The development of technologies that promote environmental stewardship while maintaining or improving the efficiency of food animal production is essential to the sustainability of producing a food supply to meet the demands of a growing population. As such, Elanco (Greenfield, IN) pursued an environmental indication for a selective β-modulator (lubabegron; LUB). LUB was recently approved by the United States Food and Drug Administration (FDA) to be fed to feedlot cattle during the last 14 to 91 d of the feeding period for reductions in gas emissions/kg of unshrunk final BW and HCW. A 4 × 2 factorial arrangement of treatments was used with the factors of dose (0.0, 1.38, 5.5, or 22.0 mg·kg-1 DM basis) and sex (steers or heifers). Three 91-d cycles were conducted (112 cattle/cycle) with each dose × sex combination being represented by a single cattle pen enclosure (CPE; 14 cattle/CPE) resulting in a total of 168 steers and 168 heifers (n = 6 replicates/dose). There were no interactions observed between dose and sex for any variable measured in the study (P ≥ 0.063). Five gases were evaluated for all pens based on CPE concentrations relative to ambient air: NH3, CH4, N2O, H2S, and CO2. Cumulative NH3 gas emissions were reduced by feeding cattle 5.5 and 22.0 mg·kg-1 LUB (P ≤ 0.023) and tended (P = 0.076) to be lower for the cattle fed 1.38 mg·kg-1 LUB compared with the negative controls (CON). The cumulative NH3 gas emission reductions of 960 to 1032 g, coupled with HCW increases (P ≤ 0.019) of 15 to 16 kg for all LUB doses vs. CON, led to reductions in NH3 gas emissions/kg HCW for all three LUB treatments (P ≤ 0.004). Similar to HCW, reductions in NH3 gas emissions/kg of unshrunk final BW were observed for all LUB doses (P ≤ 0.009) and were attributable to both decreases in NH3 gas emissions and numerical increases in BW. Dose had no effect on cumulative emissions or emissions standardized by BW or HCW for the other four gases (P ≥ 0.268). LUB is a novel tool to reduce emissions of NH3 gas per kilogram of unshrunk live BW and hot carcass weight.

With a growing global population and increased environmental concerns around animal agriculture, it is essential to humanely maximize animal performance and reduce environmental emissions. This study aims to determine the efficacy of feeding ractopamine hydrochloride (RAC), an orally active, β 1-adrenergic agonist (β1AA), to feedlot steers in the last 42 d of finishing to reduce ammonia (NH3) emissions and improve animal performance. A randomized complete block design was used to allocate 112 Angus and crossbred Angus steers (initial body weight [BW] = 566.0 ± 10.4 kg) to 8 cattle pen enclosures. Pens (n = 4 per treatment, 14 steers per pen, and 56 steers per treatment) were randomly assigned to one of two treatments: 1) CON; finishing ration containing no RAC, 2) RAC; finishing ration containing 27.3 g/907 kg dry matter (DM) basis RAC. Steers were weighed on day -1 and 0 before treatment and day 14, 28, and 42 during treatment. Treatment rations were mixed and delivered daily by masked personnel. Measured emissions included NH3, nitrous oxide (N2O), methane (CH4), hydrogen sulfide (H2S), and carbon dioxide (CO2). The primary response variables assessed were emissions standardized by live weight (LW) and hot carcass weight (HCW). Steers were harvested on day 43 and carcass data were collected on day 43 and 44. Steers fed RAC reduced NH3 emissions by 17.21% from day 0 to 28 (P = 0.032) and tended to reduce NH3 from day 0 to 42 by 11.07% (P = 0.070) vs. CON. When standardized for LW, NH3 was reduced by 23.88% from day 0 to 14 (P = 0.018), 17.80% from day 0 to 28 (P = 0.006), and 12.50% for day 0 to 42 (P = 0.027) in steers fed RAC vs. CON. Steers fed RAC had 14.05% (P = 0.013) lower cumulative NH3 emissions when standardized by HCW vs. CON. Feeding RAC to Steers reduced H2S by 29.49% from day 0 to 14 (P = 0.009) and tended to reduce H2S over day 0 to 28 by 11.14% (P = 0.086) vs. CON. When H2S emissions were standardized for LW, RAC fed steers had a 28.81% reduction from day 0 to 14 (P = 0.008) vs. CON. From day 0 to 42 the RAC fed steers tended to have a 0.24 kg/d greater average daily gain (ADG) (P = 0.066) and tended to eat 4.27% less (P = 0.069) on a DM basis vs. CON. The RAC fed steers had a 19.95% greater gain to feed ratio (G:F) compared to CON (P = 0.012). Steers fed RAC had an average of 12.52 kg greater HCW (P = 0.006) and an increase of 1.93 percentage units in dressing percent (DP) (P = 0.004) vs. CON. Ractopamine is an effective medicated feed additive for reducing NH3 and improving end product performance through HCW yields.

