3. Analysis of Legacy and Novel Neutral Per- and Polyfluoroalkyl Substances in Soils from an Industrial Manufacturing Facility

Abstract

Per- and polyfluoroalkyl substances (PFASs) have been detected in an array of environmental media due to their ubiquitous use in industrial and consumer products as well as potential release from fluorochemical manufacturing facilities. During their manufacture, many fluorotelomer (FT) facilities rely on neutral intermediates in polymer production including the FT-alcohols (FTOHs). These PFAS are known to transform to the terminal acids (perfluoro carboxylic acids; PFCAs) at rates that vary with environmental conditions. In the current study on soils from a FT facility, we employed gas chromatography coupled with conventional- and high-resolution mass spectrometry (GC–MS and GC-HRMS) to investigate the profile of these precursor compounds, the intermediary secondary alcohols (sFTOHs), FT-acrylates (FTAcr), and FT-acetates (FTAce) in soils around the former FT-production facility. Of these precursors, the general trend in detection intensity was [FTOHs] > [sFTOHs] > [FTAcrs], while for the FTOHs, homologue intensities generally were [12:2 FTOH] > [14:2 FTOH] > [16:2 FTOH] > [10:2 FTOH] > [18:2 FTOH] > [20:2 FTOH] > [8:2 FTOH] ∼ [6:2 FTOH]. The corresponding terminal acids were also detected in all soil samples and positively correlated with the precursor concentrations. GC-HRMS confirmed the presence of industrial manufacturing byproducts such as FT-ethers and FT-esters and aided in the tentative identification of previously unreported dimers and other compounds. The application of GC-HRMS to the measurement and identification of precursor PFAS is in its infancy, but the methodologies described here will help refine its use in tentatively identifying these compounds in the environment.

9. Matrix-induced defects and molecular doping in the afterglow of SiO2 microparticles

Abstract

Abstract A deep understanding of how the host matrix influences the afterglow properties of molecule dopants is crucial for designing advanced afterglow materials. Despite its appeal, the impact of defects on the afterglow performance in molecule-doped SiO2 matrices has remained largely unexplored. Herein, we detail the synthesis of monodisperse SiO2 microparticles by hydrothermally doping molecules, such as 4-phenylpyridine, 4,4’-bipyridine, and 1,4-bis(pyrid-4-yl)benzene. Our results demonstrate that hydrothermal reactions induce not only the formation of emissive defects in the SiO2 matrix but also enable molecule doping through SiO2 pseudomorphic transformation. Optical analyses reveal a remarkable afterglow activation of doped molecules, driven by a synergistic interplay of hydrogen bonding and physical fixation. Specifically, 4-phenylpyridine doping leads to an impressive 227- and 271-fold enhancement in fluorescence and afterglow, respectively, and an extraordinary 3711-fold enhancement in the afterglow lifetime of the resulting SiO2 MPs. We also document hybrid states involving molecule dopants and SiO2 defects, explaining energy transfer from molecule dopants to defects in both singlet and triplet states. The robust achievement of molecule doping provides flexibility to tailor excitation-dependent afterglow attributes while preserving angle-dependent structural colors, facilitating the creation of diverse building blocks for multiscale optical platforms for afterglow modulation and information encoding.

14. Gly-β-MCA is a potent anti-cholestasis agent against 'human-like' hydrophobic bile acid-induced biliary injury in mice

Abstract

Cholestasis is a chronic liver disease with limited therapeutic options. Hydrophobic bile acid-induced hepatobiliary injury is a major pathological driver of cholestasis progression. This study investigates the anti-cholestasis efficacy and mechanisms of action of glycine-conjugated β-muricholic acid (Gly-β-MCA). We use female Cyp2c70 KO mice, a rodent cholestasis model that does not produce endogenous muricholic acid (MCA) and exhibits a “human-like” hydrophobic bile acid pool and female-dominant progressive hepatobiliary injury and portal fibrosis. The efficacy of Gly-β-MCA and ursodeoxycholic acid (UDCA), the first line drug for cholestasis, on cholangiopathy and portal fibrosis are compared. At a clinically relevant dose, Gly-β-MCA shows comparable efficacy as UDCA in reducing serum transaminase, portal inflammation and ductular reaction, and better efficacy than UDCA against portal fibrosis. Unlike endogenous bile acids, orally administered Gly-β-MCA is absorbed at low efficiency in the gut and enters the enterohepatic circulation mainly after microbiome-mediated deconjugation, which leads to taurine-conjugated MCA enrichment in bile that alters enterohepatic bile acid pool composition and reduces bile acid pool hydrophobicity. Gly-β-MCA also promotes fecal excretion of endogenous hydrophobic bile acids and decreases total bile acid pool size, while UDCA treatment does not alter total bile acid pool. Furthermore, Gly-β-MCA treatment leads to gut unconjugated MCA enrichment and reduces gut hydrophobic lithocholic acid (LCA) exposure. In contrast, UDCA treatment drives a marked increase of LCA flux through the large intestine. In conclusion, Gly-β-MCA is a potent anti-cholestasis agent with potential clinical application in treating human cholestasis.

