (biocrusts) are widespread soil photosynthetic communities covering about 12% of Earth's land surface, and play crucial roles in terrestrial carbon (C) and nitrogen (N) cycles, yet scalable quantifications of biocrusts and their biogeochemical contributions are notably lacking. While remote sensing has enormous potential to assess, scale, and contextualize biocrusts and their functions, the applicability of hyperspectral data in predicting C‐ and N‐related biocrust traits remains largely unexplored. We address this issue by evaluating the potential of in situ hyperspectral data to predict C and N across a range of biocrust species and different environmental conditions. We found that in situ hyperspectral reflectance measurements can be used to predict biocrust tissue C/N ratios and N concentrations with relatively high accuracy but to a lesser extent for potential biocrust N2 fixation rates. Critical wavelength domains included the visible region of the spectrum from roughly 490–600 nm, which most effectively captured variations in biocrust tissue C, and the shortwave infrared region from 1,150 to 1,350 nm and 1,550–1,650 nm, which most effectively captured biocrust tissue N and N2 fixation potential. Finally, we provide evidence that multi‐ and hyperspectral missions with targeted band placement, such as the proposed 26‐band Landsat Next, could be effective in predicting biocrust traits. This work provides a critical step in understanding how to apply data from new and upcoming satellite missions to the monitoring of biocrusts. Biocrusts play an important role in the carbon (C) and nitrogen (N) cycles of terrestrial ecosystems. The C and N status of biocrusts can be examined in various ways. However, the applicability of studying C and N status of biocrusts using spectral reflectance is poorly explored. We combined statistical models and spectral reflectance measured using a spectroradiometer within the wavelength range from 400 to 2,450 nm to predict variations in C and N status of samples from 14 representative biocrust species, and under both wet and dry conditions. We found that variations in biocrust tissue C/N ratios and N concentrations could be accurately predicted. We also found that the wavelength range of 490–600 nm, and 1,150–1,350 nm as well as around 1,600 nm are the most important wavelength domains to study C and N status of biocrusts. Finally, we provide evidence that upcoming satellite missions with proper band placement, such as the proposed 26‐band Landsat Next, could be effective in predicting biocrust traits. Background: Portal vein tumor thrombosis (PVTT) signifies late-stage hepatocellular carcinoma (HCC) with high-risk progression and poor prognosis. As a standard treatment, sorafenib monotherapy has limited the efficacy in managing HCC with PVTT. Currently, both hepatic arterial infusion chemotherapy (HAIC) and the combination of camrelizumab and rivoceranib have shown favorable survival benefits for advanced HCC, surpassing the standard sorafenib treatment. In this study, we investigate the safety and efficacy of HAIC combined with camrelizumab and rivoceranib in treating HCC patients with PVTT. Methods: From January 2020 to December 2021, HCC patients with PVTT, who received either a triple regime of HAIC combined with camrelizumab and rivoceranib or a dual regime of camrelizumab and rivoceranib as their first-line treatment, were reviewed for eligibility at four hospital centers in China. To balance any intergroup differences, propensity score matching (PSM) was applied. The aim of this study is to compare the efficacy of the dual and triple combination treatment regimens based on survival prognosis and tumor response and evaluate the safety based on the occurrence of adverse reactions. Result: In this study, a total of 411 patients who received either the triple treatment regime (HAIC combined with camrelizumab plus rivoceranib, referred to as the HAICCR group, n = 292) or the dual treatment regime (camrelizumab combined with rivoceranib, referred to as the CR group, n = 119) between January 2020 and December 2021 were included. The results showed that the HAICCR group exhibited significantly better overall survival (mOS: 19.60 months vs. 11.50 months, p < 0.0001) and progression-free survival (mPFS: 10.0 months vs. 5.6 months, p < 0.0001) compared to the CR group in the overall cohort. Moreover, the HAICCR group also had a significantly higher ORR (objective response rate, 55.5% vs. 42.0%, p = 0.013) and DCR (disease control rate, 89.0% vs. 79.0%) compared to the CR group. After PSM, a final matched cohort of 83 pairs was obtained, and the survival benefits were consistent in this cohort as well (mOS: 18.70 months vs. 11.0 months, p < 0.0001; mPFS: 10.0 months vs. 5.6 months, p < 0.0001). However, there was no significant difference in the ORR between the triple and dual combination regimes. Univariate and multivariate analysis showed that CTP (Child–Turcotte–Pugh) stage, ALBI (albumin–bilirubin index) grade, tumor number, and treatment regime were significant risk factors affecting overall survival, while AFP (α-fetoprotein) level, tumor number, metastasis, and treatment regime were significant risk factors affecting progression-free survival. As for safety, hypertension and hand–foot syndrome were the two most common adverse