Overcoming the low reactivity of thioethers, a C(sp3)–H bond functionalization of thioethers with styrenes using TBHP and DBU involving a radical relay process in metal-free conditions with theoretical questions elaborated is described.

The synthesis of axially chiral compounds has attracted a great deal of attention in recent years. Herein, an efficient and economical synthetic route has been developed for 2-(3- carbamoylpyridin-2-yl) nicotinamide pyridine salts, axially chiral compounds. The starting material 1,10-phenanthroline is readily available. In this study, 2-(3-carbamoylpyridin-2-yl) nicotinamide pyridine salts are obtained in moderate to good yields. This protocol includes simple operations and has easy scalability. In addition, the axial chirality of the products is also preliminary studied.

High-nitrogen energetic compounds are widely used in military and civil fields. To cope with some special conditions, materials with high melting point and good heat resistance are required. Herein, synthetic approach of two TACOT-derived compounds with high heat resistance and strong insensitivity for friction has been optimized. Significantly, 100 g-scale of aza-TACOT(I) was prepared with a total yield of 80% and a purity of 99.9%. Meanwhile, aza-TACOT(II) was obtained with a purity of 98.9% under the optimized conditions. This practical method features scalable and simple operations. In addition, an acceptable mechanism was firstly proposed for the critical step.

Abstract: A new type of NADH model compound, which has six asymmetric carbon centers, has been synthesized by an efficient and convenient method. We used NADH models combined with enzymes in the asymmetric reduction. It was the first time that dehydrogenase combined with NADH models instead of inorganic catalysts, such as MgCl2, was used in the asymmetric reduction. Furthermore, the experimental results showed that the new NADH model has the strongest fluorescence emission properties compared to previously reported models.

The C(sp3)–H bond is found widely in organic molecules. Recently, the functionalization of C(sp3)–H bonds has developed into a powerful tool for augmenting highly functionalized frameworks in organic synthesis. The synthesis of bioactive amides has been the pursuit of chemists. Herein secondary amides incorporated with an aldehyde group were first generated using aldehydes and secondary amines. Various (hetero)aryl aldehydes and even aliphatic aldehydes (40 examples) were converted into the desired products in moderate to excellent yields (up to 89%). A plausible mechanism involving a Cu(I/II/III) catalytic cycle combined with radical rearrangement was proposed and confirmed with four key intermediates detected by high-resolution mass spectrometry.

Rare-earth catalysis has become a hot topic in the field of catalytic organic reaction. Chain ethers mostly have lower reactivity and lower boiling points which limited their reaction scope. Herein, we found a rare-earth Y(OTf)3 can catalyze the coupling reaction of ethers especially chain ethers and thioethers with azaarenes. This protocol features simple operations, a broad substrate scope (31 examples), moderate to good yields (up to 85%), and atom economy.

The first sequentially combined inorganic base promoted N-addition/[2,3]-sigmatropic rearrangement reaction of α-alkylidene pyrazolinones and propargyl sulfonium salts has been reported to construct homoallyl sulfur-containing pyrazolones with moderate to excellent yields. α-Alkylidene pyrazolinones function as N-nucleophilic agents distinguished from the reported C-addition reactions. Propargyl sulfonium salts were first involved in the [2,3]-sigmatropic rearrangement protocol differentiated from the well-established annulation reactions. The excellent regioselectivity, the broad scope of substrates, gram-scale synthesis and convenient transformation embody the synthetic superiority of this cascade process.

A copper-catalyzed decarboxylative oxidative coupling of α,β-unsaturated carboxylic acids with non-cyclic ethers is developed. This method provides a new approach for C(sp3)–H bond functionalization of non-cyclic ethers. Mechanism study shows the reaction involves a radical process.

A novel direct alkylation of thiophenes via bis-coupling with vinyl acetates has been developed. To the best of our knowledge, this represents the first report of the iron-catalysed coupling of two thiophenes with vinyl groups. Utilizing earth-abundant, inexpensive, and non-toxic iron catalysts, this methodology converts simple thiophenes to symmetrical dithienylethane derivatives under mild conditions in one step.

