Despite the great potential of anti-PD-L1 antibodies for immunotherapy, their low response rate due to an immunosuppressive tumor microenvironment has hampered their application. To address this issue, we constructed a cell membrane-coated nanosystem (mB4S) to reverse an immunosuppressive microenvironment to an immuno-supportive one for strengthening the anti-tumor effect. In this system, Epirubicin (EPI) as an immunogenic cell death (ICD) inducer was coupled to a branched glycopolymer via hydrazone bonds and diABZI as a stimulator of interferon genes (STING) agonist was encapsulated into mB4S. After internalization of mB4S, EPI was acidic-responsively released to induce ICD, which was characterized by an increased level of calreticulin (CRT) exposure and enhanced ATP secretion. Meanwhile, diABZI effectively activated the STING pathway. Treatment with mB4S in combination with an anti-PD-L1 antibody elicited potent immune responses by increasing the ratio of matured dendritic cells (DCs) and CD8+ T cells, promoting cytokines secretion, up-regulating M1-like tumor-associated macrophages (TAMs) and down-regulating immunosuppressive myeloid-derived suppressor cells (MDSCs). Therefore, this nanosystem for co-delivery of an ICD inducer and a STING agonist achieved promotion of DCs maturation and CD8+ T cells infiltration, creating an immuno-supportive microenvironment, thus potentiating the therapy effect of the anti-PD-L1 antibody in both 4T1 breast and CT26 colon tumor mice.

Abstract DNA double-strand breaks (DSBs) are repaired by a hierarchically regulated network of pathways. Factors influencing the choice of particular repair pathways, however remain poorly characterized. Here we develop an Integrated Classification Pipeline (ICP) to decompose and categorize CRISPR/Cas9 generated mutations on genomic target sites in complex multicellular insects. The ICP outputs graphic rank ordered classifications of mutant alleles to visualize discriminating DSB repair fingerprints generated from different target sites and alternative inheritance patterns of CRISPR components. We uncover highly reproducible lineage-specific mutation fingerprints in individual organisms and a developmental progression wherein Microhomology-Mediated End-Joining (MMEJ) or Insertion events predominate during early rapid mitotic cell cycles, switching to distinct subsets of Non-Homologous End-Joining (NHEJ) alleles, and then to Homology-Directed Repair (HDR)-based gene conversion. These repair signatures enable marker-free tracking of specific mutations in dynamic populations, including NHEJ and HDR events within the same samples, for in-depth analysis of diverse gene editing events.

Abstract Antibiotics are indispensable to infection management. However, use of antibiotics can cause gut microbiota dysbiosis, which has been linked to cognitive impairment by disrupting communication between the gut microbiota and the brain. We conducted a systematic review and meta-analysis on the effects of long-term antibiotic use on cognitive outcomes. We have searched PubMed, Web of Science, Embase, Cochrane Library and Scopus for English publications before March 2023 following the PRISMA guidelines. Screening, data extraction, and quality assessment were performed in duplicate. 960 articles were screened and 16 studies which evaluated the effect of any antibiotic compared to no antibiotics or placebo were included. Case-reports, in vitro and animal studies were excluded. We found that antibiotic use was associated with worse cognitive outcomes with a pooled effect estimate of − 0.11 (95% CI − 0.15, − 0.07, Z = 5.45; P

Brain metastasis is the leading cause of death in most cancer patients; thus, anti‐brain metastasis is a crucial step in cancer treatment. The unique microenvironment and pathophysiological characteristics of brain metastases hamper the development of effective treatment methods, while the advances in nanomedicine demonstrate its immense potential for addressing this challenge. In recent years, due to breakthroughs in nanotechnology, functional and complex nanomedicine is prepared and it has been extensively explored to improve the treatment of brain metastasis. Compared to traditional treatment methods, nanomedicine can combine multiple traditional treatment methods into one single medicinal format and target unique characteristics of brain metastases to enhance the treatment efficacy. This review summarizes the main obstacles in the field of brain metastasis treatment and clinical/preclinical studies and surveys the use of a variety of nanomedicine candidates as antibrain metastasis strategies for different types of brain metastases.

Abstract Combined chemotherapy and targeted therapy holds immense potential in the management of advanced gastric cancer (GC). GC tissues exhibit an elevated expression level of protein kinase B (AKT), which contributes to disease progression and poor chemotherapeutic responsiveness. Inhibition of AKT expression through an AKT inhibitor, capivasertib (CAP), to enhance cytotoxicity of paclitaxel (PTX) toward GC cells is demonstrated in this study. A cathepsin B‐responsive polymeric nanoparticle prodrug system is employed for co‐delivery of PTX and CAP, resulting in a polymeric nano‐drug BPGP@CAP. The release of PTX and CAP is triggered in an environment with overexpressed cathepsin B upon lysosomal uptake of BPGP@CAP. A synergistic therapeutic effect of PTX and CAP on killing GC cells is confirmed by in vitro and in vivo experiments. Mechanistic investigations suggested that CAP may inhibit AKT expression, leading to suppression of the phosphoinositide 3‐kinase (PI3K)/AKT signaling pathway. Encouragingly, CAP can synergize with PTX to exert potent antitumor effects against GC after they are co‐delivered via a polymeric drug delivery system, and this delivery system helped reduce their toxic side effects, which provides an effective therapeutic strategy for treating GC.

The precise wake model is crucial for accurately estimating wind farm loads and power, playing a key role in wake control within wind farms. This study proposes a segmented dual-Gaussian wake model, which is built upon existing dual-Gaussian wake models but places greater emphasis on the influence of initial wake generation and evolution processes on the wind speed profile in the near-wake region. The enhanced model optimizes the wake speed profile in the near-wake region and improves the accuracy of wake diffusion throughout the entire flow field. Furthermore, the optimized dual-Gaussian wake model is utilized to estimate the power output and blade root vibration loads in offshore wind farms. Through comparative analysis of high-fidelity simulation results and actual measurement data, the accuracy of the optimized dual-Gaussian wake model is validated. This approach offers high computational efficiency and provides valuable insights for load fluctuations and power estimation, thereby advancing the development of wake control strategies rapidly.

