Rheumatoid arthritis (RA) is the most frequent chronic inflammatory rheumatism. This disease is characterized by an auto-immunity and a hyperplasic synovitis, both responsible for peripheral joints destruction. Macrophages contribute to inflammatory aspects of RA. This cell type can present various states of activation or "polarization", reversible and dependent on its environment, notably cytokines. Biologics (bDMARDs) represented a revolution in severe RA treatment. However, data regarding their specific action on macrophage are scarce. The aim of our translational work was: (i) to assess the in vitro effect of RA bDMARDs (anti-TNF: etanercept, adalimumab, certolizumab; anti-IL-6R: tocilizumab; CTLA4-Ig: abatacept) on the phenotype of monocytes-derived-macrophages from RA patients and healthy volunteers, during differentiation and activation phases, (ii) to identify polarization markers correlated with disease activity (DAS28). Among bDMARDs, only anti-TNF modulated macrophage polarization. During differentiation, anti-TNF bDMARDs induced a bias toward the so-called alternative non-inflammatory polarization. In inflammatory context, anti-TNF bDMARDs induced a selective preservation of markers associated with IL-10 (CD16, CD163, MerTK) and an inhibition of inflammatory markers (CD40, CD80). We showed that these changes in phenotype were associated with changes in functions consistent with: (i) a polarization induced by IL10, (ii) a decrease in inflammatory cytokines production (TNF, IL-6, IL-12), (iii) and an increase in phagocytosis. We showed that this mechanism was dependant on early IL-10 production and STAT3 signaling. Moreover, we have showed that certolizumab, an anti-TNF agent, induced an anti-inflammatory response, implicating the transcription factor NRF2 (nuclear factor erythroid-2- related factor 2), a central regulator of the response to oxidative stress. We observed that CD16 expression on non-activated macrophages was negatively correlated with RA activity. This work contributes to demonstrate the relevance of macrophage targeting in RA, and enabled us to identify theranostic targets for RA treatment with anti-TNF bDMARDs.; La polyarthrite rhumatoïde (PR) est le rhumatisme inflammatoire chronique le plus fréquent. Nous avons montré que ce changement phénotypique s'accompagnait : (i) d'un changement fonctionnel concordant avec une polarisation induite par l'IL-10, (ii) d'une inhibition de la production des cytokines de l'inflammation (TNF, IL-6, IL-12), (iii) et d'une majoration de la phagocytose. Nous avons montré que ce mécanisme était dû à une production précoce d'IL-10 et dépendant de STAT3. De plus, nous avons pu montrer que le certolizumab, un anti-TNF, induisait une réponse anti-inflammatoire, impliquant le facteur de transcription NRF2 (nuclear factor erythroid-2-related factor 2), un régulateur central dans la réponse au stress oxydatif. Enfin, nous avons observé que l'expression de CD16, à la surface des macrophages non activés, était corrélée négativement à l'activité de la PR. Ces travaux concourent à montrer l'intérêt du ciblage du macrophage dans la PR et nous ont d'identifier de potentielles cibles théranostiques dans le traitement de la PR par anti-TNF.