reactions in both groups, with no significant difference in the occurrence of adverse reactions between the two groups (p < 0.05). Conclusion: In the context of advanced HCC patients with PVTT, the combination regime of HAIC and camrelizumab plus rivoceranib demonstrates more excellent capacity for prolonging survival and offers a well-tolerated safety compared to the CR dual therapy approach. To describe the epidemiological feature of schistosomiasis in China mainland. The information on national schistosomiasis case and incidence were collected, its epidemiological feature was analyzed by time, location and age. A total of 78 656 cases and 15 deaths of schistosomiasis were reported in China between 2004 and 2018. The incidence of schistosomiasis in 2009-2013 was statistical significance higher than before and after. Hubei was the most seriously afflicted among all endemic provinces in the periods of 2004-2009 and 2010-2014. In addition, the 50-59 and 60-69 age groups had significantly higher incidence. The incidence of schistosomiasis was concentrated in the middle and lower reaches of the Yangtze River and people over 50 were at higher risk. [ABSTRACT FROM AUTHOR]

Background: Recent studies have linked alterations in the gut microbiome and metabolic disruptions to the invasive behavior and metastasis of colorectal cancer (CRC), thus affecting patient prognosis. However, the specific relationship among gut microbiome, metabolite profiles, and mutated-RAS/BRAF metastatic colorectal cancer (M-mCRC) remains unclear. Furthermore, the potential mechanisms and prognostic implications of metabolic changes induced by gut microbiome alterations in patients with M-mCRC still need to be better understood. Methods: We conducted Mendelian randomization (MR) to evaluate the causal relationship of genetically predicted 196 gut microbiome features and 1400 plasma metabolites/metabolite ratios on M-mCRC-specific survival. Additionally, we identified significant gut microbiome-metabolites/metabolite ratio associations based on M-mCRC. Metabolite information was annotated, and functional annotation and pathway enrichment analyses were performed on shared proteins corresponding to significant metabolite ratios, aiming to reveal potential mechanisms by which gut microbiome influences M-mCRC prognosis via modulation of human metabolism. Results: We identified 11 gut microbiome features and 49 known metabolites/metabolite ratios correlated with M-mCRC-specific survival. Furthermore, we identified 17 gut microbiome-metabolite/metabolite ratio associations specific to M-mCRC, involving eight lipid metabolites and three bilirubin degradation products. The shared proteins corresponding to significant metabolite ratios were predominantly localized within the integral component of the membrane and exhibited enzymatic activities such as glucuronosyltransferase and UDP-glucuronosyltransferase, crucial in processes such as glucuronidation, bile secretion, and lipid metabolism. Moreover, these proteins were significantly enriched in pathways related to ascorbate and aldarate metabolism, pentose and glucuronate interconversions, steroid hormone biosynthesis, and bile secretion. High‐performance soft magnetic materials are important for energy conservation and emission reduction. One challenge is achieving a combination of reliable temperature stability, high resistivity, high Curie temperature, and high saturation magnetization in a single material, which often comes at the expense of intrinsic coercivity–a typical trade‐off in the family of soft magnetic materials with homogeneous microstructures. Herein, a nanostructured FeCoNiSiAl complex concentrated alloy is developed through a hierarchical structure strategy. This alloy exhibits superior soft magnetic properties up to 897 K, maintaining an ultra‐low intrinsic coercivity (13.6 A m−1 at 297 K) over a wide temperature range, a high resistivity (138.08 µΩ cm−1 at 297 K) and the saturation magnetization with only a 16.7% attenuation at 897 K. These unusual property combinations are attributed to the dual‐magnetic‐state nature with exchange softening due to continuous crystal ordering fluctuations at the atomic scale. By deliberately controlling the microstructure, the comprehensive performance of the alloy can be tuned and controlled. Trio exome sequencing was performed on a female fetus with an increased nuchal translucency, along with nasal bone hypoplasia, suspected cleft palate and abnormal outflow tract of the heart. A de novo heterozygous variant c.5500_5507del, p.