In this PhD thesis, fluorescent molecules or nanoparticles were investigated to probe the pore space of heterogeneous catalysts and mass transport therein. The fluorescent emitters (guests) report on the properties of the catalyst (hosts) via guest–host interactions. These interactions can be a resistance on the movement of the probes by the pore space, reversible and irreversible adsorption of probes on the pore wall, or a chemical reaction between the guest and host. It is advantageous to follow reporters via their fluorescence because it can be detected with high sensitivity, e.g., to determine the location of the reporter, which makes it possible to probe individual molecules or nanoparticles. A second advantage is that the fluorescence emission spectrum is often dependent on the chemical identity of the fluorescent reporter. Thus, chemical reactions of the guest with the host can be followed directly via the guest’s emission spectrum. Analysis of the trajectories obtained with single-molecule tracking in inorganic porous hosts is often challenging because trajectories are short and/or motion is heterogeneous. In Chapter 3, we presented the software DiffusionLab for motion analysis of such challenging datasets. This approach relies on the pooling of trajectories with a similar motion behaviour into a population. The average motion characteristics of a population can be computed and compared without losing information on the single-molecule level about, e.g., the spatial distribution of adsorption. In Chapter 4, we introduced a microfluidic device designed for the characterization of fluorescent reporters in confinement. The device consists of a two-dimensional model pore with a height of 50 nm. This design allowed for the measurement of long trajectories, which facilitated detailed probe characterization. We investigated guest–host adsorption of single quantum-dot emitters to the pore wall and guest diffusion through the model pore. Building on the probe characterization, we defined conditions that allowed for mapping of the accessible pore space of a model-pore array and a real-life catalyst particle. We studied molecular diffusion in the straight and the sinusoidal pores of ZSM-5 zeolites in Chapter 5. The pore types have a similar effective pore size but a different shape. Both the mobility, when the molecules were not adsorbed over hundreds of milliseconds, and the frequency of adsorption was different in the straight and sinusoidal pores. Additional meso- and macropores with a diameter ≥ 2 nm were etched into the zeolite material to promote diffusion. The molecular mobility increased and the adsorption frequency of the molecules decreased in the sinusoidal pores, which indicated the formation of an interconnected secondary pore network. In Chapter 6, we studied the guest–host interactions of fluorescent resorufin molecules inside the micropores of ~10 micrometre-sized zeolite-β crystals. We found that resorufin molecules were protonated after entering the zeolite, which provided contrast between the resorufin species inside the zeolite micropores and in aqueous solution. Using this effect, it was shown that the micropores in the crystals were fully accessible. Moreover, we visualised that diffusion through the straight pores of zeolite-β was impeded when diffusing through the boundaries between the zeolite subunits.

It starts with the capture of a photon by one of the pigments embedded within one of the two photosystems, Photosystem I (PSI) or II (PSII). These photosystems are large assemblies of many pigments held together by the protein scaffold. The absorption of the photon brings the pigment to an electronic excited state. The excitation energy is then transferred from pigment-to-pigment to the reaction center (RC) of the photosystem, where it is used to perform charge separation (CS). The pigment-to-pigment energy transfer within photosynthetic complexes occurs on a very fast, femtosecond (fs, 10^(-15) second) to picosecond (ps, 10^(-12) second) timescale, which ensures that the photosystems are extremely efficient in using the energy for charge separation. In this thesis, aspects of the light-harvesting of photosynthetic pigment-protein complexes were investigated. The spectroscopic properties (absorption, emission) and energy-transfer processes were studied with a variety of different techniques, including advanced ultrafast time-resolved spectroscopic methods (two-dimensional electronic spectroscopy (2DES) and time-resolved fluorescence spectroscopy). In these time-resolved experiments, the complexes are excited with ultrashort (fs temporal width) pulses of light, after which the optical response (photon-echo, fluorescence) is monitored in time. By measuring these signals, excitation energy transfer (EET) and energy trapping within these complexes can be determined. Oxygenic photosynthesis is mainly powered by visible light in the 400–700 nm range. Expanding the absorption range to 750 nm would result in 19% more photons available for photosynthesis [Chen, M. & Blankenship, R. E. (2011) Trends Plant Sci., 16, 427–431]. Moreover, improved far-red light-harvesting can be advantageous in shaded environments. For these reasons extention of the absorption beyond the 400–700 nm range is an important approach in the global aim to improve crop productivity to meet the increasing global demands for food production. This thesis focuses on the far-red light-harvesting properties of PSI from higher plants and cyanobacteria. The aim is to understand underlying aspects that are important for far-red light (FRL, 700–800 nm) absorption and EET within PSI. These aspects can be useful to enhance the far-red light-harvesting in photosynthesis of other organism, such as plants. The chapters of this thesis contain several insights that can be generally important in the goal to enhance the far-red light-harvesting abilities of photosynthetic complexes. The integration of new low-energy states, such as from long-wavelength Chlorophylls (Chls) or new Chl a red form states, is a viable strategy to enhance the absorption of far-red light of these complexes. However, additional alterations to the light-harvesting mechanism may be required to obtain a highly efficient complex with optimally enhanced far-red light absorption. Notably, as shown by the investigated natural (light-harvesting complex I and FRL-specific PSI complex) and artificial (Chl f-containing hybrid PSI) complexes the protein scaffold is a determining factor in optimization of the far-red light-harvesting properties of photosynthetic complexes. Conclusively, in this thesis we provide several important lessons for the aim to effectively enhance the far-red light-harvesting capacity of other photosynthetic organisms.

During the last decades, fluorescence spectroscopy has emerged as a powerful tool in the fields of biophysics, biotechnology, biochemistry, cellular biology and the medical sciences. These techniques are highly sensitive, and allow us to study the structure and dynamics of (bio)molecular systems (Valeur 2001). A significant advantage of fluorescence techniques is that they can often be non-invasive and measurements can be performed in real time. In this thesis different advanced fluorescence methods will be used to study two important biological processes: (1) DNA dynamics and (2) plant photosynthesis. The first part aims at improving the smFRET technique for the analysis of DNA dynamics and other fast conformational changes. This improvement is made by combining and developing instrumentation and data evaluation tools. The second part is the continuous development of time-resolved fluorescence spectroscopy methods, as well their application in the field of photosynthesis to study ultrafast processes in thylakoid membranes and leaves. The two fluorescence techniques are technically and conceptually very different, but they are both designed for analysis of biomolecular systems. In this thesis, the techniques are applied to study energy transfer and dynamical changes in DNAs, thylakoid membranes and leaves. REFERENCE: VALEUR B 2001. The basic process for crop growth and production is photosynthesis. Measuring crop photosynthesis is therefore important to monitor the status of the crop and whether the greenhouse climate is set to the needs of the crop. In this project, two monitoring systems for crop photosynthesis were developed and tested. (1) The crop photosynthesis monitor is a soft sensor that can calculate the CO2 uptake of an entire crop. The basis for these calculations are the balance between CO2 supply and CO2 loss via ventilation and crop photosynthesis. By measuring the CO2 concentration and humidity inside and outside the greenhouse, the crop photosynthesis can be calculated. (2) The CropObserver is a fluorescence sensor that measures the light use efficiency of photosynthesis of a large crop area (3 x 3 m2). The crop receives light pulses from a laser in the top of the greenhouse, the sensor measures the fluorescence signal of the crop. Both sensors were tested in a tomato crop in 2014 with promising results. The sensors functioned without problems and delivered patterns of daily photosynthesis which matched the reference measurements reasonably well up to well.

