Background: Continuous cropping of the same crop leads to land degradation. This is also called the continuous-cropping obstacle. Currently, intercropping tobacco with other crops can serve as an effective strategy to alleviate continuous cropping obstacles. Results: In this study, tobacco K326 and insectary floral plants were used as materials, and seven treatments of tobacco monoculture (CK), tobacco intercropped with Tagetes erecta, Vicia villosa, Fagopyrum esculentum, Lobularia maritima, Trifolium repens, and Argyranthemum frutescens respectively, were set up to study their effects on rhizosphere soil chemical properties and composition and structure of rhizosphere soil microbial community of tobacco. The 16 S rRNA gene and ITS amplicons were sequenced using Illumina high-throughput sequencing. tobacco/insectary floral plants intercropping can influence rhizosphere soil chemical properties, which also change rhizosphere microbial communities. The CK and treatment groups tobacco rhizosphere soil microorganisms had significantly different genera, such as tobacco intercropping with T. repens and A. frutescens significantly increased the number of Fusarium and intercropping T. erecta, V. villosa, L. maritima, T. repens, and A. frutescens significantly increased the number of Sphingomonas and unknown Gemmatimonadaceae. Additionally, intercropping T. erecta, V. villosa and L. maritima changed the rhizosphere fungal and bacteria community and composition of tobacco and the positive correlation between tobacco rhizosphere the genera of fungi and bacterial were greater than CK. The pathway of the carbohydrate metabolism, amino acid metabolism, and energy metabolism in rhizosphere bacteria were significantly decreased after continuous cropping. Fungal symbiotic trophic and saprophytic trophic were significantly increased after intercropping V. villosa, L. maritima and plant pathogen and animal pathogen were increased after intercropping T. repens and A. frutescens. Additionally, bacterial and fungal communities significantly correlated with soil chemical properties, respectively. Conclusion: This study reveals that intercropping tobacco with insectary floral plants, particularly T. erecta, V. villosa, L. maritima and A. frutescens significantly affects soil chemical properties and alters rhizosphere microbial communities, increasing the abundance of certain microbial genera. Additionally, intercropping enhances pathways related to carbohydrate, amino acid, and energy metabolism in rhizosphere bacteria. These findings suggest that intercropping could provide a promising strategy to overcome challenges associated with continuous tobacco cropping by regulating the rhizosphere environment. Tobacco bacterial wilt is a major limiting factor for tobacco production and development, and it is more likely to occur under perennial single cropping of tobacco. In recent years, the rotation of tobacco-radish has gradually become popular. Therefore, we studied the effects of years of tobacco–radish rotation on tobacco bacterial wilt occurrence and rhizosphere microorganisms. The results indicated that both SY and TY could significantly reduce the risk of tobacco bacterial wilt occurrence, and SY had the lowest disease index. The rotation of radish plants significantly increased the soil pH but decreased the contents of alkali-hydrolysed nitrogen and organic matter in the soil. Alkali-hydrolysed nitrogen and pH are the key factors affecting the composition of the bacterial community. Furthermore, radish rotation changed the composition of the soil microbial community, increased the diversity of the bacterial community, and significantly altered the bacterial community structure. At the genus level, the abundance of Sphingomonas species negatively correlated with Ralstonia increased significantly, while the relative abundance of Rhodanobacter species positively correlated with Ralstonia decreased significantly. Disease index, pH and available phosphorus were the main factors affecting the variation in different bacterial genera. The network analysis results showed that Ralstonia was less connected in the network than in the CK group, and the SY treatment group had a more complex bacterial network structure. Overall, 2 years of tobacco and radish rotation improved the bacterial community structure of the rhizosphere soil and alleviated the harm caused by tobacco bacterial wilt, which is highly important for the stability and health of the rhizosphere soil ecosystem. Long-term continuous cropping of tobacco causes dysbiosis of soil microbial communities, the imbalance of soil nutrients, and the increase of pathogenic bacteria, which will slow the growth and development of tobacco plants, reduce the production quality, and cause significant losses to tobacco production and tobacco farmers. The application of Potassium fulvic acid can not only provide nutrients, but also inhibit the propagation of pathogens in soil along with raising the amount of organic matter in the soil, which is an effective way to improve soil health. In this experiment, Tobacco variety SNT60 was used as the test material, and 6 treatments were set up by pot test, they were: no fertilisation control group (CK), tobacco special fertiliser (NPK), 3.45 g/kg of potassium fulvic acid fertiliser (T1), 4.65 g/kg of potassium fulvic acid fertiliser (T2), 5.85 g/kg of potassium fulvic acid fertiliser (T3), 7.05 g/kg of potassium fulvic acid fertiliser (T4), Ten replications were set up for each treatment and the soil and fertiliser were mixed and potted before transplanting, 70% as basal fertiliser and 30% as supplementary fertiliser. We also analyzed soil properties, soil microorganisms and agronomic traits of tobacco plants in different treatments to provide reference for mitigating tobacco succession barrier. The test results are as follows: 4.65 g/kg of potassium fulvic acid fertiliser (T2) treatment was the best, soil organic matter, quick nitrogen, phosphorus, potassium, pH, soil catalase, soil sucrase, and soil urease content, compared to CK control, increased by 22.04%, 43.12%, 96.21%, 381.79%, 25.43%, 91.69%, 262.07% and 93.16%. In terms of microbial community, application of potassium fulvic acid fertiliser significantly increased the relative abundance of Ascomycetes, Chlorobacterium, Bacillus, Proteobacteria and Tephritobacterium in the soil. Meanwhile, 4.65 g/kg of potassium fulvic acid fertiliser (T2) promoted the growth of tobacco plants, improved leaf photosynthetic capacity, and enhanced plant disease resistance. Most research on plant–microbe interactions emphasize the effects of micronutrients on the rhizosphere microbial community structure. However, the influence of seed structures, particularly the radicle sheath, on microbial diversity at the seedling root tips under varying temperatures and humidity has been less explored. This study conducted controlled indoor experiments in the northern desert of Xinjiang to assess the radicle sheath's impact on microbial community composition, diversity, and function. The results indicated no significant changes in the Chao1 index for bacteria and fungi, but notable differences were observed in the Shannon and Simpson indices (p < 0.05). Under drought conditions, the radicle sheath significantly reduced bacterial infections without affecting fungi. Genus-level analysis showed an increased abundance of specific dominant bacterial groups when the radicle sheath was retained. NMDS analysis confirmed its significant effect on both bacterial and fungal community structures. LEfSe analysis identified 34 bacterial and 15 fungal biomarkers, highlighting the treatment's impacts on microbial taxonomic composition. Functional predictions using PICRUSt 2 revealed that the radicle sheath facilitated the conversion of CH4 to CH3OH and various nitrogen cycle processes under drought. Overall, the radicle sheath plays a crucial role in maintaining rhizosphere microbial community stability and enhancing the functions of both bacteria and fungi under drought conditions. Phosphorus (P) is crucial for tree growth and development, and it significantly influences the rhizosphere microbial community. However, the effects of phosphorus addition on the microbial communities in the rhizosphere soil of Idesia polycarpa remain understudied. In this study, two-year-old "Yuji" Idesia polycarpa seedlings were used to investigate the effects of phosphorus fertilization at four different levels of 0 g (control, CK), 0.92 g (low phosphorus, LP), 1.83 g (medium phosphorus, MP), and 2.75 g (high phosphorus, HP) per plant. The fertilizers were applied every 40 days over 120 days. MiSeq high-throughput sequencing of the 16S rRNA and ITS1 genes was employed to analyze the microbial community composition and diversity of bacteria and fungi in the rhizosphere soil under different phosphorus levels. The results showed that compared with CK treatment, the application of phosphorus fertilizer changed the physicochemical properties of the soil. The LP treatment significantly increased the soil pH, while the HP treatment group exhibited the highest soil-available phosphorus (AP) content. LP treatment significantly increased the number of microbial OTUs in the early and rapid growth stages and the richness and diversity of microbial communities. In addition, the bacterial community structure was significantly correlated with soil pH and AP, while the fungal community