(Tyr1834Argfs × 58) in the MED12 gene was detected. Loss‐of‐function variants in MED12 in females are associated with Hardikar syndrome (HS). A follow‐up ultrasound at 15+5 weeks of gestation identified multiple fetal anomalies including bilateral cleft lip and palate, diaphragmatic hernia, atrioventricular septal defect, persistent truncus arteriosus, and bilateral renal pelvis dilation. Fetal autopsy confirmed the prenatal sonographic findings, and the MED12 variant was discussed by our multidisciplinary team to be the cause of fetal anomalies. Our case is the first prenatal one in which HS was diagnosed due to first trimester structural malformations. This case report presents another example of early identification of a major anomaly which allows earlier genetic diagnosis and more time for clinical management. Image and video inpainting is a classic problem in computer vision and computer graphics, aiming to fill in the plausible and realistic content in the missing areas of images and videos. With the advance of deep learning, this problem has achieved significant progress recently. The goal of this paper is to comprehensively review the deep learning-based methods for image and video inpainting. Specifically, we sort existing methods into different categories from the perspective of their high-level inpainting pipeline, present different deep learning architectures, including CNN, VAE, GAN, diffusion models, etc., and summarize techniques for module design. We review the training objectives and the common benchmark datasets. We present evaluation metrics for low-level pixel and high-level perceptional similarity, conduct a performance evaluation, and discuss the strengths and weaknesses of representative inpainting methods. For sperm cryopreservation, the conventional method, which requires glycerol, has been used for a long time. In addition, the permeable cryoprotectant-free vitrification method has been continuously studied. Although the differences of cryopreservation effects between the two methods have being studied, differences in microRNA (miRNA) profiles between them remain unclear. In this study, we investigated the differences in miRNA expression profiles among conventional freezing sperm, droplet vitrification freezing sperm and fresh human sperm. We also analyzed the differences between these methods in terms of differentially expressed miRNAs (DEmiRs) related to early embryonic development and paternal epigenetics. Our results showed no significant differences between the cryopreservation methods in terms of sperm motility ratio, plasma membrane integrity, DNA integrity, mitochondrial membrane potential, acrosome integrity, and ultrastructural damage. However, sperm miRNA-sequencing showed differences between the two methods in terms of the numbers of DEmiRs (28 and 19 with vitrification using a nonpermeable cryoprotectant and the conventional method, respectively) in postthaw and fresh sperm specimens. DEmiRs related to early embryonic development and paternal epigenetics mainly included common DEmiRs between the groups. Our results showed that the differences between conventional freezing and droplet vitrification were minimal in terms of miRNA expression related to embryonic development and epigenetics. Changes in sperm miRNA expression due to freezing are not always detrimental to embryonic development. This study compared differences in miRNA expression profiles before and after cryopreservation between cryopreservation by conventional and vitrification methods. The unique structural features of hard carbon (HC) make it a promising anode candidate for sodium‐ion batteries (SIB). However, traditional methods of preparing HC require special equipment, long reaction times, and large energy consumption, resulting in low throughputs and efficiency. In our contribution, a novel synthesis method is proposed, involving the formation of HC nanosheets (NS‐CNs) within minutes by creating an anoxic environment through flame combustion and further introducing sulfur and nitrogen sources to achieve heteroatom doping. The effect of heterogeneous element doping on the microstructure of HC is quantitatively analyzed by high‐resolution transmission electron microscopy and image processing technology. Combined with density functional theory calculation, it is verified that the functionalized HC exhibits stronger Na+ adsorption ability, electron gain ability, and Na+ migration ability. As a result, NS‐CNs as SIB anodes provide an ultrahigh reversible capacity of 542.7 mAh g−1 at 0.1 A g−1, and excellent rate performance with a reversible capacity of 236.4 mAh g−1 at 2 A g−1 after 1200 cycles. Furthermore, full cell assembled with NS‐CNs as the can present 230 mAh g−1 at 0.5 A g−1 after 150 cycles. Finally, in/ex situ techniques confirm that the excellent sodium storage properties of NS‐CNs are due to the construction of abundant active sites based on the novel synthesis method for realizing the reversible adsorption of Na+. This work provides a novel strategy to develop novel carbons and gives deep insights for the further investigation of facile preparation methods to develop high‐performance carbon anodes for alkali‐ion batteries. The unique edge states of the zigzag β-SiC7 nanoribbons aroused our attention, and therefore, based on first-principles calculations, we investigated their spin-dependent electronic transport properties by constructing controllable defects to modulate these special edge states. Interestingly, by introducing rectangular edge defects in the SiSi and SiC edge-terminated systems, not only the spin-unpolarized is successfully converted to completely spin-polarized, but also the direction of polarization can be switched, thus enabling a dual spin filter. The analyses further reveal that the two transmission channels with opposite spins are spatially separated and that the transmission eigenstates are highly concentrated at the relative edges. The specific edge defect introduced only suppresses the transmission channel at the same edge but reserves the transmission channel at the other edge. In addition, for