Abstract Background As an important forage in arid and semi-arid regions, Agropyron cristatum provides livestock with exceptionally high nutritional value. Additionally, A. cristatum exhibits outstanding genetic characteristics to endure drought and disease. Therefore, rich genetic diversity serves as a cornerstone for the improvement of major food crops. The purposes of this study were to systematically describe mitogenome of A.cristatum and preliminarily analyze its internal variations. Result The A. cristatum mitogenome was a single-ring molecular structure of 381,065 bp that comprised 52 genes, including 35 protein-coding, 3 rRNA and 14 tRNA genes. Among these, two pseudoprotein-coding genes and multiple copies of tRNA genes were observed. A total of 320 repetitive sequences was found to cover more than 10% of the mitogenome (105 simple sequences, 185 dispersed and 30 tandem repeats), which led to a large number of fragment rearrangements in the mitogenome of A. cristatum. Leucine was the most frequent amino acid (n = 1087,10.8%) in the protein-coding genes of A. cristatum mitogenome, and the highest usage codon was ATG (initiation codon). The number of A/T changes at the third base of the codon was much higher than that of G/C. Among 23 PCGs, the range of Pi values is from 0.0021 to 0.0539, with an average of 0.013. Additionally, 81 RNA editing sites were predicted, which were considerably fewer than those reported in other plant mitogenomes. Most of the RNA editing site base positions were concentrated at the first and second codon bases, which were C to T transitions. Moreover, we identified 95 sequence fragments (total length of 34, 343 bp) that were transferred from the chloroplast to mitochondria genes, introns, and intergenic regions. The stability of the tRNA genes was maintained during this process. Selection pressure analysis of 23 protein-coding genes shared by 15 Poaceae plants, showed that most genes were subjected to purifying selection during evolution, whereas rps4, cob, mttB, and ccmB underwent positive selection in different plants. Finally, a phylogenetic tree was constructed based on 22 plant mitogenomes, which showed that Agropyron plants have a high degree of independent heritability in Triticeae. Conclusion The findings of this study provide new data for a better understanding of A. cristatum genes, and demonstrate that mitogenomes are suitable for the study of plant classifications, such as those of Agropyron. Moreover, it provides a reference for further exploration of the phylogenetic relationships within Agropyron species, and establishes a theoretical basis for the subsequent development and utilization of A. cristatum plant germplasm resources.

Abstract A novel prefabricated segmental guardrail is proposed to facilitate connections between guardrails and between guardrails and bridge decks by casting ultrahigh-performance concrete (UHPC) joints in situ. Through finite element crash simulation analysis of three types of vehicles and crash tests of real vehicles, the prefabricated segmental guardrail with a UHPC connection was systematically evaluated in terms of its energy-absorbing capacity, vehicular acceleration, post-impact trajectory of the impacting vehicle, and behaviour of the guardrail upon impact. During the evaluation process, performance comparisons of the prefabricated segmental guardrails are made with the monolithic concrete guardrails. The results indicate that the performance of the prefabricated segmental guardrail with a UHPC connection was superior to that of the conventional concrete monolithic guardrails: it exhibited a higher level of crash performance, the occupants of the impacting vehicle were better protected, and the impacting vehicle exhibited better post-collision stability. Finally, the convenience of the prefabricated segmental guardrails with UHPC connections was proven in practical engineering applications.

Abstract This paper mainly introduces the emergency repair process for small- and medium-span bridges. The causes of deterioration were analysed by investigating old bridges. After comparison and selection of schemes, the scheme of beam replacement was confirmed using a lightweight steel ultra-high performance concrete (UHPC) composite beam as the superstructure. The main components of the calculations and the design are introduced in detail. Finally, through the load test of the bridge, it was shown that the lightweight steel-UHPC composite beam had good performance and met the requirements of Highway Level I bearing capacity. The lightweight steel-UHPC composite beam described in this paper has the characteristics of high strength, light weight, fast construction, excellent working performance, and remarkable social and economic benefits. It can be popularised and applied in the emergency repair of small- and medium-span bridges and new bridges.

Abstract Limited test data hinder the accurate prediction of mechanical strength and permeability of permeable cement-stabilized base materials (PCBM). Here we show a kriging-based surrogate model assisted artificial neural network (KS-ANN) framework that integrates laboratory testing, mathematical modeling, and machine learning. A statistical distribution model was established from limited test data to enrich the dataset through the combination of markov chain monte carlo simulation and kriging-based surrogate modeling. Subsequently, an artificial neural network (ANN) model was trained using the enriched dataset. The results demonstrate that the well-trained KS-ANN model effectively captures the actual data distribution characteristics. The accurate prediction of the mechanical strength and permeability of PCBM under the constraint of limited data validates the effectiveness of the proposed framework. As compared to traditional ANN models, the KS-ANN model improves the prediction accuracy of PCBM’s mechanical strength by 21%. Based on the accurate prediction of PCBM’s mechanical strength and permeability by the KS-ANN model, an optimization function was developed to determine the optimal cement content and compaction force range of PCBM, enabling it to concurrently satisfy the requirements of mechanical strength and permeability. This study provides a cost-effective and rapid solution for evaluating the performance and optimizing the design of PCBM and similar materials.

Abstract Intracranial aneurysms (IAs) are present in 2–6% of the global population and can be catastrophic upon rupture with a mortality rate of 30–50%. IAs are commonly detected through time-of-flight magnetic resonance angiography (TOF-MRA), however, this data is rarely available for research and training purposes. The provision of imaging resources such as TOF-MRA images is imperative to develop new strategies for IA detection, rupture prediction, and surgical training. To support efforts in addressing data availability bottlenecks, we provide an open-access TOF-MRA dataset comprising 63 patients, of which 24 underwent interval surveillance imaging by TOF-MRA. Patient scans were evaluated by a neuroradiologist, providing aneurysm and vessel segmentations, clinical annotations, 3D models, in addition to 3D Slicer software environments containing all this data for each patient. This dataset is the first to provide interval surveillance imaging for supporting the understanding of IA growth and stability. This dataset will support computational and experimental research into IA dynamics and assist surgical and radiology training in IA treatment.

Corrosion-erosion performances of Ti-6.5Al–2Zr–1Mo–1V (TA15) alloy fabricated by two laser additive manufacturing technologies, namely selective laser melting (SLM) and laser metal deposition (LMD) were investigated, and compared with those of traditional forged counterpart. Results show that SLM-ed TA15 possesses better corrosion, wear, and tribocorrosion resistances than LMD-ed and forged TA15. This is mainly attributed to the ultrafine-grain αʹ phase in the microstructure of SLM-ed sample, which improves the hardness and thereby enhances the wear and tribocorrosion resistance. Additionally, the complete αʹ phase formed in SLM-ed sample also avoids the galvanic corrosion that occurs at the α+β-type LMD-ed and forged samples.

