Xylanase, an enzyme capable of hydrolyzing non-starch polysaccharides found in grain structures like wheat, has been found to improve the organizational structure of dough and thus increase its volume. In our past work, one promising xylanase FXYL derived from Fusarium oxysporum Fo47 and first expressed 779.64 U/mL activity in P. pastoris. It has shown significant potential in improving the quality of whole wheat bread, making it become a candidate for development as a new flour improver. After optimization of expression elements and gene dose, the xylanase activity of FXYL strain carrying three-copies reached 4240.92 U/mL in P. pastoris. In addition, 12 factors associated with the three stages of protein expression pathway were co-expressed individually in order in three-copies strain, and the translation factor Pab1 co-expression increased FXYL activity to 8893.53 U/mL. Nevertheless, combining the most effective or synergistic factors from three stages did not exhibit better results than co-expressing them alone. To further evaluate the industrial potential, the xylanase activity and protein concentration reached 81184.51 U/mL and 11.8 g/L in a 5 L fed-batch fermenter. These engineering strategies improved the expression of xylanase FXYL by more than 104-fold, providing valuable insights for the cost-effective industrial application of FXYL in the baking field.

Abstract Background COVID-19 is a challenge to education systems worldwide. The aim of the study was to explore the impact of COVID-19-pandemic-related emotions and COVID-19-related concern for education on the study careers and professional futures of nursing students. Methods The study was completed between March and June 2023 using a multi-stage sampling design. A total of 1126 nursing students were recruited to complete the questionnaire. The self-administered questionnaire consisted of basic characteristics of the subjects, the COVID-19-pandemic-related emotions scale, the COVID-19-related concern for education scale, and the impact of the COVID-19 on study careers and professional futures scale (SCPFI-19 S). One-way ANOVA/t-test, correlation coefficient analysis, and hierarchical linear regression analysis were used to explore factors influencing changes in study careers and professional futures, and the relationship between COVID-19-pandemic-related emotions and COVID-19-related concern for education. A series of high-entropy pyrochlore ceramics, specifically (La1/3Nd1/3M1/3)2(Zn1/2Sn1/2)2O7 (M = Sm, Eu, Gd, or Dy), have been synthesized using the solid-state reaction method. Their potential as high-temperature thermistors was investigated by analyzing electrical and aging properties at elevated temperatures. Characterization using X-ray diffraction, scanning electron microscopy, and Raman spectroscopy confirms that these ceramics are dense, single-phase solid solutions with a pyrochlore structure. Electrical analysis demonstrate that these ceramics maintain high resistivity and resistance stability, exhibiting typical negative temperature coefficient features and high B values across a wide temperature range. These characteristics make (La1/3Nd1/3M1/3)2(Zn1/2Sn1/2)2O7 promising candidates for the development of high-sensitivity, long-life high-temperature thermistors suitable for applications within the temperature range of 400–1200 °C.

