Whether 7,8-dihydroxyflavone (7,8-DHF), a tyrosine kinase receptor B (TrkB) agonist, modulates colonic smooth muscle motility and/or alleviates constipation has not yet been studied. Here, we aimed to determine how 7,8-DHF influences carbachol (CCh)-stimulated contraction of colonic strips and the in vivo effect of 7,8-DHF on constipation. Muscle strips were isolated from rat colons for recording contractile tension and performing western blotting. Constipation was induced in rats with loperamide. Although it specifically activated TrkB, 7,8-DHF applied alone neither activated PLCγ1 in the colonic strips nor induced colonic strip contraction. However, 7,8-DHF enhanced CCh-stimulated PLCγ1 activation and strip contraction. The PLCγ1 antagonist U73122 suppressed both CCh-stimulated and 7,8-DHF-enhanced/CCh-stimulated contraction. While clarifying the underlying mechanism, we revealed that 7,8-DHF augmented muscarinic M3 receptor expression in the colonic strips. The M3-selective antagonist tarafenacin specifically inhibited the 7,8-DHF-enhanced/CCh-stimulated contraction of the colonic strips. Since 7,8-DHF increased Akt phosphorylation, and LY294002 (an antagonist of PI3K upstream of Akt) dramatically inhibited both 7,8-DHF-augmented M3 expression and 7,8-DHF-enhanced/CCh-stimulated contractions, we assumed that 7,8-DHF/TrkB/Akt was associated with the modulation of M3 expression in the colonic strips. ANA-12, a specific TrkB antagonist, not only inhibited TrkB activation by 7,8-DHF but also suppressed 7,8-DHF-enhanced cholinergic contraction, 7,8-DHF/CCh-mediated activation of PLCγ1/Akt, and M3 overexpression in colonic strips. In vivo 7,8-DHF, also by promoting intestinal motility and M3 expression, significantly alleviated loperamide-induced functional constipation in rats. Our results suggest that 7,8-DHF regulates colonic motility possibly via a TrkB/Akt/M3 pathway and may be applicable for alleviating constipation.

Objective: Current evidences on the association between hyperuricaemia and retinol intake remain inconsistent. Furthermore, no known studies have investigated the relationship between hyperuricaemia and retinol intake from animal food and plant food separately. This study aimed to assess the relationship between different sources of retinol intake and risk of hyperuricaemia among US adults. Design: Univariate and multivariate weighted logistic regression models and restricted cubic spline models were used to assess the associations of total, animal-derived and plant-derived retinol intakes with the risk of hyperuricaemia. Dietary retinol was measured through two 24-h dietary recall interviews. Hyperuricaemia was defined as serum uric acid level ≥7·0 and ≥6·0 mg/dl in men and women, respectively. Setting: Data from the National Health and Nutrition Examination Survey 2009–2014 were used in this cross-sectional study. Participants: Overall, 12 869 participants aged ≥20 years were included. Results: Compared with the lowest quintile, the multivariable OR of hyperuricaemia for the highest quintile intake of total, animal-derived and plant-derived retinol were 0·71 (95 % CI 0·52, 0·96), 0·76 (95 % CI 0·59, 0·96) and 0·92 (95 % CI 0·72, 1·17), respectively. The inverse association between dietary intake of total retinol and the risk of hyperuricaemia was observed in men. Dose–response analyses revealed a novel linear trend between the risk of hyperuricaemia and total, animal-derived retinol intake separately. Conclusions: Our findings indicated that intakes of total and animal-derived retinol were negatively associated with hyperuricaemia in US adults.

Background: Currently available evidence on the association between dietary iron intake and hyperuricemia is limited and inconsistent. Objective: This study aimed to examine the relationships between animal-derived dietary iron (ADDI) intake, plant-derived dietary iron (PDDI) intake, and the ratio PDDI:ADDI and hyperuricemia risk among US adults. Design: Data from the National Health and Nutrition Examination Survey (NHANES) 2009–2014 were used. Iron intake from diet was assessed through two 24-h dietary recalls. Logistic regression models and restricted cubic spline models were used to investigate the associations between dietary iron intake from different sources and hyperuricemia risk. Results: A total of 12,869 participants aged ≥20 years were enrolled in the study. After adjustment for multiple confounders, relative to the lowest quartile, the odds ratios (ORs) with 95% confidence intervals (CIs) of hyperuricemia for the highest quartile of ADDI intake, PDDI intake, and the PDDI:ADDI intake ratio were 1.11 (0.90–1.38), 0.69 (0.55–0.87), and 0.85 (0.67–1.07), respectively. Dose–response analysis revealed that the risk of hyperuricemia was negatively associated with PDDI intake in a linear manner. Conclusion: PDDI intake was inversely associated with hyperuricemia in US adults.