Abstract Background MicroRNA-200b-3p (miR-200b-3p) plays a pivotal role in inflammatory responses and is implicated in various inflammatory disorders. In this study, we aim to explore the role of miR-200b-3p in the inflammatory response in heart failure (HF). Methods Patients diagnosed with heart failure and age-matched healthy controls were studied. Peripheral blood samples from participants were collected for RNA-seq analysis to explore the expression profile of miR-200b-3p. The predictive value of miR-200b-3p and ZEB1 in the prognosis of heart failure was evaluated by analyzing the receiver operating characteristic (ROC) curve. Bioinformatics analysis and double luciferase reporter gene analysis were used to confirm the interaction between miR-200b-3p and ZEB1. Real-time quantitative polymerase chain reaction (QRT-PCR) was used to detect the expression levels of miR-200b-3p and ZEB1 in cardiopulmonary bypass. Additionally, the effects of miR-200b-3p on myocardial cell line (H9c2) injury were evaluated by enzyme-linked immunosorbent assay (ELISA). Results In the extracardiac circulation of HF patients, miR-200b-3p expression was significantly reduced, while ZEB1 levels were notably elevated. Analysis of the ROC curve revealed that miR-200b-3p and ZEB1 have predictive value in the prognosis of HF patients. The double luciferase reporter experiment demonstrated that miR-200b-3p binds to ZEB1 and inhibits its expression. Overexpression of miR-200b-3p demonstrated a remarkable ability to alleviate inflammation and inhibit the damage to myocardial cells in vivo. Conclusion MiR-200b-3p can target and inhibit ZEB1, reducing the inflammatory reaction of myocardial cells. The miR-200b-3p/ZEB1 network may be helpful in preventing and treating HF.

Introduction: Rice flowering is a major agronomic trait, determining yield and ecological adaptability in particular regions. ABA plays an essential role in rice flowering, but the underlying molecular mechanism remains largely elusive. Objectives: In this study, we demonstrated a “SAPK8-ABF1-Ehd1/Ehd2” pathway, through which exogenous ABA represses rice flowering in a photoperiod-independent manner. Methods: We generated abf1 and sapk8 mutants using the CRISPR-Cas9 method. Using yeast two-hybrid, Pull down, BiFC and kinase assays, SAPK8 interacted and phosphorylated ABF1. ABF1 directly bound to the promoters of Ehd1 and Ehd2 using ChIP-qPCR, EMSA, and LUC transient transcriptional activity assay, and suppressed the transcription of these genes. Results: Under both long day and short day conditions, simultaneous knock-out of ABF1 and its homolog bZIP40 accelerated flowering, while SAPK8 and ABF1 over-expression lines exhibited delayed flowering and hypersensitivity to ABA-mediated flowering repression. After perceiving the ABA signal, SAPK8 physically binds to and phosphorylates ABF1 to enhance its binding to the promoters of master positive flowering regulators Ehd1 and Ehd2. Upon interacting with FIE2, ABF1 recruited PRC2 complex to deposit H3K27me3 suppressive histone modification on Ehd1 and Ehd2 to suppress these genes transcription, thereby leading to later flowering. Conclusion: Our work highlighted the biological functions of SAPK8 and ABF1 in ABA signaling, flowering control and the involvement of a PRC2-mediated epigenetic repression mechanism in the transcription regulation governed by ABF1 on ABA-mediated rice flowering repression.

Abstract Background Most researches on sponge holobionts focus primarily on symbiotic microbes, yet data at the level of the sponge hologenome are still relatively scarce. Understanding of the sponge host and its microbial gene expression profiles and the host-microbes interplay in different niches represents a key aspect of sponge hologenome. Using the Hawaiian demosponge Mycale grandis in different niches as a model, i.e. on rocks, on the surface of coral Porites compressa, under alga Gracilaria salicornia, we compared the bacterial and fungal community structure, functional gene diversity, expression pattern and the host transcriptome by integrating open-format (deep sequencing) and closed-format (GeoChip microarray) high-throughput techniques. Results Little inter-niche variation in bacterial and fungal phylogenetic diversity was detected for M. grandis in different niches, but a clear niche-dependent variability in the functional gene diversity and expression pattern of M. grandis host and its symbiotic microbiota was uncovered by GeoChip microarray and transcriptome analyses. Particularly, sponge host genes related to innate immunity and microbial recognition showed a strong correlation with the microbial symbionts’ functional gene diversity and transcriptional richness in different niches. The cross-niche variability with respect to the symbiont functional gene diversity and the transcriptional richness of M. grandis holobiont putatively reflects the interplay of niche-specific selective pressure and the symbiont functional diversity. Conclusions Niche–dependent gene expression profiles of M. grandis hologenome and the host-microbes interplay were suggested though little inter-niche variation in bacterial and fungal diversity was detected, particularly the sponge innate immunity was found to be closely related to the symbiotic microbes. Altogether, these findings provide novel insights into the black box of one sponge holobiont in different niches at the hologenome level.

Abstract Background Bilateral retinal detachment and choroidal detachment in a patient are rare occurrences. The presence of bilateral diabetic retinopathy (DR) in such a case is even rarer and complicates the condition. Case presentation In this study, we document a case of unconventional VKH. Manifestations in this patient included intense peripheral retinal detachment and choroidal detachment, along with vitreous opacities akin to cotton wool spots, concurrent with DR. The diagnosis was considered as probable VKH with DR. Treatment according to VKH protocols, including high-dose corticosteroids, yielded positive results. Conclusions VKH can co-occurrence with DR. VKH manifestations vary, and early, aggressive, and long-term treatment is essential. The complexity of treatment increases with concurrent DR, necessitating the use of immunosuppressants.

Abstract Two-dimensional single crystal metals, in which the behavior of highly confined optical modes is intertwined with quantum phenomena, are highly sought after for next-generation technologies. Here, we report large area (>104 μm2), single crystal two-dimensional gold flakes (2DGFs) with thicknesses down to a single nanometer level, employing an atomic-level precision chemical etching approach. The decrease of the thickness down to such scales leads to the quantization of the electronic states, endowing 2DGFs with quantum-confinement-augmented optical nonlinearity, particularly leading to more than two orders of magnitude enhancement in harmonic generation compared with their thick polycrystalline counterparts. The nanometer-scale thickness and single crystal quality makes 2DGFs a promising platform for realizing plasmonic nanostructures with nanoscale optical confinement. This is demonstrated by patterning 2DGFs into nanoribbon arrays, exhibiting strongly confined near infrared plasmonic resonances with high quality factors. The developed 2DGFs provide an emerging platform for nanophotonic research and open up opportunities for applications in ultrathin plasmonic, optoelectronic and quantum devices.

The natural aging process is accompanied by changes in exosomes, gut microbiota, and metabolites. This study aimed to reveal the anti-aging effect and mechanisms of fecal microbiota transplantation (FMT) from young donors on the natural aging process in mice by analyzing exosomes, gut microbiota, and metabolomics. Aging-relevant telomeric length, oxidative stress indexes in brain tissue, and serum cytokine levels were measured. Flow analysis of T-regulatory (Treg), CD4+, and CD8+ cells was performed, and the expression levels of aging-related proteins were quantified. High-throughput sequencing technology was used to identify differentially expressed serum exosomal miRNAs. Fecal microbiota was tested by 16 S rDNA sequencing. Changes in fecal metabolites were analyzed by UPLC-Q-TOF/MS. The results indicated that the expression of mmu-miR-7010–5p, mmu-miR-376b-5p, mmu-miR-135a-5p, and mmu-miR-3100–5p by serum exosomes was down-regulated and the abundance of opportunistic bacteria (Turicibacter, Allobaculum, Morganella.) was decreased, whereas the levels of protective bacteria (Akkermansia, Muribaculaceae, Helicobacter.) were increased after FMT. Metabolic analysis identified 25 potential biomarkers. Correlation analysis between the gut microbiota and metabolites suggested that the relative abundance of protective bacteria was positively correlated with the levels of spermidine and S-adenosylmethionine. The study indicated that FMT corrected brain injury due to aging via lipid metabolism, the metabolism of cofactors and vitamins, and amino acid metabolism.