ObjectivePatients with chronic diseases may have some psychological problems due to their own or surrounding environmental factors, which can adversely affect the patient’s illness and life. Given that the number of chronically ill patients in China is currently increasing every year, more research is needed to determine the best ways to manage changes in psychological status and psychological stress responses in chronically ill patients. The researchers constructed a mediated moderation model to explore the impact of stigma on the quality of life of chronically ill patients, as well as the mediating role of depression and the moderating role of psychological resilience.MethodsA stratified sampling method was used to select 363 middle-aged and old-aged patients with chronic diseases aged 45 years and older from the Affiliated Hospital of Zhejiang University for the study. Data were collected from patients with chronic diseases such as cardiac, respiratory, renal, and other chronic diseases using the Cumulative Illness Rating Scale for Geriatrics (CIRS-G), the Stigma Scale for Patients with Chronic Diseases (SSCI), the Patient Health Questionaire-9 (PHQ-9), the Quality of Life Inventory (SF-12), and the Conner-Davidson Resilience Scale (CD-RISC) were collected from patients with cardiac, respiratory, renal, and other chronic diseases. A descriptive analysis was used to describe the sample. Linear regression was used to evaluate the relationship between the variables. Mediation and moderation analyses were used to explore the mediating role of depression and the moderating role of psychological resilience.ResultsThere was a moderate negative correlation between stigma and quality of life (r = -0.378, P < 0.01). There was a moderate negative correlation between depression and quality of life (r = -0.497, P < 0.01). There was a moderately positive correlation between psychological resilience and quality of life (r = 0.382, P < 0.01). There was a moderate negative correlation between psychological resilience and depression (r = -0.348, P < 0.01). There was a weak negative correlation between psychological resilience and stigma (r = -0.166, P < 0.01). There was a strong positive correlation between stigma and depression (r = 0.607, P < 0.01) The mediation study showed that stigma was a significant predictor of quality of life and that stigma and quality of life were mediated to some extent by depression, with the mediating effect accounting for 67.55% of the total effect. The direct path from stigma to depression is moderated by psychological resilience (β = -0.0018, P < 0.01).ConclusionsDepression mediates the relationship between stigma and quality of life, while psychological elasticity plays a moderating role between stigma and depression, and when the level of psychological elasticity increases, the more significant the role of stigma on depression. As a physiologically and psychologically vulnerable group, patients with chronic diseases’ overall quality of life and mental health should be taken more seriously, and clinical workers should pay timely attention to the psychological and mental conditions of patients with chronic diseases and provide timely and appropriate interventions and therapeutic measures. The relevant results of this study also provide a new perspective for clinical work on psychological intervention for patients with chronic diseases.

Abstract Background Many studies have explored the relationship between self-esteem and quality of life. However, few studies have elucidated the mechanisms underlying the relationship between self-esteem and quality of life in middle-aged and older patients with chronic diseases. The present study aimed to explore the mediating role of death anxiety in this relationship. Methods Middle-old-aged patients with chronic diseases were selected as the respondents by using a multi-stage sampling method, random number table method from October 2021 to February 2022 in the Second Affiliated Hospital of Zhejiang University School of Medicine. The Cumulative Disease Rating Scale, the Self-Esteem Scale (SES), the Chinese version of the Death Anxiety Scale (CT-DAS), and the Simplified version of the Quality of Life Scale (SF-12) were used as the researching tools to conduct the survey. SPSS26.0 was used to analysis data. AMOS 23.0 software was used to construct structural equation modeling. Results 294 valid questionnaires were collected. Abstract Background Nursing is one of the critically important disciplines in healthcare. Due to its complexity and specificity, nursing students often face additional higher education-related stress. However, there is less research on higher education-related stress among nursing students. Therefore, the purpose of this study was to investigate the effects of emotional empathy and emotional intelligence on the dimensions of higher education-related stress among nursing students. Methods This study utilized a multi-stage sampling design and was completed within the March-June 2023 period. A total of 1126 nursing students were recruited to complete the questionnaire. The self-administered questionnaire included the basic characteristics of the subjects, an emotional empathy scale, an emotional intelligence scale, and a higher education-related stress scale. One-way ANOVA/t-tests, correlation coefficients, and hierarchical linear regression analyses were used to explore the factors affecting higher education-related stress and the relationship between emotional empathy and emotional intelligence with the dimensions of higher education-related stress. Results The mean (SD) scores of challenges and dissatisfaction dimensions of higher education-related stress among nursing students were 30.052 (7.466) and 8.270 (2.205), respectively. Emotional empathy was significantly and positively related to the challenges and dissatisfaction dimensions of higher education-related stress. Emotional intelligence was negatively correlated with the challenges dimension of higher education-related stress and positively correlated with the dissatisfaction dimension. Stratified multiple regression analyses revealed that nursing students' emotional empathy and emotional intelligence were significant predictors of the dimensions of higher education-related stress. Conclusion Overall, emotional empathy and emotional intelligence were significantly correlated with all dimensions of higher education-related stress. Consequently, in future interventions, the Chinese government and education sector can develop nursing students' ability to use emotional empathy and emotional intelligence rationally through emotional regulation strategies and emotional intelligence courses, to reduce the higher education-related stress they experience.

