In recent years, concerns have been raised regarding the contamination of grapes with pesticide residues. As consumer demand for safer food products grows, regular monitoring of pesticide residues in food has become essential. This study sought to develop a rapid and sensitive technique for detecting two specific pesticides (phosmet and paraquat) present on the grape surface using the surface‐enhanced Raman spectroscopy (SERS) method. Gold nanostars (AuNS) particles were synthesized, featuring spiky tips that act as hot spots for localized surface plasmon resonance, thereby enhancing Raman signals. Additionally, the roughened surface of AuNS increases the surface area, resulting in improved interactions between the substrate and analyte molecules. Prominent Raman peaks of mixed contaminants were acquired and used to characterize and quantify the pesticides. It was observed that the SERS intensity of the Raman peaks changed in proportion to the concentration ratio of phosmet and paraquat. Moreover, AuNS exhibited superior SERS enhancement compared to gold nanoparticles. The results demonstrate that the lowest detectable concentration for both pesticides on grape surfaces is 0.5 mg/kg. These findings suggest that SERS coupled with AuNS constitutes a practical and promising approach for detecting and quantifying trace contaminants in food. Practical Application: This research established a novel surface‐enhanced Raman spectroscopy (SERS) method coupled with a simplified extraction protocol and gold nanostar substrates to detect trace levels of pesticides in fresh produce. ObjectivePolyethylene glycol-modified gold nanostar particles (GNS-PEG) were constructed to investigate whether the degradation of extracellular matrix in triple-negative breast cancer could improve the tumor delivery of GNS-PEG and enhance the efficacy of photothermal therapy. MethodsGNS-PEG were constructed and characterized for physicochemical properties as well as photothermal properties. At the cellular level, the cytotoxicity of halofuginone (HF) and the effect of photothermal therapy were detected. Mouse model of triple negative breast cancer was established by subcutaneous inoculation of 4T1 cells in BALB/c nude mice. Five injections of HF were given via tail vein (HF group), and tumor sections were stained with Masson stain and immunohistochemical staining for transforming growth factor β1 (TGFβ1), α-smooth muscle actin (α-SMA) and CD31 to observe the effect of tumor stromal degradation. Five injections of HF via tail vein followed by GNS-PEG (HF+ GNS-PEG group) were applied to determine the content of gold in tumor tissues by inductively coupled plasma mass spectrometry. The tumor sites of the mice in the GNS-PEG and HF+ GNS-PEG groups were irradiated with NIR laser and the temperature changes were recorded with an IR camera. The tumour growth and weight changes of mice in each group were observed. Ki-67 immunohistochemical staining, TdT-mediated dUTP nick-end labeling and HE staining were performed on tumor tissue sections from each group to observe tumor proliferation, apoptosis and necrosis. HE staining was performed on heart, liver, spleen, lung and kidney tissues from each group to observe the morphological changes of cells. ResultsGNS-PEG nanoparticles showed a multi-branched structure with a particle size of 73.5±1.4 nm. The absorption peak of GNS was 810 nm, which is in the near infrared region. The photothermal conversion rate of GNS-PEG was up to 79.3%, and the photothermal effect could be controlled by the laser energy. HF has a concentration-dependent cytotoxicity, with a cell survival rate being as low as (22.8±2.6)% at HF concentration of up to 1 000 nmol/L. The photothermal effect of GNS-PEG was significant in killing tumor cells, with a cell survival rate of (32.7±5.2)% at the concentration of 25 pmol/L. The collagen area fraction, TGFβ1 integrated optical density and α-SMA integrated optical density in the tumor tissues of mice in the HF group were (2.1±0.2)%, 3.1±0.4 and 5.2±1.9, respectively, which were lower than those of the control group (all P<0.01), and the vessel diameter was 8.6±2.9 μm, which was higher than that of the control group (P<0.05). In the HF+ GNS-PEG group, the concentration of gold in tissues was 52.4 μg/g, higher than that in the GNS-PEG group (15.9 μg/g, P<0.05). After laser irradiation, the temperature of the tumor site in the HF+ GNS-PEG group was significantly higher than that in the GNS-PEG group. At the 4th minute, the temperatures of the tumor site in the GNS-PEG and HF+ GNS-PEG groups were 51.5 ℃ and 57.7 ℃ respectively; the tumor volume in the HF+ GNS-PEG group was effectively suppressed. The body weights of the mice in each group did not change significantly during the monitoring period. No significant abnormalities were observed in the main organs of the mice in the GNS-PEG group, but some hepatocytes in the HF and HF+ GNS-PEG groups showed edema and degeneration. ConclusionThe remodeling of extracellular matrix in triple-negative breast cancer could significantly improve the intratumoral delivery of GNS-PEG and thus achieve better photothermal therapy effect. Gold nanostars are valuable materials for nanomedicine, energy conversation, and catalysis. Microfluidic synthesis offers a simple and controlled means to produce nanoparticles as they offer precise fluid control and improve heat and mass transfer. 3D-printed microfluidics are a good alternative to PDMS devices because they are affordable to produce and can be more easily integrated with active mixing strategies. 3D-printed microfluidics has only been applied to the production of silver and gold nanospheres, but not complex structures like gold nanostars. Synthesis of gold nanostars requires highly effective mixing to ensure uniform nucleation and growth. In this work, we present a 3D-printed microfluidic device that utilizes an efficient vibrating sharp-tip acoustic mixing system to produce high-quality and reproducible gold nanostars via a seedless and surfactant-free method. The vibrating sharp-tip mixing device can mix three streams of fluid across ~ 300 μm within 7 ms. The device operates with flow rates ranging from 10 μL/min to 750 μL/min at low power requirements (2–45 mW). The optical properties of the resulting nanotars are easily tuned from 650 to 800 nm by modulating the input flow rate. Thus, the presented 3D-printed microfluidic device produces high-quality gold nanostars with tunable optical and physical properties suitable for extensive applications. [ABSTRACT FROM AUTHOR]

