ABSTRACT. Sesquiterpenoids are a class of terpenoid compounds with a remarkable diversity of structures and biological activities. Sesquiterpenoids are primarily found in higher plants, such as the Meliaceae family's Aglaia genus. Aglaia cucullata is a species of Aglaia that is still rarely explored and can potentially contain sesquiterpenoid compounds with cytotoxic activity. Hence, the research intended to isolate sesquiterpenoids from the n-hexane extract of A. cucullata stem bark and evaluate their cytotoxic effect against A549 lung cancer cells. Compounds 1 and 2 were isolated and purified from n-hexane extracts utilizing various chromatographic techniques. The structure of compounds 1 and 2 was determined by analyzing various spectroscopic methods (IR, MS, and NMR) and comparing them to previously reported spectral data. Compound 1 was identified as an isodaucane-type sesquiterpenoid, 10-hydroxy-6,10-epoxy-7(14)-isodaucane, and was first reported in Meliaceae family. Compound 2 was confirmed as an eudesmane-type sesquiterpenoid, eudesm-4(15)-ene-1β,6α-dihydroxy, and was first reported in Aglaia cucullata. Cytotoxic activity of 1 and 2 were investigated in vitro against A549 lung cancer cells using the PrestoBlue assay and resulted in inactive and low cytotoxicity with IC50 values of 292.77 and 90.55 μM, respectively. Mangrove-associated endophytic fungi are producers of secondary metabolites in unique and diverse structures with interesting biological activities such as antiviral, antifungal, antibacterial, anti-inflammatory, and cytotoxic agents. Endophytes play an important role in the physiological activities of the host plants, influencing the improvement of resistance to stress, insects, nematodes, and diseases. In this study, arohynapene A, a polyketide compound, was successfully isolated from the mangrove-derived fungus Penicillium steckii JB-NW-2-1 obtained from mangrove plant Avicennia marina (Forssk) Vierh. The structure was determined by a spectroscopic method including IR, MS, 1D-, and 2D-NMR techniques. This compound was evaluated for cytotoxic activities using resazurin assay against four cancer cells, HeLa cervical, MCF-7 breast cancer, B16-F10 melanoma, and A549 lung adenocarcinoma. The results showed no significant activities against all cancer cells tested (IC50 > 500 µM).

The Aglaia species, which contains triterpenoids, is the most numerous in the Meliaceae family. The A. cucullata species, of which there are only a few known examples, has received scant research attention. This investigation aims to identify triterpenoids in an n-hexane preparation of A. cucullata stem bark and evaluate their effects against the A549 lung cancer cell line. Five dammarane-type triterpenoids were isolated from the A. cucullata trunk bark, which is (1) (20S)-20-hydroxydammar-24-en-3-one, (2) cabraleone, (3) cabralealactone, (4) eichlerianic acid, and (5) (+)-fouquierol. Their chemical structures were determined using infrared, high-resolution mass spectrometry, and nuclear magnetic resonance, as well as through data comparison of the reported compounds. Compound 1 was priorly separated from the Aglaia genus, compounds 2–4 were first isolated from the A. cucullata species, and compound 5 has been reportedly isolated from the Meliaceae family and the Aglaia genus. All substances were tested for their lethal potential against the A549 lung cancer cell type. A seco structure in the A ring of dammarane-type triterpenoid might play an important part in the lethal activity of component 4, which showed the greatest activity with an IC50 value of 32.17 µM against the A549 lung cancer cell line.

With about 120 species, Aglaia is one of the largest genera of the plant family Meliaceae (the mahogany plants). Various Aglaia species have been investigated since the 1960s for their phytochemical constituents and biological properties. This research objective was to find secondary metabolites that have activity as anti-breast cancer compounds from endemic Indonesian Aglaia, such as Aglaia angustifolia Miq. Two cholestan type steroids, stigmast-5en-3α-acetat (1), as a new steroid with α-sterochemistry of acetyl moiety at C-3 and 23a-homostigmast-5en-3β-ol (2), with unusual side chain were isolated for the first time from the stem bark of Aglaia angustifolia Miq or known as segara tree in Kalimantan. The chemical structures of two steroids were identified with spectroscopic data, including IR, NMR (1H, 13C, DEPT 135°, HMQC, HMBC, NOESY, 1H-1H COSY) and HRTOF-MS, as well as by comparing with previously reported spectral data. These two steroids were isolated for the first time from this genus. Steroids 1 and 2 were evaluated for cytotoxic activity against MCF-7 breast cancer cells and showed weak activity with IC50 values of 829.0 and 903.0 µg/mL, respectively.