Abstract

The phenomenon of low initial growth rates in atomic layer deposition (ALD) of various oxides on HF-treated substrates can be effectively avoided by use of OH-terminated SI(001) substrates. The effect of reducing the incubation period is confirmed in the ALD process of aluminum oxide thin films in which trimethylaluminum and water are used as sources of aluminum and oxygen.

21. COVID-19 and the Impact on Children's Mental Health

Abstract

The COVID-19 public health crisis has created significant challenges for children in British Columbia. These challenges have included most children facing restrictions in their contacts with family members and friends, as well as temporary school closures. Many children are also part of families that have experienced economic hardships. Beyond the social, educational and economic costs, there will also be mental health consequences. This rapid review therefore aimed to determine how the pandemic and its associated challenges may affect the mental health of BC’s children, including those who may be disproportionately harmed. The overarching goal was to inform and assist policymakers to support all children in BC during COVID-19 — and beyond. Our systematic review identified one relevant original study on the mental health consequences of previous pandemics and five systematic reviews on the mental health consequences of natural disasters for children. The findings showed dramatic increases in rates of anxiety, posttraumatic stress, depression and behavioural challenges compared to rates typically found in the general population of children. Other literature suggests that some groups may also be disproportionately affected, including children from socioeconomically disadvantaged families and those who have faced extreme or cumulative adversities. Racism may contribute to Asian-Canadian children facing added hardships. Indigenous children may also be particularly disadvantaged given the cumulative adversities associated with the legacies of colonialism. As well, children with neuro-diverse special needs such as autism spectrum disorder, fetal alcohol spectrum disorder, developmental delays or other disabilities may have greater mental health needs during the pandemic. On balance, the available research evidence suggests that BC’s response to the COVID-19 pandemic must make children’s mental health a high priority — ensuring that children do not experience additional avoidable adversities due to either the pandemic or the public health responses. Central to these objectives will be: providing additional necessary prevention and treatment services; ensuring that public investments go towards effective interventions; preventing avoidable childhood adversities including reducing socioeconomic disparities; and tracking child outcomes so that all British Columbians can see the progress. One GWAS usedgenotypesfrom 5895 whole-genome sequences and the other GWAS used genotyping array data from an additional 5849 individuals.Cross-validationof loci reaching genome-wide significance was sought by meta-analysis. Conditional analysis corrected for the most significant variants at each locus was used to resolve signals for multiple associations. We functionally annotated associated variants and tested associations with duration ofsurvival.All-cause mortalitywas the primary endpoint in survival analyses. Abstract

Background: In the medical literature, the nomenclature and descriptions (ND) of small bowel (SB) ulcerative and inflammatory (U-I) lesions in capsule endoscopy (CE) are scarce and inconsistent. Inter-observer variability in interpreting these findings remains a major limitation in the assessment of the severity of mucosal lesions, which can impact negatively on clinical care, training and research on SB-CE. Objective: Focusing on SB-CE in Crohn’s disease (CD), our aim is to establish a consensus on the ND of U-I lesions. Methods: An international panel of experienced SB-CE readers was formed during the 2016 United European Gastroenterology Week meeting. A core group of five CE and inflammatory bowel disease (IBD) experts established an Internet-based, three-round Delphi consensus but did not participate in the voting process. The core group built illustrated questionnaires, including SB-CE still frames of U-I lesions from patients with documented CD. Twenty-seven other experts were asked to rate and comment on the different proposals for the ND of the most frequent SB U-I lesions. For each round, we used a 6-point rating scale (varying from ‘strongly disagree’ to ‘strongly agree’). The consensus was reached when at least 80 % of the voting members scored the statement within the ‘agree’ or ‘strongly agree’ categories. Results: A 100% participation rate was obtained for all the rounds. Consensual ND were reached for the following seven U-I lesions: aphthoid erosion, deep ulceration, superficial ulceration, stenosis, edema, hyperemia and denudation. Conclusion: Considering the most frequent SB U-I lesions seen in CE in CD, a consensual ND was reached by the international group of experts. These descriptions and names are useful not only for daily practice and medical education, but also for medical research.