Introduction: Current evidence on the association between dietary vitamin E intake and hyperuricemia risk is limited and conflicting. Objective: The aim of the study was to assess the association of dietary vitamin E intake with hyperuricemia in US adults. Methods: We conducted a cross-sectional study using data from the National Health and Nutrition Examination Survey, 2009–2014. Dietary vitamin E intake was evaluated through two 24-h dietary recall interviews. Logistic regression and restricted cubic spline models were used to examine the association between dietary vitamin E intake and hyperuricemia. Results: Overall, 12,869 participants were included. The prevalence of hyperuricemia was 19.35%. After adjustment for age, gender, BMI, race, educational level, smoking status, alcohol consumption, physical activity, total daily energy intake, total cholesterol, protein intake, glomerular filtration rate, serum Cr, use of uric acid drugs, and drug abuse, the odds ratio (95% confidence interval) of hyperuricemia for the highest tertile of dietary vitamin E intake was 0.77 (0.63–0.96) compared with that of the lowest tertile. In men, dietary vitamin E intake and hyperuricemia were negatively correlated. In stratified analyses by age (20–39, 40–59, and ≥60 years), dietary vitamin E intake was inversely associated with hyperuricemia only among participants aged ≥60 years. Dose-response analyses showed that dietary vitamin E intake was inversely associated with hyperuricemia in a nonlinear manner. Conclusion: Dietary vitamin E intake was negatively correlated with hyperuricemia in US adults, especially among males and participants aged ≥60 years.

Objective:Previous studies evaluating the associations of circulating Zn and Se levels with asthma have produced inconsistent results. Therefore, we conducted a meta-analysis to summarize and quantitatively synthesize the evidence from observational research.Design:Meta-analysis.Setting:We searched PubMed, Web of Science and Scopus databases up to May 2019 for relevant available articles. Random-effects model was adopted to estimate the pooled standardized mean difference (SMD) with 95 % CI. Meta-regression analysis and ‘leave-one-out’ sensitivity analysis were used to assess heterogeneity.Participants:The meta-analysis focused on general populations.Results:A total of twenty-six studies for Zn and forty studies for Se were included in the meta-analysis. The overall analyses identified that asthma patients had lower Zn (SMD = −0·40; 95 % CI −0·77, −0·03; I2 = 94·1 %) and Se (SMD = −0·32; 95 % CI −0·48, −0·17; I2 = 90·9 %) levels in serum or plasma compared with healthy controls. After removing the studies that contributed to the heterogeneity, the pooled SMD were −0·26 (95 % CI −0·40, −0·13; I2 = 37·42 %) for Zn and −0·06 (95 % CI −0·13, 0·02; I2 = 43·54 %) for Se.Conclusions:Lower circulating Zn and Se levels might be associated with an increased risk of asthma.

The differences of dietary iron and zinc intakes between patients with nonalcoholic fatty liver disease (NAFLD) and controls remain controversial. The meta-analysis aimed to explore the differences of dietary iron and zinc intakes between NAFLD patients and healthy subjects.A systematic literature search was performed up to July 2021 in databases of PubMed, Embase, Web of Science, Cochrane Library, Chinese National Knowledge Infrastructure (CNKI), and Wanfang. Using a randomeffects model, the differences of dietary iron and zinc intakes between cases and controls were calculated as standardized mean differences (SMDs) with 95% confidence intervals (CIs). A total of 21 studies from 19 articles with 6639 cases were included.The pooled estimate showed no difference in dietary iron consumption in the NAFLD groups compared with control groups. The difference became significant in Asia (SMD=0.16; 95% CI: 0.04, 0.28; I2=89.1%; pheterogeneity0.001) as well as in cross-sectional studies (SMD=0.12; 95% CI: 0.07, 0.17; I2=4.7%; pheterogeneity=0.350). The difference in dietary zinc intake between cases and controls was not significant. We noticed a statistically significant increase of dietary zinc intake in NAFLD compared to controls in studies using food-frequency questionnaire (FFQ) to evaluate dietary intake (SMD=0.15; 95% CI: 0.10, 0.20; I2=12.2%; pheterogeneity=0.332).Our findings indicated that dietary iron intake in patients with NAFLD was higher than healthy subjects in Asia.

To research the effect of sodium alginate intervention on the nutritional intake of healthy young men.Recruited 20 healthy university students as research subjects. The experiment was divided into two periods: the dietary balance period and the sodium alginate intervening period, and each period was expected to be 28 days. During the dietary balance period, all meals in every day of experiment are provided by the research team; during the sodium alginate intervening period, based on the diet during the balanced diet period, sodium alginate was added to the staple food steamed buns(10 g sodium alginate per person per day). The experiment compares the food intake types, main nutrient intake levels and serum amino acid changes of subjects before and after intervention.Adding sodium alginate can significantly reduce the intake of energy(-242.4 kcal), protein(-11 g)and carbohydrates(-47.3 g, Plt;0.05)in healthy subjects, but there was no significant effect on the intake of fat(-2.9 g, P=0.496)and cholesterol(-14.9 mg, P=0.070), and because of the addition of alginate, the whole dietary fibers obtained a significant increase(+7.8 g, Plt;0.05). After the intervention of sodium alginate, there was no significant change in the intake of rice, soy products, poultry products and vegetable oil, while the intake of wheat products(-49.6 g), egg foods(-2.6 g), dark-colored vegetables(-29.1 g), light-colored vegetables(-63.8 g)and fruits(-37 g)decreased significantly(Plt;0.05). Most of the essential amino acids in the subjectsapos; serum increased significantly after the intervention, especially the valine in serum rise from 226.9 μmol/L to 466.4 μmol/L(Plt;0.05).Sodium alginate can play a dietary fiber-like effect, produce satiety, reduce nutrient intake of subjects, especially carbohydrates, so sodium alginate has the potential to limit energy intake and control postprandial blood sugar. And sodium alginate also has a potential positive effect on the metabolism of amino acids in healthy people, especially the metabolism of essential amino acids.

