3. First principles modelling of the ion binding capacity of finger millet.

- Abstract
Finger millet, a cereal grain widely consumed in India and Africa, has gained more attention in recent years due to its high dietary fibre (arabinoxylan) and trace mineral content, and its climate resilience. The aim of this study was to understand the interactions between potassium (K+), calcium (Ca2+) and zinc (Zn2+) ions and the arabinoxylan structure and determine its ion-binding capacity. Three variations of a proposed model of the arabinoxylan structure were constructed and first principles Density Functional Theory calculations were carried out to determine the cation-binding capacity of the arabinoxylan complexes. Zn2+-arabinoxylan complexes were highly unstable and thermodynamically unfavourable in all three models. Ca2+ and K+ ions, however, form thermodynamically stable complexes, particularly involving two glucuronic acid residues as a binding pocket. Glucuronic acid residues are found to play a key role in stabilising the cation-arabinoxylan complex, and steric effects are more important to the stability than charge density. Our results highlight the most important structural features of the millet fibre regarding ion-storage capacity, and provide valuable preliminary data for confirmatory experimental studies and for the planning of clinical trials where the bioavailability of bound ions following digestion may be tested.

5. Sumanene monolayer of pure carbon: a two-dimensional Kagome-analogy lattice with desirable band gap, ultrahigh carrier mobility and strong exciton binding energy

Design and synthesis of novel two-dimensional (2D) materials that possess robust structural stability and unusual physical properties may open up enormous opportunities for device and engineering applications. Herein we propose a 2D sumanene lattice that be regarded as a derivative of the conventional Kagome lattice. Our tight-binding analysis demonstrates sumanene lattice contains two sets of Dirac cones and two sets of flat bands near the Fermi surface, distinctively different from the Kagome lattice. Using first-principles calculations, we theoretically suggest two possible routines for realization of stable 2D sumanene monolayers (named as a phase and b phase), and a-sumanene monolayer can be experimentally synthesized with chemical vapor deposition using C21H12 as a precursor. Small binding energies on Au(111) surface signify the possibility of their peel-off after grown on the noble metal substrate. Importantly, our GW plus Bethe-Salpeter equation calculations demonstrate both monolayers have moderate band gaps (1.94 eV for a) and ultrahigh carrier mobilities (3.4*104 cm2/Vs for a). In particular, a-sumanene monolayer possesses a strong exciton binding energy of 0.73 eV, suggesting potential applications in optics.

6. Robust Sandwiched B/TM/B Structures by Metal Intercalating into Bilayer Borophene Leading to Excellent Hydrogen Evolution Reaction

Bilayer borophene, very recently synthesized on Ag and Cu, possesses extremely flat large surface and excellent conductivity. Besides, the van der Waals gap of bilayer borophene can be intercalated by metal atoms, thereby tailoring the properties of bilayer borophene. Herein, we propose that sandwiched B/TM/B (TM=Co, Ni, Cu, Pd) could be a new 2D formation by transiton metal atoms intercalated into bilayer borophene network, it is quiet robust with both energetic, structural and thermal stability, and exhibits heat resistance of at least 1300 K. Besides, it is novel platform for electrocatalytic hydrogen evolution reaction (HER). The interecalation metal atom serves as single-atomic catalyst, which acting the nonmetal boron layers. Beyond that, the transtion metal is protected by outside boron layers from being corroded by acidic/alkaline solution. B/Cux/B, B/Pdx/B and B/Alx/B with different metal coverage exhibit defect-independent extremely low HER free energy in the range of -0.162 ~ 0.179 eV, -0.134 ~ 0.183 eV and -0.082 ~ 0.086 eV which are comparable to noble metal Pt. Combining excellent conduction, high structural and thermal stability, low resistance to intercalated behaviour, effortless water splitting process, excellent defect-independent catalytic performance, cheapness and abundance of raw materials, free of corrodation, 2D sandwiched B/TM/B (TM=Co, Ni, Cu, Pd) is believed to promising for electrocatalytic HER applications.

Abstract When the solar wind interacts with the ionosphere of an unmagnetized planet, it induces currents that form an induced magnetosphere. These currents and their associated magnetic fields play a pivotal role in controlling the movement of charged particles, which is essential for understanding the escape of planetary ions. Unlike the well-documented magnetospheric current systems, the ionospheric current systems driven by solar wind and atmospheric neutral winds have not been quantitatively observed, which constrains the quantification of energy transfer from stars to these planets. Here, utilizing eight years of data from the Mars Atmosphere and Volatile EvolutioN (MAVEN) mission, we investigate the global distribution of ionospheric currents on Mars. We identify two distinct current systems in the ionosphere: one aligns with the solar wind electric field but exhibits hemispheric asymmetry perpendicular to the solar wind electric field direction; the other corresponds to the flow pattern of annually averaged neutral winds. We propose that these two current systems are driven by the solar wind and atmospheric neutral winds, respectively. Our findings reveal that Martian ionospheric dynamics are influenced by the neutral winds from below and the solar wind from above, highlighting the complex and intriguing nature of current systems on unmagnetized planets.