International audience; Introduction: Cognitive impairment that affects older adults is commonly associated with an inflammatory imbalance, resulting in decreased physical fitness. Exercise has been pointed to mitigate immunosenescence and cognitive impairment associated with aging, while increase in physical fitness. However, few studies explored the relationship between changes in cytokine concentration and improvement on cognition due to elastic band strength training. The aim of this study was to investigate the effects of strength training on pro-and anti-inflammatory cytokines, hematological markers and physical fitness of older women with cognitive impairment. Methods: Thirty-three women (82.7 +/- 5.7 years old) participated in the study and were divided in two groups: strength exercise training group (ST; n = 16) and Control Group (CG; n = 17) and were evaluated before and after 28 weeks of the exercise program. The CG did not undergo any type of exercise programs. Data for IL-10, TNF-alpha, IFN-gamma, C-Reactive Protein (CRP), white blood counts (WBC), red blood counts (RBC), Mini Mental State Examination (MMSE) and physical fitness tests were analyzed in both moments. Results: IL-10 increased in the ST group without changes in CG. TNF-alpha and CRP increased in the control group while no changes were observed for IFN-g in both groups. Strength training decreased leukocyte and lymphocyte counts and increase hemoglobin, mean cell volume and mean cell hemoglobin concentration. The MMSE score increased in strength training group but remained unchanged in the control group. A correlation between the variation of granulocyte counts and the MMSE scores was also observed within the total sample. An improvement in physical fitness was observed with strength training. Conclusion: Resistance exercise promoted better anti-inflammatory balance and physical performance simultaneously with an increase in cognitive profile in older women with cognitive impairment.

Regulatory B cells (Breg) were first reported to be interleukine-10 (IL-10) producing B cells in mice. The almost concurrent discovery of Breg cells drew interest toward potential links with transitional B cells because of phenotypic and functional similarities. In addition with IL-10 production, CD24high CD38high transitional B cells limit the proliferation of T cells and the polarization of CD4+ T cells into Th1 cells. Transitional B cells represent a central developmental stage in B-cell maturation, linking generation in the bone marrow with differentiation in periphery. In a first study, we reveal for the first time that human transitional B cells encompass not only transitional type 1 and type 2 B cells, but also distinct anergic type 3 B cells, as well as IL-10-producing CD27+ transitional B cells. Interestingly, the latter two subsets differentially regulate CD4+ T-cell proliferation and polarization toward Th1 effector cells. Additional experiments showed that type 1 and CD27+ transitional B cells are capable to differentiate into antibody secreting cells after toll-like receptor 9 engagement. In a second work, we wanted to explore the ability of B cells to target T-cell populations. We demonstrate that B cells can be suppressive cells. B cells are capable to target CD4+ memory T-cell, limiting the proliferation and inducing the death of this T-cell population. At the opposite, B cells seem to be effector of CD4+ naïve T-cell functions. These properties are probably associated with a specific transcriptional program. Thus, we observed that suppressive B cells overexpress PRDM1 and IL10, whereas effector B cells preferentially express BCL6 and NFκB1 in in vitro mixed culture. In the last part, we worked on B-cell phenotype and functions in transplanted patients. BHL (B lymphocytes in humoral rejection and alloimmunisation) is a clinical study that aims to better understand the role of B cells in the alloimmunisation and the chronic rejection occurring after renal transplantation. Donor specific antibodies (DSA) seem to limit the expansion of transitional B cells, which are probably not associated with the ability of B cells to regulate T-cell proliferation in DSA+ patients.; Le potentiel régulateur des lymphocytes B (LB), largement associé avec la production d’interleukine-10 (IL-10), a été mis en évidence dans des modèles murins de pathologies spécifiques d’Ag. Les cellules B transitionnelles (Tr.) CD24fortes CD38fortes ont été décrites comme régulatrices, au travers de la production d’IL-10, de l’inhibition de la prolifération T, ainsi que de la suppression de la réponse inflammatoire des cellules T. Les LB transitionnels représentent un stade de développement central dans la maturation des cellules B, en faisant le lien entre les cellules immatures de la moelle osseuse et celles matures situées dans les organes lymphoïdes