PUFAs play critical roles in the development of nonalcoholic fatty liver disease (NAFLD). This study examined the associations between dietary n-3 and n-6 PUFA intake and NAFLD risk in a US population.Data from the National Health and Nutrition Examination Survey (NHANES) 2007-2014 was used in this cross-sectional study. Data on dietary n-3 and n-6 PUFAs were extracted through two 24-h dietary recall interviews, and the dietary n-3 and n-6 PUFA intakes were adjusted by weight. NAFLD was defined based on the US fatty liver index (FLI) value ≥30. Multivariable logistic regression models and restricted cubic spline models were applied to investigate the associations between dietary n-3 and n-6 PUFA intakes and NAFLD risk.Dietary n-3 and n-6 PUFA intakes were inversely associated with NAFLD risk. The multivariable-adjusted OR (95% CI) of NAFLD for the highest versus lowest quartile of dietary n-3 and n-6 PUFA intakes was 0.24 (0.17-0.35) and 0.18 (0.13-0.26), respectively. In stratified analyses by sex and age, the negative associations between dietary n-3 and n-6 PUFA intakes and NAFLD risk were significant in men, women, and individuals younger and older than 45 years. Dose-response analyses indicated that NAFLD risk was associated with dietary n-3 and n-6 PUFA intakes in a nonlinear manner.Dietary n-3 and n-6 PUFA intakes were inversely associated with NAFLD risk in US adults.

To investigate the effects of lycopene on metabolism of glycolipid and inflammation in rats with non-alcoholic fatty liver disease.According to body weight, sixty male Sprague-Dawley rats were randomly divided into control group, non-alcoholic fatty liver model group, 20, 60 mg/kg lycopene intervention groups, with 15 rats in each group. The control group was fed normal diet, and the other 3 groups were fed high-fat and high-fructose diet for 4 weeks to establish the model of non-alcoholic fatty liver disease. After modeling, the intervention groups were fed lycopene of different doses by gavage. After 8 weeks of continuous feeding, the rats were sacrificed, the body weight and liver weight were weighed, and the liver index was calculated. Hematoxylin-eosin staining was used to observe the morphological changes of liver tissue. Serum was collected, fasting blood glucose(FBG), fasting insulin(INS), and insulin resistance index(HOMA-IR) were calculated; Liver function indicators alanine aminotransferase(ALT), aspartate aminotransferase(AST) were measured; Serum triglycerides(TG), total cholesterol(TC), high density lipoprotein-cholesterol(HDL-C), low density lipoprotein-cholesterol(LDL-C) and levels of inflammatory cytokines interleukin-6(IL-6), interleukin-18(IL-18) and interleukin-1β(IL-1β) were measured.Compared with the control group, the liver weight and liver index in the model group increased by 27% and 24%, respectively; And steatosis occurred in the liver tissue; The levels of serum ALT, TG, TC, LDL-C, IL-6, IL-1β were significantly increased, the level of serum HDL-C was significantly decreased(Plt;0. 05). Compared with the model group, liver weight, liver index, levels of serum ALT, TG, FBG, IL-6 and IL-1β were significantly lower in the 20, 60 mg/kg lycopene intervention groups(Plt;0. 05), the INS and HOMA-IR index showed a downward trend; Liver tissue lesions were reduced to different degrees, and the effect was more significant in the 60 mg/kg lycopene group.Lycopene can improve non-alcoholic fatty liver disease by regulating glycolipid metabolism and reducing levels of inflammatory cytokines in rats.

Non-alcoholic fatty liver disease (NAFLD) in normal-weight population is becoming an important health issue. This study aimed to assess the association between serum ferritin (SF) and NAFLD among middle-aged and elderly Chinese with normal weight.A total of 2029 Chinese adults aged 35-70 years with normal weight were involved in this cross-sectional study. General information, lifestyle factors and laboratory characteristics were collected. The concentrations of serum alanine aminotransferase (ALT) and SF were recorded. Receiver operating characteristic (ROC) was applied to assess predictive performance of SF for NAFLD. Logistic regression analysis was conducted to evaluate the associations of SF with NAFLD and elevated ALT.Compared with controls, subjects with NAFLD had higher SF concentrations (p0.001). In multivariate logistic regression analyses, the odds ratios (ORs) with 95% confidence intervals (CIs) of NAFLD were 3.19 (2.07-4.92) for the highest versus lowest quartile of SF. ROC analysis revealed a predictive ability of SF for NAFLD with an area under the curve of 0.660 (95% CI, 0.63-0.69). In addition, higher SF was significantly associated with increased risk of elevated ALT (OR=1.84, 95% CI: 1.32-2.55). In stratified analyses by gender and age, the positive associations of SF with the risk of NAFLD and elevated ALT were only observed in women and 35~49y group.SF was positively associated with the risk of NAFLD and elevated ALT among normal-weight Chinese adults. ROC analysis suggested that SF may serve as an indicator of predicting NAFLD.

