Das Nebennierenrindenkarzinom geht mit einer schlechten Prognose und hoher Letalität einher. Eine Suppression der lncRNA H19 führt zu einer Herunterregulation von Autophagiemarkern, einer erhöhten Proliferationsrate sowie gesteigertem Metastasierungspotential bei von Nebennierenrindenkarzinom betroffenen Patienten. Die Anwendung der Deacetylaseinhibitoren (DACIs) Panobinostat, Trichostatin A (TSA) und SAHA beeinträchtigt die Zellviabilität von H295R-Monolayern und Sphäroiden und führt zu einer Überexpression von H19 und Autophagietranskripten. Durch einen H19-Knockdown in H295R-Zellen war es nicht möglich, dass Expressionsniveau von Autophagietranskripten zu modulieren. Stattdessen inhibiert ein Knockdown von H19 die Fähigkeit von DACIs, das Proteinniveau von Autophagiemarkern zu modulieren. Außerdem kann durch eine höhere Konzentration von DACIs die Proteinquantität von Beclin1 und SQSTM1/p62 reduziert werden sowie die Konversion von LC3BI in LC3BII induziert werden, wodurch sich ein aktiver Autophagieprozess bestätigt. Weder die Proteinquantität noch die Aktivität von Caspase 8 und 3/7 werden durch DACis verändert, sodass die Involvierung von Apoptose am NCI-H295R-Zelluntergang ausgeschlossen werden kann. DACIs stellen die H19-Expression und die Expression von Autophagiemarkergenen wieder her und lösen Zelltod in Nebennierenrindenkarzinomzellen aus. Die Wiederaktivierung von Autophagie könnte eine neue Strategie für die Behandlung von Patienten bedeuten, die von dieser schwerbehandelbaren malignen Erkrankung betroffen sind.

La sénescence cellulaire et l'apoptose ont évolué comme des puissantes barrières protectrices contre la transformation néoplasique. La sénescence est un état d'arrêt permanent de la prolifération dans lequel les cellules restent métaboliquement actives. La sénescence cellulaire est déclenchée par différentes sources de stress, notamment les oncogènes activés, le dysfonctionnement des télomères, les dommages à l'ADN et des défauts dans la réplication provoqués par des agents génotoxiques, des espèces réactives de l'oxygène, etc. Ce processus complexe engage deux voies différentes de suppresseurs de tumeurs, les voies p53/p21 et p16INK4a/pRb, et les deux voies doivent être compromises dans les cellules humaines afin de contourner la sénescence. Par conséquent, décrire la relation entre l'activation des oncogènes, l'arrêt de la prolifération induite par la sénescence et l'échappement à l'état de sénescence est essentiel pour comprendre le processus de tumorigenèse. KDM4A est un membre de la sous-famille KDM4 des Jumonji lysine déméthylases ciblant les variantes di- et triméthylées de l'histone H3 lysine 9 (H3K9) et l'histone H3 lysine 36 (H3K36). Les trois premiers membres de la sous-famille KDM4A, KDM4B et KDM4C sont également capables de lier l'histone 4 lysine 20 di-méthyl/tri-méthyl (H4K20me2/3) et l'histone 3 lysine 4 tri-méthyl (H3K4me3), via leurs domaines Tudor consécutifs. KDM4A module négativement l'activité de la voie p53, en ciblant directement le suppresseur de tumeur CHD5, et est également un régulateur négatif de la réponse aux dommages de l'ADN. Les niveaux d'expression de KDM4A sont souvent élevés dans les cellules cancéreuses et diminués pendant la sénescence cellulaire. La motivation principale de cette thèse est d'élargir nos connaissances actuelles sur la façon dont la réorganisation de la chromatine influence la stabilité du phénotype de sénescence. Dans la première partie de ce travail, nous abordons la fonction de méthylation de H4K20 et H3K9, dans le contexte des foyers d'hétérochromatine associés à la sénescence (SAHF: senescence-associated heterochromatin foci). Nous démontrons que l'intégration de H4K20me3 dans les SAHF dépend de l'incorporation précédente de H3K9me3 et révélons les méthyltransférases H4K20 impliquées dans ce processus. Nous proposons un mécanisme moléculaire par lequel H4K20me3 et H3K9me3 coopèrent avec p53 dans la répression stable des gènes cibles de E2F au cours de la sénescence induite par l'oncogène Ras. Dans la deuxième partie de la thèse, nous présentons une voie de dégradation lysosomale (c'est-à-dire l'autophagie médiée par des chaperons) en tant que nouveau mécanisme potentiel par lequel les cellules modulent les niveaux de KDM4A pendant la sénescence. Nos résultats suggèrent que la méthylation dans les lysines des histones régule la stabilité de sénescence en réponse à l'oncogène Ras et révèlent le potentiel d'induction de la sénescence par inhibition ciblée de KDM4A dans le traitement du cancer., Cellular senescence and apoptosis have evolved as potent protective barriers against neoplastic transformation. Senescence is a state of stable arrest of proliferation in which cells remain metabolically active. Cellular senescence is triggered by different sources of stress, including activated oncogenes, telomere dysfunction, DNA damage and replication defects elicited by genotoxic agents, reactive oxygen species, etc. This complex process engages two different tumor suppressor pathways, the p53/p21 and p16INK4a/pRb pathways that need to be compromised in human cells in order to circumvent the senescence-associated growth halt. Hence, describing the relationship between oncogene activation, senescence-induced proliferation arrest and escape from the senescence state remains essential to understand tumorigenesis. KDM4A is a member of the KDM4 sub-family of Jumonji lysine demethylases targeting di- and tri-methylated histone H3 lysine 9 (H3K9) and histone H3 lysine 36 (H3K36). The first three sub-family members KDM4A, KDM4B and KDM4C are also able to bind histone 4 lysine 20 di-methyl/tri-methyl (H4K20me2/3) and histone 3 lysine 4 tri-methyl (H3K4me3), via their tandem Tudor domain. KDM4A negatively modulates the activity of the p53 pathway, by directly targeting the tumor suppressor CHD5, and is also a negative regulator of the DNA damage response. KDM4A expression levels are often elevated in cancer cells and decreased during cellular senescence. The principal motivation for this thesis is to expand our current knowledge on how chromatin reorganization influences the stability of the senescence phenotype. In the first part of this work we address the function of H4K20 and H3K9 methylation, in the context of the senescence-associated heterochromatin foci (SAHF). We demonstrate that integration of H4K20me3 into the SAHF depends on the previous incorporation of H3K9me3 and reveal the H4K20 methyltransferases involved in this process. We propose a molecular mechanism by which H4K20me3 and H3K9me3 cooperate with p53 in the stable repression of E2F target genes during oncogenic Ras-induced senescence. In the second part of the thesis we present a lysosomal-degradation pathway (i.e. chaperone-mediated autophagy) as a novel potential mechanism by which cells modulate KDM4A protein levels during senescence. Our results strongly suggest that histone lysine methylation contributes to the stability of the senescence response to the Ras oncogene and reveal the potential of senescence induction by targeted inhibition of KDM4A in the treatment of cancer.