To improve the response rate of immune checkpoint inhibitors such as anti-PD-L1 antibody in immunosuppressive cancers like triple-negative breast cancer (TNBC), induction of immunogenic cell death (ICD) at tumor sites can increase the antigenicity and adjuvanticity to activate the immune microenvironment so that tumors become sensitive to the intervention of immune checkpoint inhibitors. Herein, a self-amplified biomimetic nanosystem, mEHGZ, was constructed by encapsulation of epirubicin (EPI), glucose oxidase (Gox) and hemin in ZIF-8 nanoparticles and coating of the nanoparticles with calreticulin (CRT) over-expressed tumor cell membrane. EPI acts as an ICD inducer, Gox and hemin medicate the cascade generation of reactive oxygen species (ROS) to strengthen the ICD effect, and CRT-rich membrane as “eat me” signal promote presentation of the released antigens by dendritic cells (DCs) to invoke the tumor-immunity cycle. The biomimetic delivery system displays an amplified ICD effect via Gox oxidation, hydroxyl radical generation and glutathione (GSH) depletion. The induced potent ICD effect promotes DCs maturation and cytotoxic T lymphocytes (CTLs) infiltration, reversing an immunosuppressive tumor microenvironment to an immunoresponsive one. Treatment with the nanosystem in combination with anti-PD-L1 antibody results in distinctive inhibition of tumor growth and lung metastasis, supporting that a potent ICD effect can significantly boost the therapeutic efficacy of the anti-PD-L1 antibody. This self-amplified biomimetic nanoplatform offers a promising means of raising the response rate of immune checkpoint inhibitors.

Ethylene plays an important role in grape ripening, acting through ethylene response factors (ERFs) that are downstream of ethylene signaling pathways, yet little else is known about these molecular mechanisms. In our study, we demonstrate that ethylene treatment promotes grape ripening and that the transcription factor VvERF75 shows increased expression during grape ripening. VvERF75 was found to contain a conserved AP2/ERF domain, to be a member of the ERF IXa subfamily, and to localize to the nucleus and to have transactivation activity in yeast. Our in vitro assays showed that VvERF75 can positively regulate expression of pheophorbide a oxygenase (VvPAO1) and 1-aminocyclopropane-1-carboxylic acid synthase (VvACS5) by binding to DRE motifs in their promoters. Overexpression of VvERF75 in tomato increased ethylene production and resulted in dwarfed plants, early flowering and fruit ripening, increased SlPAO and SlACS transcript levels, and decreased chlorophyll content. In general, our study revealed that VvERF75 may regulate fruit ripening by promoting ethylene biosynthesis and chlorophyll degradation, a discovery that lays a foundation for deciphering the molecular mechanisms of grape ripening.

CRISPR-based gene-drives can carry the Cas9 and guide RNA (gRNA) components in a single-linked cassette or in separate elements inserted into different genomic loci. Here the authors genetically transform and compare full versus split drives, with the former performing less efficiently than predicted.

In this study, an ensemble forecasting model for in situ wind speed and wave height was developed using the Coupled Ocean–Atmosphere–Wave–Sediment Transport (COAWST) model. This model utilized four bias correction algorithms—Model Output Statistics (MOS), Back Propagation Neural Network (BPNN), Long Short-Term Memory (LSTM) neural network, and Convolutional Neural Network (CNN)—to construct ensemble forecasts. The training data were derived from the COAWST simulations of one year and observations from three buoy stations (Laohutan, Zhifudao, and Lianyungang) in the Yellow Sea and Bohai Sea. After the optimization of the bias correction model training, the subsequent evaluations on the ensemble forecasts showed that the in situ forecasting accuracy of wind speed and wave height was significantly improved. Although there were some uncertainties on bias correction performance levels for individual algorithms, the uncertainties were greatly reduced by the ensemble forecasts. Depending on the dynamic weight assignment, the ensemble forecasts presented a stable performance even when the corrected forecasts by three algorithms had an obvious negative bias. Specifically, the ensemble forecasting bias was found with a mean reduction of about 96%~99% and 91%~95% for wind speed and wave height, and a reduction of about 91%~98% and 16%~54% during the period of Typhoon “Muifa”. For the four correction algorithms, the performance of bias correction was not directly related to the algorithm complexity. However, the strategies with more complex algorithms (i.e., CNN) were more conservative, and simple algorithms (i.e., MOS) might have induced unstable performance levels despite their lower bias in some cases.

The Coupled Ocean–Atmosphere–Wave–Sediment Transport (COAWST) model serves as the foundation for creating a forecast model to detect lower atmospheric ducts in this study. A set of prediction tests with different forecasting times focusing on the South China Sea domain was conducted to evaluate the short-term forecasting effectiveness of lower atmospheric ducts. The assessment of sounding observation data revealed that the prediction model performed well in predicting the characteristics of all types of ducts. The mean values of the forecasting errors were slightly lower than the reanalysis data but had lower levels of correlation coefficients. At an altitude of about 2000 m, the forecasted error of modified atmospheric refractivity reached peak values and then decreased gradually with increasing altitude. The accuracy of forecasted surface ducts was higher than that of elevated ducts. Noticeable land–sea differences were identified for the spatial distributions of duct characteristics, and the occurrence rates of both the surface and elevated ducts were high at sea. As for the differences among the forecasts of 24, 48, and 72 h ahead, the differences primarily occurred at altitude levels below 20 m and 500 m~1500 m, which are consistent with the differences in the duct height.

The sea surface roughness parameterization and the universal stability function are key components of the evaporation duct prediction model based on the Monin–Obukhov similarity theory. They determine the model’s performance, which in turn affects the efficiency and accuracy of electromagnetic applications at sea. In this study, we collected layered meteorological and hydrological observation data and preprocessed them to obtain near-surface reference modified refractivity profiles. We then optimized the sea surface roughness parameterization and the universal stability function using particle swarm optimization and simulated annealing algorithms. The results show that the particle swarm optimization algorithm outperforms the simulated annealing algorithm. Compared to the original model, the particle swarm optimization algorithm improved the prediction accuracy of the model by 5.09% under stable conditions and by 9.97% under unstable conditions, demonstrating the feasibility of the proposed method for optimizing the evaporation duct prediction model. Subsequently, we compared the electromagnetic wave propagation path losses under two different evaporation duct heights and modified refractivity profile states, confirming that the modified refractivity profile is more suitable as the accuracy criterion for the evaporation duct prediction model.