secondaires. Dans une première étude, nous montrons que cette population est hétérogène, et composée de LB Tr. de type 1 (T1), T2, T3 et Tr. CD27+. Les LB T3 anergiques semblent jouer un rôle dans la tolérance périphérique en limitant la prolifération des lymphocytes T (LT) CD4+, tandis que les LB Tr. CD27+ IL-10+ nouvellement décrits inhibent la différenciation des LT CD4+ en cellules productrices d’IFN-γ et de TNF-α. Notons que les LB T1 et Tr. CD27+ se différencient rapidement en cellules productrices d’Ac suite à la reconnaissance de signaux de l’immunité innée. La production d’IL-10 est en partie dépendante des signaux perçus, provenant du microenvironnement. Nous avons décrit dans un second travail que les LB s’adaptent aux cellules avec lesquelles ils sont cultivés. En effet, les cellules B régulent spécifiquement les LT CD4+ mémoires (et non naïfs), en limitant leur prolifération avant d’induire une mort cellulaire. Ces caractéristiques fonctionnelles pourraient être associées avec une modification du programme transcriptionnel, permise par la plasticité des cellules B, qui se polarisent en LB régulateurs (Breg) de façon ciblée. L’expression des gènes PRDM1 et IL10 serait associée avec une signature Breg spécifique en culture mixte autologue, en opposition avec celle des gènes NFκB1 et BCL6. La transplantation rénale est un excellent modèle physiopathologique, pour étudier l’importance de certaines populations de LB dans la tolérance immunologique. L’étude BHL (B lymphocytes in humoral rejection and alloimmunisation) nous a permis de confirmer que les LB Tr. ont probablement un rôle important dans cette tolérance du greffon. La présence d’anticorps spécifiques du donneur (DSA) semble limiter l’émergence des LB Tr., même si le pourcentage de cellules B CD24fortes CD38fortes n’est a priori pas associé avec la capacité du compartiment lymphocytaire B à réguler la prolifération des cellules T des patients alloimmunisés.

The incidence of inflammatory disorders in industrialized countries has dramatically increased over the last decennia, which is believed to result from a change in life-style. Treatment of these inflammatory disorders relies on the intervention in immune responses thereby restoring homeostasis. For now, many inflammatory disorders are treated with broad-acting immunosuppressive drugs or monoclonal antibodies that specifically target pro-inflammatory molecules of the immune system. An alternative therapeutic approach would be to use immunomodulatory proteins that are naturally involved in re-establishing immune homeostasis. This thesis describes the plant-based expression of a variety of immunomodulatory cytokines that may be used as biopharmaceutical proteins in the future. Furthermore, this thesis contains a pioneering chapter on the plant-based expression of immunomodulatory helminth-secreted glycoproteins. In Chapter 2 we describe the plant-based expression of the immune-regulatory cytokine human transforming growth factor β1 (TGF-β1). By co-expressing human furin with latent TGF-β1 we were able to engineer the post-translational proteolytic processing of TGF-β1, which enabled the production of biologically active TGF-β1. In Chapter 3 we reveal that aggregation is a major production bottleneck for the anti-inflammatory cytokine interleukin-10 (IL-10). By protein engineering we were able to prevent aggregation and created a biologically active fusion protein of IL-10. In Chapter 4 we express biologically active IL-22 in plants. We reveal that, in contrast to current literature, its activity is independent of the presence of N-glycans or their composition. This chapter further reveals that plants offer a powerful tool to allow investigation into the role of N-glycans in protein folding and biological activity of glycoproteins. In Chapter 5 we further explore the potential of glyco-engineering in plants by engineering helminth-like N-glycans. We produce large quantities of two major egg antigens from Schistosoma mansoni and successfully engineer Lewis X, LDN and LDNF N-glycan structures. These plant biotechnological research lines are a showcase for the potential of engineering proteins as well as post-translational modifications in plants with special emphasis on N-glycan engineering. Altogether, the results presented in the first four chapters reveal the remarkable flexibility of plants as a production platform for recombinant proteins. It showcases the potential of engineering proteins as well as post-translational modifications in plants, but it especially highlights the engineering of tailor made N-glycans in plants. This, combined with the speed of transient expression by means of agroinfiltration, makes transient expression in Nicotiana benthamiana a powerful tool to study the role of N-glycans on glycoprotein function. In parallel to these plant biotechnological