Het meten van de gewasfotosynthese is dus van belang om goed te monitoren hoe het gewas er voor staat en of het kasklimaat is ingesteld naar de behoefte van het gewas. In dit project zijn twee meetsystemen ontwikkeld en getest om de gewasfotosynthese te meten. (1) De gewasfotosynthesemonitor is een soft sensor die de CO2 opname van een hele kas met gewas kan berekenen. Dit doet deze monitor op basis van de balans tussen enerzijds CO2 dosering en anderzijds verlies van CO2 door ventilatie en gewasfotosynthese. Door de CO2 concentratie en de luchtvochtigheid binnen en buiten de kas te meten kan de gewasfotosynthese berekend worden. (2) De nieuwe fluorescentiesensor die ontwikkeld is, de CropObserver, meet de efficiëntie van de lichtbenutting als maat voor de fotosynthese van een groot oppervlakte gewas (3 x 3 m2). Het gewas wordt belicht met korte laserlichtpulsen bovenin de kas, de sensor meet het terugkomende fluorescentiesignaal. Beide nieuwe sensoren zijn begin 2014 getest in een tomatengewas. De eerste testen waren veelbelovend. De sensoren hebben zonder problemen gefunctioneerd en leverden een dagverloop van de fotosynthese op, dat redelijk tot goed overeen kwam met referentiemetingen.

Terrestrial gross primary production (GPP) is an important parameter to explore and quantify carbon fixation by plant ecosystems at various scales. Remote sensing (RS) offers a unique possibility to investigate GPP in a spatially explicit fashion; however, budgeting of terrestrial carbon cycles based on this approach still remains uncertain. To improve calculations, spatio-temporal variability of GPP must be investigated in more detail on local and regional scales. The overarching goal of this study is to enhance our knowledge on how environmentally induced changes of photosynthetic light-use efficiency (LUE) are linked with optical RS parameters. Diurnal courses of sun-induced fluorescence yield (FSyield) and the photochemical reflectance index of corn were derived from high-resolution spectrometric measurements and their potential as proxies for LUE was investigated. GPP was modeled using Monteith's LUE-concept and optical-based GPP and LUE values were compared with synoptically acquired eddy covariance data. It is shown that the diurnal response of complex physiological regulation of photosynthesis can be tracked reliably with the sun-induced fluorescence. Considering structural and physiological effects, this research shows for the first time that including sun-induced fluorescence into modeling approaches improves their results in predicting diurnal courses of GPP. Our results support the hypothesis that air- or spaceborne quantification of sun-induced fluorescence yield may become a powerful tool to better understand spatio-temporal variations of fluorescence yield, photosynthetic efficiency and plant stress on a global scale

To assess the mechanisms causing genotypic differences in heat tolerance of wheat (Triticum aestivum L.), physiological responses to a heat shock in a vegetative (‘end of tillering’) or a reproductive (‘early grain filling’) stage were studied. Three cultivars — Lavett, Ciano-79 and Attila — differing in adaptation to heat were grown in a glasshouse at a day/night temperature regime of 15/10 °C and a 12-h daylength from sowing to ‘end of tillering’ and next at two day/night regimes of 25/20 and 18/13 °C under natural daylength. The heat-shock treatment consisted of an exposure of plants to temperatures raised gradually over a time-span of 12 hours to above 30 °C with a maximum of 38 °C during three hours at midday for three days either at the ‘end of tillering’ or at ‘grain filling’. A heat shock at the ‘end of tillering’ strongly reduced the rate of leaf photosynthesis. A similar heat shock during ‘grain filling’ decreased both rate of photosynthesis (source) and grain growth (sink). The rate of leaf photosynthesis was decreased by 40 to 70%, depending on cultivar and developmental stage. Photosynthesis fully recovered within 4 days after the heat-shock treatment was ended. The effects of the heat shock on biomass yield were more pronounced for treatments at ‘early grain filling’ than at ‘end of tillering’. However, the impact of a 3-day heat shock on biomass yield was less than the effects of the pre- and post-treatment growing temperature.

Summary The possible mechanisms for reorganisation of outer LHCs of PSII (LHCII) upon state transitions in Chlamydomonas reinhardtii have been discussed for several decades [38, 43-54]. For a long time people adhered to the opinion that upon the transition from state 1 to state 2, 80% of LHCII detaches from PSII and attaches completely to PSI in Chlamydomonas reinhardtii [38, 45]. This thesis provides new insights for the mechanism of state transitions in Chlamydomonas reinhardtii. In the remainder of this thesis, the role of minor light-harvesting complexes in excitation energy transfer to reaction centers of photosystem II are discussed as well as multiexciton dynamics of the alloyed ZnCdTe quantum dots are studied in detail. In chapter 2, we demonstrate with picosecond-fluorescence spectroscopy on C. reinhardtii cells that although LHCs indeed detach from Photosystem II in state-2 conditions only a fraction attaches to Photosystem I. The detached antenna complexes become protected against photodamage via shortening of the excited-state lifetime. It is discussed how the transition from state 1 to state 2 can protect C. reinhardtii in high-light conditions and how this differs from the situation in plants. In chapter 3, we study the picosecond fluorescence properties of Chlamydomonas reinhardtti over a broad range of wavelengths at 77K. It is observed that upon going from state 1 (relatively high 680nm/720nm fluorescence ratio) to state 2 (low ratio), a large part of the fluorescence of LHC/PSII becomes substantially quenched, probably because of LHC detachment from PSII, whereas the fluorescence of PSI hardly changes. These results are in agreement with the proposal in chapter 2 that the amount of LHC moving from PSII to PSI upon going from state 1 to state 2 is very limited. In chapter 4, we used picosecond-fluorescence spectroscopy to study excitation-energy transfer (EET) in thylakoids membranes isolated from A. thaliana wild-type plants and knockout lines depleted of either two (koCP26/24 and koCP29/24) or all minor Lhcs (NoM). In the absence of all minor Lhcs, the functional connection of LHCII to the PSII cores appears to be seriously impaired whereas the “disconnected” LHCII is substantially quenched. For both double knock-out mutants, excitation trapping in PSII is faster than in NoM thylakoids but slower than in WT thylakoids. In NoM thylakoids, the loss of all minor Lhcs is accompanied by an over-accumulation of LHCII, suggesting a compensating response to the reduced trapping efficiency in limiting light, which leads to a photosynthetic phenotype resembling that of low-light-acclimated plants. Finally, fluorescence kinetics and biochemical results show that the missing minor complexes are not replaced by other Lhcs, implying that they are unique among the antenna subunits and crucial for the functioning and macro-organization of PSII. In chapter 5, we have performed picosecond fluorescence measurements on ZnCdTe ternary quantum dots at room temperature by using a streak-camera setup in order to investigate in detail the fluorescence kinetics for ZnCdTe quantum dots with different size and structure by using different excitation laser intensities. Our data show that the changes in fluorescence kinetics are mostly related to the changes in structure and size. In heterogeneous structured ZnCdTe quantum dots, the fluorescence kinetics become faster as compared to homogeneous structured ZnCdTe quantum dots. Also, in both homogeneous and heterogeneous ZnCdTe quantum dots, a new peak is observed in the high-energy region of the emission spectrum when using high excitation intensities, which shows that the radiative processes that occur from higher energy states become more favoured as the excitation intensity increases.

