Microstructural alterations of brain structure in haemophilic boys were found in our previous study.We investigated alterations of brain function in school-age boys with severe haemophilia A (HA) with resting-state functional magnetic resonance imaging (rs-fMRI).We obtained rs-fMRI scans from 24 boys with HA and 25 demographically matched healthy children. Spontaneous brain activity parameters were calculated. Graph theoretical analyses on rs-fMRI data at the global and regional levels were performed. Two-sample t tests were used to analyze differences, and correlation analyses identified relationships between altered neural properties and psychological characteristics.Children with severe HA showed small-worldness organization but with an increased efficiency and compactness in functional segregation. The whole brain showed an overtight connection pattern. At the regional level, significantly increased nodal efficiency in the salience network (SN), default mode network (DMN) and executive control network was found. Social Anxiety Scale for Children (SASC) scores were positively correlated with these alterations. Spontaneous brain activity alterations in regions including the cerebellum, frontal gyrus (orbital part), temporal gyrus and thalamus were observed; some of these regions have been closely related to social anxiety and family or social support.Our study is the first to evaluate the neurological functional changes in school-age boys with severe HA. Disruptions in topographic characteristics and abnormal activity were closely related to social conditions. These data could help us to understand early neurological alterations in haemophilic children, improve the traditional view of family support and strengthen normal school life at an early stage.

Corynoline has been reported to have anti-inflammatory and antioxidative effects. In the present study, the potential protective effects of corynoline against zearalenone (ZEA)-induced liver injury were investigated. ZEA was administered daily for 5 days. Then, liver tissues were used for subsequent experiments. Corynoline attenuated liver histopathological changes induced by ZEA. The production of tumor necrosis factor-α and interleukin-1β in liver tissues, as well as aspartate aminotransferase and alanine aminotransferase in serum, was also inhibited by corynoline. Meanwhile, ZEA-induced MPO activity and MDA content were both attenuated by corynoline. ZEA-induced NF-κB p65 and IκBα phosphorylation were inhibited by corynoline. Furthermore, SIRT1, Nrf2, and HO-1 expression were increased by corynoline. In addition, the protective effects of corynoline against liver injury were reversed by the SIRT1 inhibitor EX-527. Taken together, corynoline protected against ZEA-induced liver injury by activating the SIRT1/Nrf2 signaling pathway.

To figure out the evolution of the heterogeneous microstructure across the molten pool boundaries (MPBs) and its effects on tensile properties of selective laser melted (SLM) AlSi10Mg alloy, two of the most commonly used scanning strategies, i.e., bidirectional scanning within a layer and with a 90° and 67° rotation for the successive layers (zigzag-90 and zigzag-67) were adopted. Grain morphology, solidification microstructure, texture and room temperature tensile properties of the SLMed AlSi10Mg alloy under the two scanning strategies were studied. The coarse columnar and fine equiaxed grains were observed in a single solidified track under the two scanning strategies. The columnar grains dominated the bulk of the molten pool (MP) while the equiaxed grains mainly distributed along the MPBs. The equiaxed grains along MPBs break the continuous growth of the columnar grains and lead to a weak texture. A new mechanism based on heterogeneous nucleation and constitutional supercooling was proposed to explain the formation of heterogeneous microstructure across the MPBs. The tensile test was performed in the horizontal (perpendicular to the build direction) and the vertical (parallel to the build direction) directions fabricated under the two scanning strategies. The mechanical properties showed obvious fluctuation, and the yield strength of the horizontal direction is generally higher than that of the vertical direction. The distribution of pores and MPBs is the main factors to influence the tensile properties.

Materials with unique structures can exhibit different properties and are widely studied in the preparation of new materials. Herein we reported a hydrothermal method to fabricate a layered nickel silicate/reduced graphene oxide (NiSiO/RGO) nanocomposite with an interesting dandelion-like structure. The morphology, composition, and electrochemical performance of RGO, NiSiO, and NiSiO/RGO were comparatively investigated in the current work. The results showed that the NiSiO/RGO nanocomposite has a dandelion-like hollow core–shell structure and shows good electrochemical performance. Compared with NiSiO, the original discharge capacity of NiSiO/RGO increased from 1291.6 mA h g−1 to 1653.9 mA h g−1; meanwhile, the reversible specific capacity of NiSiO/RGO increased from 649.6 mA h g−1 to 691.4 mA h g−1 after testing at a current density of 100 mA g−1 for 100 cycles. Moreover, the prepared NiSiO/RGO maintained a coulombic efficiency of about 97% after the initial charging and discharging cycle. This unique hollow dandelion-like structure enhanced the electrical conductivity and further resulted in lower diffusion resistance and higher reversible capacity. This work demonstrated that the layered NiSiO/RGO with an interesting dandelion-like structure can act as an alternative anode material for lithium-ion batteries.