Objective Restless legs syndrome (RLS) has been reported to occur more frequently in obese individuals than in those with normal weight. In this study, a systematic review and meta-analysis was performed to explore the relationship between obesity and RLS. Methods Published articles were identified through a comprehensive review of PUBMED and EMBASE from inception to the 16th December 2017. Data from studies reporting relative risks, odds ratios, or hazard ratios comparing the risk of RLS among individuals with obesity versus people with normal weight were analyzed. A random-effect model was used to calculate pooled odds ratios and 95% confident intervals (CI) for the association between obesity and RLS. Results Findings for a total of 197,204 participants were pooled across 15 studies and were examined. Our analyses demonstrated a positive association between obesity and RLS, with an odds ratio (OR) of 1.44 (95%CI: 1.31–1.58, I2 = 62.3%). Overweight subjects were also likely to have RLS, with an odds ratio of 1.29 (95%CI: 1.22–1.36, I2 = 0). In subgroup analysis, women (OR = 1.42) with obesity were more likely to have RLS as compared with men (OR = 1.19). Conclusions Adults with obesity are more likely to suffer from RLS, with women at higher risk.

Current evidence on the relationship between dietary fiber intake and risk of hyperuricemia is limited. The aim of the present study was to examine their associations in the U.S. general adult population. Data from the National Health and Nutrition Examination Survey (NHANES) 2009 to 2014 were used. Dietary fiber intake was extracted through two 24 h dietary recall interviews. Hyperuricemia was defined by cut-off values of 7.0 mg dL−1 for men and 6.0 mg dL−1 for women. Multivariable logistic regression models and restricted cubic spline models were applied to explore the associations between dietary intakes of total, cereal, fruit and vegetable fiber and the risk of hyperuricemia. A total of 12 869 participants aged 20 years or older were included in the present study. The multivariable odds ratio (OR) and 95% confidence interval (CI) of hyperuricemia for the highest vs. lowest quartile intake of total fiber were 0.58 (0.46–0.74), 0.61 (0.52–0.74) for cereal fiber, 0.94 (0.76–1.16) for fruit fiber and 0.95 (0.76–1.18) for vegetable fiber. Objectives To evaluate the dose-response relationships between alcohol consumption and functional limitations in older European men, and explore the role of muscle strength as a mediator of these relationships. Design Cross-sectional study of older men participating in the Survey of Health, Aging and Retirement in Europe (SHARE). Setting Urban and rural households in 17 European countries and Israel. Participants A total of 17 870 men aged 65 years and older from the SHARE (Wave 6, 2015) were included in this study. Measurements Outcome variables were functional limitations: mobility limitation, arm function limitation, and fine motor limitation. Main exposure variable was alcohol consumption. Mediating factor was grip strength. Basic demographics, life habits, and health status were considered as potential confounders. Dose-response analyses with restricted cubic splines and the Karlson/Holm/Breen method were conducted. Results A total of 17 870 participants were included in this study. Dose-response analyses revealed that moderate alcohol consumption was related to the lower odds of reporting mobility limitation (≤35 units/wk) and arm function limitation (≤41 units/wk), with a minimum odds ratio (OR) occurring at 10 units/week drinks for mobility limitation (OR = .71; 95% confidence interval [CI] = .62-.81) and arm function limitation (OR = .66; 95% CI = .59-.75). The odds of reporting the fine motor limitation monotonically increased with alcohol consumption when alcohol consumption was beyond 15 units/week. No significant mediating effect of grip strength on the relationships between alcohol consumption and mobility limitation and arm function limitation was found. Conclusion Moderate alcohol consumption has a protective role in mobility and arm function limitation in older European men. Grip strength is not the main mediator of these associations, suggesting that the protective effect is independent of muscle strength. The relationship between vitamin C intake and hyperuricemia among the general US adult population has seldom been reported; thus, the present study examined the associations of total vitamin C (dietary vitamin C plus supplementary vitamin C) and dietary vitamin C intake with the risk of hyperuricemia.Pooled data from three 2-year cycles (2007-2012) of the cross-sectional National Health and Nutrition Examination Survey were used in the present study. Dietary intake data were extracted from two 24-hour dietary recall interviews. Logistic regression models were used to determine the associations between vitamin C intake and hyperuricemia risk.A total of 14885 adults aged 20 years or older (7269 men and 7616 women) were registered in the present study. The prevalence of hyperuricemia was 19.1%. Based on the lowest quartile of dietary vitamin C intake, multivariate adjusted odds ratios with 95% confidence intervals of hyperuricemia for quartiles 2-4 were 0.84 (0.74-0.95), 0.83 (0.73-0.94), and 0.72 (0.63-0.82), and those for total vitamin C intake were 0.87 (0.77-0.99), 0.85 (0.75-0.96), and 0.66 (0.58-0.76). Inverse associations between vitamin C intake and hyperuricemia were discovered in both men and women, even with or without covariate adjustments.Total vitamin C and dietary vitamin C intake are inversely associated with hyperuricemia in the general US adult population.

