Objective: Given the association of inflammatory conditions with the development of comorbidities, particularly cardiovascular diseases, it is crucial to monitor inflammation in hemodialysis patients. This study aimed to evaluate plasma gelsolin and myeloperoxidase (MPO) levels before and after dialysis sessions and to assess their relationship with inflammation. Materials and Methods: The study included 16 healthy volunteers and 30 patients receiving regular hemodialysis treatment. Along with routine biochemical analyses, plasma gelsolin and MPO levels were measured in blood samples taken from the study group before and after the sessions. Results: Plasma gelsolin levels were found to be statistically higher both before and after dialysis compared to the control group (p=0.000); however, there was no significant change during the session (p=0.094). Conversely, plasma MPO activity, which was significantly higher before dialysis, increased at the end of the session (p=0.000). Conclusion: It can be concluded that elevated levels of gelsolin are associated with a chronic inflammatory response, as indicated by high-sensitivity C-reactive protein (hs- CRP) and MPO levels. Consequently, gelsolin could be considered a supportive treatment strategy for these patients. [ABSTRACT FROM AUTHOR]

11. The Effects of Genistein as Supplement to Oral/Enteral Nutrition on Inflammatory Cytokines in Septic ICU patients: A Prospective, Single-center, Randomized Controlled Pilot Study.

Objective: Genistein has an anti-inflammatory effect that may be beneficial in many inflammatory diseases. The aim of this study was to determine the effects of supplementation of oral/enteral nutrition (EN) with genistein on the level of inflammatory cytokines in septic patients. Materials and Methods: This prospective, randomized controlled study included critically ill adult patients with sepsis receiving EN or oral/EN. The patients were randomly divided into a genistein or a control group. Genistein (60 mg/day) was administered as a supplement to EN in the genistein group and the control group received only EN or oral/EN. Serum interleukin 1 beta (IL-1β), interleukin 6 (IL-6), high mobility group box 1 protein (HMGB-1), tumor necrosis factor alfa (TNF-α), procalcitonin (PCT), and C-reactive protein (CRP) concentrations were serially measured at the baseline, 24th hour, and 72
nd hour after admission. Results: Thirty-two participants (genistein group: 16 patients, control group: 16 patients) were included. The mean age was 56±17 years. The serum IL-1β concentration in the genistein group was significantly higher than that of the control group in follow-up (p=0.001). The control group had a significantly lower serum IL-6 value at the 72nd hour compared with the baseline and 24th -hour values (p=0.001). The TNF-α concentration was significantly greater (p<0.001, both groups), while the PCT values were lower in follow-up measurements (genistein group: p=0.031; control group: p=0.004). The CRP level was higher in the genistein group than in the control group at the baseline (p=0.019) and significantly lower in follow-up measurements (p=0.028). At all of the study time points, the serum IL-6, TNF-α, HMGB-1, and PCT level of the genistein group was similar to that of the control group. Conclusion: Genistein supplementation may add to the inflammation process and worsen the prognosis of sepsis patients in the acute period. [ABSTRACT FROM AUTHOR]- Published

BACKGROUND: Very high mortality rate in sepsis may be related to oxidative stress. This study was conducted on the rats to investigate the presence of oxidative stress and also the potential protective effects of the ß-glucan in the intra-abdominal sepsis model formed by cecal ligation-perforation (CLP). METHODS: In this study, 30 Male rats were equally divided into three groups as “Sham”, “Sepsis” and “ß-Glucan”. Only laparotomy was performed in the Sham group, and sepsis was induced by CLP in Sepsis and ß-Glucan groups. Following CLP, a single dose of 4 mg ß-glucan/kg was also intraperitoneally administered to the β-Glucan group. Blood and tissue (liver, lung and kidney) samples were taken from Sepsis and ß-Glucan groups after sepsis development determined at the end of the 48th hour, also from the Sham group. The levels of myeloperoxidase (MPO) and advanced oxidation protein products (AOPP) were determined in plasma samples, and the malondialdehyde (MDA) was measured in plasma and tissues. - Abstract
Urine may be a waste product, but it contains an enormous amount ofinformation. Well-standardized procedures for collection, transport,sample preparation and analysis should become the basis of an effective diagnostic strategy for urinalysis. As reproducibility of urinalysis hasbeen greatly improved due to recent technological progress, preanalytical requirements of urinalysis have gained importance and have becomestricter. Since the patients themselves often sample urine specimens,urinalysis is very susceptible to preanalytical issues. In order to increasethe reliability of the test results and to reduce the cost, the precautionarymeasure and prevention is very important in the preanalytical phase. Thelaboratory must monitor and evaluate the overall quality of the preanalytic systems and correct identified problems for each specialty and subspecialty of testing performed. The laboratory must establish and followwritten policies and procedures for an ongoing mechanism to monitorassess, and when indicated, correct problems identified in the preanalytic systems. The preanalytic systems assessment must include a review ofthe effectiveness of corrective actions taken to resolve problems, revisionof policies and procedures necessary to prevent recurrence of problems,and discussion of preanalytic systems assessment reviews with appropriate staff. The laboratory must document all preanalytic systems assessment activities. An effective diagnostic strategy from urine should bebased on standard procedures for collection, transport and analysis. Thetime of specimen collection must be recorded both on the examinationrequest and on the subsequent report to aid in the correct interpretationof findings.

