Pulmonary fibrosis (PF) is characterised by inflammation and collagen deposition in the alveolar interstitium, leading to dyspnoea and death. However, the comprehensive pathogenesis of PF remains unclear due to the lack of spontaneous fibrosis mouse models. Here, we found that Ifngr1-/-Rag2-/- mice, lacking the mechanisms to suppress group 2 and 3 innate lymphoid cells (ILC2s and ILC3s), developed severe PF spontaneously. In Ifngr1-/-Rag2-/- mice, Il1rl1hiIl13hi-ILC2 subpopulation was increased at disease-onset phase before collagen production began. Further, defects in ILCs or IL-33, the strong ILC2 activator, prevented PF development. ILC2s directly induce collagen production by fibroblasts in vitro, and fibroblasts started to produce IL-33 in the chronic phase, presumably forming a positive feedback loop between fibroblasts and ILC2s leading to irreversible fibrosis. Moreover, the increased IL1RL1 and IL13 and decreased IFNGR1 expression levels in ILC2s from human idiopathic PF patients highlight the significance of the novel mouse model in fibrosis research.

CD4(+) T cells that express the forkhead box P3 (FOXP3) transcription factor function as regulatory T (Treg) cells and hinder effective immune responses against cancer cells. Abundant Treg cell infiltration into tumors is associated with poor clinical outcomes in various types of cancers. However, the role of Treg cells is controversial in colorectal cancers (CRCs), in which FOXP3(+) T cell infiltration indicated better prognosis in some studies. Here we show that CRCs, which are commonly infiltrated by suppression-competent FOXP3(hi) Treg cells, can be classified into two types by the degree of additional infiltration of FOXP3(lo) nonsuppressive T cells. The latter, which are distinguished from FOXP3(+) Treg cells by non-expression of the naive T cell marker CD45RA and instability of FOXP3, secreted inflammatory cytokines. Indeed, CRCs with abundant infiltration of FOXP3(lo) T cells showed significantly better prognosis than those with predominantly FOXP3(hi) Treg cell infiltration. Development of such inflammatory FOXP3(lo) non-Treg cells may depend on secretion of interleukin (IL)-12 and transforming growth factor (TGF)-β by tissues and their presence was correlated with tumor invasion by intestinal bacteria, especially Fusobacterium nucleatum. Thus, functionally distinct subpopulations of tumor-infiltrating FOXP3(+) T cells contribute in opposing ways to determining CRC prognosis. Depletion of FOXP3(hi) Treg cells from tumor tissues, which would augment antitumor immunity, could thus be used as an effective treatment strategy for CRCs and other cancers, whereas strategies that locally increase the population of FOXP3(lo) non-Treg cells could be used to suppress or prevent tumor formation.

Clostridium species belonging to clusters XIVa and IV are predominant organisms in the gut microbiota. They are known to affect various aspects of host biology, including the optimal breakdown of foods, competition with pathogenic microorganisms, maintenance of the intestinal epithelial barrier, and immune system development. We have recently shown that a defined mixture of 46 strains of mouse-derived Clostridium species belonging to clusters XIVa and IV affects the development and function of colonic Foxp3+ CD4+ regulatory T (Treg) cells in mice. Clostridium-induced Treg cells are likely to play a role in the maintenance of immune homeostasis in the gut.

The purpose of this study is to achieve decentralized processing of the finite-element method (FEM) by making use of matrix decomposition. For structural analysis of FEM, generally, the global stiffness matrix is a very large matrix which reflects the physical properties of the structure. The matrix decomposition can be used to transpose the global stiffness matrix with simple band matrices. The object of analysis in this study is a truss. The matrix decomposition is effective for the high-dimensional equation which decomposes a reduced-order equation. The computational process with analysis of FEM can be optimized such that the time complexity is minimized due to parallel processing. In this study, the matrix decomposed algorithm is derived for the analysis of a truss, and the time complexity and the arithmetic complexity are compared numerically. Results show that, the time complexity and the arithmetic complexity can be reduced to 29% and 26%, respectively.

