Abstract

Introduction: Mesenchymal stromal/stem cells (MSCs) are multipotent, self-renewing cells that are extensively used in tissue engineering. Dedifferentiated fat (DFAT) cells are derived from adipose tissues and are similar to MSCs. Three-dimensional (3D) spheroid cultures comprising MSCs mimic the biological microenvironment more accurately than two-dimensional cultures; however, it remains unclear whether DFAT cells in 3D spheroids possess high osteogenerative ability. Furthermore, it is unclear whether DFAT cells from 3D spheroids transplanted into calvarial bone defects are as effective as those from two-dimensional (2D) monolayers in promoting bone regeneration. Methods: We compared the in vitro osteogenic potential of rat DFAT cells cultured under osteogenic conditions in 3D spheroids with that in 2D monolayers. Furthermore, to elucidate the ability of 3D spheroid DFAT cells to promote bone healing, we examined the in vivo osteogenic potential of transplanting DFAT cells from 3D spheroids or 2D monolayers into a rat calvarial defect model. Results: Osteoblast differentiation stimulated by bone morphogenetic protein-2 (BMP-2) or osteogenesis-inducing medium upregulated osteogenesis-related molecules in 3D spheroid DFAT cells compared with 2D monolayer DFAT cells. BMP-2 activated phosphorylation in the canonical Smad 1/5 pathways in 3D spheroid DFAT cells but phosphorylated ERK1/2 and Smad2 in 2D monolayer DFAT cells. Regardless of osteogenic stimulation, the transplantation of 3D DFAT spheroid cells into rat calvarial defects promoted new bone formation at a greater extent than that of 2D DFAT cells. Conclusions: Compared with 2D DFAT cells, 3D DFAT spheroid cells promote osteoblast differentiation and new bone formation via canonical Smad 1/5 signaling pathways. These results indicate that transplantation of DFAT cells from 3D spheroids, but not 2D monolayers, accelerates bone healing.

Abstract

Objective: To investigate the criteria, based on fetal TR waveforms in late gestation, to predict biventricular circulation (BV) after birth in cases of tricuspid valve dysplasia (TVD) or Ebstein's anomaly diagnosed during the fetal period. Methods: We included 35 consecutive cases diagnosed with TVD or Ebstein's anomaly during the fetal period between January 2008 and December 2021 at Kanagawa Children's Medical Center, Kanagawa, Japan. The maximum velocity and change in pressure over time of tricuspid regurgitation (TR) jet (dP/dt), estimated using TR waveforms obtained during the late‐gestation period (gestational age ≥ 28 weeks), were collected from patient records. dP/dt was calculated by dividing the change in estimated right ventricular pressure obtained using Bernoulli's principle by the time taken for the TR maximum velocity to change from one‐third to two‐thirds of its peak value. The outcome was divided into four categories: BV, single ventricular circulation, neonatal death and fetal death. Patients with BV were included in the BV group, while patients with single ventricular circulation, neonatal death or fetal death were included in the non‐BV (NBV) group. Results: Overall, 19 and 16 patients were included in the BV and NBV groups, respectively. The median TR maximum velocity was 3.3 (range, 2.4–3.6) m/s in the BV group and 1.9 (range, 1.0–3.3) m/s in the NBV group. There were no cases of postnatal BV in fetuses with TR maximum velocity < 2.4 m/s; cases with TR maximum velocity of 2.4–3.3 m/s were observed in both BV and NBV groups. Receiver‐operating‐characteristics‐curve analysis was performed on the 11 patients in the BV group and five patients in the NBV group with a TR maximum velocity of 2.4–3.3 m/s. dP/dt ≥ 350 mmHg/s and TR maximum velocity ≥ 2.9 m/s were identified as criteria for predicting the outcome in such cases. The performance of dP/dt ≥ 350 mmHg/s in predicting BV after birth in fetuses with TVD or Ebstein's anomaly was higher compared to that of TR maximum velocity ≥ 2.9 m/s (sensitivity, 90.9% vs 72.3% and specificity, 80.0% vs 80.0%, respectively). Conclusions: In fetuses with TVD or Ebstein's anomaly, the postnatal outcome may be BV or NBV when the TR maximum velocity is 2.4–3.3 m/s. In such cases, by combining the TR maximum velocity with dP/dt ≥ 350 mmHg/s, BV after birth may be predicted with greater accuracy. © 2022 International Society of Ultrasound in Obstetrics and Gynecology. Abstract