Aims: The effect of algae and its extract supplementation on glycolipid metabolism has not been finalized. Therefore, the purpose of the meta-analyses was to assess the effects of its supplementation on glycolipid metabolism concentration. Methods: We have systematically searched PubMed, Web of Science, the Cochrane Library and Embase to identify randomized controlled trials (RCTs) that investigated the impact of algae and its extracts supplementation on glycolipid metabolism. Effect size analysis was performed using weighted mean difference (WMD) and 95% CI between the methods of the experiment group and the control group. Subgroup analyses were performed to explore the possible influences of study characteristics. Publication bias and sensitivity analysis were also performed. Results: A total of 27 RCTs (31 trials) with 1221 participants were finally selected for the meta-analysis. The algae and its extract intervention significantly decreased glycosylated hemoglobin (HbA1c, WMD = −0.18%; 95% CI: −0.27 to −0.10; p < 0.001), high-density lipoprotein cholesterol (HDL-C, WMD = −0.22 mmol/L; 95% CI: −0.38 to −0.06; p = 0.008), and triglycerides (TC, WMD = −0.31 mmol/L; 95% CI: −0.37 to −0.25; p < 0.001) levels and increased insulin (WMD = 6.05 pmol/mL; 95% CI: 4.01 to 8.09; p < 0.001) levels. It did not significantly change the blood glucose, homeostasis model assessment-insulin resistance index (HOMA-IR), 2-h post-meal blood glucose (2hPBG) and other lipid profiles. Subgroup analyses based on the duration of intervention and subjects demonstrated that the intervention of algae and its extracts for 10 weeks or fewer and more than 40 subjects decreased TC levels (p < 0.05). Moreover, the intervention reduced TC and 2hPBG concentrations for East Asians (p < 0.05). Conclusions: Our findings provided evidence that algae and its extract interventions were beneficial for the regulation of human glycolipid metabolism. More precise RCTs on subjects are recommended to further clarify the effect of algae, seaweed polysaccharide, seaweed polypeptide, algae polyphenol and its products intervention on glycolipid metabolism.

To analyze the effect of vitamin E( Vit E) and magnesium( Mg) supplementation on stability of erythrocyte and antioxidant capacity in obese rats.Seventy Wistar rats were randomly divided into five groups. Each group was supplied defferent doses of Vit E and Mg. All rats were sacrificed after 67 weeks. Erythrocyte osmotic fragility, fluidity of erythrocyte membrane, erythrocytic hemolyses, superoxide dismutase( SOD), malondialdehyde( MDA) and glutathione peroxidase( GSH-Px) were detected. Results of each group were analyzed by One-Way ANOVA andanalysis of variance of factorial design.The values of fluorescence polarization( P) and microviscosity( η) of obese model were( 0. 096 ± 0. 024) and( 0. 543 ±0. 179), respectively, which were significantly higher than other groups( P0. 05), it could indicate the fluidity of erythrocyte membrane of obese model was lower than other goups. The erythrocytic hemolyses of obese model was( 33. 57 ± 7. 50) %. Compared with other groups, it was increased by 11. 18%, 10%, 11. 4% and 18. 13%, respectively. The level of SOD of obese model was( 97. 54 ± 24. 81) U/mL which was lower than other groups( P0. 05). The level of MDA of obese model was( 17. 18 ± 12. 73) nmol/mL which was higher than other groups( P0. 05). Compared with normal controls and obese model, the level of GSH-Px of the group which was adding vitamin E was increased by859. 26 U, 955. 42 U, respectively.Both of Vit E and Mg could improve fluidity of erythrocyte membrane and erythrocytic hemolyses of obese rats. Combined use of Vit E and Mg could effectively improve fluidity of erythrocyte membrane. Vit E or Mg can improve the antioxidant capacity in obese rats.

The associations between sarcopenia and metabolic syndrome (MetS) in non-obese middle-aged and older adults remain controversial. Thus, this meta-analysis aimed to evaluate the overall prevalence of MetS and the correlations between sarcopenia and MetS in middle-aged and older non-obese adults. We performed a systematic searched strategy using PUBMED, EMBASE and Web of Science databases for relevant observational studies investigating sarcopenia and MetS up to 11 May 2017. The polled prevalence of MetS and odds ratios with 95% confidence intervals (CI), as well as subgroup analyses were calculated using a random effects model. Twelve articles with a total of 35,581 participants were included. The overall prevalence of MetS was 36.45% (95% CI, 28.28–45.48%) in middle-aged and older non-obese adults with sarcopenia. Our analysis demonstrated a positive association between sarcopenia and MetS (OR = 2.01, 95% CI, 1.63–2.47). The subgroup analysis showed that both larger cohort size and the use of dual-energy X-ray absorptiometry to measure body composition can enhance the relationship. Our study revealed that a higher proportion of MetS in middle-aged and older non-obese people with sarcopenia. Moreover, sarcopenia was positively associated with MetS in this population. Further large-scale prospective cohort studies are needed to investigate the causality between sarcopenia and MetS.

PURPOSE We evaluated the association between age-related macular degeneration (AMD) and the risk of all-cause and cardiovascular mortality by meta-analyses of data from prospective studies. METHODS A literature search was performed in PubMed, Web of Science, Embase, Cocharne Library, and China National Knowledge Infrastructure for relevant articles published up to December 2016. We estimated hazard ratios with 95% confidence intervals with fixed-effect models and conducted meta-regression to explore the potential sources of heterogeneity. Small-study effect was estimated by Egger's test and funnel plot. RESULTS We identified 13 population-based prospective cohort studies that examined the relationship between AMD and all-cause and cardiovascular mortality. Overall, the hazard ratios (95% confidence intervals) of all-cause mortality and cardiovascular mortality associated with any AMD were 1.15 (1.05-1.27) and 1.05 (95% confidence intervals: 0.87-1.26), respectively. The risk of all-cause mortality and cardiovascular mortality associated with early AMD were 1.08 (1.00-1.18) and 1.05 (0.89-1.24), and the associations with late AMD were 1.23 (1.11-1.36) and 1.28 (1.04-1.57), respectively. No evidence of small-study effect was found. CONCLUSION This meta-analysis indicated that AMD, especially late AMD, was associated with increased risk of all-cause mortality and cardiovascular mortality based on comparisons with people who did not have AMD and who were of similar age and sex.