The South Atlantic Anomaly (SAA) is one of the most remarkable structural features of the Earth's magnetic field. The region is characterized by weak magnetic field intensity. Because of this weakening, particles within the inner radiation belt can mirror at lower altitudes, consequently increasing the local particle flux. Over centuries, the SAA has displayed notable secular variation (SV) characteristics, with a consistent decline in strength, expansion in area, and an overall westward drift. Both paleomagnetic and modern magnetic data indicate that SAA is currently undergoing rapid changes, with a rapid expansion in area and a rapid decreasing in intensity. These changes are coinciding with an increasing field asymmetry. Recent observations suggest that the evolution of SAA has become increasingly anisotropic, showing two magnetic intensity minima. This may indicate that the evolution of SAA has entered a new stage, which has aroused people's attention and discussion on the long-term evolution and future of SAA. The manifestation of the SAA on the Earth’s surface corresponds to the response of an inverse flux path at the core-mantle boundary (CMB) to the radial component of the geomagnetic field, positioned approximately beneath the South Atlantic Ocean. Consequently, studying the SV of SAA can understand the details of the Earth's internal dynamics. Moreover, these SV scenarios provide some speculative predictions for the SAA. The evolutionary details of SAA not only serve as a reference for future trends but also imposes additional constraints on Earth's core dynamic model, rendering it a compelling subject for scholarly inquiry. In this paper, we review the results of paleomagnetic and modern geomagnetic models concerning the past, present, and future evolution of the SAA, while also summarizing advancements in understanding the origin of this anomaly.

A majority of the planets in the Solar System are characterized by intrinsic magnetic fields that co-rotate with these celestial bodies. With ongoing advances in human deep-space exploration, magnetic field models have been established for all known Solar System bodies with intrinsic magnetic fields. Planetary magnetic field modeling serves as a fundamental basis for a diverse range of research objectives, including investigations of planetary internal dynamical processes, examinations of planetary evolutionary histories, determinations of the interactions between planetary magnetic fields and the solar wind, and estimations of the shielding effects of planetary magnetic fields against high-energy charged particles. Furthermore, the modeling of planetary magnetic fields represents an interdisciplinary research endeavor, encompassing aspects such as the theories of magnetic field modeling, separation of internal and external magnetic field sources, parameter inversion methods, and equation optimization. In this paper, we provide a comprehensive review of the progress in magnetic field modeling for the Earth, other planets in the Solar System, and also the Moon and Ganymede, one of Jupiter’s moon. We describe the characteristics of the intrinsic magnetic fields of planets and present details of the most accurate global magnetic field models for Solar System planets. Starting with mathematical modeling methods for planetary magnetic fields, we outline the development of magnetic field modeling techniques since the era of Gauss and identify the current hot topics in this field. The review concludes with a forward-looking perspective on the trends in magnetic field modeling research and the potential contributions of China’s future deep-space exploration to the study of planetary magnetic fields.

11. Fifty-year investigation of the correlation between the geomagnetic field and climate

After 50 years of research on the correlation between the geomagnetic field and climate, we have gained a deeper understanding that the regulation of climate by geomagnetic field changes is far more complex than previously imagined. In this paper, we comprehensively review the research connecting the geomagnetic field and climate over the past 50 years, examining different spatial and temporal scales. Our results demonstrate a significant correlation between geomagnetic field changes and climate at various scales, including millennial-scale geomagnetic reversals, thousand-year-scale geomagnetic shifts, centennial-scale geomagnetic abrupt, and even decadal-scale variations in geomagnetic activity. The Matuyama–Brunhes geomagnetic reversal is associated with the cold climate. The increase in galactic cosmic rays (GCR) during this period gave rise to the formation of low-altitude cloud "umbrella effects", subsequently weakening the East Asian summer monsoon and intensifying the East Asian winter monsoon. Archaeomagnetic data from Mesopotamia remarkably illustrates a consistent correlation between four cold periods in the last three thousand years and a sudden increase in geomagnetic anomalies. Exploring the possibility that these geomagnetic anomalies correspond to extreme tilts of the Earth's dipole, it is hypothesized that auroral ovals and sub-auroral regions may have expanded to lower latitudes. In these regions, cosmic rays potentially interacted with a more humid troposphere, resulting in increased cloud cover and, consequently, observed atmospheric cooling. Changes in GCR flux, induced by solar activity and variations in the geomagnetic field, are proposed to have significant implications for alterations in temperate pressure systems, precipitation patterns, and atmospheric electric fields. These effects play diverse roles within the intricately coupled system of the Earth's atmosphere and the near-Earth space environment. However, it's important to note that correlation does not imply causation, and the potential mechanisms by which the geomagnetic field influences climate change are still a subject of debate. The cosmic ray-cloud mechanism is the most promising avenue for understanding how the geomagnetic field regulates climate, operating effectively across different time scales, but its exact physical mechanisms and relative importance remain unresolved issues. We propose that understanding the correlation between the geomagnetic field and climate from the perspective of solar-terrestrial multisphere coupling is crucial, with a specific focus on the response of regional climate systems to changes in regional magnetic fields. By integrating knowledge from ancient times to the present, from regional to global perspectives, we aim to form a comprehensive understanding of the correlation between the geomagnetic field and climate change. The recent successful launch of Aoke-1, which can delicately characterize magnetic field changes in the South Atlantic Anomaly (SAA), the first discovery of the centennial-scale western Pacific anomaly (WPA), and the rapid development of paleomagnetic/archaeomagnetic and paleoclimate data reconstruction have provided new historical opportunities for research on the geomagnetic field and climate. This progress indicates that the study of the impact of geomagnetic field changes on climate will be propelled to new heights.