A multicolor visual method for the detection of hydrogen peroxide (H2O2) was reported based on the iodide-mediated surface etching of gold nanostar (AuNS). First, AuNS was prepared by a seed-mediated method in a HEPES buffer. AuNS shows two different LSPR absorbance bands at 736 nm and 550 nm, respectively. Multicolor was generated by iodide-mediated surface etching of AuNS in the presence of H2O2. Under the optimized conditions, the absorption peak Δλ had a good linear relationship with the concentration of H2O2 with a linear range from 0.67~66.67 μmol L−1, and the detection limit is 0.44 μmol L−1. It can be used to detect residual H2O2 in tap water samples. This method offered a promising visual method for point-of-care testing of H2O2-related biomarkers. [ABSTRACT FROM AUTHOR]

Abstract A multifunctional nanoplatform with core–shell structure was constructed in one-pot for the synergistic photothermal, photodynamic, and chemotherapy against breast cancer. In the presence of gambogic acid (GA) as the heat-shock protein 90 (HSP90) inhibitor and the gold nanostars (AuNS) as the photothermal reagent, the assembly of Zr4+ with tetrakis (4-carboxyphenyl) porphyrin (TCPP) gave rise to the nanocomposite AuNS@ZrTCPP-GA (AZG), which in turn, further coated with PEGylated liposome (LP) to enhance the stability and biocompatibility, and consequently the antitumor effect of the particle. Upon cellular uptake, the nanoscale metal − organic framework (NMOF) of ZrTCPP in the resulted AuNS@ZrTCPP-GA@LP (AZGL) could be slowly degraded in the weak acidic tumor microenvironment to release AuNS, Zr4+, TCPP, and GA to exert the synergistic treatment of tumors via the combination of AuNS-mediated mild photothermal therapy (PTT) and TCPP-mediated photodynamic therapy (PDT). The introduction of GA serves to reduce the thermal resistance of the cell to re-sensitize PTT and the constructed nanoplatform demonstrated remarkable anti-tumor activity in vitro and in vivo. Simple Summary: Recent advances in the state of the art for early cancer detection and therapy have led to a paradigm shift in the way the disease is detected and treated. In particular, treatments have become much more targeted and localized to minimize systemic body-wide side effects common in traditional treatment methods such as chemotherapy. At the forefront of this revolution is the utilization of plasmonic gold nanoparticles, which have gained increasing attention as a highly effective nanoplatform ranging from drug delivery to plasmonics-enhanced treatments such as photothermal therapy. In this paper, we highlight several advances our laboratory has developed in the pursuit of cancer diagnostics and therapeutics by integrating plasmonics, photonics, and nanotechnology. We discuss the development and applications of plasmonics-active gold nanostar (GNS), a uniquely shaped nanoparticle with numerous branches that serve to greatly amplify the thermal generation at resonant wavelengths. GNS has also been successfully used in tumor imaging contexts from two-photon fluorescence to surface-enhanced Raman scattering (SERS) sensing and imaging. Finally, GNS has been coupled with immunotherapy applications to serve as an effective adjuvant to immune checkpoint inhibitors. This combination of GNS and immunotherapy, the so called synergistic immuno photo nanotherapy (SYMPHONY), has been shown to be effective at controlling long-lasting cancer immunity and metastatic tumors. A novel ratiometric electrochemiluminescence (ECL) system based on gold nanostars (AuNSs) support was constructed for the determination of hypotonicity-induced ATP release from HepG2 cells. AuNS@Lu nanoprobe was used as anodic luminophore and K2S2O8 as cathodic luminophore as well as anodic co-reactant. AuNS with the large specific surface was adopted to adsorb plentiful luminol to form solid-state probe and as affinity support to immobilize ATP aptamer (Apt). The obtained nanocomposite (Apt-AuNS@Lu) generated a strong ECL signal at + 0.4 V (vs. Ag/AgCl) with co-reactant K2S2O8, because of excellent conductivity and catalytic activity of AuNS. Furthermore, graphene oxide was reduced onto indium tin oxide (ITO) electrodes to facilitate the electron transfer. Following, polydopamine (PDA) film was formed via self-polymerization, improving stability and adhesion of the electrode surface. To immobilize ATP capture aptamer (AptC), abounding AuNSs were attached to RGO/PDA surface. When the sensor was incubated in the mixture solution of Apt-AuNS@Lu and target ATP, the ECL signal of Apt-AuNS@Lu increased with the increase of ATP concentration, meanwhile, the signal of K2S2O8 declined. The ratio of the two luminophores was used for the quantitative determination of ATP. The linear range was 5 to 250 nM, and the limit of detection was 1.4 nM at (3σ)/S. The method was successfully applied to analyze ATP release from HepG2 cells stimulated by 0.45% NaCl hypotonic solution. The