Background Uric acid (UA) is a powerful antioxidant that may have neuroprotective properties, yet it is also a risk factor of vascular disease that predisposes individuals to cognitive impairment. Results from longitudinal studies on UA and cognitive decline remain controversial. Objective We examined the associations of baseline plasma UA level with follow-up cognitive function as well as cognitive decline over time among a large sample of middle-aged and older Chinese. Methods Data from China Health and Retirement Longitudinal Study (CHARLS) were used. Cognitive function, including episodic memory, mental intactness, and global cognition, were tested twice with 2-year interval. Plasma UA was measured at baseline. Basic demographics, life habits, and health status were considered as potential confounders. Multiple linear regression models and mixed-effects regression models were fitted. Results A total of 12,798 individuals aged above 45 years were eligible with the follow-up time ranging from 1.33 to 2.42 years. Both global cognitive function and mental intactness declined, while episodic memory remained stable over time. In multiple linear regression models, compared with the lowest baseline UA level, 3rd baseline UA quartile was associated with better follow-up global cognitive function (b = 0.425, p = 0.041) and episodic memory (b = 0.413, p = 0.004), and highest baseline UA quartile was associated with better follow-up mental intactness (b = 0.253, p = 0.041) in males; highest baseline UA level was associated with better follow-up cognition for each measure (b = 0.281∼0.768, p≤0.046) in females. Mixed-effects regression models suggested no significant baseline UA-by-time interactions on any cognitive measure. Conclusion Higher baseline UA level was associated with better cognition in later life but not with rates of cognitive decline among middle-aged and older Chinese.

Elderly people are subject to higher levels of oxidative stress than are young people. Vitamin E, as a powerful antioxidant residing mainly in biomembranes, may provide effective protection against oxidative membrane damage and resultant age-related deterioration, especially in the elderly. We hypothesized that appropriate levels of vitamin E supplementation would protect erythrocyte membranes from oxidative stress and thus improve membrane fluidity in healthy middle-aged and elderly people. To test this, we conducted a 4-month double-blind, randomized trial in which 180 healthy subjects (55-70 years old) were randomly divided into 4 groups: group C (control), and 3 treatment groups in which daily doses of 100 mg (VE1), 200 mg (VE2), and 300 mg (VE3) dl-α-tocopheryl acetate were administered. We measured plasma α-tocopherol concentration, malondialdehyde, and superoxide dismutase levels, erythrocyte hemolysis, and erythrocyte membrane fluidity at the beginning and end of the trial. After 4 months supplementation, plasma α-tocopherol concentrations in the 3 treatment groups had increased by 71%, 78%, and 95%, respectively (all P < .01), and significant decreases in plasma malondialdehyde concentrations were observed in these groups (all P < .05). Erythrocyte hemolysis was decreased by 20% to 38% after vitamin E supplementation (all P < .05), and in addition, groups VE2 and VE3 showed dramatic improvements in erythrocyte membrane fluidity (P < .01). Surprisingly, superoxide dismutase activity also decreased significantly in the treatment groups (all P < .05). In summary, vitamin E supplementation apparently alleviates oxidative stress in healthy middle-aged to elderly people, at least in part by improving erythrocyte membrane fluidity and reducing erythrocyte hemolysis.

The objective of the present study was to investigate age-related differences in erythrocyte membrane fluidity (EMF) and changes in antioxidant capacity following supplementation. A total of seventy-four children were randomly divided into two groups: group A1 was the placebo-controlled group and group A2 was supplemented daily with 600 μg retinol, 1·0 mg β-carotene, 100 mg tocopherol, 300 mg ascorbic acid and 200 μg Se. A total of ninety young people were randomly divided into B1 and B2 groups, and ninety-one elderly subjects were divided into C1 and C2 groups. Groups B1 and C1 were placebo-controlled groups, and groups B2 and C2 were daily supplemented with 900 μg retinol, 1·5 mg β-carotene, 200 mg tocopherol, 500 mg ascorbic acid and 400 μg Se. Results showed that plasma malondialdehyde (MDA) was 5·35 μmol/l in children, which was lower than in young and elderly people. The MDA levels of the young and elderly individuals in the treated groups were significantly lower compared with the control groups, but the supplementation did not alter MDA levels in children. At baseline, there was a lower value of polarisation (ρ) and microviscosity (η) in children, indicating a higher EMF, than in both the young and elderly subjects. After the 2-month trial, the ρ and η values of young and elderly subjects in the treated groups decreased significantly in comparison with the placebo groups, indicating an increase in EMF. In conclusion, there was a background of higher MDA levels and lower EMF in young and elderly people than in children, which could be improved by antioxidant supplementation.

