The Dong Pipa, a traditional Chinese musical instrument, is a significant element of the Dong ethnic group's cultural heritage. The objective is to investigate the historical development of the Dong Pipa, a traditional Chinese musical instrument, in literacy functions and educational occasions. The research was conducted in Liping, Congjiang, and Rongjiang counties in Guizhou Province, Tongdao County in Hunan Province, and Sanjiang in the Guangxi Zhuang Autonomous Region. Two key informants and four casual informants, including national transmitters and experts in Dong Pipa music, were interviewed, and data were gathered through fieldwork, interviews, and observations. The analysis focused on the integration of the Dong Pipa into educational settings and its role in fostering cultural literacy. The results indicate that the Dong Pipa has played a dynamic role both historically and in contemporary times, contributing significantly to cultural transmission and education. The study suggests that incorporating traditional instruments like the Dong Pipa into modern educational practices can enhance cultural literacy and support preservation efforts.

The provenance of the Upper Carboniferous Benxi Formation in North China Craton (NCC) has been considered as the northern margin of the NCC, not the North Qinling Orogenic Belt. To understand the provenance and the tectonic-sedimentary evolution during the sedimentary period of the Benxi Formation, the zircon U–Pb geochronology analysis was conducted on eleven clastic sandstone samples. The south of the NCC received clastic sediments from the North Qinling Orogenic Belt. The orogenic movements around the NCC in the Late Carboniferous period had significant impacts on the changes in paleotopography. During the early sedimentary period of the Hutian member of the Benxi Formation, the north of the Qinling Orogenic Belt was rapidly uplifted, and paleotopography was south-uplifting and north-dipping; thus, the clastic source was the North Qinling Orogenic Belt. From the late sedimentary period of the Benxi Formation Hutian member to the sedimentary period of the Jinci member, paleotopography was reversed. The northern margin of the NCC quickly uplifted, and paleotopography was north-uplifting and south-dipping. Two distinct provenances were present in the sediments of the Benxi Formation. The sediments were supplied predominately by the provenance in the north.

Abstract The multi-lead electrocardiogram (ECG) is widely utilized in clinical diagnosis and monitoring of cardiac conditions. The advancement of deep learning has led to the emergence of automated multi-lead ECG diagnostic networks, which have become essential in the fields of biomedical engineering and clinical cardiac disease diagnosis. Intelligent ECG diagnosis techniques encompass Recurrent Neural Networks (RNN), Transformers, and Convolutional Neural Networks (CNN). While CNN is capable of extracting local spatial information from images, it lacks the ability to learn global spatial features and temporal memory features. Conversely, RNN relies on time and can retain significant sequential features. However, they are not proficient in extracting lengthy dependencies of sequence data in practical scenarios. The self-attention mechanism in the Transformer model has the capability of global feature extraction, but it does not adequately prioritize local features and cannot extract spatial and channel features. This paper proposes STFAC-ECGNet, a model that incorporates CAMV-RNN block, CBMV-CNN block, and TSEF block to enhance the performance of the model by integrating the strengths of CNN, RNN, and Transformer. The CAMV-RNN block incorporates a coordinated adaptive simplified self-attention module that adaptively carries out global sequence feature retention and enhances spatial–temporal information. The CBMV-CNN block integrates spatial and channel attentional mechanism modules in a skip connection, enabling the fusion of spatial and channel information. The TSEF block implements enhanced multi-scale fusion of image spatial and sequence temporal features. In this study, comprehensive experiments were conducted using the PTB-XL large publicly available ECG dataset and the China Physiological Signal Challenge 2018 (CPSC2018) database. The results indicate that STFAC-ECGNet surpasses other cutting-edge techniques in multiple tasks, showcasing robustness and generalization.

Abstract Lymphocyte receptors independently evolved in both jawed and jawless vertebrates with similar adaptive immune responses. However, the diversity of functional subtypes and molecular architecture in jawless vertebrate lymphocytes, comparable to jawed species, is not well defined. Here, we profile the gills, intestines, and blood of the lamprey, Lampetra morii, with single-cell RNA sequencing, using a full-length transcriptome as a reference. Our findings reveal higher tissue-specific heterogeneity among T-like cells in contrast to B-like cells. Notably, we identify a unique T-like cell subtype expressing a homolog of the nonlymphoid hematopoietic growth factor receptor, MPL-like (MPL-L). These MPL-L+ T-like cells exhibit features distinct from T cells of jawed vertebrates, particularly in their elevated expression of hematopoietic genes. We further discovered that MPL-L+ VLRA+ T-like cells are widely present in the typhlosole, gill, liver, kidney, and skin of lamprey and they proliferate in response to both a T cell mitogen and recombinant human thrombopoietin. These findings provide new insights into the adaptive immune response in jawless vertebrates, shedding new light on the evolution of adaptive immunity.