had no significant effect. The primary metabolic pathway function of bacteria in the rhizosphere soil of Idesia polycarpa seedlings is mainly metabolism, while fungi are mainly biosynthesis. Compared with CK treatment, 20 differential metabolic pathways were screened out in the bacterial community. Only two differential metabolic pathways were screened out in the fungal community by LP treatment at 120 days. In summary, applying low-level phosphate fertilizer is conducive to promoting the diversity of rhizosphere soil microorganisms. BACKGROUND: Aphis gossypii Glover is a prevalent phytophagous insect that inflicts significant damage on cucumber plants. Recent studies have provided insights into plant communication and signal transduction within conspecifics. However, understanding of the effect of these communication mechanisms on adjacent cucumbers and their resident aphids, especially in the context of an aphid infestation, is still in its early stages. RESULTS: Utilizing a partitioned root configuration, a tendency for aphids to gather on nearby cucumber leaves of non‐infested plants was observed. Furthermore, neighboring plants near aphid‐infested cucumber plants showed a reduction in aphid reproduction rates. Concurrently, these plants exhibited a significant increase in reactive oxygen species (ROS) levels, along with enhanced defensive and antioxidant enzymatic responses. Analysis of the microbial community in the rhizosphere showed significant differences in species composition among the samples. Among these, the bacterial families Microbacteriaceae and Rhizobiaceae, along with the fungal species Leucocoprinus ianthinus and Mortierella globalpina, exhibited increases in their relative abundance in cucumber seedlings located near aphid‐infested plants. Significantly, this study unveiled robust correlations between dominant microbial phyla and physiological indicators, primarily associated with aphid resistance mechanisms in plants. CONCLUSION: The results show that aphid‐infested cucumber plants trigger oxidative stress responses in adjacent seedlings through complex interplant communication mechanisms. In addition, these plants cause changes in the composition of the rhizospheric microbial community and the physiological activity of neighboring plants, consequently boosting their natural resistance to aphids. Introduction: Microorganisms exhibit intricate interconnections with tea plants; however, despite the well-established role of microorganisms in crop growth and development, research on microbes within the tea plant remains insufficient, particularly regarding endophytic microorganisms. Methods: In this study, we collected samples of leaves and rhizosphere soils from 'Zhuyeqi', 'Baojing Huangjincha#1', 'Baiye#1', and 'Jinxuan' varieties planted. Results: Our analyses revealed significant variations in tea polyphenol contents among tea varieties, particularly with the 'Zhuyeqi' variety exhibiting higher levels of tea polyphenols (>20% contents). Microbiome studies have revealed that endophytic microbial community in tea plants exhibited higher host specificity compared to rhizospheric microbial community. Analyses of across-ecological niches of the microbial community associated with tea plants revealed that soil bacteria serve as a significant reservoir for endophytic bacteria in tea plants, Bacillus may play a crucial role in shaping the bacterial community acrossecological niche within the tea plants with higher tea polyphenol levels. In the aforementioned analyses, the microbial community of 'Zhuyeqi' exhibited a higher degree of host specificity for leaf endophytic microorganisms, the topological structure of the co-occurrence network is also more intricate, harboring a greater number of potential core microorganisms within its nodes. A closer examination was conducted on the microbial community of 'Zhuyeqi', further analyses of its endophytic bacteria indicated that its endophytic microbial community harbored a greater abundance of biomarkers, particularly among bacteria, and the enriched Methylobacterium and Sphingomonas in 'Zhuyeqi' may play distinct roles in disease resistance and drought resilience in tea plants. Conclusion: In summary, this study has shed light on the intricate relationships of tea plant varieties with their associated microbial communities, unveiling the importance of microorganisms and tea varieties with higher tea polyphenols, and offering valuable insights to the study of microorganisms and tea plants. In agricultural production, it is crucial to increase the availability of phosphorus (P) in cultivated soil to solve the P limitation. Arbuscular mycorrhizal fungi (AMF) have been proven to promote crop nutrient absorption effectively, while biochar can lead to improvements in soil properties. However, the possible synergistic effect of AMF and biochar on P uptake by crops as well as its underlying mechanisms are unclear. In this study, we conducted a pot experiment to explore the effects of biochar and AMF (Glomus etunicatum) on the community of rhizospheric phosphate-solubilizing microorganisms (PSMs) of maize (Zea mays L. Xianyu-335) using metagenomic methods. The experiment used 0 mg P2O5 g·kg−1 soil (P0) and 30 mg P2O5 g·kg−1 soil (P30) application rates. Each P application rate included 0 (NC), 20 g·kg−1 biochar (BC) addition, inoculation AMF, and without AMF treatments (NM) for a total of eight treatments. During the experiment, both the P uptake and the biomass of maize were measured. The study found that the combination of AMF and biochar significantly increased the mycorrhizal colonization rate of maize roots, regardless of P application level. It was observed that the P uptake by maize was significantly increased when exposed to a combination of AMF and biochar. The increase in P uptake in P0 treatments was 67% higher than the sum of the effects of biochar and AMF inoculation alone. The increase was only 35% higher in P30 treatments, demonstrating a substantially higher interactive effect under P0 than under P30 conditions. The AM-BC treatments significantly increased the abundance of Streptomyces, Bacillus, and Pseudomonas, genera that are known to contain PSMs. In addition, the abundance of genes related to P-cycling (gcd, phoD, and ugpQ) in PSMs increased significantly by 1.5–1.8 times in AM-BC treatments compared with NM-BC and AM-NC treatments under P0 conditions. This increase was significantly and positively correlated with the P uptake. Overall, the results suggest that biochar can help AMF colonize the roots, increasing the functional roles of PSMs in the rhizosphere, which in turn promotes P uptake and biomass in maize. In the present study, experiments were conducted to assess the influence of nanoscale sulfur in the microbial community structure of metallophytes in Hg-contaminated rhizosphere soil for planting rapeseed. The results showed that the richness and diversity of the rhizobacteria community decreased significantly under Hg stress, but increased slightly after SNPs addition, with a reduction in the loss of Hg-sensitive microorganisms. Moreover, all changes in the relative abundances of the top ten phyla influenced by Hg treatment were reverted when subjected to Hg + SNPs treatment, except for Myxococcota and Bacteroidota. Similarly, the top five genera, whose relative abundance decreased the most under Hg alone compared to CK, increased by 19.05%–54.66% under Hg + SNPs treatment compared with Hg alone. Furthermore, the relative abundance of Sphingomonas, as one of the dominant genera for both CK and Hg + SNPs treatment, was actively correlated with plant growth. Rhizobacteria, like Pedobacter and Massilia, were significantly decreased under Hg + SNPs and were positively linked to Hg accumulation in plants. This study suggested that SNPs could create a healthier soil microecological environment by reversing the effect of Hg on the relative abundance of microorganisms, thereby assisting microorganisms to remediate heavy metal-contaminated soil and reduce the stress of heavy metals on plants. In this manuscript, we first comprehensively investigated the changes in the rhizosphere microbial community structure of metallophytes in Hg-contaminated soil with SNPs addition, as well as the relationship between soil microbiology and plant resistance to Hg stress. Our results demonstrated that SNPs exhibit a significant advantage in improving rhizosphere microecology by increasing the abundance of beneficial rhizobacteria, thereby alleviating heavy metal toxicity, and promoting plant growth. This study is the first study describing the response of soil microorganisms coexposed to heavy metals and SNPs, providing valuable information for the potential use of SNPs to assist phytoremediation of toxic metal pollution and its impact on soil microbial communities. Subsurface flow garden constructed wetland (SFGCW) is a type of constructed wetland with garden characteristics. This study explores the efficiency of SFGCW in the removal of ammonia nitrogen (NH4+-N) and total nitrogen (TN) from domestic sewage and examines the structure of the rhizosphere microbial community and prevalence of nitrifying and denitrifying bacteria. An L4(23) orthogonal experiment was conducted using different factors such as substrates, plants, and hydraulic retention times (HRT). The results of range and variance analyses revealed that HRT and substrate considerably influenced nitrogen