the CSi and CC edge-terminated systems, an additional spin-down band exists due to spin splitting in the spin-up band at EF, so that besides the original spatially separated two spin-opposite channels, an extra spin channel is distributed at the upper edge, resulting in unidirectional fully spin-polarized transport. The peculiar spatially separated edge states and excellent spin filtering properties could open up further possibilities for β-SiC7-based electronic devices in spintronics applications. Introduction: The safety and efficacy of CRS + HIPEC combined with urinary tract resection and reconstruction are controversial. This study aims to summarize the clinicopathological features and to evaluate the safety and survival prognosis of CRS + HIPEC combined with urinary tract resection and reconstruction. Methods: The patients who underwent urinary tract resection and reconstruction as part of CRS surgery were retrospectively selected from our disease-specific database for analysis. The clinicopathological characteristics, treatment-related variables, perioperative adverse events (AEs), and survival outcomes were studied using a descriptive approach and the K-M analysis with log-rank comparison. Results: Forty-nine patients were enrolled. Perioperative serious AEs (SAEs) were observed in 11 patients (22.4%), with urinary SAEs occurring in 3 patients (6.1%). Additionally, there were 23 cases (46.8%) involving urinary adverse events (UAEs). The median overall survival (OS) in the entire cohort was 59.2 (95%CI: 42.1–76.4) months. The median OS of the UAE group and No-UAE group were 59.2 months (95%CI not reached), and 50.5 (95%CI: 11.5 to 89.6) months, respectively, with no significant difference (P = 0.475). Furthermore, there were no significant differences in OS based on the grade of UAEs or the number of UAEs (P = 0.562 and P = 0.622, respectively). Conclusion: The combination of CRS + HIPEC with urinary tract resection and reconstruction is associated with a high incidence of Grade I-II UAEs, which do not have an impact on OS. The safety profile of this combined technique is acceptable. However, this is a retrospective single-center single-arm analysis, with limitations of generalizability and potential selection bias. Background: The aim of the study was to investigate the muscle differences in children with osteogenesis imperfecta (OI) using opportunistic low-dose chest CT and to compare different methods for the segmentation of muscle in children. Methods: This single center retrospective study enrolled children with OI and controls undergoing opportunistic low-dose chest CT obtained during the COVID pandemic. From the CT images, muscle size (cross-sectional area) and density (mean Hounsfield Units [HU]) of the trunk muscles were measured at the mid-T4 and the mid-T10 level using two methods, the fixed thresholds and the Gaussian mixture model. The Bland-Altman method was also used to compute the strength of agreement between two methods. Comparison of muscle results between OI and controls were analyzed with Student t tests. Results: 20 children with OI (mean age, 9.1 ± 3.3 years, 15 males) and 40 age- and sex-matched controls were enrolled. Mean differences between two methods were good. Children with OI had lower T4 and T10 muscle density than controls measured by the fixed thresholds (41.2 HU vs. 48.0 HU, p < 0.01; 37.3 HU vs. 45.9 HU, p < 0.01). However, children with OI had lower T4 muscle size, T4 muscle density, T10 muscle size and T10 muscle density than controls measured by the Gaussian mixture model (110.9 vs. 127.2 cm2, p = 0.03; 44.6 HU vs. 51.3 HU, p < 0.01; 72.6 vs. 88.0 cm2, p = 0.01; 41.6 HU vs. 50.3 HU, p < 0.01, respectively). Conclusions: Children with OI had lower trunk muscle density indicating that OI might also impair muscle quality. Comprehensive Summary: The conversion of CF3‐alkenes to gem‐difluoroalkenes using reductive cross‐coupling strategy has received much attention in recent years, however, the use of green and readily available reducing salt to mediate these reactions remains to be explored. In this work, a concise construction of gem‐difluoroalkenes, which requires neither a catalyst nor a metal reducing agent, was established. Rongalite, a safe and inexpensive industrial product, was employed as both a radical initiator and reductant. This procedure was compatible with both linear and cyclic diaryliodonium salts, enabling a wide variety of substrates (>70 examples). Spinel oxides are affordable transition-metal electrocatalytic materials, but their low conductivity and intrinsic activity restrict their wide application in electrolyzed water catalysts. In this study, a series of heterojunction samples comprising phosphorus-doped spinel oxides NiCo2O4 in collaboration with g-C3N4 (NCPx@C3N4, x = 0.25, 0.50, 0.75) were fabricated through a straightforward mixture method for water splitting. Therein, the NCP0.5@C3N4 electrocatalyst exhibits remarkable performance compared to the other synthesized compounds, demonstrating a small overpotential of 247 mV at 10 mA cm–2 for the oxygen evolution reaction (OER), a favorable Tafel slope of 48 mV dec–1, and decent durability. The collaborative interaction between phosphorus doping and g-C3N4 results in the creation of numerous oxygen defects, increasing the electrochemically