Cable-driven serial robots have emerged with high potential for wide applications due to their compact size and low inertia properties. However, developing this type of robot encounters a motion coupling issue that the movement of one joint leads to the motion of other joints, resulting in complex control. In this paper, we proposed a novel approach for motion decoupling based on a noncircular pulley. The length change of the driving cable caused by the motion coupling problem is resolved by using the noncircular pulley. The calculation process of the profile for the noncircular pulley is illustrated in detail. An optimization process based on the brute force method is presented to identify the optimal parameters to minimize the compensation error. A cable-driven serial robot based on the decoupling method is prototyped for assessments. Experiments are conducted to evaluate the performance of the proposed motion decoupling method. The results reveal that the proposed method can effectively resolve the motion coupling issue by maintaining almost constant cable length with a maximum accumulative error only as 0.086 mm, demonstrating the effectiveness of the method. This article discusses the topic of acute abdominal pain caused by ovarian endometriosis, a common gynecologic disease. The study examined the characteristics and outcomes of patients who underwent surgical intervention for this condition. The findings suggest that certain symptoms, such as sudden onset of pain, enlargement of the adnexal mass with tenderness, and a history of dysmenorrhea requiring painkillers, are important indicators of ovarian endometriosis manifesting as acute abdominal pain. Patients with progressive ovarian endometriosis are more likely to experience severe pain, fever, and leukocytosis. The study recommends early intervention and elective surgery for stable patients, but acknowledges the limitations of its retrospective design and single-center data. Addressing the optimization problem during the simultaneous energy and information transmission stage in the simultaneous wireless information and power transfer (SWIPT), an optimal energy efficiency strategy based on mmWave cooperative communication small cell is proposed to maximize the link energy efficiency. This strategy, under the joint constraints of minimum link transmission rate and minimum collected energy, employs a power splitting mode at the receding end of energy-constrained user devices. By optimizing transmit power control and power splitting factors, it aims to maximize the systems link energy efficiency. Recognizing the original problem as a nonconvex fractional programming problem with NP-hard characteristics, the Dinkelbach method is utilized to transform the objective function into a convex optimization problem that is easier to solve. An iterative algorithm is designed to find the optimal solution through cross iterations. Simulation results demonstrate that the proposed strategy outperforms traditional power control methods and maximum transmit power approaches in optimizing system energy efficiency. Purpose: To compare bilateral lateral rectus recession (BLR) with unilateral recession and resection (RR) for divergence excess intermittent exotropia (IXT). Methods: Retrospective analysis of 66 patients with divergence excess IXT who underwent either BLR or RR from January 2013 to December 2020 was conducted. Data on demographics, pre- and postoperative deviations, fusion, stereopsis, control, and accommodative convergence/accommodation ratio were collected. Success was defined as esodeviation =5 PD (prism diopter) to exodeviation =10 PD with a follow-up time of at least 8.0 ± 2.0 weeks. Results: BLR (42 cases) and RR (24 cases) groups had the same success rate (83.3%, P = 0.688) and similar reduced postoperative deviations both in distance and at near (P > 0.05). Near-distance disparity decreased significantly in both groups (P = 0.000) with no intergroup difference (P = 0.193). Androgen receptor (AR) signaling is crucial for driving prostate cancer (PCa), the most diagnosed and the second leading cause of death in male patients with cancer in the United States. Androgen deprivation therapy is initially effective in most instances of AR-positive advanced or metastatic PCa. However, patients inevitably develop lethal castration-resistant PCa (CRPC), which is also resistant to the next-generation AR signaling inhibitors. Most CRPCs maintain AR expression, and blocking AR signaling remains a main therapeutic approach. GATA2 is a pioneer transcription factor emerging as a key therapeutic target for PCa because it promotes AR expression and activation. While directly inhibiting GATA2 transcriptional activity remains challenging, enhancing GATA2 degradation is a plausible therapeutic strategy. How GATA2 protein stability is regulated in PCa remains unknown. Here, we show that constitutive photomorphogenesis protein 1 (COP1), an E3 ubiquitin ligase, drives GATA2 ubiquitination at K419/K424 for degradation. GATA2 lacks a conserved [D/E](x)xxVP[D/E] degron but uses alternate BR1/BR2 motifs to bind COP1. By promoting GATA2 degradation, COP1 inhibits AR expression and activation and represses PCa cell and xenograft growth and castration resistance. Accordingly, GATA2 overexpression or COP1 mutations that disrupt COP1-GATA2 binding block COP1 tumor-suppressing activities. We conclude that GATA2 is a major COP1 substrate in PCa and that COP1 promotion of GATA2 degradation is a direct mechanism for regulating AR expression and activation, PCa growth, and castration resistance. Prostate cancer (PCa) is the second leading cause of cancer death in American men. Androgen receptor (AR) signaling is essential for PCa cell growth/survival and remains a key therapeutic target for lethal castration-resistant PCa (CRPC). GATA2 is a pioneer transcription factor crucial for inducing AR expression/activation. We recently reported that MAPK4, an atypical MAPK, promotes tumor progression via noncanonical activation of AKT. Here, we demonstrated that MAPK4 activated AR by enhancing GATA2 transcriptional expression and stabilizing GATA2 protein through repression of GATA2 ubiquitination/degradation. MAPK4 expression correlated with AR activation in human CRPC. Concerted activation of both GATA2/AR and AKT by MAPK4 promoted PCa cell proliferation, anchorage-independent growth, xenograft growth, and castration