Sesquiterpenoids are derivatives of terpenoid compound that have a diverse skeleton and broad spectrum of biological activities, particularly anticancer activity. This research reported the isolation and elucidation structure of sesquiterpenoid from Chisocheton lasiocarpus stem bark, as well as their cytotoxic activity. Four sesquiterpenoid compounds, namely eudesm-4(15)-ene-1β,6β-diol (1), allo-aromadendrane-10α,14-diol (2), allo-aromadendrane-10β,14-diol (3) and guaianediol (4), were isolated for the first time from Chisocheton lasiocarpus stem bark. The chemical structure of all compounds was identified by spectroscopic analysis, including IR, MS, 1H-NMR, 13C-NMR and DEPT, and compared with previous reported spectral data. The cytotoxic activity of the isolated compounds against MCF-7 breast cancer line were examined by using the resazurin method. The results showed compound 1 shown the highest cytotoxic activity with IC50 108.08 ± 0.58 µM.

A new limonoid, belonging to the havanensin-type, called pentandricine E (1), alongside four other known types, i.e., trichilenone acetate (2), toonaciliatones C (3), 11α-acetoxyazadirone (4) and ceramicine I (5), were found in Chisocheton pentandrus stem bark. The structures of the isolated 1-5 were identified by NMR spectroscopy and HRTOFMS. Pentandricine E (1) displayed the highest cytotoxicity toward the MCF-7 breast cancer line with an IC50 value of 63.27 ± 0.21 mM.

Limonoid is derivatives of triterpenoid compound that have a wide variety of structures due to various ring-opening, rearrangements, and high-degree of oxidation. Limonoid is known as compounds that have wide-range of biological activities, including anticancer activity. This research was aimed to determine the chemical structure and cytotoxic activity of limonoid in the n-hexane extract of Chisocheton lasiocarpus fruit. Dried powder of C. lasiocarpus fruit was extracted using methanol followed by fractionated using n-hexane, ethyl acetate, and n-butanol. Five azadirone-type limonoids, 6α-(acetoxy)-14β,15β-epoxyazadirone (1), dysobinin (2), 7α-acetylneotricilenone (3), 6α-O-acetyl-7-deacetylnimocinol (4), and 7α-hyroxyneotricilenone (5), were isolated from the n-hexane extract of Chisocheton lasiocarpus fruit. The chemical structure of all compounds was identified by spectroscopic analysis, including 1D, 2D-NMR, IR, UV and HR-TOF MS as well as by comparison with previous reported spectra data. Compounds 1-5 were investigated from this plant for first time. The cytotoxic activity of the isolated compounds against MCF-7 breast cancer line were examined and the results showed that 7α-hyroxyneotricilenone (5) showed the moderate activity with IC50 values of 53 μM

Endophytic fungi reside within their host plants with no obvious symptoms. They have been proven as abundant sources of new bioactive compounds with diverse structures. In our continuous search of the new compound from endophytic fungi, a new compound, 5,7-dihydroxy-3(R)-methylphthalide (1), was isolated from Xylaria brevipes PGR1, and three known compounds, (–)-altenuene (2), alternariol (3), and altertoxin I (4), were isolated from Alternaria alternata D-8. Both endophytic fungi were isolated from the branch of an Indonesian medicinal plant, Psidium guajava. Their structures were elucidated using extensive spectroscopy analyses, including 1D/2D NMR, HRESITOFMS, and data comparisons with the literature. Compounds 1–3 exhibited phytotoxicity at IC50 (µg/ml) values of 4.6, 23.07, and 77.28, respectively. Additionally, 1–4 showed significant cytotoxicity in the brine shrimp lethality test (BSLT), with mortality rates at 75%, 50%, 90%, and 80% at the concentration of 0.19 μg/ml, respectively.

