Abstract Autophagy is a highly conserved eukaryotic pathway and plays a crucial role in cell survival under stress conditions. Here, we applied a full-length transcriptome approach to study an Arabidopsis autophagy mutant (atg5-1) subjected to nitrogen-starvation, using Oxford Nanopore Technologies. A total of 39,033 transcripts were identified, including 11,356 new transcripts. In addition, alternative splicing (AS) events and lncRNAs were also detected between Col-0 (WT) and atg5-1. Differentially expressed transcript enrichment showed that autophagy upregulates the expression of many stress-responsive genes and inhibits the transcription of photosynthesis-associated genes. The qRT-PCR results showed that the expression patterns of photosynthesis-related genes in the atg5-1 differed under the conditions of nitrogen starvation and carbon starvation. Under nitrogen starvation treatment, many genes related to photosynthesis also exhibited AS. Chlorophyll fluorescence images revealed that the Fv/Fm and ΦPSII of old atg5-1 leaves were significantly reduced after nitrogen starvation treatment, but the Y(NPQ) indices were significantly increased compared to those of the WT plants. The results of qRT-PCR suggest that autophagy appears to be involved in the degradation of genes related to photodamage repair in PSII. Taken together, the full-length transcriptiome sequencing provide new insights into how new transcripts, lncRNAs and alternative splicing (AS) are involved in plant autophagy through full-length transcriptome sequencing and suggest a new potential link between autophagy and photosynthesis.

Seagrasses demonstrate complex ecological functions, and their morphological and physiological indicators are affected by various environmental factors. In this study, we conducted six sampling surveys of a Thalassia hemprichii seagrass bed in Xincun Bay, Lingshui, Hainan in 2019. The morphological and physiological characteristics of seagrasses in January, March, May, July, September, and November of a single year were studied, and the effects of various environmental factors on the morphological and physiological characteristics of T. hemprichii were analyzed. The results demonstrated markedly differences in leaf length and width, rhizome length and thickness, and root length across different months. The highest value [(15.05±6.13) cm] of leaf length occurred in May, and the lowest value [(7.19±2.55) cm] in September. The highest [(11.93±1.68) mm] and the lowest [(8.73±1.96) mm] values for leaf width were recorded in November and September, respectively. The highest value [(5.22± 1.71) mm] for rhizome diameter occurred in November, and the lowest value [(4.06±0.74) mm] in March. The C content of the underground seagrass tissues was significantly different across months, with the highest value (31.23%±0.94%) in November and the lowest value (24.90%±3.48%) in March. Seawater temperature had a significantly positive correlation with leaf width and C content of the underground tissues. Salinity was significantly positively correlated with the leaf and rhizome lengths of T. hemprichii and had a negative relationship with the C content of the underground tissues. There was a significant positive correlation between pH and leaf length, leaf width, and C and N content of the underground tissues. The ammonium level in seawater was significantly positively correlated with the N content of aboveground tissues. Nitrate level of seawater was negatively correlated with the C/N ratio of the aboveground parts. There was a significant positive correlation between the dissolved inorganic nitrogen (DIN) in seawater and the N content of the underground parts. Sediment organic carbon had a negative relationship with leaf length and width, and rhizome growth. The results demonstrated that the growth of T. hemprichii in Xincun Bay was greatly affected by temperature, salinity, and pH. A high nutrient level in Xincun Bay may have a negative impact on T. hemprichii. In order to protect the T. hemprichii meadow, measures should be taken to limit the input of nutrients in Xincun Bay. The results of this study may provide data support and serve as a scientific basis for the restoration of declining seagrass.

The distribution of seagrass meadows in intertidal zone is determined by the intensity of desiccation stress. The physiological characteristics of plants are the important basis to reflect their environmental stress effects. In order to explore the effects of the duration of photothermal desiccation stress on the physiological characteristics of seagrass during the tidal cycle, taking the tropical dominant seagrass Thalassia hemprichii as the research object, a simulation experiment was carried out in a solar greenhouse with high temperature (41.0-44.5℃) and strong light (633-1 682 μmol·m-2·s-1) environmental conditions. The effects of different desiccation time (0, 10, 20, 40, 60, 90 min·d-1, continuous for 7 d) on the growth rate, photosynthetic pigment content and photosynthetic fluorescence parameters of the leaves, as well as on the activities of antioxidant enzymes and the contents of nonstructural carbohydrates and secondary metabolites of leaves and roots were studied. The results were as follows: (1) The diurnal periodic photothermal desiccation of ≤10 min·d-1 had no significant effect on the growth rate and physiological characteristics of T. hemprichii (P>0.05). (2) Under the photothermal desiccation conditions, the damage of T. hemprichii leaves increased, the color of leaves changed from green to brown, and the growth rate of leaves decreased with the extension of desiccation time. Meanwhile, the photosynthetic pigment content, chlorophyll a/chlorophyll b and the chlorophyll fluorescence parameters of leaves showed a downward trend; the contents of MDA and soluble sugar and the activities of POD, SOD and CAT of leaves and roots showed an upward trend; whereas the contents of starch, total phenol and total flavonoids in leaves increased first and then decreased. (3) The diurnal periodic photothermal desiccation of 90 min·d-1 was the upper limit of incomplete inactivation for T. hemprichii chloroplast PS Ⅱ photosynthetic reaction center. In summary, a long term photothermal desiccation stress during tidal cycle has negative impacts on the growth and physiological characteristics for T. hemprichii. The results provide the reference for understanding the distribution of tropical seagrasses in the intertidal zone and the location selection of seagrass meadows for ecological restoration.