The internal structure and dynamics of different planets are different, so some planets have an intrinsic magnetic field, and some have not yet found an intrinsic magnetic field. A planet's intrinsic magnetic field or atmosphere interacts with the solar wind forming a magnetosphere or induced magnetosphere, respectively. Different magnetic fields, atmospheres and moons in planetary systems will form different space environments around the planets, which will have different effects on the evolution of the planets. The detection of planetary magnetic field is of great significance for understanding the internal structure and dynamics, space environment and evolution process of planets, so the planetary magnetic field is often one of the main physical quantities concerned by planetary exploration. In the detection of Jupiter's magnetic field, it was found that Jupiter has the largest magnetic moment and magnetosphere among the planetary systems in the solar system, and the magnetic field of Jupiter shows obvious hemispherical difference, magnetic anomaly areas (the Great Blue Spot) and other non-dipole features. Besides, the internal structure of Jupiter is significantly different from the Earth, which makes the structure and dynamics of Jupiter's dynamo difficult to confirm. This paper reviews the research history of Jupiter's magnetic field, introduces the current cognition of Jupiter's magnetic field mainly based on the research in the Juno era, and prospects the future research and exploration of Jupiter's magnetic field. It is suggested that the probe into Jupiter's atmosphere should be carried in the future Jupiter exploration, and the magnetic field detection instrument should be loaded on the probe. This enabled the first in-situ detection of Jupiter's magnetic field in the atmosphere.

13. Disulfidptosis-related subtype and prognostic signature in prostate cancer

Abstract Background Disulfidptosis refers to cell death caused by the accumulation and bonding of disulfide in the cytoskeleton protein of SLC7A11-high level cells under glucose deprivation. However, the role of disulfidptosis-related genes (DRGs) in prostate cancer (PCa) classification and regulation of the tumor microenvironment remains unclear. Methods Firstly, we analyzed the expression and mutation landscape of DRGs in PCa. We observed the expression levels of SLC7A11 in PCa cells through in vitro experiments and assessed the inhibitory effect of the glucose transporter inhibitor BAY-876 on SLC7A11-high cells using CCK-8 assay. Subsequently, we performed unsupervised clustering of the PCa population and analyzed the differentially expressed genes (DEGs) between clusters. Using machine learning techniques to select a minimal gene set and developed disulfidoptosis-related risk signatures for PCa. We analyzed the tumor immune microenvironment and the sensitivity to immunotherapy in different risk groups. Finally, we validated the accuracy of the prognostic signatures genes using single-cell sequencing, qPCR, and western blot. Results Although SLC7A11 can increase the migration ability of tumor cells, BAY-876 effectively suppressed the viability of prostate cancer cells, particularly those with high SLC7A11 expression. Based on the DRGs, PCa patients were categorized into two clusters (A and B). The risk label, consisting of a minimal gene set derived from DEGs, included four genes. The expression levels of these genes in PCa were initially validated through in vitro experiments, and the accuracy of the risk label was confirmed in an external dataset. Cluster-B exhibited higher expression levels of DRG, representing lower risk, better prognosis, higher immune cell infiltration, and greater sensitivity to immune checkpoint blockade, whereas Cluster A showed the opposite results. These findings suggest that DRGs may serve as targets for PCa classification and treatment. Additionally, we constructed a nomogram that incorporates DRGs and clinical pathological features, providing clinicians with a quantitative method to assess the prognosis of PCa patients. Conclusion This study analyzed the potential connection between disulfidptosis and PCa, and established a prognostic model related to disulfidptosis, which holds promise as a valuable tool for the management and treatment of PCa patients.

14. Quercetin promotes the proliferation, migration, and invasion of trophoblast cells by regulating the miR‐149‐3p/AKT1 axis