resistance. Conversely, knockdown of MAPK4 decreased activation of both AR and AKT and inhibited PCa cell and xenograft growth, including castration-resistant growth. Both GATA2/AR and AKT activation were necessary for MAPK4 tumor-promoting activity. Interestingly, combined overexpression of GATA2 plus a constitutively activated AKT was sufficient to drive PCa growth and castration resistance, shedding light on an alternative, MAPK4-independent tumor-promoting pathway in human PCa. We concluded that MAPK4 promotes PCa growth and castration resistance by cooperating parallel pathways of activating GATA2/AR and AKT and that MAPK4 is a novel therapeutic target in PCa, especially CRPC. RET is a transmembrane growth factor receptor. Aberrantly activated RET is found in several types of human cancer and is a target for treating RET aberration-associated cancer. Multiple clinically relevant RET protein-tyrosine kinase inhibitors (TKIs) have been identified, but how TKIs bind to RET is unknown except for vandetanib. Nintedanib is a RET TKI that inhibits the vandetanib-resistant RET(G810A) mutant. Here, we determined the X-ray co-crystal structure of RET kinase domain-nintedanib complex to 1.87 Å resolution and a RET(G810A) kinase domain crystal structure to 1.99 Å resolution. We also identified a vandetanib-resistant RET(L881V) mutation previously found in familial medullary thyroid carcinoma. Drug-sensitivity profiling of RET(L881V) revealed that it remains sensitive to nintedanib. The RET-nintedanib co-crystal structure disclosed that Leu-730 in RET engages in hydrophobic interactions with the piperazine, anilino, and phenyl groups of nintedanib, providing a structural basis for explaining that the p.L730V mutation identified in nine independently isolated cell lines resistant to nintedanib. Comparisons of RET- nintedanib, RET(G810A), and RET-vandetanib crystal structures suggested that the solvent-front Ala-810 makes hydrophobic contacts with a methyl group and aniline in nintedanib and blocks water access to two oxygen atoms of vandetanib, resulting in an energetic penalty for burying polar groups. Of note, even though the p.L881V mutation did not affect sensitivity to nintedanib, RET(L881V) was resistant to nintedanib analogs lacking a phenyl group. MAPK4 is an atypical MAPK. Currently, little is known about its physiological function and involvement in diseases, including cancer. A comprehensive analysis of 8887 gene expression profiles in The Cancer Genome Atlas (TCGA) revealed that MAPK4 overexpression correlates with decreased overall survival, with particularly marked survival effects in patients with lung adenocarcinoma, bladder cancer, low-grade glioma, and thyroid carcinoma. Interestingly, human tumor MAPK4 overexpression also correlated with phosphorylation of AKT, 4E-BP1, and p70S6K, independent of the loss of PTEN or mutation of PIK3CA. This led us to examine whether MAPK4 activates the key metabolic, prosurvival, and proliferative kinase AKT and mTORC1 signaling, independent of the canonical PI3K pathway. We found that MAPK4 activated AKT via a novel, concerted mechanism independent of PI3K. Mechanistically, MAPK4 directly bound and activated AKT by phosphorylation of the activation loop at threonine 308. It also activated mTORC2 to phosphorylate AKT at serine 473 for full activation. MAPK4 overexpression induced oncogenic outcomes, including transforming prostate epithelial cells into anchorage-independent growth, and MAPK4 knockdown inhibited cancer cell proliferation, anchorage-independent growth, and xenograft growth. • Nucleic acid-based vaccines may play a role in the pathogenesis of autoimmune diseases, rheumatic diseases, and immune-mediated diseases involving a variety of organs and systems. • Awareness to new-onset or flare-up of immune-mediated diseases in individuals receiving nucleic acid-based vaccines for COVID-19 within at least 120 days. Immune-mediated diseases (IMDs) after nucleic acid-based vaccines have been sporadically reported since their introduction during the worldwide COVID-19 crisis. Confirming their cause-effect association remains challenging. We analysed the effects of AZD1222 (ChAdOx1 nCoV-19), BNT-162b2, and/or mRNA-1273 on the development &/or deterioration of IMDs in terms of the time of clinical onsets of IMDs after exposure to these vaccines. We retrospectively reviewed 78 in-patients in Taipei Veterans General Hospital, who presented with IMDs within 120 days after receiving AZD1222, BNT-162b2, &/or mRNA-1273 vaccinations in Taiwan from May 2021 to April 2022. The duration from inoculation to development of IMD was analysed by two-tailed Kolmogorov-Smirnov (K-S) test for goodness of fit. The average time to new IMDs or flare-up of the diseases following vaccinations was 36 ± 26 days for all 91 events in these 78 patients. The onset time of IMDs after vaccinations was not haphazard as analysed by two-tailed K-S test for overall 91 events (40 new and 51 deteriorating episodes, p < 0.001). The IMDs presenting as non-connective tissue diseases (non-CTDs) have a shorter duration of incubation after vaccinations than those of CTDs (<14.7 days, 95 % confidence interval [CI], 3.0 to 26.4, p = 0.014). Furthermore, systemic vasculitis and type 2 inflammatory diseases were observed exclusively in those receiving AZD1222. AZD1222, BNT-162b2, or mRNA-1273 influence the activities of IMDs in ways yet to be explored. Protein phosphatase 5 (PP5), a serine/threonine phosphatase, has a wide range of biological functions and exhibits elevated expression in tumor cells. We previously reported that pp5-deficient mice have altered ataxia-telangiectasia mutated (ATM)- mediated signaling and function. However, this regulation was likely indirect, as ATM is not a known PP5 substrate. In the current study, we found that pp5-deficient mice are hypersensitive to genotoxic stress. This hypersensitivity was associated with the marked up-regulation of the tumor suppressor tumor protein p53 and its downstream targets cyclin-dependent kinase inhibitor 1A (p21),MDM2proto-oncogene (MDM2), and phosphatase and tensin homolog (PTEN) in pp5-deficient tissues and cells. These observations suggested that PP5 plays a role in regulating p53 stability and function. Experiments conducted with p53+/-pp5+/- or p53+/-pp5-/- mice revealed that complete loss of PP5 reduces tumorigenesis in the p53+/- mice. Biochemical analyses