IntroductionThe transplantation of seagrass fragments with shoots and rhizomes is the most common method for the ecological restoration of damaged seagrass meadows.MethodsThe aim of this study was to explore the effects of exogenous addition (10 mg per month for 3 months) of three commonly used plant growth regulators (PGRs), including indoleacetic acid (IAA), gibberellin (GA3), and paclobutrazol (PP333) on the growth and physiology of transplanted Thalassia hemprichii fragments (TTFs), with and without the rhizome apex (RA), using a simulation test. [Results] IAA and GA3 elevated the photosynthetic capacity and growth rate of TTF leaves but did not significantly alter leaf length and width. In contrast, PP333 reduced the leaf photosynthetic capacity and growth rate, while greatly increasing the leaf width and root viability. Additionally, PP333 treatment led to an increase in antioxidant enzyme activities (peroxidase [POD], superoxide dismutase [SOD], and catalase [CAT]), as well as malondialdehyde (MDA) and total phenol contents in TTFs, indicating some stress effects on the seagrass. Furthermore, IAA and GA3 decreased soluble sugar and protein contents and increased starch content in TTF tissues, whereas PP333 treatment elevated both nonstructural carbohydrate and soluble protein contents. The presence of RA positively affected the growth and physiology of T. hemprichii compared with TTFs without RA. There was a significant interaction between the PGRs and RA on leaf growth rate, chlorophyll fluorescence variables, and photosynthetic pigment content of the TTFs (p < 0.05).DiscussionThis study provides guidelines for the application of PGRs during the transplantation of T. hemprichii fragments for the restoration of seagrass meadows.

The vegetation of the ecological substrate plays a crucial role in restoring shoreline ecology. This study focused on using zeolite and biochar as substrate modifiers, specifically utilizing the Cynodon dactylon from Central China for vegetation. A pot vegetation experiment was carried out to compare the effects of different ratios of zeolite and biochar. The vegetation indices, including germination index, plant height, and coverage rate, were analyzed and discussed. The results revealed that zeolite primarily influenced the germination index of Cynodon dactylon, while biochar had a more significant impact on germination percentage, germination energy, plant height, and coverage rate. This study discovered that the seed germination effect of the improved substrate initially increased with zeolite content and then decreased. The average germination percentage was 63.96%. Conversely, it decreased with an increase in biochar content, resulting in an average germination percentage of 55.45%. Zeolite and biochar caused a decrease and increase in substrate pH by −0.11 and 0.4 on average, respectively. The germination of each substrate showed a negative correlation with pH. Additionally, the average coverage and plant height decreased with an increase in biochar content. However, the inclusion of 6% zeolite led to an increase in coverage and plant height. Specifically, the average plant height increased by 3.92 cm and the coverage by 7.48%. Our research identified the optimal ratio of zeolite and biochar as 6% zeolite and 0% biochar, showcasing good overall vegetative properties. These findings offer insights for further understanding the vegetative effects of zeolite–biochar-modified substrates and optimizing substrate schemes for ecological vegetation projects.

Using different fast charging strategies for lithium-ion batteries can affect the degradation rate of the batteries. In this case, predicting the capacity fade curve can facilitate the application of new batteries. Considering the impact of fast charging strategies on battery aging, a battery capacity degradation trajectory prediction method based on the TM-Seq2Seq (Trend Matching—Sequence-to-Sequence) model is proposed. This method uses data from the first 100 cycles to predict the future capacity fade curve and EOL (end of life) in one-time. First, features are extracted from the discharge voltage-capacity curve. Secondly, a sequence-to-sequence model based on CNN, SE-net, and GRU is designed. Finally, a trend matching loss function is designed based on the common characteristics of capacity fade curves to constrain the encoding features of the sequence-to-sequence model, facilitating the learning of the underlying relationship between inputs and outputs. TM-Seq2Seq model is verified on a public dataset with 132 battery cells and multiple fast charging strategies. The experimental results indicate that, compared to other popular models, the TM-Seq2Seq model has lower prediction errors.

Under the combined action of soil evaporation and vegetation transpiration, the law of soil water dissipation at different depths of vegetated slopes is unknown and the related influencing factors are unclear. In this paper, six large-scale slope models were constructed for long-term dynamic monitoring of soil water. The effects of slope ratio and vegetation on the dynamic changes in soil water at different depths were analyzed. Pearson correlation analysis was used to analyze the relationship between slope conditions, meteorological factors, and soil water dissipation. The results show that under the condition of natural evaporation, slope ratio has little effect on the dynamic change in soil water in bare slopes. However, the greater the slope ratio of vegetated slopes, the faster the soil water decreases in the 40 cm depth range. Additionally, soil water dissipation follows a logarithmic functional relationship with evaporation time in both bare and vegetated slopes. The correlation between slope conditions and soil water dissipation is stronger than that of meteorological factors. The research results can provide some theoretical support for exploring the hydrological effects of vegetated slopes.

Eutrophication due to aquaculture can cause the decline of seagrasses and impact their carbon storage capacity. This study explored the effects of aquaculture on the sediment organic carbon (SOC) in Thalassia testudinum seagrass beds using enzyme activity and microorganisms as indicators. Our results showed that the distance to aquaculture significantly increased the SOC and TN of sediments; the C/N ratio of sediments was reduced by the distance to aquaculture. Distance to aquaculture and seagrasses significantly impacted the δ13C of sediments, and their significant interactive effects on the δ13C of sediments were found. Distance to aquaculture and seagrasses had significantly interactive effects on the cellulase activity of sediments. Distance to aquaculture and seagrasses separately reduced the invertase activity of sediments. SOC in the seagrass bed was significantly positively impacted by cellulase activity and polyphenol oxidase activity in sediments. Firmicutes, Desulfobacterota and Chloroflexi were the dominant taxa in the S1 and S2 locations. From the S1 location to the S2 location, the relative abundances of Firmicutes and Desulfobacterota increased. The functional profiles of COG were relatively similar between the S1 and S2 locations. BugBase phenotype predictions indicated that the microbial phenotypes of all the seagrass sediment samples were dominated by anaerobic bacteria in terms of oxygen utilizing phenotypes. FAPROTAX functional predictions indicated that aquaculture affects functions associated with seagrass bed sediment bacteria, particularly those related to carbon and nitrogen cycling. This study can provide an important basis for understanding the response mechanism of global carbon sink changes to human activities such as aquaculture and supply more scientific data for promoting the conservation and management of seagrass beds.