The photosynthetic process can hardly be bettered. But the utilisation of natural or artificial light certainly leaves room for improvement. In recent years our understanding of light has grown considerably and this has major implications on how we deal with light in horticulture.

Oral coatings are residues of food and beverages that coat the oral mucosa after consumption. Several studies have reported on the lubrication properties in mouth, and the after-feel and after-taste impact of oral coatings. Further, oral coatings have been suggested to influence subsequent taste perception. Although it is well known that oral coatings can influence sensory perception, there was little information available on the chemical composition and physical properties of oral coatings. As such, the aim of this thesis was to understand which factors influence the composition of oral coatings and their sensory perception. This study started with the development of an appropriate calibration method for an already described methodology to quantify oil oral coatings: in vivo fluorescence. Further, the samples studied were shifted from pure oil (used on previous studies) to a more realistic food beverage: o/w emulsions. Pig´s tongues are known to be a good model of human tongue. As such, Chapter 2 used pig´s tongues on the calibration of the method, to mimic the fluorescence in mouth of oil coatings. On chapter 2, Confocal Scanning Laser Microscopy images showed that stable o/w emulsions (1-20% (w/w)) stabilised by Na-caseinate created individual oil droplets on the surface of the pigs tongue, as such a new descriptor for oil coatings was developed. Oil fraction, i.e. mass of oil per surface area of the tongue, was shown to be higher on the back compared to the front anterior part of the tongue. This is thought to be due to the morphology of the tongue and abrasion of the oil coating owed to the rubbing with the palate. Further, in vivo measurements showed that oil fraction deposited on the tongue increased linearly with oil content of o/w emulsions. Coating clearance from the tongue was a fast process with around 60% of the oil being removed on the first 45s. After-feel perception (Fatty Film and Flavour Intensity) was shown to be semi-logarithmic related to oil fraction on the tongue. Chapter 3, further investigated different properties of 10% (w/w) o/w emulsions that influence the oil fraction deposited on the tongue, its clearance and after-feel perception. Three different properties were studied: protein type, protein content and viscosity of the o/w emulsions. To study the influence of protein type, two different proteins which behave differently in-mouth were studied: Na-caseinate - creates emulsions which do not flocculate under in mouth conditions, and lysozyme – creates emulsions which flocculate under in mouth conditions. To study the influence of protein content, three concentrations of Na-caseinate and lysozyme were used (0.2, 3, 5.8% (w/w) all in excess to stabilize the water/oil interface). To study the influence of viscosity of o/w emulsions, three o/w emulsions stabilized with 3% (w/w) Na-caseinate were thickened with varying concentrations of xanthan gum (0-0.5%) (w/w). Generally, the irreversible flocculation of lysozyme stabilized emulsions with saliva did not create a significant difference on oil deposition compared to emulsions stabilized with Na-caseinate, immediately after expectoration of the emulsions. Nevertheless, lysozyme stabilised emulsions caused slower oil clearance from the tongue surface compared to emulsions stabilized with Na-caseinate. Protein content had a negative relation with oil fraction on the tongue for lysozyme stabilized emulsions and no relation for Na-caseinate stabilized emulsions. The presence of thickener decreased deposition of oil on tongue, although viscosity differences (i.e., thickener content) did not affect oil fraction. After-feel perception of creaminess and fatty-film was strongly influenced by the presence of thickener likely due to lubrication in-mouth, i.e., the higher the concentration of thickener in the emulsions the stronger was the perception. Oral coatings perception was further influenced by the protein used in the emulsions, with Na-caseinate stabilised emulsions creating coatings with higher perception on creaminess and fatty-film. Chapter 2 and chapter 3 provided knowledge on the deposition and clearance of oil coatings, but little was known on the formation of oil coatings. Chapter 4 focused on the formation of oil coatings formed by Na-caseinate stabilised o/w emulsions (1-20% (w/w)). The formation of oil coatings was a rapid process, where the maximum oil deposition was achieved at normal drinking behaviour (~3s). Further, in Chapter 4 we investigated the hypothesis often referred on literature, in which oil coatings form a physical barrier which prevents tastants to reach the taste buds, and thus create a reduction on taste perception. It was concluded that oil coatings formed by emulsions within one sip did not affect subsequent sweetness perception of sucrose solutions. We suggested that the oil droplets deposited on the tongue (as seen on chapter 2) did not form a hydrophobic barrier that is sufficient to reduce the accessibility of sucrose to the taste buds and consequently does not suppress taste perception. Previous chapters focused on oral coatings formed by liquid o/w emulsions, however studies describing oral coatings formed by semi-solids and solids are scarce. As such, chapter 5 focused on the formation, clearance and sensory perception of fat coatings from emulsion-filled gels. Four emulsion-filled gelatin gels varying in fat content and type of emulsifier (whey protein isolate - created fat droplets bound to matrix; tween 20 - created fat droplets unbound to matrix) were studied. As in for oil coatings formed by liquid o/w emulsions, fat coatings formed by emulsion-filled gels reach their maximum deposition in the first seconds of mastication. This suggests that the first bites are the most relevant for the formation of fat coatings on the tongue. Further, fat fraction deposited on tongue increased when oral processing time of the gels increased. This trend was clearer for gels with higher fat content (15%) compared to gels with lower fat content (5%). Fatty perception increased with increasing mastication time, and decreased after expectoration with increasing clearance time. Fat fraction deposited on tongue and fatty perception are higher in gels with unbound droplets compared to bound droplets, as well as in gels with 15% fat compared to 5% fat. To elucidate the role of protein on oral coatings, Chapter 6 focused on the development of a method to quantify protein in the oral coatings. Further, Chapter 6 studied the influence of protein content, in-mouth protein behaviour (lysozyme - protein which creates flocs with saliva vs. Na-Caseinate - protein which does not create flocs with saliva) and presence of thickener on the formation of protein oral coatings and sensory perception of protein coatings. Protein coatings were collected from the front and middle part of the anterior tongue using cotton swabs after subjects orally processed protein solutions for different time periods. Protein concentration of the coating (mass protein/mass coating) was quantified with the Lowry method. Similarly to oil/fat coatings, results show protein coatings are formed rapidly, reaching maximum deposition on the first seconds of the samples´ oral processing. Further, different protein in mouth-behaviour (Na-caseinate vs. lysozyme) did not create differences on protein deposition on the tongue. Presence of xanthan-gum in the processed samples decreased protein deposition on the tongue, compared to when samples without xanthan-gum were processed. The perception of protein coatings was strongly influenced by the viscosity and protein used in the samples. Higher viscosity of the samples lead to higher intensity on creaminess and thickness. Lysozyme samples created coatings with high sweetness and astringent intensity, which is related to the molecular structure of the protein. Changes in the viscosity of beverages can cause changes in thickness perception. The changes in thickness perception can be accompanied by differences in other sensory properties, such as sweetness and creaminess which might be undesirable when reformulating beverages or developing new products. Knowledge on the differences by which viscosity of beverages can be modified to create a difference in sensory perception is currently lacking. Chapter 7 focus on the determination of the Just Noticeable Difference (the minimal difference that can be detected between two stimuli) for thickness perception of beverages. Oral thickness sensitivity (K=0.26) was found to be comparable to literature values for kinesthetic food firmness and spreadability, creaminess, sourness and bitterness perception. The aim of this thesis was to determine and characterize factors influencing oral coatings and their sensory perception. For this purpose, reliable methods to quantify oil and protein deposited on the tongue had to be developed to later study the macronutrients deposition. Further, the influence of stimulus properties on the formation and clearance dynamics of oral coatings and their impact on sensory perception were investigated.