results showed that the release kinetics profile of ATP had a sigmoidal shape with rapid release within 10 min and then slowed. Compared to the isotonic groups, the intracellular ATP concentration was 3.7 ± 0.3 µM (n = 3) decreasing by 40.3% and the extracellular was 23.4 ± 1.2 nM (n = 3) increasing by 9.2 times in the hypotonicity for 10 min, which showed ATP release from cells and good agreement with commercial ELISA test. High sensitivity and a wide detection range are always the pursuit of sensor design. In this work, gold nanostars (Au NSs) featuring the shape of sea urchins with an absorption peak at the near infrared region (822 nm) were prepared. We proposed a Au NSs-based plasmonic colorimetric sensing platform for ultrasensitive catechol (CC) detection with a wide detection range from 3.33 nM to 107 μM and a limit of detection (LOD) at 1 nM. The target analyte, CC, was used to reduce silver ions (Ag+) to form silver (Ag) coating on the surface of Au NSs, which caused a blue-shift in the localized surface plasmon resonance (LSPR) of Au NSs. With the gradual increase in CC concentration, the Ag coating on the surface was gradually nucleated, and the LSPR blue-shift carried on. This strategy yields a wide LSPR shift by as much as 276 nm for plasmonic effects, enabling an ultra-wide range and the ultrasensitive detection of CC. Octopuses are notable creatures that can dynamically adhere to a variety of substrates owing to the efficient pressure control within their suction cups. An octopus' suckers are sealed at the rim and function by reducing the pressure inside the cavity, thereby creating a pressure difference between the ambient environment and the inner cavity. Inspired by this mechanism, we developed a plasmonic smart adhesive patch (Plasmonic AdPatch) with switchable adhesion in response to both temperature changes and near-infrared (NIR) light. The AdPatch incorporates an elastic, nanohole-patterned elastomer that mimics the structure of octopus suckers. Additionally, a monolayer of gold nanostars (GNSs) is coated on the patch, facilitating a NIR light-responsive photothermal effect. A musclelike, thermoresponsive poly( N -isopropylacrylamide) (PNIPAM) hydrogel functions as a volumetric actuator to regulate cavity pressure. When exposed to heat or light, the PNIPAM hydrogel shrinks, enabling the AdPatch to achieve strong suction adhesion (134 kPa at 45 °C, 71 kPa at 85 mW cm -2 ). Owing to its capability to achieve light-triggered remote adhesion without the need for external pressure, the Plasmonic AdPatch can be employed to transfer ultrathin films and biosensors to fragile organs without causing damage.

Capping agents (organic ligands, polymers, and surfactants) are pivotal for stabilizing nanoparticles; however, they may influence the surface chemistry, as well as the physico-chemical and biological characteristics, of gold nanostar (AuNS)-based biosensors. In this study, we proved that various capping agents affected capped and bioconjugated AuNS stability, functionality, biocatalysis, and colorimetric readouts. Capped and bioconjugated AuNSs were applied as localized surface plasmon resonance (LSPR)-based H2O2 sensors using glucose oxidase (GOx) as a model enzyme. Furthermore, our analyses revealed that the choice of capping agent influenced the properties of the AuNSs, their stability, and their downstream applications. Our analyses provide new insights into factors governing the choice of capping agents for gold nanostars and their influences on downstream applications with conjugated enzymes in confined environments. The main goal of this work was to modify the previously developed blade-type planar structure using plasmonic gold nanostars in order to stimulate photofield emission and provide efficient laser control of the electron current. Localization and enhancement of the field at the tips of gold nanostars provided a significant increase in the tunneling electron current in the experimental sample (both electrical field and photofield emission). Irradiation at a wavelength in the vicinity of the plasmon resonance (red laser) provided a gain in the photoresponse value of up to 5 times compared to irradiation far from the resonance (green laser). The prospects for transition to regimes of structure irradiation by femtosecond laser pulses at the wavelength of surface plasmon resonance, which lead to an increase in the local optical field, are discussed. The kinetics of the energy density of photoinduced hot and thermalized electrons is estimated. Photon upconversion nanomaterials exhibit captivating ability to convert low energy photons to high energy ones, enabling enhanced sensitivity in sensing application. In this perspective, we develop a new photon upconversion based immunosensor for the detection of a heart failure biomarker, Brain Natriuretic Peptide (BNP) by Resonance Energy Transfer (RET) between Polyacrylic acid capped Mn2+ ions doped NaYF 4 :Yb/Er Upconversion Nanoparticles (PAA@Mn-NaYF 4 :Yb/Er UCNPs) as the donor and gold nanostar (AuNS) as the acceptor. BNP antibody was conjugated on the surface of PAA@Mn-NaYF 4 :Yb/Er UCNPs. The immunosensor can produce strong turn on luminescence intensity in proportional to increasing BNP antigen concentration through specific antigen–antibody interactions. The proposed photon upconversion based immunosensor can achieve turn on behaviour in the linear range from 30 pg/mL to 303 pg/mL with a Limit of Detection (LOD) of 1.76 pg/mL. The gold nanostars (GNSs) with sharp branches have shown tunable strong localized surface plasmon resonance (LSPR) peak in the NIR region by varying the size and the shape of the branches which cause to use them for applications such as surface-enhanced Raman spectroscopy (SERS), intracellular imaging and sensing. In this paper, plasmonic nanoprobes based on the GNSs with different morphology have been designed and embedded in the human skin tissue as a surrounding media. The strong Raman scattering has been calculated for GNSs. Furthermore, the high refractive index sensitivity ( RIS ), the effective figure of merit (FOM) enhancement with minimum cellular damage in intracellular imaging, are observed. The RIS and FOM are found to be dependent on the shape and the size of the GNS and the dielectric constant of the surrounding medium. Our results briefly demonstrate the existence of electromagnetic hotspot in the sharp branches of the GNS with the high LSPR peak in the tissue diagnostic window. Moreover, we obtained the high RIS of 856 nm RIU , and a large FOM around 2.1 R I U - 1 with the wide resonance spectra in comparison with previously presented nanoparticles. • A highly sensitive star-shaped SERS substrate was synthesized. • A fast and convenient phthalate detection method was proposed. • LSTM model was established to qualitatively and quantitatively analyze phthalates added to EVOO. • A rapid detection of two phthalates was achieved. Phthalates are commonly used plasticizers in the plastics industry, and have received extensive attention due to their reproductive toxicity. Since phthalates are lipophilic solutions, phthalates can easily migrate from packaging to edible oils. This study synthesized stable and sensitive Gold Nanostars as SERS substrates to conduct qualitative and quantitative analysis of two common phthalates, dibutyl phthalate and di(2-ethylhexyl) phthalate. Two ethanol standard solutions and actual oil solutions of phthalates at different concentrations (10, 5, 1, 0.5, 0.1, 0.02 mg/kg) were prepared. After dimension reduction, LSTM achieved the accuracy of 98% for pure EVOO and EVOO adulterated with different types of phthalates. In terms of quantification, LSTM demonstrates great predictive performance with R p 2 greater than 0.97 and the ratio of performance to deviation greater than 5. The proposed laser-controlled matrix current source is promising for use in X-ray computed tomography systems.

We propose a convenient and easy colorimetric assay for highly sensitive detection of iodide by using gold nanostars (GNSs) as probes. The assay relies on that iodide directly changes the morphology of GNSs and alters their longitudinal localized surface plasmon resonance (LSPR) without surface modifications and the use of other reagents. Upon increasing iodide concentration, GNSs gradually transformed to sphere gold nanoparticles, the absorbance at longitudinal LSPR decreased, and solution color varied from greenish blue to red, as confirmed by the UV-Vis absorption spectroscopy and transmission electron microscopy. With this strategy, as low as 0.005 µM of iodide can be determined due to the specific properties of GNS with plenty of tips and corners and high surface-to-volume ratio. The detection was simply achieved by mixing testing samples and GNS solution. Many ions like CO32-, S2- did not interfere with iodide detection since only iodide can trigger GNS geometry change through an electron injection process. The iodide contents in river water, table salt, seaweed, and complex vitamin tablet were quantified with great accuracy. The proposed assay shows great promises for environment protection and food safety. Moreover, GNSs are useful in developing colorimetric assays for biochemical analysis or clinical diagnosis. Phototherapy is a type of medical treatment that involves exposure to ultraviolet-visible (UV–Vis) region or near-infrared (NIR) region light to treat certain medical conditions. Compared to other treatment methods such as chemotherapy and radiotherapy, it attracts a lot of attention today due to its lower cost, more effect on the tumor cell, and less side effects. Today's clinical therapy research shows that combining chemotherapeutic methods and phototherapeutic methods on a single nanostructure increases the therapeutic effect that each method alone has, and this is even more effective in destroying cancer cells. The emergence of nanomaterials recently opens up new opportunities to increase therapeutic efficacy and overcome limitations in current phototherapy techniques. Abstract Lateral flow immunoassay (LFIA) is a widely used express method and offers advantages such as a short analysis time, simplicity of testing and result evaluation. However, an LFIA based on gold nanospheres lacks the desired sensitivity, thereby limiting its wide applications. In this study, spherical nanogold labels along with new types of nanogold labels such as gold nanopopcorns and nanostars were prepared, characterized, and applied for LFIA of model protein antigen procalcitonin. It was found that the label with a structure close to spherical provided more uniform distribution of specific antibodies on its surface, indicative of its suitability for this type of analysis. LFIA using gold nanopopcorns as a label allowed procalcitonin detection over a linear range of 0.5–10 ng mL−1 with the limit of detection of 0.1 ng mL−1, which was fivefold higher than the sensitivity of the assay with gold nanospheres. Another approach to improve the sensitivity of the assay included the silver enhancement method, which was used to compare the amplification of LFIA for procalcitonin detection. The sensitivity of procalcitonin determination by this method was 10 times better the sensitivity of the conventional LFIA with gold nanosphere as a label. The proposed approach of LFIA based on gold nanopopcorns improved the detection sensitivity without additional steps and prevented the increased consumption of specific reagents (antibodies).