Der abskopale Effekt beschreibt ein Phänomen, bei dem bei Krebspatienten nach einer Strahlentherapie Tumorzellen abgetötet werden, die sich nicht in der bestrahlten Region befinden. Es wird angenommen, dass dies aufgrund einer immunologischen Reaktion geschieht, die durch die Strahlentherapie ausgelöst wird. Eine mögliche Erklärung könnte der cGAS/STING-Signalweg sein. Dieser Signalweg ist Teil des angeborenen Immunsystems und wird durch dsDNA im Zytosol induziert. Da zytosolische dsDNA über Autophagie abgebaut wird, kann man davon ausgehen, dass eine Autophagie Inhibierung zu einer erhöhten cGAS/STING-Aktivität führt. Um diese Theorie zu untersuchen, wurden dreifach-negative Brustkrebszellen mit vier verschiedenen Inhibitoren behandelt. Bafilomycin A1 und SAR405, zwei Autophagie-Inhibitoren, sowie PX-478 und Echinomycin, Inhibitoren von HIF-1α, wurden verwendet. Die Hemmung von HIF-1α wurde untersucht, da Hypoxie zu einer erhöhten Autophagie-Aktivität in Zellen führen kann. Die Inhibitoren wurden in Kombination mit Torin 1, einem Autophagie-Induktor, verwendet. Es wurden Western Blots durchgeführt, um zu analysieren, ob die Autophagie gehemmt wurde. Zu diesem Zweck wurden LC3 und p62 als Autophagie-Marker verwendet. Die durch die ausgewählten Inhibitoren induzierten Auswirkungen auf die cGAS/STING-Signalisierung wurden anhand der an der Signalkaskade beteiligten Proteine und ihres Phosphorylierungsstatus untersucht. Darüber hinaus wurden auch STAT1 und seine Phosphorylierung untersucht, da dies ein nachgeschalteter Effektor des cGAS/STING-Signalweges ist. Die direkte Hemmung der Autophagie zeigte weitgehend die erwartete Wirkung. Die genauen Ergebnisse unterschieden sich jedoch stark zwischen den verschiedenen Zelllinien. Auch die beiden Inhibitoren an sich ergaben unterschiedliche Resultate, wobei Bafilomycin A1 häufiger den cGAS/STING-Signalweg induzierte. Die Behandlung mit SAR405 führte in den meisten Fällen zu schwächeren Ergebnissen, in zwei Fällen kam es jedoch zu einem stärkeren Anstieg der phosphorylierten cGAS/STING-Proteine. Im Vergleich zu den direkten Autophagie-Inhibitoren zeigte die Behandlung mit HIF-1α-Inhibitoren weniger konsistente Wirkungen. Da die Experimente jedoch nur in 2D-Zellkulturen durchgeführt wurden, wurde nie eine hypoxische Umgebung geschaffen, was die Interpretation der gesammelten Daten erschwerte. Experimente in 3D- oder organoiden Zellkulturen könnten in diesem Zusammenhang zu weiteren Erkenntnissen führen. The abscopal effect describes a phenomenon in which tumour cells located away from the irradiated region are killed in cancer patients after radiotherapy. It is assumed that this happens due to an immunological reaction triggered by radiotherapy. One possible explanation could be the cGAS/STING signalling pathway. This signalling pathway is part of the innate immune system and is induced by dsDNA in the cytosol. Since cytosolic dsDNA is cleared via autophagy, it can be assumed that autophagy inhibition leads to increased cGAS/STING activity. To investigate this theory, triple-negative breast cancer cells were treated with four different inhibitors. Bafilomycin A1 and SAR405, two autophagy inhibitors, as well as PX-478 and Echinomycin, inhibitors of HIF-1α, were used. HIF-1α inhibition was investigated since hypoxia can lead to increased autophagy activity in cells. The inhibitors were used in combination with Torin 1, an autophagy inducer. Western Blots were performed to analyse whether autophagy was inhibited. For this purpose, LC3 and p62 were used as autophagy markers. The effects induced by the chosen inhibitors on cGAS/STING signalling were investigated using proteins involved in the signalling cascade and their phosphorylation status. In addition, STAT1 and its phosphorylation were also investigated, as this is a downstream effector of the cGAS/STING pathway. The direct inhibition of autophagy largely showed the expected effect. However, the exact outcomes differed greatly between the different cell lines. The two inhibitors per se also gave different results, with Bafilomycin A1 showing more frequent induction of the cGAS/STING pathway. Treatment with SAR405 gave weaker results in most cases, however, in two cases a stronger increase in phosphorylated cGAS/STING proteins occurred. Compared to the direct autophagy inhibitors, the treatment with HIF-1α inhibitors showed less consistent effects. However, as the experiments were only performed in 2D cell cultures, a hypoxic environment was never established, making interpretation of the gathered data difficult. Experiments in 3D or organoid cell cultures could provide better results in this context.

Autophagy is an auto-digestive process targeting cytoplasmic cargo for lysosomal degradation. It is crucial for maintaining cellular homeostasis, and dysregulation of autophagy is associated with a number of malignancies, including cancer and neurodegenerative diseases. Notably, autophagy plays an emerging role in anti-viral innate immune defenses. However, successful viral pathogens have evolved mechanisms to avoid restriction by autophagy. For example, the pandemic coronavirus SARS-CoV-2 suppresses autophagic turnover in infected cells, but the viral determinants responsible were unknown. To investigate which proteins of SARS-CoV-2 counteract autophagy, I established a method to rapidly and accurately quantify changes in autophagic flux using flow cytometry. I analyzed the impact of 29 proteins of SARS-CoV-2 on autophagy and identified Non-structural protein 2 (Nsp2), Nsp5, Nsp15, envelope (E), membrane (M), ORF3a and ORF7a as candidate autophagy antagonists. The five most potent modulators (Nsp15, E, M, ORF3a and ORF7a) were subsequently explored in more detail. We found that Nsp15 inhibits the induction of autophagy, likely via mTORC1, whereas E, ORF3a, and ORF7a inhibit turnover of autophagosomes. Mechanistically, ORF3a inhibits the fusion of autophagosomes and lysosomes, and ORF7a prevents the acidification of (auto-)lysosomes. Finally, M redirects autophagic membranes to the perinuclear space but does not impact autophagic flux. The reporter system to analyze the impact of proteins on autophagy was also applied to discover novel cellular proteins that regulate autophagy. Here, I identified three members of the tripartite motif (TRIM) protein family, TRIM2, 3, and 71, as novel inhibitors of autophagic flux. My data shows that these TRIMs use their filamin-NHL domain to reduce autophagy induction. CK2 was found to be a major interaction partner of TRIM2/3 and 71 which also regulates autophagy. Overexpression of CK2 reduces autophagic flux, conversely inhibition of CK2 activates autophagy. In silico, we identified a putative CK2 phosphorylation site in the NHL repeats of the TRIMs. Strikingly, mutation of this site reduced the functionality of TRIM3, suggesting that CK2 activates filamin-NHL TRIMs by phosphorylation in their NHL domain to inhibit autophagy. In summary, I established a method for rapid quantification of changes in autophagy, allowing me to define novel viral and host autophagy regulating proteins. Characterization of autophagy antagonists of SARS-CoV-2 gave insight on how this pathogen avoids restriction by autophagy. In addition, my data proposes the cellular CK2-TRIM2/3/71 axis as a novel autophagy inhibiting pathway to maintain autophagy homeostasis.