Abstract Background The rapid growth of annual shoots is detrimental to peach production. While gibberellin (GA) promotes the rapid growth of peach shoots, there is limited information on the identity and expression profiles of GA-metabolism genes for this species. Results All six GA biosynthetic gene families were identified in the peach genome, and the expression profiles of these family members were determined in peach shoots. The upstream biosynthetic gene families have only one or two members (1 CPS, 2 KSs, and 1 KO), while the downstream gene families have multiple members (7 KAOs, 6 GA20oxs, and 5 GA3oxs). Between the two KS genes, PpKS1 showed a relatively high transcript level in shoots, while PpKS2 was undetectable. Among the seven KAO genes, PpKAO2 was highly expressed in shoots, while PpKAO1 and − 6 were weakly expressed. For the six GA20ox genes, both PpGA20ox1 and − 2 were expressed in shoots, but PpGA20ox1 levels were higher than PpGA20ox2. For the five GA3ox genes, only PpGA3ox1 was highly expressed in shoots. Among these biosynthesis genes, PpGA20ox1 and PpGA3ox1 showed a gradual decrease in transcript level along shoots from top to bottom, and a similar trend was observed in bioactive GA1 and GA4 distribution. Among the GA-deactivation genes, PpGA2ox6 was highly expressed in peach shoots. PpGA2ox1 and − 5 transcripts were relatively lower and showed a similar pattern to PpGA20ox1 and PpGA3ox1 in peach shoots. Overexpression of PpGA20ox1, − 2, or PpGA2ox6 in Arabidopsis or tobacco promoted or depressed the plant growth, respectively, while PpGA3ox1 did not affect plant height. Transient expression of PpGA20ox1 in peach leaves significantly increased bioactive GA1 content. Conclusions Our results suggest that PpGA20ox and PpGA2ox expression are closely associated with the distribution of active GA1 and GA4 in peach annual shoots. Our research lays a foundation for future studies into ways to effectively repress the rapid growth of peach shoot.

In this study, a forecasting model was developed based on the COAWST and atmospheric 3D EnVar module to investigate the effects of assimilation of the sounding and COSMIC–2 data on the forecasting of the revised atmospheric refraction. Three groups of 72 h forecasting tests, with assimilation of different data obtained for a period of one month, were constructed over the Yellow Sea. The results revealed that the bias of the revised atmospheric refraction was the lowest if both the sounding and COSMIC–2 data were assimilated. As a result of the assimilation of the hybrid data, the mean bias reduced by 6.09–6.28% within an altitude of 10 km, and the greatest reduction occurred below the altitude of 3000 m. In contrast, the test that assimilated only the sounding data led to an increase in bias at several levels. This increased bias was corrected after the introduction of the COSMIC–2 data, with the mean correction of 1.6 M within the middle and lower troposphere. During the typhoon period, the improvements in the assimilation were more significant than usual. The improved forecasts of the revised atmospheric refraction were mainly due to the moisture changes within the middle and lower troposphere, while the changes in the upper troposphere were influenced by multiple factors.

Evaporation ducts are abnormal states of the atmosphere in the air–sea boundary layer that directly affect the propagation trajectory of electromagnetic (EM) waves. Therefore, an accurate diagnosis of the evaporation duct height (EDH) is important for studying the propagation trajectory of EM waves in evaporation ducts. Most evaporation duct models (EDMs) based on the Monin–Obukhov similarity theory are empirical methods. Different EDMs have different levels of environmental adaptability. Evaporation duct diagnosis methods based on machine learning methods only consider the mathematical relationship between data and do not explore the physical mechanism of evaporation ducts. To solve the above problems, this study observed the meteorological and hydrological parameters of the five layers of the low-altitude atmosphere in the East China Sea on board the research vessel Xiangyanghong 18 in April 2021 and obtained the atmospheric refractivity profile. An evaporation duct multimodel fusion diagnosis method (MMF) based on a library for support vector machines (LIBSVM) is proposed. First, based on the observed meteorological and hydrological data, the differences between the EDH diagnosis results of different EDMs and MMF were analyzed. When ASTD ≥ 0, the average errors of the diagnostic results of BYC, NPS, NWA, NRL, LKB, and MMF are 2.57 m, 2.92 m, 2.67 m, 3.27 m, 2.57 m, and 0.24 m, respectively. When ASTD < 0, the average errors are 2.95 m, 2.94 m, 2.98 m, 2.99 m, 2.97 m, and 0.41 m, respectively. Then, the EM wave path loss accuracy analysis was performed on the EDH diagnosis results of the NPS model and the MMF. When ASTD ≥ 0, the average path loss errors of the NPS model and MMF are 5.44 dB and 2.74 dB, respectively. When ASTD < 0, the average errors are 5.21 dB and 3.46 dB, respectively. The results show that the MMF is suitable for EDH diagnosis, and the diagnosis accuracy is higher than other models.

Evaporation ducts are a phenomenon that occurs with extremely high frequency at the boundary between the atmosphere and the ocean. Because they directly affect the propagation of electromagnetic waves, it is necessary to study their various characteristics. Since it is difficult to conduct large-scale observations at sea, many researchers use reanalysis data for this task instead of observation data. However, there have been no studies verifying accuracy of this analysis method for the diagnosis of evaporation ducts. Therefore, in this work, observations of the low-altitude atmospheric refractivity profile were carried out over the East China Sea on board the research vessel Xiangyanghong 18 in April 2021. First, the differences between different evaporation duct models were examined based on the meteorological and hydrological data obtained at different heights. It was concluded that the diagnostic accuracy of the evaporation duct model is low in stable conditions and a low-wind-speed environment. Under the same conditions, the Naval Postgraduate School (NPS) model showed high diagnostic accuracy when compared with other models. Second, Taylor plots were used to verify the accuracy of the reanalysis data and the observation data. It was concluded that the single-parameter precision of the reanalysis data is relatively high, and there were strong correlations with the observation data. Finally, the observation and reanalysis data were used to compare and analyze the false-report rate, the missing-report rate, and the accuracy of the diagnosed evaporation duct height using the NPS model. The false-report rate and missing-report rate were found to be 1.93% and 1.52%, respectively. The average diagnosis deviation was 3.34 m. The Pearson correlation coefficient was found to be close to 1. The results indicate that it is basically feasible to analyze the characteristics of evaporation ducts based on the NPS model using reanalysis data.

Complement receptor 3 (CD11b/CD18) is an important receptor that mediates adhesion, phagocytosis and chemotaxis in various immunocytes. The conidia of the medically-important pathogenic fungus, Aspergillus fumigatus can be internalized into alveolar epithelial cells to disseminate its infection in immunocompromised host; however, the role of CR3 in this process is poorly understood. In the present study, we investigated the potential role of CR3 on A. fumigatus internalization into type II alveolar epithelial cells and its effect on host intracellular PA content induced by A. fumigatus. We found that CR3 is expressed in alveolar epithelial cells and that human serum and bronchoalveolar lavage fluid (BALF) could improve A. fumigatus conidial internalization into A549 type II alveolar epithelial cell line and mouse primary alveolar epithelial cells, which were significantly inhibited by the complement C3 quencher and CD11b-blocking antibody. Serum-opsonization of swollen conidia, but not resting conidia led to the increase of cellular phosphatidic acid (PA) in A549 cells during infection. Moreover, both conidial internalization and induced PA production were interfered by CD11b-blocking antibody and dependent on FAK activity, but not Syk in alveolar epithelial cells. Overall, our results revealed that CR3 is a critical modulator of Aspergillus fumigatus internalization into alveolar epithelial cells.