In this experiment, a gold nanostar immunochromatographic test strip for detecting glycinin was established. The detection limit of the test strip was 0.23 ng / mL, the recovery rate was 91.5–96.6 %, and the coefficient of variation was 1.61–6.15 %. In addition, the test strip had no cross reaction with whey protein, wheat protein, peanut protein, sesame protein and β-conglycinin, indicating that the detection method had good specificity and the ability to avoid false positive results. Yolk-shell Au@POP nanocomposites have been constructed and regulated at the atomic level, as high-efficiency electrocatalysts for N 2 -to-NH 3 transformation under mild conditions. Inspired by the catalytic microenvironment of enzymes, a series of yolk-shell nanostructures of monodispersed Au nanostars within porous organic polymers (POPs) have been successfully prepared via a self-sacrificing template strategy. The obtained yolk-shell nanocomposites have highly active Au nanostar cores and permeable POP shells for ambient electrocatalytic NRR to NH 3. The hydrophobic properties of yolk-shell nanoreactors can be tailored accurately by engineering the POP shells at an atomic level, which greatly improves the N 2 fixation performance by inhibiting the hydrogen evolution reaction, resulting in the unprecedented NH 3 yield rate (∼109.1 μg h−1 mg cat. −1) and Faradaic efficiency (∼68.3 %). Objective: Polyethylene glycol-modified gold nanostar particles (GNS-PEG) were constructed to investigate whether the degradation of extracellular matrix in triple-negative breast cancer could improve the tumor delivery of GNS-PEG and enhance the efficacy of photothermal therapy. Methods: GNS-PEG were constructed and characterized for physicochemical properties as well as photothermal properties. At the cellular level, the cytotoxicity of halofuginone (HF) and the effect of photothermal therapy were detected. Mouse model of triple negative breast cancer was established by subcutaneous inoculation of 4T1 cells in BALB/c nude mice. Five injections of HF were given via tail vein (HF group), and tumor sections were stained with Masson stain and immunohistochemical staining for transforming growth factor β1 (TGFβ1), α-smooth muscle actin (α-SMA) and CD31 to observe the effect of tumor stromal degradation. Five injections of HF via tail vein followed by GNS-PEG (HF+ GNS-PEG group) were applied to determine the content of gold in tumor tissues by inductively coupled plasma mass spectrometry. The tumor sites of the mice in the GNS-PEG and HF+ GNS-PEG groups were irradiated with NIR laser and the temperature changes were recorded with an IR camera. The tumour growth and weight changes of mice in each group were observed. Ki-67 immunohistochemical staining, TdT-mediated dUTP nick-end labeling and HE staining were performed on tumor tissue sections from each group to observe tumor proliferation, apoptosis and necrosis. HE staining was performed on heart, liver, spleen, lung and kidney tissues from each group to observe the morphological changes of cells. Results: GNS-PEG nanoparticles showed a multi-branched structure with a particle size of 73.5±1.4 nm. The absorption peak of GNS was 810 nm, which is in the near infrared region. The photothermal conversion rate of GNS-PEG was up to 79.3%, and the photothermal effect could be controlled by the laser energy. HF has a concentration-dependent cytotoxicity, with a cell survival rate being as low as (22.8±2.6)% at HF concentration of up to 1 000 nmol/L. The photothermal effect of GNS-PEG was significant in killing tumor cells, with a cell survival rate of (32.7±5.2)% at the concentration of 25 pmol/L. The collagen area fraction, TGFβ1 integrated optical density and α-SMA integrated optical density in the tumor tissues of mice in the HF group were (2.1±0.2)%, 3.1±0.4 and 5.2±1.9, respectively, which were lower than those of the control group (all P <0.01), and the vessel diameter was 8.6±2.9 μm, which was higher than that of the control group ( P <0.05). In the HF+ GNS-PEG group, the concentration of gold in tissues was 52.4 μg/g, higher than that in the GNS-PEG group (15.9 μg/g, P <0.05). After laser irradiation, the temperature of the tumor site in the HF+ GNS-PEG group was significantly higher than that in the GNS-PEG group. At the 4th minute, the temperatures of the tumor site in the GNS-PEG and HF+ GNS-PEG groups were 51.5 ℃ and 57.7 ℃ respectively; the tumor volume in the HF+ GNS-PEG group was effectively suppressed. The body weights of the mice in each group did not change significantly during the monitoring period. No significant abnormalities were observed in the main organs of the mice in the GNS-PEG group, but some hepatocytes in the HF and HF+ GNS-PEG groups showed edema and degeneration. Conclusion: The remodeling of extracellular matrix in triple-negative breast cancer could significantly improve the intratumoral delivery of GNS-PEG and thus achieve better photothermal therapy effect.