research lines, we also developed an in vitro model system based on mouse bone marrow-derived cells to study immunological responses. We used this model to obtain clues on why IL-10 therapy has not been as successful as previously anticipated. In Chapter 6 we have set-up biological activity assays based on bone marrow-derived cells and reveal that IL-10 activity is dependent on both IL-10R1 and IL-10R2, but not IL-10R2-associated signalling via Tyk2. We also show that interactions between IL-10R1 and IL-10R2 (both intracellular and extracellular) reduce cellular binding of IL-10, but are crucial to initiate IL-10 mediated signalling. Furthermore, we observed that macrophages and dendritic cells respond differently to IL-10. This was further investigated in Chapter 7 where we reveal that GM-CSF (the cytokine used to differentiate dendritic cells) is responsible for negatively regulating early IL-10-mediated responses. Strikingly, GM-CSF does not strongly affect the IL-10-induced activation of the transcription factor STAT3. Instead, GM-CSF induces strong constitutive phosphorylation of GSK-3β, a signalling component downstream of the PI3K/Akt pathway. These immunological chapters give novel insights on the mechanism of initiating IL-10-induced signalling and on the possible integration of signal transduction pathways elicited by different cytokines. Ultimately this knowledge could provide us with new therapeutic strategies to treat inflammatory disorders.

It showcases the potential of engineering proteins as well as post-translational modifications in plants, but it especially highlights the engineering of tailor made N-glycans in plants. This, combined with the speed of transient expression by means of agroinfiltration, makes transient expression in Nicotiana benthamiana a powerful tool to study the role of N-glycans on glycoprotein function. In parallel to these plant biotechnological research lines, we also developed an in vitro model system based on mouse bone marrow-derived cells to study immunological responses. We used this model to obtain clues on why IL-10 therapy has not been as successful as previously anticipated. In Chapter 6 we have set-up biological activity assays based on bone marrow-derived cells and reveal that IL-10 activity is dependent on both IL-10R1 and IL-10R2, but not IL-10R2-associated signalling via Tyk2. We also show that interactions between IL-10R1 and IL-10R2 (both intracellular and extracellular) reduce cellular binding of IL-10, but are crucial to initiate IL-10 mediated signalling. Furthermore, we observed that macrophages and dendritic cells respond differently to IL-10. This was further investigated in Chapter 7 where we reveal that GM-CSF (the cytokine used to differentiate dendritic cells) is responsible for negatively regulating early IL-10-mediated responses. Strikingly, GM-CSF does not strongly affect the IL-10-induced activation of the transcription factor STAT3. Instead, GM-CSF induces strong constitutive phosphorylation of GSK-3β, a signalling component downstream of the PI3K/Akt pathway. These immunological chapters give novel insights on the mechanism of initiating IL-10-induced signalling and on the possible integration of signal transduction pathways elicited by different cytokines. Ultimately this knowledge could provide us with new therapeutic strategies to treat inflammatory disorders.

Preventing untoward immune responses against a specific antigen is a major challenge in different clinical settings such as gene therapy, transplantation, or autoimmunity. Following intramuscular delivery of recombinant adeno-associated virus (rAAV)-derived vectors, transgene rejection can be a roadblock to successful clinical translation. Specific immunomodulation strategies potentially leading to sustained transgene expression while minimizing pharmacological immunosuppression are desirable. Tolerogenic dendritic cells (TolDC) are potential candidates but have not yet been evaluated in the context of gene therapy, to our knowledge. Following intramuscular delivery of rAAV-derived vectors expressing an immunogenic protein in the nonhuman primate model, we assessed the immunomodulating potential of autologous bone marrow-derived TolDC generated in the presence of IL10 and pulsed with the transgene product. TolDC administered either intradermally or intravenously were safe and well tolerated. While the intravenous route showed a modest ability to modulate host immunity against the transgene product, intradermally delivery resulted in a robust vaccination of the macaques when associated to intramuscular rAAV-derived vectors-based gene transfer. These findings demonstrate the critical role of TolDC mode of injection in modulating host immunity. This study also provides the first evidence of the potential of TolDC-based immunomodulation in gene therapy.