Die bedingt essentielle Aminosäure Taurin ist Bestandteil nahezu aller Gewebe des menschlichen Körpers und ist eine von wenigen Aminosäuren, die der Proteinsynthese nicht beiträgt. Als wichtiger organischer Osmolyt spielt Taurin eine bedeutende Rolle bei der Regulierung des Zellvolumens sowie der Immunmodulation. Außerdem ist Taurin eine der am häufigsten in Leukozyten vorkommenden Aminosäuren. Diese Datenlage, sowie der Mangel an Literatur zu Taurin in T-Zellen, bilden den Ausgangspunkt der vorliegenden Studie, in welcher der Fragestellung nachgegangen wird, inwiefern das Fehlen von Taurin einen Einfluss auf metabolische Vorgänge in CD8+ T- Zellen hat. Ein besonderes Augenmerk wurde dabei auf Glykolyse, mitochondriale Aktivität und Autophagie gelegt. Zur Beantwortung der Fragestellung wurden ausschließlich humane Blutprodukte herangezogen, aus denen mittels magnetischer Zellseparation naive CD8+ T-Zellen isoliert wurden. Anschließend erfolgte die Deaktivierung der intrazellulären Taurinsynthese in den naiven CD8+ T-Zellen. Zur Darstellung möglicher veränderter metabolischer Vorgänge der manipulierten Zellen wurden von jedem/jeder Spender*in eine Kontrollgruppe bestehend aus unbehandelten naiven CD8+ T-Zellen, die demnach über eine intakte Taurinsynthese verfügten, mitgeführt. 72 Stunden nach der in vitro Aktivierung der Zellen folgten Färbungen mit ausgewählten Fluoreszenzfarbstoffen sowie deren durchflusszytometrische Analysen. Die Analyse der erhobenen Daten zeigt, dass CD8+ T-Zellen mit deaktivierter Taurinsynthese mehr mitochondriale reaktive Sauerstoffspezies (ROS) produzieren, als jene Zellen, die unbehandelt blieben. Dies deutet auf eine erhöhte mitochondriale Aktivität hin. The semi-essential amino acid taurine can be found in most cells and tissues throughout the human body. However, despite its frequent occurrence, taurine is not integrated into protein sequences. Since taurine functions as osmolyte, it is considered to be important for cell volume regulation and immune functions. Moreover, taurine is one of the most abundant amino acids in leukocytes. These facts, as well as little existing literature on the effects of taurine in T cells are the main reasons for the following study. The aim of this bachelor thesis is to assess the influence of deactivated taurine synthesis on cellular metabolism of CD8+ T cells. For this purpose, glycolysis, mitochondrial activation and autophagy were measured with flow cytometry. First, naïve CD8+ T cells were isolated from human blood samples, followed by deactivation of intracellular taurine synthesis. Each of the samples were then split into a study group, consisting of manipulated cells, and a control group, consisting of cells without deactivation of taurine synthesis, in order to visualize a possible difference. To measure the metabolic parameters of interest, the naïve cells were activated and then incubated with fluorescent dyes 72 hours later. The cells were then analyzed with flow cytometry. The results of this study show that a deactivation of taurine synthesis in CD8+ T cells leads to increased production of mitochondrial reactive oxygen species (ROS) which is indicative of enhanced mitochondrial activation.

Le cancer de la prostate (CP) est le cancer le plus diagnostiqué chez les hommes en Amérique du Nord. Même si les méthodes de détection et les traitements s’améliorent au fil des années, le phénomène de résistance reste encore un problème majeur pour les thérapies utilisées de nos jours. En effet, une majorité des patients vont développer une résistance à l’hormonothérapie et seront alors traités avec une variété d’agents de type hormonal et avec de la chimiothérapie. Malheureusement, ces derniers, individuellement, rallongent la durée de vie des patients que de quelques mois seulement. De ce fait, de nouvelles thérapies, comme les inhibiteurs de PARP (PARPi ; olaparib, niraparib, talazoparib et rucaparib), sont à l’heure actuelle en essais cliniques. Ces composés ciblent la réparation de l’ADN dans le but d’induire une mort cellulaire lorsque des mutations au niveau de cette voie sont présentes. Il est bien connu dans le cancer de l’ovaire et du sein que plusieurs formes de résistances peuvent se mettre en place au cours du temps. Cette résistance aux PARPi dans le CP est encore trop peu étudiée et comprise. Ce projet de thèse s’est ainsi intéressé à la résistance innée mais aussi acquise à l’olaparib dans le CP. Nous avons démontré dans notre première étude que les cellules du CP sont plus résistantes à l’olaparib quand leur niveau basal d’autophagie est plus élevé. Cette résistance médiée par l’autophagie est due, en partie, par la régulation de la localisation nucléaire de la protéine sequestrosome 1 (SQSTM1/p62), capable de dégrader la filamine A (FLNA), essentielle dans le recrutement de Rad51 lors de la recombinaison homologue (HR). Notre étude suggère donc une prise en compte de l’autophagie avant le traitement à l’olaparib pour y déterminer l’efficacité de ce PARPi. Dans notre deuxième étude, nous avons développé des lignées du CP résistantes à l’olaparib pour y déterminer les voies moléculaires les plus affectées par cette transformation et pouvant réguler la résistance acquise. Nous avons mis en évidence que certaines voies, comme les mécanismes de réparation de l’ADN et l’autophagie, pouvaient jouer un rôle essentiel dans la mise en place de résistance acquise à l’olaparib. Cibler certains membres de ces voies comme BRCC3, ATG2B ou ROCK2, pourrait permettre d’inverser la résistance et ainsi permettre d’augmenter l’efficacité de l’olaparib sur le long terme. Ces travaux permettent ainsi de mieux caractériser le phénomène de résistance aux PARPi dans le CP en donnant de nouvelles cibles pour anticiper son apparition et l’inverser. Ils ont aussi permis d’illustrer l’importance du mécanisme d’autophagie dans la réponse aux PARPi dans le CP., Prostate cancer (CP) is the most diagnosed cancer in North America. Even if detection and treatments have improved over the years, the phenomenon of resistance remains a major problem for the therapies used today. Indeed, most patients will develop resistance to hormone therapy and will subsequently be treated with a variety of hormonally based agents and chemotherapeutic agents. Unfortunately, these agents extend survival by only a few months each. As a result, new therapies, such as PARP inhibitors (PARPi; olaparib, niraparib, talazoparib and rucaparib), are currently in clinical trials. These compounds target DNA repair with the aim of inducing cell death when mutations in this pathway are present. It is well known in ovarian and breast cancer that several forms of resistance will develop over time. Unfortunately, this resistance to PARPi in CP is not well studied and therefore understood. This thesis project focused on innate but also acquired resistance to olaparib in CP. We demonstrated in our first study that CP cells are more resistant to olaparib when their basal level of autophagy is higher. This resistance mediated by autophagy is due, in part, by the regulation of the nuclear localization of the protein sequestrosome 1 (SQSTM1/p62), which degrades filamin A (FLNA), essential in the recruitment of Rad51 during the homologous recombination (HR). Our study therefore suggests the importance of considering autophagy before olaparib treatment to predict the effectiveness of a PARPi. In our second study, we developed CP lines resistant to olaparib to determine the molecular pathways most affected by this transformation and able to contribute to resistance. We have shown that different pathways, such as DNA repair mechanisms and autophagy, could play an essential role in the establishment of acquired resistance to olaparib. Targeting members of these pathways, such as BRCC3, ATG2B or ROCK2, may reverse resistance and thus increase the efficacy of olaparib. This work thus makes it possible to better characterize the phenomenon of resistance to PARPi in CP by identifying new targets to counter its appearance. Together these results support the importance of autophagy and DNA repair in the response to PARPi in PC.