In order to study the deformation localization failure characteristics of sandstone under the coupling action of seepage and stress, the triaxial compression tests of sandstone under different drainage conditions were carried out using the visualized triaxial servo control test system combined with three-dimensional digital image correlation (3D-DIC). The rock mechanics, seepage and deformation localization characteristics were analyzed, and the micro-morphology of fracture surface after sandstone failure was analyzed using electron microscope scanning. The results shown that the peak strength and elastic modulus of sandstone under drainage condition were higher than those under undrained condition, the peak strength, elastic modulus and Poisson’s ratio of sandstone increased with the increasing seepage water pressure, the time point of penetration crack and maximum permeability would be advanced. When the seepage water pressure was the same, the localized zone of deformation field cloud map of the sand surface was wider in the undrained condition than that in the drained condition, i.e., the macroscopic crack of rock was more obvious. Water flow under drained condition took away the mineral particles inside the rock and formed holes, the rupture surface of which was smoother than that under undrained condition, whereas flakes cuttings were obviously attached to the surface of the particles under undrained condition. The initiation point of radial deformation localization was consistently higher than that of axial deformation localization for all drainage conditions, with an average increase of 1.23%. The initiation stress levels of the radial and axial deformation localization increased with increasing seepage water pressure, i.e., the initiation time point was earlier. The radial and axial initiation stress level of sandstone under drainage condition were higher than those under undrained condition, with an average increase of 1.85% and 2.21% respectively. When the water pressure was the same, the initiation stress and stress level were more significantly affected by water pressure in the undrained condition than in the drained condition.

Thin-film composite (TFC) polyamide (PA) RO membrane modified with a layer of tethered poly(acrylic acid) (PAA) chains displayed intrinsic rejections of nitrate, boron, As(III), and As(V) of 98.0%, 90.7%, 96%, and 99.6%, respectively. The solute permeability coefficients for nitrate, boron, As(III), and As(V) by the SNS-PAA-PA membrane were lower by 31–38%, 49–63%, 57–72% and 87–93% relative to tested commercial membranes. The above results indicate that the SNS-PAA-PA membrane should be suitable for purification of brackish source water contaminated with nitrate, boron, As(III), and As(V) to levels 1219 ppm, 2 ppm, 95 ppb, and 948 ppb, respectively. The study results suggest that there is merit in further exploration of the potential of the present approach for enhancing RO membranes performance for targeted solute removal.

In recent years, there has been a significant increase in the incidence of Breast cancer (BC), making it the most common cancer among women and a major threat to women’s health. Consequently, there is an urgent need to discover new and effective strategies for treating BC. Ferroptosis, a novel form of cell death characterized by the accumulation of iron-dependent lipid reactive oxygen species, has emerged as a distinct regulatory pathway separate from necrosis, apoptosis, and autophagy. It is widely recognized as a crucial factor in the development and progression of cancer, offering a promising avenue for BC treatment. While significant progress has been made in understanding the mechanisms of ferroptosis in BC, drug development is still in its early stages. Numerous compounds, including phytochemicals derived from dietary sources and medicinal plants, as well as synthetic drugs (both clinically approved medications and laboratory reagents), have shown the ability to induce ferroptosis in BC cells, effectively inhibiting tumor growth. This comprehensive review aims to examine in detail the compounds that target ferroptosis in BC and elucidate their potential mechanisms of action. Additionally, the challenges associated with the clinical application of ferroptosis-inducing drugs are discussed, offering valuable insights for the development of novel treatment strategies for BC.