To investigate the effects of iron overload on the liver tissue and function of maternal rats' offspring.Forty pregnant rats were randomly divided into four groups, high-dose group, middle-dose group, low-dose group (120, 60 and 30 mg/kg BW) and control group. Iron detran was administered by intraperitoneal injection every other day and the entire trial lasted for 6 weeks. After 6 weeks, the iron serum levels, GOT and GPT of rats' offspring were determined. Histological changes of the liver injury were measured. The expressions of Bcl-2 and Bax in liver were measured by immunohistochemistry method. In addition, MDA, SOD and GSH-Px in liver were assessed by spectrophotometry.After 6 weeks, the level of serum iron in control group was significantly lower than in middle-dose and high-dose group (P0.05), and the result of level of liver iron was similar with that. According to the HE staining, it showed that liver cell gradually was damaged with the increasing of accumulation of hepatic iron. The expression of Bcl-2/Bax was significantly higher in control group than in middle-dose and high-dose groups (P0.05). The levels of serum GPT and GOT in high-dose group were significantly higher in control group (P0.05). The levels of Liver MDA in middle-dose and high-dose groups were both significantly higher than in control group (P0.05). Moreover, the levels of liver GSH-Px and SOD in high-dose group were significantly lower than in control group (P0.05).Iron overload in maternal rats may result in excessive iron deposition and oxidative damage of liver tissue and cell function in maternal rats' offspring.

Background There is a high burden of both diabetes and tuberculosis in China. Diabetes depresses the immunologic response that facilitates the development of infectious diseases, including infection by Mycobacterium tuberculosis, the agent of tuberculosis. Tuberculosis is the third cause of death among subjects with non-communicable diseases, and among the non-communicable diseases, diabetes is one of the most important. The relationship between diabetes and tuberculosis has already been object of many investigations but the association between these two diseases is not fully understood. The aim of this study was to determine whether relative qualitative and quantitative differences in protein expression of plasma could be related to active pulmonary tuberculosis complicated with diabetes. Methods Biological parameters are useful tools for understanding and monitoring complicated disease processes. Our study employed two-dimensional gel electrophoresis and mass spectrometry to analyze the proteins associated with active pulmonary tuberculosis complicated with diabetes. Results Under the baseline condition, we found that the levels of α-1 antitrypsin precursor, vitamin D-binding protein precursor, CD5 antigen like precursor, clusterin precursor, apolipoprotein A-I precursor, haptoglobin, and fibrinogen γ-chain differed between patients with active pulmonary tuberculosis and active pulmonary tuberculosis complicated with diabetes subjects. Western blotting results confirmed differential expression of clusterin. Conclusions We identified active pulmonary tuberculosis complicated with diabetes-associated proteins in plasma. C-terminal haptoglobin is a possible candidate protein of interest, which might be a link between active pulmonary tuberculosis and diabetes. The dynamics of protein expression during disease progression may improve our understanding of the pathogenesis of active pulmonary tuberculosis complicated with diabetes.

To analyze the correlation of band 3 protein of erythrocytes with physiological conditions of middle-aged and elderly people, and to investigate the expression of band 3 protein under different physiological status.Cluster randomized sampling method for selecting subjects . Physical examination data and blood samples of 218 middle-aged and elderly people (80 males and 138 females) were collected. SDS-polyacrylamide gel electrophoresis was used to analyze band 3 protein in erythrocyte membrane. The difference of band 3 protein in different groups was analyzed by T Test and One-Way ANOVA. Correlation and regression of band 3 protein with physiological status was analyzed by Pearson correlation and linear regression.There was a significantly positive correlation (r =0.149) between the level of band 3 protein and physical activity (P0.05), and a negative correlation (r = -0.156) between the level of band 3 protein and systolic blood pressure (P0.05). The correlation of band 3 protein with gender, age, BMI and diastolic blood pressure (P0.05) is not significant. The band 3 protein level of workers with medium activity was 24. 09% , which was significantly higher than that of light activity workers (23.42%) (P0.05). The subjects with hypertension were proved to have a significantly lower level of band 3 protein (23.33%) than normal individuals (24.20%) (P0.05).The level of band 3 protein was positively correlated with physical activity and negatively correlated with systolic blood pressure.

Background The merging epidemics of pulmonary tuberculosis (PTB) and diabetes mellitus (DM) have been raised concerns by many experts but no large scale screening and intervention have been launched yet, especially in low-income areas. The current study aims to understand the prevalence of DM in active PTB patients and evaluate the outcomes of diet and living habit intervention in poverty zones in China. Methods/design A cross-sectional investigation and intervention study will be carried out. At least 7000 active PTB patients will be recruited, together with 7000 nonTB persons from the same community. The project will be divided into two stages. The first stage is to train TB workers on DM screening and regular treatment. Screening and related investigation will be carried out afterwards. The second stage is focussed on intervention. A comprehensive strategy will be utilized to conduct health promotion among the patients, the health providers and the lay public. Discussion To our knowledge, this is the first and largest study which focuses on the prevalence of DM in PTB in China. We hypothesize that the current prevalence of DM in PTB in China will be understood and the results of our study will provide important evidence for preventing and controlling DM and PTB.