Sumalactones A–D (1–4), four new curvularin-type macrolides, together with two known analogues, curvularin (5) and dehydrocurvularin (6), were isolated from Penicillium Sumatrense, a marine fungus isolated from deep-sea sediments. Sumalactones C (3) and D (4) are unprecedented curvularin-type macrolides bearing a rare 11-membered macrolide skeleton. Their structures were elucidated on the basis of intensive spectroscopic analysis. The absolute configurations of compounds 1–4 were determined by CD spectra and modified Mosher's method. Compound 6 showed significant inhibition activity towards LPS-induced nitric oxide production in RAW 264.7 macrophages with IC50 value of 0.91 μM.

Studying NADH molecules is one of the most active areas in biomimetic research. It is important to design novel and efficient chiral NADH model molecules. Herein, a cage-like NADH model with multi-chiral centers was designed, and key intermediates have been synthesized. In this study, we found that pentafluorophenoxy group is an excellent leaving group for our synthetic route.

4,5-seco-Probotryenols A–C (1–3), a new type of sesquiterpenoids named seco-probotryane-type sesquiterpenoid, have been obtained from Stachybotrys bisbyi (PYH05-7), along with five new botryane skeleton sesquiterpenoids (4, 5, and 8–10), and two known ones (6 and 7). Their structures were determined by NMR analyses, chemical derivatization, and X-ray crystallography.

C(sp3)–H Bonds are the most common structures in organic molecules. In recent years, the direct functionalization of C(sp3)–H bonds has attracted wide attention and made significant progress. One new dihydroisocoumarin, pericocin A (1), one new chromone, pericocin B (2), and two new α-pyrone derivatives, pericocins C-D (3-4), together with two known compounds, 3-(2-oxo-2H-pyran-6-yl)propanoic acid (5) and (E)-3-(2-oxo-2H-pyran-6-yl)acrylic acid (6), were isolated from the culture of the endolichenic fungus Periconia sp.. Their structures were elucidated by spectroscopic methods. All these compounds are derived from the polyketone biosynthetic pathway. Compound 1 was obtained as a mixture of enantiomers. The antimicrobial activity of compounds 1-5 was tested against Escherichia coli, Staphylococcus aureus, Aspergillus niger, and Candida albicans. Compounds 1-5 showed moderate antimicrobial activity against A. niger and weak activity against C. albicans.

Fusicoccane diterpenoids usually possess a fused 5-8-5 tricyclic ring system, which are biogenetically generated from geranylgeranyl diphosphate (GGDP). In our report, three novel diterpenoid alkaloids with fusicoccane skeleton, pericolactines A–C (1–3), were isolated from Periconia sp.. Their structures with absolute configurations were determined by spectroscopic analyses and quantum chemical ECD calculation. Pericolactines A–C (1–3) are a new class of diterpenoid alkaloids with an unusual fused 5-5-8-5 tetracyclic ring system, which derive from a geranylgeranyl diphosphate (GGDP) and serine conjugated biosynthesis. They belong to the atypical diterpenoid alkaloids.

Pericoterpenoid A (1), a new cadinane-type sesquiterpene, was isolated from an endolichenic fungal strain Periconia sp. (No. 19-4-2-1). Its structure was characterized by analyzing the spectroscopic data (IR, MS, 1D- and 2D-NMR). The antimicrobial activity against Escherichia coli, Staphylococcus aureus, Aspergillus niger, and Candida albicans was evaluated. Pericoterpenoid A showed moderate antimicrobial activity against A. niger and weak activity against C. albicans. This is the first report of the presence of cadinane-type sesquiterpene in Periconia sp.

Stachybisbins A (1) and B (2), two new meroterpenoids with unprecedented seco-bisabosqual skeleton, together with three biogenetically related metabolites (3-5), were isolated from a wetland fungal strain of Stachybotrys bisbyi. The structures of the new compounds were determined by spectroscopic analyses, modified Mosher's method, and quantum chemical CD method. Cytoglobosins H (1) and I (2), together with seven known cytochalasan alkaloids (3–9), were isolated from the deep-sea-derived fungus Chaetomium globosum. The structures of new compounds 1 and 2 were elucidated by extensive 1D and 2D NMR and mass spectroscopic data. All the compounds were evaluated for their antiproliferative activities against MDA-MB-231 human breast cancer cells, LNCaP human prostate cancer cells, and B16F10 mouse melanoma cells. Compound 6 showed significant antiproliferative activity against LNCaP and B16F10 cell lines with IC50 values of 0.62 and 2.78 μM, respectively. Further testing confirmed that compound 6 inhibited the growth of LNCaP cells by inducing apoptosis.