removal by SFGCWs, with plant factors playing a notable role. The use of fluidized bed slag as the substrate enhanced nitrogen removal, particularly when HRT was set at 3 or 6 days. At the phylum level, Proteobacteria predominated the rhizosphere microbial abundance, comprising 42.58%–55.38% of the microbial population, followed by Chloroflexi (7.19%–17.16%). It exhibited higher counts in winter than in autumn. Anaerolineaceae, which belongs to Chloroflexi, was predominant in each wetland group. Seasonal variations significantly impacted the abundance of ammonia-oxidizing and nitrite-oxidizing bacteria among nitrifying bacteria in the rhizosphere microbial community, with higher levels observed in autumn than in winter, and played a crucial role in NH4+-N transformation. Additionally, a correlation was observed between the nitrogen removal effectiveness and abundance of rhizosphere microbial nitrifying bacteria, providing technical insights for further optimization of the structure and operational parameters of SFGCW. Rhizosphere environments play a vital role in the nutrient cycling of crops and soil organic nitrogen mineralization. Sugar beet is a highly nitrogen (N)-demanding crop, and it is necessary to explore the relationship between the sugar beet root exudates, the microbial community, and nitrogen utilization. In this study, a special separation method was employed to create rhizosphere (H3) and non-rhizosphere (H2 and H1) environments for sugar beet. After 50 d of cultivation in nearly inorganic-free soil, the microbial diversity and its correlation with root metabolites and N were examined. The results showed that in H3, the inorganic N content was over 23 times higher than in H1 and H2, with a 13.1% higher relative abundance of ammonia-oxidizing bacteria compared to H2 and a 32% higher abundance than H1. The relative abundance of nitrite-oxidizing bacteria was also 18.8% higher than in H1. Additionally, a significant positive correlation was observed between inorganic nitrogen content and serine (Ser) and isoleucine (Ile). The organic nitrogen content exhibited positive correlations with glycine (Gly), alanine (Ala), and tyrosine (Tyr) but displayed negative correlations with certain amino acids, organic acids, and glucose. The co-linearity network indicated that the microbial composition in H3 also exhibited higher node connectivity. It can be inferred that under the influence of sugar beet root exudates, the changes in the rhizosphere's microbial diversity were more intricate, thereby benefiting soil nitrogen cycling and inorganic N accumulation. These findings provide profound insight into sugar beet soil organic nitrogen mineralization and contribute to the sustainable and environmentally friendly development of modern agriculture. Improving the quality of tree planting material for carbon sequestration through reforestation can help solve environmental problems, including the need to reduce the concentration of carbon dioxide in the atmosphere. The purpose of this study was to investigate the possibility of using humic substances in combination with rhizosphere microorganisms Pseudomonas protegens DA1.2 and Pseudomonas sp. 4CH as a means to stimulate the growth of seedlings of pine, poplar, large-leaved linden, red oak, horse chestnut, and rowan. Humic substances stimulated the growth of shoots and roots of pine, large-leaved linden, and horse chestnut seedlings. The effects of bacteria depended on both plant and bacteria species: Pseudomonas protegens DA1.2 showed a higher stimulatory effect than Pseudomonas sp. 4CH on pine and linden, and Pseudomonas sp. 4CH was more effective in the case of chestnut. An additive effect of humates and Pseudomonas protegens DA1.2 on the growth rate of pine and linden saplings was discovered. Poplar, red oak, and rowan seedlings were unresponsive to the treatments. The growth-stimulating effects of the treatments are discussed in connection with the changes in carbon, chlorophyll, and nitrogen contents in plants. Medicinal plants have aroused considerable interest as an alternative to chemical drugs due to the beneficial effects of their active secondary metabolites. However, the extensive use of chemical fertilizers and pesticides in pursuit of yield has caused serious pollution to the environment, which is not conducive to sustainable development in the field of medicinal plants. Microbial fertilizers are a type of "green fertilizer" containing specific microorganisms that can improve the soil microbial structure, enhance plant resistance to biological and abiotic stresses, and increase the yield of medicinal plants. The root exudates of medicinal plants attract different microorganisms to the rhizosphere, which then migrate further to the plant tissues. These microbes can increase the levels of soil nutrients, and improve the physical and chemical properties of soil through nitrogen fixation, and phosphorus and potassium solubilization. In addition, soil microbes can promote the synthesis of hormones that increase plant growth and the accumulation of active compounds, eventually improving the quality of medicinal plants. In 2022, the total value of the global microbial fertilizer market was $4.6 billion and is estimated to reach $10.36 billion by 2030. In this review, we have summarized the types of microbial fertilizers, the colonization and migration of microorganisms to plant tissues, and the beneficial effects of microbial fertilizers. In addition, the prospects of developing microbial fertilizers and their application for medicinal plants have also been discussed. It aims to provide a reference for the rational application of microbial fertilizers in the field of medicinal plants and the green and sustainable development of medicinal plant resources. Continuous cropping in greenhouse cultivation often leads to increased pest and disease problems, reducing crop quality and yield. Crop rotation is a common strategy to address these issues. This study compared the growth of Chinese cabbage (Brassica rapa var. chinensis) following rotations with ginger (Zingiber officinale) and sponge gourd (Luffa aegyptiaca). The Chinese cabbage exhibited normal growth following ginger rotation but showed abnormal growth after sponge gourd rotation. The study investigated the underlying causes by analyzing soil physicochemical properties and rhizosphere microbial communities of Chinese cabbage using 16S rRNA and ITS sequencing. The results revealed that soil from ginger–Chinese cabbage rotation had higher levels of soil organic carbon (SOC) and available phosphorus (AP), but lower total nitrogen (TN) and available potassium (AK). Despite similar alpha-diversity for both bacterial and fungal communities, distinct bacterial and fungal community structures between two rotation cropping systems were observed. This suggests that even if the alpha-diversity does not change, the composition of the microbial community can shift in ways that might influence soil health and plant growth. Furthermore, redundancy analysis revealed a significant correlation between microbial community structures and soil physicochemical properties of two rotation cropping systems. The SOC and TN were revealed to be the most significant of the investigated soil physicochemical parameters with respect to the variation of both bacterial and fungal assemblages, respectively. The identified biomarkers in bacterial community composition further emphasize the potential for specific microbes to influence crop health positively or negatively. We found that the indicator genera of the bacterial community composition of the ginger–Chinese cabbage rotation system were Amycolatopsis (genus), Pseudonocardiales (order), Pseudonocardiaceae (family), and Amycolatopsis mediterranei, which are known as producers of secondary metabolites, such as antibiotics. These findings highlight the importance of crop selection in rotation strategies for optimizing agricultural outcomes.

Potatoes (Solanum tuberosum L.) are the third largest food crop globally and are pivotal for global food security. Widespread N fertilizer waste in potato cultivation has caused diverse environmental issues. This study employed microbial metagenomic sequencing to analyze the causes behind the declining N use efficiency (NUE) and escalating greenhouse gas emissions resulting from excessive N fertilizer application. Addressing N fertilizer inefficiency through breeding has emerged as a viable solution for mitigating overuse in potato cultivation. In this study, transcriptome and metabolome analyses were applied to identify N fertilizer-responsive genes. Metagenomic sequencing revealed that excessive N fertilizer application triggered alterations in the population dynamics of 11 major bacterial phyla, consequently affecting soil microbial functions, particularly N metabolism pathways and bacterial secretion systems. Notably, the enzyme levels associated with NO3- increased, and those associated with NO and N2O increased. Furthermore, excessive N fertilizer application enhanced soil virulence factors and increased potato susceptibility to diseases. Transcriptome and metabolome sequencing revealed significant impacts of excessive N fertilizer use on lipid and amino acid metabolism pathways. Weighted gene co‑expression network analysis (WGCNA) was adopted to identify two genes associated with N fertilizer response: PGSC0003DMG400021157 and PGSC0003DMG400009544.