further revealed that PP5 directly interacts with and dephosphorylates p53 at multiple serine/threonine residues, resulting in inhibition of p53-mediated transcriptional activity. Interestingly, PP5 expression was significantly up-regulated in p53-deficient cells, and further analysis of pp5 promoter activity revealed that p53 strongly represses PP5 transcription. Background/Aims: Researches have showed that cardiac shock wave therapy (CSWT) could improve left ventricular function and attenuate LV remodeling of the ischemic heart. Apoptosis plays an important role in myocardial infarction and determines heart function and prognosis. However, it is still not clear whether CSWT is sufficient to attenuate acute myocardial infarction (AMI) induced cardiomyocyte apoptosis in vivo. In this study, we used a rat model to examine whether CSWT could attenuate cardiomyocyte apoptosis after AMI and to explore potential mechanisms. Methods: We generated an AMI rat model to investigate the function and possible regulatory mechanisms of CSWT. All rats were randomly divided into four groups: the sham-operated only group, sham-operated with SW treatment group, AMI only group, and AMI treated with SW treatment group. The rats were treated with a left anterior descending coronary artery ligation for 12h and then treated with or without CSWT (800 shots at 0.1 mJ/ mm2). Cytochrome c release was measured to analyze mitochondrial function and integrity. The apoptotic cell rate was determined by TUNEL assay. Western blot was used to analyze the cell apoptosis-, inflammation-, and survival-related signaling pathways. Results: First, the methodology of CSWT in the rat model of AMI was established. Second, CSWT attenuated the cardiomyocyte apoptosis rate in the infarct border zone. Third, CSWT suppressed the expression of apoptosis and inflammation molecules after AMI. Fourth, CSWT inhibited activation of the JNK pathway, which indicated inhibition of the cell inflammatory pathways and promotion of cardiomyocyte survival after AMI. Conclusion: These results indicate that CSWT exerts a protective effect against AMI-induced cardiomyocyte apoptosis, potentially by attenuating cytochrome c release from the mitochondria and inhibiting of the mitochondrialdependent intrinsic apoptotic pathway. Tremella fuciformis polysaccharide (TFPS), which is the extract of Tremella fuciformis Berk, has previously been demonstrated to exhibit potent anti-oxidative, anti-inflammatory and anti-aging effects. However, the mechanisms underlying these protective and therapeutic effects remain to be elucidated. The aim of the present study was to investigate the protective effects of TFPS on hydrogen peroxide-induced injury of human skin fibroblasts and to elucidate the aforementioned underlying mechanisms. A hydrogen peroxide-induced human skin fibroblast injury model was firstly established. MTT and reactive oxygen species (ROS) production assays, in addition to terminal deoxynucleotidyl transferase dUTP nick end labeling, reverse transcription-quantitative polymerase chain reaction and western blotting, were performed to investigate the protective effects of TFPS. Hydrogen peroxide decreased human skin fibroblast viability with a concurrent increase in ROS generation and cell apoptosis. Treatment with 0-400 µg/ml TFPS alone for up to 48 h did not result in alteration in cell viability. Notably, TFPS pre-treatment reduced oxidative stress and cell apoptosis in hydrogen peroxide-treated skin fibroblasts. In addition, there was profound inhibition of p16, p21, p53 and caspase-3 expression, and activation of extracellular-signal regulated kinase and Akt serine/threonine kinase 1, following TFPS pre-treatment. Furthermore, it was revealed that TFPS additionally protected fibroblasts via the upregulation of SIRT1 expression, and this was abrogated by the SIRT1 inhibitor niacinamide. These results indicated that TFPS alleviated hydrogen peroxide-induced oxidative stress and apoptosis in skin fibroblasts via upregulation of SIRT1 expression, indicating that TFPS may act as a potential therapeutic agent for oxidative-stress-associated skin diseases and aging. We report the growth of Ce-substituted yttrium iron garnet (CeYIG) thin films deposited by pulsed laser deposition on quartz substrates. CeYIG exhibits a large Faraday rotation in the near-IR region. Upon investigating the phase composition and the magnetic and magneto-optical properties of the films, we present optimal process conditions for growing CeYIG thin films on quartz substrates. A saturation magnetization (Ms) as high as 159 emu/cm was achieved by depositing a Ce Y Fe O thin film at a substrate temperature of 650C and a 2-Hz laser pulse rate. Under the same depositing conditions, CeYIG thin films annealed at 800C for 40 min provided the largest Faraday rotation (FR) angle of −6000/cm by applying an out-of-plane magnetic field. Similar to traditional wireless sensor networks (WSN), the nodes only have limited memory and energy in low-duty-cycle sensor networks (LDC-WSN). However, different from WSN, the nodes in LDC-WSN often sleep most of their time to preserve their energies. The sleeping feature causes serious data transmission delay. However, each source node that has sensed data needs to quickly disseminate its data to other nodes in the network for redundant storage. Otherwise, data would be lost due to its source node possibly being destroyed by outer forces in a harsh environment. The quick dissemination requirement produces a contradiction with the sleeping delay in the network. How to quickly disseminate all the source data to all the nodes with limited memory in the network for effective preservation is a challenging issue. In this paper, a low-latency and energy-efficient data preservation mechanism in LDC-WSN is proposed. The mechanism is totally distributed. The data can be disseminated to the network with low latency by using a revised probabilistic broadcasting mechanism, and then stored by the nodes with LT (Luby Transform) codes, which are a famous rateless erasure code. After the process of data dissemination and storage completes, some nodes may die due to being destroyed by outer forces. If a mobile sink enters the network at any time and from any place to collect the data, it can recover all of the source data by visiting a small portion of survived nodes in the network. In order to reduce the