Summary The possible mechanisms for reorganisation of outer LHCs of PSII (LHCII) upon state transitions in Chlamydomonas reinhardtii have been discussed for several decades [38, 43-54]. For a long time people adhered to the opinion that upon the transition from state 1 to state 2, 80% of LHCII detaches from PSII and attaches completely to PSI in Chlamydomonas reinhardtii [38, 45]. This thesis provides new insights for the mechanism of state transitions in Chlamydomonas reinhardtii. In the remainder of this thesis, the role of minor light-harvesting complexes in excitation energy transfer to reaction centers of photosystem II are discussed as well as multiexciton dynamics of the alloyed ZnCdTe quantum dots are studied in detail. In chapter 2, we demonstrate with picosecond-fluorescence spectroscopy on C. reinhardtii cells that although LHCs indeed detach from Photosystem II in state-2 conditions only a fraction attaches to Photosystem I. The detached antenna complexes become protected against photodamage via shortening of the excited-state lifetime. It is discussed how the transition from state 1 to state 2 can protect C. reinhardtii in high-light conditions and how this differs from the situation in plants. In chapter 3, we study the picosecond fluorescence properties of Chlamydomonas reinhardtti over a broad range of wavelengths at 77K. It is observed that upon going from state 1 (relatively high 680nm/720nm fluorescence ratio) to state 2 (low ratio), a large part of the fluorescence of LHC/PSII becomes substantially quenched, probably because of LHC detachment from PSII, whereas the fluorescence of PSI hardly changes. These results are in agreement with the proposal in chapter 2 that the amount of LHC moving from PSII to PSI upon going from state 1 to state 2 is very limited. In chapter 4, we used picosecond-fluorescence spectroscopy to study excitation-energy transfer (EET) in thylakoids membranes isolated from A. thaliana wild-type plants and knockout lines depleted of either two (koCP26/24 and koCP29/24) or all minor Lhcs (NoM). In the absence of all minor Lhcs, the functional connection of LHCII to the PSII cores appears to be seriously impaired whereas the “disconnected” LHCII is substantially quenched. For both double knock-out mutants, excitation trapping in PSII is faster than in NoM thylakoids but slower than in WT thylakoids. In NoM thylakoids, the loss of all minor Lhcs is accompanied by an over-accumulation of LHCII, suggesting a compensating response to the reduced trapping efficiency in limiting light, which leads to a photosynthetic phenotype resembling that of low-light-acclimated plants. Finally, fluorescence kinetics and biochemical results show that the missing minor complexes are not replaced by other Lhcs, implying that they are unique among the antenna subunits and crucial for the functioning and macro-organization of PSII. In chapter 5, we have performed picosecond fluorescence measurements on ZnCdTe ternary quantum dots at room temperature by using a streak-camera setup in order to investigate in detail the fluorescence kinetics for ZnCdTe quantum dots with different size and structure by using different excitation laser intensities. Our data show that the changes in fluorescence kinetics are mostly related to the changes in structure and size. In heterogeneous structured ZnCdTe quantum dots, the fluorescence kinetics become faster as compared to homogeneous structured ZnCdTe quantum dots. Also, in both homogeneous and heterogeneous ZnCdTe quantum dots, a new peak is observed in the high-energy region of the emission spectrum when using high excitation intensities, which shows that the radiative processes that occur from higher energy states become more favoured as the excitation intensity increases.