Severe invasive aspergillosis infection occurs when human immune function is impaired. The interaction between Aspergillus fumigatus (A. fumigatus) conidia and type II lung epithelial cells serves an important role in disease progression. The present study compared the proteomes of A549 human lung epithelial cells with and without A. fumigatus infection. Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) and protein interaction analyses were performed, and differential protein expression was verified by western blotting and reverse transcription-quantitative PCR (RT-qPCR). Background Edible mushrooms have been used as food and medicine materials for thousands of years, and the yield of cultivatable edible mushrooms has increased in recent years. The increased consumption of edible mushrooms is not only due to their nutritional value, but also to their unique taste and specific flavor. As consumer awareness of food sensory qualities increases, umami taste and aroma have become important factors affecting consumer choices. There are many factors affecting umami taste and aroma of edible mushrooms, such as cultivation conditions, species, maturity, grading, parts of mushrooms, and processing and storage methods. However, the mechanisms underlying the variations in umami taste and aroma components of different mushrooms are still unclear. Scope and approach In this review, traditional umami components, novel umami peptides, and aroma compounds are discussed, as well as the perception of umami taste and aroma. Based on a combination of human sensory evaluation and instrumental analysis, changes in the composition of these components in edible mushrooms are summarized, and the metabolic pathways and biochemical reactions involved in these changes are also discussed. Key findings and conclusions The umami taste and aroma of edible mushrooms were closely related to nucleotide metabolism, amino acid metabolism, fatty acid metabolism, and the Maillard reaction. Umami peptides and the synergy between these compounds contribute to overall umami taste. There are differences in umami taste and aroma between cultivated and wild mushrooms. The selection of processing and storage techniques is, therefore, based on the established demand for umami taste and aroma of edible mushrooms.

Ras-associated autoimmune leukoproliferative disease (RALD) is a clinical syndrome, also known as autoimmune lymphoproliferative syndrome (ALPS) type IV, which is a primary immunodeficiency disease. However, there is still controversy about whether RALD is a chronic benign lymphoproliferative disease or a precancerous lesion. The clinical characteristics, laboratory examination results, treatment and prognosis of children with RALD treated in Beijing Children's Hospital were analyzed retrospectively. We reported 13 patients with RALD in our single center. The male-to-female ratio was 1.6:1, and the median age of onset was 6 months (range 2 months- 29 months). Eight patients (61.5%) had KRAS somatic mutations, and five patients (38.5%) had NRAS somatic mutations. The most common features were splenomegaly (12/13 cases), autoimmune cytopenia (9/13 cases), mononucleosis (10/13 cases), recurrent infection (7/13 cases), skin involvement (4/13 cases) and intestinal lesions (4/13 cases). Thirteen children were treated with immunotherapy, including glucocorticoids, intravenous immunoglobulin, cyclosporine, and sirolimus. The median follow-up time was 50 months (range 3 months-62 months). Four children died (2 cases of respiratory failure, 1 case of intestinal perforation and bleeding, and 1 case of sepsis with sudden respiratory and cardiac arrest). Nine children survived, seven children were treated with sirolimus, and no children developed malignant tumors of the hematopoietic system. In conclusion, RALD requires long-term follow-up to monitor malignant tumors, and sirolimus is effective for treatment.