active surface area in NCP0.5@C3N4 and demonstrating a direct enhancement of the OER. Density functional theory (DFT) calculations verified that the introduction of phosphorus and its collaboration with g-C3N4 effectively regulated the electronic structure and optimized the d-band center position. N-glycosylation is a highly conserved glycan modification that plays crucial roles in various physiological processes, including protein folding, trafficking, and signal transduction. Porcine deltacoronavirus (PDCoV) poses a newly emerging threat to the global porcine industry. The spike protein of PDCoV exhibits a high level of N-glycosylation; however, its role in viral infection remains poorly understood. In this study, we applied a lentivirus-based entry reporter system to investigate the role of N-glycosylation on the viral spike protein during PDCoV entry stage. Our findings demonstrate that N-glycosylation at positions 652 and 661 of the viral spike protein significantly reduces the infectivity of PDCoV pseudotyped virus. Broadband near‐infrared (NIR) emissions in the long wavelength region with simultaneous high efficiency and thermal stability are desirable for NIR phosphor‐converted light‐emitting diodes (NIR pc‐LEDs) light sources. Herein, a novel broadband NIR‐emitting phosphor is reported, Ba5La3MgAl3O15:Cr3+ (BLMAO:Cr3+), which exhibits a NIR emission band spanning 700–1300 nm with maximum peak wavelength (λmax) at 925 nm and full width at half maximum of 160 nm. The optimized phosphor achieves an internal quantum efficiency of 66%, and the integrated emission intensity at 423 K remains 61% of that at room temperature, which surpasses most Cr3+‐activated phosphors emitting in the long‐wavelength NIR region (λmax > 900 nm). The NIR luminescence properties are elucidated on the basis of density functional calculations of the multiple Cr3+ local structures as generated by the Ba/La occupation disorder in the host material. The applications of the NIR phosphor in pc‐LED devices are demonstrated, showing potential in night‐vision and nondestructive examination. NASICON‐type cathode with remarkable ionic conductivity is perspective candidate for fast‐charging sodium‐ion battery. However, severely restricted by low electrical conductivity and poor interfacial kinetics, it usually delivers poor charge transfer kinetics. Different from traditional carbon compositing with high carbon contents, herein, a trace carbon incorporation tactic is proposed based on a typical NASICON‐structured Na3V2(PO4)3. First, particle‐growth process of Na3V2(PO4)3 is regulated via incorporating carbon dot, significantly reducing its particle size to shorten charge diffusion path. Second, electrical conductivity of Na3V2(PO4)3 is improved without sacrificing its high electrochemical activity due to the incorporated trace carbon content (0.76 wt.%). Third, Na3V2(PO4)3‐electrolyte interface structure is optimized by abundant functional groups from the incorporated carbon dot, enabling a thin and stable NaF‐rich CEI layer to boost interface kinetics. As a result, carbon dot endows Na3V2(PO4)3 with ultrastable cyclability up to 20 k cycles (capacity retention of 98.4%) and excellent rate capability (up to 200 C) in half cell, as well as high energy density (368.7 Wh kg−1) and fast charging property (≈110.2 s per charging with 250.8 Wh kg−1 input) in full cell. Cocaine use disorder is a significant public health issue without an effective pharmacological treatment. Successful treatments are hindered in part by an incomplete understanding of the molecular mechanisms that underlie long-lasting maladaptive plasticity and addiction-like behaviors. Here, we leverage a large RNA sequencing dataset to generate gene coexpression networks across six interconnected regions of the brain's reward circuitry from mice that underwent saline or cocaine self-administration. We identify phosphodiesterase 1b (Pde1b), a Ca2+/calmodulin-dependent enzyme that increases cAMP and cGMP hydrolysis, as a central hub gene within a nucleus accumbens (NAc) gene module that was bioinformatically associated with addiction-like behavior. Chronic cocaine exposure increases Pde1b expression in NAc D2 medium spiny neurons (MSNs) in male but not female mice. Viral-mediated Pde1b overexpression in NAc reduces cocaine self-administration in female rats but increases seeking in both sexes. In female mice, overexpressing Pde1b in D1 MSNs attenuates the locomotor response to cocaine, with the opposite effect in D2 MSNs. Overexpressing Pde1b in D1/D2 MSNs had no effect on the locomotor response to cocaine in male mice. At the electrophysiological level, Pde1b overexpression reduces sEPSC frequency in D1 MSNs and regulates the excitability of NAc MSNs. Lastly, Pde1b overexpression significantly reduced the number of differentially expressed genes (DEGs) in NAc following chronic cocaine, with discordant effects on gene transcription between sexes. Together, we identify novel genemodules across the brain's reward circuitry associated with addiction-like behavior and explore the role of Pde1b in regulating the molecular, cellular, and behavioral responses to cocaine. Ferroelectricity in two-dimensional (2D) systems generally arises from phonons and has been widely investigated. On the contrary, electronic ferroelectricity in 2D systems has