The integrated carbon capture and utilization (ICCU) technology has been considered a prospective strategy for mitigating carbon emissions issues. Compared to conventional CO2 capture and utilization, the ICCU process reduces transporting, product purification, construction, and operation costs. However, few works focus on investigating the effect of in-situ CO2 conversion on the sintering of CaO at high temperatures. In this work, Cu/CaO dual functional materials (DFMs) were synthesized and used in the Calcium Looping-Reverse Water-Gas Shift (CaL-RWGS) process at 650 °C. The results showed that it is thermodynamically feasible to couple the CaL with the RWGS reaction. In the cyclic CO2 capture test, Ca1Cu0.1 DFMs showed desirable CO2 capture performance (11.34 mmol/gDFMs) and self-activated phenomenon in the first 15 cycles. Moreover, Cu nanoparticle catalysts in the DFMs effectively inhibited the sintering of CaO by accelerating the desorption of CO2 from the CaO surface and converting it to CO during the conversion stage. In-situ DRIFTS of Ca1Cu0.1 DFMs revealed that formates might be the RWGS intermediates in CaL-RWGS.

This investigation's primary objective is to analyze the fluid dynamics and heat transfer enhancement in an irregular enclosure. Several physical phenomena such as Alumina-water nanofluids, magnetic field, non-uniform heating and amplitude of non-uniform heating are taken into account. A thorough investigation using machine learning algorithms (MLAs) and computational fluid dynamics (CFD) is carried out. Initially, for CFD phase the coupled non-linear dimensionless governing equations of the considered problem are solved numerically by adapting a Finite Element method (FEM) using appropriate boundary conditions. The analysis considers a range of physical parameters, such as the Hartmann number (0–20), nanoparticle volume friction (0–0.04), Ryleigh number (103-105), offset temperature (0.1–1), frequency (1-10) and non-uniform heating amplitude (0.1–1). The numerical results are explicated in the form of streamlines, isotherms, heatlines structures. The impact of these factors on entropy generation is also explored. In addition, three popular MLAs, Random Forest (RF), Artificial Neural Network (ANN) and Support Vector Machine (SVM) were used to save the computational costs. The finding explore Hartmann number and nanoparticle volume friction has decreasing trend on total entropy production and heat transfer rate. The entropy production decreases up to 42.02 % for Hartman number and for nanoparticles it decreases 12.61 %. However, the non-uniform heating amplitude has opposite effect on irreversibility factor. Also, compared to other MLAs, the ANN model demonstrates more accurate predictions. The objective is to propose novel concept for enhancing the performance of cooling systems and optimizing energy utilization across diverse technical domain.

A novel precast segmental barrier system was proposed to facilitate the connections between barriers and between the barrier and bridge deck by casting ultra-high-performance concrete joints in situ. To assess the performance of the barrier with this connection, a full-scale pseudo-static tests were conducted. At the same time, a comparative assessment was considered the failure mode and load capacity of the precast barrier for two different bridge deck stiffnesses. The results indicated that the ultimate capacity of the precast barrier was comparable to that of a traditional cast-in-place barrier and emphasised the critical role of the deck slab stiffness in the overall load capacity. Based on the damage patterns observed in the barrier and deck slab during the pseudo-static tests, a refined trapezoidal yield-line model considering the deck slab stiffness contribution was derived to more accurately evaluate the load capacity of a precast segmental barrier. Moreover, the calibrated numerical model of the barrier system was employed to investigate the structural dynamic performance under varying scenarios, specifically examining the effects of different impact masses and velocities. Numerical results demonstrated that upon impact by a high-mass, high-velocity object, the barrier experienced a shift in damage mode from flexural–shear to more critical punching shear. This transition concentrated the dynamic load exclusively on the impacted barrier segment, which in turn markedly diminished its dynamic load capacity.

Abstract Cancer is associated with a high degree of heterogeneity, encompassing both inter- and intra-tumor heterogeneity, along with considerable variability in clinical response to common treatments across patients. Conventional models for tumor research, such as in vitro cell cultures and in vivo animal models, demonstrate significant limitations that fall short of satisfying the research requisites. Patient-derived tumor organoids, which recapitulate the structures, specific functions, molecular characteristics, genomics alterations and expression profiles of primary tumors. They have been efficaciously implemented in illness portrayal, mechanism exploration, high-throughput drug screening and assessment, discovery of innovative therapeutic targets and potential compounds, and customized treatment regimen for cancer patients. In contrast to conventional models, tumor organoids offer an intuitive, dependable, and efficient in vitro research model by conserving the phenotypic, genetic diversity, and mutational attributes of the originating tumor. Nevertheless, the organoid technology also confronts the bottlenecks and challenges, such as how to comprehensively reflect intra-tumor heterogeneity, tumor microenvironment, tumor angiogenesis, reduce research costs, and establish standardized construction processes while retaining reliability. This review extensively examines the use of tumor organoid techniques in fundamental research and precision medicine. It emphasizes the importance of patient-derived tumor organoid biobanks for drug development, screening, safety evaluation, and personalized medicine. Additionally, it evaluates the application of organoid technology as an experimental tumor model to better understand the molecular mechanisms of tumor. The intent of this review is to explicate the significance of tumor organoids in cancer research and to present new avenues for the future of tumor research.

Abstract Background Corpus callosum glioblastoma (ccGBM) is a specific type of GBM and has worse outcomes than other non-ccGBMs. We sought to identify whether en-bloc resection of ccGBMs based on T2-FLAIR imaging contributes to clinical outcomes and can achieve a satisfactory balance between maximal resection and preservation of neurological function. Methods A total of 106 adult ccGBM patients (including astrocytoma, WHO grade 4, IDH mutation, and glioblastoma) were obtained from the Department of Neurosurgery in Nanfang Hospital between January 2008 and December 2018. The clinical data, including gender, age, symptoms, location of tumor, involvement of eloquent areas, extent of resection (EOR), pre- and postoperative Karnofsky Performance Status (KPS) scales, and National Institute of Health stroke scale (NIHSS) scores were collected. Propensity score matching (PSM) analysis was applied to control the confounders for analyzing the relationship between the en-bloc technique and EOR, and the change in the postoperative KPS scales and NIHSS scores. Results Applying the en-bloc technique did not negatively affect the postoperative KPS scales compared to no-en-bloc resection (P = 0.851 for PSM analysis) but had a positive effect on preserving or improving the postoperative NIHSS scores (P = 0.004 for PSM analysis). Abstract The pandemic caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has had a profound impact on the global health and economy. While mass vaccination for herd immunity is effective, emerging SARS-CoV-2 variants can evade spike protein-based COVID-19 vaccines. In this study, we develop a new immunization strategy by utilizing a nanocarrier, dendritic mesoporous silica nanoparticle (DMSN), to deliver the receptor-binding domain (RBD) and conserved T-cell epitope peptides (DMSN-P-R), aiming to activate both humoral and cellular immune responses in the host. The synthesized DMSN had good uniformity and dispersion and showed a strong ability to load the RBD and peptide antigens, enhancing their uptake by antigen-presenting cells (APCs) and promoting antigen delivery to lymph nodes. The DMSN-P-R vaccine elicited potent humoral immunity, characterized by highly specific RBD antibodies. Neutralization tests demonstrated significant antibody-mediated neutralizing activity against live SARS-CoV-2. Crucially, the DMSN-P-R vaccine also induced robust T-cell responses that were specifically stimulated by the RBD and conserved T-cell epitope peptides of SARS-CoV-2. The DMSN demonstrated excellent biocompatibility and biosafety in vitro and in vivo, along with degradability. Our study introduces a promising vaccine strategy that utilizes nanocarriers to deliver a range of antigens, effectively enhancing both humoral and cellular immune responses to prevent virus transmission.