Background: Inflammatory bowel diseases (IBD) are intestinal disorders characterized by inflammation in the gastrointestinal tract. Interleukin-10 is one of the most important anti-inflammatory cytokines involved in the intestinal immune system and because of its role in downregulating inflammatory cascades, its potential for IBD therapy is under study. We previously presented the development of an invasive strain of Lactococcus lactis (L. lactis) producing Fibronectin Binding Protein A (FnBPA) which was capable of delivering, directly to host cells, a eukaryotic DNA expression vector coding for IL-10 of Mus musculus (pValac:il-10) and diminish inflammation in a trinitrobenzene sulfonic acid (TNBS)-induced mouse model of intestinal inflammation. As a new therapeutic strategy against IBD, the aim of this work was to evaluate the therapeutic effect of two L. lactis strains (the same invasive strain evaluated previously and the wild-type strain) carrying the therapeutic pValac:il-10 plasmid in the prevention of inflammation in a dextran sodium sulphate (DSS)-induced mouse model. Results: Results obtained showed that not only delivery of the pValac:il-10 plasmid by the invasive strain L. lactis MG1363 FnBPA+, but also by the wild-type strain L. lactis MG1363, was effective at diminishing intestinal inflammation (lower inflammation scores and higher IL-10 levels in the intestinal tissues, accompanied by decrease of IL-6) in the DSS-induced IBD mouse model. Conclusions: Administration of both L. lactis strains carrying the pValac:il-10 plasmid was effective at diminishing inflammation in this murine model of experimental colitis, showing their potential for therapeutic intervention of IBD. Les lymphocytes B (LB) jouent un rôle essentiel dans la physiopathologie des maladies auto-immuneset notamment de la polyarthrite rhumatoïde (PR) à cause de leur capacité à produire des anticorps etdes cytokines pro-inflammatoires. Un traitement visant à éliminer les LB, le rituximab (anti-CD20) esttrès efficace pour lutter contre la PR.Toutefois, un sous-type de lymphocytes B, les LB régulateurs, a été récemment mis en évidence chezla souris. Ce sous-type de lymphocytes B aurait plutôt un rôle protecteur. Les LB régulateurs appelésB10 semblent capables de réguler le système immunitaire via la sécrétion d’une cytokineanti-inflammatoire, l’interleukine 10 (IL10), et l’induction de lymphocytes T régulateurs (Tregs). Ilspeuvent ainsi inhiber les lymphocytes T effecteurs de type Th1 et Th2. Il a été montré, chez la souris,que le transfert de ces cellules permettait de prévenir le développement de l’arthrite. En janvier 2010,ces B10 ont été mis en évidence chez l’homme. Des anomalies qualitatives (diminution de laproduction d’IL10) ont été retrouvées chez les patients lupiques.Dans un premier temps, nous avons mis au point la culture des cellules mononucléées du sang afin degénérer des B10. Nous avons ensuite recherché le ou les précurseurs des B10 grâce à des triscellulaires. Nous avons ensuite comparé les B10 d’un groupe de sujets sains avec des patients atteintsde PR.À l’issue de ce travail de recherche, nous avons mis en évidence une diminution des B10 dans la PR etnotamment les PR de moins de 5 ans ainsi qu’une corrélation inverse des B10 avec l’activité de lamaladie chez ces derniers. Chez les patients qui présentent des auto-anticorps de type facteurrhumatoïde ou anticorps anti-protéines citrullinées (ACPA), nous avons démontré une corrélationinverse entre le taux sérique de ces auto-anticorps et les B10.