disadvantages of current biological product quality testing methods, taking the quality testing in cocoon trade markets as an example, this paper has proposed a quality nondestructive testing method for biological products based on the analysis of vibration signal from acceleration sensors. According to the wavelet transformation analysis on the random vibration signal acquired from the acceleration sensor, the random vibration signal related to the silkworm chrysalis quality has been analyzed and reconstructed; then the characteristic values such as: mean value, variance, mean square root, waveform index, pulse factor, and so on of the quality signal have been extracted also; and then the characteristic values of the quality have been optimally selected within a fuzzy clustering method; at the end, a RBF neural network testing model with characteristic values from the silkworm chrysalis quality as an input signal was built. With these procedures, this paper has established a quality nondestructive testing system for silkworm chrysalis quality based on acceleration sensor signal measurement. The results from the application experiments demonstrated the effectiveness and applicability of this quality nondestructive system for quality testing of biological products. This quality nondestructive testing system has many advantages, including shortening the testing time, avoiding sample waste from traditional testing method, increasing the accuracy and reliability, which shows many bright social and economic benefits. Stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), threatens global wheat production. Development of cultivars with increased resistance to stem rust by identification, mapping, and deployment of resistance genes is the best strategy for controlling the disease. In this study, we performed fine mapping and characterization of the all-stage stem rust resistance (Sr) gene Sr8155B1 from the durum wheat line 8155-B1. In seedling tests of biparental populations, Sr8155B1 was effective against six Chinese Pgt races tested. In a segregating population of 5060 gametes, Sr8155B1 was mapped to a 0.06-cM region flanked by markers Pku2772 and Pku43365, corresponding to 1.5- and 2.7-Mb regions in the Svevo and Chinese Spring reference genomes. Both regions include several typical nucleotide-binding leucine-rich repeat (NLR) and protein kinase genes that represent candidate genes. Among them, three NLR genes and three receptor-like protein kinases were highly polymorphic between the parental lines and their transcripts were upregulated in the homozygous resistant line TdR2 relative to its susceptible sister line TdS4. Four markers (Pku2772, Pku43365, Pku2950, and Pku3721) developed in this study, together with seedling resistance responses, correctly predicted Sr8155B1 absence or presence in 78 tetraploid wheat genotypes tested. The presence of Sr8155B1 in tetraploid wheat accessions CItr 14916, PI 197492, and PI 197493 was confirmed by mapping in three F2 populations. Aimed to solve the problem of weak anti-resonance performance and poor working stability in current anti-resonance vibrating machines, this paper presents a nonlinear dynamic model that reflects the actual working state of the anti-resonance machine. Under the material mass fluctuation condition, the dynamic response of the anti-resonance vibrating system has been discussed, and the dynamic parameters selection problem of the anti-resonance vibrating system has been analyzed which could be used to improve working performance stability. In the paper, the influence of nonlinear factors of the anti-resonance vibrating machine on the driving body and the working body has been analyzed under the material fluctuation conditions also. Wettability is an important surface property that deserves to further explore the factors on its alteration. Series of bis-N-heterocyclic quaternary ammonium salts with different spacer length and N-heterocyclic headgroups (morpholinium (BMMB, BMMD and BMMH), piperidinium (BPMH) and piperazinium (BMPMH)) have been synthesized and employed for altering the wettability of vermiculite and its derivates (Vts) treated by Liþ-saturated heating method. The results of X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TG-DTG), scanning electron microscopy (SEM) and N2 adsorption/desorption isotherms indicate that all of the bis-N-heterocyclic quaternary ammonium salts have been successfully inserted into the vermiculite layers, leading to the organic monolayer. The results of capillary rise tests combined with Lipophilic to Hydrophilic Ratio (LHR) values unveil the wettability alteration of the organo-Vts. As the layer charge decreases, the hydrophilicity of the organo-Vts gradually increases, which is probably caused by the decline in binding sites. As the result of the change in spacer length of modifier, the wetting properties of morpholinium-based organo-Vts change in order of BMMD-Vts > BMMH-Vts > BMMB-Vts, and difference in N-heterocyclic headgroups leads to the sequence of wettability: BMPMH-Vts > BPMH-Vts > BMMH-Vts. Venous return is a major determinant of cardiac output. Adjustments within the venous system are critical for maintaining venous pressure during loss in circulating volume. This article reviews two factors that are thought to enable the venous system to compensate during acute hemorrhage: 1) changes in venous elastance and 2) mobilization of unstressed blood volume into stressed blood volume. We show that mobilization of unstressed blood volume is the predominant and more effective mechanism in preserving venous pressure. The lentiviruses HIV and simian immunodeficiency virus (SIV) subvert intracellular membrane traffic as part of their replication cycle. The lentiviral Nef protein helps viruses evade innate and adaptive immune defenses by hijacking the adaptor protein 1 (AP-1) and AP-2 clathrin adaptors. We found that HIV-1 Nef and the guanosine triphosphatase Arf1 induced trimerization and activation of AP-1. Here we report the cryo-electron microscopy structures of the Nef- and Arf1-bound AP-1 trimer in the active and inactive states. A central nucleus of three Arf1 molecules organizes the trimers. We combined the open trimer with a known dimer structure and thus predicted a hexagonal assembly with inner and outer faces that bind the membranes and clathrin, respectively. Hexagons were directly visualized and the model validated