However, the effect of white grub (Maladera verticalis) larvae feeding on the structure and function of rhizosphere microbial communities of aerobic rice (Oryza sativa L.) is unclear. Results In this study, we compared physicochemical properties, enzyme activities, and microbial communities using 18 samples under healthy and M. verticalis larvae-feeding aerobic rice rhizosphere soils at the Yunnan of China. 16 S rRNA and ITS amplicons were sequenced using Illumina high throughput sequencing. M. verticalis larvae feeding on aerobic rice can influence rhizosphere soil physicochemical properties and enzyme activities, which also change rhizosphere microbial communities. The healthy and M. verticalis larvae-feeding aerobic rice rhizosphere soil microorganisms had distinct genus signatures, such as possible_genus_04 and Knoellia genera in healthy aerobic rice rhizosphere soils and norank_f__SC − I−84 and norank_f__Roseiflexaceae genera in M. verticalis larvae-feeding aerobic rice rhizosphere soils. The pathway of the metabolism of terpenoids and polyketides and carbohydrate metabolism in rhizosphere bacteria were significantly decreased after M. verticalis larvae feeding. Fungal parasite–wood saprotroph and fungal parasites were significantly decreased after M. verticalis larvae feeding, and plant pathogen–wood saprotroph and animal pathogen–undefined saprotroph were increased after larvae feeding. [Objective]It is to study the composition and diversity of rhizosphere soil microbial communities in tea gardens of different habitats, to identify the main driving factors affecting rhizosphere soil microbial communities, and to provide theoretical basis for the study of the relationship between rhizosphere soil microbial communities and pest control of tea trees, as well as for the function and utilization of soil microbials.[Method]We used Illumina-MiSeq high-throughput sequencing technology to investigate the community composition and diversity of soil microorganisms in the rhizosphere soil communities of Jingmai Mountain Ancient Tea Garden(GS)and Jingmai Mountain Modern Organic Tea Garden(TS), and used redundancy analysis to explore the relationship between soil nutrients and microorganisms.[Result]The cation exchange capacity, organic matter, humus, ammonia nitrogen, ammonium nitrogen, available phosphorus, and water content in the rhizosphere soil in TS were higher than those in the GS, showing significant differences; the dominant bacterial populations in both gardens were Actinobacteria, Acidobacteria, and Planctomycetes, and the dominant fungal populations are Ascomycota, Mortierellomycota, and Basidiomycota, except for the unclassified genera. The dominant bacterial genera in GS are very similar to those in TS, but the relative abundance of each genus varies greatly in two gardens. Redundancy analysis shows that cation exchange capacity, organic matter, humus, ammonia nitrogen, and ammonium nitrogen were the main environmental factors causing differences in the microbial community structure and diversity in the rhizosphere soil of two tea gardens.[Conclusion]The nutrient and species richness of the rhizosphere soil of TS was significantly higher than that of Gs, and the composition of the dominant bacterial genera in the rhizosphere soil of two tea gardens was highly similar, but their abundance varied significantly, showing a high degree of correlation with soil nutrients. Introduction: Rhizosphere microorganisms are influenced by vegetation. Meanwhile, they respond to vegetation through their own changes, developing an interactive feedback system between microorganisms and vegetation. However, it is still unclear whether the functional diversity of rhizosphere soil microorganisms varies with different carbon storage levels and what factors affect the functional diversity of rhizosphere soil microorganisms. Methods: In this study, the Biolog-Eco microplate technique was used to analyze the metabolic diversity of carbon source of rhizosphere soil microorganisms from 6 Pinus massoniana provenances with three levels of high, medium and low carbon storage. Results: The results showed that the average well color development(AWCD) value of rhizosphere microorganisms was significantly positive correlated with carbon storage level of Pinus massoniana (p < 0.05). The AWCD value, Simpson and Shannon diversity of high carbon sequestrance provenances were 1.40 (144h incubation) 0.96 and 3.24, respectively, which were significantly higher (p < 0.05) than those of other P. massoniana provenances. The rhizosphere microbial AWCD, Shannon and Simpson diversity of the 6 provenances showed the same variation trend (SM>AY>QJ>SX>HF>SW). Similarly, microbial biomass carbon (MBC) content was positively correlated with carbon storage level, and there were significant differences among high, medium and low carbon storage provenances. The PCA results showed that the differences in the carbon source metabolism of rhizosphere microorganisms were mainly reflected in the utilization of amino acids, carboxylic acids and carbohydrates. Pearson correlation analysis showed that soil organic carbon (SOC), total nitrogen (TN) and pH were significantly correlated with rhizosphere AWCD (p < 0.05). Conclusion: Soil properties are important factors affecting rhizosphere microbial carbon source metabolism. The study confirmed that the microorganisms of high carbon storage provenances had relatively high carbon metabolic activity. Among them, the carbon metabolic activity of rhizosphere microorganisms of SM provenance was the highest, which was the preferred provenances in effective ecological service function. Introduction: Suaeda salsa is a typical wetland plant species in coastal areas that plays an important role in protecting the marine eco-environment. The rhizosphere microorganisms of S. salsa are responsible for its growth and development. Method: Eighteen samples were collected from three areas, including the natural S. salsa-growing area (YDJ), artificial S. salsa restoration area (YDB), and nonrestoration area (BKS), and high-throughput sequencing technology was employed to explore the characteristics of the rhizosphere microorganisms of S. salsa in the Yellow River Delta. Results: The results illustrated that the abundance and diversity of soil bacteria were highest in the YDJ group, fungal abundance was highest in the YDJ group, and fungal diversity was greatest in the YDB group. In total, 26,663 operational taxonomy units (OTUs) were found in soil bacteria, among which 9,095, 8,023, and 11,001 were detected in the BKS, YDB, and YDJ groups, respectively. 