Four sesquiterpenoid derivatives, i.e., 4β,10α-dihydroxyaromadendrane (1), caryophyllenol-II (2), senecrassidiol (3), and clovane-2β,9α-diol (4) have been isolated from the stem bark of Aglaia simplicifolia. The chemical structures of compounds 1-4 were determined based on spectroscopic data, including one and two-dimensional NMR and mass spectroscopy. In addition, these sesquiterpenoids 1-4, were also tested for their cytotoxic activity against B16-F10 melanoma skin cancer cell lines through in vitro assay. Among the isolated compounds 1-4, compound 1 showed the highest activity with an IC50 value of 44.8 μg/mL, suggesting the presence of a cyclopropane ring that plays an essential role in cytotoxic activity against B16-F10 melanoma skin cancer cell lines.

Amomum compactum Soland Ex Maton is one of the Zingiberaceae family plants which is the endemic plants from West Java, Indonesia. This study was aimed to determine the chemical structure of flavonoid compounds from n-hexane extract of A.compactum Sol. Ex Maton roots. Dried powder of the roots was extracted consecutively with n-hexane, ethyl acetate, and methanol solvents. Three flavonoids, 5-hydroxy-3,7,4`-trimethoxy kaempferol (1), 5-hydroxy-3,7,3',4'-tetra methoxy kaempferol (2) and 4'-hydroxy-3,5,7-trimethoxy kaempferol (3), have been isolated from the roots of A. compactum Sol. Ex Maton. The chemical structures of compounds 1-3 were identified by spectroscopy data including infrared 1D-NMR, 2D-NMR and HRTOF-MS as well as by comparison with previously reported spectral data. Compounds 1-3 were isolated from this plant for the first time and showed free radical DPPH scavenging activity.

Limonoids (tetranortriterpenoids) are triterpenoid compounds that lose four terminals in their structural framework. These compounds have a wide variety of structures and interesting activities including anti-inflammatory, anti-cancer, anti-tumor and anti-malarial properties. The purpose of this study was to find limonoid compounds from the Indonesian Chisocheton plant, and one of which is Chisocheton macrophyllus. The dried and powdered seeds of C. macrophyllus (4.5 kg) were extracted with methanol and partitioned successively with n-hexane, ethyl acetate and n-butanol. Evaporation of each extract resulted in the crude extracts of n-hexane (346.6 g), ethyl acetate (60.8 g) and n-butanol (14.6 g). The n-hexane fraction was subjected to a silica gel vacuum-liquid chromatography (VLC) column packed with silica gel 60 using gradient of n-hexane, ethyl acetate and methanol (10% stepwise) to afford thirteen fractions (A-M). Fraction F (4.4 g) was subjected to silica gel column chromatography using gradient of n-hexane and ethyl acetate (5% stepwise). Subfraction F5 (1.2 g) was chromatographed on a column of silica gel eluted with n-hexane: CH2Cl2: EtOAc (2:7.5:0.5) to give compound 1 (19.7 mg) and fraction H (1.8 g) was subjected to silica gel column chromatography using gradient of n-hexane and ethyl acetate (5% stepwise) as eluting solvent to give 2 (12.0 mg). Chemical structures of 1 and 2 were elucidated by spectroscopic methods and determined as 6α-acetoxyepoxyazadiradione (1) and Dysobinin(2). Dysobinin (2) showed weak cytotoxic activity against MCF-7 breast cancer cells with an IC50 value of 228.15 μM

Three steroids, 3α-hydroxystigmast-5(6), 22-diene-7-one (1), stigmasterol (2) and 3-hydroxy-7β-methoxystigmast-5(6)-ene (3), were isolated from the stem bark of Dysoxylum nutans. The chemical structures were identified by spectroscopic data, which includes IR, 1D-NMR, 2D-NMR, and HR-TOFMS as well as by comparing previously reported spectral data. Compounds 1-3 were tested for cytotoxic effect against MCF-7 breast cancer cell lines and compound 1 showed the strongest cytotoxic activity with an IC50 value of 20.13 ± 0.06 μM.