Abstract Recurrent spontaneous abortion (RSA) has a complex pathogenesis with an increasing prevalence and is one of the most intractable clinical challenges in the field of reproductive medicine. Quercetin (QCT) is an effective active ingredient extracted from Semen Cuscutae and Herba Taxilli used in traditional Chinese medicine for tonifyng the kidneys and promoting fetal restoration. Although QCT helps improve adverse pregnancy outcomes, the specific mechanism remains unclear. The trophoblast cell line HTR‐8/SVneo cultured in vitro was treated with different concentrations of QCT, and the cell counting kit‐8 assay, wound healing assay, transwell assay, and western blotting were used to evaluate the effects and mechanisms of QCT on the proliferation, migration, and invasion of HTR‐8/SVneo cells, respectively. To assess the expression levels of miR‐149‐3p and AKT serine/threonine kinase 1 (AKT1), quantitative real‐time polymerase chain reaction (qRT‐PCR) and western blotting analysis were performed. A dual‐luciferase reporter assay was used to investigate the potential regulatory relationship between miR‐149‐3p and AKT1. Our results showed that QCT promoted the proliferation, migration, and invasion of trophoblast cells, promoted the expression of MMP2, MMP9, and vimentin, and downregulated the expression of E‐cadherin. Mechanistically, QCT downregulated the expression of miR‐149‐3p and upregulated the expression of AKT1, and miR‐149‐3p directly targets AKT1, negatively regulating its expression. Overexpression of miR‐149‐3p and silencing of AKT1 counteracted the promotional effects of QCT on trophoblast proliferation, migration, and invasion. Taken together, QCT regulates the migration and invasion abilities of HTR‐8/SVneo cells through the miR‐149‐3p/AKT1 axis, which may provide a promising therapeutic approach for RSA.

15. Study on the Effect of Solid-state Fermentation with Compound Probiotics on Removing Deoxynivalenol from Wheat and Corn

The study aimed to identify an effective probiotic combination for the removal of deoxynivalenol (DON) from wheat and corn, and to verify the detoxification effect of solid-state fermentation. The content of DON removed, adsorbed, and degraded by the selected strains was measured using high-performance liquid chromatography (HPLC). The results showed that in the liquid matrix, the detoxification rate increased with the reaction time and was higher for Streptococcus pentosus, Saccharomyces cerevisiae, and Lactobacillus plantarum among all the bacteria. After 24 hours, the maximum detoxification rates of the individual strains reached 79.62%, 62.22%, and 82.75%, respectively. Measurement of DON content in different treatment solutions revealed that the removal of DON by the three strains was primarily due to adsorption, with a DON desorption rate of less than 15%. In the solid-state fermentation system, the highest detoxification rate of DON-contaminated wheat could reach 79.49%, when the inoculation amount of three strains were 1×106, 1×107 and 1×108 CFU/mL. The highest detoxification rates for DON- and zearalenone-contaminated corn achieved 61.54% and 78.15%, respectively, when the inoculation amount of three strains were 1×106, 1×108 and 1×107 CFU/mL. In addition, the combined solid-state fermentation of the three strains increased the crude protein and reduced the crude fibre content of toxin-contaminated wheat and corn. This study identified compound probiotics that can efficiently remove DON in liquid substrates and solid-state fermentation systems, and preliminarily explored their detoxification mechanisms, which could provide a theoretical and practical basis for the detoxification of grains contaminated with mycotoxins through solid-state fermentation.

16. Influence of upstream solar wind on magnetic field distribution in the Martian nightside ionosphere

Using over eight years of Mars Atmosphere and Volatile EvolutioN (MAVEN) data, from November 2014 to May 2023, we have investigated the Martian nightside ionospheric magnetic field distribution under the influence of upstream solar wind drivers, including the interplanetary magnetic field intensity (\begin{document}$ \left|{{\boldsymbol{B}}}_{\mathrm{I}\mathrm{M}\mathrm{F}}\right| $\end{document}), solar wind dynamic pressure (\begin{document}$ {P}_{\mathrm{S}\mathrm{W}} $\end{document}), solar extreme ultraviolet flux (EUV), and Martian seasons (\begin{document}$ {L}_{\mathrm{s}} $\end{document}). Our analysis reveals pronounced correlations between magnetic field residuals and both \begin{document}$ \left|{{\boldsymbol{B}}}_{\mathrm{I}\mathrm{M}\mathrm{F}}\right| $\end{document} and \begin{document}$ {P}_{\mathrm{S}\mathrm{W}} $\end{document}. Correlations observed with EUV flux and \begin{document}$ {L}_{\rm{s}} $\end{document} were weaker — notably, magnetic field residuals increased during periods of high EUV flux and at Mars perihelion. We find that the IMF penetrates to an altitude of 200 km under a wide range of upstream conditions, penetrating notably deeper under high \begin{document}$ \left|{{\boldsymbol{B}}}_{{\mathrm{IMF}}}\right| $\end{document} and \begin{document}$ {P}_{{\mathrm{SW}}} $\end{document} conditions. Our analysis also indicates that EUV flux and IMF cone angle have minimal impact on IMF penetration depth. Those findings provide useful constraints on the dynamic nature of Martian atmospheric escape processes and their evolution, suggesting that historical solar wind conditions may have facilitated deeper IMF penetration and higher rates of ionospheric escape than are observed now. Moreover, by establishing criteria for magnetic ‘quiet’ conditions, this study offers new insights into the planet’s magnetic environment under varying solar wind influences, knowledge that should help refine models of the Martian crustal magnetic field.