BackgroundThe present study aimed to explore the application value of random survival forest (RSF) model and Cox model in predicting the progression-free survival (PFS) among patients with locoregionally advanced nasopharyngeal carcinoma (LANPC) after induction chemotherapy plus concurrent chemoradiotherapy (IC+CCRT).MethodsEligible LANPC patients underwent magnetic resonance imaging (MRI) scan before treatment were subjected to radiomics feature extraction. Radiomics and clinical features of patients in the training cohort were subjected to RSF analysis to predict PFS and were tested in the testing cohort. The performance of an RSF model with clinical and radiologic predictors was assessed with the area under the receiver operating characteristic (ROC) curve (AUC) and Delong test and compared with Cox models based on clinical and radiologic parameters. Further, the Kaplan-Meier method was used for risk stratification of patients.ResultsA total of 294 LANPC patients (206 in the training cohort; 88 in the testing cohort) were enrolled and underwent magnetic resonance imaging (MRI) scans before treatment. The AUC value of the clinical Cox model, radiomics Cox model, clinical + radiomics Cox model, and clinical + radiomics RSF model in predicting 3- and 5-year PFS for LANPC patients was [0.545 vs 0.648 vs 0.648 vs 0.899 (training cohort), and 0.566 vs 0.736 vs 0.73 vs 0.861 (testing cohort); 0.556 vs 0.604 vs 0.611 vs 0.897 (training cohort), and 0.591 vs 0.661 vs 0.676 vs 0.847 (testing cohort), respectively]. Delong test showed that the RSF model and the other three Cox models were statistically significant, and the RSF model markedly improved prediction performance (P<0.001). Additionally, the PFS of the high-risk group was lower than that of the low-risk group in the RSF model (P<0.001), while comparable in the Cox model (P>0.05).ConclusionThe RSF model may be a potential tool for prognostic prediction and risk stratification of LANPC patients.

Temperature is one of the most important factors for drying edible mushrooms. To evaluate the effects of different hot-air drying (HAD) temperatures on the umami taste and aroma profile of Suillus granulatus (S. granulatus) mushrooms, we measured umami substances and volatile compounds of S. granulatus dried at 40°C, 50°C, 60°C, 70°C, and 80°C. Results showed that when dried at 60°C, S. granulatus exhibited significantly higher (p 0.05) equivalent umami concentration, taste activity values of glutamic acid (Glu) and 5'-guanosine monophosphate (5'-GMP), and electronic tongue umami sensory scores. The results identified a total of 71 volatile components of which geranylacetone, benzaldehyde, phenylethyl alcohol, and 3-methylbutanoic acid were the dominant compounds. Sensory evaluation and relative odor activity values (ROAVs) revealed that 16 volatile compounds were the key volatile organic compounds contributing mushroom-like and sweet odor to the overall aroma of S. granulatus; these included 1-octen-3-ol (ROAV: 15.11-62.06) and ethyl phenylacetate (ROAV: 13.62-79.11). The drying temperature changed the aroma profile of S. granulatus. Furthermore, the mushroom dried at 60°C had a more desirable mushroom-like and almond odor. It was, therefore, proposed that HAD at 60°C was optimal for retaining a pleasant flavor in S. granulatus. This study provides a theoretical basis for the optimal drying condition selection for the mushroom processing industry. PRACTICAL APPLICATION: Hot-air drying at 60°C can significantly retain the flavor of S. granulatus and is an optimal temperature for mushroom drying.

Pretreatment individualized assessment of tumor response to induction chemotherapy (ICT) is a need in locoregionally advanced nasopharyngeal carcinoma (LANPC). Imaging method plays vital role in tumor response assessment. However, powerful imaging method for ICT response prediction in LANPC is insufficient.To establish a robust model for predicting response to ICT in LANPC by comparing the performance of back propagation neural network (BPNN) model with logistic regression model.Retrospective.A total of 286 LANPC patients were assigned to training (N = 200, 43.8 ± 10.9 years, 152 male) and testing (N = 86, 43.5 ± 11.3 years, 57 male) cohorts.TPredictive clinical factors were selected by univariate and multivariate logistic models. Radiomic features were screened by interclass correlation coefficient, single-factor analysis, and the least absolute shrinkage selection operator (LASSO). Four models based on clinical factors (ModelStudent's t-test, Mann-Whitney U-test, and Chi-square test or Fisher's exact test were used for comparison analysis. The performance of models was assessed by area under the receiver operating characteristic (ROC) curve (AUC) and Delong test. P 0.05 was considered statistical significance.Three significant clinical factors: Epstein-Barr virus-DNA (odds ratio [OR] = 1.748; 95% confidence interval [CI], 0.969-3.171), sex (OR = 2.883; 95% CI, 1.364-6.745), and T stage (OR = 1.853; 95% CI, 1.201-3.052) were identified via univariate and multivariate logistic models. Twenty-four radiomics features were associated with treatment response. In this study, a rapid, low-cost and facile method for detecting exosomes was developed by engineering DNA ligands on the surface of an iron-based metal-organic framework (Fe-MOF). Aptamers of exosomal transmembrane CD63 protein (CD63-aptamers) were utilized as both the optically active layer and the exosome-specific recognition element to engineer an Fe-MOF bio-interface for high-efficiency regulation of the catalytic behavior of Fe-MOF toward the chromogenic substrate. The effective enhancement of the intrinsic peroxidase-like catalytic activity was confirmed via the self-assembly of CD63-aptamers on the surface of Fe-MOF. The specific binding of exosomes with CD63-aptamers altered the conformation of DNA ligands on the surface of Fe-MOF, contributing to sensitive variation in Fe-MOF catalytic activity. This directly produced a distinct color change and enabled the visual detection of exosomes. Via one-step "mixing-and-detection", the Fe-MOF bio-interface exhibited excellent performance in quantitative analysis of exosomes derived from human breast cancer cell lines ranging from 1.1 × 10