Abstract

Abstract The advancement of contemporary X-ray imaging heavily depends on discovering scintillators that possess high sensitivity, robust stability, low toxicity, and a uniform size distribution. Despite significant progress in this field, the discovery of a material that satisfies all of these criteria remains a challenge. In this study, we report the synthesis of monodisperse copper(I)-iodide cluster microcubes as a new class of X-ray scintillators. The as-prepared microcubes exhibit remarkable sensitivity to X-rays and exceptional stability under moisture and X-ray exposure. The uniform size distribution and high scintillation performance of the copper(I)-iodide cluster microcubes make them suitable for the fabrication of large-area, flexible scintillating films for X-ray imaging applications in both static and dynamic settings.

Abstract

Background and aims: Several bile acids-based monotherapies have been developed for non-alcoholic steatohepatitis (NASH) treatment but clinical trial findings suggest that they do not satisfactorily improve NASH and liver fibrosis in many patients. Recently, we have shown that combining a gut-restricted apical sodium-bile acid transporter (ASBT) inhibitor GSK2330672 (GSK) with adeno-associated virus (AAV)-mediated liver fibroblast growth factor 15 (FGF15) overexpression provides significantly improved efficacy than either single treatment against NASH and liver fibrosis in a high fat, cholesterol, and fructose (HFCFr) diet-induced NASH mouse model. The beneficial effects of the combined treatment can be attributed to the markedly reduced bile acid pool that reduces liver bile acid burden and intestinal lipid absorption. The aim of this study is to further investigate if combining GSK treatment with the orally bioavailable obeticholic acid (OCA), which induces endogenous FGF15 and inhibits hepatic bile acid synthesis, can achieve similar anti-NASH effect as the GSK + AAV-FGF15 co-treatment in HFCFr-diet-fed mice. Materials and methods: Male C57BL/6J mice were fed HFCFr diet to induce NASH and liver fibrosis. The effect of GSK, OCA, and GSK + OCA treatments on NASH development was compared and contrasted among all groups. Results: Findings from this study showed that the GSK + OCA co-treatment did not cause persistent reduction of obesity over a 12-week treatment period. Neither single treatment nor the GSK + OCA co-treatment reduce hepatic steatosis, but all three treatments reduced hepatic inflammatory cytokines and fibrosis by a similar magnitude. The GSK + OCA co-treatment caused a higher degree of total bile acid pool reduction (∼55%) than either GSK or OCA treatment alone. However, such bile acid pool reduction was insufficient to cause increased fecal lipid loss. The GSK + OCA co-treatment prevented GSK-mediated induction of hepatic cholesterol 7alpha-hydroxylase but failed to induce ileal FGF15 expression. GSK did not reduce gallbladder OCA amount in the GSK + OCA group compared to the OCA group, suggesting that ASBT inhibition does not reduce hepatic OCA distribution. Conclusions: Unlike the GSK + AAV-FGF15 co-treatment, the GSK + OCA co-treatment does not provide improved efficacy against NASH and liver fibrosis than either single treatment in mice. The lack of synergistic effect may be partly attributed to the moderate reduction of total bile acid pool and the lack of high level of FGF15 exposure as seen in the GSK + AAV-FGF15 co-treatment.

Abstract

The incidence of small bowel tumors is increasing over time. Until recently, their diagnosis was delayed and it was often reached only at the time of surgery. New diagnostic tools, such as capsule endoscopy, device-assisted enteroscopy, and dedicated small bowel cross-sectional imaging techniques, have been introduced recently in clinical practice. The combination of these tools allows medical practitioners to detect small bowel tumors at an early stage and to reach a definite diagnosis before surgery, thus enabling minimally invasive treatments.