Abstract As a recognized special resource, tar-rich coal can extract the country's scarce oil and gas resources and generate semi-coke that can replace anthracite and coking coal. The tar-rich coal in northern Shaanxi is prominent, but due to the dense structure and high strength of tar-rich coal, it is easy to cause frequent dynamic disasters in coal mining. Therefore, the realization of pressure relief and disaster reduction has become the primary problem in mining tar-rich coal. There are many shortcomings in conventional pressure relief methods, so a new method of microwave-weakening coal is proposed. Through different water saturation treatments of tar-rich coal samples, the longitudinal wave velocity degradation trend and surface crack expansion law of water-bearing coal after microwave irradiation were analyzed, and the strength softening characterization and energy evolution relationship under the combined action of microwave and water were studied. Fractal dimension and its internal correlation based on the equivalent side length-mass of coal sample fragments. The experimental results show that: (1) Under the same microwave radiation condition, with the increase of water saturation, the deterioration trend of physical and mechanical parameters such as longitudinal wave velocity and peak strength is obvious. (2) After microwave radiation, the uniaxial compression results show that the coal sample is damaged by load, there is still a high residual strength, the ratio of elastic energy to dissipation energy decreases, and the possibility of rockburst of the coal sample decreases. The strength softening degree of coal specimen under the degradation of microwave and water is the highest, followed by microwave and water. (3) The fractal dimension is inversely proportional to the moisture content and microwave radiation intensity, and the fractal dimension has a significant positive correlation with the peak intensity and longitudinal wave velocity. The mechanical damage law of water-bearing tar-rich coal under microwave action is revealed, which aims to solve the problem of weakening and reducing the impact of hard coal on-site to a certain extent, ensure the safety of working face, and improve the mining efficiency of tar-rich coal. It provides basic theoretical support for microwave-assisted hydraulic fracturing technology and effective weakening measures for hard roof treatment.

Digital Image Correlation (DIC) has been widely used as a non-contact deformation measurement technique. Nevertheless, its accuracy is greatly affected by the speckle pattern on the specimen. To systematically evaluate how speckle deformability affects the precision of DIC algorithms. In this study, a test dataset of 2D speckle patterns with various prescribed deformation fields was numerically generated, containing two categories of speckles, i.e., the deformable and the non-deformable (rigid) ones. This dataset was used to evaluate the performance of inverse compositional Gauss-Newton (ICGN)-based DIC algorithms with two types of shape function (first-order and second-order), in the different scenarios of the deformation field. The results showed that imaging noise had a significant influence on the DIC algorithm. The first-order shape function (ICGN-1) performed better when tracking the simple linear deformation field. While the second-order shape function (ICGN-2) was proved to perform better on non-linear deformations. Moreover, the deformability of the speckle was found to have an obvious impact on the performance of the DIC algorithm. ICGN-2 could effectively reduce so-called speckle rigidity induced (SRI) error. Conclusively, ICGN-2 should be chosen as priority, because of its feasibility on non-linear deformation fields and speckle rigidity. While in the linear deformation scenarios, ICGN-1 was still a robust and efficient method.

Aim: To investigate the clinical features, diagnosis and treatment of lymphoepithelioma-like carcinoma (LELC).Materials & methods: The clinical data of 114 LELC patients were retrospectively analyzed.Results: Ninety-eight patients (86.0%) were Epstein-Barr virus-encoded small RNA (EBER) positive detected by situ hybridization. A 67.1% (51/76) patients had PD-L1 expression. The 5-year overall survival rate of EBER negative patients was 51.6% while the rate of positive patients was 84.8% (p = 0.015). The 5-year progression free survival rate of EBER negative patients was 40.2% while the rate of positive patients was 70.2% (p = 0.004).Conclusion: The progression of LELC is relatively slow and present a better prognosis. The occurrence of tumor is closely related to Epstein–Barr virus infection and PD-L1 is highly expressed in tumor cells.

Introduction: Aged-related brain damage and gut microbiome disruption are common. Research affirms that modulating the microbiota-gut-brain axis can help reduce age-related brain damage.Methods: Ginseng, esteemed in traditional Chinese medicine, is recognized for its anti-aging capabilities. However, previous Ginseng anti-aging studies have largely focused on diseased animal models. To this end, efforts were hereby made to explore the potential neuroprotective effects of fecal microbiota transplantation (FMT) from Ginseng-supplemented aged mice to those pre-treated with antibiotics.Results: As a result, FMT with specific modifications in natural aging mice improved animal weight gain, extended the telomere length, anti-oxidative stress in brain tissue, regulated the serum levels of cytokine, and balanced the proportion of Treg cells. Besides, FMT increased the abundance of beneficial bacteria of Lachnospiraceae, Dubosiella, Bacteroides, etc. and decreased the levels of potential pathogenic bacteria of Helicobacter and Lachnoclostridium in the fecal samples of natural aged mice. This revealed that FMT remarkably reshaped gut microbiome. Additionally, FMT-treated aged mice showed increased levels of metabolites of Ursolic acid, β-carotene, S-Adenosylmethionine, Spermidine, Guanosine, Celecoxib, Linoleic acid, etc., which were significantly positively correlated with critical beneficial bacteria above. Additionally, these identified critical microbiota and metabolites were mainly enriched in the pathways of Amino acid metabolism, Lipid metabolism, Nucleotide metabolism, etc. Furthermore, FMT downregulated p53/p21/Rb signaling and upregulated p16/p14, ATM/synapsin I/synaptophysin/PSD95, CREB/ERK/AKT signaling in brain damage following natural aging.Discussion: Overall, the study demonstrates that reprogramming of gut microbiota by FMT impedes brain damage in the natural aging process, possibly through the regulation of microbiota-gut-brain axis.

Introduction: Nearly vertical coal seams present a significant challenge for the coal mining industry due to their difficult accessibility. However, these seams account for a substantial proportion of the world’s coal reserves. Therefore, it is vital to conduct research on disaster control techniques for safe mining of these seams.Method: The research team used experimental research, theoretical analysis, and numerical calculation methods to investigate the creep characteristics and failure mechanisms of layered sandstone roadway in nearly vertical coal seam.Results and discussion: These findings revealed that the maximum moment and concentrated stress of the sandstone located on the side of the roadway roof was positioned in the middle of the nearly vertical structure, making it more susceptible to transverse shear failure. On the other hand, the nearly vertical structure on the floor side was prone to shear slip failure initiated from the bottom of the structure. This led to the asymmetric instability of the roadway. The practical implications of this research are significant for the safe mining of nearly vertical coal seams. The results could help inform the development of disaster control techniques.

The gene family of thaumatin-like proteins (TLPs) plays a crucial role in the adaptation of organisms to environmental stresses. In recent years, fungal secreted proteins (SP) with inducing disease resistance activity in plants have emerged as important elicitors in the control of fungal diseases. Identifying SPs with inducing disease resistance activity and studying their mechanisms are crucial for controlling sheath blight. In the present study, 10 proteins containing the thaumatin-like domain were identified in strain AG4-JY of Rhizoctonia solani and eight of the 10 proteins had signal peptides. Analysis of the TLP genes of the 10 different anastomosis groups (AGs) showed that the evolutionary relationship of the TLP gene was consistent with that between different AGs of R. solani. Furthermore, it was found that RsTLP3, RsTLP9 and RsTLP10 were regarded as secreted proteins for their signaling peptides exhibited secretory activity. Prokaryotic expression and enzyme activity analysis revealed that the three secreted proteins possess glycoside hydrolase activity, suggesting they belong to the TLP family. Additionally, spraying the crude enzyme solution of the three TLP proteins could enhance maize resistance to sheath blight. Further analysis showed that genes associated with the salicylic acid and ethylene pathways were up-regulated following RsTLP3 application. The results indicated that RsTLP3 had a good application prospect in biological control.

