Background: Plants comprise great antioxidant sources as a result of their redox and biochemical components, which are rich in secondary metabolites such as phenolic acids, flavonoids, and other constituents. Haplophyllum obtusifolium from polygonaceae is widely used for preventing and managing diabetes. This study investigated the antibacterial and antioxidant activities of silver nanoparticles (AgNPs) biosynthesized by H. obtusifolium. Methods: The aerial parts of H. obtusifolium were gathered from the north of Khorasan Razavi province, Iran and desiccated at the chamber temperature. The shoots were powdered by grinding, 5 g of the powder was mixed with 250 mL of deionized water, and the resultant blend was then filtered. Bactericidal properties and antioxidant activity of the nanoparticles were assessed using disk diffusion and DPPH (2, 2-diphenyl-1-picrylhydrazyl) tests, respectively. Results: The results of this study showed that the biosynthesized nanoparticles exhibited antibacterial activity against a gram-negative (Klebsiella pneumoniae) bacterium, but they had no effects on gram-positive Staphylococcus epidermidis. Antioxidant test results showed that these nanoparticles were capable of eliminating DPPH radicals in a concentration-dependent manner so that a more potent antioxidant activity was seen in higher concentrations of the nanoparticles. Conclusion: Our results suggested that H. obtusifolium can be used as a key source of antioxidants/ antimicrobial agents in food and pharmaceutical industries.

Family Rutaceae is presented in Serbian flora with 3 genera and 4 species: Ruta graveolens L., Dictamnus albus L., Haplophyllum suaveolens (DC.) G. Don and H. boissieranum Vis. & Pančić. Comparative study of the phytochemical variability and antioxidant activity of ethanolic and methanolic extracts, obtained from these species’ areal parts, was performed. Total flavonoid content was determined spectrophotometrically and HPLC method was employed for quantification of coumarins. Fast antioxidant screening of samples was done in DPPH test. Flavonoid content of the tested samples varied from 2.82±0.27 to 27.72±0.15 mg of rutin equivalent (Ru)/g of extract. Psoralen, bergapten and xanthotoxin were detected and quantified only in R. graveolens extracts. All the tested samples exhibited notable potential in DPPH radicals scavenging assay. Yet, considerably lower activity was measured for Haplophyllum extracts. The present study brings forward the new data regarding the chemical profiles and antiradical activities of named Rutaceae species.