Es wird vermutet, dass das Zusammenspiel von NF-κB-Signalen und Autophagie die Entzündung in verschiedenen zellulären Kontexten und als Reaktion auf unterschiedliche Stimuli reguliert. Der molekulare Mechanismus, durch den diese beiden Signalwege bei der Regulierung der Entzündungsreaktion zusammenwirken, ist jedoch noch nicht bekannt. Mithilfe biochemischer Analysen und bildgebender Verfahren haben wir zum ersten Mal die Interaktion zwischen dem autophagischen Marker LC3 und der NF-κB/p65-Untereinheit als Reaktion auf verschiedene Stressbedingungen charakterisiert. Wir konnten zeigen, dass die Anhäufung von LC3 im Zellkern nach der NF-κB-Aktivierung mit p65 interagiert, was durch die Ubiquitinierung des p65-Proteins gefördert und durch den Cargo-Rezeptor p62 erkannt wird. Zusammengenommen weisen diese Daten auf eine neue Rolle von p62 beim Transport von im Kern ubiquitiniertem p65 zu Autophagosomen hin, wo es abgebaut wird, um die entzündungsbedingte NF-κB-Hyperaktivierung zu kontrollieren. Diese Erkenntnisse sind wichtig für die Entwicklung neuer therapeutischer Strategien gegen Krankheiten, die mit einer gestörten Autophagie und einer konstitutiven NF-κB-Aktivität einhergehen. Die NF-κB-Signalübertragung spielt nicht nur eine entscheidende Rolle bei Entzündungen und der Tumorbildung, sondern ist auch für Entwicklungsprozesse wichtig. Durch die Etablierung von 3D-Organoid-Kulturen aus dem Dünndarm und unter Verwendung verschiedener Mauslinien weisen wir im zweiten Teil der Arbeit nach, dass NF-κB eine wichtige Funktion bei der Zelldifferenzierung und der Erhaltung von Stammzellen in vivo und ex-vivo spielt. Wir konnten zeigen, dass die Proliferation und das Absterben von Darmepithelzellen (IEC) bei Mäusen mit ubiquitärer Unterdrückung der NF-κB-Aktivität unverändert sind, während die Zahl der Becherzellen auf Kosten der Paneth-Zellen zunimmt. Zusammenfassend lässt sich sagen, dass unsere Ergebnisse eine neue IEC-immanente Rolle von NF-κB bei Entscheidungen über das Zellschicksal und die Differenzierung aufzeigen, die über die Regulierung der Wnt-Signale und der Sox9-Expression stromabwärts von NF-κB erfolgt. Die hier beschriebenen Erkenntnisse verbessern unser Verständnis der NF-κB-Funktionen in der Stammzellbiologie, die, wenn sie dereguliert sind, auch Auswirkungen auf die Entzündung des Darms und die Tumorentstehung haben., The interplay between NF-κB signaling and autophagy has been suggested to regulate inflammation in different cellular contexts and in response to different stimuli. However, the molecular mechanism by which these two pathways interact to regulate the inflammatory response remains elusive. By using biochemical analysis and imaging techniques, we characterized for the first time the interaction of autophagic marker LC3 and NF-κB/p65 subunit in response to different stress conditions. We demonstrated that the accumulation of LC3 within the nucleus interacts with p65 following NF-κB activation, which is promoted by ubiquitination of p65 protein and recognized by the cargo receptor p62. Together, these data identify a novel role for p62 in trafficking nuclear-ubiquitinated p65 to autophagosomes for degradation to control inflammation-driven NF-κB hyperactivation. These findings are important for developing novel therapeutic strategies against diseases involving defective autophagy and constitutive NF-κB activity. In addition to its critical role in inflammation and tumor formation, NF-κB signaling is essential in developmental processes. Establishing 3D organoid culture from the small intestine and using different mouse lines, we prove in the second part of the thesis that NF-kB plays an important function in cell differentiation and stem cell maintenance in vivo and in ex-vivo. We demonstrated that while intestinal epithelial cell (IEC) proliferation and death are unaltered in mice with ubiquitous suppression of NF-κB activity, goblet cell numbers increase at the expense of Paneth cells. In summary, our results revealed a novel IEC-intrinsic role of NF-κB in cell fate decisions and differentiation which occur via regulation of Wnt signaling and Sox9 expression downstream of NF-κB. The findings described here improve our understanding of NF-κB functions in stem cell biology which, when deregulated, also have an impact on intestinal inflammation and tumorigenesis.