Abstract

The phenomenon of low initial growth rates in atomic layer deposition (ALD) of various oxides on HF-treated (thus H-terminated) Si(001) substrates, which is termed the incubation effect or incubation period, can be effectively avoided by use of OH-terminated Si(001) substrates. Two ways of preparing OH-terminated Si(001) were devised in this work: one from atomically clean Si(001) and the other from H-terminated Si(001). The effect of reducing the incubation period was confirmed in the ALD process of aluminum oxide (Al2O3) thin films in which trimethylaluminum and water were used as sources of aluminum and oxygen, respectively. The use of OH-terminated Si(001) substrates has another beneficial effect of producing thin films with very smooth surface morphology. We propose the use of OH-terminated Si(001) substrates for growing thin films of many other metal oxides that have shown the incubation period on H-terminated Si(001) substrates.

Abstract

Therapeutic reduction of hydrophobic bile acids exposure is considered beneficial in cholestasis. The Cyp2c70 KO mice lack hydrophilic muricholic acids and have a human-like hydrophobic bile acid pool resulting in hepatobiliary injury. This study investigates if combining an apical sodium-dependent bile acid transporter inhibitor GSK2330672 (GSK) and fibroblast growth factor-15 (FGF15) overexpression, via simultaneous inhibition of bile acid synthesis and gut bile acid uptake, achieves enhanced therapeutic efficacy in alleviating hepatobiliary injury in Cyp2c70 KO mice. The effects of GSK, adeno-associated virus (AAV)-FGF15, and the combined treatment on bile acid metabolism and cholangiopathy were compared in Cyp2c70 KO mice. In female Cyp2c70 KO mice with more severe cholangiopathy than male Cyp2c70 KO mice, the combined treatment was more effective in reversing portal inflammation, ductular reaction, and fibrosis than AAV-FGF15, while GSK was largely ineffective. The combined treatment reduced bile acid pool by ∼80% compared to ∼50% reduction by GSK or AAV-FGF15, and enriched tauro-conjugated ursodeoxycholic acid in the bile. Interestingly, the male Cyp2c70 KO mice treated with AAV-FGF15 or GSK showed attenuated cholangiopathy and portal fibrosis but the combined treatment was ineffective despite reducing bile acid pool. Both male and female Cyp2c70 KO mice showed impaired gut barrier integrity. AAV-FGF15 and the combined treatment, but not GSK, reduced gut exposure to lithocholic acid and improved gut barrier function. In conclusion, the combined treatment improved therapeutic efficacy against cholangiopathy than either single treatment in the female but not male Cyp2c70 KO mice by reducing bile acid pool size and hydrophobicity.

One of the key technological events of the 1980's in papermaking was the development and implementation of the soft nip calendering, on or off-line, The technology found many applications and is gaining acceptance around the world. When some of the present technical problems are resolved, it may become a standard feature of all paper machines.As the CD profile control technology continues to improve, the primary role of the calender has been shifting from the traditional reel building tool to that of surface finishing. During the second half of the 1980's, we examined the technology for various paper and paperboard operations. In this paper, I would like to share with you some of our findings from pilot trials and laboratory evaluations.High temperature soft nip calender (HTSNC) has a nip that is hot, long and of low specific pressure. It works primarily on the small scale roughness of the paper surface. This may or may not be of practical value depending on the paper grades and their end-use requirements. Benefits from new technology can never be scrutinized too much.

We studied the contribution of image parameters to visualization quality and their effect on diagnostic certainty of small bowel (SB) lesions.Methods: Five clear CE images of common SB pathology – two vascular lesions, two inflammatory, one polyp – were processed for three image parameters to simulate poor SB conditions: opacity (color-matched to luminal content; 10– 90%, 10% increments); blurriness (radius 1– 10 pixels; one pixel increments); and contrast (− 50-50%; 10% increments). Nine expert readers evaluated whether images were adequate for diagnosis. Points where perception of image quality changed significantly were determined for each parameter. Three further sets of SBCE images (vascular, inflammatory, and neoplastic lesions; nine images/set) were processed for four points/parameters. Twenty experienced/expert CE readers reviewed these images.Results: The negative effects of opacity in diagnostic certainty were mostly evident in images of neoplasia; images of vascular and inflammatory lesions were less affected. Similar results were observed with increasing blur radius, simulating movement, and poor focus. The proportions of readers finding vascular and inflammatory images adequate for diagnosis did not drop significantly at wider blur radii, while images of neoplasia were quickly deemed inadequate. Low contrast had a greater negative effect than high, most consistently in neoplastic lesions.Conclusion: Poor visualization quality in all parameters affected mostly neoplastic lesions. Software to increase contrast and sharpen images can improve visualization quality; smart frame rate adaptation could improve the number of high-quality frames obtained.