Oral coatings are residues of food and beverages that coat the oral mucosa after consumption. Several studies have reported on the lubrication properties in mouth, and the after-feel and after-taste impact of oral coatings. Further, oral coatings have been suggested to influence subsequent taste perception. Although it is well known that oral coatings can influence sensory perception, there was little information available on the chemical composition and physical properties of oral coatings. As such, the aim of this thesis was to understand which factors influence the composition of oral coatings and their sensory perception. This study started with the development of an appropriate calibration method for an already described methodology to quantify oil oral coatings: in vivo fluorescence. Further, the samples studied were shifted from pure oil (used on previous studies) to a more realistic food beverage: o/w emulsions. Pig´s tongues are known to be a good model of human tongue. As such, Chapter 2 used pig´s tongues on the calibration of the method, to mimic the fluorescence in mouth of oil coatings. On chapter 2, Confocal Scanning Laser Microscopy images showed that stable o/w emulsions (1-20% (w/w)) stabilised by Na-caseinate created individual oil droplets on the surface of the pigs tongue, as such a new descriptor for oil coatings was developed. Oil fraction, i.e. mass of oil per surface area of the tongue, was shown to be higher on the back compared to the front anterior part of the tongue. This is thought to be due to the morphology of the tongue and abrasion of the oil coating owed to the rubbing with the palate. Further, in vivo measurements showed that oil fraction deposited on the tongue increased linearly with oil content of o/w emulsions. Coating clearance from the tongue was a fast process with around 60% of the oil being removed on the first 45s. After-feel perception (Fatty Film and Flavour Intensity) was shown to be semi-logarithmic related to oil fraction on the tongue. Chapter 3, further investigated different properties of 10% (w/w) o/w emulsions that influence the oil fraction deposited on the tongue, its clearance and after-feel perception. Three different properties were studied: protein type, protein content and viscosity of the o/w emulsions. To study the influence of protein type, two different proteins which behave differently in-mouth were studied: Na-caseinate - creates emulsions which do not flocculate under in mouth conditions, and lysozyme – creates emulsions which flocculate under in mouth conditions. To study the influence of protein content, three concentrations of Na-caseinate and lysozyme were used (0.2, 3, 5.8% (w/w) all in excess to stabilize the water/oil interface). To study the influence of viscosity of o/w emulsions, three o/w emulsions stabilized with 3% (w/w) Na-caseinate were thickened with varying concentrations of xanthan gum (0-0.5%) (w/w). Generally, the irreversible flocculation of lysozyme stabilized emulsions with saliva did not create a significant difference on oil deposition compared to emulsions stabilized with Na-caseinate, immediately after expectoration of the emulsions. Nevertheless, lysozyme stabilised emulsions caused slower oil clearance from the tongue surface compared to emulsions stabilized with Na-caseinate. Protein content had a negative relation with oil fraction on the tongue for lysozyme stabilized emulsions and no relation for Na-caseinate stabilized emulsions. The presence of thickener decreased deposition of oil on tongue, although viscosity differences (i.e., thickener content) did not affect oil fraction. After-feel perception of creaminess and fatty-film was strongly influenced by the presence of thickener likely due to lubrication in-mouth, i.e., the higher the concentration of thickener in the emulsions the stronger was the perception. With the present developments in CA technology it becomes possible to fine tune the storage conditions to the specific needs of the product. This generates the need to know the exact quality conditions of the product before storage starts. By measuring the initial quality we can determine these conditions optimally. At present the most likely candidates to assess the initial quality with fast and non-destructive measurements are colour, chlorophyll fluorescence, and maybe NIR spectroscopy. Two examples are presented where initial colour measurements on all products in a batch can be shown to be indicative for the keeping quality of that batch. The first example focuses on how initial colour measurements using a 3CCD video camera can be utilised to predict the keeping quality of a batch of cucumbers where colour itself is regarded as the most important quality attribute. The second example focuses on how colour measurements can be used to predict the keeping quality of a batch of strawberries where the ability to suppress a Botrytis cinerea infection is the most important quality attribute. Furthermore, attention is given to the use of modulated chlorophyll fluorescence imaging as a possible initial quality indicator for rose leafy stem cuttings. The level of inhomogeneity in the quantum yield of photochmistry od PSII of leaves of rose cuttings may be an indictor of the capability of the cutting to recover from severance, and to form roots and generate regrowth

Hart- en vaatziekten vormen een belangrijk gezondheidsprobleem en risicofactoren komen veel voor. Voor dit proefschrift zijn de opsporing en behandeling van risicofactoren onderzocht in Nederlandse huisartsenpraktijken. Allereerst is een persoonsgerichte screeningsmethode onderzocht in Oude Pekela, een gebied met een lage sociaaleconomische status, wat is gerelateerd aan het hebben van meer risicofactoren en een lagere deelname aan preventie-activiteiten. Deze aanpak gaf een veel hogere opkomst en opbrengst dan het landelijk gepropageerde “PreventieConsult” en kan daarom bijdragen aan het verbeteren van de huidige preventieprogramma’s. Daarnaast is onderzocht of de ”AGE-reader”, een apparaat dat via UV-licht versuikerde eiwitten in de huid meet, kan bijdragen aan de opsporing van risicopatiënten. AGE-waarden bleken na drie jaar verhoogd bij mensen die reeds hart- en vaatziekten hadden doorgemaakt. De bruikbaarheid hiervan in de praktijk wordt nog verder onderzocht. Verder is gekeken naar de behandeling van risicofactoren door praktijkondersteuners in de huisartsenpraktijk. Deze behandeling bleek duidelijk effect te hebben op het risico na één jaar. Er was geen extra effect waarneembaar bij de behandelgroep waarbij na loting aanvullende thuismeting van bloeddruk, gewicht en/of lichaamsbeweging was ingezet. De kosten van risicobehandeling in de huisartsenpraktijk waren relatief laag en werden grotendeels veroorzaakt door werkverzuim ten gevolge van praktijkbezoek. De kosten van aangepaste medicatie vielen mee, terwijl dit wel gerelateerd was aan een groter eindeffect. Omdat mensen steeds jonger met risicofactoren kampen en er bezuinigd moet worden op zorgkosten, kunnen deze resultaten eraan bijdragen dat de politiek en zorgverzekeraars hierin verstandige beleidsbeslissingen nemen.

In de chirurgie worden verschillende beeldvormingstechnieken gebruikt, zowel voor diagnostiek rondom een operatie, als begeleidende beeldvorming tijdens een operatie. Voorbeelden van veelgebruikte beeldvormingstechnieken in de kliniek zijn standaard röntgendiagnostiek, computed tomography (CT), magnetic resonance imaging (MRI), positron emission tomography (PET), single-photon emission computed tomography (SPECT) en echografie. Hoewel deze beeldvormingstechnieken geavanceerd zijn, zijn ze niet specifiek. Hierdoor is het vaak lastig om bacteriële infectie of kwaadaardige tumoren te onderscheiden van andere weefsels. Dit proefschrift beschrijft de ontwikkeling van specifieke bacteriële infectie- en kankergerichte beeldvormingstechnieken, waardoor deze pathologie specifiek van andere processen onderscheiden kan worden. De nadruk ligt op de ontwikkeling van doelgerichte fluorescentiebeeldvorming. Fluorescentiebeeldvorming is bij uitstek geschikt voor oppervlakte beeldvorming, en daarom erg bruikbaar tijdens operaties om de chirurg “live” van extra informatie te voorzien. Het proefschrift behandelt de verschillende mogelijkheden van doelgerichte beeldvorming bij bacteriële infecties en bij kwaadaardige tumoren en beschrijft de toepassing van verschillende doelgerichte fluorescentieprobes in diermodellen. Tot slot wordt aangetoond dat doelgerichte fluorescentiebeeldvorming van substantiële meerwaarde is voor de detectie van deze ziekteprocessen.