Objective To construct a RAW264.7 macrophage cell strain with stable knockout of Mcart-1 by using CRISPR/Cas9 gene editing technique, and to detect its biological function. Methods Cas9 and sgRNA lentivirus were used to infect RAW264.7 macrophages in two steps. Positive cells were screened with puromycin and hygromycin, and single cells were plated by flow cytometry to obtain monoclonal cells; Expression of Cas9 and Mcart-1 was detected by qPCR and Western blot, and the mutation site was confirmed by sequence analysis. Cells number counting and carboxyfluorescein diacetate succinimdyl ester (CFSE) staining were used to detect cell proliferation; ATP detection kit was used to measure total ATP content in cells; Seahorse bioenergy analyzer was used to detect cell oxygen consumption rate(OCR) and glycolysis rate. Results Successfully constructed RAW264.7 cell strain with Mcart-1 stably knocked out, denoted as Mcart-1-/--RAW264.7; A frameshift mutation occurred in Mcart-1 gene in this cell strain; Compared with the wild-type RAW264.7 cell line (RAW264.7), the OCR increased and the extracellular acidification rate(ECAR)of Mcart-1-/--RAW264.7 cells decreased; The total ATP content in Mcart-1 knocked out cells decreased(P＜0.05),mitochondrial ATP production rate decreased (P＜0.01), while glycolytic ATP production rate increased (P＜0.01); The proliferation activity of RAW264.7 cells decreased after Mcart-1 was knocked out(P＜0.001). Conclusions The proliferation activity and oxidative phosphorylation level of RAW264.7 cells were both down-regulated after the Mcart-1 was stably knocked out. This cell strain is an important tool for exploring the function of cells in the tumor microenvironment.

BackgroundAnal cancer, mainly attributed to human papillomavirus (HPV) infection, is rising in prevalence among the general population in Pakistan. This study aimed to examine the knowledge, attitudes, and practices (KAP) towards anal cancer screening and HPV of the general population in Pakistan.MethodWe surveyed anal cancer KAP using social media and snowball sampling from December 2022 to May 2023. The questionnaire had 16 knowledge, 12 attitudes, 6 practice questions, and socio-demographic variables. We applied validity criteria for inclusion and exclusion and used cutoffs ≥50% for each KAP category. We analyzed data in R with Guttman’s λ2 for reliability, did univariate and bivariate analysis, and reported frequencies, percentages, p-values, coefficients, odds ratios, and 95% confidence intervals.ResultsWe surveyed 1620 people and discovered low awareness of HPV and anal cancer causes prevention, and screening (11%-24%), high stigma and embarrassment for screening (54%-70%), strong moral beliefs (89%), condom nonuse (91%), and low engagement in health services and programs (9.1%-14%). Knowledge (75.23%, OR = 1.0984, p = 0.05) was shaped by socio-demographic factors, attitude, and practice, with higher education enhancing knowledge (OR = 1.0984, p = 0.05). Attitude (78.45%, OR = 6.6052, p< 0.001) was influenced by socio-demographic factors, practice, and knowledge as well. Younger females, single, unemployed, students, living with more family members, earning more income, and residing in Islamabad had a more positive attitude (ORs from 1.0115 to 6.6052, p< 0.05), while religion did not affect attitude (p = 0.51). Practice (9.16%, OR = 0.1820, p< 0.001) was determined by socio-demographic factors, knowledge, and attitude. Older males, employed teachers, living with more family members, earning less income, and residing in Islamabad had better practice (ORs from 0.1323 to 3.8431, p< 0.05), but marital status and religion did not influence practice (p > 0.05).ConclusionPakistani young adults need more education, awareness, health services, and programs on HPV and anal cancer, as they have low awareness, high stigma, and socio-cultural challenges. In addition, it is recommended for more research and policy initiatives are needed to address socio-cultural factors and increase anal Pap to overcome anal cancer.