Abstract Background Macromoleculization of nitroxides has been an effective strategy to improve low relaxivities and poor in vivo stability, however, nitroxides-based metal-free magnetic resonance imaging (MRI) macromolecular contrast agents (mCAs) are still under-performed. These mCAs do not possess a high nitroxides content sufficient for a cumulative effect. Amphiphilic nanostructures in these mCAs are not stable enough for highly efficient protection of nitroxides and do not have adequate molecular flexibility for full contact of the paramagnetic center with the peripheral water molecules. In addition, these mCAs still raise the concerns over biocompatibility and biodegradability due to the presence of macromolecules in these mCAs. Results Herein, a water-soluble biodegradable nitroxides-based mCA (Linear pDHPMA-mPEG-Ppa-PROXYL) was prepared via covalent conjugation of a nitroxides (2,2,5,5-tetramethyl-1-pyrrolidinyl-N-oxyl, PROXYL) onto an enzyme-sensitive linear di-block poly[N-(1, 3-dihydroxypropyl) methacrylamide] (pDHPMA). A high content of PROXYL up to 0.111 mmol/g in Linear pDHPMA-mPEG-Ppa-PROXYL was achieved and a stable nano-sized self-assembled aggregate in an aqueous environment (ca. 23 nm) was formed. Its longitudinal relaxivity (r 1 = 0.93 mM− 1 s− 1) was the highest compared to reported nitroxides-based mCAs. The blood retention time of PROXYL from the prepared mCA in vivo was up to ca. 8 h and great accumulation of the mCA was realized in the tumor site due to its passive targeting ability to tumors. Thus, Linear pDHPMA-mPEG-Ppa-PROXYL could provide a clearly detectable MRI enhancement at the tumor site of mice via the T1WI SE sequence conventionally used in clinical Gd3+-based contrast agents, although it cannot be compared with DTPA-Gd in the longitudinal relaxivity and the continuous enhancement time at the tumor site of mice. Additionally, it was demonstrated to have great biosafety, hemocompatibility and biocompatibility. Conclusions Therefore, Linear pDHPMA-mPEG-Ppa-PROXYL could be a potential candidate as a substitute of metal-based MRI CAs for clinical application. Graphic Abstract

Abstract Background In order to address the potential toxicity of metal-based magnetic resonance imaging (MRI) contrast agents (CAs), a concept of non-metallic MRI CAs has emerged. Currently, paramagnetic nitroxides (such as (2,2,5,5-tetramethylpyrrolidine-1-oxyl, PROXYL), (2,2,6,6-tetramethylpiperidine-1-oxide, TEMPO), etc.) are being extensively studied because their good stability and imaging mechanism are similar to metal-based contrast agents (such as Gd3+ chelate-based clinical CAs). However, a lower relaxivity and rapid in vivo metabolism of nitroxides remain to be addressed. Previous studies have demonstrated that the construction of macromolecular nitroxides contrast agents (mORCAs) is a promising solution through macromolecularization of nitroxides (i.e., use of large molecules to carry nitroxides). Macromolecular effects not only increase the stability of nitroxides by limiting their exposure to reductive substances in the body, but also improve the overall 1H water relaxation by increasing the concentration of nitroxides and slowing the molecular rotation speed. Results Branched pDHPMA-mPEG-Ppa-PROXYL with a high molecular weight (MW = 160 kDa) and a nitroxides content (0.059 mmol/g) can form a nanoscale (~ 28 nm) self-assembled aggregate in a water environment and hydrophobic PROXYL can be protected by a hydrophilic outer layer to obtain strong reduction resistance in vivo. Compared with a small molecular CA (3-Carboxy-PROXYL (3-CP)), Branched pDHPMA-mPEG-Ppa-PROXYL displays three prominent features: (1) its longitudinal relaxivity (0.50 mM− 1 s− 1) is about three times that of 3-CP (0.17 mM− 1 s− 1); (2) the blood retention time of nitroxides is significantly increased from a few minutes of 3-CP to 6 h; (3) it provides long-term and significant enhancement in MR imaging of the tumor, liver, kidney and cardiovascular system (heart and aortaventralis), and this is the first report on nitroxides-based MRI CAs for imaging the cardiovascular system. Conclusions As a safe and efficient candidate metal-free magnetic resonance contrast agent, Branched pDHPMA-mPEG-Ppa-PROXYL is expected to be used not only in imaging the tumor, liver and kidney, but also the cardiovascular system, which expands the application scope of these CAs. Multi-modal therapeutics are emerging for simultaneous diagnosis and treatment of cancer. Polymeric carriers are often employed for loading multiple drugs due to their versatility and controlled release of these drugs in response to a tumor specific microenvironment. A theranostic nanomedicine was designed and prepared by complexing a small gadolinium chelate, conjugating a chemotherapeutic drug PTX through a cathepsin B-responsive linker and covalently bonding a fluorescent probe pheophorbide a (Ppa) with a branched glycopolymer. The branched prodrug-based nanosystem was degradable in the tumor microenvironment with overexpressed cathepsin B, and PTX was simultaneously released to exert its therapeutic effect. The theranostic nanomedicine, branched glycopolymer-PTX-DOTA-Gd, had an extended circulation time, enhanced accumulation in tumors, and excellent biocompatibility with significantly reduced gadolinium ion (Gd3+) retention after 96 h post-injection. Enhanced imaging contrast up to 24 h post-injection and excellent antitumor efficacy with a tumor inhibition rate more than 90% were achieved from glycopolymer-PTX-DOTA-Gd without obvious systematic toxicity. This branched polymeric prodrug-based nanomedicine is very promising for safe and effective diagnosis and treatment of cancer.

The plant growth regulator ethylene influences rachis browning in grape (Vitis vinifera L.). Although the ethylene signaling pathway is well defined, there is limited knowledge on its mode of action during rachis browning. Here, we show that an ethylene response factor (VvERF111) positively regulates chlorophyll degradation in rachis by binding to a DRE motif in the promoter of VvCLH1. The expression of VvERF111 and VvCLH1 in rachis was induced by ethylene and inhibited by 1-methylcyclopropene (1-MCP). VvERF111 belongs to the ERF IX subfamily of the APETALA2/ethylene responsive factor (AP2/ERF) superfamily, shows transcriptional activity in yeast, and is localized in the nucleus and membrane. The transient overexpression of VvERF111 or chlorophyllase (VvCLH1) in grape leaves accelerated chlorophyll degradation. In VvERF111-overexpressing leaves, transcript levels of VvCLH1 were also increased. Our findings offer a deeper understanding of the transcriptional regulation of chlorophyll degradation during the rachis browning of grape.