Three aromadendrane-type sesquiterpenoids, spathulenol (1), 4β,10α-dihydroxyaromadendrane (2), and 4α,10α-dihydroxyaromadendrane (3) were isolated from the stem bark of Aglaia harmsiana (Meliaceae). Compound 3 was isolated for the first time from Aglaia genus. The chemical structures of isolated compounds were elucidated by various spectroscopic methods, including one and two-dimensional NMR, as well as mass spectroscopy analysis. These sesquiterpenoids 1-3 were evaluated for their cytotoxic activity against MCF-7 breast cancer cell lines. The IC50 value of compound 1-3 were 31.65 ± 0.1, 8.41 ± 0.04 and 2.80 ± 0.02 µM, respectively. Among the aromadendrane-type sesquiterpenoids, compounds 2 and 3, which do not have a double bond, showed higher activity than compound 1. Whereas, compound 3 showed the strongest activity indicate that α configuration of hydroxyl group increases the cytotoxic activity.

Two new dammarane-type triterpenoid fatty acid ester derivatives, 3β-oleate-20S-hydroxydammar-24-en (1) and 3β-oleate-20S,24S-epoxy-25-hydroxydammarane (2) with a known dammarane-type triterpenoid compound, such as 20S-hydroxydammar-24-en-3-on (3), were isolated from the stem bark of Aglaiaelliptica (C.DC.) Blume. The chemical structures were determined by spectroscopic methods, including FTIR, NMR (one and two-dimensional), and HRESITOF-MS analysis, as well as chemical derivatization and comparison with previous literature. Furthermore, the synthetic analog resulting from transesterification of 1 and 2 also obtained 3β,20S-dihydroxy-dammar-24-en (4) and 20S,24S-epoxy-3β,25-dihydroxydammarane (5), respectively. The cytotoxic effect of all isolated and synthetic analog compounds was evaluated using PrestoBlue reagent against MCF-7 breast cancer cell and B16-F10 melanoma cell lines. The 20S-hydroxydammar-24-en-3-on (3) showed the strongest activity against MCF-7 breast cancer and B16-F10 melanoma cell, indicating that the ketone group at C-3 in 3 plays an essential role in the cytotoxicity of dammarane-type triterpenoid. On the other hand, compounds 1 and 2 had very weak cytotoxic activity against the two cell lines, indicating the presence of fatty acid, significantly decreasing cytotoxic activity. This showed the significance of the discovery to investigate the essential structural feature in dammarane-type triterpenoid, specifically for the future development of anticancer drugs.

In the course of our continuing search for anticancer compounds from Chisocheton species, three steroids, stigmast-5-en-3β-ol (1), stigmast-5-en-3β-ol-3-O-β-D-glucopyranoside (2) and stigmast-5,22-dien-3β-ol-3-O-β-D-glucopyranoside (3), were obtained from the stembark of Chisocheton celebicus. The structures of compound 1-3 were identified with spectroscopic data including IR, 1D-NMR, 2D-NMR and TOF-MS, as well as by comparing with those spectral data previously. Compounds 1-3, were evaluated for their cytotoxic effects against P-388 murine leukemia cells and displayed the cytotoxicity activity with IC50 values of 12.45 ± 0.050, 52.27 ± 0.031 and 62.52 ± 0.076 µg/mL, respectively.