11,619 OTUs were found in soil fungi, among which 4,278, 4,552, and 5,100 were detected in the BKS, YDB, and YDJ groups, respectively. The YDJ group had the highest number of OTUs for bacteria and fungi among the three groups. Discussion: S. salsa in natural wetland conditions tended to be similar to artificially restored S. salsa. The composition of fungi in the S. salsa rhizosphere had greater similarities than that of the bacteria. Proteobacteria had the highest abundance among bacterial communities, and Ascomycota, Basidiomycota, and Olpidiomycota were dominant in the fungal communities of the three groups. The correlation results found that power of hydrogen (pH) was significantly and negatively correlated with the abundance of Acidobacteriota and Proteobacteria. Meanwhile, electrical conductivity (EC) was significantly and positively correlated with the abundance of Firmicutes and negatively correlated with that of Proteobacteria. Regarding fungi, pH and EC were significantly and negatively correlated with the abundance of Chytridiomycota. Herbicides are known to affect the soil nitrogen cycle by shaping soil microorganisms. However, it is not clear how herbicides regulate diverse transformation processes of soil nitrogen cycling by altering rhizosphere microorganisms, subsequently influencing the feedback to plant nitrogen metabolism. Here, we investigated how imazethapyr (IM) enantiomers drive plant-soil feedback on nitrogen metabolism by altering the rhizosphere microorganisms. The results indicated that (R)- and (S)-IM significantly changed the composition and structure rhizosphere microbiome with enantioselectivity and functional changes in microbial communities were associated with soil nitrogen circulation. The determination of nitrogen-cycling functional genes further supported the above findings. The results revealed that (R)- and (S)-IM could change the abundance of nitrogen-cycling functional genes by changing specific bacteria abundances, such as Bacteroidetes, Proteobacteria, and Acidobacteria, thus resulting in diverse nitrogen transformation processes. The alternation of nitrogen transformation processes indicated (R)-IM exhibited a more notable tendency to form a nitrogen cycling pattern with lower energy cost and higher nitrogen retention than (S)-IM. Sterilization experiments demonstrated changes in soil nitrogen cycling drive plant nitrogen metabolism and rhizosphere microorganisms are responsible for the above process of plant-soil feedback for nitrogen metabolism. Under IM enantiomer treatments, rhizosphere microorganisms might stimulate glutamate synthesis by promoting NH4+ uptake and glutamine-glutamate synthesis cycling in roots, thus contributing to positive feedback, with (R)-IM treatments showing more pronounced positive feedback on nitrogen metabolism than (S)-IM treatments. Our results provide theoretical support for determining the mechanism by which IM enantiomers drive plant-soil nitrogen metabolism by changing the rhizosphere microbial communities. 【Objective] The cultivation way of Shanlan upland rice is single, and the traditional slash and burn cultivation method damages the environment. Cultivation in water can be a new way for Shanlan upland rice. This work is to explore the changes in rhizosphere microorganisms of Shanlan upland rice under water cultivation environment in large fields.[Method] The cultivation treatment of Shanlan upland rice was set up with normal irrigation and drying management, and 16S rRNA gene amplification sequencing was performed on the bacterial community in the rhizosphere soil of Shanlan upland rice. The sequencing data was combined with soil physicochemical properties for comprehensive analysis.[Result] The changes in cultivation methods have significantly affected the composition of rhizosphere bacterial communities. In particular, the relative abundance of Nitrosprota and Proteobacteria in the normal irrigation is lower than that in the drying management, and the relative abundance of Firmicutes is higher than that in drying management. The top 10 bacterial communities with relative abundance are correlated with soil environmental factors. The correlation network analysis shows that more bacterial communities interact in normal irrigation, and there is a more complex bacterial network under normal irrigation.[Conclusion] Significant changes in bacterial community composition occure in the rhizosphere of Shanlan upland rice under different water management cultivation methods. The bacteria groups of Nitrospirota and Proteobacteria may improve the stress resistance of Shanlan upland rice by promoting nitrogen uptake at the root of rice. The application of beneficial microbial consortium can effectively improve plant disease resistance and its growth. Various fungi were compounded with Bacillus velezensis LJ02 and applied to watermelon plants in this paper. The results showed that the microbial consortium T2 (compounded Bacillus velezensis LJ02 with Aspergillus aculeatus 9) can effectively control gummy stem blight and powdery mildew in watermelon, while the control effect reached 83.56% and 70.93%, respectively (p < 0.05). Compound treatment improved the diversity and richness of the rhizosphere microbial community structure, and the relative abundance of Caulobacterales and Xanthomonadaceae significantly increased after applying T2 to the soil. Meanwhile, the internode length was significantly decreased 28% (p < 0.05), and the maximum leaf length increased 10.33% (p < 0.05). In addition, the microbial consortium delays the maturity of watermelon vegetables. By studying the effects of microbial consortium on watermelon seedlings, our study provides a theoretical basis for the popularization and application of the compound inoculant. Abstract Purpose The health of rhizosphere soil microorganisms is an important indicator to evaluate soil quality. Therefore, understanding the response of rhizosphere soil microorganisms to tobacco crop succession is crucial for promoting the sustainable development of agriculture. Methods The microbial diversity and community structure of rhizosphere soil in continuous cropping and non-cropped tobacco for 7 years were analyzed by the Illumina platform. Result (1) Continuous cropping tobacco cause rhizosphere soil acidification and reduction in alkaline nitrogen (AN) and soil organic matter (SOM). (2) Continuous cropping tobacco reduces the diversity of rhizosphere soil microbial communities, increasing harmful functional microorganisms and declining beneficial ones. (3) The abundance of bacteria that perform nitrification and saprophytic fungi in the rhizosphere soil of continuous cropping areas decreases, inhibiting carbon and nitrogen cycling processes. (4) The composition and diversity of the soil rhizosphere microbial community are affected by the imbalance in the physicochemical property of the rhizosphere. Conclusion Continuous cropping tobacco cause rhizosphere soil acidification and nutrient imbalance, and the carbon and nitrogen cycles involved in microorganisms were damaged. Furthermore, the decreased diversity of rhizosphere soil microorganisms and the increased abundance of pathogenic fungi contribute to the continuous cropping obstacles of tobacco.

Most research on plant–microbe interactions emphasize the effects of micronutrients on the rhizosphere microbial community structure. However, the influence of seed structures, particularly the radicle sheath, on microbial diversity at the seedling root tips under varying temperatures and humidity has been less explored. This study conducted controlled indoor experiments in the northern desert of Xinjiang to assess the radicle sheath’s impact on microbial community composition, diversity, and function. The results indicated no significant changes in the Chao1 index for bacteria and fungi, but notable differences were observed in the Shannon and Simpson indices (p < 0.05). Under drought conditions, the radicle sheath significantly reduced bacterial infections without affecting fungi. Genus-level analysis showed an increased abundance of specific dominant bacterial groups when the radicle sheath was retained. NMDS analysis confirmed its significant effect on both bacterial and fungal community structures. LEfSe analysis identified 34 bacterial and 15 fungal biomarkers, highlighting the treatment’s impacts on microbial taxonomic composition. Functional predictions using PICRUSt 2 revealed that the radicle sheath facilitated the conversion of CH4 to CH3OH and various nitrogen cycle processes under drought. Overall, the radicle sheath plays a crucial role in maintaining rhizosphere microbial community stability and enhancing the functions of both bacteria and fungi under drought conditions.