by reconstituting clathrin cage assembly. Arf1 and Nef thus play interconnected roles in allosteric activation, cargo recruitment, and coat assembly, revealing an unexpectedly intricate organization of the inner AP-1 layer of the clathrin coat. The properties of MoS 2 quantum dots are mainly ruled by the edge structures and shapes. We systematically explore structural stability, shapes and magnetic properties of different MoS 2 quantum dots with different chemical potential of sulfur using the first principles. Three possible stable edge structures corresponding to different chemical potential of sulfur are confirmed. These edge structures are combined into different shapes of MoS 2 quantum dots which is changed with sulfur chemical potential. The electronic structure calculation and the partial charge analysis indicate that the MoS 2 quantum dot with armchair edge might have better catalyze ability. These MoS 2 quantum dots show different magnetic behavior, which can adapt to different needs of spin devices. Given the recent achievement of graphene quantum dot structures and MoS 2 quantum dots experimental study, we believe that our calculated results are suitable for experimental verification and can be useful to preparing different shapes of MoS 2 quantum dots. This study examines the possible effect of the record ice jam of 1984 in the St. Clair River on river bed changes and conveyance. Numerical simulations were made to examine the flow and bed shear stresses during the jam formation and release periods. Simulation results indicate that the ice jam in the river did not cause a significant increase in bed shear stress compared to pre- and post-jam open water conditions. The insignificant change on bed shear stress during the ice jam period is the result of the large flow depth and the limited jam thickness. The bed shear stresses are much less than the critical shear stress for bed particle movement. This implies that the jam may not have had a significant impact on the channel conveyance. Improved regulation of the wintertime flow from Lake Superior is needed to improve the balance of water levels of Lake Superior and Lake Michigan-Huron to decrease the frequency of extreme levels without unduly affecting Lake Superior interest. The wintertime outflow limit is set as 2410 m³/s by Lake Superior Regulation Plan 1977-A as a result of ice jam flooding during the 1916-1917 winter. This paper presents a numerical model study on the ice conditions in the St. Marys River to assess the maximum allowable Lake Superior wintertime outflow. Freeze-up, frazil transport and accumulation, and breakup were simulated with a thermal-ice dynamic model. The highest potential for flooding exists during ice breakup and simulations were run to determine a safe discharge limit for the breakup period. First principles calculations have been performed to investigate the structure, electronic and optical properties of Y3Fe5O12. Both the cubic and trigonal phases have been considered in our calculation. The calculated structural parameters are slightly larger than the experimental values. The band structures show that Y3Fe5O12 in cubic and trigonal phases have direct band gaps of 0.65 and 0.17 eV. The genetic defects underlying approximately half of all retinitis pigmentosa (RP) cases are unknown. A number of genes responsible for Leber congenital amaurosis (LCA) may also cause RP when they are mutated. Our previous study revealed that variants in the most frequently mutated nine exons accounted for approximately half of the mutations detected in a cohort of patients with LCA. The aim of the present study was to detect mutations in LCA-associated genes in patients with RP using two different strategies. Sanger sequencing was used to screen mutations in the nine exons in 293 patients with RP and exome sequencing was used to detect variants in 12 LCA-associated genes in 157 of the 293 patients with RP and then to validate the variants by Sanger sequencing. Potential pathogenic mutations were identified in four patients with early onset RP, including homozygous CRB1 mutations in two patients, compound heterozygous CRB1 mutations in one patient and compound heterozygous CEP290 mutations in one patient. The present study indicated that mutations in CEP290 may also be associated with RP but not with LCA. Castration-resistant prostate cancer (CRPC) poses a major clinical challenge with the androgen receptor (AR) remaining to be a critical oncogenic player. Several lines of evidence indicate that AR induces a distinct transcriptional program after androgen deprivation in CRPCs. However, the mechanism triggering AR binding to a distinct set of genomic loci in CRPC and how it promotes CRPC development remain unclear. We demonstrate here that atypical ubiquitination of AR mediated by an E3 ubiquitin ligase TRAF4 plays an important role in this process. TRAF4 is highly expressed in CRPCs and promotes CRPC development. It mediates K27-linked ubiquitination at the C-terminal tail of AR and increases its association with the pioneer factor FOXA1. Consequently, AR binds to a distinct set of genomic loci enriched with FOXA1- and HOXB13-binding motifs to drive different transcriptional programs including an olfactory transduction pathway. Through the surprising upregulation of olfactory receptor gene transcription, TRAF4 increases intracellular cAMP levels and boosts E2F transcription factor activity to promote cell proliferation under androgen deprivation conditions. Altogether, these findings reveal a posttranslational mechanism driving AR-regulated transcriptional reprogramming to provide survival advantages for prostate cancer cells under castration conditions. Background The measurement of hepatitis C virus core antigen (HCV-coreAg) has been shown to be an indicator of active HCV infection. The aim of the present study was 1) to investigate the stability and effectiveness of HCV-coreAg and HCV-RNA quantification in HCV infection with or without HIV-1 coinfection, 2) to explore the association between the HCV-coreAg/ HCV-RNA (Ag/RNA) ratio and the immune status in chronic HCV/HIV-1- coinfected patients. Methods A longitudinal investigation comprised of 227 HCV-monoinfected (n = 129) and HCV/HIV- 1-coinfected (n = 98) patients was initiated in August 2009, and 139 (73 with HCV monoinfection and 66 with HCV/HIV-1 coinfection) were followed up in August 2012. Both