Jupiter is one of the top priorities for deep space exploration in China and other countries. The structure of Jupiter’s interior, in particular, is a crucial but still unclear scientific topic. This paper discusses current scientific understanding of Jupiter’s interior by summarizing the history of past and current exploration and data analysis. We review recent space-based and ground-based observation methods and analyze their feasibility. To gain new insight into the internal structure of Jupiter, we propose to study Jupiter’s innards by planetary seismology. Ground-based observation, namely the Jupiter Seismologic Interferometer Polarization Imager (SIPI) in Lenghu, will be developed to obtain the Doppler velocity distribution on the surface of Jupiter and identify oscillation signals. Lenghu has observation conditions that are not only exceptional in China but even in the world, capable of providing novel insight into the interior of Jupiter. This will also be the first study in China of the interior of Jupiter using asteroseismology, which has significant implications for China’s plans to explore Jupiter via spacecraft-mounted instruments.

Magnetic reconnection and dipolarization are crucial processes driving magnetospheric dynamics, including particle energization, mass circulation, and auroral processes, among others. Recent studies have revealed that these processes at Saturn and Jupiter are fundamentally different from the ones at Earth. The reconnection and dipolarization processes are far more important than previously expected in the dayside magnetodisc of Saturn and potentially Jupiter. Dayside magnetodisc reconnection was directly identified by using Cassini measurements (Guo RL et al., 2018b) and was found to be drizzle-like and rotating in the magnetosphere of Saturn (Delamere et al., 2015b; Yao ZH et al., 2017a; Guo RL et al., 2019). Moreover, magnetic dipolarization could also exist at Saturn’s dayside (Yao ZH et al., 2018), which is fundamentally different from the terrestrial situation. These new results significantly improve our understanding of giant planetary magnetospheric dynamics and provide key insights revealing the physics of planetary aurorae. Here, we briefly review these recent advances and their potential implications for future investigations.

Background and objective: The purpose-built SHURUI single-port (SP) robotic platform has recently been introduced for several procedures in urology, general surgery, and gynecology. However, comparative evidence on its performance in relation to earlier models such as the da Vinci SP is lacking. Our aim was to compare the step-by-step techniques and 1-yr outcomes for radical prostatectomy (RP) between the SHURUI SP and da Vinci SP robots. Methods: Data were retrieved from two prospectively maintained databases. The SHURUI SP robot was used to perform RP in 34 patients in China (September 2021 to August 2022); the da Vinci SP robot was used to perform 100 consecutive RP cases in the USA (June 2019 to October 2020). A comparative analysis was conducted before and after 1:1 propensity score matching for age, body mass index, American Urological Association symptom score, prostate size, prostate-specific antigen (PSA) levels, biopsy grade group, and D’Amico risk group. Intraoperative performance and short-term oncological and continence outcomes were compared between the groups. Biochemical recurrence was defined as two consecutive postoperative PSA levels >0.2 ng/ml. Continence was defined as full recovery of urinary control without the use of pads. The Kaplan-Meier method was used to estimate continence recovery curves, and a log-rank test for trend was used to detect ordered differences in continence recovery between the SHURUI SP and da Vinci SP groups after surgery. Key findings and limitations: For the matched SHURUI and da Vinci groups, median age (69 vs 69 yr), median PSA (8.4 vs 7.1 ng/ml), and the proportion of patients with low-risk (33.3% vs 29.6%), intermediate-risk (66.7% vs 63%), and high-risk disease (0% vs 7.4%) were comparable (all p > 0.05). The low-degree gravity field and rotation parameters of terrestrial planets are functions of the planet's interior structure, providing insights into their composition and evolution, which serves them as crucial targets throughout deep-space exploration. Particularly, the measurement of rotation state can directly reveal the density and size of most untouchable planetary core. With the accumulation of data and the recent high-precision deep-space missions, substantial scientific advances have been achieved in understanding the internal structure of terrestrial planets, especially the core, using the low-degree gravity field and rotation measurements. In this study, we provide a summary of the fundamental theory and method for determining the principal moment of inertia tensor of planets, and then the basic theory of planetary rotation dynamics and rotational normal modes closely related to internal structures, and the methods for solving principal moments of inertia tensors of planets using the low-degree gravitational potential coefficients. We review the up-to-date researches on determining the mean bulk density and mean moment of inertia factor through limited rotation observations and the low-degree gravity field measurements, and the basic approaches to invert interior structures. Finally, we present existing challenges in revealing interior structures from low-degree gravity field and rotation normal modes for terrestrial planets and asteroids with a hope to provide scientific references and theoretical support for Chinese future deep-space missions.

Jupiter is the biggest and fastest-rotating planet in our solar system. As a gas giant without land–sea distribution or topography, Jupiter is an ideal natural laboratory for studying atmospheric dynamics. Jupiter's atmospheric composition, atmospheric circulation, and internal structure are all topics of scientific significance. On December 4, 1973, Pioneer 10 achieved its closest approach to Jupiter, marking the first successful exploration mission to the Jovian system. Since then, Jupiter exploration has gathered more than 50 years of experience with 10 missions, including 7 flyby missions, 2 orbiting missions, Galileo and Juno, and the still en route mission Jupiter Icy moons Explorer (JUICE). This review takes a brief look-back to these 10 Jupiter missions and their science results, especially the progress on Jupiter's atmospheric composition, waves and zonal jet streams, the Great Red Spot, and polar vortices, as well as the unsolved scientific problems. China plans to launch Tianwen-4 by 2030, targeting the Jovian system. To shed some light on the scientific payloads and target design of Tianwen-4, reviewing the past Jupiter missions has certain significance.