Two doses of the inactivated influenza vaccine (IIV) are generally recommended for children under 9 years old. This study assessed the necessity for a second dose of quadrivalent IIV (IIV4) in children aged 3–8 years. In this randomized, open-label, paralleled-controlled study, 400 children aged 3–8 years who were vaccine-unprimed were randomly assigned at a 1:1 ratio to receive a two-dose (Group 1) or one-dose (Group 2) regimen of IIV4, and 200 who were vaccine-primed received one dose of IIV4 (Group 3). A serum sample was collected before and 28 days after the last dose to determine the hemagglutination inhibition (HI) antibody level. Adverse events were collected within 28 days after each dose. One-dose or two-doses of IIV4 were well tolerated and safe in children aged 3–8 years, and no serious adverse events related to the vaccine were reported. The seroconversion rates (SCRs) of HI antibody ranged from 61.86% to 95.86%, and the post-vaccination seroprotection rates (SPRs) were all >70% in three groups against the four virus strains. The two-dose regimen in vaccine-unprimed participants (Group 1) achieved similar SPRs in comparison with the one-dose in the vaccine-primed group (Group 3), and the SPRs in Group 1 and Group 3 were higher in vaccine-unprimed participants of the one-dose regimen (Group 2). The present study supports the recommendations of a two-dose regimen for IIV4 use in children aged 3–8 years.

Through whole-genome bisulfite sequencing and RNA-seq, we determined the potential impact of autophagy in regulating DNA methylation in Arabidopsis, providing a solid foundation for further understanding the molecular mechanism of autophagy and how plants cope with nitrogen deficiency. A total of 335 notable differentially expressed genes (DEGs) were discovered in wild-type Arabidopsis (Col-0-N) and an autophagic mutant cultivated under nitrogen starvation (atg5-1-N). Among these, 142 DEGs were associated with hypomethylated regions (hypo-DMRs) and were upregulated. This suggests a correlation between DNA demethylation and the ability of Arabidopsis to cope with nitrogen deficiency. Examination of the hypo-DMR-linked upregulated DEGs indicated that the expression of MYB101, an ABA pathway regulator, may be regulated by DNA demethylation and the recruitment of transcription factors (TFs; ERF57, ERF105, ERF48, and ERF111), which may contribute to the growth arrest induced by abscisic acid (ABA). Additionally, we found that DNA methylation might impact the biosynthesis of salicylic acid (SA). The promoter region of ATGH3.12 (PBS3), a key enzyme in SA synthesis, was hypomethylated, combined with overexpression of PBS3 and its potential TF AT3G46070, suggesting that autophagy defects may lead to SA-activated senescence, depending on DNA demethylation. These findings suggest that DNA hypomethylation may impact the mechanism by which Arabidopsis autophagy mutants (atg5-1) respond to nitrogen deficiency, specifically in relation to ABA and SA regulation. Our evaluation of hormone levels verified that these two hormones are significantly enriched under nitrogen deficiency in atg5-1-N compared to Col-0-N.

This study quantified the absorption ability of the seagrass Zostera japonica and the macroalgae Ulva pertusa for dissolved inorganic nitrogen (DIN) (ammonium and nitrate) and dissolved organic nitrogen (DON) (urea and glycine) under different light conditions. The plants were cultured in filtered seawater (31‰, 25°C) for 2 weeks under three light levels. Macroalgae and the above- and belowground parts of seagrasses were separately placed into four different manmade seawater solutions with DIN (ammonium and nitrate) and DON (urea and glycine) stable isotopes for 1 h. The results showed that macroalgae had higher absorption rates for ammonium and nitrate after higher light (14.67 ± 2.50 and 1.29 ± 0.16 mg−1 dry weight (DW) h−1) than after lower light (4.52 ± 0.95 and 0.18 ± 0.12 mg−1 DW h−1) treatments. Compared to the belowground seagrass portions that had previously been grown in high and low light conditions, the seagrass leaves assimilated ammonium more quickly. Z. japonica preferred glycine to nitrate and urea after the high- and low-light treatments; that is, the absorption rates of the belowground seagrass parts for glycine were 14.71 ± 1.85 and 6.38 ± 0.52 mg−1 DW h−1 after the high- and low-light treatments, respectively, which were higher than those of ammonium, nitrate, and urea. The absorption rates of algae were lower than those for ammonium previously grown under medium- and low-light treatments. These results indicate that light reduction can impact the assimilation of DIN by Z. japonica and U. pertusa, and both have the ability to directly assimilate DON. This study provides information that could help reduce the negative effects of eutrophication on macroalgae and seagrasses in order to protect seagrass meadows.

Early detection of tumors has great significance for formative detection and determination of treatment plans. However, cancer detection remains a challenging task due to the interference of diseased tissue, the diversity of mass scales, and the ambiguity of tumor boundaries. It is difficult to extract the features of small-sized tumors and tumor boundaries, so semantic information of high-level feature maps is needed to enrich the regional features and local attention features of tumors. To solve the problems of small tumor objects and lack of contextual features, this paper proposes a novel Semantic Pyramid Network with a Transformer Self-attention, named SPN-TS, for tumor detection. Specifically, the paper first designs a new Feature Pyramid Network in the feature extraction stage. It changes the traditional cross-layer connection scheme and focuses on enriching the features of small-sized tumor regions. Then, we introduce the transformer attention mechanism into the framework to learn the local feature of tumor boundaries. Extensive experimental evaluations were performed on the publicly available CBIS-DDSM dataset, which is a Curated Breast Imaging Subset of the Digital Database for Screening Mammography. The proposed method achieved better performance in these models, achieving 93.26% sensitivity, 95.26% specificity, 96.78% accuracy, and 87.27% Matthews Correlation Coefficient (MCC) value, respectively. The method can achieve the best detection performance by effectively solving the difficulties of small objects and boundaries ambiguity. The algorithm can further promote the detection of other diseases in the future, and also provide algorithmic references for the general object detection field.

