SCN8A variants cause a spectrum of epilepsy phenotypes ranging from self‐limited infantile epilepsy (SeLIE) to developmental and epileptic encephalopathy. SeLIE is an infantile onset focal epilepsy, occurring in developmentally normal infants, which often resolves by 3 years. Our aim was to ascertain when epilepsy resolves in SCN8A‐SeLIE. We identified unpublished individuals with SCN8A‐SeLIE and performed detailed phenotyping. Literature was searched for published SCN8A‐SeLIE cases. Nine unpublished individuals from four families were identified (age at study = 3.5–66 years). Six had their last seizure after 3 years (range = 4–21 years); although drug‐responsive and despite multiple weaning attempts (1–5), five of six remain on antiseizure medications (carbamazepine, n = 3; lamotrigine, n = 2). We identified 29 published individuals with SCN8A‐SeLIE who had data on seizure progression. Of the 22 individuals aged at least 10 years, reported here or in the literature, nine of 22 (41%) had seizure offset prior to 3 years, five of 22 (23%) had seizure offset between 3 and 10 years, and eight of 22 (36%) had seizures after 10 years. Our data highlight that more than half of individuals with SCN8A‐SeLIE continue to have seizures into late childhood. In contrast to SeLIE due to other etiologies, many individuals have a more persistent, albeit drug‐responsive, form of epilepsy. [ABSTRACT FROM AUTHOR]

White-footed mice (Peromyscus leucopus) are important host reservoirs for Borrelia burgdorferi and Rickettsia rickettsii. These bacteria are responsible for the tick-borne illnesses Lyme disease and Rocky Mountain spotted fever, respectively. These illnesses infect Ohio citizens annually, as well others in the larger range where these ticks are found. The goal of this study was to test white-footed mice and ticks for B. burgdorferi and R. rickettsii at Capital University's Primmer Outdoor Learning Center located in Logan, Ohio during the summer of 2020. Blood samples and ticks were collected from mice (n = 16) over the span of six live-trapping sessions in a woodlot and fencerow on the property. Ticks were also collected from four other locations using the drag-cloth method. DNA was extracted from the blood samples (n = 13) and ticks (n = 16) and tested for the presence of B. burgdorferi and Rickettsia sp. using conventional PCR and agarose gel electrophoresis. Relative abundance of mice was slightly higher in the fencerow than in the woodlot. Only American dog ticks (Dermacentor variabilis) were found in this study. B. burgdorferi was not detected, while Rickettsia sp. were confirmed for one mouse and 14 ticks. The results from this study increase our understanding of tick-borne pathogens within the state of Ohio and suggest we need to take steps to minimize risk of disease exposure at the University field site. In the future, this work can be used to inform models to simulate tick-borne pathogen dynamics in southeastern Ohio for both Lyme disease and Rocky Mountain spotted fever. [ABSTRACT FROM AUTHOR]

Kinship caregivers raise relative children who are unable to be cared for by their parents. Most kinship caregiving takes place outside the child welfare system ("informal" care), meaning that caregivers do not have access to foster care payments to offset the costs of child rearing. The main source of financial support for informal kinship caregivers is Child-Only TANF, which is inadequate to meet the full range of caregiver needs. To address this funding gap, the authors present a comparative policy analysis of seven programs in six states that have implemented supplemental state funds for informal kinship caregivers. The authors analyze key features of each program and compare the socioeconomic and political characteristics of states that offer supplemental caregiver benefits to those of states that do not. The International Society for Physical Activity and Health advocates for increased capability of the physical activity workforce as a key ingredient to a system-based approach. Young leader programmes are gaining traction globally as peers are a primary influence on young people and positive role models are important for increasing or maintaining physical activity. Yet, there is limited understanding of 'what works' for training young physical activity leaders. This study aims to develop a consensus on how to identify and support young people to become physical activity leaders. An iterative three-phased mixed methods Delphi consensus approach. A rapid review focused on the feasibility, acceptability and impact of existing young leader physical activity training (phase one); focus groups (n = 3) and interviews (n = 6) with 15 practitioners and young leaders to examine young physical activity leader training needs (phase two); and a three-round questionnaire process (phase three). Stakeholders (n = 43) from across the public, voluntary and education sectors, academics and young leaders completed all questionnaires. A consensus was reached for 75 statements related to: young leader traits prior to and following training, recruitment methods, training content, delivery format and context, relationships, incentives, and skill development. The Delphi process, combining insight from multi-sectoral stakeholders, identified a range of factors that underpin young leader training programmes. These factors should be applied to develop a curriculum and comprehensive training programme to provide young leaders with the required capability to be effective within their roles, and ultimately support an increase in physical activity amongst children and young people. Cellulomonas flavigena KU (ATCC 53703) produces an extracellular matrix involved in the degradation of microcrystalline cellulose. This extracellular material is primarily composed of the gel-forming, β-1,3-glucan known as curdlan and associated, cellulose-degrading enzymes. In this study, the effects of various forms of nutrient limitation on cellulose attachment, cellular aggregation, curdlan production, and biofilm formation were investigated throughout a 7-day incubation period by using phase-contrast microscopy. Compared to cultures grown in non-limiting media, nitrogen-limitation promoted early attachment of C. flavigena KU cells to the cellulose surface, and cellulose attachment was congruent with cellular aggregation and curdlan production. Over the course of the experiment, microcolonies of attached cells grew into curdlan-producing biofilms on the cellulose. By contrast, bacterial cells grown on cellulose in non-limiting media remained unattached and unaggregated throughout most of the incubation period. By 7 days of incubation, bacterial aggregation was ninefold