Abstract Despite the well-established significance of transcription factors (TFs) in pathogenesis, their utilization as pharmacological targets has been limited by the inherent challenges in modulating their protein interactions. The lack of defined small-molecule binding pockets and the nuclear localization of TFs do not favor the use of traditional tools. Aptamers possess large molecular weights, expansive blocking surfaces and efficient cellular internalization, making them compelling tools for modulating TF interactions. Here, we report a structure-guided design strategy called Blocker-SELEX to develop inhibitory aptamers (iAptamers) that selectively block TF interactions. Our approach leads to the discovery of iAptamers that cooperatively disrupt SCAF4/SCAF8-RNAP2 interactions, dysregulating RNAP2-dependent gene expression, which impairs cell proliferation. This approach is further applied to develop iAptamers blocking WDR5-MYC interactions. Overall, our study highlights the potential of iAptamers in disrupting pathogenic TF interactions, implicating their potential utility in studying the biological functions of TF interactions and in nucleic acids drug discovery.

Abstract Background This study investigated the molecular mechanism of long intergenic non-protein coding RNA 1605 (LINC01605) in the process of tumor growth and liver metastasis of pancreatic ductal adenocarcinoma (PDAC). Methods LINC01605 was filtered out with specificity through TCGA datasets (related to DFS) and our RNA-sequencing data of PDAC tissue samples from Renji Hospital. The expression level and clinical relevance of LINC01605 were then verified in clinical cohorts and samples by immunohistochemical staining assay and survival analysis. Loss- and gain-of-function experiments were performed to estimate the regulatory effects of LINC01605 in vitro. RNA-seq of LINC01605-knockdown PDAC cells and subsequent inhibitor-based cellular function, western blotting, immunofluorescence and rescue experiments were conducted to explore the mechanisms by which LINC01605 regulates the behaviors of PDAC tumor cells. Subcutaneous xenograft models and intrasplenic liver metastasis models were employed to study its role in PDAC tumor growth and liver metastasis in vivo. Results LINC01605 expression is upregulated in both PDAC primary tumor and liver metastasis tissues and correlates with poor clinical prognosis. Loss and gain of function experiments in cells demonstrated that LINC01605 promotes the proliferation and migration of PDAC cells in vitro. In subsequent verification experiments, we found that LINC01605 contributes to PDAC progression through cholesterol metabolism regulation in a LIN28B-interacting manner by activating the mTOR signaling pathway. Furthermore, the animal models showed that LINC01605 facilitates the proliferation and metastatic invasion of PDAC cells in vivo. Conclusions Our results indicate that the upregulated lncRNA LINC01605 promotes PDAC tumor cell proliferation and migration by regulating cholesterol metabolism via activation of the mTOR signaling pathway in a LIN28B-interacting manner. These findings provide new insight into the role of LINC01605 in PDAC tumor growth and liver metastasis as well as its value for clinical approaches as a metabolic therapeutic target in PDAC.

Abstract Traditional catalytic techniques often encounter obstacles in the search for sustainable solutions for converting CO2 into value-added products because of their high energy consumption and expensive catalysts. Here, we introduce a contact-electro-catalysis approach for CO2 reduction reaction, achieving a CO Faradaic efficiency of 96.24%. The contact-electro-catalysis is driven by a triboelectric nanogenerator consisting of electrospun polyvinylidene fluoride loaded with single Cu atoms-anchored polymeric carbon nitride (Cu-PCN) catalysts and quaternized cellulose nanofibers (CNF). Mechanistic investigation reveals that the single Cu atoms on Cu-PCN can effectively enrich electrons during contact electrification, facilitating electron transfer upon their contact with CO2 adsorbed on quaternized CNF. Furthermore, the strong adsorption of CO2 on quaternized CNF allows efficient CO2 capture at low concentrations, thus enabling the CO2 reduction reaction in the ambient air. Compared to the state-of-the-art air-based CO2 reduction technologies, contact-electro-catalysis achieves a superior CO yield of 33 μmol g−1 h−1. This technique provides a solution for reducing airborne CO2 emissions while advancing chemical sustainability strategy.