Abstract In nature, biological compartments such as cells rely on dynamically controlled permeability for matter exchange and complex cellular activities. Likewise, the ability to engineer compartment permeability is crucial for in vitro systems to gain sustainability, robustness, and complexity. However, rendering in vitro compartments such a capability is challenging. Here, a facile strategy is presented to build permeability‐configurable compartments, and marked advantages of such compartmentalization are shown in reconstituting sustained synthetic biology systems in vitro. Through microfluidics, the strategy produces micrometer‐sized layered microgels whose shell layer serves as a sieving structure for biomolecules and particles. In this configuration, the transport of DNAs, proteins, and bacteriophages across the compartments can be controlled an guided by a physical model. Through permeability engineering, a compartmentalized cell‐free protein synthesis system sustains multicycle protein production; ≈100 000 compartments are repeatedly used in a five‐cycle synthesis, featuring a yield of 2.2 mg mL−1. Further, the engineered bacteria‐enclosing compartments possess near‐perfect phage resistance and enhanced environmental fitness. In a complex river silt environment, compartmentalized whole‐cell biosensors show maintained activity throughout the 32 h pollutant monitoring. It is anticipated that permeability‐engineered compartmentalization should pave the way for practical synthetic biology applications such as green bioproduction, environmental sensing, and bacteria‐based therapeutics.

In recent years, molecular dynamics (MD) simulation has been used to study the deformation behaviors of glass under nanoindentation, mainly using ideal geometries like a spherical indenter or a 2.5-D sample geometry to simplify post-analysis and save computational costs. To generate stress/strain fields that can be directly compared with experiments, we developed a 3-D nanoindentation protocol in this work to study the deformation behaviors of a model metallic glass under sharp contact loading in MD. Our studies show that the indenter sharpness controls the shear band formation, and the interaction between shear bands dictates the crack initiation in the model metallic glass. Shear bands and residual stress fields in the model metallic glass from our simulated nanoindentation tests are consistent with observations in soda-lime silicate (SLS) glass from the instrumented indentation in experiments, as both of them favor shear deformation under sharp contact loading.

Summary: Background: BNT162b2, an mRNA vaccine against COVID-19, is being utilised worldwide, but immunogenicity and safety data in Chinese individuals are limited. Methods: This phase 2, randomised, double-blind, placebo-controlled trial included healthy or medically stable individuals aged 18–85 years enrolled at two clinical sites in China. Participants were stratified by age (≤55 or >55 years) and randomly assigned (3:1) by an independent randomisation professional to receive two doses of intramuscular BNT162b2 30 μg or placebo, administered 21 days apart. Study participants, study personnel, investigators, statisticians, and the sponsor's study management team were blinded to treatment assignment. Primary immunogenicity endpoints were the geometric mean titers (GMTs) of neutralising antibodies to live severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and seroconversion rates (SCR) 1 month after the second dose. Safety assessments included reactogenicity within 14 days of vaccination, adverse events (AEs), and clinical laboratory parameters. Randomised participants who received at least one dose were included in the efficacy and safety analyses on a complete case basis (incomplete/missing data not imputed). Results up to 6 months after the second dose are reported. Findings: Overall, 959 participants (all of Han ethnicity) who were recruited between December 5th, 2020 and January 9th, 2021 received at least one injection (BNT162b2, n=720; placebo, n=239). At 1 month after the second dose, the 50% neutralising antibody GMT was 294.4 (95% CI; 281.1–308.4) in the BNT162b2 group and 5.0 (95% CI; 5.0–5.0) in the placebo group. Support vector machine (SVM) is one of the most powerful technologies of machine learning, which has been widely concerned because of its remarkable performance. However, when dealing with the classification problem of large-scale datasets, the high complexity of SVM model leads to low efficiency and become impractical. Due to the sparsity of SVM in the sample space, this paper presents a new parallel data geometry analysis (PDGA) algorithm to reduce the training set of SVM, which helps to improve the efficiency of SVM training. The PDGA introduce Mahalanobis distance to measure the distance from each sample to its centroid. And based on this, proposes a method that can identify non support vectors and outliers at the same time to help remove redundant data. When the training set is further reduced, cosine angle distance analysis method is proposed to determine whether the samples are redundant data, ensure that the valuable data are not removed. Different from the previous data geometry analysis methods, the PDGA algorithm is implemented in parallel, which greatly saving the computational cost. Experimental results on artificial dataset and 6 real datasets show that the algorithm can adapt to different sample distributions. Which significantly reduce the training time and memory requirements without sacrificing the classification accuracy, and its performance is obviously better than the other five competitive algorithms.

Abstract Background Psittacosis, which is also known as parrot fever, is Chlamydia psittaci (C. psittaci) caused infectious disease. The clinical manifestations vary from asymptomatic infection to severe atypical pneumonia or even fatal meningitis. Early recognition of psittacosis is difficult because of its nonspecific clinical manifestations. Culture and gene probe techniques for C. psittaci are not available for routine clinical use, which makes the diagnosis difficult too. Although psittacosis has increasingly been recognized and reported in recent years, cure of severe pneumonia complicated with meningitis, with etiologic diagnosis aided by the use of metagenomic next-generation sequencing (mNGS), is still uncommon. So, it is necessary to report and review such potentially fatal case. Case presentation This report describes a 54-year-old woman with C. psittaci caused severe atypical pneumonia and meningitis. She presented with symptoms of fever, dry cough and dyspnea, accompanied by prominent headache. Her condition deteriorated rapidly to respiratory failure and lethargy under the treatment of empirical antibacterial agents, and was treated with invasive mechanical ventilation soon. She denied contact with birds, poultry or horses, but unbiased mNGS of both the bronchoalveolar lavage fluid (BALF) and the cerebrospinal fluid (CSF) identified sequence reads corresponding to C. psittaci infection, and there was no sequence read corresponding to other probable pathogens. Combined use of targeted antimicrobial agents of tetracyclines, macrolides and fluoroquinolones was carried out, and the patient’s condition improved and she was discharged home 28 days later. Her status returned close to premorbid condition on day 60 of follow-up. Conclusions When clinicians come across a patient with atypical pneumonia accompanied by symptoms of meningitis, psittacosis should be taken into consideration. mNGS is a promising detection method in such condition and is recommended.