Construction of the JT-60SA tokamak was completed on schedule in March 2020. Manufacture and assembly of all the main tokamak components satisfied technical requirements, including dimensional accuracy and functional performances. Development of the plasma heating systems and diagnostics have also progressed, including the demonstration of the favourable electron cyclotron range of frequency (ECRF) transmission at multiple frequencies and the achievement of long sustainment of a high-energy intense negative ion beam. Development of all the tokamak operation control systems has been completed, together with an improved plasma equilibrium control scheme suitable for superconducting tokamaks including ITER. For preparation of the tokamak operation, plasma discharge scenarios have been established using this advanced equilibrium controller. Individual commissioning of the cryogenic system and the power supply system confirmed that these systems satisfy design requirements including operational schemes contributing directly to ITER, such as active control of heat load fluctuation of the cryoplant, which is essential for dynamic operation in superconducting tokamaks. The integrated commissioning (IC) is started by vacuum pumping of the vacuum vessel and cryostat, and then moved to cool-down of the tokamak and coil excitation tests. Transition to the super-conducting state was confirmed for all the TF, EF and CS coils. The TF coil current successfully reached 25.7 kA, which is the nominal operating current of the TF coil. For this nominal toroidal field of 2.25 T, ECRF was applied and an ECRF plasma was created. The IC was, however, suspended by an incident of over current of one of the superconducting equilibrium field coil and He leakage caused by insufficient voltage holding capability at a terminal joint of the coil. The unique importance of JT-60SA for H-mode and high-β steady-state plasma research has been confirmed using advanced integrated modellings. These experiences of assembly, IC and plasma operation of JT-60SA contribute to ITER risk mitigation and efficient implementation of ITER operation.

Abstract

Broiler chickens are homoeothermic animals, i.e., animals capable of maintaining their body temperature within quite narrow limits; therefore, climate change poses a great challenge to poultry. With this in mind, this research aims to evaluate the performance of broilers submitted to different commercial production systems and exposed to different future scenarios, taking into account the climate change trends. To achieve this objective, we developed and validated a fuzzy model able to predict the performance of a broiler as a function of enthalpy along its life stages. This model was developed and validated in part I of this article based on experimental data collected for one year in three aviaries: conventional, negative pressure, and dark house systems. A Mann?Kendall nonparametric test and linear regression analysis were applied to the enthalpy values, which were calculated as a function of the ambient air temperature and relative humidity in order to study the climate change trends. Later, simulations were performed using the fuzzy model for 2025, 2050, 2075, and 2100 future scenarios. Specific improvements were observed when the heating trends coincided with the initial stages of breeding; however, in general, the productive responses of broilers in the different evaluated systems worsened with the climate change trends. Abstract

The selection of the type of fuzzy systems pertinence curve allows a better representation of the mathematical model and a smaller simulation error. We aimed to study the effect of pertinence curves in fuzzy modeling of broiler performance, created in different production systems. For the development and testing of fuzzy models, three commercial aviaries (conventional, tunnel with negative pressure, and dark house) were evaluated over one year, totaling six lots per system. For the development of the model, the input variables were enthalpy in each rearing phase (initial: phases 1, 2, and 3; growth: phase 4; and final: phase 5) and the output variables were feed intake (FE), weight gain (GP), feed conversion (FE), and the productive efficiency index (PEI). Triangular, trapezoidal, and Gaussian pertinence curves were combined and applied to represent the input and output fuzzy sets, totaling nine fuzzy models for each output variable. The combinations of pertinence curves provided adequate responses for the prediction of AL, GP, RC, and PEI. However, the selection of the types of curves should be studied on a case-by-case basis, so that the smallest possible simulation errors are obtained. Abstract

The preparation of p-(In,Mn)As/n-InAs/p-(In,Mn)As trilayer structures by molecular beam epitaxy on GaSb/GaAs/GaAs (001) substrates is examined. Ferromagnetic coupling of the magnitude J = 7.6 * 10(super -3) mJ/m(super 2) is observed between the two magnetic p-(In,Mn)As layers with a thick n-InAs layer.

Abstract

ABSTRACT Brazilian electric power is mostly based on hydraulics sources through hydropower plant reservoirs that are fed by rivers located in Southeast Brazil. Possible changes in climate can affect the energy supply of the country. The objective of this work is to assess the possible changes in the hydrology of the Upper Grande River Basin ( UGRB) under a future climate change scenario, using the Lavras Simulation of Hydrology ( LASH) model forced by the outputs of the Eta model, a regional climate model, which was driven by HadCM3 A1B scenario for three time slices in the period between 2011 and 2099. Owing to the surface heterogeneity of the region, the hydrologic model was previously calibrated for four watersheds of the UGRB, which are located in its headwater region and evaluated against observational time series of the present climate period, 1961-1990. In the first future time slice, 2011-2040, the results showed a small reduction in the annual run-off, but for the other time slices, the trend changed to strong increase in most of the watersheds. The water budget in the region calculated from the Eta model agreed with the run-off trends, as water excess reduced in the first time slice and gradually increased towards the end of the century. The run-off components showed intraseasonal variability. The reduction of rains in the end of winter, the dry period in the region, and in the beginning of spring, may cause a change in the dynamics of the groundwater recharge, affecting the base flow, which can extend and intensify the flow recession period, and therefore affect the availability of water resources. On the other hand, the increase of rains during the summer, the rainy season of the region, caused an increase of the direct surface run-off, which can modify the flood regimes of the rivers in the region.