Abstract The intelligent appearance quality classification method for Auricularia auricula is of great significance to promote this industry. This paper proposes an appearance quality classification method for Auricularia auricula based on the improved Faster Region-based Convolutional Neural Networks (improved Faster RCNN) framework. The original Faster RCNN is improved by establishing a multiscale feature fusion detection model to improve the accuracy and real-time performance of the model. The multiscale feature fusion detection model makes full use of shallow feature information to complete target detection. It fuses shallow features with rich detailed information with deep features rich in strong semantic information. Since the fusion algorithm directly uses the existing information of the feature extraction network, there is no additional calculation. The fused features contain more original detailed feature information. Therefore, the improved Faster RCNN can improve the final detection rate without sacrificing speed. By comparing with the original Faster RCNN model, the mean average precision (mAP) of the improved Faster RCNN is increased by 2.13%. The average precision (AP) of the first-level Auricularia auricula is almost unchanged at a high level. The AP of the second-level Auricularia auricula is increased by nearly 5%. And the third-level Auricularia auricula AP is increased by 1%. The improved Faster RCNN improves the frames per second from 6.81 of the original Faster RCNN to 13.5. Meanwhile, the influence of complex environment and image resolution on the Auricularia auricula detection is explored.

The combination of thermodynamic calculation and machine learning was used to reverse design nickel-based superalloys for thermodynamic performance requirements. The results show that the thermodynamic calculation dataset of nickel-based superalloys is successfully constructed by high-throughput thermodynamic calculations, which provides the data basis for the reverse design of nickel-based superalloys with thermodynamic performance requirements by using machine learning methods. Several C2P models are established for the thermodynamic target performance, and the accuracy of the models is higher than 99%. MLDS method is used to reverse design the alloy composition, and the eight alloys are recommended to meet the performance requirements (Vγ',1100 ℃≥60%,Vγ,1100 ℃+Vγ',1100 ℃≥99% and Tγ′≥1300 ℃). The experimental verification of the three alloys with the smallest prediction error of thermomechanical properties shows that the Vγ',1100 ℃ are greater than 80%, Vγ,1100 ℃+Vγ',1100 ℃≥99% in the microstructure after aging, and Tγ′≥1300 ℃, which meet the design requirements.

Abstract Higher serum cholesterol levels have been associated with an increased risk of dry eye disease (DED). The relationship between statin (HMG‐CoA reductase inhibitor) use and DED in patients with hyperlipidemia remains unclear. To investigate the association between statin use and the risk of DED in patients with hyperlipidemia, we conducted a population‐based retrospective cohort study utilizing data from Taiwan's Longitudinal Generation Tracking Database. Patients were categorized into statin users and nonusers, with a 5‐year follow‐up period. The study identified patients with newly diagnosed hyperlipidemia, excluding those with prior DED diagnoses. Matching and adjustments for covariates resulted in 41,931 individuals in each group. Patients receiving statin therapy were compared with those unexposed. Cumulative exposure doses were also evaluated to assess dose–response relationships. The primary outcome was the incidence of DED diagnosed during the follow‐up period. Cox proportional hazards regression models estimated the risk of DED, and conditional logistic regression analyzed the dose–response effect of statin exposure. Objective Coronary artery disease (CAD) is the leading cause of morbidity and mortality in the world. The proprotein convertase subtilisin/kexin type 9 ( PCSK9 ) E670G polymorphism has been reported to be associated with variability in levels of low density lipoprotein cholesterol, a risk factor for CAD. However, the relationship between PCSK9 E670G and CAD is still not fully elucidated. Methods A total of 225 patients and 189 control subjects were recruited in this study. DNA was extracted from peripheral blood samples and was genotyped by mass array method. In addition, we also conducted a meta-analysis of case-control studies to elucidate the relationship of CAD and polymorphism. Results The GG genotype of PCSK9 E670G was associated with a higher risk of CAD [odds ratio (OR) 2.994, 95% confidence interval (CI): 1.174–7.631], even adjusting for risk factors (OR 2.794, 95% CI: 1.215–7.460). Logistic regression analysis showed that the dominant genetic model increased the CAD risk (OR 2.313, 95% CI: 1.070–6.983) after adjusting the confounding factors. Meta-analysis results of 13 studies revealed that PCSK9 E670G polymorphism was correlated with CAD risk under different genetic models. Conclusion Our results demonstrated that PCSK9 E670G genotype was associated with a high risk of CAD.

Strong evidence indicates that environmental stressors are the risk factors for male testosterone deficiency (TD). However, the mechanisms of environmental stress-induced TD remain unclear. Based on our all-cause male reproductive cohort, we found that serum ferrous iron (Fe2⁺) levels were elevated in TD donors. Then, we explored the role and mechanism of ferroptosis in environmental stress-reduced testosterone levels through in vivo and in vitro models. Data demonstrated that ferroptosis and lipid droplet deposition were observed in environmental stress-exposed testicular Leydig cells. Pretreatment with ferrostatin-1 (Fer-1), a specific ferroptosis inhibitor, markedly mitigated environmental stress-reduced testosterone levels. Through screening of core genes involved in lipid droplets formation, it was found that environmental stress significantly increased the levels of perilipins 4 (PLIN4) protein and mRNA in testicular Leydig cells. Further experiments showed that Plin4 siRNA reversed environmental stress-induced lipid droplet deposition and ferroptosis in Leydig cells. Additionally, environmental stress increased the levels of METTL3, METTL14, and total RNA m6A in testicular Leydig cells. Mechanistically, S-adenosylhomocysteine, an inhibitor of METTL3 and METTL14 heterodimer activity, restored the abnormal levels of Plin4, Fe2⁺ and testosterone in environmental stress-treated Leydig cells. Collectively, these results suggest that Plin4 exacerbates environmental stress-decreased testosterone level via inducing ferroptosis in testicular Leydig cells.