Hepatocellular carcinoma is the second most worldwide cancer-related death. Due to the absence of symptoms in the early stage of the disease and the late diagnosis, a curative treatment seems to be mostly ineffetctive. The multi-kinase-inhibitors Sorafenib (Nexavar) and Lenvatinib (Lenvima) are the only evidence-based drugs with a first-line use approved by the European Medicines Ageny (EMA) for systemic treatment of advanced local or even metastatic stage of hepatocellular carcinoma within the Child-Pugh-Score A. Despite many attempts of the current investigations to identify molecular-targeting drugs beside sorafenib, the results have been unsatisfying so far. Hopefully, the induction of epigenetic modifications could represent a promising approach for the development of a new systemic therapy. The application of deacetylase inhibitors (DACis) results in an increase of endoplasmatic-reticulum stress and autophagic process, which trigger liver cancer cell death. The study here proposed focused on the role exerted by the highly evolutionary conserved transcription factor CUX1 in DACi-mediated endoplasmic reticulum stress/autophagic mechanisms in HCC cells. CUX1 is responsible for promoting cell motility, invasion and cell cycle progression as well as for causing cell death resistance. Clinical studies associate a high level of CUX1 transcript in several cancers. The aim of this study was to elucidate the oncogenic and/or tumor suppressive potential of CUX1 in HCC cells. CUX1 transient knockdown was performed in two human HCC-cell lines Hep3B (p53-deficient) and HepG2 (p53-wildtype). After a 6 hour treatment (RT-qPCR) and 24 hour treatment (RT-qPCR and Western Blot) with Thapsigargin and the DACis Trichostatin A (TSA), SAHA and Panobinostat the expression of autophagic markers was detected by RT-qPCR and Western Blot. Fluorescent microscopy was performed to visualize the autophagosome maturation after autophagy induction via Panobinostat-treatment in the absence of CUX1. The effect of transient CUX1-knockdown on cell proliferation was detected by real time cell viability assays. CUX1 transient knockdown reduced the efficacy of short-term (6h) administration of especially thapsigargin and panobinostat in promoting the expression of ER-stress-markers. These findings could reveal oncogenic potential of CUX1-regulation regarding ER-Stress-mediated drug resistance. The real time cell viability assay pointed out an altered cell proliferation with the most significant effect in the absence of CUX1 and the administration of panobinostat in Hep3B cells. Regarding a possible resistance to apoptosis, CUX1 exerts an oncogenic function. Decreased mRNA-expression of autophagic markers after CUX1-Knockdown suggests impaired autophagic mechanism, depending on the time of administration with the DACis and the p53-levels of the examined cell lines. The measurement of increased Beclin1 and MAP1LC3B protein levels in the Western Blot analysis contributed to a reduced efficacy of the compounds to perform autophagy after CUX1-silencing. Autophagy, as a catabolic process, leads to a massive protein degradation. An increase of the protein levels within a reduction of the mRNA expression of autophagic markers therefore supports an impairment of autophagy in the absence of CUX1. A reduced maturation of the autophagosome vesicles (Hep3B-cells) as well as impaired autophagosome-lysosome-fusion (HepG2-cells) was observed under fluorescence microscopy in the absence of CUX1 after the administration of panobinostat. These findings suggest a tumor suppressive potential of CUX1 on HCC cells performing autophagic cell death after DACi treatment. Regarding to current studies implicating the contribution of CUX1 to resistance towards apoptosis highlights a possible interaction between autophagic-related cell death and apoptosis. CUX1 could either inhibit, promote or necessitate apoptosis and autophagy. While showing its pro-tumorigenic potential in HCC, CUX1 simultaneously promotes autophagy-related cell death and therefore exerts anti tumorigenic role too. Ein zunächst eher symptomarmer Krankheitsverlauf und eine somit mehrheitliche Diagnosestellung in fortgeschrittenen Krankheitsstadien erschweren eine kurative Behandlung. In lokoregionär fortgeschrittenen oder fernmetastasierten Krankheitsstadien des HCCs der Child-Pugh-Klassifikation A gelten die Multikinaseinhibitoren Sorafenib (Nexavar) und Lenvatinib (Lenvima ) als bislang einzige von der European Medicines Agency (EMA) zugelassene, medikamentöse Therapieoptionen der Erstlinientherapie. Eine Überlegenheit anderer molekularer, therapeutischer Ansätze gegenüber diesen Multikinaseinhibitoren konnte bisher nicht gezeigt werden. Die Induktion epigenetischer Modifikation in Krebszellen stellt währenddessen einen neuen Ansatz bezüglich der Systemtherapie des metastasierten HCCs dar. Die Applikation von Histondeacetylaseinhibitoren (DACis) führt zu vermehrten Stressreaktionen des Endoplasmatischen Retikulums und einer konsekutiven Induktion autophagischer Prozesse, welche die HCC-Zellen in einen protrahierten Zelltod senden. Basierend auf dieser Grundlage wurde in dieser experimentellen Arbeit der evolutionär hoch konservierte und bislang unzureichend erforschte Transkriptionsfaktor CUX1 auf sein etwaiges regulatorisches Potential hinsichtlich der ER-Stress- und Autophagie-Induktion nach DACi-Behandlung von HCC-Zellen untersucht. Bisher werden CUX1 in maligne transformierten Zellen regulatorische Fähigkeiten bezüglich der Zellzyklusprogression, Zellmotilität und Zellinvasion sowie Induktion einer Apoptoseresistenz attestiert. Es galt als Ziel dieser Arbeit, das onkogene aber auch das tumorsuppressive Potential von CUX1 in HCC-Zellen unter Substanzbehandlung zu analysieren. Anhand zweier humaner HCC-Zelllinien mit p53-wildtyp Status (HepG2) und p53-defizientem Status (Hep3B) wurde über die Transfektion mit einer siRNA für CUX1 ein Knockdown erzielt. Nach 6- und nach 24-stündiger Applikation der DACis TSA, SAHA und Panobinostat sowie des Kalziumkanal-Inhibitors Thapsigargin wurde in Gegenüberstellung der CUX1-Knockdown-Gruppe und einer Kontrollgruppe die Expression der ER-Stress-Indikatoren BIP, CHOP, ATF4 und AT6 sowie die Expression der Autophagie-Marker BECN1, MAP1LC3B, TFEB und UVRAG durch qRT-PCR-Versuche und Western Blot-Analysen untersucht. Die fluoreszenzmikroskopische Darstellung des Effekts eines CUX1-Knockdowns auf die Autophagosomen-Reifung sowie die Echtzeit-Analyse der Zellproliferation unter CUX1-Defizienz nach ER-Stress- und Autophagie-Induktion sollten die Experimente vervollständigen. Die Ergebnisse der genannten Versuche lassen regulatorisches Potential von CUX1 bezüglich Thapsigarin- und DACi-getriggerter ER-Stress und Autophagie-Mechanismen vermuten. Onkogene Eigenschaften des Transkriptionsfaktors werden einerseits durch eine defizitäre Expression der ER-Stress-Marker BIP, ATF4 und ATF6 unter CUX1-Knockdown und kurzzeitiger Substanzbehandlung deutlich. CUX1 könnte die Bildung von Chemo-oder Strahlenresistenzen durch die Stimulation des