Lactic acid bacteria (LAB) represent a heterogeneous group of microorganisms that are naturally present in many foods and possess a wide range of therapeutic properties. The aim of this paper is to present an overview of the current expanding knowledge of one of the mechanisms by which LAB and other probiotic microorganisms participate in the prevention and treatment of gastrointestinal inflammatory disease through their immune-modulating properties. A special emphasis will be placed on the critical role of the anti-inflammatory cytokine IL-10, and a brief overview of the uses of genetically engineered LAB that produce this important immune response mediator will also be discussed. Die chronisch obstruktive Lungenerkrankung (COPD) und das Bronchialkarzinom (BC) sind weltweit häufige Erkrankungen von großer volkswirtschaftlicher Bedeutung, mit weiter stegender Inzidenz. Bei derzeit fehlenden kurativen Therapiemöglichkeiten für die COPD und der weiterhin schlechten Prognose der Bronchialkarzinome scheinen Untersuchungen zur weiteren Klärung ätiopathogenetischer Mechanismen dieser beiden Erkrankungen sinnvoll. In der vorgelegten Arbeit werden Polymorphismen der Gene der Entzün-dungsmediatoren Tumor Nekrose Faktor alpha (TNF-α) und beta (TNF-β), Interleukin 6 (IL-6) und Interleukin 10 (IL-10) bei Patienten mit COPD und Bronchialkarzinomen im Vergleich mit Kontrollgruppen in einer Fall-Kontroll Studie untersucht. Bei Patienten mit einer COPD und bei einer biologischen Untergruppe der Bronchialkarzinome, den kleinzelligen Karzinomen (SCLC), fand sich eine signifikant höhere Frequenz des IL-10 –1082_G Polymorphismus. Die Korrelation des Polymorphismus des IL-10 Gens mit der Entwicklung einer COPD oder eines Bronchialkarzinoms, weist auf einen Zusammenhang von immunologischen und antiinflammatorischen Prozessen in den Atemwegen und der Pathogenese von COPD und Bronchialkarzinomen hin. Ebenso könnte der assoziierte Einzelbasenaustausch (SNP) auch zur Risikostratifikation, oder möglicherweise zur serologischen Typisierung im klinischen Bereich dienen., COPD and lung cancer are important diseases with high economic impact and increasing incidence worldwide. Caused by missing curative therapeutic options for COPD and at least bad prognosis for lung cancer patients it seems to be point of interest in looking for etiopathogenetic factors of this diseases. In this work polymorphisms of genes of the cytokines TNF-alpha and beta, interleukin 6 and 10 has been investigated in patients with COPD and lung cancer in a case control study. In patients with COPD an in a lung cancer subgroup there was a significant higher frequency of the IL-10 -1082_G polymorphism. The correlation of this polymorphism with the developement of COPD or lung cancer demonstrates a connection between immunological and antiinflamatory processes in the airways with the pathogenesis of these diseases. Also this SNP association could be a factor for risk stratification as well as for serological typing in clinical practice.