BackgroundBreast Cancer (BC) stands out as the widely prevalent malignancy among all the types of cancer affecting women worldwide. There is significant evidence that the pathogenicity of BC may be altered by Human Papillomavirus (HPV) infection; however, conclusive data are not yet available.MethodsBy searching five databases, including EMBASE, IBECS, PubMed, Scopus, Science Direct, Google Scholar, and Web of Science, a thorough systematic analysis was conducted on the prevalence of HPV in BC patients from 1990 to June 30, 2022. After applying extensive eligibility criteria, we selected 74 publications for further analysis based on the prevalence of HPV infections in breast tissues. All of the data were analyzed using a random-effects meta-analysis, Cochran Q test and I2 statistic were used to calculate the heterogeneity of the prevalence among these studies using subgroup analysis. Variations in the HPV prevalence estimates in different subgroups were evaluated by subgroup meta-analysis.ResultsIn total, 3156 studies were initially screened, resulting in 93 full-text studies reviewed, with 74 meeting inclusion criteria. Among a total of 7156 BC biopsies, the pool prevalence of HPV was 25.6% (95% CI= 0.24-0.33, τ2 = 0.0369 with significant heterogeneity between estimates (I2 = 97% and p< 0.01). Consequently, 45 studies with available controls were further studied, and the prevalence of HPV in case-control studies was 26.2% with overall odds 5.55 (95% CI= 3.67-8.41, I2 = 38%, τ2 = 1.4878, p< 0.01). Further subgroup analysis of HPV revealed HPV-16 had a maximum prevalence of 9.6% (95% CI= 3.06-11.86, I2 = 0%, τ2 = 0.6111, p< 0.01). Among different geographical regions, Europe reported the maximum prevalence of HPV, i.e., 39.2% (95% CI=1.29-7.91, I2 = 18%, τ2 = 1.2911, p< 0.01). Overall distribution showed HPV-18 was a frequent HPV subtype reported in Australia.ConclusionCurrent study provides a global estimate of HPV prevalence in BC patients and demonstrates a significant association between this virus and BC etiology. Nevertheless, we recommend further investigation into the underlying mechanism is essential to validate this hypothesis.

Aerosol and tropospheric ozone transport to North America has important implications for regional air quality and climate. We present data from Chews Ridge, a remote mountaintop site in California, in February–September 2012, which identifies separate Asian dust and Asian combustion aerosol signals and quantifies exogenous ozone. We use positive matrix factorization of size-fractionated aerosol X-ray fluorescence data and 206Pb/207Pb and 208Pb/207Pb isotope measurements to isolate aerosol signals of Asian origins and find good agreement between both types source attribution techniques. Power spectra of Asian dust and Asian combustion signals reveal two distinct peaks, one seasonal (~90 days) and one monthly (~25–30 days). Overall, 89% of the Asian dust signal occurs in March–May, compared with June–September, whereas only 51% of the Asian combustion signal occurs in March–May. Nearly half the Pb observed at Chews Ridge was derived from Asia (46%; 8.3% from Asian dust, 23% from Asian combustion, and 15% of the Asian Pb signal was not explained by the positive matrix factorization). A positive correlation between deseasonalized ozone concentrations and the Asian factors suggests that ozone is elevated by 6.3 ± 0.8 ppb due to transpacific transport. Scanning mobility particle size spectra were also measured from May to September and indicate that new particles are often produced and grow throughout the afternoon during onshore flow. We hypothesize that particle nucleation on the windward edge of the continent is an important source of new particles.