Fruit ripening includes several metabolic changes that lead to sweeter and softer fruit. Pectin depolymerization is one of major factors that softens developing grape berries. Pectin lyases (PLs) play important roles in pectin degradation in the grape berry. However, little is known about the temporal and spatial expression of grapevine (Vitis spp.) pectin lyase genes (VvPLs) or their function during fruit ripening and softening. In this study, 18 individual VvPL genes were identified in the grape genome. All VvPL genes were sorted into group I and group II, except VvPL12 which demonstrated higher and similar expression trends in different tissues and organs. In grape berry, VvPL1, 5, 7, 11 and 16 were highly expressed, whereas VvPL18, 15, 2, 13, 10, 14, 17, 6 and 8 showed lower expression levels at different berry developmental stages. Expression of VvPL11 firstly increased and then decreased, and the highest expression was shown at 6 weeks after full bloom (WAFB) during berry development. Over-expression of the VvPL11 gene in tomato caused higher ethylene production and lower firmness compared to wild-type fruit. Moreover, decreased propectin and increased water-soluble pectin (WSP) levels were observed in VvPL11 transgenic tomato fruit. Consistent with this result, the expression levels of SlPG2, SlEXP, and SlPME1, all of which are genes involved in fruit softening, were up-regulated in VvPL11-OE tomato fruit, which supported the idea that VvPL11 plays an important role in fruit ripening and softening. This study provided a comprehensive analysis of the grapevine PL family and advanced our knowledge of the functions of VvPLs during fruit softening.

The domesticated silkworm, Bombyx mori, is an economically important insect that synthesizes large amounts of silk proteins in its silk gland to make cocoons. In recent years, germline transformation strategies advanced the bioengineering of the silk gland as an ideal bioreactor for mass production of recombinant proteins. However, the yield of exogenous proteins varied largely due to the random insertion and gene drift caused by canonical transposon-based transformation, calling for site-specific and stable expression systems. In the current study, we established a targeted in-fusion expression system by using the transcription activator-like effector nuclease (TALEN)-mediated targeted insertion to target genomic locus of sericin, one of the major silk proteins. We successfully generated chimeric Sericin1-EGFP (Ser-2A-EGFP) transformant, producing up to 3.1% (w/w) of EGFP protein in the cocoon shell. With this strategy, we further expressed the medically important human epidermal growth factor (hEGF) and the protein yield in both middle silk glands, and cocoon shells reached to more than 15-fold higher than the canonical piggyBac-based transgenesis. This natural Sericin1 expression system provides a new strategy for producing recombinant proteins by using the silkworm silk gland as the bioreactor.

Abstract Background Ubiquitin ligases (E3) are the enzymes in the ubiquitin/26S proteasome pathway responsible for targeting proteins to the degradation pathway and play major roles in multiple biological activities. However, the E3 family and their functions are yet to be identified in the fruit of peach. Results In this study, genome-wide identification, classification and characterization of the E3 ligase genes within the genome of peach (Prunus persica) was carried out. In total, 765 E3 (PpE3) ligase genes were identified in the peach genome. The PpE3 ligase genes were divided into eight subfamilies according to the presence of known functional domains. The RBX subfamily was not detected in peach. The PpE3 ligase genes were not randomly distributed among the 8 chromosomes, with a greater concentration on the longer chromosomes. The primary mode of gene duplication of the PpE3 ligase genes was dispersed gene duplication (DSD). Four subgroups of the BTB subfamily never characterized before were newly identified in peach, namely BTBAND, BTBBL, BTBP and BTBAN. The expression patterns of the identified E3 ligase genes in two peach varieties that display different types of fruit softening (melting flesh, MF, and stony hard, SH) were analyzed at 4 different stages of ripening using Illumina technology. Among the 765 PpE3 ligase genes, 515 (67.3%) were expressed (FPKM > 1) in the fruit of either MF or SH during fruit ripening. In same-stage comparisons, 231 differentially expressed genes (DEGs) were identified between the two peach cultivars. The number of DEGs in each subfamily varied. Most DEGs were members of the BTB, F-box, U-box and RING subfamilies. PpE3 ligase genes predicted to be involved in ethylene, auxin, or ABA synthesis or signaling and DNA methylation were differentially regulated. Eight PpE3 ligase genes with possible roles in peach flesh texture and fruit ripening were discussed. Conclusions The results of this study provide useful information for further understanding the functional roles of the ubiquitin ligase genes in peach. The findings also provide the first clues that E3 ligase genes may function in the regulation of peach ripening.

In order to diagnose an incipient fault in rotating machinery under complicated conditions, a fast sparse decomposition based on the Teager energy operator (TEO) is proposed in this paper. In this proposed method, firstly, the TEO is employed to enhance the envelope of the impulses, which is more sensitive to frequency and can eliminate the low-frequency harmonic component and noise; secondly, a smoothing filtering algorithm was adopted to suppress the noise in the TEO envelope; thirdly, the fault signal was reconstructed by multiplication of the filtered TEO envelope and the original fault signal; finally, sparse decomposition was used based on a generalized S-transform (GST) to obtain the sparse representation of the signal. The proposed preprocessing method using the filtered TEO can overcome the interference of high-frequency noise while maintaining the structure of fault impulses, which helps the processed signal perform better on sparse decomposition; sparse decomposition based on GST was used to represent the fault signal more quickly and more accurately. Simulation and application prove that the proposed method has good accuracy and efficiency, especially in conditions of very low SNR, such as impulses with anSNR of −8.75 dB that are submerged by noise of the same amplitude.

The wake management of offshore wind farms (OWFs) mainly considers the wake effect. Wake effects commonly occur in offshore wind farms, which cause a 5–10% reduction in power production. Although there have been many studies on wake management, many methods are not accurate enough; for instance, look-up table and static wake model control methods do not consider the time-varying wake state. Dynamic wake management is based on the real-time dynamic wake, so it can increase the energy of the OWFs effectively. For OWFs, dynamic wake control is the main method of dynamic wake management. In this paper, the existing wake model and control progress are discussed, mainly emphasizing the dynamic wake model and the dynamic wake control method, solving the gap of the review for dynamic wake management. This paper presents a digital twins (DT) framework for power and fatigue damage for the first time.. The structure of this paper is as follows: (1) the mechanism of wind farm wake interference is described and then the dynamic wake model is reviewed and summarized; (2) different control methods are analyzed and the dynamic wake management strategies for different control methods are reviewed; (3) in order to solve the problems of dynamic wake detection and real-time effective control, the technology of DT is applied to the dynamic wake control of OWFs. This new DT frame has a promising application prospect in improving power and reducing fatigue damage.