Chrysanthemum wilt is a plant disease that exerts a substantial influence on the cultivation of Chrysanthemum zawadskii (Herb.) for tea and beverage production. The rhizosphere microbial population exhibits a direct correlation with the overall health of plants. Therefore, studying the rhizosphere microbial community of Chrysanthemum zawadskii (Herb.) Tzvel. is of great significance for finding methods to control this disease. This study obtained rhizosphere soil samples from both diseased and healthy plant individuals and utilized high-throughput sequencing technology to analyze their microbial composition. The results showed that the rhizosphere microbial diversity decreased significantly, and the microbial community structure changed significantly. In the affected soil, the relative abundance of pathogenic microorganisms such as rhizospora and Phytophthora was greatly increased, while the relative abundance of beneficial microorganisms such as antagonistic fungi and actinomyces was greatly decreased. In addition, this study also found that soil environmental variables have an important impact on plant resistance; the environmental factors mainly include soil properties, content of major microorganisms, and resistance characteristics of samples. Redundancy analysis showed that the drug-resistant population had a greater impact on the 10 species with the highest abundance, and the environmental factors were more closely related to the sensitive population. In the fungal community, the resistant sample group was more sensitive to the influence of environmental factors and high-abundance fungi. These findings provide a theoretical basis for improving microbial community structure by optimizing fertilization structure, thus affecting the distribution of bacteria and fungi, and thus improving the disease resistance of chrysanthemum. Reducing greenhouse gas emissions while improving productivity is the core of sustainable agriculture development. In recent years, rice ratooning has developed rapidly in China and other Asian countries, becoming an effective measure to increase rice production and reduce greenhouse gas emissions in these regions. However, the lower yield of ratooning rice caused by the application of a single nitrogen fertilizer in the ratooning season has become one of the main reasons limiting the further development of rice ratooning. The combined application of nitrogen and phosphorus plays a crucial role in increasing crop yield and reducing greenhouse gas emissions. The effects of combined nitrogen and phosphorus application on ratooning rice remain unclear. Therefore, this paper aimed to investigate the effect of combined nitrogen and phosphorus application on ratooning rice. Two hybrid rice varieties, 'Luyou 1831' and 'Yongyou 1540', were used as experimental materials. A control treatment of nitrogen-only fertilization (187.50 kg·ha−1 N) was set, and six treatments were established by reducing nitrogen fertilizer by 10% (N1) and 20% (N2), and applying three levels of phosphorus fertilizer: N1P1 (168.75 kg·ha−1 N; 13.50 kg·ha−1 P), N1P2 (168.75 kg·ha−1 N; 27.00 kg·ha−1 P), N1P3 (168.75 kg·ha−1 N; 40.50 kg·ha−1 P), N2P1 (150.00 kg·ha−1 N; 13.50 kg·ha−1 P), N2P2 (150.00 kg·ha−1 N; 27.00 kg·ha−1 P), and N2P3 (150.00 kg·ha−1 N; 40.50 kg·ha−1 P). The effects of reduced nitrogen and increased phosphorus treatments in ratooning rice on the yield, the greenhouse gas emissions, and the community structure of rhizosphere soil microbes were examined. The results showed that the yield of ratooning rice in different treatments followed the sequence N1P2 > N1P1 > N1P3 > N2P3 > N2P2 > N2P1 > N. Specifically, under the N1P2 treatment, the average two-year yields of 'Luyou 1831' and 'Yongyou 1540' reached 8520.55 kg·ha−1 and 9184.90 kg·ha−1, respectively, representing increases of 74.30% and 25.79% compared to the N treatment. Different nitrogen and phosphorus application combinations also reduced methane emissions during the ratooning season. Appropriately combined nitrogen and phosphorus application reduced the relative contribution of stochastic processes in microbial community assembly, broadened the niche breadth of microbial communities, enhanced the abundance of functional genes related to methane-oxidizing bacteria and soil ammonia-oxidizing bacteria in the rhizosphere, and decreased the abundance of functional genes related to methanogenic and denitrifying bacteria, thereby reducing greenhouse gas emissions in the ratooning season. The carbon footprint of ratooning rice for 'Luyou 1831' and 'Yongyou 1540' decreased by 25.82% and 38.99%, respectively, under the N1P2 treatment compared to the N treatment. Salt stress as one of the common abiotic stresses can affect plant growth and development. Root exudates are considered as an important medium for“information”exchange between plants and rhizosphere environment. When plants sufferred in salt stress, the composition and content of root exudates would change to affect the growth and development of plants. This article summarizes the components, detection methods, mechanism of action and changing trends. Root exudates mainly contained amino acids, sugars, organic acids and phenolic acids, etc. The high performance liquid chromatography (HPLC), gas chromatography-mass spectrometry (GC-MS), liquid chromatographymass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) are widely used to identify and detect compositions and contents of root exudates. Combined with previous studies, the changes of composition and content of amino acids, sugars, organic acids, etc. under salt stress were summarized. From the aspect of physical and chemical environment and rhizosphere microorganisms, the putative roles of root exudates under salt stress were also summarized, including: 1) acting as signal molecules in plant suffering to salt tolerance; 2) maintaining the homeostasis of root cells; and 3)affecting rhizosphere environment. Cut chrysanthemum, known as a highly favored floral choice globally, experiences a significant decline in production due to continuous cropping. The adverse physiological effects on cut chrysanthemums result from the degradation of a soil's physical and chemical properties, coupled with the proliferation of pathogens. The "Guangyu" cultivar in Xinxiang, Henan Province, China, has been specifically influenced by these effects. First, the precise pathogen accountable for wilt disease was effectively identified and validated in this study. An analysis was then conducted to examine the invasion pattern of the pathogen and the physiological response of chrysanthemum. Finally, the PacBio platform was employed to investigate the dynamic alterations in the microbial community within the soil rhizosphere by comparing the effects of 7 years of monocropping with the first year. Findings indicated that Fusarium solani was the primary causative agent responsible for wilt disease, because it possesses the ability to invade and establish colonies in plant roots, leading to alterations in various physiological parameters of plants. Introduction: Tobacco root-knot nematode (RKN) is a highly destructive soil-borne disease worldwide. However, there is a lack of research on the relationship between RKN and tobacco root microbial community composition under large-scale geographical conditions in China. Methods: In this study, we collected 65 samples from 28 main tobacco-growing areas across 10 provinces in China and conducted 16S rDNA sequencing to investigate the dynamic microbial changes in tobacco soil infected by RKN compared to healthy tobacco soil. Based on the analysis of rhizosphere soil bacterial communities, changes after RKN infection, and soil environmental factors. Results: We found the 28 tobacco-growing areas could be divided into two distinct groups with different microbial compositions and varying responses to RKN infection. In group1 of the provinces of Anhui, Henan, Shanxi, and Heilongjiang, Vicinamibacteria dominated the bacterial community, while Acidobacteriae was present in low abundance. In contrast, group2 of the other six provinces (Yunnan, Guizhou, Chongqing, Guangxi, Hubei, and Shandong) exhibited an opposite pattern. After infected by RKN, the genera Chitinophaga increased significant in group 1, while the genera Rhodococcus in group 2 exhibited a substantial increase. Alpha-diversity analysis revealed that RKN-infected tobacco exhibited a richer and more diverse rhizosphere soil bacterial community compared to healthy tobacco in most growing areas. A total of 12 kinds of soil environmental factors were measured in healthy and RKN-infected tobacco soil, and based on the co-occurrence and correlation analysis between environmental factors and microbial species, the pH level, calcium (Ca), magnesium (Mg), phosphorus (P), iron (Fe), and sodium (Na) were identified as key environmental factors influencing the population composition of rhizosphere microorganisms during RKN infection. We observed that RKN infection further increased the pH in weakly alkaline group 1 soil, while weakly acidic group 2 soil experienced a further decrease in pH. Furthermore, we identified three genera as potential biocontrol or plant growth-promoting bacteria for tobacco. Vegetation root exudates have the ability to shape soil microbial community structures, thereby enhancing CH 4 bio-oxidation capacity in landfill cover systems. In this study, the CH 4 oxidation capacity of indigenous vegetation rhizosphere microorganisms within operational landfill covers in Chongqing, China, was investigated for the first time, with the objective of identifying suitable plant candidates for CH 4 mitigation initiatives within landfill cover systems. Furthermore, a multi-omics methodology was employed to explore microbial community structures and metabolic variances within the rhizospheric environment of diverse vegetation types. The primary aim was to elucidate the fundamental factors contributing to divergent CH 4 oxidation capacities observed in rhizosphere soils. The findings demonstrated that herbaceous vegetation predominated in landfill covers. Notably, Rumex acetosa exhibited the highest CH 4 oxidation capacity in the rhizosphere soil, approximately 20 times greater than that in non-rhizosphere soil. Root exudates played a crucial role in inducing the colonization of CH 4 -oxidizing functional microorganisms in the rhizosphere, subsequently prompting the development of specific metabolic pathways. This process, in turn, enhanced the functional activity of the microorganisms while concurrently bolstering their tolerance to microbial pollutants. Increasing soil salinization can severely restrict local agricultural production. Planting alfalfa is considered an effective measure to ameliorate saline–alkali soil. However, it remains unclear how alfalfa planting years affect the sustained impact on soil and