Par leur capacité de moduler la réponse immune, les rayons ultraviolets (UV) ont trouvé des applications dans le traitement de diverses maladies immunes. Leurs mécanismes d’action sont encore incomplètement définis. L’un d’entre eux comporte l’induction de cytokines immunosuppressives et antiinflammatoires. Ce processus peut être provoqué par la phagocytose de corps apoptotiques, l’apoptose constituant une des lésions cellulaires élémentaires provoquée par les UV. Le but de notre travail a été de préciser les cytokines impliquées dans la réponse aux UV, de définir certains mécanismes de leur production et de la potentialiser par des agents pharmacologiques.Notre étude a comporté deux parties: (1) l’une in vivo chez des malades souffrant de GVH chronique résistante aux traitements conventionnels et traités par photochémothérapie extracorporelle, procédure dans laquelle les leucocytes du malades, prélevés par leucaphérèse puis traités par un psoralène et par UVA lui sont finalement réinjectés; (2) l’autre in vitro où des PBMC de volontaires sains ont été irradiés avec 10J/m2 de rayons UVC qui ne nécessitent pas de photosensibilisation par psoralène. Deux cytokines, l’IL-10 et l’IL-1Ra ont été évaluées par RT-PCR dans un système de coculture autologue entre PBMC et PBL rendus apoptotiques par irradiation. L’évolution du processus apoptotique déclenché par les UV a été mesurée par cytomètrie de flux. Celui-ci concernait essentiellement les lymphocytes, les monocytes/macrophages révélant une résistance relative à l’apoptose, il était progressif, culminant entre la 24ème et la 48ème heures. Lors des cocultures entre PBMC et PBL irradiés, un accroissement très significatif du nombre de copies d’ARNm, concernant les deux cytokines étudiées, l’IL-10 et l’IL-1Ra était observé. L’induction d’IL-1Ra était dépendante de l’IL-10. Une préactivation par du LPS était nécessaire pour la révélation du phénomène. Ensuite, nous avons évalué l’implication sur la synthèse de cytokines du processus de phagocytose de lymphocytes rendus apoptotiques par irradiation UV et divers moyens pharmacologiques pour la potentialiser. La préincubation du matériel irradié pendant une nuit (16h) à 37° dans le but d’accroître la proportion de cellules en voie d’apoptose avant mise en contact avec les PBMC a permis d’obtenir un accroissement très marqué sans nécessiter de LPS, portant essentiellement sur la production d’IL-1Ra tant sur l’ARNm que la protéine secrétée; l’induction d’IL-10 était cette fois négligeable. L’implication de la phagocytose dans le processus a été démontrée par deux agents bloquants (a) l’anticorps monoclonal anti-CD36 (corécepteur avec l’intégrine &61537;V&61538;3 de la thrombospondine) activant la production d’IL-1Ra et mimant par ce fait le processus phagocytaire et (b) la cytochalasine E la bloquant.Nous avons testé diverses substances pharmacologiques dont l’action activatrice de l’IL-1Ra est connue, en l’occurrence les immunoglobulines G à usage IV (IgIV) et le GM-CSF. L’adjonction d’IgIV (1mg/ml) ou GM-CSF (10 ng/ml) une heure après le début de la coculture exerce sur la sécrétion d’IL-1Ra un effet additif avec les UV. Selon la concentration utilisée, les IgIV peuvent agir par deux mécanismes. Outre l’effet d’activation macrophagique lié au récepteur Fc, nous avons démontré à haute concentration un mécanisme nouveau, du à la présence dans les IgIV d’anticorps naturels antiFas induisant l’apoptose des lymphocytes. Une incubation de 16h des lymphocytes avec 25 mg/ml d’IgIV avant mise en culture provoque outre une apoptose importante une augmentation significative de l’IL-1Ra. Dans ce cas, le processus est indépendant du fragment Fc, la fraction F(ab’)2 gardant la capacité d’induire l’apoptose et de provoquer la production d’IL-1Ra. En conclusion, nous avons mis en évidence un mécanisme nouveau d’induction d’IL-1Ra, non décrit auparavant et défini diverses modalités qui pourraient accroître sa production: - L’incubation de 16h du matériel irradié permet d’orienter le système en accroissant la production de l’IL-1Ra sans que la production de l’IL-10 soit modifiée et sans nécessiter de LPS. Nous attribuons cet effet à l’accroissement du processus apoptotique qui en résulte.