Accurate extraction of aggregates, cement bridges, and voids in permeable cement-stabilized base materials (PCBM) is a challenging task. This paper proposed a segmentation and reconstruction paradigm for raw CT slice images using a mask regional convolutional neural network (Mask R-CNN). The results demonstrate that the proposed method effectively segments and reconstructs unclear raw CT slice images, providing a convenient and advanced tool for accurately assessing the meso-scale structures of cement bridges and voids in PCBM prepared with different combinations of cement content and compaction force. An increased cement content or compaction force results in the migration of cement paste, leading to thicker localized cement bridges and blockage of permeable pore space. This phenomenon further disrupts the homogeneous distribution of cement bridges and voids. An increased compaction force tends to elongate and flatten pore throats, while an increased cement content may clog voids, rendering them impermeable. The vertical permeability of the PCBM is significantly greater than the horizontal permeability, which should be considered during material design optimization. The proposed method could serve as a reference for microscopic analysis and structural design optimization of PCBM.

Abstract Mixing is not much used in the high-frequency literature so far. However, mixing is a common weakly dependent property of continuous and discrete stochastic processes, such as Gaussian, Ornstein–Uhlenberck (OU), Vasicek, CIR, CKLS, logistic diffusion, generalized logistic diffusion, and double-well diffusion processes. So, long-span high-frequency data typically have weak dependence, and using mixing to study them is also an alternative approach. In this paper, we give some moment inequalities for long-span high-frequency data with ϕ-mixing, ρ-mixing, and α-mixing. These inequalities are effective tools for studying asymptotic properties. Applying these inequalities, we investigate the strong consistency of parameter estimation for the OU-integrated diffusion process. We also derive the mean square error of the estimation of the OU process and the optimal interval for the drift parameter estimator.

Mastering fracture and movement of far-field high-position hard-and-thick strata plays an important role in safety prevention and control of disasters including severe surface subsidence and strong mine seismic events. The fracture mechanical model of high-position hard-and-hick strata is established based on the panels 2201 and 2202 in Yingpanhao Coal Mine. The fracture and movement of the Cretaceous sandstone formation and its influencing factors are analyzed by using the theory of Vlasov thick plate, which is verified and analyzed by surface subsidence and microseismic events. The conclusions are as follows: ①In view of the occurrence characteristics of Cretaceous sandstone group that is far away from the coal seam, with large thickness and good integrity, a high-position hard-and-hick strata fracture mechanical model is established. And the critical mechanical conditions for the first fracture of high-position thick-and-hard rock stratum are calculated by Vlasov thick plate theory. ② With the increase of tensile strength, the fracture overhang length of hard-and-hick stratum slowly grow, linearly grow, and exponentially grow; As the thickness of the stratum increases, the fracture overhang length of the thick and hard rock stratum increases exponentially; With the increase of the overhang length along the dip (width of panel), the fracture overhang length of thick and hard rock stratum increases first and then decreases. ③ During the extraction of panel 2201 in Yingpanhao Coal Mine, the hard-and-hick stratum did not break; And when the panel 2202 advanced about 960 m, the vertical "O-X" fracture occured; When the panel continues to advance about 188 m, the hard-and-hick stratum breaks periodically. ④ The monitoring results of surface subsidence show that the surface subsidence value of panel 2201 is small as a whole, and there are many small energy micro-seismic events. The surface subsidence of panel 2202 has experienced slow subsidence, rapid subsidence and stable stages in turn, and the frequency of high-energy mine seismic event has increased significantly. The location of the high energy mine seismic events and the area of the maximum surface subsidence are highly coincident, both in the O-X break area; The surface subsidence velocity reached the maximum of the 9.87 mm/d at 960 m away from the setup room, inducing the “6·8” strong mine seismic event. The above phenomenons verified the correctness of the far-field thick and hard rock stratum fracturing behavior. The research results of this paper have guiding significance for the study on the fracture of thick and hard rock stratum and disaster pre-control under similar engineering geological conditions.

Abstract Background Temozolomide (TMZ) treatment efficacy in glioblastoma (GBM) patients has been limited by resistance in the clinic. Currently, there are no clinically proven therapeutic options available to restore TMZ treatment sensitivity. Here, we investigated the potential of albumin-bound paclitaxel (ABX), a novel microtubule targeting agent, in sensitizing GBM cells to TMZ and elucidated its underlying molecular mechanism. Methods A series of in vivo and in vitro experiments based on two GBM cell lines and two primary GBM cells were designed to evaluate the efficacy of ABX in sensitizing GBM cells to TMZ. Further proteomic analysis and validation experiments were performed to explore the underlying molecular mechanism. Finally, the efficacy and mechanism were validated in GBM patients derived organoids (PDOs) models. Results ABX exhibited a synergistic inhibitory effect on GBM cells when combined with TMZ in vitro. Combination treatment of TMZ and ABX was highly effective in suppressing GBM progression and significantly prolonged the survival oforthotopic xenograft nude mice, with negligible side effects. Further proteomic analysis and experimental validation demonstrated that the combined treatment of ABX and TMZ can induce sustained DNA damage by disrupting XPC and ERCC1 expression and nuclear localization. Additionally, the combination treatment can enhance ferroptosis through regulating HOXM1 and GPX4 expression. Preclinical drug-sensitivity testing based on GBM PDOs models confirmed that combination therapy was significantly more effective than conventional TMZ monotherapy. Conclusion Our findings suggest that ABX has the potential to enhance TMZ treatment sensitivity in GBM, which provides a promising therapeutic strategy for GBM patients.

Abstract Background Genitourinary small cell carcinoma is rare, and has a poor prognosis. However, effective treatment options for this disease are limited. We present a study to assess the efficacy of chemotherapy alone or combined with immunotherapy for locally advanced or metastatic genitourinary small cell carcinoma (GSCC). Methods We performed a retrospective analysis of patients with locally advanced or metastatic GSCC from Jan 2013 to September 2022 at Sun Yat-sen University Cancer Center. The survival and safety profiles were analyzed. Results Forty-two GSCC patients were enrolled, which included 20 with chemotherapy plus immunotherapy and 22 with chemotherapy alone. The median follow-up time was 15.13 months (95% CI, 8.84–21.42). The addition of immunotherapy to chemotherapy demonstrated no significant difference in median progression-free survival (p = 0.37). However, the median overall survival (OS) was 22.97 and 14.03 months with immunotherapy plus chemotherapy and chemotherapy alone, respectively (HR = 0.69, 95%CI 0.08–0.55, p = 0.017). Two patients with immunotherapy plus chemotherapy achieved clinical complete remission. The overall response rate for patients receiving chemotherapy combined with immunotherapy was 65%, which was higher in comparison to those treated with chemotherapy alone (50%). Univariate and multivariate analyses demonstrated that chemotherapy combined with immunotherapy independently achieved favorable OS. Four patients experienced immunotherapy-related adverse events, with one developing grade 3 hypothyroidism. Conclusions Among patients with locally advanced or metastatic GSCC, immunotherapy combined with chemotherapy might be thought of as a potentially effective treatment option for patients with GSCC.

Abstract When conducting relay protection research, research costs can be significantly reduced if protection principle development, protection parameter verification and debugging can be carried out without relying on actual protection devices. The concept of ‘digital twin’ has made this possible, but the existing research has shortcomings in real-time data interaction ability, protection logic transparency, interface standardization, human–computer interaction etc., and consequently, mirror operation of relay protection in digital space has not been fully realized. Therefore, referring to the characteristics of digital twin, and combining with the practical application requirements in relay protection, this paper proposes the concept and characteristics of relay protection mirror operation based on digital twin. Key solutions are proposed to address the difficulties that may be encountered in the implementation of relay protection mirror operation in terms of protection principles, interfaces, real-time operation of the system, and human–computer interaction function simulation. Finally, an example of 110 kV double-bus and double-branch bus protection is used to verify the feasibility and progressiveness of the scheme proposed in this paper by comparing the action behavior and external characteristics of the twin protection and the actual protection device. The presented research can provide a reference for further in-depth research and application of relay protection using digital means.