Sesquiterpenoid is a class of terpenoid compounds that have the most abundant diversity of structures and biological activitiesthat can be found in natural resources. Tropical plants are main source of sesquiterpenoid compounds such as Aglaiagenus belong to Meliaceae family. A. harmsianais a species from Aglaiathat only has few previous researchs. Therefore, the purpose of this study was to isolate and determine the structure of sesquiterpenoid compounds from stem barkA. harmsianaalong withtheir cytotoxic activity against MCF-7 breast cancer cells. The isolation process begins by extracting powder from A. harmsianastembark using n-hexane, ethyl acetate and methanol. All extracts were evaluated for their cytotoxic activity against MCF-7 breast cancer cells, and n-hexane extractsshowed significant cytotoxic activitywith IC50values of 117.86 µg/mL. Therefore, n-hexane extracts were further separated and purified by various chromatographic techniquesto obtain compounds 1and 2. Compounds 1and 2were elucidated their chemical structures by spectroscopic methods includingIR, NMR, and MS as well as bycomparison of data with literatures and identified ascaryophyllene-typesesquiterpenoids, β-caryophylleneoxide (1) and senecrassidiol (2). Compounds 1and 2were submitted for cytotoxic eveluation on MCF-7 breast cancer cells and as a result, β-caryophyllene oxide (1) showed the stronger activity compared to senecrassidiol (2). These finding indicatedthat the cytotoxic activity of caryophyllene-typesesquiterpenoid areinfluenced by the presence of double bonds and configuration of methyl groups.

Gambir (Uncaria gambir, Roxb.) contains catechins that is often empirically used to treat various diseases. Catechins can reduce cholesterol levels by inhibiting coenzyme HMG-CoA reductase that plays a role in cholesterol metabolism. Research has been carried out covering the optimization of transethosomal catechins, the formulation of Transethosomal Catechin Gel (TCG) and Non-Transethosomal Catechin Gel (NTCG), which were then tested for catechin permeation from these gel preparations in vitro using Franz’s diffusion cell with PTFE membranes. The anti-hypercholesterol activity test was carried out with Simvastatin orally as a positive control using 25 male Wistar rats (Rattus norvegicus). The catechin transetosomes have a size of 176.1 ± 5.8 nm, Zeta potential −11.6 ± 5.28, and Entrapment Efficacy of 96.77% ± 0.05. The result of cumulative catechins that permeated from TCG and NTCG were and 172.454 ± 5.287 and 112.741 ± 2.241 μg respectively. Permeation test graphs showed similar permeation and flux profiles. TCG can reduce total cholesterol and LDL (Low Density Lipoprotein) values in rats by 39.77% and 51.52% respectively during 14 days of use.

A new propylcholesterol-type steroid, namely (22E,24S)-24-propylcholest-5en-3α-acetate (1), has been isolated from the stembark of Aglaia angustifolia (Miq.). The structure of 1 was determined on the basis of spectroscopic data including 1D- and 2D-NMR as well as high resolution mass spectroscopy analysis. Compound 1 showed weak activity against the MCF-7 breast cancer cell line.

A known naphthalene, isoeleutherol (1), was isolated from the herb of Lygodium microphyllum. The chemical structure of 1 was determined on the basis of spectroscopic data mainly UV, IR, HRTOFMS, 1D- and 2D-NMR spectroscopy, as well as by comparing with compounds previously reported. Isoeleutherol was isolated from this plant for the first time and showed moderate antioxidant activity against DPPH (2,2-diphenyl-1-picrylhydrazyl) with IC50 value of 53.96 ± 2.87 µg/mL.

Six flavonoid compounds, kaempferol (1), quercetin (2), acacetin (3), quercetin-3-O-β-D-glucopyranoside (4), kaempferol-3-O-β-D-glucopyranoside (5), and isorhamnetin-3-O-β-D-glucopyranoside (6), were isolated from the methanol extract of the Lygodium microphyllum herb. The chemical structures of compounds 1-6 were identified on the basis of spectroscopic evidence and comparison with previously reported spectral data. Compounds 1-6 were isolated for first time from this plant. Their antioxidant activity against DPPH (2,2-Diphenyl-1-picrylhydrazyl) was evaluated. Among the compounds, quercetin (2) showed high antioxidant activity with IC50 values of 6.94 ± 0.03 μg/mL.