Energy-Coupling Factor (ECF) transporters vormen een subgroep van de ATP-binding cassette (ABC) transporters en zijn betrokken bij de opname van vitamines door prokaryoten. Ze maken hiervoor gebruik van membraaneiwitten (S - componenten) die hun substraten met een hoge affiniteit binden. S-componenten kunnen een complex vormen met de ECF-module zodat de substraten actief getransporteerd kunnen worden. De energie hiervoor wordt geleverd door hydrolyse van ATP. In veel gevallen kan de ECF module een complex vormen met verschillende S -componenten, die verschillende vitamines binden. Op deze manier kunnen chemisch zeer diverse substraten via een vergelijkbare route in de cel worden opgenomen. De eerste twee experimentele hoofdstukken beschrijven de karakterisatie van twee verschillende S-componenten (voor biotine (BioY) en thiamine (ThiT)). In hoofdstuk 2 presenteerden we een hoge-resolutie kristalstructuur van BioY uit L. lactis en biochemische analyse van dit eiwit en zijn homoloog uit Rhodobacter capsulatus. In hoofdstuk 3 hebben we gebruik gemaakt van EPR spectroscopie, stopped-flow fluorescentie spectroscopie en molecular dynamics simulaties om de structurele veranderingen die plaatsvinden in ThiT uit L. lactis na de binding van thiamine te bepalen. In de twee volgende experimentele hoofdstukken werden de volledige ECF transporters onderzocht. Hart- en vaatziekten vormen een belangrijk gezondheidsprobleem en risicofactoren komen veel voor. Voor dit proefschrift zijn de opsporing en behandeling van risicofactoren onderzocht in Nederlandse huisartsenpraktijken. Allereerst is een persoonsgerichte screeningsmethode onderzocht in Oude Pekela, een gebied met een lage sociaaleconomische status, wat is gerelateerd aan het hebben van meer risicofactoren en een lagere deelname aan preventie-activiteiten. Deze aanpak gaf een veel hogere opkomst en opbrengst dan het landelijk gepropageerde “PreventieConsult” en kan daarom bijdragen aan het verbeteren van de huidige preventieprogramma’s. Daarnaast is onderzocht of de ”AGE-reader”, een apparaat dat via UV-licht versuikerde eiwitten in de huid meet, kan bijdragen aan de opsporing van risicopatiënten. AGE-waarden bleken na drie jaar verhoogd bij mensen die reeds hart- en vaatziekten hadden doorgemaakt. De bruikbaarheid hiervan in de praktijk wordt nog verder onderzocht. Verder is gekeken naar de behandeling van risicofactoren door praktijkondersteuners in de huisartsenpraktijk. Deze behandeling bleek duidelijk effect te hebben op het risico na één jaar. Er was geen extra effect waarneembaar bij de behandelgroep waarbij na loting aanvullende thuismeting van bloeddruk, gewicht en/of lichaamsbeweging was ingezet. De kosten van risicobehandeling in de huisartsenpraktijk waren relatief laag en werden grotendeels veroorzaakt door werkverzuim ten gevolge van praktijkbezoek. De kosten van aangepaste medicatie vielen mee, terwijl dit wel gerelateerd was aan een groter eindeffect. Omdat mensen steeds jonger met risicofactoren kampen en er bezuinigd moet worden op zorgkosten, kunnen deze resultaten eraan bijdragen dat de politiek en zorgverzekeraars hierin verstandige beleidsbeslissingen nemen.

Tryptofaan toepassen in fluorescentiespectroscopie Dejan Pétrovic verrichtte onderzoek naar de vraag hoe het aminozuur tryptofaan kan worden toegepast in fluorescentiespectroscopie, een onderzoeksmethode die in de laatste decennia belangrijk is geworden in de levenswetenschappen. Tryptofaan (Trp) heeft eigenschappen die maken dat het erg geschikt is voor het bestuderen van eiwitstructuren en eiwitdynamica. Deze eigenschappen kunnen in de praktijk echter alleen optimaal benut worden als gezuiverd eiwit met slechts 1 of 2 Trp-residuen gebruikt wordt. Eén mogelijkheid om een mengsel van eiwitten m.b.v. fluorescentie te bestuderen is het vervangen van een specifieke Trp in één van de eiwitten door een Trp-analogon. Vanwege de aanwezigheid van een onnatuurlijk aminozuur worden deze eiwitten ook wel alloproteïnes genoemd. Eén van de doelen van het promotieproject was te onderzoeken of Trp-analogon bevattende eiwitten gebruikt kunnen worden als moleculair herkenningselement in een biosensor. Daarnaast zijn een aantal nieuwe procedures ontwikkeld voor de productie van alloproteïnes. Tot nu toe was er geen gevoelige spectroscopische methode voorhanden om de interactie tussen LysM domeinen en de celwand direct te kunnen volgen. In hoofdstuk 2 wordt een fluorescentiemethode gepresenteerd waarmee deze interactie gevolgd kan worden. In hoofdstuk 3 wordt een Lactococcus lactis Trp auxotroof expressie systeem beschreven dat de biosynthetische inbouw van Trp-analoga met grotere substituenten mogelijk maakt. In hoofdstuk 4 worden experimentele data gepresenteerd over de emissietoestand van vrij 5HI en 5HW, en van 5HW ingebouwd in een eiwit. Hieruit blijkt dat in de meeste gevallen 1Lb de emissietoestand is. Echter, één eiwit vertoont zowel 1La en 1Lb emissie.

In de chirurgie worden verschillende beeldvormingstechnieken gebruikt, zowel voor diagnostiek rondom een operatie, als begeleidende beeldvorming tijdens een operatie. Voorbeelden van veelgebruikte beeldvormingstechnieken in de kliniek zijn standaard röntgendiagnostiek, computed tomography (CT), magnetic resonance imaging (MRI), positron emission tomography (PET), single-photon emission computed tomography (SPECT) en echografie. Hoewel deze beeldvormingstechnieken geavanceerd zijn, zijn ze niet specifiek. Hierdoor is het vaak lastig om bacteriële infectie of kwaadaardige tumoren te onderscheiden van andere weefsels. Dit proefschrift beschrijft de ontwikkeling van specifieke bacteriële infectie- en kankergerichte beeldvormingstechnieken, waardoor deze pathologie specifiek van andere processen onderscheiden kan worden. De nadruk ligt op de ontwikkeling van doelgerichte fluorescentiebeeldvorming. Fluorescentiebeeldvorming is bij uitstek geschikt voor oppervlakte beeldvorming, en daarom erg bruikbaar tijdens operaties om de chirurg “live” van extra informatie te voorzien. Het proefschrift behandelt de verschillende mogelijkheden van doelgerichte beeldvorming bij bacteriële infecties en bij kwaadaardige tumoren en beschrijft de toepassing van verschillende doelgerichte fluorescentieprobes in diermodellen. Tot slot wordt aangetoond dat doelgerichte fluorescentiebeeldvorming van substantiële meerwaarde is voor de detectie van deze ziekteprocessen.

The photosynthetic process can hardly be bettered. But the utilisation of natural or artificial light certainly leaves room for improvement. In recent years our understanding of light has grown considerably and this has major implications on how we deal with light in horticulture.