The challenge of effectively managing the formation and recovery of traffic cone robots (TCRs) is addressed by proposing a linear time-varying model predictive control (MPC) strategy. This problem involves coordinating multiple TCR formations within a work area to reach a target location, which is a huge challenge due to the complexity of dynamic coordination. Unlike conventional approaches, our method decomposes the formation control problem into two main components: leader TCR motion planning and follower formation tracking control. The motion planning component involves path and velocity planning to achieve leader trajectory control, which serves as a reference trajectory for the follower. The formation tracking task extends to formation control among multiple robots to achieve the traffic cone robot formation placement and recovery task. To address the TCR input limitation problem, input constraints are considered during the design process of the MPC controllers. The effectiveness and practicality of the proposed control strategy are validated through a series of numerical simulations and physical experiments with TCRs.

Utilizing deep learning for semantic segmentation of cropland from remote sensing imagery has become a crucial technique in land surveys. Cropland is highly heterogeneous and fragmented, and existing methods often suffer from inaccurate boundary segmentation. This paper introduces a UNet-like boundary-aware compensation model (BAFormer). Cropland boundaries typically exhibit rapid transformations in pixel values and texture features, often appearing as high-frequency features in remote sensing images. To enhance the recognition of these high-frequency features as represented by cropland boundaries, the proposed BAFormer integrates a Feature Adaptive Mixer (FAM) and develops a Depthwise Large Kernel Multi-Layer Perceptron model (DWLK-MLP) to enrich the global and local cropland boundaries features separately. Specifically, FAM enhances the boundary-aware method by adaptively acquiring high-frequency features through convolution and self-attention advantages, while DWLK-MLP further supplements boundary position information using a large receptive field. The efficacy of BAFormer has been evaluated on datasets including Vaihingen, Potsdam, LoveDA, and Mapcup. It demonstrates high performance, achieving mIoU scores of 84.5%, 87.3%, 53.5%, and 83.1% on these datasets, respectively. Notably, BAFormer-T (lightweight model) surpasses other lightweight models on the Vaihingen dataset with scores of 91.3% F1 and 84.1% mIoU.

Visible near-infrared spectroscopy (VNIR) is extensively researched for obtaining soil property information due to its rapid, cost-effective, and environmentally friendly advantages. Despite its widespread application and significant achievements in soil property analysis, current soil prediction models continue to suffer from low accuracy. To address this issue, we propose a convolutional neural network model that can achieve high-precision soil property prediction by creating 2D multi-channel inputs and applying a multi-scale spatial attention mechanism. Initially, we explored two-dimensional multi-channel inputs for seven soil properties in the public LUCAS spectral dataset using the Gramian Angular Field (GAF) method and various preprocessing techniques. Subsequently, we developed a convolutional neural network model with a multi-scale spatial attention mechanism to improve the network’s extraction of relevant spatial contextual information. Our proposed model showed superior performance in a statistical comparison with current state-of-the-art techniques. The RMSE (R²) values for various soil properties were as follows: organic carbon content (OC) of 19.083 (0.955), calcium carbonate content (CaCO3) of 24.901 (0.961), nitrogen content (N) of 0.969 (0.933), cation exchange capacity (CEC) of 6.52 (0.803), pH in H2O of 0.366 (0.927), clay content of 4.845 (0.86), and sand content of 12.069 (0.789). Our proposed model can effectively extract features from visible near-infrared spectroscopy data, contributing to the precise detection of soil properties.

The accumulation of years imbues historical buildings within universities with a profound sense of heritage, evident not only in the temporal imprints within their internal spaces but also in the evolution of their external surroundings. This cultural legacy subtly enriches students’ spatial awareness of history and fosters a collective memory of campus context. Current scholarly inquiry into university historical buildings primarily revolves around comprehensive considerations encompassing the preservation of these edifices, the overarching planning of academic institutions, and the safeguarding of the distinctive features inherent to historical structures. However, the predominant focus lies on qualitative analyses, leaving a pressing need for quantitative assessments and the establishment of an evaluation framework to gauge the efficacy of historical building preservation in academia. Addressing this gap, this study employs the Analytic Hierarchy Process (AHP) and Fuzzy Comprehensive Evaluation (FCE) to formulate the University Historical Building Protection Evaluation Framework (UHBPEF). Drawing from the examination and categorization of the primary instructional edifices within the Yujiatou campus of Wuhan University of Technology through the Research Classification Method (RCM), along with the consolidation of insights from experts and academic stakeholders, this study underscores the imperative of enhancing the scientific precision and pertinence of the university’s strategy for preserving historical buildings. By employing both qualitative and quantitative methodologies, this study offers innovative insights into the challenges facing historical building preservation in university settings, therefore propounding effective preservation strategies and offering a roadmap for future endeavors in this domain.

ABSTRACT Global warming is a key issue that causes coral bleaching mainly because of the thermosensitivity of zooxanthellae. Compared with the well-studied zooxanthellae Symbiodiniaceae in coral holobionts, we rarely know about other coral symbiotic algae, let alone their thermal tolerance. In this study, a zoochlorellae, Symbiochlorum hainanensis, isolated from the coral Porites lutea, was proven to have a threshold temperature of 38°C. Meanwhile, unique high-temperature tolerance mechanisms were suggested by integrated transcriptomics and real-time quantitative PCR, physiological and biochemical analyses, and electron microscopy observation. Under heat stress, S. hainanensis shared some similar response strategies with zooxanthellae Effrenium sp., such as increased ascorbate peroxidase, glutathione peroxidase, superoxide dismutase activities and chlorophyll a, thiamine, and thiamine phosphate contents. In particular, more chloroplast internal layered structure, increased CAT activity, enhanced selenate reduction, and thylakoid assembly pathways were highlighted for S. hainanensis’s high-temperature tolerance. Notably, it is the first time to reveal a whole selenate reduction pathway from SeO42− to Se2− and its contribution to the high-temperature tolerance of S. hainanensis. These unique mechanisms, including antioxidation and maintaining photosynthesis homeostasis, efficiently ensure the high-temperature tolerance of S. hainanensis than Effrenium sp. Compared with the thermosensitivity of coral symbiotic zooxanthellae Symbiodiniaceae, this study provides novel insights into the high-temperature tolerance mechanisms of coral symbiotic zoochlorellae S. hainanensis, which will contribute to corals’ survival in the warming oceans caused by global climate change.IMPORTANCEThe increasing ocean temperature above 31°C–32°C might trigger a breakdown of the coral-Symbiodiniaceae symbioses or coral bleaching because of the thermosensitivity of Symbiodiniaceae; therefore, the exploration of alternative coral symbiotic algae with high-temperature tolerance is important for the corals’ protection under warming oceans. This study proves that zoochlorellae Symbiochlorum hainanensis can tolerate 38°C, which is the highest temperature tolerance known for coral symbiotic algae to date, with unique high-temperature tolerance mechanisms. Particularly, for the first time, an internal selenium antioxidant mechanism of coral symbiotic S. hainanensis to high temperature was suggested.