Two dammarane-type triterpenoids, 20S,24S-epoxy-3α,25-dihydroxydammarane (1) and 3α-acetyl-20S,24S-epoxy-3α,25-dihydroxydammarane (2), have been isolated from the stembark of Aglaia argentea. The chemical structure of compounds (1 and 2) were identified by spectroscopic evidences including UV, IR, 1D-NMR, 2D-NMR and MS as well as by comparing with previously reported spectral data. Those compounds were isolated from this plant for first time. Compounds (1 and 2) showed cytotoxic activity against P-388 murine leukemia cells with IC50 values of 23.96 and 8.14 mM, respectively. Four dammarane-type triterpenoid compounds, dammar-24-en-3a-ol (1), 3-epi-cabraleahydroxy lactone (2), (E)-25-hydroperoxydammar-23-en-3b,20-diol (3), and dammar-24-en-3β,20-diol (4), were isolated from the methanolic extract of the stembark of Aglaia argentea. Compounds 1-4 were isolated for first time from this plant. The structure of isolated compounds were elucidated by spectroscopic methods including one and two-dimensional NMR as well as mass spectrometric analysis. Compounds, 1-4, along with a known synthetic analog, 20-hydroxy-dammar-24-en-3-on (5), were evaluated their cytotoxic activity against P-388 murine leukemia cells in vitro. The IC50 values of compounds, 1-5 were 9.09 ± 0.10, 68.53 ± 0.08, 5.89 ± 0.08, 22.40 ± 0.11, and 11.53 ± 0.08 µg/mL, respectively. Among the dammarane-type triterpenoids, compounds 1, 3, 4 and 5 having opened side chain showed the stronger activity, wheres compound 2 with cyclic side chain showed weak or no activity. In addition, compound 3 showed strongest activity, indicate that hydroperoxy group at side chain increase cytotoxic activity.

Two mixtures of flavanoid compounds (1 and 2), a mixture of catechin (1a) and epicatechin (1b), and a mixture of gallocatechin (2a) and epigallocatechin (2b), were isolated from the active fraction of the stem bark of Aglaia elliptica methanol extract. The chemical structure of the compounds was identified with spectroscopic data, including UV, IR, NMR (1H, 13C, DEPT 135°, HMQC, HMBC, 1H-1H COSY), and MS, and additionally compared with previously reported spectral data. All compounds were evaluated for their cytotoxic effects against P-388 murine leukemia cells. Compound 2 showed cytotoxicity against the P-388 murine leukemia cell, with an IC50 value of 7.79 µg/mL, but compound 1 was found not to be active (more than 100 µg/mL).

The Aglaia genus belong to Meliceae family is unique plant species because the presence of rocaglate and rocaglamide which is so far isolated only from Aglaia genus, indicate that type of this compound as a chemical marker for the genus of Aglaia. This type of compound known to have strong activity, such as insecticide and cytotoxic. This study describe the isolation, structure elucidation, and cytotoxic activity of an isolated rocaglate compound. Dried stembark of A. argentea extracted with methanol and partition between n-hexane, ethyl acetate, and n-butanol, respectively The extracts were tested against P-388 murine leukemia cells and the ethyl acetat showed strongest activity with IC50 value of 15.5 mg/mL. The ethyl acetate then was separated and purified with chromatography technique to obtain isolated compound 1. The chemical structure of isolated compounds were elucidated by spectroscopic methods including one and two-dimensional NMR as well as high-resolution mass spectrometric analysis and identified as a methyl rocaglate. Compound 1 showed strong cytotoxic activity with an IC50 value of < 0.1 μg/mL.

Two flavonoid compounds, quercetin (1) and quercetin-3-O-glucoside (2) have been isolated from the bark of Sonneratia alba (Lythraceae). Chemical structure of both compounds were determined on the basis of spectroscopic data and comparison with those spectra data previously reported. Compound 1 and 2 exhibited antibacterial activity against Gram-positive bacteria, Staphylococcus aureus and Streptococcus mutans with MIC values of 51.2; 48.8; 72.5; dan 100.7 µg/mL respectively. Methyl 3,4-dicarboxy-3-hydroxyeicosanoate (1) was isolated from the acetone extract of the fruiting bodies of Ciborinia camelliae Kohn. The planar structure of 1 was elucidated by spectroscopic methods.