Abstract P2X receptors, a subfamily of ligand-gated ion channels activated by extracellular ATP, are implicated in various physiopathological processes, including inflammation, pain perception, and immune and respiratory regulations. Structural determinations using crystallography and cryo-EM have revealed that the extracellular three-dimensional architectures of different P2X subtypes across various species are remarkably identical, greatly advancing our understanding of P2X activation mechanisms. However, structural studies yield paradoxical architectures of the intracellular domain (ICD) of different subtypes (e.g., P2X3 and P2X7) at the apo state, and the role of the ICD in P2X functional regulation remains unclear. Here, we propose that the P2X3 receptor’s ICD has an apo state conformation similar to the open state but with a less tense architecture, containing allosteric sites that influence P2X3’s physiological and pathological roles. Using covalent occupancy, engineered disulfide bonds and voltage-clamp fluorometry, we suggested that the ICD can undergo coordinated motions with the transmembrane domain of P2X3, thereby facilitating channel activation. Additionally, we identified a novel P2X3 enhancer, PSFL77, and uncovered its potential allosteric site located in the 1α3β domain of the ICD. PSFL77 modulated pain perception in P2rx3 +/+, but not in P2rx3 −/−, mice, indicating that the 1α3β, a “tunable” region implicated in the regulation of P2X3 functions. Thus, when P2X3 is in its apo state, its ICD architecture is fairly ordered rather than an unstructured outward folding, enabling allosteric modulation of the signaling of P2X3 receptors.

Understanding the hydrochemical characteristics and genesis mechanisms of strontium-rich groundwater is pivotal for supporting the exploitation and utilization of natural strontium-rich groundwater. In this research, 27 groundwater samples were collected. By analyzing major ion chemistry and strontium isotope data, and considering the hydrogeological context, various analytical approaches, including multivariate statistics, ion ratios, and isotopes, were used to reveal the characteristics and genesis mechanisms of strontium-rich groundwater in the study area. The findings indicate that the predominant hydrochemical type of groundwater is HCO3-Ca, with Ca2+ and HCO3− as the primary cations and anions. The hydrochemistry of the strontium-rich groundwater is predominantly influenced by rock weathering processes. A combination of factors, including ion exchange, and anthropogenic activities, shapes the compositional characteristics of the groundwater in the region. The dissolution of calcite due to weathering emerges as the principal source of strontium in the groundwater. While ion exchange processes are not conducive to strontium enrichment in groundwater, their effect is relatively limited. The impact of human activities on the groundwater is minor.

The metastability of the dendritic phase is crucial in determining the mechanical properties and mechanisms of the Ti-based in-situ dendrite-reinforced bulk metallic glass composites (BMGCs). However, tailoring the metastability effectively remains challenging. In this study, we propose a straightforward and effective strategy to tailor the dendrites composition and metastability using conventional Titanium alloys. Initially, six common Titanium alloys with increasing content of β-stabilizing elements such as Ti, Ti–6Al–4V, Ti–13Nb–13Zr, Ti–15Mo, Ti–12Mo–6Zr–2Fe and Ti-4.5Fe-6.8Mo-1.5Al were selected. Subsequently, a series of in-situ dendrite-reinforced BMGCs denoted as T1-T6 were designed by alloying these Titanium alloys with a specific amorphous alloy. It was found that the microstructure, stability and mechanical behaviors of these composites change significantly as the content of β-stabilizing elements in the dendritic phase increases monotonically. Based on their microstructure and mechanical behaviors, these composites can be classified into three categories: T1, T2 and T3, T4-T6. The T1 alloy exhibits brittle deformation characteristics due to the significantly higher elastic modulus of the dendritic phase. In contrast, the T2 and T3 alloys demonstrate good tensile plasticity and remarkable work-hardening ability, attributed to the metastable β phase undergoing stress-induced phase transformation during deformation. On the other hand, the T4-T6 alloys, consisting of stable β phase, deform via dislocation slip, resulting in a higher yielding stress and compression plasticity. The composition design strategy presented in this study, along with the correlation between mechanical behaviors and dendrite stability, may offer a novel perspective for the development of Ti-based in-situ dendrite-reinforced BMGCs.