Nowadays, substantial research efforts are being devoted to technologies which could potentially increase propulsion efficiency and reduce emissions of aircrafts. For this purpose, a waste heat recovery (WHR) combined system based on recuperator and organic Rankine cycle (ORC) is proposed in this paper for rotorcraft powerplant application. The concept is systematically analyzed within an integrated multidisciplinary simulation framework targeting the implicit coupling between rotorcraft-engine system and WHR module under designated flight conditions, also taking the additional weight penalty into consideration. Obtained results through quantification of the potential benefits suggest a considerable improvement in engine fuel economy with the adoption of highly effective recuperator, and ORC can significantly increase the power output, representing more than 24% of the engine power with benzene as working fluid. Reducing the power-to-weight ratio of ORC from 4 to 0.5 kW/kg, the added weight of WHR unit grows exponentially from 80 to 450 kg, which heavily penalizes the valuable carrying capability of the rotorcraft and consequently results in an increase of approximately 3.35%–4.2% in fuel consumption for the designated cruise mode of the generic mission scenario. The overall methodology can be effectively deployed to assess the implementation of WHR technologies for rotorcraft powerplant applications under realistic flight operations.

Abstract The desert ecosystem of the Qinghai–Tibet Plateau (QTP) is an important component of China's desert ecosystem. Studying the mechanisms shaping the taxonomic, phylogenetic, and functional beta diversity of plant communities in the QTP desert will help us to promote scientific conservation and management of the region's biodiversity. This study investigated the effects of environmental (including altitude, climate factors, and soil factors) and geographic distances on three facets of beta diversity as well as their turnover and nestedness components based on field survey data. The results showed that turnover components dominate the three facets of beta diversity. However, the turnover contributions to phylogenetic and functional beta diversity were lower than for taxonomic beta diversity. Environmental distance had a greater influence than geographic distance, with the former uniquely explaining 15.2%–22.8% of beta diversity and the latter explaining only 1.7%–2.4%. Additionally, the explanatory power of different factors for beta diversity differed between herbs and shrubs, with environmental distance being more important for the latter. Distance‐based redundancy analysis suggested that soil total potassium content had a substantial impact on the beta diversity of three dimensions, with mean temperature of the coldest month and soil total phosphorus content having a substantial impact on taxonomic and functional beta diversity as well. Our results support that environmental sorting plays a predominant role in shaping plant community composition across QTP desert ecosystems. To maintain the plant diversity of this region, it is crucial to prioritize the conservation of its diverse environmental conditions and actively mitigate its degradation by anthropogenic pressures.

Livestock presence impacts plant biodiversity (species richness) in grassland ecosystems, yet extent and direction of grazing impacts on biodiversity vary greatly across inter-annual periods. In this study, an 8-year (2014-2021) grazing gradient experiment with sheep was conducted in a semi-arid grassland to investigate the impact of grazing under different precipitation variability on biodiversity. The results suggest no direct impact of grazing on species richness in semi-arid Stipa grassland. However, increased grazing indirectly enhanced species richness by elevating community dominance (increasing the sheltering effect of Stipa grass). Importantly, intensified grazing also regulates excessive community biomass resulting from increased inter-annual wetness (SPEI), amplifying the positive influence of annual humidity index on species richness. Lastly, we emphasize that, in water-constrained grassland ecosystems, intra-annual precipitation variability (PCI) was the most crucial factor driving species richness. Therefore, the water-heat synchrony during the growing season may alleviate physiological constraints on plants, significantly enhancing species richness as a result of multifactorial interactions. Our study provides strong evidence for how to regulate grazing intensity to increase biodiversity under future variable climate patterns. We suggest adapting grazing intensity according to local climate variability to achieve grassland biodiversity conservation.

This article employs an artificial neural network technique to approximate the solution for the stagnation point flow with velocity and thermal slip effects, as well as radiative heat transfer. The PDE governing system is transformed into a set of coupled ordinary differential systems by incorporating similarity variables. The shooting method is used to obtain a dataset in Mathematica. To test the precision of the suggested model, the operations of training, testing, and validation are performed, and the results are compared to a reference dataset. The Levenberg-Marquardt backpropagation neural network model is utilized to solve the system of equations under different scenarios, and its output is evaluated using mean square error, state transition dynamics, error histograms analysis, and regression illustrations. The findings indicate that the neural network approach achieves a high level of accuracy when predicting thermal analysis. Moreover, the current Artificial Neural Network model has an advantage over other numerical techniques in that it can handle more intricate mathematical models while minimizing the resources required for problem-solving.

Learners' anxiety and emotional engagement have attracted a surge of scholarly interest due to their considerable relevance to students' academic performance and psychological well-being. However, the two variables await more exploration in the English as a Foreign Language (EFL) context. To fill this gap, we aimed to investigate these two variables in online and offline English-speaking classes, respectively, at a university in China. We adopted a sequential mixed-methods research approach. First, we used questionnaires to measure and compare the level of anxiety and emotional engagement in the two classes. Then, we conducted semi-structured interviews to gain a deeper understanding of the factors affecting learners' online and offline learning experiences. Quantitative results showed that students had higher levels of anxiety and lower levels of emotional engagement in the online environment than in the offline context. The subsequent qualitative findings revealed a complexity of positive and negative factors affecting anxiety and emotional engagement in the two instructional contexts. Our findings can shed light on research and practices concerning learner emotions in online and offline EFL instruction. Strategies were also provided to help teachers mitigate learners' anxiety and enhance their emotional engagement, which includes formulating a learning community, providing emotional support, alleviating students' technical stress and distraction, and incorporating group-based interactive activities into the course.

Abstract Background Gene expression signatures can be used as prognostic biomarkers in various types of cancers. We aim to develop a gene signature for predicting the response to radiotherapy in glioma patients. Methods Radio-sensitive and radio-resistant glioma cell lines (M059J and M059K) were subjected to microarray analysis to screen for differentially expressed mRNAs. Additionally, we obtained 169 glioblastomas (GBM) samples and 5 normal samples from The Cancer Genome Atlas (TCGA) database, as well as 80 GBM samples and 4 normal samples from the GSE7696 set. The “DESeq2” R package was employed to identify differentially expressed genes (DEGs) between the normal brain samples and GBM samples. Combining the prognostic-related molecules identified from the TCGA, we developed a radiosensitivity-related prognostic risk signature (RRPRS) in the training set, which includes 152 patients with glioblastoma. Subsequently, we validated the reliability of the RRPRS in a validation set containing 616 patients with glioma from the TCGA database, as well as an internal validation set consisting of 31 glioblastoma patients from the Nanfang Hospital, Southern Medical University. Results Based on the microarray and LASSO COX regression analysis, we developed a nine-gene radiosensitivity-related prognostic risk signature. Patients with glioma were divided into high- or low-risk groups based on the median risk score. The Kaplan–Meier survival analysis showed that the progression-free survival (PFS) of the high-risk group was significantly shorter. The signature accurately predicted PFS as assessed by time-dependent receiver operating characteristic curve (ROC) analyses. Stratified analysis demonstrated that the signature is specific to predict the outcome of patients who were treated using radiotherapy. Univariate and multivariate Cox regression analysis revealed that the predictor was an independent predictor for the prognosis of patients with glioma. The prognostic nomograms accompanied by calibration curves displayed the 1-, 2-, and 3-year PFS and OS in patients with glioma. Conclusion Our study established a new nine-gene radiosensitivity-related prognostic risk signature that can predict the prognosis of patients with glioma who received radiotherapy. The nomogram showed great potential to predict the prognosis of patients with glioma treated using radiotherapy.