Tillage modes and straw returning influence soil aggregate stability and the distribution of organic carbon (C) and nitrogen (N) in aggregates of different particle sizes. In the typical hilly regions of southwest China, the predominant soil type is purple soil, characterized by heavy texture and high stickiness, with relatively lower soil fertility compared to other soil types. The improper use of fertilizers and field management practices further exacerbates soil compaction. However, abundant straw resources in the region provide an opportunity for comprehensive straw utilization. The effective utilization of straw resources is of significant importance for stabilizing agricultural ecological balance, improving resource utilization efficiency, and alleviating ecological pressure. Previously, most studies have focused on the impact of different mechanized tillage systems on the physical and chemical properties of soil in hilly areas, while research on the preservation of water-stable aggregates’ organic C and N content remains limited. In this study, the soil properties of fields under a winter pea–summer corn rotation for two years were studied with regards to the effects of straw returning on its water-stable aggregate distribution, macroaggregate content (R0.25), mean weight diameter (MWD), geometric mean diameter (GMD), and the organic C and N content in soil aggregates of different particle sizes and at different depths. The effects of five different tillage modes were assessed, namely rotary tillage with straw mixed retention (RTM), conventional tillage with straw burial retention (CTB), no-tillage with straw covered retention (NTC), subsoiling with straw covered retention (STC), and no-tillage without straw retention (NT). Based on the study results, under different tillage modes, straw returning effectively enhanced the soil organic carbon (SOC) and total nitrogen (TN) reserves at the plow layer (0–30 cm), SOC increased by 17.2% to 88%, and TN increased by 8.6% to 85.9%. At the same time, the content of 0.25–2 mm aggregates increased under the straw-return treatments under different tillage patterns. The NT treatment had the lowest R0.25 and MWD and GMD values for soil aggregates at different depths, which were significantly different (p < 0.05) from the other treatment modes. The correlation coefficients between SOC and soil aggregate stability indices ranged from 0.68 to 0.90, with most of them showing highly significant positive correlations (p < 0.01). In conclusion, straw returning under different tillage systems has improved soil aggregate stability and promoted soil structure stability. Specifically, the STC treatment has shown more pronounced effects on soil improvement in the upper soil layer of the hilly regions in southwest China, while the RTM treatment is beneficial for improving the lower soil layer. Therefore, the comprehensive experimental results indicate that the combination of STC and RTM treatments represents the most promising mechanized tillage and straw returning practices for the hilly regions in southwest China.

Mice lacking Claudin11 (Cldn11) manifest reduced trabecular bone mass. However, the impact of Cldn11 expression in osteoblasts in vivo remains understudied. Herein, we generated osteoblast-specific transgenic (Tg) mice expressing Cldn11 and characterized their skeletal phenotype. Micro-CT analyses of the distal metaphysis of the femur showed a 50% and a 38% increase in trabecular bone mass in Tg male and female mice, respectively, due to a significant increase in trabecular number and a reduction in trabecular separation. Histomorphometry and serum biomarker studies uncovered that increased trabecular bone mass in Cldn11 Tg mice was the consequence of enhanced bone formation. Accordingly, an abundance of bone formation (Alp, Bsp), but not bone resorption (Ctsk), markers were augmented in the femurs of Cldn11 Tg mice. Since the trabecular bone density is known to inversely correlate with the amount of marrow adipose tissue (MAT), we measured the MAT in osmium-tetroxide-labeled bones by micro-CT scanning. We found 86% less MAT in the proximal tibia of the Tg males. Consistently, the expression levels of the adipogenic markers, adiponectin and leptin, were 50% lower in the femurs of the Tg males. Our data are consistent with the possibility that claudin11 exerts anabolic effects in osteoblastic lineage cells that act via promoting the differentiation of marrow stem cells towards osteoblasts at the expense of adipocytes.