Background: This case-control study was conducted to investigate the relationship between serum nesfatin-1 levels and nutritional status and blood parameters in patients diagnosed with metabolic syndrome.Methods: Thirty patients (case) diagnosed with metabolic syndrome according to National Cholesterol Education Program-Adult Treatment Panel III criteria were included. Thirty healthy subjects (control) matched with patients with metabolic syndrome in terms of age, gender and body mass index were included. Three-day food consumption records were obtained. Anthropometric indices were measured and body composition was determined by bioelectrical impedance method. Biochemical parameters and serum nesfatin-1 levels were measured after 8 hours of fasting.Results: Serum nesfatin-1 levels were 0.245±0.272 ng/mL in the case group and 0.528±0.987 ng/mL in the control group (p>0.05). There was a positive significant correlation between serum nesfatin-1 levels and body weight, waist and hip circumferences in the case group (p<0.05). Each unit increase in hip circumference measurement affects the levels of nesfatin by 0.014 times. In the control group, there was a positive significant correlation between body weight and serum nesfatin-1 levels (p<0.05). A significant correlation was detected between HbA1c and serum nesfatin-1 levels in the case group (p<0.05). A significant relationship was detected between dietary fibre intake and the serum nesfatin-1 levels in the case group (p<0.05).Conclusions: Anthropometric indices and blood parameters were correlated with serum nesfatin-1 levels in patients with metabolic syndrome. 24. Effects of thymoquinone in prevention of experimental contrast-induced nephropathy in rats.

Objective(s): This study aimed to show the effects of thymoquinone, which is known for its antioxidant, anti-inflammatory, and renal protective effects in contrast-induced nephropathy. Materials and Methods: This is an experimental study in rats. 7 groups were included within the scope of our study: sham-vehicle (n=3), premedication-control (n=6), model (n=6), isolated thymoquinone (n=3+3), low-dose thymoquinone (n=6), and high-dose thymoquinone (n=7). In addition to 48 hr of water deprivation, we pre-medicated the rats with intra-peritoneal indomethacin and L-NAME administration. After premedication, 12.5 ml/kg dose of a high osmolar contrast agent-diatrizoat (Urografin %76) was administrated. Thymoquinone was administrated in two different doses of 1 mg/kg and 1.75 mg/kg for four days intraperitoneally. Renal functions, histopathological differences, oxidative stress parameters, and inflammatory indicators of rats were evaluated at the end of the study. Results: Significant decreases were observed in levels of serum creatinine and serum BUN with low-dose thymoquinone (1 mg/kg) administration. In light microscopy, significantly less histopathological damage was observed in the low-dose thymoquinone group compared to the contrast agent group. While high-dose thymoquinone is accepted as ineffective biochemically, toxic evidence was identified histopathologically. There were no significant differences between M and TA groups for serum MDA and SOD levels, which were compared to evaluate oxidative stress (P:0.99, P:0.98; respectively). TNF-α, iNOS, and NF-кB gene expressions were not significantly different between all groups (P:0.748, P:0.531, P:0.910; respectively). Conclusion: This experimental study has demonstrated for the first time the protective effect of the TQ substance for CIN in 1 mg/kg dose, in the accompaniment of biochemical and histopathological data in rats. Objective To investigate the effect of pretreatment with obestatin (OB), an endogenous hormone also found in mother's milk, in experimental necrotizing enterocolitis (NEC). Study Design Pups were randomized into four groups: control, OB-control, NEC, and OB-NEC. NEC was induced by asphyxia and hypothermia in the NEC and OB-NEC groups. OB was administered to the OB-control and OB-NEC groups. Macroscopic scoring of the intestinal tract was evaluated and tissue samples were obtained for histopathological and biochemical examination on the fourth day. Results OB improved the macroscopic appearance of the gut and the clinical score during the experiment (p < 0.05). The rate of occurrence of NEC in the OB-NEC group was lower than the NEC group (p = 0.001). OB prevented necrosis and reduced the number of apoptotic cells in the OB-NEC group compared with the NEC group (p = 0.006). Furthermore, interleukin-6 and malondialdehyde levels in the OB-NEC group were lower than the NEC group (p < 0.05). Conclusion OB reduced intestinal damage and prevented necrosis through anti-inflammatory and antiapoptotic effects in experimental NEC. This effect of OB should be confirmed in clinical studies. Furthermore, future research should investigate whether OB plays a role in NEC pathogenesis or NEC is associated with OB levels in the serum and in breast milk. [ABSTRACT FROM AUTHOR]

Background and Design: Mycosis fungoides (ME) is a cutaneous T cell lymphoma. The clinical and histopathological diagnosis of early mycosis fungoides is usually difficult. There is no special laboratory method for the diagnosis of ME and this is the most important problem in diagnosis and follow up the effectiveness of treatment. Adenosine deaminase (ADA) activity is a non-specific marker of T cell activation. In the present study, we aimed to investigate the levels of plasma and tissue ADA in patients with mycosis fungoides and to determine if ADA is an activation criterion for this disease.