been rarely studied. Using first-principles calculations, the ferroelectric behavior of the buckled blue SiSe monolayer under strain are explored. It is found that the direction of the out-of-plane ferroelectric polarization can be reversed by applying an in-plane strain. And such polarization switching is realized without undergoing geometric inversion. Besides, the strain-triggered polarization reversal emerges in both biaxial and uniaxial strain cases, indicating it is an intrinsic feature of such a system. Further analysis shows that the polarization switching is the result of the reversal of the magnitudes of the positive and negative charge center vectors. And the variation of buckling is found to play an important role, which results in the switch. Moreover, a non-monotonic variation of band gap with strain is revealed. Coherent precipitation, a common strengthening approach, is typically subjected to spatial non-uniformity due to microscopic segregation, leading to multi-scale stress heterogeneity. Such heterogeneity remains poorly characterized because unavailable local strain-free lattice parameters invalidate traditional diffraction-based stress measurement techniques. To overcome these limitations, we demonstrate a submicron-resolved synchrotron X-ray diffraction method to map coherency stress distribution based on the γ/γ′ lattice misfits in Ni-based superalloys. Assisted by finite element analysis, sub-dendritic stresses are deduced from heterogeneous coherency stresses, confirmed by the diffraction experiments. The methodology offers a comprehensive framework to assess stress heterogeneity at multi-scales for all coherent precipitation strengthened alloys. Iterative learning control (ILC) is a popular scheme in the trajectory tracking of manipulators, greatly improving tracking accuracy despite often requiring multiple iterations over identical trajectories. This research introduces an optimization technique for ILC parameters, enhanced with proportional-derivative (PD) feedback control, which aims to significantly reduce tracking errors within a single iteration. In the proposed approach, a PD feedback controller is utilized in the first run, collecting error data. An ILC controller is then incorporated in the second run to minimize the tracking error. Utilizing the dynamic model of the system, the transcription method transforms the continuous-form optimization problem concerning the ILC parameters into a discrete form, enabling its solution via standard numerical optimization algorithms. To demonstrate the effectiveness of the proposed approach in reducing tracking errors, we compared the tracking errors for the first and second runs of the system using frequency-domain analysis and conducted simulations and experiments on two different trajectory types. The operating speed of the commercial electromagnetic suspension (EMS) maglev train has been over 430 km/h, making it the fastest means of land transportation. With the increasing demands of people traveling, it is necessary to further improve the operating speed of maglev trains. Aiming to examine whether the existing EMS system can meet the requirements of ultra-high-speed operation (more than 600 km/h), the electromagnetic characteristics of an EMS linear motor under the operating speed of 600–1000 km/h fed by a square-wave voltage supply is investigated in this article. First, an electromagnetic field model of the EMS system under the square-wave voltage supply is established to investigate its electromagnetic performance, e.g., the characteristics of phase current, the magnetic flux density, and the electromagnetic force. Second, the relationship between the harmonic components of the air-gap magnetic flux density and electromagnetic force is investigated using the two-dimensional fast Fourier transform (2D-FFT) to reveal the mechanism of electromagnetic force ripple. Third, to address the issues of excessive armature current density and significant electromagnetic force ripple, the linear motor is re-designed by enlarging the stator slot area and reshaping the mover's main magnetic poles. Furthermore, the Taguchi method is used to further improve the electromagnetic characteristics of the linear electric motor. Porcine reproductive and respiratory syndrome virus (PRRSV) critically threatens the pig industry in China. Lineage 1 PRRSV, which is divided into L1A–L1F and L1H–L1J, is widely recognized as the most extensively genetically diverse lineage globally. L1A (NADC34-like) and L1C (NADC30-like) PRRSVs have become the current major circulating strains in China. Notably, strains from other branches of L1 have not been reported in China. During our epidemiological investigation of PRRSV, we identified a new strain named HLJDZD55. Phylogenetic analysis of the ORF5 gene revealed that HLJDZD55 belongs to the L1B branch. Alignment of deduced amino acid sequences based on the Nsp2 gene indicated that HLJDZD55 has a discontinuous deletion of 131 amino acids (111 + 1 + 19). We further sequenced the whole genome of HLJDZD55, and phylogenetic analysis based on the whole-genome sequence revealed that HLJDZD55 belongs to the L1C branch. Recombination analysis of the whole genome demonstrated that HLJDZD55 is a recombinant strain of TJZH-1607 (L1C, identified in China) and Minnesota 14 (L1B, identified