ObjectiveTamoxifen is an effective anti-tumor medicine, but evidence has been provided on tamoxifen-related inflammation as well as its impact on gut microbiota. In this study, we aimed to investigate tamoxifen-induced gut microbiota and inflammation alteration.MethodsWe established a BC xenograft mouse model using the MCF-7 cell line. 16S rRNA gene sequencing was used to investigate gut microbiota. qRT–PCR, western blotting, and cytometric bead array were used to investigate inflammation-related biomarkers. Various bioinformatic approaches were used to analyze the data.ResultsSignificant differences in gut microbial composition, characteristic taxa, and microbiome phenotype prediction were observed between control, model, and tamoxifen-treated mice. Furthermore, protein expression of IL-6 and TLR5 was up-regulated in tamoxifen-treated mice, while the mRNA of Tlr5 and Il-6, as well as protein expression of IL-6 and TLR5 in the model group, were down-regulated in the colon. The concentration of IFN-γ, IL-6, and IL12P70 in serum was up-regulated in tamoxifen-treated mice. Moreover, correlation-based clustering analysis demonstrated that inflammation-negatively correlated taxa, including Lachnospiraceae-UCG-006 and Anaerotruncus, were enriched in the model group, while inflammation-positively correlated taxa, including Prevotellaceae_UCG_001 and Akkermansia, were enriched in the tamoxifen-treated group. Finally, colon histologic damage was observed in tamoxifen-treated mice.ConclusionTamoxifen treatment significantly altered gut microbiota and increased inflammation in the breast cancer xenograft mice model. This may be related to tamoxifen-induced intestinal epithelial barrier damage and TLR5 up-regulation.

Mas-related G-protein-coupled receptors X1-X4 (MRGPRX1-X4) are 4 primate-specific receptors that are recently reported to be responsible for many biological processes, including itch sensation, pain transmission, and inflammatory reactions. MRGPRX1 is the first identified human MRGPR, and its expression is restricted to primary sensory neurons. Due to its dual roles in itch and pain signaling pathways, MRGPRX1 has been regarded as a promising target for itch remission and pain inhibition. Here, we reported a cryo-electron microscopy (cryo-EM) structure of Gq-coupled MRGPRX1 in complex with a synthetic agonist compound 16 in an active conformation at an overall resolution of 3.0 Å via a NanoBiT tethering strategy. Compound 16 is a new pain-relieving compound with high potency and selectivity to MRGPRX1 over other MRGPRXs and opioid receptor. MRGPRX1 was revealed to share common structural features of the Gq-mediated receptor activation mechanism of MRGPRX family members, but the variable residues in orthosteric pocket of MRGPRX1 exhibit the unique agonist recognition pattern, potentially facilitating to design MRGPRX1-specific modulators. Together with receptor activation and itch behavior evaluation assays, our study provides a structural snapshot to modify therapeutic molecules for itch relieving and analgesia targeting MRGPRX1.

Fertilization plays a crucial role in enhancing tea production. However, it has been demonstrated that the long-term single application of chemical fertilizer will reduce soil nutrient content and the formation of soil aggregates, which is not conducive to the sustainable development of soil and agriculture. Many studies have shown that partial substitution of chemical fertilizer with organic fertilizer can improve soil physicochemical properties and soil nutrient content. This study compared the effects of different organic materials as substitutes for chemical fertilizer. We partially replaced chemical fertilizer with rabbit manure, wine lees and rapeseed cake, amounting to 30% of the total annual nitrogen application in the field experiment, and we set nine different fertilization methods to assess and analyze the soil nutrient condition, aggregate stability and enzyme activity. The results showed that the experimental soil aggregate mean weight diameter (MWD) and geometric mean diameter (GMD) were significantly increased compared with control (p < 0.05); the aforementioned fertilization methods also decreased the soil aggregate fractal dimension (D), disruption rate (PAD), average weight-specific surface area (MWSSA) and soil erodibility factor (K). The application of the fertilizer containing organic materials and microbial agent increased soil organic carbon (SOC) by 20.7% to 22.6% and total nitrogen (TN) by 34.6% to 38.1%; it also significantly promoted sucrase, urease and protease activities in all aggregate sizes (p < 0.05) and increased the 2–5 mm aggregate content. The correlation coefficients between the SOC and the enzyme activities were 0.18–0.95, and most of them showed an extremely significant positive correlation (p < 0.01). In conclusion, the application of fertilizers containing organic materials and microbial agents can improve soil aggregate stability, aggregate enzyme activity and soil structural stability.