rhizosphere microecology. This study analyzed the effects of alfalfa planted 3, 6, and 8 years ago on soil physicochemical properties and key soil enzyme activities and investigated the rhizosphere microbial community structure and diversity. The results indicate that cultivating alfalfa plants for six years can improve soil physicochemical properties and enhance soil fertility to a certain extent. This is attributed to a higher abundance of plant growth-promoting bacteria, such as Bradyrhizobium and Allorhizobium, as well as degradation bacteria, such as Flavobacterium, Stenotrophomonas, Brevundimonas, and Massilia, in the rhizosphere of alfalfa plants. These microorganisms promote alfalfa growth, improve soil quality, and inhibit the accumulation of autotoxins. This not only maintains high alfalfa yields but also optimizes soil physicochemical properties and enzyme activity, facilitating more effective nutrient cycling and metabolic processes in the soil. [Objective] The study aimed to reveal the microbial community structure and soil physicochemical properties of apple rhizosphere under the intercropping system of persimmon and apple trees. [Method] In the study, rhizosphere soil of intercropping (I) and non-intercropping (NI) apples were used as materials to analyze the rhizosphere microbial community structure by high-throughput sequencing technology. [Result] lnter-cropping of persimmon significautly increased the contents of organic matter (OM), available nitrogen (AN), total phosphorus (TP), decreased the contents of available potassium (AK) in apple rhizosphere soil. The Alpha diversity analysis showed that intercropping had no significant effect on the bacterial richness index and diversity index of apple rhizosphere soil, whereas significantly reduced the Ace index of rhizosphere soil fungi (P = 0. 03). β diversity analysis showed that there were significant differences in bacterial and fungal community structure between intercropped and non-intercropped soils. Further LEfSe analysis showed that intercropping significantly increased the abundance of 17 bacterial genera in apple rhizosphere soil, of which, 9 were known genera, and 8 were unknown genera. These nine known genera included Sporacetigenium, Tepidisphaera, Agromyces, Alkanindiges, Phycisphaera, Pseudoxanthomonas, Terrimonas, Pseudohaliea, Paenisporosarcina; The abundances of four bacterial genera, namely Sporichthya, Methylocystis, Devosia and Alter-erythrobacter were significantly decreased. Intercropping significantly increased the abundance of 1 fungal genus Arthrographis and decreased the abundance of 3 fungal genera, namely Cylindrocarpon, Metacordyceps and Exophiala. Spearman's correlation analysis showed that Pseudomonas was positively correlated with available phosphorus (AP) and TP, while Naganishia was positively correlated with AP. The fungal genera Polyscytalum and Minimedusa showed a significant negative correlation with AP. PICRUSt2 functional prediction analysis showed that intercropping significantly decreased the abundance of carbohydrate transport and metabolism, and extracellular structure in rhizosphere bacteria. FUNGuild analysis showed that intercropping significantly reduced the abundance of pathogen-saprophytic transitional fungi in apple rhizosphere soil. Salinity is one of the biggest harmful stress factors that limit the stability of plants and their productivity, as well as reduce the fertility of soils. Therefore, the research on plant protection mechanisms against high salt concentration in the environment and the search for ways to increase their resistance to this stress factor are relevant today. The presented literature review describes the peculiarities of legume response to salt stress, in particular during the establishment of relationship with nodule bacteria. High concentrations of salt in soil lead to the interruption of some vital processes in legumes and thus cause a significant decrease in both crop quality and harvest size. So, the results of studies which indicate a negative effect of salt stress on growth and development, hormonal status, photosynthesis and carbon assimilation, osmotic processes, maintaining the ion homeostasis and the formation of reproductive organs are given. Special attention is paid to the question of the influence of salinity on the interaction between plants and rhizobia during nodule formation and their further functioning. It is noteworthy that the presence of certain adaptive mechanisms as well as the peculiarities of growth and development of legumes, in particular their capability to form symbiotic nitrogen fixation systems with nodule bacteria, suggest a possibility of using certain species of this family for the remediation of saline soils. The importance of the selection of salt-resistant rhizobia strains and the effectiveness of rhizobia in combination with other beneficial microorganisms for agriculture are noted.

Abstract Background Panax ginseng cultivated under the forest is popular because its shape and effective ingredients are similar to wild ginseng. The growth of P. ginseng in the larch forest is generally better than in the broad-leaved forest, and the incidence rate of diseases is low. Therefore, the selection of forest species is one of the basic factors in the successful cropping of P. ginseng. Methods Illumina HiSeq high-throughput sequencing was used to analyze the 16S rRNA/ITS gene sequence of P. ginseng rhizosphere soil under larch forest to study the rhizosphere microbiome's diversity and community composition structure. Results The species classification and richness of rhizosphere bacterial and fungal communities in the same-aged P. ginseng were similar. Consistent with the soil system of commonly cultivated crops, Proteobacteria, Actinobacteriota, Acidobacteriota, Verrucomicrobiota, Chloroflexi, and Basidiomycota, Ascomycota were the dominant phylum of bacteria and fungi, respectively. Compared with the soil without planting P. ginseng, the diversity of microorganisms and community structure of continuous planting for 2 years, 5 years, and 18 years of P. ginseng rhizosphere soil had little change. The accumulation levels of Ilyonectria, Fusarium, Gibberella, and Cylindrocarpon were not significantly increased with planting P. ginseng and the increased age of cropping P. ginseng. Conclusions The results of this study showed that the soil function of the larch forest was good, which provided a theoretical basis for the land selection and soil improvement of cultivating P. ginseng under the larch forest.

Abstract Background Rapeseed cake is an important agricultural waste. After enzymatic fermentation, rapeseed cake not only has specific microbial diversity but also contains a lot of fatty acids, organic acids, amino acids and their derivatives, which has potential value as a high-quality organic fertilizer. However, the effects of fermented rapeseed cake on tea rhizosphere microorganisms and soil metabolites have not been reported. In this study, we aimed to elucidate the effect of enzymatic rapeseed cake fertilizer on the soil of tea tree, and to reveal the correlation between rhizosphere soil microorganisms and nutrients/metabolites. Results The results showed that: (1) The application of enzymatic rapeseed cake increased the contents of soil organic matter (OM), total nitrogen (TN), total phosphorus (TP), available nitrogen (AN), and available phosphorus (AP); increased the activities of soil urease (S-UE), soil catalase (S-CAT), soil acid phosphatase (S-ACP) and soil sucrase (S-SC); (2) The application of enzymatic rapeseed cake increased the relative abundance of beneficial rhizosphere microorganisms such as Chaetomium, Inocybe, Pseudoxanthomonas, Pseudomonas, Sphingomonas, and Stenotrophomonas; (3) The application of enzymatic rapeseed cake increased the contents of sugar, organic acid, and fatty acid in soil, and the key metabolic pathways were concentrated in sugar and fatty acid metabolisms; (4) The application of enzymatic rapeseed cake promoted the metabolism of sugar, organic acid, and fatty acid in soil by key rhizosphere microorganisms; enzymes and microorganisms jointly regulated the metabolic pathways of sugar and fatty acids in soil. Conclusions Enzymatic rapeseed cake fertilizer improved the nutrient status and microbial structure of tea rhizosphere soil, which was beneficial for enhancing soil productivity in tea plantations. These findings provide new insights into the use of enzymatic rapeseed cake as an efficient organic fertilizer and expand its potential for application in tea plantations.

In agricultural production, it is crucial to increase the availability of phosphorus (P) in cultivated soil to solve the P limitation. Arbuscular mycorrhizal fungi (AMF) have been proven to promote crop nutrient absorption effectively, while biochar can lead to improvements in soil properties. However, the possible synergistic effect of AMF and biochar on P uptake by crops as well as its underlying mechanisms are unclear. In this study, we conducted a pot experiment to explore the effects of biochar and AMF (Glomus etunicatum) on the community of rhizospheric phosphate-solubilizing microorganisms (PSMs) of maize (Zea mays L. Xianyu-335) using metagenomic methods. The experiment used 0 mg P2O5 g·kg−1 soil (P0) and 30 mg P2O5 g·kg−1 soil (P30) application rates. Each P application rate included 0 (NC), 20 g·kg−1 biochar (BC) addition, inoculation AMF, and without AMF treatments (NM) for a total of eight treatments. During the experiment, both the P uptake and the biomass of maize were measured. The study found that the combination of AMF and biochar significantly increased the mycorrhizal colonization rate of maize roots, regardless of P application level. It was observed that the P uptake by maize was significantly increased when exposed to a combination of AMF and biochar. The increase in P uptake in P0 treatments was 67% higher than the sum of the effects of biochar and AMF inoculation alone. The increase was only 35% higher in P30 treatments, demonstrating a substantially higher interactive effect under P0 than under P30 conditions. The AM-BC treatments significantly increased the abundance of Streptomyces, Bacillus, and Pseudomonas, genera that are known to contain PSMs. In addition, the abundance of genes related to P-cycling (gcd, phoD, and ugpQ) in PSMs increased significantly by 1.5–1.8 times in AM-BC treatments compared with NM-BC and AM-NC treatments under P0 conditions. This increase was significantly and positively correlated with the P uptake. Overall, the results suggest that biochar can help AMF colonize the roots, increasing the functional roles of PSMs in the rhizosphere, which in turn promotes P uptake and biomass in maize. This study provides a new way to improve P-use efficiency and reduce the need for P-fertilizer application in agricultural production.