The progression of disease caused by fungal infection is closely associated with the human immune system. Macrophages and natural killer cells (NK cells) are two important types of innate immune cells that serve an important role in anti‑infection immunity. There has been limited research into the interactions between fungi and macrophages. In the present in vitro study, reverse transcription‑quantitative PCR, ELISA and flow cytometry were performed to reveal that the interaction between macrophages and NK cells, regulated by Aspergillus fumigatus conidia, induced macrophages to polarize into M1 macrophages by secreting large quantities of tumor necrosis factor‑α, interleukin‑18 and Galectin‑9. In addition, when NK cells were co‑cultured with the conidia of A. fumigatus‑stimulated M1 macrophages, they exhibited increased activation levels and secretion of interferon‑γ (IFN‑γ). It was further demonstrated via antibody neutralization and gene silencing experiments that galectin‑9 served an important role in the interaction between macrophages and NK cells regulated by A. fumigatus. In conclusion, it was demonstrated that A. fumigatus induced the polarization of macrophages into M1 macrophages by secreting Galectin‑9, which then promoted NK cell activity and IFN‑γ secretion. The results provided improved understanding of the role of innate immune cells in invasive fungal infections. The present study also provided novel insight into the study of macrophages and NK cells in inflammatory infections caused by A. fumigatus and potential strategies to control the progression of inflammation.

Umami and aroma are important flavor qualities of edible mushrooms, and packaging can maintain or improve the flavor during storage. This study explored the effects of light-proof packaging (LPP), light-transparent packaging (LTP), vacuum light-proof packaging (VLPP), and vacuum light-transparent packaging (VLTP) on umami taste and aroma of dried Suillus granulatus. Monosodium glutamate-like amino acid content, equivalent umami concentration, and electronic tongue umami sensory scores in VLTP were higher at 2, 4, and 6 months and higher in LTP at 8 and 10 months. Principal component analysis of aroma components showed that the comprehensive scores were higher for the transparent packaging. Ketones and pyrazines were more abundant in transparent packaging. Flavor quality was better at 4–6 months, based on the equivalent umami concentration and the concentration of eight-carbon compounds that contribute to aroma. Transparent packaging is a promising way to optimize the flavor of dried Suillus granulatus.

Objective To analyze the effects of escalating treatment strategy in children with severe chronic immune thrombocytopenia (SCITP). Methods This was a single-center, retrospective cohort study. Data from children with SCITP who received escalating treatment strategy in our center were collected between June 2017 and August 2019. The escalating strategy included three steps: Step I (six courses of high-dose dexamethasone [HDD]), Step II (HDD combined with low-dose rituximab), and Step III (eltrombopag). Results A total of 30 cases (18 males and 12 females) were included, with duration of immune thrombocytopenia (ITP) of 20.5 (12.0-96.0) months. After treatment, the remission rate was 36.7% (11/30) and the sustained response (SR) rate was 68.2% (15/22). The distribution (remission rates) from Step I to III was as follows: nine of 30 (33.3%, 3/9); four of 30 (50%, 2/4); 17/30 (29.4%, 5/17), respectively. In eltrombopag (Step III) cases, 47.5% (8/17) maintained a platelet count of ≥50 × 109 /L, 37.5% (3/8) had dose tapering, and 25% (2/8) have successfully discontinued the medication. The number of patients at 12, 24, and 36 months were 30, seven, and two, with a total response and remission rates of 80% (36.7%), 57.1% (28.6%), and 50% (50%), respectively. The total relapse rate was 26.7% (8/30), and three cases from Step II and five cases from Step III. Conclusion The escalating strategy for children SCITP showed an effective improvement rate with 36.7% remission and 68.2% SR, and 30% could benefit and retain SR from HDD treatment. Combined treatment with eltrombopag can reduce the relapse rate of low-dose rituximab.