This study was made to research the effect of vitamin E on muscle damage, oxidant and antioxidant levels of runners. 28 students participated in the study as the experiment group at the age of 20,75±1,84 (n=16) and the control group (n=12) at the age of 20,42±1,78 who are physically active, have the similar physical features and education at Erciyes University. In this study, the same exercises were performed on both groups three times a week for thirty days. 400 IU (268 mg) vitamin E was given to the athletes in experiment group in every day for 30 days. Nothing was given to the control group. The body weights and the percentage of body fat were measured at the pre and post one month exercise. Total antioxidant capacity, malondialdehyde, creatine kinase and lactate dehydrogenase were measured by making biochemical analyses. While any significant difference on pre-exercise CK and LDH levels, pre and post exercise TAC and MDA levels of experiment and control groups was not found, there was a significant difference on post exercise CK and LDH levels as a result of the intergroup comparison. As for the intragroup comparison, there was a significant increase on post exercise MDA level of both experiment and control group. This result was thought that the antioxidant capacity remains incapable; despite usage of vitamin E counter to oxidative stress increasing connectedly exercise. The reason of this inability in the antioxidant defense system is thought that this is because of the amount of daily vitamin E. Bu çalışma, atletlerde E vitamini kullanımının kas hasarı, oksidan ve antioksidan kapasite üzerine etkisinin araştırılması amacıyla yapılmıştır. Calışmaya Erciyes Universitesi Beden Eğitimi ve Spor Yüksekokulunda okuyan aktif olarak spor yapan, benzer fiziksel özelliklere sahip, yaşları 20,75 ± 1,84 olan (n=16) deney grubu ve yaşları 20,42 ± 1,78 olan (n=12) kontrol grubu olmak üzere 28 kişi katıldı. Calışmada her iki gruba da aynı egzersizler haftada 3 gün olmak üzere 30 gün sureyle uygulandı. Deney grubundaki atletlere 30 gün boyunca her gün 400 IU (268mg) E vitamini verildi. Kontrol grubundaki atletlere herhangi bir şey verilmedi. Bir aylık antrenman öncesi ve sonrası atletlerin vücut ağırlığı ve vücut yağ yüzdesi değerleri ölçüldü. Biyokimyasal analizler ile total antioksidan kapasite, malondialdehit, kreatin kinaz ve laktat dehidrojenaz parametrelerine bakıldı. Deney ve kontrol gruplarının gruplar arası karşılaştırmaları sonucunda egzersiz öncesi CK ve LDH değerlerinde, egzersiz öncesi ve sonrası TAC ve MDA değerlerinde anlamlı farklılık bulunamazken (p>0,05), egzersiz sonrası CK ve LDH değerlerinde anlamlı farklılık meydana geldi (p≤0.05) Grup ici karşılaştırmada ise, hem deney grubunun hem de kontrol grubunun egzersiz sonrası MDA değerinde anlamlı derecede artış saptanmıştır. Bu sonuç, egzersize bağlı olarak artan oksidatif strese karşı, E vitamini kullanımına rağmen antioksidan kapasitenin yetersiz kaldığını düşündürmektedir. Antioksidan savunma sistemindeki bu yetersizliğin nedeni olarak gunluk alınan E vitamini miktarı düşünülmektedir.

Objective: To investigate the changes in otoacoustic emissions (OAEs) for detecting the ototoxicity of cisplatin and to study the possible preventive effect of N-acetylcysteine (NAC) in cisplatin ototoxicity. Materials and Methods: This study was conducted on 21 Wistar albino rats in four groups. Cisplatin, cisplatin plus NAC, NAC alone, and saline were intraperitoneally administered. The rats were anesthetized to study the otoacoustic emissions before and after the experiment. Results: The OAEs were attenuated in the cisplatin group: (-1.31/-4.90), (6.28/4.58), (4.00/0.93), (2.73/-3.74), (2.66/-4.53). The group that received NAC in addition to cisplatin had better OAEs. Conclusion: Cisplatin ototoxicity can be detected by OAE testing in rats, and NAC may reduce the ototoxic effect. [ABSTRACT FROM AUTHOR]

En yaygın görülen ve klinik kullanımını sınırlayan yan etkisi nefrotoksisitedir. Son yıllarda, nefrotoksisite gelişimine katılan en önemli mekanizmaların oksidatif stres ve enflamasyon olduğu öne sürülmektedir.N-asetilsistein (NAC), doğrudan radikal yakalayıcı olarak ve/veya hücre içi redükte glutatyon düzeylerini artırarak etki gösteren bir antioksidandır. Bu çalışmada, NAC'ın ratlarda sisplatinle indüklenen nefrotoksisite üzerine etkilerinin araştırılması amaçlandı. Bu amaçla, ratlar her grupta sekiz rat olmak üzere, dört gruba ayrıldı: KONT, NAC-250, CP ve CP+NAC. Sisplatin uygulaması intraperitoneal (ip) tek doz, 10 mg/kg rat ağırlığı ve NAC uygulaması ip, ardışık 3 gün, 250 mg/kg rat ağırlığı şeklinde yapıldı. Nefrotoksisite gelişimi plazma BUN, kreatinin ve idrar mikroalbümin seviyeleri ile belirlendi. Ayrıca böbrek dokularında histolojik değerlendirme yapılarak da nefrotoksisite tanısı desteklendi. Kan ve doku örneklerinde miyeloperoksidaz (MPO), nükleer faktör-kappa B (NF-B) ve `high mobility group box-1` (HMGB-1) seviyeleri ölçüldü. Sisplatinin plazma ve doku örneklerinde MPO, NF-B ve HMGB-1 düzeyleri üzerinde etkili olmadığı, ancak NAC'ın MPO ve HMGB-1 doku seviyelerini anlamlı derecede artırdığı gözlendi. Bununla birlikte, NAC böbreklerdeki fonksiyonel ve histolojik değişiklikleri iyileştirdi. The most common side effect that limits its clinical usage is nephrotoxicity. In recent years it has been suggested that the most important mechanisms that contribute to nephrotoxicity are oxidative stress and inflammation. N-acetylcysteine (NAC) is an antioxidant, that acts directly as a free radical scavengers and/or increase intracellular reduced glutathione. In the present study, the effects of NAC is investigated, on cisplatin induced nephrotoxicity in rats. For this purpose, rats were divided into four groups each group including eight rats: CONT, NAC-250, CP, CP+NAC. Cisplatin was administrated intraperitoneally (ip), single dose,10 mg/kg body weight and NAC was administrated ip, 250 mg/kg body weight for three consequitive days. Nephrotoxicity was determined by plasma BUN, creatinine and urine microalbumin levels. Nephrotoxicity was also supported by histological analysis in kidney tissue samples. In blood and tissue samples myeloperoxidase (MPO), nuclear factor-kappa B (NF-B) and high mobility group box-1 (HMGB-1) levels were measured. It was found out that cisplatin was not effective on MPO, HMGB-1 and NF-B levels in blood and tissue samples, however, NAC elevated MPO and HMGB-1 levels significantly. Nevertheless NAC ameliorated histological and functional changes in kidney tissues. In conclusion, it can be said that inflammation has limited effect on cisplatin nephrotoxicity in this experimental design. However NAC, which can ameliorate renal function without effecting the levels of NF-kB and HMGB-1 that have versatile and opposite effects on different cancer cell lines, is suggested to be safely used to reduce the side effects of cisplatin without altering chemotherapy efficacy for all cancer types.Keywords: Rats, cisplatin, oxidative stress, inflammation, N-acetylcysteine 116