Abstract Aims/Introduction To evaluate the differences in cardiac autonomic function, cardiac structure and diastolic function between individuals with diabetic foot (DF) and those with diabetes but without DF. Materials and Methods A total of 413 individuals with DF and 437 without DF who underwent a 24‐h electrocardiogram Holter and a Doppler echocardiogram were included. The heart rate variability parameters to evaluate cardiac autonomic function, and the indices for the assessment of cardiac structure and left ventricular (LV) diastolic function, including left atrium, LV posterior wall thickness, interventricular septum and E/e′ ratio, were measured or calculated. Propensity score matching was used for the sensitivity analysis to minimize potential imbalance. Objective This study investigated the efficacy of different concentrations of cholesterol-loaded cyclodextrin (CLC) on cryopreservation in boar sperm quality. Methods In this study, we treated boar sperm with different concentrations of CLC before freezing and analyzed the sperm cholesterol concentration, plasma membrane, acrosome integrity rate and total motility rate before and after freeze-thawing. Antarctic trace metal records provide important information for grasping the influence of human activities on the environment over the last centuries. The CA2016-75 ice core is located along the East Antarctic Zhongshan Station–Dome A, enhances the record of metals research in the East Antarctic region, and its high-resolution supplies data support for the study of high-frequency climatic drivers and the effect of human activities on the Antarctic environment. A thorough dataset on seven trace metals (Al, Fe, Mn, Cu, Zn, Ba and Pb) in a coastal ice core in eastern Antarctica during the previous 102 years (1915–2016) is presented in this study. Pb has the lowest concentration (9.51 ± 20.95 pg g−1), and Ba has the highest concentration (36.57 ± 51.35 ng g−1). Notable variations are observed between the pre-1968 AD and post-1968 AD phases for Mn, Zn and Ba. The abrupt, remarkable increase in the concentrations coincided with the change of metal smelting production in the southern hemisphere. In addition to this, it may also be related to local Antarctic scientific research activities. Al and Fe, the primary crustal elements, are essentially obtained from soil dust; Cu shows high crustal enrichment factors (EFc, >10), indicating that it is notably affected by anthropogenic activities. Moreover, the anthropogenic activities in the Southern Hemisphere have had an impact on lead deposition in Antarctica. This study not only enriches the trace metal historical record along the Zhongshan Station–Dome A but also provides a high-resolution ice core record, which is very crucial for the reconstruction of trace metal concentration changes in the last 100 years.

Abstract Background In this study, we aimed to evaluate the clinical utility of Metagenomic Next-Generation sequencing (mNGS) on bronchoalveolar lavage fluid (BALF) in diagnosis of Lower Respiratory Tract Infections (LRTIs). Methods In this study, we retrospectively analyzed 186 hospitalized patients who were suspected with LRTIs and performed mNGS (DNA) test of BALF simultaneously at The Fifth Affiliated Hospital of Sun Yat-Sen University from March 2023 to August 2023. Suspected LRTI was based on LRTI related clinical manifestations or imaging examination. Among them, 155 patients had undergone conventional culture and mNGS (DNA) testing simultaneously. Finally, 138 cases (89.03%,138/155) were diagnosed as LRTI and 17 cases (10.97%,17/155) were diagnosed as non-LRTI. Both detecting rate and diagnostic efficacy of mNGS and conventional culture were compared. Results The positive detection rates of pathogens between mNGS and conventional culture were significant different (81.29% VS 39.35%, P 0.05). Furthermore, 77.54% and 35.51% of LRTI cases were being etiologically diagnosed by mNGS and culture respectively. Importantly, mNGS directly led to a change of treatment regimen in 58 (37.42%) cases, including antibiotic adjustment (29.68%) and ruling out active infection (7.74%). Moreover, treatment regimen remained unchanged in 97 (62.58%) cases, considering the current antibiotic therapy already covered the detected pathogens (36.13%) or empirical treatment was effective (11.61%). Conclusions mNGS can identify a wide range of pathogens in LRTIs, with improved sensitivity and being more superior at diagnosing LRTIs etiologically. mNGS has the potential to enhance clinical outcomes by optimizing the treatment regimens.

Abstract Background Plerocercoid larvae of the tapeworm Spirometra mansoni can infect both humans and animals, leading to severe parasitic zoonosis worldwide. Despite ongoing research efforts, our understanding of the developmental process of S. mansoni remains inadequate. To better characterize posttranslational regulation associated with parasite growth, development, and reproduction, a comparative phosphoproteomic study was conducted on the plerocercoid and adult stages of S. mansoni. Methods In this study, site-specific phosphoproteomic analysis was conducted via 4D label-free quantitative analysis technology to obtain primary information about the overall phosphorylation status of plerocercoids and adults. Results A total of 778 differentially abundant proteins (DAPs) were detected between adults and plerocercoids, of which 704 DAPs were upregulated and only 74 were downregulated. DAPs involved in metabolic activity were upregulated in plerocercoid larvae compared with adults, whereas DAPs associated with binding were upregulated in adults. Gene Ontology (GO) and Kyoto Encyclopedia of Genes (KEGG) analyses indicated that most DAPs involved in signal transduction and environmental information processing pathways were highly active in adults. DAPs upregulated in the plerocercoid group were enriched mainly in metabolic activities. The kinases PKACA, GSK3B, and smMLCK closely interact, suggesting potential active roles in the growth and development of S. mansoni. Conclusions The dataset presented in this study offers a valuable resource for forthcoming research on signaling pathways as well as new insights into functional studies on the molecular mechanisms of S. mansoni. Abstract Nucleocytoplasmic large DNA viruses (NCLDVs; also called giant viruses), constituting the phylum Nucleocytoviricota, can infect a wide range of eukaryotes and exchange genetic material with not only their hosts but also prokaryotes and phages. A few NCLDVs were reported to encode genes conferring resistance to beta‑lactam, trimethoprim, or pyrimethamine, suggesting that they are potential vehicles for the transmission of antibiotic resistance genes (ARGs) in the biome. However, the incidence of ARGs across the phylum Nucleocytoviricota, their evolutionary characteristics, their dissemination potential, and their association with virulence factors remain unexplored. Here, we systematically investigated ARGs of 1416 NCLDV genomes including those of almost all currently available cultured isolates and high-quality metagenome-assembled genomes from diverse habitats across the globe. We reveal that 39.5% of them carry ARGs, which is approximately 37 times higher than that for phage genomes. A total of 12 ARG types are encoded by NCLDVs. Phylogenies of the three most abundant NCLDV-encoded ARGs hint that NCLDVs acquire ARGs from not only eukaryotes but also prokaryotes and phages. Two NCLDV-encoded trimethoprim resistance genes are demonstrated to confer trimethoprim resistance in Escherichia coli. The presence of ARGs in NCLDV genomes is significantly correlated with mobile genetic elements and virulence factors.