Aiming at the problems of early weak fault feature extraction of bearings in rotating machinery, an improved stochastic resonance (SR) is proposed combined with the advantage of SR to enhance weak characteristic signals with noise energy. Firstly, according to the characteristics of the large parameters of the actual fault signal, the amplitude transform coefficient and frequency transform coefficient are introduced to convert the large parameter signal into small parameter signal which can be processed by SR, and the relationship of second-order parameters are introduced. Secondly, a comprehensive evaluation index (CEI) consisted of power spectrum kurtosis, correlation coefficient, structural similarity, root mean square error, and approximate entropy, is constructed through BP neural network. Moreover, this CEI is adopted as fitness function to search the optimal damping coefficient and amplitude transform coefficient with adaptive weight particle swarm optimization (PSO). Finally, according to the improved optimal SR system, the weak fault feature can be extracted. The simulation and experiment verify the effectiveness of the proposed method compared with traditional second-order general scale transform adaptive SR.

Abstract Background The glyoxalase system usually comprises two enzymes, glyoxalase I (GLYI) and glyoxalase II (GLYII). This system converts cytotoxic methylglyoxal (MG) into non-toxic D-lactate in the presence of reduced glutathione (GSH) in two enzymatic steps. Recently, a novel type of glyoxalase III (GLYIII) activity has observed in Escherichia coli that can detoxify MG into D-lactate directly, in one step, without a cofactor. Investigation of the glyoxalase enzymes of a number of plant species shows the importance of their roles in response both to abiotic and to biotic stresses. Until now, glyoxalase gene families have been identified in the genomes of four plants, Arabidopsis, Oryza sativa, Glycine max and Medicago truncatula but no similar study has been done with the grapevine Vitis vinifera L. Results In this study, four GLYI-like, two GLYII-like and three GLYIII-like genes are identified from the genome database of grape. All these genes were analysed in detail, including their chromosomal locations, phylogenetic relationships, exon-intron distributions, protein domain organisations and the presence of conserved binding sites. Using quantitative real-time PCR analysis (qRT-PCR), the expression profiles of these genes were analysed in different tissues of grape, and also when under infection stress from downy mildew (Plasmopara viticola). The study reveals that most VvGLY-like genes had higher expressions in stem, leaf, tendril and ovule but lower expressions in the flower. In addition, most of the VvGLY-like gene members were P. viticola responsive with high expressions 6-12 h and 96-120 h after inoculation. However, VvGLYI-like1 was highly expressed 48 h after inoculation, similar to VvPR1 and VvNPR1 which are involved in the defence response. Conclusions This study identified the GLYI-like, GLYII-like and GLYIII-like full gene families of the grapevine. Based on a phylogenetic analysis and the presence of conserved binding sites, we speculate that these glyoxalase-like genes in grape encode active glyoxalases. Moreover, our study provides a basis for discussing the roles of VvGLYI-like, VvGLYII-like and VvGLYIII-like genes in grape’s response to downy mildew infection. Our results shed light on the selection of candidate genes for downy mildew tolerance in grape and lay the foundation for further functional investigations of these glyoxalase genes.

Aiming at the parachute recovery of fixed-wing unmanned aerial vehicles, a method of parachute deployment by tractor rocket is proposed. First, the tensile tests were carried out on high-strength polyethylene and brocade silk-weaved composite materials. The dynamic property parameters of the materials were obtained, which was the input for the dynamic model of the parachute deployment phase. Second, the model was verified by the experiment results. Finally, parachute weight and rocket launch temperature during the deployment phase were studied. The results showed that the dynamic model has good accuracy; as the parachute weight increases, the maximum snatch force of the extraction line and the sling decreases as the force on the suspension lines increases and the deployment effect worsens. With the temperature rise, the maximum snatch force on the extraction line, sling, and suspension lines increases and the deployment length changes slightly.

Atmospheric reactive oxygen species (ROS) play a key role in the process of air pollution and oxidative damage to organisms. The analysis of ROS was carried out by the capture-derivative method. Therefore, it is necessary to prepare an effective molecular membrane to trap and detect ROS. Electrospinning membranes were prepared by combining the electrospinning technique with chrysin, baicalein, scutellarin, genistein, quercetin, and baicalin. By comparing the structures of the membranes before and after the reaction, the fluorescence enhancement characteristics of the reactive molecular membranes and the atmospheric radicals were studied. The ability of the active molecular membranes to trap atmospheric radicals was also studied. It was found that the genistein active molecular membrane had good trapping ability in four environments. The fluorescence enhancement rates in ROS, OH radical and O3 simulated environments were 39.32%, 7.99% and 11.92%, respectively. The fluorescence enhancement rate in atmospheric environment was 16.16%. Indeed, the sites where the atmospheric radicals react with the active molecular membranes are discussed. It is found that it is mainly related to the 5,7 phenolic hydroxyl of ring A, catechol structure and the coexistence structure of 4′ phenolic hydroxyl of ring B and 7 phenolic hydroxyl of ring A. Therefore, the genistein molecular membrane has shown great potential in its trapping ability and it is also environmentally friendly.

Background Heat shock factors (HSFs) play important roles during normal plant growth and development and when plants respond to diverse stressors. Although most studies have focused on the involvement of HSFs in the response to abiotic stresses, especially in model plants, there is little research on their participation in plant growth and development or on the HSF (PpHSF) gene family in peach (Prunus persica). Methods DBD (PF00447), the HSF characteristic domain, was used to search the peach genome and identify PpHSFs. Phylogenetic, multiple alignment and motif analyses were conducted using MEGA 6.0, ClustalW and MEME, respectively. The function of PpHSF5 was confirmed by overexpression of PpHSF5 into Arabidopsis. Results Eighteen PpHSF genes were identified within the peach genome. The PpHSF genes were nonuniformly distributed on the peach chromosomes. Seventeen of the PpHSFs (94.4%) contained one or two introns, except PpHSF18, which contained three introns. The in silico-translated PpHSFs were classified into three classes (PpHSFA, PpHSFB and PpHSFC) based on multiple alignment, motif analysis and phylogenetic comparison with HSFs from Arabidopsis thaliana and Oryza sativa. Dispersed gene duplication (DSD at 67%) mainly contributed to HSF gene family expansion in peach. Promoter analysis showed that the most common cis-elements were the MYB (abiotic stress response), ABRE (ABA-responsive) and MYC (dehydration-responsive) elements. Transcript profiling of 18 PpHSFs showed that the expression trend of PpHSF5 was consistent with shoot length changes in the cultivar ‘Zhongyoutao 14’. Further analysis of the PpHSF5 was conducted in 5-year-old peach trees, Nicotiana benthamiana and Arabidopsis thaliana, respectively. Tissue-specific expression analysis showed that PpHSF5 was expressed predominantly in young vegetative organs (leaf and apex). Subcellular localization revealed that PpHSF5 was located in the nucleus in N. benthamiana cells. Two transgenic Arabidopsis lines were obtained that overexpressed PpHSF5. The root length and the number of lateral roots in the transgenic seedlings were significantly less than in WT seedlings and after cultivation for three weeks. The transgenic rosettes were smaller than those of the WT at 2–3 weeks. The two transgenic lines exhibited a dwarf phenotype three weeks after transplanting, although there was no significant difference in the number of internodes. Moreover, the PpHSF5-OE lines exhibited enhanced thermotolerance. These results indicated that PpHSF5 might be act as a suppresser of growth and development of root and aerial organs.