Amaç: Sisplatin (cis-diamindikloroplatinum (II), CP) nefrotoksisitesi geliştirilen ratlara uygulanan N-asetilsistein (NAC)'in kalp dokusu üzerine koruyucu etkilerinin, çeşitli biyokimyasal ve histopatolojik belirteçlerle gösterilmesi amaçlanmıştır.Bu çalışma CP kardiyotoksisitesinin patogenezinde oksidatif ve nitrozatif stresin ortaya konulması, NAC'ın olası yan etkiler üzerine etkilerini ve sisplatin kardiyotoksisitesinin belirlenmesinde yeni biyomarkerların kullanılabilirliğini değerlendirmek amacıyla planlandı. Gereç ve yöntem: Deneysel ve Klinik Araştırma Merkezi'nden temin edilen 32 adet 300-400 gr ağırlığında Wistar albino erkek rat dört gruba ayrıldı. Tüm uygulamalar kg rat ağırlığı/gün, ip olacak şekilde: Kontrol grubuna SF (1 ml); CP, (10 mg) tek doz; NAC, (250 mg) üç doz; CP-NAC (tek doz 10 mg CP ve ardışık 3 doz 250 mg NAC, CP uygulamasından 4 saat sonra) verildi. Son uygulamayı izleyen 72. saatte, anestezik ratların heparinize kanları ve kalpleri alındı. Kalp dokuları nötral formalinle tespit edildi, histopatolojik olarak değerlendirildi. cTnI (kardiyak troponin I), kardiyak miyozin hafif zincir-1 (CMLC-1) ve iskemi modifiye albumin (İMA) plazmada; total lipid hidroperoksit (ROOH), 4-hidroksinoneal (4-HNE), 3-Nitrotirozin (3-NT), tiyol, total antioksidan (TAK) ve total oksidan (TOK) kapasite kalp dokusunda ölçüldü.Bulgular: Plazma, cTnI, CMLC-1, İMA ve doku ROOH, 4-HNE, 3-NT, TOK, düzeylerinin Kontrole göre CP grubunda arttığı; doku tiyol ve TAK düzeylerinin ise azaldığı belirlendi. Histolojik olarak CP grubunda hemoraji, interstisyel ödem ve vakuolizasyon gözlemlendi. CP grubuyla kıyaslandığında CP-NAC grubunda TAK ve tiyol değerlerinin arttığı belirlendi. Azalan cTnI, CMLC-1, İMA ve doku ROOH, 4-HNE, 3-NT, TOK düzeyleri ve histopatolojik skorlar istatiksel olarak anlamlı bulunamadı. Sonuç: CP kardiyotoksisitesi gelişiminde lipid peroksidasyonun (yüksek 4-HNE, ROOH düzeyleri) ve protein oksidasyonun (yüksek 3-NT düzeyleri) rol oynadığı söylenebilir. Düşük TAK ve tiyol ve yüksek TOK düzeyleri de bu görüşü desteklemektedir. CP kardiyotoksisitesini önlemede, bu doz ve sürede uygulanan NAC'ın antioksidan kapasiteyi artırmasına rağmen (yüksek tiyol ve TAK); gerek plazma gerekse doku düzeyinde başarısız olduğu söylenebilir. Aim: Objective is to demonstrate the protective effects of N-acetylcysteine (NAC) on heart tissue, which is applied to rats which developed cisplatin (cis-diaminedichloroplatinum (II), CP) induced nephrotoxicity, through various biochemical and histopathological indicators.The study thereof has been planned in order to evaluate the usability of new biomarkers in determining CP cardiotoxicity, and to evaluate the effects of NAC on possible side effects, and to demonstrate the oxidative and nitrosative stress regarding the CP cardiotoxicity's pathogenesis. Material and method: 32 male Wistar albino rats, each one weighing 300-400 grams and which are procured from Experimental and Clinical Research Center, have been separated into 4 groups. All applications being kg rat weight/day, ip: Control group has been applied: saline (1 ml); CP, (10 mg) single dose; NAC, (250 mg) three doses; CP-NAC (single dose 10 mg CP and successive 3 doses 250 mg NAC, 4 hours after CP application). In the 72.hour following the last practice, heparinized bloods and hearts of the rats have been taken. Heart tissues have been fixed by neutral formalin, and have been evaluated histopathologically. cTnI (cardiac troponin I), cardiac myosin light chain-1 (CMLC-1) and ischemia modified albumin(IMA) in plasma; total lipid hydroperoxide(ROOH), 4-hydroxynonenal (4-HNE), 3-Nitrotyrosine(3-NT), thiol, total antioxidant (TAC) and total (TOC) capacity has been measured on heart tissue.Results: Plasma, cTnI, CMLC-1, IMA and tissue ROOH, 4-HNE, 3-NT, TOC, levels are found to have increased in CP group; tissue thiol and TAC levels are found to have reduced. Histologically on CP group; haemorrhage, interstitial oedema and vacuolization have been observed. When compared to CP group; CP-NAC group is found to have increased in TAC and thiol values. Reduced cTnI, CMLC-1, IMA and tissue ROOH, 4-HNE, 3-NT, TOC levels and histopathological scores did not yield any histopathologically meaningful results. Conclusion: In development of CP cardiotoxicity; lipid peroxidation (high 4-HNE, ROOH levels) and protein oxidation(high 3-NT levels) can be said to have been effective. Low TAC and thiol and high TOC levels as well support this opinion. In preventing CP cardiotoxicity, despite the NAC which was applied at specified dose and duration increasing the antioxidant capacity (high thiol and TAC); it can be said to have failed. 110