Abstract Predicting material’s microstructure under new processing conditions is essential in advanced manufacturing and materials science. This is because the material’s microstructure hugely influences the material’s properties. We demonstrate an elegant machine learning algorithm that faithfully predicts the microstructure under new conditions, without the need of knowing the governing laws. We name this algorithm, RCWGAN-GP, which is regression-based conditional generative adversarial networks with Wasserstein loss function and gradient penalty. This algorithm was trained with experimental SEM micrographs from laser-sintered alumina under various laser powers. The RCWGAN-GP realistically regenerates the SEM micrographs under the trained laser powers. Impressively, it also faithfully predicts the alumina’s microstructure under unexplored laser powers. The predicted microstructure features, including the morphology of the sintered particles and the pores, match the experimental SEM micrographs very well. We further quantitatively examined the prediction accuracy of the RCWGAN-GP. We trained the algorithm with computer-created micrograph datasets of secondary-phase growth governed by the well-known Johnson–Mehl–Avrami (JMA) equation. The RCWGAN-GP accurately regenerates the micrographs at the trained time series, in terms of the grains’ shapes, sizes, and spatial distributions. More importantly, the predicted secondary phase fraction accurately follows the JMA curve.

In this study, a column experiment was employed to evaluate the nuclide migration behavior in the surrounding rock medium of a near-surface disposal site in China and to investigate the advection–dispersion behavior of tritium (H-3) and plutonium-238 (Pu-238) in highly weathered argillaceous shale. A reasonable numerical model was selected to fit the experimental breakthrough curves (BTCs) and to obtain the relevant migration parameters. The results show the following: (1) the internal structure of the highly weathered argillaceous shale exhibited heterogeneity, and the nuclide migration BTC showed characteristics of a “curve peak moving forward” and a “tail curve trailing”; (2) compared with other models, the stream tube mode could better fit the BTCs and obtain the average dispersion coefficient , average distribution coefficient , and other parameters; (3) compared to the results of the batch experiment, the distribution coefficient Kd obtained from the column experiment was smaller than that obtained from the batch experiment, which is speculated to be due to the influence of contact time and the contact area between the nuclide and the medium.

Congestion is a kind of expression of instability of traffic network. Traffic signal control keeping traffic network stable can reduce the congestion of urban traffic. In order to improve the efficiency of urban traffic network, this study proposes a decentralized traffic signal control strategy based on backpressure algorithm used in Wi-Fi mesh networks for packets routing. Backpressure based traffic signal control algorithm can stabilize urban traffic network and achieve maximum throughput. Based on original backpressure algorithm, the variant parameter and penalty function are considered to balance the queue differential and capacity of downstream links in urban traffic network. For each traffic phase of intersections, phase weight is computed using queue differential and capacity of downstream links, which fixed the deficiency of infinite queue capacity in original backpressure algorithm. It is proved that the extended backpressure traffic signal control algorithm can maintain stability of urban traffic network, and also can prevent queue spillback, so as to improve performance of whole traffic network. Simulations are carried out in Vissim using Vissim COM programming interface and Visual Studio development tools. Evaluation results illuminate that it can get better performance than the backpressure algorithm just based on queue length differential in average queue length and delay of traffic network.

Abstract Background Computerized provider order entry (CPOE) can help providers deliver better quality care. We aimed to understand recent trends in use of CPOE by health system-affiliated ambulatory clinics. Methods We analyzed longitudinal data (2014–2016) for 19,109 ambulatory clinics that participated in all 3 years of the Healthcare Information and Management Systems Society Analytics survey to assess use of CPOE and identify characteristics of clinics associated with CPOE use. We calculated descriptive statistics to examine overall trends in use, location of order entry (bedside vs. clinical station), and system-level use CPOE across all clinics. We used linear probability models to explore the association between clinic characteristics (practice size, practice type, and health system type) and two outcomes of interest: CPOE use at any point between 2014 and 2016, and CPOE use beginning in 2015 or 2016. Results Between 2014 and 2016, use of CPOE increased more than 9 percentage points from 58 to 67%. Larger clinics and those affiliated with multi-hospital health systems were more likely to have reported use of CPOE. We found no difference in CPOE use by primary care versus specialty care clinics. When used, most clinics reported using CPOE for most or all of their orders. Health systems that used CPOE usually did so for all system-affiliated clinics. Conclusions Small practice size or not being part of a multi-hospital system are associated with lower use of CPOE between 2014 and 2016. Less than optimal use in these environments may be harming patient outcomes.

In this paper, the effects of sodium sulfosalicylate on hemimorphite activation were studied. Sulfosalicylic acid ions enhanced the sulfidation-flotation process, which increased the flotation recovery by 49.70%, and after lead ion assisted activation, the flotation recovery subsequently increased by 13.11%. Time-of-flight secondary ion spectrometry (TOF-SIMS) surface analysis revealed that the surface structure of the hemimorphite was greatly damaged by sodium sulfosalicylate, which could promote ZnS film formation for obtaining hemimorphite hydrophobicity. The ZnS film thickness on the hemimorphite surface was approximately 41.78 nm with direct sulfidation. After activating sulfidation, excellent flotation effects were obtained as the ZnS film reached about 418.19 nm. 3D images showed a heterogeneous and homogeneous ZnS film on the hemimorphite surface after direct sulfidation and activating sulfidation, respectively. The X-ray photoelectron spectroscopy (XPS)-based analysis of the substances on the hemimorphite surface further indicated that the atomic concentration of sulfur on the surface of the activating sulfidation hemimorphite nearly double that of the direct sulfidation hemimorphite (1.57% versus 2.38%, respectively). After activating sulfidation, the XPS spectra indicated that a deep complexation reaction occurred with sodium sulfosalicylate on the hemimorphite surface with various elements and significantly influenced elemental distribution on the hemimorphite surface.