Manipulation of the gut microbiota holds great promise for the treatment of inflammatory and allergic diseases. Although numerous probiotic microorganisms have been identified, there remains a compelling need to discover organisms that elicit more robust therapeutic responses, are compatible with the host, and can affect a specific arm of the host immune system in a well-controlled, physiological manner. Here we use a rational approach to isolate CD4(+)FOXP3(+) regulatory T (Treg)-cell-inducing bacterial strains from the human indigenous microbiota. Starting with a healthy human faecal sample, a sequence of selection steps was applied to obtain mice colonized with human microbiota enriched in Treg-cell-inducing species. From these mice, we isolated and selected 17 strains of bacteria on the basis of their high potency in enhancing Treg cell abundance and inducing important anti-inflammatory molecules--including interleukin-10 (IL-) and inducible T-cell co-stimulator (ICOS)--in Treg cells upon inoculation into germ-free mice. Genome sequencing revealed that the 17 strains fall within clusters IV, XIVa and XVIII of Clostridia, which lack prominent toxins and virulence factors. The 17 strains act as a community to provide bacterial antigens and a TGF-β-rich environment to help expansion and differentiation of Treg cells. Oral administration of the combination of 17 strains to adult mice attenuated disease in models of colitis and allergic diarrhoea. Use of the isolated strains may allow for tailored therapeutic manipulation of human immune disorders.

Foxp3+ CD4+ cells are prominent immune regulatory T (Treg) cells that are most abundant in the intestine. Recent studies have suggested that intestinal Treg cells consist of thymically and extrathymically developed cells that have unique characteristics. A fraction of intestinal Treg cells express T cell receptors that recognize antigens that are derived from the gut microbiota. The presence of the gut microbiota, particularly the Clostridium species, affects the development and function of Treg cells. These intestinal bacteria-induced Treg cells are likely to play a role in the tolerance toward the gut microbiota. These recent advances provide new insight into how T cells are educated in the intestine to maintain homeostasis with the gut microbiota.

A coupled enzyme system of orotidine-5′-phosphate pyrophosphorylase and orotidine-5′-phosphate decarboxylase has been purified approximately 30-fold from cell-free extract of Micrococcus glutamicus 534 Co–147 by means of acid treatment and fractionations with ammonium sulfate and ethanol addition, and properties of the enzyme system have been studied.Optima of pH, temperature and substrate concentrations for the activity of the purified enzyme system have been investigated, and compared with those of the same enzyme system from dried brewer’s yeast. Furthermore, effects of various inhibitors on the enzyme activity have been examined and it has become evident that the enzyme system is completely inactivated by addition of chelating agent such as EDTA, and regenerated by further addition of magnesium ion.

An enzymatic method for determination of 5-phosphoribosylpyrophosphate (PRPP) was investigated. The method was based on a coupled reaction of orotidine-5ʹ-monophosphate pyrophosphorylase and or otidine-5ʹ-monophosphate decarboxylase obtained as cell-free extract from a mutant strain of Micrococcus glutamicus (Syn. Corynebacterium glutamicum). The enzymes catalyzed quantitative conversion of orotic acid to uridinc-5ʹ-monophosphate. In the method, the consumption of orotic acid was determined by measuring optical density at 295 mµ and then PRPP was calculated from the amount of orotic acid consumed. The conditions of the enzyme reaction were examined in detail, and a modified method for determination of PRPP involving termination of the enzyme reaction by EDTA was established. According to the present method, 10-1 µmol. of PRPP is determined with accuracy of 100±3% within 30 minutes. As an application of the method, chemical stability of PRPP was examined and the results were described.

SummaryIntestinal Th17 cells are induced and accumulate in response to colonization with a subgroup of intestinal microbes such as segmented filamentous bacteria (SFB) and certain extracellular pathogens. Here, we show that adhesion of microbes to intestinal epithelial cells (ECs) is a critical cue for Th17 induction. Upon monocolonization of germ-free mice or rats with SFB indigenous to mice (M-SFB) or rats (R-SFB), M-SFB and R-SFB showed host-specific adhesion to small intestinal ECs, accompanied by host-specific induction of Th17 cells. Citrobacter rodentium and Escherichia coli O157 triggered similar Th17 responses, whereas adhesion-defective mutants of these microbes failed to do so. Moreover, a mixture of 20 bacterial strains, which were selected and isolated from fecal samples of a patient with ulcerative colitis on the basis of their ability to cause a robust induction of Th17 cells in the mouse colon, also exhibited EC-adhesive characteristics.

When Lysine-rice was washed with water for ten miniutes, 24% of the total Lysine were lost in the water. The results of the cooking test employing the mixed rice of Lysine-rice with white polished rice at the ratio of 1:100 indicated that Lysine was uniformly distributed in the cooked rice but 18% of the total Lysine were lost during cooking. There were no significant differences in the color and flavor of cooked rice between the mixed rice and white polished rice. Storage test showed that the amount of Lysine in Lysine-rice did not decrease within 6 months at 40°C.