Abstract The diagnosis of neurodegenerative diseases (NDDs) remains challenging, and existing therapeutic approaches demonstrate little efficacy. NDD drug delivery can be achieved through the utilization of nanostructures, hence enabling multimodal NDD theranostics. Nevertheless, both biomembrane and non-biomembrane nanostructures possess intrinsic shortcomings that must be addressed by hybridization to create novel nanostructures with versatile applications in NDD theranostics. Hybrid nanostructures display improved biocompatibility, inherent targeting capabilities, intelligent responsiveness, and controlled drug release. This paper provides a concise overview of the latest developments in hybrid nanostructures for NDD theranostics and emphasizes various engineering methodologies for the integration of diverse nanostructures, including liposomes, exosomes, cell membranes, and non-biomembrane nanostructures such as polymers, metals, and hydrogels. The use of a combination technique can significantly augment the precision, intelligence, and efficacy of hybrid nanostructures, therefore functioning as a more robust theranostic approach for NDDs. This paper also addresses the issues that arise in the therapeutic translation of hybrid nanostructures and explores potential future prospects in this field.

Abstract Federated learning (FL) enables users to train the global model cooperatively without exposing their private data across the engaged parties, which is widely used in privacy-sensitive business. However, during the life cycle of FL models, both adversaries’ attacks and ownership generalization threaten the FL models’ copyright and affect the models’ reliability. To address these problems, existing model watermarking techniques can be used to verify FL model’s ownership. However, due to the lack of credible binding from “model extracted watermarks” to “ownership verification”, it is difficult to form a closed-loop watermarking framework for copyright protection. Therefore, starting from the shortcomings of the current watermark verification scheme, this article proposed WFB, a blockchain-empowered watermarking framework for ownership verification of federated models. Firstly, we propose a improved watermark generation algorithm to solve the credibility issue of watermarks. Secondly, we propose a watermark embedding method in federated learning, while blockchain technology is used to ensure the credible storage of watermark information throughout the process. Thirdly, the credibility of ownership verification is improved because of the watermark authenticity. Experimental results demonstrate the fidelity, effectiveness and robustness of WFB, with other superiorities such as improving process security and traceability.

Abstract Time-modulated array antenna (TMAA) is a new type of array antenna based on time modulation technology. By introducing "time" as the fourth dimensional design freedom into the design of conventional array antennas in three-dimensional space, the array antenna has time modulation characteristics, which better controls the radiation characteristics of the array antenna and achieves the best far-field radiation pattern synthesis. This paper designs a Time-modulated linear array (TMLA) with low sidelobe level (SLL) and low sideband level (SBL) based on the chaotic exchange nonlinear dandelion optimization (CENDO) algorithm. Three optimization methods are studied: firstly, determining the optimal on-time (τ nn ) for each array element; The second is to determine the optimal on-time (τ nn ) and optimal uniform array element spacing (d) for each array element; The third is to determine the optimal opening time (t on ), closing time (t off ), and optimal uniform array element spacing (d) for each array element. To achieve simultaneous reduction of sidelobe level and suppression of harmonic interference. The same array model contains different harmonic frequency radiation. In this article, we only considered two harmonic frequencies, namely the first sideband frequency and the second sideband frequency. Because the harmonic of other sideband frequencies has a very small impact on the radiation of the fundamental wave, it can be ignored. To demonstrate the stronger ability of the CENDO algorithm in optimizing Time-modulated array antennas, and in line with the principle of fairness and impartiality, this paper also simulates different Time-modulated array models and compares the results of the CENDO algorithm with other published literature. It is concluded that this study shows lower SLL and lower SBL in different models. This provides a more scientific and accurate explanation of the superiority of the CENDO algorithm compared to other algorithms in the field of antenna optimization in electromagnetics. At the same time, this also provides great research value and fundamental support for designing high-performance Time-modulated array antennas in subsequent engineering applications.