ER-Stresses als Initiator zelleigener Reparaturmechanismen begünstigen. Andererseits wird onkogenes Potential von CUX1 in den Zellproliferationsmessungen deutlich. Die Hep3B-Zelllinie wies hierbei unter CUX1-Knockdown und Panobinostat-Behandlung eine signifikante Abnahme der Zellproliferation auf. Die Förderung von Apoptose-Resistenzen durch CUX1 könnte diesen Proliferations-Stopp in den xCELLigence®-Analysen begründen. Tumorsuppressive Eigenschaften von CUX1 wurden durch den Nachweis defizitärer Autophagie-Mechanismen und einer möglichen Verzögerung des autophagic cell death als antikanzerogener Mechanismus deutlich. In Abhängigkeit des p53-Status der untersuchten Zelllinie sowie der Inkubationsdauer mit den DACis konnte dabei eine herunterregulierte Transkript-Expression der Autophagie-Marker nachgewiesen werden. Die Akkumulation der autophagischen Proteine MAP1LC3B und Beclin1 in den Western Blot Analysen sowie Defizite der Autophagosomen-Formation und der Autolysosomen-Bildung in der Fluoreszenzmikroskopie stützten die These defizitärer Autophagie-Mechanismen unter CUX1-Knockdown. Bezüglich jüngster Erkenntnisse zur CUX1-vermittelten Resistenzentwicklung gegen die Apoptose erscheint die Zusammenschau des programmierten Zelltodes und des Autophagie-bedingten Zelltodes sinnvoll. Durch ein komplexes Netzwerk an Signalkaskaden können sich Apoptose und Autophagie gegenseitig verstärken, hemmen oder wechselseitig bedingen, sodass CUX1 als multifunktionaler Transkriptionsfaktor und regulatorischer Mediator agieren könnte. Dabei soll die mehrheitliche Auffassung von CUX1 als Onkogen keineswegs negiert werden, sondern vielmehr das Potential einer simultanen tumorsuppressiven Wirkung im Autophagie-bezogenen Therapieregime im HCC betont werden. Bereits initiierte klinische Studien zur Kombinationstherapie aus Sorafenib und Panobinostat im metastasierten Krankheitsstadium des HCCs bilden eine Grundlage für klinische Forschungsansätze unter Einbezug von CUX1.

As part of innate immune defenses, macroautophagy/autophagy targets viruses and viral components for lysosomal degradation and exposes pathogen-associated molecular patterns to facilitate recognition. However, viruses evolved sophisticated strategies to antagonize autophagy and even exploit it to promote their replication. In our recent study, we systematically analyzed the impact of individual SARS-CoV-2 proteins on autophagy. We showed that E, M, ORF3a, and ORF7a cause an accumulation of autophagosomes, whereas Nsp15 prevents the efficient formation of autophagosomes. Consequently, autophagic degradation of SQSTM1/p62 is decreased in the presence of E, ORF3a, ORF7a, and Nsp15. Notably, M does not alter SQSTM1 protein levels and colocalizes with accumulations of LC3B-positive membranes not resembling vesicles. Infection with SARS-CoV-2 prevents SQSTM1 degradation and increases lipidation of LC3B, indicating overall that the infection causes a reduction of autophagic flux. Our mechanistic analyses showed that the accessory proteins ORF3a and ORF7a both block autophagic degradation but use different strategies. While ORF3a prevents the fusion between autophagosomes and lysosomes, ORF7a reduces the acidity of lysosomes. Das klassische Hodgkin Lymphom (cHL) kann oft erfolgreich behandelt werden, die Radio- und Chemotherapie sind jedoch stark toxisch und haben viele Langzeitwirkungen. Eine Behandlungsmöglichkeit mit weniger Nebenwirkungen ist der DNA-Methyltransferase Inhibitor 5-Aza-2'-desoxycytidin (5-Aza-dC). Dieser wird bereits in der Akuten Myeloischen Leukämie eingesetzt. Unsere Arbeitsgruppe konnte in einer früheren Arbeit zeigen, dass 5-Aza-dC zytotoxisch auf cHL Zelllinien wirkt, gleichzeitig aber auch überlebensfördernde Signalwege induziert. In der vorliegenden Arbeit untersuchten wir die Wirkung von 5-Aza-dC auf Autophagie und oxidativen Stress in cHL Zelllinien. Wir beobachteten mit Hilfe von LC3B eine Hochregulierung von Autophagie nach 5-Aza-dC Behandlung. Durch Inhibition der Autophagie mittels synthetischen Inhibitoren und small hairpin RNAs (shRNA) für ATG5 konnten wir lediglich einen additiven Effekt der Autophagie Inhibition zur 5-Aza-dC Behandlung feststellen. Unsere Gene-Expression-Profiling Daten zeigten eine Induktion von PRDX2 nach 5-Aza-dC Behandlung. Dies konnten wir mittels quantitativer Echtzeit Polymerase-Kettenreaktion (Q-RT-PCR) bestätigen. Wir erstellten zwei shRNAs für PRDX2 und ermittelten die Wachstumsdynamiken nach 5-Aza-dC Behandlung. Hier konnten wir eine Wachstumshemmung der PRDX2 herunterregulierten Zellen im Vergleich zu den Kontrollzellen beobachten. PRDX2 könnte dadurch im cHL ein geeignetes Therapieziel für eine Kombinationstherapie mit 5-Aza-dC sein.

Inflammatory bowel diseases (IBD) are chronic, idiopathic disorders of the gastrointestinal tract that affect mainly the young population. Disease-targeted, tolerable, cost-effective medications to treat IBD are still awaited. The discovery of the crucial involvement of the autophagy pathway in the genetic predisposition of IBD opened a novel avenue for exploration in IBD therapeutics. Autophagy is a vital process that plays a central role in maintaining cellular homeostasis and pharmacological regulation of autophagy has proven to be beneficial in several diseases. Hence, we evaluated the efficacy of P140 - a therapeutic phosphopeptide known to modulate autophagy processes in other autoimmune and inflammatory conditions - in murine models of IBD. We have demonstrated that the peptide exerts protective effects on colitis models and corrects the pathological dysfunctions in different autophagy pathways. Thus, after the era of drugs classified as "disease-modifying" therapeutics, emerging "mechanism-guided" pharmaceuticals seem to hold a lot of promises for treating inflammatory diseases., Les maladies inflammatoires de l'intestin (MICI) sont des troubles chroniques idiopathiques du tractus gastro-intestinal qui touchent principalement la population jeune. On attend toujours des médicaments ciblés sur la maladie, tolérables et rentables pour traiter les MICI. La découverte de l'implication cruciale de la voie de l'autophagie dans la prédisposition génétique des MICI a ouvert une nouvelle voie d'exploration pour la thérapie des MICI. L'autophagie est un processus vital qui joue un rôle central dans le maintien de l'homéostasie cellulaire et la régulation pharmacologique de l'autophagie s'est avérée bénéfique dans plusieurs maladies. Nous avons donc évalué l'efficacité du P140 - un phosphopeptide thérapeutique connu pour moduler les processus d'autophagie dans d'autres conditions auto-immunes et inflammatoires - dans des modèles murins de MICI. Nous avons démontré que le peptide exerce des effets protecteurs sur les modèles de colite et corrige les dysfonctionnements pathologiques dans différentes voies d'autophagie. Ainsi, après l'ère des médicaments classés comme thérapeutiques "modificatrices de la maladie", les nouveaux produits pharmaceutiques "guidés par les mécanismes" semblent très prometteurs pour le traitement des maladies inflammatoires.