Aim The Irano-Turanian ( IT) floristic region is characterized by high levels of endemicity. Despite its potential role as a cradle of xerophytic taxa for neighbouring areas, its biogeographical history remains poorly studied. Haplophyllum, a diagnostic element of the IT region, was used as a model to discriminate between alternative biogeographical scenarios for the evolution of the region and, more specifically, to investigate whether it served as a source of xerophytes for the colonization of the Mediterranean Basin. Location Irano-Turanian floristic region (Central Asia and West Asiatic areas) and Mediterranean floristic region (western and eastern parts of the Mediterranean Basin). Methods Three chloroplast DNA regions were sequenced in 77 accessions of Haplophyllum and 37 accessions from other subfamilies of Rutaceae. To elucidate the temporal and spatial evolution of Haplophyllum in the IT and Mediterranean regions, we performed Bayesian molecular dating analyses with four fossil constraints and ancestral range reconstructions, respectively. Results Our molecular dating and ancestral area reconstruction analyses suggest that Haplophyllum originated in the Central Asian part of the IT region during the early Eocene and started to diversify in situ during the early Oligocene, soon after the vanishing of the Tethys Ocean. Our results further imply that Haplophyllum later invaded the eastern Mediterranean Basin in the middle-to-late Miocene, concomitantly with the Paratethys Salinity Crisis and rapid palaeobiogeographical changes in the proto-Mediterranean. Finally, Haplophyllum diversified in the western Mediterranean in the early Pliocene at the end of the Messinian Salinity Crisis. Main conclusions The IT floristic region can serve as a 'donor' of xerophytic taxa to 'recipient' neighbouring regions, including the Mediterranean floristic region. The climatic/geological processes during the Miocene-Pliocene, by increasing aridity and topographic heterogeneity, facilitated range shifts and allopatric speciation in the region. Haplophyllum A. Juss. is one of the most species-rich, but poorly-known genera of Rutaceae (citrus family), reaching maximum species diversity in Turkey. Iran, and Central Asia. Many of its species exhibit a narrow geographic range ("narrow endemics"), which makes them particularly vulnerable to extinction. Despite its importance for the characterization of the Irano-Turanian floristic region, the evolution of species diversity in Haplophyllum has never been examined in a phylogenetic and biogeographic context. We generated gene trees from DNA sequences of four regions of the chloroplast genome for 118 accessions, representing 66% of the species diversity of the genus. Additionally. Haplophyllum was examined morphologically. The phylogenetic analyses showed that several species of the genus do not form reciprocally monophyletic groups. Optimization of morphological characters on the chloroplast DNA phylogeny indicated that most of the species, in particular those with a widespread geographic distribution, can only be diagnosed by combinations of homoplasious character states. Homoplasy notwithstanding, the main morphological characters traditionally used to classify the genus are consistent with the molecular phylogeny of Haplophyllum. Finally, the Mediterranean representatives of Haplophyllum were found to be embedded within a clade that includes primarily Irano-Turanian species, suggesting multiple invasions of the Mediterranean basin from the east. Total extracts of the aerial parts of four Haplophyllum species (Rutaceae) including Haplophyllum stapfianum Hand.-Mazz., H. tuberculatum (Forssk.) Adr. Juss., H. acutifolium (DC.) G. Don, and H. viridulum Soják were tested for in vitro cytotoxicity against 11 tumor cell lines and normal peripheral blood mononuclear cells (PBMCs). Cytotoxicity of Haplophyllum species were also compared with Ruta graveolens L. (Rutaceae) extract. Cytotoxic activity of extracts was evaluated by the WST-1 assay and expressed as IC50 values. The results obtained indicated that among hematopoietic tumor cell lines, strong cytotoxic activity was observed from H. stapfianum, H. tuberculatum, and H. acutifolium against RAMOS cell line with IC50 values of 12.3, 25.3, and 23.7 µg/mL, respectively; H. stapfianum and H. tuberculatum against U937 cell line with IC50 values of 15.6 and 29.3 µg/mL; and H. tuberculatum against RPMI-8866 cell line with IC50 = 31.8 µg/mL. Testing plant extracts on solid tumor cell lines revealed that all Haplophyllum species have markedly high cytotoxicity against LNCap-FGC-10, a prostate adenocarcinoma cell line, except H. viridulum. H. tuberculatum also showed strong cytotoxic effect against 5637, a bladder carcinoma cell line (IC50 = 23.3 µg/mL). Investigating cytotoxic activity of R. graveolens resulted in higher IC50 values against all tumor cell lines except LNCap-FGC-10 (IC50 = 27.6 µg/mL). Cytotoxicity evaluation of Haplophyllum species on PBMCs revealed that resulting IC50 values for H. stapfianum and H. acutifolium on tumor cell lines with high cytotoxic activity were significantly lower than the IC50 values obtained for PBMCs (p < 0.05). Abstract: Further to a systematic chemotaxonomic study of Uzbek Haplophyllum A. Juss. plants selected on ethnopharmacological data, 14 alkaloids were screened for their cytotoxic properties. As a first selection for interesting compounds, each alkaloid was tested against two human cancer cell lines (HeLa and HCT-116), using WST-1 reagent. Of the 14 alkaloids, 5 were cytotoxic when tested against the HeLa line with an IC50 <100μM. These five compounds consisted of three furoquinolines: skimmianine; haplopine and γ-fagarine and two pyranoquinolones: flindersine and haplamine. Only haplamine was active against the HCT-116 line. The cytotoxic properties of these five alkaloids were further investigated against five additional human cancer cell lines. Their structure–activity relationships will be discussed. Of these five pre-selected alkaloids, only haplamine showed significant cytotoxic activity against all the tested cell lines. This is the first report of the cytotoxic activity of haplamine. Finally, this pyranoquinolone alkaloid was tested here against 14 different cancer cell lines and against normal skin fibroblasts. [Copyright &y& Elsevier]

• Background and Aims The discovery of a new species, Haplophyllum bastetanum F.B. Navarro, V.N. Suárez-Santiago & Blanca sp. nov., in the south-east of Spain has prompted the comparative study of species of the Iberian Peninsula, and others related, through morphological, cytogenetic, molecular, distributional and ecological characterization.• Methods The morphological study involved a quantitative analysis of the species present in the Iberian Peninsula and a comparative analysis of the morphological characteristics between H. bastetanum and other related species. Mitotic analyses were made with root meristems taken from germinating seeds. Phylogenetic analyses of the internal transcribed spacer sequences of nuclear ribosomal DNA were performed using neighbour-joining (NJ) and maximum-parsimony methods.• Key Results Haplophyllum bastetanum is a diploid species (2n = 18) distinguished primarily for its non-trifoliate glabrous leaves, lanceolate sepals, dark-green petals with a dorsal band of hairs, and a highly hairy ovary with round-apex locules. The other two Iberian species (H. linifolium and H. rosmarinifolium) are tetraploid (2n = 36) and have yellow petals. Both phylogenetic methods generated a well-supported clade grouping H. linifolium with H. rosmarinifolium. In the NJ tree, the H. linifolium–H. rosmarinifolium clade is a sister group to H. bastetanum, while in the parsimony analysis this occurred only when the gaps were coded as a fifth base and the characters were reweighted according to the rescaled consistency index. This latter group is supported by the sequence divergence among taxa.• Conclusions The phylogenies established from DNA sequences together with morphological and cytogenetic analyses support the separation of H. bastetanum as a new species. The results suggest that the change in the number of chromosomes may be the key mechanism of speciation of the genus Haplophyllum in the Iberian Peninsula. An evolutionary scheme for them is propounded. [ABSTRACT FROM AUTHOR]

