Trametes versicolor glutathione transferase Omega 3S (TvGSTO3S) catalyzes the conjugation of isothiocyanates (ITC) with glutathione (GSH).Previously, this isoform was investigated in depth both biochemically and structurally. Structural analysis of complexes revealed the presence of a GSH binding site (G site) and a deep hydrophobic binding site (H site) able to bind plant polyphenols. In the present study, crystals of apo TvGSTO3S were soaked with glutathionyl-phenethylthiocarbamate, the product of the reaction between GSH and phenethyl isothiocyanate (PEITC).On the basis of this crystal structure, we show that the phenethyl moiety binds in a new site at loop beta(2)-alpha(2) while the glutathionyl part exhibits a particular conformation that occupies both the G site and the entrance to the H site. This binding mode is allowed by a conformational change of the loop beta(2)-alpha(2) at the enzyme active site. It forms a hydrophobic slit that stabilizes the phenethyl group at a distinct site from the previously described H site.Structural comparison of TvGSTO3S with drosophila DmGSTD2 suggests that this flexible loop could be the region that binds PEITC for both isoforms. These structural features are discussed in a catalytic context.

International audience; Exudates from rectus femoris pork’s tumbling were characterized by quantification of proteins, lipids and indicators of bonds between proteins (hydrophobicity, protein carbonyl groups, free thiols). Effect of salt content (112, 165 and 220 g/l in the brine) and cooking time were studied. No influence of salt was observed on protein content and composition, on lipids and on free thiol groups. Protein surface hydrophobicity was dependent on salt content, with a maximum with 165g/l of salt in brine. Anti-oxidant effect of salt on carbonyl content appeared when exudate was heated. Heating exudate led to an increase in protein hydrophobicity and carbonyl content and to a decrease in free thiols.

Dans ce travail, après une revue bibliographique de l'allergie médicamenteuse, nous mettons en évidence les IgE spécifiques aux myorelaxants. Pour cela, nous avons développé une technique radioimmunologique en couplant les myorelaxants au gel Epoxy-Sepharose. Nous avons montré que l'épitope impliqué est l'ammonium quaternaire, groupement qui entre dans la structure de tous les myorelaxants et qui explique la fréquence élevée de l'allergie croisée abordée dans cette étude. Nous avons amélioré la sensibilité des techniques radioimmunologiques en modifiant la méthode de couplage de l'ammonium quaternaire sur gel. Nous avons montré que la liaison IgE spécifique myorelaxant est de type hydrophobe favorisée par un mécanisme d'interaction de charge