Objective: To analyze the effect of a novel second-line escalating treatment strategy (high-dose dexamethasone (HDD), low-dose rituximab to eltrombopag) for children with severe chronic immune thrombocytopenia (SCITP). Materials and Methods: This study was a single-center, retrospective cohort study. The second-line escalating strategy included 3 steps: Step I (6 courses high-dose dexamethasone: HDD), Step II (HDD combined with low-dose rituximab), and Step III (eltrombopag). Results: A total of 30 cases (18 males and 12 females) were included; the median age was 8.83 (1.42-13.9) year-old, the duration time of ITP was 20.5 (12.0-96.0) months, and the platelet counts were 15 (3-29) ×109/L. After the median 14 (12-37) months’ treatment, the remission rate was 36.7% (11/30), and the sustained response (SR) rate was 68.2% (15/22). In eltrombopag (step III) cases, 47.5% (8/17) maintained platelet ≥50×109/L, 37.5% (3/8) dose tapering, and 25% (2/8) were successfully discontinued from medication. The number of patients at 12th, 24th, and 36th months was 30, 7, and 2, with the total response (TR) and remission rates of 80% (36.7%), 57.1% (28.6%), and 50% (50%), respectively. The total relapse rate was 26.7% (8/30)，three cases(75%, 3/4)from Step II and 5 cases (41.7% ,5/12)from Step III, none in Step I. Conclusion: The new second-line escalating strategy for children SCITP has an effective improving rate with 36.7% remission and 68.2% SR; 30% could benefit and retain stable response from HDD treatment. Combined treatment with eltrombopag can reduce the relapse rate of low-dose rituximab.

Based on the results of rate-controlled mercury-injection experiments, the microscopic pore-throat structure characteristics of tight sandstone in Sha-1 Section and tight limestone in Da’anzhai Section of Sichuan Basin were quantitatively characterized. The results show that the pore radius distribution characteristics of tight oil reservoirs are similar. The main distribution is between 100~190 μm, and the average pore radius is 160 μm. While the distribution of the throat radius of tight sandstone and limestone is quite different, the distribution of the throat of sandstone samples is relatively concentrated, and the distribution of the throat of limestone samples is relatively sparse. There is a good positive correlation between the average throat radius and permeability, but the correlation between fractal dimension and permeability is not obvious. This indicates that the permeability is mainly affected by the radius of the throat. The pore-throat ratio in tight oil reservoirs is relatively large, and the resistance to seepage is greater during development. Therefore, during the development of tight oil, measures should be taken to increase the radius of the throat, reduce the ratio of pore radius to pore-throat radius, and improve the seepage capacity of the reservoir, thereby improving the development of tight oil.

This work is concerned with a stochastic wave equation driven by a white noise. Borrowing from the invariant random cone and employing the backward solvability argument, this wave system is approximated by a finite dimensional wave equation with a white noise. Especially, the finite dimension is explicit, accurate and determined by the coefficient of this wave system; and further originating from an Ornstein-Uhlenbek process and applying Banach space norm estimation, this wave system is approximated by a finite dimensional wave equation with a smooth colored noise.