Following the change of dietary structure and living style, metabolic syndrome (MetS) has become increasingly common in China, especially in women, who have abnormal plasma lipid profiles with increased levels of oxidative stress. Vitamin E (VitE) is a powerful chain-breaking antioxidant, which may be a protective factor against oxidative stress-related diseases. This study investigated the effects of three different dosages of tocopherol supplementation (100 IU /day, 200 IU /day, 300 IU /day) for 4 months in Chinese women with MetS. The plasma VitE concentrations increased significantly after the 4 months of supplementation (p < 0.01). The protective decreases in plasma total cholesterol were significant in 200 IU/day and 300 IU/day VitE groups (p < 0.05), but decreases in high-density lipoprotein cholesterol were also significant in all the supplementation groups (p < 0.05). Plasma triglycerides were unaltered (p > 0.05). The indicators of oxidative stress decreased substantially in all of the VitE supplementation groups: malondialdehyde (MDA) was reduced by nearly 50 percent (all groups, p < 0.001), erythrocyte hemolysis was decreased by nearly 40 percent (all groups, p < 0.05); among which the 300IU/day VitE group showed the most significant effect. However, the activity of superoxide dismutase (SOD) decreased after the trial (p < 0.001). VitE provided marked benefits in reducing oxidative stress levels and improving lipid status in women with MetS. Although no dose-effect relationship was observed, 300 IU VitE per day showed the optimal effect. Research is needed to identify potential protective mechanisms or utilization of vitamin E during MetS.

To evaluate the status and dietary intake of magnesium in mid-old people in a rural area of China, a total of 324 mid-old people (111 males and 213 females) aged 55-70 were randomly selected. All the subjects were divided into four groups: a normal group (N), a hypertension group (H), an impaired fasting glucose group (IFG) and a diabetic group (D). Magnesium, sodium, potassium, calcium, iron, zinc, and copper concentrations in red blood cells (RBC) were measured by emission spectroscopy (ICP-OES), and dietary intakes were surveyed by 24-hr recall questionnaires. The data were analyzed between groups and also between genders. The average body weight and BMI were 59.4 kg and 22.1 in subjects with IFG, and 62.4 kg and 23.5 in the diabetics, which were higher than those of the normal group (p0.05 and p0.01). The average magnesium concentration in RBC was significantly lower in the hypertension group than in the normal group (2.0 mmol/L vs. 2.2 mmol/L, p0.05). Groups H, IFG and D took less magnesium than group N (315.9 mg/d, 331.6 mg/d and 323.3 mg/d vs. 371.54 mg/d, p0.05). Women took less magnesium than men (326.2 mg/d vs. 373.7 mg/d, p0.05). There were no significant differences in other mineral concentrations in RBC and dietary intakes. Results of this study show that magnesium concentration in RBC is lower in mid-old people suffering hypertension as compared to healthy subjects. Dietary intakes of magnesium are lower in mid-old people suffering hypertension, IFG and diabetes as compared to healthy subjects. This indicates that an increase in the magnesium supply would be beneficial to mid-old people.

Anemia is a major nutrition related problem in China. In addition to iron deficiency this may be due to deficiencies of other micronutrients.To describe the micronutrient status of anemic and non-anemic pregnant women in China.734 clinically normal pregnant women in the third trimester aged 20-35, were randomly recruited from the population of pregnant women regularly receiving pregnant examination in community medical centers. Serum concentrations of vitamins A, B12 and C, iron and zinc status parameters, and vitamin B2 in urine were determined. Subjects were categorized according to the presence or absence of anemia and compared according to micronutrient status.Serum concentrations of iron and micronutrients were significantly lower in anemic women than non-anemic women: serum iron 909 microg/L versus 1109 microg/L, ferritin 13.8 microg/L versus 19.6 microg/L, vitamin C 308.9 microg/L versus 388.1 microg/dL, and retinol 50.0 microg/dL versus 59.3 microg/dL. Zinc concentrations were also lower in anemic women. Subnormal serum iron (700 microg/L) and iron depletion (ferritin12 microg/L) were 39.7% and 52.6%, significantly more frequent in anemic than 23.9% and 35.0% in non-anemic subjects, as were subnormal vitamin A and ascorbic acid. Subnormal vitamin B2 and B12 were frequent in both anemic and non-anemic groups.Subnormal concentrations of iron and micronutrients in combination may contribute to this situation. Further studies on food-based or supplement-based approaches trying to increase intake of iron and certain vitamins are warranted to decrease anemia in pregnant Chinese women in the third trimester.

To estabish a high performance liquid chromatographic (HPLC) method for the determination of dehydroaetic acid in food determinated.Separation was carried out on a column of C18 (51 microm, 250 x 4.60mm) at the temperature of 25 degrees C with methanol/0.02mol/L ammonium acetate (5:95, V/V) as mobile phase, and elution flow rate was 1.0ml/min. The detection wavelength was 306nm and the injection volume was 10 microl, quantitation was used by external standard method.The results showed a good linear relationship between the peak area and the concentration (r = 0.9997), the recoveries for Dehydroaetic acid products were 96.5% - 99.5%, and the relative standard deviation was 1.09% - 5.85% for different kinds of food.The results showed that the method may be simple, rapid, accurate and applicable for the determination of dehydroaetic acid in food.

ObjectiveTo investigate whether there is an association between diameter of bacille Calmette-Guérin (BCG) scars and effect of purified protein derivative (PPD) reaction and to determine whether vitamin A (VA) combined vitamin D (VD) supplementation influences the immune response to BCG revaccinated in Chinese infants.MethodsA cross-section and 3-month community-randomised trial was conducted.