The process of producing sustainable aviation fuel (SAF) from organic waste involves the use of volatile fatty acids (VFAs) as intermediates that are obtained via arrested anaerobic digestion (AAD) and VFA recovery. This recovery process often requires several steps, including dewatering, filtration, extraction, and purification. The recovery of VFAs is crucial for their upgrading and can pose a challenge in the production of SAF from organic waste due to high costs and compatibility issues. This review discusses various dewatering methods, including centrifuges, belt filter presses, and screw presses, and explores conditioning technologies that can improve dewatering performance. It also introduces filtration technologies, with a focus on dynamic filtration, which shows promise in addressing the issue of membrane fouling. Additionally, the review describes extraction technologies such as electrodialysis, adsorption, and liquid–liquid extraction (LLE). By providing insights into these different techniques, the review aims to contribute to the development of an integrated VFA recovery process with low carbon footprint.

Due to its renewable nature, its inherent strength, and many other favorable attributes, nanocellulose (NC) has drawn increasing attention for many potential applications. A diverse and complex assortment of NC products have been reported, and these are most commonly classified based on some contrasting procedures of preparation. The research community is facing a continuing challenge to adequately measure and quantify morphological features of various NC products. In principle, it ought to be possible to quantify and name NC based on such attributes as “degree of branching,” “breadth of particle size,” and “aspect ratio distribution,” etc. However, the ability to measure and compute such quantities still lies beyond what can be achieved in practical amounts of time in typical laboratories. Meanwhile, there has been tension between researchers proposing additional descriptive names, while at the same time there have been efforts at achieving uniformity and simplicity in nomenclature. It is proposed in this essay that this state of affairs is largely a reflection of complexity itself, such that NC products that have the same nominal description can be very different from each other when examined closely. The diversity itself may turn out to be a good thing, as researchers work to come up with varieties of NC that can survive an expected relentless competition from existing plastic-based or cellulose-based materials.

To study the role of Claudin (CLDN)12 in bone, we developed mice with a targeted deletion of exon2 in the Cldn12 gene for skeletal phenotype analysis. Micro-CT analysis of the secondary spongiosa of distal femurs of mice with targeted disruption of the Cldn12 gene and control littermates showed no significant genotype-specific differences in either cortical or trabecular bone parameters for either gender in 13-week-old mice. Immunohistochemistry revealed that while CLDN12 was expressed in both differentiating chondrocytes and osteoblasts of the secondary spongiosa of 3-week-old wild-type mice, its expression was restricted to differentiating chondrocytes in the articular cartilage and growth plate in adult mice. Articular cartilage area at the knee were increased by 47% in Cldn12 knockout (KO) mice compared to control littermates. Micro-CT analyses found that while the trabecular number was increased by 9% and the trabecular spacing was reduced by 9% in the femoral epiphysis of Cldn12 KO mice, neither bone volume nor bone volume adjusted for tissue volume was different between the two genotypes. The expression levels of Clusterin, Lubricin and Mmp13 were increased by 56%, 46%, and 129%, respectively, in primary articular chondrocytes derived from KO compared to control mice. Our data indicate that targeted deletion of the Cldn12 gene in mice increases articular cartilage, in part, by promoting articular chondrocyte phenotype.