Abstract The induction of type-I interferons (IFN-Is) is important for the efficacy of chemotherapy. By investigating the role of amino acids in regulation of IFN-I production under chemo-drug treatment in bladder cancer (BC) cells, we find an inherent AhR-dependent negative feedback to restrain STING signaling and IFN-I production. Mechanistically, in a ligand dependent manner, AhR bridges STING and CUL4B/RBX1 E3 ligase complex, facilitating STING degradation through ubiquitin-proteasome pathway. Inhibition of AhR increases STING levels and reduces tumor growth under cisplatin or STING agonist treatment. Endogenous AhR ligands are mainly consisted of tryptophan (Trp) metabolites; dietary Trp restriction, blocking the key Trp metabolism rate-limiting enzyme IDO1 or inhibition of cellular Trp importation also show similar effect as AhR inhibition. Clinically, BC patients with higher intratumoral expression of AhR or stronger intratumoral Trp metabolism (higher IDO1 or Kyn levels) that lead to higher AhR activation show worse response rate to neoadjuvant chemotherapy (NAC).

Background: The survival benefit of partial nephrectomy (PN) in pT3a RCC patients is controversial. Here we aimed to explore the potential benefit of PN for pT3aN0M0 renal cell carcinoma (RCC). Material and methods: Data of patients with pT3aN0M0 RCC who were diagnosed between 2010 and 2012 in the National Cancer Institute Surveillance, Epidemiology, and End Results (SEER) database were retrospectively collected. Overall survival (OS) and cancer specific survival (CSS) were compared using a Cox proportional hazards model between PN and radical nephrectomy (RN) in pT3aN0M0 RCC. Propensity score (–adjusted, –stratified, –weighted, and –matched) analyses were performed to control for imbalances in individual risk factors. Results: A total of 1277 patients with pT3aN0M0 RCC were identified, of whom 200 patients were treated with PN and 1077 patients were RN. PN showed favorable OS and CSS in 0–4 cm pT3aN0M0 RCC (P

Disulfide zirconium (ZrS2) is a two-dimensional (2D) transition metal disulfide and has given rise to extensive attention because of its distinctive electronic structure and properties. However, mass production of high quality of ZrS2 nanosheets to realize their practical application remains a challenge. Here, we have successfully exfoliated the bulk ZrS2 powder with the thickness of micron into single and few-layer nanosheets through liquid-phase exfoliation in N-methylpyrrolidone (NMP) assisted via aliphatic amines as intercalators. It is found that the exfoliation yield is as high as 27.3%, which is the record value for the exfoliation of ZrS2 nanosheets from bulk ZrS2 powder, and 77.1% of ZrS2 nanosheets are 2–3 layers. The molecular geometric size and aliphatic amine basicity have important impact on the exfoliation. Furthermore, the ZrS2 nanosheets have been used as catalyst in the electrocatalytic dinitrogen reduction with the NH3 yield of 57.75 μg h−1 mgcat.−1, which is twice that by ZrS2 nanofibers reported in literature and three times that by the bulk ZrS2 powder. Therefore, the liquid phase exfoliation strategy reported here has great potential in mass production of ZrS2 nanosheets for high activity electrocatalysis.

Abstract Background Oral squamous cell carcinoma (OSCC) is a disease with increasing incidence worldwide that leads to deformity and death. In OSCC, fascin actin-bundling protein 1 (FSCN1) is an oncogene involved in the tumorigenesis process. However, the functions and potential mechanisms of FSCN1 in the OSCC tumorigenesis process have not been reported thus far. Methods We used qRT‒PCR to detect the expression of FSCN1 in 40 paired OSCC tumor tissues (tumor) and neighboring noncancerous tissues. The role of FSCN1 was also assessed in vitro through colony formation, CCK-8, and transwell assays. Moreover, glucose consumption was detected. Western blotting was used to confirm the interaction of FSCN1, IRF4 and AKT. Results FSCN1 was remarkably overexpressed in OSCC tissues and cell lines compared to corresponding controls. In addition, colony formation, CCK-8, and transwell assays revealed a notable reduction in OSCC growth and invasion when FSCN1 was silenced. FSCN1 silencing remarkably suppressed OSCC glycolysis. Mechanistic studies showed that FSCN1 achieves its function partially by activating interferon regulatory factor 4 (IRF4) and the AKT pathway in OSCC. Conclusion In conclusion, our study investigated the functions and mechanisms of the FSCN1/IRF4/AKT pathway in OSCC progression. In OSCC, FSCN1 is likely to be a biomarker and therapeutic target.

Abstract Background Cognitive behavior therapy (CBT) has been applied in intervention research in diabetes patients with satisfying results. However, there was no research on type 2 diabetes (T2DM) patients with comorbidities. This study aimed to investigate the effectiveness of CBT on psychological variables, behavior variables, quality of life, sleep quality, and physical variables among adult T2DM patients with comorbid metabolic syndrome (MS). Methods 281 patients aged 18–75 years were recruited from Ningbo First Hospital in China from October 2021 to March 2022. Patients were randomized to the intervention group (IG, N = 148) or control group (CG, N = 133). Patients in the IG received 12 CBT-based sessions during a six-month intervention time. Patients in the CG received the usual care only. Univariate General Linear Model was used to analyze the effect of CBT-based interventions. The analysis was conducted by SPSS Version 28. Results Results indicated that CBT-based intervention was superior in the following aspects: relieving depression symptoms: IG (4.11 ± 4.35 vs. 1.99 ± 2.12), CG (3.40 ± 3.26 vs. 2.32 ± 1.88), interaction effect (F = 4.074, P = 0.044); enhancing diabetes self-care behaviors: IG (26.79 ± 12.18 vs. 37.49 ± 10.83), CG (25.82 ± 13.71 vs. 31.96 ± 11.72), interaction effect (F = 5.242, P = 0.022); promoting the efficacy of CBT: IG (47.45 ± 6.83 vs. 50.76 ± 4.98), CG (46.74 ± 6.94 vs. 47.87 ± 5.11), interaction effect (F = 5.198, P = 0.023); improving subjective sleep quality: IG (0.93 ± 0.68 vs. 0.69 ± 0.63), CG (1.03 ± 0.72 vs. 1.01 ± 0.68), interaction effect (F = 3.927, P = 0.048). Conclusions The CBT-based intervention was beneficial in improving depression symptoms, diabetes self-care behaviors, the efficacy of CBT, and sleep quality in T2DM patients with comorbid MS. The downtrend of body mass index, systolic blood pressure, diastolic pressure, and glycated hemoglobin was larger in the intervention group but not to a significant level. The idling of automobiles at street intersections can lead to pollutant accumulation which impacts the health of residents in street-facing buildings. Previous research focused on pollutant dispersion within street canyons and did not consider the coupling of indoor and outdoor pollutants. This paper employs the computational fluid dynamics (CFD) method to simulate the dispersion characteristics of vehicle emission pollutants in street canyons, primarily investigating the indoor and outdoor pollutant dispersion patterns under various window opening configurations (single-sided ventilation, corner ventilation, and different positions of the glass under corner ventilation). Additionally, the study considers the impacts of the aspect ratio and ambient wind speed. Studies have shown that corner ventilation is effective in reducing indoor pollutant levels. When the two window glass positions are far away from the center of the intersection, the average CO mass fraction in the single-sided ventilation room is reduced by 87.1%. The average indoor CO mass fraction on the leeward side decreases with the increasing wind speed and aspect ratio. At a wind speed of 8 m/s, the average indoor CO mass fraction on the leeward side decreases to 2.45 × 10−8. At an aspect ratio of 2, the indoor CO mass fraction on the leeward side decreases with increasing floors before stabilizing at approximately 4.77 × 10−9. This study suggests optimal window opening strategies to reduce indoor pollutant levels in street-facing buildings at street intersections, offering guidance to indoor residents on window ventilation practices.