Voor het meten van kwikgehalten in urine die in het kader van de studie naar de blootstelling van de bevolking aan milieucontaminanten worden aangeboden werd de methode geoptimaliseerd en gevalideerd. De optimalisering van een methode voor kwik in urine was gebaseerd op de operationele methode 'bepaling van kwik in regenwater'. In ongewijzigde vorm bleek broomconcentratie te laag om alle oxideerbare stoffen inclusief kwik (verbindingen) om te zetten. De methode werd geoptimaliseerd door het monster zodanig te verdunnen dat de beoogde aantoonbaarheidsgrens van 0.1 mug Hg/l nog mogelijk is. Bij een 50-voudige verdunning en een broomconcentratie van 2,5 mmol/l werd experimenteel een aantoonbaarheidsgrens van 0.08 mug Hg/l vastgesteld. De herhaalbaarheid en reproduceerbaarheid op een meetniveau van 3,44 mug Hg/l is respectievelijk 0,07 en 0.27 mug Hg/l. Addities, van anorganisch gebonden kwik en organisch gebonden kwik, worden kwantitatief teruggevonden.

Single molecule electronics: Een systematische aanpak om de eigenschappen van enkele porfyrine moleculen te bestuderen Porfyrinen zijn zeer interessante en belangrijke complexe organische moleculen, die bestudeerd worden in prototypische moleculaire elektronische apparaten. We waren geïnteresseerd om het geleidingsgedrag van porfyrine moleculen op het single molecule-niveau te bestuderen. We hebben de vereiste trans-amine en thiol gefunctionaliseerde porfyrinen gesynthetiseerd en vervolgens hun binding met goud nanodeeltjes als model oppervlak voor nano-elektroden bestudeerd om te leren over hun interactiesterkte en -dynamiek. Van een reeks gefunctionaliseerde porfyrinen die werd ontwikkeld voor deze studie, toonde de verbinding met vier amino functionaliteiten de hoogste bindingsinteractie met gouden nanodeeltjes. We namen fotogeïnduceerde ladingsoverdracht waar van porphyrins naar quantum dots in hybride systemen, tijdens het onderzoek naar de bindingsinteractie van porphyrinen met PbSe quantum dots. Dit resultaat geeft aan dat deze porfyrinen goede kandidaten zijn voor single molecule elektronische studies. Tot slot hebben we ons gericht op het single molecule elektronische gedrag van porphyrinen met platina electromigration electrodes en mechanisch regelbare breekjunctie technieken. Porphyrinen met amino (-NH2) en thiol (-SH) verankering groepen waren respectievelijk gebonden aan de platina-elektroden (verkregen met electromigration) en goud-elektroden (MCBJ) We onderscheiden moleculair transport van achtergrond-tunneling conductances in platina electromigration techniek. Een verandering in geleiding werd waargenomen, wanneer porfyrine moleculen met en zonder thiol opzeggingen, vrije base, Zn-en Ru-porphyrins aanwezig in MCBJs waren. Onze porfyrinederivaten liet zowel vlak en loodrecht gesondeerd configuraties. Deze studies vergroten ons begrip van metalen - molecuul interacties en elektronen transport in een enkel molecuul, wat cruciaal is voor de verdere ontwikkeling op nanoschaal elektronica.

Dit is deels onderdeel van het normale verouderingsproces, maar bij meerdere ziektebeelden is de opstapeling van AGE versneld en verhoogd. Dit was al aangetoond voor diabetes mellitus, nierfalen en meerdere systeemziekten. De opstapeling van AGE blijkt een belangrijk rol te spelen in het ziekteproces met nadelige gevolgen op lange termijn (complicaties) en vroegtijdige sterfte. De mate van AGE stapeling in ons lichaam kan geschat worden door een geavanceerde lichttechniek (autofluorescentie van de huid, afgekort SAF). Mijn proefschrift heeft 2 belangrijke conclusies. Allereerst zijn er meerdere factoren die de SAF meting beïnvloeden. Deze storende factoren moeten vermeden worden om een betrouwbare meting te waarborgen. Ten tweede spelen AGE een grote rol in het ziekteproces van aderverkalking (atherosclerose). SAF, als maat voor AGE stapeling, is consequent verhoogd bij meerdere uitingen van atherosclerose. Een verhoogde SAF werd gevonden bij patiënten met vernauwde halsslagaders (carotisstenose), bij patiënten met vernauwde beenvaten (perifeer arterieel vaatlijden) en bij patiënten met diabetes mellitus type 2 met vaatproblemen (macrovasculair lijden). Uit eerder onderzoek bij patiënten met type 2 diabetes bleek de SAF meting een goede risico-inschatting te geven op toekomstige hart-en vaatziekten en sterfte, bovenop en onafhankelijk van de klassieke risicofactoren (bloeddruk, cholesterol, roken, overgewicht etc). Door de makkelijke en snelle uitvoerbaarheid zou de SAF meting in de toekomst een brede toepasbaarheid kunnen hebben bij het inschatting van het cardiovasculaire risico van patiënten.

Borstkanker is de meest vóórkomende vorm van kanker bij vrouwen. Uiteindelijk zal circa 1 op de acht vrouwen borstkanker ontwikkelen gedurende haar leven. In het geval van relatief kleine tumoren kunnen patiënten doorgaans goed worden behandeld door middel van borstsparende chirurgie en bestraling van de borst. Tijdens de borstsparende procedure verwijdert de chirurg de tumor inclusief een dun randje van omliggend gezond borstweefsel. Desondanks blijken er in een aanzienlijk deel van de patiënten tumorcellen te reiken tot in het snijvlak. Deze zogeheten positieve snijvlakken vormen een belangrijk risico op het recidiveren van de tumor. Het verwijderen van een grotere hoeveelheid borstweefsel verlaagt weliswaar de kans op positieve snijvlakken, maar verslechtert tegelijkertijd het cosmetisch resultaat van de in opzet borstsparende behandeling. Nieuwe technieken zijn daarom gewenst om beter onderscheid te kunnen maken tussen tumorweefsel en gezond weefsel tijdens de operatie. In het onderhavige proefschrift wordt een methode beschreven, genaamd nabij-infrarood fluorescentie beeldvorming. De tumorcellen worden hierbij gemerkt met een fluorescente kleurstof, waardoor de tumor oplicht tijdens de operatie. Omdat het uitgezonden licht niet kan worden waargenomen met het menselijk oog, wordt er gebruik gemaakt van een speciale camera waarmee de tumor tijdens de operatie zichtbaar gemaakt kan worden. De verwachting is dat deze techniek in de toekomst een bijdrage zal gaan leveren aan de chirurgische behandeling van patiënten met borstkanker. Daarnaast wordt momenteel onderzocht of dezelfde techniek kan worden toegepast om uitzaaiingen in de lymfeklieren op te sporen tijdens de operatie.