Reliable and sensitive methods to monitor mercury levels in real samples are highly important for environment protection and human health. Herein, a label-free colorimetric sensor for Hg2+ quantitation using gold nanostar (GNS) has been demonstrated, based on the formation of Au-Hg amalgamate that leads to shape-evolution of the GNS and changes in its absorbance. Addition of ascorbic acid (AA) to GNS solution is important for quantitation of Hg2+, mainly because it can reduce Hg2+ to Hg to enhance amalgamation on the GNSs and stabilize GNSs. In addition to transmission electron microscopy images, the distribution of circular ratios of GNSs in the presence of 2 mM AA and various concentrations of Hg2+ are used to show the morphology changes of the GNSs. Upon increasing the concentration of Hg2+, the average circular ratio of GNSs decreases, proving GNS is approaching to sphere. The morphology change alters the longitudinal localized surface plasmonic resonance (LSPR) absorbance of the GNSs significantly. Under the optimum conditions, our sensor exhibits a dynamic response for Hg2+ in the range of 1–4,000 nM with a detection limit of 0.24 nM. Upon Increasing Hg2+ concentration, the solution color changes from greenish-blue, purple to red, which can be distinguished by the naked eye when the Hg2+ concentration is higher than 250 nM. Owing to having a high surface-to-volume ratio and affinity toward Hg0, the GNS is sensitive and selective (at least 50-fold over tested metal ions like Pb2+) toward Hg2+ in the presence of AA. Practicality of this assay has been validated by the analysis of water samples without conducting tedious sample pretreatment.

An important direction in the development of X-ray computed tomography sensors in systems with increased scanning speed and spatial resolution is the creation of an array of miniature current sources. In this paper, we describe a new material based on gold nanostars (GNS) embedded in nanoscale diamond-like carbon (DLC) films (thickness of 20 nm) for constructing a pixel current source with photoinduced electron emission. The effect of localized surface plasmon resonance in GNS on optical properties in the wavelength range from UV to near IR, peculiarities of localization of field and thermal sources, generation of high-energy hot electrons, and mechanisms of their transportation in vacuum are investigated. The advantages of the proposed material and the prospects for using X-ray computed tomography in the matrix source are evaluated.

Abstract We report on the development of a new range-extended bisphenol A (BPA) detection method that uses a surface-enhanced Raman scattering lateral flow assay (SERS-LFA) binary system. The assay includes high-SERS-performance gold nanoparticles, i.e., 40 nm gold nanostars (GNSs) and 4-aminothiolphenol (4-ATP) as a Raman reporter. To demonstrate the performance of this SERS-LFA kit in BPA detection, we tested BPA stock solutions with concentrations ranging from 0.05 to 60 ppb. The GNSs were coated with 4-ATP and then anti-BPA antibody serving as a biorecognition reagent to acquire both visual and Raman signals after application in the BPA SERS-LFA kit. The limit of detection for the SERS signals was 0.073 ppb, which is 205 and 20 times more sensitive than those of visual intensity and color intensity quantifications, respectively. The range of quantification of the SERS signals was doubled that of color intensity quantification. Graphene-mediated surface-enhanced Raman scattering (SERS) substrates are currently explored for ultrasensitive detection and expected to provide uniform SERS response by the virtue of graphene and plasmonic metal nanostructures. Here, we integrated graphene oxide (GO) and anisotropic metal nanostructures to create a novel multiple coupling system, in which ultrathin GO as nanospacer was sandwiched between two layers of closely packed gold nanostars (AuNSts). The sandwiched hybrid was prepared through alternative loading of AuNSts and graphene oxide (GO) film via filtration and spin-coating methods in the absence of any polymer stabilizer or organic linkage agent. The morphologies and plasmon resonance of AuNSts could be tuned by simply adjusting the synthesis parameters. Due to the multi-dimensional plasmonic coupling in horizontally and vertically patterned AuNSts, extra chemical enhancement and outstanding molecule harvesting capability from GO interlayer, the as-prepared AuNSt-GO-AuNSt sandwich structures manifested ultrahigh sensitivity and excellent reproducibility (the signal variations <6%). A detection limit of rhodamine-6G (R6G) as low as 10 −13  M and a high enhancement factor of 6.64 × 10 7 were achieved. Particularly, the AuNSt-GO-AuNSt system was applied to detect erythrosine B and chrysoidin down to 10 −9  M as well as offer spectroscopic identification in complicated solutions, indicating great potential practical applications for the rapid and sensitive on-site monitoring, especially for food and environmental safety. Folic acid targeting moiety facilitated the selective accumulation in cancer cells via receptor-mediated endocytosis. Upon photo irradiation, AuNS@probe demonstrated excellent photothermal effect and induced cell death with apoptosis related mechanism. The typical apoptosis-effector proteases, caspase-3, was subsequently activated and terminated intramolecular fluorescent quenching process. Obvious fluorescence recovery could be applied to precisely assess the activated caspase-3 expression and the real time therapeutic efficacy. Light-triggered nanotheranostics opens a fascinating but challenging avenue to achieve simultaneous and highly efficient anticancer outcomes for multimodal therapeutic and diagnostic modalities. Herein, a multifunctional phototheranostics based on a