We previously demonstrated that mice with targeted deletion of the leucine repeat rich kinase 1 (Lrrk1) gene were osteopetrotic due to the failure of osteoclasts to resorb bone. To determine how LRRK1 regulates osteoclast activity, we examined the intracellular and extracellular acidification with an acidotropic probe, acridine orange, in live osteoclasts on bone slices. We examined lysosome distribution in osteoclasts by localization of LAMP-2, cathepsin K, and v-ATPase by immunofluorescent staining with specific antibodies. We found that both vertical and horizontal cross-sectional images of the wild-type (WT) osteoclasts showed orange-staining of the intracellular acidic vacuoles/lysosomes dispersed to the ruffled border. By contrast, the LRRK1 deficient osteoclasts exhibited fluorescent orange staining in the cytoplasm away from the extracellular lacunae because of an altered distribution of the acidic vacuoles/lysosomes. In addition, WT osteoclasts displayed a peripheral distribution of LAMP-2 positive lysosomes with a typical actin ring. The clustered F-actin constitutes a peripheral sealing zone and a ruffled border which was stretched out into a resorption pit. The LAMP-2 positive lysosomes were also distributed to the sealing zone, and the cell was associated with a resorption pit. By contrast, LRRK1-deficient osteoclasts showed diffused F-actin throughout the cytoplasm. The sealing zone was weak and not associated with a resorption pit. LAMP-2 positive lysosomes were also diffuse in the cytoplasm and were not distributed to the ruffled border. Although the LRRK1-deficient osteoclast expressed normal levels of cathepsin K and v-ATPase, the lysosomal-associated cathepsin K and v-ATPase were not accumulated at the ruffled border in Lrrk1 KO osteoclasts. Our data indicate that LRRK1 controls osteoclast activity by regulating lysosomal distribution, acid secretion, and protease exocytosis.

There is little evidence regarding the effectiveness of procedural interventions for treatment of pain after an acute herpes zoster (AHZ) infection before the development of postherpetic neuralgia (PHN). In our case, a 61-year-old male presented with 1 month of left foot pain following an AHZ infection. After 1 month of pain refractory to treatment and admission to our hospital for acute pain management, a nerve stimulator was placed at the left first sacral (S1) dorsal root ganglion (DRG), which significantly decreased his pain despite his ongoing dermatologic manifestations of AHZ. In conclusion, we describe a case of nerve stimulator placement at the S1 DRG as a successful treatment for intractable pain following an AHZ infection.

The gypsum and calcite scaling propensities of the thin-film composite polyamide (PA-TFC) reverse osmosis (RO) membrane, modified with a tethered surface layer of polyacrylic acid (PAA) chains, was evaluated and compared to the scaling of selected commercial RO membranes. The tethered PAA layer was synthesized onto a commercial polyamide membrane (i.e., base-PA) via atmospheric pressure plasma-induced graft polymerization (APPIGP). The PAA nano-structured (SNS) base-PA membrane (SNS-PAA-PA) was scaled to a lesser degree, as quantified by a lower permeate flux decline and surface imaging, relative to the tested commercial membranes (Dow SW30, Toray SWRO, and BWRO). The cleaning of gypsum-scaled membranes with D.I. water flushing achieved 100% water permeability recovery for both the SNS-PAA-PA and Dow SW30 membranes, relative to 92–98% permeability restoration for the Toray membranes. The calcium carbonate scaling of SNS-PAA-PA membranes was also lower relative to the commercial membranes, but permeability recovery after D.I. water cleaning was somewhat lower (94%) but consistent with the level of surface scale coverage. In contrast, the calcite and gypsum-scaled membrane areas of the commercial membranes post-cleaning were significantly higher than for the SNS-PAA-PA membrane but with 100% permeability recovery, suggesting the potential for membrane damage when mineral scaling is severe.