The effects of short-time partial dehydration (STPD) on quality of shiitake mushroom were evaluated. Quality properties included respiration, texture, color and umami taste. Results showed that STPD treatment (STPD-2 and STPD-3) can slow down respiration rate and delay respiration peak, thus providing more than 7 days of storage time stored at 20 ± 1 °C. Mushroom subjected to STPD-2 and STPD-3 treatments also maintained better firmness. The STPD-2 treatment effectively delayed the browning of fresh mushroom. Compared to other groups, the content of flavor 5′-nucleotides and total 5′-nucleotides in STPD-2 and STPD-3 samples were higher, and STPD-2 sample had higher total amino acid content during storage. Higher equivalent umami concentration (EUC) values were found in STPD-2 sample (17.58 – 38.69 g MSG 100 g − 1) compared to STPD-3 sample (13.80 – 27.51 g MSG 100 g − 1) and STPD-1 sample (5.11 – 14.19 g MSG 100 g − 1). Thus, it indicated that STPD-2 treatment could effectively preserve monosodium glutamate-like (MSG-like) components and nucleotides in mushroom. Results suggested that STPD treatment, especial STPD-2, could maintain mushroom overall quality.

Developing a facile and reliable approach for tumor marker detection is crucial in early diagnosis of cancer. Here, a multicolor and photothermal dual signal readout immunosensor was proposed based on a nanoparticle-mediated transformation strategy and used for sensitive detection of prostate specific antigen (PSA). To construct such a platform, Fe3O4 nanoparticles (NPs) -functionalized graphene oxide was modified with partially complementary DNA and served as the signal probes. In the absence of PSA, the signal probes were captured via hybridization reaction between DNA and PSA aptamer. Subsequently, Fe3O4 NPs anchored on the probes were transformed into a photothermal agent, Prussian blue NPs, which converted biological signal into heat via the near-infrared laser irradiation, and thus sensitive quantitative detection of PSA was realized by using a thermometer with a lower detection limit of 0.31 ng/mL. Meanwhile, Prussian blue NPs displayed multiply colors by mixing with potassium ferricyanide, and rapid qualitative detection by naked eyes was realized. The two sets of data mutually validate each other, which greatly improved the accuracy and reliability of PSA detection. More strikingly, graphene oxide as the enrichment carrier of Fe3O4 NPs significantly enhanced the accumulation of Prussian blue NPs in situ, and thus the signal amplification was effectively accomplished. Besides, the sensing strategy avoided the use of enzyme and simplified analysis process. Such a cost-effective and sensitive dual-readout visual protocol opens up new perspectives for personalized diagnosis and on-site detection.

This study aimed to identify preoperative MR imaging features for predicting early recurrence after curative resection of solitary hepatocellular carcinoma (HCC) without microvascular invasion (MVI).124 patients with MVI-negative HCC who underwent preoperative dynamic contrast-enhanced 1.5-T MR imaging before surgical resection were included. Liver Imaging Reporting and Data System (LI-RADS v2018) imaging features and three non-LI-RADS MR imaging features for predicting early recurrence (intrahepatic recurrence2 years) were identified by univariable and multivariable analyses. A nomogram was constructed for individualized risk estimation, and its predictive accuracy and discriminative ability were identified by concordance index (C-index) and calibration curve.In multivariable analysis, tumor size (p = 0.045), nonsmooth tumor margin (p = 0.013), and presence of mosaic architecture (p = 0.035) were independent significant variables associated with early recurrence. These were all incorporated to establish the nomogram. The C-index of the nomogram was 0.743 (95 % CI: 0.697-0.788).At dynamic contrast-enhanced MR imaging, tumor size, nonsmooth tumor margin, and presence of mosaic architecture may be helpful to predict early recurrence of solitary HCC without MVI after curative resection.

Ni2SiO4, as a new anode material for lithium–ion batteries, was prepared by the high-temperature calcination method in this work. The MgO-coated NSO was prepared by melt injection method. Electrochemical properties, including voltammogram (CV), electrochemical impedance spectroscopy (EIS), charge/discharge curves and cycle performance were tested. The structure and morphology of materials were further characterized by XRD and SEM. The results demonstrated that the MgO-coated Ni2SiO4 materials exhibited higher cycle charge capacity and coulombic efficiency than that of Ni2SiO4. When the MgO coating amount is 1%, the first cycle charge capacity and coulombic efficiency were 584.2 mAh/g and 66.25%, respectively. After 50 cycles, the charge capacity was still maintained at 359.7 mAh/g when the current density was 100 mAh/g, which was 162.7 mAh/g higher than the NSO. The crystal structure of the materials belongs to an orthorhombic system, and the morphological structure presented cubic particles. Therefore, the NSO anode material has a better cycle stability and high capacity when the MgO coating amount is 1%.