greater in N-limited cultures compared to nutritionally complete cultures. In a similar way, phosphorus- and vitamin-limitation (i.e., yeast extract-limitation) also resulted in early cellulose attachment and biofilm formation. Furthermore, nutrient limitation promoted more rapid and efficient fragmentation and degradation of cellulose, with cellulose fragments in low-N media averaging half the size of those in high-N media after 7 days. Two modes of cellulose degradation are proposed for C. flavigena KU, a “planktonic mode” and a “biofilm mode”. Similar observations have been reported for other curdlan-producing cellulomonads, and these differing cellulose degradation strategies may ultimately prove to reflect sequential stages of a multifaceted biofilm cycle important in the bioconversion of this abundant and renewable natural resource. A vascular anastomosis is a critical surgical skill that involves connecting blood vessels. Traditional handsewn techniques can be challenging and resource intensive. To address these issues, we have developed a unique sutureless anastomotic device called Vaso-Lock. This intraluminal device connects free vascular ends using anchors to maintain traction and enable a rapid anastomosis. We tested the anastomotic capability of Vaso-Locks in a pig common carotid-internal jugular arteriovenous model. The use of Vaso-Lock allowed us to accomplish this procedure in less than 10 min, in contrast to the approximately 40 min required for a handsewn anastomosis. The Vaso-Lock effectively maintained patency for at least 6 weeks without causing significant tissue damage. Histological analysis revealed that the device was successfully incorporated into the arterial wall, promoting a natural healing process. Additionally, organ evaluations indicated no adverse effects from using the Vaso-Lock. Our findings support the safety and effectiveness of the Vaso-Lock for arteriovenous anastomosis in pigs, with potential applicability for translation to humans. While there is now strong evidence that many factors can shape dispersal, the mechanisms influencing connectivity patterns are species‐specific and remain largely unknown for many species with a high dispersal potential. The rock lobsters Jasus tristani and Jasus paulensis have a long pelagic larval duration (up to 20 months) and inhabit seamounts and islands in the southern Atlantic and Indian Oceans, respectively. We used a multidisciplinary approach to assess the genetic relationships between J. tristani and J. paulensis, investigate historic and contemporary gene flow, and inform fisheries management. Using 17,256 neutral single nucleotide polymorphisms we found low but significant genetic differentiation. We show that patterns of connectivity changed over time in accordance with climatic fluctuations. Historic migration estimates showed stronger connectivity from the Indian to the Atlantic Ocean (influenced by the Agulhas Leakage). In contrast, the individual‐based model coupled with contemporary migration estimates inferred from genetic data showed stronger inter‐ocean connectivity in the opposite direction from the Atlantic to the Indian Ocean driven by the Subtropical Front. We suggest that the J. tristani and J. paulensis historical distribution might have extended further north (when water temperatures were lower) resulting in larval dispersal between the ocean basis being more influenced by the Agulhas Leakage than the Subtropical Front. As water temperatures in the region increase in accordance with anthropogenic climate change, a southern shift in the distribution range of J. tristani and J. paulensis could further reduce larval transport from the Indian to the Atlantic Ocean, adding complexity to fisheries management. Antarctic krill (Euphausia superba) is a key species in the marine ecosystem of the Southern Ocean, but is also the target of a commercial fishery, with an important fishing ground in the South Orkney Islands region. The potential for competition for krill between predators and the fishery requires risk management strategies for the fishery, underpinned by an understanding of the key physical and behavioural drivers of krill movement and retention in target areas. Here, we present the results of a regional modelling study, combining a high-resolution ocean-sea ice model and an individual-based model parameterised for krill, to elucidate the roles of oceanographic variability and krill behaviour on patterns of transport and retention on and around the South Orkney Plateau. Simulations suggest that oceanic transport from sources around the Antarctic Peninsula is restricted by the northward flowing Antarctic Slope Current. Around the South Orkney Plateau, anticyclonic flows associated with the Weddell Front and the shelf edge transport krill rapidly towards the main fishing grounds to the northwest of the plateau. Transport onto the shelf and subsequent retention are influenced by the strength and direction of regional winds; weaker on-shelf transport and shorter retention times are associated with stronger westerly and northerly winds. The incorporation of sea-ice associated behaviour, whereby krill are moved with sea ice when sea ice is present, significantly modifies the patterns from purely oceanic transport; it reduces the influence of strong regional oceanic flows and increases transport of krill to the South Orkneys region from the Antarctic Peninsula. The inclusion of diel vertical migration also modifies predicted patterns from oceanic transport, but to a lesser extent, and moderates the impact of including sea-ice associated behaviour. This paper details a small-scale research project that explores the student transition into higher education (HE), delivered in a further education college (FE), and considers whether the requirements of transition at this level impacts on emotional wellbeing. As such, it aims to contribute to the growing body of research on HE-in-FE from the perspective of student transition and emotional wellbeing. The data drawn on in this paper were collected by a questionnaire, administered at two points in the first semester, to measure levels of anxiety in students, alongside a further questionnaire designed to evaluate the effectiveness of bespoke academic skills input aimed at supporting the transition into HE. From quantitative and qualitative data, key findings identify factors that influence students' emotional responses to HE, including those that exacerbate an those that mitigate negative emotional responses. Significantly, the research identifies academic factors, rather than personal or social issues, as being critical to students' emotional experiences during the transitionary period.