Abstract Global climate change and the collection of environmental protection taxes are accelerating the green transformation of thermal power enterprises. This study selected Chinese thermal power listed companies as samples and used a dynamic three-stage (operational, green transformation, and market performance) network DEA model to evaluate their transformation efficiency and corporate performance. This paper incorporates targeted indicators such as ESG (environment, society, governance) and stock prices into the model and conducts a comparative study on the basis of macro policies and the geographical location of the enterprise. A comparative analysis was conducted on the efficiency of enterprises before and after the adjustment of the environmental tax burden, using the environmental tax burden as an exogenous variable. Thus, the following conclusions can be drawn: there is a certain positive correlation between the collaborative efficiency of the two links of thermal power enterprises and the economic development of their respective regions. Moreover, the green transformation efficiency of most thermal power enterprises is superior to the market performance efficiency. The environmental tax burden mainly improves the overall efficiency of thermal power enterprises by improving their operational efficiency and efficiency in the green transformation stage without affecting market performance. To further improve efficiency, thermal power enterprises should actively communicate with stakeholders to strive for more financial relief.

Abstract Background Cobalt (Co) is a metal which is widely used in the industrial production. The previous studies found the toxic effects of environmental Co exposure on multiple organs. However, the correlation of blood Co concentration with lung function was inconsistent in patients with chronic obstructive pulmonary disease (COPD). Methods All 771 stable COPD patients were recruited. Peripheral blood and clinical information were collected. The levels of blood Co and serum CC16 were measured. Results Cross-sectional study suggested that the level of blood Co was inversely and dose-dependently related to lung function parameters. Each 1 ppm elevation of blood Co was related to 0.598 L decline in FVC, 0.465 L decline in FEV1, 6.540% decline in FEV1/FVC%, and 14.013% decline in FEV1%, respectively. Moreover, higher age, enrolled in winter, current-smoking, higher smoking amount, and inhaled corticosteroids prominently exacerbated the negative correlation between blood Co and lung function. Besides, serum CC16 content was gradually reduced with blood Co elevation in COPD patients. Besides, serum CC16 was positively correlated with lung function, and inversely related to blood Co. Additionally, decreased CC16 substantially mediated 11.45% and 6.37% Co-triggered downregulations in FEV1 and FEV1%, respectively. Conclusion Blood Co elevation is closely related to the reductions of pulmonary function and serum CC16. CC16 exerts a significantly mediating role of Co-related to pulmonary function decrease among COPD patients.

As a green and environmentally friendly fire gold test method for efficient separation and enrichment of trace precious metals in ore, bismuth gold test effectively avoids the problem of toxic pollution of lead test gold. In this article, Bi2O3 with low toxicity is used as the fire test gold trap of Au element, and Bi2O3 is reduced to Bi by the reducing agent flour in the test ingredient during the high-temperature melting process, and Au2Bi alloy is formed with Au in the samples, and the Ag protective ash blowing method is used to make Au and Ag form about 1mg of Ag zygote. The Ag zygoteum is heated and dissolved by acid solution method to completely enter the solution of Au. In this test, the Au element content in the national standard material GBW 07205 was used as a reference, and the effective pixels of the CCD detector of the continuous light source flame atomic absorption spectrometer were optimized, and 7 were selected as the effective pixels of the CCD detector based on their sensitivity and stability. In the mass concentration range of 0～20 μg/mL, the corresponding absorbance was fitted by the quadratic equation least squares method, the fitting coefficient of the calibration curve was 0.999 8, the characteristic concentration was 0.069 97 μg/mL, and the detection limit of the method was 0.012 7 g/mL. According to the selected experimental method and optimized instrument parameters, Au in the national standard material was determined, and the measured value was in good agreement with the standard value, and the relative standard deviation (RSD, n=6) was 2.23%～4.54%. The established method was applied to the test of Au in actual ore samples, and the recovery rate of spiked was 92.6%～106%. The relative standard deviation (n=6) was 2.53%～4.70%, which met the requirements of the National Geological and Mineral Resources Industry Standard DZ/T 0130-2006.

Abstract End turn grading with resistive–capacitive coupling experiences severe electrothermal stress when subjected to pulse width modulation (PWM) voltage. In this paper, several experiments and simulations were carried conducted for four types of end turn grading. First of all, the temperature rise in the end turn grading increased with a decrease in rise time. When the rise time was less than 500 ns, the temperature rise at the terminal was higher owing to the increased capacitive current coupled from the main wall insulation. Further, the current in the linear region exhibited minimal variation at different fundamental frequencies resulting in synchronized the temperature rise at the terminal and overlap. Furthermore, the jump voltage was the key factor influencing temperature rise in end turn grading, confirmed by comparing different voltage magnitudes. Finally, the transient behaviour of the maximum field in the stress grading material was determined at rise time. The experimental and simulation results indicate that balancing and interdependently addressing the electrical and thermal stress protection in end turn grading is crucial. The study aims to provide an experimental and theoretical foundation for an insulation system of inverter‐fed rotating machinery operating under PWM voltage.