42. Deneysel hipertiroidi oluşturulan ratlarda karnozinin antioksidan özelliklerinin incelenmesi

Tiroid hormonlarının (L-tiroksin, L-triiyodotironin) aşırı üretimi sonucu oluşan hipertiroidinin, mitokondriyal solunumu hızlandırarak serbest radikal üretimini indüklediği ve oksidatif hasara yol açarak çeşitli hastalıkların etiyopatogenezinde rol oynadığı ileri sürülmektedir. Endojen bir dipeptid olan L-karnozinin antioksidan etkiler gösterdiği bildirilmiştir. Yapılan literatür taramasında, hipertiroidi oluşturulan ratlarda L-karnozinin antioksidan özelliklerini inceleyen bir çalışmaya rastlanılamamıştır.Bu çalışmada; deneysel hipertiroidi oluşturulan ratlarda, hipertiroidinin oksidan/antioksidan sistemler üzerine etkisi ve karnozin varlığında bu etkinin ne şekilde değişebileceği araştırıldı. Bu amaçla her biri 12 rattan (Wistar albino) oluşan sekiz çalışma grubu oluşturuldu. Deney gruplarındaki ratlara 10/20 gün süreyle L-tiroksin (intraperitoneal: ip, 300 µg /kg vücut ağırlığı/gün) ile pre-, post- ve kombine L-karnozin (ip, 300 mg/kg vücut ağırlığı/gün) uygulandı. Ratlardan elde edilen plazma ve dokularda (karaciğer, böbrek kalp); malondialdehit (MDA), protein karbonil bileşikleri (PCC), protein ileri oksidasyonu ürünleri (AOPP) (sadece plazmada) ve tiyol seviyeleri ölçüldü. Hipertiroidinin, yüksek MDA, PCC, AOPP ve düşük tiyol düzeyleriyle yansıtıldığı üzere lipid ve protein oksidasyonu üzerinden oksidatif strese neden olduğu, karnozin varlığında ise MDA, PCC ve AOPP düzeylerinin düştüğü ve tiyol düzeylerinin arttığı tespit edildi. Sonuç olarak dokularda oksidatif hasara yol açabilen hipertiroidi gibi patolojilerde; antioksidan özellikler gösteren karnozinin standart tedavi protokollerine ilave bir terapötik ajan olarak, hastalıktan önce ve/veya sonra kullanılması önerilebilir. Anahtar kelimeler: Rat, hipertiroidizm, oksidatif stres, karnozin, antioksidan It is suggested that hyperthyroidism which is formed as a result of excessive production of thyroid hormones (L-thyroxin, L-triiodotyronin) induces free radical production by accelerating mitochondrial respiration and plays a role in etiopathogenesis of several diseases by causing oxidative damage.It is reported that L-carnosine, a dipeptid, shows antioxidant effects. In literature scanning there isn't a study investigating the antioxidant characteristics of L-carnosine in rats with experimentally induced hyperthyroidism.In this study, it was investigated the effect of hyperthyroidism on oxidant/antioxidant systems, and how this effect would change in the presence of carnosine on experimentally induced hyperthyroidism in rats.For this purpose, eight study groups were formed and each group included twelve rats (Wistar albino). The rats in experimental groups were applied L-thyroxine (intraperitoneal: ip 300 µg/kg body weight/day) and pre-, post- and combined L-carnosine (ip, 300 mg/kg body weight/day), for 10/20 days. Malondialdehyde (MDA), protein carbonyl compounds (PCC), advanced oxidation protein products (AOPP) (only in plasma) and thiol levels were measured in the plasma, liver, kidney and heart obtained from the rats. It was found out that hyperthyroidism caused oxidative stress via lipid and protein oxidation provided by high MDA, PCC, AOPP and low thiol levels; and the levels of MDA, PCC and AOPP decreased and the levels of thiol increased in the presence of carnosine.As a result, in patologies such as hyperthyroidism which causes oxidative damages in tissues; it is suggested that carnosine which has antioxidant characteristics can be used as an additional therapeutic agent to standard treatment protocols before/after illness.Keywords: Rats, hyperthyroidism, oxidative stress, carnosine, antioxidant 172