HCV core antigen and HCV RNA quantification were determined on this cryopreserved plasma. HCV core antigen and HCV RNA quantification were performed subsequently. In addition, an in vitro experiment investigating the possibility of degradation of HCV components (core antigen and RNA) were conducted. Results Significant and stable correlations (p < 0.001) were observed both in chronic HCV-monoinfected and HCV/HIV-1-coinfected patients over the 3-year observation. Coinfected patients with immunocompromised condition had a significantly higher (p < 0.05) HCVcoreAg/ HCV-RNA ratios than those patients with immunocompetent condition both at two time points (2009 and 2012). Moreover, the Ag/RNA ratios were negatively correlated with CD4+ T-cell counts (p < 0.001). An in vitro experiment investigated the possibility of the slower degradation of HCV particles under HIV-related immunocompromised condition was conducted and the data demonstrated that the Ag/RNA ratios were significantly higher in HIV-1-positive plasma than in healthy plasma (p = 0.005) in this study. Conclusions Our longitudinal study indicated that the HCV-coreAg presented comparable dynamics over time as HCV RNA in chronic HCV-infected patients. The scission of biological membranes is facilitated by a variety of protein complexes that bind and manipulate lipid bilayers. ESCRT-III (endosomal sorting complex required for transport III) filaments mediate membrane scission during the ostensibly disparate processes of multivesicular endosome biogenesis, cytokinesis, and retroviral budding. However, mechanisms by which ESCRT-III subunits assemble into a polymer remain unknown. Using cryogenic electron microscopy (cryo-EM), we found that the full-length ESCRT-III subunit Vps32/CHMP4B spontaneously forms single-stranded spiral filaments. The resolution afforded by two-dimensional cryo-EM combined with molecular dynamics simulations revealed that individual Vps32/CHMP4B monomers within a filament are flexible and able to accommodate a range of bending angles. In contrast, the interface between monomers is stable and refractory to changes in conformation. We additionally found that the carboxyl terminus of Vps32/CHMP4B plays a key role in restricting the lateral association of filaments. A dynamic model of gear transmission system of wind turbine is built with consideration of randomness of loads and parameters. The dynamic response of the system is obtained using the theory of random sampling and the Runge-Kutta method. According to rain flow counting principle, the dynamic meshing forces are converted into a series of luffing fatigue load spectra. The amplitude and frequency of the equivalent stress are obtained using equivalent method of Geber quadratic curve. Moreover, the dynamic reliabilitymodel of components and systemis built according to the theory of probability of cumulative fatigue damage. The system reliability with the random variation of parameters is calculated and the influence of random parameters on dynamic reliability of components is analyzed. In the end, the results of the proposed method are compared with that of Monte Carlo method. A numerical study on the effectiveness of the proposed ice boom to be installed near the entrance of Lake Notoro, Hokkaido, Japan to prevent sea ice moving into the lake is presented. A two-dimensional hydro-ice dynamics model was modified to allow for the treatment of ice-boom interaction with the effect of tidal current. The numerical model is a coupled hydrodynamic and ice dynamic model. The ice dynamic component uses a Lagrangian discrete parcel method based on smoothed particle hydrodynamics. The model was used to assess the dynamics of sea-ice transport into the lake, the effectiveness of the boom on ice retention, and the ice load on boom spans. The microtubule (MT) and actin cytoskeletons drive many essential cellular processes, yet fairly little is known about how their functions are coordinated. One factor that mediates important cross talk between these two systems is WHAMM, a Golgi-associated protein that utilizes MT binding and actin nucleation activities to promote membrane tubulation during intracellular transport. Using cryoelectron microscopy and other biophysical and biochemical approaches, we unveil the underlying mechanisms for how these activities are coordinated. We find that WHAMM bound to the outer surface of MT protofilaments via a novel interaction between its central coiled-coil region and tubulin heterodimers. Upon the assembly of WHAMM onto MTs, its N-terminal membrane-binding domain was exposed at the MT periphery, where it can recruit vesicles and remodel them into tubular structures. In contrast, MT binding masked the C-terminal portion of WHAMM and prevented it from promoting actin nucleation. Neurotransmitters are released through nascent fusion pores, which ordinarily dilate after bilayer fusion, preventing consistent biochemical studies. We used lipid bilayer nanodiscs as fusion partners; their rigid protein framework prevents dilation and reveals properties of the fusion pore induced by SNARE (soluble N-ethylmaleimide--sensitive factor attachment protein receptor). We found that although only one SNARE per nanodisc is required for maximum rates of bilayer fusion, efficient release of content on the physiologically relevant time scale of synaptic transmission apparently requires three or more SNARE complexes (SNAREpins) and the native transmembrane domain of vesicle-associated membrane protein 2 (VAMP2). We suggest that several SNAREpins simultaneously zippering their SNARE transmembrane helices within the freshly fused bilayers provide a radial force that prevents the nascent pore from resealing during synchronous neurotransmitter release. Human mutations in or variants of TBX20 are associated with congenital heart disease, cardiomyopathy, and arrhythmias. To investigate whether cardiac disease in patients with these conditions results from an embryonic or ongoing requirement for Tbx20 in myocardium, we ablated Tbx20 specifically in adult cardiomyocytes in mice. This ablation resulted in the onset of severe cardiomyopathy accompanied by arrhythmias, with death ensuing within 1 to 2 weeks of Tbx20 ablation. Accounting for this dramatic phenotype, we identified molecular signatures that posit Tbx20 as a central integrator of a genetic program that maintains cardiomyocyte function in the adult