BackgroundHuman parvovirus B19 (B19) can cause acute hepatitis and is attributed to the high mortality of alcoholic hepatitis (AH). B19 infection is generally self-healing in previously healthy people, but it can cause fatal effects in some high-risk groups and increase its virulence and infectivity. Disseminated B19 infection-induced multiple organ dysfunction syndrome (MODS) in patients with AH has not been reported yet. Here, we described B19 viremia in an adult patient with AH accompanied by hemolytic anemia (HA), leading to disseminated infection and secondary MODS, as well as self-limiting B19 infections in seven nurses caring for him. Meanwhile, we reviewed the literature on AH and B19 infection.Case PresentationA 43-year-old male patient with AH accompanied by HA was transferred to the Third Affiliated Hospital of Sun Yat-sen University, Guangzhou, China, on March 31, 2021. After supportive treatment, his transaminase and bilirubin levels were reduced, but his anemia worsened. He received a red blood cell (RBC) infusion on April 9 for hemoglobin (Hb) lower than 6 g/dl. On April 13, he suddenly had a high fever. Under empirical anti-infection, his high fever dropped and maintained at a low fever level; however, his anemia worsened. On April 25, he was transferred to the medical intensive care unit (MICU) due to severe pneumonia, acute respiratory distress syndrome (ARDS), acute aplastic crisis (AAC), and hemophagocytic syndrome (HPS), which were subsequently confirmed to be related to B19 infection. After methylprednisolone, intravenous immunoglobulin (IVIG), empirical anti-infection, and supportive treatment, the lung infection improved, but hematopoietic and liver abnormalities aggravated, and systemic B19 infection occurred. Finally, the patient developed a refractory arrhythmia, heart failure, and shock and was referred to a local hospital by his family on May 8, 2021. Unfortunately, he died the next day. Fourteen days after he was transferred to MICU, seven nurses caring for him in his first two days in the MICU developed self-limiting erythema infectiosum (EI).ConclusionsB19 infection is self-limiting in healthy people, with low virulence and infectivity; however, in AH patients with HA, it can lead to fatal consequences and high contagion.

The safety of deep geological repositories is important in the disposal of high-level radioactive waste (HLW). In this study, advection–dispersion experiments were designed to build a transport model through a calibration/validation process, and the transport behavior of tritiated water (HTO) and various iodine species (iodide: I− and iodate: IO3−) was studied on a dynamic compacted granite column. Breakthrough curves (BTCs) were plotted under various flow rates (1–5 mL/min). BTCs showed that the non-sorption effect by anion exclusion was observed only in I− transport because the retardation factor (R) of I− was lower than that of HTO (R = 1). Moreover, equilibrium and nonequilibrium transport models were used and compared to identify the mobile/immobile zones in the compacted granite column. The anion exclusion effect was influenced by the immobile zones in the column. The non-sorption effect by anion exclusion (R < 1) was only observed for I− at 5.0 ± 0.2 mL/min flow rate, and a relatively higher Coulomb’s repulsive force may be caused by the smaller hydration radius of I−(3.31 Å) than that of IO3−(3.74 Å).

Background: Previous infectious or inflammatory events may be involved in the pathogenesis of neuromyelitis optica (NMO), potentially by triggering an autoimmune response. Cytomegalovirus (CMV)-related NMO (CMV-NMO) is rarely reported. Acute hemorrhagic rectal ulcer (AHRU) is a rare disease with a largely unknown pathogenesis. Herein, we reported a co-NMO and AHRU case associated with CMV infection. In addition, we review previously reported cases of CMV-NMO and CMV-AHRU.Case presentation: A 40-year-old female diagnosed with aquaporin4 (AQP4)-IgG+ NMO and a poor response to high-dose intravenous methylprednisolone and immunoglobulin, followed by three rounds of plasma exchange was transferred to Third Affiliated Hospital of Sun Yat-sen University, Guangzhou, China. She developed repeated acute lower gastrointestinal hemorrhage from the third day of admission. Abdominal computed tomography angiography (CTA) and interventional angiography did not detect any bleeding vessel. Bedside colonoscopy revealed a large ulcer-like lesion at 10 cm above the anus. Rectal biopsy pathology confirmed a CMV infection on day 23 post-admission, and cerebrospinal fluid (CSF) pathogen gene sequencing detected CMV gene copies on day 25 post-admission. After 2 weeks of treatment with ganciclovir and sodium phosphinate, the patient's lower gastrointestinal bleeding stopped, and her limb muscle strength and visual acuity gradually improved. After 4 weeks of antiviral therapy, colonoscopy showed that the intestinal wall of the original lesion was smooth. Hematoxylin and eosin (HE) staining and immunohistochemistry (IHC) of a biopsy specimen was negative for CMV, her right eye vision was normal, and limb muscle strength had recovered. Serum AQP4-IgG was negative, and lesions on brain magnetic resonance imaging (MRI) manifested shrinkage.Conclusions: The benefits of antiviral therapy remain unclear; however, clinicians should be aware of the possibility of CMV-related NMO, if NMO was refractory to high-dose intravenous methylprednisolone, immunoglobulin, and plasma exchange. Moreover, clinicians should consider the possibility of CMV-related AHRU when recurrent acute lower gastrointestinal bleeding occurs in a patient.