Diminished testosterone levels have been documented as a key factor in numerous male health disorders. Both human and animal studies have consistently demonstrated that cadmium (Cd), a pervasive environmental heavy metal, results in decreased testosterone levels. However, the exact mechanism through which Cd interferes with testosterone synthesis remains incompletely elucidated. This research sought to examine the impact of cellular senescence on Cd-suppressed testosterone synthesis. We also investigated the related m6A modification mechanism. The results demonstrated that Cd (100 mg/L) led to a decrease in testosterone levels, along with downregulated expression of testosterone synthase in C57BL/6 N male mice. Furthermore, Cd significantly increased β-galactosidase staining intensity, senescence-related proteins, and senescence-related secretory phenotypes in mouse testicular Leydig cells. Subsequent investigations revealed that Cd decreased the mRNA and protein levels of NAD-dependent deacetylase Sirtuin-1 (SIRT1) in Leydig cells. Mechanistically, mice treated with resveratrol (50 mg/kg), a specific SIRT1 activator, mitigated Leydig cell senescence and reversed Cd-reduced testosterone levels in mouse testes. These effects were also restored by SIRT1 overexpression in Leydig cells. Additionally, we found that Cd increased the level of methyltransferase enzyme METTL3 and Sirt1 m6A modification in Leydig cells. Mettl3 siRNA effectively restored Cd-enhanced Sirt1 m6A level and reversed Cd-downregulated Sirt1 mRNA expression in Leydig cells. Overall, our findings suggest that Cd exposure inhibits testosterone synthesis via Sirt1 m6A modification-mediated senescence in mouse testes. These results offer an experimental basis for investigating the causes and potential treatments of hypotestosteronemia induced by environmental factors.

BackgroundLymphovascular invasion (LVI) is a significant risk factor for lymph node metastasis in gastric cancer (GC) and is closely related to the prognosis and recurrence of GC. This study aimed to establish clinical models, radiomics models and combination models for the diagnosis of GC vascular invasion.MethodsThis study enrolled 146 patients with GC proved by pathology and who underwent radical resection of GC. The patients were assigned to the training and validation cohorts. A total of 1,702 radiomic features were extracted from contrast-enhanced computed tomography images of GC. Logistic regression analyses were performed to establish a clinical model, a radiomics model and a combined model. The performance of the predictive models was measured by the receiver operating characteristic (ROC) curve.ResultsIn the training cohort, the age of LVI negative (−) patients and LVI positive (+) patients were 62.41 ± 8.41 and 63.76 ± 10.08 years, respectively, and there were more male (n = 63) than female (n = 19) patients in the LVI (+) group. Diameter and differentiation were the independent risk factors for determining LVI (−) and (+). A combined model was found to be relatively highly discriminative based on the area under the ROC curve for both the training (0.853, 95% CI: 0.784–0.920, sensitivity: 0.650 and specificity: 0.907) and the validation cohorts (0.742, 95% CI: 0.559–0.925, sensitivity: 0.736 and specificity: 0.700).ConclusionsThe combined model had the highest diagnostic effectiveness, and the nomogram established by this model had good performance. It can provide a reliable prediction method for individual treatment of LVI in GC before surgery.

When the atomic layers in a non-centrosymmetric van der Waals structure slide against each other, the interfacial charge transfer results in a reversal of the structures spontaneous polarization. This phenomenon is known as sliding ferroelectricity and it is markedly different from conventional ferroelectric switching mechanisms relying on ion displacement. Here, we present layer dependence as a new dimension to control sliding ferroelectricity. By fabricating 3R MoS2 of various thicknesses into dual-gate field-effect transistors, we obtain anomalous intermediate polarization states in multilayer 3R MoS2. Using results from ab initio density functional theory calculations, we propose a generalized model to describe the ferroelectric switching process in multilayer 3R MoS2 and to explain the formation of these intermediate polarization states. This work reveals the critical roles that layer number and interlayer dipole coupling play in sliding ferroelectricity and presents a new strategy for the design of novel sliding ferroelectric devices., Comment: 25 pages, 4 figures

Purpose: To study the clinical effectiveness of the topical application of tranexamic acid in hand tendon release. Methods: This was a randomized controlled trial conducted after receiving approval from the local ethics committee according to guidelines from the Helsinki Declaration. Eighty patients who underwent hand tendon release operation in our hospital from January 2021 to December 2022 were included and randomly divided into 2 groups. Patients in the tranexamic acid group (40 cases) received intraoperative topical application of 2 g of tranexamic acid after tendon release, while patients in the conventional group (40 cases) did not receive topical application of tranexamic acid during operation. The operation time, perioperative hemoglobin changes, total blood loss, incidence of early postoperative complications, and total active movement (TAM) before surgery and 6 months after surgery were compared between the 2 groups.