Amaç: Kronik böbrek yetersizliği (KBY)'nin patogenezinde yer alan oksidatif stres için, hemodiyaliz (HD) ortamı bir model olarak düşünülmektedir. Lipoprotein metabolizması bozuklukları, hiperhomosisteinemi ve kronik enflamasyon sonrası gelişen oksidatif stresin yol açtığı patolojik değişikliklerin, ateroskleroz gelişimini hızlandırarak KBY ve HD hastalarında gözlenen artmış kardiyovasküler hastalık riskine neden olduğu belirtilmektedir.Bu çalışma, HD işleminin, oksidatif stresle ilişkisini göstermek; tedavi protokolüne eklenen N-asetilsistein (NAC)'in ölçülen parametreler üzerine muhtemel etkilerini değerlendirmek amacıyla planlandı.Gereç ve Yöntem: En az altı aydır, düzenli HD tedavisi (2-3 seans/hafta, 4 saat) alan 13 hasta çalışmaya alındı. Hastalardan birinci HD seansı öncesi kan alındı ve seans boyunca 500 mL % 0.9 sodyum klorür çözeltisi intravenöz (iv) olarak verildi (Plasebo grubu). Bu seansı takip eden 2. (NAC-1 grubu) ve 3. (NAC-2 grubu) HD seanslarında, hastalara rutin tedavilerine ek olarak, 500 mL % 0.9 sodyum klorür içinde 50 mg/kg NAC verildi.Diyaliz öncesi ve sonrası olmak üzere, toplam altı kez alınan kanlardan elde edilen serum/plazma örneklerinde; miyeloperoksidaz (MPO), tümör nekrozis faktör ? (TNF-?), neopterin, proteinlerin ileri oksidasyon ürünleri (AOPP), protein karbonil bileşikleri (PCC), tiyol, paraoksonaz (PON), homosistein ve okside LDL (oxLDL) düzeyleri ölçüldü.Bulgular: HD'nin MPO, TNF-?, AOPP, PCC, tiyol ve PON1 düzeylerini artırdığı; neopterin, homosistein düzeyini düşürdüğü; oxLDL'yi etkilemediği; iki ardışık doz NAC ilavesi ile AOPP, PCC ve homosistein düzeylerinin düştüğü; ölçülen diğer parametrelerin ise değişmediği belirlenmiştir.Sonuç: Yüksek MPO ve TNF-? seviyeleriyle gösterildiği gibi, HD'de aktifleşen nötrofillerin oksidatif strese neden olduğu; güçlü bir antioksidan olan NAC'ın, rutin HD tedavi protokolüne eklenerek oksidatif stresle ilişkili bazı patolojik değişiklikleri düzeltebileceği söylenebilir.Anahtar kelimeler: Antioksidan, hemodiyaliz, N-asetilsistein, oksidatif stres. Aim: Hemodialysis (HD) medium is considered as a model for oxidative stress found in pathogenesis of chronic renal failure (CRF). It is stated that pathological changes caused by oxidative stress developing after chronic inflammation, lipoprotein metabolism disorders and hyperhomocysteinaemia expedite the emergence of atherosclerosis and, therefore, induces increased for cardiovascular disease risk observ ed in the patients with CRF and HD.This study was planned to exhibit the relationship between HD process and the oxidative stress; to evaluate the possible effects of N-acetylcysteine (NAC), which is added to treatment protocol, on measured parameters.Materials and methods:Thirteen patients receiving HD treatment for at least six months (2-3 sessions /week, 4 hours) were included in the study. Blood samples from the patients taken prior to the first HD session and 500 mL of 0.9 % sodium chloride solution was administrated intravenously (iv) to the patients during the session (Placebo Group). In the second (NAC-1 Group) and third sessions (NAC-2 Group) following this session,the patients given 50 mg/kg NAC (iv) in 500 mL of 0.9 % sodium chloride solution in addition to their routine treatment.The serum/plasma samples obtained from blood samples taken prior to and after the HD treatment, six times in total, are analyzed for myeloperoxidase (MPO), tumor necrosis factor ? (TNF-?), neopterin, advanced oxidation protein products (AOPP), protein carbonyl content (PCC), thiol, paraoxonase (PON), homocysteine and oxidized LDL (oxLDL) levels.Results: It is identified that HD increased the MPO activity and TNF-?, AOPP, PCC, thiol and PON1 levels; while causing a decrease in the levels of neopterin and homocysteine, and causing no change with oxLDL. AOPP, PCC and homocysteine levels declined and other parameters did not change with a supplementary double dose of NAC.Conclusion: It may be stated, as exhibited with high levels of MPO and TNF-?, that the neutrophils activated in HD process causes oxidative stress and that some of pathological changes related to oxidative stress may be prevented by adding NAC, which is a very strong antioxidant, to the routine HD treatment protocol.Key Words: Antioxidant, hemodialysis, N-acetylcysteine, oxidative stress. 113