Abstract Microflow catalysis is a cutting-edge approach to advancing chemical synthesis and manufacturing, but the challenge lies in developing efficient and stable multiphase catalysts. Here we showcase incorporating amine-containing metal-organic cages into automated microfluidic reactors through covalent bonds, enabling highly continuous flow catalysis. Two Fe4L4 tetrahedral cages bearing four uncoordinated amines were designed and synthesized. Post-synthetic modifications of the amine groups with 3-isocyanatopropyltriethoxysilane, introducing silane chains immobilized on the inner walls of the microfluidic reactor. The immobilized cages prove highly efficient for the reaction of anthranilamide with aldehydes, showing superior reactivity and recyclability relative to free cages. This superiority arises from the large cavity, facilitating substrate accommodation and conversion, a high mass transfer rate and stable covalent bonds between cage and microreactor. This study exemplifies the synergy of cages with microreactor technology, highlighting the benefits of heterogenous cages and the potential for future automated synthesis processes

Abstract Fiddler crabs, as coastal ecosystem engineers, play a crucial role in enhancing biodiversity and accelerating the flow of material and energy. Here we show how widespread crab burrows modify the carbon sequestration capacity of different habitats across a large climatic gradient. The process of crab burrowing results in the reallocation of sediment organic carbon and humus. Crab burrows can increase more greenhouse gases emissions compared to the sediment matrix (CO2: by 17–30%; CH4: by 49–141%). Straightforward calculations indicate that these increased emissions could offset 35–134% of sediment carbon burial in these two ecosystems. This research highlights the complex interactions between crab burrows, habitat type, and climate which reveal a potential lower carbon sink function of blue carbon ecosystems than previously expected without considering crab burrows.

Abstract Mutations in fibrillin 1 (FBN1) is the main cause of Marfan syndrome (MFS) with thoracic aortic aneurysm (TAA) as the main complication. Activation of the complement system plays a key role in the formation of thoracic and abdominal aortic aneurysms. However, the role of the complement system in MFS-associated aortic aneurysms remains unclear. In this study, we observed increased levels of complement C3a and C5a in the plasma of MFS patients and mouse, and the increased deposition of the activated complement system product C3b/iC3b was also observed in the elastic fiber rupture zone of 3-month-old MFS mice. The expression of C3a receptor (C3aR) was increased in MFS aortas, and recombinant C3a promoted the expression of cytokines in macrophages. The administration of a C3aR antagonist (C3aRA) attenuated the development of thoracic aortic aneurysms in MFS mice. The increased inflammation response and matrix metalloproteinases activities were also attenuated by C3aRA treatment in MFS mice. Therefore, these findings indicate that the complement C3a/C3aR inhibition alleviates the formation of aortic aneurysm in Marfan syndrome mice.

Abstract Brucella spp. are facultative intracellular pathogens that cause zoonosis- brucellosis worldwide. There has been a trend of the re-emergence of brucellosis worldwide in recent years. The epidemic situation of brucellosis is serious in Xinjiang. To analyze the epidemic situation of Brucella spp. in Xinjiang among humans and animals, this study identified 144 Brucella isolates from Xinjiang using classical identification and 16 S rRNA sequencing. MLVA, drug resistance testing, and wgSNP detection were also performed. At the same time, analysis was conducted based on the published data of Brucella isolates worldwide. The results showed that the dominant species was B. melitensis biovar 3, which belonged to GT42 (MLVA-8 typing) and the East Mediterranean lineage. The correlation among isolates was high both in humans or animals. The isolates in Xinjiang exhibited higher polymorphism compared to other locations in China, with polymorphism increasing each year since 2010. No amikacin/kanamycin-resistant strains were detected, but six rifampicin-intermediate isolates were identified without rpoB gene variation. The NJ tree of the wgSNP results indicated that there were three main complexes of the B. melitensis epidemic in Xinjiang. Based on the results of this study, the prevention and control of brucellosis in Xinjiang should focus on B. melitensis, particularly strains belonging to B. melitensis bv.3 GT42 (MLVA-8 typing) and East Mediterranean lineage. Additionally, the rifampicin- and trimethoprim-sulfamethoxazole- resistance of isolates in Xinjiang should be closely monitored to avoid compromising the therapeutic efficacy and causing greater losses. These results provide essential data for the prevention and control of brucellosis in Xinjiang and China. Although the isolates from Xinjiang have significant characteristics among Chinese isolates and can reflect the epidemiological situation of brucellosis in China to some extent, this study cannot represent the characteristics of isolates from other regions.