in the USA). These findings suggested that HLJDZD55 is a newly emerged lineage 1 PRRSV in China and is closely related to L1B PRRSV in the US, which may have been introduced from the U.S. strain and subsequently recombined with the local Chinese strain and underwent evolution. Background: Inflammation has been reported to be related to anemia. As a novel inflammatory marker, Systemic immune-inflammation index (SII) has not been studied with Anemia. The aim of this study was to investigate the possible relationship between SII and anemia. Methods: This retrospective cross-sectional survey was conducted using data from the 2005-2018 National Health and Nutrition Examination Survey (NHANES) population. In total, 19851 American adults aged =18 years were included. SII was calculated as the platelet count×neutrophil count/lymphocyte count. Anemia was defined as hemoglobin (Hgb) levels of < 13 g/dL in males and < 12 g/dL in females. Logistic regression analyses, subgroup analyses and sensitivity analyses were performed to investigate the relationship between SII and anemia. Results: Our study included a total of 19851 patients, of which 1501 (7.6%) had anemia. After adjusting for all covariates, the multivariate logistic regression analysis showed that a higher SII (In-transform) level was associated with increased likelihood of anemia (OR=1.51, 95% CI: 1.36-1.68, P<0.001). The association between SII and anemia exhibited a nonlinear manner. The positive correlation between SII and anemia was related to the severity of anemia. Subgroup analysis showed that there was no significant dependence on age, family income, body mass index, hypertension, kidney disease and cancer except gender on this positive association. Furthermore, sensitivity analyses confirmed the robustness of our results. Conclusion: Our study demonstrated that SII was positively associated with anemia especially among female participants. Introduction: CD8+T cell tolerance plays an important role in tumor escape. Recent studies have shown that CD45+ erythroid progenitor cells (CD45+EPCs) generated through splenic extramedullary erythropoiesis suppress tumor immunity. However, the mechanism underlying how CD45+EPCs mediate CD8+T cell tolerance remains incompletely understood and requires further research. Methods: In this study, the antigen-processing abilities of CD45+EPCs was verified through both in vitro and in vivo experiments. We have used the method of co-culture in vitro and adoptive transfer experiments in vivo to explore the effects of CD45+EPCs on CD8+T cell tolerance. RNA-sequencing analysis and blocking experiments were used to evaluate the role of ROS in the CD45+EPC mediated tolerance of CD8+T cells. Finally, we incorporated uric acid into the adoptive transfer experiments to rescue the CD45+EPC mediated tumor-promoting effect. Results and discussion: We found that CD45+EPCs take up soluble proteins, present antigenic epitopes on their surface, and induce antigen-specific CD8+T cell anergy. In addition, we found that CD45+EPC directly nitrates tyrosine within the TCR/CD8 complex via the production of reactive oxygen species and peroxynitrite, preventing CD8+ T cells from responding to their specific peptide antigens. Furthermore, uric acid treatment effectively abolished the immunosuppressive effects of CD45+EPCs during CD8+T cell adoptive transfer, thereby enhancing the anti-tumor efficacy. In this study, a Ti3C2 MXene@g-C3N4 composite powder (TM-CN) was prepared by the ultrasonic self-assembly method and then loaded onto a carbon nanofiber membrane by the self-assembly properties of MXene for the treatment of organic pollutants in wastewater. The characterization of the TM-CN and the C-TM-CN was conducted via X-ray diffraction (XRD), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared spectrometer (FTIR) to ascertain the successful modification. The organic dye degradation experiments demonstrated that introducing an appropriate amount of Ti3C2 MXene resulted in the complete degradation of RhB within 60 min, three times the photocatalytic efficiency of a pure g-C3N4. The C-TM-CN exhibited the stable and outstanding photocatalytic degradation of the RhB solution over a wide range of pH values, indicating the characteristics of the photodegradation of organic pollutants in a wide range of aqueous environments. Furthermore, the results of the cyclic degradation experiments demonstrated that the C-TM-CN composite film maintained a degradation efficiency of over 85% after five cycles, thereby confirming a notable improvement in its cyclic stability. Consequently, the C-TM-CN composite film exhibits excellent photocatalytic performance and is readily recyclable, making it an auspicious eco-friendly material in water environment remediation. Senecavirus A (SVA) is an important emerging swine pathogen that causes vesicular lesions in swine and acute death in newborn piglets. VP2 plays a significant role in the production of antibodies, which can be used in development of diagnostic tools and vaccines. Herein, the aim of the current study was to identify B-cell epitopes (BCEs) of SVA for generation of epitope-based SVA marker vaccine. Three monoclonal antibodies (mAbs), named 2E4, 1B8, and 2C7, against the SVA VP2 protein were obtained, and two novel linear BCEs, 177SLGTYYR183 and 