We previously showed that conditional disruption of the Phd2 gene in chondrocytes led to a massive increase in long bone trabecular bone mass. Loss of Phd2 gene expression or inhibition of PHD2 activity by a specific inhibitor resulted in a several-fold compensatory increase in Phd3 expression in chondrocytes. To determine if expression of PHD3 plays a role in endochondral bone formation, we conditionally disrupted the Phd3 gene in chondrocytes by crossing Phd3 floxed (Phd3flox/flox) mice with Col2α1-Cre mice. Loss of Phd3 expression in the chondrocytes of Cre+; Phd3flox/flox conditional knockout (cKO) mice was confirmed by real time PCR. At 16 weeks of age, neither body weight nor body length was significantly different in the Phd3 cKO mice compared to Cre−; Phd3flox/flox wild-type (WT) mice. Areal BMD measurements of total body as well as femur, tibia, and lumbar skeletal sites were not significantly different between the cKO and WT mice at 16 weeks of age. Micro-CT measurements revealed significant gender differences in the trabecular bone volume adjusted for tissue volume at the secondary spongiosa of the femur and the tibia for both genotypes, but no genotype difference was found for any of the trabecular bone measurements of either the femur or the tibia. Trabecular bone volume of distal femur epiphysis was not different between cKO and WT mice. Histology analyses revealed Phd3 cKO mice exhibited a comparable chondrocyte differentiation and proliferation, as evidenced by no changes in cartilage thickness and area in the cKO mice as compared to WT littermates. Consistent with the in vivo data, lentiviral shRNA-mediated knockdown of Phd3 expression in chondrocytes did not affect the expression of markers of chondrocyte differentiation (Col2, Col10, Acan, Sox9). Our study found that Phd2 but not Phd3 expressed in chondrocytes regulates endochondral bone formation, and the compensatory increase in Phd3 expression in the chondrocytes of Phd2 cKO mice is not the cause for increased trabecular bone mass in Phd2 cKO mice.

This paper reports on the preparation of cellulose/reduced graphene oxide (rGO) aerogels for use as chemical vapour sensors. Cellulose/rGO composite aerogels were prepared by dissolving cellulose and dispersing graphene oxide (GO) in aqueous NaOH/urea solution, followed by an in-situ reduction of GO to reduced GO (rGO) and lyophilisation. The vapour sensing properties of cellulose/rGO composite aerogels were investigated by measuring the change in electrical resistance during cyclic exposure to vapours with varying solubility parameters, namely water, methanol, ethanol, acetone, toluene, tetrahydrofuran (THF), and chloroform. The increase in resistance of aerogels on exposure to vapours is in the range of 7 to 40% with methanol giving the highest response. The sensing signal increases almost linearly with the vapour concentration, as tested for methanol. The resistance changes are caused by the destruction of the conductive filler network due to a combination of swelling of the cellulose matrix and adsorption of vapour molecules on the filler surfaces. This combined mechanism leads to an increased sensing response with increasing conductive filler content. Overall, fast reaction, good reproducibility, high sensitivity, and good differentiation ability between different vapours characterize the detection behaviour of the aerogels.

Chronic pain begins with acute pain. Physicians tend to classify pain by duration (acute vs chronic) and mechanism (nociceptive, neuropathic and nociplastic). Although this taxonomy may facilitate diagnosis and documentation, such categories are to some degree arbitrary constructs, with significant overlap in terms of mechanisms and treatments. In clinical practice, there are myriad different definitions for chronic pain and a substantial portion of chronic pain involves mixed phenotypes. Classification of pain based on acuity and mechanisms informs management at all levels and constitutes a critical part of guidelines and treatment for chronic pain care. Yet specialty care is often siloed, with advances in understanding lagging years behind in some areas in which these developments should be at the forefront of clinical practice. For example, in perioperative pain management, enhanced recovery protocols are not standardized and tend to drive treatment without consideration of mechanisms, which in many cases may be incongruent with personalized medicine and mechanism-based treatment. In this educational document, we discuss mechanisms and classification of pain as it pertains to commonly performed surgical procedures. Our goal is to provide a clinical reference for the acute pain physician to facilitate pain management decision-making (both diagnosis and therapy) in the perioperative period. [ABSTRACT FROM AUTHOR]