Detection of telomerase activity is crucial for the telomerase-related early diagnosis of cancer and drug screening. Herein, a multicolor visual telomerase detection method with high sensitivity was developed based on the etching of hexadecyl trimethyl ammonium bromide-stablized Au nanorods (Au NRs) by the oxidation state 3,3',5,5'-tetramethylbenzidine sulfate (TMB2+). In order to meet the demand of bare-eye inspection, an enzyme-free signal amplification strategy of catalytic hairpin assembly (CHA) was incorporated. After the introduction of telomerase, telomerase extension products specifically triggered cyclic CHA and led to the successive formation of G-quadruplex/hemin DNAzyme, which catalyzed the H2O2-mediated oxidation of TMB. The Au NRs were gradually etched as the concentration of catalysate TMB2+ increased, resulting in the decrease of aspect ratio of the Au NRs. Correspondingly, the longitudinal localized surface plasmon resonance peak of Au NRs was blue-shifted, with the concomitant generation of a series of color transition. Under optimal conditions, a highly sensitive detection toward telomerase was realized down to 15 HeLa cells. Compared to previous colorimetric method for telomerase determination, multiple colors corresponding to telomerase activity was the most attractive virtue of our approach. More strikingly, telomerase-induced cyclic CHA significantly enhanced the accumulation of G-quadruplex/hemin DNAzyme, thus the signal amplification was effectively realized. Our approach exhibited excellent sensitivity and convenient signal readout, which is expected to provide great potential application in cancer diagnosis and therapy.

In order to research on the physical characteristics of tight oil reservoirs, pulse permeameter is adopted to measure the permeability and porosity of the tight rock cores, which proved that permeability acquired via pulse method being approximate to Klinkenberg-corrected permeability value, and rock porosity measured by gas is consistent to that measured by water. To represent the development potential of tight oil reservoirs, centrifugal and nuclear magnetic resonance (NMR) experiments are applied to determine the movable fluids inside the reservoirs, and the premium centrifugal force to measure T2 cutoff value of tight reservoirs is 417 psi (1 psi = 0.006895 MPa), with T2 spectrum of tight cores being classified and explained. The comparison of the saturation and pressure fields during two decades based on the nonlinear numerical software in the cases of depletion, water injection, and gas injection were analyzed which indicated that the method of gas injection and producing via horizontal well was optimal because of its highest recovery. Based on the characteristics of tight oil, this assay carried out the exploration of new testing methods, which provides basis for the effective production of tight oil and offers thoughts and suggestions for future analysis of tight oil reservoirs.

Thrombotic storm (TS) is a rare disease, especially with thrombus in the heart of pediatric patient. We present a case of a 4-year-old boy, who was diagnosed with TS during his first hospitalization due to lower extremity deep venous thrombosis, pulmonary embolism, and thrombosis of the inferior vena cava, cerebral, left internal jugular, portal, renal, and iliac veins. He was eventually prescribed with rivaroxaban to control thrombosis after 30 days of successive use of low-molecular-weight heparin, unfractionated heparin, and warfarin, which were demonstrating little effect on preventing thrombosis, and the patient was intolerant to argatroban. While his lupus anticoagulant ratio was slightly above the normal range and no other potential causes such as congenital thrombophilia, severe infection, malignancy, and trauma were confirmed, we suspected antiphospholipid antibody syndrome and prescribed glucocorticoid and rituximab to control the disease. After 36 days of admission, ultrasonography showed recanalization of the former thrombus. One month after discharge, a tumor embolus resembling a mass emerged in his right atrium under effective anticoagulant therapy. During his second admission, he underwent surgical thrombectomy, and pathological examination confirmed the mass to be a platelet-rich thrombus rather than tumor embolus or infection. Considering the suspected antiphospholipid antibody syndrome as the cause of the TS, we prescribed aspirin combined with rivaroxaban to prevent thrombosis. In this case, surgery and pathology shed light on the type of thrombus that emerged from the inferior vena cava and traveled to the heart, which is the possible potential cause of TS. It also changed our therapeutic strategy to antiplatelet therapy combined with anticoagulant therapy to control the disease.