Abstract We used molecular dynamics simulation to investigate the friction of a single asperity against a rigid substrate, while generating debris. In the low wear regime (i.e., non-linear wear rate dependence on the contact stress, via atom-by-atom attrition), the frictional stress is linearly dependent on the normal stress, without any lubrication effect from the wear debris particles. Both the slope (friction coefficient) and friction at zero normal stress depend strongly on asperity-substrate adhesion. In the high wear regime (i.e., linear wear rate dependence on the contact stress, via plastic flow), the friction-normal stress curves deviate from a linear relation merging toward plastic flow of the single asperity which is independent of the interfacial adhesion. One can further link wear and friction by considering debris generation as chemical reaction, driven by both normal and frictional forces. The coupling between wear and friction can then be quantified by a thermodynamic efficiency of the debris generation. While the efficiency is less than 5% in the low wear regime, indicating poor mechanochemical coupling, it increases with normal stress toward 50% in the high wear regime.

An accurate and effective method combining ion chromatography (IC) and inductively coupled plasma optical emission spectrometry (ICP-OES) was applied in this work to qualitatively and quantitatively analyze individual and co-existing iodide (I−) and iodate (IO3−) at various concentrations. More specifically, a very strong linear relationship for the peak area for the co-existing I− and IO3− ions was reached, and a high resolution value between two peaks was observed, which proves the effectiveness of our combined IC-ICP-OES method at analyzing iodine species. We observed lower accessible porosity for the diffusion of both I− and IO3− in samples of bentonite clay using IC-ICP-OES detection methods, where the effective diffusion coefficient varied based on the anion exclusion effect and the size of the diffusing molecules. In fact, the distribution coefficients (Kd) of both I− and IO3− were close to 0, which indicates that there was no adsorption on bentonite clay. This finding can be explained by the fact that no change in speciation took place during the diffusion of I− and IO3− ions in bentonite clay. Our IC-ICP-OES method can be used to estimate the diffusion coefficients of various iodine species in natural environments.

Advection-dispersion experiments (ADE) were effectively designed for inadequate transport models through a calibration/validation process. HTO, selenium (Se), and cesium (Cs) transport in crushed granite were studied using a highly reliable, dynamic column device in order to obtain the retardation factors (R) and the dispersion coefficients (D) by fitting experimental breakthrough curves (BTCs) for various path lengths. In order to conduct a safety assessment (SA) of a deep geological repository for high-level radioactive waste, radionuclide transport in rock systems is necessary to clarify and establish a suitable model. A dynamic column with a radiotracer (HTO, Se(IV), and Cs) was applied to 2, 4, and 8 cm path lengths using a STANMOD simulation. The results showed similar results between the BTCs of Se and Cs by fitting a non-equilibrium sorption model due to the retardation effect. In fact, there was a relatively obvious sorption of Se and Cs in the BTCs obtained by fitting a retardation factor (R) value higher than 1. In addition, a two-region (physical) and a two-site (chemical) non-equilibrium model with either the lowest sum of squared residuals (SSQ) or the root mean square error (RMSE) were applied to determine the Se and Cs sorption mechanisms on granite.

Various numerical methods have been being extensively used to estimate the diffusion parameters of pertechnetate (99TcO4−) in compacted MX80 bentonite clay using through-diffusion (TD) techniques. In this study, an improved TD column test was applied, and the diffusion fluxes of tritium (HTO) as a non-reactive radionuclide, and 99TcO4− with various dry densities, were compared under the same experimental conditions. Similar results were obtained for the apparent diffusion coefficients of HTO and 99TcO4− using three estimation methods: a graphical method applying the asymptote calculation, an analytical solution using Lsqcurvefit installed in MATLAB, and the Marquardt–Levenberg optimization algorithm in the HYDRUS-1D inverse method. The statistical analysis showed that the densities using a one-way analysis of variance (ANOVA) for the three estimation methods ranged from 1200 to 2000 kg/m3, which indicated that there were no obvious differences between HTO and 99TcO4−. In general, the diffusion parameters of 99TcO4− were lower than those for HTO due to anion exclusion effects and lower accessible porosity.

Clay rock in the Tamusu (TMS) area in the Inner Mongolia province is one candidate as a geological disposal site for highly radioactive waste in China. The diffusion behavior of HTO and Se(IV) in Tamusu clay rock was studied by through-diffusion (TD) experiments and numerical analysis to determine the spatial and temporal variation. A minimum error analysis was conducted to determine the HTO and Se(IV) diffusion coefficients in compacted TMS clay. The TD experimental results and numerical analysis showed that the diffusion of HTO and Se reached a steady state within 7 and 50 days, respectively, and the apparent diffusion coefficients (Da) decreased with the increases in the compacted density. In fact, there was retardation of Se diffusion in intact TMS clay rock. A two-site sorption model for Se was applied to simulate fast and slow sorption behavior quantitatively.

The strength of nanocrystalline (NC) metal has been found to be sensitive to strain rate. Here, by molecular dynamics simulation, we explore the strain rate effects on apparent Young’s modulus, flow stress and grain growth of NC gold with small size. The simulation results indicate that the apparent Young’s modulus of NC gold decreases with the decrease of strain rate, especially for strain rates above 1 ns−1. The rearrangement of atoms near grain boundaries is a response to the decrease of apparent Young’s modulus. Indeed, the flow stress is also sensitive to the strain rate and decreases following the strain rate’s decrease. This can be found from the change of strain rate sensitivity and activation volume with the strain rate. Temperature has little effect on the activation volume of NC gold with small grain size, but has an obvious effect on that of relatively large grain size (such as 18 nm) under low strain rate (0.01 ns−1). Finally, grain growth in the deformation process is found to be sensitive to strain rate and the critical size for grain growth increases following the decrease of strain rate.