SUMMARY: Lotus (Nelumbo nucifera G.) is a perennial aquatic horticultural plant with diverse architectures. Distinct plant architecture (PA) has certain attractive and practical qualities, but its genetic morphogenesis in lotus remains elusive. In this study, we employ genome‐wide association analysis (GWAS) for the seven traits of petiole length (PLL), leaf length (LL), leaf width (LW), peduncle length (PLF), flower diameter (FD), petal length (PeL), and petal width (PeW) in 301 lotus accessions. A total of 90 loci are identified to associate with these traits across 4 years of trials. Meanwhile, we perform RNA sequencing (RNA‐seq) to analyze the differential expression of the gene (DEG) transcripts between large and small PA (LPA and SPA) of lotus stems (peduncles and petioles). As a result, eight key candidate genes are identified that are all primarily involved in plant cell wall remodeling significantly associated with PA traits by integrating the results of DEGs and GWAS. To verify this result, we compare the cell wall compositions and structures of LPA versus SPA in representative lotus germplasms. Intriguingly, compared with the SPA lotus, the LPA varieties have higher content of cellulose and hemicellulose, but less filling substrates of pectin and lignin. Additionally, we verified longer cellulose chains and higher cellulose crystallinity with less interference in LPA varieties. Taken together, our study illustrates how plant cell wall remodeling affects PA in lotus, shedding light on the genetic architecture of this significant ornamental trait and offering a priceless genetic resource for future genomic‐enabled breeding. Abstract. The largest 7 Southeast Asian Studies (7SEAS) operations period within the Maritime Continent (MC) occurred in the August–September 2012 biomass burning season. Data included were observations aboard the M/Y Vasco, dispatched to the Palawan Archipelago and Sulu Sea of the Philippines for September 2012. At these locations, the Vasco observed MC smoke and pollution entering the southwest monsoon (SWM) monsoonal trough. Here we describe the research cruise findings and the finer-scale aerosol meteorology of this convectively active region. This 2012 cruise complemented a 2-week cruise in 2011 and was generally consistent with previous findings in terms of how smoke emission and transport related to monsoonal flows, tropical cyclones (TC), and the covariance between smoke transport events and the atmosphere's thermodynamic structure. Biomass burning plumes were usually mixed with significant amounts of anthropogenic pollution. Also key to aerosol behavior were squall lines and cold pools propagating across the South China Sea (SCS) and scavenging aerosol particles in their path. However, the 2012 cruise showed much higher modulation in aerosol frequency than its 2011 counterpart. Whereas in 2011 large synoptic-scale aerosol events transported high concentrations of smoke into the Philippines over days, in 2012 measured aerosol events exhibited a much shorter-term variation, sometimes only 3–12 h. Strong monsoonal flow reversals were also experienced in 2012. Nucleation events in cleaner and polluted conditions, as well as in urban plumes, were observed. Perhaps most interestingly, several cases of squall lines preceding major aerosol events were observed, as opposed to 2011 observations where these lines largely scavenged aerosol particles from the marine boundary layer. Combined, these observations indicate pockets of high and low particle counts that are not uncommon in the region. These perturbations are difficult to observe by satellite and very difficult to model. Indeed, the Navy Aerosol Analysis and Prediction System (NAAPS) simulations captured longer period aerosol events quite well but largely failed to capture the timing of high-frequency phenomena. Ultimately, the research findings of these cruises demonstrate the real world challenges of satellite-based missions, significant aerosol life cycle questions such as those the future Aerosol/Clouds/Ecosystems (ACE) will investigate, and the importance of small-scale phenomena such as sea breezes, squall lines, and nucleation events embedded within SWM patterns in dominating aerosol life cycle and potential relationships to clouds.