photosensitizer-assembled graphene/gold nanostar hybrid (GO/AuNS-PEG) was developed for cancer synergistic photodynamic (PDT) and photothermal therapy (PTT) as well as effective photothermal imaging. The stable and biocompatible GO/AuNS-PEG composite displayed a high photothermal conversion efficiency due to the enhanced optical absorbance of both graphene and gold nanostars in the near-infrared (NIR) range. By tuning the absorption wavelength of GO/AuNS-PEG to that of Chlorin e6 (Ce6), GO/AuNS-PEG/Ce6 completely eliminated the EMT6 xenograft tumors by the tremendous synergistic anticancer efficiency of simultaneous PDT and PTT under a single NIR laser irradiation (660 nm) in vivo . The underlying mechanism may be the enhanced cytoplasmic uptake and accumulation of GO/AuNS-PEG/Ce6 and the subsequent photodestruction of the lysosomal membrane and mitochondria. Moreover, GO/AuNS-PEG/Ce6 exhibited negligible side-effects on the body and other organs. These results demonstrate that the graphene/gold nanostar nanoconstruct provides a versatile and reliable integrated platform for the photo-controlled cancer theragnostic applications. Statement of Significance This work demonstrated the application of graphene-Au Nanostars hybridized system (denoted as GO/AuNS-PEG) in single wavelength laser induced synergistic photodynamic (PDT) and photothermal therapy (PTT) and effective cancer photothermal/fluorescence multimode imaging. GO/AuNS-PEG showed excellent biocompatibility and high dual-enhanced photothermal efficiency under the near-infrared laser irradiation that was very promise for deep tumor imaging. By combining with the photosensitizer Chlorin e6, both in vitro and in vivo data confirmed the efficient photoablation of the EMT6 tumors through the synergistic PDT and PTT effect under the activation of a single wavelength laser. In this work, mitochondria-targeting gold nanostar (AuNS) and anticarcinogen DOX were co-encapsulated in hyaluronic acid (HA) protective shell for tumor-targeting synergistic photothermal/chemo-therapy. Cationic peptide R 8 and mitochondria-targeting pro-apoptotic peptide TPP-KLA were co-decorated on AuNS to form AuNS-pep via Au-S bonds. Then, electronegative HA was further coated on the surface via electrostatic interaction for cancer cell targeting. During the coating process, DOX was also introduced via electrostatic interaction to obtain a versatile nanoplatform AuNS-pep/DOX@HA. It was found that the nanoplatform could be internalized into tumor cells via CD44 receptor-mediated recognition. Followed digestion by hyaluronidase (HAase), the therapeutic nanoplatform was able to release DOX for chemotherapy and mitochondria-targeting nanoheater AuNS-pep for near infrared (NIR) light triggered subcellular photothermal therapy (PTT). Bacterial pathogens represent a safety concern in the food industry, and this is amplified by the lack of sensing devices that can be applied on-site by non-trained personnel. In this study, peroxidase-mimicking activity of gold nanostars was exploited to develop a user-friendly colourimetric sensor. A smartphone was exploited as an image reader and analyser, empowered with a novel App developed in-house. The mobile App was evaluated and compared with a commercial smartphone App for its capability to quantify generated colourimetric signals. A major obstacle found with sensors relying on gold nanozymes is the fact that modification of the surface of gold nanoparticles with biorecognition elements generally lead to a suppression of their nanozyme activity. This drawback was overcome by introducing an autocatalytic growth step, which successfully restored the peroxidase-mimicking activity through generation of new gold nanoseeds acting as catalytic centres. A proof-of-concept using this sensing mechanism was developed targeting Mycobacterium bovis , a zoonotic pathogen primarily found in cattle but that can be transmitted to humans by consumption of contaminated food and cause tuberculosis disease. The resulting smartphone-based immunological sensor has shown promising results with a linear response between 104 – 106 CFU/mL, enabling detection of M. bovis at concentrations as low as 7.2·103 CFU/mL in buffer conditions. Graphene aerogel has attracted increasing attention due to its large specific surface area, high-conductivity and electronic interaction. The paper reported a facile synthesis of nitrogen-doped multiple graphene aerogel/gold nanostar (termed as N-doped MGA/GNS) and its use as the electrochemical sensing platform for detection of double stranded (dsDNA). On the one hand, the N-doped MGA offers a much better electrochemical performance compared with classical graphene aerogel. Interestingly, the performance can be enhanced by only increasing the cycle number of graphene oxide gelation. On the other hand, the hybridization with GNS further enhances the electrocatalytic activity towards F e ( C N ) 6 3 − / 4 − . In addition, the N-doped MGA/GNS provides a well-defined three-dimensional architecture. The unique structure make it is easy to combine with dsDNA to form the electroactive bioconjugate. The integration not only triggers an ultrafast DNA electron and charge transfer, but also realizes a significant synergy between N-doped MGA, GNS and dsDNA. As a result, the electrochemical sensor based on the hybrid exhibits highly sensitive differential pulse voltammetric response (DPV) towards dsDNA. The DPV signal linearly increases with the increase of dsDNA concentration in the range from 1.0×10 − 21 g ml − 1 to 1.0×10 −16 g ml −1 with the detection limit of 3.9×10 −22 g ml −1 ( S / N =3). The sensitivity is much more than that of all reported DNA sensors. The analytical method was successfully applied in the electrochemical detection of circulating free DNA in