266SPYFNGL272, were identified by peptide scanning. The epitope 177SLGTYYR183 was recognized by the mAb 1B8 and was fully exposed on the VP2 surface, and alanine scanning analysis revealed that it contained a high continuity of key amino acids. Importantly, we confirmed that 177SLGTYYR183 locates on "the puff" region within the VP2 EF loop, and contains three key amino acid residues involved in receptor binding. Moreover, a single mutation, Y182A, blocked the interaction of the mutant virus with the mAb 1B8, indicating that this mutation is the pivotal point for antibody recognition. Excessive extracellular matrix (ECM) deposition is a defining feature of cardiac fibrosis. Most notably, it is characterized by a significant change in the concentration and volume fraction of collagen I, a disproportionate deposition of collagen subtypes, and a disturbed ECM network arrangement, which directly affect the systolic and diastolic functions of the heart. Immune cells that reside within or infiltrate the myocardium, including macrophages, play important roles in fibroblast activation and consequent ECM remodeling. Through both direct and indirect connections to fibroblasts, monocyte-derived macrophages and resident cardiac macrophages play complex, bidirectional, regulatory roles in cardiac fibrosis. In this review, we discuss emerging interactions between fibroblasts and macrophages in physiology and pathologic conditions, providing insights for future research aimed at targeting macrophages to combat cardiac fibrosis. In this paper, a single‐layer filtering antenna based on half‐mode patches is proposed. It consists of a main driven patch and a parasitic patch. A U‐shaped slot is first etched on the main driven patch to generate a new resonant mode and a radiation null simultaneously. Four pairs of shorting pins are introduced on the main driven patch to raise its operating frequency to near that of the U‐shaped slot, thus broadening the impedance bandwidth (IBW). Additionally, a parasitic patch is introduced near the driven patch to introduce an extra half‐TM10 mode and an extra radiation null, significantly enhancing the IBW and the upper‐edge selectivity of the antenna. A prototype was fabricated and tested for validation. It achieves a peak realized gain of 6.55 dBi and −10‐dB IBW of 8.8%, and exhibits good out‐of‐band rejection performance while maintaining a low profile of 0.017 λ0 and a compact transverse size of 0.29 λ02. In the present study, the complete mitochondrial genome (mitogenome) of the Papilio macilentus (Lepidoptera: Papilionoidea: Papilionidae) was sequenced by next-generation sequencing method. The mitochondrial genome is a circular DNA molecule of 15,264 bp in size with 80.7% AT content, including 37 genes (13 protein-coding genes, 2 rRNA genes, and 22 tRNA genes), and a long non-coding region (Control region). All protein-coding genes are initiated by ATN codons, and terminated with TAA, TAG, or single T. All tRNAs can be folded into common clover leaf secondary structure, except trn-S1. Phylogenetic analyses based on 13 protein-coding genes and 2 rRNA genes using maximum likelihood and Bayesian inference confirmed that P. macilentus and Papilio memnon are clustered into a clade, and revealed the relationships between Papilionini, Troidini, Teinopaippini and Leptocircini. Robotic manipulators play a pivotal role in modern intelligent manufacturing and unmanned production systems, often tasked with executing specific paths accurately. However, the input of the robotic manipulators is trajectory which is a path with time information. The resulting core technology is trajectory planning methods which are broadly classified into two categories: maximum velocity curve (MVC) methods and multiphase direct collocation (MPDC) methods. This paper concentrates on addressing challenges associated with the latter methods. In MPDC methods, the solving efficiency and accuracy are greatly influenced by the number of discretization nodes. When dealing with systems with complex dynamics, such as robotic manipulators, striking a balance between solving time and path discretization errors becomes crucial. We use a mesh refinement (MR) algorithm to find a suitable number of nodes under the premise of ensuring the path discretization error. So, the actual device can effectively implement the planned solutions. Nonetheless, the conventional application of the MR algorithm requires solving the original problem in each iteration; these processes are extremely time-consuming and may fail to solve when dealing with a complex dynamic system. As a result, we propose a sequential optimal trajectory planning scheme to solve the problem efficiently by dividing the original optimal control (OC) problem into two stages: path planning (PP) and trajectory planning (TP). In the PP stage, we employ a DC method based on arc length and an MR algorithm to identify key nodes along the specified path. This aims to minimize the approximation error introduced during discretization. In the TP stage, the identified key nodes serve as boundary conditions for an MPDC method based on time. This facilitates the generation of an optimal trajectory that maximizes motion performance, considering constant velocity in Cartesian space and dynamic constraints while keeping the path discretization error.