The Rotating Drum Impactor (RDI) is designed to collect samples of airborne particulate matter that are resolved by aerodynamic particle diameter as a function of time. The purpose of this study is to characterize the accuracy and precision of measurements for particulate matter less than 2.5 μm (PM2.5) from the latest generation of the RDI sampler for use in relatively low concentration environments. Nominal RDI size cuts were used to interpret the results; a laboratory calibration of the RDI was not undertaken. Airborne particulate matter was collected on the campus of the University of California at Davis during the winter and summer of 2013. Two RDI samplers operating with 1 h resolution, two IMPROVE samplers operating 21 h per day, and one MOUDI sampler operating 42 h every two days were deployed for both seasons to enable a comparison of co-located measurements. The RDI samples were scanned using synchrotron X-rays from the Advanced Light Source (ALS), Lawrence National Berkeley Laboratory, to obtain X-ray Fluorescence (XRF) spectra. IMPROVE and MOUDI samples were analyzed using Panalytical XRF instruments using the IMPROVE protocol, then analyzed using Inductively Coupled Plasma Mass Spectrometry (ICP-MS) using an Agilent 7500i. Repeated measurements from the same samplers and analytical techniques were compared to elucidate precision; inter-comparisons between measurements made with different sampler and analytical technique combinations were used to indirectly evaluate accuracy. The IMPROVE XRF analysis technique gave similar repeated measurements for 78% of the elements present above stated Minimum Detection Limits (MDLs) in winter and 42% of elements above MDL in summer. The MOUDI XRF analysis technique gave similar repeated measurements for 33% and 37% of the elements above stated MDLs in winter and summer, respectively. The MOUDI, ICP-MS analysis technique was the most repeatable method tested, yielding similar repeated measurements for 62% and 91% of elements above stated MDLs in winter and summer, respectively. The RDI, XRF analysis technique was the least repeatable method tested, yielding similar repeated measurements for 35–40% and 5–20% of elements above stated MDLs in winter and summer, respectively. PM2.5 measurements from IMPROVE samplers were in better agreement with integrated size-resolved measurements made by MOUDI samplers than RDI samplers in the summer season, however with higher concentrations in the winter season both instruments performed with similar accuracy when using the XRF analysis method. MOUDI samples analyzed by ICP-MS had the best agreement overall with IMPROVE samples analyzed by ICP-MS. These results suggest that MDLs should be reassessed for all methods and a longer collection time should be considered under low concentration conditions.

Atmospheric particles (total suspended particles, TSPs) mass samples (288) were collected by high volume samplers at nine sampling locations in Faisalabad, Pakistan from May 2012 to April 2013. These TSP mass samples were subjected to gravimetric and quantitative chemical analyses for determining trace elements (Pb, Cd, Ni, Zn, Cu, Fe) using atomic absorption spectroscopy and water-soluble cations (Ca2+, Mg2+, Na+ and K+, NH4+) and anions (Cl−, SO42−, and NO3−) by ion chromatography. The average TSP mass and elemental concentrations at all locations were found to be highest during the dry and lowest during the wet season. The crustal elements Ca, Fe, Mg, and Na were the largest contributors to TSP mass while elements of anthropogenic origin Pb, Cd, Ni, Cu, and Zn had relatively lower concentrations and also showing a high spatial variation. The concentration of TSP and elements exhibited the maxima at the sampling locations characterized by intensive industrial and vehicular activities. The wind rose analysis and the UNMIX model applied to chemical speciation data both identified the same three primary sources of TSP: power plant/refinery, brick kilns, and roadways. The normalized dot product was successfully used to quantify the similarity between different source profiles extracted from UNMIX model. The coupling of UNMIX with wind direction analysis complemented each other and provided a complete assessment of source contributions and locations.

The EPA regulates ambient particulate matter (PM) because substantial associations have been established between PM and health impacts. Presently, regulatory compliance involves broad control of PM emission sources based on mass concentration rather than chemical composition, although PM toxicity is likely to vary depending upon PM physicochemical properties. The overall objective of this study was to help inform source-specific PM emission control regulations. For the first time, source-oriented PM was collected from the atmosphere in Fresno, CA, onto 38 source/size substrates. Mice were exposed via oropharyngeal aspiration to equivalent mass doses [50 μg] of two size fractions: ultrafine (Dp < 0.17μm) and submicron fine (0.17 < Dp < 1 μm) during summer and winter seasons. At 24 hours post-exposure, cellular and biochemical indicators of pulmonary inflammation were evaluated in the bronchoalveolar lavage fluid. Significant inflammatory responses were elicited by vehicle, regional background, and cooking PM sources that were dependent on season and particle size. This is the first study of source-oriented toxicity of atmospheric PM and supports source-specific emissions control strategies.