Abstract The establishment of discrete element method (DEM) model for simulating the behavior of viscous soil can significantly contribute to the development of agricultural machinery, particularly in hilly areas worldwide. There is limited research on the parameter calibration of the DEM for purple soil in the Southwest China and its interaction mechanism between very narrow tine. Piling up test, rotating drum test and design-expert 12.0 software were used to design Plackett–Burman test, steepest climb test and Box–Behnken test successively. Combined with soil intrinsic parameter measurement test, soil simulation parameter calibration and optimization were completed. Based on the optimized DEM simulation parameters, the average relative errors of static repose angle and dynamic repose angle were 1.934 and 2.289%, respectively. The accuracy of parameter calibration results of DEM simulation was proved. Three kinds of very narrow tines were designed and processed. The interaction model and mechanism between soil-touch parts and soil was established. The average relative errors between the DEM simulation results of the forward resistance and disturbance area of the three soil-touch parts and the soil bin test results were 7.22% and 11.24%, respectively, which further proved that the calibration results of DEM parameters of purple soil and the interaction model with the soil-touch parts in this study were reliable. The physical and interaction parameters of slope cultivated purple soil in Southwest China were determined based on static-dynamic repose angle, providing more accurate parameters for study of soil-touch parts.

RECIST 1.1 criteria play a crucial role in mitigating this variability by standardizing evaluations, aiming to establish an accepted “truth” confirmed by histology or patient survival. Clinical trials utilize Blind Independent Centralized Review (BICR) techniques to manage variability, employing double reads and adjudicators to address inter-observer discordance effectively. It is essential to dissect the root causes of variability in response assessments, with a specific focus on the factors influencing RECIST evaluations. We propose proactive measures for radiologists to address variability sources such as radiologist expertise, image quality, and accessibility of contextual information, which significantly impact interpretation and assessment precision. Adherence to standardization and RECIST guidelines is pivotal in diminishing variability and ensuring uniform results across studies. Variability factors, including lesion selection, new lesion appearance, and confirmation bias, can have profound implications on assessment accuracy and interpretation, underscoring the importance of identifying and addressing these factors. Delving into the causes of variability aids in enhancing the accuracy and consistency of response assessments in oncology, underscoring the role of standardized evaluation protocols and mitigating risk factors that contribute to variability. Access to contextual information is crucial. Abstract Introduction A dynamic molecular biomarker that can identify early efficacy of immune checkpoint inhibitor (ICI) therapy remains an unmet clinical need. Here we evaluate if a novel circulating tumor DNA (ctDNA) assay, xM, used for treatment response monitoring (TRM), that quantifies changes in ctDNA tumor fraction (TF), can predict outcome benefits in patients treated with ICI alone or in combination with chemotherapy in a real-world (RW) cohort. Methods This retrospective study consisted of patients with advanced cancer from the Tempus de-identified clinical genomic database who received longitudinal liquid-based next-generation sequencing. Eligible patients had a blood sample ≤ 40 days prior to the start of ICI initiation and an on-treatment blood sample 15–180 days post ICI initiation. TF was calculated via an ensemble algorithm that utilizes TF estimates derived from variants and copy number information. Patients with molecular response (MR) were defined as patients with a ≥ 50% decrease in TF between tests. In the subset of patients with rw-imaging data between 2 and 18 weeks of ICI initiation, the predictive value of MR in addition to rw-imaging was compared to a model of rw-imaging alone. Results The evaluable cohort (N = 86) was composed of 14 solid cancer types. Patients received either ICI monotherapy (38.4%, N = 33) or ICI in combination with chemotherapy (61.6%, N = 53). Patients with MR had significantly longer rw-overall survival (rwOS) (hazard ratio (HR) 0.4, P = 0.004) and rw-progression free survival (rwPFS) (HR 0.4, P = 0.005) than patients with molecular non-response (nMR). Similar results were seen in the ICI monotherapy subcohort; HR 0.2, P = 0.02 for rwOS and HR 0.2, P = 0.01 for rwPFS. In the subset of patients with matched rw-imaging data (N = 51), a model incorporating both MR and rw-imaging was superior in predicting rwOS than rw-imaging alone (P = 0.02). Conclusions xM used for TRM is a novel serial quantitative TF algorithm that can be used clinically to evaluate ICI therapy efficacy.