The traffic behaviours of commuters may cause traffic congestion during peak hours. Advanced Traffic Information System can provide dynamic information to travellers. Due to the lack of timeliness and comprehensiveness, the provided information cannot satisfy the travellers’ needs. Since the assumptions of traditional route choice model based on Expected Utility Theory conflict with the actual situation, a route choice model based on Game Theory is proposed to provide reliable route choice to commuters in actual situation in this paper. The proposed model treats the alternative routes as game players and utilizes the precision of predicted information and familiarity of traffic condition to build a game. The optimal route can be generated considering Nash Equilibrium by solving the route choice game. Simulations and experimental analysis show that the proposed model can describe the commuters’ routine route choice decision exactly and the provided route is reliable.

A new strategy for preparing amphibious ZnO quantum dots (QDs) with blue fluorescence within hyper-branched poly(ethylenimine)s (HPEI) was proposed in this paper. By changing [Zn2+]/[OH−] molar ratio and heating time, ZnO QDs with a quantum yields (QY) of 30% in ethanol were obtained. Benefiting from the amphibious property of HPEI, the ZnO/HPEI nanocomposites in ethanol could be dissolved in chloroform and water, acquiring a QY of 53%, chloroform and 11% in water. By this strategy, the ZnO/HPEI nano-composites could be applied in not only in optoelectronics, but also biomedical fields (such as bio-imaging and gene transfection). The bio-imaging application of water-soluble ZnO/HPEI nanocomposites was investigated and it was found that they could easily be endocytosed by the COS-7 cells, without transfection reagent, and they exhibited excellent biological imaging behavior.

A new strategy for nanocrystal encapsulation, release and application based on pH-sensitive covalent dynamic hyperbranched polymers is described. The covalent dynamic hyperbranched polymers, with multi-arm hydrophobic chains and a hydrophilic hyperbranched poly(amidoamine) (HPAMAM) core connected with pH-sensitive imine bonds (HPAMAM−DA), could encapsulate CdTe quantum dots (QDs) and Au nanoparticles (NPs). Benefiting from its pH response property, CdTe QDs and Au NPs encapsulated by HPAMAM−DA could be released to aqueous phase after imine hydrolysis. The released CdTe/HPAMAM and Au/HPAMAM nanocomposites exhibited excellent biological imaging behavior and high catalytic activities on p-nitrophenol hydrogenation, respectively.

Commercially available lattices contain various kinds of morphological imperfections which result in great degradation in lattices’ mechanical properties, therefore, to obtain imperfection insensitive lattice structure is obviously a practical research subject. Hierarchical structure materials were found to be a class of promising anti-defect materials. This paper builds hierarchical lattice by adding soft adhesion to lattice’s cell edges and numerical results show that its imperfection sensitivity to missing bars is minor compared with the classic lattice. Soft adhesion with appropriate properties reinforce cell edge’s bending stiffness and thus reduce the bending deformation in lattice caused by missing bars defect, which is confirmed by statistical analysis of normalized node displacements of imperfect lattices under hydrostatic compression and shear loads.

Leojaponin (2), a labdane diterpene, was isolated from the EtOH extract of the herb of Leonurus japonicus together with a new halimane diterpene named isoleojaponin (1). Isoleojaponin has a new diterpene skeleton with a unique cross-conjugated α,β-unsaturated ketone system, Their structures were elucidated by physical and spectroscopic analysis, and the relative configuration of the chiral C-9 carbon was determined by a computational method, and analysis of its possible biogenesis pathways.

A series of laboratory incubation experiments was conducted to evaluate the effect of biochar application on the efficacy of the nitrification inhibitor (NI) dicyandiamide (DCD) in Cambisol (pH 7.14) and Latosol (pH 4.83). The feedstocks (eucalyptus wood, coconut coir, and rice straw), pyrolysis temperatures (350, 500, and 650 ˚C), and application rates (0.5, 1.0, 2.5, and 5.0% of 200 g soil) were identified as influential factors. The results showed that biochar could significantly reduce the effectiveness of DCD on nitrification inhibition. Biochar produced from eucalyptus wood with a large surface area (426.4 m2 g−1) had the strongest ability to reduce the inhibitory effect of DCD in nearly neutral Cambisol, while biochar from rice straw with a high pH had the greatest influence on acidic Latosol. Increasing pyrolysis temperature and application rates can strengthen the ability of biochar to reduce the inhibitory effect of DCD. Generally, the decrease of the DCD nitrification inhibitory effect on nearly neutral soil was controlled by the surface area of the applied biochar; meanwhile, the rise of soil pH caused by biochar application was also an important influencing factor in acid soil.

The effect of fatigue on ZrCuAl amorphous metals induced by mechanical cyclic loading is investigated using inelastic neutron scattering and the pair density function analysis of neutron diffraction data. With cooling, the local atomic structure undergoes reorganization under fatigue that is directly related to the number of fatigue cycles. Also under fatigue, suppression in the atomic dynamics is observed as well. A structural restructuring occurs within a 4 Å radius and intensifies with increasing the compression cycles, whereas the vibrational density of states is attenuated as the intensity shifts towards the elastic, zero-energy transfer peak. The combined static and dynamic structural effects are a signature of the microscopic changes brought about by fatigue, and together may be the onset for subsequent behaviors following extended cyclic loading such as fracture. Even after the load is removed, the structural changes described here remain and increase with repeated cyclic loading which is an indication that the lattice deforms even before shear bands are formed.

With a hyperbranched poly(amidoamine) core and many water-soluble poly(ethylene glycol) monomethyl ether arms connected by pH-sensitive acylhydrazone bonds, multiarm hyperbranched polymer was used as nanoreactor and reductant to prepare metal nanoparticles endowed with intelligence and biocompatibility. The multiarm hyperbranched polymer encapsulated nanoparticles (NPs) showed excellent catalytic activity for hydrogenation, thus an excellent catalyst system for hydrogenation was established. The rate constants could reach as high as 3.48 L·s−1·m−2, which can be attributed to the lack of surface passivation afforded by the multiarm hyperbranched polymer.