Rhizosphere environments play a vital role in the nutrient cycling of crops and soil organic nitrogen mineralization. Sugar beet is a highly nitrogen (N)-demanding crop, and it is necessary to explore the relationship between the sugar beet root exudates, the microbial community, and nitrogen utilization. In this study, a special separation method was employed to create rhizosphere (H3) and non-rhizosphere (H2 and H1) environments for sugar beet. After 50 d of cultivation in nearly inorganic-free soil, the microbial diversity and its correlation with root metabolites and N were examined. The results showed that in H3, the inorganic N content was over 23 times higher than in H1 and H2, with a 13.1% higher relative abundance of ammonia-oxidizing bacteria compared to H2 and a 32% higher abundance than H1. The relative abundance of nitrite-oxidizing bacteria was also 18.8% higher than in H1. Additionally, a significant positive correlation was observed between inorganic nitrogen content and serine (Ser) and isoleucine (Ile). The organic nitrogen content exhibited positive correlations with glycine (Gly), alanine (Ala), and tyrosine (Tyr) but displayed negative correlations with certain amino acids, organic acids, and glucose. The co-linearity network indicated that the microbial composition in H3 also exhibited higher node connectivity. It can be inferred that under the influence of sugar beet root exudates, the changes in the rhizosphere’s microbial diversity were more intricate, thereby benefiting soil nitrogen cycling and inorganic N accumulation. These findings provide profound insight into sugar beet soil organic nitrogen mineralization and contribute to the sustainable and environmentally friendly development of modern agriculture.

Background and Aim: Increased soil salinization caused by natural or human activities is the leading cause of soil degradation. Cultivating salt-tolerant crops is an effective way to cope with soil salinity. However, the key salinity-tolerance traits and regulation mechanisms in salt-tolerant crops and the relevant ecological functions of the microbiome in the rhizosphere remain largely unexplored. Methods: The phenotypic, physiological, morphological, and molecular responses of salt-sensitive (SS 212) and salt-tolerant (ST 47) broomcorn millet (Panicum miliaceum L.) to salinity were dissected, and the composition and variation in the rhizospheric microorganisms under saline and non-saline conditions were established. Results: Salinity tolerance was achieved in ST 47 by maintaining Na+/K+ balance and intact cell surface and internal structures and regulating the expression of PmCNGC (Na+ uptake), PmNHX (Na+ sequestration), and PmSOS1 (Na+ extrusion). Moreover, the genotype mediated the differences in the rhizospheric microbial community. The ST 47 rhizosphere had a stronger and more stable fungal composition under saline conditions compared to SS 212. In addition, ST 47 reshaped the microorganisms in the rhizosphere by recruiting specific beneficial microbes, namely Haliangium, Marispirillum, RB41, and Saccharospirillum bacteria, which promote soil nutrient cycling, and Acremonium, Blastobotrys, Calostoma, Chaetomium, Chrysosporium, Fusarium, and Scytalidium fungi, which are involved in stress tolerance and resource uptake. Conclusion: The findings of this study provide an empirical basis for improving crop performance through utilizing a key salt-tolerance mechanism and soil microbial management practices for sustainable agriculture in salinized soil. The modified material at the bottom of lakes plays a key role when restoring and reconstructing the ecology of waterbodies. However, the effects of different substrate types on the growth and development of submerged plants and rhizosphere microorganisms remain unclear. Therefore, this study analyzed the effects of five substrates (attapulgite, biochar, cinder, maifanite, and quartz sand) on the growth and rhizosphere microorganisms of Vallisneria natans (Lour.) Hara (V. natans). The results showed that during the culture period, the mean plant height of V. natans peaked in the maifanite group (26.75 cm), the mean leaf number peaked in the biochar group (12.13), and the mean root activity peaked in the cinder group (83.42 U g−1 min). There was no significant difference in the total chlorophyll content among the groups during culture (P > 0.05). The malondialdehyde content of V. natans leaves in all groups, except quartz sand group, peaked early in the culture period (10 days). Moreover, the superoxide dismutase activity of the five groups increased first and then decreased; however, the catalase activity in the maifanite group decreased significantly compared with the levels in the other groups (P<0.05). This indicates that maifanite supplementation can accelerate the adaptation of V. natans to changes in the environment. After the addition of substrate, the species of dominant bacteria in the rhizosphere remained nearly unchanged, but their relative abundance was different. Specifically, the relative abundance of Desulfobacterota increased in each group, peaking at 9.77% in the maifanite group. This study showed that different substrates had different growth-promoting effects on submerged plants, with maifanite exerting the greatest effect. Introduction: Rhizosphere microorganisms can effectively promote the stress resistance of plants, and some beneficial rhizosphere microorganisms can significantly promote the growth of crops under salt stress, which has the potential to develop special microbial fertilizers for increasing the yield of salinealkali land and provides a low-cost and environmentally friendly new strategy for improving the crop yield of saline-alkali cultivated land by using agricultural microbial technology. Methods: In May 2022, a field study in a completely randomized block design was conducted at the Heilongjiang Academy of Agricultural Sciences to explore the correlation between plant rhizosphere microorganisms and soybean growth in saline-alkali soil. Two soybean cultivars (Hening 531, a salt-tolerant variety, and 20_1846, a salt-sensitive variety) were planted at two experimental sites [Daqing (normal condition) and Harbin (saline-alkali conditions)], aiming to investigate the performance of soybean in saline-alkali environments. Results: Soybeans grown in saline-alkali soil showed substantial reductions in key traits: plant height (25%), pod number (26.6%), seed yield (33%), and 100 seed weight (13%). This underscores the unsuitability of this soil type for soybean cultivation. Additionally, microbial analysis revealed 43 depleted and 56 enriched operational taxonomic units (OTUs) in the saline-alkali soil compared to normal soil. Furthermore, an analysis of ion-associated microbes identified 85 mOTUs with significant correlations with various ions. A cooccurrence network analysis revealed strong relationships between specific mOTUs and ions, such as Proteobacteria with multiple ions. In addition, the study investigated the differences in rhizosphere species between salt-tolerant and salt-sensitive soybean varieties under saline-alkali soil conditions. Redundancy analysis (RDA) indicated that mOTUs in saline-alkali soil were associated with pH and ions, while mOTUs in normal soil were correlated with Ca2+ and K+. Comparative analyses identified significant differences in mOTUs between salttolerant and salt-sensitive varieties under both saline-alkali and normal soil conditions. Planctomycetes, Proteobacteria, and Actinobacteria were dominant in the bacterial community of saline-alkali soil, with significant enrichment compared to normal soil. The study explored the functioning of the soybean rhizosphere key microbiome by comparing metagenomic data to four databases related to the carbon, nitrogen, phosphorus, and sulfur cycles. A total of 141 KOs (KEGG orthologues) were identified, with 66 KOs related to the carbon cycle, 16 KOs related to the nitrogen cycle, 48 KOs associated with the phosphorus cycle, and 11 KOs linked to the sulfur cycle. Significant correlations were found between specific mOTUs, functional genes, and phenotypic traits, including per mu yield (PMY), grain weight, and effective pod number per plant. Conclusion: Overall, this study provides comprehensive insights into the structure, function, and salt-related species of soil microorganisms in salinealkali soil and their associations with salt tolerance and soybean phenotype.