Peach (Prunus persica L. Batsch) trees grow vigorously and are subject to intense pruning during orchard cultivation. Reducing the levels of endogenous gibberellins (GAs) represents an effective method for controlling branch growth. Gibberellin 2-oxidases (GA2oxs) deactivate bioactive GAs, but little is known about the GA2ox gene family in peach. In this study, we identified seven PpGA2ox genes in the peach genome, which were clustered into three subgroups: C19-GA2ox-I, C19-GA2ox-II, and C20-GA2ox-I. Overexpressing representative genes from the three subgroups, PpGA2ox-1, PpGA2ox-5, and PpGA2ox-2, in tobacco resulted in dwarf plants with shorter stems and smaller leaves than the wild type. An analysis of the GA metabolic profiles of the transgenic plants showed that PpGA2ox-5 (a member of subgroup C19-GA2ox-II) is simultaneously active against both C19-GAs and C20-GAs,which implied that C19-GA2ox-II enzymes represent intermediates of C19-GA2oxs and C20-GA2oxs. Exogenous GA3 treatment of shoot tips activated the expression of all seven PpGA2ox genes, with different response times: the C19-GA2ox genes were transcriptionally activated more rapidly than the C20-GA2ox genes. GA metabolic profile analysis suggested that C20-GA2ox depletes GA levels more broadly than C19-GA2ox. These results suggest that the PpGA2ox gene family is responsible for fine-tuning endogenous GA levels in peach. Our findings provide a theoretical basis for appropriately controlling the vigorous growth of peach trees.

In this study, an experimental hydrometeorological forecasting system was developed based on the Coupled Ocean-Atmosphere-Wave-Sediment Transport (COAWST) model. The system downloads global real-time ocean, atmosphere, and wave forcing data, producing regional forecasts every day. A coastal area in South China, encompassing Hainan Island, Leizhou Peninsula, and surrounding sea areas, was chosen as the study domain. A series of 72-hour forecasting simulations were conducted in the area, lasting from July 27 to August 31, 2019. The forecasts throughout August were chosen for evaluation with station observations, along with two sets of reanalysis data, ERA5 and CLDAS. The evaluation results revealed that the COAWST model had high potential for routine forecasting operations. The 24 h forecasts, with a lead time of 24 hours, had high accuracy, while the 48 h and 72 h forecasts did not differ greatly in terms of performance. The distributions of bias between forecast and reanalysis data showed obvious differences between land and sea, with more forecasted precipitation and lower temperatures in land grids than in sea grids. In most cases, the forecasts were closer to ERA5 in terms of means and other statistical measures. The forecasts enlarged the land-sea differences of temperature when compared with ERA5 and strengthened summer monsoon with more moisture transported to land areas. Resulting from that, a forecasted bias of lower surface pressure, higher air humidity, stronger south wind, and so forth was also detected over the domain but at low values.

Abstract Herein, two water‐soluble PROXYL‐based magnetic resonance imaging (MRI) macromolecular organic contrast agents (mORCAs) are designed and synthesized: linear and cross‐linked PCE‐mPEG‐Ppa‐PROXYL. They are prepared by conjugating linear and cross‐linked poly(carboxylate ester) (PCE) with poly(ethylene glycol) (mPEG2000)‐modified nitroxides (PROXYL), respectively. Both mORCAs form self‐assembled aggregates in an aqueous phase and PROXYL is protected inside a hydrophobic core to achieve great resistance to reduction in the physiological environment, and they have low toxicity. Since cross‐linked PCE‐mPEG‐Ppa‐PROXYL possess a branched architecture, its self‐assembled aggregate is more stable and compact with a greater particle size. Cross‐linked PCE‐mPEG‐Ppa‐PROXYL outperform the linear one in the following aspects: 1) its longitudinal relaxivity (r1 = 0.79 mm−1 s−1) is higher than that of the linear one (r1 = 0.64 mm−1 s−1) and both excel the best mORCA reported so far (r1 = 0.42 mm−1 s−1); 2) its blood retention time (≈48 h) is longer than that of its linear counterpart (≈10 h); 3) cross‐linked PCE‐mPEG‐Ppa‐PROXYL provided better MR imaging contrast resolution in normal organs (liver and kidney) and tumor of mice than the linear one. Overall, cross‐linked PCE‐mPEG‐Ppa‐PROXYL may have great potential to be a novel metal‐free macromolecular contrast agent for MR imaging.

Abstract A stimuli‐responsive polymeric prodrug‐based nanotheranostic system with imaging agents (cyanine5.5 and gadolinium‐chelates) and a therapeutic agent paclitaxel (PTX) is prepared via polymerization and conjugating chemistry. The branched polymeric PTX‐Gd‐based nanoparticles (BP‐PTX‐Gd NPs) demonstrate excellent biocompatibility, and high stability under physiological conditions, but they stimuli‐responsively degrade and release PTX rapidly in a tumor microenvironment. The in vitro behavior of NPs labeled with fluorescent dyes is effectively monitored, and the NPs display high cytotoxicity to 4T1 cells similar to free PTX by impairing the function of microtubules, downregulating anti‐apoptotic protein Bcl‐2, and upregulating the expression of Bax, cleaved caspase‐3, cleaved caspase‐9, cleaved‐PARP, and p53 proteins. Great improvement in magnetic resonance imaging (MRI) is demonstrated by these NPs, and MRI accurately maps the temporal change profile of the tumor volume after injection of NPs and the tumor treatment process is also closely correlated with the T1 values measured from MRI, demonstrating the capability of providing real‐time feedback to the chemotherapeutic treatment effectiveness. The imaging‐guided chemotherapy to the 4T1 tumor in the mice model achieves an excellent anti‐tumor effect. This stimuli‐responsive polymeric nano‐agent opens a new door for efficient breast cancer treatment under the guidance of fluorescence/MRI.