Background and purpose: Alterations of neuronal excitability has been recently characterized as an important vulnerability modulator of motoneurons (MNs) in multiple neurodegenerative diseases including amyotrophic sclerosis (ALS). The serum response factor (SRF) has been known as a neuronal activity dependent transcription factor regulating various genes which are beneficial for neuronal outgrowth, regeneration and survival. The role of SRF in ALS MNs during disease progression is poorly understood so far. In this thesis, SRF expression and its function in MNs were investigated during the process of MN degeneration in ALS. Methods: Transgenic mice with a point mutation (G93A) in the human superoxide dismutase gene (huSOD1G93A) was used as an established ALS mouse model. In these mice, Srf was specifically deleted in MNs. Behavior tests (body weight, neurological score, grip strength, inverted grid test, pole test and survival analysis) were carried out to evaluate the motor ability, disease progression and survival. Mice were fixed with perfusion at multiple timepoints and immunofluorescence was performed for protein detection and colocalization. Mice lumbar spinal cords were harvested and frozen in liquid nitrogen for RNA isolation and gene expression detection with qPCR. Human spinal cords of ALS patients were used for immunohistochemistry (IHC). Fluorescence images were acquired on confocal microscopy and IHC images were acquired on brightfield microscopy. Images were quantified in ImageJ. Statistical analysis was performed in GraphPad. Bioinformatic analysis was performed using Eukaryotic Promoter Database. Results: Expression analysis revealed higher SRF levels in a subpopulation of MNs which were more vulnerable to ALS and are typically earlier affected. SRF levels in the vulnerable population of MNs were decreased in the SOD1G93A model of ALS over time. MN-specific SRF deletion in SOD1G93A model led to an earlier disease onset, characterized by a lower body weight, worse neurological score, lower grip strength, and worse outcomes from inverted test and pole test. Furthermore, a stronger microgliosis, a more pronounced neuromuscular junction (NMJ) denervation and an increased misfolded protein burden were observed after motoneuronal SRF ablation in the SOD1G93A mouse model. Interestingly, the effect of SRF deficiency vanished at later phase of disease progression, characterized by no change of motor disability or NMJ denervation. SRF was recognized as a potential regulator of autophagy related genes to maintain the intracellular proteostasis in MNs in the SOD1G93A model of ALS. The significantly different level of proteins involved in the macro-autophagy pathway in MNs in early disease phase indicated an impairment of autophagy in the SRF-deficient MNs in SOD1G93A. The bioinformatic analysis revealed that SRF could be directly binding at the cytosine adenine rich guanine (CArG) boxes in the promoters of some autophagy related target genes. SRF was expressed in MNs of ALS patient samples and SRF appeared to be mislocalized in some human ALS MNs. This supports a high translational relevance of mouse data obtained to the human disease condition. Conclusion: SRF was recognized as a modulator of vulnerability in ALS MNs. A beneficial effect of SRF was recognized by a loss of function model in ALS mice which showed faster disease progression and NMJ denervation after SRF ablation. Impaired autophagy was noticed in ALS MNs with Srf deficiency at protein level. Several autophagy related genes containing CArG boxes in their promoters were predicted to be target genes of SRF. These data suggest a new connection of SRF and autophagy.

The objective was to further elucidate one part of the complicated pathomechanism of Parkinson's disease (PD). On the one hand, it consists of a dopamine deficiency in the motor loop of the basal ganglia, especially the nigrostriatal pathway. On the other hand, Lewy bodies form from misfolded α-synuclein which accumulates in the dorsolateral substantia nigra pars compacta and leads to neuronal death. α-synuclein aggregates can be degraded by autophagy. This process ends with the fusion of the autophagosome which contains the protein to be degraded with a lysosome to form an auto(phago)lysosome. To date, there are no known causal therapeutic approaches in the treatment of the very common neurodegenerative disease PD. In the first part of my experiments, I investigated the distribution of α-synuclein aggregates under the influence of FYCO1. It revealed that FYCO1 causes the α- synuclein particles to move toward the distal end in their neurites. This suggests that binding to the microtubuli network in dendrites occurs via kinesins and FYCO1.In the second part of my experiments, I analyzed the distribution of autophagosomes and autolysosomes under the influence of FYCO1. I made use of the principle of GFP quenching to distinguish the vesicles. I found that FYCO1 caused accumulation of autophagosomes in neurons. Again, the accumulation occurred mainly in the neurites. One can speculate that these neutral particles in this case are not autophagosomes but pH-neutral precursors of autolysosomes. Thus, FYCO1 mediates the binding of autophagosomes to lysosomes in neurons as well, fueling the cell's autophagy process. I have also been able to show that FYCO1 enhances the transport of autolysosomes and autophagosomes to the soma periphery and of autolysosomes to the dendrites’ tips. Implant-associated infections are severe complications in orthopaedic surgery. Beneath S. epidermidis, S. aureus is the most prevalent pathogen responsible for orthopaedic infection. Treatments using antibiotics are often unsuccessful due to biofilm formation or the intracellular persistence of S. aureus. In the literature, few reports have taken S. aureus as a facultative intracellular pathogen. This study focuses on adhesion, invasion and intracellular survival of clinical isolates of S. aureus on/in osteoblasts. Isolates obtained from implant-associated infection were determined by API® Staph. test and 16S rRNA sequencing. Further characterization was carried out using clear zone formation, collagenase and oxidase test, growth analysis, antibiotic resistance and biofilm formation analysis. For bacteria-osteoblasts interaction studies, S. aureus ATCC 29213 and eight S. aureus isolates (Pat 2, 4, 6, 9, 28, 36, 42, 45), as well as confluent layers of osteoblast like cell lines SaOS2 and MG63 were used. Standard infection model was established using S. aureus ATCC 29213 to infect SaOS2 and MG63. Based on this model, adhesion and invasion assay with S. aureus isolates were processed. MTT and respiration assay were performed to get the metabolic activity results of the infected osteoblasts. PCR and qPCR were applied to identify genes involved in adhesion and the corresponding expression level. Mass spectrometry was used for