- Nous avons potentialisé la production d’IL-1Ra par deux agents pharmacologiques, le GM-CSF et les IgIV. Les mécanismes d’action des IgIV dépendent de la concentration utilisée.1. Aux concentrations de l’ordre de 1mg/ml, les IgIV exercent, avec les UV un effet additif sur l’induction d’IL-1Ra par une action dépendant du fragment Fc. 2. Aux concentrations élevées de 25mg/ml, un effet apoptotique attribuable à l’action d’anticorps anti-Fas agonistes est observé. Une préincubation de 16h de lymphocytes avec cette concentration d’ IgIV avant mise en culture avec les PBMC autologues provoque outre l’apoptose importante des lymphocytes un accroissement significatif de la production d’IL-1Ra. L'étude des interactions hôte-virus contribue à une meilleure compréhension du fonctionnement du système immunitaire et de sa régulation. Dans un contexte de latence, le génome viral est maintenu dans les cellules infectées grâce à une expression antigénique limitée. Nombreux sont les virus qui ont développé différentes stratégies pour échapper au système immunitaire de l'hôte : on parlera alors d'immunosubversion virale. Le virus d'Epstein-Barr (EBV) a été le premier à être associé à des cancers humains. 90% de la population est infectée et il semble que cette "cohabitation" ait favorisé, tout au long de l'évolution humaine, une capture de certains gènes cellulaires par le virus. L'EBV s'est ainsi "armé" de protéines virales, homologues ou non aux protéines humaines, afin de moduler le système immunitaire de l'hôte. Ainsi, le virus est capable de bloquer les effets antiviraux de l'IFN, de limiter l'apoptose des cellules infectées, ou encore de bloquer la présentation d'antigènes aux cellules effectrices de la réponse immunitaire. Les pathologies associées à l'EBV, dans un contexte d'immunodépression ou d'immunocompétence, résultent d'un déséquilibre dans l'interaction hôte-virus. La détection d'interleukine-10 chez les patients présentant un lymphome post-transplantation, un lymphome de Burkitt ou encore un lymphome non hodgkinien a révélé une nouvelle stratégie du virus visant à favoriser sa dissémination : le virus possède une protéine homologue à l'interleukine-10 humaine (hIL-10), nommée vIL-10, synthétisée au cours de la phase productive de virions. Cette vIL-10, qui possède les mêmes propriétés que l'hIL-10, exerce cependant des effets 3 à 10 fois plus faibles. L'infection de lymphocytes B par EBV entraîne une augmentation de l'hIL-10. Cette cytokine favorise d'une part la croissance et la prolifération des lymphocytes B et induit d'autre part une immunosuppression visant les monocytes/macrophages et les lymphocytes T. La protéine LMP-1 induirait le gène de l'hIL-10 au cours de la phase de latence du virus. Lors de la réactivation virale, l'expression de ZEBRA est associée à l'augmentation de la sécrétion d'hIL-10 par une lignée lymphocytaire B. Au vu des propriétés transactivatrices de ZEBRA sur des gènes viraux et cellulaires nous avons suggéré que ZEBRA est capable d'induire l'hIL-10. Nous avons donc étudié l'action de ZEBRA sur les différentes étapes de la production de l'hIL-10 en analysant son effet sur l'activité du promoteur de l'hIL-10 (pIL-10) en expression transitoire dans des lignées lymphocytaires B, sur la sécrétion d'hIL-10 et sur le taux d'ARNm hIL-10 dans ces mêmes lignées. Nous montrons que la protéine ZEBRA active ces trois étapes, qu'elle soit surexprimée dans les cellules (endogène) ou ajoutée dans le milieu de culture (exogène) ; ce dernier point impliquerait un passage transmembranaire de ZEBRA, ce qui reste à démontrer. Une application clinique peut être envisagée : une protéine ZEBRA recombinante (complète ou partielle) augmente, in vitro, la production d'hIL-10 qui peut être ensuite utilisée en thérapeutique. En effet, les propriétés immunosuppressives de l'hIL-10 peuvent être mises à profit pour induire une tolérance post-greffe ; par ailleurs, les propriétés anti-inflammatoires de la cytokine peuvent être exploitées dans la prévention de pathologies cardiovasculaires. D'autres modulateurs de l'expression de l'IL-10 ont été identifiés : parmi ceux-ci, la tétracycline augmente l'expression du gène entre 10 et 1600%. Nous montrons ainsi comment une stratégie virale visant la modulation du système immunitaire peut être détournée à des fins thérapeutiques.