Ambient particulate matter (PM) originates from a range of sources and differs in composition with respect to season, time of day, and particle size. In this study, ambient PM samples in the ultrafine and submicrometer fine range were tested for the potential to exacerbate a murine model of allergic airway inflammation when exposure occurs solely during allergic sensitization, but not during subsequent allergen challenge. Temporally resolved and size-segregated PM samples were used to understand how summer or winter, day or night, and ambient ultrafine and submicrometer fine particle size influence PM's ability to exacerbate allergic inflammation. PM was collected in urban Fresno, CA. BALB/c mice were exposed to PM and house dust mite allergen (HDM) via intranasal aspiration on d 1, 3, and 5. HDM challenge occurred on d 12-14, with inflammation assessed 24 h following final challenge. While season or particle size did not predict allergic inflammation, daytime ultrafine and submicrometer fine particles significantly increased total cellular inflammation, specifically lymphocyte and eosinophil infiltration, compared to allergic controls. Further studies examined PM-mediated changes within the lung during the period where allergen sensitization occurred by measuring direct effects of PM on pulmonary oxidative stress and inflammation. Pulmonary levels of heme oxygenase-1 (HO-1), a biomarker of oxidative stress, but not cellular inflammation, demonstrated a remarkable correlation with the degree of allergic inflammation in animals sensitized to allergen and PM concomitantly, suggesting acute PM-mediated HO-1 levels may serve as a predictive indicator of a particle's ability to exacerbate allergic airway inflammation.

Airborne particulate matter with aerodynamic size less than 10μm was collected at a California dairy in fall 2006 using a Rotating Drum Impactor (RDI) sampler in 8 size ranges (i.e., 10-5, 5-2.5, 2.5-1.15, 1.15-0.75, 0.75-0.56, 0.56-0.34, 0.34-0.26, and 0.26-0.09μm). The RDI samples were analyzed using synchrotron X-ray Fluorescence (XRF) at the Advance Light Source (ALS), Lawrence National Berkeley Laboratory, to derive the concentrations of trace elements Mg, Al, Si, P, S, Cl, K, Ca, Ti, Cu, Fe, Mn, and Zn. During this study, periods of rainfall in early November provided an opportunity to isolate particle emission sources. After the rainfalls, elemental concentrations decreased by about 80% for Mg, Al, Si, and Ti, 70% for Fe and Mn, 65% for Cu, and 60% for K and Ca. By contrast, the concentrations of the elements P, S, Zn and Cl did not decrease as significantly as others. Therefore, it is very likely that the dairy drylot corrals could be a major source of the elements Mg, Al, Si, K, Ca, Ti, Cu, Fe, and Mn under dry condition, but not of elements P, S, Zn and Cl. Elements Mg, Al, Si, K, Ca, Ti, Cu, Fe, and Mn were correlated with each other under both dry and wet conditions (R2 ranging from 0.47 to 0.99), indicating that they had common emission sources. Highly correlated sub-groups of elements shifted from dry to wet conditions, R2>0.96 among elements Mg, Al, and Si, R2>0.93 among elements K, Ca, Ti, Cu, Fe, and Mn under dry condition, and R2>0.87 among elements Mg, Al, Si, Ti, Fe, and Mn under wet condition, possibly due to the change in emission sources. Size distribution of the observed elements revealed (1) a general trend that elemental concentrations increased with particle size except for S, which had a peak at 1.15μm and (2) a greater portion of coarse elements (2.5-10μm) than fine elements (less than 2.5μm) for all elements except for S and Zn under both dry and wet conditions. © 2014 Elsevier Ltd.

A field evaluation of versatile aerosol concentration enrichment system (VACES) coupled to a rapid single- particle mass spectrometer (RSMS-3) was conducted as part of the U. S. Environmental Protection Agency Supersite program in Pittsburgh during March 2002. RSMS- 3 hit rate increases were measured, and possible particle composition changes introduced by the VACES were examined in the single- particle mass spectra. The hit rates increased by 5 - 20 times at particle sizes ranging from 40 to 640 nm. VACES only enhances the hit rate by about a factor of 2 for large particle sizes because the RSMS-3 flow rates for these particles did not match the optimum operating condition of VACES. During the 3 days of measurements most of the particles were a mixture of carbonaceous material and ammonium nitrate with a variation across the spectrum from particles that were mostly carbonaceous to particles that were mostly ammonium nitrate. Both ambient and concentrated carbonaceous and ammonium nitrate composition distributions were indistinguishable with RSMS-3, suggesting that VACES introduces an insignificant artifact for those particles.