Het klimaat in een kas wordt ingesteld om een optimale gewasfotosynthese, assimilatenverdeling en plantvorm te realiseren. Om momentaan het kasklimaat aan te kunnen passen aan de behoeftes van de plant is het van groot belang inzicht te hebben in de directe gevolgen van aanpassingen in het klimaat op de plant prestaties, in het bijzonder op de fotosynthese. Dit is te doen met de volgende methodes: 1. Gasuitwisseling van bladeren: nauwkeurige metingen van de fotosynthese van een stukje blad, met draagbare meetapparatuur. 2. Plantivity: een commercieel verkrijgbare meter die de fluorescentie van een stukje blad meet. 3. Kas-in-kas: een niet-geklimatiseerde meetkamer waarin CO 2 opname van een aantal planten gemeten kan worden. 4. Fluorescentie-imaging: fluorescentie metingen op afstand aan een groter oppervlakte gewas. 5. Fotosynthese-monitor: soft-sensor waarmee de CO 2 opname van een kas berekend wordt op basis van ventilatievoud en metingen van de CO 2 concentratie binnen en buiten de kas. Uit twee workshops met telers bleek dat zij fotosynthese als een belangrijk proces beschouwen in de teelt van hun gewas, en dat zij de fotosynthese van hun gewas graag momentaan online zouden willen meten. Het is daarom wenselijk door te gaan met de ontwikkeling van een robuust en betrouwbaar meetsysteem voor de gewasfotosynthese

Fonkelende kleuren van vogels, kevers en vlinders onderzocht Miljoenen jaren van evolutionaire geschiedenis hebben tot de verbazingwekkende diversiteit van biologische organismen geleid, die tegenwoordig gebruik maken van verschillende kleurmechanismen om heldere, fonkelende kleuren te reflecteren. De meest briljante en schitterende kleuren, zoals het witste wit en het zwartste zwart van vele dier- soorten, zoals vogels, kevers en vlinders, ontstaan uit de fysieke interactie van licht met geordende, quasi-geordende en ongeordende materie, wat resulteert in structurele kleuren. Deze biofotonische nanostructuren kunnen variëren in complexiteit, gaande van 'eenvoudige' multilaag-structuren tot topologisch complexe, driedimensionale fotonische kristallen. Deze fotonische structuren veranderen de spectrale samenstelling van het invallende licht en creëren zo de fonkelende kleuren, die we iedere dag kunnen zien. Bodo Wilts onderzocht een waaier van fotonische structuren in vogels, kevers en vlinders en bekeek in detail hoe deze complexe structuren met het invallend licht wisselwerken. Hij bestudeerde hoe deze structuurkleuren afgestemd kunnen worden door de aanwezigheid van absorberende pigmentaire filters. Door een brede reeks van verschillende fysische technieken en rekenfysica te gebruiken, kreeg hij inzicht in de optische eigenschappen van deze complex gevormde composieten van structurele en pigmentaire kleuren. De biologische functies van deze structuren worden in het proefschrift besproken en kunnen variëren van interseksuele signalering tot camouflage.

Ten tweede spelen AGE een grote rol in het ziekteproces van aderverkalking (atherosclerose). SAF, als maat voor AGE stapeling, is consequent verhoogd bij meerdere uitingen van atherosclerose. Een verhoogde SAF werd gevonden bij patiënten met vernauwde halsslagaders (carotisstenose), bij patiënten met vernauwde beenvaten (perifeer arterieel vaatlijden) en bij patiënten met diabetes mellitus type 2 met vaatproblemen (macrovasculair lijden). Uit eerder onderzoek bij patiënten met type 2 diabetes bleek de SAF meting een goede risico-inschatting te geven op toekomstige hart-en vaatziekten en sterfte, bovenop en onafhankelijk van de klassieke risicofactoren (bloeddruk, cholesterol, roken, overgewicht etc). Door de makkelijke en snelle uitvoerbaarheid zou de SAF meting in de toekomst een brede toepasbaarheid kunnen hebben bij het inschatting van het cardiovasculaire risico van patiënten.

BMI-waarde belangrijke voorspeller voor sterfte onder dialysepatiënten Nierdialysepatiënten kunnen het beste een Body Mass Index (BMI) hebben van 24 kg/m2. Zij maken dan de minste kans om vroegtijdig aan nierfalen te sterven. Dat concludeert Stefan Arsov in zijn promotieonderzoek. Arsov onderzocht door welke factoren de hoeveelheid glycoseringseindproducten in de huid en het bloed van dialysepatiënten beïnvloed worden en hoe een stijging bij nierpatiënten vermeden kan worden. End Stage Renal Disease (ERSD) is een chronische en onomkeerbare nierziekte. De nieren zijn dan niet meer in staat om afval of overtollig water af te voeren. Patiënten met ERSD zijn voor overleving afhankelijk van nierdialyse of een niertransplantatie. Arsov ging na hoe snel de afvalstoffen in de huid van ERSD-patiënten stijgen. Die stijging blijkt jaarlijks maar liefst zeven tot negen maal hoger dan bij gezonde proefpersonen. Hemodialyse, waarbij het bloed buiten het lichaam gefilterd wordt door een kunstnier, verminderde wel de afvalstoffen in het bloed, maar leidde niet tot onmiddellijke, meetbare verschillen in de huid. De stijging van afvalstoffen in de huid vormt volgens Arsov een belangrijke voorspeller voor sterfte. Deze stijging blijkt samen te hangen met de BMI-waarde. De promovendus concludeert dat een BMI-waarde van 24 zorgt voor de kleinste jaarlijkse stijging. Hij concludeert dat het optimaliseren van de voeding wellicht kan leiden tot minder sterfte onder nierpatiënten.

Het klimaat in een kas wordt ingesteld om een optimale gewasfotosynthese, assimilatenverdeling en plantvorm te realiseren. Om momentaan het kasklimaat aan te kunnen passen aan de behoeftes van de plant is het van groot belang inzicht te hebben in de directe gevolgen van aanpassingen in het klimaat op de plant prestaties, in het bijzonder op de fotosynthese. Dit is te doen met de volgende methodes: 1. Gasuitwisseling van bladeren: nauwkeurige metingen van de fotosynthese van een stukje blad, met draagbare meetapparatuur. 2. Plantivity: een commercieel verkrijgbare meter die de fluorescentie van een stukje blad meet. 3. Kas-in-kas: een niet-geklimatiseerde meetkamer waarin CO 2 opname van een aantal planten gemeten kan worden. 4. Fluorescentie-imaging: fluorescentie metingen op afstand aan een groter oppervlakte gewas. 5. Fotosynthese-monitor: soft-sensor waarmee de CO 2 opname van een kas berekend wordt op basis van ventilatievoud en metingen van de CO 2 concentratie binnen en buiten de kas. Uit twee workshops met telers bleek dat zij fotosynthese als een belangrijk proces beschouwen in de teelt van hun gewas, en dat zij de fotosynthese van hun gewas graag momentaan online zouden willen meten. Het is daarom wenselijk door te gaan met de ontwikkeling van een robuust en betrouwbaar meetsysteem voor de gewasfotosynthese