A novel N-benzyl-(3E,5E)-3,5-bis(2-hydroxybenzylidene)-4-piperidone (3), was synthesized in good yield by a condensation reaction of 2-hydroxybenzaldehyde (1) and N-benzyl-4-piperidone (2) under microwave irradiation in the presence of 10% NaOH solution. The chemical structure was assigned on the basis of UV-visible, IR, 1H-NMR, 13C-NMR and mass spectral data.

The title compound, 3,5-bis(2-hydroxybenzylidene)piperidin-4-one (3), was prepared via reaction of 2-hydroxybenzaldehyde (1) and 4-piperidone (2) under microwave irradiation in the presence of 10% NaOH solution. The compound was fully characterized from its UV, IR, NMR and MS data.

This work was conducted to isolate the steroid compounds from an endophytic fungus Cladosporium anthropophilum and determine their effects against 4 strains of bacteria. The fungi were afforded from a mangrove plant Avicennia marina (Forssk.) Vierh of Acanthaceae family, growing in West Java, Indonesia. The fermentation and isolation of C. anthropophilum yielded 3 ergostane-type steroids including a highly oxidized one, identified as penicisteroid A (1), ergosterol (2), and ergosterol-5,8-peroxide (3), together with 2 stigmastanes, characterized as stigmasterol (4), and stigmasterol-5,8-peroxide (5). The elucidation structure of all isolated steroids was performed by extensive spectroscopic measurements (MS, IR, 1D, and 2D NMR) and supported by a comparison of previously reported spectral data. Subsequently, the potential of the 5 steroids as well as methanol, n-hexane, and ethyl acetate extracts were evaluated against 2 Grampositive bacteria, Staphylococcus aureus ATCC 29213 and Enterococcus faecalis ATCC 29212 and 2 Gram-negative bacteria, Vibrio harveyi ATCC 5339 and Escherichia coli ATCC 25922. The results showed that none of the isolated steroids were active, while the 3 extracts exhibited significant inhibition against all the tested bacteria's growth and enabled us to propose the synergistic effects between active compounds in these extracts and their antibacterial activity. Sesquiterpenoids are a class of terpenoid compounds with a remarkable diversity of structures and biological activities. Sesquiterpenoids are primarily found in higher plants, such as the Meliaceae family's Aglaia genus. Aglaia cucullata is a species of Aglaia that is still rarely explored and can potentially contain sesquiterpenoid compounds with cytotoxic activity. Hence, the research intended to isolate sesquiterpenoids from the n-hexane extract of A. cucullata stem bark and evaluate their cytotoxic effect against A549 lung cancer cells. Compounds 1 and 2 were isolated and purified from n-hexane extracts utilizing various chromatographic techniques. The structure of compounds 1 and 2 was determined by analyzing various spectroscopic methods (IR, MS, and NMR) and comparing them to previously reported spectral data. Compound 1 was identified as an isodaucane-type sesquiterpenoid, 10-hydroxy-6,10-epoxy-7(14)-isodaucane, and was first reported in Meliaceae family. Compound 2 was confirmed as an eudesmane-type sesquiterpenoid, eudesm-4(15)-ene-1β,6α-dihydroxy, and was first reported in Aglaia cucullata. Cytotoxic activity of 1 and 2 were investigated in vitro against A549 lung cancer cells using the PrestoBlue assay and resulted in inactive and low cytotoxicity with IC50 values of 292.77 and 90.55 μM, respectively. The phytochemical investigation of Dialium corbisieri seeds led to the isolation of five monoterpenoid indole alkaloids along with a phytoserotonin, 1-6 and among the known compounds, the spectroscopic data of (5S)-methoxy-akuammiline (1) was reported for the first time. The structures were elucidated based on nuclear magnetic resonance spectroscopic techniques such as ultraviolet, infrared, high-resolution electrospray ionization time-of-flight mass spectrometry, and electron-capture dissociation spectrum calculations.