The questionnaire had 16 knowledge, 12 attitudes, 6 practice questions, and socio-demographic variables. We applied validity criteria for inclusion and exclusion and used cutoffs ≥50% for each KAP category. We analyzed data in R with Guttman's l2 for reliability, did univariate and bivariate analysis, and reported frequencies, percentages, p-values, coefficients, odds ratios, and 95% confidence intervals. Results: We surveyed 1620 people and discovered low awareness of HPV and anal cancer causes prevention, and screening (11%-24%), high stigma and embarrassment for screening (54%-70%), strong moral beliefs (89%), condom nonuse (91%), and low engagement in health services and programs (9.1%-14%). Knowledge (75.23%, OR = 1.0984, p = 0.05) was shaped by socio-demographic factors, attitude, and practice, with higher education enhancing knowledge (OR = 1.0984, p = 0.05). Attitude (78.45%, OR = 6.6052, p< 0.001) was influenced by socio-demographic factors, practice, and knowledge as well. Younger females, single, unemployed, students, living with more family members, earning more income, and residing in Islamabad had a more positive attitude (ORs from 1.0115 to 6.6052, p< 0.05), while religion did not affect attitude (p = 0.51). Practice (9.16%, OR = 0.1820, p< 0.001) was determined by socio-demographic factors, knowledge, and attitude. Older males, employed teachers, living with more family members, earning less income, and residing in Islamabad had better practice (ORs from 0.1323 to 3.8431, p< 0.05), but marital status and religion did not influence practice (p > 0.05). Conclusion: Pakistani young adults need more education, awareness, health services, and programs on HPV and anal cancer, as they have low awareness, high stigma, and socio-cultural challenges. Background: Breast Cancer (BC) stands out as the widely prevalent malignancy among all the types of cancer affecting women worldwide. There is significant evidence that the pathogenicity of BC may be altered by Human Papillomavirus (HPV) infection; however, conclusive data are not yet available. Methods: By searching five databases, including EMBASE, IBECS, PubMed, Scopus, Science Direct, Google Scholar, and Web of Science, a thorough systematic analysis was conducted on the prevalence of HPV in BC patients from 1990 to June 30, 2022. After applying extensive eligibility criteria, we selected 74 publications for further analysis based on the prevalence of HPV infections in breast tissues. All of the data were analyzed using a random-effects meta-analysis, Cochran Q test and I2 statistic were used to calculate the heterogeneity of the prevalence among these studies using subgroup analysis. Variations in the HPV prevalence estimates in different subgroups were evaluated by subgroup meta-analysis. Results: In total, 3156 studies were initially screened, resulting in 93 full-text studies reviewed, with 74 meeting inclusion criteria. Among a total of 7156 BC biopsies, the pool prevalence of HPV was 25.6% (95% CI= 0.24-0.33, t2 = 0.0369 with significant heterogeneity between estimates (I² = 97% and p< 0.01). Consequently, 45 studies with available controls were further studied, and the prevalence of HPV in case-control studies was 26.2% with overall odds 5.55 (95% CI= 3.67-8.41, I² = 38%, t2 = 1.4878, p< 0.01). Further subgroup analysis of HPV revealed HPV-16 had a maximum prevalence of 9.6% (95% CI= 3.06-11.86, I² = 0%, t2 = 0.6111, p< 0.01). Among different geographical regions, Europe reported the maximum prevalence of HPV, i.e., 39.2% (95% CI=1.29-7.91, I² = 18%, t2 = 1.2911, p< 0.01). Overall distribution showed HPV-18 was a frequent HPV subtype reported in Australia. Conclusion: Current study provides a global estimate of HPV prevalence in BC patients and demonstrates a significant association between this virus and BC etiology.