IV METİLGLİOKSAL VERİLEN RATLARDA KARNOZİNİN ANTİOKSİDAN VE ANTİGLİKASYON ÖZELLİKLERİNİN ARAŞTIRILMASI ÖZET Oluşum mekanizmaları içinde oksidatif/nonoksidatif ve enzimatik/nonenzimatik reaksiyonlarm yer aldığı ileri glikasyon son ürünleri (advanced glycation end product; AGE), başta diyabetes mellitus (DM) komplikasyonlan olmak üzere ateroskleroz, yaşlanma gibi pek çok hastalığın etiyopatogenezinde rol oynamaktadır. Sahip olduğu karbonil grubu, yüksek reaktivitesi ve toksik etkileri nedeniyle metilglioksal (MGO)'in, önemli bir AGE prekürsörü olduğu düşünülmektedir. Diğer taraftan karnozinin antioksidan ve antiglikasyon özelliklere sahip olduğu ve bu etkileriyle, yukarıda belirtilen hastalıkların önlenmesinde faydalı olabileceği ileri sürülmektedir. Bu çalışma, rat modeli üzerinde, MGO ve karnozinin ayrı ayrı ve kombine halde uygulandıklarında, oksidan sistem üzerindeki etkilerini araştırmak amacıyla yapıldı. Ratlar her biri 10 rat içeren dört gruba ayrılarak çalışmaya alındı. Birinci gruba serum fizyolojik, ikinci gruba 200 mg/kg rat ağırlığı/gün dozunda karnozin, üçüncü gruba 50 mg/kg/gün MGO, dördüncü gruba da aynı dozlarda kombine olarak karnozin ve MGO, intraperitoneal olarak on gün süre ile uygulandı. Ratlardan elde edilen plazma örneklerinde lipid peroksidasyonu göstergesi olan malondialdehit (MDA) ve protein oksidasyonu göstergesi olan protein karbonil bileşikleri (PCC) tayin edildi. İstatistiksel karşılaştırmalarda ANOVA ve post-ANOVA (Scheffe prosedür) ve eşleştirilmiş / test kullanıldı. Çalışma gruplarının parametreleri arasında regresyon analizleri yapıldı. MGO verilen ratlarda plazma MDA ve PCC seviyelerinin kontrol grubuna göre anlamlı şekilde yükseldiği, bu değerlerin karnozin ve MGO'nun kombine olarak uygulandığı grupta. MGO verilen gruba göre anlamlı şekilde azaldığı belirlendi. Sonuç olarak, yüksek MDA ve PCC düzeyleriyle yansıtıldığı gibi, MGO'nun oluşturduğu lipid peroksidasyonu ve protein oksidasyonunun, karnozin ile önlenebildiği gösterildiğinden; etiyopatogenezinde AGE oluşumunun rol aldığı hastalıklarda karnozinin terapötik bir ajan olarak kullanımı önerilebilir. THE INVESTIGATION OF ANTIOXIDANT AND ANTIGLYCATION PROPERTIES OF CARNOSINE IN THE RAT ADMINISTRATED METHLGLYOXAL ABSTRACT The formation mechanisms in which the oxidative/nonoxidative and enzymatic/nonenzymatic reactions take place, and advanced glycation end product (AGE) play a role mainly in diabetes mellitus (DM) complications and in the etiopathogenesis of many disorders such as atherosclerosis and aging. It is believe that methylglyoxal (MGO) is a significant AGE precursor because of its carbonyl group, high reactivity and toxic effects. On the other hand carnosine is useful for preventing the disorders mentioned above due to its antioxidant and antiglycation properties. This study was done in order to investigate the effects of MGO and carnosine on oxidant system when they are applied to rats separately and jointly. The rats were studied in four groups each consisting of 10 rats. The first group was intraperitoneally applied serum physiologic, the second 200 mg carnisone/kg rat body weight per day, the third 50 mg/kg of MGO per day and the fourth group the same doses of carnosine and MGO together for ten days. We measured malondialdehyde (MDA) indicating lipid peroxidation and protein carbonyl compounds (PCC) indicating protein oxidation in a plasma specimen obtained from the rats. We used ANOVA and post-ANOVA (Scheffe procedure) and paired t test in statistical comparisons. Regression analyses were done between the parameters of the groups. We found out that the levels of plasma MDA and PCC in the rats given MGO increased significantly according to control group, and that these values in the group given carnosine together with MGO decreased significantly according to the group given MGO. In conclusion, it may be proposed that carnosine should be used as a therapeutic agent in the disorders whose etiopathogenesis AGE formation plays a role in since we showed that lipid peroxidation and protein oxidation formed by MGO, reflected in the high levels of MDA and PCC, can be prevented by using carnosine. 45. Serum vitamin D concentration is inversely associated with matrix metalloproteinase-9 level in periodontal diseases.