human serum. Gold nanomaterials possess unique physical and chemical properties, which attracted much attention in recent years. As a new type of gold nanomaterials, gold nanostars (GNSTs) have been prepared and characterized in this study. GNSTs under near-infrared (NIR) light irradiation can exert not only cancer photothermal therapy via heat production but also photodynamic therapy via generation of reactive oxygen species. GNSTs were able to enter the cytoplasm as well as nuclei of human breast michigan cancer foundation-7 (MCF-7) cells. Under NIR light irradiation, GNSTs caused more severe DNA damage, arrest the cell cycle in G0/G1 phase, and reduce more cellular glutathione level, causing more severe apoptosis and cell death in vitro. Intratumoral injection of GNSTs with NIR light irradiation significantly inhibited tumor growth in vivo. In addition, GNSTs were demonstrated to be a contrast agent for X-ray imaging. Gold nanoparticles are materials with unique optical properties that have made them very attractive for numerous biomedical applications. With the increasing discovery of techniques to synthesize novel nanoparticles such as star-shaped gold nanoparticles for biomedical applications, the safety and performance of these new nanomaterials must be systematically assessed before use. In this study, gold nanostars (AuNSTs) with multibranched surface structures were synthesized, and their influence on the cytotoxicity of human skin fibroblasts and rat fat pad endothelial cells (RFPECs) were assessed and compared with that of gold nanospheres (AuNSPs) with unbranched surfaces. Results showed that the AuNSPs with diameters of approximately 61.46 nm showed greater toxicity with fibroblast cells and RFPECs compared with the synthesized AuNSTs with diameters of approximately 33.69 nm. The AuNSPs were lethal at concentrations of 40 μg/mL for both cell lines, whereas the AuNSTs were less toxic at higher concentrations (400 μg/mL). The calculated IC50 (50% inhibitory concentration) values of the AuNSPs exposed to fibroblast cells were greater at 1 and 4 days of culture (26.4 and 27.7 μg/mL, respectively) compared with the RFPECs (13.6 and 13.8 μg/mL, respectively), indicating that the AuNSPs have a greater toxicity to endothelial cells. It was proposed that possible factors that could be promoting the reduced toxicity effects of the AuNSTs to fibroblast cells and RFPECs, compared with the AuNSPs may be size, surface chemistry, and shape of the gold nanoparticles. The three-dimensional (3D) polarization-dependent optical property is one distinctive characteristic of the anisotropic metal nanoparticles. This has the potential applications in 3D polarization information encryption to detect the polarization information and single particle 3D orientation determination. Previous studies almost focus on the condition that the polarization orientation is in-plane. In the present paper, we systematically investigate the 3D polarization-dependent near- and far-field properties of a kind of fascinating anisotropic nanoparticle, i.e., gold nanostar (AuNS). It shows that the scattering spectra of the AuNS induced by arbitrary incident polarizations can be described as a linear superposition of a set of basic scattering spectra. And we introduce a trigonometric equation to reveal the inherent relationship between the scattering intensity and the 3D polarization orientation, which is universally independent of the size, shape, or surrounding medium of the nanostar. Furthermore, it is facile to synthesize AuNS with two pairs of orthogonal different-length arms, based on the 3D polarization dependence of the scattering trait, and using coordinate transformation, we have demonstrated that such AuNS is useful for designing single particle 3D orientation and rotational tracking sensor and orientation-unlimited polarization information detector. And the self-rotation problem of the gold nanorod, i.e., rotation around the gold nanorod's long axis cannot be resolved, can be solved by replacing it with AuNS. Gold nanomaterials promise a wide range of potential applications in chemical and biological sensing, imaging, and catalysis. In this paper, we demonstrate a facile method for room-temperature synthesis of gold nanostars (AuNSs) with a size of ~50 nm via seeded growth. Significantly, the AuNSs are found to have high light-scattering properties, which are successfully used as labels for sensitive and selective detection of nucleic acids and proteins by using exonuclease III (Exo III) as a biocatalyst. For DNA detection, the binding of targets to the functionalized AuNS probes leads to the Exo III-stimulated cascade recycling amplification. As a result, a large amount of AuNSs are released from magnetic nanoparticles (MNPs) into solution, providing a greatly enhanced light-scattering signal for amplified sensing process. Moreover, a binding-induced DNA three-way junction (DNA TWJ) is introduced to thrombin detection, in which the binding of two aptamers to thrombin triggers assembly of the DNA motifs and initiates the subsequent DNA strand displacement reaction (SDR) and Exo III-assisted cascade recycling amplification. The detection limits of 89 fM and 5.6 pM are achieved for DNA and thrombin, respectively, which are comparable to or even exceed that of the reported isothermal amplification methods. It is noteworthy that based on the DNA TWJ strategy the sequences are independent on target proteins. Additionally, the employment of MNPs in the assays can not only simplify the operations but also improve the detection sensitivity. Therefore, the proposed amplified light-scattering assay with high sensitivity and selectivity, acceptable accuracy, and satisfactory versatility of analytes provides various applications in bioanalysis.