heart. Expression of a number of genes encoding critical transcription factors, ion channels, and cytoskeletal/myofibrillar proteins was downregulated consequent to loss of Tbx20. Genome-wide ChIP analysis of Tbx20-binding regions in the adult heart revealed that many of these genes were direct downstream targets of Tbx20 and uncovered a previously undescribed DNA-binding site for Tbx20. Bioinformatics and in vivo functional analyses revealed a cohort of transcription factors that, working with Tbx20, integrated multiple environmental signals to maintain ion channel gene expression in the adult heart. Development of HCV infection is typically followed by chronic hepatitis C (CHC) in most patients, while spontaneous HCV viral clearance (SVC) occurs in only a minority of subjects. Compared with the widespread application of HCV RNA testing by quantitative RT-PCR technique, HCV core antigen detection may be an alternative indicator in the diagnosis of hepatitis C virus infections and in monitoring the status of infectious individuals. However, the correlation and differences between these two indicators in HCV infection need more investigation, especially in patients coinfected by HIV-1. In this study, a total of 354 anti-HCV and/or anti-HIV serum positive residents from a village of central China were enrolled. Besides HCVrelated hepatopathic variables including clinical status, ALT, AST, anti-HCV Abs, as well as the altered CD4+/CD8+ T cell counts, HCV core antigen and HCV viral load were also measured. The concentration of serum HCV core antigen was highly correlated with level of HCV RNA in CHC patients with or without HIV-1 coinfection. Of note, HCV core antigen concentration was negatively correlated with CD4+ T cell count, while no correlation was found between HCV RNA level and CD4+ T cell count. Our findings suggested that quantitative detection of plasma HCV core antigen may be an alternative indicator of HCV RNA qPCR assay when evaluating the association between HCV replication and host immune status in HCV/HIV-1 coinfected patients. Mutations in full-length HBV isolates obtained from a chronic HBV-infected patient were evaluated at three time points: 1 day, 6 months, and 31 months.While 5 nucleotides variation, and an 18 bp deletion of preS1 have been kept in during at least the first two years, C339T mutation occurring in the hydrophilic region of HBsAg and T770C that caused polymerase V560A substitution were the new point mutations found existing in sequenced clones of the 3rd time point. Internal deletion of coding region obviously appeared in the 3rd time point. The splicers included two new 5'-splice donors and three new 3'-splice acceptors besides the reported donors and acceptors and may have produced presumptive HBV-spliced proteins or truncated preS proteins. ALT, HBeAg and viral DNA load varied during the follow-up years. These data demonstrated the diversity of genomes in HBV-infected patient during evolution. Background: Both regulatory T cells (Tregs) and PD-1/PD-L1 pathway were critically involved in HCV viral persistence. However, the association between them was not well investigated. Herein, we aimed to investigate the distributional profiles of Tregs subsets and association between PD-1 expression on these subsets and development of HCV long-term persistence. Methods: CD45RA and CD27 were employed to separate peripheral Tregs as naïve/central memory/effector memory/effector subsets. The phenotypic characteristics and PD-1 expression of Tregs were studied by flow cytometry. Results: In the present study, the majority of Tregs was identified as central memory phenotype in chronic hepatitis C patients compared with nearly equal contribution of naïve and central memory subsets in healthy individuals. PD-1 expression was elevated in all CD4+ T cell subset in chronic HCV infected patients, including Tregs. Of note, higher level of PD-1 expression was found on TEM- and effector-Treg than naïve- and TCM-Tregs subsets. The ratio of TEM-Tregs/naive-Tregs and TEM-Tregs/TCM-Tregs regarding to PD-1 MFI were significantly lower in CHC patients compared to controls. Conclusions: Our study indicated that distinctive characteristics of PD-1 expression on Tregs in HCV infection suggests associated with impaired adaptive immunity as well as viral long-term persistence. Attenuated CD4+ T-cell-mediated immune responses are involved in persistence of HCV infection, but the mechanism remains undefined. In this study, the proportions of CD4+ T cell subsets, naïve, central memory, effector memory and effector cells, along with CD25 (IL-2Rα) and CD127 (IL-7Rα) expression on different CD4+ T cell subsets, were measured by polychromatic flow cytometry in 24 chronic HCV-infected individuals and 21 healthy controls. A significant decrease in naïve CD4+ T cells and an increase of central memory and effector memory CD4+ T cells were found in HCV-infected patients compared with healthy controls. HCV-infected patients showed a lower level of CD127 expression in all CD4+ T cells subsets, especially in central memory and effector CD4+ T cells. In terms of total CD4+ T cells, an increase in CD25+ regular T cells (CD4+ CD25+ CD127lo) was found in HCV-infected patients. Interestingly, naïve CD4+ T cells showed increased CD25 expression, while effector memory and effector CD4+ T cells had lower CD25 expression. These data indicated that variations in different fractions of CD4+ T cells, including the phenotypic profile and expression level of CD25 and CD127, may be associated with low efficiency of immune response in chronic HCV infection. Background: Tight correlation between host circulating CD8+ T cell-mediated immune response and control of viral replication is classical characteristic of long-term HCV infection. CD8+ T cell maturation/activation markers are expected to be associated with viral replication and disease progression in chronic HCV infection. The aim of the present study was to explore novel markers on CD8+ T cells with ability to evaluate HCV viral replication and disease progression. Methods: PBMCs were isolated from 37 chronic HCV-infected patients and 17 healthy controls. Distributed pattern of CD8+ T cells subsets and expression of PD-1, CD38, HLA-DR and CD127 were analyzed by flow cytometry. The correlation between expression of surface markers and HCV viral load or ALT was studied. Results: Declined naïve and