During the coal mining process, a large amount of waste such as coal gangue and coal washing water is generated. The heavy metal pollutants in those wastes migrate and diffuse into the surrounding soil and water environment through sedimentation, runoff, leaching and other processes, posing a threat to the ecological environment. In order to study the heavy metal pollution in an abandoned coal mine area and to evaluate its ecological hazard risk, the contents of Cu, Pb, Zn, As, Cr, Ni, Hg, Cd elements in the coal gangue, water and soil samples of the mining area were detected. The single factor index method and potential ecological hazard index method were used to analyze the heavy metal pollution and its potential ecological hazard risk. The heavy metal pollution in the soil downstream of the coal washing plant in the mining area was mainly studied. The improved BCR continuous extraction method was used to analyze the forms of heavy metal elements (Cu, Zn, Cd) in the soil samples and the risk assessment coding method was employed to evaluate the bioavailability and environmental risks of heavy metal elements. The results show that: ① on average, the heavy metal contents in the coal gangue of the coal mine area are relatively low, with only Cr element content being slightly higher than that in soil. Moreover, the heavy metal element contents in the coal gangue do not exceed the risk intervention values for the soil of agricultural land, which has a little impact on heavy metal pollution in the soil. ② The heavy metal contents in the mine area’ exceed the standard, including that of Cu, Zn, Cd, Ni and As elements, and the Cd pollution is relatively serious. The pollution areas of Cu, Zn, and Ni elements are mainly concentrated in the western and southeastern parts of the gangue hill, with a certain overlap in spatial distribution. The As element pollution is distributed in the northeastern part of the gangue hill, while the Cd element pollution is widely distributed in the mine area, with moderate to severe pollution. ③ The Cd and Hg elements have potential ecological hazard risk, with a higher risk for Cd element. The comprehensive potential ecological hazard risk (RI) of heavy metal elements is mainly moderate to strong. ④ The contents of heavy metal elements in the soil downstream of the coal washing plant show an overall decreasing trend along the water flow direction, with the Cd element pollution being more severe, and the Cu, Zn, Ni, As element pollution concentrated within 200 m downstream of the coal washing plant. The heavy metal element contents of the profile sample shows a decreasing trend in the vertical direction, with no accumulation in the vertical direction. ⑤ The groundwater quality in the mine area is good, and the pH and the contents of heavy metal elements in the mine water meet the limit values of Class III surface water quality standards. The discharge of mine water drainage has a little impact on heavy metal pollution in the soil. ⑥ The proportions of acid extractable Cu and Zn elements are low, with low bioavailability, while that of Cd element is high, presenting a high-extremely high risk, with strong bioavailability and high environmental risk. Based on the current situation of heavy metal pollution in the mining area, it is recommended to further strengthen the prevention and control of heavy metal pollution, strengthen the source control, monitoring of agricultural food safety, and the investigation and research on the Cd element pollution.

Abstract Diabetes accelerates vascular senescence, which is the basis for atherosclerosis and stiffness. The activation of NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome and oxidative stress are closely associated with the deteriorative senescence in endothelial cells (ECs) and vascular smooth muscle cells (VSMCs). For decades, Sodium Tanshinone IIA Sulfonate (STS) has been utilized as a cardiovascular medicine with acknowledged anti-inflammatory and anti-oxidative properties. Nevertheless, the impact of STS on vascular senescence remains unexplored in diabetes. Diabetic mice, primary ECs and VSMCs were transfected with the NLRP3 overexpression/knockout plasmid, the tumor necrosis factor alpha-induced protein 3 (TNFAIP3/A20) overexpression/knockout plasmid, and treated with STS to detect senescence-associated markers. In diabetic mice, STS treatment maintained catalase (CAT) level and vascular relaxation, reduced hydrogen peroxide probe (ROSgreen) fluorescence, p21 immunofluorescence, Senescence β-Galactosidase Staining (SA-β-gal) staining area, and collagen deposition in aortas. Mechanistically, STS inhibited NLRP3 phosphorylation (serine 194), NLRP3 dimer formation, NLRP3 expression, and NLRP3-PYCARD (ASC) colocalization. It also suppressed the phosphorylation of IkappaB alpha (IκBα) and NFκB, preserved A20 and CAT levels, reduced ROSgreen density, and decreased the expression of p21 and SA-β-gal staining in ECs and VSMCs under HG culture. Our findings indicate that STS mitigates vascular senescence by modulating the A20-NFκB-NLRP3 inflammasome-CAT pathway in hyperglycemia conditions, offering novel insights into NLRP3 inflammasome activation and ECs and VSMCs senescence under HG culture. This study highlights the potential mechanism of STS in alleviating senescence in diabetic blood vessels, and provides essential evidence for its future clinical application.

Abstract Soil extractable nitrate, ammonium, and organic nitrogen (N) are essential N sources supporting primary productivity and regulating species composition of terrestrial plants. However, it remains unclear how plants utilize these N sources and how surface-earth environments regulate plant N utilization. Here, we establish a framework to analyze observational data of natural N isotopes in plants and soils globally, we quantify fractional contributions of soil nitrate (f NO3-), ammonium (f NH4+), and organic N (f EON) to plant-used N in soils. We find that mean annual temperature (MAT), not mean annual precipitation or atmospheric N deposition, regulates global variations of f NO3-, f NH4+, and f EON. The f NO3- increases with MAT, reaching 46% at 28.5 °C. The f NH4+ also increases with MAT, achieving a maximum of 46% at 14.4 °C, showing a decline as temperatures further increase. Meanwhile, the f EON gradually decreases with MAT, stabilizing at about 20% when the MAT exceeds 15 °C. These results clarify global plant N-use patterns and reveal temperature rather than human N loading as a key regulator, which should be considered in evaluating influences of global changes on terrestrial ecosystems.