It is of prime importance to develop anode electrocatalysts for direct methanol fuel cells (DMFCs) with good performance, which is critical for their commercial applications. Metal-organic framework (MOF)-derived carbon materials are extensively developed as supports of catalysts. Herein, Co embedded nitrogen-doped carbon nanotube hollow porous carbon (Co-NCNT-HPC) derived from MOFs have been fabricated, which were synthesized by pyrolyzing at an optimized temperature of 800 °C using ZIF-8@ZIF-67 as a precursor. The presence of ZIF-8@ZIF-67 ensures the doping of nitrogen and the large specific surface area of the support materials at high temperatures. A Pt/Co-NCNT-HPC800 sample, which was synthesized using Co-NCNT-HPC800 as a support, showed an enhanced mass activity of 416.2 mA mg−1Pt for methanol oxidation reaction (MOR), and the onset potential of COad oxidation of 0.51 V, which shifted negatively about 0.13 V compared with Pt/C (20%). Moreover, the Pt/Co-NCNT-HPC800 sample exhibits high stability. This work provides a facile strategy for MOF-derived carbon materials to construct advanced electrocatalysts for MOR.

Abstract Background The HD-Zip family has a diversity of functions during plant development. In this study, we identify 33 HD-Zip transcription factors in grape and detect their expressions in ovules and somatic embryos, as well as in various vegetative organs. Results A genome-wide survey for HD-Zip transcription factors in Vitis was conducted based on the 12 X grape genome (V. vinifera L.). A total of 33 members were identified and classified into four subfamilies (I-IV) based on phylogeny analysis with Arabidopsis, rice and maize. VvHDZs in the same subfamily have similar protein motifs and intron/exon structures. An evaluation of duplication events suggests several HD-Zip genes arose before the divergence of the grape and Arabidopsis lineages. The 33 members of HD-Zip were differentially expressed in ovules of the stenospermic grape, Thompson Seedless and of the seeded grape, Pinot noir. Most have higher expressions during ovule abortion in Thompson Seedless. In addition, transcripts of the HD-Zip family were also detected in somatic embryogenesis of Thompson Seedless and in different vegetative organs of Thompson Seedless at varying levels. Additionally, VvHDZ28 is located in the nucleus and had transcriptional activity consistent with the typical features of the HD-Zip family. Our results provide a foundation for future grape HD-Zip gene function research. Conclusions The identification and expression profiles of the HD-Zip transcription factors in grape, reveal their diverse roles during ovule abortion and organ development. Our results lay a foundation for functional analysis of grape HDZ genes.

Sex determination is a hierarchically-regulated process with high diversity in different organisms including insects. The W chromosome-derived Fem piRNA has been identified as the primary sex determination factor in the lepidopteran insect, Bombyx mori, revealing a distinctive piRNA-mediated sex determination pathway. However, the comprehensive mechanism of silkworm sex determination is still poorly understood. We show here that the silkworm PIWI protein BmSiwi, but not BmAgo3, is essential for silkworm sex determination. CRISPR/Cas9-mediated depletion of BmSiwi results in developmental arrest in oogenesis and partial female sexual reversal, while BmAgo3 depletion only affects oogenesis. We identify three histone methyltransferases (HMTs) that are significantly down-regulated in BmSiwi mutant moths. Disruption one of these, BmAsh2, causes dysregulation of piRNAs and transposable elements (TEs), supporting a role for it in the piRNA signaling pathway. More importantly, we find that BmAsh2 mutagenesis results in oogenesis arrest and partial female-to-male sexual reversal as well as dysregulation of the sex determination genes, Bmdsx and BmMasc. Mutagenesis of other two HMTs, BmSETD2 and BmEggless, does not affect piRNA-mediated sex determination. Histological analysis and immunoprecipitation results support a functional interaction between the BmAsh2 and BmSiwi proteins. Our data provide the first evidence that the HMT, BmAsh2, plays key roles in silkworm piRNA-mediated sex determination.

Brain tumors in the corticospinal tract (CST) region are more likely to cause motor dysfunction. The aim of this study is to evaluate the effect of gliomas located in the CST region on motor function with diffusion tensor imaging (DTI) preoperatively.Forty-five patients with histopathologically confirmed gliomas were included in this pilot study, in all cases (low-grade n = 13, high-grade n = 32) CST but not the motor cortex were involved by the tumor. DTI image were acquired and the posterior limb of the internal capsule fractional anisotropy (FA) and relative FA (rFA = affected FA/contralateral FA) were measured. Injury of the CST from tumor was divided into three grades (grade 1: displacement, grade 2: displacement and infiltration, grade 3: displacement and disruption). The fiber density index (FDi) and relative FDi (rFDi = affected FDi/contralateral FDi) of the injured and contralateral CST were measured. The correlations between muscle strength and the CST injury grade and the rFA, affected FDi, rFDi values were calculated using Spearman rank correlation analysis. rFA and rFDi values of muscle strength groups (MMT2-5) were compared with one-way analysis of variance (ANOVA). The difference of muscle strength between low- and high-grade glioma groups were analysed with the Mann-Whitney U-test.Muscle strength was negatively correlated with the injury grade of the CST (r = -0.840, P0.05).Preoperative DTI and diffusion tensor tractography may quantify the injury degrees of CST and the extent of motor dysfunction in patients with brain glioma.

DNA double-strand breaks (DSBs) are repaired by a hierarchically regulated network of pathways. Factors influencing the choice of particular repair pathways, however remain poorly characterized. Here we develop an Integrated Classification Pipeline (ICP) to decompose and categorize CRISPR/Cas9 generated mutations on genomic target sites in complex multicellular insects. The ICP outputs graphic rank ordered classifications of mutant alleles to visualize discriminating DSB repair fingerprints generated from different target sites and alternative inheritance patterns of CRISPR components. We uncover highly reproducible lineage-specific mutation fingerprints in individual organisms and a developmental progression wherein Microhomology Mediated End-Joining (MMEJ) or Insertion events predominate during early rapid mitotic cell cycles, switching to distinct subsets of Non-Homologous End-Joining (NHEJ) alleles, and then to Homology-Directed Repair (HDR)- based gene conversion. These repair signatures enable marker-free tracking of specific mutations in dynamic populations, including NHEJ and HDR events within the same samples, for in-depth analysis of diverse gene editing events.