quantitative protein analysis. Induced and inhibited autophagy, Western blot were carried out to analyse the role of autophagy in bacteria-osteoblasts interactions. Based on the characterization and sequence analysis from the perspective of homology, isolates Pat 2, 4, 6, 9, 28, 36, 42, 45 were classified as S. aureus. In the infection model, a significant stronger adherence of S. aureus ATCC 29213 to SaOS2, rather than MG63, was observed. Bacteria harvested in stationary growth phase showed a stronger adherence to osteoblasts as compared to bacteria harvested in the logarithmic growth phase. Three S. aureus isolates (Pat 36, 42, 45) were found to adhere to and invade in osteoblasts strongly. PCR showed that eno, fnbA, clfA, clfB, ebpS, psmA, psmB, eap, sdrD, fmtB and ebh genes were expressed in all S. aureus isolates. Furtherly, qPCR showed that clfA might be a candidate gene related to bacterial adherence, because it was overexpressed only in in the adhesive S. aureus (Pat36, Pat42, Pat45) and S. aureus ATCC 29213. MTT assay showed that metabolic activity was independent of the infection with S. aureus. Respiration assay indicated that metabolic activity of both SaOS2 and MG63 cell lines were stimulated after being infected with selected S. aureus isolates (Pat 9, 36 and S. aureus ATCC 29213). The relation of cell adhesion and the gene expression under biofilm condition was not closely correlated. Proteome analysis of two different osteoblast cell lines with or without infection with S. aureus showed that genes involved in adhesion were mostly not expressed in osteoblasts. Only fibronectin and collagen were detectable by mass spectrometry. To conclude, it can be seen that neither the presence of the respective genes nor the gene products can be regarded as a sufficient predictive marker for adhesion. Gene expression analysis suggests that there may not be a uniform mechanism behind the adhesion of the examined S. aureus isolates, but that different adhesion proteins are involved. The increased adhesion is not correlated with increased invasion, which makes adhesion uninteresting as a possible therapeutic target. With respect to intracellular survival, autophagy in the host cells was turned on after infection with the S. aureus isolates tested here, indicating an onset of autophagy, which was regardless of the outcome. This study highlights a controversial role of the autophagy pathways with respect to clearance of S. aureus indicating that autophagy is necessary but not sufficient for infection. Furthermore, bacteria-induced autophagy differs from a starvation-induced one, making autophagy an interesting treatment option for the implant-associated infection.

Charcot-Marie-Tooth (CMT) ist die weltweit häufigste vererbte periphere Neuropathie. Mutationen in mehr als 90 Genen wurden mit dem Auftreten von CMT in Verbindung gebracht, die für ein breites Spektrum an klinischen Phänotypen verantwortlich sind. CMT ist nach wie vor eine unheilbare neurologische Erkrankung mit verheerenden Auswirkungen auf die Lebensqualität der Patienten. Das Verständnis der zugrundeliegenden molekularen Mechanismen ist entscheidend für Fortschritte in der CMT-Therapie. In dieser Arbeit konzentrieren wir uns auf eines der am häufigsten mutierten Proteine bei axonaler CMT, das Hitzeschockprotein Familie B Mitglied 1 3 (HSPB1). HSPB1 ist ein ubiquitär exprimiertes molekulares Chaperon, das für die Proteinfaltung und für Abbauprozesse wie die Autophagie unerlässlich ist. In Zellen führt ein Fehlen von HSPB1 zu einer Abnahme der Autophagie-Aktivität, die durch die Wiederexpression von HSPB1 wiederhergestellt wird. Darüber hinaus wurde bereits berichtet, dass HSPB1 mit dem Autophagie-Rezeptor SQSTM1 interagiert und dass HSPB1-Mutanten unterschiedliche Bindungskapazitäten aufweisen. Wie HSPB1 an der Autophagie beteiligt ist, ist noch nicht ganz klar. Die hier vorgeschlagene Methode soll einen besseren Einblick in die Beziehung zwischen HSPB1 und einer Reihe von Ubiquitin-abhängigen selektiven Autophagie-Rezeptoren (SARs) geben. Es wurden Co-Immunopräzipitationen durchgeführt, um die direkte Bindung von HSPB1 an die SARs nachzuweisen und zu testen, wie sich HSPB1-Mutationen auf die direkte SAR-Bindung auswirken. Dies wurde durch Immunzytochemie validiert, um die Kolokalisierung von HSPB1 und SARs sichtbar zu machen. Wir haben auch getestet, ob andere SARs unterschiedliche Bindungskapazitäten an HSPB1-Mutanten aufweisen. Um zu verstehen, wie die Abwesenheit von HSPB1 einen Defekt in der Autophagie verursacht, wurde die Expression der SARs in Abwesenheit von HSPB1 zu verschiedenen Zeitpunkten und unter verschiedenen Belastungen quantifiziert. Bei der Autophagie ist die Funktion der selektiven Autophagie-Rezeptoren nicht völlig unabhängig von HSPB1, da ein Fehlen von HSPB1 zu einer Verringerung der SAR-Expression führt und HSPB1-Mutanten im Vergleich zum Wildtyp-Protein veränderte Interaktionen aufweisen. Charcot-Marie-Tooth (CMT) is the most common inherited peripheral neuropathy worldwide. Mutations in more than 90 genes have been associated with the onset of CMT that are responsible for the generation of a wide spectrum of clinical phenotypes. CMT is still and uncurable neurological disorder with devastating impact on the patients quality of life. The understanding of the underlying molecular mechanisms is essential to progress in the CMT therapy. In this thesis, we will focus on one of the most frequently mutated proteins in axonal CMT, heat shock protein family B member 1 (HSPB1). HSPB1 is an ubiquitously expressed molecular chaperone essential in protein folding and in degradation pathways like autophagy. In cells HSPB1 depletion leads to a decrease of the autophagy activity which is restored upon HSPB1 re-expression. In addition, it was already reported that HSPB1 interacts with the autophagy receptor SQSTM1 and that HSPB1 mutants have different binding capacities. How HSPB1 is involved in autophagy is not yet well understood. The methodology proposed here tries to give a better insight into its relationship between HSPB1 and a set of ubiquitin-dependent selective autophagic receptors (SARs). Co-immunoprecipitations were performed in order to prove the direct binding of HSPB1 to the SARs and to test how HSPB1 mutations impact on the direct SARs binding. This was validated by immunocytochemistry to visualize the HSPB1-SARs colocalization. We also tested whether other SARs prove to have different binding capacities to HSPB1 mutants. In order to understand how HSPB1 depletion causes defect in autophagy, the expression of the SARs in absence of HSPB1 was quantified at different time points and under different stress. In autophagy the function of the selective autophagy receptors is not fully independent from HSPB1, since HSPB1 depletion causes a reduction in the SARs expression and mutant HSPB1 show altered interactions compared to the wild-type protein.