- Abstract
Background: The aim of this study was to investigate the potential association between vitamin D deficiency and matrix metalloproteinase-9 (MMP-9) levels in gingival crevicular fluid (GCF) across various periodontal health and disease statuses., Methods: A total of 200 volunteers were divided into two groups according to serum vitamin D concentration (25(OH)D < 10 ng/mL and 25(OH)D ≥ 10 ng/mL). Periodontal health status was determined based on a full-mouth periodontal examination and radiographic evaluation. Participants in both groups were categorized according to periodontal diagnoses, encompassing periodontal health, gingivitis, and periodontitis. Following sampling, the MMP-9 levels in GCF were determined by the enzyme-linked immunosorbent assay (ELISA) method., Results: The GCF MMP-9 levels were found to be higher in individuals with serum 25(OH)D < 10 ng/mL, in both the healthy and gingivitis and periodontitis groups, compared to those with 25(OH)D ≥ 10 ng/mL. Nevertheless, a statistically significant distinction was observed exclusively within the gingivitis and periodontitis groups. Correlation analysis and robust regression analyses provided additional evidence supporting the predictive role of periodontal disease status and vitamin D concentration in local MMP-9 levels. These associations remained significant after adjusting for age and sex in robust regression analysis (p = 0.002). Furthermore, the inclusion of periodontal clinical parameters in the regression analysis revealed notable associations of clinical attachment loss with local MMP-9 levels, along with periodontal disease status and serum vitamin D concentration (p < 0.001)., Conclusion: The findings of our study suggest a potential mechanistic relationship between serum vitamin D levels and periodontitis., Plain Language Summary: Vitamin D deficiency is a widespread issue globally due to urban living, less outdoor time, seasonal changes, aging, and sunscreen use, leading to inadequate sun exposure. Low vitamin D levels are linked to several health problems, including hypertension, diabetes, heart diseases, and periodontal diseases, which affect the gums and bones around teeth and can cause tooth loss if untreated. Although the link between vitamin D and periodontal disease is unclear, it may involve the enzyme matrix metalloproteinase-9 (MMP-9). Our study examined 200 people, dividing them into two groups based on vitamin D levels. We assessed their gum health and measured MMP-9 levels in their gingival crevicular fluid, a liquid that seeps out from the tiny space between gums and teeth. We found that people with lower vitamin D levels had higher MMP-9 levels, especially those with gum disease. Our analysis showed that both vitamin D levels and gum health significantly impact MMP-9 levels, with gum health being the more influential factor. Maintaining good gum health and adequate vitamin D levels is crucial for managing MMP-9, an enzyme critical for tissue remodeling during healing and inflammation. 46. Does ß-Glucan can have protective role against oxidative stress in experimental sepsis model in rats?

- Abstract
Background: Very high mortality rate in sepsis may be related to oxidative stress. This study was conducted on the rats to investigate the presence of oxidative stress and also the potential protective effects of the ß-glucan in the intra-abdominal sepsis model formed by cecal ligation-perforation (CLP)., Methods: In this study, 30 Male rats were equally divided into three groups as 'Sham', 'Sepsis' and 'ß-Glucan'. Only laparotomy was performed in the Sham group, and sepsis was induced by CLP in Sepsis and ß-Glucan groups. Following CLP, a single dose of 4 mg ß-glucan/kg was also intraperitoneally administered to the β-Glucan group. Blood and tissue (liver, lung and kidney) samples were taken from Sepsis and ß-Glucan groups after sepsis development determined at the end of the 48th hour, also from the Sham group. The levels of myeloperoxidase (MPO) and advanced oxidation protein products (AOPP) were determined in plasma samples, and the malondialdehyde (MDA) was measured in plasma and tissues., Results: MPO and AOPP were higher in both the Sepsis and ß-Glucan groups; however, plasma and tissue MDA levels were higher only in the Sepsis group than the Sham group (p<0.05). However, when compared to the Sepsis group, all parameters measured, except kidney MDA, were significantly lower in the ß-Glucan group (p<0.05)., Conclusion: To our knowledge, this is the first study to investigate the AOPP levels in the CLP sepsis model, ROS produced by the reaction of MPO derived from neutrophils may form oxidative damage to the proteins, compared to the lipids, and ß-glucan may be used as an alternative agent for sepsis treatment.

- Abstract
Background/aim: It is not always easy to diagnose pulmonary neuroendocrine tumors (PNETs). The aim of the present study is to make a differential diagnosis by studying the same markers in patients with non-small-cell lung carcinoma (NSCLC), patients with benign lung disease (chronic obstructive pulmonary disease and pneumonia), and healthy volunteers to determine the roles of these markers in pulmonary neuroendocrine tumor diagnosis and to identify their power., Materials and Methods: A total of 100 participants including 23 PNET patients and 28 NSCLC patients who were pathologically di-agnosed but not yet treated, 25 participants with benign disease, and 24 healthy volunteers were included in this cross-sectional study., Results: No significant difference was found between the chromogranin A (CgA) and squamous cell carcinoma antigen 1 (SCCA1) values among the groups (PNET, NSCLC, benign, healthy volunteers), but the difference in progesterone-releasing peptide (ProGRP), neuron-specific enolase (NSE), and adjusted NSE was statistically significant (P values were respectively ProGRP, P = 0.006; NSE, P = 0.015; NSE adjusted, P = 0.09). In a comparison of the PNET and NSCLC groups, having a ProGRP value higher than 84.6 pg/mL re-vealed PNET with 60.9% sensitivity and 89.3% specificity (P = 0.001)., Conclusion: The ProGRP value is the only indicator that distinguishes the PNET group from the other 3 groups., (This work is licensed under a Creative Commons Attribution 4.0 International License)

- Abstract
Purpose: This study examined the feasibility of using the serum intestinal fatty acid binding protein (I-FABP) level for the early diagnosis of acute mesenteric ischemia, and investigated whether it contributes to the clinical decision-making process., Method: Thirty patients diagnosed with acute mesenteric ischemia, 27 patients with other types of acute abdomen who presented with acute abdomen symptoms but were not diagnosed with acute mesenteric ischemia, and 20 healthy people were included in the study. Mesenteric ischemia was confirmed by a pathological evaluation in patients who underwent intestinal resection due to detection of mesenteric ischemia during surgery., Results: There was no significant difference in the leukocyte counts and D-dimer levels between subjects with mesenteric ischemia and acute abdomen due to other causes (p > 0.05). There was a significant difference in the serum I-FABP level between these groups (p < 0.001)., Conclusion: The I-FABP level is a more reliable parameter for diagnosing acute mesenteric ischemia compared to leukocytosis and D-dimer elevation.