Anahtar kelimeler; Karaciğer kanseri, Haplophyllum ptilostylum L., apoptozis, antikanser Nowadays, among the cancers that cause death, liver cancer is the second in males and 6th in females. There is no effective treatment for hepatocellular carcinoma (HCC), the most common type of liver cancer. Chemotherapy treatment in HCC is generally used for metastatic and recurrent diseases, whereas therapeutic agents such as doxorubicin, platinum, fluoropyrimidine and gemcitabine can provide less than 10% response when used alone, but when these drugs are administered in combination, treatment is only responded to around 20%. Because of the side effects of chemotherapy and radiotherapy applications in HCC treatment is limited, therefore, new alternative treatment methods are being researched. It is observed that herbal treatment methods take the first place among the alternative treatment methods due to the low side effects and have an important place in cancer treatment. Intensive research is being carried out for the detection of new plant-derived compounds that can be used as an alternative to chemotherapies which are used in cancer treatment and have high side effects. This study was conducted to investigate the anticancer activity of the extract obtained from Haplophyllum ptilostylum plant which is endemic in Şanlıurfa on liver cancer cell (SNU-423).In this study, the cytotoxic effect of plant extract on normal and cancer cell MTT, apoptotic effect (flow cytometric Annexin V and acridin oranhe / ethidiumbormide staining) on the cell cycle effect on flow cytometric and mitochondrial membrane potential was investigated by JC-1 method. As a result of the study, it was observed that the cytotoxic dose (IC50: 3,55 g / ml) of plant extract on liver cancer cell (SNU-423) showed no toxic effect on normal cells. It was.In this study, it has been concluded that Haplophyllum ptilostylum has anticancer potential and therefore new studies should be conducted to determine new specific active substances that can be used in cancer treatment of this plant extract. Keywords; Liver cancer, Haplophyllum ptilostylum L., apoptosis, anticancer 59

In this regard, a new identification key for the studied taxa was given. Pollen size and surface sculpturing pattern were studied with using Scanning Electron Microscopy (SEM) and Light Microscopes (LM). Pollen shapes of the taxa were found as suboblate, oblate- spheroidal, prolate- spheroidal and sub-prolate and, their aperture types are tricolporate and trizonolcolporate. Pollen surface sculpturing patterns were determined as striate, striate-perforate, striate-microreticulate, rugulate and rugulate-perforate. For anatomic studies, cross sections from stem and leaves were taken. Stem anatomy was provided some diagnostic features for distinguishing some species. Within the scope of the molecular researches, all Haplophyllum and Ruta species from Turkey and additional taxa from the other genera in the Rutaceae family and Spathelia species were used as out group to construct phylogenetic tree and to show taxonomic relationships among the taxa. On the purpose of morphological variations and phylogenetic relationships among different populations of the same species, sequence analysis of the chloroplast regions, i.e. matK, rpl16 and trnL-F were used. QIAGEN DNA extraction kit was used for DNA isolation in molecular studies. For constructing phylogenetic tree, PAUP and MrBayes softwares were utilized. Chorologies of the species were determined based on the field and herbarium studies, and their distribution maps were given. In this study, Haplophyllum suaveolens var. cilicicum and Haplophyllum suaveolens var. glabrum which were treated as a variety level in the Flora of Turkey have been raised and treated as a species level, as H. cilicicum and H. hatayensis, respectively. Haplophyllum bourgaei and H. villosulum, which were treated as synonymous in Flora of Turkey, have been resurrected. In addition, Haplophyllum ermenekensis Ulukus & Tugay and Haplophyllum megalanthum Bornm. subsp. manisaensis Ulukus & Tugay have been described as new taxa in this study. In consequence of this revision, the genus Haplophyllum is represented with 20 taxa in Turkey, 11 of which are endemic and their endemism ratio is 55 %. As for the genus Ruta, it is represented with 2 taxa as R.. montana and R.. chalepensis in Turkey., Bu tez çalışması Selçuk Üniversitesi, BAP Koordinatörlüğü tarafından 13201023 nolu proje ile desteklenmiştir.

33 5. SUMMARY In this investigation, morphological and anatomical features of species, Ruta uontana(l),Ruta chalepeneie(L) and Haplophyllura buxbaumii sep.buxbauaii, which belong to the Rutaceae family ana which hâs quite dispersed groving area in our country, have been given* Differences such as morphologically pubescent or glabrous of the stem, shape of the leaves, undulate or flat of petals, shape of sepals, position of pedicel in comparison to the capsule, bare or pubescent of the ovarium or size of fruit, have been observed» Anatomically no certain difference has been observed, but on the steffl and root, on starch and drus content of cortex cells, differences have been seen and it has been determined that starch and drus content of cortex cells on the stem and root show similarities* Together with source data, discussion of our findings have been done. 37