increased TEMRA CD8+ T subsets were found in HCV-infected individuals compared with healthy controls. Percentage and MFI of PD-1, CD38 and HLA-DR on all CD8+ T cell subsets were higher in HCV-infected patients than healthy controls. In contrast, CD127 expression on CD8+ TCM showed an opposite trend as PD-1, CD38 and HLA-DR did. In chronic HCV infection, MFI of PD-1 on CD8+ TEM (p < 0.0001) and TEMRA (p = 0.0015) was positively correlated with HCV viral load while HLA-DR expression on non-naive CD8+ T cell subsets (p < 0.05) was negatively correlated with HCV viral load. Conclusion: PD-1 level on peripheral CD8+ TEM/TEMRA was highly correlated with HCV viral load in chronic HCV-infected patients, which made PD-1 a novel indicator to evaluate HCV replication and disease progression in chronic hepatitis C patients. Hepatitis B virus (HBV) infection is highly prevalent in China. To identify the genotypes of HBV in the southern Yunnan Province of China, full-length HBV genomes were extracted from 1 Dai and 4 Hani HBV carriers and linked with the pMD T-18 vector. For each patient, 3–10 clones were sequenced directly and a consensus sequence was created. Genotypic and serotypic analysis revealed 4 HBV/B (2 B2 with adw2 and 2 new subgenotypes with ayw1) and 1 HBV/C (C1 with adrq+) genotypes. The divergences of the entire genome sequences of the new subgenotype were 0–0.9% and 2.99–6.48% between other known HBV/B. Divergences in other coding regions revealed that it was more similar to B3 and B4 in the precore/core gene (2.02 and 2.09%, respectively), and similar to B3 and B5 in the preS1/S2/S gene (2.24 and 2.78%, respectively). Phylogenetic trees using the precore/core and X genes both revealed a new clad separating from the major trunk of genotype B with a 99% bootstrap value. These results show that the 2 consensus isolates are a mosaic of B3–B5, which we designated to subgenotype B6. Considering the geographical distances, the relationship between B6 and other HBV/B subgenotypes (B3–B5) and HBV evolution needs to be further studied. Copyright © 2009 S. The macrocyclic compounds consisting of photosensitive units as parts of the frame have been extensively studied to mimic photoregulated functions in nature. In this paper, controlled assembly of well- ordered arrays of photosensitive macrocyclic rectangles is demonstrated by using a host-guest molecular template. 4NN-Macrocycle molecules are observed to photoisomerize from trans-trans-trans-trans (t,t,t,t) to a range of isomers including trans-trans-trans-cis (t,t,t,c) and trans-cis-trans-cis (t,c,t,c) isomers after irradiation of UV light. The photoisomers are also observed to affect the guest-host network characteristic appreciably. In the STM observations we can distinguish three (t,t,t,t) conformational isomers, three (t,t,t,c) conformational isomers, and one (t,c,t,c) isomer, which self-assemble into different adlayers with TCDB on a HOPG surface. This study examined the joint impact of experiencing both interparental violence and child physical maltreatment on young adults' self-esteem. It also tested the hypothesis of parental and peer relationship qualities as mediators in the relationship between childhood histories of family violence and adult self-esteem. Data were collected from a national probability sample of 1,924 college students in Taiwan. Research results demonstrated that experiencing both interparental violence and physical maltreatment during childhood have long-term and detrimental impact on adult self-esteem. This impact was statistically independent of other potential confounding factors. Moreover, participants experiencing dual violence during childhood reported lower self-esteem than those experiencing only one type of family violence or none at all. Male participants who experienced dual violence reported lower self-esteem than female participants who experienced dual violence. Further analyses revealed that parental and peer relationship qualities mediated the joint impact of interparental violence and physical maltreatment on adult self-esteem. [ABSTRACT FROM AUTHOR]

Recently, video clips have become very popular online. The massive influx of video clips has created an urgent need for video search engines to facilitate retrieving relevant clips. Different from traditional long videos, a video clip is a short video often expressing a moment of significance. Due to the high complexity of video data, efficient video clip search from large databases turns out to be very challenging. We propose a novel video clip representation model called the Bounded Coordinate System (BCS), which is the first single representative capturing the dominating content and content—changing trends of a video clip. It summarizes a video clip by a coordinate system, where each of its coordinate axes is identified by principal component analysis (PCA) and bounded by the range of data projections along the axis. The similarity measure of BCS considers the operations of translation, rotation, and scaling for coordinate system matching. Particularly, rotation and scaling reflect the difference of content tendencies. Compared with the quadratic time complexity of existing methods, the time complexity of measuring BCS similarity is linear. The compact video representation together with its linear similarity measure makes real-time search from video clip collections feasible. To further improve the retrieval efficiency for large video databases, a two-dimensional transformation method called Bidistance Transformation (BDT) is introduced to utilize a pair of optimal reference points with respect to bidirectional axes in BCS. Our extensive performance study on a large database of more than 30,000 video clips demonstrates that BCS achieves very high search accuracy according to human judgment. This indicates that content tendencies are important in determining the meanings of video clips and confirms that BCS can capture the inherent moment of video clip to some extent that better resembles human perception. A new 3014→13abeodammarane triterpenoid, myrrhasin 1, a new 1410→1abeoeudesmane sesquiterpenoid, myrrhanolide A 2, and two new cadinane sesquiterpenoids, myrrhanolides B and C 3and 4, resp., as well as nine known sesquiterpenoids, 5–13, were isolated from the resinous exudates of Commiphora myrrha.Their structures were determined on the basis of extensive MS and NMR spectroscopic analyses.
